Myasthenia gravis (MG) presents with symptoms that significantly affect patients' daily lives. Long-term MG therapies may lead to substantial side effects, predominantly due to prolonged immune suppression. Sirt6, which plays a vital role in maintaining cellular homeostasis and is recognised for its involvement in cytokine production in immune cells, has not yet been explored in relation to MG. PBMCs and CD4 + T cells were isolated from blood samples. RT-qPCR, western blot and ELISA were used to assess the expression of target genes and proteins. Flow cytometry was used to identify the subsets of T helper cells. Co-IP was conducted to investigate the interaction between USP9X and Sirt6. Finally, the experimental autoimmune myasthenia gravis (EAMG) model was established. In MG patients, Sirt6 levels were downregulated compared to healthy controls. Sirt6 overexpression led to a reduction in Th1 and Th17 cell populations while augmenting Treg cells in PBMCs. USP9X interacted with Sirt6, leading to its deubiquitination and stabilisation. Elevated Sirt6 levels subsequently mitigated symptoms in the EAMG model. The stabilisation of Sirt6, mediated by USP9X, has been found to relieve symptoms of EAMG by influencing the subtypes of T helper cells. With the rapid development of Internet of Things (IoT) technology, embedded devices in various computer vision scenarios can realize real-time target detection and recognition tasks, such as intelligent manufacturing, automatic driving, smart home, and so on. YOLOv8, as an advanced deep learning model in the field of target detection, has attracted much attention for its excellent detection speed, high precision, and multi-task processing capability. However, since IoT embedded devices typically own limited computing resources, direct deployment of YOLOv8 is a big challenge, especially for real-time detection tasks. To address this vital issue, this work proposes and deploys an optimized lightweight real-time detection network model that well-suits for IoT embedded devices, denoted as FRYOLO. To evaluate its performance, a case study based on real-time fresh and defective fruit detection in the production line is performed. Characterized by low training cost and high detection performance, this model accurately detects various types of fruits and their states, as the experimental results show that FRYOLO achieves 84.7% in recall and 89.0% in mean Average Precision (mAP), along with a precision of 92.5%. In addition, it provides a detection frame rate of up to 33 Frames Per Second (FPS), satisfying the real-time requirement. Finally, an intelligent production line system based on FRYOLO is implemented, which not only provides robust technical support for the efficient operation of fruit production processes but also demonstrates the availability of the proposed network model in practical IoT applications., Competing Interests: Declarations. Piezoelectric catalysis possesses the potential to convert ocean wave energy into and holds broad prospects for extracting uranium from seawater. Herein, the Z-type ZnO@COF heterostructure composite with excellent piezoelectric properties was synthesized through in situ growth of covalent organic frameworks (COFs) on the surface of ZnO and used for efficient uranium extraction. The designed COFs shell enables ZnO with stability, abundant active sites and high-speed electron transport channels. Meanwhile, the interface electric field established in the heterojunctions stimulates electron transfer from COFs to ZnO, which break the edge shielding effect of the ZnO's metallic state. Additionally, the polarization of ZnO is enhanced by heterogeneous engineering, which ensures the excellent piezocatalytic performance. As a result, ZnO@COF achieves an ultra-high efficiency of 7.56 mg g -1 d -1 for uranium extraction from natural seawater driven by waves. Background: Uterine injury can cause uterine scarring, leading to a series of complications that threaten women's health. Uterine healing is a complex process, and there are currently no effective treatments. Although our previous studies have shown that bone marrow mesenchymal stem cells (BMSCs) promote uterine damage repair, the underlying mechanisms remain unclear. However, exploring the specific regulatory roles of BMSCs in uterine injury treatment is crucial for further understanding their functions and enhancing therapeutic efficacy., Aim: To investigate the underlying mechanism by which BMSCs promote the process of uterine healing., Methods: In in vivo experiments, we established a model of full-thickness uterine injury and injected BMSCs into the uterine wound. Transcriptome sequencing was performed to determine the enrichment of differentially expressed genes at the wound site. In in vitro experiments, we isolated rat uterine smooth muscle cells (USMCs) and cocultured them with BMSCs to observe the interaction between BMSCs and USMCs in the microenvironment., Results: We found that the differentially expressed genes were mainly related to cell growth, tissue repair, and angiogenesis, while the phosphoinositide 3-kinase (PI3K)/protein kinase B (AKT) pathway was highly enriched. Quantitative reverse-transcription polymerase chain reaction was used to validate differentially expressed genes, and the results demonstrated that BMSCs can upregulate genes related to regeneration and downregulate genes related to inflammation. Coculturing BMSCs promoted the migration and proliferation of USMCs, and the USMC microenvironment promoted the myogenic differentiation of BMSCs. Eutectic-based polymer electrolytes have emerged as promising solid electrolytes because of their ionic liquid-like properties, while modifications are essential to further increase their ionic conductivity at room temperature and solve their compatibility with lithium anode. In this work, an in situ polymerized composite electrolyte is modified by the addition of fluoroethylene carbonate (FEC) whose beneficial effect is systematically investigated in different contents. Poly(ethylene glycol) diacrylate (PEGDA), deep eutectic solvent (LiTFSI: N -methylacetamide = 1:3), and alumina fiber work as the monomer, solvent, and three-dimensional skeleton, respectively. In adjusting FEC content, ionic conductivity at room temperature is dramatically raised by three times to 8.93 × 10 -4 S cm -1 , with a 4-fold increase in lithium-ion transference number to 0.405. Meanwhile, the electrochemical window is widened from 3.5 to 4.8 V. The FEC addition also helps in improving the stability with Li anode, which comes from LiF-rich interphases formed at interfaces. The dynamics of LiFePO 4 is significantly enhanced with higher reversibility in full cells, so that fast capacity decay is inhibited with a specific capacity of 124.1 mAh g -1 obtained after 300 cycles at 1 C. These results provide an effective modification for the deep eutectic electrolyte, which will boost its development in solid-state batteries.

