Terrestrial plant and soil organic carbon stocks are critical for regulating climate change, enhancing soil fertility, and supporting biodiversity. While a global-scale decoupling between plant and soil organic carbon has been documented, the hotspots and interconnections between these two carbon compartments across Africa, the second-largest continent on the planet, have been significantly overlooked. Here, we have compiled over 10,000 existing soil organic carbon observations to generate a high-resolution map, illustrating the distribution pattern of soil organic carbon in Africa. We then showed that above- and below-ground plant carbon are significantly and positively correlated with soil organic carbon across Africa. Both soil and plant carbon compartments shared major hotspots in the tropical regions. Ecosystem functioning depends on biodiversity at multiple trophic levels, yet relationships between multitrophic diversity and ecosystem multifunctionality have been poorly explored, with studies often focusing on individual trophic levels and functions and on specific ecosystem types. Here, we show that plant diversity can affect ecosystem functioning both directly and by affecting other trophic levels. Using data on 13 trophic groups and 13 ecosystem functions from two large biodiversity experiments-one representing temperate grasslands and the other subtropical forests-we found that plant diversity increases multifunctionality through elevated multitrophic diversity. Across both experiments, the association between multitrophic diversity and multifunctionality was stronger than the relationship between the diversity of individual trophic groups and multifunctionality. Our results also suggest that the role of multitrophic diversity is greater in forests than in grasslands. Artificial neural networks (ANNs) have proven to be a useful tool for complex questions that involve large amounts of data. Our use case of predicting soil maps with ANNs is in high demand by government agencies, construction companies, or farmers, given cost and time intensive field work. However, there are two main challenges when applying ANNs. In their most common form, deep learning algorithms do not provide interpretable predictive uncertainty. This means that properties of an ANN such as the certainty and plausibility of the predicted variables, rely on the interpretation by experts rather than being quantified by evaluation metrics validating the ANNs. Further, these algorithms have shown a high confidence in their predictions in areas geographically distant from the training area or areas sparsely covered by training data. To tackle these challenges, we use the Bayesian deep learning approach "last-layer Laplace approximation", which is specifically designed to quantify uncertainty into deep networks, in our explorative study on soil classification. It corrects the overconfident areas without reducing the accuracy of the predictions, giving us a more realistic uncertainty expression of the model's prediction. In our study area in southern Germany, we subdivide the soils into soil regions and as a test case we explicitly exclude two soil regions in the training area but include these regions in the prediction. Our results emphasize the need for uncertainty measurement to obtain more reliable and interpretable results of ANNs, especially for regions far away from the training area. Moreover, the knowledge gained from this research addresses the problem of overconfidence of ANNs and provides valuable information on the predictability of soil types and the identification of knowledge gaps. The microbiota is attributed to be important for initial soil formation under extreme climate conditions, but experimental evidence for its relevance is scarce. To fill this gap, we investigated the impact of in situ microbial communities and their interrelationship with biocrust and plants compared to abiotic controls on soil formation in initial arid and semiarid soils. Additionally, we assessed the response of bacterial communities to climate change. Topsoil and subsoil samples from arid and semiarid sites in the Chilean Coastal Cordillera were incubated for 16 weeks under diurnal temperature and moisture variations to simulate humid climate conditions as part of a climate change scenario. Our findings indicate that microorganism-plant interaction intensified aggregate formation and stabilized soil structure, facilitating initial soil formation. Interestingly, microorganisms alone or in conjunction with biocrust showed no discernible patterns compared to abiotic controls, potentially due to water-masking effects. Arid soils displayed reduced bacterial diversity and developed a new community structure dominated by Proteobacteria , Actinobacteriota , and Planctomycetota , while semiarid soils maintained a consistently dominant community of Acidobacteriota and Proteobacteria . This highlighted a sensitive and specialized bacterial community in arid soils, while semiarid soils exhibited a more complex and stable community. We conclude that microorganism-plant interaction has measurable impacts on initial soil formation in arid and semiarid regions on short time scales under climate change. Plants modulate their phosphorus (P) acquisition strategies (i.e., change in root morphology, exudate composition, and mycorrhizal symbiosis) to adapt to varying soil P availability. However, how community- and species-level P-acquisition strategies change in response to nitrogen (N) supply under different P levels remains unclear. To address this research gap, we conducted an 8-year fully factorial field experiment in an alpine grassland on the Qinghai-Tibet Plateau (QTP) combined with a 12-week glasshouse experiment with four treatments (N addition, P addition, combined N and P addition, and control). In the field experiment (community-level), when P availability was low, N addition increased the release of carboxylate from roots and led to a higher percentage of colonisation by arbuscular mycorrhizal fungi (AMF), along with decreased root length, specific root length (SRL), and total root length colonised by AMF. When P availability was higher, N addition resulted in an increase in the plant's demand for P, accompanied by an increase in root diameter and phosphatase activity. In the glasshouse experiment (species-level), the P-acquisition strategies of grasses and sedge in response to N addition alone mirrored those observed in the field, exhibiting a reduction in root length, SRL, and total root length colonised, but an increased percentage of AMF colonisation. Forbs responded to N addition alone with increased investment in all P-acquisition strategies, especially increased root biomass and length. P-acquisition strategies showed consistent changes among all species in response to combined N and P addition. Our results suggest that increased carboxylate release and AMF colonisation rate are common P-acquisition strategies of plants in alpine grasslands under N-induced P limitation. Drought and the impacts of climate change have led to an escalation in soil salinity and alkalinity across various regions worldwide, including Iran. The Chahardowli Plain in western Iran, in particular, has witnessed a significant intensification of this phenomenon over the past decade. Consequently, modeling of soil attributes that serve as indicators of soil salinity and alkalinity became a priority in this region. To date, only a limited number of studies have been conducted to assess indicators of salinity and alkalinity through spectrometry across diverse spectral ranges. The spectral ranges encompassing mid-infrared (mid-IR), visible, and near-infrared (vis-NIR) spectroscopy were employed to estimate soil properties including sodium adsorption ratio (SAR), exchangeable sodium ratio (ESR), exchangeable sodium percentage (ESP), pH, and electrical conductivity (EC). Five distinct models were employed: Partial Least Squares Regression (PLSR), bootstrapping aggregation PLSR (BgPLSR), Memory-Based Learning (MBL), Random Forest (RF), and Cubist. The calibration and assessment of model performance were carried out using several key metrics including Ratio of Performance to Deviation (RPD) and the coefficient of determination (R 2 ). Analysis of the outcomes indicates that the accuracy and precision of the mid-IR spectra surpassed that of vis-NIR spectra, except for pH, which exhibited a superior RPD compared to other properties. Notably, in the prediction of pH utilizing vis-NIR reflectance spectra, the BgPLSR model exhibited the highest accuracy and precision, boasting an RPD value of 2.56. In the domain of EC prediction, the PLSR model yielded an RPD of 2.64. For SAR, the MBL model achieved an RPD of 2.70, while ESR prediction benefited from the MBL model with an impressive RPD of 4.36. Likewise, the MBL model demonstrated remarkable precision and accuracy in ESP prediction, garnering an RPD of 4.41. The MBL model's efficacy in forecasting with limited datasets was notably pronounced among the models considered. This study underscores the valuable role of spectral predictions in facilitating the work of soil surveyors in gauging salinity and alkalinity indicators. Carbon-focused climate mitigation strategies are becoming increasingly important in forests. However, with ongoing biodiversity declines we require better knowledge of how much such strategies account for biodiversity. We particularly lack information across multiple trophic levels and on established forests, where the interplay between carbon stocks, stand age, and tree diversity might influence carbon-biodiversity relationships. Using a large dataset (>4600 heterotrophic species of 23 taxonomic groups) from secondary, subtropical forests, we tested how multitrophic diversity and diversity within trophic groups relate to aboveground, belowground, and total carbon stocks at different levels of tree species richness and stand age. Our study revealed that aboveground carbon, the key component of climate-based management, was largely unrelated to multitrophic diversity. By contrast, total carbon stocks-that is, including belowground carbon-emerged as a significant predictor of multitrophic diversity. Relationships were nonlinear and strongest for lower trophic levels, but nonsignificant for higher trophic level diversity. Tree species richness and stand age moderated these relationships, suggesting long-term regeneration of forests may be particularly effective in reconciling carbon and biodiversity targets. Multi-scale contextual modelling is an important toolset for environmental mapping. It accounts for spatial dependence by using covariates on multiple spatial scales and incorporates spatial context and structural dependence to environmental properties into machine learning models. For spatial soil modelling, three relevant scales or ranges of scale exist: quasi-local soil formation processes that are independent of the spatial context, short-range catenary processes, and long-range processes related to climate and large-scale terrain settings. Recent studies investigated the spatial dependence of topsoil properties only. We hypothesize that soil properties within a soil profile were formed due to specific interactions between different features and scales of the spatial context, and that there are depth gradients in spatial and structural dependencies. The results showed that for topsoil, features at small to intermediate scales do not increase model accuracy, whereas large scales increase model accuracy. In contrast, subsoil models benefit from all scales-small, intermediate, and large. Based on the differences in relevance, we conclude that the relevant ranges of scales do not only differ in the horizontal domain, but also in the vertical domain across the soil profile. Research in global change ecology relies heavily on global climatic grids derived from estimates of air temperature in open areas at around 2 m above the ground. These climatic grids do not reflect conditions below vegetation canopies and near the ground surface, where critical ecosystem functions occur and most terrestrial species reside. Here, we provide global maps of soil temperature and bioclimatic variables at a 1-km 2 resolution for 0-5 and 5-15 cm soil depth. These maps were created by calculating the difference (i.e. offset) between in situ soil temperature measurements, based on time series from over 1200 1-km 2 pixels (summarized from 8519 unique temperature sensors) across all the world's major terrestrial biomes, and coarse-grained air temperature estimates from ERA5-Land (an atmospheric reanalysis by the European Centre for Medium-Range Weather Forecasts). We show that mean annual soil temperature differs markedly from the corresponding gridded air temperature, by up to 10°C (mean = 3.0 ± 2.1°C), with substantial variation across biomes and seasons. Over the year, soils in cold and/or dry biomes are substantially warmer (+3.6 ± 2.3°C) than gridded air temperature, whereas soils in warm and humid environments are on average slightly cooler (-0.7 ± 2.3°C). The observed substantial and biome-specific offsets emphasize that the projected impacts of climate and climate change on near-surface biodiversity and ecosystem functioning are inaccurately assessed when air rather than soil temperature is used, especially in cold environments. The global soil-related bioclimatic variables provided here are an important step forward for any application in ecology and related disciplines. Soil bacteria play a fundamental role in pedogenesis. However, knowledge about both the impact of climate and slope aspects on microbial communities and the consequences of these items in pedogenesis is lacking. Therefore, soil-bacterial communities from four sites and two different aspects along the climate gradient of the Chilean Coastal Cordillera were investigated. Using a combination of microbiological and physicochemical methods, soils that developed in arid, semi-arid, mediterranean, and humid climates were analyzed. Proteobacteria , Acidobacteria , Chloroflexi , Verrucomicrobia , and Planctomycetes were found to increase in abundance from arid to humid climates, while Actinobacteria and Gemmatimonadetes decreased along the transect. Bacterial-community structure varied with climate and aspect and was influenced by pH, bulk density, plant-available phosphorus, clay, and total organic-matter content. Higher bacterial specialization was found in arid and humid climates and on the south-facing slope and was likely promoted by stable microclimatic conditions. The presence of specialists was associated with ecosystem-functional traits, which shifted from pioneers that accumulated organic matter in arid climates to organic decomposers in humid climates. These findings provide new perspectives on how climate and slope aspects influence the composition and functional capabilities of bacteria, with most of these capabilities being involved in pedogenetic processes.

