Dies sind die Hauptfragen, die ich in dieser Arbeit zu beantworten versuchen werde. Ich werde versuchen, verschiedene Bilder von statistischen Diagrammen, die aus der Open-Source-Bibliothek Wikimedia Commons stammen, zu klassfizieren. Insgesamt enthält diese Bibliothek fast 100 Millionen Medienelemente. Eine laissez - faire Politik bezüglich der Erstellung von Kategorien und dem Hochladen von Bildern, hat zu vielen unbeschrifteten oder falsch klassifizierten Bildern sowie zum Teil sehr kleinen Nischenklassen geführt. Ich werde diese Probleme angehen, indem ich Bildklassifizierungsalgorithmen auf bereinigten Datensätzen von Instanzen ausgewählter Kategorien trainiere. Ich werde daraufhin die Trainings- und Validierungssets mit variierend vielen generitren Bildern erweitern und untersuchen wie sich verschiedene Performanzmetriken in Abhängigkeit von der Anzahl bzw. dem Anteil der generierten Bilddaten verändern. Ich werde einige Ergebnisse präsentieren. I will try to label different images of statistical charts that originate from the open-source library Wikimedia Commons. In total, this library contains almost 100 million media elements. A loose policy regarding the creation of categories and the uploading of images has led to a lot of unlabeled or falsely classified images. Another challenge when labeling the images on Wikimedia Commons is that some niche classes are very small. I will tackle these problems by training image classification algorithms on cleaned data sets of selected categories. I will enhance the training and validation sets with different amounts of artificial images. While there are many exciting results, the most important one is that generative data augmentation seems to work well for major categories but not that well for niche categories, which are similar conceptually.

3. Der Einfluss der europäischen Struktur- und Investitionsfonds im Kontext des Problems der Heterogenität der Entwicklung in der Europäischen Union

Abstract: Eines der grundlegenden Prinzipien der Verträge der Europäischen Union war die Förderung einer nachhaltigen und gerechten Entwicklung ihrer Mitgliedsländer. Um Disparitäten in Wohlstand, Einkommen und Chancen anzugehen, führte die EU die Europäischen Struktur- und Investitionsfonds (ESIFs) ein, eine Reihe finanzieller Instrumente, die aus fünf Hauptprogrammen bestehen. In meiner Bachelorarbeit habe ich mich auf die Rolle der ESIFs bei der Bewältigung der Heterogenität in den Entwicklungsstufen innerhalb der EU konzentriert. Mein Ziel war es, die Wirksamkeit der ESIFs durch eine gründliche Analyse von Paneldaten zu bewerten. Meine Forschung ergab, dass sie einen positiven Einfluss haben, um Disparitäten zwischen den Mitgliedsländern zu mildern., One of the fundamental principles of the European Union’s treaties was to promote a sustainable and equitable development of its member countries. To address disparities in wealth, income, and opportunities, the EU introduced the European Structural and Investment Funds (ESIFs), a set of financial tools consisting of five main programmes. In my Bachelor’s thesis, I concentrated on the role of ESIFs in tackling the issue of heterogeneity in the levels of development within the EU. My objective was to assess the effectiveness of ESIFs by conducting a thorough panel data analysis. My research revealed that they have a favorable impact on mitigating disparities among the members.

4. Avaliação das propriedades físicas do solo, relevo e uso do solo para mapeamento de potenciais áreas para recarga de água subterrânea nas margens do Rio Mogi Guaçu

