Recent technical advances allowing affordable whole genome sequencing, as well as a better inventory of species-by-species chemical diversity, have greatly advanced our understanding as to how flavonoid biosynthesis pathways vary across species. In parallel, reverse genetics combined with detailed molecular phenotyping is currently allowing us to elucidate the functional importance of individual genes and metabolites and by this means to provide further mechanistic insight into their biological roles. Here we provide an inventory of current knowledge of pathways of flavonoid biosynthesis in both the model plant Arabidopsis thaliana and a range of crop species, including tomato, maize, rice, and bean.

Background: Gentiana lutea (GL), commonly known as yellow gentian, bitter root, and bit-terwort, belongs to family Gentianaceae. GL belongs to genus Gentiana, which is a rich natural source of iridoids, secoiridoids, xantones, flavonoids, triterpenoids, and carbohydrates. Medicinal plants from Gentiana species have anti-oxidant, anti-inflammatory, anti-mitogenic, anti-proliferative, and lipid-lowering effects, as well as a cardioprotective, hypotensive, vasodilator and anti-platelet activities. Objective: We reviewed the recent literature related to the effects of Gentiana species, and their active components on vascular diseases. Methods: Data used for this review were obtained by searching the electronic database [PUB-MED/MEDLINE 1973-February 2020]. The primary data search terms of interest were: Gentiana lutea, Gentienacea family, phytochemistry, vascular diseases, treatment of vascular diseases, anti-oxidant, anti-inflammatory, anti-atherogenic. Conclusion: Gentiana species and their constituents affect many different factors related to vascular disease development and progression. Therefore, Gentiana-based therapeutics represent potentially use-ful drugs for the management of vascular diseases. © 2021 Bentham Science Publishers.

Alterations of skin homeostasis are widely diffused in our everyday life both due to accidental injuries, such as wounds and burns, and physiological conditions, such as late-stage diabetes, dermatitis, or psoriasis. These events are locally characterized by an intense inflammatory response, a high generation of harmful free radicals, or an impairment in the immune response regulation, which can profoundly change the skin tissue’ repair process, vulnerability, and functionality. Moreover, diabetes diffusion, antibiotic resistance, and abuse of aggressive soaps and disinfectants following the COVID-19 emergency could be causes for the future spreading of skin disorders. In the last years, hydroxycinnamic acids and derivatives have been investigated and applied in several research fields for their anti-oxidant, anti-inflammatory, and anti-bacterial activities. First, in this study, we give an overview of these natural molecules’ current source and applications. Afterwards, we review their potential role as valid alternatives to the current therapies, supporting the management and rebalancing of skin disorders and diseases at different levels. Also, we will introduce the recent advances in the design of biomaterials loaded with these phenolic compounds, specifically suitable for skin disorders treatments. Lastly, we will suggest future perspectives for introducing hydroxycinnamic acids and derivatives in treating skin disorders.

Mango is one of the choicest fruits in the world because of its good flavour, attractive colour and fruity fragrance. It belongs to family Anacardiaceae having wide range of varieties and health benefits. It grows in tropical region and has good nutritional profile. Innumerable researches have been done around the world that demonstrated diversified nutritional and pharmacological benefits of Mangifera indica Linn. It has particular composition of nutrients and phytochemicals. Different parts of mango trees have different wide ranges of benefits. The flower, seeds, leaves, bark, raw as well as ripe fruits provide extensive variety of pharmacological, medicinal and many health benefits. In addition to the higher values of macronutrients as well as micronutrients, mangoes have a wide range of bioactive compounds present in different portions of plant for example, the primary active constituent present in mango is mangiferin. In this review article we reviewed a number of beneficial characteristics of mango including its antioxidant, anti-diabetic, anti-bacterial, gastric health, anti-cancer and anti-inflammatory activities.

