An extended calving interval (CInt) by extending the voluntary waiting period (VWP) could be associated with altered metabolism in dairy cows. The aim of this study was first to evaluate the effects of VWP on metabolism and body condition during the first 305 d after the first calving in the experiment (calving 1), around the end of the VWP, and during pregnancy (280 d before calving 2). Second, the effects of the VWP on metabolism were determined from 2 wk before until 6 wk after calving 2. Third, individual cow characteristics were used to predict milk production and body condition of cows after different VWP. Holstein-Friesian cows (N = 154, 41 primiparous [PP], 113 multiparous [MP]) were blocked for parity, milk production, and lactation persistency, randomly assigned to a VWP of 50, 125, or 200 d (VWP50, VWP125, or VWP200) and followed from calving 1 until 6 wk after calving 2. In the first 6 wk after calving 1 and from 2 wk before until 6 wk after calving 2, weekly plasma samples were analyzed for nonesterified fatty acids (NEFA), β-hydroxybutyrate, glucose, insulin, and insulin-like growth factor 1 (IGF-1). From wk 7 after calving 1 until 2 wk before calving 2, insulin and IGF-1 were analyzed every 2 wk. Fat- and protein-corrected milk (FPCM) and body weight (BW) gain were measured weekly. Cows were divided in two parity classes based on calving 1 (PP and MP) and remained in these classes after calving 2. During pregnancy, MP cows in VWP200 had greater plasma insulin and IGF-1 concentration and lower FPCM compared with MP cows in VWP125 (insulin: 18.5 vs. 13.9 µU/mL, CI 13.0-19.7, P < 0.01; IGF-1: 198.5 vs. 175.3 ng/mL ± 5.3, P = 0.04; FPCM: 22.6 vs. 30.0 kg/d ± 0.8, P < 0.01) or VWP50 (insulin: 15.8 µU/mL, P < 0.01; IGF-1: 178.2 ng/mL, P < 0.01; FPCM: 26.6 kg/d, P < 0.01) and had a greater daily BW gain compared with cows in VWP50 (3.6 vs. 2.5 kg/d ± 0.2; P < 0.01). After calving 2, MP cows in VWP200 had greater plasma NEFA concentration (0.41 mmol/liter) compared with MP cows in VWP125 (0.30 mmol/liter, P = 0.04) or VWP50 (0.26 mmol/liter, P < 0.01). For PP cows, the VWP did not affect FPCM or body condition during the first lactation in the experiment, or metabolism after calving 2. Independent of the VWP, higher milk production and lower body condition before insemination were associated with higher milk production and lower body condition at the end of the lactation. Natural autoantibodies (NAAb) have a role in maintaining physiological homeostasis and prevention of infections, and have been found in mammalian species tested so far. Albeit NAAb levels rise with age, little is known about the origin, function, regulation and initiation of NAAb in young animals. The present study addressed the presence of IgM and IgG NAAb binding glutamate dehydrogenase (GD), carbonic anhydrase (CA), myosin (MYO) and transferrin (TRANS) from before drinking colostrum until the first 12 weeks of life in plasma of female calves. In addition, NAAb to these four self-antigens were also measured in colostrum and in plasma of their mothers during three weeks before calving. Titers of NAAb binding GD, CA, MYO and TRANS were detected in plasma of cows before calving, in colostrum, and in plasma of calves before and after drinking of colostrum. Levels of NAAb in colostrum were positively related with levels of NAAb in plasma of cows. Before colostrum intake, levels of NAAb in plasma of calves were not related with levels of NAAb in plasma of their mother but were influenced by parity of their mother. After colostrum intake, levels of NAAb in plasma of calves in the first week of life were positively related with levels of NAAb in colostrum. Low NAAb levels in colostrum were related with low NAAb in plasma of calves in the first week of life, but after two weeks of life the relation between colostrum and plasma of calves was absent. In conclusion, NAAb are already present in the unborn calf, and levels of neonatal NAAb during the early weeks of life are affected by levels of maternal NAAb obtained via colostrum., (Copyright © 2016 Elsevier B.V. In earlier studies, natural antibodies (NAb) were related not only to the energy balance (EB) of dairy cows, but also to somatic cell count (SCC) and clinical mastitis (CM). The first objective of our study was to evaluate the effects of dry period length and dietary energy source on titers of NAb binding keyhole limpet hemocyanin (KLH) and lipopolysaccharide (LPS) in plasma and milk, SCC and CM occurrence in dairy cows in two subsequent lactations. Our second objective was to study the relationship between NAb levels and mammary health. Holstein-Friesian dairy cows (N=167) were randomly assigned to three dry period lengths (0, 30 or 60-d) and two early lactation rations (glucogenic or lipogenic). Treatments were repeated during two subsequent lactations (years 1 and 2). In year 2, 19 cows which were planned to have 0-d dry period dried off naturally and were assigned to an additional group 0→30-d dry period. In year 1, cows with a 0-d dry period had a higher SCC, a higher titer of immunoglobulin G (IgG) binding LPS in plasma, and higher titers of IgG and IgM binding KLH and LPS in milk compared with cows with a 30-d or 60-d dry period. In year 