Endometrium fibrosis is the leading cause of uterine infertility. Macrophages participated in the occurrence and development of endometrial fibrosis. We previously reported that human umbilical cord multipotent stromal cells (hUC-MSCs) exerted their therapeutic effect in a macrophage-dependent manner in endometrial fibrosis. However precise mechanisms by which hUC-MSCs may influence macrophages in endometrial fibrosis remain largely unexplored. Here, we demonstrated that abnormal iron and lipid metabolism occurred in patients with intrauterine adhesions (IUA) and murine models. Ferroptosis has been proven to contribute to the progression of fibrotic diseases. Our results revealed that pharmacological activation of ferroptosis by Erastin aggravated endometrial fibrosis, while inhibition of ferroptosis by Ferrostatin-1 ameliorated endometrial fibrosis in vivo. Moreover, ferroptosis of macrophages was significantly upregulated in endometria of IUA murine models. Of note, transcriptome profiles revealed that CD36 gene expression was significantly increased in patients with IUA and immunofluorescence analysis showed CD36 protein was mainly located in macrophages. Silencing CD36 in macrophages could reverse cell ferroptosis. Dual luciferase reporter assay revealed that CD36 was the direct target of activation transcription factor 3 (ATF3). Furthermore, through establishing coculture system and IUA murine models, we found that hUC-MSCs had a protective role against macrophage ferroptosis and alleviated endometrial fibrosis related to decreased CD36 and ATF3. The effect of hUC-MSCs on macrophage ferroptosis was attributed to the upregulation of amphiregulin (AREG). Our data highlighted that macrophage ferroptosis occurred in endometrial fibrosis via the ATF3-CD36 pathway and hUC-MSCs protected against macrophage ferroptosis to alleviate endometrial fibrosis via secreting AREG. Background: Acute exacerbation (AE) of interstitial lung disease (ILD) is one of the most serious complications during perioperative period of lung cancer resection. This study aimed to investigate the correlation between preoperative 2- deoxy-2-[18F]fluoro-D-glucose ( 18 F-FDG) PET/CT findings and AE in lung cancer patients with ILD., Methods: We retrospectively reviewed the data of 210 patients who underwent lung resection for non-small cell lung cancer. Relationships between clinical data and PET images and AE were evaluated. The patients were divided into an AE(+) and an AE(-) group for multivariate logistic regression analysis. Receiver operating characteristic (ROC) curve analysis was conducted and the area under curve (AUC) was used to assess the predictive values., Results: Among 210 patients, 48 (22.8%) were diagnosed with ILD based on chest CT. Among them, 9 patients (18.75%) developed AE after lung resection and were defined as AE(+) group. The course of ILD was longer in AE(+) group compared to AE(-) group. More patients in AE(+) group had a history of AE and chronic obstructive pulmonary disease (COPD) than in AE(-) group. The maximum standardized uptake value (SUVmax) of the noncancerous interstitial pneumonia (IP) area and cancers in AE(+) group was significantly higher compared to AE(-) group. Univariate logistic regression analysis showed that AE, COPD, SUVmax of the noncancerous IP area, SUVmax of cancer, surgical method were significantly correlated with AE. The course of ILD[OR(95%CI) 2.919; P = 0.032], SUVmax of the noncancerous IP area[OR(95%CI) 7.630;P = 0.012] and D-Dimer level[OR(95%CI) 38.39;P = 0.041] were identified as independent predictors for AE in patients with ILD after lung cancer surgery. When the three indicators were combined, we found significantly better predictive performance for postoperative AE than that of SUVmax of the noncancerous IP area alone [0.963 (95% CI 0.914-1.00); sensitivity, 100%, specificity 87.2%, P < 0.001 vs. 0.875 (95% CI 0.789 ~ 0.960); sensitivity, 88.9%, specificity, 76.9%, P = 0.001; difference in AUC = 0.088, Z = 1.987, P = 0.04]., Conclusion: The combination of the course of ILD, SUVmax of the noncancerous IP area and D-Dimer levels has high predictive value for the occurrence of AE in patients with concomitant interstitial lesions., (© 2024. Sepsis is defined as a dysregulated host response to infection that leads to multiorgan failure. Innate immune memory, i.e., "trained immunity", can result in stronger immune responses and provide protection against various infections. Many biological agents, including β-glucan, can induce trained immunity, but these stimuli may cause uncontrolled inflammation. Oroxylin A (OA) is an active flavonoid compound that is derived from Scutellaria baicalensis. OA is an agonist for inducing trained immunity in vivo and in vitro, and β-glucan was used as a positive control. The protective effects of OA-induced trained immunity were evaluated in mouse models that were established by either lipopolysaccharide (LPS) administration or caecal ligation and puncture (CLP). The expression of inflammatory factors and signaling pathway components involved in trained immunity was evaluated in vitro using qRT‒PCR, western blotting (WB) and enzyme-linked immunosorbent assay (ELISA). Flow cytometry and confocal microscopy were used to examine reactive oxygen species (ROS) levels and phagocytosis in trained macrophages. A PCR array was used to screen genes that were differentially expressed in trained macrophages. Here, we revealed that OA alleviated sepsis via trained immunity. OA-treated macrophages displayed increased glycolysis and mTOR phosphorylation, and mTOR inhibitors suppressed OA-induced trained immunity by effectively reprogramming macrophages. The PCR array