This research study is centered on the sono-assisted photocatalytic degradation of a well-known antibiotic (ciprofloxacin; CIP) in aquatic media using a g-C 3 N 4 /NH 2 -UiO-66 (Zr) catalyst under visible light irradiation. Initially, the catalyst was prepared by a simple method, and its physiochemical features were thoroughly analyzed by XRD, FT-IR, FE-SEM, EDX, EDS-Dot-Mapping, and UV-Vis analytical techniques. After that, the impact of several influential factors affecting the performance of the applied sono-assisted photocatalytic process such as the initial concentration of CIP, solution pH, catalyst dosage, light intensity, and ultrasound power was fully assessed, and the optimal conditions were established. After 75 min of the sono-assisted photocatalytic treatment, the complete degradation of CIP (10 mg/L) was accomplished under the condition as follows: g-C 3 N 4 /NH 2 -UiO-66 (Zr), 0.6 g/L; pH, 5.0, and ultrasound power, light intensity 75 mw/cm 2 , 200 W/m 2 . Meanwhile, the photocatalytic degradation of CIP followed the pseudo-first-order kinetic model. In addition, the scavenger experiments demonstrated that OH˚ and O 2 ° - radicals played a key role in the sono-assisted photocatalytic degradation process. It is also acknowledged that the applied catalyst was reused for five consecutive runs with a minor loss observed in its degradation efficiency. In a further experiment, a significant synergistic effect with regard to the degradation of CIP was observed once all three major parameters (visible light, ultrasound waves, and catalyst) were used in combination compared to each used alone. To sum up, it is thought that the integration of g-C 3 N 4 /MOF-based catalyst, ultrasound waves, and visible light irradiation could be potentially applied as a promising strategy for the degradation of various pharmaceuticals on account of high degradation performance, simple operation, excellent reusability, and eco-friendly approach., (© 2024. Sarcoidosis presents as a systemic granulomatous disease of unknown etiology, characterized by the development of non-caseating granulomas that commonly affect the lungs, lymph nodes, skin, and eyes. Manifestations of various conditions such as infections, neoplasms, autoimmune, cardiovascular, and drug-induced diseases can bear resemblance to sarcoidosis. Coccidiosis, attributed to protozoan parasites of the Coccidia genus, primarily affects the intestinal tract but may also display systemic symptoms akin to those of sarcoidosis. In this particular case, we present a 46-year-old immunocompetent gentleman who had an extensive disease despite the patient's well-controlled diabetes and absence of residency in an endemic area with fungal infection, his only pertinent part of the history was his travel to endemic areas for short periods that raised the possibility of thinking about the disseminated fungal infection. The patient's symptoms initially attributed to and treated as sarcoidosis, which later did not respond to steroids, led us to consider other potential causes, including systemic fungal infection  Misdiagnosis of the sarcoidosis bears the risk of inappropriate treatment, potentially leading to exacerbated patient outcomes. There is increasing demand for natural food preservative in food manufacturing industry as it is the key to meet consumers' preferences toward healthier food choice. Plant is listed among the most important resources of bioactive components to be utilized as the green and natural food preservatives. There are more than 10,000 kinds of bioactive components in plants that possess antioxidant and antimicrobial properties. Artemisia with potential antimicrobial and antioxidant attributes, as well as functional and medicinal properties, is one of the most important plant species. The manuscript presents a comprehensive review of the potential of the Artemisia species as natural food preservatives. The current challenges and ways forward in using Artemisia EOs and extracts as food preservatives are also discussed. Background: Bone Morphogenetic Proteins and the related Growth and Differentiation Factors (GDFs) are much conserved signaling proteins. GDF5 is pivotal for skeletal development. Several skeletal dysplasia and malformation syndromes are known as a result of mutations in GDF5 . Multiple Synostosis Syndrome2 (SYNS2) is characterized by tarsal-carpal coalition, humeroradial synostosis, brachydactyly, and proximal symphalangism. In this study, we analyzed a large Iranian pedigree affected with a new type of SYNS2 (Farhud Type) in five successive generations., Methods: In this family-based study (1982-2022), Genetic linkage analysis of the pedigree (58affected, 62healthy) excluded the locus on chromosome 17q21-q22 in our previous study. Thus, we focused on 20q11.22 locus and GDF5 gene. Genetic investigations were performed on 16 patients with SYNS2 and 40 healthy individuals., Results: Whole-exome-sequencing results identified a heterozygote missense mutation in exon2 of GDF5 (NG_008076.1:g.9239G>A, NM_000557.2:c.1424G>A, S475N, rs121909347). This mutation was found in all patients but not in the unaffected individuals. This missense mutation is notable because S475 is strictly conserved among different species, and it is located in a highly conserved and active mature domain of GDF5 (phyloP100way=7.64). The corresponding defect in GDF5 may have unknown interaction with normal active 3 rd and 4 th structure of the product. Further bioinformatics study (amino acid multiple alignments) showed that the S475 is a much-conserved residue in many different species., Conclusion: These results introduce a new role of GDF5 in pathogenesis of a SYNS2 (Farhud Type), considered in genetic counseling, prenatal diagnosis, and as a potential target for molecular therapy, if possible., (Copyright© 2024 Farhud et al. Herein, a new MOF-based modified adsorbent for the efficient removal of Hg(ii) ions from water media was successfully prepared. Initially, a MOF nanocomposite was synthesized and applied as an efficient adsorbent for the removal of the target heavy metal ion. Following the synthesis, the MOF-based modified adsorbent was identified and characterized by SEM, XRD and FT-IR analytical instruments. The impact of several key variables such as pH of aqueous solution, adsorbent dosage, contact time, and initial concentration of the analyte of interest on the adsorption efficiency was also investigated in detail. Under the optimal conditions established (pH, 3; dose of adsorbent, 0.4 g L -1 ; contact time, 40 min and the analyte's concentration of 1 mg L -1 ) the removal efficiency of 96.3% for Hg(ii) was obtained. The results of the studies on the isotherm and kinetics of adsorption revealed that the adsorption process of Hg(ii) matched with the Langmuir isotherm ( R 2 > 0.990) and the pseudo 2nd-order kinetic models ( R 2 > 0.998). Additionally, reuse of the applied adsorbent for five consecutive tests exhibited a small percentage of drop (about 8%) in the removal efficiency of the target ion. The pervasive presence of poly- and perfluoroalkyl substances (PFAS) in diverse products has led to their introduction into wastewater systems, making wastewater treatment plants (WWTPs) significant PFAS contributors to the environment. Despite WWTPs' efforts to mitigate PFAS impact through physicochemical and biological means, concerns persist regarding PFAS retention in generated biosolids. While numerous review studies have explored the fate of these compounds within WWTPs, no study has critically reviewed their presence, transformation mechanisms, and partitioning within the sludge. Therefore, the current study has been specifically designed to investigate these aspects. Studies show variations in PFAS concentrations across WWTPs, highlighting the importance of aqueous-to-solid partitioning, with sludge from PFOS and PFOA-rich wastewater showing higher concentrations. Research suggests biological mechanisms such as cytochrome P450 monooxygenase, transamine metabolism, and beta-oxidation are involved in PFAS biotransformation, though the effects of precursor changes require further study. Carbon chain length