2, cows with a 60-d dry period had a lower SCC than cows with a 30-d and 0→30-d dry periods. In year 2, dry period length did not affect NAb titers in plasma or milk. The CM occurrence was 17 percent in year 1 of the experiment and 25 percent in year 2, and did not differ according to dry period lengths or rations. For both years, an increasing titer of IgG binding LPS in plasma was associated with decreased odds of a high SCC and decreased odds of CM occurrence. Also up to three weeks before the CM occurrence, an increasing titer of IgM binding KLH and LPS in plasma was associated with a decreased odds of CM occurrence. In conclusion, omitting the dry period increased SCC, NAb titers in milk and IgG binding LPS in plasma compared with a short (30-d) or conventional (60-d) dry period. The effects on NAb titers, however, were only present in the first year after omitting the dry period and disappeared after repeated omitting the dry period. The objectives of the present study were to evaluate the effects of dry period length on ovarian activity in cows fed a lipogenic or a glucogenic diet within 100 days in milk (DIM) and to determine relationships between ovarian activity and energy balance and metabolic status in early lactation. Holstein-Friesian dairy cows (n = 167) were randomly assigned to one of three dry period lengths (0, 30, or 60 days) and one of two diets in early lactation (glucogenic or lipogenic diet) resulting in a 3 × 2 factorial design. Cows were monitored for body condition score, milk yield, dry matter intake, and energy balance from calving to week 8 postpartum, and blood was sampled weekly from 95 cows from calving to week 8 postpartum. Milk samples were collected three times a week until 100 DIM postpartum for determination of progesterone concentration. At least two succeeding milk samples with progesterone concentration of 2 ng/mL or greater were used to indicate the occurrence of luteal activity. Normal resumption of ovarian cyclicity was defined as the onset of luteal activity (OLA) occurring at 45 DIM or less, followed by regular ovarian cycles of 18 to 24 days in length. Within 100 DIM postpartum, cows with a 0-day dry period had greater incidence of normal resumption of ovarian cyclicity (53.2%; 25 out of 47 cows) compared with cows with a 60-day dry period (26.0%; 13 out of 50 cows, P = 0.02). Independent of dry period length or diet, cows with OLA at less than 21 DIM had a greater body condition score during weeks 1 and 2 (P = 0.01) and weeks 1 through 8 (P = 0.01) postpartum compared with cows with OLA at greater than 30 DIM. Cows with the first ovarian cycle of medium length (18-24 days) had greater energy balance (P = 0.03), plasma concentrations of insulin (P = 0.03), glucose (P = 0.04), and insulin-like growth factor I (P = 0.04) than cows with long ovarian cycle lengths (>24 days) but had lower plasma β-hydroxybutyrate (P < 0.01) and liver triacylglycerol (P = 0.02) concentrations than cows with short ovarian cycle lengths (<18 days) during weeks 1 through 8 postpartum. Diet did not affect the measured ovarian activity variables within 100 DIM. In conclusion, omitting the dry period (0 days) increased the incidence of normal resumption of ovarian cyclicity in dairy cows within 100 DIM compared with a conventional dry period (60 days). Short (<18 days) or long (>24 days) ovarian cycles during the first ovarian cycle postpartum were associated with severe negative energy balance and poor metabolic status in early lactation., (Copyright © 2015 Elsevier Inc. Lying behavior is a relevant indicator for the evaluation of cow welfare. Lying can be recorded automatically by data loggers attached to one of the hind legs of a cow. A threshold for the duration of a lying bout (LB) record is required, however, to discard false records caused by horizontal leg movements, such as scratching. Previously determined thresholds for similar sensors ranged from 25s to 4min. We aimed to validate LB recorded by the IceQube sensor (with IceManager software; IceRobotics, South Queensferry, UK) and to determine a threshold to distinguish true from false LB records in lactating dairy cows. A novel method of validation, which does not require time-consuming behavioral observations, was used to generate a larger data set with potentially more incidental short LB records. Both hind legs of 28 lactating dairy cows were equipped with an IceQube sensor for a period of 6d and used as each other's validation. Classification of LB records as true (actual LB) or false (recorded while standing) was based on 3 assumptions. First, all standing records (absence of LB records) were assumed to occur while standing. Second, false LB records due to short leg movements could not occur in both hind legs simultaneously. Third, true LB only occurred if the LB records of the paired sensors coincided. False LB records constituted 4% of the records. Based on a maximum accuracy of 0.99, a minimum duration of LB records of 33 s was determined, implying a sensitivity of 0.99 and a specificity of 0.98. Applying this threshold of 33 s hardly affected estimates of daily lying time, but improved estimates of frequency and mean duration of LB for individual cows. The importance of