Background: The present study aimed to investigate possible influences of body weight and sex on adrenal gland size in endocrinologically healthy dogs. Possible factors influencing the adrenal size are discussed in relation to a universal upper reference value from the literature of 7.4 mm as the thickness in the caudal pole of the adrenal gland. The adrenal size was measured by computed tomography (CT) from 66 normal dogs of six different breeds (Labrador Retriever (n = 16), German Shepherd (n = 10), Boxer (n = 8), Beagle (n = 14), Dachshund (n = 6) and Jack Russell Terrier (n = 12); male n = 38 (thereof neutered n = 23), female n = 28 (thereof neutered n = 17)) based on volume quantification and linear measurements using the data processing software Amira. For interbreed comparability, a ratio consisting of the third root of adrenal volume to aortic diameter (Ratio volume-aorta, RVA) was introduced. Additionally, breed-related attenuation values in contrast-enhanced CT data sets were measured., Results: The measured volumes ranged from 0.34 to 1.93 cm 3 for the right and from 0.39 to 2.23 cm 3 for the left adrenal gland. The present study was able to demonstrate a body weight effect on the adrenal volume as well as on length and height. In terms of adrenal size, no significant differences between male and female, nor between intact and neutered dogs were obtained due to the RVA. In addition, for the weight classes, a breed independent threshold for dogs less (left 1.4; right gland 1.5) or more than 20 kg body weight (left 1.1; right gland 1.2) based on RVA was defined. Breed-related significant differences with respect to attenuation were determined only for the left adrenal gland, with lower attenuation values in large dog breeds., Conclusion: The present study points out the importance of weight-related data when assessing CT data of the canine adrenal gland regarding volume, size and attenuation. The use of a universal reference value for the assessment of adrenal size appears unsuitable considering weight-related volume and linear measurements. Objective: Aim was to establish an individually adapted endurance test for dogs on a treadmill, which takes the individual's physical condition into account. To check the applicability of the test, two age groups of clinically healthy beagles were examined., Methods: A total of 10 clinically healthy Beagles were enrolled and divided in a younger (1-3 years, n = 5) and older group (> 8 years, n = 5). The individual comfort gait speed of each dog was determined on a treadmill with integrated force plates. A maximal time of 20 minutes at trot was set for the endurance test. The test was terminated prematurely if the dog showed signs of fatigue (massive panting, unwillingness to move further). Blood samples were taken at general examination (G), prior to (B) and post exercise (P) for determination of lactate level (LL), oxygen and carbon dioxide partial pressure (pO2, pCO 2 ), bicarbonate (HCO 3 ), base excess (BE) and pH. On each occasion (G, B, P) heart rate (HR) and respiratory rate (RR) were recorded. Additionally, vertical ground reaction forces (Fz) were analysed.- ), base excess (BE) and pH. Additionally, vertical ground reaction forces (Fz) were analysed., Results: The older dogs (age: 10.4 ± 0.89 years) completed the test with less speed and duration compared to the younger dogs (age: 2.4 ± 0.89 years), which managed to complete the maximum time. Lactate levels in the older dogs were higher than in the younger dogs at all timepoints of examination. Contrary to the younger dogs, there was no significant increase in the heart rate of the older dogs. Ground reaction forces were not significantly different between the groups., Conclusion and Clinical Relevance: Whereas standardised endurance tests allow for the comparison of fitness levels between dogs, an individually adjusted endurance test aims at objectively determining the physical fitness of the single dog taking into account its individual performance. Such a test allows to examine the individual performance development over time and to evaluate medicinal therapies or dietary measures, e. g. in aging dogs. Background: Aim was to investigate age-dependent changes in the prostate of castrated dogs in computed tomographic (CT) examination. Thirty-six canine prostates were evaluated in pre- and post-contrast CT scans. Dogs were divided in groups with homogenous prostatic tissue (25/36) and with tissue alterations (11/36). Prostatic attenuation in Hounsfield Units (HU) and prostatic size were measured and a ratio of the prostatic size to the sixth lumbar vertebra was calculated. Additionally, the CT images of the prostate were compared with ultrasound examination., Results: In pre-contrast CT scans no significant differences were found in prostatic size between homogenous and altered prostatic tissue groups whereas prostatic attenuation differed significantly in post-contrast CT between these groups. The homogenous tissue pattern of homogeneous prostates could be confirmed in CT images and in ultrasound examination. Concerning prostates with alterations, the results differed between ultrasound and CT examination in four cases of 11 dogs with tissue alterations., Conclusions: CT is beneficial to examine the prostate of castrated dogs. The prostatic attenuation is characteristic for the prostatic morphology, which can vary due to ageing processes. Differences in attenuation and size can be found between prostates of castrated and intact dogs. Using contrast agent, CT can visualize prostatic alterations, which were not seen in ultrasound. The presented results should be considered preliminary until a study with larger sample size and histologic examination of the prostates is performed.

In dogs, decreasing telomere length is a biomarker for cellular aging. On a systemic level, aging affects the locomotor system in particular, leading to restricted joint mobility. As aging is thought to be related to oxidative stress, it may be counteracted by a diet enriched with antioxidants, mitochondrial cofactors and omega-3 fatty acids. This randomized, blinded and placebo-controlled study examined the influence of an accordingly enriched diet compared to a control diet on 36 young and 38 old shepherd dogs. At the outset, after 3 and after 6 months, mean and minimum telomere lengths were measured. Furthermore, minimum and maximum joint angles and range of motion of the shoulder, elbow, carpal, hip, stifle and tarsal joints were measured by computer-assisted gait analysis. A positive influence of the enriched diet on old dogs could be verified for minimum telomere length and all three parameters of the shoulder joint on the side with the higher vertical ground reaction force after 6 months. In the other joints there were less significant differences; in some cases they indicated a contrary influence of the enriched diet on young dogs, probably due to its reduced protein content. The greater effect of the enriched diet on minimum than on mean telomere length may be due to the higher preference of telomerase for short telomeres. The greater effect on shoulder joint mobility is explained by the greater influence of musculature and connective tissue in this joint. Background: Since most prostatic diseases are associated with the organ's enlargement, evaluation of prostatic size is a main criterion in the diagnosis of prostatic state of health. While enlargement is a non-uniform process, volumetric measurements are believed to be advantageous to any single dimensional parameter for the diagnosis of prostatomegaly. In a previous study, volume was analysed with a slice addition technique (SAT), which was validated as highly accurate. Irrespective of high accuracy, SAT represents a complex and time-consuming procedure, which limits its clinical use. Thus, demand exists for more practical volume assessment methods. In this study, the prostatic volume of 95 canine patients (58 intact males, 37 neutered males) were analysed retrospectively by using the ellipsoid formula (Formula) and an imaging "wrap" function tool (Wrap) to help assess accuracy and applicability. Accuracy was checked against phantom measurements and results were compared to SAT measurements of the same patient pool obtained from a previously published paper. Patients were grouped according to prostatic structure (H = homogeneous, I = inhomogeneous, C = cystic) and volume using the SAT (volume group = vg: 1, 2 and 3)., Results: High correlation between the Formula or Wrap volume and the phantom volume was found, the values being higher for the Formula. Mean Formula volumes (vg 1: 2.2 cm 3 , vg 2: 14.5 cm 3 , vg 3: 109.4 cm 3 , respectively) were significantly underestimated, while mean Wrap volumes (vg 1: 3.8 cm 3 , vg 2: 19.5 cm 3 , vg 3: 159.2 cm 3 ) were statistically equivalent to SAT measurements (vg 1: 3.1 cm 3 , vg 2: 18.6 cm 3 , vg 3: 157.2 cm 3 , respectively). Differences between Formula and SAT volumes ranged from 22.4-31.1%, while differences between Wrap and SAT volumes were highest in small prostates (vg 1: 22.1%) and fell with increasing prostatic size (vg 3: 1.3%)., Conclusion: The Wrap function is highly accurate, less time-consuming and complex compared to SAT and could serve as beneficial tool for measuring prostatic volume in clinical routine after further validation in future studies. The Formula method cannot be recommended as an alternative for volumetric measurements of the prostate gland due to its underestimation of volumes compared to SAT results.