Abstract: In Brazil, Mogi Guaçu River is inserted in the one of the most important hydrogeological provinces, populous and economically developed. Although essential to freshwater sustainably manage, analysis of water recharge zones is difficult, complex, time-consuming and expensive, because require further information regarding soils and the relationship with the landscape, rainfall and vegetation. This studyaims to develop fuzzy logic models for the groundwater recharge zoning in a sub-basin located in the Mogi Guaçu River banks. The spatial variability of soil attributes was determined through ordinary kriging. For the topography, the Topographic Wetness Index (TWI) was considered. The analysis of vegetation cover was represented by the Normalized Difference Vegetation Index (NDVI). Were generated and tested models with 3 different groups of input variables: 1 –soil indexes; 2 –soil indexes + TWI; 3 –soilindexes + TWI + NDVI. The development of fuzzy logic models with input variables representing the soil, landscape and vegetation was the most effective in the complex recharge mapping, characterizing consistently the saturated hydraulic conductivity obtained from field, No Brasil, o Rio Mogi Guaçu está inserido em uma das províncias hidrogeológicas mais importantes, populosas e economicamente desenvolvidas. Embora essencial para a gestão sustentável da água doce, a análise de zonas de recarga de água é difícil, complexa, demorada e dispendiosa, porque requer informações adicionais sobre solos e a relação com a paisagem, precipitação e vegetação. Este estudo tem como objetivo desenvolver modelos de lógica fuzzy para zoneamento de áreas de recarga de água subterrânea em uma sub-bacia localizada às margens do Rio Mogi Guaçu. A variabilidade espacial dos atributos do solo foi determinada através da krigagem ordinária. Para a topografia foi considerado o Índice de Umidade Topográfica (TWI). A análise da cobertura vegetal foi representada pelo Índice de Vegetação por Diferença Normalizada (NDVI). Foram gerados e testados modelos com 3 diferentes grupos de variáveis de entrada: 1 – índices de solo; 2 – índices de solo + TWI; 3 – índices de solo + TWI + NDVI. O desenvolvimento de modelos via lógica fuzzy com variáveis de entrada representando o solo, a paisagem e a vegetação foi o mais eficaz no complexo mapeamento da recarga, caracterizando de forma consistente a condutividade hidráulica saturada obtida em campo.

7. The effects of an obesogenic diet on behavior and cognition in zebrafish (Danio rerio): Trait average, variability, repeatability, and behavioral syndromes

Abstract: The obesity epidemic, largely driven by the accessibility of ultra-processed high-energy foods, is one of the most pressing public health challenges of the 21st century. Consequently, there is increasing concern about the impacts of diet-induced obesity on behavior and cognition. While research on this matter continues, to date, no study has explicitly investigated the effect of obesogenic diet on variance and covariance (correlation) in behavioral traits. Here, we examined how an obesogenic versus control diet impacts means and (co-)variances of traits associated with body condition, behavior, and cognition in a laboratory population of ~160 adult zebrafish (Danio rerio). Overall, an obesogenic diet increased variation in several zebrafish traits. Zebrafish on an obesogenic diet were significantly heavier and displayed higher body weight variability; fasting blood glucose levels were similar between control and treatment zebrafish. During behavioral assays, zebrafish on the obesogenic diet displayed more exploratory behavior and were less reactive to video stimuli with conspecifics during a personality test, but these significant differences were sex-specific. Zebrafish on an obesogenic diet also displayed repeatable responses in aversive learning tests whereas control zebrafish did not, suggesting an obesogenic diet resulted in more consistent, yet impaired, behavioral responses. Where behavioral syndromes existed (inter-class correlations between personality traits), they did not differ between obesogenic and control zebrafish groups. By integrating a multifaceted, holistic approach that incorporates components of (co-)variances, future studies will greatly benefit by quantifying neglected dimensions of obesogenic diets on behavioral changes.

8. Vergleich von Vorhersagemodellen zu Stornierungen von Hotelbuchungen

Abstract: In dieser Arbeit werden Stornierungen von Hotelbuchungen mithilfe verschiedener Methoden maschinellen Lernens vorhergesagt. Buchungsstornierungen stellen für die Hotelindustrie ein Problem dar, da sie die Planbarkeit von Einnahmen und Kosten stark erschweren. Unter Verwendung von logistischen Regressionen, Random Forests und künstlichen neuronalen Netzen werden Modelle erstellt, die diese Stornierungen vorhersagen sollen. Diese verschiedenen Modelle werden anschließend miteinander verglichen, um die effektivste Methode zu finden. Vorhersagemodelle könnten bestehende Probleme, die durch Stornierungen entstehen, lösen, in dem Maßnahmen wie Überbuchungen effektiver eingesetzt werden könnten. Bei der Analyse wird ein Datensatz, der von zwei portugiesischen Hotels stammt, mit fast 120.000 realen Beobachtungen und 31 Variablen zu Hilfe genommen., The aim of this thesis is to predict hotel booking cancellations with different machine learning methods. Hotel booking cancellations are a problem for hotels, because they diminish revenue and make it harder to plan future income. Using logistic regressions, Random Forests and artificial neural networks different prediction models are computed. The models are being compared to eachother to find the most effective method. These models could solve existing problems caused by cancellation, because measures such as overbooking could be used more efficiently. The analysis is based on a real dataset from two portuguese hotels and has almost 120,000 observations and 31 variables.