distinguishing short LB was demonstrated specifically for detection of calving. The 2-sensor approach, using sensor outputs on both hind legs as each other's validation, is a time-efficient method to validate LB records that can be applied to different sensors and husbandry conditions., (Copyright © 2015 American Dairy Science Association. The aim of this study was to assess the potential of milk fatty acids as diagnostic tool for hyperketonemia of 93 dairy cows in a 3×2 factorial arrangement. Cows were fed a glucogenic or lipogenic diet and originally were intended to be subjected to a 0-, 30-, or 60-d dry period. Nevertheless, some of the cows, which were intended for inclusion in the 0-d dry period group, dried off spontaneously. Milk was collected in wk 2, 3, 4, and 8 of lactation for milk fat analysis. Blood was sampled from wk 2 to 8 after parturition for β-hydroxybutyrate (BHBA) analysis. Cases were classified into 2 groups: hyperketonemia (BHBA ≥1.2mmol/L) and nonhyperketonemia (BHBA <1.2mmol/L). Concentrations of 45 milk fatty acids and ratios of anteiso C15:0-to-anteiso C17:0 and C18:1 cis-9-to-C15:0 were subjected to a logistic regression analysis (stepwise forward method). The milk fat C18:1 cis-9-to-C15:0 ratio revealed the most discriminating factor for diagnosis of hyperketonemia. Ninety percent of nonhyperketonemia cases showed a milk fat C18:1 cis-9-to-C15:0 ratio of 40 or lower, whereas 70% of cows suffering from hyperketonemia showed milk fat C18:1 cis-9-to-C15:0 ratios exceeding 40. Additionally, cows with a milk fat ratio C18:1 cis-9-to-C15:0 of at least 45 in wk 2 after parturition had about 50% chance to encounter blood plasma BHBA values of 1.2mmol/L or more during the first 8 wk of lactation. Of the cows not suffering from hyperketonemia during the first 2 mo of lactation, only 9% exceeded this wk 2 threshold. Practical implementation requires routine analysis of both milk fatty acids, which currently is lacking for C15:0. The objective was to study the effect of dry period length in dairy cows on immunoglobulin content and natural antibodies (NAb) titers in colostrum, growth, and plasma natural and specific antibody titers in plasma of calves. Holstein-Friesian dairy cows (n=167) were randomly assigned to 3 dry period lengths (0, 30, or 60 d). Colostrum production, concentration of colostrum IgG and IgM, and titers of NAb (isotypes IgG and IgM) binding keyhole limpet hemocyanin (KLH) and human serum albumin (HuSA) in colostrum were measured. Female calves were immunized with both KLH and HuSA at wk 6 and 10 of life. Titers of NAb and specific antibody (SpAb) for isotypes IgG, IgM, and total immunoglobulin (IgT) binding KLH or HuSA were determined in plasma of female calves. Primary and secondary antibody responses to KLH or HuSA from wk 6 and 10 were expressed as the increase in antibody titers to wk 10 and 11 of life after primary and secondary challenges, respectively. Pregnancy length for cows with a 0-d dry period was 3d shorter compared with cows with a 30- or 60-d dry period. Birth weight of calves from cows with a 0-d dry period was lower compared with calves from cows with a 30-d dry period. Growth of calves until 12 wk of life was not affected by dry period length. Colostrum production and IgG and IgM concentration in colostrum were lower for cows with a 0-d dry period than a 60-d dry period. Natural IgG and IgM titers binding KLH or HuSA were lower in colostrum from cows with a 0-d dry period compared with cows with a 60-d dry period. Natural antibody titers (IgG, IgM, and IgT) binding KLH or HuSA in plasma were lower during the first 2 wk of life for calves from cows with a 0-d dry period compared with calves from cows with a 30- or 60-d dry period. After primary and secondary immunization of calves with KLH and HuSA, SpAb titers of calves were not affected by dry period length. After secondary immunization, the response of IgG and IgT binding KLH was higher in plasma of calves from cows with a 0-d dry period. The results of this study demonstrate that, although omission of the dry period of dairy cows leads to lower plasma NAb titers in calves during the first 2 wk of life, SpAb titers in calves were not affected and even the secondary antibody responses were enhanced compared with calves from cows with a 30- or 60-d dry period., (Copyright © 2015 American Dairy Science Association. In a prior study, we observed that cows with a 0-d dry period had greater energy balance and lower milk production compared with cows with a 30- or 60-d dry period in early lactation. The objective of the current study was to evaluate the influence of dry period length on metabolic status and hepatic gene expression in cows fed a lipogenic or glucogenic diet in early lactation. Holstein-Friesian dairy cows (n=167) were assigned randomly to 3×2 factorial design with 3 dry period lengths (n=56, 55, and 56 for 0-, 30-, and 60-d dry, respectively) and 2 early lactation diets (n=84 and 83 for glucogenic and lipogenic diet, respectively). Cows were fed a glucogenic or lipogenic diet from 10d before the expected calving date and onward. The main ingredient for a glucogenic concentrate was corn, and the main ingredients for a lipogenic concentrate were sugar beet