In the present study, the taxonomic position of Salisediminibacterium haloalkalitolerans was evaluated by determining the 16S rRNA gene sequence similarity, genome relatedness, and phylogenetic analyses. The 16S rRNA gene sequences extracted from the genomes of Salisediminibacterium haloalkalitolerans 10nlg T and Salisediminibacterium halotolerans DSM 26530 T showed 100% similarity, supporting their classification as the same species. The average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values between S. haloalkalitolerans and S. halotolerans were 97.7 and 80.2%, respectively, above the cutoff value (95-96% ANI; 70% dDDH) for species delineation. S. haloalkalitolerans and S. halotolerans should be classified as the same species because the 16S rRNA gene phylogeny and phylogenomic tree (based on 71 bacterial single-copy genes) showed no genetic divergence between them. Based on the above results and the rule of priority in nomenclature, we propose to reclassify Salisediminibacterium haloalkalitolerans Sultanpuram et al. 2015 as a later heterotypic synonym of Salisediminibacterium halotolerans Jiang et al. 2012., Competing Interests: Declarations. Hypoxia is a key feature of the tumor microenvironment that leads to the failure of many chemotherapies and induces more aggressive and resistant cancer phenotypes. Up to date, there are very few compounds and treatments that can target hypoxia. BE-43547A 2 from Streptomyces sp. was one of the most hypoxia-selective compounds against PANC-1, MCF-7, and K562 cell lines. In this study, we investigated the molecular mechanism underlying the hypoxia selectivity of BE-43547A 2 in human pancreatic cancer cells. We showed that BE-43547A 2 displayed hypoxia-selective cytotoxicity in five pancreatic cancer cells (PANC-1, Capan-2, MIA PaCa-2, AsPC-1, and PaTu8988T) with IC 50 values under hypoxia considerably lower than those under normoxia. We demonstrated that BE-43547A 2 is directly bound to eEF1A1 protein in PaTu8988T cells under hypoxia. Furthermore, we revealed that hypoxia significantly elevated the expression levels of HIF1α, FoxO1, and eEF1A1 in the five pancreatic cancer cells; eEF1A1 interacted with FoxO1 in the cytoplasm, which was disrupted by BE-43547A 2 followed by the nuclear translocation of FoxO1 and ultimate inhibition of JAK/STAT3 signaling pathway under hypoxia. This study reveals that BE-43547A 2 , targeting eEF1A1, disrupts its interaction with FoxO1 in human pancreatic cancer cells under hypoxia. Purpose: To determine the dosimetric effects of set-up errors on boost coverage, and compares skin toxicity of sequential and simultaneous boost techniques for left-sided breast cancer., Materials and Methods: This retrospective study included 23 early-stage breast cancer cases. Single isocenter HFWBI-SIB(s-SIB), single isocenter HFWBI-SB(s-SB) and dual isocenter HFWBI-SB(d-SB) were planing. Rotations of 0.5°, 1°, and 2° coupled with translationals of 0.5 mm, 1.0 mm, and 2.0 mm were applied along three orthogonal axes. The dose to 95% of the PTV (D95) and the volume covered by 95% of the prescribed dose (V95) were evaluated using GEE univariate analysis to determine how PTV coverage was related to 1/CI RTOG , PTVboost volume, PTVboost separation to isocenter. The relationship between the high-dose regions within the PTVbreast and Ratio_V was evaluated using univariate analysis., Results: The s-SIB had optimal target coverage and lower high-dose volume, but it increased the risk of compromised coverage to tumor bed. For the s-SB technique, V95 exceeded 95% under all setup errors. At 2.0° coupled with 2.0 mm, s-SIB and d-SB exhibited V95 values below 95% in 34.8% and 8.7% of cases, respectively. At other setup errors, both s-SIB and d-SB demonstrated V95 values greater than 95%. Objective: To investigate the physicochemical properties, structural characteristics and in vitro activities of Shancigu polysaccharides to identify the authenticity of Shancigu, safeguard its medicinal value, and ensure the safety of its market distribution., Methods: Eleven polysaccharides from different sources of Shancigu were obtained using water extraction and alcohol precipitation. Physicochemical properties were determined using colorimetry. Preliminary structural characterization was achieved using instrumental analyses, including infrared spectroscopy and gas chromatography. Anti-breast cancer and pro-inflammatory activities were compared using in vitro assays., Results: Shancigu polysaccharides mainly consist of pyranose sugars with α-type glycosidic bonds, including uronic acids, and sulfate groups. The most important monosaccharides are mannose and glucose. Glycosidic bonds were found to comprise (1 → 2) or (1 → 6) bond types. The molecular weight of Shancigu polysaccharides from different sources varied, with Cremastra appendiculata polysaccharides having the highest average molecular weight (352.713 kDa). Polysaccharides from the 11 different sources of Shancigu could all inhibit MCF-7 human breast cancer cells. Except for S2-S4, which had no significant effect, all other samples were able to promote the inflammatory response of RAW264.7., Conclusions: This study provides a theoretical reference for further in-depth research and application of Shancigu polysaccharides. Based on the physicochemical properties, structural characteristics, and biological activities, this study can accurately and rapidly identify the authenticity of Shancigu. Background: The causative genes for over 60% of inherited peripheral neuropathy (IPN) remain unidentified. This study endeavours to enhance the genetic diagnostic rate in IPN cases by conducting screenings focused on non-coding repeat expansions., Methods: We gathered data from 2424 unrelated Japanese patients diagnosed with IPN, among whom 1555 cases with unidentified genetic causes, as determined through comprehensive prescreening analyses, were selected for the study. Screening for CGG non-coding repeat expansions in LRP12 , GIPC1 and RILPL1 genes was conducted using PCR and long-read sequencing technologies., Results: We identified CGG repeat expansions in LRP12 from 44 cases, establishing it as the fourth most common aetiology in Japanese IPN. Most cases (29/37) exhibited distal limb weakness, without ptosis, ophthalmoplegia, facial muscle weakness or bulbar palsy. Neurogenic changes were frequently observed in both needle electromyography (97%) and skeletal muscle tissue (100%). In nerve conduction studies, 28 cases primarily showed impairment in motor nerves without concurrent involvement of sensory nerves, consistent with the phenotype of hereditary motor neuropathy. In seven cases, both motor and sensory nerves were affected, resembling the Charcot-Marie-Tooth (CMT) phenotype. Importantly, the mean CGG repeat number detected in the present patients was significantly shorter than that of patients with LRP12 -oculopharyngodistal myopathy (p<0.0001). Pseudouridine (Ψ) is the most abundant RNA modification in nature; however, not much is known about the biological functions of this modified nucleoside. Employing an unbiased quantitative proteomics method, we identified multiple candidate reader proteins of Ψ in RNA, including a cytoskeletal protein profilin-1 (PFN1). We demonstrated that PFN1 binds directly and selectively to Ψ-containing RNA. Additionally, we discovered approximately 4000 binding sites of PFN1 in human cells, including a known dyskerin (DKC1)-installed Ψ site in TPI1 mRNA, which encodes triosephosphate isomerase. Moreover, we showed that PFN1 and DKC1 are crucial for regulating the stability and translation efficiency of TPI1 mRNA through modulating PFN1-Ψ interaction. Together, we identified PFN1 as a reader protein of Ψ in RNA and illustrated a potential role of PFN1-Ψ interaction in post-transcriptional regulation. These findings provide new insights into the functions of Ψ in RNA biology and in modulating the expression of an important metabolic enzyme.

Fast and accurate determination of persistent organic pollutants (POPs) plays a crucial role in addressing concerns related to public security and environmental safety. Herein, a unique thin film based solid phase microextraction (denoted as TF-SPME) method was reported and used for on-site analysis of POPs via loading the TFs into a homemade sampling device and equipped on a drone, which can load up to 6 pieces of TFs at the same time. The parallel 6 pieces of TFs offered significant advantages in terms of efficiency, accuracy, cost-effectiveness and comparability of sampling. The detection limit for polychlorinated biphenyls and polyaromatic hydrocarbons was as low as 0.03 ng L -1 , far below the regulatory thresholds for drinking water prescribed by the United States Environmental Protection Agency. The standard deviations were ranged between 2.7 % and 9.9 %, showcasing its remarkable precision on POPs analysis. Then, by facilely equipping TF-SPME on a drone, remotely controlled sampling and on-site analysis in real water samples was realized. The concentrations were determined to be from 0.12 ng L -1 to 1.01 ng L -1 for PCBs and 0.53 ng L -1 to 19.93 ng L -1 for PAHs in the river water of Guangzhou downtown area. Kidney renal clear cell carcinoma (KIRC) is the most prevalent subtype of kidney cancer. Although multiple therapeutic agents have been proven effective in KIRC, their clinical application has been hindered by a lack of reliable biomarkers. This study focused on the prognostic value and function of drug absorption, distribution, metabolism, and excretion- (ADME-) related genes (ARGs) in KIRC to enhance personalized therapy. The critical role of ARGs in KIRC microenvironment was confirmed by single cell RNA-seq analysis and spatial transcriptome sequencing analysis for the first time. Then, an ADME-related prognostic signature (ARPS) was developed by the bulk RNA-seq analysis. The ARPS, created through Cox regression, LASSO, and stepAIC analyses, identified eight ARGs that stratified patients into high-risk and low-risk groups. High-risk patients had significantly poorer overall survival. Multivariate analysis confirmed the independent predictive ability of ARPS, and an ARPS-based nomogram was constructed for clinical application. Gene ontology and KEGG pathway analyses revealed immune-related functions and pathways enriched