significantly affects PFAS partitioning, with longer chains leading to greater adsorption in sludge. The wastewater's organic and inorganic content is crucial for PFAS adsorption; for instance, higher sludge protein content and divalent cations like calcium and magnesium promote adsorption, while monovalent cations like sodium impede it. In conclusion, these discoveries shed light on the complex interactions among factors affecting PFAS behavior in biosolids. Residual pharmaceuticals in the environment are a class of emerging pollutants that endanger human health. Tetracycline's family, including oxytetracycline (OTC), are known as one of the most produced and consumed antibiotics worldwide. The g-C 3 N 4 /Fe 3 O 4 nanocomposite with high level of catalytic efficiency features suitable performance in water/wastewater treatment. Therefore, in the present study, this nanocomposite was applied to remove the oxytetracycline from the aqueous environment. In this research study, g-C 3 N 4 /Fe 3 O 4 nanocomposite (serving as catalyst) was initially synthesized by a simple hydrothermal method. The effect of key operating parameters such as initial solution pH, dose of catalyst, contact time and initial concentration of OTC in aqueous solutions was investigated under UV irradiation. In addition, COD and TOC tests, the kinetics and the effect of radical scavengers on the applied photocatalytic process were all evaluated. The maximum removal efficiency of OTC (99.8 %) was achieved under the following conditions: neutral solution pH 7; catalyst dose, 0.7 g/L; and an initial OTC concentration of 5 mg/L. The data showed that the kinetics of the reaction followed the first-order model with R 2 of 0.9755. The respective COD and TOC efficiency values for the applied photocatalytic process were determined to be 87 and 59 %, respectively. In addition, the lowest removal efficiency of OTC was observed in the presence of tert-butanol radical scavengers, and OH radicals played a main role. Cells are very important to researchers due to their use in various biological studies in in vitro and in vivo settings. This importance stems from the short lifespan of most cells under laboratory conditions, which can pose significant challenges, such as the difficulties associated with extraction from the source tissue, ethical concerns about separating cells from human or animal models, limited cell passage ability, and variation in results due to differences in the source of the obtained cells, among other issues. In general, cells in laboratory conditions can divide into a limited number, known as the Hayflick limit, due to telomere erosion at the end of each cellular cycle. Given this problem, researchers require cell lines that do not enter the senescence phase after a limited number of divisions. This can allow for more stable studies over time, prevent the laborious work associated with cell separation and repeated cultivation, and save time and money in research projects. The aim of this review is to summarize the function and effect of immortalization techniques, various methods, their advantages and disadvantages, and ultimately the application of immortalization and cell line production in various research fields.

Background: We aimed to investigate two polymorphisms, rs8106922 and rs157580 of TOMM40 in Alzheimer's disease (AD)., Methods: In the present case-control research, we collected blood samples from 117 AD patients and 130 controls from Alzheimer's Hospital, residents of Tehran, Iran during the winter 2020 to autumn 2022. Following extraction of DNA, Genotyping of TOMM40 polymorphisms rs8106922 and rs157580 were examined by sequencing and ARMS/PCR approaches. We compared distributions of genotypes in both patient and healthy groups using the Chi-Square test., Results: Regarding rs157580, a statistically significant difference was observed in the GA genotype frequency between patient and healthy groups, in both univariate and multivariate modes with these results that have come respectively, and it can be regarded as a protection factor P <0.05).. No significant difference was observed in the frequency of A and G alleles between patient and healthy groups. Besides, concerning rs8106922, the AG genotype frequency in research groups in both univariate and multivariate cases, with these results that have come respectively was significantly different ( P =0.003) & ( P =0.009). Regarding GG genotype, a statistically significant difference was observed between the patient and healthy groups in both univariate and multivariate cases, respectively ( P =0.419) & ( P =0.425). Significant differences were observed in the G allele frequency for rs8106922 in the healthy and patient groups ( P =0.007), it can be regarded as a potential protective factor., Conclusion: It is possible to consider the TOMM40 gene as one of the potential genes concerning Alzheimer's disease., (Copyright© 2024 Hajilou et al. Background: In Iran, there is a lack of information and studies on acute rheumatic fever (ARF), a global health issue. The limited understanding of ARF's prevalence and primary clinical symptoms has led to confusion. This research investigates the characteristics of children aged 3-17 years who experience ARF with monoarthritis as their initial symptom., Methods: A retrospective evaluation of medical records of children diagnosed with ARF was conducted. The study aimed to determine the prevalence of monoarthritis as the first manifestation of ARF and its association with age, gender, family history, and cardiac involvement. Categorical variables were analyzed using the chi-square test with a significance level of < 0.05 and a confidence interval of 95%, using SPSS software (Version 23)., Results: The study included 62 patients with ARF, comprising 41 (66.1%) boys with an average age of 8.48±3.27 years. Among these patients, 12 exhibited cardiac involvement according to the revised Jones criteria, with 5 clinical carditis and 7 cases of subclinical carditis. Monoarthritis was the initial symptom in seven patients (11.29%); five (71.4%) also had carditis. There was a significant association ( p<0.001 ) between monoarthritis and carditis., Conclusion: The study concludes that monoarthritis may be an early sign of ARF in children and correlates significantly with cardiac involvement. However, more extensive research with more significant participant numbers is necessary to understand ARF in Iran comprehensively. Background: Renal involvement is the most damaging long-term complication of Immunoglobulin-A (IgA) vasculitis. In the lack of a definite predictive biomarker for renal involvement, antiphospholipid antibodies (aPL) have been proposed in recent years., Methods: In this prospective cohort of 48 pediatric patients who were admitted with IgA vasculitis from September 2015 to June 2017, two serum samples were taken 12 weeks apart to detect Anti-Phospholipid antibodies. All patients were followed-up for renal involvement for six months., Results: Renal involvement occurred in 14 out of 48 patients with IgA vasculitis (29.16%). APLs were positive in nine out of 14 patients with IgA vasculitis and renal involvement (64.28%), in contrast to only six out of 34 patients with IgA vasculitis without renal involvement (17.64%). The presence of aPL antibodies was statistically associated with renal involvement (P=0.002). Although, the relationship between both sex (P=0.025) and age (P=0.046) with aPL positivity was statistically significant, performing a modified logistic regression test, the odds ratio was significant between the groups with and without renal involvement only in term of age and aPL positivity)., Conclusion: The presence of aPL antibodies was statistically associated with renal involvement. We found a significant relationship between the age and aPL positivity. Hence, we need multicenter, more extensive cohort studies to reach a better and more accurate conclusion on the relationship between serum aPLs and renal involvement in IgA vasculitis patients., Competing Interests: The authors report no conflicts of interest regarding this manuscript. Background: Consecutive droughts and