Medial coronoid disease and elbow incongruence are common orthopedic problems causing forelimb lameness in large breed dogs. Therefore, the objective of this retrospective clinical study is to radiographically quantify the incongruity of humeroradial, humeroulnar, and radioulnar joints in dogs with confirmed medial coronoid disease compared to controls. Sixty-six (93 elbows) control and 73 (113 elbows) diseased Labrador and Golden Retrievers were enrolled. Our objective was achieved by measuring the central humeroradial, humeroulnar, and radioulnar distances and calculating the corresponding subluxation index of each joint on the extended mediolateral elbow radiograph. The traditional radioulnar step was also measured on the same mediolateral projection. Variables were compared between control and diseased groups, and the significance level was P < .05. The central humeroradial, humeroulnar, and radioulnar distances and corresponding subluxation indices increased significantly (P ≤ .007) in elbows with confirmed medial coronoid disease compared to controls. Radioulnar step increased significantly (P < .0001) in diseased elbows. Elbow incongruence, manifested by increased humeroradial, humeroulnar, and radioulnar subluxation indices, appeared to be consistent with medial coronoid disease in Labrador and Golden Retrievers. An elbow with a humeroradial, humeroulnar, or radioulnar subluxation index >0.15, 0.10, or 0.14, respectively, or radioulnar step >1.4 mm, is more likely to be incongruent and may have a concomitant coronoid disease. Background: Technical failures and incorrect usage of digital X-ray systems may lead to a decreasing image quality, artefacts and a higher dose exposure of staff and patients. Although there are no regulations regarding constancy testing in veterinary radiology all operators are required to avoid unnecessary exposure. The aim of this study was to develop a reasonably inexpensive zoomorphic 3D-printed test specimen for constancy testing that allows the detection of changing image quality by visual analysis. Primarily, a calibration curve of the attenuation factor of the 3D-printing material (ZP150) was determined. MATLAB converted every pixel value of a thorax X-ray image of a Beagle dog into an equivalent thickness of printing material. The thickness distribution was printed using a 3D-printer. This printed test specimen was additionally provided with five thin aluminium discs to simulate lung nodules. To evaluate the usability for constancy testing 12 X-ray images of the test specimen were made. Two images (reference and control) were taken with the minimum dose in order to obtain images suitable for diagnosis purposes. Eight images were taken with a dose differing 30-140% from the reference dose by varying current-time product (mAs) or tube voltage (kVp). Two images were taken with the same parameters as the reference image but edited with different image processing. Six veterinarians (general practitioners) evaluated ten chosen structures in the X-ray images in a Visual Grading Analysis and scored the image quality of these structures for every image in comparison to the reference image. A Visual Grading Analysis Score was calculated and statistically analysed., Results: A higher current-time product led to a negligibly better evaluation of the X-ray image. The lower the current-time product the worse the X-ray images were scored. Likewise, both increasing and decreasing of the tube voltage led to lower scores., Conclusions: A zoomorphic test specimen can be used for constancy testing of digital X-ray systems in veterinary medicine. Especially a lower dose can be recognised due to deviation in the image quality when compared to the reference image. The 3D-printed test specimen is less expensive than test equipment used in human medicine.

Background: Prostatic diseases are common and mostly associated with enlargement of the accessory gland. Thus, determining the prostate size has become a main criterion for evaluating prostate health status. Computed tomography (CT) is recommended as a beneficial tool for evaluating prostate size, morphology and surrounding tissues. The purpose of this study was to establish an accurate procedure for volume estimation and afterwards evaluate the prostate volume in CT. Data of 95 dogs were analysed (58 male intact, 37 male neutered) using the slice addition technique with the Amira program. Accuracy of volumetric measurements by CT was validated by comparing them with those of phantoms of known volume. Patients were grouped according to age (< 4 yrs., 4-8 yrs., > 8 yrs) and prostate morphology in CT (H = homogeneous, I = inhomogeneous, C = cystic). The length of the sixth lumbar vertebra was measured to relate prostate volume to body size. This ratio was generated to compare prostate volume between the groups, irrespective of body size (ratio volume = Rv)., Results: A high correlation between the CT-derived and phantom volume was found. Overall, the mean prostate volume was 58.6 cm 3 . The mean ratio volume was 1.3 in intact male dogs, this being significantly higher than in neutered dogs (0.7). The lowest ratio volume values were found in group H for intact (Rv = 0.9) and neutered dogs (Rv = 0.6), followed by group I (intact: Rv = 1.1; neutered: Rv = 0.7) and C (intact: Rv = 1.4; neutered: Rv = 0.8). The length of the sixth lumbar vertebra was well correlated with the prostate volume (intact: r = 0.63, p < 0.001; neutered: r = 0.48, p = 0.003), while age exhibited a correlation only in intact dogs (r = 0.52, p < 0.001)., Conclusion: The present study is pioneering in applying a slice addition technique to volumetric measurements of the prostate gland in CT, resulting in a highly precise method. Volumetric measurements of the canine prostate gland in CT images provide information about the prostate structure, castration status, age and body size of the patients. Therefore, prostate volume is a relevant parameter for evaluating prostate health status.

Objectives:  The purpose of this study was to objectively estimate humeroradial (HR), humeroulnar (HU) and radioulnar (RU) congruity on mediolateral elbow radiographs of Labrador and Golden Retrievers without and with medial coronoid disease (MCD), and to determine the correlation between traditional and modified RU-step assessment techniques., Materials and Methods:  Extended mediolateral elbow radiographs of Labrador and Golden Retrievers without MCD (control group) and with confirmed MCD (diseased group) were investigated. Absolute and average HR and HU distances were determined and standardized by the radius of the corresponding humeral condyle. Traditional RU-step was measured, and a modified procedure of RU-step assessment was generated. The correlation between the two RU-step assessment procedures was tested., Results:  A total of 131 (197 elbows) Labrador and Golden Retrievers met the criteria for inclusion in the control and diseased groups. The normalized HR and HU distances increased significantly ( p ≤ 0.002) in dogs with MCD. There was a significant increase ( p  < 0.0001) in the traditional and modified RU-step in dogs with MCD. A significant correlation ( r s  = 0.74, p  < 0.0001) was identified between traditional and modified RU-step calculated for control and diseased elbows., Clinical Relevance:  Dogs with confirmed MCD had quantitative radiographic evidences of elbow incongruity. Modified RU-step procedure may be an alternative to the traditional technique and can be utilized during routine quantification of HR and HU congruity. Background: Nephropathies are among the most common diseases in dogs. Regular examination of the kidney function plays an important role for an adequate treatment scheme. The determination of the glomerular filtration rate (GFR) is seen as the gold standard in assessing the kidney status. Most of the tests have the disadvantage that only the complete glomerular filtration rate of both kidneys can be assessed and not the single kidney glomerular filtration rate. Imaging examination techniques like dynamic contrast-enhanced magnetic resonance imaging have the potential to evaluate the single kidney GFR. There are studies in human medicine describing the determination of the single kidney GFR using this technique. To our knowledge there are no such studies for dogs., Results: An exponential fit was found to describe the functional interrelation between signal intensity and contrast medium concentrations. The changes of contrast medium concentrations during the contrast medium bolus propagation were calculated. The extreme values of contrast medium concentrations in the kidneys were reached at nearly the same time in every individual dog (1st maximum aorta 8.5 s, 1st maximum in both kidneys after about 14.5 s; maximum concentration values varied between 17 and 125 µmol/mL in the aorta and between 4 and 15 µmol/mL in the kidneys). The glomerular filtration rate was calculated from the concentration changes of the contrast medium using a modified Rutland-Patlak plot technique. The GFR was 12.7 ± 2.9 mL/min m 2 BS for the left kidney and 12.0 ± 2.2 mL/min/m 2 BS for the right kidney. The mean values of the coefficient of determination of the regression lines were averagely 0.91 ± 0.08., Conclusions: The propagation of contrast medium bolus could be depicted well. The contrast medium proceeded in a similar manner for every individual dog. Additionally, the evaluation of the single kidney function of the individual dogs is possible with this method. A standardized examination procedure would be recommended in order to minimize influencing parameters.

