The rise of agro-industrial activities over recent decades has exponentially increased lignocellulose biomasses (LCB) production. LCB serves as a cost-effective source for fermentable sugars and other renewable chemicals. This study explores the use of microbial consortia, particularly thermophilic consortia, for LCB deconstruction. Thermophiles produce stable enzymes that retain activity under industrial conditions, presenting a promising approach for LCB conversion. This research focused on two microbial consortia (i.e., microbiomes) that were analyzed for enzyme production using a cheap medium, i.e., a mixture of spent mushroom substrate (SMS) and digestate. The secreted xylanolytic enzymes were characterized in terms of temperature and pH optima, thermal stability, and hydrolysis products from LCB-derived polysaccharides. These enzymes showed optimal activity aligning with common biorefinery conditions and outperformed a formulated enzyme mixture in thermostability tests in the digestate. Phylogenetic and genomic analyses highlighted the genetic diversity and metabolic potential of these microbiomes. Bacillus licheniformis was identified as a key species, with two distinct strains contributing to enzyme production. The presence of specific glycoside hydrolases involved in the cellulose and hemicellulose degradation underscores these consortia's capacity for efficient LCB conversion. These findings highlight the potential of thermophilic microbiomes, isolated from an industrial environment, as a robust source of robust enzymes, paving the way for more sustainable and cost-effective bioconversion processes in biofuel and biochemical production and other biotechnological applications.

Background: Laboratory-scale experiments have shown that treatment with selective lignin-degrading white-rot fungi improves the nutritional value and ruminal degradability of lignocellulosic biomass (LCB). However, the lack of effective field-applicable pasteurization methods has long been recognized as a major obstacle for scaling up the technique for fungal treatment of large quantities of LCB for animal feeding. In this study, wheat straw (an LCB substrate) was subjected to four field-applicable pasteurization methods - hot-water, formaldehyde fumigation, steam, and hydrated lime - and cultured with Pleurotus ostreatus grain spawn for 10, 20, and 30 days under solid-state fermentation. Samples of untreated, pasteurized but non-inoculated and fungus-treated straws were analyzed for chemical composition, aflatoxin B 1 (AFB 1 ), and in vitro dry matter digestibility (IVDMD), in vitro total gas (IVGP), methane (CH 4 ), and volatile fatty acid (VFA) production., Results: During the 30-day fungal treatment, steam and lime pasteurized straws had the greatest loss of lignin, resulting in marked improvements in crude protein (CP), IVDMD, IVGP, and total VFAs. Irrespective of the pasteurization method, the increase in IVDMD during fungal treatment was linearly (R 2  = 0.77-0.92) related to lignin-loss in the substrate during fungal treatment. The CH 4 production of the fungus-treated straw was not affected by the pasteurization methods. Aflatoxin B 1 was within the safe level (<5 μg kg -1 ) in all pasteurized, fungus treated straws., Conclusion: Steam and lime were promising field-applicable pasteurization techniques to produce nutritionally improved fungus-treated wheat straw to feed ruminants. Biomass conversion aims at degrading the structural polysaccharides of lignocellulose into reducing sugars. Pretreatment is necessary to overcome the recalcitrance of lignocellulose. The DES La/ChCl in this paper was selected based on our previous study. To examine cellulase adsorption of lignocellulose after DES pretreatment, sorghum straw was pretreated with DES under different condition. The adsorption improvement of cellulase on lignocellulose after DES pretreatment has positive impact on reducing sugar production of biomass. After DES pretreatment, 1. pore corrosion caused the upward trend of pore radius and the downward trend of SSA. 2. the hydrogen bounding force of pretreated sorghum straw and MCC decreased, the hydrogen bounding force of pretreated lignin increased. 3. although the unsaturation of pretreated lignin increased, DES pretreatment is helpful for the removal of lignin. 4. The decrease in the hydrophobicity of sorghum straw make it easier to disperse. 5. the Zeta potential of pretreated sorghum straw shifted towards the positively charged region, while pretreated lignin shifted towards the negatively charged region. 6. different adsorption behaviors were observed in specific components of cellulase mixtures (BGs, CBHs, EGs and xlylanase). Creating renewable energy from lignocellulosic biomass is essential for a sustainable future. Due to their abundance and the possibility of producing cheap and clean energy, non-lignocellulosic wastes like sewage sludge from industrial and municipal wastes have drawn attention as a feasible alternative to fossil fuels. These abundant, cost-effective resources may help minimize the effects of climate change since they produce less pollution. Several drawbacks are associated with using sewage sludge in thermal conversion procedures. These issues encompass suboptimal energy yield, elevated ash levels in the final product, and subpar biomass quality. Using these scraps in conjunction with coal might enhance energy conversion processes. This study has revealed the necessity for further investigation into how various combinations of residues interact with each other, influencing synergistic effects and degradation processes. The study's underlying objective was to provide a centralized database on the synergistic effects of mixing biomass and sewage sludge for bioenergy production, coal and biomass, and coal and sewage sludge through thermochemical processes like combustion, pyrolysis, gasification, and hydrolysis with Aspen Plus. Lignocellulosic biomass is available in abundance as a renewable resource, but the major portion of it is often discarded as waste without utilizing its immense potential as an alternative renewable energy resource. To overcome recalcitrance of lignocellulosic biomass, various pretreatment methods are applied to it, so that the