Background: Benign prostatic hyperplasia (BPH) is a global age-related disease. It has been reported that over half of the Chinese male population aged 70 years or older are experiencing BPH. Solid fuel, which is the major source of household air pollution, has been reportedly associated with several adverse events, including sex hormone disorders. Due to the certain relationship between sex hormone levels and prostate disease, the relationship between solid fuel use and lower urinary tract symptoms (LUTSs) suggestive of BPH (LUTS/BPH) deserves further exploration., Objective: This study mainly aimed to investigate the association between solid fuel use and LUTS/BPH., Methods: The data used in this study were obtained from the West China Natural Population Cohort Study. Household energy sources were assessed using questionnaires. LUTS/BPH was evaluated based on participant self-reports. We performed propensity score matching (PSM) to reduce the influence of bias and unmeasured confounders. The odds ratio (OR) and 95% CI of LUTS/BPH for the solid fuel group compared with the clean fuel group were calculated. We also conducted stratified analyses based on BMI, metabolic syndrome, waist to hip ratio, drinking status, smoking status, and age., Results: A total of 5463 participants were included in this study, including 399 solid fuel users and 5064 clean fuel users. After PSM, the solid fuel group included 354 participants, while the clean fuel group included 701 participants. Solid fuel use was positively correlated with LUTS/BPH before and after PSM (OR 1.68, 95% CI 1.31-2.15 and OR 1.81, 95% CI 1.35-2.44, respectively). In stratified analyses, the OR of the nonsmoking group was higher than that of the smoking group (OR 2.56, 95% CI 1.56-4.20 and OR 1.47, 95% CI 0.99-2.18, respectively). Similarly, the OR of the nondrinking group was higher than that of the drinking group (OR 2.70, 95% CI 1.46-4.99 and OR 1.48, 95% CI 1.01-2.17, respectively)., Conclusions: A positive correlation between solid fuel use and LUTS/BPH was observed. Background: Testicular cancer (TC) is currently the most common malignancy in young and middle-aged men. A comprehensive assessment of TC burden is in lack., Method: Global incidence, deaths, and disability-adjusted life-years (DALYs) of TC from 1990 to 2019 were obtained. Estimated annual percentage change (EAPC) was calculated to quantify trends in TC changes during the period. Relationships between disease burden and age, socio-demographic index (SDI) levels, human development index (HDI) were further analyzed., Results: Globally, incident cases of TC more than doubled from 1990 to 2019, together with an increasing of global age-standardized incidence rates (ASIR) of TC from 1.9 to 2.8. The age-standardized deaths rates (ASDR) remained stable from 0.31 to 0.28. The similar results were reflected in the DALYs. In 2019, the highest ASIR were found in Southern Latin America, Central Europe and Western Europe. Analogously, the highest ASDR were found in Southern Latin America followed by Central Latin America and Central Europe. The burden of incidence increased with SDI, appropriately reached a peak at about 0.78, and then declined. Similarly, the burden of deaths increased with SDI, met a maximum at about 0.7., Conclusions: From 1990 to 2019, the ASIR of TC has increased significantly, while the ASDR has been relatively stable and slightly decreased. The disease burden of TC is shifting to regions and countries with moderate to high levels of development. Lexical databases are essential tools for studies on language processing and acquisition. Most previous Chinese lexical databases have focused on materials for adults, yet little is known about reading materials for children and how lexical properties from these materials affect children's reading comprehension. In the present study, we provided the first large database of 2999 Chinese characters and 2182 words collected from the official textbooks recently issued by the Ministry of Education (MOE) of the People's Republic of China for most elementary schools in Mainland China, as well as norms from both school-aged children and adults. The database incorporates key orthographic, phonological, and semantic factors from these lexical units. A word-naming task was used to investigate the effects of these factors in character and word processing in both adults and children. The results suggest that: (1) as the grade level increases, visual complexity of those characters and words increases whereas semantic richness and frequency decreases; (2) the effects of lexical predictors on processing both characters and words vary across children and adults; (3) the effect of age of acquisition shows different patterns on character and word-naming performance. Herein, SnO 2 QDs (<10 nm) with small size instead of