Trauma or the tourniquet used in orthopedic surgery is often associated with ischemia-reperfusion (I/R) injury with a consequent decrease of muscle power. To explore whether components of the neuromuscular junction (NMJ) are involved in this muscle dysfunction, NMJs were ultrastructurally characterized in the extensor digitorum longus muscle of rats at reperfusion times of 1, 24, 72, and 168 h after a 120-min arterial occlusion. Disorganization of the presynaptic membrane and mitochondrial injury was noted at 1 h, followed by fragmentation and partial engulfment of nerve terminals by Schwann cells at 24 and 72 h. The magnitude of degenerative changes declined at 168 h, suggesting the commencement of regeneration. The postsynaptic membrane remained intact throughout the whole period. In our previous study, deafferentation with pretreatment of the sciatic nerve with capsaicin, which reduces neurogenic inflammation and has a selective effect on nociceptive fibers, improved functional recovery of the muscle after I/R. The present results document a significantly delayed structural regeneration of the motor nerve terminals after combined capsaicin and I/R treatment. Since capsaicin treatment alone had no discernible effect on the structure of NMJs, the findings point to a possibly indirect effect of capsaicin on the motor nerves, which may predispose them to increased susceptibility unmasked only by a subsequent injury. The mismatch between the enhanced functional improvement of the muscle and delayed regeneration of the nerve after capsaicin pretreatment questions the efficient use of such deafferentation to protect the integrity of neuromuscular junctions in I/R injury.

The anatomical connection of the magnocellular isthmic nucleus with the optic tectum was investigated with the axonal tracer biotinylated dextran amine. Following iontophoretic injection of this tracer into different areas of the chick optic tectum, neurones of both magno- and parvocellular isthmic nuclei were labelled together in a topographical arrangement. The number of labelled neurones in the parvocellular nucleus was generally higher than in magnocellular. Using different locations of the tracer injections, systematic shifts in the location of the labelled neurones were detected. The labelled axons were seen to course along the shortest possible distance between the injection site and the cells of origin, i.e., the ventral part of the tectum received projections from neurones located ventrally in the isthmic nuclei, the dorsal tectum from neurones in the dorsal part, and the lateral extension of the tectum from neurones lying midway along the nuclei. This parallel and topographic projection of the two nuclei was primarily observed in sagittal sections. After tracer injections into the magnocellular nucleus, the terminal arbours were seen to extend from the deep layers (11-12) to layer 2 of the tectum. The projections observed appeared to be topographically organised, and furthermore appeared to be parallel with and complimentary to previously described projections of the parvocellular isthmic nucleus.

The optic tectum is reciprocally connected to the nuclei isthmi pars magnocellularis (Imc) and pars parvocellularis (Ipc), which have different modulatory effects on optic transmission. We studied the axon arbourisation of these isthmic nuclei in the optic tectum in order to differentiate between them using Golgi-impregnated preparations both in chickens and pigeons. In addition, sections from animals injected with the anterograde tracer biotinylated dextran-amine (BDA) into the Imc were examined in the bright-field and electron microscope to identify the axon arbourisations and terminals. Also, GABA immunogold stained sections were examined in the electron microscope. In Golgi preparations, slab-like (or poplar tree-like) axon terminal arbourisations of both magnocellular and parvocellular isthmic nuclei neurons were found extending to the tectal surface, with similar branching patterns, but different lengths. The axon arbourisations extending from layer 5 of the optic tectum to the surface were termed type 1, whereas those extending from the internal (12-11) layers to the tectal surface were termed type 2. Type 2 arbourisations very closely matched arbourisations observed in BDA injected material, indicating that Imc neurons gave rise to type 2 arbourisations. The two kinds of axon arbourisation in the external tectal layers were alike in both types of bird, except for the width, which was about 10 mum larger in the type 2 axon arbour. Controlling for size, there was no significant difference between chicks and pigeons. The significance of these afferents in the optic tectum is discussed.

Different types of neurons in the ventral geniculate nucleus of the thalamus of chicks were visualised by Golgi impregnation. The dendritic tree of projection neurons branched in a sphere-like territory in both the ventral and middle areas of the lamina externa. The dendrites of projection neurons in the lamina interna descended into the lamina externa and entered both the ventral and middle dendritic areas. One or two dendrites of the lamina interna neurons also emitted branches that developed a dorsal sphere-like dendritic territory. Optic terminals labelled by Golgi impregnation or injection of biotinylated dextran amine were found in these dendritic territories gathered into groups. They established synapses in these areas (synaptic islands or fields without a glial sheath) with different dendritic profiles, and a few gamma-aminobutyric acid (GABA)-positive terminals synapsed with them. No glomerulus-like synaptic complexes ensheathed by glial processes were found. Optic terminals also contacted the stem dendrites of projection neurons and GABA-positive neuron cell bodies and dendrites. Numerous synapses established by both optic and GABA-positive terminals were found on the proximal dendritic stems of the lamina interna projection neurons.