Ecosystems provide multiple services to humans. However, agricultural systems are usually evaluated on their productivity and economic performance, and a systematic and quantitative assessment of the multifunctionality of agroecosystems including environmental services is missing. Using a long-term farming system experiment, we evaluated and compared the agronomic, economic, and ecological performance of the most widespread arable cropping systems in Europe: organic, conservation, and conventional agriculture. We analyzed 43 agroecosystem properties and determined overall agroecosystem multifunctionality. We show that organic and conservation agriculture promoted ecosystem multifunctionality, especially by enhancing regulating and supporting services, including biodiversity preservation, soil and water quality, and climate mitigation. In contrast, conventional cropping showed reduced multifunctionality but delivered highest yield. Organic production resulted in higher economic performance, thanks to higher product prices and additional support payments. It has been shown that reactive soil minerals, specifically iron(III) (oxyhydr)oxides, can trap organic carbon in soils overlying intact permafrost, and may limit carbon mobilization and degradation as it is observed in other environments. However, the use of iron(III)-bearing minerals as terminal electron acceptors in permafrost environments, and thus their stability and capacity to prevent carbon mobilization during permafrost thaw, is poorly understood. We have followed the dynamic interactions between iron and carbon using a space-for-time approach across a thaw gradient in Abisko (Sweden), where wetlands are expanding rapidly due to permafrost thaw. We show through bulk (selective extractions, EXAFS) and nanoscale analysis (correlative SEM and nanoSIMS) that organic carbon is bound to reactive Fe primarily in the transition between organic and mineral horizons in palsa underlain by intact permafrost (41.8 ± 10.8 mg carbon per g soil, 9.9 to 14.8% of total soil organic carbon). During permafrost thaw, water-logging and O 2 limitation lead to reducing conditions and an increase in abundance of Fe(III)-reducing bacteria which favor mineral dissolution and drive mobilization of both iron and carbon along the thaw gradient. By providing a terminal electron acceptor, this rusty carbon sink is effectively destroyed along the thaw gradient and cannot prevent carbon release with thaw.

Current analyses and predictions of spatially explicit patterns and processes in ecology most often rely on climate data interpolated from standardized weather stations. This interpolated climate data represents long-term average thermal conditions at coarse spatial resolutions only. Hence, many climate-forcing factors that operate at fine spatiotemporal resolutions are overlooked. This is particularly important in relation to effects of observation height (e.g. vegetation, snow and soil characteristics) and in habitats varying in their exposure to radiation, moisture and wind (e.g. topography, radiative forcing or cold-air pooling). Since organisms living close to the ground relate more strongly to these microclimatic conditions than to free-air temperatures, microclimatic ground and near-surface data are needed to provide realistic forecasts of the fate of such organisms under anthropogenic climate change, as well as of the functioning of the ecosystems they live in. To fill this critical gap, we highlight a call for temperature time series submissions to SoilTemp, a geospatial database initiative compiling soil and near-surface temperature data from all over the world. Currently, this database contains time series from 7,538 temperature sensors from 51 countries across all key biomes. Diversity of producers (e.g. plants) usually increases the diversity of associated organisms, but the scale (i.e. the spatial area of plant diversity considered) at which plant diversity acts on other taxa has rarely been studied. Most evidence for cross-taxon diversity relations come from above-ground consumers that directly interact with plants. Experimental tests of plant diversity effects on elusive organisms inhabiting the leaf litter layer, which are important for nutrient cycling and decomposition, are rare. Using a large tree diversity experiment, we tested whether tree diversity at the larger plot (i.e. community) or the smaller neighbourhood scale relates to the abundance, species richness, functional and phylogenetic diversity of leaf litter ants, which are dominant organisms in brown food webs. Contrary to our expectations of scale-independent positive tree diversity effects, ant diversity increased only with plot but not neighbourhood tree diversity. While the exact causal mechanisms are unclear, nest relocation or small-scale competition among ants may explain the stronger tree diversity effects at the plot scale. Our results indicate that even for small and less mobile organisms in the leaf litter, effects of tree diversity are stronger at relatively larger scales. Spatial autocorrelation in the residuals of spatial environmental models can be due to missing covariate information. In many cases, this spatial autocorrelation can be accounted for by using covariates from multiple scales. Here, we propose a data-driven, objective and systematic method for deriving the relevant range of scales, with distinct upper and lower scale limits, for spatial modelling with machine learning and evaluated its effect on modelling accuracy. We also tested an approach that uses the variogram to see whether such an effective scale space can be approximated a priori and at smaller computational cost. Results showed that modelling with an effective scale space can improve spatial modelling with machine learning and that there is a strong correlation between properties of the variogram and the relevant range of scales. Hence, the variogram of a soil property can be used for a priori approximations of the effective scale space for contextual spatial modelling and is therefore an important analytical tool not only in geostatistics, but also for analyzing structural dependencies in contextual spatial modelling.