Medial coronoid process disease is a common leading cause of thoracic limb lameness in dogs. Computed tomography and arthroscopy are superior to radiography to diagnose medial coronoid process disease, however, radiography remains the most available diagnostic imaging modality in veterinary practice. Objectives of this retrospective observational study were to describe the prevalence of medial coronoid process disease in lame large breed dogs and apply a novel method for quantifying the radiographic changes associated with medial coronoid process and subtrochlear-ulnar region in Labrador and Golden Retrievers with confirmed medial coronoid process disease. Purebred Labrador and Golden Retrievers (n = 143, 206 elbows) without and with confirmed medial coronoid process disease were included. The prevalence of medial coronoid process disease in lame large breed dogs was calculated. Mediolateral and craniocaudal radiographs of elbows were analyzed to assess the medial coronoid process length and morphology, and subtrochlear-ulnar width. Mean grayscale value was calculated for radial and subtrochlear-ulnar zones. The prevalence of medial coronoid process disease was 20.8%. Labrador and Golden Retrievers were the most affected purebred dogs (29.6%). Elbows with confirmed medial coronoid process disease had short (P < 0.0001) and deformed (∼95%) medial coronoid process, with associated medial coronoid process osteophytosis (7.5%). Subtrochlear-ulnar sclerosis was evidenced in ∼96% of diseased elbows, with a significant increase (P < 0.0001) in subtrochlear-ulnar width and standardized grayscale value. Radial grayscale value did not differ between groups. Periarticular osteophytosis was identified in 51.4% of elbows with medial coronoid process disease. Medial coronoid process length and morphology, and subtrochlear-ulnar width and standardized grayscale value varied significantly in dogs with confirmed medial coronoid process disease compared to controls. Age-related involution in dogs involves loss of muscle mass and changes in connective tissue and articular cartilage. The aim of this study was to examine whether an age-related influence on joint mobility can be detected in the absence of disease. Five young (mean age 2.0 years) and five old (mean age 10.4 years) healthy and sound Beagle dogs underwent computer-assisted gait analysis during locomotion on a treadmill. Shoulder, elbow, carpal, hip, stifle, and tarsal joint angles including joint angle progression curves, minimum and maximum joint angles, and range of motion (ROM) in degrees were analyzed. The old group had a smaller maximum joint angle ( p = 0.037) and ROM ( p = 0.037) of the carpal joint; there were similar tendencies in the shoulder, elbow, and carpal joints. Descriptive analysis of the progression curves revealed less flexion and extension of the forelimb joints. The results indicate restricted joint mobility of the forelimb in old dogs, primarily of the carpal joint. Results in the joints of the hindlimb were inconsistent, and the contrasting alterations may be due to a compensatory mechanism. As most alterations were found in the distal joints, these should receive particular attention when examining elderly dogs.

Bone density measurements using computed tomography (CT) instead of dual-energy X-ray absorptiometry (DEXA) are currently of great interest in human and veterinary medical research as it would be beneficial to use CT scans obtained for other indications also for determining bone density. For Hounsfield units (HU) measured with CT in specific regions of interests (ROIs) in one or several slice/s a correlation with bone mineral density (BMD) measured by DEXA in humans and dogs of between 0.44 and 0.77 is reported in the literature. In the present study, instead certain volumes of interest (VOIs) obtained by CT scan and the corresponding HU to the respective VOIs were compared with the bone mineral density of the corresponding areas measured by DEXA. The aim of the study was to investigate whether this procedure gives more accurate information about bone density of the bones as three-dimensional objects of the respective patient. Correlation between measured HU in the respective VOI and BMD measured with DEXA in the corresponding ROI showed a very good correlation of 0.93. Linear regression with R 2  = 0.85 (p = 0.0262) was calculated. Except for VOI5, similar distribution of values and significant differences (p < 0.0001-0.0087) between ROIs/VOIs were detected. Determining HU for assessing bone mineral density in a certain volume provides more accurate results than those previously reported from two-dimensional (2D) CT measurements. © 2017 Orthopaedic Research Society. Background: Aseptic loosening due to bone remodelling processes after total hip replacement is one common cause for revision surgery. In human medicine, dual-energy X-ray absorptiometry (DEXA) is the gold standard for quantitative evaluation of bone mineral density, whereas in veterinary medicine conventional radiography is used for follow-up studies. Recently, a method has been described using digital X-ray images for quantitative assessment of grey scale values of bone contrast. Therefore, the aim of the present study was to evaluate the correlation of bone mineral density (BMD) measured by DEXA with grey scale values (GV) measured in digital X-ray images (RX50, RX66) ex vivo., Results: The measured GV in the chosen X-ray settings showed on average a good correlation (r = 0.61) to the measured BMD with DEXA. Correlation between the two X-ray settings was very good (r = 0.81). For comparisons among regions of interests (ROIs) a difference of 8.2% was found to be statistically significant, whereas in the case of RX50 and RX66 differences of 5.3% and 4.1% were found to be statistically significant., Conclusions: Results indicate that measuring absolute changes in bone mineral density might be possible using digital radiography. Not all significant differences between ROIs detectable with DEXA can be displayed in the X-ray images because of the lower sensitivity of the radiographs. However, direct comparison of grey scale values of the periprosthetic femur in one individual patient during the follow-up period, in order to predict bone remodelling processes, should be possible, but with a lesser sensitivity than with DEXA. It is important that the same X-ray settings are chosen for each patient for follow-up studies.

Background: Prostatic diseases in intact male dogs are common. However, studies about the computed tomographic (CT) examination of the prostate in dogs are rare. The aim of the present study was to evaluate age related-changes in the canine prostate with the help of the CT and to evaluate whether measuring Hounsfield Units (HUs) in different morphological conditions of the prostate is of diagnostic value. Fifty pre- and post-contrast CT scans of the prostate of dogs were evaluated and divided into three groups according to the tissue structure: Group1 dogs with homogenous prostate tissue (16/50); group 2 with prostate cysts (26/50) and group 3 with inhomogeneous prostate tissue (8/50). The prostatic dimensions were measured and the ratio between length, height and width and the sixth lumbar vertebra was calculated. Median values of prostatic attenuation measured in HUs, using regions of interests (ROIs) were determined on pre- and post- contrast scans over the whole length of the prostate. The results were compared to the dog's age. Furthermore, the CT Images were compared with the results of ultrasonography (47/50)., Results: On pre-contrast scans HUs within ROIs placed in the prostate did not differ statistically significantly between the different morphological groups (1: 37.7; 2: 36.3; 3: 39.8 HU). HUs within on the post- contrast scans showed statistically significant differences between the groups. Group one had a mean density of 93.6 HU, group two had a mean density of 106.1 HU and group three had one of 138.2 HU. The prostatic size in the first group was smaller than in the other groups, whereas the largest prostates were found in the second group. In six cases the post-contrast CT scan showed results that differed from the ultrasound examination. Dogs had a homogenous tissue in ultrasonography while the CT scan revealed an inhomogeneous tissue structure., Conclusions: The CT examination can be a beneficial diagnostic tool for examining the prostatic size and for evaluating the prostatic tissue. The different HUs reflected age-related changes and alterations in the prostate while measuring the density of the prostate. Contrast agent application enables a more specific analysis of the prostate to be carried out and for precise changes in tissue structure to be observed.

Background: Coxofemoral osteoarthritis is a chronic, disabling condition affecting people and dogs, with THA providing an excellent return to function in severely affected joints. The principal role of THA is to restore an adequate range of motion to the hip joint while transferring load from the acetabulum in order to improve the survival of the implant and enhance the limb function in the short and long terms. The objectives of the study reported here were, therefore, to radiographically evaluate periprosthetic acetabular bone GV and to assess prosthetic head acetabular coverage after 4 months of uncemented and cemented THA in dogs. Means periprosthetic acetabular GV for each and combined 3 regions of interest (zones 1, 2 and 3) were calculated immediately and 4 months after THA. Prosthetic head Norberg (PHN) angle was also measured to assess the degree of prosthetic head acetabular coverage after 4 months of surgery., Results: Zones 2 and 3 showed a significant increase in the mean bone GV after 4 months of uncemented THA. No differences in zones 1-3 after 4 months of cemented THA. Combined zones showed a significant increase in overall mean bone GV 4 months after uncemented THA; whereas, no changes were identified after 4 months of cemented THA. The PHN angles did not change after 4 months of uncemented and cemented THA and did not differ significantly between the 2 designs of hip arthroplasty., Conclusions: Regional periprosthetic acetabular bone GV varies with the design of THA. None of the designs showed periprosthetic acetabular bone lucency. No differences identified in the degree of prosthetic head acetabular coverage in both designs, indicating proper implant stability after 4 months of surgery. Further longer-term investigation on larger population is however still warranted.

Background: Functional magnetic resonance imaging (fMRI) is an advanced and frequently used technique for studying brain functions in humans and increasingly so in animals. A key element of analyzing fMRI data is group analysis, for which valid spatial normalization is a prerequisite. In the current study we applied normalization and group analysis to a dataset from an auditory functional MRI experiment in anesthetized beagles. The stimulation paradigm used in the experiment was composed of simple Gaussian noise and regular interval sounds (RIS), which included a periodicity pitch as an additional sound feature. The results from the performed group analysis were compared with those from single animal analysis. In addition to this, the data were examined for brain regions showing an increased activation associated with the perception of pitch., Results: With the group analysis, significant activations matching the position of the right superior olivary nucleus, lateral lemniscus and internal capsule were identified, which could not be detected in the single animal analysis. In addition, a large cluster of activated voxels in the auditory cortex was found. The contrast of the RIS condition (including pitch) with Gaussian noise (no pitch) showed a significant effect in a region matching the location of the left medial geniculate nucleus., Conclusion: By using group analysis additional activated areas along the canine auditory pathways could be identified in comparison to single animal analysis. It was possible to demonstrate a pitch-specific effect, indicating that group analysis is a suitable method for improving the results of auditory fMRI studies in dogs and extending our knowledge of canine neuroanatomy.

