Gestational Diabetes Mellitus (GDM) is a medical complication during the gestational period in which woman who had never been diagnosed with diabetes develops hyperglycemia. Prior studies have demonstrated that the advancement of GDM and its consequences arises from a disparity between oxidants and antioxidants in the cells. The observed outcomes can be attributed to an excessive formation of reactive oxygen species (ROS) within the cells, coupled with a reduced activity of anti-oxidative enzymes. Glutathione S-transferase (GSTs) is recognized as an antioxidant enzyme that is belong to as a phase II family member of detoxifying enzymes. These metabolic multigene catalysts are found into the cytoplasm of the cell. GSTs play a vital part in the elimination of cellular ROS or free radicals. The study involves total 300 pregnant women, (150 GDM cases and 150 healthy controls). The polymorphism study of GSTs genes (GSTM1 and GSTT1) was determined by conventional Polymerase Chain Reaction (PCR). The mRNA expression study of GSTM1 and GSTT1 genes analysed by qPCR/ RT-PCR (quantitative PCR/Real-Time PCR) followed by statistical analysis done using Prism8 software (version 8.01). The study revealed statistically significant variations in biochemical parameters between GDM cases and controls. It was found GSTM1-null (GSTM1-/-) polymorphism significantly (P < 0.0001) most prevalent in GDM cases (56.7%) when compared to healthy control (28%). However, no significant difference was observed for GSTT1 null and present polymorphism (P = 0.906). The gene expression levels of both GSTM1 and GSTT1 were found considerably downregulated in individuals with GDM as compared to the control group (P < 0.0001). The downregulation of gene expression has a significant (P<0.0001) association with the null/deletion polymorphism of both GSTM1/ GSTT1 genes respectively. Null/deletion genotype of GSTM1 gene and its expression showed significant association with GDM. Therefore, this gene variant has the potential to be used as a prognostic biomarker for GDM. Supercapacitors currently hold a prominent position in energy storage systems due to their exceptionally high power density, although they fall behind batteries and fuel cells in terms of energy density. This paper examines contemporary approaches aimed at enhancing the energy density of supercapacitors by adopting hybrid configurations, alongside considerations of their power density, rate capability, and cycle stability. Given that electrodes play a pivotal role in supercapacitor cells, this review focuses on the design of hybrid electrode structures with elevated specific capacitance, shedding light on the underlying mechanisms. Factors such as available surface area, porosity, and conductivity of the constituent materials significantly influence electrode performance, prompting the adoption of strategies such as nanostructuring. Additionally, the paper delves into the impact of novel bio-based hybrid electrolytes, drawing upon literature data to outline the fabrication of various hybrid electrode materials incorporating conducting polymers like polyaniline and polypyrrole, as well as metal oxides, carbon compounds, and hybrid electrolytes such as ionic liquids, gel polymers, aqueous, and solid polymer electrolytes. Lipophagy is a ubiquitous mechanism for degradation of lipid droplets (LDs) in lysosomes. Autophagy receptors selectively target organelles for lysosomal degradation. The selective receptor for lipophagy remains elusive. Using mouse liver phosphoproteomics and human liver transcriptomics, we identify vacuolar-protein-sorting-associated protein 4A (VPS4A), a member of a large family AAA+ ATPases, as a selective receptor for lipophagy. We show that phosphorylation of VPS4A on Ser 95,97 and its localization to LDs in response to fasting drives lipophagy. Imaging/three-dimensional (3D) reconstruction and biochemical analyses reveal the concomitant degradation of VPS4A and LDs in lysosomes in an autophagy-gene-7-sensitive manner. Either silencing VPS4A or targeting VPS4A S95,S97 phosphorylation or VPS4A binding to LDs or LC3 blocks lipophagy without affecting other forms of selective autophagy. Background: The impact of duration of ex-vivo heart perfusion (EVHP) on patient outcomes following donation after circulatory death (DCD) heart transplantation has not been established., Methods: Adult first-time DCD heart transplants using EVHP were identified in the Organ Procurement & Transplant Network database (12/2019-09/2023). Total out of body time (OBT) was dichotomized based on perfusion duration exceeding the 90 th percentile of EVHP hearts in the study. The primary outcome of 6-month mortality was assessed using Kaplan Meier curves and multivariable Cox regression. 30-day, 1-year, and 3-year mortality were also assessed. Secondary postoperative outcomes of index hospitalization length of stay, acute rejection, dialysis, and stroke were assessed using univariable linear or logistic regression., Results: Among 575 recipients of DCD transplantations using EVHP, 58 hearts had extended perfusion times based on an OBT cutoff of 8.3 hours which identified OBT greater than the 90 th percentile. Extended perfusion heart recipients had worse overall mortality at 6 months compared to standard perfusion hearts after adjusting for critical donor and recipient factors [aHR=2.48(1.25,4.93),p=0.009]. Early 30-day mortality was comparable between the groups (p=0.592). However, 1-year and 3-year outcomes showed worse mortality in recipients of extended perfusion hearts (both p<0.05). Post-transplant dialysis requirement and increased length of stay was more likely in the extended perfusion group (both p<0.05). There was no difference in acute rejection(p=0.163), and stroke(p=0.170)., Conclusions: There is a potential detrimental effect of extended EVHP duration on DCD heart recipient survival. Objectives: Neighborhood social cohesion or living in communities characterized by trust and social ties may mitigate sleep disparities among sexual minoritized vs. heterosexual persons; but its relation to sleep health is understudied among sexual minoritized groups. To investigate associations between perceived neighborhood social cohesion and sleep health among adult US men and women who identified as "lesbian or gay, bisexual, or something else," we used cross-sectional National Health Interview Survey data (2013-2018)., Methods: Participants reported neighborhood social cohesion (categorized as low or medium vs. high) and sleep characteristics. Adjusting for sociodemographic, health, and residential characteristics, we used Poisson regression with robust variance to estimate prevalence ratios (PRs) and 95% confidence intervals for poor sleep., Results: Among 4666 sexual minoritized adults, 44% reported low, 32% medium, and 24% high neighborhood social cohesion. Women, minoritized racial/ethnic groups, and young adults disproportionately reported low neighborhood social cohesion. Overall, low vs. high neighborhood social cohesion was associated with a higher prevalence of short sleep (PR=1.27 [95% confidence interval:1.11-1.45]) and all sleep disturbances (e.g., PR insomnia symptoms =1.36 [1.19-1.55]). PRs were often higher as intersectionality or membership to multiple minoritized groups increased., Conclusions: Lower perceived neighborhood social cohesion was associated with poorer sleep. Magnetic resonance imaging (MRI) provides detailed structural information of the internal body organs and soft tissue regions of a patient in clinical diagnosis for disease detection, localization, and progress monitoring. MRI scanner hardware manufacturers incorporate various post-acquisition image-processing techniques into the scanner's computer software tools for different post-processing tasks. These tools provide a final image of adequate quality and essential features for accurate clinical reporting and predictive interpretation for better treatment planning. Different post-acquisition image-processing tasks for MRI quality enhancement include noise removal, motion artifact reduction, magnetic bias field correction, and eddy electric current effect removal. Recently, deep learning (DL) methods have shown great success in many research fields, including image and video applications. DL-based data-driven feature-learning approaches have great potential for MR image denoising and image-quality-degrading artifact correction. Recent studies have demonstrated significant improvements in image-analysis tasks using DL-based convolutional neural network (CNN) techniques. The promising capabilities and performance of DL