Hepatic fibrosis is an important pathological manifestation of chronic schistosome infection. Patients with advanced schistosomiasis show varying degrees of abnormalities in liver fibrosis indicators and bilirubin metabolism. However, the relationship between hepatic fibrosis in schistosomiasis and dysregulated bilirubin metabolism remains unclear. In this study, we observed a positive correlation between total bilirubin levels and the levels of ALT, AST, LN, and CIV in patients with advanced schistosomiasis. Additionally, we established mouse models at different time points following S. japonicum infection. As the infection time increased, liver fibrosis escalated, while liver UGT1A1 consistently exhibited a low expression, indicating impaired glucuronidation of bilirubin metabolism in mice. In vitro experiments suggested that SEA may be a key inhibitor of hepatic UGT1A1 expression after schistosome infection. Furthermore, a high concentration of bilirubin activated the NF-κB signaling pathway in L-O2 cells in vitro. These findings suggested that the dysregulated glucuronidation of bilirubin caused by S. japonicum infection may play a significant role in schistosomiasis liver fibrosis through the NF-κB signaling pathway.

Combustion and explosion accidents of the mixture may occur after the adsorption of volatile organic compounds (VOCs) by coal-based activated carbon (CBAC). It is of great significance to explore the oxidation and combustion performance of CBAC before and after adsorbing VOCs in order to prevent the reoccurrence of fire and explosion. Based on the CBAC sample commonly used in industrial production, three types of CBAC samples after adsorbing VOCs, i.e., acetone, cyclohexane, and butyl acetate, were prepared. The oxidation and combustion characteristics of the samples before and after adsorbing VOCs are measured and analyzed by thermal analyzer and cone calorimeter. Thermal analysis results indicate that during the oxidation process, the VOCs in the adsorbed samples will burn in the early stage, generating amounts of heat which may accelerate the oxidation and combustion of CBAC. According to the combustion performance experiments by cone calorimeter, it is also found that the combustion rate of CBAC after adsorbing VOCs is significantly enhanced. The time to ignition is shortened, the heat release rate becomes larger, and the time to reach the peak of heat release rate is significantly moved forward. In addition, the CO yield of the adsorbed sample is significantly improved. Schistosomiasis is an immunopathogenic disease characterized by egg granuloma and fibrosis. The hepatic fibrosis of schistosomiasis is caused by the coordinated action of local immune cells, liver-resident cells and related cytokines around the eggs of the liver. B-cell-activating factor (BAFF), expressed in many cells, is an essential factor for promoting the survival, differentiation, and maturation of cells. The overexpression of BAFF is closely related to many autoimmune diseases and fibrosis, but has not been reported to play a role in liver fibrosis caused by schistosomiasis. In the study, we found that, during Schistosoma japonicum ( S. japonicum ) infection in mice, the level of BAFF and its receptor BAFF-R progressively increased, then decreased with the extension of infection time, which was consistent with the progression of hepatic granuloma and fibrosis. Anti-BAFF treatment attenuated the histopathological damage in the liver of infected mice. The average areas of individual granulomas and liver fibrosis in anti-BAFF treatment mice were significantly lower than those in control mice, respectively. Anti-BAFF treatment increased the IL-10, decreased IL-4, IL-6, IL-17A, TGF-β, and downregulated the antibody level against S. japonicum antigens. These results suggested that BAFF acts a strong player in the immunopathology of schistosomiasis. Anti-BAFF treatment may influence Th2 and Th17 responses, and reduce the inflammatory reaction and fibrosis of schistosomiasis liver egg granuloma. It is suggested that BAFF might be a prospective target for the development of new methods to treat schistosomiasis liver fibrosis.

The paper elaborates the effects of ionic liquids (ILs) on the phase equilibrium temperature, induction time, gas consumption, gas consumption rate, and water to hydrate conversion in the presence of 0.25, 0.63, 0.95, 1.25, 3.75, 6.25, and 10.00 wt % ethyltributylphosphonium hexafluorophosphate ([P 2 4 4 4 ][PF 6 ]), tributylhexylphosphonium hexafluorophosphate ([P 6 4 4 4 ][PF 6 ]), tetraethylammonium bromide ([N 2 2 2 2 ]Br), tetraethylammonium bistrifluoromethanesulfonimide ([N 2 2 2 2 ][NTf 2 ]), and tetraethylammonium hexafluorophosphate ([N 2 2 2 2 ][PF 6 ]) under a pressure of 2 MPa. The results indicate that all five ILs could increase CO 2 consumption and enhance the water to hydrate conversion. Compared with the pure water system, [P 2 4 4 4 ][PF 6 ] and [P 6 4 4 4 ][PF 6 ] shifted the phase equilibrium temperature of CO 2 hydrates to a slightly higher temperature with reduced induction times by boosting CO 2 hydrate nucleation, showing the dual function promotion effects. In contrast, [N 2 2 2 2 ]Br, [N 2 2 2 2 ][NTf 2 ], and [N 2 2 2 2 ][PF 6 ] shifted the phase equilibrium temperature of CO 2 hydrates to a lower temperature and prolonged the induction time by slowing down CO 2 hydrate nucleation. The inhibition effects of anions on CO 2 hydrates follow an order of Br - > [NTf 2 ] - > [PF 6 ] - . Besides, the density functional theory and molecular dynamic calculations were conducted to explain the inconsistent influences of [N 2 2 2 2 ]Br and [N 4 4 4 4 ]Br on CO 2 hydrate formation. It was found that the anion-cation interaction of [N 2 2 2 2 ]Br was stronger than that of [N 4 4 4 4 ]Br, and Br - in [N 2 2 2 2 ]Br is less likely to participate in the formation of hydrate cages in the [N 2 2 2 2 ]Br + H 2 O + CO 2 system according to the intermolecular anion-water, anion-CO 2 , and water-water radial distribution function in [N 2 2 2 2 ]Br + H 2 O + CO 2 and [N 4 4 4 4 ]Br + H 2 O + CO 2 systems., Competing Interests: The authors declare no competing financial interest., (© 2022 The Authors. Most gas hydrates on the earth are in sediments and permafrost areas, and porous media are often used industrially as additives to improve gas hydrate formation. For further understanding its exploration and exploitation under natural conditions and its application in industry, it is necessary to study the effect of porous media on hydrate formation. The results show that the stacked porous media affects the phase