In the present study the terminals of retinal fibres and those of internal layer cells in ventral geniculate nucleus of chicks were labelled with the anterograde tracer biotinylated dextran amine. The tracer showed the connections from the internal cell layers of ventral geniculate nucleus to the medial part of the dorsal lateral geniculate nucleus. The labelled retinal terminals were located exactly in the lateral part of nucleus. The labelled terminals in the two parts of the nucleus were analysed with the electron microscope and showed a different synaptic organisation in the two parts of the dorsal lateral geniculate nucleus. In the lateral part, two kinds of synaptic glomeruli were found mostly in the vicinity of large dendrites, which are proximal dendrites of projection neurons. One type is a simple glomerulus containing a large dendrite, a large optic terminal and a large and/or series of asymmetrical synapses surrounded by glial processes. The other type is a complex synaptic unit with several pre- and postsynaptic components, among them synapses of GABA-positive axon terminals and/or dendraxons. No glomeruli were found in the medial part of the nucleus. In the medial part of the lateral geniculate nucleus, the terminals of internal layer cell axons established asymmetrical synapses with dendrites. Often, a large terminals and large dendritic profiles established serial asymmetrical synapses. GABA-positive myelinated fibres entered and ramified in both parts of the dorsal lateral geniculate nucleus, and GABA-positive terminals were seen to form synapses on the same dendrite near to the asymmetrical contacts. The ramification patterns and terminals of optic fibres in layers 2 and 3 of the optic tectum were studied in Golgi-stained and immunolabelled preparations made from the brains of chicks and pigeons. The different neuronal structures of layers 2 and 3 were also examined. In Golgi preparations, two types of optic fibre were found both in chick and pigeon tectum according to their thickness and terminal branching patterns. The same types of optic fibres were also found to be present in the anterograde tracer experiments after iontophoresis of biotinylated dextran amine into the optic nerve. The varicose terminals of thin fibres mostly terminated on terminal dendritic sections of radiate and pyramidal-like neurons, contacting them on their apical dendrites. The medium-thick fibre terminals in layer 2 mainly established synapses with horizontally extending dendrites, which may therefore be contacts with inhibitory local circuit neurons. The medium-thick optic fibre bushy-like arborisation in layer 3 established synapses with larger dendrites and also stem dendrites. Their terminals formed groups with different dendritic profiles, some of which were partly covered by glial processes, and/or were located among converging dendrites. The presence of these glomerular-like synapses in layer 3 proves that the optic terminals in layer 3 also take part in the transmission of optic impulses to the nucleus rotundus., (Copyright 2003 S. The nucleus lentiformis mesencephali magnocellularis (nLMmc) is an essential part of the accessory optic nuclei and is responsible for stabilization of the horizontal eye movement. The morphology of this nucleus and its intrinsic structural connectivity were studied with Golgi, biotinylated dextran amine anterograde immunotracer and GABA immunostaining methods by light and electron microscopy. In the Golgi preparations neurons of large, medium-large, medium and small sizes were distinguished. The small neurons are GABA-immunopositive local circuit neurons, the others are proposed to be partly projection, partly local circuit neurons. The large and medium-large projection neurons are located in a tight topographical relationship observed in the Golgi preparations. The dendrites of the large and medium-large cells are also observed to be in close proximity with each other, and also with retinal fibre terminals. The morphological arrangement suggests that the retinal fibres make synaptic contacts with dendrites from both types of cell, and this is confirmed by the examination of retinal fibre terminals using electron microscopy. The optic fibre terminals establish synaptic contacts with small dendritic branches, dendritic processes and dendritic spines of large and medium-large neurons in the nLMmc. This arrangement allows the two types of nLMmc neuron access to very similar, if not identical, inputs, which may facilitate some of the different aspects of visual processing. Optic transmission by these cells may be modulated by the GABA-immunopositive terminals from various local circuit neurons, and very probably from GABAergic myelinated fibres as well, which may originate from the contralateral nLMmc and/or the visual Wulst., (Copyright 2003 S. The intrinsic neuronal organisation in the nucleus of the basal optic root of chickens was investigated. The divergent connections with various areas and the functional complexity of the nucleus require a complex intrinsic structural arrangement. Therefore, an analysis of Golgi impregnated material, ultrastructure, GABA-immunocytochemistry and biotinylated dextran-amine anterograde tracer analysis of the nucleus was carried out. In the Golgi analysis, a characteristic dendritic ramification pattern of two types of putative projection neurons was observed. These neurons form dendritic nests with their overlapping dendritic terminal sections, that develop synaptic fields with the optic fibre terminals. These synaptic fields were confirmed by electron microscopy. GABA-immunopositive terminals synapse with distinct loci of the dendritic trees of projection neurons; they may therefore play an important role in the inhibitory-modulatory system of the nucleus of the basal optic root. The GABA-immunopositive terminals derive from small and/or elongated local circuit neurons which receive retinal afferents, and from myelinated fibres afferents to the nucleus of unknown origin.