complex and rigid polymeric structure can be broken down into fractions susceptible for enzymatic hydrolysis. Effective and efficient biomass processing is the goal of pretreatment methods, but none of the explored pretreatment methods are versatile enough to fulfil the requirement of biomass processing with greater flexibility in terms of operational cost and desired output efficiency. Deployment of green solvents such as ionic liquids for the pretreatment of lignocellulosic biomass has been a topic of discussion amongst the scientific community in recent times. The presented work provides a detailed overview on the deployment of ionic liquid for the pretreatment of lignocellulosic biomass coupled with a brief discussion on other pretreatments methods. The recyclability and reusability along with other unique properties makes an ionic liquid pretreatment different from the other traditional pretreatment methods. Also, this study explores diverse critical parameters that governs the dissolution process of biomass. Hazardous properties of ionic liquids have also been explored. The efficient valorization of biomass for energy-derived biocrudes is essential for effective waste management. However, the production of biocrudes with high energy and reduced oxygen contents during the liquefaction process requires further insight. Therefore, the impact of reaction temperature, residence time, and ethanol: acetone on the energy compositions and bioproduct's yield enhancement were investigated. The biocrudes obtained were characterized using elemental analysis, GC-MS, FTIR, GPC and TGA to understand the effects of process parameters on the biocrudes' compositions. An improved HHV (38.18 MJ/kg) and lower O/C ratio (0.11) were obtained at 430 °C, 35 min and 50% ethanol with a significant improvement in the enhancement factor, deoxygenation, and percentage hydrogenation of 2.63, 36.88%, and 77.87%, respectively. 5-hydroxymethylfurfural (HMF) oxidation in aqueous media using visible photocatalysis is a green and sustainable route for the valorization of lignocellulosic biomass derivatives. Several semiconductors have already been applied for this purpose; however, the use of Poly(heptazine imides), which has high crystallinity and a special cation exchange property that allows the replacement of the cation held between the layers of C 3 N 4 structure by transition metal ions (TM), remains scarce. In this study, PHI(Na) was synthesized using a melamine/NaCl method and used as precursor to prepare metal (Fe, Co, Ni, or Cu)-doped PHI catalysts. The catalysts were tested for selective oxidation of HMF to 2,5-diformylfuran (DFF) in water and O 2 atmosphere under blue LED radiation. The catalytic results revealed that the 0.1 wt% PHI(Fe) catalyst is the most efficient photocatalyst while higher Fe loading (1 and 2 wt%) favors the formation of Fe 3+ clusters, which are responsible for the drop in HMF oxidation. Moreover, the 0.1 wt% PHI(Fe) photocatalyst has strong oxidative power due to its efficiency in H 2 O 2 production, thus boosting the generation of nonselective hydroxyl radicals ( ● OH) via different pathways that can destroy HMF. We found that using 50 mM, the highest DFF production rate (393 μmol·h -1 ·g -1 ) was obtained in an aqueous medium under visible light radiation.

A hydrothermal process is a sustainable approach for biorefinery leading to conversion of lignocellulosic (LC) biomass into value-added products. This study is based on the production of xylooligosaccharides (XOS) from pineapple leaves (PL) waste by using mild acid like gluconic acid (GA). GA, when used as catalyst in hydrothermal process to produce XOS the yield improved. The above process can be integrated with bacterial cellulose (BC) production bioprocess via Komagataeibacter europaeus 14,148 where gluconic acid is produced as by-product. Maximum XOS (2-5 degree of polymerisation) yield of 67.79 % in the liquid fraction was obtained via hydrothermal treatment at 160 °C for 60 min with 5% gluconic acid concentration. It is based on the selective solubilization of hemicellulose fraction. Paunch is comprised of the partially digested feed contained in cattle or sheep and contributes 20-50% of organic waste produced at red meat processing facilities. Anaerobic digestion has been identified as a promising technology for paunch treatment, however treatment times can be long and when combined with the moderate degradability of paunch this results in high treatment costs that need to be improved. Pre-treatment was investigated as a strategy to improve AD of paunch, alkaline treatment (NaOH or KOH) was selected due to the high lignin content. A range of alkaline loadings (1-20 g 100gTS -1 ) were tested with an equivalent hydroxide molar concentration of 9-250 mM [OH - ]. Alkaline pre-treatment improved both the hydrolysis rate and the overall degradability of paunch solid by up to 4.4 times and 60%, respectively. The enhanced hydrolysis rate and methane yield was correlated to changes in material composition during pre-treatment. This work aimed to study an integrated pretreatment technology employing p-toluenesulfonic acid (TsOH)-catalyzed liquid hot water (LHW) and short-time ball milling for the complete conversion of poplar biomass to xylooligosaccharides (XOS), glucose, and native-like lignin. The optimized TsOH-catalyzed LHW pretreatment solubilized 98.5% of hemicellulose at 160 °C for 40 min, releasing 49.8% XOS. Moreover, subsequent ball milling (20 min) maximized the enzymatic hydrolysis of cellulose from 65.8% to 96.5%, owing to the reduced particle sizes and cellulose crystallinity index. The combined pretreatment reduced the crystallinity by 70.9% while enlarging the average pore size and pore volume of the substrate by 29.5% and 52.4%, respectively. The residual lignin after enzymatic hydrolysis was rich in β-O-4 linkages (55.7/100 Ar) with less condensed structures. This lignin exhibited excellent antioxidant activity (RSI of 66.22) and ultraviolet absorbance. The concept of biorefinery has been advancing globally and organosolv pretreatment strategy has seen an upsurge in research due to its efficiency in removing the recalcitrant lignin and dissolution of cellulose. The high-performance organosolv system