quantitative and qualitative reduction of surface and underground water resources have caused an increase in greenhouse and hydroponic cultivation for most garden crops, including strawberries, in Iran. On the other hand, most of the inputs of greenhouse crops in Iran are imported. To possibility of replacing vermicompost with peat moss under hydroponic cultivation, an experiment was done in a split plot based on randomized complete blocks design in three replications in Isfahan (Iran) Agricultural and Natural Resources Research Center in 2019. The main treatment was substrate at four levels included different levels of vermicompost (30 and 50%) and peat moss (30 and 50%) in combination with perlite and sub-treatment were Selva and Camarosa cultivars., Results: The results showed that Camarosa cultivar and Selva cultivar in (perlite/ peat moss 50:50) and Selva cultivar in (perlite / vermicompost 70:30) had maximum yield. Leaf number and chlorophyll index were maximum in Camarosa cultivar in peat moss substrates. Strawberry cultivars had the highest root fresh weight, the content of vitamin C and total soluble solids (TSS) in substrates containing vermicompost. Camarosa cultivar in (perlite / peat moss50:50) and Selva cultivar in (perlite /vermicompost 50:50) had maximum root dry weight. Also, the highest number of inflorescences was related to substrates containing peat moss and (perlite /vermicompost 70:30). Maximum amount of fresh and dry weight of shoots were observed in (perlite/ peat moss70:30). Selva cultivar had more inflorescences (16.5%) than Camarosa cultivar and Camarosa cultivar produced more fresh and dry weight of shoots (16.5%, 23.01%) than Selva cultivar., Conclusion: Expriment results highlighted the importance of considering both main and sub-treatments in agricultural research, as they interacted to influence various growth and yield parameters. 50% vermicompost treatment combined with perlite had a positive impact on plant growth and in quality index such as vitamin C content and TSS was highest. while the choice of cultivar affected different aspects of plant development. Selva cultivar was known to be more tolerant to salinity caused by vermicompost. Background: Applying modern educational methods for digital native students seems necessary. Active learning strategies promote students' skills and knowledge. This study was conducted to design and evaluate active learning methods by teaching psychopharmacotherapy to pharmacy students., Methods: This was a quasi-experimental study with three randomized study groups (control, game, and multimedia), using a pre-and post-test design, conducted on 155 students of 5-year pharmacy in 2022 at the Faculty of Pharmacy of Tehran University of Medical Sciences, Iran. Overall, 18 clinical cases were designed for the basic structure of interventions. After teaching psychopharmacotherapy contents through lecturing, the pre-test was held. The next steps were playing the educational game, studying the multimedia case-based learning files, and then completing questionnaires, respectively. Then, a post-test was held., Results: 65.33% of participants were female and 34.66% were male. The pre-test and post-test scores comparison showed no difference in control group ( P =0.409). However, in the serious game and multimedia groups, the average score of pre-test and post-test had a statistically significant difference ( P <0.001, P =0.002 respectively), this difference was higher in the serious game group. Questionnaire evaluation showed substantial differences between game and multimedia groups., Conclusion: The educational interventions were able to improve student's knowledge and skills so they can better help patients and promote public health. Holoprosencephaly, a complicated brain abnormality arising from incomplete prosencephalon cleavage, affects both the forebrain and the face. Holoprosencephaly Type 11, with variable expression or partial penetrance, is caused by CDON pathogenic variants associated with the disrupted Sonic Hedgehog ( SHH )-pathway. Herein, we aimed to describe a family with genetic nose problems. After counselling and drawing pedigree in Farhud's Genetic Clinic, Tehran, Iran in 2021, DNA extraction of a proband and a few members of his family (patient and control) was conducted. Whole exome sequencing was utilized for detecting the gene and its variant in the proband with a nose deformity. The results were confirmed with Sanger sequencing. This variant was checked in other members by Sanger sequencing. Analysis of the Exome data showed a heterozygous splicing variant in the CDON gene (NM&#95;016952; c.3276+1G>T) in the proband who had a nose deformity and then the results were confirmed with Sanger sequencing. Such a variant was observed in Proband's brother with a nose deformity and was not observed in Proband's cousin with no abnormal phenotype. Ionizable lipid nanoparticles (LNPs) pivotal to the success of COVID-19 mRNA (messenger RNA) vaccines hold substantial promise for expanding the landscape of mRNA-based therapies. Nevertheless, the risk of mRNA delivery to off-target tissues highlights the necessity for LNPs with enhanced tissue selectivity. The intricate nature of biological systems and inadequate knowledge of lipid structure-activity relationships emphasize the significance of high-throughput methods to produce chemically diverse lipid libraries for mRNA delivery screening. Here, we introduce a streamlined approach for the rapid design and synthesis of combinatorial libraries of biodegradable ionizable lipids. This led to the identification of iso-A11B5C1, an ionizable lipid uniquely apt for muscle-specific mRNA delivery. It manifested high transfection efficiencies in muscle tissues, while significantly diminishing off-targeting in organs like the liver and spleen. Moreover, iso-A11B5C1 also exhibited reduced mRNA transfection potency in lymph nodes and antigen-presenting cells, prompting investigation into the influence of direct immune cell transfection via LNPs on mRNA vaccine effectiveness. In comparison with SM-102, while iso-A11B5C1's limited immune transfection attenuated its ability to elicit humoral immunity, it remained highly effective in triggering cellular immune responses after intramuscular administration, which is further corroborated by its strong therapeutic performance as cancer vaccine in a melanoma model. Collectively, our study not only enriches the high-throughput toolkit for generating tissue-specific ionizable lipids but also encourages a reassessment of prevailing paradigms in mRNA vaccine design. Environmental destruction, water crisis, and clean energy are among the very important challenges worldwide based on sustainable development goals. Photocatalytic fuel cell, a potential candidate for converting chemical energy into electrical energy through a pollution-free method, holds promise in addressing these challenges. In this regard, we investigated the response of a photoanode covered with UiO66-NH 2 -TiO 2 /NiF on a porous nickel foam as an attractive electrochemical response to remove antibiotics from aqueous solution and simultaneously produce electricity using a one-step hydrothermal synthesis. Nickel foam with its fine structure provides a suitable space for the interaction of light, catalyst, and efficient mass transfer of reactive molecules. It appears that it can be used as a competitive electrode in fuel cells. In order to investigate the properties of the photocatalyst, structural analyses including XRD, FESEM, FTIR, and UV-vis DRS were utilized. Additionally, polarization and electrochemical tests such as chronoamperometry and EIS were measured to further examine the electrochemical features of the PFC photoanode system. The obtained results under optimal conditions (SMZ concentration = 20 ppm, pH = 6, irradiation time = 120 min) were as follows: removal efficiency of 91.7%, P max  = 16.98 μW/cm 2 , J sc  = 96.75 μA/cm 2 , V oc  = 644 mV. The light-induced current flow in UiO66-NH 2 -TiO 2 /NiF exhibited prominent and reproducible photocurrent responses, indicating efficient and stable charge separation in TiO 2 /NiF composite materials, which