conventional nanoparticles was employed to modify ZnFe 2 O 4 to synthesize porous and heterogeneous SnO 2 /ZnFe 2 O 4 (ZFSQ) composites for gas sensing. By an immersion process combined with calcination treatment, the resultant porous ZFSQ composites with different contents of SnO 2 QDs were obtained, and their sensing properties were investigated. Compared with bare ZnFe 2 O 4 and SnO 2 QDs, porous ZFSQ composites based-sensors showed much improved sensor response to acetone. For contrast, the sensor performance of ZFSQ composites was also compared with that of ZnFe 2 O 4 sphere modified by SnO 2 nanoparticles with different size. The porous ZFSQ composite with 5 wt% SnO 2 QDs (ZFSQ-5) showed a better acetone sensing response than that of other ZFSQ composites, and it exhibited a high response value of 110-100 ppm of acetone and a low detection limit of 0.3 ppm at 240 °C. In addition to the rich heterojunctions and porous structure, the size effect of SnO 2 QDs was other indispensable reasons for the improved sensor performance. Finally, the ZFSQ-5 composite sensor was attempted to be applied for acetone sensing in exhaled breath, suggesting its great potential in monitoring acetone., (© 2024 IOP Publishing Ltd.)

Numerous functional magnetic resonance imaging (fMRI) studies have examined the neural mechanisms of negative emotional words, but scarce evidence is available for the interactions among related brain regions from the functional brain connectivity perspective. Moreover, few studies have addressed the neural networks for negative word processing in bilinguals. To fill this gap, the current study examined the brain networks for processing negative words in the first language (L1) and the second language (L2) with Chinese-English bilinguals. To identify objective indicators associated with negative word processing, we first conducted a coordinate-based meta-analysis on contrasts between negative and neutral words (including 32 contrasts from 1589 participants) using the activation likelihood estimation method. Results showed that the left medial prefrontal cortex (mPFC), the left inferior frontal gyrus (IFG), the left posterior cingulate cortex (PCC), the left amygdala, the left inferior temporal gyrus (ITG), and the left thalamus were involved in processing negative words. Next, these six clusters were used as regions of interest in effective connectivity analyses using extended unified structural equation modeling to pinpoint the brain networks for bilingual negative word processing. Brain network results revealed two pathways for negative word processing in L1: a dorsal pathway consisting of the left IFG, the left mPFC, and the left PCC, and a ventral pathway involving the left amygdala, the left ITG, and the left thalamus. We further investigated the similarity and difference between brain networks for negative word processing in L1 and L2. The findings revealed similarities in the dorsal pathway, as well as differences primarily in the ventral pathway, indicating both neural assimilation and accommodation across processing negative emotion in two languages of bilinguals., (© The Author(s) 2023. The cognitive function of the human cerebellum could be characterized as enigmatic. However, researchers have attempted to detail the comprehensive role of the cerebellum in several cognitive processes in recent years. Here, using functional magnetic resonance imaging (fMRI) and transcranial direct current stimulation (tDCS), we revealed different functions of bilateral cerebellar lobules in bilingual language production. Specifically, brain activation showed the bilateral posterolateral cerebellum was associated with bilingual language control, and an effective connectivity analysis built brain networks for the interaction between the cerebellum and the cerebral cortex. Furthermore, anodal tDCS over the right cerebellum significantly optimizes language control performance in bilinguals. Together, these results reveal a precise asymmetrical functional distribution of the cerebellum in bilingual language production, suggesting that the right cerebellum is more involved in language control. Background: It has been reported for several years that polycyclic aromatic hydrocarbons (PAHs) could disturb human endocrine function. However, there is still a short of consistent conclusion about the relationship between PAH exposure and levels of sexual hormones. The aim of our study is to explore whether exposure to PAHs and how PAHs affect the levels of serum testosterone (T) and estradiol (E2) in adults, hoping to fulfill the knowledge gap., Materials and Methods: This study included adults aged 20 and above who participated in the National Health and Nutrition Examination Survey (NHANES) from 2011 to 2016. We included 10 PAH metabolites in this study. The levels of urinary PAH metabolites were