Background: Problem-Based Learning (PBL) education can improve nursing students' thinking skills and overall quality of life, but its effectiveness depends largely on the level of student engagement within the group. Finding an intervention to increase nursing students' classroom engagement is critical to improving learning outcomes., Objectives: To explore the impact of a grouping intervention using the Felder-Silverman Learning Style Model (FSLSM) on PBL in nursing education, including classroom engagement, critical thinking, self-directed learning skills, and academic performance., Design: A single-center, single-blind, double-arm, parallel randomized controlled trial., Settings: One university school of nursing., Participants: The sample consisted of 56 fourth-year undergraduate nursing students., Methods: Students were randomly assigned to experimental group (n = 28) and control group (n = 28). The experimental group received PBL education based on FSLSM grouping and control group received traditional PBL education. Data collection was done using Demographic questionnaire, Index of Learning Style scale, Classroom Engagement Questionnaire for Students, Critical Thinking Disposition Inventory-Chinese Version, and Self-Directed Learning Skill scale. The data were analyzed using descriptive statistics, paired t-test, independent t-test, and chi-square test., Results: The FSLSM-based grouping significantly enhanced classroom engagement and critical thinking in the experimental group compared to the control group. While both groups improved in self-directed learning skills, the experimental group showed greater gains in learning motivation and information quality. Mitochondria are organelles involved in energy metabolism and biosynthesis. As the metabolites released from mitochondria are raw materials used for lipid synthesis, mitochondria also play important roles in lipid metabolism. Here we report that Slc1, a 1-acylglycerol-3-phosphate O-acyltransferase in the fission yeast Schizosaccharomyces pombe, is required to maintain tubular mitochondrial morphology and normal mitochondrial functions. The absence of Slc1 causes mitochondrial fragmentation, increases mitochondrial fission frequency, reduces mitochondrial respiration, and slows down nitrogen starvation-induced mitophagy. In addition, the absence of Slc1 significantly increases the protein level of Ptl2, which is the triacylglycerol lipase localized on lipid droplets. The phenotypes caused by the absence of Slc1 depend on its acyltransferase enzymatic activity. The polycrystalline nature of perovskites, stemming from their facile solution-based fabrication, leads to a high density of grain boundaries (GBs) and point defects. However, the impact of GBs on perovskite performance remains uncertain, with contradictory statements found in the literature. We developed a machine learning force field, sampled GB structures on a nanosecond time scale, and performed nonadiabatic (NA) molecular dynamics simulations of charge carrier trapping and recombination in stoichiometric and doped GBs. The simulations reveal long, microsecond carrier lifetimes, approaching experimental data, stemming from charge separation at the GBs and small NA coupling, 0.01-0.1 meV. Stoichiometric GBs exhibit transient trap states, which, however, are not particularly detrimental to the carrier lifetime. Halide dopants form interstitial defects in the bulk, but have a stabilizing influence on the GB structure by passivating undersaturated Pb atoms and reducing the transient trap state formation. On the contrary, excess Pb destabilizes GBs, allowing formation of persistent midgap states that trap charges. Still, the charge carrier lifetime reduces relatively little, because the midgap states decouple from the bands, and charges are more likely to escape back into bands upon a GB structural fluctuation. The atomistic study into the structural dynamics of perovskite GBs and its influence on charge carrier trapping and recombination provides valuable insights into the complex properties of perovskites and the intricate role of GBs in the material performance.

Vanadium oxide (VO 2 ) is an exotic phase-change material with diverse applications ranging from thermochromic smart windows to thermal sensors, neuromorphic computing, and tunable metasurfaces. Nonetheless, the mechanism responsible for its metal-insulator phase transition remains a subject of vigorous debate. Here, we investigate the ultrafast dynamics of the photoinduced phase transition in VO 2 under low perturbation conditions. By experimentally examining carrier relaxation dynamics at energy levels near the VO 2 band gap (0.6-0.92 eV), we note that numerous optical features do not correspond to the first-order phase transition. Previous studies indeed induced such a phase transition, but they relied on fluences at least an order of magnitude higher, leading to temperature increases well above the transition threshold (340 K). Instead, for excitation fluences that correspond to lattice temperatures only in slight excess of the phase transition (absolute temperatures < 500 K), we find that the marked changes in optical properties are dominated by a shift in the electronic density of states/Fermi level. We find that this effect is a lattice-driven process and does not occur until sufficient energy has been transferred from the excited electrons into the phonon subsystem.

Objective: To explore the performance of deep learning-based segmentation of infarcted lesions in the brain magnetic resonance imaging (MRI) of patients with acute ischemic stroke (AIS) and the recurrence prediction value of radiomics within 1 year after discharge as well as to develop a model incorporating radiomics features and clinical factors to accurately predict AIS recurrence., Materials and Methods: To generate a segmentation model of MRI lesions in AIS, the deep learning algorithm multiscale residual attention UNet (MRA-UNet) was employed. Furthermore, the risk factors for AIS recurrence within 1 year were explored using logistic regression (LR) analysis. In addition, to develop the prediction model for AIS recurrence within 1 year after discharge, four machine learning algorithms, namely, LR, RandomForest (RF), CatBoost, and XGBoost, were employed based on radiomics data, clinical data, and their combined data., Results: In the validation set, the Mean Dice (MDice) and Mean IOU (MIou) of the MRA-UNet segmentation model were 0.816 and 0.801, respectively. In multivariate LR analysis, age, renal insufficiency, C-reactive protein, triglyceride glucose index, prognostic nutritional index, and infarct volume were identified as the independent risk factors for AIS recurrence. Furthermore, in the validation set, combining radiomics data and clinical data, the AUC was 0.835 (95%CI:0.738, 0.932), 0.834 (95%CI:0.740, 0.928), 0.858 (95%CI:0.770, 0.946), and 0.842 (95%CI:0.752, 0.932) for the LR, RF, CatBoost, and XGBoost models, respectively., Conclusion: The MRA-UNet model can effectively improve the segmentation accuracy of MRI. Aim: To review the experiences of family caregivers using home care services, the perception of home care services and the positive and negative effects on family caregivers., Design: A scoping review of the literature was undertaken using JBI guidance., Data Sources: Databases were searched in November 2023, including PubMed, Web of Science, Embase, CINAHL, Cochrane Library and CNKI (China National Knowledge Infrastructure)., Review Methods: Eligible articles included peer-reviewed literature describing experience using home care services. Reviewers screened titles and abstracts of located sources, and to screen full-text articles. A table was used to extract data and synthesise results., Results: We found that 22 articles met the inclusion criteria, most of which were published since 2010 and this study found inconsistent findings across studies of the impact of home care services on care burden. In addition, family caregivers still have negative experiences in the process of use, such as the difficulty in obtaining service information, believing that the service duration and content cannot meet their needs, and the lack of continuity of care is the most involved negative experience., Conclusion: Current home care services cannot fully meet the needs of family caregivers. Family caregivers have different expectations of policies, institutions and nursing staff. Future home care service practices should fully consider the unmet needs of caregivers and provide them with higher-quality nursing services., Impact: This study provides new evidence that there are still unmet needs for family caregivers in the use of home care services. Metal halide perovskites (MHPs) have attracted strong interest for a variety of applications due to their low cost and excellent performance, attributed largely to favorable defect properties. MHPs exhibit complex dynamics of charges and ions that are coupled in unusual ways. Focusing on a combination of two common MHP defects, i.e. , a grain boundary (GB) and a Pb interstitial, we developed a machine learning model of the interaction potential, and studied the structural and electronic dynamics on a nanosecond timescale. We demonstrate that point defects at MHP GBs can create new chemical species, such as Pb-Pb-Pb trimers, that are less likely to occur with point defects in bulk. The formed species create structural instabilities in the GB and prevent it from healing towards the pristine structure. Pb-Pb-Pb trimers produce deep trap states that can persist for hundreds of picoseconds, having a strong negative influence on the charge carrier mobility and lifetime. Such stable chemical defects at MHP GBs can only be broken by chemical means, e.g. , the introduction of excess halide, highlighting the importance of proper defect passivation strategies. Long-lived GB structures with both deep and shallow trap states are found, rationalizing the contradictory statements in the literature regarding the influence of MHP GBs on performance.