Layer 7 is one of the retinorecipient layers of the avian optic tectum. However, little information is available about the neuronal organization of this layer and its implications for visual function. Golgi impregnation was used to investigate the retinal input to and the neuronal architecture of layer 7 of the chick optic tectum, which forms a narrow band between the two cell-dense layers 6 and 8. Anterograde tracers were also used to investigate the afferent and efferent connections of layer 7, in both the light and the electron microscope, together with GABA immunogold labelling. Three types of radial neuron were defined according to the origin and course of their axons. The perikarya of these neurons were situated in tectal layers 10-11. The principal dendrites of these radial neurons ascended to the tectal surface and gave rise to dendritic side-branches in layer 7. These dendritic side-branches received asymmetric synapses from the terminations of retinal fibre arborisations. Type 2 radial neurons, whose axons arose from the deep pole of the perikaryon or occasionally from a basal dendrite, were shown to project to the nucleus isthmi pars magnocellularis, which has previously been demonstrated to be GABAergic and to project to glomerulus-like complexes in tectal layers 4-5. In these layers, the dendritic branches of layer 13 neurons that project to the nucleus rotundus have previously been shown to receive retinal fibre input. Therefore, the retinal input to layer 7 may be able to modulate the transmission of information to the visual thalamus, by way of a feed-back loop to layers 4-5 of the tectum involving the nucleus isthmi pars magnocellularis.

Ischemia followed by reperfusion of skeletal muscle frequently takes place in trauma surgery. Anoxia followed by reoxygenation leads to reperfusion injury, which damages the involved tissues. However, no information is available about how the neuromuscular junction is affected by ischemia-reperfusion. Tourniquet ischemia of the left hind limb was applied in the anesthetized rat for 2 h. Reperfusion lasted for 2 and 24 h and for 1, 2 and 4 weeks. The extensor digitorum longus and the soleus muscles from both legs were prepared for electron-microscopic analyses. Morphological changes of the neuromuscular junction were investigated. In all cases only the nerve endings (terminals) were affected. The postsynaptic structures were not affected. Changes can be grouped in two categories: degeneration and recovery. Degeneration consisted of the loss of synaptic vesicles, disruption of the presynaptic membrane, degeneration of the mitochondria and the development of vacuoles. It was most severe at 24 h and was still present at 4 weeks of reperfusion. Recovery started at 1 week of reperfusion and lasted at least for 4 weeks. It consisted of the slow reappearance of synaptic vesicles and mitochondria, and restoration of the presynaptic membrane with active zones. Ultrastructure of the skeletal muscle fibers did not show pathological changes. The recovery of the structures may be regulated by the Schwann cells and also by the postsynaptic membrane which is not affected by 2 h of ischemia followed by reperfusion., (Copyright 2002 S. Three types of local circuit neurons have recently been reported in the homing pigeon hippocampus. The principal type appears to be constituted by the medium-sized angular or ovoid local circuit neurons that occur in all layers of the hippocampus. The current Golgi study has revealed that these neurons can be classified according to their axonal arborisation extension: (1) in all directions, (2) principally medio-laterally, or (3) dorso-ventrally. The local circuit neurons with dorso-ventral axon arborisation are present only in the subpyramidal layer. Serial sections of a Golgi-impregnated medium-sized, multiangular local circuit neuron in the pyramidal layer and a small, ovoid neuron in the suprapyramidal layer were investigated in the electron microscope. Some of these sections were processed for GABA immunogold cytochemistry. The soma and large dendrites of both neurons displayed GABA immunogold labelling. On the soma of medium-sized local circuit neuron there were numerous terminals; on the soma of the small one relatively fewer terminals were observed. The terminals contained round and/or flat synaptic vesicles. The long axonal branches of the neurons exhibited varicosities containing flattened or pleomorphic vesicles. Axo-dendritic, axo-somatic and a few axo-axonic synapses were observed. The large dense axon arborisation field of medium-sized local circuit neurons is properly situated to modulate intrinsic hippocampal activity and that of the small local circuit neurons is well situated to modulate the hippocampal input in the suprapyramidal layer.

In the present study the efferent connections of the ectostriatal core were investigated with biotinylated dextran-amine anterograde tracer in the chicken using light microscopy. The efferents of the ectostriatal core were labelled anterogradely, but retrograde labelling was also observed, which displayed the afferents of the same region. The ectostriatal belt received a few thin varicose fibres; retrogradely labelled cells also appeared. The most extended projection ended in the surrounding neostriatum, which turned out to be reciprocally connected to the ectostriatal core. On the basis of these connections, the neostriatum is said to be an important visual associative center. Efferent fibres reached the motor areas as well. A significant projection entered the paleostriatum augmentatum, especially the ventral part. The paleostriatum primitivum also received a few fibres. The other motor center, the medial part of the anterior archistriatum, was proved to be directly connected to the ectostriatal core as well. Considering that the archistriatum is also connected indirectly to the Wulst, the movements are able to be guided by well processed visual information.

Neurons and fibres in the chick and homing pigeon hippocampus were described following Golgi impregnation. Two principal classes of neurons were distinguished: projection neurons with distant projecting axons and spiny dendrites, and local circuit neurons. In the homing pigeon and chicken hippocampus there are three types of projection neurons: pyramidal, pyramidal-like and multipolar. The pyramidal and pyramidal-like neurons are only found in the central 'pyramidal' layer of the hippocampus whereas multipolar neurons are present in the suprapyramidal, pyramidal and infrapyramidal layers. The axon of projection neurons typically emits several varicose collaterals from the initial section. Most of these collaterals extend along the infrapyramidal layer of the hippocampus, while others ascend to the pyramidal and suprapyramidal layers where they branch. The number of impregnated axon collaterals was higher in the homing pigeon than in the chick hippocampus. A variety of multi-angular/ovoid local circuit neurons ranging from small to large size are found in the homing pigeon and chick hippocampus. Their axons develop local arborization of varicose branches, the extent of which varies with the type of local circuit neurons. The density of GABA immunopositive local circuit neurons was found to be greater in the homing pigeon than in the chick. The profuse arborization of projection neuron axon collaterals and the higher density of GABA-immunopositive local circuit neurons in the homing pigeon hippocampus may underlie the differences in hippocampal function between the homing pigeon and chick, and this complex local connectivity may contribute to the ability of spatial orientation and memory.