techniques in various problem-solving domains have motivated researchers to adapt DL methods to medical image analysis and quality enhancement tasks. This paper presents a comprehensive review of DL-based state-of-the-art MRI quality enhancement and artifact removal methods for regenerating high-quality images while preserving essential anatomical and physiological feature maps without destroying important image information. Mongoose hair is used to prepare fine brushes, which increases the demand for mongooses to be poached from the wild and brutally bludgeoned to death. Mongooses were listed as Schedule I species under the Indian Wildlife (Protection) Act 1972. Species identification of wildlife case-related samples is necessary to convict a person under this legislation. Microscopy and DNA-based techniques are commonly used to identify mongoose hair in seized brushes. However, in painting brushes, the roots, and the lower part of the hair are mostly trimmed, and only the upper part is used to make the brushes. In addition, brushes are often prepared with mixed hair from mongoose, domestic cattle, human hair, and synthetic fibre. Therefore, the identification of mongoose hair by microscopy and DNA-based techniques is restricted due to the lack of complete strands of hair and the absence of hair roots. Therefore, there is an urgent need to develop an alternative methodology for the identification of mongoose hair from seized articles. FTIR spectroscopy for forensic analysis has gained significant attention over the years because of its sensitivity, specificity, and non-destructive nature. The present study aimed to discriminate Indian grey mongoose ( Herpestes edwardsii ) hair from domestic cattle hair (domestic water buffalo and domestic cow), human hair, and synthetic fiber based on their chemical composition using FTIR spectroscopy and chemometric analysis. We have taken hair from four individuals for each species, namely Indian grey mongoose, domestic cattle, human hair, and synthetic fibre. The FTIR spectrum was recorded, and partial least-squares discriminant analysis (PLS-DA) was used to discriminate hair and synthetic fiber. The established PLS-DA model showed an R-square value and an RMSE (root mean square error) value of 0.9 and 0.13 respectively. Our preliminary findings have shown that FTIR spectroscopy combined with chemometrics can quickly discriminate Indian grey mongoose hair, domestic cattle hair, human hair, and synthetic fiber, providing crucial evidence for judicial proceedings., Competing Interests: The authors declare that they have no conflicts of interest. This work is based on probing photosensitization in carbon nanotubes (CNTs) by organotin(IV) compounds to fabricate a hybrid material with excellent photocatalytic activity and generation of reactive oxygen species. Two organotin(IV) compounds (compounds 1 and 2) were synthesized and characterized by spectroscopic and spectrometric studies, elemental analysis and single crystal X-ray diffraction followed by their impregnation inside the CNTs. The so obtained hybrid materials (1@CNT and 2@CNT) were characterized by FTIR, TGA, FE-SEM, HR-TEM, PXRD and XPS analysis, and assessed for photosensitization and generation of reactive oxygen species. The enhanced photocatalytic activity of the fabricated materials in comparison to bare CNTs is attributed to the reduction of band gap and suppression of rapid recombination rates due to the encapsulation of photogenerated electrons. The generation of reactive species in photocatalyst 1@CNT was validated by the degradation of Amoxicillin (AMX) under optimized conditions for catalytic dosage, H 2 O 2 concentration, response time and pH. The material 1@CNT could degrade ca. 83% of AMX by generating free radicals (˙OH and ˙O 2 - ) under visible light irradiation at pH 6 as investigated by UV-visible spectroscopy and supported by EPR and DFT studies. Furthermore, the structural stability and sustained photocatalytic properties of 1@CNT over four cycles highlight its potential as an eco-friendly solution for degrading environmental toxins.

Machine learning has gained attention in the medical field. Continuous efforts are being made to develop robust models for early prognosis purposes. The brain is the most pivotal organ in the human body. A brain stroke is generally caused by a blockage in the brain arteries. A brain stroke is one of the primary reasons for death. Therefore, early prediction of diseases like brain stroke, heart attack can significantly help in making decisions for doctors. The research study aims to find a robust and potential technique for the early prediction of brain stroke, Alzheimer's, heart attack, cancer, Parkinson's and potentially reducing the incidence of severe post complications of the mentioned diseases. By considering the five datasets as input, machine learning models have been trained for the research study. Early prediction of brain stroke has been done using eight individual classifiers along with 56 other models which are designed by merging the pairs of individual models using soft and hard voting for brain stroke and eight individual classifiers have been used for early prediction of heart attack, cancer, Alzheimer and Parkinson's. After analyzing the results of each classifier for each disease, the proposed method, which is a pair of random forest and decision tree using a hard voting method for early brain stroke prediction, achieves the highest accuracy of 99%, which is better than all classifiers. Along with accuracy, the proposed method attained a value of 98% in precision, an outstanding 100% in recall, and 99% in F1 score. Rationale: Lower airway enrichment with oral commensals has been previously associated with grade 3 severe primary graft dysfunction (PGD) after lung transplantation (LT). We aimed to determine whether this dysbiotic signature is present across all PGD severity grades, including milder forms, and whether it is associated with a distinct host inflammatory endotype., Methods: Lower airway samples from 96 LT recipients with varying degrees of PGD were used to evaluate the lung allograft microbiota via 16S rRNA gene sequencing. Bronchoalveolar lavage (BAL) cytokine concentrations and cell differential percentages were compared across PGD grades. In a subset of samples, we evaluated the lower airway host transcriptome using RNA sequencing methods., Results: Differential analyses demonstrated lower airway enrichment with supraglottic-predominant taxa (SPT) in both moderate and severe PGD. Dirichlet Multinomial Mixtures (DMM) modeling identified two distinct microbial clusters. A greater percentage of subjects with moderate-severe PGD were identified within the dysbiotic cluster (C-SPT) than within the no PGD group (48 and 29%, respectively) though this difference did not reach statistical significance (p=0.06). PGD severity associated with increased BAL neutrophil concentration (p=0.03) and correlated with BAL concentrations of MCP-1/CCL2, IP-10/CXCL10, IL-10, and TNF-α (p<0.05). Furthermore, microbial signatures of dysbiosis correlated with neutrophils, MCP-1/CCL-2, IL-10, and TNF-α (p<0.05). C-SPT exhibited differential expression of TNF, SERPINE1 (PAI-1), MPO, and MMP1 genes and upregulation of MAPK pathways, suggesting that dysbiosis regulates host signaling to promote neutrophilic inflammation., Conclusions: Lower airway dysbiosis within the lung allograft is associated with a neutrophilic inflammatory endotype, an immune profile commonly recognized as the hallmark for PGD pathogenesis. Two major antigen processing pathways represent protein Ags through major histocompatibility complexes (MHC class I and II) or lipid Ags through CD1 molecules influence the tumor immune response. Invariant Natural Killer T cells (iNKT) manage a significant role in cancer immunotherapy. CD1d, found on antigen-presenting cells (APCs), presents lipid Ags to iNKT cells. In many cancers, the number and function of iNKT cell are compromised, leading to immune evasion. Additionally impaired motility of iNKT cells may contribute to poor tumor prognosis. Emerging evidences suggest that CD1d, itself also influences cancer progression. Patient databases further highlight the importance of CD1d expression in different cancers and its correlation with patient survival outcomes. The ability of iNKT cells to activate and enhance the immune response renders them an attractive target for cancer immunotherapy. Shifting cultivation, an age-old agricultural practice, is a major factor in forest cover change across Southeast Asia, where repeated cycles of vegetation disturbance and