Heavy metal pollution poses a serious threat to crops growth and yield. Recently, nanoparticles (NPs) have emerged as a promising strategy to mitigate the negative effect of heavy metal on crop growth. This study investigated the beneficial effects of copper oxide nanoparticles (CuO NPs) on the morphological and physiological-biochemical traits of rice seedlings (Oryza sativa L.) under cadmium (Cd) stress. The results demonstrated that the application of CuO NPs increased the contents of nutrition elements in shoots and roots as well as photosynthetic pigments, consequently improving the growth of rice seedlings under Cd stress, especially at low level of Cd stress. Meanwhile, CuO NPs obviously decreased the Cd accumulation in the rice seedlings and immobilized Cd in less toxic chemical forms and subcellular compartments. Moreover, CuO NPs modulated the antioxidant system, ameliorating oxidative damage and membrane injury caused by Cd. Multivariate analysis established correlations between physio-biochemical parameters and further revealed the mitigation of Cd damage to rice seedlings by CuO NPs was associated with inhibition Cd accumulation, altering Cd chemical form and subcellular distribution, increasing the contents of mineral nutrients, photosynthetic pigments and secondary metabolites and antioxidant enzyme activities, and reducing oxidative damage. As widely acknowledged, wastewater treatment plants (WWTPs) stand as significant contributors to the presence of microplastics in surface water. Nonetheless, there exists a notable research gap regarding the extent of potential pollution resulting from the concurrent and uninterrupted discharges originating from multiple WWTPs into small-scale receiving water bodies. This study endeavors to address this knowledge deficit by conducting a thorough investigation into the prevalence of microplastics in surface water. The research encompasses seven distinct locations within the Changzhou section of the Beijing-Hangzhou Grand Canal and the effluent of three WWTPs situated along the tributary. The results indicate differences in the distribution of microplastics in surface waters of mainstream and tributaries. While the microplastic abundance and composition showed little variation along the main stream, the tributaries displayed an overall increasing trend in microplastic abundance from upstream to downstream. Notably, the major contributors to this increase were fragments, fiber particles, and microplastics with particle sizes ranging from 100 to 300 μm. Considering that the primary distinction between the tributaries and the mainstream is the presence of the three WWTPs along the tributaries, the study conducted a correlation analysis between river surface water and effluents from these plants. The results indicated a stronger correlation between the tributaries and the effluents, suggesting that WWTPs are one of the primary factors contributing to the elevated levels of microplastics in the tributaries. Finally, a comparative analysis of microplastic abundance in the Beijing-Hangzhou Grand Canal's Changzhou section and other regions was conducted. The findings revealed that the microplastic pollution level in the Beijing-Hangzhou Grand Canal's Changzhou section is higher than that in most other rivers. A chitosan-based nanocomposite film (CSC) was developed by mixing chitosan (CS, 2 %, v/v) and copper oxide nanoparticles (CuO NPs, 500 μg∙mL -1 ) synthesized using Alpinia officinarum extract for the safe storage of mango fruit. The effects of CuO NPs on the morphological, mechanical, thermal, physical and antifungal properties of the CS films and postharvest quality of mango fruit were determined. Scanning electron microscopy (SEM) analysis confirmed that CuO NPs were uniformly dispersed into the CS matrix. X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR) profiles showed that intermolecular H-bondings occurred between CS and CuO NPs, accompanied by decreased crystallinity and increased amorphous structure. In comparison to the pure CS film, addition of CuO NPs obviously improved the morphological, mechanical, thermal, physical and antifungal properties of CSC film. CSC coating treatment obviously delayed the fruit decay and yellowing, as well as reduced losses of weight and firmness of mango (Mangifera indica L.) fruit during the storage, when compared with the control and CS coating treatment. Meanwhile, it significantly decreased the respiration rate and ethylene generation and maintained high level of ascorbic acid (AsA), titratable acid (TA) and soluble sugar content (SSC) of the fruit during the storage. Notably, Cu presented in the CSC film was restrained to the peel, indicating that the CSC coated mango fruit had good edible safety. Principal component analysis (PCA) confirmed that CSC coating played a positive role in mango preservation. Anthracnose decay caused by Colletotrichum gloeosporioides greatly shortens the shelf life and commercial quality of mango fruit. Putrescine (1,4-Diaminobutane) is involved in modulating plant defense to various environmental stresses. In this research, in vivo and in vitro tests were used to explore the antifungal activity and the underlying mechanism of putrescine against C. gloeosporioides in mango fruit after harvested. In vivo tests suggested that putrescine markedly delayed the occurrence of disease and limited the spots expansion on inoculated mango fruit. Further analysis exhibited that putrescine treatment enhanced disease resistance, along with enhanced activities of chitinase (CHI), β-1,3-glucanase (GLU), phenylalanine ammonia-lyase (PAL), cinnamate-4-hydroxylase (C4H), 4-coumarate coenzyme A ligase (4CL), polyphenol oxidase (PPO) and the accumulation of lignin, flavonoid, phenolics, and anthocyanin in infected mango fruit. In addition, in vitro tests showed that putrescine exerted strongly antifungal activity against C. gloeosporioides. Putrescine induced the production of reactive oxygen species (ROS) and severe lipid peroxidation damage in C. gloeosporioides mycelia, resulting in the leakage of soluble protein, soluble sugar, nucleic acids, K + and Ca 2+ of C. gloeosporioides mycelia. The mycelium treated with putrescine showed severe deformity and shrinkage, and even cracking. Dental caries is one of the most common chronic diseases worldwide. Streptococcus mutans and Candida albicans are two major pathogens associated with dental caries. Several recent studies revealed that Lactobacillus plantarum inhibits S. mutans and C. albicans in biofilms and in a rodent model of dental caries. The aim of this study was to investigate the dose-dependent effect of L. plantarum against S. mutans and C. albicans in a planktonic model that simulated a high-caries-risk clinical condition. Mono-, dual-, and multi-species models were utilized, with five doses of L. plantarum (ranging from 1.0 × 10 4 to 1.0 × 10 8 CFU/mL). Real-time PCR was used to assess the expression of the virulence genes of C. albicans and S. mutans and the genes of L. plantarum . Student's t -tests and one-way ANOVA, followed by post hoc tests, were employed to compare the cell viability and gene expression among groups. A dose-dependent inhibition on C. albicans and S. mutans was observed with increased dosages of L. plantarum . L. plantarum at 10 8 CFU/mL demonstrated the highest antibacterial and antifungal inhibitory effect in the dual- and multi-species models. Specifically, at 20 h, the growth of C. albicans and S. mutans was suppressed by 1.5 and 5 logs, respectively ( p < 0.05). The antifungal and antibacterial effects were attenuated in lower doses of L. plantarum (10 4 -10 7 CFU/mL). The expression of C. albicans HWP1 and ECE 1 genes and S. mutans lacC and lacG genes were significantly downregulated with an added 10 8 CFU/mL of L. plantarum ( p < 0.05). The addition of 10 8 CFU/mL L. plantarum further inhibited the hyphae or pseudohyphae formation of C. albicans . In summary, L. plantarum demonstrated dose-dependent antifungal and antibacterial effects against C. albicans and S. mutans . L. plantarum emerged as a promising candidate for the creation of novel antimicrobial probiotic products targeting dental caries prevention. Further research is warranted to identify the functional metabolites produced by L. plantarum at different dosages when interacting with C. albicans and S. mutans .