revealed key genes in the mTOR signaling pathway in OA-treated macrophages. Furthermore, OA targeted the Dectin-1-syk axis to promote LC3-associated phagocytosis (LAP) by trained macrophages, thereby enhancing the ability of these macrophages to protect against infection. This ability could be transferred to a new host via the adoptive transfer of peritoneal macrophages. This study is the first to provide new insights into the potential of OA-induced trained immunity to be used as a strategy to protect mice against sepsis by promoting LAP by macrophages., (© 2024. Background: Endometria are one of the important components of the uterus, which is located in the peritoneal cavity. Endometrial injury usually leads to intrauterine adhesions (IUA), accompanied by inflammation and cell death. We previously reported that both the endometrial ferroptosis was increased and monocytes/macrophages were involved in endometrial injury of IUA. Large peritoneal macrophages (LPMs) are recently reported to migrate into the injured tissues and phagocytose dead cells to repair the tissues. We previously demonstrated that mesenchymal stromal cells (MSCs) had made excellent progress in the repair of endometrial injury. However, it is unclear whether MSCs regulate the LPM efferocytosis against ferroptotic monocytes/macrophages in the injured endometria., Methods: Here, endometrial injury in IUA mouse model was conducted by uterine curettage and LPS injection surgery and the samples were collected at different times to detect the changes of LPMs and ferroptotic monocytes/macrophages. We conducted LPMs depletion assay in vivo and LPMs and Erastin-induced ferroptotic THP-1 cells coculture systems in vitro to detect the LPM efferocytosis against ferroptotic monocytes/macrophages. The IUA model was treated with MSCs, and their effects on LPMs and endometrial repair were analyzed. Flow cytometry, western blotting, quantitative real-time PCR, immunohistochemical analysis, ELISA, and RNA-sequencing were performed., Results: We found that LPMs migrated to the injured uteri in response to the damage in early phase (3 h), and sustained to a later stage (7 days). Astonishingly, we found that ferroptotic monocytes/macrophages were significantly increased in the injured uteri since 12 h after injury. Moreover, LPMs cocultured with Erastin-induced ferroptotic THP-1 cells in vitro, efferocytosis of LPMs against ferroptotic monocytes/macrophages was emerged. The mRNA expression profiles revealed that LPM efferocytosis against ferroptotic monocytes/macrophages was an induction of glycolysis program and depended on the PPARγ-HK2 pathway. Importantly, we validated that MSCs promoted the efferocytic capability and migration of LPMs to the injured uteri via secreting stanniocalcin-1 (STC-1)., Conclusion: The data collectively demonstrated first the roles of LPMs via removal of ferroptotic monocytes/macrophages and provided a novel mechanism of MSCs in repairing the endometrial injury., (© 2024. Pyrolysis is a promising method for recovering resources and energy products from municipal solid waste (MSW). Predicting MSW pyrolysis products is crucial for establishing an efficient pyrolysis system for resource recovery. In this study, a database was established based on MySQL to record relevant information on MSW pyrolysis, which includes the MSW ultimate analysis results, proximate analysis results, parameters of pyrolysis operation and yields of pyrolysis products, etc. Based on the database and with help of a deep artificial neural network (ANN) which contains 10 hidden layers, a prediction model was successfully established to predict the yield of char, liquid and gas products from MSW pyrolysis. The results showed that the coefficients of determination for predicting the yields of char, liquid and gas from the MSW pyrolysis are 0.841, 0.84, and 0.85, respectively; these values demonstrate an accuracy comparable to that achieved for product prediction from single biomass, indicating a successful model performance. The results also show that ash content and temperature are the most important input factors influencing the outputs, namely, yields of char, liquid and gas. Neutral oxygen evolution reaction (OER) with unique reactive environments exhibits extremely slow reaction kinetics, posing significant challenges in the design of catalysts. Herein, a built-in electric field between the tungstate (Ni-FeWO 4 ) with adjustable work function and Lewis acid WO 3 is elaborately constructed to regulate asymmetric interfacial electron distribution, which promotes electron accumulation of Fe sites in the tungstate. This decelerates the rapid dissolution of Fe under the OER potentials, thereby retaining the active hydroxyl oxide with the optimized OER reaction pathway. Meanwhile, Lewis acid WO 3 enhances hydroxyl adsorption near the electrode surface to improve mass transfer. As expected, the optimized Ni-FeWO 4 @WO 3 /NF self-supporting electrode achieves a low overpotential of 235 mV at 10 mA cm -2 in neutral media and maintains stable operation for 200 h. Furthermore, the membrane electrode assembly constructed by such self-supporting electrode exhibits robust stability for 250 h during neutral seawater electrolysis. Phenol is one of the important ingredients of pyrolysis oil, contributing to the high biotoxicity of pyrolysis oil. To promote the degradation and conversion of phenol during anaerobic digestion, cheap hydro-chars with high phenol adsorption capacity were produced. The phenol adsorption capabilities of the plain hydro-char, plasma modified hydro-char at 25 °C (HC-NH 3 -P-25) and 500 °C (HC-NH 3 -P-500) were evaluated, and their adsorption kinetics and