uses green solvents and its reusability contributes concurrently to the biorefinery sector and sustainability. The major advantage of the current system involves the continuous removal of lignin to enhance cellulose accessibility, thereby easing the later biorefinery steps, which were immensely restricted due to the recalcitrant lignin. The current system process can be further explored and enhanced via the amalgamation of new technologies, which is still a work in progress. Thus, the current review summarizes organosolv pretreatment and the range of solvents used, along with a detailed mechanistic approach that results in efficient pretreatment of LCB. The latest developments for designing high-performance pretreatment systems, their pitfalls, and advanced assessments such as Life Cycle Assessment along with Techno-Economic Assessment have also been deliberated to allow an insight into its diverse potential applicability towards a sustainable future., (© 2023. The complex and heterogeneous nature of the lignin macromolecule has presented a lasting barrier to its utilization. To achieve high lignin yield, the technical lignin extraction process usually severely modifies and condenses the native structure of lignin, which is a critical drawback for its utilization in conversion processes. In addition, there is no method capable of separating lignin from plant biomass with controlled structural properties. Here, we developed an N-heterocycle-based deep eutectic solvent formed between lactic acid and pyrazole (La-Py DES) with a binary hydrogen bonding functionality resulting in a high affinity toward lignin. Up to 93.7% of lignin was extracted from wheat straw biomass at varying conditions from 90 °C to 145 °C. Through careful selection of treatment conditions as well as lactic acid to pyrazole ratios, lignin with controlled levels of ether linkage content, hydroxyl group content, and average molecular weight can be generated. Under mild extraction conditions (90 °C to 120 °C), light-colored native-like lignin can be produced with up to 80% yield, whereas ether linkage-free lignin with low polydispersity can be obtained at 145 °C. Overall, this study offers a new strategy for native lignin extraction and generating lignin with controlled structural properties.

Co-hydrothermal carbonization (co-HTC) of lignocellulose biomass (LB) and chlorinated waste could produce value-added co-hydrochar while simultaneously removing inorganic metal salts and organic chlorine to the liquid phase. However, there is a lack of understanding of the influence of LB feedstocks on the fuel properties and combustion behaviors of co-hydrochars. Therefore, co-hydrochars derived from co-HTC of pine, bamboo, corncob, wheat stalk, and corn stalk with polyvinyl chloride (PVC) at the mass ratio of 9:1 under 260 °C for 30 min were tested. PVC facilitated the hydrolysis, dehydration, and polymerization of LB compositions (hemicellulose, cellulose, soluble lignin, and insoluble lignin). In turn, these LB compositions could prevent PVC aggregation and promote PVC substitution. Hydrochar fragments could coat the PVC surface and hinder its hydrolysis. Interactions between LB compositions and PVC improved the fuel properties and combustion behaviors of co-hydrochars derived from bamboo, corncob, wheat stalk, and corn stalk while decreasing the fuel properties and combustion behaviors of co-hydrochar derived from pine (HC-PPE). Except for HC-PPE, the fuel ratio (fixed carbon/volatile matter) of co-hydrochars increased to 0.90-1.18 and their HHVs reached approximately 17.5-32.45 MJ/kg without an increased risk of chlorine corrosion. The combustion of co-hydrochars was easier and more stable due to their higher ignition and burnout temperatures and lower activation energies. Co-hydrothermal carbonization (co-HTC) of lignocellulose biomass (LB) and chlorinated waste can simultaneously remove organic chlorine and inorganics, however, the interaction mechanisms are unclear owing to the variety of operating conditions and complexity of biomass compositions. Pine, bamboo, corncob, corn stalk, and wheat straw were co-hydrothermally carbonized with polyvinyl chloride (PVC) at the mass ratio of 9:1 for 30 min under 260 °C to explore the fundamental interactions. The synergistic index (SI) of dechlorination efficiency ranged from -20.3 % to 19.9 %, indicating the interaction depended on the content and composition of cellulose, hemicellulose, and lignin in the LB feedstocks. Hydroxyl functional groups in cellulose and soluble lignin dehydration intermediates promoted PVC substitution. The LB fragments prevented PVC aggregation while promoted PVC fragmentation, thereby facilitating dechlorination. The polyaromatic hydrochar derived from insoluble lignin and polymeric hydrochar derived from hemicellulose, cellulose, and soluble lignin can coat the surface of molten PVC and act as significant dechlorination inhibitors. All SI of removal efficiency of inorganics (RE) were positive, ranging from 0.74 % to 154 %, with large variations for different inorganics, indicating that inorganics contents in LB influenced RE significantly. A large amount of water-insoluble/acid-soluble inorganics was removed via a metathesis reaction. One of the main strategies for sustainable human society progress is the development of efficient strategies to limit waste production and maximize renewable resource utilization. In this context, this review highlights the opportunity to transform vegetable biomass residues into valuable commercial products. Biomass conversion entails the depolymerization of lignocellulosic biomass towards biopolyols and the synthesis and characterization of the valuable products obtained by using them. The influence of the reaction parameters in both acid and basic catalysis is highlighted, respectively the influence of microwaves on the liquefaction reaction versus conventional heating. Following the depolymerization reaction, polyols are employed to produce polyurethane foams. As a special characteristic, the addition of flame-retardant properties was emphasized. Another interesting topic is the biodegradability of these products, considering the negative consequences that waste accumulation has on the environment.