regrowth lead to far-reaching environmental and socio-economic impacts. The present study aims to assess the spatio-temporal patterns of vegetation disturbance and regrowth caused by shifting cultivation in Tripura state of India, over the past three decades, utilizing temporal segmentation of time-series Landsat data. The study analyzed vegetation disturbance and regrowth patterns in a shifting cultivation landscape from 1991 to 2020 using normalized burn ratio trends through LandTrendr, validated by the TimeSync tool. Six land use and land cover classes, viz., forests and trees outside forests, rubber plantation, shifting cultivation, water bodies, agriculture, and settlements, were mapped with an accuracy of 83.08% using a random forest classifier. This classification enabled the identification of the effective study area, which included forest areas with shifting cultivation patches. The analysis revealed that 2533.96 km 2 of the study area remained undisturbed, 568.43 km 2 experienced low-magnitude disturbance, 1501.11 km 2 were moderately disturbed, and 184.82 km 2 were highly disturbed. The shifting cultivation cycles in the study area show considerable variation. Low-magnitude disturbance indicates a single slash-and-burn event in the past three decades, moderate-magnitude disturbance involves fallow periods of over 10 years, and high-magnitude disturbance occurs with fallow periods of less than 8 years. The study revealed a shift from traditional shifting cultivation to more permanent agricultural practices in parts of Tripura. In response to incentives for commercial crops, cultivators are increasingly adopting long-term cultivation, including the growing of pineapple, areca nut, and rubber, as well as intercropping with papaya, banana, lemon, tapioca, pepper, and ginger. These plots are, however, abandoned if the yields become economically unviable. The expansion of monoculture cultivation significantly reduces the available area for shifting cultivation, thereby compelling jhum cultivators to revisit their jhum patches more frequently. Adeno-Associated Virus (AAV) is often selected as the vector of choice for gene therapy due to its superior clinical performance compared to other gene delivery systems. Currently the characterization of AAV degradation, especially the chemical degradation of capsid, has been limited due to lack of suitable methods. Our study using AAV9 as a model molecule shows that anion exchange chromatography (AEX) as a charge-based separation method has limitations in monitoring the chemical degradation of AAV9 capsid due to a confounding effect from DNA cargo ejection. We developed a hydrophobic interaction chromatography (HIC) method, free from DNA interference, that could serve as a quick and reliable alternative to resource-demanding peptide mapping method for monitoring AAV capsid chemical degradation. Background: Chikungunya (CHIK) is an underdiagnosed acute febrile illness (AFI) and an important cause of acute encephalitis syndrome (AES). Unavailaibility of rapid and sensitive molecular point-of-care tests (PoCTs) for CHIK at grass-root level, results in increased hospital burden, due to delayed diagnosis or misdiagnosis with other clinically relevant diseases. Since, no therapeutic intervention is readily available, accurate and differential diagnosis of CHIK is the only available option to initiate early supportive treatment. Thus, we aimed to develop a one-pot reverse transcription recombinase polymerase amplification (RT-RPA) mediated CRISPR/Cas12a based detection platform for rapid, specific, and ultrasensitive detection of chikungunya virus (CHIKV) in clinical samples., Results: We have successfully integrated CRISPR/Cas12a technology with reverse transcription recombinase polymerase amplification (RT-RPA) for the detection of Chikungunya virus (CHIKV). The developed assay enabled rapid detection of CHIKV within 35 min, requiring minimal handling process and instrumentation. Next, this assay demonstrated dual mode end-point detection capabilities, employing both fluorescence and lateral flow detection within a reaction. Our one-pot system allows the entire process to be completed without the need to open the reaction tube, thereby eliminating the risk of cross-contamination. Remarkably, the assay exhibits an analytical sensitivity of 412 zg μL -1 (≈1 copy), and 100 % clinical sensitivity and specificity for CHIKV. Furthermore, the developed assay demonstrated limit of detection of 8 gene copies of CHIKV. The assay demonstrates precise detection of CHIKV without any cross-reactivity with other pathogens commonly associated AFI or AES., Significance: The overall findings of this study indicate that the RT-RPA:CRISPR/Cas12a detection assay, with one-pot dual-mode detection approach enables rapid, specific and ultrasensitive molecular detection of CHIKV. This advancement holds significant potential for CHIKV detection in resource-limited settings, providing a robust tool for diagnosis and management of the disease. Aim: Metabolic comorbidities such as obesity type 2 diabetes, insulin resistance, glucose intolerance, dyslipidemia are the major contributors for lower life expectancy and reduced patient compliance during antipsychotic therapy in patients with severe mental illnesses such as schizophrenia, bipolar disorder, and depression. TRPM8 activation by menthol is also reported to alleviate high fat diet-induced obesity in mice. Additionally, this TRPM8 activation leads to increase in gene expression of thermogenic genes in white adipocytes and dietary menthol was found to increase browning of WAT along with improved glucose utilization. Therefore, we aimed to evaluate the plausible role of TRPM8 channels in olanzapine-induced metabolic alterations in female balb/c mice., Methods: 6 weeks olanzapine (6 mg kg -1 , per oral) model was used in female balb/c mice. Pharmacological manipulation of TRPM8 channel was done using menthol and N-(3-aminopropyl)-2-[(3-methylphenyl)methoxy]-N-(2-thienylmethyl)-benzamide (AMTB), the agonist and antagonist respectively., Key Results: Menthol co-treatment for six weeks prevented olanzapine-induced metabolic alterations such as weight gain, increased food intake, decreased energy expenditure, adiposity, liver lipid accumulation, systemic inflammation and insulin resistance. Copper oxide nanosheets (CuO NSs) have been successfully obtained by exploiting an effective one-step approach of sugar-blowing method followed by calcination. The nanosheets were characterized by several techniques like X-ray powder diffraction (XRD), Transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS) and Fourier transform infrared spectroscopy (FTIR). Impressively, CuO NSs display haloperoxidase (HPO) like catalytic activity which catalyses the oxidation of chloride ions by H 2 O 2 giving rise to reactive chlorine species (RCS). A sensitive and selective colorimetric sensor was then demonstrated via the oxidation of chromogenic substrate 3,3',5,5'- tetramethylbenzidine (TMB) by the novel nanoenzyme CuO NSs through the generation of RCS for H 2 O 2 and glucose detection with limit of detection of 109 nM and 21 nM in the linear ranges of 4.6 µM to 769 µM and 0.22 µM to 19.57 µM respectively. Stripe or yellow rust (YR) caused by Puccinia striiformis tritici ( Pst ) is an important foliar disease affecting wheat production globally. Resistant varieties are the most economically and environmentally effective way to manage this disease. The common winter wheat ( Triticum aestivum L.) cultivar Luomai 163 exhibited resistance to the Pst races CYR32 and CYR33 at the seedling stage and showed a high level of adult plant resistance in the field. To understand the genetic basis of YR resistance in this cultivar, 142 F 5 recombinant inbred lines (RILs) derived from cross Apav#1 × LM163 and both parents were genotyped with the 16K SNP array and bulked segregant analysis sequencing. The analysis detected a major gene, YrLM163 , at the seedling stage associated with the 1BL.1RS translocation. Additionally, three genes for resistance at the adult plant stage were detected on chromosome arms 1BL ( Lr46/Yr29/Pm39/Sr58 ), 6BS, and 6BL in Luomai 163, whereas Apav#1 contributed resistance at a quantitative trait locus (QTL) on 2BL. These QTL explained YR disease severity variations ranging from 6.9 to 54.8%. The kompetitive allele-specific PCR (KASP) markers KASP-2BL , KASP-6BS , and KASP-6BL for the three novel loci QYr.hzau-2BL , QYr.hzau-6BS , and QYr.hzau-6BL were developed and validated. QYr.hzau-1BL , QYr.hzau-2BL , and QYr.hzau-6BS showed varying degrees of resistance to YR