The nucleus rotundus receives GABA-like immunoreactive fibres from the nuclei subpretectalis and postero-ventralis thalami. This result was confirmed by Phaseolus vulgaris leucoagglutinin (PhA-L) anterograde tracer and with electron microscopic (EM) gamma-aminobutiric acid (GABA)-immunogold staining. The detailed electron microscopic analysis of the structure of the neurons in these nuclei revealed that the neurons in the nucleus subpretectalis displayed GABA-like immunoreactivity. In the postero-ventral thalamic nucleus a group of neurons was GABA-positive. The surface of the neurons was covered both with numerous GABA-negative and GABA-like immunoreactive terminals that established asymmetrical and symmetrical synapses, respectively, with the GABA-positive neurons. The GABA-like immunonegative terminals are supposed to be the axon terminals of the collaterals of tecto-rotundal fibres in the subpretectal nucleus and the collateral terminal branches of contralateral tecto-rotundal fibres in the postero-ventralis thalami. In both nuclei, the GABA-like immunoreactive terminals may be developed by the collaterals of local neurons that establish symmetrical synapses. In the Phaseolus lectin-stained preparations these terminals may be labelled. The morphological characteristics of the neurons in the subpretectal and partly, in the posteroventral nuclei are similar to those of interneurons (local circuit neurons) and the numerous asymmetrical and symmetrical axo-somatic synapses, respectively. But these neurons locate outside of their target nucleus, and exert their modulatory effect on rotundo-ectostriatal transmission. Also, a contralateral influence is present in the nucleus rotundus that may interact in the cooperation of the eyes. The neurons of the subpretectal and posteroventral nuclei, similarly to the neurons of isthmic nuclei, are a special group of modulatory neurons with effects at a distance.

The principal afferent fibres of the avian optic tectum are the optic fibres of retinal origin. They terminate on the contralateral side, in the external layers (2-7) of the optic tectum (called optic layers) turning into these layers from the external surface. The terminal branchings of the optic fibres develop four densely innervated areas in layers 2, 3, 4-5 and 7. Their terminals are large and of various appearance in the different areas. In the middle third of the optic layer (in layers 4-5), thin dendritic terminal sections of tectal ganglion cells (according to Ramòn y Cajal) of layer 13 terminate into bunches. Phaseolus vulgaris lectin immunotracer corroborates these dendritic endings (further: dendritic terminals) of tectal ganglion cells. The direct connections between these dendritic terminals and the supposed optic fibres were studied under electron microscope and it was found that the large terminals of optic fibres containing round synaptic vesicles establish asymmetrical synapses with several dendritic profiles, among them Phaseolus lectin labelled dendritic terminals of ganglion cells. This result morphologically supports the former physiological observation of a direct synaptic transmission between optic fibres and ganglion cells of layer 13. In addition, on the dendritic terminals of ganglion cells, symmetrical synapses established by GABA-positive terminals were found. The optic terminals, the GABA-immunopositive terminals and the dendritic terminals of ganglion cells form complex synaptic units surrounded with glial sheath, and thus they establish glomerulus-like synaptic units. The size of the dendritic tree and the branching pattern of the dendrites of ganglion cells point to divergence and convergence in visual transmission.

In the present study the telencephalic connections between the centres of the tectofugal and thalamofugal pathways were investigated with Phaseolus vulgaris leucoagglutinin and biotinylated dextran-amine anterograde tracers in chicken using light and electron microscopy. No direct connection was found between the visual Wulst and the ectostriatum or the telencephalic centres of the tectofugal and thalamofugal pathways. Besides other projections, the visual Wulst emitted fibres also to the middle and lateral parts of the hyperstriatum ventrale. Further experiments revealed that the middle part of the hyperstriatum ventrale projected to the ectostriatum centrale and periphericum and established an indirect connection between the visual Wulst and the ectostriatum. The lateral part of the hyperstriatum ventrale sent a few efferent fibres toward the diencephalon and brainstem, but projected massively to the ectostriatum periphericum, neostriatum intermedium pars laterale, the ventral part of the neostriatum caudale and the archistriatum dorsale. Considering that these areas are structures of the tectofugal circuitry as well, the presence of these connections may result in a more elaborate visual processing. The neostriatum may be an associative visual center and possibly a modulatory area toward the archistriatum intermedium dorsale, by which visual information may mediate, modulate and control the movements.