Extremophiles provide a one-of-a-kind source of enzymes with properties that allow them to endure the rigorous industrial conversion of lignocellulose biomass into fermentable sugars. However, the fact that most of these organisms fail to grow under typical culture conditions limits the accessibility to these enzymes. In this study, we employed a functional metagenomics approach to identify carbohydrate-degrading enzymes from Ethiopian soda lakes, which are extreme environments harboring a high microbial diversity. Out of 21,000 clones screened for the five carbohydrate hydrolyzing enzymes, 408 clones were found positive. Cellulase and amylase, gave high hit ratio of 1:75 and 1:280, respectively. A total of 378 genes involved in the degradation of complex carbohydrates were identified by combining high-throughput sequencing of 22 selected clones and bioinformatics analysis using a customized workflow. Around 41% of the annotated genes belonged to the Glycoside Hydrolases (GH). Multiple GHs were identified, indicating the potential to discover novel CAZymes useful for the enzymatic degradation of lignocellulose biomass from the Ethiopian soda Lakes. More than 73% of the annotated GH genes were linked to bacterial origins, with Halomonas as the most likely source. Biochemical characterization of the three enzymes from the selected clones (amylase, cellulase, and pectinase) showed that they are active in elevated temperatures, high pH, and high salt concentrations. Carotenoids are widely utilized in the food, pharmaceutical and nutraceutical industries. Here, Kluyveromyces marxianus was engineered to overproduce carotenoids from corncob hydrolysate or xylose mother liquid (XML, a byproduct of commercial xylose purification). First, the toxicity of fat-soluble carotenoids to cells was reduced by employing xylose inducible promoters using with a two-temperature strategy to separate cell growth and product accumulation. Then, through further engineering and optimization of the carotenoid biosynthesis pathway, 1506.7 mg/L lycopene, 988.5 mg/L β-carotene or 142.9 mg/L astaxanthin were produced with glucose and xylose by K. marxianus. Finally, 397.7 mg/L and 279.7 mg/L lycopene, 297.3 mg/L and 108.8 mg/L β-carotene, and 86.4 mg/L and 56.8 mg/L astaxanthin were produced with nonsterilized andnondetoxified XML or corncob hydrolysate after nitrogen source optimization. A thermophilic Geobacillus bacterial strain, WSUCF1 contains different carbohydrate-active enzymes (CAZymes) capable of hydrolyzing hemicellulose in lignocellulosic biomass. We used proteomic, genomic, and bioinformatic tools, and genomic data to analyze the relative abundance of cellulolytic, hemicellulolytic, and lignin modifying enzymes present in the secretomes. Results showed that CAZyme profiles of secretomes varied based on the substrate type and complexity, composition, and pretreatment conditions. The enzyme activity of secretomes also changed depending on the substrate used. The secretomes were used in combination with commercial and purified enzymes to carry out saccharification of ammonia fiber expansion (AFEX)-pretreated corn stover and extractive ammonia (EA)-pretreated corn stover. When WSUCF1 bacterial secretome produced at different conditions was combined with a small percentage of commercial enzymes, we observed efficient saccharification of EA-CS, and the results were comparable to using a commercial enzyme cocktail (87% glucan and 70% xylan conversion). It also opens the possibility of producing CAZymes in a biorefinery using inexpensive substrates, such as AFEX-pretreated corn stover and Avicel, and eliminates expensive enzyme processing steps that are used in enzyme manufacturing. Anaerobic digestion (AD) is a bioprocess technology that integrates into circular economy systems, which produce renewable energy and biofertilizer whilst reducing greenhouse gas emissions. However, improvements in biogas production efficiency are needed in dealing with lignocellulosic biomass. The state-of-the-art of AD technology is discussed, with emphasis on feedstock digestibility and operational difficulty. Solutions to these challenges including for pre-treatment and bioaugmentation are reviewed. This article proposes an innovative integrated system combining alkali pre-treatment, temperature-phased AD and bioaugmentation techniques. The integrated system as modelled has a targeted potential to achieve a biodegradability index of 90% while increasing methane production by 47% compared to conventional AD. The methane productivity may also be improved by a target reduction in retention time from 30 to 20 days. Glycerol waste was combined with microwave to pretreat lignocellulose before fast pyrolysis. After pretreatment, most alkali and alkaline earth metals (87.9%) and lignin (52.6%) were removed, and a higher crystallinity was obtained. Comparatively, glycerol waste combined with microwave was proven to be more efficient than