in these groups, with low-risk patients showing better responses to immunotherapy. Finally, the expression of ARGs was validated by qRT-PCR and Western blotting experiments. Mitochondria are important organelles that regulate cellular energy and biosynthesis, as well as maintain the body's response to environmental stress. Their dynamics and autophagy influence occurrence of cellular function, particularly under stressful conditions. They can generate reactive oxygen species (ROS) which is a major contributor to inflammatory diseases such as ulcerative colitis (UC). In this review, we discuss the key effects of mitochondrial dynamics and mitophagy on the pathogenesis of UC, with a particular focus on the cellular energy metabolism, oxidative stress, apoptosis, and immunoinflammatory activities. The therapeutic efficacy of existing drugs and phytochemicals targeting the mitochondrial pathway are discussed to reveal important insights for developing therapeutic strategies for treating UC. In addition, new molecular checkpoints with therapeutic potential are identified. Objectives: Cisplatin (DDP) resistance remains a primary cause of chemotherapy failure and recurrence of non-small cell lung cancer (NSCLC). Abnormal high microsomal glutathione transferase 1 (MGST1) expression has been found in DDP-resistant NSCLC cells. This study aimed to explore the function and mechanism of MGST1 in DDP resistance of NSCLC cells., Materials and Methods: The expression levels of target molecules were assessed by quantitative real-time polymerase chain reaction (RT-qPCR) and western blotting. Cell proliferation was evaluated by cell counting kit-8 (CCK-8) and colony formation assays. Ferroptosis was determined by malondialdehyde (MDA), glutathione (GSH), Fe 2+ , and reactive oxygen species (ROS) levels. The interaction between proteins was confirmed by Co-immunoprecipitation (Co-IP). The effect of MGST1 on DDP resistance was evaluated using the tumor xenograft assay in vivo. Immunohistochemical staining was performed to measure Ki-67 and p-H2A.X expression in tumor tissues., Results: MGST1 expression was higher, while arachidonate lipoxygenase 5 (ALOX5) expression was lower in DDP-resistant NSCLC patients and cells. MGST1 ablation sensitized NSCLC cells to DDP therapy through inducing ferroptosis. MGST1 protein directly interacted with ALOX5 protein to restrain ALOX5-triggered ferroptosis. Ferroptosis inhibitor or sh-ALOX5 reversed the promotive effect of MGST1 silencing on the DDP sensitivity of NSCLC cells. Finally, MGST1 depletion sensitized NSCLC cells to DDP therapy in nude mice in vivo ., Conclusion: MGST1 high expression contributed to DDP resistance of NSCLC cells by inhibiting ALOX5-induced ferroptosis. It has been more than 40 years since the beginning of exploring the genetic composition of ancient organisms from the perspective of ancient DNA. In the recent 20 years, with the development and application of high-throughput sequencing technology platforms and the improved efficiency of retrieving highly fragmented DNA molecules, ancient DNA research moved forward to a brand-new era of deep-time paleogenomics. It not only solved many controversial phylogenetic problems, enriched the migration and evolution details of various organisms including humans, but also launched exploration of the molecular responses to climate changes in terms of "whole genomic-big data-multi-species" level. Moreover, it expanded the sample age from no more than 100,000 years to the Early Pleistocene, ~2 million years ago. Recently, Chinese scientists have made many influential breakthroughs in evolution and migration integration of East Asian populations and thus filled an important gap in the evolutionary process of modern human. Compared to the situation in human paleogenomic studies, less attention has been paid to the study of ancient DNA from vertebrates remains. In this review, we introduce a series of advances in ancient DNA investigations of large mammals in Late Quaternary in China, summarize the research breakthroughs in revealing the systematic evolutionary relationship between ancient and extant groups, gene flow, and molecular responses of mammalian populations to climate change, and explore the opportunities and key challenges in the field of mammalian paleogenomics.

Piezoelectric catalysis is an emerging green strategy, but the existing piezoelectric heterostructures are not sufficient in performance for catalytic reduction of low-reduction potential uranium under harsh conditions. This study innovatively employs a defect heterogeneous engineering strategy, wherein covalent organic frameworks (COFs) are grown in situ on the surface of zinc oxide (ZnO) via Schiff base reactions, and defects are introduced into the COF shell layer via imine exchange reactions to construct D-COF@ZnO for piezoelectric catalytic uranium removal. The comprehensive study shows that defect heterogeneous engineering increases the asymmetry induced polarization of the material to promote charge redistribution, and thus significantly improves the activity of piezoelectric catalysis. In addition, defect engineering optimizes the nanosize of D-COF@ZnO to expose a richer array of active sites, resulting in ultra-fast U(VI) removal kinetics and ultra-high removal capacity. In the actual nuclear wastewater settings, D-COF@ZnO demonstrates outstanding selective removal efficacy for uranium, manifesting its considerable application potential and efficiency superiority. Insufficient activation of stimulator of interferon genes (STING) signaling pathway in tumor-associated dendritic cells limits the efficiency of tumor immunotherapy. Herein, the "three-in-one" IAHA-LaP/siPTPN6 NPs containing lanthanum ions (La 3+ ), cGAMP, and PTPN6 siRNA are developed for triple amplification of the STING pathway. In vitro results demonstrate that La 3+ significantly promotes cGAMP-mediated activation of the STING pathway by enhancing the phosphorylation of STING, TBK1, IRF3, and NF-κB p65. Moreover, the IAHA-LaP/siPTPN6 NPs further significantly enhance the phosphorylation of STING and NF-κB p65 and augment K63-linked ubiquitination of STING protein via siPTPN6-mediated downregulation of SHP-1 protein. Furthermore, NPs improve the secretion of IFNβ (2.4-fold), IL-6 (1.5-fold), and TNF-α (1.4-fold), thereby promoting DCs maturation compared to the mixture of La 3+ and cGAMP. In vivo results show that the IAHA-LaP/siPTPN6 NPs remarkably inhibit primary tumor growth by increasing the percentage of mature DCs in tumor-draining lymph nodes, polarizing M2/M1 phenotype in TME, and promoting the infiltration of CD8 + T cells into tumors. Moreover, these NPs dramatically prevent the growth of distal tumor by inducing systemic anti-tumor immunity and generating a long-term anti-tumor memory for protection against tumor recurrence in mice bearing bilateral B16F10. Background: Spontaneous pneumothorax (SP) has been reported to be associated with osteosarcoma lung metastases, but the computed tomography (CT) characteristics of lung metastases in patients with pneumothorax have not been comprehensively described. The study aimed to describe the CT characteristics of lung metastases in patients with osteosarcoma and to identify factors associated with SP., Methods: This study comprised 123 patients diagnosed with osteosarcoma lung metastasis at our hospital between January 2016 and December 2021. Demographics, tumor characteristics at initial diagnosis, the time interval between diagnosis of lung metastases and pneumothorax, and features of lung metastases on chest CT were documented., Results: A total of 31 patients experienced SP (20 unilateral and 11 bilateral cases). Lung metastases were identified as multiple nodules or masses (39/42 vs. 123/204, P<0.001), with higher rates of cavitation and pleural involvement (23/42 vs. 19/178, P<0.001 and 38/42 vs. 63/178, P<0.001, respectively) in patients with pneumothorax. A considerable prevalence of calcification (123/220) was observed in lung metastases. However, no statistically significant difference was found between patients with and without pneumothorax. The duration of detection of lung metastases was shorter in patients with pneumothorax than in those without it (6 vs. 11 months, P=0.039). Binary logistic regression analysis revealed that only the presence of cavitation and pleural involvement of lung metastases were significantly associated with pneumothorax, with odds ratios of 12.430 [95% confidence interval (CI): 3.011-51.320; P<0.001] and 15.480 (95% CI: 4.038-59.348; P<0.001), respectively., Conclusions: SP can occur in the region of lung metastases near the pleura following the formation and bursting of cavities. This study presents a facile one-pot synthesis method to fabricate BiFeO 3 -Bi 25 FeO 40 -Bi 2 O 3 heterojunction photocatalysts with controllable compositions and pure phases. Three different binary heterojunctions (BiFeO 3 /Bi 25 FeO 40 , BiFeO 3 /Bi 2 O 3 , and Bi 25 FeO 40 /Bi 2 O 3 ) and a ternary BiFeO 3 /Bi 25 FeO 40 /Bi 2 O 3 heterojunction were formed, all exhibiting significantly enhanced photocatalytic performance for the degradation of methylene blue (MB) and phenol under visible light irradiation, outperforming the individual compositions. Notably, the BiFeO 3 /Bi 25 FeO 40 heterojunction achieved the highest degradation efficiency (93.68% and 83.3% for MB and phenol, respectively) as well as excellent stability. Impressively, the phenol degradation efficiency of BiFeO 3 /Bi 25 FeO 40 was even over twice that of BiFeO 3 and Bi 25 FeO 40 , and four times higher than that of Bi 2 O 3 . The enhanced photocatalytic activity of the BiFeO 3 /Bi 25 FeO 40 heterojunction is primarily attributed to the advantageous S-scheme band alignments that facilitate efficient charge separation and enhance redox capabilities. While other heterojunctions also exhibited improved MB and phenol degradation efficiency, each unique combination of materials led to distinct electronic structures and diverse reaction mechanisms. The simplicity and scalability of the synthesis method, combined with the remarkable photocatalytic performance of these BiFeO 3 -Bi 25 FeO 40 -Bi 2 O 3 heterojunction materials, position them as highly promising candidates for applications in environmental remediation and solar energy conversion.

