Acmella radicans (Asteraceae) is a plant native to America. Despite it having medicinal attributes, studies on its phytochemical properties are scarce, and biotechnological studies do not exist for this species. In this study, we established an adventitious root culture from A. radicans internodal segments in shake flasks with indole-3-butyric acid (IBA), and then elicited it with jasmonic acid (JA) and salicylic acid (SA). The total phenolic content and antioxidant activity were evaluated, and a comparison was made using in vitro plantlets and wild plants. Internodal segments with 0.1 mg/L IBA showed 100% root induction and exhibited better growth after transfer to shake flasks with MS liquid culture medium. JA had a significant effect on biomass increase compared to unelicited roots, mainly with 50 µM JA (28%), while SA did not show significant results. Root elicited with 100 µM (SA and JA) showed a 0.34- and 3.9-fold increase, respectively, in total phenolic content (TPC) compared to the control. The antioxidant activity was also significant, and a lower half-maximal inhibitory concentration (IC 50 ) was observed as the AJ concentration increased. Roots elicited with AJ (100 µM) exhibited high antioxidant activity with DPPH (IC 50 = 9.4 µg/mL) and ABTS (IC 50 = 3.3 µg/mL) assays; these values were close to those for vitamin C (IC 50 = 2.0 µg/mL). The TPC and antioxidant activity of in vitro plants and root cultured in shake flasks showed the lowest values in most cases; even the root cultures without elicitation were better than those of a wild plant. In this study, we demonstrated that A. radicans root culture is capable of producing secondary metabolites, while its production and antioxidant activity can be enhanced using jasmonic acid.

Background: One of the main recommendations of the Paleo Diet is that individuals replicate the whole-diet macronutrient ranges of hunter-gatherer diets. These are suggested to be 19%-35% protein, 22%-40% carbohydrate, and 28%-58% fat, by energy. However, the plant food contribution to these ranges was estimated exclusively from Australian data, which is a potential problem., Objectives: We investigated whether estimates of the contribution of protein, carbohydrate, and fat to hunter-gatherer diets are impacted by using plant data from other regions of the world., Methods: The values that form the basis of the Paleo Diet' s recommended macronutrient ranges were generated with a set of equations. We combined these equations with published plant macronutrient data from a multi-region sample of 5 hunter-gatherer groups to generate new estimated macronutrient percentages and then statistically compared the old and new estimates. Subsequently, we collated plant macronutrient data for a sample of 10 hunter-gatherer groups from several regions and repeated the exercise., Results: The whole-diet macronutrient percentages we calculated are significantly different from those that underpin the Paleo Diet's recommendations. Additionally, the ranges derived from our whole-diet macronutrient percentages (14%-35% protein, 21%-55% carbohydrate, 12%-58% fat) are markedly wider than those recommended by the Paleo Diet., Conclusions: The estimated whole-diet macronutrient percentages that form the basis of the Paleo Diet's macronutrient recommendations are not robust. Using plant data from multiple regions leads to significantly different estimates. Additionally, the macronutrient ranges derived from our whole-diet macronutrient percentages overlap with those recommended by the US Department of Agriculture and the WHO. This undercuts one of the main justifications for adopting the Paleo Diet-namely that because it is vastly different from Western diets, it can reduce the probability of experiencing noncommunicable diseases. Nutritional, soluble carbohydrates and aroma volatile profiles of Tunisian wild Swiss chard leaves (Beta vulgaris L. var. cicla) have been characterized. The chemical composition of an ethanol chard leaves extract, as well as its in vitro antioxidant, α-amylase and α-glucosidase inhibition activities were carefully evaluated. The results of the proximate composition showed that total carbohydrate fraction, mainly as dietary fiber were the major macronutrient (2.43 g/100 g fw), being the insoluble dietary fiber the predominant fraction (2.30 g/100 g). In addition, leaves of Beta vulgaris L. were especially rich in Mg, Fe and Ca (4.54, 2.94 and 2.28 mg/100 g fw) and very poor in Na (0.09 mg/100 g fw). Volatile profile revealed that non-terpene derivatives and sesquiterpene hydrocarbons were the essential classes of volatiles in the chard leaves. Myricitrin, p-cumaric acid and rosmarinic acid were characterized. Moreover, the ethanol extract of wild Swiss chard leaves revealed significant antioxidant capacity. Furthermore, a good enzyme inhibitory effects on α-glucosidase and α-amylase activities were observed. Wadi Maryut Lake is one of the remaining parts of the ancient Lake Mareotis and is not mentioned in the scientific literature. For the first time, physical and chemical characteristics as well as elemental concentrations in sediment, water, soil, and plants were determined. The lowest metal pollution indices are in the northeastern end of the lake and tend to increase toward the other end. In lake aquatic environment, Al, Fe, K, Mn, Na, B, and Cr are more likely to exist in insoluble form in the southwestern part and in soluble form in the northeastern part. Using different approaches to assess sediment contamination demonstrates that sediments can be categorized as unpolluted. However, the geo-accumulation index suggests that two locations have low anthropogenic influence of Pb and the enrichment factors and the degree of contamination indicate that Co and Pb may be enriched in sediment of some locations. Comparisons with consensus-based sediment quality guidelines revealed that no sample exceeded the probable effect concentration for Cr, Cu, Ni, Pb, and Zn. The most mobile elements in sediment-water and soil-plant systems are Na, K, and Pb. In sediment-water system, Al, Fe, and Mn oxides have the major influence on scavenging of B, K, and Cr. Similarity in the occurrences of Mn, K, and Fe in soil and Mn and Fe in wild plant was found. This work demonstrates the state of decontamination of the lake and confirms its importance as a reference and comparative case for south Mediterranean coastal water bodies.