thermodynamics were explored. Experimental results indicate that the phenol adsorption capability of HC-NH 3 -P-500 was the highest. The phenol adsorption kinetics of all samples followed the pseudo-second-order equation and interparticle diffusion model, indicating that the adsorption rate of phenol was controlled by interparticle diffusion and chemistry adsorption simultaneously. By DFT calculations, π-π stacking and hydrogen bond are the main interactions for phenol adsorption. It was observed that an enriched graphite N content decreased the average vertical distance between hydro-chars and phenol in π-π stacking complex, from 3.5120 to 3.4532 Å, causing an increase in the negative adsorption energy between phenol and hydro-char from 13.9330 to 23.4181 kJ/mol. For hydrogen bond complex, the average vertical distance decreased from 3.4885 to 3.3386 Å due to the increase in graphite N content; causing the corresponding negative adsorption energy increased from 19.0233 to 19.9517 kJ/mol. Additionally, the presence of graphite N in the hydro-char created a positive diffusion region and enhanced the electron density between hydro-char and phenol. Analyses suggest that enriched graphite N contributed to the adsorption complex stability, resulting in an improved phenol adsorption capacity., (© 2023. Electronic structure manipulation with regard to active site coordination is an effective strategy to improve the electrocatalytic oxygen evolution reaction (OER) activity. Herein, we present the structure-activity relationship between oxygen-atom-mediated electron rearrangement and active site coordination asymmetry. Ni 2+ ions are introduced to FeWO 4 on Ni foam (NF) via self-substitution to break the symmetry of the FeO 6 octahedron and regulate d-electron structure of Fe sites. Structural regulation optimizes the adsorption energy of hydroxyl on the Fe sites and promotes the partial formation of hydroxyl oxide with high OER activity on the tungstate surface. Fe 0.53 Ni 0.47 WO 4 /NF with the asymmetric FeO 6 octahedron of Fe sites can achieve an ultralow overpotential of 170 mV at 10 mA cm -2 and 240 mV at 1000 mA cm -2 with robust stability for 500 h at high current density under alkaline conditions. Intrauterine adhesion (IUA) is manifested by endometrial fibrosis and inflammation, which seriously affects female reproductive health. Macrophages are mainly inflammatory cells and have been reported to participate in the fibrosis of IUA. Oroxylin A (OA), a kind of flavonoid compounds, was showed to possess the inhibitory effects on inflammation and fibrosis. However, the role of OA in IUA remains unclear. In the present study, we found that OA effectively alleviated the level of inflammation and uterine fibrosis in IUA mice. OA also decreased the macrophage pyroptosis which increased in uteri of IUA mice. Pyroptosis is a programmed cell death accompanied by an inflammatory response. Moreover, OA repressed the mediators of pyroptosis including the expression of NOD-like receptor family pyrin domain containing 3 (NLRP3), caspase-1 and Gasdermin D (GSDMD) and the release of IL-1β, IL-18 and cleaved-caspase-1 in J774A.1 cells induced by LPS/ATP in vitro. Mechanistically, the alleviation of OA on uterine fibrosis is achieved by inhibiting macrophage pyroptosis via SIRT3-SOD2-ROS pathway. The effective disposal of municipal solid wastes (MSW) and its incineration-derived fly ash (IFA), which contains large amounts of heavy metals (HMs) and chlorine (Cl), is an urgent task. In this study, IFA was used to reform MSW pyrolysis volatiles within 500-800 °C. The changes of reformed pyrolysis products, the migration characteristics of HMs and Cl between IFA and pyrolysis products were investigated. The results indicated that the O- and Cl-containing compounds in pyrolysis oil tended to decrease, light hydrocarbons and its calorific value increased accordingly after reforming; more CH 4 and H 2 gases were produced concurrently. The increase in reforming temperature enhanced these trends. The IFA absorbed Cl from volatiles during reforming, which reduced HCl in the gas product. The toxicity equivalent (TEQ) of PCDD/Fs in IFA decreased dramatically from 0.47 μg/kg to 0.0055 μg/kg after reforming at 500 °C, and it decreased with increasing reforming temperature. Some of the HMs' concentrations in the used IFAs increased, but their leaching capacity all decreased significantly at 800 °C except for Cr. The used IFA at 800 °C (IFA-800) corresponded to the lowest HMs leaching concentrations and could meet the landfill requirements; while the used IFA at 500 °C (IFA-500) corresponded to the maximum carbon deposition of 14.63 wt%, providing the energy source for its melting. Therefore 800 °C was recommended for harmless disposal of IFA, and 500 °C was better for a further melting of IFA., The contamination of pyrolysis liquid caused by inorganic Cl-containing compounds at 500 and 800 °C with much lower levels than the original. LS-007, an inhibitor of cyclin-dependent kinase 9 (CDK9), exhibits potential antitumor activity against chronic lymphocytic leukemia and ovarian cancer, but its effect on melanoma and tumor microenvironment (TME) has not been reported yet. This study aimed to investigate the role of LS-007 in B16F10 melanoma and relevant mechanisms. LS-007 significantly inhibited viability and induced apoptosis of B16F10 cells in a dose-dependent manner, which were accompanied with the increased ratio of Bax to Bcl-2 and decreased Mcl-1 mRNA level. Western blot analysis showed that LS-007 increased the expression of cleaved caspase-3 and poly ADP-ribose polymerase (PARP). Furthermore, flow cytometry analysis and qRT-PCR results showed