Objective: To investigate the association between Oxidative Balance Score (OBS) and glaucoma risk., Methods: Using data from the National Health and Nutrition Examination Survey (2005-2008), we analyzed 2,615 participants aged ≥40 years. OBS was calculated from 15 antioxidant and 5 pro-oxidant components, including dietary nutrients and lifestyle factors. Glaucoma was identified through self-reported diagnosis and retinal imaging. Survey-weighted logistic regression and restricted cubic splines (RCS) were used to assess associations., Results: Higher OBS was associated with lower glaucoma risk (adjusted OR = 0.97, 95% CI: 0.94-1.00). Participants in the highest OBS quartile showed 51% lower odds of glaucoma compared to the lowest quartile (adjusted OR = 0.49, 95% CI: 0.27-0.90). Introduction: Social security, as a core component of the national welfare system, has consistently played a crucial role in ensuring the basic livelihood of citizens and promoting social equity and justice. Against this backdrop, this study explores the association between social security satisfaction and acceptance of vulnerable groups., Methods: This study involved 9923 participants. Generalized linear regression and smooth curve fitting were used to assess the association between social security satisfaction and acceptance of vulnerable groups. Threshold effect was examined by piecewise regression. We conducted subgroup analyses and assessed the potential interaction effect., Results: A non‑linear association was detected between social security satisfaction and inclusion of vulnerable groups with an inflection point of 45.00. When social security satisfaction was < 45.00, inclusion of vulnerable groups increased with increasing social security satisfaction score up to inflection point. The association between social security satisfaction and inclusion of vulnerable groups differed across gender, education level, and spouse subgroups., Discussion: The study reveals the importance of social security satisfaction on their acceptance of vulnerable groups. Osteomyelitis has gradually become a catastrophic complication in orthopedic surgery due to the formation of bacterial biofilms on the implant surface and surrounding tissue. The therapeutic challenges of antibiotic resistance and poor postoperative osseointegration provide inspiration for the development of bioactive implants. We have strategically designed bioceramic scaffolds modified with Lactobacillus reuteri (LR) and bacteriophages (phages) to achieve both antibacterial and osteogenic effects. Leveraging the tendency of bacteria to adhere to the surface of implants, bioceramics have been modified with LR biofilm to promote bone repair. The LR biofilm, sterilized by pasteurization, prevents sepsis caused by live bacteria and is biocompatible with phages. Phages, being natural enemies of bacteria, not only effectively kill bacteria and inhibit biofilm formation but also readily adsorb onto the surface of bioceramics. Hence, this scaffold, loaded with a phage cocktail, lysates specific bacterial populations, namely Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). More importantly, the inactivated LR biofilm stimulates macrophages RAW264.7 to polarize towards an anti-inflammatory M2 phenotype, creating an immune microenvironment favorable for inducing osteogenic differentiation of rat mesenchymal stem cells in vitro . In a rat model of infectious cranial defects, the scaffold not only effectively eliminated S. aureus and alleviated associated inflammation but also mediated macrophage-mediated immunoregulation, thus resulting in effective osteogenesis. Background: Giant cell arteritis (GCA) is a prevalent artery and is strongly correlated with age. The role of CD4+ Memory T cells in giant cell arteritis has not been elucidated., Method: Through single-cell analysis, we focused on the CD4+ Memory T cells in giant cell arteritis. eQTL analysis and mendelian randomization analysis identified the significant genes which have a causal effect on giant cell arteritis risk. CD4+ Memory T cells were subsequently divided into gene-positive and gene-negative groups, then further single-cell analysis was conducted. Mendelian randomization of plasma proteins, blood-urine biomarkers and metabolites were also performed. Eventually, the PMA induced Jurkat cell lines were used for biological experiments to explore the specific functions of significant causal genes in CD4+ Memory T cells., Results: Similarity of CD4+ Memory T cells in GCA and old samples were explored. DDIT4 and ARHGAP15 were identified as significant risk genes via mendelian randomization. The CD4+ Memory T cells were then divided into DDIT4 ± or ARHGAP15 ± groups, and further single-cell analysis indicated the differences in aspects involving intercellular communication, functional pathways, protein activity, metabolism and drug sensitivity between positive and negative groups. In vitro experiments, including overexpression and knockdown, demonstrated that DDIT4 leading to a chronic, low-intensity inflammatory state in CD4+ Memory T cells, eventually promoting the development of GCA., Conclusion: DDIT4 and ARHGAP15 have significant causal effects on giant cell arteritis risk. To investigate the gas diffusion mechanism in coal particles under the coupled effects of gas pressure and temperature, a low-temperature liquid nitrogen adsorption experiment was first conducted to obtain the pore structure characteristics and pore size distribution patterns of the coal samples. Subsequently, a methane diffusion experiment was performed on coal particles with simultaneous increases in gas pressure and temperature using a unipore model to solve the diffusion coefficient. Finally, models for the diffusion coefficient of free-state methane and adsorbed-state methane were established, with their applicability discussed. The results indicate that the pore structure of the coal samples exhibits an approximately unimodal distribution, predominantly consisting of mesopores. As the gas pressure and temperature increase, the tortuosity of the coal sample pores also increases. Both the desorption volume and desorption rate of gas rise with the simultaneous increase in gas pressure and temperature, and overall, the diffusion coefficient of coal particles tends to increase under these conditions. Methane diffusion in coal particles is primarily governed by the surface diffusion of adsorbed methane, with gas pressure exerting a greater influence on surface diffusion than temperature. Cisplatin is a widely used anticancer drug, but its accumulation in renal tubular epithelial cells (TECs) can cause acute kidney injury. Phosphoseryl-tRNA kinase (PSTK) is an intermediate product produced under oxidative stress conditions. This study aimed to elucidate whether PSTK could protect TECs and its possible mechanisms. We found that PSTK levels decreased after cisplatin treatment, but PSTK overexpression using lentivirus vectors protected TEC viability. Overexpression of PSTK increased selenoprotein concentrations and reduced intracellular ROS levels. Additionally, PSTK overexpression inhibited the BAX/BCL2/Caspase 3 pathway after cisplatin stimulation, suggesting its potential role in preventing cell apoptosis. Taken together, this study suggests that PSTK could protect TEC viability from cisplatin-induced injury, possibly by inhibiting mitochondrial apoptosis. The complex interactions between parasites, their hosts, and associated microbiota hold significant implications for host health and disease outcomes. Helminths like Ascaris lumbricoides and Ascaris suum can significantly alter the host's intestinal microbiota, affecting both parasite biology and host pathology. Despite extensive research on host-microbiota changes due to helminth infections, the study of helminth-associated microbiota remains limited. This study aims to characterize the microbiota associated with Ascaris larvae and surrounding host tissues at distinct developmental stages (day 4, day 8, day 14), during larval migration through the liver, lungs, and intestine, and its impact on the host's microbiota in a murine model. Twenty mice were infected with 2500 embryonated A. suum eggs via oral gavage. Five Ascaris-infected mice and age-matched naïve mice were euthanized at 4-, 8-, and 14-days post-infection (DPI). Stool, intestine, liver, and lung samples were collected. Larvae were isolated from embryonated eggs in vitro, from the liver at 4 DPI, and the lung at 