when present individually or in combination based on genotype and phenotype analysis of a panel of 570 wheat accessions. Diabetes poses a significant global health challenge, driving the search for effective management strategies. In the past years, α-amylase inhibitors have emerged as promising candidates for regulating blood sugar levels. In this concern, we have synthesized a series of novel 3-methyl-2-aroylthiazolo[3',2':2,3][1,2,4]triazino[5,6- b ]indole derivatives via the regioselective reaction of 2,5-dihydro-3 H -[1,2,4]triazino[5,6- b ]indole-3-thione and 1,3-diketones in the presence of NBS under solvent-free conditions. Subsequently, the inhibitory potential of the newly synthesized 3-methyl-2-aroylthiazolo[3',2':2,3][1,2,4]triazino[5,6- b ]indole derivatives was assessed against the α-amylase enzyme to probe their antidiabetic properties. In vitro studies revealed moderate to excellent α-amylase inhibitory activity, with IC 50 values ranging from 16.14 ± 0.41 to 27.69 ± 0.58 μg ml -1 . Furthermore, SAR analysis showed that compounds containing halogen groups exhibited superior inhibition potential, surpassing the standard drug Acarbose (IC 50 = 18.64 ± 0.42 μg ml -1 ), particularly derivatives substituted with 4-fluoro and 2,4-dichloro groups, with IC 50 values of 16.14 ± 0.41 μg ml -1 and 17.21 ± 0.15 μg ml -1 , respectively. Additionally, molecular docking unveiled the binding modes of ligands with the active site of A. oryzae α-amylase. Encouragingly, the theoretical analyses closely mirrored the experimental findings, further underlining the promise of these synthetic molecules as potent α-amylase inhibitors.

The phase singularity, a sudden phase change occurring at the reflection zero is widely explored using various nanophotonic systems such as metamaterials and thin film cavities. Typically, these systems exhibit a single reflection zero with a phase singularity at a specific incident angle, particularly at larger angles of incidence (>50 degrees). However, achieving multiple phase singularities at a single incident angle remains a formidable challenge. Here, the existence of a dual-phase singularity is experimentally demonstrated at a lower incident angle using a tunable Tamm plasmon polariton (TPP) cavity that consists of gold-coated ultralow-loss phase change material Sb 2 S 3 -based distributed Bragg reflector. It can excite narrowband TPP resonances from normal incidence to a wide angle of incidence for both s- and p-polarizations of light. Notably, this TPP cavity shows dual-phase singularity at lower angles of incidence since the excited TPP for s- and p-polarizations exhibits zero reflection at slightly different wavelengths for the same incident angle. A TPP cavity-based scalable hydrogen sensor is proposed and shows that the dual-phase singularity can further improve the sensitivity of singular phase-based sensing approaches. The increase in publicly available human single-cell datasets, encompassing millions of cells from many donors, has significantly enhanced our understanding of complex biological processes. However, the accessibility of these datasets raises significant privacy concerns. Due to the inherent noise in single-cell measurements and the scarcity of population-scale single-cell datasets, recent private information quantification studies have focused on bulk gene expression data sharing. To address this gap, we demonstrate that individuals in single-cell gene expression datasets are vulnerable to linking attacks, where attackers can infer their sensitive phenotypic information using publicly available tissue or cell-type-specific expression quantitative trait loci (eQTLs) information. We further develop a method for genotype prediction and genotype-phenotype linking that remains effective without relying on eQTL information. Purpose: Intravenous lidocaine is a non-opioid analgesic adjunct for perioperative pain relief. The aim of our study was to explore whether concurrent administration of intravenous lidocaine prolongs the duration of sensory block during total knee replacement (TKR) under spinal anaesthesia., Methods: This prospective randomized double blind controlled trial was conducted on 28 patients (14 in lidocaine group and 14 in the control group) undergoing unilateral TKR under spinal anesthesia. In the lidocaine group, intravenous lidocaine 1.5 mg·kg -1 followed by an infusion of 1.5 mg·kg -1 ·h -1 was administered intraoperatively after spinal anesthesia. The primary outcome was the duration of sensory block of spinal anesthesia. Secondary outcomes included onset time of sensory and motor block, duration of motor block, time to first postoperative analgesic, postoperative visual analog scale (VAS) scores and postoperative analgesia requirement in 24 h after surgery., Results: The duration of sensory and motor block was longer in the lidocaine group (Mean ± SD; 112.50 ± 5.80 min versus 78.21 ± 9.12 min; p < 0.001 and 237.14 ± 9.14 min versus 215.00 ± 10.12 min; p < 0.001, respectively). Time to requirement of first rescue analgesia was 184.29 ± 9.38 min in the lidocaine group and 127.14 ± 23.35 min in the control group (p < 0.001). VAS scores were lower in the lidocaine group at 4, 8, 12 and 24 h after surgery (p < 0.00001, p < 0.00001, p < 0.00006, p = 0.032, respectively). Requirement of additional analgesia in the first 24 h was higher in the control group. Inflammatory bowel disease (IBD) treatment often involves systemic administration of anti-inflammatory drugs or biologics such as anti-TNF-α antibodies. However, current drug therapies suffer from limited efficacy due to unresponsiveness and adverse side effects. To address these challenges, we developed inflammation-targeting nanoparticles (ITNPs) using biopolymers derived from the gum kondagogu (Cochlospermum gossypium) plant. These ITNPs enable selective drug delivery to inflamed regions, offering improved therapeutic outcomes. We designed ITNPs that specifically bind to inflamed regions in both human and mouse intestines, facilitating more effective, uniform, and prolonged drug delivery within the inflamed tissues. Furthermore, we demonstrated that oral administration of ITNPs loaded with urolithin A (UroA), a microbial metabolite or its synthetic analogue UAS03 significantly attenuated chemical- and immune checkpoint inhibitor- induced colitis in pre-clinical models. In conclusion, ITNPs show great promise for delivering UroA or its analogues while enhancing therapeutic efficacy at lower doses and reduced frequency compared to free drug administration. Objective: The purpose of this study was to conduct a systematic investigation of the potential of artificial intelligence (AI) models in the prediction, detection of diagnostic biomarkers, and progression of diabetic kidney disease (DKD). In addition, we compared the performance of non-logistic regression (LR) machine learning (ML) models to conventional LR prediction models., Methods: Until January 30, 2024, a comprehensive literature review was conducted by investigating databases such as Medline ( via PubMed) and Cochrane. Research that is inclusive of AI or ML models for the prediction, diagnosis, and progression of DKD was incorporated. The area under the Receiver Operating Characteristic Curve (AUROC) served as the principal outcome metric for assessing model performance. A meta-analysis was performed utilizing MedCalc statistical software to calculate pooled AUROC and assess the performance differences between LR and non-LR models., Results: A total of 57 studies were included in the meta-analysis. The pooled AUROC of AI or ML model was 0.84 (95% CI = 0.81-0.86, p  < 0.0001) for analyzing prediction of DKD, 0.88 (95%CI = 0.84-0.92, p  < 0.0001) for detecting diagnostic biomarkers, and 0.80 (95% CI = 0.77-0.82, p  < 0.0001) for analyzing progression of DKD. The pooled AUROC of LR and non-LR ML models exhibited no significant differences across all categories ( p  > 0.05), except for the random forest (RF) model, which displayed a statistically significant increase in predictive accuracy compared to LR for DKD occurrence ( p  < 0.04)., Conclusion: ML models showed solid DKD prediction effectiveness, with pooled AUROC values over 0.8, suggesting good performance. These data demonstrated that non-LR and LR models perform similarly in overall CKD management, but the RF model outperforms the LR model, particularly in predicting the occurrence of DKD. These findings highlight the promise of AI technologies for better DKD management. To improve model reliability, future study should include extended follow-up periods as well as external validation.