As limited resources, soils are the largest terrestrial sinks of organic carbon. In this respect, 3D modelling of soil organic carbon (SOC) offers substantial improvements in the understanding and assessment of the spatial distribution of SOC stocks. Previous three-dimensional SOC modelling approaches usually averaged each depth increment for multi-layer two-dimensional predictions. Therefore, these models are limited in their vertical resolution and thus in the interpretability of the soil as a volume as well as in the accuracy of the SOC stock predictions. So far, only few approaches used spatially modelled depth functions for SOC predictions. This study implemented and evaluated an approach that compared polynomial, logarithmic and exponential depth functions using non-linear machine learning techniques, i.e. multivariate adaptive regression splines, random forests and support vector machines to quantify SOC stocks spatially and depth-related in the context of biodiversity and ecosystem functioning research. The legacy datasets used for modelling include profile data for SOC and bulk density (BD), sampled at five depth increments (0-5, 5-10, 10-20, 20-30, 30-50 cm). The samples were taken in an experimental forest in the Chinese subtropics as part of the biodiversity and ecosystem functioning (BEF) China experiment. Here we compared the depth functions by means of the results of the different machine learning approaches obtained based on multi-layer 2D models as well as 3D models. The main findings were (i) that 3rd degree polynomials provided the best results for SOC and BD (R2 = 0.99 and R2 = 0.98; RMSE = 0.36% and 0.07 g cm-3). However, they did not adequately describe the general asymptotic trend of SOC and BD. In this respect the exponential (SOC: R2 = 0.94; RMSE = 0.56%) and logarithmic (BD: R2 = 84; RMSE = 0.21 g cm-3) functions provided more reliable estimates. (ii) random forests with the exponential function for SOC correlated better with the corresponding 2.5D predictions (R2: 0.96 to 0.75), compared to the 3rd degree polynomials (R2: 0.89 to 0.15) which support vector machines fitted best. We recommend not to use polynomial functions with sparsely sampled profiles, as they have many turning points and tend to overfit the data on a given profile. This may limit the spatial prediction capacities. Instead, less adaptive functions with a higher degree of generalisation such as exponential and logarithmic functions should be used to spatially map sparse vertical soil profile datasets. We conclude that spatial prediction of SOC using exponential depth functions, in conjunction with random forests is well suited for 3D SOC stock modelling, and provides much finer vertical resolutions compared to 2.5D approaches., Competing Interests: The authors have declared that no competing interests exist.

Soil properties and terrain attributes are of great interest to explain and model plant productivity and community assembly (hereafter P&CA). Many studies only sample surface soils, and may therefore miss important variation of deeper soil levels. We aimed to identify a critical soil depth in which the relationships between soil properties and P&CA were strongest due to an ideal interplay among soil properties and terrain attributes. On 27 plots in a subtropical Chinese forest varying in tree and herb layer species richness and tree productivity, 29 soil properties in six depth columns and four terrain attributes were analyzed. Soil properties varied with soil depth as did their interrelationships. Non-linearity of soil properties led to critical soil depths in which different P&CA characteristics were explained best (using coefficients of determination). The strongest relationship of soil properties and terrain attributes to most of P&CA characteristics (adj. R 2 ~ 0.7) was encountered using a soil column of 0-16 cm. Thus, depending on the biological signal one is interested in, soil depth sampling has to be adapted. Considering P&CA in subtropical broad-leaved secondary forests, we recommend sampling one bulk sample of a column from 0 cm down to a critical soil depth of 16 cm.

Deconvoluting the relative contributions made by specific biotic and abiotic drivers to soil fungal community compositions facilitates predictions about the functional responses of ecosystems to environmental changes, such as losses of plant diversity, but it is hindered by the complex interactions involved. Experimental assembly of tree species allows separation of the respective effects of plant community composition (biotic components) and soil properties (abiotic components), enabling much greater statistical power than can be achieved in observational studies. We therefore analyzed these contributions by assessing, via pyrotag sequencing of the internal transcribed spacer (ITS2) rDNA region, fungal communities in young subtropical forest plots included in a large experiment on the effects of tree species richness. Spatial variables and soil properties were the main drivers of soil fungal alpha and beta-diversity, implying strong early-stage environmental filtering and dispersal limitation. Tree related variables, such as tree community composition, significantly affected arbuscular mycorrhizal and pathogen fungal community structure, while differences in tree host species and host abundance affected ectomycorrhizal fungal community composition. At this early stage of the experiment, only a limited amount of carbon inputs (rhizodeposits and leaf litter) was being provided to the ecosystem due to the size of the tree saplings, and persisting legacy effects were observed. We thus expect to find increasing tree related effects on fungal community composition as forest development proceeds.

Biodiversity experiments have shown that species loss reduces ecosystem functioning in grassland. To test whether this result can be extrapolated to forests, the main contributors to terrestrial primary productivity, requires large-scale experiments. We manipulated tree species richness by planting more than 150,000 trees in plots with 1 to 16 species. Simulating multiple extinction scenarios, we found that richness strongly increased stand-level productivity. After 8 years, 16-species mixtures had accumulated over twice the amount of carbon found in average monocultures and similar amounts as those of two commercial monocultures. Species richness effects were strongly associated with functional and phylogenetic diversity. A shrub addition treatment reduced tree productivity, but this reduction was smaller at high shrub species richness. Forest ecosystems are an integral component of the global carbon cycle as they take up and release large amounts of C over short time periods (C flux) or accumulate it over longer time periods (C stock). However, there remains uncertainty about whether and in which direction C fluxes and in particular C stocks may differ between forests of high versus low species richness. Based on a comprehensive dataset derived from field-based measurements, we tested the effect of species richness (3-20 tree species) and stand age (22-116 years) on six compartments of above- and below-ground C stocks and four components of C fluxes in subtropical forests in southeast China. Across forest stands, total C stock was 149 ± 12 Mg ha -1 with richness explaining 28.5% and age explaining 29.4% of variation in this measure. Species-rich stands had higher C stocks and fluxes than stands with low richness; and, in addition, old stands had higher C stocks than young ones. Overall, for each additional tree species, the total C stock increased by 6.4%. Our results provide comprehensive evidence for diversity-mediated above- and below-ground C sequestration in species-rich subtropical forests in southeast China. Therefore, afforestation policies in this region and elsewhere should consider a change from the current focus on monocultures to multi-species plantations to increase C fixation and thus slow increasing atmospheric CO 2 concentrations and global warming., (© 2018 The Author(s).)

Human-induced biodiversity change impairs ecosystem functions crucial to human well-being. However, the consequences of this change for ecosystem multifunctionality are poorly understood beyond effects of plant species loss, particularly in regions with high biodiversity across trophic levels. Here we adopt a multitrophic perspective to analyze how biodiversity affects multifunctionality in biodiverse subtropical forests. We consider 22 independent measurements of nine ecosystem functions central to energy and nutrient flow across trophic levels. We find that individual functions and multifunctionality are more strongly affected by the diversity of heterotrophs promoting decomposition and nutrient cycling, and by plant functional-trait diversity and composition, than by tree species richness. Moreover, cascading effects of higher trophic-level diversity on functions originating from lower trophic-level processes highlight that multitrophic biodiversity is key to understanding drivers of multifunctionality. A broader perspective on biodiversity-multifunctionality relationships is crucial for sustainable ecosystem management in light of non-random species loss and intensified biotic disturbances under future environmental change.