8 DPI. Utilizing 16S rRNA sequencing, we analyzed bacterial diversity in samples from different Ascaris stages and host tissues. Our results revealed a total of 8040 amplicon sequence variants (ASVs) with Ascaris samples displaying the highest diversity. Notably, Ascaris-larvae associated microbiota differed significantly from that of the host, with higher diversity observed in the parasite. Differential abundance analysis identified distinct taxonomic patterns, highlighting specific genera such as Bradyrhizobium, Achromobacter, and Pseudomonas in Ascaris. Our findings suggest that Ascaris harbors a unique microbiota that potentially exchanges bacteria with the host during larval migration. Background: This study aimed to investigate the optimal surgical techniques in reduction malarplasty by comparing the difference between the L-shaped osteotomy with vertical and oblique bone resection., Methods: A total of 120 patients who visited the authors' department for L-shaped osteotomy with either vertical (group I) or oblique bone resection (group II) from 2015 to 2021 were reviewed retrospectively. The preoperative and postoperative spiral computed tomographic data were analyzed. The preoperative computed tomographic data were also used to simulate the virtual and model surgery., Results: The results showed that a broken bridge-like structure with a bony gap of 2.86 ± 1.03 mm at the zygomatic arch root was observed in group I, and a mortice-and-tenon joint structure with a bone overlap of 2.28 ± 0.58 mm was formed in group II. The zygoma in group I displaced significantly in the vertical direction and horizontal direction during the follow-up. Simultaneously, the incidence of complications was higher in group I. In the mechanism analysis through virtual and model surgery, the zygomatic segment was shifted anteriorly and inferiorly in group I, and a bony gap was detected at the zygomatic arch root. Augmented reality for laparoscopic liver resection is a visualisation mode that allows a surgeon to localise tumours and vessels embedded within the liver by projecting them on top of a laparoscopic image. Preoperative 3D models extracted from Computed Tomography (CT) or Magnetic Resonance (MR) imaging data are registered to the intraoperative laparoscopic images during this process. Regarding 3D-2D fusion, most algorithms use anatomical landmarks to guide registration, such as the liver's inferior ridge, the falciform ligament, and the occluding contours. These are usually marked by hand in both the laparoscopic image and the 3D model, which is time-consuming and prone to error. Therefore, there is a need to automate this process so that augmented reality can be used effectively in the operating room. We present the Preoperative-to-Intraoperative Laparoscopic Fusion challenge (P2ILF), held during the Medical Image Computing and Computer Assisted Intervention (MICCAI 2022) conference, which investigates the possibilities of detecting these landmarks automatically and using them in registration. The challenge was divided into two tasks: (1) A 2D and 3D landmark segmentation task and (2) a 3D-2D registration task. The teams were provided with training data consisting of 167 laparoscopic images and 9 preoperative 3D models from 9 patients, with the corresponding 2D and 3D landmark annotations. A total of 6 teams from 4 countries participated in the challenge, whose results were assessed for each task independently. All the teams proposed deep learning-based methods for the 2D and 3D landmark segmentation tasks and differentiable rendering-based methods for the registration task. The proposed methods were evaluated on 16 test images and 2 preoperative 3D models from 2 patients. In Task 1, the teams were able to segment most of the 2D landmarks, while the 3D landmarks showed to be more challenging to segment. In Task 2, only one team obtained acceptable qualitative and quantitative registration results. Electrochemical CO 2 capture driven by renewable electricity holds significant potential for efficient decarbonization. However, the widespread adoption of this approach is currently limited by issues such as instability, discontinuity, high energy demand, and challenges in scaling up. In this study, we propose a scalable strategy that addresses these limitations by transforming the conventional single-step electrochemical redox reaction into a stepwise electrochemical-chemical redox process. Specifically, the hydrogen evolution reaction (HER) at the cathode and the oxidation of a redox carrier at the anode are employed to modulate the pH of the electrolyte, thereby facilitating effective CO 2 capture. By decoupling the electrochemical swing for CO 2 capture from redox carrier regeneration in both temporal and spatial domains, this approach mitigates unwanted side reactions and enhances system stability. Our results demonstrate a stable CO 2 capture process sustained for over 200 h, with a electrical work of 49.16 kJ e mol -1 CO 2 at a current density of 10 mA cm -2 . Furthermore, a scaled-up system capable of producing approximately 0.4 kg of pure CO 2 per day maintained stable operation for 72 h, highlighting the potential feasibility of this method for large-scale decarbonization applications., Competing Interests: Competing interests: The authors declare no competing interests., (© 2024. Background: Extracellular vesicles (EVs) play a crucial role in intraspecies and interspecies communication, significantly influencing physiological and pathological processes. Outer membrane vesicles (OMVs) secreted by Gram-negative bacteria are rich in components from the parent cells and are important for bacterial communication, immune evasion, and pathogenic mechanisms. However, the extraction and purification of OMVs face numerous challenges due to their small size and heterogeneity., Results: This study proposes an innovative strategy that combines traditional differential centrifugation (DC) with one-step ultracentrifugation (ODG) to develop a dual differential gradient centrifugation (DDGC) method for extracting outer membrane vesicles from Klebsiella pneumoniae. By comparing the DC and DDGC extraction methods, we found that OMVs extracted by DDGC exhibited more typical morphology, clearer backgrounds, and more uniform particle size distribution. The lipid polysaccharide (LPS) content in OMVs extracted by DDGC was significantly higher than that obtained by DC, and the outer membrane protein content was also greater, demonstrating enhanced biological activity. Biological activity assays indicated that OMVs extracted by DDGC showed stronger cytotoxicity to A549 lung epithelial cells, a significant decrease in cell viability, and higher levels of inflammatory factor expression(IL-6, TNF-α, IL-1β, and IL-8)., Conclusion: Our study demonstrates the advantages of the DDGC method in extracting K. pneumoniae OMVs, showing improvements in morphology, particle size distribution, protein content, and biological activity. Background: In a response to the lack of public awareness of dementia, 'ageing in place' and 'dementia-friendly community' policies have been proposed, and a number of relevant studies have been carried out., Aim: To map the evidence of public help for people with dementia in the community., Design: A scoping review., Methods: A comprehensive search of nine databases was conducted to find studies from the inception of the databases to January 2024. We developed our inclusion criteria to identify studies about public help for people with dementia in the community. Data from included studies were charted (purposively designed template) and narrated synthesis in relation to the study's research questions., Results: Of 34 studies included, we found that difficult situations were encountered by people with dementia after developing cognitive, behavioural and psychological symptoms. We identified three types of helping behaviour, namely, formal planned, informal planned and spontaneous helping behaviour, which were influenced by general demographic characteristics, knowledge, attitudes, responsibilities and understanding. Objective: This study utilized two-sample Mendelian randomization (TSMR) to investigate the bidirectional causal associations between temporomandibular disorders (TMD) and five mental disorders., Methods: Single-nucleotide polymorphisms (SNPs) linked to TMD were extracted from the Genome-Wide Association Studies (GWAS) database. The SNPs selected as instrumental variables (IVs) were