Bacterial infections and biofilm formation are significant challenges for medical implants. While titanium nanotube engineering improves biocompatibility, it cannot prevent bacterial adhesion and biofilm formation. Optimizing the biomaterial's surface chemistry is vital for its desired functioning in the biological environment. This study demonstrates the covalent conjugating of the self-assembling dipeptide N-fluorenylmethyloxycarbonyl-diphenylalanine (Fmoc-FF) onto titanium nanotube surfaces (TiNTs) without altering the topography. Fmoc-FF peptides, in conjugation with TiNTs, can inhibit biofilm formation, eradicate pre-existing biofilms, and kill bacteria. This functionalization imparts antibacterial properties to the surface while retaining beneficial nanotube topography, synergistically enhancing bioactivity. Surface characterization by XPS, FT-IR, EDS, and SEM confirmed the successful functionalization. Bacterial adhesion experiments showed a significantly improved antibacterial activity of the functionalized TiNT surfaces. This study opens future possibilities for associating biomedical applications such as cell-cell interactions, tissue engineering, and controlled drug delivery of multifunctional self-assembling short peptides with implant materials through surface functionalization.

Emerging from robust coulombic interactions and diminished dielectric screening, two-dimensional transition metal dichalcogenides (2D TMDs) manifest strongly bound excitons. While these pronounced many-body effects have been observed in 2D TMDs, their impact on the materials' intrinsic photoconductive characteristics remains relatively less explored. In this study, a considerable reduction in photoconductivity in chemical vapor deposited (CVD) ReS 2 thin films has been observed. Through experimental and theoretical scrutiny, it is deduced that this anomalous phenomenon stems from the possible many-body interactions within the system, wherein photoexcited electron-hole pairs combine with excess electrons to generate trions. The formation of trions reduces the effective number of carriers contributing to photocurrent, thereby attenuating the photoconductivity. This study shows the profound influence of trions on the photoconductivity behaviour of ReS 2 thin films, thereby elucidating their significance in modulating optical and electrical properties crucial for optoelectronic applications.

Non-alcoholic fatty liver disease (NAFLD) is the main cause of chronic liver disease worldwide, affecting one-fourth of the world's population. With more than half of the world's population, the Asia-Pacific region contributed 62.6 % of liver-related fatal incidents in 2015. Currently, liver imaging techniques such as computed tomography (CT), nuclear magnetic resonance (NMR) spectroscopy, and ultrasound are non-invasive imaging methods to diagnose the disease. A liver biopsy is the gold standard test for establishing the definite diagnosis of non-alcoholic steatohepatitis (NASH). However, there are still significant problems with sample variability and the procedure's invasiveness. Numerous studies have indicated various non-invasive biomarkers for both fibrosis and steatosis to counter the invasiveness of diagnostic procedures. Metabolomics could be a promising method for detecting early liver diseases, investigating pathophysiology, and developing drugs. Metabolomics, when utilized with other omics technologies, can result in a deeper understanding of biological systems. Metabolomics has emerged as a prominent research topic, offering extensive opportunities to investigate biomarkers for liver diseases that are both sensitive and specific. Alzheimer's disease (AD) remains a pressing global health concern, necessitating comprehensive investigations into its underlying molecular mechanisms. While the late-stage pathophysiology of this disease is well understood, it is crucial to examine the role of amyloid beta oligomers (Aβo), which form in the brain during the early stages of disease development. These toxic oligomers could affect neuronal viability and generate oxidative stress in the brain. In this study, we exposed SHSY-5Y cells to Aβo. The increase in intracellular reactive oxygen species and apoptosis observed in Aβo-treated cells mimics the early stages of AD. Comprehensive proteomic profiling identified 2966 differentially expressed proteins, with 123 significantly modulated. Utilizing the NeuroPro database, we identified 80 confirmed AD-related proteins and 43 novel candidates. Seven AD-related proteins with a NeuroPro score ≥ 5 were shortlisted. Furthermore, these proteins are found to be associated with Aβ plaques in AD brains. VGF, LTF, PARP1, and MAOA have been implicated in various mechanisms underlying AD, including synaptic plasticity, iron homeostasis, DNA repair, and neurotransmitter degradation. Our study also revealed the involvement of less-explored proteins like MYH9, CISD1, and SNRNP70, which play critical roles in cytoskeletal dynamics, mitochondrial function, and RNA splicing, respectively. These findings underscore the complex pathophysiology of AD, highlighting potential biomarkers and therapeutic targets for early intervention. Social isolation affects the brain and behavior in a variety of animals, including humans. Studies in traditional laboratory rodents, including mice and rats, have supported the idea that short-term social isolation promotes affiliative social behaviors, while long-term isolation promotes anti-social behaviors, including increased aggression. Whether the effects of isolation on the social behaviors of mice and rats generalize to other rodents remains understudied. In the current study, we characterized the effects of short-term (3-days) social isolation on the social behaviors of adult prairie voles (Microtus ochrogaster) during same-sex and opposite-sex social interactions. Our experiments revealed that short-term isolation did not affect rates of ultrasonic vocalizations or time spent in non-aggressive social behaviors and huddling during same-sex and opposite-sex interactions. Unexpectedly, although short-term isolation also did not affect time spent in resident-initiated and mutually-initiated aggressive behavior, we found that short-term isolation increased time spent in visitor-initiated aggression during male-male interactions. Over a decade of randomized controlled trial data demonstrate excellent outcomes with transcatheter aortic valve replacement or surgical aortic valve replacement for patients with symptomatic severe aortic stenosis regardless of surgical risk. The 2020 American College of Cardiology/American Heart Association guidelines recommend both options for low-risk AS patients aged 65 to 80 years. However, the fastest growing population of patients receiving transcatheter aortic valve replacement in the United States is <65 years old, with little data to support the practice. The American College of Cardiology's Cardiac Surgery Team Section Leadership and Interventional Cardiology Councils, a multidisciplinary collaboration of cardiologists and cardiac surgeons, sought to summarize the relevant data into a decision-making tool for heart valve teams. Background: Vertebrobasilar dolichoectasia (VBD) is a rare disease with significant morbidity. Its propensity for posterior circulation and relationship with aneurysms is poorly understood. Here, we aimed to describe the anatomical characteristics of the vertebral arteries (VA) in patients with VBD., Methods: We conducted a multi-center retrospective cohort analysis on patients diagnosed with VBD between January 2009 and December 2022. Anatomical variables were measured after manual segmentation on CTA. Length of the VAs, basilar artery (BA), sectional diameters, and tortuosity index were measured and compared with a control group., Results: 124 patients were included: 38 patients with VBD and 86 controls. VBD patients had longer VAs (right VA 262.5 vs. 228.7 mm, p<0.01; left VA 253.4 vs. 222.1 mm, p<0.01), longer BAs (41.9 vs. 30.0 mm, p<0.01), and larger cross-sectional diameters of right V3 (5.1 vs. 4.6 mm, p<0.05) and V4 segments (4.5 vs.2.9 mm, p<0.05) and left V1-V4 segments (V1 6.1 vs. 4.2, V2 7.2 vs.4.8 mm, V3 6.4 vs.5.0 mm, V4 6.5 vs. 