pulp, palm kernel, and rumen-protected palm oil. Blood was sampled weekly from 95 cows from wk 3 precalving to wk 8 postcalving. Liver samples were collected from 76 cows in wk -2, 2, and 4 relative to calving. Liver samples were analyzed for triacylglycerol concentrations and mRNA expression of 12 candidate genes. Precalving, cows with a 0-d dry period had greater plasma β-hydroxybutyrate, urea, and insulin concentrations compared with cows with a 30- or 60-d dry period. Postcalving, cows with a 0-d dry period had lower liver triacylglycerol and plasma nonesterified fatty acids concentrations (0.20, 0.32, and 0.36mmol/L for 0-, 30-, and 60-d dry period, respectively), greater plasma glucose, insulin-like growth factor-I, and insulin (24.38, 14.02, and 11.08µIU/mL for 0-, 30-, and 60-d dry period, respectively) concentrations, and lower hepatic mRNA expression of pyruvate carboxylase, compared with cows with a 30- or 60-d dry period. Plasma urea and β-hydroxybutyrate concentrations were greater in cows fed a lipogenic diet compared with cows fed a glucogenic diet. In conclusion, cows with a 0-d dry period had an improved metabolic status in early lactation, indicated by lower plasma concentrations of nonesterified fatty acids, greater plasma concentrations of glucose, insulin-like growth factor-I, and insulin, and lower mRNA expression of pyruvate carboxylase in the liver, compared with cows with a 30- or 60-d dry period. Most cows encounter a state of negative energy balance during the periparturient period, which may lead to metabolic disorders and impaired fertility. The aim of this study was to assess the potential of milk fatty acids as diagnostic tools of detrimental levels of blood plasma nonesterified fatty acids (NEFA), defined as NEFA concentrations beyond 0.6 mmol/L, in a data set of 92 early lactating cows fed a glucogenic or lipogenic diet and subjected to 0-, 30-, or 60-d dry period before parturition. Milk was collected in wk 2, 3, 4, and 8 (n = 368) and blood was sampled weekly from wk 2 to 8 after parturition. Milk was analyzed for milk fatty acids and blood plasma for NEFA. Data were classified as "at risk of detrimental blood plasma NEFA" (NEFA ≥ 0.6 mmol/L) and "not at risk of detrimental blood plasma NEFA" (NEFA <0.6 mmol/L). Concentrations of 45 milk fatty acids and milk fat C18:1 cis-9-to-C15:0 ratio were subjected to a discriminant analysis. Milk fat C18:1 cis-9 revealed the most discriminating variable to identify detrimental blood plasma NEFA. A false positive rate of 10% allowed us to diagnose 46% of the detrimental blood plasma NEFA cases based on a milk fat C18:1 cis-9 concentration of at least 230 g/kg of milk fatty acids. Additionally, it was assessed whether the milk fat C18:1 cis-9 concentrations of wk 2 could be used as an early warning for detrimental blood plasma NEFA risk during the first 8 wk in lactation. Cows with at least 240 g/kg of C18:1 cis-9 in milk fat had about 50% chance to encounter blood plasma NEFA values of 0.6 mmol/L or more during the first 8 wk of lactation, with a false positive rate of 11.4%. Profit simulations were based on costs for cows suffering from detrimental blood plasma NEFA, and costs for preventive treatment based on daily dosing of propylene glycol for 3 wk. Given the relatively low incidence rate (8% of all observations), continuous monitoring of milk fatty acids during the first 8 wk of lactation to diagnose detrimental blood plasma NEFA does not seem cost effective. On the contrary, milk fat C18:1 cis-9 of the second lactation week could be an early warning of cows at risk of detrimental blood NEFA. The length of the dry period in commercial dairy production is under close scrutiny. While the main concern is the composition and volume of milk produced, the evaluation of colostrum quality under these new paradigms has suggested a decline in IgG concentrations, while some reports indicate no change. Colostrum quality has been defined as an adequate concentration (>50 mg/ml) of immunoglobulin in the secretions to provide the newborn with maximal disease resistance. We investigated the appearance of IgG in mammary pre- and post partum secretions in cows without a dry period (continuously milked, Dry0) and compared the secretions with cows that experienced a dry period of 60 d (Dry60). Blood was collected during the experimental period and plasma analysed for progesterone (P4) and prolactin (Prl). Approximately -6 d relative to parturition, the Dry0 animals exhibited increased concentration of IgG in their secretions to an average of ∼35 mg/ml that remained rather constant through subsequent pregnancy and following parturition. Dry0 cows were producing an average IgG concentration in parturition colostrum of 44·2±17·6 mg/ml that was not different than that of controls (66·86±16·8 mg/ml). However, Dry0 cows exhibited high variation, different peak times (day) of IgG concentration including times that occurred both pre and post parturition. IgG mass of the Dry0 cows remained rather constant pre- and post partum and did not show the same declining mass following parturition that was shown for the Dry60 cows. The change in plasma P4 and Prl were shown to have no timing effect on colostrum IgG concentration.