required to have strong associations with the exposure phenotype and to meet the assumptions of Mendelian randomization (MR) analysis. The primary method employed for TSMR analysis was the Inverse Variance-Weighted (IVW) method. Additionally, the Weighted Median, Weighted Mode, and MR Egger methods were used as secondary approaches to ensure the reliability and credibility of the results. Furthermore, heterogeneity, pleiotropy, and sensitivity tests were conducted to ensure the accuracy and robustness of the findings. After MR analysis, we performed reverse analysis to explore the bidirectional association between TMD and five mental disorders., Results: Our study provided evidence of a positive correlation between TMD and both anxiety (P = 5.94 × 10 -15 , OR = 1.5253, 95 % CI = 1.3719-1.6959) and depression (P = 3.79 × 10 -12 , OR = 1.4243, 95 % CI = 1.2890-1.5738). In the reverse TSMR analysis, a positive association was found between anxiety and TMD (P = 0.0126, OR = 1.0664, 95 % CI = 1.0139-1.1217). Background and Objective: Breast-conserving surgery (BCS) plays a crucial role in breast cancer treatment, with a primary focus on ensuring cancer-free surgical margins, particularly for patients undergoing neoadjuvant treatment. After neoadjuvant treatment, tumor regression can complicate the differentiation between breast cancer and adjacent tissues. Raman spectroscopy, as a rapid and non-invasive optical technique, offers the advantage of providing detailed biochemical information and molecular signatures of internal molecular components in tissue samples. Despite its potential, there is currently no research on using label-free Raman spectroscopy to distinguish between breast cancer tumors and adjacent tissues after neoadjuvant treatment. This study intends to distinguish between cancer and adjacent tissues after neoadjuvant treatment in breast cancer through label-free Raman spectroscopy., Methods: In this study, the intraoperative frozen samples of breast cancer tumor and adjacent tissue were collected from patients who underwent neoadjuvant treatment during surgery. The samples were examined using Raman confocal microscopy, and Raman spectra were collected by LabSpec6 software. Spectra were preprocessed by Savitz-Golay filter, adaptive iterative reweighted penalized least squares and MinMax normalization method. The differences in Raman spectra between breast cancer tumor and adjacent tissues after neoadjuvant treatment were analyzed by Wilcoxon rank-sum test, with a Bonferroni correction for multiple comparisons. Based on the support vector machine (SVM) method in machine learning, a predictive model for classification was established in the total group and subgroups of different hormone receptor (HR) status, human epidermal growth factor receptor 2 (HER2) status and Ki-67 expression level. The independent test set was used to evaluate the performance of the model, and the area under curve (AUC) of the receiver operating characteristic (ROC) curve, sensitivity, specificity and accuracy of different models were obtained., Result: This study comprised 4260 Raman spectra of breast cancer tumor and adjacent frozen tissue samples from 142 breast cancer patients treated with neoadjuvant treatment. The Raman peaks associated with nucleotides and their metabolites in the Raman spectra of breast cancer tumor tissues were higher in intensities than those of adjacent tissues after neoadjuvant therapy (676 cm-1: Bonferroni adjusted P < 0.0001; 724 cm-1: P < 0.0001; 754 cm-1: P < 0.0001), and the Raman peaks from amide III bands were more intense (1271 cm-1: P < 0.01). Multivariate curve resolution- alternating least squares (MCR-ALS) decomposition of Raman spectra revealed reduced lipid content and increased collagen and nucleic acid content in breast cancer tumor tissues compared to adjacent tissues following neoadjuvant therapy. The predictive model based on the Raman spectral signature of breast cancer tumor and adjacent tissues after neoadjuvant treatment achieved an AUC of 0.98, with accuracy, sensitivity, and specificity values of 0.89, 0.97, and 0.83, respectively. The AUC of subgroup analysis according to different status of molecular pathological biomarkers was stably around 99%., Conclusion: This study demonstrated that label-free Raman spectroscopy can differentiate cancer and adjacent tissues of breast cancer patients treated with neoadjuvant therapy thorough getting the panoramic perspective of the biochemical compounds for the first time. Background: Early-onset ischemic heart disease (IHD) is a growing burden associated with high disability and death., Objectives: This study aimed to estimate the burden of incidence, prevalence, and disability-adjusted life years (DALY) of early-onset IHD from 1990 to 2019., Methods: Data on the burden of early-onset IHD (men<55 years, women<65 years), including prevalence, incidence, DALY, and deaths, were collected from the Global Burden of Disease study for 204 countries and territories from 1990 to 2019., Results: In 2019, early-onset IHD affected 5.34 million (95% uncertainty interval [UI]: 3.96-6.96) individuals globally. This resulted in 58.48 million (95% UI: 52.65-64.69) DALY and 1.44 million (95% UI: 1.29-1.59) deaths. Countries with a middle sociodemographic index (SDI) had the highest incidence cases (1.79 million, 95% UI: 1.32-2.34) and the low-middle SDI countries had the highest age-standardized incidence rate of 179.34 per 100,000 (95% UI: 134.38-231.94). Low-middle SDI countries exhibited the highest age-standardized death rate (57.13 per 100,000, 95% UI: 48.45-66.53) and age-standardized DALY rate (2,309.67 per 100,000, 95% UI: 1962.31-2,693.93). Globally, female incidence cases (2.77 million, 95% UI: 2.04-3.64) surpassed male (2.57 million, 95% UI: 1.92-3.32). Background: There is substantial epidemiological evidence demonstrating that influenza contributes to cardiovascular events in patients who already have cardiovascular diseases. However, the efficacy of influenza vaccination on the prognosis of patients with ischemic heart disease (IHD) is unclear., Methods: We conducted a systematic search for eligible randomized controlled trials (RCTs) in PubMed, Cochrane, and Embase on September 13, 2024, to investigate the effects of the influenza vaccine on the prognosis of patients with IHD. The effect sizes were combined using random-effects models, and Trial Sequential Analysis (TSA) was used to assess the reliability and validity of the results., Results: Five RCTs with a total of 5659 patients (median age ranging from 57.1 to 66 years, 67.8% male) with IHD were included. The use of influenza vaccine reduced the risk of major adverse cardiovascular events (risk ratio [RR] = 0.67, 95% confidence interval [CI] 0.52-0.87, number-needed-to-treat [NNT] of 37, high certainty), cardiovascular death (RR = 0.55, 95% CI 0.35-0.87, moderate certainty), all-cause mortality (RR = 0.58, 95% CI 0.40-0.84, high certainty) and myocardial infarction (MI) (RR = 0.66, 95% CI 0.46-0.93, high certainty) in patients with IHD compared with control. The analysis revealed no significant benefit regarding hospitalization for heart failure (HF) (RR = 0.91, 95% CI 0.21-3.99, moderate certainty) and revascularization (RR = 0.59, 95% CI 0.10-3.45, moderate certainty). The NNT to avoid 1 event was 37 for major adverse cardiovascular events, 56 for cardiovascular death, 67 for MI, and 41 for all-cause death. The early intrauterine environment of mothers during pregnancy significantly affects the metabolic health of their offspring. Existing studies suggest that poor maternal nutrition during pregnancy increases the risk of obesity or diabetes in offspring, so it is highly important to intervene during pregnancy to prevent metabolic disorders in mothers and their offspring. Polyphenols with anti-inflammatory and antioxidant properties are found in many foods and have protective effects on obesity, diabetes, cancer, and cardiovascular disease. Furthermore, recent evidence indicates that maternal dietary polyphenols could be a potential therapy for improving pregnancy outcomes and offspring metabolism. In this review, we discuss the studies and mechanisms of different kinds of maternal dietary polyphenols during pregnancy and lactation in improving the metabolism of offspring, analyze the limitations of the current studies, and propose possible directions of further research, which provide new ideas and directions for reducing metabolic diseases in offspring.