Background: The amputation of a limb is a surgical procedure that is regularly performed in small animal practice. In spite of several clinical reports indicating high owner satisfaction after limb amputation in dogs, an amputation is still very critically seen by the owners, and even by some veterinarians, due to the lack of accurate information about the recovery of amputee patients. Thus, the objective of this study was to prospectively evaluate, both objectively and subjectively, the recovery outcome of dogs undergoing a hind limb amputation. Twelve patients in which a hind limb amputation was scheduled were studied. Kinetic and kinematic gait analyses were performed before the amputation, and 10, 30, 90 and 120 days after surgery. Magnetic resonance (MR) examination of the contralateral stifle joint was performed before and 120 days after amputation. The subjective impressions of the owners were gathered at the same examination times of the gait analyses., Results: Kinetic data showed a redistribution of the load to all remaining limbs after the amputation; ten days after the procedure patients had already established their new locomotory pattern. Kinematic data showed significant differences between sessions in the mean angle progression curves of almost all analyzed joints; however, the ranges of motion were very similar before and after the amputation, and remained constant in the subsequent sessions after the amputation. No changes in the signal intensity of the soft tissues evaluated, and no evidence of cartilage damage or osteoarthritis was seen on the MR examination of the contralateral stifle. Owners evaluated the results of the amputation very positively, both during and at the end of the study., Conclusions: Dogs had a quick adaptation after a hind limb amputation, and the adaptation process began before the amputation was performed. This happened without evidence of morphologic changes in the contralateral stifle joint, and with a very positive evaluation from the owner.

Background: Canine intervertebral disc degeneration can lead to intervertebral disc disease. Mild degenerative changes in the structure of the canine intervertebral disc can be identified in magnetic resonance images, whereas these changes are not visible in computed tomographic images. Therefore, one aim of this study was to detect whether colour-coded computed tomography enhances the visibility of mild degenerative changes in the canine disc structure compared to non-contrast computed tomography. Furthermore, the study aimed to detect if intervertebral disc degeneration could be classified with a higher reliability in colour-coded images than in non-contrast images., Results: Computed tomographic image studies of 144 canine intervertebral discs were coloured using three different lookup tables. Canine intervertebral disc degeneration was evaluated by three observers using a 5-grade classification system and compared to the evaluation of non-contrast CT and MRI images. A moderate to almost perfect intraobserver and a moderate to substantial interobserver agreement were found depending on the used colour code. On comparing non-contrast and colour-coded CT significant differences were found by one observer only. Significant differences in evaluation were found in grading intervertebral disc degeneration in MRI and colour-coded CT., Conclusions: Intervertebral disc degeneration could not be classified with a higher reliability on colour-coded images compared to non-contrast images. Furthermore, colour-coded CT did not enhance the visibility of mild degenerative changes in disc structure compared to non-contrast CT. However, the better intraobserver agreement and the subjective impression of the observers highlighted that the usage of colour encoded CT data sets with a wide range of tonal values of few primary and secondary colours may facilitate evaluation.

Backround: Real-time three-dimensional echocardiography (RT3DE) enables accurate volume determination of the left ventricle (LV), since measurements in foreshortened depicted views are avertable. Different analyzing programs are available for this RT3DE. The commonly used semi-automatic software 4D-AutLVQ™ showed underestimation of LV volumes in comparison with CMRI in healthy anesthetized dogs (Am J Vet Res 74(9):1223-1230, 2013). TomTec 4D LV-Function™ is an offline analysis program for morphological and functional analyses of the left ventricle by using manual measurement optimization, showing excellent agreement with CMRI in human medicine (Echocardiography 27(10):1263-1273, 2010; Eur J Echocardiogr 11(4):359-368, 2010; Echocardiography 24(9):967-974, 2007). The aim of the present study was to compare these different RT3DE analyzing software programs to test the possibility of one performing better than the other by assessing accuracy and reproducibility in comparison with the reference method cardiac magnetic resonance imaging (CMRI) by determining the left ventricular end-diastolic volume (EDV), end-systolic volume (ESV), stroke volume (SV) and ejection fraction (EF). RT3DE and CMRI were performed during anesthesia in 10 healthy beagles. The analyzing programs 4D-AutLVQ™ (based on semi-automated border detection) and TomTec 4D LV-Function™ (primary manual tracking with semi-automated border detection) were used for RT3DE volume analysis of the left ventricle. Left ventricular EDV, ESV, SV and EF were measured and compared to those measured by the reference method CMRI. Repeated measurements were performed to determine inter- and intra-observer variability., Results: Both, 4D-AutLVQ™ and 4D-TomTec™ showed small but significant underestimation for EDV and ESV with quite good correlation (r = 0.34-0.69) in comparison with CMRI, without significant difference between each of them. Ejection fraction (EF) measured by 4D-TomTec™ showed no significant differences compared to CMRI (p = 0.12), while 4D-AutLVQ™ significantly underestimated LV-EF (p = 0.03). Analyzing time was shorter using 4D-AutLVQ™ compared to 4D-TomTec™. The inter-observer variability was higher using 4D-TomTec™ than with 4D-AutLVQ™, whereas both methods present excellent intra-observer variability., Conclusion: 4D-TomTec™ and 4D-AutLVQ™ are feasible RT3DE analyzing programs, allowing accurate volume quantification of the left ventricle, albeit with significant underestimation of ventricular volumes in comparison with the gold standard CMRI. 4D-AutLVQ™ is performed faster with less inter-observer variability than 4D-TomTec™. Therefore, 4D-AutLVQ™ is the more practicable measurement method when comparing the different analyzing programs.

Background: Displacement of canine intervertebral disk material can be seen directly in diagnostic imaging modalities such as magnetic resonance imaging and computed tomographic imaging. Canine intervertebral disk herniation can be differentiated into Hansen type 1 and 2 categories by clinical appearance, but anular- and nuclear disk material cannot be distinguished in computed tomographic images. Therefore, we hypothesized that the "Disk extension beyond the interspace"-nomenclature that describes the displacement by the disk contour might aid diagnosis. The aim of this study was to test the reliability of the "Disk extension beyond the interspace"-nomenclature in the evaluation of canine intervertebral disks via magnetic resonance and computed tomographic imaging., Results: Magnetic resonance and computed tomographic images of 144 intervertebral disks of 43 dogs were evaluated by 3 observers with different degrees of experience from 2 institutions retrospectively. A substantial intraobserver agreement was found, while interobserver agreement was fair to moderate with significant differences in evaluation. Comparison of imaging methods showed a fair to moderate agreement without statistically significant differences in evaluation., Conclusions: DEBIT-nomenclature cannot be recommended for veterinary clinical usage yet. The largest variability was found in the evaluation of the bulged canine intervertebral disk. The observers' experience and the imaging method influenced DEBIT- evaluation only slightly, while training and working at different institutions influenced DEBIT-evaluation strongly.

Background: Right ventricular (RV) volume and function are important diagnostic and prognostic factors in dogs with primary or secondary right-sided heart failure. The complex shape of the right ventricle and its retrosternal position make the quantification of its volume difficult. For that reason, only few studies exist, which deal with the determination of RV volume parameters. In human medicine cardiac magnetic resonance imaging (CMRI) is considered to be the reference technique for RV volumetric measurement (Nat Rev Cardiol 7(10):551-563, 2010), but cardiac computed tomography (CCT) and three-dimensional echocardiography (3DE) are other non-invasive methods feasible for RV volume quantification. The purpose of this study was the comparison of 3DE and CCT with CMRI, the gold standard for RV volumetric quantification., Results: 3DE showed significant lower and CCT significant higher right ventricular volumes than CMRI. Both techniques showed very good correlations (R > 0.8) with CMRI for the volumetric parameters end-diastolic volume (EDV) and end-systolic volume (ESV). Ejection fraction (EF) and stroke volume (SV) were not different when considering CCT and CMRI, whereas 3DE showed a significant higher EF and lower SV than CMRI. The 3DE values showed excellent intra-observer variability (<3%) and still acceptable inter-observer variability (<13%)., Conclusion: CCT provides an accurate image quality of the right ventricle with comparable results to the reference method CMRI. CCT overestimates the RV volumes; therefore, it is not an interchangeable method, having the disadvantage as well of needing general anaesthesia. 3DE underestimated the RV-Volumes, which could be explained by the worse image resolution. The excellent correlation between the methods indicates a close relationship between 3DE and CMRI although not directly comparable. 3DE is a promising technique for RV volumetric quantification, but further studies in awake dogs and dogs with heart disease are necessary to evaluate its usefulness in veterinary cardiology.

