In 1956 Reines & Cowan discovered the neutrino using a liquid scintillator detector. The neutrinos interacted with the scintillator, producing light that propagated across transparent volumes to surrounding photo-sensors. This approach has remained one of the most widespread and successful neutrino detection technologies used since. This article introduces a concept that breaks with the conventional paradigm of transparency by confining and collecting light near its creation point with an opaque scintillator and a dense array of optical fibres. This technique, called LiquidO, can provide high-resolution imaging to enable efficient identification of individual particles event-by-event. A natural affinity for adding dopants at high concentrations is provided by the use of an opaque medium. With these and other capabilities, the potential of our detector concept to unlock opportunities in neutrino physics is presented here, alongside the results of the first experimental validation.

Liquid scintillators are a common choice for neutrino physics experiments, but their capabilities to perform background rejection by scintillation pulse shape discrimination is generally limited in large detectors. This paper describes a novel approach for a pulse shape based event classification developed in the context of the Double Chooz reactor antineutrino experiment. Unlike previous implementations, this method uses the Fourier power spectra of the scintillation pulse shapes to obtain event-wise information. A classification variable built from spectral information was able to achieve an unprecedented performance, despite the lack of optimization at the detector design level. Several examples of event classification are provided, ranging from differentiation between the detector volumes and an efficient rejection of instrumental light noise, to some sensitivity to the particle type, such as stopping muons, ortho-positronium formation, alpha particles as well as electrons and positrons. In combination with other techniques the method is expected to allow for a versatile and more efficient background rejection in the future, especially if detector optimization is taken into account at the design level.

We present a search for signatures of neutrino mixing of electron anti-neutrinos with additional hypothetical sterile neutrino flavors using the Double Chooz experiment. The search is based on data from 5 years of operation of Double Chooz, including 2 years in the two-detector configuration. The analysis is based on a profile likelihood, i.e.\ comparing the data to the model prediction of disappearance in a data-to-data comparison of the two respective detectors. The analysis is optimized for a model of three active and one sterile neutrino. It is sensitive in the typical mass range $5 \cdot 10^{-3} $ eV$^2 \lesssim \Delta m^2_{41} \lesssim 3\cdot 10^{-1} $ eV$^2$ for mixing angles down to $\sin^2 2\theta_{14} \gtrsim 0.02$. No significant disappearance additionally to the conventional disappearance related to $\theta_{13} $ is observed and correspondingly exclusion bounds on the sterile mixing parameter $\theta_{14} $ as function of $ \Delta m^2_{41} $ are obtained., Comment: accepted for publication by EPJC

Este producto de goma blanda, se ha elaborado intentando usar el máximo de ingredientes de un listado a petición de un cliente de Givaudan. Tambien se ha utilizado otros, que han estado seleccionados a través d’un estudio de mercado y de las recomendaciones del equipo técnico de Givaudan. Al final se ha obtenido un osito de goma con más de un 70% de reducción de azúcares añadidos en base a estevia., El ràpid desenvolupament de la internet ens ha donat la facilitat per obtenir informació, el que proporciona a la gent poder escollir decisions saludables més intel·ligents per tal de viure més anys. Actualment hi ha una tendència a reduir els sucres afegits degut a malalties com la diabetis, la deterioració de les cavitats bucals i l’obesitat. Això pot explicar perquè el sector de gominoles s’està encaminant per crear productes reduïts o lliures de sucres afegits, sempre que mantinguin i/o millorin les propietats del producte estàndard del mercat. Givaudan està liderant aquest canvi buscant innovadores solucions. En aquest intent, l’estudiant en pràctiques ha creat un prototip d’osset de goma amb la mitjana de sucre afegit del mercat espanyol i una altre reduint-li el sucre afegit. Aquest producte de goma tova, s’ha elaborat intentant fer servir el màxim d’ingredients d’un llistat a petició d’un client de Givaudan. També s’han utilitzat d’altres, que han estat seleccionats a través d’un estudi de mercat i de les recomanacions de l’equip tècnic de Givaudan. Nowadays, there is a reduced added sugar trend due to sugar related illnesses such as diabetes, dental cavity deterioration and obesity. This explains why the confectionary companies are working towards creating either a reduced sugar or free sugar prototype so as to maintain and improve the characteristics of the standard sugar product. Givaudan is leading this change by searching for an innovative solution. In this endeavor, the intern has created a gummy bear prototype with the standard added sugar average in the Spanish market and another gummy bear protype reducing the added sugar. To a large extent, it has been used most of the ingredients from one of Givaudan’s customer list. Other ingredients obtained through the gummy bear market research and recommended ingredients by the company technical team were also used. The end result gave a roughly 70% added sugar reduction on the gummy bear with stevia leaf extract.