equilibrium of hydrate formation depending on the competition water activity and large specific surface areas, while integrated porous media, such as metal foam, can transfer the hydration heat rapidly and moderate the hydrate phase equilibrium. A supersaturated metal-organic framework is able to significantly improve gas storage performance and can be used as a new material to promote hydrate formation. However, the critical particle size should be studied further for approaching the best promotion effect. In addition, together with the kinetic accelerators, porous media has a synergistic effect on gas hydrate formation. The carboxyl and hydroxyl groups on the surface of porous media can stabilize hydrate crystals through hydrogen bonding. Background: The activation of immune response driven by the eggs of Schistosoma japonicum and the subsequent secretions is the culprit behind granulomatous inflammation and liver fibrosis. Evidence suggests that PKCλ/ι participates in a variety of physiological and pathological processes, including the regulation of metabolism, growth, proliferation and differentiation of cells. However, the role of PKCλ/ι in liver disease caused by Schistosoma japonicum remains unclear., Methods: In the present study, we observe the pathological changes of egg-induced granulomatous inflammation and fibrosis in the liver of mice infected by Schistosoma japonicum by using conditional PKCλ/ι-knockout mice and wild-type control. Immune cytokines and fibrogenic factors were analyzed by performing flow cytometry and real-time fluorescence quantitative PCR., Results: The results of H&E and Masson staining show that the degree of granulomatous lesions and fibrosis in the liver of the infected PKCλ/ι-knockout mice was significantly reduced compared with those of the infected wild-type mice. The mean area of single granuloma and hepatic fibrosis in the PKCλ/ι-knockout mice was significantly lower than that of the wild-type mice (85,295.10 ± 5399.30 μm 2 vs. 1,433,702.04 ± 16,294.01 μm 2 , P < 0.001; 93,778.20 ± 8949.05 μm 2 vs. 163,103.01 ± 11,103.20 μm 2 , P < 0.001), respectively. Serological analysis showed that the ALT content was significantly reduced in the infected knockout mice compared with infected wild-type mice. RT-PCR analysis showed that IL-4 content in knockout mice was significantly increased after Schistosoma japonicum infection, yet the increase was less than that in infected wild-type mice (P < 0.05). PKCλ/ι deficiency led to reduced expression of fibrosis-related factors, including TGF-β1, Col-1, Col-3, α-SMA and liver DAMP factor HMGB1. Flow cytometry analysis showed that the increasing percentage of Th2 cells, which mainly secrete IL-4 cytokines in spleen cells, was significantly lower in PKCλ/ι-deficient mice compared with wild-type mice after infection (P < 0.05)., Conclusions: Our data demonstrate that PKCλ/ι deficiency alleviating granulomatous inflammation and fibrosis in the liver of mice with S. japonicum infection by downregulating Th2 immune response is the potential molecular mechanism behind the role of PKCλ/ι in schistosomiasis., (© 2022. Objective: This study aimed to analyze the changes in intestinal flora and metabolites in the intestinal contents of mice with inflammatory bowel disease (IBD) to preliminarily clarify the mechanism of action of Schistosoma soluble egg antigen (SEA) on IBD, thus, laying a research foundation for the subsequent treatment of IBD. Methods: A total of 40 Institute of Cancer Research (ICR) mice were divided into four groups: control, SEA 50 μg, dextran sulfate sodium salt (DSS), and SEA 50 μg + DSS. The overall state of the animals was observed continuously during modeling. The colonic length was measured after 10 days of modeling. The degree of colonic inflammation was observed by hematoxylin and eosin staining. 16srRNA and liquid chromatography-mass spectrometry sequencing techniques were used to determine the abundance of bacteria and metabolites in the intestinal contents of mice in the DSS and SEA 50 μg + DSS groups, and the differences were further analyzed. Results: After SEA intervention, the disease activity index score of mice with IBD decreased and the colon shortening was reduced. Microscopically, the lymphocyte aggregation, glandular atrophy, goblet cell disappearance, and colonic inflammation were less in the SEA 50 μg + DSS group than in the DSS group ( p < 0.0001). After SEA intervention, the abundance of beneficial bacteria prevotellaceae&#95;UCG-001 was upregulated, while the abundance of the harmful bacteria Helicobacter , Lachnoclostridium , and Enterococcus was downregulated in the intestinal tract of mice with IBD. The intestinal metabolite analysis showed that SEA intervention decreased the intestinal contents of glycerophospholipids (lysophosphatidylcholine, lysophosphatidylethanolamine, phatidylcholine, and phatidylethanolamine) and carboxylic acids (L-alloisoleucine and L-glutamate), whereas increased bile acids and their derivatives (3B,7A,12a-trihydroxy-5A-cholanoic acid and 3A,4B, 12a-trihydroxy-5b-cholanoic acid). Combined microbiota-metabolite analysis revealed a correlation between these differential microbiota and differential metabolites. At the same time, the changes in the contents of metabolites and differential metabolites in the two groups also correlated with the abundance of the gut microbiome. Conclusions: The study showed that SEA reduced DSS-induced inflammation in IBD and improved the symptoms of IBD in mice through the combined regulation of intestinal flora and intestinal metabolism. After a coal seam is mined, the coal remaining in the goaf is prone to flooding and spontaneous combustion accidents. To explore the reignition (secondary oxidation) characteristics of long-flame coal after oxidation and water immersion, the experimental methods of thermogravimetric analysis and infrared spectroscopy were used to analyze coal samples of oxidation first and then water immersion (FO) and samples of water immersion first and then oxidization (FI) at different pre-oxidation temperatures. The results showed that the content of main oxygen-containing functional groups (hydroxyl, carbonyl, and carboxyl groups) of the FO120 (oxidation 120 °C first and then water immersion) coal sample increased, and the FI 90 (water immersion first and then oxidization 90 °C) coal sample decreased. Pre-oxidation at 120 °C will slow down the decrease in the extent of low-temperature secondary oxidation TG, as the pre-oxidation temperature increases, the total heat release of the FO coal samples first increase and then decrease, and