In Golgi preparations of the chicken diencephalon, various types of cells and fibres were studied in the nucleus dorsolateralis anterior thalami (DLA). Two groups of neurons were found: projection neurons with long axons and interneurons with locally branching axons. The projection neurons varied in the different areas of the DLA. In the medial part the neurons are large cells with long, moderately spiny dendrites, and in the dorsal part there are small cells with short, wavy and moderately spiny dendrites. These neurons differ completely from those found in the rostral and lateral parts, where the neurons have medium-sized cell bodies and curving, spiny dendrites, which branch tuft-like or with bifurcations. In the rostral and lateral parts of the DLA, thick afferent fibres were impregnated which developed their terminal branchings among the neurons. Owing to their terminal branching pattern and the shape of the terminals they are thought to be optic fibres. The terminal pattern of these fibres is similar to the optic terminals in the LGB of the mammalian brain. The interneurons are GABA positive, as attested by immunostaining in light microscopic and EM specimens. There are rather few of them. The HRP-filled projection neurons and the surrounding neuropil were investigated under EM: synaptic connections around the large terminals and/or around dendrites were identified, but this synaptic arrangement does not display the characteristics of a synaptic glomerulus.

Following a demonstration of Golgi-impregnated neurons and their terminal axon arborization in the optic tectum, the neurons of the nucleus parvocellularis and magnocellularis isthmi were studied by means of postembedded electron-microscopical (EM) gamma-aminobutyric acid (GABA)-immunogold staining. In the parvocellular nucleus, none of the neuronal cell bodies or dendrites displayed GABA-like immunoreactivity in EM preparations stained by postembedded GABA-immunogold. However, numerous GABA-like immunoreactive and also unlabeled terminals established synapses with GABA-negative neurons. GABA-like immunoreactive terminals were usually found at the dendritic origin. Around the dendritic profiles, isolated synapses of both GABA-like immunoreactive and immunonegative terminals established glomerulus-like structures enclosed by glial processes. All giant and large neurons of the magnocellular nucleus of the isthmi displayed GABA-like immunoreactivity. Their cell surface was completely covered by GABA-like immunoreactive and unlabeled terminals that established synapses with the neurons. These neurons are thought to send axon collaterals to the parvocellular nucleus; their axons enter the tectum opticum. The morphological characteristics of neurons of both isthmic nuclei are like those of interneurons, because of their numerous axosomatic synapses with both asymmetrical and symmetrical features. These neurons are not located among their target neurons and exert their modulatory effect on optic transmission in the optic tectum at a distance.

The neuronal structures and their possible connections in the optic tectum of chicks were studied by using Golgi impregnation, in light microscopy, and Golgi-GABA immunogold staining in electron microscopy. The terminals of the optic fibers displayed different patterns and orientation in various laminae of the avian optic tectum. The shape of the branching and the terminals of the optic fibers conformed to the postsynaptic neuronal structures. In Golgi preparations, the terminals in tectal layers 4 and 5 seemed to be located rather densely, and they contact, among other things, the 'dendritic terminal sections' of large ganglion cells of layer 13 of the tectum. This connection could be responsible for direct optic transmission via ganglion cells to the nucleus rotundus. Additionally, the contacts established by optic terminals in these layers with the tectal neurons arranged with their dendrites parallel to the surface of the tectum, could provide the structural basis for an inhibitory system. The terminals of isthmic nuclei were found in this optic lamina, too. The optic terminals in layer 7 contacted the dendritic side branches of some radial tectal neurons.

After PhA-L injection into the tectum opticum of chickens, column-like labelled terminal arborizations were found in tectum and also labelled neurons of Ipc and Imc in the corresponding sections. The PhA-L labele proved the projection from both Imc and Ipc neurons to tectum; their terminal axonarborizations were clearly discernible. Column-like terminal arborizations of different sizes were found both in PHA-L and Golgi impregnated preparations. GABA immunostaining displayed GABA positive neurons in Imc. The EM-Golgi study corroborated that the axon-terminals of Imc neurons established symmetrical synapses in the tectum. The results of the present study support the existence of an inhibitory feedback loop from Imc neurons to the tectum.

The projection from the chick optic tectum to the nucleus rotundus was investigated by iontophoresis of the anterograde tracer Phaseolus vulgaris lectin (PHA-L) into the optic tectum and by Golgi impregnation. PHA-L labelled fibres were observed within the ipsilateral nucleus rotundus and to course through the supraoptic decussation to the contralateral nucleus rotundus. They appeared similar to fibres evident in Golgi impregnated material. Before reaching the nucleus rotundus, PHA-L labelled fibres gave off short terminal branches in the caudoventral thalamic nucleus. Within the ipsilateral nucleus rotundus, the labelled fibres formed an extensive fibre net with a dense core and a less dense periphery. PHA-L labelled fibres in the contralateral nucleus rotundus exhibited a qualitatively similar but sparser distribution. There appeared to be a topographic representation of the optic tectum within the nucleus rotundus. Electron microscopy revealed that PHA-L labelled tectal fibres established asymmetric synapses with principal neurons in the nucleus rotundus.

Having injected Phaseolus Vulgaris Lectin (PhA-L) iontophoretically to subpretectal (SP) and postero-ventralis (PV) thalamic nuclei, labelled fibers and terminals appeared in nucleus rotundus (Rt). The PhA-L immunostained fibers are divided dichotomically and they develop long terminal sections with large varicosities. In EM preparates PhA-L labelled terminals were observed establishing axo-somatic, axo-dendritic symmetrical synapses. These terminals were stained by GABA immunogold procedure and proved to be GABA-ergic, i.e. the neurons of SP and PV nuclei supply the principal neurons of Rt with GABA positive terminals, which are located on specific, exponent loci of the principal neuron.