log-transformed and divided into quartiles. The associations between PAH metabolites and both serum T levels of males and E2 levels of females were investigated using multivariate regression models. We furtherly calculated PAHs scores by sum of ranks across 10 PAHs metabolites, which represented the exposure levels of PAHs mixtures, and the association between PAHs scores and serum T and E2 levels were analyzed., Results: A total of 4,654 subjects were included in this study, including 2,460 males and 2,194 females. After adjusting for confounders, 2-hydroxynapthalene and 3-hydroxyfluorene were positively associated with serum T levels of males (p-value for trend=0.047, and p-value for trend=0.006, respectively), while 1-hydroxyphenanthrene was positively associated with serum E2 levels of females (p-value for trend=0.013). In the adjusted models, no significant association was found between PAHs scores and either T levels of males or E2 levels of females (p-value for trend=0.615, and p-value for trend=0.241, respectively)., Conclusions: This study showed urinary 2-hydroxynapthalene and 3-hydroxyfluorene were associated with increased T levels of males, and urinary 1-hydroxyphenanthrene was associated with increased E2 levels of females. The poor compatibility between the polymer matrix and complex modification processes greatly affects the excellent tribological properties of graphene in the polymer matrix. In this study, a covalent organic framework (COF)-coated graphene hybrid lubricating filler (G/COFs) was synthesized in situ using a sample one-step mechanochemical synthesis process. This was used to improve the tribological properties of bismaleimide (BMI) resin. The morphology and microstructure of the G/COFs hybrid were characterized, and the effect of the added amount on the tribological properties of the G/COFs/BMI composites was studied. The results showed that the G/COFs hybrid could improve the stability of the friction coefficient and decrease the volume wear rate of BMI composites. Compared to the neat BMI, the 0.6 wt% G/COFs/BMI composites showed optimal tribological performance, with the friction coefficient and volume wear rate decreasing from 0.35 to 0.14 and from 48 × 10 -6 to 10.6 × 10 -6 mm 3 /(N‧m), respectively. In addition, the G/COFs/BMI composites showed lower friction coefficient fluctuations and volume wear rates than G/BMI composites. This is mainly attributed to the fact that the deposition of COFs can not only effectively prevent the aggregation of graphene nanosheets, but can also significantly improve the compatibility and interfacial bond between the graphene and BMI matrix. Moreover, the good synergistic effect between the lamellar COFs and graphene nanosheets can generate high-quality self-lubricating transfer films during the friction process. The excellent dispersibility, efficient chemical functionalization, better friction reduction and wear-resistance properties, and facile preparation method make graphene/COFs hybrid nanoparticles promising as an excellent lubricating filler.

Listeners regularly comprehend continuous speech despite noisy conditions. Previous studies show that neural tracking of speech degrades under noise, predicts comprehension, and increases for non-native listeners. We test the hypothesis that listeners similarly increase tracking for both L2 and noisy L1 speech, after adjusting for comprehension. Twenty-four Chinese-English bilinguals underwent EEG while listening to one hour of an audiobook, mixed with three levels of noise, in Mandarin and English and answered comprehension questions. We estimated tracking of the speech envelope in EEG for each one-minute segment using the multivariate temporal response function (mTRF). Contrary to our prediction, L2 tracking was significantly lower than L1, while L1 tracking significantly increased with noise maskers without reducing comprehension. However, greater L2 proficiency was positively associated with greater L2 tracking. We discuss how studies of speech envelope tracking using noise and bilingualism might be reconciled through a focus on exerted rather than demanded effort., (Copyright © 2022 Elsevier Inc. Extensive studies have reported significant activation of the cerebellum in reading and reading-related tasks. However, it has remained unclear how the cerebellum contributes to reading and how reading-related regions in the cerebrum are related to those in the cerebellum. In this review, by summarizing previous literature, we observe that multiple cerebellar areas are engaged in reading and vary in their contributions to reading. Moreover, the cerebellar reading-related areas are selectively connected with the cerebral areas with the same functional specificity. Abnormalities in the cerebro-cerebellar connection are also associated with reading impairments. We thus