the heat released is high at 120 °C. The FI coal samples transfer active sites during the water immersion process, and the high pre-oxidation temperature leads to the rapid increase of the speed of the primary active site, which leads to the transformation between the secondary active site and the oxygen-containing group, resulting in the cleavage of the oxygen-containing group and increasing the heat production. Water immersion pre-oxidation performed under different conditions has the dual effects of promoting and inhibiting spontaneous coal combustion. To diminish the greenhouse effect by reducing CO 2 emission into the air based on a capture and sequestration method through hydrates, the thermodynamic and kinetic effects of additives on CO 2 hydrate formation under 1.5 MPa in the presence of 5, 6, 8, 10, and 20 wt % RNS-A (reactive SiO 2 containing amino groups) were studied, and the stirrer speed was set to 800 rpm. This paper calculated the gas consumption and explained the possible mechanisms of RNS-A on CO 2 hydrates. The results showed that RNS-A was a kinetic additive instead of a thermodynamic one. It was found that 5-10 wt % RNS-A all shortened the induction time of hydrates, but only 5 and 6 wt % RNS-A increased the gas consumption of CO 2 hydrates. Although we observed the shortest induction time at a 10 wt % RNS-A system, the lowest gas consumption indicated its weak CO 2 capture and storage ability. In addition, when the concentration was 6 wt %, RNS-A had the highest gas consumption and its reaction time was relatively short. To deeply explore the spontaneous combustion disaster of coal caused by air leakage and oxygen supply, low-temperature coal oxidation experiments under different oxygen concentrations (DOC) were carried out. Within the coal spontaneous combustion characteristic measurement system, a synchronous thermal analyzer (STA) and a Fourier transform infrared spectrometer (FTIR), the macro laws of gas and heat generation under DOC are analyzed, and the mechanism of the development of coal spontaneous combustion restricted by the lean-oxygen environment is also revealed. The results show that the change of oxygen concentration (OC) does not affect the critical temperature value and gas index change trend, but the lean-oxygen environment reduces the gas concentration and heat production rate very obviously. According to the temperature of the intersection, OC needs to be lowered to less than 5% when preventing spontaneous combustion of coal. The chain thermal reaction lags in the lean-oxygen environment, and the pyrolysis activity is significantly reduced. Meanwhile, the temperature points at T 6 and T 7 show significant differences. Furthermore, with increasing OC and temperature, the content of the aliphatic hydrocarbon presents an overall trend of first increasing, then decreasing, and continuously increasing after stage IV. It is concluded that • OH, aliphatic hydrocarbons, aromatic hydrocarbons, and carboxyl groups are the key groups for the coal spontaneous combustion evolution under DOC. To combine the spontaneous combustion reaction of coal in the DOC environment, the reaction path of the index gas in the macroscopic phenomenon and the reason for the concentration differences are revealed, the mechanism for exotherm varies caused by OC is clarified, and the microscopic inhibition affection on the chain reaction within the lean-oxygen environment is also explored. Noncovalent cross-linked hydrogels with promising mechanical properties are on demand for applications in tissue engineering, flexible electronics, and actuators. However, integrating excellent mechanical properties with facile preparation for the design of hydrogen bond cross-linked hydrogels is still challenging. In this work, an advanced hydrogel was prepared from acrylamide and N -acryloyl phenylalanine by one-pot free-radical copolymerization. Owing to hydrophobicity-assisted multiple hydrogen bonding interactions among phenylalanine derivatives, the hydrogels exhibited fascinating mechanical behaviors: tensile strength of 0.35 MPa, elongation at break of 2100%, tearing energy of 1134 J/m 2 , and compression strength of 3.56 MPa. The hydrogels also showed robust elasticity and fatigue resistance, and the compression strength did not show any decline, even after 100 successive cycles, as well as promising self-recovery property. In addition, the cytotoxicity test in vitro proved that the hydrogel showed good biocompatibility with normal human liver cells (LO2 cells). The excellent stretchability, robust elasticity, high toughness, fatigue resistance, and biocompatibility of the hydrogel demonstrated its vast potential in the biomedical field and flexible electronic devices.

Due to the reported fact that the active functional groups in coal can be dissolved and destroyed by ionic liquids, it is expected that the spontaneous combustion of coal can be affected from a thermodynamic perspective. However, ionic liquids with different thermal stabilities have distinct influences on coal combustion. Here, the thermal stability of long-flame coal in the presence of five pure ionic liquids ([Bmim][BF 4 ], [Bmim][Ac], [Bmim][NO 3 ], [Hoemim][BF 4 ], and [Pmim][BF 4 ]) was analyzed by thermogravimetric analysis, and the flammability of the raw coal, pure ionic liquids, and coal-IL mixtures (mass ratio of 1:1) were tested using a cone calorimeter according to the indexes of the time to ignition (TTI), mass loss rate (MLR), heat release rate (HRR), total heat release rate (THR), specific extinction area (SEA), and CO production. It is shown that the TTIs of mixtures containing coal-[Bmim][BF 4 ], coal-[Hoemim][BF 4 ], and coal-[Pmim][BF 4 ] are relatively long, and the MLR, HRR, THR, and SEA values are relatively low, indicating that these fluorine-containing ionic liquids have a better flame-retardant effect than the other two fluorine-free ones, which may be ascribed to their similar role to halogen inhibitors. In addition, the endothermic process of [Bmim][BF 4 ], [Hoemim][BF 4 ], and [Pmim][BF 4 ] can reduce the temperature of the coal surface and delay the ignition time of coal. In contrast, the TTI of coal-[Bmim][NO 3 ] and coal-[Bmim][Ac] mixtures is much shorter than that of coal alone, and the MLR, HRR, and THR values are larger. This may be caused by the poor thermal stability of the two nonfluorine ionic liquids that began to decompose and release heat prior to coal, providing a large amount of heat for the low-temperature oxidation of coal and thus accelerating coal oxidation and combustion. Coalfield fires during coal mining