The terminals of rotundal fibers labelled with Phaseolus Lectin anterograde tracer were studied in ectostriatum periphericum and neostriatum intermedium laterale with EM. These terminals belong to the fibers which cross the ectostriatum centrale before they enter the ectostriatum periphericum and neostriatum intermedium laterale. These terminals establish synapses with dendrites, spines and also with GABA immunogold stained somata. The EM results prove the connections of ectostriatum periphericum and neostriatum intermedium laterale with the visual system.

Investigation of the chick archistriatum revealed that it was not possible to distinguish distinct sub-regions on the basis of their Golgi architecture, despite the fact that the pigeon archistriatum has been suggested to comprise "amygdaloid" and "sensorimotor" divisions. Nine sub-types of projection neuron and four of local circuit neuron were identified in the chick archistriatum. Projection neurons were generally larger with thicker dendrites than those in other regions of the chick telencephalon. A greater proportion of these projection neurons also had highly densely spiny dendrites. These findings suggest that archistriatal neurons receive a large, possibly complex afferentation and project considerable distances. Large and medium-sized local circuit neurons with varicose axon branches and relatively large neurogliaform cells were characteristic of the archistriatum. All types of archistriatal neuron were identified in both 28 and one day old chicks. However, presumptive highly densely spiny projection neurons appeared relatively immature in the one day old chick archistriatum.

Phaseolus vulgaris leucoagglutinin extracellular injection was performed in nucleus rotundus thalami and the course of labelled fibers and their termination were examined in chicken. The labelled fibers terminate in ectostriatum centrale, ectostriatum perifericum and neostriatum intermedium. The course and termination of the fibers are analysed in detailed.

After having iontophoretically injected Phaseolus vulgaris Lectin into nucleus rotundus, labelled terminals were examined in ectostriatum centrale by EM. These terminals contained round synaptic vesicles. They were observed earlier by Golgi method and proved to be of rotundal origin by Phaseolus Lectin tracing method. The labelled terminals establish synapses, on large and small dendritic truncs of projection and local circuit neurons, and also contact spines and dendritic processes of projection neurons. They were observed on soma of GABA immunopositive local circuit neurons, and also on dendrites. Some observations completed the already known data on fine structure of the chicken ectostriatum centrale.

The EM preparates displayed in the nucleus rotundus also symmetrical type of synapses with flattened vesicles, known this type as inhibitory synapsis. To prove this supposition and to find the sources of inhibitory terminals GABA-immunostaining was applied in the diencephalon of chicks. The preparates were examined by light- and electronmicroscope. In nucl. rotundus the large terminal profiles on the cell membrane of principal neurons, on the dendrites and branching areas of terminal branchings of dendrites proved to be GABA positive. The sources of GABA-ergic fibers seemed to be the subpretectal and ventral posterior thalamic nuclei.

The analysis of EM structure of nucleus rotundus completes the results got by Golgi study. The fine structure of neurons and neuropil of the nucleus and the synaptic relations were studied by EM. The fine structural details of principal neuron were described. Several synapses of symmetrical type with flattened vesicles in large terminals contacted the cell body and also the origin and proximal part of the main dendrites. In the neuropil synaptic junctions were formed by terminals that contained (1) spherical vesicles with occasionally very few dense core vesicles, (2) flattened synaptic vesicles. Terminals that contained spherical vesicles were associated with asymmetric synaptic densities, and terminals that contained flattened vesicles formed symmetric junctions. Synapses of asymmetric type associated mostly with terminal sections of dendrites forming glomerular-like structure. Synapses of symmetric type with flattened synaptic vesicles contacted the branching areas of dendritic terminals and side-branches, the origin of main dendrites and the cell surface of principal neuron.

The structural arrangement of nucleus rotundus was studied by Golgi method. Large, multiangular principal neurons, their dendrites and dendritic terminals were observed. The long dendrites and their side-branches course parallel and/or perpendicular to each-other and they are arranged about in dorso-ventral latero-medial and rostro-caudal directions. The dendrites and their side-branches develop characteristic terminal branchings. These dendritic terminals overlap each-other and the area is called dendritic terminal field. The tecto-rotundal fibers traverse the nucleus in large bundles and the final sections of fibers join to the dendrites of principal neuron and terminate in dendritic terminal fields.

In Golgi impregnated ectostriatum centrale of telencephalon of one-day-old chicken two types of neuron: projection neuron and interneuron with large axonal field and varicose axonal preterminals and terminals, were identified and examined in EM preparates. In the serial EM sections the morphological features of the two neurons types were observed. The projection neuron is characterized by the few axo-somatic synapses on the cell membrane, which are of symmetrical type. The INs, however, have numerous axo-somatic synapses on cell membrane both of symmetrical and asymmetrical types. Also the axonterminals were analysed.