3.0 mm, p<0.01). Patients with VBD and fusiform aneurysms had longer VAs and BA than patients without aneurysms (p<0.01). The VAs tortuosity index was higher in VBD than controls (60.69 vs. 44.18, p<0.01). No cases of vertebral stenosis were detected., Conclusions: Our study offers an anatomical study of VBD. While the natural history is poorly understood, the higher tortuosity index with associated wall shear stress, provides a possible mechanism for disease progression. We present the first measurement of nuclear recoils from solar ^{8}B neutrinos via coherent elastic neutrino-nucleus scattering with the XENONnT dark matter experiment. The central detector of XENONnT is a low-background, two-phase time projection chamber with a 5.9 t sensitive liquid xenon target. A blind analysis with an exposure of 3.51  t×yr resulted in 37 observed events above 0.5 keV, with (26.4&#95;{-1.3}^{+1.4}) events expected from backgrounds. The background-only hypothesis is rejected with a statistical significance of 2.73σ. The measured ^{8}B solar neutrino flux of (4.7&#95;{-2.3}^{+3.6})×10^{6}  cm^{-2} s^{-1} is consistent with results from the Sudbury Neutrino Observatory. The measured neutrino flux-weighted CEνNS cross section on Xe of (1.1&#95;{-0.5}^{+0.8})×10^{-39}  cm^{2} is consistent with the Standard Model prediction. This is the first direct measurement of nuclear recoils from solar neutrinos with a dark matter detector.

The first search for soft unclustered energy patterns (SUEPs) is performed using an integrated luminosity of 138  fb^{-1} of proton-proton collision data at sqrt[s]=13  TeV, collected in 2016-2018 by the CMS detector at the LHC. Such SUEPs are predicted by hidden valley models with a new, confining force with a large 't Hooft coupling. In events with boosted topologies, selected by high-threshold hadronic triggers, the multiplicity and sphericity of clustered tracks are used to reject the background from standard model quantum chromodynamics. With no observed excess of events over the standard model expectation, limits are set on the cross section for production via gluon fusion of a scalar mediator with SUEP-like decays.

Schizophrenia is a severe psychiatric disorder associated with a wide range of cognitive and neurophysiological dysfunctions and long-term social difficulties. In this paper, we test the hypothesis that integration of multiple simultaneous acquisitions of neuroimaging, behavioral, and clinical information will be better for prediction of early psychosis than unimodal recordings. We propose a novel framework to investigate the neural underpinnings of the early psychosis symptoms (that can develop into Schizophrenia with age) using multimodal acquisitions of neural and behavioral recordings including functional near-infrared spectroscopy (fNIRS) and electroencephalography (EEG), and facial features. Our data acquisition paradigm is based on live face-to-face interaction in order to study the neural correlates of social cognition in first-episode psychosis (FEP). We propose a novel deep representation learning framework, Neural-PRISM, for learning joint multimodal compressed representations combining neural as well as behavioral recordings. These learned representations are subsequently used to describe, classify, and predict the severity of early psychosis in patients, as measured by the Positive and Negative Syndrome Scale (PANSS) and Global Assessment of Functioning (GAF) scores. We found that incorporating joint multimodal representations from fNIRS and EEG along with behavioral recordings enhances classification between typical controls and FEP individuals. Additionally, our results suggest that geometric and topological features such as curvatures and path signatures of the embedded trajectories of brain activity enable detection of discriminatory neural characteristics in early psychosis.

Parkinson's Disease (PD) is a chronic neurodegenerative disorder that impacts the substantia niagra region of the midbrain leading to impaired motor as well as non-motor symptoms of the central nervous system (CNS). Mitochondrial dysfunction has been characterized as the primary cause of dopaminergic neuronal loss, however, the molecular mechanisms leading to mitochondrial dysfunction are not completely understood. PARKIN, E3 ubiquitin ligase, plays a crucial role in maintaining mitochondrial quality control, albeit the role of other E3 ligases in regulating mitochondrial functions is not understood. In the current study, we explored the implication of TRIM71, E3 ubiquitin ligase, in the modulation of mitochondrial functions and neuronal death in PD stress conditions induced by rotenone and 6-OHDA. Ectopic expression of TRIM71 in SH-SY5Y dopaminergic neuronal cells sensitizes to PD stress-induced cell death, while its knock-down rescues neuronal cell death. TRIM71 turnover is enhanced in neurons under PD stress conditions. TRIM71 predominantly localizes on the outer mitochondrial membrane and translocation increases during PD stress conditions. TRIM71 regulates mitochondrial complex I and IV assembly and activity. TRIM71 knock-down decreases mitochondrial ROS and enhances ATP level as well as mitochondrial membrane potential in PD stress conditions. TRIM71-mediated mitochondrial ROS and cell death were rescued by mitoTEMPO, a mitochondrial-targeted antioxidant. Purpose: Carbon fiber-reinforced polyetheretherketone (CFR-PEEK) instrumentation is being used more frequently in the spinal oncology landscape. Better visualization with this material allows for more precise postoperative stereotactic body radiation therapy (SBRT) planning using either computed tomography (CT)-myelography or magnetic resonance imaging (MRI) studies. We compared the dosimetric planning equivalencies and outcomes., Methods and Materials: Thirty-six consecutively treated patients were reviewed who underwent spinal fusion using CFR-PEEK instrumentation for spine metastases followed by postoperative SBRT between January 1, 2022, and April 3, 2023. Patients were divided into 2 cohorts based on the imaging modality, MRI versus CT-myelogram, used for postoperative SBRT planning. Surgical, demographic, postoperative radiation dosimetry, complication, and survival data were collected. Statistical analysis was performed in SPSS (v29.0.1.0)., Results: Eleven patients underwent CT-myelograms, and 25 patients underwent MR-spine imaging for SBRT planning. The median follow-up was 145.5 days (13-530). There were no significant differences between baseline demographic, surgical characteristics, or SBRT dosimetry between the MRI spine and CT-myelogram groups. There was no significant difference between the cohorts for survival (P = .402)., Conclusions: MR scans are an effective choice for postoperative SBRT contouring patients using CFR-PEEK instrumentation for oncologic spinal fusions. This article discusses examples from an extended family of aphorisms, stories, and themes that have circulated widely in STS and remain associated with the formation and integration of the field. Drawing upon Harvey Sacks's insightful remarks about features of everyday conversation, which he related to ancient practices in oral culture, we argue that familiar citation magnets in STS operate in many respects like proverbs, parables, and an extended family of neatly and memorably packaged viral articulations in ordinary language. After discussing the contingent production of proverbial truth, the article focuses on three well-known examples that combine memorable proverbs and themes with parables: Winner's account of the low parkway bridges designed by city planner Robert Moses to show that technology has politics; Pinch and Bijker's concise history of the bicycle to illustrate the social construction of technology, and Star and Griesemer's viral two-word theme of 'boundary objects' as artifacts that sustain collaboration across organizational contexts. The discussion of these cases suggests that different elements of these examples become the focus of subsequent citations and applications, and that ambiguities about the origins and meanings of the cited items opens new avenues for critical reflection on practicing citational justice and the nature of STS as an affiliative discipline.