Background/Aim: Plants play an important role in anti-cancer drug discovery, therefore, the current study aimed to evaluate the biological activity of Alpinia zerumbet (A. zerumbet) flowers. Methods: The phytochemical and biological criteria of A. zerumbet were in vitro investigated as well as in mouse xenograft model. Results: A. zerumbet extracts, specially CH2Cl2 and MeOH extracts, exhibited the highest potent anti-tumor activity against Ehrlich ascites carcinoma (EAC) cells. The most active CH2Cl2 extract was subjected to bioassay-guided fractionation leading to isolatation of the naturally occurring 5,6-dehydrokawain (DK) which was characterized by IR, MS, H-1-NMR and C-13-NMR. A. zerumbet extracts, specially MeOH and CH2Cl2 extracts, exhibited significant inhibitory activity towards tumor volume (TV). Furthermore, A. zerumbet extracts declined the high level of malonaldehyde (MDA) as well as elevated the levels of superoxide dismutase (SOD) and catalase (CAT) in liver tissue homogenate. Moreover, DK showed anti-proliferative action on different human cancer cell lines. The recorded IC50 values against breast carcinoma (MCF-7), liver carcinoma (Hep-G2) and larynx carcinoma cells (HEP-2) were 3.08, 6.8, and 8.7 mu g/mL, respectively. Conclusion: Taken together, these findings open the door for further investigations in order to explore the potential medicinal properties of A. zerumbet.

Where several investigations highlight the mechanism and kinetics of the cross-link network formation, the exclusive effect of compounding ingredients such as filler, plasticizer, anti-oxidant, etc., is still being pursued. In this direction, we report preparation and characterization of cis-1,4-polybutadiene (PB)- graphene (G) nanocomposite, and highlight a phenomenon in which graphene induces the spontaneous chemical reactions in the cis-1,4-polybutadiene elastomer matrix triggered at 170 °C temperature. Therefore, the aims of this study were to evaluate the effects of the in vitro administration of LPHs from Lupinus angustifolius on the immunological and oxidative statuses of human peripheral blood mononuclear cells (PBMCs) from 53 healthy donors. LPHs reduced PBMCs proliferation and the levels of Th1, Th9 and Th17 pro-inflammatory cytokines without being cytotoxic. LPHs also skewed the pro-/anti-inflammatory balance towards a Th2 protective response. Additionally, LPHs increased superoxide dismutase and catalase activities, and the total antioxidant capacity (TAC). This study is the first to show that LPHs reduce T cell inflammatory responses and improve the anti-inflammatory/pro-inflammatory cytokine balance and the TAC by PBMCs. Thus, LPHs may represent an effective option for developing nutritional strategies to prevent pathologies with underlying inflammation and oxidative stress.

Furthermore, A. zerumbet extracts declined the high level of malonaldehyde (MDA) as well as elevated the levels of superoxide dismutase (SOD) and catalase (CAT) in liver tissue homogenate. Moreover, DK showed anti-proliferative action on different human cancer cell lines. The recorded IC50 values against breast carcinoma (MCF-7), liver carcinoma (Hep-G2) and larynx carcinoma cells (HEP-2) were 3.08, 6.8, and 8.7 mu g/mL, respectively. Conclusion: Taken together, these findings open the door for further investigations in order to explore the potential medicinal properties of A. zerumbet.

Context: Hypertension, a global menace requires innovative research into the use of Moringa oleifera being promoted and traditionally used as alternative therapy. Objective: To innovatively evaluate the mechanistic effect, safety and efficacy of the methanol fraction of M. oleifera (MMO) leaves on L-NAME induced hypertensive rabbits. Methods: Rabbits were divided into six groups: Control, L-NAME alone, L-NAME with 100, 200 or 400 mg/kg of MMO and enalapril. Inclusion and exclusion criteria were similar baseline parameters and Day 3 systolic blood pressure (SBP) less than baseline SBP respectively. The primary outcome was a 10% reduction of SBP on Day 21. Enalapril group was excluded from analysis. Safety was assessed with liver and renal functions, hydrogen peroxide and nitric oxide concentrations to elucidate mechanistic effect. Results: Moringa 100 mg/kg, 200 mg/kg and 400 mg/kg reduced SBP by 4.75, 18.00 and 15.25 mmHg (F=22.123, p=0.000). SBP control was achieved with MMO 200mg/kg, 14% reduction and 400mg/kg, 12% reduction. Nitric oxide concentration, 0.06, 0.094 and 0.114mmol (F= 30.255, p= 0.000) dose-dependently increased and was most predictive of SBP control (r2=0.802, p=0.000). Nitric oxide production was inversely related to heart/body weight ratio which was dose-dependently reduced. MMO reduced hydrogen peroxide and ALT level but no significant effect on urea, HDL, and TG. Conclusion: MMO reduced SBP and dose-dependently increased nitric oxide concentration in L-NAME induced hypertensive rabbits. The effect may be mediated via activation of nitric oxide pathway. MMO demonstrated a potent anti-oxidant activity and safety. Effect on ventricular hypertrophy needs further evaluation.