have become a major problem in the world today. To effectively prevent such disasters, we established an experimental platform to measure the spontaneous combustion characteristics of large-scale pressurized coal; thermal analysis experiments and microscopic analysis of briquettes under different axial pressures were carried out. It can be seen from the results that when the axial pressure is 4 MPa, the heating rate of the oxidative combustion of coal samples is accelerated, the crossing point temperature is lower (reduced by 71.09 °C), the activation energy is reduced (the second stage is decreased by 21.3 kJ/mol), and the oxidative combustion is more intense. Simultaneously, the porosity evolution process of briquettes under different axial pressures is simulated. Through calculation, it can be seen that the porosity and thermal conductivity show a linear increasing trend. The basis for the increase in the internal oxygen supply channels and increase in oxygen consumption when the axial pressure is 4 MPa is given. Through thermogravimetric-differential scanning calorimetry analysis, it is found that the maximum mass loss rate and maximum mass growth rate of residual coal after combustion under an axial pressure of 4 MPa are low, the residual rate after combustion is large, and the flammability rate is low when reoxidized, while complete combustion oxidation releases more heat. The application of axial pressure will change the combustion characteristics of briquettes, and the promotion effect is more obvious at 4 MPa. In order to improve the total sulfur removal rate in coal combustion, an acidic ionic liquid (IL) 1-carboxymethyl-3-methylimidazolium hydrogen sulfate ([HOOCCH 2 mim][HSO 4 ]) as the extractant combined with the oxidant 30% hydrogen peroxide (H 2 O 2 ) was applied to reduce the total sulfur content, and its microscopic mechanism of desulfurization was analyzed. The experimental results show that the desulfurization rate of the [HOOCCH 2 mim][HSO 4 ]-H 2 O 2 (1:10) solution was 45.12% and the organic sulfur removal rate was 16.26%, which were significantly higher than those of only H 2 O 2 or pure [HOOCCH 2 mim][HSO 4 ]. Fourier-transform infrared (FTIR) spectroscopy and X-ray photoelectron spectroscopy analyses showed that the mercaptan -SH and disulfide -S-S- in coal decreased after being treated with IL-H 2 O 2 . In particular, the results of FTIR spectroscopy indicated that the relative proportion of -S-S-and -SH treated with IL-H 2 O 2 (1:10) decreased by 31.9 and 27.2%, respectively, compared with that of a pure IL. This is due to H 2 O 2 oxidation; -SH and -S-S- were oxidized to sulfoxide and then the sulfoxide transferred from the coal phase to the IL phase, which improved organic sulfur removal from coal. Therefore, the combination of an ionic liquid and H 2 O 2 could increase the total desulfurization rate. In addition, the thermogravimetric analysis of coal is divided into four different stages; the weight loss during the combustion stage and the residues show that the IL-H 2 O 2 could improve the coal combustion because of good previous swelling and destruction of bridge bonds and hydrogen bonding of coal. Metabolomics, as an emerging technology, has been demonstrated to be a very powerful tool in the study of the host metabolic responses to infections by parasites. Schistosomiasis is a parasitic infection caused by schistosoma worm via the direct contact with the water containing cercaria, among which Schistosoma japonicum ( S. japonicum ) is endemic in Asia. In order to characterize the schistosome-induced changes in the host metabolism and further to develop the strategy for early diagnosis of schistosomiasis, we performed comprehensive LC-MS-based metabolomics analysis of serum from mice infected by S. japonicum for 5 weeks. With the developed diagnosis strategy based on our metabolomics data, we were able to successfully detect schistosomiasis at the first week post-infection, which was 3 weeks earlier than "gold standard" methods and 2 weeks earlier than the methods based on 1 H NMR spectroscopy. Our metabolomics study revealed that S. japonicum infection induced the metabolic changes involved in a variety of metabolic pathways including amino acid metabolism, DNA and RNA biosynthesis, phospholipid metabolism, depression of energy metabolism, glucose uptake and metabolism, and disruption of gut microbiota metabolism. In addition, we identified seventeen specific metabolites whose down-regulated profiles were closely correlated with the time-course of schistosomiasis progression and can also be used as an indicator for the worm-burdens. Interestingly, the decrease of these seventeen metabolites was particularly remarkable at the first week post-infection. Thus, our findings on mechanisms of host-parasite interaction during the disease process pave the way for the development of an early diagnosis tool and provide more insightful understandings of the potential metabolic process associated with schistosomiasis in mice. Background: Mounting evidence suggested a complex correlation between host lipid metabolism and Toxoplasma gondii (T. gondii) infection. However, the inherent association between T. gondii infection and host lipid state remains elusive either in mice or in human hosts., Methods: Through a study in a sample of 1045 healthy participants from eastern China, we determined the association of T. gondii infection and host lipid levels using serological methods. We then examined the host lipid levels in C57BL/6 J mice at both acute and chronic T. gondii infection stages (for a period up to 36 weeks post infection)., Results: In our case-control study, T. gondii seropositive individuals had higher low-density lipoproteins (LDL) (P = 0.0043) and total cholesterol (TC) (P = 0.0134) levels compared to seronegative individuals. Furthermore, individuals with LDL (OR = 3.25; 95% CI:1.60-6.61) and TC (OR = 2.10; 95% CI:1.22-3.63) levels above the upper limit of normal range had higher odds ratio to be T. gondii IgG positive. Consistently, in vivo data revealed that a significantly increased LDL level was first observed at early acute stage but plateaued to later time (chronic infection with T. gondii)., Conclusions: In both healthy population and T. gondii-infected mice, seropositive individuals had higher LDL level. Individuals with positive T. gondii IgG had more odds of being with LDL and TC abnormality. Latent T. gondii infection is common worldwide, potential medical interventions to host lipid metabolism may be a breakthrough point to the prevention and control of this parasite infection.