The objective of this study was to evaluate the effects of dry period length and dietary energy source in early lactation on milk production, feed intake, and energy balance (EB) of dairy cows. Holstein-Friesian dairy cows (60 primiparous and 108 multiparous) were randomly assigned to dry period lengths (0, 30, or 60 d) and early lactation ration (glucogenic or lipogenic), resulting in a 3 × 2 factorial design. Rations were isocaloric and equal in intestinal digestible protein. The experimental period lasted from 8 wk prepartum to 14 wk postpartum and cows were monitored for milk yield, milk composition, dry matter intake (DMI), energy balance, and milk fat composition. Prepartum average milk yield for 60 d precalving was 13.8 and 7.7 ± 0.5 kg/d for cows with a 0- and 30-d dry period, respectively. Prepartum DMI and energy intake were greater for cows without a dry period and 30-d dry period, compared with cows with a 60-d dry period. Prepartum EB was greater for cows with a 60-d dry period. Postpartum average milk yield until wk 14 was lower for cows without a dry period and a 30-d dry period, compared with cows with a 60-d dry period (32.7, 38.7, and 43.3 ± 0.7 kg/d for 0-, 30-, and 60-d dry period, respectively). Postpartum DMI did not differ among treatments. Postpartum EB was greater for cows without a dry period and a 30-d dry period, compared with cows with a 60-d dry period. Young cows (parity 2) showed a stronger effect of omission of the dry period, compared with a 60-d dry period, on additional milk precalving (young cows: 15.1 kg/d; older cows: 12.0 kg/d), reduction in milk yield postcalving (young cows: 28.6 vs. 34.8 kg/d; older cows: 41.8 vs. 44.1 kg/d), and improvement of the EB postcalving (young cows: 120 vs. -93 kJ/kg(0.75)·d; older cows: -2 vs. -150 kJ/kg(0.75)·d. Ration did not affect milk yield and DMI, but a glucogenic ration tended to reduce milk fat content and increased EB, compared with a more lipogenic ration. Reduced dry period length (0 and 30 d) increased the proportion of short- and medium-chain fatty acids in milk fat and omitting the dry period decreased the proportion of long-chain fatty acids in milk fat. In conclusion, shortening and omitting the dry period shifts milk yield from the postpartum to the prepartum period; this results in an improvement of the EB in early lactation. Shortening or omitting the dry period (DP) has been proposed as a management strategy to improve energy balance of dairy cows in early lactation. Both shortening and complete omission of the DP reduces milk production in the subsequent lactation compared with a conventional DP length of 60d. Some cows have less milk production loss than other cows after applying no DP or a short DP. The aim of this study is to evaluate which cow characteristics are associated with the amount of milk production losses following no DP or a short DP (30d). Daily production information from the lactation before and after the DP was available from 161 dairy cows (54 cows with a 0-d DP, 51 cows with a 30-d DP, and 56 cows with a 60-d DP) from a research herd. Daily production (milk, fat, and protein) until 305d in milk was estimated for all cows. Subsequently, total fat- and protein-corrected milk yield from 60d before the expected calving date until 305d in the following lactation (FPCMtotal) was estimated. A statistical analysis was performed to evaluate which cow characteristics were associated with limited or no production losses following no DP or a short DP, compared with a conventional DP length of 60d. Average FPCMtotal was 9,341, 10,499, and 10,795kg for cows with no DP, a 30-d DP, and a 60-d DP, respectively. The cow characteristics parity, daily milk production at 12wk before the expected calving date, and reduction in daily milk production between 16 and 12wk before the expected calving date were associated with production loss due to a short (30d) or no DP. Compared with 60d DP, multiparous cows had less production loss (987kg) following no DP than primiparous cows (2,132kg). The difference in FPCMtotal between the 3DP groups was largest for cows with a low milk production (e.g., 10kg/d) at 12wk before the expected calving date. The greater the reduction in milk production between 16 and 12wk before the expected calving date, the larger the difference in FPCMtotal between the 3DP groups. The difference in FPCMtotal between cows with no DP and 60d DP at a reduction in milk production between 16 and 12wk of 10% was 665kg, whereas this difference was 1,138kg at a reduction of 70%. The cow characteristics found can be used to select cows for specific DP lengths in a decision-support model to support the farmer on the economic optimal DP length for each individual cow. A dry period of 6-8 weeks for dairy cows is generally thought to maximise milk production in the next lactation. However, the value of such a long dry period is increasingly questioned. In particular, shortening the dry period shifts milk production from the critical period after calving to the weeks before calving. This shift in milk production could improve the energy balance (EB), health and fertility of dairy cows. The objective of this study was to systematically review the current knowledge on dry period length in relation to milk production, EB, fertility, and health of cows and calves. A meta-analysis was performed for variables where at least five studies were available. Overall, both shortening and omitting the dry period reduces milk production, increases milk protein percentage and tends to reduce the risk of ketosis in the next lactation. Individual studies reported an improvement of EB after a short or no dry period, compared with a conventional dry period. Shortening or omitting the dry period did not affect milk fat percentage and shortening the dry period did not alter the odds ratio for mastitis, metritis, or fertility measures in the next lactation. So, current evidence for an improvement of health and fertility of dairy cows is marginal and may be partly explained by the limited number of studies which have evaluated health and fertility in relation to dry period length, the limited number of animals in those studies and the variable responses reported., (Copyright © 2013 Elsevier Ltd. Shortening the dry period (DP) has been proposed as a management strategy to improve energy balance in early lactation. It is well known that both shortening and complete omission of the DP reduces milk production in the subsequent lactations. In most of these studies milk production data were obtained from planned animal experiments where cows were randomly assigned to DP length treatments, and cow management and diet composition did not differ among treatments. It may therefore be hypothesized that cows on commercial herds which apply a no-DP or short-DP-strategy, and support this by management adjustments, will have a less dramatic reduction in milk production. In this study, milk production and somatic cell count (SCC) following different DP lengths was investigated under commercial circumstances. Milk production of 342 cows (2,077 test-day records) was available from 5 Dutch commercial dairy herds which started a no DP-strategy for all cows. Test days of the year before applying the no-DP