Objective: To (1) evaluate thoracic limb loads and symmetry, and elbow function and morphology, before and after arthroscopic treatment of unilateral medial coronoid process disease (MCPD), and (2) determine if functional variables correlate with morphologic findings., Study Design: Prospective case series., Animals: Dogs (n = 14) with thoracic limb lameness., Methods: Dogs were included when unilateral MCPD was confirmed as the cause of lameness. Kinetic analysis of both thoracic limbs, along with kinematic analysis and goniometry of both elbows were carried out before, and 60, 120, and 180 days after partial coronoidectomy by arthroscopy. Radiography and computed tomography of both elbows were performed before and 180 days after arthroscopy., Results: A nonsignificant (P = .11) increase in the peak vertical loads (PFz), and a significant (P = .022) increase in the vertical impulse (iFz) applied by the affected limb were seen. Symmetry indices improved, with significant differences between sessions (PFz: P = .019; iFz: P = .003). Kinematic variables showed no significant differences, between sessions or when comparing both elbows within sessions. Goniometry revealed no significant differences between sessions, but some significant differences were identified when comparing both elbows within sessions. Osteophytosis and degree of lameness showed no correlation, before (rs  = -0.077; P = .79) or after arthroscopy (rs  = 0.27; P = .35)., Conclusions: Kinetic variables improved after arthroscopy, without full restoration of function. Kinematic variables did not change significantly. Osteoarthritis and goniometric measurements in the affected joint worsened. Objective: Although the classification of canine intervertebral disc degeneration using magnetic resonance imaging (MRI) has been described in the literature, there is no such classification using computed tomographic imaging. Because computed tomography (CT) is a frequently used diagnostic imaging tool in veterinary medicine, the aim of this study was the introduction and validation of such a scoring system. T2-weighted magnetic resonance images were available for comparative analysis., Material and Methods: A total of 43 dogs were examined using CT and MRI. Image data records of 144 intervertebral discs were blinded, randomized and evaluated twice by three observers. CT data were analyzed using a self-developed scoring system, while MRI data sets were evaluated using the Pfirrmann scoring system. Intra- and interobserver agreement were determined using Statistical Analysis Software (SAS)., Results: Intra- and interobserver agreement were mostly substantial in the Pfirrmann (0.58-0.77) and self-developed (0.60-0.81) scoring systems. A slight agreement was found between both classification systems (κ scores 0.26-0.29)., Conclusion and Clinical Relevance: The self-developed scoring system allows a reliable assessment of canine intervertebral disc degeneration using CT imaging. Therefore, further diagnostic and prognostic information can be obtained. Degenerative changes in the intervertebral discs could be identified at an earlier stage when using MRI in comparison with CT.

Background: Functional magnetic resonance imaging (fMRI) is a technique able to localize neural activity in the brain by detecting associated changes in blood flow. It is an essential tool for studying human functional neuroanatomy including the auditory system. There are only a few studies, however, using fMRI to study canine brain functions. In the current study ten anesthetized dogs were scanned during auditory stimulation. Two functional sequences, each in combination with a suitable stimulation paradigm, were used in each subject. Sequence 1 provided periods of silence during which acoustic stimuli could be presented unmasked by scanner noise (sparse temporal sampling) whereas in sequence 2 the scanner noise was present throughout the entire session (continuous imaging). The results obtained with the two different functional sequences were compared., Results: This study shows that with the proper experimental setup it is possible to detect neural activity in the auditory system of dogs. In contrast to human fMRI studies the strongest activity was found in the subcortical parts of the auditory pathways. Especially sequence 1 showed a high reliability in detecting activated voxels in brain regions associated with the auditory system., Conclusion: These results indicate that fMRI is applicable for studying the canine auditory system and could become an additional method for the clinical evaluation of the auditory function of dogs. Additionally, fMRI is an interesting technique for future studies concerned with canine functional neuroanatomy.

Background: Mammalian juveniles undergo dramatic changes in body conformation during development. As one of the most common companion animals, the time line and trajectory of a dog's development and its body's re-proportioning is of particular scientific interest. Several ontogenetic studies have investigated the skeletal development in dogs, but none has paid heed to the scapula as a critical part of the mammalian forelimb. Its functional integration into the forelimb changed the correspondence between fore- and hindlimb segments and previous ontogenetic studies observed more similar growth patterns for functionally than serially homologous elements. In this study, the ontogenetic development of six Beagle siblings was monitored between 9 and 51 weeks of age to investigate their skeletal allometry and compare this with data from other lines, breeds and species., Results: Body mass increased exponentially with time; log linear increase was observed up to the age of 15 weeks. Compared with body mass, withers and pelvic height as well as the lengths of the trunk, scapula, brachium and antebrachium, femur and crus exhibited positive allometry. Trunk circumference and pes showed negative allometry in all, pelvis and manus in most dogs. Thus, the typical mammalian intralimb re-proportioning with the proximal limb elements exhibiting positive allometry and the very distal ones showing negative allometry was observed. Relative lengths of the antebrachium, femur and crus increased, while those of the distal elements decreased., Conclusions: Beagles are fully-grown regarding body height but not body mass at about one year of age. Particular attention should be paid to feeding and physical exertion during the first 15 weeks when they grow more intensively. Compared with its siblings, a puppy's size at 9 weeks is a good indicator for its final size. Among siblings, growth duration may vary substantially and appears not to be related to the adult size. Within breeds, a longer time to physically mature is hypothesized for larger-bodied breeding lines. Similar to other mammals, the Beagle displayed nearly optimal intralimb proportions throughout development. Neither the forelimbs nor the hindlimbs conformed with the previously observed proximo-distal order of the limb segment's growth gradients. Potential factors responsible for variations in the ontogenetic allometry of mammals need further evaluation.

As part of the preparation of a large cohort study in the entire German population, this study examined the feasibility of cat and dog owners collecting nasal and oral swabs of their animals at home as a method of assessing exposure to zoonoses. In veterinary clinics in Hannover, Germany, 100 pet owners were recruited. Nasal and oral swabs of pets were taken by a veterinarian at the clinic and owners took swabs at home. Swabs were analysed regarding bacterial growth and compared (owner vs. vet) using Cohen's kappa and McNemar's test. The return rate of kits was 92%, and 77% of owners thought it unnecessary to have veterinarian assistance to swab the mouth. McNemar's test results: oral swabs 78% agreement with Gram-positive bacterial growth, 87% agreement with Gram-negative bacterial growth; with similar results for nasal swabs. Although sample quality differed, this method allowed the receipt of swabs from pets in order to obtain information about colonization with zoonotic pathogens.

Objective: To quantify left ventricle (LV) volumes by use of 1-D, 2-D, and 3-D echocardiography versus MRI in dogs., Animals: 10 healthy Beagles., Procedures: During anesthesia, each dog underwent an echocardiographic examination via the Teichholz method, performed on the basis of standard M-mode frames (1-D); the monoplane Simpson method of disk (via 2-D loops); real-time triplane echocardiography (RTTPE) with a 3-D probe; and real-time 3-D echocardiography with a 3-D probe. Afterward, cardiac MRI was performed. Values for the LV end-diastolic volume (EDV), end-systolic volume (ESV), and ejection fraction (EF) were compared between each echocardiographic method and the reference method (cardiac MRI)., Results: No significant differences for EDV, ESV, and EF were detected between RTTPE and cardiac MRI. Excellent correlations (r = 0.97, 0.98, and 0.95 for EDV, ESV, and EF, respectively) were found between RTTPE and values for cardiac MRI. The other echocardiographic methods yielded values significantly different from cardiac MRI and results correlated less well with results of cardiac MRI for EDV, ESV, and EF. Use of the Teichholz method resulted in LV volume overestimation, whereas the Simpson method of disk and real-time 3-D echocardiography significantly underestimated LV volumes., Conclusions and Clinical Relevance: Use of RTTPE yielded excellent correlations and nonsignificant differences with cardiac MRI and is a suitable method for routine veterinary cardiac examination.

Objective: To evaluate the accuracy and reproducibility of left ventricular (LV) volumetric and function variables determined via contrast-enhanced cardiac CT and cardiac MRI in healthy dogs., Animals: 10 healthy Beagles., Procedures: Cardiac MRI and cardiac CT were performed in anesthetized Beagles; both examinations were conducted within a 2-hour period. Cardiac MRI was performed with a 3.0-T magnet, and contrast-enhanced cardiac CT was performed with a 64-row detector CT machine. Data sets were acquired during apnea with simultaneous ECG gating. Short-axis images were created to determine functional variables via the Simpson method., Results: Cardiac CT values for mean end-diastolic and end-systolic LV volumes had excellent correlation (r = 0.95) with cardiac MRI measurements, whereas LV stroke volume (r = 0.67) and LV ejection fraction (r = 0.75) had good correlations. The only variable that differed significantly between imaging modalities was end-diastolic LV volume. For each pair of values, Bland-Altman analysis revealed good limits of agreement., Conclusions and Clinical Relevance: The 3-D modalities cardiac CT and cardiac MRI were excellent techniques for use in assessing LV functional variables. Similar results were obtained for LV volume and function variables via both techniques. The major disadvantage of these modalities was the need to anesthetize the dogs for the examinations.

