Cattle suffering from inflammatory infection display sickness and pain-related behaviours. As these behaviours may be transient and last only a few hours, one may miss them. The aim of this study was to assess the benefit of combining continuous monitoring of cow behaviour via collar-attached accelerometers with direct visual observations to detect sickness and pain-related behavioural responses after a systemic inflammatory challenge (intravenous lipopolysaccharide injection) in cows of two different ages, proven by clinical, physiological and blood parameters. Twelve cloned Holstein cows (six 'old' cows aged 10-15 years old and six 'young' cows aged 6 years old) were challenged and either directly observed at five time-points from just before the lipopolysaccharide injection up to 24 h post-injection (hpi) or continuously monitored using collar-attached accelerometers in either control or challenge situations. Direct observations identified specific sickness and pain behaviours (apathy, changes in facial expression and body posture, reduced motivation to feed) expressed partially at 3 hpi and fully at 6 hpi. These signs of sickness and pain behaviours then faded, and quicker for the young cows. Accelerometers detected changes in basic activities (low ingesting, low ruminating, high inactivity) and position (high time standing up) earlier and over a longer period of time than direct observations. The combination of sensors and direct observations improved the detection of behavioural signs of sickness and pain earlier on and over the whole study period, even when direct signs were weak especially in young cows. The Open Science movement aims at ensuring accessibility, reproducibility, and transparency of research. The adoption of Open Science practices in animal science, however, is still at an early stage. To move ahead as a field, we here provide seven practical steps to embrace Open Science in animal science. We hope that this paper contributes to the shift in research practices of animal scientists towards open, reproducible, and transparent science, enabling the field to gain additional public trust and deal with future challenges to guarantee reliable research. Disease and stress can disrupt the circadian rhythm of activity in animals. Sensor technologies can automatically detect variations in daily activity, but it remains difficult to detect exactly when the circadian rhythm disruption starts. Here we report a mathematical Fourier-Based Approximation with Thresholding (FBAT) method designed to detect changes in the circadian activity rhythm of cows whatever the cause of change (typically disease, stress, oestrus). We used data from an indoor positioning system that provides the time per hour spent by each cow resting, in alleys, or eating. We calculated the hourly activity level of each cow by attributing a weight to each activity. We considered 36-h time series and used Fourier transform to model the variations in activity during the first and last 24 h of these 36-h series. We then compared the Euclidian distance between the two models against a given threshold above which we considered that rhythm had changed. We tested the method on four datasets (giving a cumulative total of ~120000 cow*days) that included disease episodes (acidosis, lameness, mastitis or other infectious diseases), reproductive events (oestrus or calving) and external stimuli that can stress animals (e.g. relocation). The method obtained over 80% recall of normal days and detected 95% of abnormal rhythms due to health or reproductive events. FBAT could be implemented in precision livestock farming system monitoring tools to alert caretakers to individual animals needing specific care. The FBAT method also has the potential to detect anomalies in humans to guide healthcare intervention or in wild animals to detect disturbances. Maladjusted cubicles for dairy cattle may cause increased skin alterations, lameness, and dirtiness. The International Commission of Agricultural and Biosystems Engineering has produced several recommendations for cubicle design, but a previous study showed that not all of them seem efficient. Here, we aim to refine and complete these recommendations. We collected data on 76 dairy farms (2,404 cows). We modeled the association between combinations of cubicle properties (e.g., type of bedding litter) and dimensions (e.g., cubicle width) relative to cow size, and prevalence of cow skin alterations, lameness, and dirtiness. We used weighted multivariable logistic regression models to predict the presence of skin alteration on the carpus; the neck, shoulder, and back; the flank, side, and udder; and the tarsus or hindquarters. We also evaluated the presence of lameness as well as the dirtiness of the lower hind legs including hocks; the hindquarters, upper hind legs, and flank; the cow rear including tail; and the udder. The risk factors highlighted led us to recommend (1) position cubicles in a way that leaves more than 1 m of clearance from any obstacle in front of the cubicle; (2) if there is an obstacle on the lateral plane (i.e., where the cubicle partition is) in front ahead of the cow, put the obstacle in front of the fore knees; (3) if there is