Photorhabdus luminescens is an entomopathogenic bacterium found in symbiosis with the nematode Heterorhabditis. Dam DNA methylation is involved in the pathogenicity of many bacteria, including P. luminescens, whereas studies about the role of bacterial DNA methylation during symbiosis are scarce. The aim of this study was to determine the role of Dam DNA methylation in P. luminescens during the whole bacterial life cycle including during symbiosis with H. bacteriophora. We constructed a strain overexpressing dam by inserting an additional copy of the dam gene under the control of a constitutive promoter in the chromosome of P. luminescens and then achieved association between this recombinant strain and nematodes. The dam overexpressing strain was able to feed the nematode in vitro and in vivo similarly as a control strain, and to re-associate with Infective Juvenile (IJ) stages in the insect. No difference in the amount of emerging IJs from the cadaver was observed between the two strains. Compared to the nematode in symbiosis with the control strain, a significant increase in LT50 was observed during insect infestation with the nematode associated with the dam overexpressing strain. These results suggest that during the life cycle of P. luminescens, Dam is not involved the bacterial symbiosis with the nematode H. bacteriophora, but it contributes to the pathogenicity of the nemato-bacterial complex., SCOPUS: ar.j, info:eu-repo/semantics/published

Growing resistance of pathogenic bacteria and shortage of antibiotic discovery platforms challenge the use of antibiotics in the clinic. This threat calls for exploration of unconventional sources of antibiotics and identification of inhibitors able to eradicate resistant bacteria. Here we describe a different class of antibiotics, odilorhabdins (ODLs), produced by the enzymes of the non-ribosomal peptide synthetase gene cluster of the nematode-symbiotic bacterium Xenorhabdus nematophila. ODLs show activity against Gram-positive and Gram-negative pathogens, including carbapenem-resistant Enterobacteriaceae, and can eradicate infections in animal models. We demonstrate that the bactericidal ODLs interfere with protein synthesis. Genetic and structural analyses reveal that ODLs bind to the small ribosomal subunit at a site not exploited by current antibiotics. ODLs induce miscoding and promote hungry codon readthrough, amino acid misincorporation, and premature stop codon bypass. We propose that ODLs' miscoding activity reflects their ability to increase the affinity of non-cognate aminoacyl-tRNAs to the ribosome.

The yields and production rates of the radioisotopes Li-9 and He-8 created by cosmic muon spallation on C-12, have been measured by the two detectors of the Double Chooz experiment. The identical detectors are located at separate sites and depths, which means that they are subject to different muon spectra. The near (far) detector has an overburden of approximate to 120 m.w.e. (approximate to 300 m.w.e.) corresponding to a mean muon energy of 32.1 +/- 2.0 GeV (63.7 +/- 5.5 GeV). Comparing the data to a detailed simulation of the Li-9 and He-8 decays, the contribution of the He-8 radioisotope at both detectors is found to be compatible with zero. The observed Li-9 yields in the near and far detectors are 5.51 +/- 0.51 and 7.90 +/- 0.51, respectively, in units of 10(-8-1)g(-1)cm(2). The shallow overburdens of the near and far detectors give a unique insight when combined with measurements by KamLAND and Borexino to give the first multi-experiment, data driven relationship between the Li-9 yield and the mean muon energy according to the power law and Y-0 = (0.43 +/- 0.11) x 10(-8-1)g(-1)cm(2). This relationship gives future liquid scintillator based experiments the ability to predict their cosmogenic Li-9 background rates.

The Tetraconata (Pancrustacea) concept proposes that insects are more closely related to aquatic crustaceans than to terrestrial centipedes or millipedes. The question therefore arises whether insects have kept crustacean-specific genetic traits that could be targeted by specific toxins. Here we show that a toxin (nigritoxin), originally identified in a bacterial pathogen of shrimp, is lethal for organisms within the Tetraconata and non-toxic to other animals. X-ray crystallography reveals that nigritoxin possesses a new protein fold of the α/β type. The nigritoxin N-terminal domain is essential for cellular translocation and likely encodes specificity for Tetraconata. Once internalized by eukaryotic cells, nigritoxin induces apoptotic cell death through structural features that are localized in the C-terminal domain of the protein. We propose that nigritoxin will be an effective means to identify a Tetraconata evolutionarily conserved pathway and speculate that nigritoxin holds promise as an insecticidal protein.