Background and Objectives : Tooth whitening is a relatively conservative and effective option to treat discolored teeth. However, questions remain whether in-office or at-home tooth whitening products with short treatment durations are as effective and stable as products with longer treatment durations. Materials and Methods : Forty human third molars with intact enamel surfaces were divided into four groups of ten each, subjected to discoloration challenges with coffee for 60 h, and they were treated with four professional tooth whitening systems: two for take-home use-6% hydrogen peroxide for 30 min/d for a total of 7 h in 14 days (HP6), 10% carbamide peroxide for 10 h/d for 140 h in 14 days (CP10), as well as two for in-office use-35% HP for 10 min × 3 (HP35) for a total of 30 min and 40% HP for 20 min × 3 (HP40) for a total of 60 min. Teeth colors were assessed in the CIE L*a*b* color space with a spectrophotometer immediately and six months after whitening treatments. Surface roughness (Sa) for the treated and untreated enamel surfaces of the teeth in all groups were evaluated with a three-dimensional laser scanning microscope after six months. Results : No significant differences were found between HP6 and CP10 groups immediately after whitening (∆ E 10.6 ± 1.6 vs. 11.4 ± 1.7, p > 0.05) and at six months after treatments (∆ E 9.0 ± 1.9 vs. 9.2 ± 2.5, p > 0.05), or between HP35 and HP40 groups immediately after whitening (∆ E 5.9 ± 1.2 vs. 5.3 ± 1.7, p > 0.05) and at six months after treatments (∆ E 7.2 ± 1.6 vs. 7.7 ± 1.3, p > 0.05). The two at-home whitening systems achieved significantly better whitening outcomes than the two in-office products immediately after whitening ( p < 0.05). However, at six months after treatments, the differences between at-home and in-office treatments had narrowed significantly ( p > 0.05). There were no statistically significant differences with respect to the Sa values between the treated and untreated surfaces ( p > 0.05). Conclusions : Tooth whitening products in the same product category have similar whitening efficacies, despite significant differences in treatment durations (7 vs. 140 h, and 30 min vs. 60 min, respectively). Take-home products achieved better whitening outcomes than in-office products, but they needed 14 to 280 times longer treatment durations.

Ecological approaches can help to correct oral microbial dysbiosis and drive the advent and persistence of a symbiotic oral microbiome, which benefits long-term dental caries control. The aim of this study was to investigate the impact of the prebiotic Galacto-oligosaccharide (GOS) on the growth of probiotics L. plantarum 14,917 and its effect on the inhibitory ability of L. plantarum 14,917 against the growth of Streptococcus mutans and Candida albicans in an in vitro model. Single-species growth screenings were conducted in TSBYE broth with 1% glucose and 1-5% GOS. Interaction experiments were performed using duo- and multi-species models with inoculation of 10 5 CFU/mL S. mutans , 10 3 CFU/mL C. albicans , and 10 8 CFU/mL L. plantarum 14,917 under 1%, 5% GOS or 1% glucose. Viable cells and pH changes were measured. Real-time PCR was utilized to assess expression of C. albicans and S. mutans virulence genes. Six replicates were used for each group. Student's t -test, one-way ANOVA, and Kruskal-Wallis were employed to compare the outcomes of different groups. GOS significantly inhibited the growth of C. albicans and S. mutans in terms of growth quantity and speed when the two strains were grown individually. However, GOS did not affect the growth of L. plantarum 14,917. Moreover, 1% and 5% GOS enhanced the anti-fungal performance of L. plantarum 14,917 in comparison to 1% glucose. GOS as the carbon source resulted in a less acidic environment in the C. albicans and S. mutans duo-species model and multispecies model where L. plantarum 14,917 was added. When GOS was utilized as the carbohydrate substrate, S. mutans and C. albicans had a significant reduction in the expression of the HWP1 , ECE1 , atpD , and eno genes ( p < 0.05). To our knowledge, this is the first study that reported the ability of GOS to neutralize S. mutans-C. albicans high caries of medium pH and to disrupt virulence gene expression. Moreover, as a prebiotic, GOS augmented the inhibitory ability of L. plantarum against C. albicans in vitro. The current study revealed the anti-caries potential of prebiotics GOS and shed light on novel caries prevention strategies from the perspective of prebiotics and probiotics. These findings provide a rationale for future biofilm or clinical studies to elucidate the effect of GOS on modulating oral microbiota and caries control.

Green synthesis offers an environmentally friendly and cost-effective alternative for the synthesis of copper oxide nanoparticles (CuO NPs). In this study, the synthesis of CuO NPs was optimized by using copper sulfate (CuSO 4 ) and the aqueous extract of Alpinia officinarum and its antifungal activity were investigated. The synthesized CuO NPs were characterized by UV-visible spectroscopy (UV-vis), X-ray diffraction (XRD), Fourier-transform infrared radiation spectroscopy (FT-IR), scanning electron microscope (SEM), energy dispersive spectroscopy (EDS), dynamic light scattering (DLS), and transmission electron microscopy (TEM). The results showed that the optimized conditions for the synthesis of CuO NPs were 1:2 ratio of extract and CuSO 4 solution, pH 7, and 30 °C. The characteristic UV-vis peak of A. officinarum synthesized CuO NPs was at 264 nm. The synthesized CuO NPs had high crystallinity and purity and were spherical in morphology with the mean size of 46.40 nm. The synthesized CuO NPs reduced the fungal growth of Colletotrichum gloeosporioides in a dose-dependent manner. The minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) of the CuO NPs were 125 μg·mL -1 and 500 μg·mL -1 , respectively. The antifungal activity of CuO NPs may be attributed to its ability to deform the structure of fungal hyphae, induce excessive reactive oxygen species accumulation and lipid peroxidation in fungi, disrupt the mycelium cell membrane, and result cellular leakage., (© 2022. The effectiveness of nitric oxide (NO) for control of grey spot rot cause by Pestalotiopsis eriobotryfolia in harvested loquat fruit and its probable mechanisms have been investigated. The results showed that NO donor sodium nitroprusside (SNP) did not evidently inhibit mycelial growth and spore germination of P. eriobotryfolia , but resulted in a low disease incidence and small lesion diameter. SNP resulted in a higher hydrogen peroxide (H 2 O 2 ) level in the early stage after inoculation and a lower H 2 O 2 level in the latter period by regulating the activities of superoxide dismutase, ascorbate peroxidase and catalase. At the same time, SNP enhanced the activities of chitinase, β-1,3-glucanase, phenylalanine ammonialyase, polyphenoloxidase, and total phenolic content in loquat fruit. However, SNP treatment inhibited the activities of cell wall-modifying enzymes and the modification of cell wall components. Our results suggested that NO treatment might have potential in reducing grey spot rot of postharvest loquat fruit.