The projection from the ventral tegmental area of Tsai (VTA-A10) to the hippocampal formation (HF) has been investigated in the rat by means of the Fink-Heimer technique, after VTA destruction by electrolytic lesions or local injections of 6-hydroxydopamine (6-OHDA, 1 microgram/0.5 microliters). Degenerated axons prevail in the ventral subiculum and adjacent CA1 field. Some degenerated fibers are also observed in the dorsal subiculum and a few in the stratum oriens of the CA3, in the hilus of fascia dentata and in the fimbria. The distribution of VTA neurons projecting to the HF has also been examined by injecting retrograde fluorescent tracers in different combinations (Fast Blue, 2% and Nuclear Yellow, 1%) in several hippocampal areas. The most abundant VTA-HF projections originate from the lower third, the upper and lower edges and the lower half of the VTA. The major terminal fields of VTA projections in the HF (i.e. the ventral subiculum, the adjacent CA1 field and the dorsal subiculum) match with the HF area projecting to the nucleus accumbens. Thus, the dopaminergic meso-limbic pathway could modulate the HF-striatal projection which provides a link between the limbic and motor systems.

The study demonstrates one kind of the afferent fibers in ectostriatum in Golgi preparates. These fibers enter the ventro-medial border of the area. The primary section and further course and branches, as well as the terminal arborization of these fibers were observed. Also the maturation process of the branchings of the fibers was studied in the Golgi preparates after hatching. The fiber-networks found in one-day-old and 28-day-old chicken ectostriatum centrale were compared.

The brains of kittens prenatally exposed to ethanol were studied by the rapid-Golgi method at PN1 and PN9 days. The sizes of the alcohol damaged brains were different. Delayed gyrification was observed on the brain surface. The neurons in subcortical (thalamic) centres and cortical areas developed later than in normal animals. Between days PN1 and PN9 rapid changes occurred in the development of the structural elements of brains of prenatally alcohol exposed kittens.

The existence of Golgi type II neurons was verified in the anterior intralaminar central lateral (CL) nucleus of the cat thalamus, and its projection cell types were identified, by means of Golgi impregnation. CL principal neurons were found to display a large- or medium-sized cell body and a radiate dendritic pattern. Their primary dendrites were limited in number, and had a rather long course; they were poorly ramified. The axons of principal neurons were impregnated only occasionally and for a short distance. Projection neurons of the 'bushy' or tufted type, described in the main thalamic sensory nuclei, were not identified in the CL in the present study. Typical Golgi type II neurons were found throughout CL. They were mainly small-sized, and displayed a rich dendritic arborization characterized by dendritic appendages. The axons of Golgi type II neurons were seen to give rise to extensive local arborizations. The present findings indicate that in the cat CL, principal cells are mainly represented by radiate neurons. Typical local circuit neurons also are evident in CL, suggesting that the activity of anterior intralaminar structures is regulated by intrinsic mechanisms similar to those operating in the main thalamic relay nuclei.

The medial septal nucleus of non-experimentally treated albino rats was investigated electron microscopically. According to former light microscopical findings most of the dendrites in this region are spineless but conspicuously varicose. Besides a striking diminution of the packing density of neurotubules inside the varicosities--compared with the narrow portions of varicose dendrites--some organelles like multivesicular bodies, lamellated bodies and cisterns of the smooth endoplasmic reticulum were found to be characteristic of them. Large neuronal somata rich in RER with a broad rim of cytoplasm around the nucleus and numerous lipofuscin-like granules occur. But there are numerous small cells exhibiting a narrow rim of cytoplasm and pourly developed RER, too. At least six classes of synaptic junctions can be distinguished, three of the asymmetric and three of the symmetric type. Most of axo-somatic contacts are of the symmetric type, at the dendrites asymmetric contacts dominate. Presynaptic profiles with large dense-core vesicles form symmetrical contacts with varicose and non-varicose dendritic profiles.

In the Golgi preparation of visual Wulst of chicken four types of projection neurons were found: type 1a, 1b, 1c, and 1d neurons, which are distinguished on the basis of their dendritic ramification pattern and especially on the density of the dendritic spines. Type 1a has large dendritic tree with moderately dense spiny dendrites. Type 1b has very dense spiny dendrites. Type 1c is a small neuron with moderately dense spiny dendrites. Type 1d is a star-pyramid-like neuron with a short, bifurcating apical dendrite. The final dendritic pattern and spine density of projection neurons was found to develop at the end of the first month. In the Wulst short axon cells (interneurons-INs) of different types occur: large, medium-sized and small INs can be observed. Their axon-arborizations are significantly different from each other. Some of them are GABA-immuno-positive neurons, which are very probably inhibitory interneurons. Stellate-like neurons also occur in IHA and HIS. These neurons have sparsely spiny dendrites and locally arborizing axons.

Our investigations try to reveal the morphological changes found postnatally at different levels and centres of central nervous system of kittens of cats chronically treated with ethanol during their pregnancy and lactation. The changes found in neurons of lateral geniculate nucleus (LGN) were analyzed in Golgi-stained preparations from brains removed on the first or on the ninth postnatal days. The data were computerized. The changes measured in neurons refer to their maturation process: a significant delay of differentiation of the neurons occurs in LGN. Also a delayed gyrification of brains was observed.

The Golgi method more than hundred years old can reveal new results also in the current research of CNS. The study provides examples for using the Golgi method and demonstrates the types of neurons, the various types of interneurons, characterizes the stellate cells in layer IV of the cortex. The Golgi method is applicable for the observation of maturation process and also for demonstrating different pathological alterations of neurons in the CNS.