Predicting taxonomic classes can be challenging with dataset subject to substantial irregularities due to the involvement of many surveyors. A data pruning approach was used in the present study to reduce such source errors by exploring whether different data pruning methods, which result in different subsets of a major reference soil groups (RSG) - the Plinthosols - would lead to an increase in prediction accuracy of the minor soil groups by using Random Forest (RF). This method was compared to the random oversampling approach. Four datasets were used, including the entire dataset and the pruned dataset, which consisted of 80% and 90% respectively, and standard deviation core range of the Plinthosols data while cutting off all data points belonging to the outer range. The best prediction was achieved when RF was used with recursive feature elimination along with the non-oversampled 90% core range dataset. This model provided a substantial agreement to observation, with a kappa value of 0.57 along with 7% to 35% increase in prediction accuracy for smaller RSG. The reference soil groups in the Dano catchment appeared to be mainly influenced by the wetness index, a proxy for soil moisture distribution.

Colluvial deposits, as the correlate sediments of human-induced soil erosion, depict an excellent archive of land use and landscape history as indicators of human-environment interactions. This study establishes a chronostratigraphy of colluvial deposits and reconstructs past land use dynamics in the Swabian Jura, the Baar and the Black Forest in SW Germany. In the agriculturally favourable Baar area multiple main phases of colluvial deposition, and thus intensified land use, can be identified from the Neolithic to the Modern times. In the unfavourable Swabian Jura increased colluvial deposition began later compared to the more favourable areas in the Baar. The same holds true for the unfavourable areas of the Black Forest, but intensified land use can only be reconstructed for the Middle Ages and Early Modern times instead of for the Bronze and Iron Age as in the Swabian Jura. Land use intensity and settlement dynamics represented by thick, multilayered colluvial deposits increase in the Baar and the Black Forest during the Middle Ages. In between those phases of geomorphodynamic activity and colluviation, stable phases occur, interpreted as phases with sustainable land use or without human presence., Competing Interests: We declare we have no competing interests.

Biodiversity-ecosystem functioning (BEF) research has extended its scope from communities that are short-lived or reshape their structure annually to structurally complex forest ecosystems. The establishment of tree diversity experiments poses specific methodological challenges for assessing the multiple functions provided by forest ecosystems. In particular, methodological inconsistencies and nonstandardized protocols impede the analysis of multifunctionality within, and comparability across the increasing number of tree diversity experiments. By providing an overview on key methods currently applied in one of the largest forest biodiversity experiments, we show how methods differing in scale and simplicity can be combined to retrieve consistent data allowing novel insights into forest ecosystem functioning. Furthermore, we discuss and develop recommendations for the integration and transferability of diverse methodical approaches to present and future forest biodiversity experiments. We identified four principles that should guide basic decisions concerning method selection for tree diversity experiments and forest BEF research: (1) method selection should be directed toward maximizing data density to increase the number of measured variables in each plot. (2) Methods should cover all relevant scales of the experiment to consider scale dependencies of biodiversity effects. (3) The same variable should be evaluated with the same method across space and time for adequate larger-scale and longer-time data analysis and to reduce errors due to changing measurement protocols. (4) Standardized, practical and rapid methods for assessing biodiversity and ecosystem functions should be promoted to increase comparability among forest BEF experiments. We demonstrate that currently available methods provide us with a sophisticated toolbox to improve a synergistic understanding of forest multifunctionality. However, these methods require further adjustment to the specific requirements of structurally complex and long-lived forest ecosystems. By applying methods connecting relevant scales, trophic levels, and above- and belowground ecosystem compartments, knowledge gain from large tree diversity experiments can be optimized.

Ecosystem functioning and human well-being critically depend on numerous species interactions above- and belowground. However, unraveling the structure of multitrophic interaction webs at the ecosystem level is challenging for biodiverse ecosystems. Attempts to identify major relationships between trophic levels usually rely on simplified proxies, such as species diversity. Here, we propose to consider the full information on species composition across trophic levels, using Procrustes correlation and structural equation models. We show that species composition data of a highly diverse subtropical forest-with 5,716 taxa across 25 trophic groups- reveal strong interrelationships among plants, arthropods, and microorganisms, indicating complex multitrophic interactions. We found substantial support for top-down effects of microorganisms belowground, indicating important feedbacks of microbial symbionts, pathogens, and decomposers on plant communities. In contrast, aboveground pathways were characterized by bottom-up control of plants on arthropods, including many non-trophic links. Additional analyses based on diversity patterns revealed much weaker interrelationships. Our study suggests that multitrophic communities in our forest system are structured via top-down effects of belowground biota on plants, which in turn affect aboveground arthropod communities across trophic levels. Moreover, the study shows that the consequences of species loss will be more complex than indicated by studies based solely on diversity.

Knowledge of plant functional traits and trait-environment interactions is important for characterizing species strategies and understanding ecological processes. However, comprehensive field data on both above- and belowground traits, together with their environmental variables are scarce. Biome-scale studies are particularly lacking. Here we present two large-scale data sets that include functional traits of leaves and fine roots and their corresponding soil and climatic variables in China's grasslands. Leaf, fine root, and soil samples were collected in three biogeographic regions: temperate grassland on the Inner Mongolia Plateau, alpine grassland on the Tibetan Plateau, and mountain grassland in the Xinjiang mountain areas. Field data were collected over two periods. The first data set collected between 2003 and 2004 includes 13 foliar traits (leaf mass per area, LMA; photosynthetic nitrogen use efficiency, PNUE; water use efficiency, WUE; stomatal conductance for water vapor, Gs; transpiration rate, TR; mass- and area-based photosynthetic capacity, Amass and Aarea; mass- and area-based carbon concentrations, Cmass and Carea; nitrogen concentrations, Nmass and Narea; and phosphorus concentrations, Pmass and Parea) for 170 species at 173 sites. The second data set collected between 2006 and 2007 includes six sets of analogous traits for both leaves and fine roots (C, N, and P concentrations; leaf thickness/root diameter; specific leaf area, SLA; specific root length, SRL; and tissue density) for 139 species at 82 sites, along with soil attributes (soil total and organic carbon, STC and SOC; total and available N, STN and SAN; total and available P, STP and SAP; pH, bulk density, and moisture). Moreover, associated information was also gathered, including geographical location (latitude, longitude, and altitude), climate (mean annual temperature, MAT; mean annual precipitation, MAP; growing season temperature, GST; growing season precipitation, GSP; potential evapotranspiration, PET; and actual evapotranspiration, AET) and site descriptions (vegetation and soil types). The data sets are unique because they integrate plant above- and belowground traits, climate, and soil factors over broad regional, elevational, and taxonomic ranges in understudied regions (e.g., the Tibetan Plateau). This is the only database on China's grassland species for unrestricted global access. Changes in climatic conditions along geographical gradients greatly affect soil nutrient cycling processes. Yet how climate regimes such as changes in temperature influence soil nitrogen (N) and phosphorus (P) concentrations and their stoichiometry is not well understood. This study investigated the spatial pattern and variability of soil N and P availability as well as their coupling relationships at two soil layers (0-10 and 10-20 cm) along a 4000-km climate transect in two grassland biomes of China, the Inner Mongolian temperate grasslands and the Tibetan alpine grasslands. Our results found that in both grasslands, from cold to warm sites the amounts of soil total N, total P and available P all decreased. By contrast, the amount of available N was positively related to mean annual temperature in the Tibetan grasslands. Meanwhile, with increasing temperature ratio of available N to P significantly increased but the linear relationship between them was considerably reduced. Thus, increasing temperature may not only induce a stoichiometric shift but also loose the coupling between available N and P. This N-P decoupling under warmer conditions was more evident in the Tibetan alpine grasslands where P limitation might become more widespread relative to N as temperatures continue to rise.