is a promising strategy for pollutant removal and simultaneous electricity generation., (© 2023. In this study, a number of leaching solutions (H 2 SO 4 , CuSO 4 and NaCl) and an electrochemical method were used together for the separation of Cu from waste printed circuit boards. Secondly, the magnetic-MOF(Cu) was synthesized using the Cu recovered from waste printed circuit boards. Thereafter, TiO 2 /mag-MOF(Cu) composite was prepared and its photocatalytic activity was assessed in the photo degradation of two prominent organophosphorus pesticides, namely malathion (MTN) and diazinon (DZN). The catalytic structure of the MOF-based composite was fully characterized by various analyses such as XRD, SEM, EDAX, FT-IR, VSM and UV-vis. The obtained analyses confirmed the successful synthesis of TiO 2 /mag-MOF(Cu) composite. The synthesized composite exhibited highly efficient in the degradation of both pollutants under the following conditions: pH 7, contaminant concentration 1 mg/L, the catalyst dosage of 0.4 g/L, visible light intensity 75 mW/cm 2 and reaction time of 45 min. First order kinetic model was best suited with the experimental results (R 2 : 0.97-0.99 for different MTN and DZN concentrations). Trapping studies revealed that superoxide radicals (O 2 •- ) played an important role during the degradation process. Furthermore, the catalyst demonstrated a superb recovery as well as high stability over five cyclic runs of reuse. In addition, the total organic carbon (TOC) analysis showed over 83% and 85% mineralization for MTN and DZN, respectively. The combined system of TiO 2 /mag-MOF(Cu)/Vis also exhibited a great level of efficiency and feasibility in the treatment of tap water and treated wastewater samples. Human cerebral organoids (COs) are self-organizing three-dimensional (3D) neural structures that provide a human-specific platform to study the cellular and molecular processes that underlie different neurological events. The first step of CO generation from human pluripotent stem cells (hPSCs) is neural induction, which is an in vitro simulation of neural ectoderm development. Several signaling pathways cooperate during neural ectoderm development and in vitro differentiation of hPSCs toward neural cell lineages is also affected by them. In this study, we considered some of the known sources of these variable signaling cues arising from cell culture media components and sought to modulate their effects by applying a comprehensive combination of small molecules and growth factors for CO generation. Histological analysis demonstrated that these COs recapitulate the neural progenitor zone and early cortical layer organization, containing different types of neuronal and glial cells which was in accordance with single-nucleus transcriptome profiling results. Moreover, patch clamp and intracellular Ca 2+ dynamic studies demonstrated that the COs behave as a functional neural network. Thus, this method serves as a facile protocol for generating hPSC-derived COs that faithfully mimic the features of their in vivo counterparts in the developing human brain. Introduction: Hepatic stellate cells (HSC) become activated, differentiate to myofibroblasts and produce extracellular fibrillar matrix during liver fibrosis. The hepatic fibrogenic response is orchestrated by reciprocal interactions between HSCs and macrophages and their secreted products. SOCS1 can regulate several cytokines and growth factors implicated in liver fibrosis. Here we investigated the role of SOCS1 in regulating HSC activation., Methods: Mice lacking SOCS1 in HSCs ( Socs1 ΔHSC ) were generated by crossing Socs1 fl/fl and LratCre mice. Liver fibrosis was induced by carbon tetrachloride and evaluated by Sirius red staining, hydroxyproline content and immunostaining of myofibroblasts. Gene expression of pro-fibrogenic factors, cytokines, growth factors and chemokines were quantified by RT-qPCR. The phenotype and the numbers of intrahepatic leukocyte subsets were studied by flow cytometry. The impact of fibrosis on the development of diethyl nitrosamine-induced hepatocellular carcinoma was evaluated., Results: Socs1 ΔHSC mice developed more severe liver fibrosis than control Socs1fl/fl mice that was characterized by increased collagen deposition and myofibroblast differentiation. Socs1 ΔHSC mice showed a significant increase in the expression of smooth muscle actin, collagens, matrix metalloproteases, cytokines, growth factors and chemokines in the liver following fibrosis induction. The fibrotic livers of Socs1 ΔHSC mice displayed heightened inflammatory cell infiltration with increased proportion and numbers of Ly6ChiCCR2+ pro-inflammatory macrophages. This macrophage population contained elevated numbers of CCR2+CX3CR1+ cells, suggesting impaired transition towards restorative macrophages. Aim: Parkinson's Disease (PD) is a common age-related neurodegenerative disorder with a rising prevalence. Human pluripotent stem cells have emerged as the most promising source of cells for midbrain dopaminergic (mDA) neuron replacement in PD. This study aimed to generate transplantable mDA progenitors for treatment of PD., Materials and Methods: Here, we optimized and fine-tuned a differentiation protocol using a combination of small molecules and growth factors to induce mDA progenitors to comply with good manufacturing practice (GMP) guidelines based on our clinical-grade human embryonic stem cell (hESC) line., Key Findings: The resulting mDA progenitors demonstrated robust differentiation and functional properties in vitro. Purpose: Necrotizing enterocolitis (NEC), an inflammatory intestinal disease common in premature infants, has been associated with the development of lung damage. Toll-like receptor 4 has been shown to regulate inflammation in the NEC lungs, however, other important inflammatory mechanisms have not been thoroughly investigated. In addition, we reported that milk-derived exosomes were able to attenuate intestinal injury and inflammation in experimental NEC. This study aims to (i) investigate the role of the NLRP3 inflammasome and NF-κB pathway in regulating lung damage during experimental NEC; and (ii) evaluate the therapeutic potential of bovine milk exosomes in reducing lung inflammation and injury during NEC., Methods: NEC was induced by gavage feeding of hyperosmolar formula, hypoxia, and lipopolysaccharide administration in neonatal mice from postnatal days 5-9. Exosomes were obtained by ultracentrifugation of bovine milk and administered during each formula feed., Results: The lung of NEC pups showed increased inflammation, tissue damage, NLRP3 inflammasome expression, and NF-κB pathway activation, which were attenuated upon exosome administration., Conclusion: Our findings suggest that the lung undergoes significant inflammation and injury following experimental NEC which are attenuated by bovine milk-derived exosomes. Although nanoparticle-based chemotherapeutic strategies have gained in popularity, the efficacy of such therapies is still limited in part due to the different nanoparticle sizes needed to best accommodate different parts of the drug delivery pathway. Herein, we describe a nanogel-based nanoassembly based on the entrapment of ultrasmall starch nanoparticles (size 10-40 nm) within disulfide-crosslinked chondroitin sulfate-based nanogels (size 150-250 nm) to address this challenge. Upon exposure of the nanoassembly to the reductive tumor microenvironment, the chondroitin sulfate-based nanogel can degrade to release the doxorubicin-loaded starch nanoparticles in the tumor to facilitate improved intratumoral penetration. CT26 colon carcinoma spheroids could be efficiently penetrated by the nanoassembly (resulting in 1 order of magnitude higher DOX-derived fluorescence inside the spheroid relative to free DOX), while in vivo experiments showed that doxorubicin-loaded nanoassemblies reduced tumor sizes by 6× relative to saline controls and 2× relative to free DOX after 21 days. Together, these data suggest that nanogel-based nanoassemblies are a viable option for improving the efficacy and safety of nanoparticle-based drug delivery vehicles treating cancer.