Oxidative stress is a contributing factor to atherosclerosis. Circulating levels of plasmalogens (phospholipids with potential anti-oxidant properties) have been shown to be negatively associated with coronary artery disease, suggesting an elevated level of oxidative stress in these patients. We hypothesised that: (1) oxidative stress affects lipoprotein lipid composition and function; and (2) regulation of the level of plasmalogen can influence atherosclerosis progression and inflammation. Low density and high density lipoproteins (LDL and HDL) were oxidised with copper chloride at different time points. Liquid chromatography combined with tandem mass spectrometry analysis showed a myriad of changes in the lipid composition of lipoproteins during oxidation. Plasmalogen was one of the lipids that were most affected, early in the oxidation of lipoproteins. Incubation of THP1- derived macrophages with HDL of differing levels of oxidative stress showed that the capacity of mildly- and heavily oxidised HDL to efflux cholesterols was significantly reduced as compared to native HDL. Similarly, the ability of HDL to delay LDL oxidation and to accept oxidised lipids from oxidised LDL deteriorated progressively under mild- and heavy oxidative stress. To investigate the effect of plasmalogen enrichment in atherosclerosis, ApoE- and ApoE/glutathione peroxidase 1-deficient (ApoE-/- and ApoE-/-GPx1-/-) mice were fed a high-fat diet with or without 2% batyl alcohol (BA, precursor to plasmalogen synthesis) for 12 weeks. In this study we present an analysis of the dissemination strategies, chemical composition and nutraceutical properties of oregano. Phytochemicals present can be classified into three categories: volatile, lipids and phenolic compounds. These components present diverse neutraceutical properties, the most important of which are antioxidant activities, hypoglycemic, hypotensive, hypolypidemic and anti-carcinogenic. However, Dopa-mediated adhesive bonding is a hard act to follow in that, at least in mussels, bonding depends on Dopa in both reduced and oxidized forms, for adhesion and cohesion, respectively. Given the vulnerability of Dopa to spontaneous oxidation, the most significant challenge to using it in practical adhesion is controlling Dopa redox in a temporally- and spatially defined manner. Mussels appear to achieve such control in their byssal attachment plaques, and factors involved in redox control can be measured with precision using redox probes such as the diphenylpicryl hydrazyl (DPPH) free radical. Understanding the specifics of natural redox control may provide fundamentally important insights for adhesive polymer engineering and antifouling strategies.