The SO 2 solubility in ionic liquids and absorption mechanisms with different functionalities, including ether, halide, carboxylate, dicarboxylate, thiocynate, phenol, amino, azole groups, etc., are presented in this review. Strategies of improving SO 2 capture with low binding energy and the separation performance from CO 2 are also concluded. Generally, moderate basicity is favourable for enhancing SO 2 capacity and the water (below 6 wt%) effect on absorption is indefinite but generally slight. Introducing electron-withdrawing substituents such as nitrile, halogen, aldehyde and carboxylic groups are proposed to decrease the chemical absorption enthalpy between ionic liquid and SO 2 in order to reduce regeneration power consumption. Although it is promising, the absorption enthalpy is still much higher than the physisorption performance especially of the ether-functionalized ones. The biocompatible choline-based, betaine-based, and amino acid ionic liquids have clear trends to be applied in SO 2 capture due to their biodegradability, nontoxicity and easy accessibility. Generally, comparing to the traditional solvents, ionic liquids have made great improvement in SO 2 capacity, however, the high viscosity and desorption energy are two main obstacles for SO 2 absorption and separation. Background: Hepatic fibrosis caused by chronic infection with Schistosoma japonica remains a serious public health problem in the world. Symptoms include inflammation, liver granuloma and fibrosis, whilst treatment options are still limited. This study aims to investigate whether and how traditional Chinese medicine Xiaochaihu decoction (XCH) could mitigate liver fibrosis caused by S. japonicum infection., Methods: BALB/c mice were infected with S. japonicum cercariae and treated with XCH for 16 weeks. Liver pathological changes were assessed by H&E and Masson staining. NIH3T3 and Raw264.7 cells were treated with S. japonicum egg antigens with or without XCH treatment. Quantitative real-time PCR, western blot, immunfluorescence and ELISA were performed to determine the changes of levels of fibrogenic markers., Results: XCH protected mouse liver from injuries and fibrosis caused by S. japonicum infection and considerably reduced egg burden in a dose-dependent manner. Infection with S. japonicum caused elevation of serum ALT, AST, ALP, HA and PIIINP levels and reduction of ALB and GLOB levels, which was markedly suppressed by XCH. The upregulation of TGF-β1, Hsp47, α-SMA, Col1A1 and Col3A1 in S. japonicum-infected mouse liver was also significantly inhibited by XCH. Schistosoma japonicum egg antigens promoted the expression of Hsp47, TGF-β1, Timp-1, α-SMA, Col1A1 and Col3A1 in NIH3T3 cells, and TGF-β1, CTGF, IL-13, IL-17 and IL-6 in Raw264.7 cells, which was inhibited by XCH, LY2157299 and shRNA-Hsp47., Conclusions: These results demonstrated that the hepatic protective effects of Xiaochaihu decoction were mediated by HSP47/TGF-β axis.

It has been documented that Helicobacter hepaticus (H. hepaticus) infection is linked to hepatic inflammation and fibrosis. Interleukin 33 (IL-33) is a cytokine involved in inflammatory and fibrotic diseases, but its relevance to H. hepaticus infection-induced liver inflammation and fibrosis is unknown. In this study, we found that the expression of IL-33 in mice liver was significantly induced by H. hepaticus infection at 24 weeks post infection (WPI). Immunohistochemistry analysis revealed that IL-33 was transferred from the nucleus to the cytoplasm due to infection. The quantitation of inflammatory cytokine and histopathology evaluation showed that IL-33 knockdown attenuated the H. hepaticus-induced hepatic inflammation and fibrosis. More importantly, H. hepaticus promoted the expression of the IL-33 receptor ST2 on cell surfaces, and the expression of ST2 then activated the expression nuclear factor-κB (p65), α-SMA, and Erk1/2. Background: It has been documented that Helicobacter hepaticus (H hepaticus) infection is linked to chronic hepatitis and liver cancer. However, our understanding of the molecular mechanisms underlying progression of the H hepaticus-induced hepatic inflammation to cellular hepatocarcinoma is still limited., Materials and Methods: In our study, male BALB/c mice were infected by H hepaticus for 8, 12, 16, 20, and 24 weeks. Histopathology, H hepaticus colonization dynamics, select signaling pathways, and expression of key inflammatory cytokines in the liver were examined., Results: We found that H hepaticus was detectible in feces of mice at 7 days postinfection (DPI) by PCR, but it was not detected in the livers by PCR until 8 weeks postinfection (WPI). In addition, abundance of colonic and hepatic H hepaticus was progressively increased over the infection duration. H hepaticus-induced hepatic inflammation and fibrosis were aggravated over the infection duration, and necrosis or cirrhosis developed in the infected liver at 24 WPI H hepaticus infection increased levels of alanine aminotransferase and aspartate aminotransferase. Moreover, mRNA levels of Il-6 and Tnf-α were significantly elevated in the livers of H hepaticus-infected mice compared to uninfected control from 8 WPI to 24 WPI. Introduction: To investigate the effects of crocin on proliferation and migration of endogenous neural stem cells and the Notch1 signalling pathway in rats after cerebral ischemia reperfusion., Material and Methods: SD rats were randomly divided into the sham operation group, model group and administration group (crocin). Middle cerebral artery occlusion (MCAO/R) was used to establish the focal cerebral ischemia reperfusion model in rat. After surgical treatment, the treatment group was treated with crocin. Quantitative polymerase chain reaction (qPCR) was used to detect the changes in the expression of Notch1, Bax and bcl-2 proteins in rat endogenous neural stem cells after cerebral ischemia reperfusion. ELISA was used to detect changes in inflammatory factors. Neural stem cells were cultured in vitro, which were divided into: the normal control group, the hypoglycaemic deprivation/reoxygenation group, hypoglycaemic deprivation/reoxygenation group with a low concentration of crocin, and hypoglycaemic deprivation/reoxygenation group with a high concentration of crocin. The cell proliferation assay detects cell activity. The cell migration assay tests the cell migration ability. And flow cytometry was used to determine cell apoptosis., Results: Compared with the sham group, the Notch1 signalling pathway was activated in the model group. The expression of Notch1 in the crocin group was increased compared to the model group. Crocin can inhibit the release of inflammatory factors. The results of our experiments showed that crocin could induce the proliferation and migration of neural stem cells and inhibit the apoptosis of neural stem cells in the hypoglycaemic/reoxygenation model group., Conclusions: Crocin sufficiently promotes the proliferation and migration of neural stem cells and inhibits the apoptosis of these cells in rats after ischemia-reperfusion by manipulating the Notch signalling pathway.