Background: Exercise is a known trigger of the inhibitory pain modulation system and its analgesic effect is termed exercise-induced hypoalgesia (EIH). Previous studies have demonstrated that rats with deficient analgesic response following exercise develop more significant hypersensitivity following nerve injury compared to rats with substantial analgesic response following exercise., Objectives: A rat model of EIH as an indicator of the pain inhibitory system's efficiency was used to explore the association between EIH profiles and the effect of pharmacotherapy on rat's neuropathic pain., Methods: EIH profiles were assessed by evaluating paw responses to mechanical stimuli before and after exercise on a rotating rod. Rats with a reduction of ≤33% in responses were classified as low EIH and those with ≥67% as high EIH. Low and high EIH rats underwent sciatic nerve chronic constriction injury (CCI). Paw responses to mechanical stimuli were measured at baseline, following CCI, and after treatment with diclofenac, duloxetine or pregabalin. In a different group of low and high EIH rats, EIH was measured before and following treatment with the same medications., Results: Low EIH rats developed more significant hypersensitivity following CCI. Duloxetine and pregabalin successfully reduced hypersensitivity, although significantly more so in low EIH rats. Diclofenac had limited effects, and only on low EIH rats. Four days of duloxetine administration transformed low EIH rats' profiles to high EIH., Conclusions: The findings of this study suggest that EIH profiles in rats can not only predict the development of hypersensitivity following injury but may also support targeted pharmacological treatment., Significance: Exercise is a known trigger of the inhibitory pain modulation. Rats with deficient analgesic response following exercise develop more significant hypersensitivity following nerve injury. Pain modulation profiles in rats can also support targeted pharmacological treatment; rats with deficient analgesic response following exercise benefit more from treatment with duloxetine and gabapentin. Chronic cheilitis (CC) is a spectrum of inflammatory changes of unknown etiology that affect the vermilion of the lips. This study aimed to describe the epidemiology, clinical presentations and risk factors of CC. Patients with CC were recruited from the National Clinical Research Center for Oral Disease of China. A convenience sample of inhabitants who live in the same geographical region were recruited as the control group. The lip skin transepidermal water loss (TEWL) and capacitance of CC patients were compared with that of age- and gender-matched controls. Our results demonstrated that of the 109 patients with CC, 72 (66.1%; 95% CI: 57.0-75.1%) were female. The common clinical presentations of CC consisted of desquamation (n = 99; 90.8%), and/or chapping (n = 81; 74.3%), and/or pruritus (n = 64; 58.7%). Multivariable analysis showed that allergic dermatologic diseases (P < 0.001; OR: 4.5; 95% CI: 2.4-8.4), anemia (P = 0.001; OR: 3.3; 95% CI: 1.5-7.5), and indoor/outdoor alternate working environment (P < 0.001; OR: 2.1; 95% CI: 1.5-2.8) were the significant risk factors for CC. The mean lip skin TEWL was found to be significantly higher, while the capacitance was lower in CC patients compared to that of control individuals. Cadmium (Cd) and lead (Pb) pollution is a major environmental issue affecting plant production. Spermidine (Spd) is involved in plant response to abiotic stress. However, the role and associated mechanism of Spd under Cd + Pb combined stress are poorly understood. The potential protective role of Spd at different concentration on rice (Oryza sativa L.) seedlings exposed to Cd + Pb treatment was investigated by a hydroponic experiment in this study. The results showed that exogenous Spd enhanced the tolerance of rice seedlings to Cd + Pb stress, resulted in an increase in plant height, root length, fresh weight and dry weight of roots and shoots. Further, application of Spd decreased the contents of hydrogen peroxide, superoxide anion, malondialdehyde, and the accumulation of Cd and Pb, and increased the contents of mineral nutrient, carotenoids, chlorophyll, proline, soluble sugar, soluble protein, total phenol, flavonoid, anthocyanin, and antioxidant enzymes activities in roots and shoots of rice seedlings under Cd + Pb stress. Particularly, 0.5 mmol L -1 Spd was the most effective to alleviate the adverse impacts on growth and physiological metabolism of rice seedlings under Cd + Pb stress. Principal component analysis and heat map clustering established correlations between physio-biochemical parameters and further revealed Spd alleviated Cd + Pb damage in rice seedling was associated with inhibition of accumulation and translocation of Cd and Pb, increasing the contents of photosynthetic pigments and mineral nutrient and stimulation of antioxidative response and osmotic adjustment. Overall, our findings provide an important prospect for use of Spd in modulating Cd + Pb tolerance in rice plants. Drought is a major abiotic stress factor affecting plant growth and production, while utilizing beneficial endophytic fungi is one of the most promising strategies for enhancing plant growth and drought tolerance. In the current study, a pot experiment was conducted to investigate the beneficial effects of dark septate endophyte (DSE) (Macrophomina pseudophaseolina, Paraphoma radicina) and Trichoderma (Trichoderma afroharzianum, Trichoderma longibrachiatum) inoculum on Astragalus mongholicus grown in sterile soil under drought stress, alone, or in combination. The addition of Trichoderma enhanced the DSE colonization in roots regardless of the water condition. Under well-watered conditions, M. pseudophaseolina inoculation significantly enhanced the biomass and root length of A. mongholicus. The two DSE and Trichoderma inoculum significantly improved calycosin-7-O-β-D-glucoside content. However, M. pseudophaseolina + T. afroharzianum inoculation better promoted root growth, whereas co-inoculation had higher active ingredient contents compared with single inoculation, except for P. radicina + T. afroharzianum. Under drought stress, DSE and Trichoderma inoculum significantly improved root biomass, root length, calycosin-7-O-β-D-glucoside content, and activities of nitrate reductase and soil urease. P. radicina + T. afroharzianum and P. radicina + T. longibrachiatum better increased root length, and all combinations of DSE and Trichoderma had a greater impact on the increase in formononetin content compared with the single treatments. Additionally, Trichoderma relies on antioxidant enzymes, growth hormones, and the redox system (ascorbic acid−glutathione) to resist drought, while DSE strains have an additional osmotic regulation system in addition to the drought resistance function possessed by Trichoderma, and the effect of co-inoculation (especially M. pseudophaseolina + T. longibrachiatum and P. radicina + T. afroharzianum) on plant physiological parameters was greater than that of single inoculation. This study provides a new research direction for the effects of DSE and Trichoderma on medicinal plant cultivated in dryland.

Lead (Pb) poses an adverse effect on plant growth and development. Glycinebetaine (GB) plays an important role in plants response to stress environment. The study was performed to examine the potential of exogenous GB (0.5, 1, 2, and 5 mM) in alleviating Pb toxicity, the physiological and biochemical responses in pakchoi under 100 μM Pb stress by hydroponic experiment. Pb stress significantly decreased the growth, contents of pigment and mineral nutrient, and activities of antioxidative enzymes (CAT, SOD, and APX) in roots and shoots of pakchoi, while it caused a significant increase in Pb and ROS accumulation both in roots and shoots of pakchoi in comparison to the control. Exogenous application of GB improved leaf and root length, fresh and dry weight, mineral nutrient, and pigment contents of pakchoi under Pb stress. GB also effectively enhanced antioxidative enzyme activities and the accumulation of proline, soluble sugar, and GB and reduced the oxidative stress and Pb contents in shoots and roots of pakchoi. Principle component analysis (PCA) provided useful information on the classification of Pb tolerance according to the response to GB. Overall, the 1 mM GB was more effective to ameliorate the detrimental impacts of Pb stress. These findings suggested that GB application might be considered an effective strategy for alleviating Pb toxicity and enhancing the tolerance of pakchoi plants under Pb stress., (© 2021. To meet critical requirements on flexible electronic devices, multifunctionalized flexible sensors with excellent electromechanical performance and temperature perception are required. Herein, lignin-reinforced thermoresponsive poly(ionic liquid) hydrogel is prepared through an ultrasound-assisted synthesized method. Benefitting from the electrostatic interaction between lignin and ionic liquid, the hydrogel displays high stretchability (over 1425%), excellent toughness (over 132 kPa), and impressive stress loading-unloading cyclic stability. The hydrogel strain sensor presents excellent electromechanical performance with a high gauge factor (1.37) and rapid response rate (198 ms), which lays the foundation for human body movement detection and smart input. Moreover, owing to the thermal-sensitive feature of poly(ionic liquid), the as-prepared hydrogel displays remarkable thermal response sensitivity (0.217°C -1 ) in body temperature range and low limit of detection, which can be applied as a body shell temperature indicator. Particularly, the hydrogel can detect dual stimuli of strain and temperature and identify each signal individually, showing the specific application in human-machine interaction and artificial intelligence. In the present study, discarded pruned tea branch was used to prepare a new biochar, and the physicochemical properties and adsorption characteristics were investigated by characterization and batch experiments. With increasing pyrolysis temperature from 400 to 800 °C, the yield, specific surface area, and acidic functional groups had significant differences. The optimum adsorption conditions were determined as pH = 6 and dosage of 2 g L -1 . The pseudo-second-order kinetic and Langmuir isothermal model could fit well to the adsorption data, which showed that the adsorption process was dominated by monolayer chemical adsorption. The highest adsorption property (74.04 mg g -1 ) was obtained by the pyrolysis of tea branch biochar (TBB) at 700 °C owing to the adsorption mechanisms, including surface complexation, precipitation, metal ion exchange, and Cd 2+ -π interaction. After five cycles of desorption, biochar still showed superior adsorption (80%). Background: Pain may undergo modulation in the central nervous system prior to reaching the primary somatosensory cortex and being perceived as pain. Faulty pain modulation mechanisms have been linked to various chronic pain conditions. Cytokines such as IL-10 and IL-1beta, are known to be involved in initiation and maintenance of neuropathic pain. In this study, we investigated the association between pain modulation profile, pain intensity and cytokines (IL-10 and IL-1beta) levels in a rat model of neuropathic pain., Methods: Exercise-Induced Hypoalgesia (EIH) was assessed by evaluating the percentage of responses to a train of 60g mechanical stimuli before and after 180 seconds of exercise on a rotating rod. The differences in the response rates before and after the exercise were used to divide the rats into low and high EIH responders. Rats from low and high EIH groups underwent constriction injury of the left sciatic nerve. Pain behavior (allodynia and hyperalgesia) were assessed by measuring responses to mechanical and thermal stimuli applied to the plantar surface of the foot. Serum, sciatic nerve and the related Dorsal Root Ganglia (DRG) levels of IL-10 and IL-1beta were determined by ELISA. The DRG mRNA levels of IL-10 and IL-1beta measured with PCR. A comparison between the low and high EIH rats of all measured parameters was made., Results: The low EIH rats developed significantly more severe allodynia and hyperalgesia in the affected paw and allodynia in the contralateral paw compared to the high EIH rats, 7 days following the injury. The low EIH rats had higher IL-1beta protein levels in serum prior to and following injury, higher affected and contralateral sciatic nerve IL-1beta levels following injury and higher IL-1beta levels in the contralateral DRG (protein and mRNA) following injury when compared to high EIH rats. The high EIH rats had higher affected sciatic nerve IL-10 levels following nerve injury and higher IL-10 levels of both protein and mRNA in the affected and contralateral DRG at baseline and following injury., Conclusion: EIH profile was found to be predictive of pain behavior following nerve injury, low EIH rats developed more severe allodynia and hyperalgesia. Objectives: To describe the characteristics of major aphthous ulcers (MjOU) in children and analyze its potential risk factors., Materials and Methods: Data were collected from the National Clinical Research Center for Oral Diseases of China between 2012 and 2017. Children younger than 15 years old, who had a giant mucosa ulcer (≥ 1 cm in diameter) and met the diagnostic criteria for MjOU were included in this study. Differences were compared between two subgroups of patients based on the location of the ulcerous lesions. A measurement of ratio (TBR) between the length of the mandibular second molar tooth germ and the height of the mandible was performed in children with MjOU lesions located in the mandibular retromolar pad region (MjOU-P) and their age- and sex-matched controls., Results: A total of 1067 children were diagnosed with oral ulcers during the study period, of which 125 (11.7%, 95% CI: 9.8%-13.7%) met the diagnostic criteria for MjOU. More than half (n = 64, 51.2%) of the MjOU cases were MjOU-P, which had a male predilection (n = 52, 81.3%) with a significant majority at 7 to 9 years of age (n = 43, 67.2%). In comparison to the MjOU located in other regions, MjOU-P lasted longer in duration and had more comorbidities. Logistic regression analysis showed that MjOU-P was statistically significantly associated with TBR controlling age and gender., Conclusions: MjOU-P is a predominant form of MjOU in children and is a distinct subgroup of major ulcers that is likely associated with the development of the mandibular second molars., Clinical Relevance: This study is the first to describe the demographic and clinical features of MjOU in children, which may facilitate the identification and treatment of these patients.