that LS-007 treatment resulted in cell cycle arrest by changing cell cycle-related gene expression. Notably, in vivo evaluation showed that LS-007 significantly decreased the weight and volume of tumor and the expression of Ki67, promoted the expression of iNOS and inhibited the expression of CD206, suggesting that LS-007 might inhibit tumor growth by suppressing polarization of macrophages into tumor-associated macrophages (TAMs) in the TME. The increase in M1/M2 treated with LS-007 detected by flow cytometry hinted that macrophages were polarized towards an antitumor phenotype. Objective: The aim of the study was to investigate the clinical, neuropsychological, and regional cerebral blood flow (rCBF) perfusion changes in patients with neuropsychiatric symptoms caused by nitrous oxide (N 2 O) abuse., Methods: A total of 16 patients with neuropsychiatric symptoms caused by nitrous oxide abuse were recruited for this study. The study was carried out in the withdrawal phase of N 2 O abuse. A 925-1110 MBq 99m Tc-ECD was administered intravenously. SPECT/CT images were collected with a low-energy and high-resolution collimator. The region uptake statistics of different brain regions of interest between patients with N 2 O abuse and normal people of the databases for younger subjects from the Scenium DB Comparison software were calculated automatically., Results: The clinical manifestations of the 16 patients with neuropsychiatric symptoms were mood lability, anxiety, hallucination, delusion, agitation, confusion, and other psychiatric symptoms. In addition, 15 of the patients also complained of memory decline; 14 patients manifested numbness or paresthesia; 14 patients developed limb weakness, and their motor impairments were more severe in the lower limbs than in the upper limbs; and eight patients had urinary and defecation disturbances. In the neuropsychological examination, the BPRS score was 54.69 ± 11.48, the HAMD score was 30.00 ± 11.06, the HAMA score was 18.06 ± 5.77, the MMSE score was 28.06 ± 2.29, and the MoCA score was 25.06 ± 3.40. SPECT showed hypoperfusion in the frontal and temporal lobes, which is consistent with the clinical findings., Conclusion: This was the first study to demonstrate the obvious effect of N 2 O abuse on CBF in patients with neuropsychiatric symptoms. With the implementation of municipal solid wastes (MSW) source segregation collection policy, the combustible waste components are suitable for pyrolysis treatment to recover value-added energy such as gas or oil. In this research, the volatile compounds from MSW pyrolysis were reformed by the char obtained from the same process and the high-temperature flue gas (HTFG) of 1200 °C was supplying as heating resource and gasifying agent in a tailor-made experimental set-up. The final yields and properties of the syngas, char, and oil after reforming were investigated. Experimental studies showed that the composition of HTFG had a significant influence on the products. As the HTFG being CO 2 and its flowrate increased from 0 to 4 L/min/(kg MSW ), the energy share in the gas first increased from 8.29 to 15.1 MJ/(kg MSW ) with a higher heat value (HHV) of 19.9 MJ/Nm 3 , then it decreased slightly with a further increase in the flowrate. However, as the HTFG was H 2 O or the model flue gas (consisting of CO 2 , H 2 O, O 2 and N 2 ), the higher flowrate always increased the energy share and cold gasification efficiency (CGE) in the gas products. Both CO 2 and H 2 O participated the volatiles' reforming reactions and enhanced char gasification; while the volatiles' reforming inhibited char gasification. When the flowrate of the model flue gas increased from 0 to 12.04 L/min/(kg MSW ), the oil and char yields decreased and the CGE increased to more than 75%, which was much higher than that from direct air gasification of MSW; moreover, the gas products had much higher HHVs. Chronic inflammation caused by persistent infections and metabolic disorders is thought to contribute to the increased cancer risk and the accelerated cancer progression. Oppositely, acute inflammation induced by bacteria-based vaccines or that is occurring after cancer selectively inhibits cancer progression and metastasis. However, the interaction between inflammation and cancer may be more complex than the current explanations for the relationship between chronic and acute inflammation and cancer. In this review, we described the impact of inflammation on cancer on the basis of three perspectives, including inflammation with different durations (chronic and acute inflammation), different scopes (systemic and local inflammation) and different occurrence sequences (inflammation occurring after and before cancer). In addition, we also introduced bacteria/virus-based cancer immunotherapies. We perceive that inflammation may be a double-edged sword with cancer-promoting and cancer-suppressing functions in certain cases. Purpose: Since immune cells in the tumor microenvironment (TME) can affect the development and progression of tumors, strategies modulating immune cells are considered to have an important therapeutic effect. As a TLR7/8 agonist, R848 effectively activates the innate immune cells to exert an anti-tumor effect. Mn 2+ has been reported to strongly promote the maturation of antigen-presenting cells (APCs), thereby enhancing the cytotoxicity of CD8 + T cells. Thus, we tried to investigate whether chitosan-poly(acrylic acid) nanoparticles (CS-PAA NPs) loaded with R848 and MnCl 2 (R-M@CS-PAA NPs) could exert an anti-tumor effect by regulating the function of immune cells., Methods: R-M@CS-PAA NPs were prepared, and their basic characteristics, anti-tumor effect, and