glycerol with conventional heating. During fast pyrolysis, higher content of levoglucosan in glycerol waste-pretreated products (27.5%) was obtained, compared with those pretreated by pure glycerol (18.8%) and untreated samples (5.8%). Production of fermentative toxic aldehyde and phenol by-products was also inhibited after glycerol waste treatment. Following mechanistic study had validated that microwave in glycerol waste solvent could effectively ameliorate structure and components of lignocellulose while selectively removing lignin. Notably, under the optimal condition, the levoglucosan content in pyrolytic products was enhanced significantly from 5.8% to 32.9%. A halophilic marine bacterial strain, PS-C1, was isolated from Sekinchan beach in Selangor, Malaysia. The 16S rRNA gene sequence analysis indicated that strain PS-C1 was associated with the genus Celeribacter . To date, there have been no reports on enzymes from the genus Celeribacter . The present study reports on the cellular features of Celeribacter sp. PS-C1, its annotated genome sequence, and comparative genome analyses of Celeribacter glycoside hydrolase (GH) enzymes. The genome of strain PS-C1 has a size of 3.87 Mbp and a G+C content of 59.10%, and contains 3739 protein-coding genes. Detailed analysis using the Carbohydrate-Active enZYmes (CAZy) database revealed that Celeribacter genomes harboured at least 12 putative genes encoding industrially important GHs that are grouped as cellulases, β-glucanases, hemicellulases, and starch-degrading enzymes. Herein, the potential applications of these enzymes are discussed. Furthermore, the activities of two types of GHs (β-glucosidase and licheninase) in strain PS-C1 were demonstrated. These findings suggest that strain PS-C1 could be a reservoir of novel GH enzymes for lignocellulosic biomass degradation.

Objectives: The research aims to analyze the catabolic strength of different hydrolytic enzymes in assessing the biological conversion potential of lignocellulose parts of agricultural biomass wastes into functional edible sugars and biofuels., Materials and Methods: The enzymes' hydrolytic properties-versatile peroxidase, manganese peroxidase, and lignin peroxidase were used to identify their complexing strength with the lignin substrate, whereas endoglucanase cel12A, acidocaldarius cellulase, and Melanocarpus albomyces endoglucanase were tested on the cellulose gel substrate. Because the biodegradation properties are heavily influenced by the "enzyme-substrate complexing energy level," proper molecular optimization and energy minimization of the enzymes and substrates were carried out, as well as the identification of the enzyme's active sites prior to complexing.comprehensive molecular dynamic simulation was run to study their-alpha carbon, root-mean-square deviation ( Å ), molecular surface area ( Å2 ), root-mean-square fluctuation ( Å ), radius of gyration (nm), hydrogen bonds with hydrophobic interactions, and solvent accessible surface area ( Å2 ) values for 50 ns. The simulated data mining was conducted using advanced programming algorithms to establish the final enzyme-substrate complexing strength in binding and catalysis., Results: Among the lignin-degrading enzymes, versatile peroxidase shows promising catalytic activity with the best docking pose and significant values in all the dynamic simulation parameters. Similarly, Melanocarpus albomyces endoglucanase shows the best activity in all aspects of molecular docking and dynamics among the cellulose-degrading enzymes., Conclusion: The lignin content of biomass wastes can be degraded into cellulose and hemicellulose using lignin-degrading enzymes. The cellulose can be further degraded into glucose and xylose sugars following the cellulose-degrading enzyme activity. These sugars can be further degraded into biofuel through anaerobic fermentation. Ionic liquid (IL) pretreatment of lignocellulose is an efficient method for the enhancement of enzymatic saccharification. However, the remaining residues of ILs deactivate cellulase, therefore making intensive biomass washing after pretreatment necessary. This study aimed to develop the one-pot process combining IL pretreatment and enzymatic saccharification by using low-toxic choline acetate ([Ch][OAc]) and IL-tolerant bacterial cellulases. Crude cellulases produced from saline soil inhabited Bacillus sp. CBD2 and Brevibacillus sp. CBD3 were tested under the influence of 0.5-2.0 M [Ch][OAc], which showed that their activities retained at more than 95%. However, [Ch][OAc] had toxicity to CBD2 and CBD3 cultures, in which only 32.85% and 12.88% were alive at 0.5 M [Ch][OAc]. Based on the specific enzyme activities, the sugar amounts produced from one-pot processes using 1 mg of CBD2 and CBD3 were higher than that of Celluclast 1.5 L by 2.0 and 4.5 times, respectively, suggesting their potential for further application in the biorefining process of value-added products.