Aims: To investigate the role of exercise-induced hypoalgesia (EIH) in the development of neuropathic pain (NP) following infraorbital nerve (ION) injury and to explore possible underlying mechanisms defining the differences between rats with high and low EIH., Methods: EIH was evaluated by measuring the percentage of withdrawal responses to a series of 30 mechanical stimuli applied to the hind paw before and after 180 seconds of exercise on a rotating rod. The rats were assigned to low- and high-EIH groups based on reduction in the percent of withdrawal responses following exercise. NP was induced in high- and low-EIH rats via ION constriction injury. Rats were tested with graded nylon monofilaments to establish the withdrawal threshold. Increasingly stiff monofilaments were applied to the ION territory until there was a clear withdrawal by the rat. This was repeated a total of three times. A decreased withdrawal threshold indicates allodynia. Testing was performed at baseline and at 3, 10, and 17 days following the injury. On day 17 postinjury, IONs were harvested for the assessment of interleukin (IL)-6, IL-1β, and IL-10 levels. Samples from high-EIH and low-EIH surgically naïve rats served as control for the cytokines study. In this second part of the study, the effects of cannabinoid 1 (CB1) and cannabinoid 2 (CB2) antagonists and naltrexone on EIH profiles and on the withdrawal thresholds to mechanical stimulation were measured. EIH and withdrawal thresholds in high- and low-EIH rats were measured before and after administration of antagonists., Results: Low-EIH rats developed significantly more pronounced allodynia in the ION territory following injury compared to high-EIH rats. At 17 days postinjury, ION IL-1β levels were higher in low-EIH rats, and IL-10 levels were higher in high-EIH rats. CB1 antagonist blocked the analgesic effect induced by exercise in high- but not in low-EIH rats. The CB2 antagonist had no significant effect on high- or low-EIH rats. Naltrexone blocked the effects of EIH in both high- and low-EIH rats. Exercise induced a significant analgesic effect in high-EIH but not in low-EIH rats. CB1 or CB2 antagonist administration had no effect on pre-exercise responses to mechanical stimulation, while naltrexone administration resulted in significant allodynia in both low- and high-EIH rats., Conclusion: This study demonstrated substantial differences between rats with high and low EIH. The results suggest that following ION injury, high-EIH rats may have a more prominent or activated endocannabinoids system and that their inflammatory response is moderated, with higher levels of IL-10 and lower levels of IL-1β.

Purpose: To compare the effects of whitening toothpaste and bleaching with 6% hydrogen peroxide (H2O2) on discoloration of dental resin composite caused by cigarette smoke (CS) and electronic vapor product (EVP) aerosol., Methods: 40 resin composite discs were divided into three groups: 15 each for CS and EVP aerosol exposure and 10 for air exposure (control). Exposures were performed for 15 days, with daily brushing with regular toothpaste. Two whitening sessions, including 21 days of brushing with whitening toothpaste and 3 days of treatments with take-home bleaching (6% H2O2), were performed after the exposure. Color and gloss were assessed before exposure, at every 5 days of exposure, and after each whitening session., Results: After 15 days of exposure, marked discoloration of resin composite was observed in the CS group (ΔE = 23.66 ± 2.31), minimal color change in the EVP group ((ΔE = 2.77 ± 0.75), and no color change in the control group. Resin composites exposed to CS did not recover their original color after treatment with whitening toothpaste ((ΔE = 20.17 ± 2.68) or take-home bleaching ((ΔE = 19.32 ± 2.53), but those exposed to EVP aerosol reverted to baseline after treatment with whitening toothpaste ((ΔE = 0.98 ± 0.37), and no further change in color was observed following take-home bleaching. The gloss of resin composites exposed to CS, EVP aerosol, and air decreased equally with exposure time. Brushing with whitening toothpaste recovered the gloss similarly in all groups, but no further change was observed following take-home bleaching., Clinical Significance: Aerosol from electronic vapor products induced minimal discoloration of resin composites that can be completely reverted by brushing with whitening toothpaste alone. Bleaching with 6% H2O2 did not revert discoloration caused by cigarette smoke. The anthracnose rot of postharvest mango fruit is a devastating fungal disease often resulting in tremendous quality deterioration and postharvest losses. Nitric oxide (NO), as an important signaling molecule, is involved in the responses to postharvest fruit diseases. In the present study, the effectiveness of NO donor sodium nitroprusside (SNP) to prevent anthracnose of "Tainong" mango fruit caused by Colletotrichum gloeosporioides was evaluated through in vivo and in vitro tests. Results from in vivo test showed that SNP treatment effectively inhibited the lesion diameter and disease incidence on inoculated mango fruit during storage. SNP treatment could regulate hydrogen peroxide levels by reinforcing the activities of catalase, peroxidase, superoxide dismutase, and ascorbate peroxidase. Furthermore, SNP elevated the accumulation of lignin, total phenolics, anthocyanin, and flavonoids and the activities of chitinase and β-1,3-glucanase. In addition, in vitro tests indicated that SNP markedly suppressed mycelial growth and spore germination of C. gloeosporioides through damaging plasma membrane integrity and increasing the leakage of soluble sugar and protein. Our results suggested that SNP could suppress anthracnose decay in postharvest mango fruit, possibly by directly suppressing pathogen growth and indirectly triggering host defense responses.