potential mechanisms were explored both in vitro and in vivo., Results: R-M@CS-PAA NPs easily released MnCl 2 and R848 at low pH. In B16F10 mouse melanoma model, R-M@CS-PAA NPs exerted the most significant anti-melanoma effect compared with the control group and CS-PAA NPs loaded with R848 or MnCl 2 alone. FITC-labeled R-M@CS-PAA NPs were displayed to be accumulated at the tumor site. R-M@CS-PAA NPs significantly increased the infiltration of M1 macrophages and CD8 + T cells but reduced the number of suppressive immune cells in the TME. Moreover, in vitro experiments showed that R-M@CS-PAA NPs polarized macrophages into the M1 phenotype to inhibit the proliferation of B16F10 cells. R-M@CS-PAA NPs also enhanced the killing function of CD8 + T cells to B16F10 cells. Of note, R-M@CS-PAA NPs not only promoted the maturation of APCs such as dendritic cells and macrophages by STING and NF-кB pathways, but also enhanced the ability of dendritic cells to present ovalbumin to OT-I CD8 + T cells to enhance the cytotoxicity of OT-I CD8 + T cells to ovalbumin-expressing B16F10 cells., Conclusion: These data indicate that the administration of R-M@CS-PAA NPs is an effective therapeutic strategy against melanoma., Competing Interests: The authors report no conflicts of interest in this work., (© 2021 Liu et al.)

A technology of sewage sludge (SS) pyrolysis coupled with self-supplied steam reforming of the volatile is developed to utilize the latent heat of the steam vapor released in the SS drying & pyrolysis process. An integrated reactor consisting of a vertical free-falling pyrolysis section and a horizontal screw-moved reforming section is designed for this purpose. The performance of the reactor shows that by changing the moving speed of the char in the reforming section, high quality syngas with an H 2 /CO ratio of 4.37 and a percentage of H 2  + CO up to 66.58 vol% can be obtained at approximately 570-600 °C for the dry SS. There is an optimum moving speed of the screw for producing the highest volume of the syngas. A higher moving speed of the screw also results in a higher concentration of the aromatic compounds in the final pyrolysis oil. When the initial moisture content of SS increases from 0 to 65.50%, the H 2 /CO ratio and H 2 /CO 2 ratio in the syngas increase from 4.37 to 30.87 and from 2.1 to 2.6 correspondingly, and the final oil yield decreases from 24.03 wt% to 14.16 wt%. Moreover, the total energy recovery efficiency decreases from 88.85% to 61.92%, while the energy portion of syngas shows a peak of 44.18% of the total energy input when the initial moisture content is 41.26%. The integrated reactor also provides a good opportunity for adding a catalyst such as dolomite to make the process more effective. The gas and oil product derived from municipal solid waste (MSW) pyrolysis was upgraded by utilizing the interaction between the volatile compounds and the char and the mechanism involved is explored. The influences of operation parameters, including interaction temperature, char/volatiles mass ratio (C/V) and gas hourly space velocity (GHSV) of the volatiles on the distribution and property of the upgraded products were investigated. The results showed that the higher interaction temperature, higher C/V and lower GHSV favored the conversion of condensable volatiles into gas products, thus increasing the gas yield in the outlet stream. The highest gas yield (44.14 wt%) was obtained at 700 °C with the natural C/V ratio (0.8) and GHSV, which was twice of the gas yield in the volatiles. The chemical energy portion of gas increased to 8065 kJ/kg MSW from 3209 kJ/kg MSW at this condition. Syngas with H 2 /CO molar ratio of around 2 can be obtained at 700 °C with C/V ratio of 0.8 or at 600 °C with higher C/V ratios (C/V = 1.5-2.2). Oxygenates and acidity of the reformed oil products decreased; but monoaromatics and light polyaromatics concentration increased greatly. Heavy polycyclic aromatic hydrocarbons (PAHs) in the liquid products were degraded after volatiles/hot char interaction. In many vertebrates, the brain's right hemisphere which is connected to the left visual field specializes in the processing of information about threats while the left hemisphere which is connected to the right visual field specializes in the processing of information about conspecifics. This is referred to as hemispheric lateralization. But individuals that are too predictable in their response to predators could have reduced survival and we may expect selection for somewhat unpredictable responses. We studied hemispheric lateralization in yellow-bellied marmots Marmota flaviventer , a social rodent that falls prey to a variety of terrestrial and aerial predators. We first asked if they have lateralized responses to a predatory threat. We then asked if the eye that they used to assess risk influenced their perceptions of risk. We recorded the direction marmots were initially looking and then walked toward them until they fled. We recorded the distance that they responded to our experimental approach by looking, the eye with which they looked at us, and the distance at which they fled (i.e., flight initiation distance; FID). We found that marmots had no eye preference with which they looked at an approaching threat. Furthermore, the population was not comprised of individuals that responded in consistent ways. However, we found that marmots that looked at the approaching person with their left eye had larger FIDs suggesting that risk assessment was influenced by the eye used to monitor the threat. These findings are consistent with selection to make prey less predictable for their predators, despite underlying lateralization.