The number of surviving spiral ganglion neurons (SGNs) is important for the function of cochlear implants. Electrical stimulation has been discussed controversially regarding its protective effects on SGNs both in vivo and in vitro. The aim of this study was to develop a device for electrical stimulation of cultured SGNs. The developed device was first evaluated with cultivated fibroblasts at voltages from 10 to 60 V using biphasic rectangular pulses (pulse width: 10 ms, frequency: 50 Hz, stimulus burst: 1 s, interburst interval: 19 s). Possible toxic effects of 48 h patterned electrical stimulation were evaluated by propidium-iodide flow cytometry of the harvested fibroblasts. Second, the effects of 48-h electrical stimulation at voltages of 6 and 30 V on survival of cultivated SGNs were investigated as a single treatment and in the presence of brain-derived neurotrophic factor (BDNF, 100 ngml) in the culture medium. The results demonstrated constant pH and temperature behavior during electrical stimulation. Furthermore, no toxic effects on fibroblasts up to 40 V were found. The electrical stimulation of cultured neurons did not alter the survival rate compared to unstimulated control conditions. While BDNF application alone showed a significant effect on SGN survival, combination with electrical stimulation resulted in a not significantly increased cell survival. As a limitation of the study, the sensitivity of the described experimental setup appears to be low and only one stimulation pattern has been tested. Thus, experiments using different patterns of electrical stimulation and a more sensitive cultivation setup have to be carried out to fully investigate the effects of electrical stimulation on cultured SGNs. In this context, the developed device can be of help as it provides controlled and reproducible electrical stimulation conditions.

Background: Magnetic resonance (MR) imaging is the preferred diagnostic tool to evaluate internal disorders of many joints in humans; however, the usefulness of MR imaging in the context of osteoarthritis, and joint disease in general, has yet to be characterized in veterinary medicine. The objective of this study was to assess the diagnostic accuracy of short-duration 3 Tesla MR imaging for the evaluation of cranial and caudal cruciate ligament, meniscal and cartilage damage, as well as the degree of osteoarthritis, in dogs affected by non-traumatic, naturally-occurring cranial cruciate ligament rupture (CCLR). Diagnoses made from MR images were compared to those made during surgical exploration. Twenty-one client-owned dogs were included in this study, and one experienced evaluator assessed all images., Results: All cranial cruciate ligaments were correctly identified as ruptured. With one exception, all caudal cruciate ligaments were correctly identified as intact. High sensitivities and specificities were obtained when diagnosing meniscal rupture. MR images revealed additional subclinical lesions in both the cranial and caudal cruciate ligaments and in the menisci. There was a "clear" statistical (kappa) agreement between the MR and the surgical findings for both cartilage damage and degree of osteoarthritis. However, the large 95% confidence intervals indicated that evaluation of cartilage damage and of degree of osteoarthritis is not clinically satisfactory., Conclusions: The presence of cruciate ligament damage and meniscal tears could be accurately assessed using the MR images obtained with our protocol. However, in the case of meniscal evaluation, occasional misdiagnosis did occur. The presence of cartilage damage and the degree of osteoarthritis could not be properly evaluated.

Gold-bead implantation as a method of pain treatment in dogs suffering from osteoarthritic disease is receiving increasing attention in veterinary medicine. For the present article, publications from veterinary books and journals were collected and evaluated, together with related articles in human medicine. After providing an overview of the historical use of gold and gold compounds, the technique of implanting this noble metal is introduced. The reasons for establishing the terms gold acupuncture and gold (bead) implantation are described, considering the question whether and what kind of methodological differences exist behind these terms. Next, previous publications concerning the effects of gold implantation in tissue are summarised. In 2002 it was proven that gold ions are released from the surface of gold implants by a process termed dissolucytosis. Subsequent publications further investigated details about the interaction between gold ions and tissue as well as the distribution pattern of bio-released ions. Gold compounds were previously used for chrysotherapy in human medicine until medication with fewer side effects became established. The anti-inflammatory and immuno-modulatory properties of gold compounds were used to treat rheumatoid arthritis. Current research aims to ascertain whether the anti-inflammatory and immuno-modulating effects of gold compounds are imitated by gold ions released from gold implants at a local level. In conclusion, the present review summarises important findings about the effects of gold implanted in tissue. However, further research is necessary to estimate the limitations and benefits of this auromedication.

Objective: To evaluate the load redistribution mechanisms in walking and trotting dogs with induced forelimb lameness., Animals: 7 healthy adult Beagles., Procedures: Dogs walked and trotted on an instrumented treadmill to determine control values for peak and mean vertical force as well as vertical impulse for all 4 limbs. A small sphere was attached to the ventral pad of the right forelimb paw to induce a reversible lameness, and recordings were repeated for both gaits. Additionally, footfall patterns were assessed to test for changes in temporal gait variables., Results: During walking and trotting, peak and mean vertical force as well as vertical impulse were decreased in the ipsilateral forelimb, increased in the contralateral hind limb, and remained unchanged in the ipsilateral hind limb after lameness was induced. All 3 variables were increased in the contralateral forelimb during trotting, whereas only mean vertical force and vertical impulse were increased during walking. Stance phase duration increased in the contralateral forelimb and hind limb during walking but not during trotting., Conclusions and Clinical Relevance: Analysis of the results suggested that compensatory load redistribution mechanisms in dogs depend on the gait. All 4 limbs should be evaluated in basic research and clinical studies to determine the effects of lameness on the entire body. Further studies are necessary to elucidate specific mechanisms for unloading of the affected limb and to determine the long-term effects of load changes in animals with chronic lameness.

Deafness is one of the most common sensory deficits in cats and dogs. Determining the scope and the cause of deafness is important for a possible therapy and can enable the handling of the animal by its owner as well as influence the decision about breeding with the affected animal. Electrodiagnostic testing using the brainstem auditory evoked response (BAER) enables an objective evaluation of the hearing function. Additionally, otoscopy and various diagnostic imaging techniques, including computed tomography, can help in determining the cause of deafness. While conductive deafness can often be treated, there is normally no satisfactory treatment for sensorineural deafness. In such cases, an important task of the attending veterinarian is to advise the owner on handling his/her animal.

Gold bead implantation/gold acupuncture is becoming increasingly used in veterinary medicine as a method of pain treatment in cases of osteoarthritic diseases. Part one of the overview dealing with the use of gold implants as a treatment of canine hip joint dysplasia (cHD) introduced the method of implanting gold in tissue and publications which investigated the subsequent effects of implantation. This article focuses on publications concerning the clinical effectiveness of gold implantation within the scope of pain therapy in cHD. Due to the study design, a classification using evidence-based levels (EbL) was carried out. Three double-blind, placebo-controlled randomised studies (EbL II) were considered together with three retrospective studies on own patients (EbL IV) and five case studies (EbL IV). While the case and retrospective studies reported impressive therapeutic success in treating cHD-incurred pain with gold implantation, a pain-reducing effect through gold implantation was only demonstrated in one of the three double-blind studies. The two remaining EbL II studies found no differences between the placebo-group and the group of dogs treated with gold implantation. In one of these two studies, kinematic and kinetic gait analyses were used for objective evaluation of the effects of the treatment. Thus, the only study that carried out an objective evaluation of the therapeutic result of gold implantation came to the conclusion that the method is ineffective. For a concluding assessment of gold implantation in the case of cHD, gait analysis studies investigating the effects of gold implantation in comparison to a standard treatment with nonsteroidal anti-inflammatory drugs are currently lacking.

Purpose: The cementless Bicontact total hip arthroplasty (THA) system (AESCULAP AG, Tuttlingen, Germany) was introduced in 1986/1987 and has been in successful clinical use in an unaltered form up to today. Although good long-term results with the Bicontact stem have been published, it is questionable whether the implant provides the criteria for a state-of-the-art stem regarding proximal bone stock preservation. The purpose of the study was to monitor the periprosthetic bone mineral density (BMD) in a prospective two-year follow-up dual-energy X-ray absorptiometry (DEXA) study., Methods: After power analysis, a consecutive series of 25 patients with unilateral Bicontact stem implantation was examined clinically and underwent DEXA examinations. Scans of seven regions of interest were taken preoperatively and at one week, six months, and one and two years., Results: One patient required stem revision due to a deep infection. The Harris Hip Score increased significantly by 44 points. The most significant bone loss was observed in the calcar region (R7) in the first six months (-19.2 %). It recovered in the following 18 months to -8.5 %. The BMD in the greater trochanter dropped significantly after six months and remained stable at this level. BMD exceeded baseline values in distal regions and even more in the lesser trochanter region after two years., Conclusions: We conclude that the Bicontact stem provides adequate proximal bone stock preservation. We observed some signs of stress shielding at the tip of the stem, which is inevitable to some degree in THA with cementless straight stems. However, in this prospective DEXA investigation, we showed that proximal off-loading does not occur after THA with the Bicontact system. Thus, we believe that this stem is still a state-of-the-art implant.