The production of chemicals and fuels from renewable biomass with the primary aim of reducing carbon footprints has recently become one of the central points of interest. The use of lignocellulosic biomass for energy production is believed to meet the main criteria of maximizing the available global energy source and minimizing pollutant emissions. However, before usage in bioenergy production, lignocellulosic biomass needs to undergo several processes, among which biomass pretreatment plays an important role in the yield, productivity, and quality of the products. Acid-based pretreatment, one of the existing methods applied for lignocellulosic biomass pretreatment, has several advantages, such as short operating time and high efficiency. A thorough analysis of the characteristics of acid-based biomass pretreatment is presented in this review. The environmental concerns and future challenges involved in using acid pretreatment methods are discussed in detail to achieve clean and sustainable bioenergy production. The presence of heat, methane (CH 4 ) and oxygen in landfill sub-surface causes initiation of spontaneous waste ignition posing severe environmental impacts. A municipal solid waste (MSW) reactor (trough) was designed to monitor landfill gases (LFGs) i.e., CH 4 and CO 2 and its potential from different waste categories (synthetic waste, fresh waste, 3-month, 6-month, 3-year and 5-year-old waste) collected from open MSW dumpsite. The quantity of cellulose (C), hemicellulose (H) and lignin (L) contents (C + H: L) present in organic waste fraction of each waste category was determined. Results showed that fresh waste which has higher ratio of C + H: L is responsible for maximum CH 4 and CO 2 generation i.e., 31,660 and 46,078 ml/g of volatile solid, respectively. The ratio of C + H: L observed in fresh waste, 3-month, 6-month, 3-year and 5-year-old waste was 2.62, 1.70, 1.32, 1.21 and 1, respectively. The study also showed that LFG generation is directly proportional to lignocellulose biomass contents present in MSW. Omics have given rise to research on sparsely studied microbial communities such as the landfill, lignocellulolytic microorganisms and enzymes. The bacterial diversity of Municipal Solid Waste sediments was determined using the illumina MiSeq system after DNA extraction and Polymerase chain reactions. Data analysis was used to determine the community's richness, diversity, and correlation with environmental factors. Physicochemical studies revealed sites with mesophilic and thermophilic temperature ranges and a mixture of acidic and alkaline pH values. Temperature and moisture content showed the highest correlation with the bacteria community. The bacterial analysis of the community DNA revealed 357,030 effective sequences and 1891 operational taxonomic units (OTUs) assigned. Forty phyla were found, with the dominant phyla Proteobacteria , Firmicutes , Actinobacteria , and Bacteroidota , while Aerococcus , Stenotrophomonas , and Sporosarcina were the dominant species. PICRUSt provided insight on community's metabolic function, which was narrowed down to search for lignocellulolytic enzymes' function. Cellulase, xylanase, esterase, and peroxidase were gene functions inferred from the data. This article reports on the first phylogenetic analysis of the Pulau Burung landfill bacterial community. These results will help to improve the understanding of organisms dominant in the landfill and the corresponding enzymes that contribute to lignocellulose breakdown.

We are reporting the novel cellulase named Cel-5M from rumen metagenome. The deduced amino acid sequence and biochemical characterization suggested that Cel-5M is endoglucanase from the GH5 family with multifunctional potential. Cel-5M showed similarity to non-characterized proteins and the genus Prevotella as parental organism. The 957 bp ORF encoding Cel-5M was cloned and overexpressed in E. coli and purified. The recombinant Cel-5M showed maximum activity at pH 6.0 and 40 °C. It retained more than 80% activity between 4 and 7 pH and 65% thermostability between 30 and 70 °C. Cel-5M showed activity on various substrates like CMC, Filter paper, Avicel, Xylan, β-Mannan, and Glucopyranoside, which confirmed its multifunctional characteristics and classifies as member of subfamily 4 of GH5 (GH5&#95;4). LCB hydrolysis potential of Cel-5M was studied using wheat straw (10% w/v). Alkali-treated and steam-exploded wheat straws were inoculated with 1 mg/g Cel-5M, 2% yeast in a reaction mixture and SSF at 10% w/v loading rate. The ethanol yield 0.46 g/g and 0.43 g/g of cellulose obtained after 72 h fermentation in alkali-treated and steam-exploded wheat straw, respectively. Formaldehyde is a colorless, highly toxic, and flammable gas that is harmful to human health. Recently, many efforts have been devoted to the application of activated carbon to absorb formaldehyde. In this work, lignocellulose-based activated carbon fiber paper (LACFP) loaded with manganese dioxide (MnO 2 ) was fabricated for the adsorption and in-situ catalytic degradation of formaldehyde. LACFP was prepared by two-stage carbonization and activation of sisal hemp pulp-formed paper and was then impregnated with manganese sulfate (MnSO 4 ) and potassium permanganate (KMnO 4 ) solutions; MnO 2 then formed by in situ growth on the LACFP base by calcination. The catalytic performance of MnO 2 -loaded LACFP for formaldehyde was then investigated. It was found that the suitable carbonization conditions were elevating the temperature first by raising it at 10°C/min from room temperature to 280°C, then at 2°C/min from 280 to 400°C, maintaining the temperature at 400°C for 1 h, and then increasing it quickly from 400 to 700°C at 15°C/min. The conditions used for activation were similar to those for carbonization, with the temperature additionally being held at 700°C for 2 h. The conditions mentioned above were optimized to maintain the fiber structure and shape integrity of the paper, being conducive to loading with catalytically active substances. Regarding the catalytic activity of MnO 2 -loaded LACFP, the concentration of formaldehyde decreased by 59 ± 6 ppm and the concentration of ΔCO 2 increased by 75 ± 3 ppm when the reaction proceeded at room temperature for 10 h. Vitamins are important nutrients for many fermentations, but they are generally costly. Agricultural lignocellulose biomass contains considerable amounts of vitamin B compounds, but these water-soluble vitamins are easily lost into wastewater discharge during pretreatment or detoxification of lignocellulose in biorefinery processes. Here, we showed that the dry acid pretreatment and biodetoxification process allowed the preservation of significant amounts of vitamin B, which promoted l-lactic acid fermentation efficiency significantly. Supplementation with specific vitamin B compounds, VB 3 and VB 5 , into corn stover hydrolysate led to further increases of cellulosic l-lactic acid yield and fermentation rates. This study provided a new solution for the enhancement of biorefinery fermentation efficiency by using vitamin B compounds in lignocellulose biomass.