Objectives: Heightened anxiety among dental healthcare professionals (DHPs) during the COVID-19 pandemic stems from uncertainties about the effectiveness of personal protective equipment (PPE) against dental aerosols and risk levels of asymptomatic patients. Our objective was to assess the risks for DHPs providing dental care during the pandemic based on available scientific evidence., Methods: We reviewed the best available evidence and estimated the annualized risk (p=d a s(1-1-p 0 p 1 (1-e) y n ) for a DHP during the COVID-19 pandemic based on the following basic parameters: p 0 , the prevalence of asymptomatic patients in the local population; p 1 , the probability that a DHP gets infected by an asymptomatic patient; e, the effectiveness of the PPE; s, the probability of becoming symptomatic after getting infected from asymptomatic patient; d a , the probability of dying from the disease in age group a; n, number of patients seen per day; and y, number of days worked per year., Results: With the assumption that DHPs work fulltime and wear a N95 mask, the annualized probability for a DHP to acquire COVID-19 infection in a dental office, become symptomatic, and die from the infection is estimated at 1:13,000 (0.008 %) in a medium sized city in the US at the peak of the pandemic. The risk estimate is highly age-dependent. Risk to DHPs under the age of 70 is negligible when prevalence of asymptomatic cases is low in the local community., Conclusions: Risk of COVID-19 transmission in dental office is very low based on available evidence on effectiveness of PPE and prevalence of asymptomatic patients. Biochars were prepared by pyrolyzing mango peel waste at 300, 400, 500, 600 and 700 °C. Various characterizations were carried out to explore the effect of pyrolysis temperature on the biochars. The data indicated that the physical and chemical properties of biochar such as pH, element ratio, specific surface area and functional groups changed with the increase of pyrolysis temperature. The yield and contents of hydrogen, nitrogen and oxygen decreased, while contents of the ash and carbon, pH and specific surface area of the biochars increased. In addition, the molar ratios of H/C, O/C and (O + N)/C decreased. In this study, batch adsorption experiments for Cd(ii) adsorption were performed with initial Cd(ii) concentrations of 10-300 mg L -1 , contact times of 0-2880 min, various pH (2-8) and biochar dose (1-20 g L -1 ). Langmuir isotherm and pseudo-second-order kinetics models were better fits than other models, suggesting the dominant adsorption of mango peel biochars is via monolayer adsorption. Biochar derived at 500 °C was found to have the highest adsorption capacity of 13.28 mg g -1 among all biochars and the adsorption efficiency was still 67.7% of the initial adsorption capacity after desorption for 4 times. Based on adsorption kinetics and isotherm analysis in combination with EDS, FTIR and XRD analysis, it was concluded that cation exchange, complexation with surface functional groups and precipitation with minerals were the dominant mechanisms responsible for Cd adsorption by mango peel biochar. Tobacco smoking contributes to tooth discoloration. Pigmented compounds in the smoke generated by combustion of tobacco can cause discoloration of dental hard tissues. However, aerosols from heated tobacco products cause less discoloration than cigarette smoke (CS) in vitro. The objective of the present study was to optimize a method for extracting the colored chemical compounds deposited on tooth enamel following exposure to total particulate matter (TPM) from CS or a heated tobacco product (Tobacco Heating System [THS] 2.2), analyze the extracts by gas chromatography coupled to time-of-flight mass spectrometry, and identify the key chemicals associated with tooth discoloration. Sixty bovine enamel blocks were exposed for 2 weeks to TPM from CS or THS 2.2 aerosol or to artificial saliva as a control. Brushing without toothpaste and color measurements were performed each week. Noticeable discoloration of enamel was observed following exposure to CS TPM. The discoloration following exposure to THS 2.2 aerosol TPM or artificial saliva was not distinguishable to the eye (ΔE < 3.3). Carbon disulfide was used to extract surface-deposited chemicals. Untargeted analyses were followed by partial least squares correlation against discoloration scores (R 2  = 0.96). Eleven compounds had variable importance in projection scores greater than 2. Discriminant autocorrelation matrix calculation of their mass spectral information identified eight of the eleven compounds as terpenoids. None of the compounds were related to nicotine. Several of these compounds were also detected in THS 2.2 aerosol TPM-exposed enamel, but at lower levels, in line with our findings showing less discoloration. Wide application of nanoparticles causes considerable environmental, health, and safety problems. However, their potential impact and mechanisms on plant growth are not completely clear. In the present study, the effects of different concentration of copper oxide nanoparticles (nCuO) on seed germination and seedling growth, as well as physiological parameters of Brassica pekinensis L., were investigated. The seeds were exposed to 10-, 100-, and 1000-mg L -1 nCuO suspensions and 0.8-mg L -1 Cu 2+ released from 1000-mg L -1 nCuO for 7 day. The results showed that nCuO did not affect the germination rate, germination potential, and germination index of B. pekinensis but significantly affected the vitality index. The growth of roots and shoots of B. pekinensis was promoted at 10-mg L -1 nCuO, while they were inhibited under 1000-mg L -1 nCuO and Cu 2+ ion treatments, and roots suffered more damage than shoots. Cu content in shoots and roots of B. pekinensis increased with increasing concentrations of nCuO, which is significantly higher in roots as compared with shoots. Roots and shoots accumulated more Cu under nCuO treatments compared with Cu 2+ ion treatment. nCuO treatments led to significant lignification in roots of B. pekinensis. Furthermore, nCuO increased in the contents of soluble sugar and protein in shoots, while nCuO at 1000 mg L -1 significantly inhibited the content of soluble protein in roots. In addition, concentration-dependent augmentation of lipid peroxidation, hydrogen peroxide and superoxide generation, and antioxidant enzyme activity were noticed in shoots and roots of B. pekinensis seedlings under nCuO and Cu 2+ ion treatments. Altogether, the results strongly suggested that the phytotoxicity of nCuO in B. pekinensis was caused by both the nanoparticles itself and the released Cu 2+ ions.

Electronic skin is driving the next generation of cutting-edge wearable electronic products due to its good wearability and high accuracy of information acquisition. However, it remains a challenge to fulfill the requirements on detecting full-range human activities with existing flexible strain sensors. Herein, highly stretchable, sensitive, and multifunctional flexible strain sensors based on MXene- (Ti 3 C 2 T x -) composited poly(vinyl alcohol)/polyvinyl pyrrolidone double-network hydrogels were prepared. The uniformly distributed hydrophilic MXene nanosheets formed a three-dimensional conductive network throughout the hydrogel, endowing the flexible sensor with high sensitivity. The strong interaction between the double-network hydrogel matrix and MXene greatly improved the mechanical properties of the hydrogels. The resulting nanocomposited hydrogels featured great tensile performance (2400%), toughness, and resilience. Particularly, the as-prepared flexible pressure sensor revealed ultrahigh sensitivity (10.75 kPa -1 ) with a wide response range (0-61.5 kPa), fast response (33.5 ms), and low limit of detection (0.87 Pa). The increasing utilization of copper oxide nanoparticles (CuO NPs) and their release into the environment has made it imperative to elucidate their impact on the ecological system including plants. However, their potential toxic impact and mechanisms on plant growth are still unclear. The aim of this study was to investigate the effects of CuO NPs and released Cu ions on seed germination and early seedling growth, as well as physiological and biochemical parameters of Oryza sativa. The results showed that CuO NPs at high concentration significantly inhibited seed germination and early seedling growth. The toxicity of CuO NPs originated from the particulate NPs rather than the released Cu 2+ . The phytotoxicity of CuO NPs to rice seed germination and seedling growth probably induced by high Cu accumulation along with the lignification and oxidative damage. The work presented here will increase our knowledge of phytotoxicity of CuO NPs.

Exploring specific electrode active materials with excellent kinetic properties is important for the development of high performance supercapacitors. Herein, a novel nickel-cobalt fluoride (Ni1-xCoxF2) with a porous nanoprism structure is synthesized via step-wise recrystallization and ion-exchange reactions with a morphology control agent, namely polyvinyl pyrrolidone (PVP). The synergistic effect between the bimetallic redox centers promotes the reconstruction of the electronic coordination, leading to apparent discrepancies in the microstructure and morphology of Ni1-xCoxF2 with different stoichiometric ratios of Ni/Co. The micro-porous structure also provides sufficient interfaces and active sites for efficient electrolyte penetration and ion diffusion, thus improving its electrochemical performance. Among the as-synthesized samples, Ni0.5Co0.5F2, with an Ni/Co ratio of 1 : 1, achieved the highest specific capacity of 1979.6 F g-1 at 1.0 A g-1 and a remarkable long-term cycling stability of 900 F g-1 residual after 30 000 cycles at 20 A g-1. The supercapacitor with Ni0.5Co0.5F2 and activated carbon as the positive and negative electrodes, respectively, delivers a high specific capacitance of 107.3 F g-1 at 1 A g-1, outstanding cycling stability of 90.07% capacity retention after 30 000 cycles, and a maximum energy density of 48.3 W h kg-1 at a power density of 952.9 W kg-1. A flexible asymmetric all-solid-state supercapacitor based on a PVA/KOH gel electrolyte was assembled, which delivered a specific capacitance of 41.0 F g-1 at 1 A g-1 and showed promising applications in flexible electronic devices.