Abstract: Aims Marfan syndrome (MFS) is a connective tissue disorder caused by mutations in the Fibrillin-1 gene. It is associated with formation of thoracic aortic aneurysms that can potentially be a life-threatening condition due to aortic rupture or dissection. Excessive non-canonical transforming growth factor beta signalling, mediated by activation of extracellutar signal-regulated kinases 1/2 (ERK1/2), as well as inducible nitric oxide synthase (NOS2)-dependent nitric oxide production, have been identified to drive aortic pathology in MFS through induction of elastin fragmentation and smooth muscle cell apoptosis. Despite promising results in animal studies, specific pharmacological interventions approved for clinical use in patients with MFS-related aortic disease are rare. Nitro-oleic acid (NO2-OA) is an endogenously generated signalling modulator, which is available as an oral compound and has been shown to inhibit ERK1/2 activation and NOS2 expression in different disease models, thereby exerting promising therapeutic effects. In this study, we investigated whether NO2-OA decreases aortic dilation in MFS. Methods and results Eight-week-old MFS (Fbn1(C1041G+)) mice were treated with NO2-OA or vehicle for 4 weeks via subcutaneously implanted osmotic minipumps. Echocardiography indicated progressive ascending aortic dilation and wall stiffening in MFS mice, which was significantly attenuated by NO2-OA treatment. This protective effect was mediated by inhibition of aortic ERK1/2, Smad2 as well as nuclear factor kappa B overactivation and consequent attenuation of etastin fragmentation by matrix metattoproteinase 2, apoptosis, and collagen deposition. Critically, the therapeutic efficacy of NO2 -OA in MFS was further emphasized by demonstrating its capability to reduce lethal aortic complications in Fbn1(C1041G/+) mice challenged with Angiotensin II. Abstract: We sought to unravel pathomechanisms of the transition of maladaptive right ventricular (RV) remodeling to right heart failure (RHF) upon pressure overload. Exposure of C57BL/6J and C57BL/6N mice to pulmonary artery banding disclosed a tight relation of structural remodeling with afterload, but a dissociation from RV systolic function. Reduced release of mitochondrial reactive oxygen species in C57BL/6J mice prevented the development of RHF. In patients with left heart failure, increased oxidative damage in RV sections was asso-ciated with severely impaired RV function. 11. Domain-informed graph neural networks: a quantum chemistry case study

Abstract: We explore different strategies to integrate prior domain knowledge into the design of a deep neural network (DNN). We focus on graph neural networks (GNN), with a use case of estimating the potential energy of chemical systems (molecules and crystals) represented as graphs. We integrate two elements of domain knowledge into the design of the GNN to constrain and regularise its learning, towards higher accuracy and generalisation. First, knowledge on the existence of different types of relations (chemical bonds) between atoms is used to modulate the interaction of nodes in the GNN. Second, knowledge of the relevance of some physical quantities is used to constrain the learnt features towards a higher physical relevance using a simple multi-task paradigm. We demonstrate the general applicability of our knowledge integrations by applying them to two architectures that rely on different mechanisms to propagate information between nodes and to update node states.

12. Duration of toothbrushing with fixed appliances: a randomized crossover clinical trial

Abstract: OBJECTIVE Fixed orthodontic appliances might result in dental adverse effects, if sufficient oral hygiene is not maintained. Since recommendations on toothbrushing times are lacking scientific justification in orthodontic populations, the aim of the present study was to comparatively quantify the duration of toothbrushing with a manual and a powered toothbrush in two distinct age groups. TRIAL DESIGN Two-centre, two-period crossover balanced randomized clinical trial with computer-generated numbers and blinding at the outcome-assessing level. METHODS Children and adults with fixed appliances with at least 22 aligned teeth were recruited at a university clinic and private practice in Bavaria, Germany. Plaque was disclosed with staining tablets and participants brushed their teeth with their habitual technique in front of a mirror and without interference until staining was, to their own satisfaction, completely removed. The primary outcome was duration of toothbrushing in seconds. Secondary outcomes were percentage plaque reduction and questionnaire data on dental attitudes and behaviours. Data were analysed with linear mixed models. RESULTS Twenty-eight participants were randomized to the first and 27 to the second toothbrush sequence. The combined effect of age and toothbrush on the duration of toothbrushing was evident (n = 52, F(1, 50.8) = 5.1, P = 0.028). Children brushed 14.5 seconds less [n = 28, 95% confidence interval (CI) = -31.3 to 2.3, P = 0.090], yet adults 13.3 seconds more (n = 24, 95% CI = -4.6 to 31.3, P = 0.143) with the manual than powered toothbrush. Brushing times ranged from 2 minutes and 45 seconds to 3 minutes and 17 seconds with plaque reduction at 76% and good dental attitudes and behaviours. CONCLUSIONS Both children and adults with fixed appliances might equally reach an effective plaque reduction with either manual or powered toothbrushes and their habitual brushing technique in around three minutes, if plaque is visible. Abstract: This essay enfolds the conceptual and practical steps towards an open access project using the example of the digital version of the international art and photo journal Camera Work at the Heidelberg University Library. It problematizes digital publication in a time when clear transnational rules for digital projects are still lacking and gives suggestions towards possible solutions such as the practice of fair use for non-profit scholarly materials. The text also serves as the editorial for the journal’s digital facsimile and opens up a new understanding of Camera Work on a global scale, as a material object on the threshold between analogue and digital photography.