In this research morphological techniques were used to characterize dechlorination process of PVC when it is in the mixed waste plastics and the two important factors influencing this process, namely, the proportion of PVC in the mixed plastics and heating rate adopted in the pyrolysis process were investigated. During the pyrolysis process for the mixed plastics containing PVC, the morphologic characteristics describing PVC dechlorination behaviors were obtained with help of a high-speed infrared camera and image processing tools. At the same time emission of hydrogen chloride (HCl) was detected to find out the start and termination of HCl release. The PVC contents in the mixed plastics varied from 0% to 12% in mass and the heating rate for PVC was changed from 10 to 60°C/min. The morphologic parameters including "bubble ratio" (BR) and "pixel area" (PA) were found to have obvious features matching with PVC dechlorination process therefore can be used to characterize dechlorination of PVC alone and in the mixed plastics. It has been also found that shape of HCl emission curve is independent of PVC proportions in the mixed plastics, but shifts to right side with elevated heating rate; and all of which can be quantitatively reflected in morphologic parameters vs. temperature curves., (Copyright © 2016 Elsevier Ltd. In mammals, bitter taste is mediated by TAS2Rs, which belong to the family of seven transmembrane G protein-coupled receptors. Since TAS2Rs are directly involved in the interaction between mammals and their dietary sources, it is likely that these genes evolved to reflect species-specific diets during mammalian evolution. Here, we analyzed the amino acids responsible for the difference in sensitivities of TAS2R16s of various primates using a cultured cell expression system. We found that the sensitivity of TAS2R16 varied due to several amino acid residues. Mutation of amino acid residues at E86T, L247M, and V260F in human and langur TAS2R16 for mimicking the macaque TAS2R16 decreased the sensitivity of the receptor in an additive manner, which suggests its contribution to the potency of salicin, possibly via direct interaction. However, mutation of amino acid residues 125 and 133 in human TAS2R16, which are situated in helix 4, to the macaque sequence increased the sensitivity of the receptor. These results suggest the possibility that bitter taste sensitivities evolved independently by replacing specific amino acid residues of TAS2Rs in different primate species to adapt to species-specific food.

To compare the role of 99mTc-MIBI SPECT-CT image and US in reoperation of persistent secondary hyperparathyroidism patients. A total of 8 persistent secondary hyperparathyroidism patients underwent parathyroidectomy. The sensitivity and accuracy of US and 99mTc-MIBI images before operation were determined. 9 of 14 surgical resection tissues from 8 persistent secondary hyperparathyroidism patients were confirmed to parathyroid hyperplasia. The results showed that the sensitivities were 77.8% and 100%, respectively, for US and 99mTc-MIBI SPECT-CT images. And the accuracies of US and 99mTc-MIBI dual time planar image and SPECT-CT were 50%, 78.6%, respectively. There was significantly difference between two procedures (P=0.021). The superior and inferior localization of glands were both detected by the ultrasound and scintigraphy. Two ectopic parathyroidism nodules were found by 99mTc-MIBI SPECT-CT imaging, while US provided no consistent findings. There was no significantly difference between two procedures (P=0.3). Although two cases of them were not confirmed by pathology, the iPTH of them (800 and 1429 respectively) were much higher than other four cases (<400) pg/ml. PTX is a safe and effective treatment for the patients with persistent SHPT. 99mTc-MIBI planar and SPECT-CT imaging may provide more helps for clinician's localization the hyperparathyroidism glands accurately.

Infanticide was first observed in langurs nearly 50 years ago, and this rare phenomenon has been inferred to have either an evolutionarily adaptive function or to be a pathological and non-functional behavior. In this study, we report 5 male takeover events in one-male groups of white-headed langurs in the Nongguan Karst Hills, Guangxi, China from 1998 to 2006. We recorded 13 attacks on 9 infants by extra-group males or new resident males. During the male takeovers, all of the infants younger than 6 months (with an average age of 3.6 months [N = 11]) in the groups disappeared. The infant death rate during the 4.2 months after takeover by a new male was significantly higher than the infant death rate calculated for most of the year. Older infants that were still nursing (with an average age of 14.1 months [N = 7]) were often attacked and seriously wounded by the extra-group males or new resident males, but all of them survived. The interbirth intervals of females whose infants were assumed to be killed by males were significantly reduced relative to those of females in groups with stable male tenure (mean = 10 months vs 25 months). Our data suggest that males kill unrelated and unweaned infants during the takeover period to decrease the time until the infants' mothers resume fertility. Thus, infanticide would support sexual selection theory in white-headed langurs. The data also show that infanticidal behavior was directed toward the infants, especially those who were still nursing. Here, we investigated the sensitivities of TAS2R16s of various primates by using a cultured cell expression system, and found that the sensitivity of each primate species varied according to the ligand. Especially, the sensitivity of TAS2R16 of Japanese macaques to salicin was much lower than that of human TAS2R16, which was supported by behavioural tests. These results suggest the possibility that bitter-taste sensitivities evolved independently by replacing specific amino acid residues of TAS2Rs in different primate species to adapt to food items they use.