Background: This study evaluated the results of total hip arthroplasty (THA) via the direct anterior approach (DAA) in the lateral decubitus position combined with acetabular reconstruction via a bulk autograft for patients with developmental dysplasia of the hip (Crowe III and IV)., Patients and Methods: A retrospective study was conducted to analyse 16 patients (16 hips) with Crowe type III and IV developmental dysplasia of the hip (DDH) who underwent THA from June 2016 to July 2021. We recorded and analysed variables such as age, sex, height, body mass index (BMI), length of surgical incision, duration of surgery, intraoperative blood loss, Harris hip score, visual analogue scale (VAS) score, lower limb length, and pelvic radiograph analysis., Results: The mean age of the patients was 59.3 years, the mean BMI was 22.8 kg/m 2 , and the Crowe classification was type III in 13 hips and type IV in 3 hips. Sixteen patients were followed up with a mean follow-up time of 56.6 (32-92) months. The Harris hip score increased from 48.6 ± 7.5 points preoperatively to 89.6 ± 4.4 points at the final follow-up (t = -18.8, P < 0.001).The VAS score for activity decreased from 7.3 ± 0.8 points preoperatively to 1.0 ± 0.7 points at the final follow-up, which was a statistically significant difference compared with the preoperative score (t = 22.2, P < 0.001). The difference in the length of both lower limbs decreased from 3.0 ± 0.7 cm preoperatively to 0.9 ± 0.4 cm at the final follow-up, and the difference was statistically significant compared with the preoperative difference (t = 10.8, P < 0.001). The acetabular cup coverage was satisfactory., Conclusions: Acetabular structural bone grafting THA with lateral decubitus DAA was used to treat Crowe type III and IV DDH with satisfactory efficacy in the medium term, but the learning curve was long. Background: Despite decades of educational efforts, patients with acute ischemic stroke (AIS) remain delayed in seeking medical care, which becomes the greatest obstacle to the successful management of the condition., Objective: The objective of this study was to systematically explore the incidence and influencing factors of prehospital care-seeking delay in AIS patients., Methods: We systematically searched the PubMed, Embase, Cochrane Library, Web of Science, and Cumulative Index to Nursing and Allied Health Literature from database inception to September 30, 2023. Meta-analysis was conducted using the Stata 15.0 software package. The pooled incidence was calculated using a random-effects model. The quality of studies reporting incidence data was assessed using Joanna Briggs Institute's Critical Appraisal Checklist and Newcastle-Ottawa Scale. Subgroup analyses were performed according to study location, country income, recruitment date, and age., Results: Finally, 30 related articles were included, involving a total of 287,102 people. The estimated incidence of prehospital care-seeking delay was 68%, and there were differences in this incidence in different countries (p = 0.035). Meta-analysis results showed that the delay rate was highest in low-income countries (85%) and lowest in high-income countries (62%). Patients who live farther from hospitals, have a lower level of education, diabetes, hyperlipidemia, or a history of stroke are more likely to experience delays (all p < 0.05). Conversely, those who can recognize stroke symptom, perceive the severity of early symptom, understand thrombolysis treatment, atrial fibrillation, consciousness disturbance, visual disturbance, and symptom score at admission, emergency medical service use, and immediate help-seeking have a lower risk of delay (all p < 0.05)., Conclusion: Prehospital care-seeking delays are common among patients with AIS, especially in low-income countries. Background: The incidence, mortality, and readmission rates for acute heart failure (AHF) are high, and the in-hospital mortality for AHF patients in the intensive care unit (ICU) is higher. However, there is currently no method to accurately predict the mortality of AHF patients., Methods: The Medical Information Mart for Intensive Care Ⅳ (MIMIC-Ⅳ) database was used to perform a retrospective study. Patients meeting the inclusion criteria were identified from the MIMIC-Ⅳ database and randomly divided into a training set (n = 3,580, 70%) and a validation set (n = 1,534, 30%). The variates collected include demographic data, vital signs, comorbidities, laboratory test results, and treatment information within 24 hours of ICU admission. By using the least absolute shrinkage and selection operator (LASSO) regression model in the training set, variates that affect the in-hospital mortality of AHF patients were screened. Subsequently, in the training set, five common machine learning (ML) algorithms were applied to construct models using variates selected by LASSO to predict the in-hospital mortality of AHF patients. The predictive ability of the models was evaluated for sensitivity, specificity, accuracy, the area under the curve of receiver operating characteristics, and clinical net benefit in the validation set. To obtain a model with the best predictive ability, the predictive ability of common scoring systems was compared with the best ML model., Results: Among the 5,114 patients, in-hospital mortality was 12.5%. Comparing the area under the curve, the XGBoost model had the best predictive ability among all ML models, and the XGBoost model was chosen as the final model for its higher net benefit. Background: China has the highest number of people with dementia globally, and the responsibility of caring for people with dementia primarily falls on relatives, who bear heavy caregiving burdens and pressure. Providing care for an individual with dementia is emotionally and physically demanding, particularly due to the frequent manifestation of behavioral and psychological symptoms associated with dementia (BPSD). This underscores the crucial need to comprehend and address caregivers' emotional expression (EE)., Aim: To explore the characteristics of relatives with high expressed emotion of people with dementia and related factors in mainland China., Methods: A survey using cross-sectional questionnaires conducted with 165 relatives of individuals with dementia in China., Results: A significant number of relatives had high EE (n = 61, 39%). The variation in EE, about 37.8%, is explained by seven independent variables. The proportion of psychological distress among relatives in EE variation is 14.5%. (b = 0.387, p < 0.001). Length of care-taking, active coping, and chronic diseases accounted for 6.5% (b = 0.264, p < 0.001), 5.1% (b=-0.239, p = 0.001) and 4.1% (b = 0.211, p = 0.002) of the variance in EE, respectively., Conclusions: Chinese health care providers can identify high-risk groups for assistance based on the severity of dementia, age, health status and duration of care. Objective: Assess the safety and effectiveness of acupuncture for acute ischemic stroke (AIS)., Methods: We conducted a comprehensive search across the PubMed, Cochrane Library, Embase, Web of Science, China National Knowledge Infrastructure (CNKI), China Science and Technology Journal Database (VIP), Wanfang, and SinoMed databases from their inception until October 3, 2023. Two reviewers screened eligible randomized controlled trials (RCTs) according to criteria and extracted data using a pre-established form. Cochrane tool was used for risk of bias assessment, and Revman 5.3 was used for subsequent meta-analysis. The GRADE tool will be used to assess the quality of evidence., Results: Thirty-one RCTs were included, involving 3604 patients. Meta-analysis showed that compared with conventional treatments (CTs), acupuncture combined with CTs could improve in National Institutes of Health Stroke Scale (NIHSS) and Efficiency (MD:1.70, 95 %CI [-2.27, -1.14], P < 0.00001;RR: 1.21, 95 %CI [1.12, 1.31], P < 0.00001,);On the Chinese Stroke Scale (CSS) and effectiveness based on CSS, acupuncture showed positive effects (MD:4.33, 95 %CI [-5.67, -2.98], P < 0.00001; RR: 1.26, 95 %CI [1.13, 1.41], P < 0.0001). Furthermore, ADL, Fugl-Meyer Assessment Scale (FMA), prognosis analyses also showed the effectiveness of acupuncture. Background and Purpose: Atherosclerosis is the basis of cardiovascular disease. Ferroptosis is a form of programmed cell death characterized by lipid peroxidation, which contributes to atherogenesis. The plant extract PNS (Panax notoginseng saponins), containing the main active ingredients of Panax notoginseng, exhibits