The effect of sexual dimorphism on the metabolism of patients with Parkinson's disease has not been clarified. A group of patients with Parkinson's disease and healthy controls were recruited, and their clinical characteristics and plasma were collected. Untargeted liquid chromatography-mass spectrometry-based plasma metabolomics profiling was performed. Differentially expressed metabolites between patients and healthy controls were respectively identified in the male and female participants and metabolite set enrichment analyses were further employed. A total of 75 patients with Parkinson's disease (37 males and 38 females) and 31 healthy controls (16 males and 15 females) were enrolled while no significant differences can be discovered in clinical characteristics. The constructed male-specific metabolic model from orthogonal partial least squares-discriminant analysis can't well recognize female patients and the female-specific model also can't accurately identify male patients. There were 55 differentially expressed metabolites in the male participants, and fatty acids and conjugates and eicosanoids were the significantly enriched metabolite sets. Meanwhile, 86 metabolites were differentially expressed in the female participants while fatty acids and conjugates and glycerophosphocholines were enriched. Only 17 metabolites were simultaneously changed in both male and female patients. Expanding material types and developing two-dimensional (2D) semiconductor materials with high performance have been hotspots in the field. In this research, it is found that the 12 existing semiconductors A 2 BXY 2 (A = K, Na; B = Li, Na; X = Al, Ga, In; Y = P, As, Sb) have a pronounced layered structure. We predict their 2D structures and properties, using first-principles calculations. Lower exfoliation energies confirm the feasibility of mechanical exfoliation from their bulk phases and that the 2D structures can be stabilized independently at room temperature. Interestingly, A 2 BXY 2 has an anionic tetrahedral one-dimensional chain or two-dimensional mesh structure of [XY 2 ] 3- composed of elements III-V. All A 2 BXY 2 monolayers exhibit direct or indirect band gap features (0.78-1.94 eV). More encouragingly, the A 2 BXY 2 monolayers possess ultrahigh carrier mobilities (∼10 5 cm 2 V -1 s -1 ) at room temperature. Furthermore, the results based on the nonequilibrium Green's function indicate that 2D A 2 BXY 2 exhibits a high ON/OFF ratio (∼10 4 ). To sum up, the A 2 BXY 2 family is an outstanding promising candidate for optoelectronics application.

Intermetallic compound (IMC) catalysts have garnered significant attention due to their unique surface and electronic properties, which can lead to enhanced catalytic performance compared to traditional monometallic catalysts. However, developing IMC materials as high-performance catalysts has been hindered by the inherent complexity of synthesizing nanoparticles with well-defined bulk and surface compositions. Achieving precise control over the composition of supported bimetallic IMC catalysts, especially those with high surface area and stability, has proven challenging. This review provides a comprehensive overview of the recent progress in developing supported IMC catalysts. We first examine the various synthetic approaches that have been explored to prepare supported IMC nanoparticles with phase-pure bulk structures and tailored surface compositions. Key factors influencing the formation kinetics and compositional control of these materials are discussed in detail. Then the strategies for manipulating the surface composition of supported IMCs are delved into. Applications of high-performance supported IMCs in important reactions such as selective hydrogenation, reforming, dehydrogenation, and deoxygenation are comprehensively reviewed, showcasing the unique advantages offered by these materials. Finally, the prevailing research challenges associated with supported IMCs are identified, including the need for a better understanding of the composition-property relationships and the development of scalable synthesis methods. The prospects for the practical implementation of these versatile catalysts in industrial processes are also highlighted, underscoring the importance of continued research in this field.

Objectives: This study aimed to investigate the factors associated with the spontaneous clearance of Chlamydia trachomatis, a phenomenon that, despite a growing body of literature, remains understudied in the context of women in China., Methods: Spontaneous clearance was defined as the transition from a positive to negative Chlamydia status over time without the use of antichlamydial therapy. Data from 5935 women aged 18 years and older who participated in the Clinical-Based Health Check program were analyzed. Eligible participants had no history of Neisseria gonorrhoeae infection, pelvic inflammatory disease, recent antibiotic use, or pregnancy and had an interval between the screening and follow-up visits of more than 3 days. Logistic regression was used to identify factors influencing spontaneous clearance., Results: Spontaneous clearance occurred in 23.9% (50/209) of the participants, typically with a median interval of 27 days. Significant factors included an interval >45 days, an age >35 years, the use of an intrauterine device (IUD), and the presence of clue cells., Conclusion: Spontaneous clearance of C. trachomatis is significantly affected by age, the interval between two tests, IUD use, and the presence of clue cells. Statins therapy is efficacious in diminishing the risk of major cardiovascular events in diabetic patients. However, our research has uncovered a correlation between the prolonged administration of statins and an elevated risk of myocardial dysfunction in patients with type II diabetes mellitus (TIIDM). Here, we report the induction of sterol regulatory element-binding protein 1 (SREBP1) activation, associated lipid peroxidation, and the consequent diabetic myocardial dysfunction after statin treatment and explored the underlying mechanisms. In db/db mice, we observed that 40 weeks atorvastatin (5 and 10 mg/kg) and rosuvastatin (20 mg/kg) administration exacerbated diabetic myocardial dysfunction by echocardiography and cardiomyocyte contractility assay, increased myocardial inflammation and fibrosis as shown by CD68, IL-1β, Masson's staining and Collagen1A1 immunohistochemistry (IHC) staining, increased respiratory exchange ratio (RER) by metabolic cage system assessment, exacerbated mitochondrial structural pathological changes by transmission electron microscopy (TEM) examination, increased deposition of lipid and glycogen by TEM, Oil-red and periodic acid-schiff stain (PAS) staining, which were corresponded with augmented levels of myocardial SREBP1 protein and lipid peroxidation marked by 4-hydroxynonenal (4-HNE) staining. Comparable myocardial fibrosis was also observed in KK-ay and low-dose streptozotocin (STZ)-induced TIIDM mice. Elevated SREBP1 levels were observed in the heart tissues from diabetic patients, which was positively correlated with their myocardial dysfunction. To elucidate the role of statin induced SREBP1 in lipid peroxidation and lipid deposition and related mechanism, we cultured neonatal mouse primary cardiomyocytes (NMPCs) and treated them with atorvastatin (10 μM, 24 h), tracing with [U- 13 C]-glucose and evaluating for SREBP1 expression and localization. We found that statin treatment elevated de novo lipogenesis (DNL) and the levels of SREBP1 cleavage-activating protein (SCAP), reduced the interaction of SCAP with insulin-induced gene 1 (Insig1), and enhance SCAP/SREBP1 translocation to the Golgi, which facilitate SREBP1 cleavage leading to its nuclear trans-localization and activation in NMPCs. Ultimately, SREBP1 knockdown or l-carnitine mitigated long-term statins therapy induced lipid peroxidation and myocardial fibrosis in low-dose STZ treated SREBP1 +/- mice and l-carnitine treated db/db mice. Here, a simple method of applying dimethylformamide (DMF) as cosolvent in the sol-gel technology is used to improve the quality of ZnO bulk films. First-principles calculations show that with the addition of polar solvent DMF, the adsorption energy (E ads ) between the solvent and Zn(OH)₂ increases from -1.42 to -1.74 eV, which can stabilize the existence of Zn(OH)₂, thereby promoting the ZnO synthesis. Besides, the elimination of amine residues in the DMF-ZnO film significantly suppress the photocatalytic activity induced by amine-induced coordination or redox reactions. Inverted organic solar cells (OSCs) based on PM6:Y6 and PM6:BTP-eC9 achieves impressive power conversion efficiencies (PCE) of 17.58 and 18.14%, respectively. Furthermore, benefiting from the reduced defects of bulk ZnO, pseudo-bilayer bulk heterojunction (PBHJ) devices based on the optimized ZnO film exhibited superior stability, the PM6:Y6 devices based on DMF-ZnO ETLs can maintain 90.28% of their initial PCE after 1000 h of thermal aging at 85 °C, and 80.98% of their initial PCE after 168 h of UV aging. Sodium ion hybrid capacitors (SIHCs) address