Alantolactone is a sesquiterpene lactone extracted from Inula helenium L. plants possessing many biological activities, including anti-inflammatory, antiproliferation, and antimicrobial. The inhibitory effects and the underlying mechanisms of alantolactone on lung cancer cells NCI-H1299 and Anip973 were investigated in this study. The results showed that alantolactone could decrease cell viability and induce cell apoptosis of NCI-H1299 and Anip973. After the cells were treated with alantolactone, the expression of Bcl-2 decreased, while the expression of Bax increased, the expression of MMP-9, MMP-7, and MMP-2 gradually decreased after alantolactone treatment. Furthermore, results showed that alantolactone could activate p38 MAPK pathway and suppress NF-κB pathway, which are involving in lung cancer development. These results indicated that alantolactone was a potential agent for lung cancer treatment. PRACTICAL APPLICATIONS: Lung cancer is one of the most common contributors of cancer death in the world. Chemoprevention and chemotherapy with natural substances are prospective methods for lung cancer treatment. In recent years, the anti-cancer activity of various sesquiterpene lactones has attracted a great deal of interest. Alantolactone is the major active sesquiterpene lactones isolated from Inula helenium L, which is used as a medicine in ancient Romans due to wide range of pharmacological activities. Aldo-keto reductase family 1 member B10 (AKR1B10) is a secretory protein overexpressed in hepatocellular carcinoma (HCC). We aimed to evaluate AKR1B10 as a serum marker for detection of HCC. Herein, we conducted a cohort study that consecutively enrolled 1,244 participants from three independent hospitals, including HCC, healthy controls (HCs), benign liver tumors (BLTs), chronic hepatitis B (CHB), and liver cirrhosis (LC). Serum AKR1B10 was tested by time-resolved fluorescent assays. Data were plotted for receiver operating characteristic (ROC) curve analyses. Alpha-fetoprotein (AFP) was analyzed for comparison. An exploratory discovery cohort demonstrated that serum AKR1B10 increased in patients with HCC (1,567.3 ± 292.6 pg/mL; n = 69) compared with HCs (85.7 ± 10.9 pg/mL; n = 66; P < 0.0001). A training cohort of 519 participants yielded an optimal diagnostic cutoff of serum AKR1B10 at 267.9 pg/mL. When ROC curve was plotted for HCC versus all controls (HC + BLT + CHB + LC), serum AKR1B10 had diagnostic parameters of the area under the curve (AUC) 0.896, sensitivity 72.7%, and specificity 95.7%, which were better than AFP with AUC 0.816, sensitivity 65.1%, and specificity 88.9%. Impressively, AKR1B10 showed promising diagnostic potential in early-stage HCC and AFP-negative HCC. Combination of AKR1B10 with AFP increased diagnostic accuracy for HCC compared with AKR1B10 or AFP alone. A validation cohort of 522 participants confirmed these findings. An independent cohort of 68 patients with HCC who were followed up showed that serum AKR1B10 dramatically decreased 1 day after operation and was nearly back to normal 3 days after operation. The conversion of CO 2 into carbon-based fuels via electrochemical reduction is a promising approach to realizing the termination of carbon cycling. Copper (Cu 0 ) is considered to be an effective electrochemical catalyst for such purpose. However, it is limited by its instability and low selectivity. In this work, Cu 2+ was embedded into a polycondensation structure of resorcinol (R) and formaldehyde (F) via sol-gel reaction, and the complex was then calcined at high temperature to obtain copper nanoparticles-dispersed carbon aerogels (Cu/CA). The samples were further activated sequentially under CO 2 and N 2 atmosphere. The final products under different activation temperature of secondary N 2 atmosphere were obtained and marked as Cu/CA-CO 2 -N 2 -T. The existence of highly dispersed Cu 0 on the carbon aerogels was confirmed by XPS and HRTEM, which exhibited attractive activity towards electrochemical CO 2 reduction reaction (CO 2 RR) forming carbon product in 0.1 M KHCO 3 aqueous media. Furthermore, the CO 2 RR product distribution varied under different samples. The sample activated under 700 °C (Cu/CA-CO 2 -N 2 -700) with Cu loading c.a. 5.13 wt% showed remarkable CO faradaic efficiency (FE) (75.6%) at low overpotential of -0.49 V. Further calcination led to more exposure of copper, and CO dimerization to C2 product. The high selectivity toward CO by Cu/CA-CO 2 -N 2 -700 maybe attributed to their excellent stability in maintaining the metallic nature of Cu component within the hydrophobic carbon aerogels during electrocatalysis, which further prevents the adsorption and reduction of CO on the catalyst., (Copyright © 2019 Elsevier Inc. Cases of newly developed advanced schistosomiasis (NDAS) have occurred in areas where schistosomiasis transmission has been blocked for more than 25 years. The causes and pathogenesis of NDAS are still unknown. Diagnosis of NDAS relies on historical investigation and clinical symptoms, such as liver fibrosis, hepatic ascites and abnormal biochemical indexes in serum. It is important but difficult at this stage to develop a new tool for early screening and rapid diagnosis. In this study, serum peptides from thirty patients with NDAS and thirty healthy controls were captured with weak cation exchange magnetic beads, and subjected to MALDI-TOF mass spectrometry and ClinProTools analysis. Eleven peaks with m/z 924, 2661, 2953, 2991, 3241, 3884, 5337, 5905, 5943, 7766 and 9289 were decreased and three peaks with m/z 1945, 2082 and 4282 were increased in the NDAS group. The proteomic detection pattern (PDP) was established with 14 different peptide peaks, and its sensitivity and specificity were investigated with a blind test. The peptide mass fingerprints of sera from 50 NDAS patients and 100 healthy controls were double-blind subjected to the PDP method, and 50 patients and 92 healthy controls were classified as NDAS and healthy separately, which showed 100% sensitivity and 92% specificity. Aim: This study sought to determine the rate of recurrence of gestational diabetes mellitus (GDM) recurrence during the second pregnancies of women who were diagnosed with GDM during their first pregnancies, to identify risk factors associated with the probability of such recurrence and to evaluate the influence of GDM recurrence on pregnancy outcomes in north Zhejiang, China, after the recent adjustment to the nation's childbirth policy., Methods: A retrospective longitudinal study was performed in north Zhejiang, China (at Jiaxing Maternal and Child Health Hospital). A total of 128 women who delivered two sequential live singleton infants and were diagnosed with diet-treated GDM during their first pregnancies were included., Results: According to the 2013 World Health Organization diagnostic criteria for diabetes during pregnancy, the prevalence of gestational diabetes was 11.02% in northern Zhejiang. The recurrence rate of GDM in northern Zhejiang was 43.75% (56/128). The age at second pregnancy, weight gain during pregnancy, interpregnancy interval and macrosomia during the index