Purpose: To study the mechanism of vascular endothelial growth factor (VEGF) regulated by miR-375 and Sonic hedgehog (SHH) signaling pathways on the characteristics of trophoblast cells (TCs) in preeclampsia (PE) rats., Methods: Female (100) Wistar rats in rut were mated with males (50) at a ratio of 2:1, and 60 pregnant rats were assigned to a normal group (Nor, n=10) and a model group (Mod, n=50) in a random manner. Rat TCs of placental villus were isolated and were divided into 8 groups. The related indicators in each group were detected, respectively., Results: In contrast to the NC group, miR-375 expression greatly elevated in the miR-375 mimic group (mimic group); in the mimic and cyclopamine groups, VEGF, proliferating cell nuclear antigen（PCNA, N- cadherin and Bcl-2 expression, cell proliferation, S phase cells, and migration and invasion ability were significantly decreased and E-cadherin and Bax expression, G1 phase cells and apoptosis rate were increased significantly, but in Over expressing-VEGF（ov-VEGF） group, the above indicators presented contrary results (all P< 0.05). In contrast to the mimic group, the expression of SHH, VEGF, PCNA, N- cadherin and Bcl-2, cell proliferation, S phase cells and the invasion and migration activity in the miR-375 mimic + oe-VEGF group were evidently increased., Conclusions: Over-expressing miR-375 can inhibit the expression of VEGF, thus slow down the invasion and proliferation of TCS in PE rats, which is realized by regulating SHH signal pathway., (Copyright 2020 The Author(s).)

While primary care providers in New York State (NYS) are mandated to offer all patients a HIV test, still many NYS residents miss the HIV screening opportunity. To fill the gap, and as the CDC recommends, this study aimed to examine the feasibility of implementing HIV screening in dental setting, identify patient characteristics associated with acceptance of HIV rapid testing, and discuss best practices of HIV screening in dental setting. New York State Department of Health (NYSDOH) collaborated with the Northeast/Caribbean AIDS Education and Training Center (NECA AETC) and three dental schools in New York State to offer free HIV screening tests as a component of routine dental care between February 2016 and March 2018. Ten clinics in upstate New York and Long Island participated in the study. HIV screening was performed using the OraQuick™ In-Home HIV Test. 14,887 dental patients were offered HIV screening tests; 9,057 (60.8%) were screened; and one patient (0.011%) was confirmed HIV positive and linked to medical care. Of all dental patients, 33% had never been screened for HIV; and 56% had not had a primary care visit or had not been offered an HIV screening test by primary care providers in the previous 12 months. Multi-level generalized linear modeling analysis indicated that test acceptance was significantly associated with patient's age, race/ethnicity, gender, country of origin, primary payer (or insurance), past primary care visits, past HIV testing experiences, and the poverty level of patient's community. HIV screening is well accepted by dental patients and can be effectively integrated into routine dental care. Recently, self-healing hydrogel bioelectronic devices have raised enormous interest for their tissue-like mechanical compliance, desirable biocompatibility, and tunable adhesiveness on bioartificial organs. However, the practical applications of these hydrogel-based sensors are generally limited by their poor fulfillment of stretchability and sensitivity, brittleness under subzero temperature, and single sensory function. Inspired by the fiber-reinforced microstructures and mechano-transduction systems of human muscles, a self-healing (90.8%), long-lasting thermal tolerant and dual-sensory hydrogel-based sensor is proposed, with high gauge factor (18.28) within broad strain range (268.9%), low limit of detection (5% strain), satisfactory thermosensation (-0.016 °C -1 ), and highly discernible temperature resolution (2.7 °C). Especially by introducing a glycerol/water binary solvent system, desirable subzero-temperature self-healing performance, high water-retaining, and durable adhesion feature can be achieved, resulting from the ice crystallization inhibition and highly dynamic bonding. On account of the advantageous mechanoreception and thermosensitive capacities, a flexible touch keyboard for signature identification and a "fever indicator" for human forehead's temperature detection can be realized by this hydrogel bioelectronic device.

The germination and seedling vigor of crops is negatively affected by soil salinity. Nitric oxide (NO) has emerged as a key molecule involved in many physiological events in plants. The objective of present study was to evaluate the impact of exogenous sodium nitroprusside (SNP, a NO donor) at different concentrations on the seed germination and early seedling growth characteristics of pakchoi (Brassica chinensis L.) under NaCl stress. 100 mM NaCl stress markedly inhibited the seed germination potential, germination index, vitality index and growth of radicles and plumules. SNP pretreatment attenuated the salt stress effects in a dose-dependent manner, as indicated by enhancing the characteristics of seed germination and early seedling growth parameters, and the mitigating effect was most pronounced at 10 μM SNP. Efficient antioxidant systems were activated by SNP pre-treatment, and which effectively increased the activities of superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX), and reduced contents of malondialdehyde (MDA) and hydrogen peroxide (H 2 O 2 ) and the production rate of superoxide anion radical (O 2 ·- ) in radicles and plumules, thereby preventing oxidative damage from NaCl stress. SNP pre-treatment also increased the contents of proline and soluble sugar in radicles and plumules under NaCl stress. In addition, SNP pre-treatment significantly increased the K + contents and decreased Na + contents in radicles and plumules, resulting in the increased level of K + /Na + ratio. Our results demonstrated that SNP application on pakchoi seeds may be a good option to improve seed germination and seedling growth under NaCl stress by modulating the physiological responses resulting in better seed germination and seedling growth., (Copyright © 2019 Elsevier Inc. Objectives: To compare the relative effects of cigarette smoke (CS), electronic cigarette (EC), red wine, coffee, and soy sauce on the color of enamel, dentin, and composite resin restorations, as well as the effects of whitening treatments., Methods: Seventy premolars with composite restorations were exposed to CS, EC aerosol (a novel EC device with MESH™ technology [P4M3 version 1.0, Philip Morris International]), red wine, coffee, and soy sauce for 56 min/day for 15 days. Two whitening sessions with 6% and 35% hydrogen peroxide (H 2 O 2 ) were performed on the exposed samples. Teeth exposed to CS and EC aerosol were also brushed with whitening toothpaste for 3 weeks. Color match of resin restorations was assessed, and color changes were compared after exposure and after whitening treatments., Results: Discolorations in enamel, dentin, and composite resin were observed in the order of red wine > CS > soy sauce > coffee > EC. Color mismatch between enamel and resin restorations occurred only in red wine and CS groups. Brushing with whitening toothpaste removed discoloration caused by EC aerosol; H 2 O 2 treatments were necessary to eliminate discolorations caused by coffee and soy sauce. Discolorations of dentin and resin restorations could not be completely removed by whitening treatments, and color mismatch remained in teeth exposed to red wine and CS., Conclusion: Red wine and CS cause significant tooth discoloration and color mismatch in enamel and resin restorations that are not reversible by whitening treatments. Tooth discoloration associated with EC aerosol was minimal and could be removed by brushing with whitening toothpaste., Clinical Significance: Red wine drinkers and cigarette smokers have increased risks for tooth discoloration and color mismatch between enamel and composite resin restorations. Objectives: To test if cigarette smoke (CS) causes discoloration of enamel, dentin, and composite resin restorations and induces color mismatch between dental hard tissues and the restorations, and to compare the findings with the effects of aerosol generated by the tobacco heating system (THS) 2.2., Method and Materials: Twenty-two human premolars were prepared with Class V cavities restored with Filtek Supreme Ultra (3M Espe) composite resin. Teeth were divided into two groups and exposed to either CS from 20 reference cigarettes (3R4F) or aerosol from 20 THS 2.2 tobacco heat sticks 4 days a week for 3 weeks. CIE L*a*b* color was assessed before and after exposure and brushing at 1, 2, and 3 weeks. Color match, marginal discoloration, marginal integrity, and surface texture of the Class V restoration were assessed according to a modified US Public Health Service (USPHS) criterion., Results: Marked discoloration of enamel and dentin was observed following 3 weeks of CS exposure (ΔE = 8.8 ± 2.6 and 21.3 ± 4.4, respectively), and color mismatch occurred between the composite resin restorations (ΔE = 25.6 ± 3.8) and dental hard tissues. Discoloration was minimal in the enamel, dentin, and composite resin restorations in the THS 2.2 group, and no color mismatch was observed after 3 weeks of THS 2.2 aerosol exposure., Conclusion: CS causes significant tooth discoloration and induces color mismatch between dental hard tissues and composite resin restorations. Reducing or eliminating the deposits derived from tobacco combustion could minimize the impact of tobacco products on tooth discoloration.