anti-atherogenic properties. Herein, we determined whether PNS and its major components could attenuate atherosclerosis by suppressing ferroptosis and revealed the underlying mechanism(s)., Experimental Approach: The anti-atherogenic effects of PNS and their association with inhibition of ferroptosis was determined in apoE -/- mice. In vitro, the anti-ferroptotic effect and mechanism(s) of PNS components were demonstrated in the presence of ferroptosis inducers. Expression of ferroptosis markers and the ubiquitination of Keap1 were evaluated in USP2 -/- macrophages. Finally, the anti-atherogenic effect of USP2 knockout was determined by using USP2 -/- mice treated with high-fat diet (HFD) and AAV-PCSK9., Key Results: PNS inhibited ferroptosis and atherosclerosis in vivo. PNS suppressed ferroptosis and ferroptosis-aggravated foam cell formation and inflammation in vitro. Mechanistically, PNS and its components activated Nrf2 by antagonizing Keap1, which was attributed to the inhibition of USP2 expression. USP2 knockout antagonized ferroptosis and ferroptosis-aggravated foam cell formation and inflammation, thus mitigating atherosclerosis. USP2 knockout abolished inhibitory effects of PNS on foam cell formation and inflammation in vitro., Conclusion and Implications: PNS reduced USP2-mediated Keap1 de-ubiquitination and promoted Keap1 degradation, thereby activating Nrf2, improving iron metabolism and reducing lipid peroxidation, thus contributing to an anti-atherosclerotic outcome. Compared with rigid or elastic microspheres, the synthetic viscoelastic T-cell-activating cells (SynVACs) led to substantial enhancements in the expansion of human CD8 + T cells and to the suppression of the formation of regulatory T cells. Notably, activating and expanding chimaeric antigen receptor (CAR) T cells with SynVACs led to a CAR-transduction efficiency of approximately 90% and to substantial increases in T memory stem cells. The engineered CAR T cells eliminated tumour cells in a mouse model of human lymphoma, suppressed tumour growth in mice with human ovarian cancer xenografts, persisted for longer periods and reduced tumour-recurrence risk. Background: Recent individual studies have indicated that ultra-processed food (UPF) consumption may be associated with the incidence of chronic kidney disease (CKD). We conducted a systematic review and meta-analysis based on those longitudinal studies evaluating the relationship between UPF consumption and the risk of incident CKD, and synthesizing the results., Method: PubMed, Embase, The Cochrane Library, Web of Science, and Scopus were searched from inception through 22 March 2023. Any longitudinal studies evaluating the relationship between UPF consumption and the risk of incident CKD were included. Two researchers independently conducted the literature screening and data extraction. RR and its 95% CI were regarded as the effect size. The Newcastle-Ottawa Scale (NOS) was applied to assess the quality of the studies included, and the effect of UPF consumption on the risk of incident CKD was analyzed with STATA version 15.1. This study's protocol was registered in PROSPERO (CRD42023411951)., Results: Four cohort studies with a total of 219,132 participants were included after screening. The results of the meta-analysis suggested that the highest UPF intake was associated with an increased risk of incident CKD (RR = 1.25; 95% CI: 1.18-1.33)., Conclusions: High-dose UPF intake was associated with an increased risk of incident CKD. However, the underlying mechanisms remain unknown. Thus, more standardized clinical studies and further exploration of the mechanisms are needed in the future.

Engineered transcription factors (eTFs) binding diversed functional nucleic acids (dFNAs), as innovative biorecognition systems, have gradually become indispensable core elements for building synthetic biosensors. They not only circumvent the limitations of the original TF-based biosensing technologies, but also inject new vitality into the field of synthetic biosensing. This review aims to provide the first comprehensive and systematic dissection of the eTF-dFNA synthetic biosensor concept. Firstly, the core principles and interaction mechanisms of eTF-dFNA biosensors are clarified. Next, we elaborate on the construction strategies of eTF-dFNA synthetic biosensors, detailing methods for the personalized customization of eTFs (irrational design, rational design, and semi-rational design) and dFNAs (SELEX, modifying and predicting), along with the exploration of strategies for the flexible selection of signal amplification and output modes. Furthermore, we discuss the exceptional performance and substantial advantages of eTF-dFNA synthetic biosensors, analyzing them from four perspectives: recognition domain, detection speed, sensitivity, and construction methodology. Building upon this analysis, we present their outstanding applications in point-of-care diagnostics, food-safety detection, environmental monitoring, and production control. In recent years, substantial advancements have been achieved in understanding the diversity of the human virome and its intricate roles in human health and diseases. Despite this progress, the field of human virome research remains nascent, primarily hindered by the lack of effective methods, particularly in the domain of computational tools. This perspective systematically outlines ten computational challenges spanning various types of virome studies. These challenges arise due to the vast diversity of viromes, the absence of a universal marker gene in viral genomes, the low abundance of virus populations, the remote or minimal homology of viral proteins to known proteins, and the highly dynamic and heterogeneous nature of viromes. For each computational challenge, we discuss the underlying reasons, current research progress, and potential solutions. The resolution of these challenges necessitates ongoing collaboration among computational scientists, virologists, and multidisciplinary experts. Visualizing RNA is critical for understanding RNA expression patterns and spatial organization within cells, offering valuable insights into gene regulation and cellular functions. High-resolution RNA imaging techniques are therefore indispensable for revealing the complexities of cellular pathways and physiological processes. Traditional RNA imaging methods, however, face significant limitations, such as high background noise resulting from labeling or cell fixation, which can impede the accurate tracking of RNA dynamics in live cells. Fluorescent light-up RNA aptamers (FLAPs) have emerged as a powerful alternative, distinguished by their capacity for enhanced fluorescence activation, reduced background interference, and advantages such as label-free imaging, small molecular size, and customizable structures. In this review, we provide an overview of the development of FLAPs, explore recent advancements in FLAP-based RNA imaging strategies, and discuss both the challenges and future directions in the field. This study introduces a novel method that integrates artificial neural networks (ANNs) with the Particle Swarm Optimization (PSO) algorithm to enhance the efficiency and precision of parameter optimization for the small-signal equivalent model of dual-field-plate GaN HEMT devices. We initially train an ANN model to predict the S-parameters of the device, and subsequently utilize the PSO algorithm for parameter optimization. Comparative analysis with the NSGA2 and DE algorithms, based on convergence speed and accuracy, underscores the superiority of the PSO algorithm. Ultimately, this ANN-PSO approach is employed to automatically optimize the internal parameters of a 4 × 250 μm dual-field-plate GaN HEMT equivalent circuit model within the frequency range of 1-18 GHz. The method's effectiveness under varying bias conditions is validated through comparison with traditional physical formula analysis methods. The results demonstrate that the ANN-PSO method significantly enhances the automation and efficiency of parameter optimization while maintaining model accuracy, providing a reference for the optimization of other device models.

This study introduces a novel approach of precisely depositing In 2 O 3 nanoislands on the edges of BN sheets using selective atomic layer deposition. The modified catalysts demonstrated a significant improvement and characterization results revealed that the presence of In 2 O 3 nanoislands, when free of BO x overlayers, is crucial in mitigating side reactions.