In soils halogens (fluorine, chlorine, bromine, iodine) are cycled through the transformation of inorganic halides into organohalogen compounds and vice versa. There is evidence that these reactions are microbially driven but the key enzymes and groups of microorganisms involved are largely unknown. Our aim was to uncover the diversity, abundance and distribution of genes encoding for halogenating and dehalogenating enzymes in a German forest soil by shotgun metagenomic sequencing. Metagenomic libraries of three soil horizons revealed the presence of genera known to be involved in halogenation and dehalogenation processes such as Bradyrhizobium or Pseudomonas. We detected a so far unknown diversity of genes encoding for (de)halogenating enzymes in the soil metagenome including specific and unspecific halogenases as well as metabolic and cometabolic dehalogenases. Genes for non-heme, no-metal chloroperoxidases and haloalkane dehalogenases were the most abundant halogenase and dehalogenase genes, respectively. The high diversity and abundance of (de)halogenating enzymes suggests a strong microbial contribution to natural halogen cycling. This was also confirmed in microcosm experiments in which we quantified the biotic formation of chloroform and bromoform. Knowledge on microorganisms and genes that catalyze (de)halogenation reactions is critical because they are highly relevant to industrial biotechnologies and bioremediation applications.

Any release of anthrax spores in the U.S. would require action to decontaminate the site and restore its use and operations as rapidly as possible. The remediation activity would require environmental sampling, both initially to determine the extent of contamination (hazard mapping) and post-decon to determine that the site is free of contamination (clearance sampling). Whether the spore contamination is within a building or outdoors, collecting and analyzing what could be thousands of samples can become the factor that limits the pace of restoring operations. To address this sampling and analysis bottleneck and decrease the time needed to recover from an anthrax contamination event, this study investigates the use of composite sampling. Pooling or compositing of samples is an established technique to reduce the number of analyses required, and its use for anthrax spore sampling has recently been investigated. However, use of composite sampling in an anthrax spore remediation event will require well-documented and accepted methods. In particular, previous composite sampling studies have focused on sampling from hard surfaces; data on soil sampling are required to extend the procedure to outdoor use. Further, we must consider whether combining liquid samples, thus increasing the volume, lowers the sensitivity of detection and produces false negatives. In this study, methods to composite bacterial spore samples from soil are demonstrated. B. subtilis spore suspensions were used as a surrogate for anthrax spores. Two soils (Arizona Test Dust and sterilized potting soil) were contaminated and spore recovery with composites was shown to match individual sample performance. Results show that dilution can be overcome by concentrating bacterial spores using standard filtration methods. This study shows that composite sampling can be a viable method of pooling samples to reduce the number of analysis that must be performed during anthrax spore remediation.

While functional diversity (FD) has been shown to be positively related to a number of ecosystem functions including biomass production, it may have a much less pronounced effect than that of environmental factors or species-specific properties. Leaf and wood traits can be considered particularly relevant to tree growth, as they reflect a trade-off between resources invested into growth and persistence. Our study focussed on the degree to which early forest growth was driven by FD, the environment (11 variables characterizing abiotic habitat conditions), and community-weighted mean (CWM) values of species traits in the context of a large-scale tree diversity experiment (BEF-China). Growth rates of trees with respect to crown diameter were aggregated across 231 plots (hosting between one and 23 tree species) and related to environmental variables, FD, and CWM, the latter two of which were based on 41 plant functional traits. The effects of each of the three predictor groups were analyzed separately by mixed model optimization and jointly by variance partitioning. Numerous single traits predicted plot-level tree growth, both in the models based on CWMs and FD, but none of the environmental variables was able to predict tree growth. In the best models, environment and FD explained only 4 and 31% of variation in crown growth rates, respectively, while CWM trait values explained 42%. In total, the best models accounted for 51% of crown growth. The marginal role of the selected environmental variables was unexpected, given the high topographic heterogeneity and large size of the experiment, as was the significant impact of FD, demonstrating that positive diversity effects already occur during the early stages in tree plantations.

Soil erosion is a key threat to many ecosystems, especially in subtropical China where high erosion rates occur. While the mechanisms that induce soil erosion on agricultural land are well understood, soil erosion processes in forests have rarely been studied. Throughfall kinetic energy (TKE) is influenced in manifold ways and often determined by the tree's leaf and architectural traits. We investigated the role of species identity in mono-specific stands on TKE by asking to what extent TKE is species-specific and which leaf and architectural traits account for variation in TKE. We measured TKE of 11 different tree species planted in monocultures in a biodiversity-ecosystem-functioning experiment in subtropical China, using sand-filled splash cups during five natural rainfall events in summer 2013. In addition, 14 leaf and tree architectural traits were measured and linked to TKE. Our results showed that TKE was highly species-specific. Highest TKE was found below Choerospondias axillaris and Sapindus saponaria, while Schima superba showed lowest TKE. These species-specific effects were mediated by leaf habit, leaf area (LA), leaf pinnation, leaf margin, stem diameter at ground level (GD), crown base height (CBH), tree height, number of branches and leaf area index (LAI) as biotic factors and throughfall as abiotic factor. Among these, leaf habit, tree height and LA showed the highest effect sizes on TKE and can be considered as major drivers of TKE. TKE was positively influenced by LA, GD, CBH, tree height, LAI, and throughfall amount while it was negatively influenced by the number of branches. TKE was lower in evergreen, simple leaved and dentate leaved than in deciduous, pinnated or entire leaved species. Our results clearly showed that soil erosion in forest plantations can be mitigated by the appropriate choice of tree species.

As CO2 concentrations continue to rise and drive global climate change, much effort has been put into estimating soil carbon (C) stocks and dynamics over time. However, the inconsistent methods employed by researchers hamper the comparability of such works, creating a pressing need to standardize the methods for soil organic C (SOC) quantification by the various methods. Here, we collected 712 soil samples from 36 sites of alpine grasslands on the Tibetan Plateau covering different soil depths and vegetation and soil types. We used an elemental analyzer for soil total C (STC) and an inorganic carbon analyzer for soil inorganic C (SIC), and then defined the difference between STC and SIC as SOCCNS. In addition, we employed the modified Walkley-Black (MWB) method, hereafter SOCMWB. Our results showed that there was a strong correlation between SOCCNS and SOCMWB across the data set, given the application of a correction factor of 1.103. Soil depth and soil type significantly influenced on the recovery, defined as the ratio of SOCMWB to SOCCNS, and the recovery was closely associated with soil carbonate content and pH value as well. The differences of recovery between alpine meadow and steppe were largely driven by soil pH. In addition, statistically, a relatively strong correlation between SOCCNS and STC was also found, suggesting that it is feasible to estimate SOCCNS stocks through the STC data across the Tibetan grasslands. Therefore, our results suggest that in order to accurately estimate the absolute SOC stocks and its change in the Tibetan alpine grasslands, adequate correction of the modified WB measurements is essential with correct consideration of the effects of soil types, vegetation, soil pH and soil depth.