Despite the advancement of early childhood caries (ECC) prediction and treatment, ECC remains a significant public health burden in need of more effective preventive strategies. Pregnancy is an ideal period to promote ECC prevention given the profound influence of maternal oral health and behaviors on children's oral health. However, studies have shown debatable results with respect to the effectiveness of ECC prevention by means of prenatal intervention. Therefore, this study systematically reviewed the scientific evidence relating to the association between prenatal oral health care, ECC incidence, and Streptococcus mutans carriage in children. Five studies (3 randomized control trials, 1 prospective cohort study, and 1 nested case-control study) were included for qualitative assessment. Tested prenatal oral health care included providing fluoride supplements, oral examinations/cleanings, oral health education, dental treatment referrals, and xylitol gum chewing. Four studies that assessed ECC incidence reduction were included in meta-analysis using an unconditional generalized linear mixed effects model with random study effects and age as a covariate. The estimated odds ratio and 95% confidence intervals suggested a protective effect of prenatal oral health care against ECC onset before 4 years of age: 0.12 (0.02, 0.77) at 1 year of age, 0.18 (0.05, 0.63) at 2 years of age, 0.25 (0.09, 0.64) at 3 years of age, and 0.35 (0.12, 1.00) at 4 years of age. Children's S. mutans carriage was also significantly reduced in the intervention group. O-Acetylated pectins are abundant in the primary cell wall of plants and growing evidence suggests they have important roles in plant cell growth and interaction with the environment. Despite their importance, genes required for O-acetylation of pectins are still largely unknown. In this study, we showed that TRICHOME BIREFRINGENCE LIKE 10 (AT3G06080) is involved in O-acetylation of pectins in Arabidopsis (Arabidopsis thaliana). The activity of the TBL10 promoter was strong in tissues where pectins are highly abundant (e.g. leaves). Two homozygous knock-out mutants of Arabidopsis, tbl10-1 and tbl10-2, were isolated and shown to exhibit reduced levels of wall-bound acetyl esters, equivalent of ~50% of the wild-type level in pectin-enriched fractions derived from leaves. Further fractionation revealed that the degree of acetylation of the pectin rhamnogalacturonan-I (RG-I) was reduced in the tbl10 mutant compared to the wild type, whereas the pectin homogalacturonan (HG) was unaffected. The degrees of acetylation in hemicelluloses (i.e. xyloglucan, xylan and mannan) were indistinguishable between the tbl10 mutants and the wild type. The mutant plants contained normal trichomes in leaves and exhibited a similar level of susceptibility to the phytopathogenic microorganisms Pseudomonas syringae pv. tomato DC3000 and Botrytis cinerea; while they displayed enhanced tolerance to drought. These results indicate that TBL10 is required for O-acetylation of RG-I, possibly as an acetyltransferase, and suggest that O-acetylated RG-I plays a role in abiotic stress responses in Arabidopsis., (© 2018 The Authors The Plant Journal © 2018 John Wiley & Sons Ltd.)

Ovarian cancer is the most lethal malignancy of the women genital tract. Exploring novel factors involved in the development of ovarian cancer and characterizing the molecular mechanisms by which regulate the tumorigenesis of ovarian cancer are quite necessary. Here, we found that miR-19a was highly expressed in ovarian cancer tissues and cell lines. Overexpression of miR-19a promoted the viability of ovarian cancer cells, while down-regulation of miR-19a inhibited the growth of ovarian cancer cells. To further understand the underlying molecular mechanism of miR-19a in regulating ovarian cancer cell growth, the downstream targets of miR-19a were predicted. The bioinformatics analysis showed that the tumor suppressor PTEN was found as one of the targeting candidates of miR-19a. MiR-19a bound the 3'-UTR of PTEN and highly expressed miR-19a decreased both the mRNA and protein levels of PTEN in ovarian cancer cells. Overexpression of PTEN suppressed the promoting effect of miR-19a on regulating the growth of ovarian cancer cells. Notably, the expression of miR-19a and PTEN was inversely correlated in ovarian cancer tissues. These results demonstrated the potential oncogenic role of miR-19a in ovarian cancer, which suggested that miR-19a might be a promising target in the diagnosis and treatment of ovarian cancer., (Copyright © 2018 Elsevier B.V. Purpose: To study the effects of cigarette smoke (CS) on the discoloration of dental resin composite compared with the aerosol from a heat-not-burn tobacco product, the Tobacco Heating System 2.2 (THS2.2)., Methods: A total of 60 discs were prepared from three commercial resin composites: Durafill VS (DVS), Filtek Supreme Ultra (FSU) and Tetric EvoCeram BulkFill (TEC). Twenty discs of each composite were divided into two groups and exposed to CS from 20 reference cigarettes (3R4F) or aerosol from 20 THS2.2 tobacco sticks per day for 3 weeks. Color, gloss and surface roughness of the composite discs were measured at baseline and after exposure and brushing with toothpaste at 1, 2 and 3 weeks., Results: Color differences from the baseline (ΔE) were on average 27.1 (±3.6) in 3R4F and 3.9 (±1.5) in the THS2.2 group after 3 weeks of exposure (P< 0.0001). TEC (30.4±1.4 and FSU (28.0 ±2.5) exhibited more discoloration than DVS (23.0±1.2) in the 3R4F group (P< 0.0001). FSU (2.6 ±0.5) showed significantly less discoloration than TEC (5.3±1.5) in the THS2.2 group (P< 0.0001). Surface roughness of resin composites was not affected by either CS or THS2.2 aerosol, while surface gloss increased in the composite discs with more severe discoloration., Clinical Significance: Cigarette smoke caused significant discoloration of dental composite resins. Background: The human microbiota is postulated to affect cancer risk, but collecting microbiota specimens with prospective follow-up for diseases will take time. Buccal cell samples have been obtained from mouthwash for the study of human genomic DNA in many cohort studies. Here, we evaluate the feasibility of using buccal cell samples to examine associations of human microbiota and disease risk., Methods: We obtained buccal cells from mouthwash in 41 healthy participants using a protocol that is widely employed to obtain buccal cells for the study of human DNA. We compared oral microbiota from buccal cells with that from eight other oral sample types collected by following the protocols of the Human Microbiome Project. Microbiota profiles were determined by sequencing 16S rRNA gene V3-V4 region., Results: Compared with each of the eight other oral samples, the buccal cell samples had significantly more observed species (P < 0.002) and higher alpha diversity (Shannon index, P < 0.02). The microbial communities were more similar (smaller beta diversity) among buccal cells samples than in the other samples (P < 0.001 for 12 of 16 weighted and unweighted UniFrac distance comparisons). Buccal cell microbial profiles closely resembled saliva but were distinct from dental plaque and tongue dorsum., Conclusions: Stored buccal cell samples in prospective cohort studies are a promising resource to study associations of oral microbiota with disease., Impact: The feasibility of using existing buccal cell collections in large prospective cohorts allows investigations of the role of oral microbiota in chronic disease etiology in large population studies possible today. Background: The goal of the study was to investigate whether cigarette smoking alters oral and nasal microbial diversity, composition, and structure. Twenty-three current smokers and 20 never smokers were recruited. From each subject, nine samples including supra and subgingiva plaque scrapes, saliva, swabs from five soft oral tissue sites, and one nasal swab from both the anterior nares were collected. 16S rRNA V3-V4 region was sequenced for microbial profiles., Results: We found that alpha diversity was lower in smokers than in nonsmokers in the buccal mucosa, but in other sample sites, microbial diversity and composition were not significantly different by smoking status. Microbial profiles differed significantly among eight oral sites., Conclusions: This study investigates the effect of cigarette smoking on different sites of the oral cavity and shows a potential effect of cigarette smoking on the buccal mucosa microbiota. The marked heterogeneity of the oral microbial ecosystem that we found may contribute to the stability of the oral microbiota in most sites when facing environmental perturbations such as that caused by cigarette smoking.