an obstacle in front of the cow on the median plane (e.g., neck or front rail), the position the obstacle between 1.25 and 1.5 of the cow length from the curb and between 1.0 and 1.25 of its height; (4) use curb height between 0.11 and 0.15 of cow height with no sharp edges on the curb; (5) use round or at least has no sharp edges brisket board; (6) use a stone-free soil instead of concrete or use a mattress thicker than 1 cm, with microrelief, and a soft fixing area at the curb, (7) litter with straw (rather than nothing or sawdust) and keep it dry. This risk factor analysis should be followed by experiments in controlled environments to further validate these conclusions and used to update the International Commission of Agricultural and Biosystems Engineering recommendations., (Copyright © 2021 American Dairy Science Association. Although there now exists a wide range of policies, instruments and regulations, in Europe and increasingly beyond, to improve and safeguard the welfare of farmed animals, there remain persistent and significant welfare issues in virtually all types of animal production systems ranging from high prevalence of lameness to limited possibilities to express natural behaviours. Protocols and indicators, such as those provided by Welfare Quality, mean that animal welfare can nowadays be regularly measured and surveyed at the farm level. However, the digital revolution in agriculture opens possibilities to quantify animal welfare using multiple sensors and data analytics. This allows daily monitoring of animal welfare at the group and individual animal level, for example, by measuring changes in behaviour patterns or physiological parameters. The present paper explores the potential for developing innovations in digital technologies to improve the management of animal welfare at the farm, during transport or at slaughter. We conclude that the innovations in Precision Livestock Farming (PLF) offer significant opportunities for a more holistic, evidence-based approach to the monitoring and surveillance of farmed animal welfare. To date, the emphasis in much PLF technologies has been on animal health and productivity. This paper argues that this emphasis should not come to define welfare. What is now needed is a coming together of industry, scientists, food chain actors, policy-makers and NGOs to develop and use the promise of PLF for the creative and effective improvement of farmed animal welfare.

Design of cubicles and self-locking barriers can affect cow skin alterations, lameness, and dirtiness. We investigated whether the International Commission of Agricultural and Biosystems Engineering (CIGR, Gainesville, FL)-recommended cattle housing design and dimensions actually improve cow welfare. We recorded individual cow body dimensions and assessed skin alterations, dirtiness, and lameness on 3,841 cows from 131 loose housing dairy farms (76 farms with cubicles and 55 straw-yard systems). We recorded the dimensions of cubicles (e.g., width, length, and so on) and of the self-locking barrier (e.g., top rail height and so on) for each farm. We then compared whether these dimensions would match with the individual cow body dimensions and whether compliance was associated with the occurrence of skin alterations, lameness, and dirtiness. Most cows (69.2%) had at least one skin alteration, on the tarsus (41.2%); neck, shoulder, or back (28.2%); hindquarters (22.0%); carpus (21.2%); and flank, side, or udder (11.6%). Lameness prevalence reached 17.9%. Most cows (83.1%) were scored dirty in at least one body region, mainly on the lower hind legs including hocks (81.4%), hindquarters (41.8%), and udder (21.0%). Cubicle recommendations were mostly met for cubicle resting length (75.9% of cubicles) and neck rail distance (NRD, 60.7%) but less so for overall cubicle length (CL, 38.8%), cubicle width (CW, 30.9%), neck rail height (NRH, 22.5%), head space (HS, 15.8%), partition zone for controlling lying position (ii zone, 15.7%), head and lunging space (HLS, 10.6%), partition head zone (i zone, 9.4%), and partition zone for pelvis freedom (iii zone, 6.0%). Compliance with design recommendations was associated with fewer skin alterations on neck (recommendation met for CW, CL, HS, and NRD and i zone), hindquarters (CW, CL, NRD), tarsus (CW, i and ii zones), and carpus (CW, HS, i and ii zones); less lameness (CW, NRH, i zone); and less dirtiness on the hindquarters (CW, HLS, NRD), lower hind legs (iii zone), and udder (CW, CL, HLS, NRD). Compliance with recommended i zone and ii zone design was associated with less injury and lameness but more dirtiness, whereas the opposite was true for the iii zone. Self-locking barrier recommendations were mostly met for bottom rail height (68.2%), separation wall width (SWW, 68.3%), and top rail height (TRH, 56.9%) and less often met for separation wall height (36.3%) and height difference between feeding floor and walking alley (26.5%). Risk for skin alterations on carpus and neck only decreased when SWW and TRH met recommendations. In conclusion, compliance with CIGR recommendations for some cubicle dimensions and neck rail position was associated with lower risk for cow welfare. However, the CIGR recommendations