strategy are used as control (323 cows, 1,717 test-day records). Six other herds applied an individual cow approach and have different preplanned DP lengths within one herd. From these herds, information on 81 cows (482 test-day records) with a DP length between 0 and 20 d, 127 cows (925 test-day records) with a DP length between 21 and 35 d, and 143 cows (1,075 test-day records) with a DP length of more than 35 d was available. A generalized linear model incorporating an autoregressive covariance structure accounting for repeated test-day yields within cow was developed to estimate the daily yield (milk, fat and protein) and SCC of all cows. Applying no DP for all cows in the herd resulted in a reduction in postpartum milk production compared with within-herd control lactations (until 305 DIM) between 3.2 and 9.1 kg/d, which was a reduction of 12 and 32%, respectively. For the 6 herds that applied an individual cow approach with different preplanned DP lengths, the cow-specific DP strategy was based on milk production and SCC approximately 2 mo before calving. Cows with a preplanned DP length ranging between 0 and 20 d had a reduction in postpartum milk production between 5.7 and 13 kg/d compared with cows with a DP length of >35 d. Cows with a preplanned DP length ranging from 21 to 35 d had a numerically lower milk production (between 0.6 and 5.3 kg/d) than cows with a preplanned DP length of >35 d, but this difference was significant in only one herd. Natural antibodies (NAb) are defined as antibodies that circulate in normal healthy individuals under the absence of deliberate antigenic stimulation. Two types of NAb are distinguished: NAb towards exogenous antigens and NAb towards autoantigens (N(A)Ab). The objectives of the current study were threefold. First, we studied the relation between metabolic health and concentrations of NAb binding keyhole limpet hemocyanin (KLH) or lipopolysaccharide (LPS) in milk and plasma of dairy cows in early lactation. Second, we determined the presence of N(A)Ab binding transferrin, myosin and thyroglobulin in bovine milk. Third, we studied the relation between N(A)Ab in bovine milk and mammary health. For the first objective, dairy cows were either fed a control (C) (n=8) or a diet where 2 kg of concentrates were replaced by an iso-energetic concentrate containing marine algae (ALG) from week -3 till 8 postpartum (experiment 1). Plasma and milk samples were analyzed weekly for NAb binding either KLH or LPS. Plasma was analyzed for glucose, non-esterified fatty acids (NEFA) and β-hydroxybutyrate (BHBA). For the second and third objective, milk samples were collected weekly from 96 dairy cows from week 2 till 9 postpartum and analyzed for milk composition and N(A)Ab binding myosin, transferrin and thyroglobulin (experiment 2). For both datasets, N(A)Ab titers are expressed as (2)log values of the highest dilution giving a positive reaction. Data are expressed as means ± SEM. Repeated observations were analyzed in a mixed model. In experiment 1, no diet effect (P>0.05) was observed on NAb binding LPS in plasma or milk, NAb binding KLH in milk was greater (P=0.05) for cows fed the control diet. Concentration of NAb binding KLH and LPS in plasma was negatively related to plasma NEFA concentration (P<0.05). In experiment 2, NAb binding myosin (5.66 ± 0.06), thyroglobulin (4.85 ± 0.06), and transferrin (5.76 ± 0.07) were identified in milk. Clinical mastitis incidence (9%) tended to be positively related to concentration of NAb binding myosin (P=0.06) and negatively related to Nab binding transferrin (P=0.08). In conclusion, NAb binding KLH and LPS in plasma and milk are related to metabolic health, as indicated by plasma NEFA concentration. Furthermore, this study demonstrates the presence of N(A)Ab in bovine milk and shows trends for a relation between N(A)Ab binding auto-antigens and mastitis. The objective of this study was to evaluate Fourier transform infrared (FTIR) spectrometry to measure milk ketone bodies to detect hyperketonemic cows and compare this method with milk fat to protein ratio to detect hyperketonemia. Plasma and milk samples were obtained weekly from calving to wk 9 postpartum from 69 high-producing dairy cows. The reference test for hyperketonemia was defined as plasma concentration of beta-hydroxybutyrate (BHBA) >or=1,200 micromol/L. The weekly prevalence of hyperketonemia during the first 9 wk of lactation was, on average, 7.1%. Both BHBA and acetone in milk, determined by FTIR, had a higher sensitivity (80%) to detect hyperketonemia compared with milk fat to protein ratio (66%). Specificity was similar for the 3 diagnostic tests (71, 70, and 71%). In conclusion, FTIR predictions of BHBA or acetone in milk can detect cows with hyperketonemia in early lactation with a higher accuracy compared with the use of milk fat to protein ratio. Because of the high proportion of false-positive tests, there are concerns about the practical applicability of FTIR predictions of acetone, BHBA, and fat to protein ratio in milk to detect hyperketonemic cows., (Copyright (c) 2010 American Dairy Science Association. The objective of this study was to determine whether concentrations of specific fatty acids in milk fat are a candidate for the early detection of subclinical ketosis. The case study included multiparous cows fed a lipogenic diet or a mixed glucogenic:lipogenic diet during the first 9 wk of lactation. Milk fatty acid profiles of cows classified as healthy (n = 8) or as subclinically ketotic (n = 8) based on a blood plasma beta-hydroxybutyrate threshold concentration of 1.2 mmol/L were compared. Subclinically ketotic cows showed an elevated proportion of C18:1 cis-9 in milk fat during the whole registration period.

For several decades, researchers worldwide report a decrease in fertility in high-yielding dairy cows, most probably based on conflicting metabolic and reproductive needs. The dairy herd manager's success at improving milk production has been accompanied by a negative trend for the most visible reproductive parameters such as calving intervals, number of days open and number of inseminations needed per pregnancy. In parallel, many research groups studied the metabolic and endocrine factors that influence follicular growth and the developmental competence of oocytes and embryos. In the past, herd managers and reproductive biologists each tried to tackle the same problems with limited consultation. More recently, the situation has improved significantly and theriogenologists, nutritionists and veterinarians now conduct research in multidisciplinary teams. This review paper starts in a general way by discussing nutrient prioritization towards the udder to guarantee milk production and by describing interactions between the somatotropic and gonadotropic axis. It then focuses on the consequences of the negative energy balance on follicular growth and environment, oocyte and embryo quality, not only by summarizing the currently accepted hypotheses but also based on clear scientific evidence at the follicular level. All this, with one question in mind: is there a mismatch between metabolism and fertility and what can the dairy manager learn from research to tackle the problem of reduced fertility?