SOCS1 deficiency, which increases susceptibility to hepatocellular carcinoma (HCC), promotes CDKN1A expression in the liver. High CDKN1A expression correlates with disease severity in many cancers. Here, we demonstrate a crucial pathogenic role of CDKN1A in diethyl nitrosamine (DEN)-induced HCC in SOCS1-deficient mice. Mechanistic studies on DEN-induced genotoxic response revealed that SOCS1-deficient hepatocytes upregulate SOCS3 expression, SOCS3 promotes p53 activation, and Cdkn1a induction that were abolished by deleting either Socs3 or Tp53 . Previous reports implicate CDKN1A in promoting oxidative stress response mediated by NRF2, which is required for DEN-induced hepatocarcinogenesis. We show increased induction of NRF2 and its target genes in SOCS1-deficient livers following DEN treatment that was abrogated by the deletion of either Cdkn1a or Socs3 . Loss of SOCS3 in SOCS1-deficient mice reduced the growth of DEN-induced HCC without affecting tumor incidence. In the TCGA-LIHC dataset, the SOCS1 -low/ SOCS3 -high subgroup displayed increased CDKN1A expression, enrichment of NRF2 transcriptional signature, faster disease progression, and poor prognosis. Overall, our findings show that SOCS1 deficiency in hepatocytes promotes compensatory SOCS3 expression, p53 activation, CDKN1A induction, and NRF2 activation, which can facilitate cellular adaptation to oxidative stress and promote neoplastic growth. Thus, the NRF2 pathway represents a potential therapeutic target in SOCS1 -low/ SOCS3 -high HCC cases.

Hypothesis: The cluster formation and self-assembly of floating colloids at a fluid/fluid interface is a delicate force balance involving deterministic lateral interaction forces, viscous resistance to relative colloid motion along the surface and thermal (Brownian) fluctuations. As the colloid dimensions get smaller, thermal forces and associated drag forces become important and can affect the self assembly into ordered patterns and crystal structures that are the starting point for various materials applications., Numerics: Langevin dynamic simulations for particle pairs straddling a liquid-liquid interface with a high viscosity contrast are presented to describe the lateral interfacial assembly of particles in Brownian and non-Brownian dominated regimes. These simulations incorporate capillary attraction, electrostatic repulsion, thermal fluctuations and hydrodynamic interactions (HI) between particles (including the effect of the particle immersion depth). In this paper, we investigate the generation and controlling of the optical vortex beam using a dye-doped liquid crystal (DDLC) cell. The spatial distribution of the quasi-sinusoidal orientation of the liquid crystal molecules creates a quasi-sinusoidal phase grating (PG) in the DDLC cell. Depending on the incident light pattern, Trans to Cis photoisomerization of the dye molecules affects the orientation of the liquid crystal molecules. To do so, an amplitude fork grating (FG) is used as a mask, and its pattern is stored in the cell by a pattern printing method as the PG. One of the particular features of the stored grating in the cell is its capability in the diffraction efficiency controlled by the applied electric field. The results show, based on the central defect in the FG pattern, the diffracted probe beam in different orders is optical vortices. As a new technique, this type of stored pattern acts like an amplitude grating but according to the results, its structure is in fact a PG. This technique leads to the vortex beam switching capability by applying an electric field to the cell. The results show that by applying 22 V, all the diffraction orders vanish. Meanwhile, the vortex beams reappear by removing the applied voltage. The diffraction efficiency of the vortex beams as well as its generation dependency on the polarization of the incident beam studied. The maximum efficiency of the first diffraction order for linear polarized incident beam was obtained at 0 V, about 8%. Based on the presented theory, a simulation has been done which shows the Cis form of the dye molecules has been able to change the angle of LC molecules on average about 12.7°. Herein, a facile dispersive micro-solid phase extraction (d-µSPE) procedure using carboxylated multi-walled carbon nanotubes modified with silver nanoparticles (Ag/MWCNTs-COOH) was successfully developed for the adsorption and subsequent determination of low levels of two well-known contaminants, namely bisphenol A and S (BPA and BPS) in water and soft drink samples. The detection and measurement of the above-mentioned compounds were performed by HPLC-UV instrument. The applied d-µSPE procedure has several advantages such as rapidity, high degree of sensitivity, precision and efficiency. A combination of polar/non-polar interactions seems to play a key role in the adsorption process. Under the optimized conditions, the calibration curves were linear over the concentration range of 1-500 µg/L for the both targets. The practical limit of quantifications (LOQ) for the both analytes were determined to be 1.0 µg/L. Background: Medullary Thyroid Cancer (MTC) is a very aggressive type of thyroid carcinoma. Mutation in RET proto-oncogene is demonstrated in MTC development. We aimed to knock-out of RET -oncogene using CRISPR/Cas9 genome editing method in MTC cell-lines., Methods: This research was conducted in Shahid Beheshti University of Medical Sciences, Tehran, Iran during 2019-2020. Four different sgRNAs were designed to target exons one, two, and four of RET -oncogene in TT and MZ-CRC-1 cell-lines using bioinformatics tools, then the CRISPR/Cas9 constructs was made. About 72-hours after cell transfection, T7EI method and DNA sequencing were used to confirm the knock-out of RET- oncogene. Expression of RET , Calcitonin genes and RET protein were evaluated by Real-time PCR and ELISA, respectively., Results: The results of T7E1, and DNA sequencing of transfected cells confirmed RET gene knock-out by CRISPR/Cas9. There was a significant decrease in RET gene expression and RET protein in transfected TT and MZ cells compared to controls. The rate of cell apoptosis in transfected cells was significantly increased. Calcitonin gene expression was also significantly reduced in transfected cells. p-RET, p-PI3K, p-AKT, p-MEK, p-ERK protein levels were significantly reduced in TT and MZ transfected cells., Conclusion: For the first time, knock-out of RET gene was performed and confirmed using CRISPR/Cas9. Inhibition of this gene leads to inhibition of the tyrosine kinase RET signal transduction pathway. Objective: Using anticoagulants and antiplatelet drugs in patients with cardiovascular and medical comorbidities is prevalent. Because of hyper vascular nature of kidney, physicians tend to stop using aspirin before percutaneous nephrolithotomy (PCNL). We have shown the effects of remaining on low dose aspirin in complete supine PCNL (csPCNL)., Material and Methods: The records of 643 patients who underwent csPCNL between 2012 and 2018 were analyzed. Surgical outcomes and complications of patients who were on aspirin therapy and continued it daily (group A) were compared with those not taking