on cubicle partitions and self-locking barriers still leave welfare at risk and so need to be refined through further research with close observation of cow lying and feeding behavior., (Copyright © 2019 American Dairy Science Association. Many biological studies seek to confirm the effect of a treatment on the levels of chemical markers in biological tissues. This requires choosing the analytical method best able to detect the difference between basal levels and those found after treatment. We propose a new approach to calculate a criterion we call the 'discrimination threshold' (DT), and we applied it to an analytical method that we developed to determine cortisol in cattle plasma to detect stress. DT is derived from the measurement uncertainty (MU), and combines the variabilities of both the analytical method and the marker. The uncertainty of the analytical method comes from the method validation study. The marker variability (here cortisol) is modeled from a literature review. The graphical representation of DT allows estimating the applicability of the method. Under temperate climates, cattle are often at pasture in summer and are not necessarily provided with shade. We aimed at evaluating in a temperate region (Belgium) to what extent cattle may suffer from heat stress (measured through body temperature, respiration rate and panting score, cortisol or its metabolites in milk, and feces on hot days) and at assessing the potential benefits of shade. During the summer of 2012, 20 cows were kept on pasture without access to shade. During the summer of 2011, ten cows had access to shade (young trees with shade cloth hung between them), whereas ten cows had no access. Climatic conditions were quantified by the Heat Load Index (HLI). In animals without access to shade respiration rates, panting scores, rectal temperatures, and milk cortisol concentrations increased as HLI increased in both 2011 and 2012. Fecal cortisol metabolites varied with HLI in 2011 only. When cattle had access to shade, their use of shade increased as the HLI increased. This effect was more pronounced during the last part of the summer, possibly due to better acquaintance with the shade construction. In this case, shade use increased to 65% at the highest HLI (79). Shade tempered the effects on respiration, rectal temperature, and fecal cortisol metabolites. Milk cortisol was not influenced by HLI for cows using shade for > 10% of the day. Therefore, even in temperate areas, cattle may suffer from heat when they are at pasture in summer and providing shade can reduce such stress.

Biological rhythms are an essential regulator of life. There is evidence that circadian rhythm of activity is disrupted under chronic stress in animals and humans, and it may also be less marked during diseases. Here we investigated whether a detectable circadian rhythm of activity exists in dairy cows in commercial settings using a real-time positioning system. We used CowView (GEA Farm Technologies) to regularly record the individual positions of 350 cows in a Danish dairy farm over 5 mo and to infer the cows' activity (resting, feeding, in alley). We ran a factorial correspondence analysis on the cows' activities and used the first component of this analysis to express the variations in activity. On this axis, the activities obtained the following weights: resting = -0.15; in alleys = +0.12; feeding = +0.34. By applying these weights to the proportions of time each cow spent on each of the 3 activities, we were able to chart a circadian rhythm of activity. We found that average level of activity of a cow on a given day and its variations during that day varied with specific states (i.e., estrus, lameness, mastitis). More specifically, circadian variations in activity appeared to be particularly sensitive and to vary 1 to 2 d before the farmer detected a disorder. Human-animal relationships are essential for dairy farming. They affect work comfort and efficiency, as well as milk production. A poor human-animal relationship can result in stress and accidents to both animals and caretakers and needs to be improved. However, many studies have demonstrated the multifactoriality of these relationships. We aimed at assessing the relative importance of the various factors expected to be associated with poor human-animal relationships. On 118 dairy farms, we applied a standardized avoidance distance test to cows at the feeding rack. The sample of farms covered a wide range of situations: lowland versus highland, small versus medium size farms, cubicles versus deep-bedded systems, milking parlor versus automatic milking systems, and Holstein versus Montbéliarde breeds. We used Poisson regression to analyze the links between the number of cows that accepted being touched, and farm characteristics, animals, management, and farmers' attitudes. A multivariate analysis yielded a final model that explained 32.7% of the variability between farms. Calving conditions ("Main calving location" and "Cleaning or adding litter after calving") accounted for a significant part of the variability observed (respectively 25.8 and 13.6% of variability explained by the model, SSB). Fewer cows accepted being touched on farms where