Introduction: Ear reconstruction is a tedious and demanding surgical procedure and the implant framework used is essential for the esthetic result. The outcome of a reconstructed ear, however, is not necessarily limited to the implant shape but rather to the available options of transplantable tissue for coverage. Apart from the visual aesthetics, ear reconstruction subsequently also requires implant dimensions to be adapted to the surgical possibilities. In this article, we have brought different disciplines together to develop a customizable ear model for 3D printing of ear implants., Material and Methods: Computed tomography (CT) scans were made of 4 human cadaver ears before and after soft tissue dissection using a Discovery 750 High Definition Freedom Edition scanner (GE, Milwaukee, WI, USA) and subsequently converted into an STL data set using Mimics Software (Materialise, Leuven, Belgium). These scans were then used to develop a fully adjustable parametric model based on the essential ear anatomy using Rhinoceros and Grasshopper software., Results: To determine the quality of the developed models, directed Hausdorff distance (DHD) was applied as the basis for measuring the similarity between the parametric model and the ear cartilage scanning data. Two methods were used. The mean directed Haussdorff distance (MDHD) was calculated based on the distribution of point sets showing an average similarity of 0.8 mm (±0.05 mm). The mean similarity coefficient (SC) of the model and scan surfaces was 94% with a 2-mm threshold., Conclusion: This study shows that a parametric standard model could be used as a feasible method to generate custom implants based on existing ear images., (Copyright © 2015 European Association for Cranio-Maxillo-Facial Surgery. Environmental selection and dispersal limitation are two of the primary processes structuring biotic communities in ecosystems, but little is known about these processes in shaping soil microbial communities during secondary forest succession. We examined the communities of ectomycorrhizal (EM) fungi in young, intermediate and old forests in a Chinese subtropical ecosystem, using 454 pyrosequencing. The EM fungal community consisted of 393 operational taxonomic units (OTUs), belonging to 21 EM fungal lineages, in which three EM fungal lineages and 11 EM fungal OTUs showed significantly biased occurrence among the young, intermediate and old forests. The EM fungal community was structured by environmental selection and dispersal limitation in old forest, but only by environmental selection in young, intermediate, and whole forests. Furthermore, the EM fungal community was affected by different factors in the different forest successional stages, and the importance of these factors in structuring EM fungal community dramatically decreased along the secondary forest succession series. Nitrous oxide (N2O) contributes 8% to global greenhouse gas emissions. Agricultural sources represent about 60% of anthropogenic N2O emissions. Most agricultural N2O emissions are due to increased fertilizer application. A considerable fraction of nitrogen fertilizers are converted to N2O by microbiological processes (that is, nitrification and denitrification). Soil amended with biochar (charcoal created by pyrolysis of biomass) has been demonstrated to increase crop yield, improve soil quality and affect greenhouse gas emissions, for example, reduce N2O emissions. Despite several studies on variations in the general microbial community structure due to soil biochar amendment, hitherto the specific role of the nitrogen cycling microbial community in mitigating soil N2O emissions has not been subject of systematic investigation. We performed a microcosm study with a water-saturated soil amended with different amounts (0%, 2% and 10% (w/w)) of high-temperature biochar. By quantifying the abundance and activity of functional marker genes of microbial nitrogen fixation (nifH), nitrification (amoA) and denitrification (nirK, nirS and nosZ) using quantitative PCR we found that biochar addition enhanced microbial nitrous oxide reduction and increased the abundance of microorganisms capable of N2-fixation. Soil biochar amendment increased the relative gene and transcript copy numbers of the nosZ-encoded bacterial N2O reductase, suggesting a mechanistic link to the observed reduction in N2O emissions. Our findings contribute to a better understanding of the impact of biochar on the nitrogen cycling microbial community and the consequences of soil biochar amendment for microbial nitrogen transformation processes and N2O emissions from soil.

In densely populated countries like China, clean water is one of the most challenging issues of prospective politics and environmental planning. Water pollution and eutrophication by excessive input of nitrogen and phosphorous from nonpoint sources is mostly linked to soil erosion from agricultural land. In order to prevent such water pollution by diffuse matter fluxes, knowledge about the extent of soil loss and the spatial distribution of hot spots of soil erosion is essential. In remote areas such as the mountainous regions of the upper and middle reaches of the Yangtze River, rainfall data are scarce. Since rainfall erosivity is one of the key factors in soil erosion modeling, e.g., expressed as R factor in the Revised Universal Soil Loss Equation model, a methodology is needed to spatially determine rainfall erosivity. Our study aims at the approximation and spatial regionalization of rainfall erosivity from sparse data in the large (3,200 km(2)) and strongly mountainous catchment of the Xiangxi River, a first order tributary to the Yangtze River close to the Three Gorges Dam. As data on rainfall were only obtainable in daily records for one climate station in the central part of the catchment and five stations in its surrounding area, we approximated rainfall erosivity as R factors using regression analysis combined with elevation bands derived from a digital elevation model. The mean annual R factor (R a) amounts for approximately 5,222 MJ mm ha(-1) h(-1) a(-1). With increasing altitudes, R a rises up to maximum 7,547 MJ mm ha(-1) h(-1) a(-1) at an altitude of 3,078 m a.s.l. At the outlet of the Xiangxi catchment erosivity is at minimum with approximate R a=1,986 MJ mm ha(-1) h(-1) a(-1). The comparison of our results with R factors from high-resolution measurements at comparable study sites close to the Xiangxi catchment shows good consistance and allows us to calculate grid-based R a as input for a spatially high-resolution and area-specific assessment of soil erosion risk.

Fungal diversity and community composition are mainly related to soil and vegetation factors. However, the relative contribution of the different drivers remains largely unexplored, especially in subtropical forest ecosystems. We studied the fungal diversity and community composition of soils sampled from 12 comparative study plots representing three forest age classes (Young: 10-40 yrs; Medium: 40-80 yrs; Old: ≥80 yrs) in Gutianshan National Nature Reserve in South-eastern China. Soil fungal communities were assessed employing ITS rDNA pyrotag sequencing. Members of Basidiomycota and Ascomycota dominated the fungal community, with 22 putative ectomycorrhizal fungal families, where Russulaceae and Thelephoraceae were the most abundant taxa. Analysis of similarity showed that the fungal community composition significantly differed among the three forest age classes. Forest age class, elevation of the study plots, and soil organic carbon (SOC) were the most important factors shaping the fungal community composition. We found a significant correlation between plant and fungal communities at different taxonomic and functional group levels, including a strong relationship between ectomycorrhizal fungal and non-ectomycorrhizal plant communities. Our results suggest that in subtropical forests, plant species community composition is the main driver of the soil fungal diversity and community composition.

The Tibetan Plateau reacts particularly sensitively to possible effects of climate change. Approximately two thirds of the total area is affected by permafrost. To get a better understanding of the role of permafrost on soil organic carbon pools and stocks, investigations were carried out including both discontinuous (site Huashixia, HUA) and continuous permafrost (site Wudaoliang, WUD). Three organic carbon fractions were isolated using density separation combined with ultrasonic dispersion: the light fractions (<1.6 g cm(-3)) of free particulate organic matter (FPOM) and occluded particulate organic matter (OPOM), plus a heavy fraction (>1.6 g cm(-3)) of mineral associated organic matter (MOM). The fractions were analyzed for C, N, and their portion of organic C. FPOM contained an average SOC content of 252 g kg(-1). Higher SOC contents (320 g kg(-1)) were found in OPOM while MOM had the lowest SOC contents (29 g kg(-1)). Due to their lower density the easily decomposable fractions FPOM and OPOM contribute 27% (HUA) and 22% (WUD) to the total SOC stocks. In HUA mean SOC stocks (0-30 cm depth) account for 10.4 kg m(-2), compared to 3.4 kg m(-2) in WUD. 53% of the SOC is stored in the upper 10 cm in WUD, in HUA only 39%. Highest POM values of 36% occurred in profiles with high soil moisture content. SOC stocks, soil moisture and active layer thickness correlated strongly in discontinuous permafrost while no correlation between SOC stocks and active layer thickness and only a weak relation between soil moisture and SOC stocks could be found in continuous permafrost. Consequently, permafrost-affected soils in discontinuous permafrost environments are susceptible to soil moisture changes due to alterations in quantity and seasonal distribution of precipitation, increasing temperature and therefore evaporation.