A carotenogenesis gene cluster from the purple nonsulfur photosynthetic bacterium Rhodobacter azotoformans CGMCC 6086 was cloned. A total of eight carotenogenesis genes ( crtA , crtI , crtB , tspO , crtC , crtD , crtE , and crtF ) were located in two separate regions within the genome, a 4.9 kb region containing four clustered genes of crtAIB - tspO and a 5.3 kb region containing four clustered genes of crtCDEF . The organization was unusual for a carotenogenesis gene cluster in purple photosynthetic bacteria. A gene encoding phytoene desaturase ( CrtI ) from Rba. azotoformans was expressed in Escherichia coli. The recombinant CrtI could catalyze both three- and four-step desaturations of phytoene to produce neurosporene and lycopene, and the relative contents of neurosporene and lycopene formed by CrtI were approximately 23% and 75%, respectively. Even small amounts of five-step desaturated 3,4-didehydrolycopene could be produced by CrtI . This product pattern was novel because CrtI produced only neurosporene leading to spheroidene pathway in the cells of Rba. azotoformans. In the in vitro reaction, the relative content of lycopene in desaturated products increased from 19.6% to 62.5% when phytoene reduced from 2.6 to 0.13 μM. Asian leaf-eating monkeys have flexible, environmentally adaptable feeding strategies. The diet and food choices of white-headed langurs (Trachypithecus leucocephalus) have, however, not yet been studied in the Nongguan Karst Hills in China, where one of the two main surviving populations of this endangered species lives. From 2000 to 2002 inclusive, we adopted an innovative video playback analysis method to identify the food species used by the langurs and to calculate the corresponding feeding frequency for each species. The video-aided methodology was shown to be efficient. It provided high-quality images, and the plant species could be identified in 98.3% of the feeding records. Based on this analysis, we found that the langurs in Nongguan fed on 70 species of plants, among which 24 were identified as their staple food species. The cumulative curve for the staple food species reached a plateau at 255 (43.7%) feeding records. It has been demonstrated that the development of NKT cells requires CD1d. The contribution of costimulatory molecules in this process has not been studied. Here we show that in mice with targeted mutations of B7-1/2 and CD28, the TCRbeta(+)alpha-Galcer/CD1d(+) (iValpha14 NKT) subset is significantly reduced in the thymus, spleen and liver. This is mainly due to decreased cell proliferation; although increased cell death in the thymi of CD28-deficient mice was also observed. Moreover, in the B7-1/2- and CD28-deficient mice, we found a decreased percentage of the CD4(-)NK1.1(+) subset and a correspondingly increased portion of the CD4(+)NK1.1(-) subset. In addition, the mice with a targeted mutation of either B7 or CD28 had a reduced susceptibility to Con A induced hepatitis, which is known to be mediated by NKT cells. Our results demonstrate that the development, maturation and function of NKT cell are modulated by the costimulatory pathway and thus expand the horizon of costimulation into NKT, which is widely viewed as a bridge between innate and adaptive immunity. As such, costimulation may modulate all major branches of cell-mediated immunity, including T cells, NK cells and NKT cells.

Despite negative selection in the thymus, significant numbers of autoreactive T cells still escape to the periphery and cause autoimmune diseases when immune regulation goes awry. It is largely unknown how these T cells escape clonal deletion. In this study, we report that CD24 deficiency caused deletion of autoreactive T cells that normally escape negative selection. Restoration of CD24 expression on T cells alone did not prevent autoreactive T cells from deletion; bone marrow chimera experiments suggest that CD24 on radio-resistant stromal cells is necessary for preventing deletion of autoreactive T cells. CD24 deficiency abrogated the development of experimental autoimmune encephalomyelitis in transgenic mice with a TCR specific for a pathogenic autoantigen. The role of CD24 in negative selection provides a novel explanation for its control of genetic susceptibility to autoimmune diseases in mice and humans.

The impact of timing of antigen introduction into fetus and neonates leads to the suggestion that pre-existing antigens are tolerogenic to immunocompetent cells generated thereafter. This hypothesis predicts that in patients with cancer who are undergoing bone marrow transplantation, newly produced T cells with specificity for pre-existing tumor cells will be inactivated by the tumor antigens in the host. Because the effect of tumor cells on developing cancer-reactive T cells has not been investigated, we set out to systematically analyze the impact of tumor cells in the periphery on the development of tumor-reactive T cells in the thymus and their immunocompetence in the periphery. Our data demonstrate that in the host in which a tumor is established in the periphery, the cancer-reactive T cells develop normally, remain fully immunocompetent, become activated in the periphery, and cause regression of large established tumors. The immunocompetence of T cells generated in an antigen-bearing host is also confirmed in a skin graft transplantation model.