the main calving location was in the barn, and where farmers cleaned or added litter after calving. The proportion of cows that accepted being touched increased with the proportion of lean cows in the herd (18.8%), with worker/cow ratio on the farm (11.7%), when farmers considered "health" or "human-cow relationships" as most important issues for farm success (10.4%), and with farmers' years of experience (10.8%). Farmers with more negative behavioral attitudes toward cows had a lower proportion of cows that accepted being touched (8.9%). In conclusion, the human-animal relationship was not found to be associated with farm characteristics (e.g., housing or milking system) but varied with farmers' attitudes and management. We confirm that cows' fear of people is linked to negative attitudes displayed by caretakers toward cows, and is reduced in farms where several caretakers are present. Under the assumption that milk yield may be reduced in herds with impaired welfare, the present study aimed at investigating whether milk yield could be used as a reliable indicator of welfare. In 125 commercial French dairy herds, the association between the welfare of the herd (evaluated using the Welfare Quality assessment protocol) and cow milk yield was investigated using linear mixed models. Positive associations were identified between milk yield and low aggressions between cows and good emotional state of the herd but there was a negative association with good health assessed through the occurrence of diseases and injuries. These opposite associations resulted in no association with the overall welfare of the herd. Rumenocentesis is commonly used to collect rumen fluid to screen for acidosis. This study was designed to investigate whether rumenocentesis induced pain and stress and, if so, whether local anaesthesia could limit this. Twenty-four dairy cows were assigned to one of three treatments: (1) rumenocentesis with local anaesthesia (AR); (2) rumenocentesis without local anaesthesia (R); and (3) local anaesthesia only (A). Treatments were performed in a restraining cage. The cows were placed in the cage on three consecutive days and anaesthesia and/or rumenocentesis was performed on the second day. Blood samples for cortisol determination and heart rate were recorded from 0.25 h before treatment until 4h after. Behaviour was noted while the cows were caged. Feed intake and milk production were measured the week before treatment, on the day of treatment, and the day after. With all three treatments, cortisol concentrations and heart rate were increased while cows were in the cage. Cortisol, cardiac and behavioural responses were not significantly higher in the R and AR treatments than the A group. Cortisol concentrations and heart rate did not change between days. Feed intake and milk production were unaffected by the treatments. Background: Beef suckler farms (194 farms throughout 13 counties) were assessed once with housed cattle and once with cattle at grass using an animal welfare index (AWI). Twenty-three of the 194 farms were revisited a year later and re-evaluated using the AWI and the Tier-Gerechtheits-Index 35L/2000 (TGI35L/2000). Thirty-three indicators were collected in five categories: locomotion (5 indicators); social interactions (between animals) (7), flooring (5), environment (7) and Stockpersonship (9). Three indicators relating to the size of the farm were also collected.Improving animal welfare is an increasingly important aspect of livestock production systems predominantly due to increased consumer concern about the source of animal products. The objectives were (i) to evaluate animal welfare of Irish beef suckler herds using an animal welfare index (AWI), (ii) to examine correlations between parameters, how they influence the AWI and investigate the applicability of the parameters used, (iii) to investigate the impact of the activity of the farmer (full-time or part-time), the interest of the farmer and the number of animals on the AWI., Results: The mean AWI was 65% and ranged from 54% to 83%. The grazing period represented 16.5% of the total points of the AWI. Seventy percent of the farms were rated as "Very Good" or "Excellent". There was no difference (P > 0.05) in AWI between full-time and part-time farmers. Part-time farmers had greater (P = 0.01) "social interactions": calving (P = 0.03) and weaning (P < 0.001) scores. Full-time farmers had cleaner animals (P = 0.03) and their animals had less lameness (P = 0.01). The number of animals on-farm and the interest of the Stockperson were negatively and positively correlated (P = 0.001), respectively, with the AWI. A hierarchical classification was performed to examine how the indicators influenced the AWI., Conclusion: The AWI was easily applicable for an on-farm evaluation of welfare. The Stockpersonship was an important factor in determining the AWI (11% of the total variation) more specifically, the interest of the farmer. Part and full-time farming did not differ (P > 0.05) in AWI scores. This method could, with further development, be used in countries with both intensive and/or extensive production systems and would require substantially less resources than animal-based methods.