Throughfall kinetic energy (TKE) plays an important role in soil erosion in forests. We studied TKE as a function of biodiversity, functional diversity as well as structural stand variables in a secondary subtropical broad-leaved forest in the Gutianshan National Nature Reserve (GNNR) in south-east China, a biodiversity hotspot in the northern hemisphere with more than 250 woody species present. Using a mixed model approach we could identify significant effects of all these variables on TKE: TKE increased with rarefied tree species richness and decreased with increasing proportion of needle-leaved species and increasing leaf area index (LAI). Furthermore, for average rainfall amounts TKE was decreasing with tree canopy height whereas for high rainfall amounts this was not the case. The spatial pattern of throughfall was stable across several rain events. The temporal variation of TKE decreased with rainfall intensity and increased with tree diversity. Our results show that more diverse forest stands over the season have to cope with higher cumulative raindrop energy than less diverse stands. However, the kinetic energy (KE) of one single raindrop is less predictable in diverse stands since the variability in KE is higher. This paper is the first to contribute to the understanding of the ecosystem function of soil erosion prevention in diverse subtropical forests.

The Tibetan Plateau is an essential area to study the potential feedback effects of soils to climate change due to the rapid rise in its air temperature in the past several decades and the large amounts of soil organic carbon (SOC) stocks, particularly in the permafrost. Yet it is one of the most under-investigated regions in soil respiration (Rs) studies. Here, Rs rates were measured at 42 sites in alpine grasslands (including alpine steppes and meadows) along a transect across the Tibetan Plateau during the peak growing season of 2006 and 2007 in order to test whether: (1) belowground biomass (BGB) is most closely related to spatial variation in Rs due to high root biomass density, and (2) soil temperature significantly influences spatial pattern of Rs owing to metabolic limitation from the low temperature in cold, high-altitude ecosystems. The average daily mean Rs of the alpine grasslands at peak growing season was 3.92 µmol CO(2) m(-2) s(-1), ranging from 0.39 to 12.88 µmol CO(2) m(-2) s(-1), with average daily mean Rs of 2.01 and 5.49 µmol CO(2) m(-2) s(-1) for steppes and meadows, respectively. By regression tree analysis, BGB, aboveground biomass (AGB), SOC, soil moisture (SM), and vegetation type were selected out of 15 variables examined, as the factors influencing large-scale variation in Rs. With a structural equation modelling approach, we found only BGB and SM had direct effects on Rs, while other factors indirectly affecting Rs through BGB or SM. Most (80%) of the variation in Rs could be attributed to the difference in BGB among sites. BGB and SM together accounted for the majority (82%) of spatial patterns of Rs. Our results only support the first hypothesis, suggesting that models incorporating BGB and SM can improve Rs estimation at regional scale.

Background: No standard treatment for locally advanced pancreatic cancer (LAPC) is defined., Patients and Methods: Within a multi-centre, randomised phase II trial, 95 patients with LAPC were assigned to three different chemoradiotherapy (CRT) regimens: patients received conventionally fractionated radiotherapy of 50 Gy and were randomised to concurrent 5-fluorouracil (350 mg m(-2) per day on each day of radiotherapy, RT-5-FU arm), concurrent gemcitabine (300 mg m(-2)), and cisplatin (30 mg m(-2)) on days 1, 8, 22, and 29 (RT-GC arm), or the same concurrent treatment followed by sequential full-dose gemcitabine (1000 mg m(-2)) and cisplatin (50 mg m(-2)) every 2 weeks (RT-GC+GC arm). Primary end point was the overall survival (OS) rate after 9 months., Results: The 9-month OS rate was 58% in the RT-5-FU arm, 52% in the RT-GC arm, and 45% in the RT-GC+GC arm. Corresponding median survival times were 9.6, 9.3, and 7.3 months (P=0.61) respectively. The intent-to-treat response rate was 19, 22, and 13% respectively. Median progression-free survival was estimated with 4.0, 5.6, and 6.0 months (P=0.21). Grade 3/4 haematological toxicities were more frequent in the two GC-containing arms, no grade 3/4 febrile neutropaenia was observed., Conclusion: None of the three CRT regimens tested met the investigators' definition for efficacy; the median OS was similar to those previously reported with gemcitabine alone in LAPC.

Background: Economic aspects and patient-reported outcomes play an increasing role in the choice of therapeutic options. The aim of the LIVE-DE study (Long-acting insulin glargine versus NPH insulin cost evaluation in Germany[DE]) was to assess expenditures incurred in the care of diabetic patients, as well treatment satisfaction of patients with type 2 diabetes treated with insulin glargine (GLAR) or NPH insulin (NPH)., Patients and Methods: A retrospective, non-interventional, cross-sectional study was undertaken in Germany of 1,602 insulin-treated patients (982 on GLAR, 620 on NPH), enrolled from 199 randomly selected general practitioner or internal medicine specialist practices. Total cost of diabetes care (insulins, oral antidiabetic drugs, glucagon use, consumables for insulin administration and blood glucose self-monitoring devices) were calculated from total recorded expenditures, for a period of six months, from the perspective of statutory health insurance. Cost data were obtained from publicly available sources, based on the prices in the year 2007. Patient treatment satisfaction was assessed using previously validated questionnaires (SF-12, PAID, DTSQ, ITEQ)., Results: Physicians prescribed GLAR more often than NPH combined with oral antidiabetic drugs (43 % vs 16%), whereas NPH was more often used in an intensified insulin regimen compared to GLAR (79 % vs 49%). The mean total costs per patient over six months were lower in GLAR than NPH treated patients (658258 vs 685242 Euros [EUR]; p<0.001). The higher drug costs for basal insulin in the GLAR group (19497 vs 11674 EUR) were counterbalanced by lower costs for bolus insulin (96133 vs 158133 EUR), test strips (287137 vs 321142 EUR) and needles (4031 vs 4640 EUR). Only in the NPH group was glucagon use documented (in four patients). Patients treated with GLAR reported significantly higher treatment satisfaction. After adjustment of empirical results (by analysis of covariance), mean total costs of diabetes were higher in GLAR patients (+73.1 EUR; p<0.001). But treatment satisfaction remained significantly higher with GLAR., Conclusion: Based on the comparison of total diabetes treatment costs under real-life conditions between glargine and NPH insulin based treatment regimens, these results indicate that the choice of a given treatment should be determined by medical advantages and patients' preferences. Because of a lower injection rate and a higher patient treatment satisfaction, the use of glargine as first-line therapeutic approach is justified in order to achieve target glycemic control in insulin dependent type 2 diabetics.

Background: Advanced pancreatic cancer, in addition to its high mortality, is characterized by one of the highest rates of venous thromboembolic events (VTE) as compared to other types of cancer. Enoxaparin, a low molecular weight heparin (LMWH), has proven to be effective for the prevention and treatment of VTE in surgical and general medical patients. Results of some small studies suggest that this benefit might extend to patients with cancer, however, enoxaparin is not currently indicated for this use. This phase IIb study was designed to analyze the efficacy of enoxaparin in patients with locally advanced or metastatic pancreatic cancer undergoing systemic chemotherapy., Methods: The aim of this prospective multicenter trial is to compare concomitant treatment with enoxaparin to no anticoagulation in 540 patients. Primary endpoint is the incidence of clinically relevant VTE (symptomatic deep venous thrombosis (DVT) of the leg and/or pelvic and/or pulmonary embolism (PE)) within the first 3 months. Secondary endpoints include the incidence of symptomatic and asymptomatic VTE after 6, 9 and 12 months as well as remission at 3, 6, 9 and 12 months, overall survival and bleeding., Trial Registration: isrctn.org identifier CCT-NAPN-16752, controlled-trials.com identifier: ISRCTN02140505., Results: An interim analysis for safety performed after inclusion of 152 patients revealed no increased risk of bleeding (5 pts vs. 6 pts, Chi2: 0.763)., Conclusion: PROSPECT is a pivotal study in elucidating the role of low molecular weight heparins in advanced pancreatic cancer. Its results will lead to a new understanding of the role of heparins in the prevention of venous thromboembolism and of their effect on survival, remission rates and toxicity of chemotherapeutic regimens.