pregnancy were risk factors for GDM recurrence. Among those women with recurrent GDM, GDM developed earlier and caesarean section was more frequently required during the second pregnancy; in addition, the second pregnancy was associated with more premature and low birthweight infants but less macrosomia., Conclusion: The recurrence rate of GDM is high in northern Zhejiang. Asthma is a chronic airway inflammation in which Th2 and Th17 cells play critical roles in its pathogenesis. We have reported that atypical protein kinase (PKC) λ/ι is a new regulator for Th2 differentiation and function. However, the role of PKCλ/ι for Th17 cells remains elusive. In this study, we explored the effect of PKCλ/ι on Th17 cells in the context of ex vivo cell culture systems and an in vivo murine model of allergic airway inflammation with the use of activated T cell-specific conditional PKCλ/ι-deficient mice. Our findings indicate that PKCλ/ι regulates Th17 cells. The secretion of Th17 effector cytokines, including IL-17, IL-21 and IL-22, were inhibited from PKCλ/ι-deficient T cells under non-skewing or Th17-skewing culture conditions. Moreover, the impaired Th17 differentiation and function by the PKCλ/ι-deficiency was associated with the downregulation of Stat3 and Rorγt, key Th17 transcription factors. We developed a model of Th17 and neutrophil-involved allergic airway inflammation by intratracheal inoculation of house dust mites. PKCλ/ι-deficiency significantly inhibited airway inflammations. The infiltrating cells in the lungs and bronchoalveolar lavage fluids were significantly reduced in conditional PKCλ/ι-deficient mice. Th17 effector cytokines were reduced in the bronchoalveolar lavage fluids and lungs at protein and mRNA levels. A home-made near-infrared laser tweezers Raman spectroscopy (LTRS) system was applied to detect hemoglobin variation in red blood cells (RBCs) from diabetes without exogenous labeling. Results showed significant spectral differences existed between the diabetic and normal RBCs, including the peaks dominated by protein components (e.g. 1003 cm -1 ) and heme groups (e.g. 753 cm -1 ) in RBCs, and accurate classification results for diabetes detection were obtained by linear discriminant analysis with 100% sensitivity (i.e. no false negatives in the study). The "hygiene hypothesis" is a theory try to explain the dramatic increases in the prevalence of autoimmune and allergic diseases over the past two to three decades in developed countries. According to this theory, reduced exposure to parasites and microorganisms in childhood is the main cause for the increased incidences of both T helper 1 (Th1)-mediated autoimmunity and Th2-mediated allergy. In this study, we investigated the impact of Schistosoma japonicum infection on the allergic airway inflammation induced by repeated intracheal inoculations of house dust mites (HDM), which is a Th17 and neutrophils dominant murine asthma model, mimicking severe asthma. We found that S. japonicum infection downregulated airway hyperresponsiveness. The infiltrating cells, Th17 and Th2 effector cytokines in the bronchoalveolar lavage (BAL) fluids and lungs were significantly reduced in the infected mice. Our findings indicated that S. japonicum infection was able to effectively inhibit host's allergic airway inflammation, which may be related to the upregulated Treg cells upon infection. To our knowledge, it is the first study to reveal the impact of S. japonicum infection on house dust mite induced severe asthma. More in depth investigation is need to elucidate the underlying mechanisms.

MicroRNAs (miRNAs) play critical roles in tumorigenesis and metastasis by negatively regulating gene expression through complementary binding to the 3'-untranslated region of target mRNAs. The role of miRNAs in expression of the tumor suppressor DAB2IP in bladder cancer (BC) remains unknown. The aim of the present study was to identify miRNAs targeting DAB2IP and determine their expression and function in BC. We predicted candidate miRNAs targeting DAB2IP using TargetScan software. Dual-luciferase reporter assays confirmed that miRNA-556-3p directly regulated DAB2IP expression. Quantitative RT-PCR and RNase protection assays showed that endogenous miRNA-556-3p expression was significantly upregulated in clinical samples of BC patients and BC cell lines and western blot analysis indicated that DAB2IP expression in BC tissues and BC cell lines was concurrently downregulated. Gain or loss of function studies showed that upregulation of miRNA-556-3p promoted proliferation, invasion, migration and colony formation of BC cells, whereas downregulation resulted in opposite effects. Importantly, restoration of DAB2IP expression rescued the effects induced by miRNA-556-3p. Overexpression of miRNA-556-3p in BC cells not only decreased DAB2IP expression, but also markedly increased Ras GTPase activity and ERK1/2 phosphorylation level. These findings suggest that DAB2IP is a direct target of miRNA-556-3p, and endogenous miRNA-556-3p expression shows inverse correlation with simultaneous DAB2IP expression in BC tissues and cells. miRNA-556-3p functions as a tumor promoter in tumorigenesis and metastasis of BC by targeting DAB2IP. Moreover, miRNA-556-3p-mediated DAB2IP suppression plays an oncogenic role by partial activation of the Ras-ERK pathway.

The cationic starch flocculants were synthesized by the reaction of maize starch which was activated by a ball-milling treatment with 2,3-epoxypropyl trimethyl ammonium chlorides (ETMAC) using the dry method. The cationic starches were characterized by several approaches including scanning electron microscope (SEM), degree of substitution (DS), infrared spectrum (IR), X-ray diffraction (XRD), flocculating activity, electron spin resonance (ESR), and solid-state nuclear magnetic resonance (NMR). The effect of mechanical activation on starch etherifying modification was investigated. The mechanical activation cracked starch granules and destructed their crystal structures. This resulted in enhancements to the reaction activity and reaction efficiency, which was approved by ESR and solid state NMR. The starch flocculants, synthesized by the reaction of mechanically activated starches at 90°C for 2.5h with ETMAC at molar ratio of 0.40:1.00, showed good flocculation activity. The substitution degree (0.300) and reaction efficiency (75.06%) of starch flocculants synthesized with mechanically activated starches were significantly greater than those of starch flocculants with native starches (P<0.05)., (Copyright © 2016 Elsevier B.V. The interactions between the three kinds of naphthalimide-based anti-tumor drugs (NADA, NADB, NADC) and human serum albumin (HSA) under simulated physiological conditions were investigated by fluorescence spectroscopy, circular dichroism spectroscopy and molecular modeling. The results of the fluorescence quenching spectroscopy showed that the quenching mechanisms for different drugs were static and their affinity was in a descending order of NADA > NADB > NADC. The relative thermodynamic parameters indicated that hydrophobic force was the predominant