Tumor evasion of T-cell immunity remains a significant obstacle to adoptive T-cell therapy. It is unknown whether the mode of immune evasion is dictated by the cancer cells or by the tumor antigens. Taking advantage of the fact that multiple lineages of tumor cells share the tumor antigen P1A, we adoptively transferred transgenic T cells specific for P1A (P1CTL) into mice with established P1A-expressing tumors, including mastocytoma P815, plasmocytoma J558, and fibrosarcoma Meth A. Although P1CTL conferred partial protection, tumors recurred in almost all mice. Analysis of the status of the tumor antigen revealed that all J558 tumors underwent antigenic drift whereas all P815 tumors experienced antigenic loss. Interestingly, although Meth A cells are capable of both antigenic loss and antigenic drift, the majority of recurrent Meth A tumors retained P1A antigen. The ability of Meth A to induce apoptosis of P1CTL in vivo alleviated the need for antigenic drift and antigenic loss. Our data showed that, in spite of their shared tumor antigen, different lineages of cancer cells use different mechanisms to evade T-cell therapy.

Many tissue-specific antigens are expressed in specialized cells called peripheral antigen-expressing cells (PAE) in the thymus and can induce central tolerance. While thymic medullary epithelial cells are the prototypic PAE that express peripheral antigens via an aire-dependent mechanism, some studies also describe bone marrow (BM)-derived dendritic cells (DC) and macrophages as PAE in both the thymus and secondary lymphoid organs. However, the role of these cells in development of tolerance to tissue-specific antigens has not been elucidated. Here we use BM radiation chimeric mice to study the existence of hematopoietic PAE and their contribution to tolerance to tissue-specific antigens. Our results reveal that BM-derived PAE exist in both central and secondary lymphoid organs and that the expression of peripheral antigens in the BM-derived cells does not correlate with aire expression. Using double-transgenic mice expressing TCR specific for a model antigen expressed under the control of a prostate-specific promoter, we show that expression of self antigen in PAE of non-hematopoietic origin is both necessary and sufficient to induce clonal deletion. Surprisingly, while BM-derived PAE fail to induce clonal deletion, they do cause the activation-induced cell death of autoreactive cells in the secondary lymphoid organs. Thus, BM-derived PAE have a distinct function in the maintenance of tolerance to tissue-specific antigens.

Recent work demonstrates that costimulatory molecules play a critical role for clonal deletion of autoreactive T cells in the thymus. The role of CD28 in the survival of autoreactive T cells in the periphery, however, has not been reported. Here we demonstrate that while mutation of the CD28 gene consistently increased the burden of autoreactive T cells in the thymus, such an increase was not always found in the periphery, as the CD28(-/-) autoreactive T cells disappeared in the spleen over a period between 4 and 10 weeks. The disappearance of autoreactive T cells associates with a diminished induction of Bcl-2 protein by the self antigen and an increased proportion of apoptotic cells in the periphery. Moreover, the elimination of autoreactive T cells in the periphery requires chronic stimulation by the self antigen, as adoptive transfer analysis revealed no enhancement of apoptosis in CD28(-/-) T cells in antigen-bearing hosts over a 3 day period. Thus, CD28 plays a significant role in both clonal deletion and survival of autoreactive T cells after chronic exposure to autoantigens, resulting in opposite effects on the burden of autoreactive T cells.

Multiple sclerosis (MS) is a chronic neurological disease of unknown etiology, but a genetic basis for the disease is undisputed. We have reported that CD24 is required for the pathogenicity of autoreactive T cells in experimental autoimmune encephalomyelitis, the mouse model of MS. Here we investigate the contribution of CD24 to MS by studying single-nucleotide polymorphism in the ORF among 242 MS patients and 207 population controls. This single-nucleotide polymorphism results in replacement of alanine (CD24a) with valine (CD24v) in the mature protein. We found that the CD24v/v renders a >2-fold increase in the relative risk of MS in the general population (P = 0.023). Among familial MS, the CD24v allele is preferentially transmitted into affected individuals (P = 0.017). Furthermore, 50% of CD24v/v patients with expanded disability status scale 6.0 reached the milestone in 5 years, whereas the CD24a/v (P = 0.00037) and CD24a/a (P = 0.0016) patients did so in 16 and 13 years, respectively. Moreover, our data suggest that the CD24v/v patients expressed higher levels of CD24 on peripheral blood T cells than did the CD24a/a patients. Transfection with CD24a and CD24v cDNA demonstrated that the CD24v allele can be expressed at higher efficiency than the CD24a alleles. Thus, CD24 polymorphism is a genetic modifier for susceptibility and progression of MS in the central Ohio cohort that we studied, perhaps by affecting the efficiency of CD24 expression on the cell surface.

B7H1 (PDL1) and B7DC (PDL2) are two new members of the B7 family that can interact with PD-1, a putative negative regulator for immune function. Recent studies have provided evidence for inhibitory functions of both members via PD-1. Meanwhile, compelling evidence exists for costimulatory function of both members. Here we demonstrate that expression of B7DC on the tumor cells promotes CD8 T cell-mediated rejection of tumor cells, at both the induction and effector phase of antitumor immunity. Moreover, B7DC binds to PD-1(-/-) cells and enhances T cell killing in a PD-1-independent mechanism. Our results demonstrate a novel pathway for B7DC to promote tumor immunity and may reconcile the apparently contradictory findings on the function of B7DC.