Biorefinery has been suggested to provide relevant substitutes to a number of fossil products. Feedstocks and conversion technologies have, however, been the bottleneck to the realization of this concept. Herein, feedstocks and bioconversion technologies under biorefinery have been reviewed. Over the last decade, research has shown possibilities of generating tens of new products but only few industrial implementations. This is partly associated with low production yields and poor cost-competitiveness. This review addresses the technical barriers associated with the conversion of emerging feedstocks into chemicals and bioenergy platforms and summarizes the developed biotechnological approaches including advances in metabolic engineering. It is envisaged that the utilization of lignocellulosic biomass for ethanol production for transport sector, would make cellulases the most demanded industrial enzyme. The greatest potential of cellulolytic enzymes lies in ethanol production from biomass by enzymatic hydrolysis of cellulose but low thermostability and low titer of cellulase production resulting into high cost of the enzyme which is the major set-back. A number of research groups are working on cellulase to improve its thermostability so as to be able to perform hydrolysis at elevated temperatures which would eventually increase the efficiency of cellulose hydrolysis. The technologies developed from lignocellulosic biomass via cellulose hydrolysis promise environmental and economical sustainability in the long run along with non-dependence on nonrenewable energy source. A total of 123 Trichoderma strains were isolated from different habitats and tested for their ability to degrade cellulose and xylan by simple plate screening method. Among strains, more than 34 and 45% respectively, exhibited higher cellulolytic and xylanolytic activity, compared to the reference strain T. reesei QM 9414. For strains efficiently degrading cellulose, a highest enzyme activity was confirmed using filter paper test, and it resulted in a range from 1.01 to 7.15 FPU/ml. Based on morphological and molecular analysis, the isolates were identified as Trichoderma. The most frequently identified strains belonged to Trichoderma harzianum species. Among all strains, the most effective in degradation of cellulose and xylose was T. harzianum and T. virens, especially those isolated from forest wood, forest soil or garden and mushroom compost. The results of this work confirmed that numerous strains from the Trichoderma species have high cellulose and xylan degradation potential and could be useful for lignocellulose biomass conversion e.g. for biofuel production.

The mixture of Eucalyptus globulus wood (EGW) and cheese whey powder (CWP) was proposed for intensification of simultaneous saccharification and fermentation (SSF) at high temperature and solid loadings using the industrial Saccharomyces cerevisiae Ethanol Red® strain. High ethanol concentration (93 g/L), corresponding to 94% ethanol yield, was obtained at 35 °C from 37% of solid mixture using cellulase and β-galactosidase enzymes (24.2 FPU/g and 20.0 U/g, respectively). The use of CWP mixed with pretreated EGW increased the ethanol concentration in 1.5-fold, in comparison with SSF experiments without CWP for both Ethanol Red® and CEN.PK113-7D strains. Moreover, 1.4-fold higher ethanol concentration was obtained with Ethanol Red®, in comparison with CEN.PK113-7D strain. Ethanol Red® strain was genetically engineered for β-galactosidase production in order to advance towards a fully integrated process. Bio-refinery processes require use of the most suitable lignocellulosic biomass for enzymatic saccharification and microbial fermentation. Glucose yield from biomass solid fractions obtained after dilute sulfuric acid (1%) pretreatment (at 180 °C) was investigated using 14, 8, and 16 varieties of rice, wheat, and sorghum, respectively. Biomass solid fractions of each crop showed similar cellulose content. However, glucose yield after enzymatic hydrolysis (cellulase loading at 6.6 filter paper unit/g-biomass) was different among the varieties of each crop, indicating genotypic differences for rice, wheat, and sorghum. Nuclear magnetic resonance method revealed that the high residual level of lignin aromatic regions decreased glucose yield from solid fraction of sorghum.