intermolecular force in the binding of NAD to HSA, while van der Waals interactions and hydrogen bonds could not be ignored. The results of site marker competitive experiment confirmed that the binding site of HSA primarily took place in site I. Furthermore, the molecular modeling study was consistent with these results. Background: Toll-like receptors (TLRs) are key factors in the innate immune system and initiate the inflammatory response to foreign pathogens such as bacteria, fungi and viruses. In the microenvironment of tumorigenesis, TLRs can promote inflammation and cell survival. Toll-like receptor 2/6 (TLR2/6) signaling in tumor cells is regarded as one of the mechanisms of chronic inflammation but it can also mediate tumor cell immune escape and tumor progression. However, the expression of TLR2 and its biological function in the development and progression of hepatocarcinoma have not been investigated. This study aimed to determine the expression of TLRs 1-10 in the established human hepatocellular carcinoma cell line BLE-7402, to investigate the biological effect of TLR2 on cell growth and survival., Methods: TLR expression in BLE-7402 cells was assayed by RT-PCR, real-time PCR and flow cytometry (FCM). To further investigate the function of TLR2 in hepatocarcinoma growth, BLE-7402 cells were transfected with recombinant plasmids expressing one of three forms of TLR2 siRNA (sh-TLR2 RNAi(A, B and C)). TLR2 knockdown was confirmed using RT-PCR, real-time PCR and fluorescence microscopy. Tumor cell proliferation was monitored by MTT assay and secreted cytokines in the supernatant of transfected cells were measured by bead-based FCM, the function of TLR2 siRNA was also investigated in vivo., Results: The BLE-7402 cell line expressed TLRs 2 to 10 at both mRNA and protein levels. TLR2 was the most highly expressed TLR. While all the three siRNAs inhibited TLR2 mRNA and protein expression, sh-TLR2 RNAi(B) had the strongest knockdown effect. TLR2 knockdown with sh-TLR2 RNAi(B) reduced cell proliferation. Furthermore, secretion of IL-6 and IL-8 was also reduced. The result showed a drastic reduction in tumor volume in mice treated with sh-TLR2 RNAi(B)., Discussion: These results suggest that TLR2 knockdown inhibit proliferation of cultured hepatocarcinoma cells and decrease the secretion of cytokines. It is suggested that TLR2 silencing may worth further investigations for siRNA based gene therapy in treatment of hepatocarcinoma.

Duck astroviruses (DAstVs) are known to cause duck viral hepatitis; however, little is known regarding their molecular biology. Here, we report the complete sequence of a DAstV associated with a recent outbreak of fatal hepatitis in ducklings in China. Sequence analyses indicated that the genome of DAstV possessed a typical astrovirus organization and also exhibited two unique features. The polyadenylated genome comprised 7722 nt, which is the largest among astroviruses sequenced to date. The ORF2 of DAstV was not in the same reading frame as either ORF1a or ORF1b, which was distinct from all other astroviruses. Sequence comparisons and phylogenetic analyses revealed that DAstV was more closely related to turkey astrovirus (TAstV) type 2, TAstV-3 and TAstV/MN/01 (a possible new TAstV serotype) than to TAstV-1 or other astroviruses. These findings suggest that astroviruses may transmit across ducks and turkeys.

To develop a new approach for the detection and typing of duck hepatitis A virus (DHAV), a pair of non-degenerate primers was designed to amplify a approximately 250-bp genomic region in the 5'UTR. 3 reference strains and 6 duck embryo-derived isolates from various regions in China, involving 2 serotypes, were successfully amplified with the primer set. By determining the nucleotide sequence of the amplicon, a molecular typing method was developed. If isolate sequences were compared to DHAV 5'UTR sequences available in public databases, nucleotide identity was > or =94% with homologous serotype and < or =73% with heterologous serotypes. Phylogenetic analysis revealed monophyletic clustering of 5'UTR sequences of a homologous serotype, confirming the new classification of DHAV (serotype 1 and the two new serotypes recently described in Taiwan and South Korea, respectively) into three genotypes (A, B and C) defined by the capsid coding region. Analysis of the results showed that the primer pair should aid in the detection of DHAV, and that the amplicon sequence contains type-specific information and can be used for effective and rapid molecular typing. The molecular methods proved their utility through the detection and typing of DHAV directly from 28 liver specimens collected from dead ducklings during duck viral hepatitis outbreaks in different regions of China between 2001 and 2007. The results confirmed the presence of DHAV in all of the 28 samples and demonstrated that genotypes A (13/28) and C (15/28) of DHAV are co-circulating in China.

The schistosome integral membrane protein Sm/Sj23 was initially shown to induce protection in mice as a synthetic peptide vaccine and further, as a plasmid DNA vaccine to induce protection in mice, sheep and water buffalo. In this study we asked if we could induce protection against challenge infection in pigs against Schistosoma japonicum by vaccinating them with a plasmid DNA vaccine encoding the S. japonicum Chinese strain 23 kDa membrane protein. Further, we asked if we could enhance protective efficacy of this vaccine by the addition of IL-12. We compared vaccination with SjC23 plasmid DNA alone or with IL-12 plasmid DNA in pigs. Pigs were immunized three times at three weekly intervals. Thirty Chinese Songjang native pigs were divided into three groups. In group A, each pig was immunized with 500 microg of SjC23 plasmid DNA by intramuscular (i.m.) injection in both buttocks. In group B each pig was immunized with 500 microg of SjC23 plasmid DNA, and 500 microg of each of pcDNA3.1-p35 and 500 microg of pcDNA3.1-p40 DNA by i.m. injection. In group C each pig was immunized with 500 microg of pcDNA3.1 as the control. Thirty days post-vaccination, pigs were challenged with S. japonicum cercariae and adult and egg burdens and granuloma size determined 45 days post-challenge. The results showed that worm reduction rates in SjC23 group compared with control group were 29.2% and in the SjC23 + IL-12 group reduced 58.6%. Similarly the female worm reduction rates were 50.8 and 58.8%, the hepatic egg reduction rates were 48.2 and 56.4%, and the mean square measure reduction rates of hepatic egg granulomas were 48.6 and 44.4%, the mean diameter reduction rates of granulomas were 27.6 and 22.8% in pigs vaccinated with SjC23 or SjC23 + IL-12 compared to plasmid vaccinated pigs, respectively. Analysis of sera from pigs vaccinated with SjC23 showed that 4 of 10 pigs had anti-Sj23 antibody responses; with 5 of 10 pigs positive for anti-Sj23 in the SjC23+IL-12 group. These results suggest that vaccination with Sj23 DNA vaccine induces not only a significant reduction in worm and egg burdens, but also significantly reduces the size of egg granulomas, thus is also anti-pathology.