Abstract: Enhanced signaling via RTKs in pulmonary hypertension (PH) impedes current treatment options because it perpetuates proliferation and apoptosis resistance of pulmonary arterial smooth muscle cells (PASMCs). Here, we demonstrated hyperphosphorylation of multiple RTKs in diseased human vessels and increased activation of their common downstream effector phosphatidylinositol 3 '-kinase (PI3K), which thus emerged as an attractive therapeutic target. Systematic characterization of class IA catalytic PI3K isoforms identified p110 alpha as the key regulator of pathogenic signaling pathways and PASMC responses (proliferation, migration, survival) downstream of multiple RTKs. Smooth muscle cell-specific genetic ablation or pharmacological inhibition of p110 alpha prevented onset and progression of pulmonary hypertension (PH) as well as right heart hypertrophy in vivo and even reversed established vascular remodeling and PH in various animal models. These effects were attributable to both inhibition of vascular proliferation and induction of apoptosis. Since this pathway is abundantly activated in human disease, p110 alpha represents a central target in PH.

Abstract: Nitro-oleic acid (NO2-OA), a nitric oxide (NO)- and nitrite (NO2-)-derived electrophilic fatty acid metabolite, displays anti-inflammatory and anti-fibrotic signaling actions and therapeutic benefit in murine models of ischemia-reperfusion, atrial fibrillation, and pulmonary hypertension. Muscle LIM protein-deficient mice (Mlp(-/-)) develop dilated cardiomyopathy (DCM), characterized by impaired left ventricular function and increased ventricular fibrosis at the age of 8 weeks. This study investigated the effects of NO2-OA on cardiac function in Mlp(-/-) mice both in vivo and in vitro. Mlp(-/-) mice were treated with NO2-OA or vehicle for 4 weeks via subcutaneous osmotic minipumps. Wildtype (WT) littermates treated with vehicle served as controls. Mlp(-/-) mice exhibited enhanced TGF beta signalling, fibrosis and severely reduced left ventricular systolic function. NO2-OA treatment attenuated interstitial myocardial fibrosis and substantially improved left ventricular systolic function in Mlp(-/-) mice. In vitro studies of TGF beta-stimulated primary cardiac fibroblasts further revealed that the anti-fibrotic effects of NO2-OA rely on its capability to attenuate fibroblast to myofibroblast transdifferentiation by inhibiting phosphorylation of TGF beta downstream targets. In conclusion, we demonstrate a substantial therapeutic benefit of NO2-OA in a murine model of DCM, mediated by interfering with endogenously activated TGF beta signaling.

Abstract: The six-transmembrane protein of prostate 2 (Stamp2) acts as an anti-inflammatory protein in macrophages by protecting from overt inflammatory signaling and Stamp2 deficiency accelerates atherosclerosis in mice. Herein, we describe an unexpected role of Stamp2 in polymorphonuclear neutrophils (PMN) and characterize Stamp2's protective effects in myocardial ischemic injury. In a murine model of ischemia and reperfusion (I/R), echocardiography and histological analyses revealed a pronounced impairment of cardiac function in hearts of Stamp2-deficient- (Stamp2(-/-)) mice as compared to wild-type (WT) animals. This difference was driven by aggravated cardiac fibrosis, as augmented fibroblast-to-myofibroblast transdifferentiation was observed which was mediated by activation of the redox-sensitive p38 mitogen-activated protein kinase (p38 MAPK). Furthermore, we observed increased production of reactive oxygen species (ROS) in Stamp2(-/-) hearts after I/R, which is the likely cause for p38 MAPK activation. Although myocardial macrophage numbers were not affected by Stamp2 deficiency after I/R, augmented myocardial infiltration by polymorphonuclear neutrophils (PMN) was observed, which coincided with enhanced myeloperoxidase (MPO) plasma levels. Primary PMN isolated from Stamp2(-/-) animals exhibited a proinflammatory phenotype characterized by enhanced nuclear factor (NF)-kappa B activity and MPO secretion. To prove the critical role of PMN for the observed phenotype after I/R, antibody-mediated PMN depletion was performed in Stamp2(-/-) mice which reduced deterioration of LV function and adverse structural remodeling to WT levels. These data indicate a novel role of Stamp2 as an anti-inflammatory regulator of PMN and fibroblast-to-myofibroblast transdifferentiation in myocardial I/R injury.