The anthracycline quinone, doxorubicin (Adriamycin) is an antineoplastic agent that has substantial therapeutic activity against a broad variety of human cancers. Unfortunately, the use of this agent is limited by its cardiac toxicity, which is associated with free radical formation leading to apoptotic cell death. The goal of this work is to improve our understanding about doxorubicin induced cardiomyopathy and to identify compounds to limit doxorubicin induced cardiomyopathy. The knowledge gained here will have a generalized impact on all cardiac diseases involving oxidative stress and apoptosis. We show that doxorubicin induced apoptosis in primary neonatal rat cardiomyocytes can be attenuated by progesterone (PG). The anti-apoptotic action of PG was blocked by a progesterone receptor antagonist, Mifepristone (MF), indicating a progesterone receptor dependent pathyway. Affymetrix gene analyses found that PG treated cardiomyocytes increased the expression of 180 genes. Among the genes upregulated is NAD(P)H: Quinone Oxidoreductase-1 (NQO1) gene. NQO1 is a flavo-enzyme that can catalyze a two-electron reduction of Dox to a more stable hydroquinone, thereby acting as a defense mechanism against oxidative stress. The induction of NQO1 mRNA and NQO1 activity in cardiomyocytes was observed in a dose and time-dependent manner with PG treatment and was blocked by MF. Induction of NQO1 by b-naphoflavone, an inducer of NQO1, resulted in a decrease in caspase-3 activity. However, inhibition of NQO1 by dicoumarol did not attenuate the cytoprotective effect of PG. This data indicates that although induction of NQO1 can decrease Dox induced apoptosis, this is not the primary mechanism of cytoprotection induced by PG. Microarray analyses revealed that PG induced an increase of Bcl-XL mRNA. The overall objective of this study was to develop and characterise new normoxic polymer gel formulations for evaluation of complex 3-D treatment volumes for application in radiotherapy dosimetry. Throughout this thesis, the essential characteristics of normoxic polymer gels have been extensively investigated. Studies were performed on the chemical components of the MAGIC gel and an improved formulation was proposed. Various anti-oxidants were studied and different versions of the MAGIC gel with fewer chemicals were developed and named MAGAS and MAGAT gel dosimeters. The ascorbic acid anti-oxidant was found to have a slow oxygen scavenging rate and therefore a delay period between manufacture and irradiation of the MAGAS gel was necessary before the gel became radiation sensitive. Vacuum pumping on the MAGAS gel solution to remove dissolved oxygen was shown to initially increase the R2-dose response and sensitivity of the dosimeter, reducing the time between manufacture and irradiation. Studies of the MAGAS gel for measurement of depth dose showed that MAGAS gel has potential as a clinical radiotherapy dosimetry tool. The radiological properties of MAGIC, MAGAS and MAGAT gels were investigated. Due to their high gelatine and monomer concentration, differences with water were observed for the cross-section ratios for attenuation, energy absorption and collision stopping power coefficient ratios through the therapeutic energy range. Macro-angiopathy has alarming signs and symptoms such as those seen with stroke or heart attack, however the presentation of small vessel disease is generally more subtle and therefore usually unnoticed for a long period of time. While it may affect any organ, complications involving the nervous system such as diabetic poly-neuropathy (DPN) are especially debilitating, and it may also be a risk factor for other brain disorders such as dementia. The underlying mechanisms are likely to be multi-faceted, but piling evidence indicates oxidative stress as one of the crucial factors. Here we evaluate the oxidative profile of patients with DM2. The total anti-oxidant capacity appears to be reduced in DM2 with or without complications. Of the specific bio-markers studied, the levels of tissue-damage indicator malon-dialdehyde (MDA) were significantly lower in the DM2 + DPN population only. These results suggest that diabetic patients present with wavering oxidative status, and the low MDA concentrations in patients with complications such as DPN may represent either an exhausted anti-oxidative defense system or a response to anti-inflammatory medications. The findings may also support the use of anti-oxidants such as vitamins A and E.

Bacteria contamination by pathogenic species can be a fatal outbreak and risking worldwide population. Meanwhile, anthracnose development on bell pepper can contribute to substantial product loses which will substantially affect world economy and food availability. Current postharvest treatment such as the use of chlorine and fungicide poses harmful effects on human and environment due to the production of carcinogenic by-products. This leads to urgency to develop a safe postharvest treatment which leads to the objective of this study to develop a new postharvest treatment; ozone fumigation which has high potential to reduce bacterial contamination and anthracnose development on bell pepper. Ozone fumigation treatment is safe to human and environment and very practical. This technology is promising hence, worth to study. This study investigated the effect of ozone fumigation on 1) growth of Escherichia coli O157, Salmonella enterica sv. Typhimurium and Listeria monocytogenes; selected pathogens that contributed to food poisoning in fruit 2) development of anthracnose disease on bell pepper caused by Colletotrichum capsici 3) activity of defense related enzymes 4) antioxidant capacity of bell pepper 5) physico chemical, physiology and sensory qualities of bell pepper. Antibacterial studies was conducted on fresh cut bell pepper with treatments of 0, 1, 3, 5, 7 and 9 ppm ozone for 0.5, 3, 6 and 24 h at 18 - 20 ̊C, 95% RH. The results showed that ozone reduced growth of E. coli O157, Salmonella Typhimurium and L. monocytogenes populations where optimal dosage was 9 ppm ozone for 6 h. This ozone dosage resulted in 2.89, 2.56 and 3.06 log reduction of E. coli O157, Salmonella Typhimurium and L. monocytogenes populations, respectively.