the high power and energy requirements in energy storage devices but face significant challenges arising from the slow kinetics and cycling instability of the anode side. Introducing atomic disorder and employing structural engineering in anode materials proves to be effective strategies for achieving rapid charge storage. Here, it is demonstrated that N-doped MXene encapsulated amorphous vanadium oxide hollow spheres (VO x @N-MXene HSs) offer multidirectional open pathways and sufficient vacancies, enabling reversible and fast Na + insertion/extraction. Machine learning potentials, coupled with molecular simulation techniques, confirm the presence of more abundant pores within the amorphous vanadium oxide (VO x ) structure. The simulation of the charging/discharging process elucidates the authentic reaction path and structural evolutions of the VO x @N-MXene HSs, providing sufficient insight into the atomic-scale mechanisms associated with these structural superiorities. The full SIHCs devices demonstrate a high energy density of 198.3 Wh kg -1 , along with a long-term cycling lifespan of 8000 cycles. Context: The use of traditional Chinese medicine (TCM) for breast cancer patients inhibits tumor cell growth and proliferation, alleviates adverse reactions, and inhibits tumor recurrence and metastasis post-surgery. An assessment of its historical efficacy and an examination of the latest research trends are imperative to thoroughly leverage the potential of TCM for breast cancer treatment., Objective: This study analyzes the published literature on TCM for breast cancer treatment using bibliometric analysis to determine the current state, identify hot spots, and discern trends, providing insight into research in this field., Methods: TCM-based breast cancer treatment publications between 2003 and 2022 were retrieved from the Web of Science, China National Knowledge Infrastructure, Wanfang, and Duxiu databases. Visual analysis was performed using VOSviewer (V1.6.19) and CiteSpace (V6.3.R1) software. Examined metrics included the annual publication count, literature and journal, national and institutional contributions, author co-occurrence, keyword co-occurrence, keywords timeline, and keywords with citation bursts in this research field., Results and Conclusion: A total of 1080 English publications and 2617 Chinese publications were included in the analysis. China was the leading contributor of publications. High-frequency keywords such as 'apoptosis', 'expression', ' in vivo ', 'chemotherapy', 'triple-negative breast cancer', and 'lymphedema' were identified from English and Chinese publications; 'epithelial mesenchymal transition' and 'network pharmacology' emerged as hotspots. The development of modern science, technology, and in-depth research can result in broader prospects for the research and application of TCM in breast cancer treatment, resulting in more effective solutions for the treatment of breast cancer and other malignant tumors.

A sensitive, straightforward, and environmentally sustainable method was developed and validated for the quantification of 13 PBDEs in chicken eggs using gas chromatography coupled with triple quadrupole mass spectrometry (GC-MS/MS). The QuEChERS approach, combined with dispersive liquid-liquid microextraction (DLLME), was applied for sample preparation. Egg samples were extracted with acetonitrile and purified using Z-Sep + adsorbent. Tetrachloroethane was selected as the extraction solvent for the DLLME step. All PBDEs exhibited strong linearity, with R > 0.999 within the range of 0.20-10.0 ng/mL, and the limits of quantification (LOQ) were determined to be 10.0 pg/g. Apparent recoveries of PBDEs ranged from 85.2 % to 105.7 %. A total of 119 chicken eggs were analyzed using the developed method. BDE-47 and BDE-99 showed relatively high detection rates of 38.6 % and 24.4 %, respectively. Long-term biomonitoring of urinary metal ions is an essential tool for the epidemiological assessment of chronic exposure levels, enabling us to track changes in metal exposure over time and better understand its health implications. In this study, we evaluated the temporal trends of urinary metal ions among 1962 residents of Guangzhou, China, from 2018 to 2022. The total metal ion concentrations in the urine of the population did not change significantly between 2018 and 2019. With the onset of the COVID-19 pandemic in 2020, urinary total metal ion concentrations began to decline dramatically, reaching their lowest level in 2021. A rebound in concentrations was observed in 2022, which returned to the initial levels observed in 2018. Urine chromium and cadmium concentrations peaked in 2020, while urinary lead levels were the highest in 2021, and urinary nickel concentrations were the highest in 2022. Males consistently displayed higher urinary concentrations of lead and arsenic throughout each year of the study. Furthermore, minors consistently had higher urinary nickel levels than adults, whereas adults consistently had higher urinary cadmium concentrations than minors. Cluster analyses were conducted annually on urinary metal ions to examine the differences in their distribution and to evaluate changes in metal exposure patterns over time. The Monte Carlo simulations indicate that the whole population exhibits a high non-carcinogenic risk from arsenic exposure and significant carcinogenic risks associated with exposure to nickel, arsenic, chromium, and cadmium. Background and Objectives: Despite extensive efforts, the mechanisms underlying pain after axonal injury remain incompletely understood. Pain following corneal refractive surgery offers a valuable human model for investigating trigeminal axonal injury because laser-assisted in situ keratomileusis (LASIK) severs axons of trigeminal ganglion neurons innervating the cornea. While the majority of patients are pain-free shortly after surgery, a minority endure persistent postoperative ocular pain. Through genomic analysis of patients experiencing persistent postoperative ocular pain, we identified rare variants in genes encoding ion channels and receptors, including TRPM8, which codes for the menthol-sensitive and cold-sensing transient receptor potential cation channel., Methods: We conducted a profiling of 2 TRPM8 mutant variants, D665N and V915M, which were identified in patients suffering from persistent pain after LASIK surgery. We used patch-clamp and multielectrode array (MEA) recordings to investigate the biophysical and pharmacologic properties of mutant vs wild-type (WT) channels., Results: Patch-clamp analysis shows that these mutations shift the activation curves of TRPM8 in a hyperpolarized direction, with this effect being significantly different between WT and D665N channels. In addition, both mutations significantly increase channel sensitivity to the canonical ligand, menthol. MEA recordings from transfected rat trigeminal ganglion neurons indicate that expression of D665N and V915M mutant channels increases spontaneous activity compared with WT channels. Consistent with patch-clamp results, neuronal activity in MEA recordings was increased on exposure to menthol., Discussion: Collectively, our findings suggest that proexcitatory mutations of TRPM8, in the context of axonal injury within the cornea, can produce trigeminal ganglion neuron hyperexcitability that contributes to persistent postoperative ocular pain. Humans are exposed to DNA alkylating agents through endogenous metabolism, environmental exposure and cancer chemotherapy. The resulting alkylated DNA adducts may elicit genome instability by perturbing DNA replication and transcription. R-loops regulate various cellular processes, including transcription, DNA repair, and telomere maintenance. However, unscheduled R-loops are also recognized as potential sources of DNA damage and genome instability. In this study, by employing fluorescence microscopy and R-loop sequencing approaches, we uncovered, for the first time, that minor-groove N2-alkyl-dG lesions elicit elevated R-loop accumulation in chromatin and in plasmid DNA in cells. We also demonstrated that the N2-alkyl-dG-induced R-loops impede transcription elongation and compromise genome integrity. Moreover, genetic depletion of DDX23, a R-loop helicase, renders cells more sensitive toward benzo[a]pyrene diolepoxide, a carcinogen that induces mainly the minor-groove N2-dG adduct. Together, our work unveiled that unrepaired minor-groove N2-alkyl-dG lesions may perturb genome integrity through augmenting R-loop levels in chromatin. Cadmium (Cd) is a detrimental heavy metal propagated from soil to the food chain via plants, posing a great risk to human health upon consumption. Despite the understanding of Cd tolerance mechanisms in plants, whether and how plants actively respond to Cd and in turn restrict its uptake and accumulation remain elusive. Here, we identify a cell wall-associated receptor-like kinase 4 (WAKL4) involved in specific tolerance to Cd stress. We show that Cd rapidly and exclusively induces WAKL4 accumulation by promoting WAKL4 transcription and blocking its vacuole-dependent