Background: Cell lines represent a key tool in cancer research allowing the generation of neoplasias which resemble initial tumours in in-vivo animal models. The characterisation of early tumour development is of major interest in order to evaluate the efficacy of therapeutic agents. Magnetic resonance imaging (MRI) based in-vivo characterisation allows visualisation and characterisation of tumour development in early stages prior to manual palpation. Contrast agents for MRI such as superparamagnetic iron oxide nanoparticles (SPIOs) and manganese chloride (MnCl2) represent powerful tools for the in-vivo characterisation of early stage tumours. In this experimental study, we labelled prostate cancer cells with MnCl2 or SPIOs in vitro and used 1 T MRI for tracing labelled cells in-vitro and 7 T MRI for tracking in an in-vivo animal model., Methods: Labelling of prostate cancer cells CT1258 was established in-vitro with MnCl2 and SPIOs. In-vitro detection of labelled cells in an agar phantom was carried out through 1 T MRI while in-vivo detection was performed using 7 T MRI after subcutaneous (s.c.) injection of labelled cells into NOD-Scid mice (n = 20). The animals were scanned in regular intervals until euthanization. The respective tumour volumes were analysed and corresponding tumour masses were subjected to histologic examination., Results: MnCl2in-vitro labelling resulted in no significant metabolic effects on proliferation and cell vitality. In-vitro detection-limit accounted 105 cells for MnCl2 as well as for SPIOs labelling. In-vivo 7 T MRI scans allowed detection of 103 and 104 cells. In-vivo MnCl2 labelled cells were detectable from days 4-16 while SPIO labelling allowed detection until 4 days after s.c. injection. MnCl2 labelled cells were highly tumourigenic in NOD-Scid mice and the tumour volume development was characterised in a time dependent manner. The amount of injected cells correlated with tumour size development and disease progression. Histological analysis of the induced tumour masses demonstrated characteristic morphologies of prostate adenocarcinoma., Conclusions: To the best of our knowledge, this is the first study reporting direct in-vitro MnCl2 labelling and 7 T based in-vivo MRI tracing of cancer cells in a model of prostate cancer. MnCl2 labelling was found to be suitable for in-vivo tracing allowing long detection periods. The labelled cells kept their highly tumourigenic potential in-vivo. Tumour volume development was visualised prior to manual palpation allowing tumour characterisation in early stages of the disease.

Purpose: On the basis of positive clinical results with mid- and long-term follow-up using the Mayo short stem, the Metha neck-preserving stem (BBraun, Aesculap, Tuttlingen, Germany) was introduced. The purpose of this study was to validate the implant design by direct acquisition of bone remodelling data from total hip arthroplasty (THA) recipients using dual-energy X-ray absorptiometry (DEXA)., Methods: After power analysis, 25 patients were included in this prospective study. Patients were examined clinically and underwent DEXA examinations preoperatively and postoperatively at one week, six months and one and two years after THA. Gruen zones were adapted to the short stem design (R1-R7)., Results: The Harris Hip Score (HHS) increased significantly by 31 points. No stem had to be revised. Bone mineral density (BMD) in the greater trochanter decreased significantly from 0.78 g/cm(2) postoperatively to 0.72 g/cm(2) two years after surgery. Marginal changes were seen in the lateral distal regions (R4-R5). In the minor trochanter region, BMD increased significantly after two years by 12.9%. In the calcar region, BMD exceeded the baseline value by 6.1% two years after implantation., Conclusions: Stress shielding seems to occur at the greater trochanter due to the vast cross-section of the implant. However, the aim of proximal load transfer of the Metha stem seems to be partially achieved. DEXA analysis revealed a concentrated load distribution on the medial portion of the femur, which is an important region to guarantee long-term implant survival.

Objective: To evaluate the use of retrospectively ECG-gated, contrast-enhanced, multi-detector row computed tomography (MDCT) for assessment of left ventricular function in dogs and to compare the results with those obtained by use of 2-D and M-mode echocardiographc techniques., Animals: 10 healthy Beagles., Procedures: Dogs underwent MDCT (performed by use of a 64-detector row CT system) and echocardiography under general anesthesia. Left ventricular end-systolic volume (ESV), end-diastolic volume (EDV), and ejection fraction (EF) were determined in MDCT-generated multiplanar reformatted images by use of Simpson and biplane area-length calculation methods. Results were compared with left ventricular ESV, EDV, and EF determined in echocardiographc images by use of Teichholz and bullet method calculations. Results were evaluated via Deming regression analysis and Pearson correlation tests. Bland-Altman analysis was used to assess limits of agreement and systematic errors between the 2 methods., Results: Mean values for EDV and ESV determined by use of MDCT were highly correlated with those determined by use of echocardiography, regardless of the calculation methods compared (r = 0.91 to 0.96); volumes determined by use of MDCT appeared to be higher than those determined by use of echocardiography, although most differences were nonsignificant. Mean EF determined by use of MDCT with the Simpson calculation method was highly correlated with that determined by use of echocardiography with bullet method calculations (r = 0.90)., Conclusions and Clinical Relevance: Results suggested that assessment of left ventricular volume and function in dogs is feasible with MDCT. To estimate left ventricular EF with MDCT. use of the Simpson calculation method is advised.

The design of the Bicontact® stem (BBraun, Aesculap, Tuttlingen, Germany) and the implantation technique have undergone no major alterations in the last 20 years leading, and good clinical results have been reported. The aim of our study was to investigate whether the implant encourages beneficial bone remodelling. Twenty-four patients were included in a prospective dual-energy X-ray absorptiometry (DEXA) study of this stem, after appropriate statistical power analysis. Preoperative and postoperative (1 week, 6 months, and 12 months) clinical and DEXA examinations were performed. The Harris Hip Score increased significantly by 39 points. The strongest decreases in BMD were observed in the greater trochanter region (-11%) and the calcar (-12%). In the second half of the study period the bone mineral density recovered slightly and even returned to baseline values in the lesser trochanter region. Therefore, proximal load transfer and physiological bone remodelling around the Bicontact® stem appeared to be achieved.

Subject: Etiologic diagnosis of a retrobulbar process is essential for the initiation of a causative therapy. Penetrating foreign bodies and tooth root abscesses, but also neoplastic processes have to be considered as a differential diagnosis for a retrobulbar swelling. The objective of this case report of a tiger with a retrobulbar process was to demonstrate the diagnostic value of computed tomography for making a causative diagnosis., Material and Methods: General examination of the tiger as well as subsequent computed tomography and surgical extraction of a fractured fourth premolar tooth of the left upper jaw was carried out under general anaesthesia. Collected alveolar tissue samples underwent cytological, histopathological and microbiological analyses., Results and Clinical Relevance: Computed tomography was able to allocate the origin of the retrobulbar abscess to the left 4th premolar tooth of the upper jaw. The cytological, histopathological and microbiological examination of the recovered alveolar material confirmed the diagnosis of purulent-necrotic abscess-forming inflammation as a result of a mixed bacterial infection. The present case report demonstrates the diagnostic accuracy of computed tomography for the diagnosis of a retrobulbar process of unknown origin.

To date it is unclear whether cementless total hip replacement (THR) in dogs is of clinical advantage in comparison to cemented THR with regard to lameness improvement. Thus the aim of this study was to compare objectively the development of the gait pattern after cemented and cementless THR in dogs. For this purpose, 18 adult dogs with hip dysplasia underwent computer-based gait analysis on an instrumented treadmill prior to unilateral THR and then again ten days, four weeks and four months after surgery. Analysed kinetic parameters were symmetry indices (SI) of vertical ground reaction forces (GRF), which included peak vertical forces (PFz), mean vertical forces (MFz), vertical impulse (IFz), and vertical ground reaction forces of the arthroplasty limbs only. Analysed kinematic parameters were range-of-motion and the flexion and extension angles of hip, stifle and hock joints. The symmetry indice for PVF, MFz and IFz decreased to a value less than six in both THR groups four months after surgery, which is defined as not lame. Improvement in lameness of the arthroplasty limbs during the examination period of four months was not significantly different between the cemented and cementless groups. The results suggest that within a short-term observation period of four months after surgery, neither cementless nor cemented THR have a greater advantage with regard to lameness improvement. Additional studies with larger pools of subjects and longer time periods for follow-up examinations are necessary to verify these findings.