The impact of Bariatric Surgery (BS) on maternal and neonatal outcomes among pregnant women is not fully understood, especially in comparison to different weight categories. The primary aim of this study is to assess the factors associated to women who have undergone BS. The study also investigates the maternal and neonatal outcomes amongst this group in comparison to the three Body Mass Index (BMI) groups (women with obesity, overweight and normal weight). A 12-month population-based retrospective study was conducted using registry data from the PEARL-Peristat Study at the Women's Wellness and Research Center (WWRC) in Qatar from January 1, 2017, through December 31, 2017. Both univariate and multivariable regression analyses were employed to scrutinize risk factors and maternal and neonatal outcomes. The study included 6212 parturient women, of which 315 had a history of BS, while 5897 with no BS history. Qatari women, aged 35 and higher, with parity > 1, diabetes, and hypertension were more likely to be in the post-BS group. Women in the post-BS group were found to be more likely to have a cesarean delivery (37.5% vs. 24%, Adjusted Odds Ratio (aOR) = 1.59, CI 1.18-2.14), preterm babies (10% vs. 7%, aOR = 1.66, CI 1.06-2.59), and stillbirth (1.6% vs. 0.4%, aOR = 4.53, CI 1.33-15.50) compared to the normal weight women group. Moreover, post-BS women had a higher risk of low-birth-weight neonates than women with obesity (15% vs. 8%, aOR = 1.77, CI 1.153-2.73), overweight (15% vs. 7%, aOR = 1.63, CI 1.09-2.43), and normal weight (15% vs. 8%, aOR = 1.838, CI 1.23-2.75). Finally, women in the post-BS group were more likely to have low-birth-weight neonates amongst term babies than women with obesity and overweight. Pregnancies with post-BS should be considered a high-risk group for certain medical outcomes and should be monitored closely. Background: Visceral leishmaniasis (VL) is a neglected tropical disease primarily affecting Brazil, East Africa, and India, with India accounting for 18% of the global burden. While VL typically presents with systemic symptoms like fever, weight loss, and splenomegaly, it can occasionally manifest atypically, posing significant diagnostic challenges. Neurological presentations of VL are extremely rare, making them difficult to suspect and diagnose. Cases where VL predominantly presents with neurological symptoms are particularly novel, underscoring the need for heightened awareness of such atypical manifestations in endemic regions., Clinical Case: A 38-year-old man with history of recurrent atypical VL presented with diffuse lower back pain, progressive tingling, numbness, weakness in the lower extremities, and double vision for one month. Clinical and radiological evaluations suggested cauda equina syndrome and cranial nerve palsy, accompanied by generalized lymphadenopathy, subcutaneous nodules, and skin papules. The differential diagnosis initially included disseminated tuberculosis, histoplasmosis, and lymphoma. Cerebrospinal fluid (CSF) analysis revealed an inflammatory syndrome. Histopathology of lymph node and bone marrow revealed Leishmania amastigotes and subcutaneous nodule and skin biopsy revealed inflammatory cells with granulomas. Furthermore, the qPCR test on DNA from a subcutaneous nodule, lymph node, and CSF was positive for Leishmania kinetoplast DNA. The species was further confirmed as Leishmania donovani through ITS-based PCR amplification and sequencing. Finally, a diagnosis of relapse of VL with lymph node, cutaneous, and neurological involvement, including abducens nerve palsy and cauda equina syndrome, was established. He was treated with combination of liposomal amphotericin B and miltefosine, along with intrathecal hyaluronidase, resulting in significant improvement., Conclusion: Unlike previously reported cases with both systemic and neurological symptoms, our patient predominantly presented with neurological manifestations, making this a unique and novel presentation of VL. Background: Few implementation science (IS) measures have been evaluated for validity, reliability and utility - the latter referring to whether a measure captures meaningful aspects of implementation contexts. We present a real-world case study of rigorous measure development in IS that assesses Barriers and Facilitators in Implementation of Task-Sharing in Mental Health services (BeFITS-MH), with the objective of offering lessons-learned and a framework to enhance measurement utility., Methods: We summarize conceptual and empirical work that informed the development of the BeFITS-MH measure, including a description of the Delphi process, detailed translation and local adaptation procedures, and concurrent pilot testing. As validity and reliability are key aspects of measure development, we also report on our process of assessing the measure's construct validity and utility for the implementation outcomes of acceptability, appropriateness, and feasibility., Results: Continuous stakeholder involvement and concurrent pilot testing resulted in several adaptations of the BeFITS-MH measure's structure, scaling, and format to enhance contextual relevance and utility. Adaptations of broad terms such as "program," "provider type," and "type of service" were necessary due to the heterogeneous nature of interventions, type of task-sharing providers employed, and clients served across the three global sites. Item selection benefited from the iterative process, enabling identification of relevance of key aspects of identified barriers and facilitators, and what aspects were common across sites. Program implementers' conceptions of utility regarding the measure's acceptability, appropriateness, and feasibility clustered across several common categories., Conclusions: This case study provides a rigorous, multi-step process for developing a pragmatic IS measure. The process and lessons learned will aid in the teaching, practice and research of IS measurement development. The importance of including experiences and knowledge from different types of stakeholders in different global settings was reinforced and resulted in a more globally useful measure while allowing for locally-relevant adaptation. To increase the relevance of the measure it is important to target actionable domains that predict markers of utility (e.g., successful uptake) per program implementers' preferences. Purpose: CLN3 Batten disease (also known as juvenile neuronal ceroid lipofuscinosis) is a lysosomal storage disorder that typically initiates with retinal degeneration but is followed by seizure onset, motor decline and premature death. Patient-derived CLN3 disease induced pluripotent stem cell-RPE cells show defective phagocytosis of photoreceptor outer segment (POS). Because modifier genes are implicated in CLN3 disease, our goal here was to investigate a direct link between CLN3 mutation and POS phagocytosis defect., Methods: Isogenic control and CLN3 mutant stem cell lines were generated by CRISPR-Cas9-mediated biallelic deletion of exons 7 and 8. A transgenic CLN3Δ7-8/Δ7-8 (CLN3) Yucatan miniswine was also used to study the impact of CLN3Δ7-8/Δ7-8 mutation on POS phagocytosis. POS phagocytosis by cultured RPE cells was analyzed by Western blotting and immunohistochemistry. Electroretinogram, optical coherence tomography and histological analysis of CLN3Δ7-8/Δ7-8 and wild-type miniswine eyes were carried out at 6, 36, or 48 months of age., Results: CLN3Δ7-8/Δ7-8 RPE (CLN3 RPE) displayed decreased POS binding and consequently decreased uptake of POS compared with isogenic control RPE cells. Furthermore, wild-type miniswine RPE cells phagocytosed CLN3Δ7-8/Δ7-8 POS less efficiently than wild-type POS. Consistent with decreased POS phagocytosis, lipofuscin/autofluorescence was decreased in CLN3 miniswine RPE at 36 months of age and was followed by almost complete loss of photoreceptors at 48 months of age., Conclusions: CLN3Δ7-8/Δ7-8 mutation (which affects ≤85% of patients) affects both RPE and POS and leads to photoreceptor cell loss in CLN3 disease. Furthermore, both primary RPE dysfunction and mutant POS independently contribute to impaired POS phagocytosis in CLN3 disease.