proteolysis in roots. The accumulated WAKL4 next interacts with and phosphorylates the Cd transporter NRAMP1 at Tyr488, leading to the enhanced ubiquitination and vacuole-dependent degradation of NRAMP1, and consequently reducing Cd uptake. Our findings therefore uncover a mechanism conferred by the WAKL4-NRAMP1 module that enables plants to actively respond to Cd and limit its uptake, informing the future molecular breeding of low Cd accumulated crops or vegetables., (© 2024. This study aims to develop and validate a robust analytical method for the quantification of polybrominated diphenyl ethers (PBDEs) in human serum using gas chromatography-tandem mass spectrometry. We compared procedural blanks, recoveries, and operational convenience of liquid-liquid extraction and supported liquid extraction for the determination of serum PBDEs. We evaluated different extraction solvents for their effect on PBDE recoveries. Supported liquid extraction was selected for method validation due to its operational convenience. The method demonstrated satisfactory linearity, sensitivity, and reproducibility, with the range of 0.10-5.00 µg/L for most PBDE congeners and 0.20-10.0 µg/L for PBDE-154 and PBDE-183, with limits of detection ranging from 2 to 48 ng/L, and with matrix effects ranging from 94% to 113%. Quality control assessments indicated that recoveries ranged from 85% to 110% and relative standard deviations of less than 11%. The proposed method was applied to biomonitoring of 111 healthy adults, revealing detectable levels of PBDEs in over 90% of the samples. Aims: The current recognition of mitral annular disjunction (MAD) as an anatomical abnormality potentially associated with ventricular arrhythmias has sparked controversy regarding its prevalence and clinical implications. This study aimed to investigate the prevalence and extent of MAD in individuals with no significant structural abnormalities involving the left heart using enhanced cardiac CT, while also exploring potential factors, such as age, that may be associated with MAD., Methods: Systolic datasets of cardiac CT from 742 subjects were retrospectively included. MAD was determined by rotating orthogonal multiplanar reconstruction images around the central axis of the mitral annulus. The maximal distance of disjunction (D MAD ) and segments involved (SI, 0 to 5 basal segments at left ventricular wall) was quantified to evaluate the extent of separation., Results: In total, 449 (60.5%) had MAD. Subjects with MAD were significantly older (51.3 ± 19.9 years vs. 29.6 ± 20.3 years, P < 0.001). Age was found to be an independent relevant factor for MAD (OR = 1.059; 95%CI: 1.033, 1.085; P < 0.001). Subjects with MAD were then divided into 4 subgroups (G1: ≤20 years, G2: 21-40 years, G3: 41-60 years, G4: ≥61 years). D MAD and SI of each age subgroup were 1.9 ± 0.2 mm, 2.0 ± 1.2 (G1, n = 36), 2.7 ± 0.8 mm, 2.9 ± 1.3 (G2, n = 51), 3.0 ± 0.8 mm, 3.0 ± 1.3 (G3, n = 183), and 3.0 ± 1.0 mm, 3.7 ± 1.1 (G4, n = 179). Age was an independent relevant factor associated with D MAD (R 2  = 0.132; β = 0.014; 95%CI: 0.004, 0.024; P = 0.007) and SI (OR = 1.030; 95%CI: 1.005, 1.055; P = 0.016)., Conclusions: MAD is a common finding on cardiac CT. In the prodrug-based self-assembled nanoassemblies, prodrugs usually consist of drug modules, response modules, and modification modules. Modification modules play a critical role in regulating the nano-assembly ability of the prodrugs. Herein, we carried out a "fatty alcoholization" strategy and chose various lengths of aliphatic alcohol chains (AC) as modification modules to construct disulfide bond bridged paclitaxel (PTX) prodrug nanoassemblies. The PTX-AC prodrugs would self-assemble into nanoassemblies (PTX-AC PNs) with higher drug loading, stability, and tumor selectivity than commercial preparations. After comprehensive exploration, we found the chain length (AC 12 , AC 16 , AC 20 , AC 24 ) of modification modules affected the assembly of PTX-AC PNs, further leading to disparate in vivo fate and antitumor efficacy. With the increase of the chain length of the modification modules (from AC 12 to AC 20 ), the assembly ability of the nanoassemblies was improved, attributed to the appropriate enhancement of hydrophobic force. When the chain length was further increased to AC 24 , the excessive hydrophobic force will lead to the aggregation of prodrugs and weaken the assembly ability. Therefore, PTX-AC 20 PNs with proper chain length may solve the paradox of efficacy and tolerance in 4 T1 breast tumor owing to their optimal nano-assembly stability and modest redox-sensitivity. Bruton's tyrosine kinase inhibitors (BTKi) exhibit superior efficacy in relapsed/refractory primary central nervous system lymphoma (PCNSL), but few studies have evaluated patients with newly diagnosed PCNSL, and even fewer studies have evaluated differences in efficacy between treatment with BTKi and traditional chemotherapy. This study retrospectively analyzed the clinical characteristics of 86 patients with PCNSL and identified predictors of poor prognosis for overall survival (OS). After excluding patients who only received palliative care, 82 patients were evaluated for efficacy and survival. According to the induction regimen, patients were divided into the traditional chemotherapy, BTKi combination therapy, and radiotherapy groups; the objective response rates (ORR) of the three groups were 71.4%, 96.2%, and 71.4% (P = 0.037), respectively. Both median progression-free survival and median duration of remission showed statistically significant differences (P = 0.019 and P = 0.030, respectively). The median OS of the BTKi-containing therapy group was also longer than that of the traditional chemotherapy group (not reached versus 47.8 (32.5-63.1) months, P = 0.038).Seventy-one patients who achieved an ORR were further analyzed, and achieved an ORR after four cycles of treatment and maintenance therapy had prolonged OS (P = 0.003 and P = 0.043, respectively). Background: The Asteraceae family, the largest and one of the most diverse families of angiosperms, presents significant challenges in taxonomic classification and systematic research due to its vast species diversity and morphological complexity. A comprehensive understanding of the chloroplast genomes within this family is essential for refining taxonomic classifications and advancing phylogenetic studies., Methods and Results: In this study, we sequenced the complete chloroplast genomes of 14 Asteraceae species and conducted a thorough bioinformatic analysis of their characteristics. The chloroplast genomes, ranging from 150,907 bp to 152,858 bp, exhibit a typical quadripartite structure: a large single-copy (LSC) region (83,044 bp to 84,625 bp), a small single-copy (SSC) region (18,223 bp to 18,673 bp), and a pair of inverted repeats (IRs) (24,806 bp to 25,201 bp). These genomes encode 87 to 89 protein-coding genes (PCGs), 36 to 37 tRNA genes, and 8 rRNA genes, with high conservation in size, structure, gene content, and order. Comparative analysis with other Asteraceae species' chloroplast genomes revealed notable similarities and structural variations, particularly in the IR regions. Nucleotide polymorphism analysis identified four genes-trnY-GUA, trnE-UUC, ycf1, and rrn23-with higher Pi values, suggesting potential hotspots for evolutionary studies. Phylogenetic analysis using maximum likelihood (ML) and Bayesian inference (BI) approaches provided new insights, proposing the reclassification of Himalaiella auriculata and Jacobaea raphanifolia as independent genera, distinct from Saussurea and Senecio., Conclusions: This study presents a comprehensive analysis of the chloroplast genome structures and phylogenetic relationships of 14 Asteraceae species, offering critical data for future molecular identification, evolutionary biology, and population genetics research. Introduction: Non-small cell lung cancer (NSCLC) is a prominent lung cancer disease worldwide. Currently, commonly used methods, such as surgery and radiotherapy, have significant side effects. Traditional Chinese medicine (TCM) has become a research hotspot because of its safe and effective characteristics. The branches and leaves of Taxus media are abundant in antitumor active compounds, and there has been no research conducted as yet regarding its anti-lung cancer molecular mechanism., Objective: The aim of this study is to investigate the antitumor activity of two samples before and after fermentation of T. media , and to research the molecular mechanism of its inhibitory effect on NSCLC., Methods: The chemical composition of pre-fermentation T. media (TM) and post-fermentation T. media qu (TMQ) were investigated using UHPLC-Q-Qrbitrap HRMS and high-performance liquid chromatography (HPLC). The anti-lung cancer activities of TM and TMQ were compared using an A549-induced tumor mouse model. An enzyme-linked immunosorbent assay (ELISA), hematoxylin and eosin (H&E) staining, immunohistochemistry, and immunofluorescence were used to determine the of TMQ mechanism of action., Results: The results indicated that TM and TMQ contained 