The objectives of this study were to determine the presence of natural antibodies (NAb) in plasma and milk of individual dairy cows and to study the relation between NAb concentrations and energy balance (EB) and dietary energy source. Cows (n = 76) were fed a mainly glucogenic, lipogenic, or a mixture of both diets (50:50 dry matter basis) from wk 3 before the expected calving date until wk 9 postpartum. Diets were isocaloric (net energy basis) and equal in intestinal digestible protein. Blood and milk were sampled weekly. Liver biopsies were taken in wk -2, 2, 4, and 6 relative to calving. Data are expressed as LSM +/- SEM. The NAb titers are expressed as the (2)log values of the highest dilution giving a positive reaction. The NAb concentration in plasma binding either keyhole limpet hemocyanin (KLH) or Escherichia coli lipopolysaccharide (LPS) increased with parity. The NAb concentration binding KLH was greater for cows fed the glucogenic diet (9.63 +/- 0.08) compared with the lipogenic diet (9.26 +/- 0.08). In milk, cows fed the glucogenic diet had smaller NAb concentrations binding KLH (3.98 +/- 0.18) and LPS (2.88 +/- 0.17) compared with cows fed the mixed diet (KLH: 4.93 +/- 0.18; LPS: 3.70 +/- 0.17). The NAb concentration in plasma had a positive relation with energy balance variables: EB, dry matter intake, milk yield, and plasma cholesterol, whereas NAb concentration in milk had a negative relation with energy balance variables: EB, dry matter intake, and plasma cholesterol. Additionally, NAb concentrations in milk had a positive relation with plasma nonesterified fatty acid concentration and milk fat and protein percentage. There was a tendency for a positive relation of NAb concentration binding LPS in plasma and somatic cell count in milk. No significant relations were detected between NAb concentrations in milk or plasma and plasma beta-hydroxybutyrate concentration and liver triacyl glyceride content. In conclusion, NAb are present in both milk and plasma of dairy cows peripartum and NAb concentrations increase with parity. Furthermore, our data indicate that a negative energy balance in dairy cows in early lactation can be associated with compromised innate immune function as indicated by decreased NAb concentration in plasma.

This paper summarizes three recent studies by the same authors with the objective to study the effect of dietary energy source on the energy balance (EB) and risk for metabolic and reproductive disorders in dairy cows in early lactation. The first study, a literature survey, illustrated that feeding extra glucogenic nutrients relative to lipogenic nutrients, decreased milk fat and seems to decrease plasma non-esterified fatty acid (NEFA) and beta-hydroxybutyrate (BHBA) concentration. However studies are scarce and mostly confound the effect of energy source with level of energy intake, compromising eventual effects on EB and fertility. Therefore, in the second study, 16 dairy cows were either fed a glucogenic or a lipogenic diet (isocaloric) and EB was determined in climate-controlled respiration chambers from week 2 until 9 of lactation. Glucogenic diet decreased milk fat yield and milk energy and tended to decrease body fat mobilization compared with lipogenic diet. The objective of the third study was to study the effect of dietary energy source on EB, metabolites and reproduction variables. Dairy cows (n=111) were fed glucogenic, lipogenic or mixed diet from week 3 until week 9 relative to calving. Multiparous cows fed glucogenic diet had lower milk fat yield, higher calculated EB, and lower plasma NEFA, BHBA and liver triacylglyceride concentration and tended to have fewer days to first postpartum ovulation. In conclusion, increasing the glucogenic nutrient availability improved the EB and had potential to reduce the risk for metabolic disorders and to improve reproductive performance in dairy cows.

Increasing the availability of glucogenic nutrients relative to lipogenic nutrients has been hypothesized to decrease the production of milk fat, to improve the energy balance (EB), and to decrease the incidence and severity of metabolic and reproductive disorders in dairy cows in early lactation. Therefore, our objective was to evaluate the effects of a glucogenic, lipogenic, or mixed diet on EB, plasma metabolites and metabolic hormones, liver triacylglycerides (TAG), and reproductive variables in high-producing dairy cows in early lactation. Cows (n = 114) were randomly assigned to 1 of 3 diets and were fed either a mainly lipogenic diet, a mainly glucogenic diet, or a mixture of both diets (50:50 dry matter basis) from wk 3 before the expected calving date until 9 wk postpartum. Diets were isocaloric (net energy basis) and equal in intestinal digestible protein. Dry matter intake, net energy intake, milk yield, and milk protein percentage did not differ among diets. Milk lactose percentage was less for cows fed the lipogenic diet. Milk fat percentage was less for multiparous cows fed the glucogenic diet compared with cows fed the mixed or lipogenic diet (3.69 vs. 4.02 vs. 4.22 +/- 0.07%, respectively). The calculated EB was less negative for multiparous cows fed the glucogenic diet compared with cows fed the mixed or lipogenic diet [-33 vs. -125 vs. -89 +/- 21 kJ/(kg(0.75) x d), respectively]. Postpartum, the glucogenic diet decreased plasma nonesterified fatty acids, beta-hydroxybutyrate, and liver TAG concentrations and increased insulin concentration in multiparous cows. The glucogenic diet tended to decrease the number of days until first milk progesterone rise in multiparous cows compared with the mixed or lipogenic diet (20.4 vs. 24.4 vs. 26.4 +/- 2.1 d, respectively). Diet had no effect on any of the above-mentioned variables in primiparous cows, except that milk lactose percentage was greater for primiparous cows fed the glucogenic diet. We concluded that the glucogenic diet was effective in improving the calculated EB and decreasing plasma beta-hydroxybutyrate and liver TAG concentrations, suggesting a reduced risk of metabolic disorders in multiparous dairy cows fed a glucogenic diet.