propose the cerebro-cerebellar mapping hypothesis, which suggests that the cerebellum might have another reading-related network rather than serving as a neural hub. This network maps to and collaborates with its functionally corresponding network in the cerebrum. This framework heightens the importance of the cerebellum to reading and provides new insights into the relationship between the cerebellum, cerebrum, and reading development., (Copyright © 2022 The Authors. Domain-general cognitive control is closely related to language control during bilingual language production. Previous neural imaging studies have revealed a highly overlapped but rewired brain network for language control and nonverbal cognitive control. In the present study, we examined this issue from a training perspective. Two groups of participants performed the language switching task at pre-and post-tests during functional magnetic resonance imaging (fMRI) scanning. After the pre-test, the experimental group received 8-day training in a non-verbal switching task, while the control group performed an unrelated color judgement task. We found that only the experimental group but not the control group showed decreased strength of connectivity from the ventral lateral frontal cortex to the left caudate nucleus and from the medial surface of the frontal lobe to the left thalamus. These results indicate an increased efficiency after nonverbal training for the frontal cortex to implement domain-general suppression and monitoring in a domain-specific conflict context during bilingual language and lexical selections. Background: Till date, the optimal treatment strategy for delivering adjuvant androgen deprivation therapy (ADT) in localized and locally advanced prostate cancer (PCa), as a lower stage in PCa progression compared with metastatic PCa, is still unclear. This study compares the efficacy of castration alone with complete androgen blockade (CAB) as adjuvant ADT in patients with localized and locally advanced PCa undergoing radical prostatectomy (RP)., Methods: Patients diagnosed with PCa, without lymph node or distant metastasis, who received RP in West China Hospital between January 2009 and April 2019, were enrolled in this study. We performed survival, multivariable Cox proportional hazard regression, and subgroup analyses., Results: A total of 262 patients were enrolled, including 107 patients who received castration alone and 155 patients who received CAB. The survival analysis revealed that there was no significant difference between the two groups (hazard ratios [HR] = 1.07, 95% confidence intervals [95% CI] = 0.60-1.90, P = 0.8195). Moreover, the multivariable Cox model provided similarly negative results before and after adjustment for potential covariant. Similarly, there was no significant difference in the clinical recurrence between the two groups in both non-adjusted and adjusted models. Furthermore, our subgroup analysis showed that CAB achieved better biochemical recurrence (BCR) outcomes than medical castration alone as adjuvant ADT for locally advanced PCa (P for interaction = 0.0247, HR = 0.37, 95% CI = 0.14-1.00, P = 0.0497)., Conclusion: Combined androgen blockade achieved better BCR outcomes compared with medical castration alone as adjuvant ADT for locally advanced PCa without lymph node metastasis., (Copyright © 2022 The Chinese Medical Association, produced by Wolters Kluwer, Inc. under the CC-BY-NC-ND license.)

Background: Diminished sensitivity towards chemotherapy remains the major impediment to the clinical treatment of bladder cancer. However, the critical elements in control of chemotherapy resistance remain obscure., Methods: We adopted improved collagen gels and performed cytotoxicity analysis of doxorubicin (DOX) and mitomycin C (MMC) of bladder cancer cells in a 3D culture system. We then detected the expression of multidrug resistant gene ABCB1 , dormancy-associated functional protein chicken ovalbumin upstream-transcription factor 1 (COUPTF1), cell proliferation marker Ki-67, and cellular senescence marker senescence-associated β-galactosidase (SA-β-Gal) in these cells. We further tested the effects of integrin blockade or protein kinase B (AKT) inhibitor on the senescent state of bladder cancer. Also, we examined the tumor growth and survival time of bladder cancer mouse models given the combination treatment of chemotherapeutic agents and integrin α2β1 ligand peptide TFA (TFA)., Results: Collagen gels played a repressive role in bladder cancer cell apoptosis induced by DOX and MMC. In mechanism, collagen activated the integrin β1/AKT cascade to drive bladder cancer cells into a premature senescence state via the p21/p53 pathway, thus attenuating chemotherapy-induced apoptosis. In addition, TFA had the ability to mediate the switch from senescence to apoptosis of bladder