aspirin (group B)., Results: Of the 643 csPCNLs, 40 (6%) were performed in patients of group A and the rest of 603 (94%) cases were in group B. The differences between the mean age of groups were statistically significant (60.08±9.45, group A and 48.66±12.32, group B) (P<0.001). Thirty-nine (97.5%) of patients in group A and 548 (90.9%) of group B were stone free by the end of the study which was not statistically significant (P=0.118). The mean operative time between groups A and B (43.20±21.37 and 44.83±16.83, respectively) was not considered significant (P=0.561). There was also no significant difference between 2 groups in any types of complications. Multivariate analysis showed that, perioperative aspirin use was not a significant predictor of transfusion, Hb drop, operative time and other complications., Conclusions: Remaining on aspirin does not increase the risk of bleeding, transfusionand other complications. Herein, the efficient degradation of a highly consumed antibiotic known as metronidazole (MNZ) in aqueous solutions using Co/g-C 3 N 4 /Fe 3 O 4 nanocomposite under visible light irradiation was accomplished. Initially, the photocatalyst (Co/g-C 3 N 4 /Fe 3 O 4 ) was synthesized by a simple hydrothermal method and then characterized by several analytical techniques, namely EDS, SEM, XRD, UV-vis DRS, and FTIR. The efficiency of the synthesized photocatalyst with regard to the degradation of the studied antibiotic (MNZ) under visible light irradiation was fully evaluated. The influential operational parameters affecting the efficiency of the degradation process such as pH (2-10), nanocomposite dosage (0.2-1 g/L), MNZ concentration (5-20 mg/L), and irradiation time (0-80 min) were optimized. The results revealed that the maximum degradation efficiency for MNZ was obtained under the following conditions: irradiation time of 60 min, pH = 8, MNZ concentration of 5 mg/L, and photocatalyst dosage of 0.7 g/L. In addition, the degradation of MNZ followed the pseudo-first-order kinetic model. The best rate constant (k) value was determined to be 0.0102 min -1 with the correlation coefficient (R 2 ) of 0.992. According to the results of the quenching tests, it was found out that hydroxyl radicals (OH°) were the main species responsible for the MNZ degradation. Furthermore, the applied photocatalyst (Co/g-C 3 N 4 /Fe 3 O 4 ) exhibited a high level of recovery and stability after five cycles of reuse. Co/g-C 3 N 4 /Fe 3 O 4 /Vis system exhibited an excellent performance in the treatment of wastewater and real water samples. Background: Calcium is a necessary mineral for life to keep the body and bones healthy. Various factors including hormones, diet, age, and gender affect serum calcium status. The aim of this sturdy was to assess the serum calcium level (SCL) of Tehran population, which has about 10 million multi-Ethnic populations and represents from the whole country., Methods: In this retrospective study, the measured SCL of 105,128 individuals referred to different laboratories of Tehran, Iran were evaluated and its relationship with the age, gender, seasons, and different years during 2009-2018, were analyzed., Results: After excluding outliers, 91,257samples remained, which 61162 (58.64%) and 30,095 (41.36%) were female and male, respectively. The mean SCL was 9.36 (9.35, 9.37) mg/dl (95%CI). The highest and lowest SCLs were 3.1 and 18.2mg/dl, respectively. From the total study population, 74127 (81.23%) had normal SCLs, 14110 (15.46%) had hypocalcemia, and 3020 (3.31%) had hypercalcemia. SCLs were normal in 83.6% of men and 79.66% of women. Women had a significantly higher frequency of hypocalcemia compared to men (17.2% vs. 12.83%, p<0.0001)., Conclusion: Normal and abnormal SCLs were significantly different in age groups and in both genders. It means that gender and age affect SCLs. Every year of increasing age, reduces the chance of hypercalcemia by 40%, significantly. Age seems to affect hypercalcemia more than hypocalcemia. Age in men increases the risk of hypocalcemia, and reduces the risk of hypocalcemia in women. Background: Epigenetic alterations such as DNA methylation are known as the main cause of different types of cancers through inactivation of tumor suppressor genes, especially thyroid cancer. Identification of novel and effective markers are important in diagnosis and prevention of thyroid cancer. In the present study, the expression and methylation of Solute carrier family 5 member 8 ( SLC5A8 ) in Papillary Thyroid Carcinoma (PTC) in comparison to multinodular goiter (MNG) have been studied., Methods: Overall, 41 patients with PTC and 36 patients affected by MNG were recruited from four hospitals in Tehran and Qazvin, Iran in 2018. Thyroid tissues were obtained during thyroidectomy. RNA and DNA were extracted from thyroid tissues. Quantitative RT-PCR assay was performed for determining the mRNA level of SLC5A8 while Methylation-Sensitive High-Resolution Methylation was applied for assessing the methylation status., Results: Methylation status of three regions composed of 52 CpG islands in the promoter of SLC5A8 gene was studied by HRM assay. SLC5A8 level in PTC tissues was significantly downregulated in average 0.4 fold in comparison with MNG tissues ( P =0.05). The aberrant methylation of SLC5A8 (b) region was remarkably different in PTC and MNG cases. The promoter methylation of SLC5A8 (c) was significantly related to BRAF mutations and vascular invasion in PTC patients., Conclusion: The aberrant promoter hyper methylation of SLC5A8 was related to aggressive PTC. There are conflicting data regarding the association between plasma concentration of voriconazole (VCZ) and both efficacy and safety. This study investigates the association of VCZ trough plasma level with clinical efficacy and hepatotoxicity in the Iranian population suffering hematological malignancies. This cross-sectional study was performed on adult Iranian patients (age ≥ 18 years) with hematological malignancies undergoing treatment with oral or intravenous VCZ for proven or probable invasive aspergillosis. Plasma concentrations of VCZ were measured at two time points on day 4 and 14 during the study period. A total of 60 VCZ trough concentrations of 30 patients were drawn on days 4 and 14 after the initiation of treatment. There was no definite correlation between the mean plasma concentration of VCZ and VCZ dosage ( p = 0.134, r = 0.280). In multivariable model, only plasma concentration of VCZ on day 14 was associated with the incidence of hepatotoxicity ( p = 0.013; OR = 1.42, 95% CI = 1.07-3.24). Plasma trough concentration neither on day 4 nor on day 14 was related to the treatment response. No significant association was observed between the mean plasma concentration of VCZ and 3-month patients' survival ( p = 0.696). To conclude, VCZ trough concentration may not be a predictor of treatment response or 3-month patients' survival. However, the wide inter- and intra-patient variability of VCZ plasma concentration coupled with the observed association between VCZ trough level and the incidence of hepatotoxicity would pose the question regarding the potential benefit of VCZ concentration monitoring.