Calves can develop long-lasting social relationships with peers. We examined the strength of the relationships between calves according to the time they had been together. Twenty-four female dairy calves were assigned to six groups of four animals (Type-1 partners) at 0.5 month of age. At 3.5 months of age, they were mixed with other calves (Type-2 partners) to form groups of 14. Type-3 partners were calves added to the experimental groups after 5.25 months. The calves stayed together until 1.5 years of age. Social preferences between the three partner types were examined in a Y-maze, and the position and activity of animals in the barn and pasture were followed in three periods. Behavioural synchrony, distance between animals, proximity and nearest neighbour were analysed. The calves more frequently butted Type-3 than Type-1 partners in the Y-maze (P<0.05). They spent more time in proximity to Type-1 partners, and these were more often the nearest neighbours than other partners (P<0.001). Synchrony and distance between animals were greater at pasture than in the barn (P<0.01). Calves seem to form preferential relationships before 3.5 months of age. The presence of the caregiver around feeding favors the development of a human-animal relationship. To understand the underlying mechanism, we tested various temporal associations between food distribution and human presence: from an early age, a person was repeatedly present for 2 min just before milk distribution ("Forward"), during milk distribution ("Simultaneous"), and 20 min afterwards ("Delayed"). The "Control" group received no human contacts. During the treatments, "Forward" and "Delayed" lambs had more physical contacts with the person than "Simultaneous" lambs. When tested in unfamiliar environments, they stood longer near the person than did "Control" or "Simultaneous" lambs, which did not differ. Only "Forward" and "Delayed" lambs bleated when separated from the person. Fasting before testing had no effect. "Forward" and "Delayed" seemed to produce the same human-animal relationship, showing that this did not rely only on a conditioning process associating the caregiver with food. The study of emotions in animals can be approached thanks to a framework derived from appraisal theories developed in cognitive psychology, according to which emotions are triggered when the individual evaluates challenging events. This evaluation is based on a limited number of criteria such as the familiarity and the predictability of an event. If animals are able to experience emotions rather than simply displaying reflex responses to their environment, then their appraisal of events should, as in humans, modulate their emotional responses. We tested this hypothesis by comparing vocalisations, feeding behaviour, and the startle and cardiac responses of lambs submitted to a sudden event that could or could not be predicted. Lambs able to predict the sudden event thanks to a light cue (associative predictability) showed weaker suddenness-induced startle and cardiac responses and spent more time feeding than their counterparts, thus supporting the existence of an emotional experience in these animals. Furthermore, lambs submitted to the regular appearance of the sudden event (temporal regularity) vocalised less and left less unconsumed food deliveries than lambs submitted to random appearances of the sudden event (controls). These results underline that the cognitive abilities of animals should be taken into account when assessing their emotional experiences and more generally their mood states, which are underlying factors of animal welfare.

Measuring HPA axis activity is the standard approach to the study of stress and welfare in farm animals. Although the reference technique is the use of blood plasma to measure glucocorticoid hormones (cortisol or corticosterone), several alternative methods such as the measurement of corticosteroids in saliva, urine or faeces have been developed to overcome the stress induced by blood sampling itself. In chronic stress situations, as is frequently the case in studies about farm animal welfare, hormonal secretions are usually unchanged but dynamic testing allows the demonstration of functional changes at several levels of the system, including the sensitization of the adrenal cortex to ACTH and the resistance of the axis to feedback inhibition by corticosteroids (dexamethasone suppression test). Beyond these procedural aspects, the main pitfall in the use of HPA axis activity is in the interpretation of experimental data. The large variability of the system has to be taken into consideration, since corticosteroid hormone secretion is usually pulsatile, follows diurnal and seasonal rhythms, is influenced by feed intake and environmental factors such as temperature and humidity, age and physiological state, just to cite the main sources of variation. The corresponding changes reflect the important role of glucocorticoid hormones in a number of basic physiological processes such as energy metabolism and central nervous system functioning. Furthermore, large differences have been found across species, breeds and individuals, which reflect the contribution of genetic factors and environmental influences, especially during development, in HPA axis functioning. Usually, these results will be integrated with data from behavioral observation, production and pathology records in a comprehensive approach of farm animal welfare.