Objective: Determine the frequency of granulomatosis with polyangiitis (GPA) associated with non-identified ANCA (non-MPO, non-PR3 ANCA) and secondarily compare their clinic with GPA associated with MPO-positive or PR3-positive ANCA. Methods: In a monocentric retrospective observational study, clinical data of 398 patients with non-identified ANCA (titer of ANCA at least 1/80 by immunofluorescence on ethanol fixed PMN) was gathered over a period of 6 years. GPA patients from this population were compared with GPA patients with identified ANCA on the basis of clinical, biological, immunological and histological features. Results: The most common diseases associated with non-identified ANCA were inflammatory bowel diseases accounting for 17% of diseases. GPA accounted for only 1.8% of cases. There were no significant differences in terms of clinical and histological characteristics between GPA with non-identified ANCA and GPA with identified ANCA, but significantly higher CRP levels were observed in GPA patients with identified ANCA (p = 0.005). Localized disease (ear, nose and throat and/or lung involvement without any other systemic involvement) was more frequent in the group of GPA with nonidentified ANCA (p = 0.047) as compared to GPA with identified ANCA. This explains that the former group of patients was less frequently treated by cyclophosphamide than the latter (p = 0.016). Conclusion: GPA with non-MPO, non-PR3 ANCAs is relatively rare. A study on cosmic muons has been performed for the two identical near and far neutrino detectors of the Double Chooz experiment, placed at $\sim$120 and $\sim$300 m.w.e. underground respectively, including the corresponding simulations using the MUSIC simulation package. This characterization has allowed to measure the muon flux reaching both detectors to be (3.64 $\pm$ 0.04) $\times$ 10$^{-4}$ cm$^{-2}$s$^{-1}$ for the near detector and (7.00 $\pm$ 0.05) $\times$ 10$^{-5}$ cm$^{-2}$s$^{-1}$ for the far one. The seasonal modulation of the signal has also been studied observing a positive correlation with the atmospheric temperature, leading to an effective temperature coefficient of $\alpha_{T}$ = 0.212 $\pm$ 0.024 and 0.355 $\pm$ 0.019 for the near and far detectors respectively. These measurements, in good agreement with expectations based on theoretical models, represent one of the first measurements of this coefficient in shallow depth installations., Comment: 20 pages, 11 figures