Abstract: The concept of risk has received scholarly attention from a variety of angles in the social, technical, and natural sciences. However, public policy scholars have not yet generated a comprehensive overview, shared understanding and conceptual framework of the main problem-solving approaches applied by governments in coping with risks. In this regard, our main aim is to examine existing perspectives on prevailing risk coping strategies, find a common denominator among them and contribute to current policy and risk science literature through providing a conceptual framework that systematically spans the spectrum of risk coping strategies and incorporates the essence of the most relevant insights. To this end, we first examine the concept of risk in-depth by exploring various definitions and types of risk. We then review different approaches proposed by different strands of research for addressing risk. Finally, we assess current knowledge and develop an amalgamated perspective for examining how risks can be addressed by classifying them into six general types of response (no response; prevention; control; precaution; toleration; and adaptation) as well as indicators to identify these responses. Extinction phase was identical to acquisition, but without US delivery. Importantly, for Study 1 these phases were conducted on one day, while for Study 2 on two separated days. Successful fear acquisition was indicated by aversive ratings and startle potentiation to CS+ versus CS− in both studies. Interestingly, participants stressed 10 days earlier showed impaired extinction on the implicit level (startle potentiation to CS+ vs. CS−) in Study 1 and only in the acquisition context on the explicit level (aversive ratings for CS+ vs. CS−) in Study 2. In sum, distal stress may strengthen later acquired fear memories and thereby impair fear extinction. This finding could have clinical implications, showing that prior stress exposure sensitizes later aversive processing and impairs therapy.

Abstract: Nitro-fatty acids are electrophilic anti-inflammatory mediators which are generated during myocardial ischemic injury. Whether these species exert anti-arrhythmic effects in the acute phase of myocardial ischemia has not been investigated so far. Herein, we demonstrate that pretreatment of mice with 9- and 10-nitro-octadec-9-enoic acid (nitro-oleic acid, NO2-OA) significantly reduced the susceptibility to develop acute ventricular tachycardia (VT). Accordingly, epicardial mapping revealed a markedly enhanced homogeneity in ventricular conduction. NO2-OA treatment of isolated cardiomyocytes lowered the number of spontaneous contractions upon adrenergic isoproterenol stimulation and nearly abolished ryanodine receptor type 2 (RyR2)-dependent sarcoplasmic Ca2+ leak. NO2-OA also significantly reduced RyR2-phosphorylation by inhibition of increased CaMKII activity. Thus, NO2-OA might be a novel pharmacological option for the prevention of VT development.

Herein, we demonstrate that pretreatment of mice with 9- and 10-nitro-octadec-9-enoic acid (nitro-oleic acid, NO2-OA) significantly reduced the susceptibility to develop acute ventricular tachycardia (VT). Accordingly, epicardial mapping revealed a markedly enhanced homogeneity in ventricular conduction. NO2-OA treatment of isolated cardiomyocytes lowered the number of spontaneous contractions upon adrenergic isoproterenol stimulation and nearly abolished ryanodine receptor type 2 (RyR2)-dependent sarcoplasmic Ca2+ leak. NO2-OA also significantly reduced RyR2-phosphorylation by inhibition of increased CaMKII activity. Thus, NO2-OA might be a novel pharmacological option for the prevention of VT development.

Abstract: We present a type-based analysis ensuring memory safety and object protocol completion in the Java-like language Mungo. Objects are annotated with usages, typestates-like specifications of the admissible sequences of method calls. The analysis entwines usage checking, controlling the order in which methods are called, with a static check determining whether references may contain null values. The analysis prevents null pointer dereferencing and memory leaks and ensures that the intended usage protocol of every object is respected and completed. The type system has been implemented in the form of a type checker.