cancer cells in xenograft mice. Meanwhile, TFA combined with chemotherapeutic drugs produced a substantial suppression of tumor growth as well as an extension of survival time in vivo ., Conclusions: Based on our finding that integrin β1/AKT acted primarily to impart premature senescence to bladder cancer cells cultured in collagen gel, we suggest that integrin β1 might be a feasible target for bladder cancer eradication., (© The Author(s) 2022. Introduction: We aimed to develop an easy-to-use individual survival prognostication tool based on competing risk analyses to predict the risk of 5-year cancer-specific death after radical prostatectomy for patients with prostate cancer (PCa). Methods: We obtained the data from the Surveillance, Epidemiology, and End Results (SEER) database (2004-2016). The main variables obtained included age at diagnosis, marital status, race, pathological extension, regional lymphonode status, prostate specific antigen level, pathological Gleason Score. In order to reveal the independent prognostic factors. The cumulative incidence function was used as the univariable competing risk analyses and The Fine and Gray's proportional subdistribution hazard approach was used as the multivariable competing risk analyses. With these factors, a nomogram and risk stratification based on the nomogram was established. Concordance index (C-index) and calibration curves were used for validation. Results: A total of 95,812 patients were included and divided into training cohort ( n = 67,072) and validation cohort ( n = 28,740). Seven independent prognostic factors including age, race, marital status, pathological extension, regional lymphonode status, PSA level, and pathological GS were used to construct the nomogram. In the training cohort, the C-index was 0.828 (%95CI, 0.812-0.844), and the C-index was 0.838 (%95CI, 0.813-0.863) in the validation cohort. The results of the cumulative incidence function showed that the discrimination of risk stratification based on nomogram is better than that of the risk stratification system based on D'Amico risk stratification. Conclusions: We successfully developed the first competing risk nomogram to predict the risk of cancer-specific death after surgery for patients with PCa. Writing sequences play an important role in handwriting of Chinese characters. However, little is known regarding the integral brain patterns and network mechanisms of processing Chinese character writing sequences. The present study decoded brain patterns during observing Chinese characters in motion by using multi-voxel pattern analysis, meta-analytic decoding analysis, and extended unified structural equation model. We found that perception of Chinese character writing sequence recruited brain regions not only for general motor schema processing, i.e., the right inferior frontal gyrus, shifting, and inhibition functions, i.e., the right postcentral gyrus and bilateral pre-SMA/dACC, but also for sensorimotor functions specific for writing sequences. More importantly, these brain regions formed a cooperatively top-down brain network where information was transmitted from brain regions for general motor schema processing to those specific for writing sequences. These findings not only shed light on the neural mechanisms of Chinese character writing sequences, but also extend the hierarchical control model on motor schema processing.

Many studies have examined the cognitive and neural mechanisms of bilingual language control, but few of them have captured the pattern information of brain activation. However, language control is a functional combination of both cognitive control and language production which demonstrates distinct patterns of neural representations under different language contexts. The first aim of the present study was to explore the brain activation patterns of language control using multivoxel pattern analysis (MVPA). During the experiment, Chinese-English bilinguals were instructed to name pictures in either Chinese or English according to a visually presented cue while being scanned with functional magnetic resonance imaging (fMRI). We found that patterns of neural activity in frontal brain regions including the left dorsolateral prefrontal cortex, left inferior frontal gyrus, left supplementary motor area, anterior cingulate cortex, bilateral precentral gyri, and the left cerebellum reliably discriminated between switch and non-switch conditions. We then modeled causal interactions between these regions by applying effective connectivity analyses based on an extended unified structure equation model (euSEM). The results showed that frontal and fronto-cerebellar connectivity were key components of the language control network. These findings further reveal the engagement of the cognitive control network in bilingual language production.