Gestational diabetes mellitus (GDM) is a common complication of pregnancy that has short- and long-term adverse effects. Therefore, further exploration of the pathophysiology of GDM and related biomarkers is important. In this study, we performed a systematic review and meta-analysis to investigate the associations between metabolites in blood detected via metabolomics techniques and the risk of GDM and to identify possible biomarkers for predicting the occurrence of GDM. We retrieved case‒control and cohort studies of metabolomics and GDM published in PubMed, Embase, and Web of Science through March 29, 2024; extracted metabolite concentrations, odds ratios (ORs), or relative risks (RRs); and evaluated the integrated results with metabolites per-SD risk estimates and 95% CIs for GDM. We estimated the results via the random effects model and the inverse variance method. Our study is registered in PROSPERO (CRD42024539435). We included a total of 28 case‒control and cohort studies, including 17,370 subjects (4,372 GDM patients and 12,998 non-GDM subjects), and meta-analyzed 67 metabolites. Twenty-five of these metabolites were associated with GDM risk. Some amino acids (isoleucine, leucine, valine, alanine, aspartate, etc.), lipids (C16:0, C18:1n-9, C18:1n-7, lysophosphatidylcholine (LPC) (16:0), LPC (18:0), and palmitoylcarnitine), and carbohydrates and energy metabolites (glucose, pyruvate, lactate, 2-hydroxybutyrate, 3-hydroxybutyrate) were discovered to be associated with increased GDM risk (hazard ratio 1.06-2.77). Glutamine, histidine, C14:0, and sphingomyelin (SM) (34:1) were associated with lower GDM risk (hazard ratio 0.75-0.84). Previous research has established that the formation of Gardnerella vaginalis (GV) biofilm is one of the primary reasons for bacterial vaginosis (BV) recurrence. This study was the first to explore the impact of Streptococcus agalactiae (group B Streptococcus, GBS) on GV biofilm in a co-culture scenario. The results revealed that GBS could significantly increased the GV biomass in 48-hours dual-species biofilms. The luxS gene of GBS was significantly higher in dual-species biofilm, while knockdown of the luxS gene resulted in a significant decrease in mono- and dual-species biofilms. Meanwhile, in vitro addition of AI-2 (product of luxS gene) substantially increased biofilm biomass. Furthermore, we found that the expression of two genes related to biofilm formation was notably elevated in GV after receiving AI-2 signals. To evaluate the differences in the agreement between wedged hepatic venous pressure (WHVP) and portal venous pressure (PVP) at different hepatic venous pressure gradient (HVPG) levels to identify specific HVPG thresholds where WHVP can reliably estimate PVP, thus enhancing the accuracy of risk stratification and treatment decision-making for portal hypertension (PHT) patients. A multicenter study of 616 patients with PHT from three centers was stratified into five groups by their HVPG: HVPG < 12 (group A), 12 ≤ HVPG < 16 mmHg (group B), 16 ≤ HVPG < 20 mmHg (group C), 20 ≤ HVPG < 24 mmHg (group D), HVPG ≥ 24 mmHg (group E). Concordance was analyzed using Pearson's correlation coefficient (R), the intraclass correlation coefficient (ICC), and Bland‒Altman analysis in each HVPG stratum. Correlation and agreement between WHVP and PVP varied by HVPG group. Highest agreement was observed in the range of 20 ≤ HVPG < 24 mmHg. (R = 0.55, ICC = 0.68). The proportion of patients with a discrepancy between WHVP and PVP that was greater than 10% of the PVP value was highest in group A (95.7%) and lowest in group D (48.4%). Overestimation of PVP was more common in group E (44.5%), and underestimation of PVP was more common in group A (94.6%). This study does not confirm the usefulness of hepatic vein pressure measurements to predict the PVP and PPG. A concise synthesis of (-)-quinocarcinol methyl ester was accomplished with an overall yield of 39% through a 9-step longest linear sequence (LLS). Our synthesis features a two-step ester reduction/reductive amination sequence, a stereoselective [3 + 2] intramolecular cross-cycloaddition for the construction of bicyclo[3.2.1]octane skeletons, four simultaneous hydrogenolysis reactions in a one-pot process, and a stereoselective Krapcho decarboxylation. By following this protocol, (-)-oxa-quinocarcinol methyl ester was also achieved.

This study aimed to investigate the benefits of using three-dimensional (3D)-printed hip joint fixators after intra-arterial chemotherapy (IAC) by inguinal femoral artery puncture in children with retinoblastoma. Overall, 79 cases of retinoblastoma who had undergone IAC through the femoral artery were selected and divided into an observation group of 50 cases and an intervention group of 29 cases according to the hemostasis method employed. The patients in the observation group were treated with sandbags for hemostasis, while those in the intervention group were given 3D-printed hip joint fixators to help immobilize the hips and sandbags. We used the Face, Legs, Activity, Cry and Consolability scale (FLACC), the Wong-Baker Facial Expression Pain Scale, and self-made questionnaires to evaluate demographics, clinical characteristics, pain, complications, satisfaction, and other indicators of the two groups. There were no significant differences in general data, such as age, gender, height, weight, manual compression time, diseased eye, tumor stage, platelet count, puncture times, pain distribution, and total score, between the groups. There was a positive correlation between FLACC pain and the total Wong-Baker pain score (r = 0.599, p < 0.001). During the 2 h of sandbag compression, sandbags were dislodged in the observation group as many as ten times, which was significantly higher than that in the intervention group (up to four times; p < 0.001). This was correlated with a very high score of satisfaction (92.34 ± 19.96 out of 100). The 3D-printed hip fixator is easy to operate, has a low incidence of complications, and saves time and effort. It effectively reduces the incidence of sandbags falling off after IAC in children with retinoblastoma and does not increase the patient's pain. The contradiction of near-infrared II (NIR-II) emission and photothermal effects limits the development of phototheranostic agents (PTAs) in many emerging cutting-edge applications. Organic aggregates present a promising opportunity for the balance of competitive relaxation processes through the manipulation of molecular structure and packing. Herein, side chain phenyl isomerization-induced spatial conjugation was proposed for constructing A-D-A type NIR-II PTAs with simultaneous enhancement of fluorescence brightness and photothermal properties. Three pairs of mutually isomeric fluorophores, whose phenyls respectively located at the outside (o-series) and inside (i-series) of the side chain, were designed and synthesized. The positional isomerization of the phenyl endows the o-series crystals with strong spatial conjugation between the phenyl group on the side chain and the backbone, as well as interlocked planar network, which is different to that observed in the i-series. Thus, all o-series nanoparticles (NPs) exhibit red-shifted absorption, enhanced NIR-II emission, and superior photothermal properties than their i-series counterparts. A prominent member of the o-series, o-ITNP NPs, demonstrated efficacy in facilitating NIR-II angiography, tumor localization, and NIR-II imaging-guided tumor photothermal therapy. Oct4 is a master regulator of pluripotency. Potential Oct4 interactors have been cataloged extensively but the manner and significance of these interactions are incompletely defined. Like other POU domain proteins, Oct4 is capable of binding to DNA in multiple configurations, however the relationship between these configurations and cofactor recruitment (and hence transcription output) are unknown. Here, we show that Oct4 interacts with common and unique proteins when bound to DNA in different configurations. One of these proteins is Jade1, a component of the HBO histone acetyltransferase complex. Jade1 preferentially associates with Oct4 when bound to M ore palindromic O ctamer- R elated E lement (MORE) DNA sequences that bind Oct4 dimers and are associated with strong gene expression. Surprisingly, we find that the Oct4 N-terminal activation domain, rather than facilitating Jade1 binding, serves as an autoinhibitory domain that dampens the interaction. ChIP-seq using HBO1, the enzymatic component of the complex, identifies a preference for binding adjacent to Oct4 at MORE sites. Using purified recombinant proteins and nucleosome complexes, we show that the HBO1 complex acetylates histone H3K9 within nucleosomes more efficiently when Oct4 is co-bound to a MORE site. Histone acetylation is further increased when Oct4 is mutated to favor dimeric MORE binding. Cryo-electron microscopy reveals that Oct4 bound to a MORE near the nucleosome entry/exit site partially unwinds DNA from nucleosome core particles, and identifies additional mass associated with the HBO1 complex. These results identify a novel mechanism of transcriptional regulation by Oct4.