Introduction: Candida albicans has been detected together with Streptococcus mutans in high numbers in plaque-biofilm from children with early childhood caries (ECC). The goal of this study was to examine the C. albicans carriage in children with severe early childhood caries (S-ECC) and the maternal relatedness., Methods: Subjects in this pilot cross-sectional study were recruited based on a convenient sample. DMFT(S)/dmft(s) caries and plaque scores were assessed during a comprehensive oral exam. Social-demographic and related background information was collected through a questionnaire. Saliva and plaque sample from all children and mother subjects were collected. C. albicans were isolated by BBL™ CHROMagar™ and also identified using germ tube test. S. mutans was isolated using Mitis Salivarius with Bacitracin selective medium and identified by colony morphology. Genetic relatedness was examined using restriction endonuclease analysis of the C. albicans genome using BssHII (REAG-B). Multilocus sequence typing was used to examine the clustering information of isolated C. albicans. Spot assay was performed to examine the C. albicans Caspofungin susceptibility between S-ECC children and their mothers. All statistical analyses (power analysis for sample size, Spearman's correlation coefficient and multiple regression analyses) were implemented with SAS 9.4., Results: A total of 18 S-ECC child-mother pairs and 17 caries free child-mother pairs were enrolled in the study. Results indicated high C. albicans carriage rate in the oral cavity (saliva and plaque) of both S-ECC children and their mothers (>80%). Spearman's correlation coefficient also indicated a significant correlation between salivary and plaque C. albicans and S. mutans carriage (p<0.01) and caries severity (p<0.05). The levels of C. albicans in the prepared saliva and plaque sample (1ml resuspension) of S-ECC children were 1.3 ± 4.5 x104 cfu/ml and 1.2 ± 3.5 x104 cfu/ml (~3-log higher vs. caries-free children). Among 18 child-mother pairs, >60% of them demonstrated identical C. albicans REAG-B pattern. C. albicans isolated from >65% of child-mother pairs demonstrated similar susceptibility to caspofungin in spot assay, while no caspofungin resistant strains were seen when compared with C. albicans wild-type strain SC5314. Interestingly, the regression analysis showed that factors such as antibiotic usage, birth weight, inhaler use, brushing frequency, and daycare attendance had no significant effect on the oral carriage of C. albicans in the S-ECC children., Conclusions: Our results reveal that both the child with S-ECC and the mother were highly infected with C. albicans, while most of the strains were genetically related, suggesting that the mother might be a source for C. albicans acquisition in the oral cavity of children affected by the disease., Competing Interests: The authors have declared that no competing interests exist.

ATP induces pain via activation of purinergic receptors in nociceptive sensory nerves. ATP signaling is terminated by ATP hydrolysis mediated by cell surface-localized ecto-nucleotidases. Using enzymatic histochemical staining, we show that ecto-ATPase activity is present in mouse trigeminal nerves. Using immunofluorescence staining, we found that ecto-NTPDase3 is expressed in trigeminal nociceptive neurons and their projections to the brainstem. In addition, ecto-ATPase activity and ecto-NTPDase3 are also detected in the nociceptive outermost layer of the trigeminal subnucleus caudalis. Furthermore, we demonstrate that incubation with anti-NTPDase3 serum reduces extracellular ATP degradation in the nociceptive lamina of both the trigeminal subnucleus caudalis and the spinal cord dorsal horn. These results are consistent with neuronal NTPDase3 activity modulating pain signal transduction and transmission by affecting extracellular ATP hydrolysis within the trigeminal nociceptive pathway. Introduction: Adenosine 5'-triphosphate (ATP) is a potent signaling molecule that regulates diverse biological activities in cells. Its effects on human dental pulp cells (HDPCs) remain unknown. This study aimed to examine the effects of ATP on proliferation and differentiation of HDPCs., Methods: Reverse transcription polymerase chain reaction was performed to explore the mRNA expression of P2 receptor subtypes. Cell Counting Kit-8 test and flow cytometry analysis were used to examine the effects of ATP on proliferation and cell cycle of HDPCs. The effects of ATP on differentiation of HDPCs were examined by using alizarin red S staining, energy-dispersive x-ray analysis, Western blot analysis, and real-time polymerase chain reaction., Results: The purinoceptors P2X3, P2X4, P2X5, P2X7, and all P2Y receptor subtypes were confirmed to present in HDPCs. ATP enhanced HDPC proliferation at 10 μmol/L concentration. However, it inhibited cell proliferation by arresting the cell cycle in G0G1 phase (P < .05 versus control) and induced odontoblastic differentiation, ERK/MAPK activation, and dentin matrix protein 1 (DMP1) and dentin sialophosphoprotein (DSPP) mRNA transcriptions at 800 μmol/L concentration. Suramin, an ATP receptor antagonist, inhibited ERK/MAPK activation and HDPC odontoblastic differentiation (P < .05 versus control)., Conclusions: Extracellular ATP activates P2 receptors and downstream signaling events that induce HDPC odontogenic differentiation. Thus, ATP may promote dental pulp tissue healing and repair through P2 signaling. Objective: Bone grafts (sinus lift and/or ridge augmentation) may become an obstacle for some patients who desire implant treatment. The objective of this study was to evaluate the success of six- and eight-millimeters rough surface design short dental implants, for up to 2 years in function, when compared to conventional length (11 mm) implants., Materials and Methods: A total of 25.6-, 20.8- and 35.11-mm length implants were placed and restored in 30 subjects (11 males, 19 females) between the age of 22 and 80, following a standard protocol. Implant mobility, crestal bone loss as well as periodontal parameters were evaluated immediately after restoration placement, at 6, 12 and 24 months., Results: There was one failure of one 6-mm implant during the healing phase and one restorative failure. The median crestal bone loss at 24 months was 0.45 mm for the 6-mm implants, 0.55 mm for the 8 mm implants and 0.65 mm for the 11-mm implants. The success rate for 6-mm implants was 97% and for 8-mm and 11-mm implants 100%., Conclusions: Based on this preliminary data, we conclude that rough surface design short dental implants (6 and 8 mm in length) have similar success rate when compared to 11-mm implants. Corrosion of dental alloys is a major concern in dental restorations. Streptococcus mutans reduces the pH in oral cavity and induces demineralization of the enamel as well as corrosion of restorative dental materials. The rough surfaces of dental alloys induced by corrosion enhance the subsequent accumulation of plaque. In this study, the corrosion process of nickel-chromium (Ni-Cr) and cobalt-chromium (Co-Cr) alloys in a nutrient-rich medium containing S. mutans was studied using inductively coupled plasma atomic emission spectrometry (ICP-AES), X-ray photoelectron spectroscopy (XPS) and electrochemical corrosion test. Our results showed that the release of Ni and Co ions increased, particularly after incubation for 3 days. The electrochemical corrosion results showed a significant decrease in the corrosion resistance (Rp) value after the alloys were immersed in the media containing S. mutans for 3 days. Correspondingly, XPS revealed a reduction in the relative dominance of Ni, Co, and Cr in the surface oxides after the alloys were immersed in the S. mutans culture. After removal of the biofilm, the pre-corroded alloys were re-incubated in S. mutans medium, and the expressions of genes associated with the adhesion and acidogenesis of S. mutans, including gtfBCD, gbpB, fif and ldh, were evaluated by detecting the mRNA levels using real-time reverse transcription polymerase chain reaction (RT-PCR). We found that the gtfBCD, gbpB, ftf and Idh expression of S. mutans were noticeably increased after incubation with pre-corroded alloys for 24 h. This study demonstrated that S. mutans enhanced the corrosion behavior of the dental alloys, on the other hand, the presence of corroded alloy surfaces up-regulated the virulent gene expression in S. mutans. Compared with smooth surfaces, the rough corroded surfaces of dental alloys accelerated the bacteria-adhesion and corrosion process by changing the virulence gene expression of S. mutans.