An attempt is made on the definition of proportional distribution of the different types of neurons in the layers of LGN by means of quantitative method. On the basis of nuclear volume, nuclear and cell diameters the different types of neurons were separated according to their size and shape. The ratio of relay and interneurons proved to be 2:1 in all layers of LGN of cat. The proportional distribution of further subgroups of neurons was also defined. The bilateral connections between the relay and interneurons based on initial axon collateral of TCR neurons and on axon terminals and dendritic appendages of INs, are also calculated. The interneurons--in the LGN--have widespread contacts with both types of relay neurons, similarly the relay neurons contact the interneurons. The numerical data of morphological observations are accord with the results of physiologists.

Electron microscopical studies of the thalamic AV-AM nuclei substantiated the presence of two main types of neurons, i.e. principal (or relay) cells and Golgi type II interneurons. Characteristic synaptic islands are found in abundance in the AV-AM. Four different types of synaptic terminals have been identified in these islands: RL-boutons = large axonal terminals with round synaptic vesicles; RS-boutons = small axonal terminals with round synaptic vesicles; F1-boutons = small axonal profiles containing flattened synaptic vesicles, and F2-profiles interpreted as presynaptic dendrite appendages, bearing pleomorphic vesicles, both belonging to the Golgi type II interneurons. --The synaptic relations were studied in normal preparations and after lesions in the mamillary body, limbic cortex and hippocampus. The specific afferents (RL-boutons)--originating from the medial mamillary nucleus--are presynaptic to both relay cell dendrites and "presynaptic" dendrite profiles of Golgi type II interneurons, which in turn are presynaptic to the same relay dendrites (synaptic triads). RS-boutons originate mainly from limbic cortex and hippocampus.

In the hippocampus, antibody raised against vasoactive intestinal polypeptide (VIP) labeled perikarya and processes of non-pyramidal neurons whereas these structures remained unlabeled in pyramidal cells and granule cells. In the present study, VIP-immunostaining was used to investigate the fine structure and synaptic connections of identified non-pyramidal neurons and of immunoreactive axon terminals in the CA1 region of the rat hippocampus by means of electron microscopic immunocytochemistry. From a number of cells studied, two VIP-like immunoreactive non-pyramidal neurons in the regio superior were selected for an electron microscopic analysis of serial thin sections. These cells were different with regard to the location of their cell bodies and the orientation of their dendrites. One cell was located in the stratum lacunosum-moleculare with dendritic processes oriented parallel to the hippocampal fissure. The second neuron was found in the inner one-third of the stratum radiatum. The dendrites of this cell ran nearly parallel to the ascending apical dendrites of the pyramidal cells. Both cells had a round or ovoid perikaryon and an infolded nucleus. The aspinous dendrites of both neurons were densely covered with synaptic boutons. These terminals were small, filled with spherical vesicles, and established asymmetric synaptic contacts. No variations in the fine structure of the presynaptic boutons were found along the course of the labeled dendrites through the various hippocampal layers, although different afferents are known to terminate in these layers. Vasoactive intestinal polypeptide-like immunopositive axon terminals course through all layers of the hippocampus. In the stratum pyramidale they established symmetric synaptic contacts with the perikarya of pyramidal cells. In the stratum radiatum they made symmetric contacts with the shafts of apical dendrites of pyramidal cells but never contacted dendritic spines. The symmetric contacts with pyramidal cell perikarya suggest an involvement of the VIP-like immunoreactive axon terminals in pyramidal cell inhibition.

Retrograde axonal transport of horseradish peroxidase was used to identify the afferent projection systems of the anterior thalamic nuclei in the cat. The results indicate that fibres projecting to the three anterior nuclei of the thalamus arise from the hippocampal region, septum and cingulate cortex unilaterally and from the hypothalamus (mamillary nuclei) both bilaterally and unilaterally. The cells of the medial mamillary nuclei send their axons to the ipsilateral AV and AM nuclei whereas the lateral mamillary nucleus projects bilaterally to the AD nuclei.

This study has been carried out by light microscopy on 3 Golgi-Kopsch impregnated brains of young adult rabbits. It is shown that chandelier cells exist within the rabbits' cerebral cortex. In the rabbit, the chandelier cell is a medium ranged bipolar interneuron in layer II/III with a characteristic axon which forms a plexus with a diameter of about 350-500 micrometers in the horizontal and 200-350 micrometers in the vertical direction; the end of each ramulus forms the typical "candlestick", a little vertical string of 1-6 boutons on an axon fibre. These boutons form contacts with all parts of pyramidal cells in layer II and the upper part of layer III. Similarities and differences with respect to previous descriptions of these cells in other species are discussed.

The ultrastructure of interneurons was studied in the cat and it was found to differ from that of the pyramidal and granule cells. The size, shape and topographic situation of the cell body of interneurons were the criteria for approaching their identification with the cells found in Golgi material. Some interneuron dendrites are also described. The varicose and spindle-shaped dendritic profiles belonging to interneurons are different in size. In Golgi material the interneurons have beaded (varicose) or spindle-shaped dendrites. The arrangement of synaptic terminals on the interneuron dendritic surface seems to be their characteristic feature.

The Golgi architecture of the Fascia dentata and hippocampus is described in the cat. The main cell types are like those found in other species. The initial collaterals of granule cell axons were demonstrated and are commented on. The interneurons proved to be of several types and subtypes not observed in lower species. The intrinsic fibers and few of the afferents -- both in the dentate fascia and in the hippocampus -- are discussed.