Multipotency and self-renewal are considered as most important features of stem cells to persist throughout life in tissues. In this context, the role of HMGA proteins to influence proliferation of adipose-derived mesenchymal stem cell (ASCs) while maintaining their multipotent and self-renewal capacities has not yet been investigated. Therefore, extracellular HMGA1 and HMGA2 application alone (10-200 ng/mL) and in combination with each other (100, 200 ng/mL each) was investigated with regard to proliferative effects on canine ASCs (cASCs) after 48 hours of cultivation. Furthermore, mRNA expression of multipotency marker genes in unstimulated and HMGA2-stimulated cASCs (50, 100 ng/mL) was analyzed by RT-qPCR. HMGA1 significantly reduced cASCs proliferation in concentrations of 10-200 ng/mL culture medium. A combination of HMGA1 and HMGA2 protein (100 and 200 ng/mL each) caused the same effects, whereas no significant effect on cASCs proliferation was shown after HMGA2 protein application alone. RT-qPCR results showed that expression levels of marker genes including KLF4, SOX2, OCT4, HMGA2, and cMYC mRNAs were on the same level in both HMGA2-protein-stimulated and -unstimulated cASCs. Extracellular HMGA protein application might be valuable to control proliferation of cASCs in context with their employment in regenerative approaches without affecting their self-renewal and multipotency abilities.

Animals alter their locomotor mechanics to adapt to a loss of limb function. To better understand their compensatory mechanisms, this study evaluated the changes in the fore-aft ground forces to forelimb lameness and tested the hypothesis that dogs unload the affected limb by producing a nose-up pitching moment via the exertion of a net-propulsive force when the lame limb is on the ground. Seven healthy Beagles walked and trotted at steady speed on an instrumented treadmill while horizontal force data were collected before and after a moderate lameness was induced. Peak, mean and summed braking and propulsive forces as well as the duration each force was exerted and the time to reach maximum force were evaluated for both the sound and the lame condition. Compared with the sound condition, a net-propulsive force was produced by the lame diagonal limbs due to a reduced braking force in the affected forelimb and an increased propulsive force in the contralateral hindlimb when the dogs walked and trotted. To regain pitch stability and ensure steady speed for a given locomotor cycle, the dogs produced a net-braking force when the sound diagonal limbs were on the ground by exerting greater braking forces in both limbs during walking and additionally reducing the propulsive force in the hindlimb during trotting. Consistent with the proposed mechanism, dogs maximize their double support phases when walking. Likely associated with the fore-aft force adaptations to lameness are changes in muscle recruitment that potentially result in short- and long-term effects on the limb and trunk muscles.

Objectives: To compare the improvement in degree of lameness following surgical repair of cranial cruciate ligament rupture in dogs using computer-assisted gait analysis., Methods: Two groups of 14 dogs were used. One group was treated by a capsular-fascial imbrication method, and the other group by tibial plateau levelling osteotomy (TPLO). All dogs underwent gait analysis prior to surgery, as well as at four days, four weeks, and four months after surgery. Symmetry indices of vertical ground reaction forces as well as vertical ground reaction forces in % body weight, joint angles and certain gait cycle parameters were evaluated., Results: Four months after surgery, the degree of lameness expressed as symmetry index for peak vertical force for the TPLO group (5.83%) was not significantly different to the capsular-fascial imbrication group (19.05%). Within the TPLO group, there was a significantly increased ability to extend the stifle joint four months after surgery. The stifle motion pattern of the capsular-fascial imbrication group as well as the range-of-motion in both groups showed very little change at the time of the last gait analysis. The complication rate was greater in the TPLO group than in the capsular-fascial group., Clinical Significance: In conclusion the results suggest that the TPLO method leads to a faster recovery and improved limb function in comparison to the capsular-fascial imbrication method four months after surgery. Further analyses are needed to determine if the TPLO method is superior concerning long-term joint stability.

Background/aim: Administration of stem cells is a promising novel approach for treatment of neurodegenerative diseases. For in vivo monitoring of transplanted cells, non-invasive imaging modalities are needed. In this study we determined the tracking efficiency of a superparamagnetic iron oxide (SPIO)-labelled canine cell line (MTH53A) in vitro as well as the human CD34(+) umbilical cord blood stem cells (hUCBCs) in vitro and in vivo efficiency by magnetic resonance imaging (MRI)., Materials and Methods: SPIO-labelled MTH53A cells and hUCBCs were scanned in agar gel phantoms at 1.0 T or 7.0 T. For in vivo detection, 100,000 labelled hUCBCs were injected into the spinal cord of a transgenic amyotrophic lateral sclerosis (ALS) mouse and scanned at 7.0 T., Results: In vitro, 100,000 MTH53A cells and 250,000 hUCBCs were visible at 1.0 T. Scanning with 7.0 T revealed 25,000 detectable MTH53A cells. In vivo, 7.0 T MRI showed clear signals of 100,000 implanted cells., Conclusion: MRI combined with SPIO nanoparticles provides valuable potential for non-invasive, non-toxic in vivo tracking of cells implanted into the spinal cord.

Objective: To radiographically evaluate periprosthetic femoral bone contrast and assess alignment of the prosthetic stem after uncemented and cemented total hip replacement (THR)., Study Design: Prospective clinical study., Animals: Dogs (n = 15)., Methods: Dogs were classified into uncemented (n = 8) and cemented (n = 7) THR groups. Radiographs were analyzed using image processing software to evaluate femoral bone contrast (gray scale value, GV) for each and combined modified Gruen zone(s) immediately and 4 months after THR. Modified Gruen zones were classified into 5 zones to analyze retrospectively the regional radiographic GV of the femur around uncemented and cemented prosthetic stem. Alignment of prosthetic stem was assessed immediately and 4 months postoperatively. Variables were compared by use of 2-tailed t-test, with P < .05 considered significant., Results: Zone 1 showed significant decrease in the mean bone GV 4 months after uncemented THR. No differences in zones 1-5 after 4 months of cemented THR. Combined zones showed significant decrease in overall mean bone GV 4 months after uncemented THR. No changes were observed 4 months after cemented THR. Number of limbs with varus-aligned femoral stem markedly increased after 4 months of uncemented THR., Conclusions: Regional bone contrast and prosthetic stem alignment vary with the design of THR., (© Copyright 2011 by The American College of Veterinary Surgeons.)

Background: Diagnosis of extracardiac intrathoracic vascular anomalies is of clinical importance, but remains challenging. Traditional imaging modalities, such as radiography, echocardiography, and angiography, are inherently limited by the difficulties of a 2-dimensional approach to a 3-dimensional object. We postulated that accurate characterization of malformations of the aorta would benefit from 3-dimensional assessment. Therefore, multidetector-row computed tomography (MDCT) was chosen as a 3-dimensional, new, and noninvasive imaging technique. The purpose of this study was to evaluate patients with 2 common diseases of the intrathoracic aorta, either patent ductus arteriosus or vascular ring anomaly, by contrast-enhanced 64-row computed tomography., Results: Electrocardiography (ECG)-gated and thoracic nongated MDCT images were reviewed in identified cases of either a patent ductus arteriosus or vascular ring anomaly. Ductal size and morphology were determined in 6 dogs that underwent ECG-gated MDCT. Vascular ring anomalies were characterized in 7 dogs and 3 cats by ECG-gated MDCT or by a nongated thoracic standard protocol. Cardiac ECG-gated MDCT clearly displayed the morphology, length, and caliber of the patent ductus arteriosus in 6 affected dogs. Persistent right aortic arch was identified in 10 animals, 8 of which showed a coexisting aberrant left subclavian artery. A mild dilation of the proximal portion of the aberrant subclavian artery near its origin of the aorta was present in 4 dogs, and a diverticulum analogous to the human Kommerell's diverticulum was present in 2 cats., Conclusions: Contrast-enhanced MDCT imaging of thoracic anomalies gives valuable information about the exact aortic arch configuration. Furthermore, MDCT was able to characterize the vascular branching patterns in dogs and cats with a persistent right aortic arch and the morphology and size of the patent ductus arteriosus in affected dogs. This additional information can be of help with regard to improved diagnoses of thoracic anomalies and the planning of surgical interventions.

Background: Disorders of histiocytic origin affecting humans and dogs share various similarities. Canine disseminated histiocytic sarcoma (DHS) (formerly known as malignant histiocytosis) is an aggressive neoplasm of interstitial dendritic cells (DCs). The receptor for glycation end products (RAGE) and the high mobility group box1 protein (HMGB1) have been shown to be required for the maturation and migration of DCs. Thus, deregulation of the expression of these genes could have a major effect on the progression of histiocytic disorders., Materials and Methods: Neoplastic canine DHS samples and non-neoplastic control samples were analysed immunohistochemically and via real-time PCR., Results: Significant down-regulation of RAGE in the lung tumour samples and down-regulation of HMGB1 in the lung, lymph node and spleen tumour samples were detected compared to their non-neoplastic counterparts., Conclusion: RAGE and HMGB1 expression down-regulation in canine DHS points to a role in the progression of histiocytic disorders.