Salmonella Typhimurium (STM), a zoonotic pathogen, can adjust its metabolic pathway according to the variations in the partial pressure of atmospheric oxygen and nitrate via fumarate nitrate reductase regulator (Fnr) and NarL, the response regulator for nitrate reductase. Both Fnr and NarL have been individually reported to be the contributors of virulent phenotypes of STM. Hypoxia along with nitrate-rich environment are prevalent in macrophages and the Salmonella-induced inflammatory lumen of the host's large intestine activates both fnr and narL genes. In this study, the double (fnr and narL) knockout STM showed a synergistic reduction in the swimming (62%), swarming (84%) and biofilm density (86%) phenotypes anaerobically in association with its significant aerobic attenuation. The intracellular replication of the double mutant was reduced by 2.3 logs in chicken monocyte-derived macrophages. Furthermore, the competitive index of the double mutant in liver and spleen was found to be 0.3 and 0.44 respectively at 120 h post-infection (PI) in mice. Surprisingly, no double mutant could be recovered from the infected mouse liver 3 days PI. Histopathological findings showed moderate infiltration of mononuclear cells in the large intestine of mice infected with double mutant, but severe infiltration was seen with the wild-type strain., (© 2024. Introduction. Mucormycosis is an aggressive, angioinvasive infection associated with high morbidity and mortality. The disease remains difficult to treat, with limited available antifungal drugs. Consequently, there is an urgent need to develop alternate therapeutics against mucormycosis. In an earlier study, we demonstrated that the non-steroidal anti-inflammatory drug diflunisal impacted the actin cytoskeleton and quorum sensing and inhibited the formation of filopodia-/cytoneme-like extensions in Rhizopus arrhizus . Hypothesis. The non-steroidal anti-inflammatory drug diflunisal could exhibit potential antifungal activity. Aim. This study aimed to investigate the plausible antifungal activity of diflunisal against a range of medically important Mucorales and its combination effect with antifungal drugs. Methodology. The antifungal activity of diflunisal against Rhizopus arrhizus, Lichtheimia corymbifera, Rhizomucor pusillus , Cunninghamella bertholletiae , Mucor indicus, Mucor irregularis and Apophysomyces elegans was evaluated by broth microdilution assay. Allied salicylates were also screened. A combination assay with amphotericin B deoxycholate and posaconazole was performed by fractional inhibitory concentration test. Results. Exposure to diflunisal inhibited Rhizopus arrhizus spore germination in a dose-dependent manner. MICs of diflunisal against different Mucorales ranged from 64 to 2048 µg ml -1 . Remarkably low levels of diflunisal (0.03-2 µg ml -1 ), depending on the strain/species tested, improved the antifungal activity of amphotericin B against mucoralean fungi by twofold (ΣFIC ≈ 0.5-0.508; P< 0.01). Field-emission scanning electron micrographs further confirmed these observations. MICs of posaconazole were unchanged by this compound. Conclusion. Considering that amphotericin B remains the first-line drug against mucormycosis and exhibits dose-dependent side effects in clinical practice, especially nephrotoxicity, the observed additive interaction at remarkably low, clinically achievable levels of diflunisal demonstrates its potential utility as an adjunct therapy against mucoralean fungi.

Congenital infiltrating lipomatosis of facial (CIL-F) is a rare tumour of adipose tissue resulting in a progressive craniofacial deformity with involvement of the underlying bones, muscles and joints. We report a very rare case of CIL-F with left sided temporomandibular joint ankylosis in a young patient who presented with facial swelling and reduced mouth opening. This case highlights the imaging features, differential diagnosis and brief management of CIL-F with special emphasis on CT and MRI findings. Ethnopharmacological Relevance: Astilbe rivularis Buch.-Ham. ex D. Don is a rare medicinal plant, traditionally employed for treating several disorders. The juice, decoction or powder of the roots, rhizomes, leaves and even the entire plant, are used for managing peptic ulcer, diarrhoea, jaundice, sprains and muscular swellings, bone fracture and dislocation of joints, postpartum bleeding and other menstrual disorders. These conventional medicinal uses make Astilbe rivularis a promising candidate for further research., Aim of the Study: This study was designed to explore the neuroprotective potential of hydroethanolic extract of Astilbe rivularis (ARHE) in diabetic neuropathy (DN) in rats., Materials and Methods: GC-MS analysis was used to identify the phytoconstituents present in the plant extract. DN was induced by administration of STZ (55 mg/kg, i.p.), 15 min after NAD (230 mg/kg, i.p.) injection. The rats with fasting blood glucose (FBG) level >250 mg/dl were included in the study. DN was assessed by estimating the level of FBG, lipid profile, and invitro and invivo oxidative stress parameters. Additionally, behavioural parameters like, mechanical hyperalgesia, hot and cold allodynia were estimated to assess diabetic neuropathy. Furthermore, the level of antioxidant enzymes like SOD, GSH, and TBARS in sciatic nerve and inflammatory markers like, TGF-β and IL-6 were measured., Results: Altogether, 30 phytoconstituents were identified including heptafluorobutyric acid, hexadecanoic acid, and beta-sitosterol depicting antioxidant, antidiabetic, and anticancer properties, respectively. Administration of different doses (100, 200, and 400 mg/kg) of ARHE to diabetic rats attenuated elevated blood glucose level and restored lipid profile, body weight, food and water intake, and antioxidant level. Moreover, elevated level of inflammatory markers like, TGF-β and IL-6 was also found to be attenuated in sciatic nerve. Furthermore, ARHE attenuated the pain response assessed by mechanical hyperalgesia and hot and cold allodynia in diabetic neuropathy rats. ARHE also showed inhibitory activity on ALR enzyme and erythrocyte sorbitol accumulation, and ameliorated oxidative stress.