Background: The tumor suppressor genes play a critical role in cellular and molecular mechanisms such as cell cycle processes, cell differentiation and apoptosis. Aberrant DNA methylation of tumor suppressor genes and subsequent gene expression changes have shown to be involved in the initiation and progression of various malignancies including thyroid malignancies. In this review, we investigated what is known about the impact of promoter hypermethylation on the key tumor suppressor genes known to be involved in cell growth and/or apoptosis of thyroid cancer., Methods: The most important databases were searched for research articles until June 2020 to identify reported tumor suppressor genes that are modulated by methylation modulation changes in thyroid carcinoma. Following the inclusion and exclusion criteria, 26 studies were reviewed using the full text to meet the inclusion and exclusion criteria., Results: The tumor suppressor genes reviewed here are suggestive biomarkers and potential targetable drugs. Inactivation of RASSF1A , DAPK1 , SLCFA8 , and TSHR through aberrant epigenetic methylation could activate BRAF/MEK/ERK kinase pathways with potential clinical implications in thyroid cancer patients. RARβ2 and RUNX3 could suppress cell cycle and induce apoptosis in malignant cells. TIMP3 and PTEN could prevent angiogenesis and invasion through PIP3 pathway and arrest VEFG activity., Conclusion: The methylation status of key genes in various types of thyroid malignancies could be used in early diagnosis as well as differentiation of malignant and benign thyroid. 2,4-Dinitrophenol (DNP) was listed as a priority pollutant; accordingly, DNP-contaminated effluent must be treated before discharging to the receiving resources. In the present study, the hybrid ultrasound-assisted GO-Fe 3 O 4 system was employed to decontaminate DNP solution. Ultrasound irradiation makes the mass transfer of adsorbate improved and Fe 3 O 4 enables GO separation from liquid phase under external magnetic field. The as-synthesized GO-Fe 3 O 4 composite was characterized by SEM, TEM, XRD, FTIR, BET and VSM. A response surface methodology based central composite design (RSM-CCD) was used to estimate and optimize of various variables on DNP removal percentage. Under optimal conditions (pH: 4.45, adsorbent dose: 0.178 g/L, ultrasound frequency: 40.02 kHz and DNP concentration: 50.10 mg/L, maximum adsorption capacity was calculated to be 425.58 mg/g for the ultrasound system, higher than the simple system 309.40 mg/g, indicating the importance of synergistic effect between the ultrasound waves and the adsorption process. The ultrasound-assisted adsorption system showed the better agreement with the Langmuir isotherm (R 2 > 0.997), while the results of the stirring system were more consistent with the Freundlich model (R 2 > 0.991). The experimental results indicated that the pseudo-second-order kinetic model well fitted by experiment data and rate constant was calculated to be 0.000148 min -1 and 0.000002 min -1 under ultrasound and silent systems, respectively. The rate of desorption under ultrasound was more favorable and reuse of the adsorbent in both systems after 10th consecutive cycles reduced by about 22%. Thermodynamic calculations also confirmed the endothermicity and spontaneity of both systems. Electrostatic attraction, hydrogen bonding, and π -π interactions played key roles during the adsorption of DNP onto the MGO. In conclusion, the outcomes of this study provide valuable information of the ultrasound-assisted GO-Fe 3 O 4 system for practical applications., (Copyright © 2021 Elsevier B.V. This study aimed to address the hypothesis that the expression of PTEN and KLLN tumor suppressor genes could diminish in papillary thyroid cancer (PTC) compared to paired normal tissue (PNT) and multinodular goiter (MNG). PTEN and KLLN expressions were assessed at both mRNA and protein levels in 82 tissue samples, including 30 PTC, 30 PNT, and 26 MNG using SYBR-Green Real-Time PCR and enzyme-linked immunosorbent assay (ELISA), respectively. Bioinformatics studies were performed to evaluate the genomic location and the genes promoter region. The mRNA expression of PTEN and KLLN in PTC was significantly lower than PNT (PTEN, P = 0.0033; KLLN, P = 0.0005). A significant decrease in the mRNA level of KLLN was also observed in PTC than MNG (P = 0.0304). Decreased level of PTEN mRNA (odds ratio=0.391; P = 0.013) or KLLN mRNA (odds ratio=0.023; P = 0.025) was associated with an increased risk of PTC tumorigenesis. Areas under the ROC curve for PTEN and KLLN were 0.69 and 0.78, respectively. PTEN and KLLN protein expressions in PTC compared to PNT or MNG were not significantly different. The bioinformatics studies revealed the sequence near the promoter region is lowly conserved across species. Four GC boxes were found upstream of the PTEN transcription start site (TSS), and one TATA box and one GC box were found upstream of KLLN TSS. The results suggest PTEN and KLLN are the two tumor suppressor genes that decreasing or loss of both of them occurs in sporadic PTC tumorigenesis. Background: Although, preliminary reports of Severe Acute Respiratory Syndrome (SARS)-CoV-2 infection suggest that the infection causes a less severe illness in children, there is now growing evidence of other rare or even serious complications of disease., Case Presentation: During the recent COVID-19 pandemic in Kerman, Iran, two children (an 8 year-old boy and a 6 year-old girl) were referred to outpatient Clinic of Pediatric Rheumatology with complaints of limping. Both children had experienced fever and mild respiratory tract infection. At the beginning of the second week of infection, they developed joint effusion. They both tested positive for coronavirus infection and were therefore diagnosed with post Coronavirus reactive arthritis. Both children were treated successfully with rest and Non-Steroidal Anti-Inflammatory Drugs (NSAID). They did not have any medical problems in the two months fallow up., Conclusions: These two cases suggest that COVID-19 may be rheumatogenic. Highlighting the need for awareness of physicians, especially pediatricians, regarding the pathogenesis margins of this virus, as late presentations are of great importance.