Because earthworms sustain soil functioning and fertility, there is a need to advance the knowledge of their adaptation potential to chemicals in anthropogenic landscapes. Our hypothesis is that there is acclimation to organic chemicals (pesticides) in earthworms that durably persist under conventional farming in anthropogenic landscapes. The adaptation capability of two populations of earthworms (Aporectodea caliginosa) having a different chemical exposure history, - one originating from 20 years of organic farming (naïve population) and another from 20 years of conventional farming (pre-exposed population) - to cope with soil organic pollutant (Opus®, epoxiconazole a worldwide used fungicide) were investigated. Several complementary metabolic and energetic endpoints were followed, and cast production was assessed as a behavioural biomarker related to earthworms ecological role for the soil. Basal metabolism reflected by respiration rate was increased in both fungicide-exposed worms compared to controls. Glycogen resources were decreased in the same proportion in the two populations but more rapidly for the naïve (7 days) than for the pre-exposed population (28 days). Soluble protein and most amino-acids contents increased in the pre-exposed population only, suggesting a detoxification mechanism. Metabolomic profiles showed a cut-off between fungicide-exposed and control groups in the pre-exposed earthworms only, with an increase in most of the metabolites. Exposure to a low dose of epoxiconazole increased cast production of pre-exposed earthworms, and this resulted in an increase in pesticide disappearance. As far as we know, this is the first study which evidenced there is an acclimation to an agricultural chemical in earthworms derived from conventional farming that also relates to a change in their burrowing behaviour, and for which larger consequences for the soil ecosystem need to be addressed. The adaptation capability of two populations of earthworms (Aporectodea caliginosa) having a different chemical exposure history, - one originating from 20 years of organic farming (naïve population) and another from 20 years of conventional farming (pre-exposed population) - to cope with soil organic pollutant (Opus®, epoxiconazole a worldwide used fungicide) were investigated. Several complementary metabolic and energetic endpoints were followed, and cast production was assessed as a behavioural biomarker related to earthworms ecological role for the soil. Basal metabolism reflected by respiration rate was increased in both fungicide-exposed worms compared to controls. Glycogen resources were decreased in the same proportion in the two populations but more rapidly for the naïve (7 days) than for the pre-exposed population (28 days). Soluble protein and most amino-acids contents increased in the pre-exposed population only, suggesting a detoxification mechanism. Metabolomic profiles showed a cut-off between fungicide-exposed and control groups in the pre-exposed earthworms only, with an increase in most of the metabolites. Exposure to a low dose of epoxiconazole increased cast production of pre-exposed earthworms, and this resulted in an increase in pesticide disappearance. As far as we know, this is the first study which evidenced there is an acclimation to an agricultural chemical in earthworms derived from conventional farming that also relates to a change in their burrowing behaviour, and for which larger consequences for the soil ecosystem need to be addressed. An antimicrobial peptide (AMP) of the cecropin family was isolated by HPLC from plasma of the insect pest, Spodoptera frugiperda. Its molecular mass is 3910.9 Da as determined by mass spectrometry. Thanks to the EST database Spodobase, we were able to describe 13 cDNAs encoding six different cecropins which belong to the sub-families CecA, CecB, CecC and CecD. The purified peptide identified as CecB1 was chemically synthesized (syCecB1). It was shown to be active against Gram-positive and Gram-negative bacteria as well as fungi. Two closely related entomopathogenic bacteria, Xenorhabdus nematophila F1 and Xenorhabdus mauleonii VC01(T) showed different susceptibility to syCecB1. Indeed, X. nematophila was sensitive to syCecB1 whereas X. mauleonii had a minimal inhibitory concentration (MIC) eight times higher. Interestingly, injection of live X. nematophila into insects did not induce the expression of AMPs in hemolymph. This effect was not observed when this bacterium was heat-killed before injection. On the opposite, both live and heat-killed X. mauleonii induced the expression of AMPs in the hemolymph of S. frugiperda. The same phenomenon was observed for another immune-related protein lacking antimicrobial activity. Altogether, our data suggest that Xenorhabdus strains have developed different strategies to supplant the humoral defense mechanisms of S. frugiperda, either by increasing their resistance to AMPs or by preventing their expression during such host-pathogen interaction. Members of the genus Xenorhabdus are entomopathogenic bacteria that associate with nematodes. The nematode-bacteria pair infects and kills insects, with both partners contributing to insect pathogenesis and the bacteria providing nutrition to the nematode from available insect-derived nutrients. The nematode provides the bacteria with protection from predators, access to nutrients, and a mechanism of dispersal. Members of the bacterial genus Photorhabdus also associate with nematodes to kill insects, and both genera of bacteria provide similar services to their different nematode hosts through unique physiological and metabolic mechanisms. We posited that these differences would be reflected in their respective genomes. To test this, we sequenced to completion the genomes of Xenorhabdus nematophila ATCC 19061 and Xenorhabdus bovienii SS-2004. As expected, both Xenorhabdus genomes encode many anti-insecticidal compounds, commensurate with their entomopathogenic lifestyle. Despite the similarities in lifestyle between Xenorhabdus and Photorhabdus bacteria, a comparative analysis of the Xenorhabdus, Photorhabdus luminescens, and P. asymbiotica genomes suggests genomic divergence. These findings indicate that evolutionary changes shaped by symbiotic interactions can follow different routes to achieve similar end points.

Vibrio splendidus, strain LGP32, is an oyster pathogen associated with the summer mortalities affecting the production of Crassostrea gigas oysters worldwide. Vibrio splendidus LGP32 was shown to resist to up to 10 mu M Cg-Def defensin and Cg-BPI bactericidal permeability increasing protein, two antimicrobial peptides/proteins (AMPs) involved in C. gigas immunity. The resistance to both oyster Cg-Def and Cg-BPI and standard AMPs (polymyxin B, protegrin, human BPI) was dependent on the ompU gene. Indeed, upon ompU inactivation, minimal bactericidal concentrations decreased by up to fourfold. AMP resistance was restored upon ectopic expression of ompU. The susceptibility of bacterial membranes to AMP-induced damages was independent of the ompU-mediated AMP resistance. Besides its role in AMP resistance, ompU proved to be essential for the adherence of V. splendidus LGP32 to fibronectin. Interestingly, in vivo, ompU was identified as a major determinant of V. spiendidus pathogenicity in oyster experimental infections. Indeed, the V. splendidus-induced oyster mortalities dropped from 56% to 11% upon ompU mutation (Kaplan Meier survival curves, P < 0.01). Moreover, in co-infection assays, the ompU mutant was out competed by the wild-type strain with competitive indexes in the range of 0.1-0.2. From this study, ompU is required for virulence of V. splendidus. Contributing to AMP resistance, conferring adhesive properties to V. splendidus, and being essential for in vivo fitness, the OmpU porin appears as an essential effector of the C. gigas/V. splendidus interaction.