Abstract: Solution-processable two-dimensional (2D) semiconductors with chemically tunable thickness and associated tunable band gaps are highly promising materials for ultrathin optoelectronics. Here, the properties of free charge carriers and excitons in 2D PbS nanosheets of different thickness are investigated by means of optical pump-terahertz probe spectroscopy. By analyzing the frequency-dependent THz response, a large quantum yield of excitons is found. The scattering time of free charge carriers increases with nanosheet thickness, which is ascribed to reduced effects of surface defects and ligands in thicker nanosheets. The data discussed provide values for the DC mobility in the range 550-1000 cm2 V-1 s-1 for PbS nanosheets with thicknesses ranging from 4 to 16 nm. Abstract: Cardiac dysfunctions dramatically increase with age. Revealing a currently unknown contributor to cardiac ageing, we report the age-dependent, cardiac-specific accumulation of the lysosphingolipid sphinganine (dihydrosphingosine, DHS) as an evolutionarily conserved hallmark of the aged vertebrate heart. Mechanistically, the DHS-derivative sphinganine-1-phosphate (DHS1P) directly inhibits HDAC1, causing an aberrant elevation in histone acetylation and transcription levels, leading to DNA damage. Accordingly, the pharmacological interventions, preventing (i) the accumulation of DHS1P using SPHK2 inhibitors, (ii) the aberrant increase in histone acetylation using histone acetyltransferase (HAT) inhibitors, (iii) the DHS1P-dependent increase in transcription using an RNA polymerase II inhibitor, block DHS-induced DNA damage in human cardiomyocytes. Importantly, an increase in DHS levels in the hearts of healthy young adult mice leads to an impairment in cardiac functionality indicated by a significant reduction in left ventricular fractional shortening and ejection fraction, mimicking the functional deterioration of aged hearts. These molecular and functional defects can be partially prevented in vivo using HAT inhibitors. Together, we report an evolutionarily conserved mechanism by which increased DHS levels drive the decline in cardiac health.

Abstract: A future upgrade of the European XFEL (E-XFEL) foresees an additional CW operation mode, which will increase the flexibility in the photon beam time structure [1, 2, 3]. One of the challenges of this operational mode is the need for a CW operating photo injector. We believe that using an SRF gun is the preferred approach as the beam parameters of normal conducting pulsed guns can be potentially met by SRF guns operating CW. For more than a decade DESY, in collaboration with TJNAF, NCBJ, BNL, HZB and HZDR, has performed R&D to develop an all superconducting RF gun with a lead cathode. In the frame of E-XFEL CW upgrade feasibility studies, the SRF-gun R&D program gained more attention and support. Within the next few years we would like to demonstrate the performance of the all superconducting injector required for the E-XFEL upgrade. The selected approach offers advantages w.r.t. the cleanliness of the superconducting surface, but requires a complete disassembly of a cryostat and stripping the gun cavity in a clean room to exchange the cathode. Thus it is practical only when the life time of the cathode is at least several months. In this paper we present the actual status of the R&D program, next steps and the longer term plans.

Abstract: The last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles (“MISEV”) guidelines for the field in 2014. We now update these “MISEV2014” guidelines based on evolution of the collective knowledge in the last four years. An important point to consider is that ascribing a specific function to EVs in general, or to subtypes of EVs, requires reporting of specific information beyond mere description of function in a crude, potentially contaminated, and heterogeneous preparation. For example, claims that exosomes are endowed with exquisite and specific activities remain difficult to support experimentally, given our still limited knowledge of their specific molecular machineries of biogenesis and release, as compared with other biophysically similar EVs. The MISEV2018 guidelines include tables and outlines of suggested protocols and steps to follow to document specific EV-associated functional activities. Abstract: The last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles ("MISEV") guidelines for the field in 2014. We now update these "MISEV2014" guidelines based on evolution of the collective knowledge in the last four years. An important point to consider is that ascribing a specific function to EVs in general, or to subtypes of EVs, requires reporting of specific information beyond mere description of function in a crude, potentially contaminated, and heterogeneous preparation. For example, claims that exosomes are endowed with exquisite and specific activities remain difficult to support experimentally, given our still limited knowledge of their specific molecular machineries of biogenesis and release, as compared with other biophysically similar EVs. The MISEV2018 guidelines include tables and outlines of suggested protocols and steps to follow to document specific EV-associated functional activities. Finally, a checklist is provided with summaries of key points.