Bilingual language experience, such as switching between languages, has been shown to shape both cognitive and neural mechanisms of non-linguistic cognitive control. However, the neural adaptations induced by language switching remain unclear. Using fMRI, the current study examined the impact of short-term language switching training on the neural network of domain-general cognitive control for unbalanced Chinese-English bilinguals. Effective connectivity maps were constructed by using the extended unified structural equation models (euSEM) within 10 common brain regions involved in both language control and domain-general cognitive control. Results showed that, the dorsal anterior cingulate cortex/pre-supplementary motor area (dACC/pre-SMA) lost connection from the right thalamus after training, suggesting that less neural connectivity was required to complete the same domain-general cognitive control task. These findings not only provide direct evidence for the modulation of language switching training on the neural interaction of domain-general cognitive control, but also have important implications for revealing the potential neurocognitive adaptation effects of specific bilingual language experiences., Competing Interests: NO authors have competing interests.

It has been documented that conflict adaptation (conflict resolution in a task enhanced by that in a previous task) exists not only in the same domain but also across different domains with shared cognitive control mechanisms. For the first time, the present study adopted a cross-task adaptive blocked design to examine the relationship between bilingual language control and cognitive control from the perspective of the immediately adjacent, mutual influence on the neural connectivity level. The results showed that the conflict setting induced by previous tasks changed the nodal degrees of the anterior cingulate cortex/presupplementary motor area and the right thalamus, and connectivity strength of shared links between adjacent language and cognitive control tasks. In addition, pre-activation of the cognitive control network affected the transitivity of the successive use of the language control network. These findings not only indicate a cross-task adaptation effect on the neural connectivity level, but also provide evidence for similarities in conflict detection and inhibition control between language-specific control and domain-general cognitive control. When bilinguals intend to speak in their first (L1) or second language (L2) according to the environment, a brain network involving cortical-subcortical regions is recruited to resolve cross-language interference. Research has found that the activation of these brain regions varies with language contexts. However, previous studies have not yet examined adaptive changes in the interactions of brain regions for different language contexts. To address this gap, we adopted extended unified structural equation modeling (euSEM) to identify the connectivity patterns of the bilingual control network. Twenty-one unbalanced Chinese-English bilinguals were instructed to name pictures in L1-single, L2-single, and dual-language contexts while undergoing functional magnetic resonance imaging (fMRI). We found that unbalanced bilinguals relied on a more functional integrated brain network, which was reflected by clearer core-periphery structures and increased global efficiency, in dual-language and L2-single contexts compared to L1-single context. Furthermore, the pattern of brain connectivity in the dual-language context was more similar to that in the L1-single context than the L2-single context. More importantly, we found more similarities between the connectivity patterns of dual-language and L1-single contexts in bilinguals with lower inhibitory control abilities. Purpose: To study prognostic values of the clinicopathological characteristics and survival outcomes in micropapillary urothelial carcinoma (MPUC) of the urinary bladder., Method: We used the national Surveillance, Epidemiology, and End Results database (2004-2016) to compare MPUC with transitional cell carcinoma (TCC) and to investigate prognostic values of clinicopathological characteristics, as well as survival outcomes, in MPUC of the urinary bladder. A multivariable Cox proportional hazard model, subgroup analyses, and propensity score matching were used., Results: In all, 519 patients with MPUC and 154 453 patients with TCC were enrolled. Compared with TCC, patients with MPUC had a higher rate of muscle invasive disease (P < .001), lymph node metastasis (P < .001), and distal metastasis (P < .001), as well as higher tumor grade (P < .001). According to the survival analyses, the MPUC group also had lower survival probability in both cancer-specific mortality (CSM) (P < .0001) and overall mortality (OM) analyses (P < .0001). Cox proportional hazard regression