83 compounds, consisting primarily of flavonoids, organic acids, and taxanes. Both taxanes and flavonoids in TMQ were higher than that in TM. Both TM and TMQ effectively inhibited the tumor growth in non-small cell lung cancer (NSCLC), and the inhibition rate was greater in TMQ (57.24%) than in TM (49.62%). TMQ administration downregulated the tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and the glutathione (GSH) level and upregulated interferon-γ (IFN-γ), reactive oxygen species (ROS), and malondialdehyde (MDA) levels in the serum of tumor mice. TMQ treatment also increased the protein expression of Bax, Caspase-3, and Beclin-1 in tumor tissues. In contrast, the bcl-2, PI3K, Ki67, ULK1, and mTOR protein levels were suppressed by TMQ. Protein assay analyses reemphasized the superior antitumor effect of TMQ over TM. These cumulative findings demonstrated that the mechanism of action of TMQ was closely related to the activation of transcriptional misregulation in the cancer pathway that inhibited the cholinergic synaptic, AMPK, and PI3K/Akt/mTOR signaling pathways., Conclusion: This study demonstrated that fermentation increased the active ingredient contents and antitumor effects of T. media . Background: According to the degree of intradermal neoplasia in the colorectal exhalation, it can be divided into two grades: Low-grade intraepithelial neoplasia (LGIN) and high-grade intraepithelial neoplasia (HGIN). Currently, it is difficult to accurately diagnose LGIN and HGIN through imaging, and clinical diagnosis depends on postoperative histopathological diagnosis. A more accurate method for evaluating HGIN preoperatively is urgently needed in the surgical treatment and nursing intervention of colorectal polyps., Aim: To explore the characteristics and risk factors of HGIN in older patients with colorectal polyps., Methods: We selected 84 older patients diagnosed with HGIN as the HGIN group ( n = 95 colonic polyps) and 112 older patients diagnosed with LGIN as the LGIN group ( n = 132 colonic polyps) from Shandong Provincial Hospital Affiliated to Shandong First Medical University. The endoscopic features, demographic characteristics, and clinical manifestations of the two patient groups were compared, and a logistic regression model was used to analyze the risk factors for HGIN in these patients., Results: The HGIN group was older and had a higher number of sigmoid colon polyps, rectal polyps, pedunculated polyps, polyps ≥ 1.0 cm in size, polyps with surface congestion, polyps with surface depression, and polyps with villous/tubular adenomas, a higher proportion of patients with diabetes and a family history of colorectal cancer, patients who experienced rectal bleeding or occult blood, patients with elevated carcinoembryonic antigen (CEA) and cancer antigen 199 (CA199), and lower nutritional levels and higher frailty levels. Background: Recent evidence links the prognosis of traumatic brain injury (TBI) to various factors, including baseline clinical characteristics, TBI specifics, and neuroimaging outcomes. This study focuses on identifying risk factors for short-term survival in severe traumatic brain injury (sTBI) cases and developing a prognostic model., Methods: Analyzing 430 acute sTBI patients from January 2018 to December 2023 at the 904th Hospital's Neurosurgery Department, this retrospective case-control study separated patients into survival outcomes: 288 deceased and 142 survivors. It evaluated baseline, clinical, hematological, and radiological data to identify risk and protective factors through univariate and Lasso regression. A multivariate model was then formulated to pinpoint independent prognostic factors, assessing their relationships via Spearman's correlation. The model's accuracy was gauged using the Receiver Operating Characteristic (ROC) curve, with additional statistical analyses for quantitative factors and model effectiveness. Internal validation employed ROC, calibration curves, Decision Curve Analysis (DCA), and Clinical Impact Curves (CIC) to assess model discrimination, utility, and accuracy. The International Mission for Prognosis and Analysis of Clinical Trials in TBI (IMPACT) and Corticosteroid Randomization After Significant Head injury (CRASH) models were also compared through multivariate regression., Results: Factors like unilateral and bilateral pupillary non-reactivity at admission, the derived neutrophil to lymphocyte ratio (dNLR), platelet to lymphocyte ratio (PLR), D-dimer to fibrinogen ratio (DFR), infratentorial hematoma, and Helsinki CT score were identified as independent risk factors (OR > 1), whereas serum albumin emerged as a protective factor (OR < 1). The model showed superior predictive performance with an AUC of 0.955 and surpassed both IMPACT and CRASH models in predictive accuracy. Internal validation confirmed the model's high discriminative capability, clinical relevance, and effectiveness., Conclusions: Short-term survival in sTBI is significantly influenced by factors such as pupillary response, dNLR, PLR, DFR, serum albumin levels, infratentorial hematoma occurrence, and Helsinki CT scores at admission. Continuous human biomonitoring and predictive modelling of urinary pesticide metabolites are critical for evaluating pesticide exposure trends and associated health risks. We conducted repeat cross-sectional surveys to determine the urinary concentrations of eight pesticide metabolites in the residents of Guangzhou, China, from 2018 to 2022. We longitudinally analyzed the changes in these metabolite concentrations over the years and assessed the potential non-carcinogenic risks by calculating the hazard quotient and hazard index. No significant differences were observed in the total urinary pesticide metabolite concentrations over the 5 years (9.16-12.99 μg/L). The urinary concentrations of 3,5,6-trichloro-2-pyridinol and 2,4-dichlorophenoxyacetic acid reached their lowest levels in 2020 (1.47 and 0.11 μg/L). Conversely, urinary para-nitrophenol concentrations exhibited an inverse trend, peaking in 2020 (6.16 μg/L). The composition profiles of urinary pesticide metabolites showed that para-nitrophenol consistently constituted the largest proportion each year. Males consistently showed higher median concentrations of total urinary pesticide metabolites and individual metabolites of 3,5,6-trichloro-2-pyridinol, trans-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropane-1-carboxylic acid, and para-nitrophenol than females. The concentrations of cis-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropane-1-carboxylic acid in adults' urine were significantly higher than those in minors' urine each year. The total pesticide metabolite concentrations in adults' urine were significantly higher than those in minors' urine in 2018 and 2020, whereas no significant differences were observed in other years. No significant differences in urinary pesticide metabolite concentrations were observed among different BMI groups. Results showed that 14.17% of the population had hazard index values above 1, indicating a higher risk of health hazards. Three predictive models were employed to predict urinary pesticide metabolite concentrations for 2023-2024, revealing an increasing trend in 3,5,6-trichloro-2-pyridinol concentrations while other metabolites are expected to decrease. Improving fluorescence emission efficiency is essential to develop novel luminescent materials. However, the low water solubility of conventional fluorescent dyes is a serious obstacle to broadening the application scope. Herein, a green protocol have been proposed: Two poorly water-soluble naphthalimide derivatives MONI and MANI with high fluorescent quantum yields (larger than 0.95 in toluene solution) were loaded in three different sizes of cyclodextrin (CD; α, β, γ-CD) with high water solubility. To further check the feasibility of the proposal, density functional theory (DFT) and time dependent-DFT (TD-DFT) methods combining the Own N-layer Integrated molecular Orbital molecular Mechanics (ONIOM) model with dispersion correction were employed to investigate the geometric and electronic structures of complexes CD·MXNI (X = N, O) in the excited-state process. TD-DFT calculations predict that the fantastic emission behavior of MXNI can be reserved after binding with CD, even improving fluorescent intensity in aqueous solution. Basis set superposition error (BSSE) correction and symmetry adapted perturbation theory (SAPT) were adopted to estimate the complexation energies and weak noncovalent interactions. The middle-sized β-CD is the perfect candidate to allow fluorescent molecules to settle into its cavity, forming an inclusion complex. Energy decomposition analysis (EDA) indicates that dispersion is superior to electrostatics interaction in embedding-type β-CD·MXNI, while it is contrary in α,γ-CD·MXNI. NMR calculations further prove the existence of a strong intermolecular hydrogen bond interaction between host and guest. Weak interactions that limited molecular vibration and hampered the nonradiative inactivation channel are conducive to the enhanced emission intensity.