Negative energy balance-related metabolic disorders suggest that the balance between available lipogenic and glucogenic nutrients is important. The objectives of this study were to compare the effects of a glucogenic or a lipogenic diet on liver triacylglycerides (TAG), metabolites, and metabolic hormones in dairy cows in early lactation and to relate metabolite concentrations to the determined energy retention in body mass (ER). Sixteen dairy cows were fed either a lipogenic or glucogenic diet from wk 3 prepartum to wk 9 postpartum (pp) and were housed in climate respiration chambers from wk 2 to 9 pp. Diets were isocaloric (net energy basis). Postpartum, cows fed a lipogenic diet tended to have higher nonesterified fatty acid concentration (NEFA; 0.46 +/- 0.04 vs. 0.37 +/- 0.04 mmol/L) and lower insulin concentration (4.0 +/- 0.5 vs. 5.5 +/- 0.6 microIU/mL). No difference was found in plasma glucose, beta-hydroxybutyrate, insulin-like growth factor-I, and thyroid hormones. Liver TAG was equal between both diets in wk -2 and 2 pp. In wk 4 pp cows fed the glucogenic diet had numerically lower TAG levels, although there was no significant dietary effect. Negative relationships were detected between ER and milk fat and between ER and NEFA. A positive relationship was detected between ER and insulin concentration. Overall, results suggest that insulin plays a regulating role in altering energy partitioning between milk and body tissue. Feeding lactating dairy cows a glucogenic diet decreased mobilization of body fat compared with a lipogenic diet. The relative abundance of lipogenic nutrients, when feeding a more lipogenic diet, is related to more secretion of lipogenic nutrients in milk, lower plasma insulin, and higher plasma NEFA concentration.

Metabolic problems related to negative energy balance suggest a role for the balance in supply of lipogenic and glucogenic nutrients. To test the effect of lipogenic and glucogenic nutrients on energy partitioning, energy balance and nitrogen balance of 16 lactating dairy cows were determined by indirect calorimetry in climate respiration chambers from wk 2 to 9 postpartum. Cows were fed a diet high in lipogenic nutrients or a diet high in glucogenic nutrients from wk 3 prepartum until wk 9 postpartum. Diets were isocaloric (net energy basis) and equal in intestinal digestible protein. There was no effect of diet on metabolizable energy intake and heat production. Cows fed the lipogenic diet partitioned more energy to milk than cows fed the glucogenic diet [1,175 +/- 18 vs. 1,073 +/- 12 kJ/(kg(0.75) x d)] and had a higher milk fat yield (1.89 +/- 0.02 vs. 1.67 +/- 0.03 kg/d). The increase in milk fat production was caused by an increase in C16:0, C18:0, and C18:1 in milk fat. No difference was found in energy retained as body protein, but energy mobilized from body fat tended to be higher in cows fed the lipogenic diet than in cows fed the glucogenic diet [190 +/- 23 vs. 113 +/- 26 kJ/(kg(0.75) x d)]. Overall, results demonstrate that energy partitioning between milk and body tissue can be altered by feeding isocaloric diets differing in lipogenic and glucogenic nutrient content.

The pathway for oxidation of energy involves a balanced oxidation of C2 and C3 compounds. During early lactation in dairy cattle this C2/C3 ratio is out of balance, due to a high availability of lipogenic (C2) products and a low availability of glycogenic (C3) products relative of the C2 and C3 products required for milk production. This review compares studies which manipulated dietary energy source and shows that dietary energy source can affect the balance of the C2/C3 ratio, as indicated by plasma NEFA, beta-hydroxybutyrate (BHBA) and glucose levels. It is shown that glycogenic nutrients increase glucose and insulin concentrations and decrease NEFA and BHBA plasma levels. Extra lipogenic nutrients elevate NEFA and BHBA and decrease plasma glucose concentrations. Lipogenic nutrients generally increase milk fat percentage and decrease milk protein percentage, suggesting a surplus of C2 compounds. The inverse is the case for feeding extra glycogenic nutrients, implying reduced deamination and oxidation of glycogenic amino acids. Feeding extra glycogenic nutrients improved the energy balance (EB), in contrast to ambiguous results of lipogenic nutrients on EB. Moreover, glycogenic feed may reduce the severity of ketosis and fatty liver, but increased the incidence of (sub)clinical acidosis. Since studies are scarce, it seems difficult to draw conclusions on the effects of dietary energy source on reproduction. However, lipogenic nutrients decrease glucose and increase NEFA and BHBA plasma levels. High plasma NEFA and BHBA and low plasma glucose levels are associated with decreased reproductive performance, which might imply the C2/C3 compound balance to be important for reproductive function.