Eucalyptus grandis and Eucalyptus globulus are among the most widely cultivated trees, differing in lignin composition and plantation areas, as E. grandis is mostly cultivated in tropical regions while E. globulus is preferred in temperate areas. As temperature is a key modulator in plant metabolism, a large-scale proteome analysis was carried out to investigate changes in the antioxidant system and the lignification metabolism in plantlets grown at different temperatures. Our strategy allowed the identification of 3111 stem proteins. A total of 103 antioxidant proteins were detected in the stems of both species. Hierarchical clustering revealed that alterations in the antioxidant proteins are more prominent when Eucalyptus seedlings were exposed to high temperature and that the superoxide isoforms coded by the gene Eucgr.B03930 are the most abundant antioxidant enzymes induced by thermal stimulus. Regarding the lignin biosynthesis, our proteomics approach resulted in the identification of 13 of the 17 core proteins involved in this metabolism, corroborating with gene predictions and the proposed lignin toolbox. Quantitative analyses revealed significant differences in 8 protein isoforms, including the ferulate 5-hydroxylase isoform F5H1, a key enzyme in catalyzing the synthesis of sinapyl alcohol, and the cinnamyl alcohol dehydrogenase isoform CAD2, the last enzyme in monolignol biosynthesis. Activated carbon fibers (ACFs) with different pore structure have been prepared from wood sawdust using the KOH activation method. A study was conducted to examine the influence of the activation parameters (temperature, alkali/carbon ratio, and time) on the morphology and structure of the as-prepared ACFs developed in the process of pore generation and evolution. Activation temperature was very essential for the formation of utramicropores (<0.6 nm), which greatly contributed to the electric double layer capacitance. The significance of metallic potassium vapor evolved when the temperature was above 800 °C, since the generation of 0.8- and 1.1 nm micropores cannot be ignored. When the the KOH/fiber ratio was increased and the activation time was prolonged, to some extent, the micropores were enlarged to small mesopores within 2-5 nm. The sample with the optimal condition exhibited the highest specific capacitance (225 F g(-1) at a current density of 0.5 A g(-1)). Its ability to retain capacitance corresponding to 10 A g(-1) and 6 M KOH was 85.3%, demonstrating a good rate capability. With 10 000 charge-discharge cycles at 3 A g(-1), the supercapacitor kept 94.2% capacity, showing outstanding electrochemical performance as promising electrode material.

Interest in developing a sustainable technology for fuels and chemicals has unleashed tremendous creativity in metabolic engineering for strain development over the last few years. This is driven by the exceptionally recalcitrant substrate, lignocellulose, and the necessity to keep the costs down for commodity products. Traditional methods of gene expression and evolutionary engineering are more effectively used with the help of synthetic biology and -omics techniques. Compared to the last biomass research peak during the 1980s oil crisis, a more diverse range of microorganisms are being engineered for a greater variety of products, reflecting the broad applicability and effectiveness of today's gene technology. We review here several prominent and successful metabolic engineering strategies with emphasis on the following four areas: xylose catabolism, inhibitor tolerance, synthetic microbial consortium, and cellulosic oligomer assimilation.

Plant biomass is an abundant renewable natural resource that can be transformed into chemical feedstocks. Enzymes used in saccharification of lignocellulosic biomass are a major part of biofuel production costs. A cocktail of cellulolytic and xylanolytic enzymes are required for optimal saccharification of biomass. Accordingly, thirty-two fungal pure cultures were obtained from surface soil-biomass mixtures collected from Black Belt sites in Alabama by culturing on lignocellulosic biomass medium. The fungal strains were screened for the coproduction of cellulolytic and xylanolytic enzymes. Strains that displayed promising levels of cellulolytic and xylanolytic enzymes were characterized by molecular analysis of DNA sequences from the large subunit and internal transcribed spacer (ITS) of their ribosomal RNA gene. Nucleotide sequence analysis revealed that two most promising isolates FS22A and FS5A were most similar to Penicillium janthinellum and Trichoderma virens. Production dynamics of cellulolytic and xylanolytic enzymes from these two strains were explored in submerged fermentation. Volumetric productivity after 120 h incubation was 121.08 units/L/h and 348 units/L/h for the filter paper cellulase and xylanase of strain FS22A, and 90.83 units/L/h and 359 units/L/h, respectively for strain FS5A. Assays with 10 times dilution of enzymes revealed that the activities were much higher than that observed in the crude culture supernatant. Additionally, both FS22A and FS5A also produced amylase in lignocellulose medium. Lignocellulose biomass, one of the most abundant renewable resources on the planet, is an alternative sustainable energy source for the production of second-generation biofuels. Energy in the form of simple or complex carbohydrates can be extracted from lignocellulose biomass and fermented by microorganisms to produce bioethanol. Despite 40 years of active and cutting-edge research invested into the development of technologies to produce bioethanol from lignocellulosic biomass, the process remains commercially unviable. This review describes the achievements that have been made in generating microorganisms capable of utilizing both simple and complex sugars from lignocellulose biomass and the fermentation of these sugars into ethanol. We also provide a discussion on the current "roadblocks" standing in the way of making second-generation bioethanol a commercially viable alternative to fossil fuels., (Copyright © 2015 Elsevier Inc.