showed that the MPUC group had a higher risk of OM (hazard ratios [HR] = 1.39, 95% confidence intervals [CI] = 1.22-1.57, P < .0001), although the CSM (HR = 1.18, 95% CI = 1.00-1.40, P = .0505) in that group was fair. In the subgroup analysis, only MPUC patients without distal metastasis faced a higher risk of CSM (HR = 1.33, 95% CI = 1.101.61, P < .0001)., Conclusions: Micropapillary urothelial carcinoma prognosis is poorer than that of TCC. Objective: To investigate the diagnostic role of magnetic resonance imaging (MRI)-targeted biopsy (TB) for prostate cancer (PCa) in biopsy-naïve men., Materials and Methods: Own control studies and randomized controlled trials (RCTs) up to December 2018 were identified via a systematic search of PubMed, Embase, Ovid, and the Cochrane Library. We pooled relative sensitivity (or risk ratio [RR]) to compare diagnostic efficiency for PCa and clinically significant PCa (csPCa) between TB and systematic biopsy (SB). The independent role of either biopsy pathway was evaluated for participants with positive/negative MRI., Results: Thirty-one studies consisting of 25 own control studies and 6 RCTs were included. We identified 4,020 biopsy-naïve men with positive MRI who underwent two biopsies concurrently, with PCa/csPCa detection rates of 65.90 and 45.13%, respectively. TB and SB did not differ in the detection of any PCa (RR 0.98, 95% confidence interval [CI] 0.92-1.05). However, TB detected more csPCa (RR 1.19, 95% CI 1.10-1.30) and more PCa with a Gleason score ≥3+4 (RR 1.20, 95% CI 1.07-1.34). Using a combined test as a reference, omitting SB resulted in detecting 12.81% less csPCa and 20.76% less clinically insignificant PCa (cinsPCa), and omitting TB resulted in detecting 25.69% less csPCa and 10.8% more cinsPCa. For patients with negative MRI, omitting SB led to underdetection of 30.29% of any PCa (10.9% of csPCa)., Conclusions: Combining TB and SB increased the diagnostic accuracy of csPCa for biopsy-naïve men with positive MRI, and omitting SB for patients with a negative MRI would lead to the underdetection of nearly 10% of csPCa., (© 2019 S. Fusarium graminearum (teleomorph: Ascomycota, Hypocreales, Gibberella , Gibberella zeae ) is a destructive fungal pathogen that threatens the production and quality of wheat and barley worldwide. Controlling this toxin-producing pathogen is a significant challenge. In the present study, the commercially available strain Bacillus amyloliquefaciens ( Bacteria , Firmicutes , Bacillales , Bacillus ) FZB42 showed strong activity against F. graminearum The lipopeptide bacillomycin D, produced by FZB42, was shown to contribute to the antifungal activity. Purified bacillomycin D showed strong activity against F. graminearum , and its 50% effective concentration was determined to be approximately 30 μg/ml. Analyses using scanning and transmission electron microscopy revealed that bacillomycin D caused morphological changes in the plasma membranes and cell walls of F. graminearum hyphae and conidia. Fluorescence microscopy combined with different dyes showed that bacillomycin D induced the accumulation of reactive oxygen species and caused cell death in F. graminearum hyphae and conidia. F. graminearum secondary metabolism also responded to bacillomycin D challenge, by increasing the production of deoxynivalenol. Biological control experiments demonstrated that bacillomycin D exerted good control of F. graminearum on corn silks, wheat seedlings, and wheat heads. In response to bacillomycin D, F. graminearum genes involved in scavenging reactive oxygen species were downregulated, whereas genes involved in the synthesis of deoxynivalenol were upregulated. Phosphorylation of MGV1 and HOG1, the mitogen-activated protein kinases of F. graminearum , was increased in response to bacillomycin D. Taken together, these findings reveal the mechanism of the antifungal action of bacillomycin D. IMPORTANCE Biological control of plant disease caused by Fusarium graminearum is desirable. Bacillus amyloliquefaciens FZB42 is a representative of the biocontrol bacterial strains. In this work, the lipopeptide bacillomycin D, produced by FZB42, showed strong fungicidal activity against F. graminearum Bacillomycin D caused morphological changes in the plasma membrane and cell wall of F. graminearum , induced accumulation of reactive oxygen species, and ultimately caused cell death in F. graminearum Interestingly, when F. graminearum was challenged with bacillomycin D, the deoxynivalenol production, gene expression, mitogen-activated protein kinase phosphorylation, and pathogenicity of F. graminearum were significantly altered. These findings clarified the mechanisms of the activity of bacillomycin D against F. graminearum and highlighted the potential of B. amyloliquefaciens FZB42 as a biocontrol agent against F. graminearum ., (Copyright © 2017 American Society for Microbiology.)