Background: Non-small-cell lung cancer (NSCLC) remains a deadly malignancy worldwide. Resistance to cisplatin (DDP) is a significant obstacle that limits the therapeutic efficacy in NSCLC patients., Objectives: This study investigated the role and mechanism of 24-dehydrocholesterol reductase (DHCR24) in DDP resistance in NSCLC cells., Methods: 24-dehydrocholesterol reductase levels, ferroptosis-related molecules, and proteins involved in the PI3K/AKT/GSK3β pathway were measured. The growth capacity of the cells was evaluated, and ferroptosis was assessed by measuring MDA, GSH, Fe 2+ , and ROS levels. The impact of DHCR24 on NSCLC DDP resistance was analyzed using a tumor xenograft assay in vivo. Ki-67 and DHCR24 expression in tumors were evaluated through immunohistochemical staining., Results: 24-dehydrocholesterol reductase expression was elevated in DDP-resistant cells, indicating a poorer prognosis for NSCLC patients. Down-regulation of DHCR24 inhibited the growth of DDP-resistant cells and induced ferroptosis. Inhibition of DHCR24 led to the inactivation of the PI3K/AKT/GSK3β pathway and subsequent induction of ferroptosis. Inhibition of ferroptosis or activation of the PI3K/AKT/GSK3β pathway counteracted the increased DDP sensitivity induced by DHCR24 knockdown in NSCLC cells. Transition metal chalcogenides (TMCs) hold great potential for sodium-ion batteries (SIBs) owing to their multielectron conversion reactions, yet face challenges of poor intrinsic conductivity, sluggish diffusion kinetics, severe phase transitions, and structural collapse during cycling. Herein, a self-templating strategy is proposed for the synthesis of a class of metal cobalt-doped NiSe nanoparticles confined within three-dimensional (3D) N-doped macroporous carbon matrix nanohybrids (Co-NiSe/NMC). The cation defect engineering within the developed Co-NiSe and 3D N-doped carbon plays a crucial role in enhancing intrinsic conductivity, reinforcing structural stability, and reducing the barrier to sodium ion diffusion, which are verified by a series of electrochemical kinetic analyses and density functional theory calculations. Significantly, such cation defect engineering not only reduces overpotential but also accelerates conversion reaction kinetics, ensuring both exceptional high-rate capability and extended durability. Consequently, the optimally engineered Co-NiSe/NMC demonstrates a remarkable rate performance, delivering 390 mAh g -1 at 10 A g -1 . Moreover, it exhibits an unprecedented lifespan, maintaining a remarkable capacity of 403 mAh g -1 after 1400 cycles and 318 mAh g -1 after 4000 cycles, even at high rates of 1.0 and 2.0 A g -1 , respectively. Developing a convenient method to efficiently determine the size of nanoplastics in the environment is urgent in terms of ecological or human health protection. In this work, a novel strategy for discriminating the size of polystyrene (PS)-based nanoplastics was reported via regulating the radiative transition efficiency of NH 2 -UIO-66 (NU) with benzoic acid (BA) as the auxiliary ligand. The elaborately doped BA capped the defect sites and triggered nonradiative transition efficiency of NU. As a result, the formed composite (denoted as BA-NU) was more sensitive to interaction among neighboring NU and nanoplastics. The interaction between particles limited the rotation and vibration of the benzene ring within the BA-NU molecule, thus increasing the BA-NU fluorescence. The sensitivity of BA-NU on nanoplastics was well controlled by manipulating the doping contents of BA, leading to precisely tunable physicochemical properties for this structure. Deriving from the exquisitely designed nanostructures, the composite of BA-NU was successfully used to discriminate different size PS as an ultrasensitive turn-on probe. This work highlights the possibility of boosting the detection performance by regulating the main structure with guest molecules at the molecular level.

The factors that contribute to pain after nerve injury remain incompletely understood. Laser-assisted in situ keratomileusis (LASIK) and photorefractive keratectomy (PRK) are common surgical techniques to correct refractive errors. After LASIK or PRK, a subset of patients suffers intense and persistent pain, of unknown origin, described by patients as feeling like shards of glass in their eye. Here, we evaluated a TRPV1 variant, p.V527M, found in a 49-y-old woman who developed corneal pain after LASIK and subsequent PRK enhancement, reporting an Ocular Surface Disease Index score of 100. Using patch-clamp and Ca 2+ imaging, we found that the V527M mutation enhances the response to acidic pH. Increasing proton concentration induced a stronger leftward shift in the activation curve of V527M compared to WT, resulting in channel activity of the mutant in acidic pH at more physiological membrane potentials. Finally, comparing the responses to consecutive applications of different agonists, we found in V527M channels a reduced capsaicin-induced desensitization and increased sensitization by the arachidonic acid metabolite 12-hydroxyeicosatetraenoic acid (12-HETE). Abnormalities in coagulation and fibrinolytic status have been demonstrated to be relevant to inflammatory bowel disease. Nevertheless, there is no study to methodically examine the role of the coagulation and fibrinolysis-related genes in the diagnosis of ulcerative colitis (UC). UC-related datasets (GSE169568 and GSE94648) were originated from the Gene Expression Omnibus database. The biomarkers related to coagulation and fibrinolysis were identified through combining differentially expressed analysis and machine learning algorithms. Moreover, Gene Set Enrichment Analysis and immune analysis were carried out. A total of 4 biomarkers (MAP2K1, CREBBP, TAF1, and HP) were identified, and biomarkers were markedly enriched in pathways related to immunity, such as T-cell receptor signaling pathway, primary immunodeficiency, chemokine signaling pathway, etc. In total, the infiltrating abundance of 4 immune cells between UC and control was markedly different, namely eosinophils, macrophage M0, resting mast cells, and regulatory T cells. And all biomarkers were significantly relevant to eosinophils. Context: Revealing the mechanism of intermolecular interactions in dinitroamine ammonium (ADN)-based liquid propellants and exploring the reasons for their performance changes, multi-perspective interaction analyses of ADN and ADN-water (H 2 O)-methanol (CH 3 OH) solutions have been conducted via theoretical methods. The band structure, density of states (DOS), surface electrostatic potential (ESP), Hirshfeld surface, reduced density gradient (RDG), AIM topological analysis, and detonation performance were studied and the results showed that both the ADN and ADN-H 2 O-CH 3 OH solutions had hydrogen bonds and van der Waals interactions. By introducing the small molecules H 2 O and CH 3 OH, the detonation performance of the ADN-H 2 O-CH 3 OH solution slightly decreased, but its sensitivity also decreased. Overall, the comprehensive performance of the ADN-H 2 O-CH 3 OH solution has improved, and the application range has expanded. These results are helpful for obtaining a deeper understanding of ADN-based liquid propellants at the atomic level and contribute to the development of new liquid propellants., Methods: The ADN and ADN-H 2 O-CH 3 OH solutions were constructed by Amorphous cell module and optimized via GGA with PBE methods in the Dmol3 module of the Materials Studio, and their electronic properties were calculated. Hirshfeld surfaces were generated with CrystalExplorer 3.0. A topological analysis of a variety of molecular clusters was performed via QTAIM. Leaves play a crucial role in photosynthesis and respiration, ultimately affecting the final grain yield of crops, including wheat ( Triticum aestivum L.); however, the molecular mechanisms underlying wheat leaf development remain largely unknown. Here, we isolated a narrow-leaf gene, TaWAK2-A , through a map-based cloning strategy. TaWAK2-A encodes a wall-associated kinase (WAK), for which a single Ala-to-Val amino acid substitution reduces the protein stability, leading to a narrow-leaf phenotype in wheat. Further investigation suggests that TaWAK2 directly interacts with and phosphorylates TaNAL1, a trypsin-like serine/cysteine protease. The phosphorylated TaNAL1 is then involved in the degradation of the zinc finger transcription factor TaDST, which acts as a repressor of leaf expansion by activating the expression of the cytokinin oxidase gene TaCKX9 and triggering in vivo cytokinin degradation. Therefore, our findings elucidate a signaling cascade involving TaWAK2-TaNAL1-TaDST that sheds light on the regulation of wheat leaf development.