It is widely believed that an increasing trend in the production and consumption of vegetables has led to a dramatic rise in the use of pesticides potentially threatening the health of consumers around the world. This systematic study along with meta-analysis has mainly centered on the evaluation of the quantity of three well-known pesticides namely, Malathion (MLT), Diazinon (DZN) and Chlorpyrifos (CPF) in vegetables. In this regard, a comprehensive literature search has been performed over the last decade (January 1, 2011 to June 21, 2020) within the scientific databases including PubMed, Web of Science, and Scopus. Of 1239 articles identified through the database screening, 22 plus 37 data report were retained and included in the meta-analysis phase. Additionally, the probabilistic human health risks for the consumers due to the intake of CPF, DZN and MLT from eating vegetables were estimated by the Monte Carlo Simulated (MCS) method. According to the findings, the maximum quantities of MLT, DZN and CPF in the vegetables were observed in Pakistan (222 μg/kg, 95%CI = 214.94-229.08), Thailand (245.00, 95% CI = 235.2-254.8) and South Korea (440 μg/kg, 95% CI = 437.19-442.81), while the lowest concentration levels were reported in China (1.7 μg/kg, 95% CI = 1.56-1.84), Poland (0.57, 95% CI = 0.46-0.68) and Poland (5.78 μg/kg, 95% CI = 4.40-7.12), respectively. The results of the Egger's and the Begg's tests revealed that no bias with regard to the potential publication was observed. Background: Von Hippel Lindau (VHL) disease is a hereditary disorder characterized by the development of benign or malignant tumors in the brain, spinal cord, eyes, adrenal medulla, kidney, pancreas, and many other organs. Advances in molecular diagnosis have led to the identification of the affected members of families at earlier stages. We present the clinical, laboratory, and genetic characteristics of five generations of a large Iranian kindred with VHL., Methods: The proband, a 52-year-old Iranian man, was recognized with VHL. All family members underwent clinical, laboratory, imaging, and genetic evaluation. Medical files and histopathology reports of patients who had been operated on before were also reviewed. Diagnosis of the disease was based on clinical findings, positive family history of VHL, and development of a central nervous system or retinal hemangioblastoma or pheochromocytoma., Results: Based on diagnostic criteria, our initial evaluations revealed that 10 members of the family had already been affected by the disease. Among them, nine had pheochromocytoma, and one had retinal hemangioblastoma. Background: Due to the increasing prevalence of infertility, the number of referrals to infertility treatment centers has also increased. Nowadays, assisted reproductive technology (ART), including in vitro fertilization (IVF), is a treatment for infertility or genetic problems. Considering the possible consequences of this method among women undergoing in vitro fertilization (IVF) and kids conceived by IVF, extensive research has been conducted in this regard., Methods: Overall, 100 articles were entered into the study, and relevant articles were searched and extracted from PubMed, Springer, and Google Scholar databases. In IVF procedure, medications such as Clomiphene citrate and gonadotropins are used to stimulate and mature follicles and thus increase ovulation., Results: There are conflicting opinions on this issue. Some findings report a slight increase in cancer risk for hormone-sensitive cancers including breast cancer. The long-term use of IVF medications can increase estrogen hormones and cause excessive expression of genes, resulting in an increased risk of breast cancer, which is one of the most frequent cancers among women., Conclusion: There are some risks to be aware of, which followed the hypothesis that long IVF treatment process may lead to breast cancer among IVF candidates. Furthermore, the risk of breast cancer may be increased in those women with a positive family history and related inherited genes. Cigarette butt is known as hazardous waste with numerous toxic and carcinogenic pollutants which impose serious concern for both the environment and human. Heavy metals are recognized as the most common pollutant in the cigarette butts. The concentration of some heavy metals (cadmium, chromium, nickel, lead and zinc) in leachate obtained from the pilot landfill with commingled waste and freshly smoked cigarettes butts were analyzed. The results showed that the addition of 0.76% (in weight) freshly smoked cigarette butts in landfilled waste increased total heavy metal concentration by 4.8%, while addition of 1.3% (in weight) freshly smoked cigarette butts leads to increased 3.72% of total heavy metals concentrations. An increased 10.52% and 3.43% health risk values were found from the leachate of the landfill pilot, where 1% freshly smoked cigarette butt and a littered cigarette were added, respectively. Background: Medullary thyroid cancer (MTC) accounts for 5%-10% of all thyroid cancers, but causes 13% of all thyroid cancer related deaths. MicroRNAs (miRs) have key functions in the development and progression of MTC. Altered expression of some miRs has been reported in many human cancers, including Thyroid cancer. Therefore, we aimed to analyze the expression of miR-154, miR-183 and miR-127 in MTC tumor tissues., Methods: In this case-control study, 15 MTC Formalin-fixed, paraffin-embedded (FFPE) tissue samples and 15 adjacent normal thyroid FFPE tissues, as a control group, were collected from Taleghani, and Loghman Hakim Hospitals, Tehran, Iran since 2005 till 2015. After RNA extraction and cDNA synthesis, the expression of miR-127, miR-154 and miR-183 was measured by quantitative Real-Time Polymerase Chain Reaction (qRT-PCR)., Results: Our data showed a significant increase in the expression of miR-127 in MTC samples in comparison with the control group ( P <0.05). Background: Investigating the complexity of movement systems can be insightful in clinical decision making. The study conducted to evaluate whether complexity of ground reaction force is affected by hallux valgus grades and footwear or foot side conditions., Methods: A sample of 120 females including 30 participants in each group of healthy and mild, moderate, and severe hallux valgus were recruited in this case-control study. Hallux valgus grades were assessed using Manchester scale. Ground reaction force were measured in bare foot and shod conditions for both feet and weighted permutation entropy was used to calculate the complexity. Nonparametric tests were employed for statistical inference., Findings: The first important result was that hallux valgus caused loss of complexity of ground reaction force with a medium to large effect size. However, we did not find any difference between three grades of hallux valgus entropy. The second finding was that entropy of healthy group decreased from barefoot to shod condition with a large effect size and the third finding was that ground reaction force entropy were similar in both feet., Interpretation: The findings are aligned with the theory that loss of complexity appears when the biological systems become functionally impaired. As soon as hallux valgus initiates, all potential risks related to the reduction of complexity appear. So, we suggest to administer conservative treatments in the early stages of hallux valgus. Background: Little attention has been paid to foot structural factors and ground reaction force in hallux valgus. Investigating the structural factors in detail and determining their biomechanical effects can help to better manage hallux valgus., Methods: In this case-control study, 120 females, 90 hallux valgus and 30 healthy, were recruited. Hallux valgus grades (mild, moderate, severe), first metatarsal mobility, foot pronation, hallux rotation, and pain were assessed by clinical tests. Ground reaction forces were measured in barefoot and shod conditions for both feet and analyzed using mixed within-between MANOVA. The association between structural factors and pain with force was analyzed using Spearman correlation coefficient., Results: The frequency of foot structural factors and pain severity were reported in hallux valgus grades. A significant difference was seen in the force values between groups (P<0.001). Bonferroni post hoc test indicated that the mean of the first peak for the severe group was significantly higher than the mild group (P=0.013) and the mean of second peak for the moderate group was higher than that of the healthy group (P=0.009). The force values were affected by wearing shoe (P<0.001) but not by foot side (P=0.086). There was a medium, positive correlation between the hallux rotation and force in the moderate group (r=0.39, P=0.03) and also between the pronation and force in the severe group (r=0.36, P=0.04)., Conclusion: Foot structural factors, pain, and force were different in each hallux valgus grads. Similar force in both feet, and increased force by wearing shoe were seen. Over the last few years, the development of genome editing has revolutionized research on the human genome. Recent advances in developing programmable nucleases, such as meganucleases, ZFNs, TALENs and Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)/Cas, has greatly expedited the progress of gene editing from concept to clinical practice. The CRISPR has advantages over other nuclease-based genome editing tools due to its high accuracy, efficiency, and strong specificity. Eight years after CRISPR application for human genome edition by Emmanuelle Charpentier and Jennifer A.