Gastroesophageal reflux disease (GERD) is a chronic, relapsing disease that can progress to major complications. Affected patients have poorer health-related quality of life than the general population. As GERD requires continued therapy to prevent relapse and complications, most patients with erosive esophagitis require long-term acid suppressive treatment. Thus GERD results in a significant cost burden and poor health-related quality of life. The effective treatment of GERD provides symptom resolution and high rates of remission in erosive esophagitis, lowers the incidence of GERD complications, improves health-related quality of life, and reduces the cost of this disease. Proton pump inhibitors are accepted as the most effective initial and maintenance treatment for GERD. Oral pantoprazole is a safe, well tolerated and effective initial and maintenance treatment for patients with nonerosive GERD or erosive esophagitis. Oral pantoprazole has greater efficacy than histamine H(2)-receptor antagonists and generally similar efficacy to other proton pump inhibitors for the initial and maintenance treatment of GERD. In addition, oral pantoprazole has been shown to improve the quality of life of patients with GERD and is associated with high levels of patient satisfaction with therapy. GERD appears to be more common and more severe in the elderly, and pantoprazole has shown to be an effective treatment for this at-risk population.

Objective: The efficacy of pantoprazole as on-demand therapy for the long-term management of patients with mild gastro-oesophageal reflux disease (GORD) has been demonstrated in clinical studies. In this study, the efficacy of pantoprazole 20mg and esomeprazole 20mg as on-demand therapy for relief of symptoms of mild GORD was compared., Methods: Patients with reflux oesophagitis grade A or B (Los Angeles classification) or endoscopy-negative reflux disease (enGORD) were treated with pantoprazole 20mg once daily for 28 days during the acute phase (AP, n = 236). Patients without heartburn during the final 3 days of the AP entered the long-term phase (LTP, n = 199) and were randomised to either pantoprazole 20mg or esomeprazole 20mg as on-demand treatment for 6 months. Antacids were provided as rescue medication during this phase. The mean intensities of the symptoms of heartburn, acid eructation and pain on swallowing, both separately and as a combined symptom score, together with the mean duration of these symptoms during on-demand treatment, were compared between the two treatment groups. The number of tablets taken was also compared., Results: After 4 weeks of treatment with pantoprazole, 87.3% of patients had relief from heartburn, 74.1% from epigastric pain and 80.8% from acid eructation, according to the investigator assessment. A total of 236 patients were eligible for the on-demand phase. Based on patient diary data, on-demand treatment with pantoprazole resulted in significantly lower mean intensity of heartburn compared with that in the esomeprazole group (1.12 for pantoprazole and 1.32 for esomeprazole, respectively [p = 0.012], in the intention-to-treat [ITT] population). The mean symptom intensities of acid eructation and pain on swallowing, together with the duration of these symptoms, were comparable in the two treatment groups. The combined symptom score of the three symptoms heartburn, acid eructation and pain on swallowing was numerically lower in the pantoprazole group compared with the esomeprazole group (1.72 vs 1.99, respectively, in the ITT population). Tablet intake was comparable in both groups. Relief of symptoms in Helicobacter pylori-positive and -negative patients was also similar in both treatment groups. Both treatments were well tolerated with a good safety profile., Conclusion: On-demand therapy with either pantoprazole 20mg or esomeprazole 20mg is a comparably effective treatment strategy for the long-term treatment of non-erosive and mild GORD. However, the mean intensity of heartburn was significantly lower with pantoprazole treatment.

Background: Few clinical studies have demonstrated the efficacy of proton pump inhibitors (PPIs) as on-demand therapy for the long-term management of patients with mild gastro-oesophageal reflux disease (GORD). On-demand therapy with pantoprazole 20mg may offer an effective strategy for the management of mild GORD., Methods: Patients with mild GORD grade 0 or 1 and heartburn of at least moderate intensity at the start of the study were treated with pantoprazole 20mg once daily for 28 days (n = 254). Patients with no heartburn during the final 3 days of the acute treatment phase entered the long-term treatment phase and took pantoprazole 20mg on demand for 6 months. Antacids were provided as rescue medication during this phase. The primary endpoint was the percentage of patients unwilling to continue the study because of insufficient control of symptoms. In addition, the intensities and the relapse rates for heartburn, acid eructation and epigastric pain were determined., Results: At the end of the acute treatment phase, 234 of 254 patients (92.1%) were free from heartburn and entered the long-term treatment phase (intention-to-treat [ITT]). The time until study discontinuation because of inadequate control of heartburn during the on-demand treatment phase ranged from 9 to 147 days. In the ITT population, only 2.8% of patients were unwilling to continue the study because of insufficient heartburn control. Relapse rates were 4.3% for heartburn, 2.3% for acid eructation and 1.0% for epigastric pain. After 6 months of on-demand treatment, 82% of patients stated that improvements in their symptoms were maintained. For symptom control during the on-demand phase, patients took one tablet of pantoprazole 20mg every 2.3 days. Additionally, patients took one magaldrate tablet every 2.4 days., Conclusion: Results of this pilot study show that on-demand therapy with pantoprazole 20mg appears to be an effective treatment strategy for the long-term treatment of mild GORD. Treatment acceptance by patients was high.

Aims: To compare safety and efficacy of on-demand pantoprazole 20 mg/40 mg versus placebo in the long-term management of patients with mild gastroesophageal reflux disease (GERD) after heartburn relief., Methods: A total of 634 patients with endoscopically confirmed GERD grade 0/I and heartburn were included. During the acute phase, patients were treated with pantoprazole 20 mg once daily for 4 weeks. Those patients relieved from heartburn entered the long-term phase, and were randomly assigned to either treatment group pantoprazole 20 mg, 40 mg or placebo. Over 6 months, patients took study medication on demand (antacids as rescue medication) and discontinued the drug once symptoms abated., Results: After 4 weeks a total of 87.1%/90.0% of patients were free of heartburn (ITT/PP), and entered the subsequent long-term phase. The perceived average daily symptom load (placebo: 3.93, pantoprazole 20 mg: 2.91, pantoprazole 40 mg: 2.71, ITT) and the number of antacid tablets taken (average number, placebo: 0.68, pantoprazole 20 mg: 0.45, pantoprazole 40 mg: 0.33, ITT) were significantly higher in the placebo than in both pantoprazole groups (p<0.0001), with no statistically significant difference between the two pantoprazole groups. The discontinuation rate due to insufficient control of heartburn was significantly lower in both pantoprazole groups compared to placebo (placebo: 10.9, pantoprazole 20 mg: 2.8, pantoprazole 40 mg: 0.9, ITT)., Conclusions: Our findings favor on-demand treatment with pantoprazole 20 mg for the long-term management of heartburn in patients with uncomplicated GERD (grade 0/I) with superiority to placebo., (Copyright (c) 2005 S. Aim: To compare the efficacy of pantoprazole and esomeprazole for the treatment of gastro-oesophageal reflux disease- (GERD-) related symptoms., Methods: In this multicentre, randomized, double-blind study 217 patients [intention-to-treat (ITT) population] diagnosed with endoscopically proven GERD grade B/C received pantoprazole (40 mg once daily (o.d.), n = 112] or esomeprazole (40 mg o.d/, n = 105) for 4 weeks. Patients recorded GERD-related symptoms (daytime and night-time) using diaries (daily), and/or by telephone interviews (every third day) and completed the Gastrointestinal Symptom Rating Scale (GSRS) questionnaire. The area under the time curve (AUC) for the sum score of GERD-related symptoms (symptom load of each patient during the treatment) and the time to reach adequate relief from GERD-related symptoms were calculated., Results: Patients reported first adequate relief from daytime GERD-related symptoms after a mean of 3.7 (pantoprazole) and 5.9 days (esomeprazole) (P = 0.034); the values for the night-time were 1.7 and 3.5 days, respectively (P = 0.012, ITT). The AUCs for the single symptoms and the sum scores were comparable., Conclusions: Treatment with pantoprazole resulted in significantly faster first-time relief from daytime and night-time GERD-related symptoms than esomeprazole. Pantoprazole and esomeprazole were similar with respect to reduction of load of GERD-related symptoms.