Measurement of heart rate variability (HRV) is a non-invasive technique that can be used to investigate the functioning of the autonomic nervous system, especially the balance between sympathetic and vagal activity. It has been proven to be very useful in humans for both research and clinical studies concerned with cardiovascular diseases, diabetic autonomic dysfunction, hypertension and psychiatric and psychological disorders. Over the past decade, HRV has been used increasingly in animal research to analyse changes in sympathovagal balance related to diseases, psychological and environmental stressors or individual characteristics such as temperament and coping strategies. This paper discusses current and past HRV research in farm animals. First, it describes how cardiac activity is regulated and the relationships between HRV, sympathovagal balance and stress and animal welfare. Then it proceeds to outline the types of equipment and methodological approaches that have been adapted and developed to measure inter-beats intervals (IBI) and estimate HRV in farm animals. Finally, it discusses experiments and conclusions derived from the measurement of HRV in pigs, cattle, horses, sheep, goats and poultry. Emphasis has been placed on deriving recommendations for future research investigating HRV, including approaches for measuring and analysing IBI data. Data from earlier research demonstrate that HRV is a promising approach for evaluating stress and emotional states in animals. It has the potential to contribute much to our understanding and assessment of the underlying neurophysiological processes of stress responses and different welfare states in farm animals.

It is now widely accepted that good welfare is not simply the absence of negative experiences, but rather is primarily the presence of positive experiences such as pleasure. However scientific investigation of positive emotions has long been neglected. This paper addresses two main issues: first, it reviews the current state of scientific knowledge that supports the existence of positive affective states in animals and, second, it suggests possible applications of this knowledge that may enhance quality of life under animal management conditions. In the first part of the paper, recent advances in psychology and neuroscience are reviewed to provide pragmatic frameworks based on cognitive processes (such as positive anticipation, contrast and controllability) for further investigations of positive emotions in animals. Thereafter, the neurobiological bases of positive emotions are highlighted in order to identify behavioral and physiological expressions of positive experiences in animals. Monitoring both the autonomic nervous system (via heart rate and its variability) and the immune system could offer relevant tools to better assess emotional states in animals, complementary to classical adrenocortical measures. In the second part of the paper, useful strategies for enhancing positive experiences (such as physical, social and cognitive enrichment or putative genetic selection) are outlined. Then this paper emphasizes practical applications for assessing and promoting positive emotions that may help in providing animals with a better quality of life. Play, affiliative behaviors and some vocalizations appear to be the most promising convenient indicators for assessing positive experiences in laboratory and farm animals under commercial conditions.

Appraisal theories state that an emotion results from an individual's evaluation of a stimulating event, which is based on a set of criteria. Unfamiliarity induced an orientation response. The combination of suddenness and unfamiliarity enhanced both the heart rate increase and the behavioral orientation response. Appraisal theories provide a framework that gives insight into emotions and could allow comparisons across species. According to these theories, events are first evaluated on their suddenness, novelty, and unpredictability. The authors examined the ability of lambs (Ovis aries) to evaluate an event according to these 3 criteria through 3 tests. The lambs responded to suddenness with a startle response coupled with an increase in heart rate and to novelty with an orientation response coupled with an increase of vagal activity. There was no clear evidence that lambs can detect the predictability of an event. One of the major topics of applied ethology is the welfare of animals reared by humans. Welfare can be defined as a state of harmony between an individual and its environment. Any marked deviation from this state, if perceived by the individual, results in a welfare deficit due to negative emotional experiences. In humans, verbal language helps to assess emotional experiences. In animals, only behavioural and physiological measurements help to detect emotions. However, how to interpret these responses in terms of emotional experiences remains an open question. The information on the cognitive abilities of farm animals, which are available but scattered, could help the understanding of their emotions. We propose a behavioural approach based on cognitive psychology: emotions can be investigated in farm animals in terms of the individual's appraisal of the situation. This evaluative process depends on: (a) the intrinsic characteristics of the eliciting event (suddenness, novelty, pleasantness); (b) the degree of conflict of that event with the individual's needs or expectations; and (c) the individual's coping possibilities offered by the environment. The result of such an evaluation determines the negative versus positive emotions. We propose an analysis of the emotional repertoire of farm animals in terms of the relationship between the evaluative process of the event on the one hand and the behavioural and physiological responses on the other hand.