Introduction: GBS (group B streptococcus) is an opportunistic pathogen that can colonize healthy individuals but presents significant challenges in clinical obstetrics and gynecology, as it can cause miscarriage, preterm birth, and invasive infections in newborns. To develop specific and personalized preventative strategies, a better understanding of the epidemiological characteristics and pathogenic features of GBS is essential., Methods: We conducted a comprehensive strain-level genomic analysis of GBS, examining serotype and genotype distributions, as well as the composition and correlations of virulence genes using the blastn-short mode of the BLAST program(v2.10.0+), mlstsoftware (https://github.com/tseemann/mlst), Snippy (v4.6.0), FastTree (v2.1.11) and iTOL. The coding sequence region of virulence factors was annotated by Prodigal (v2.6.3) and Glimmer(v3.02b). We further identified host protein interacting with Srr2 by mass spectrometry analysis., Results: While certain genotypes showed strong serotype consistency, there was no significant association between overall serotypes and genotypes. However, the composition of virulence genes was more closely related to the phylogeny of GBS, among which simultaneous presence of Srr2 and HygA exhibit significant association with hypervirulence. Tubulin emerged as the most distinct and abundant hit. The specific interaction of Tubulin with Srr2-BR, rather than Srr1-BR, was further confirmed by immunoblotting., Discussion: Considering the impact of cytoskeleton rearrangement on GBS pathogenesis, this observation offers a plausible explanation for the hypervirulence triggered by Srr2. Background: Stress urinary incontinence (SUI), the most common form of urinary incontinence, is a condition that affects many women. It is characterized by involuntary urine leakage during activities that increase abdominal pressure, such as sneezing, coughing, or physical exertion, according to the International Continence Society. SUI affects patients' quality of life and causes depression and emotional disorders, which negatively influences physical and mental health. The participants in the studies in this review comprised women with mild to moderate SUI, because there are more female patients than male patients, and most patients with severe SUI are treated surgically. Moreover, after retrieval, there were no systematic reviews or network meta-analyses (NMAs) of conservative treatments, such as electroacupuncture (EA), in women with mild to moderate SUI., Objective: This study aims to investigate the efficacy of electroacupuncture among women with mild to moderate SUI using an NMA., Methods: Randomized clinical trials related to conservative treatments for SUI will be searched in 5 English and 3 Chinese literature databases: EMBASE, PubMed, Cochrane, Web of Science, ClinicalTrials.gov, Chinese National Knowledge Infrastructure (CNKI), WanFang, and the Chinese BioMedical Literature Database. The search period for these 8 electronic databases will be from 2002 to 2022. The PROSPERO database and the International Platform of Registered Systematic Review and Meta-Analysis Protocols (INPLASY) database will also be searched. Two reviewers will independently complete the research selection. After screening the studies, 2 other researchers will extract the data, and the quality of the included studies will be evaluated according to the quality standards specified in the Cochrane Collaboration Tool (version 2). The primary outcomes will be the change in urine leakage determined by a 1-hour pad test and International Consultation on Incontinence Questionnaire Short Form (ICIQ-SF) scores at baseline and at the conclusion of the follow-up. The secondary outcomes will be 72-hour incontinence episodes, residual bladder volume, effective rate, urodynamic indexes, and other reported measurements. Stata (version 14.0; StataCorp) and Review Manager (RevMan version 5.3; Cochrane) will be implemented for data synthesis and meta-analysis., Results: The results are not yet accessible because this is a protocol for a systematic review and meta-analysis. Aims: Tumorigenicity is a significant concern in stem cell-based therapies. However, traditional tumorigenicity tests using animal models often produce inaccurate results. Consequently, a more sensitive method for assessing tumorigenicity is required. This study aimed to enhance sensitivity by exposing functional progenitors derived from human pluripotent stem cells (hPSCs) to the tumor microenvironment (TME) in vitro before transplantation, potentially making them more prone to abnormal proliferation or tumorigenicity., Methods: Midbrain dopamine (mDA) cells derived from hPSCs were exposed to the TME by coculturing with medulloblastoma. The cellular characteristics of these cocultured mDA cells were evaluated both in vitro and in vivo, and the mechanisms underlying the observed alterations were investigated., Results: Our findings demonstrated increased proliferation of cocultured mDA cells both in vitro and in vivo. Moreover, these proliferating cells showed a higher expression of Ki67 and SOX1, suggesting abnormal proliferation. MicroRNAs (miRNAs) control 60% of genes expressed in the human body, but their role in malaria pathogenesis is incompletely understood. Here, we demonstrate cell type-specific alterations to the miRNA profiles during the early response to malaria infection in brain and lung endothelial cells (ECs). In brain ECs, incubation with Plasmodium falciparum -infected red blood cells in the ring stage (iRBCs) most significantly affected endocytosis-related miRNAs and mRNAs. Contrastingly, in lung ECs, iRBCs altered electron transport chain-related miRNAs and mRNAs. We present a dataset of inherent differences between microRNA profiles in brain and lung ECs and their extracellular vesicles (EVs). We demonstrated that shear stress affected multiple pathways in brain ECs, which were controlled by numerous human miRNAs. Together, these findings indicate that host miRNAs respond to parasite exposure, accompanied by stimulation of downstream signaling pathways within the ECs. This study investigated the expression of Zip6, a gene predominantly located in the placenta, breast, and prostate tissues, in patients with Kashin-Beck disease (KBD). Environmental risk factor models for KBD were developed using low selenium (Se) feeding (with a Se content of 0.02 mg Se/kg in the feed) and exposure to T-2 toxin (200 ng/g*BW/D). Additionally, the study examined the alterations in Se and Zn 2+ levels, along with the mRNA and protein expression levels of Zip6 and KBD related genes, including Mtf1, Mmp3, Mmp13, Adamts5, and Col2a1. Differentially expressed genes (DEGs) were examined by transcriptome sequencing to elucidate the mechanism by which Zip6 induces metabolic disorder of the extracellular matrix (ECM), subsequently leading to cartilage injury under the influence of Se deficiency and T-2 toxin. The findings indicated that the expression levels of Zip6 in adult and pediatric KBD chondrocytes were not synchronized. In the animal study, there was a notable increase in the Zn 2+ level in the comprehensive exposure (CE) group. Moreover, in both the T-2 exposure (T-2) and CE groups, there was a significant decrease in the expression of Zip6 in each zone, and the expression of Adamts5 in the middle zone exhibited a significant increase (P < 0.05) correlating with varying degrees of cartilage tissue damage in each group. Sequencing results revealed that the significantly up-regulated DEGs in the CE group included Zimz2. This study suggested that Se and T-2 toxin may influence the expression of Zip6, and it investigated the role of Zn 2+ in the pathogenesis of KBD, thereby providing a novel scientific foundation for understanding the pathogenesis of KBD., (© 2024.
