Magnesium (Mg) and zinc (Zn) are essential nutrients for plants. Mg deficiency often occurs in rice plants grown in Zn-polluted soil. However, the mechanism for this correlation is unclear. Here, we performed culture experiments on rice plants (Oryza sativa L.) and used Mg isotopes to investigate mechanisms of Zn stress on plant Mg deficiency. Our results show that excess Zn can significantly reduce the uptake of Mg in rice tissues. The root displays positive Δ 26 Mg plant-nutrient values (δ 26 Mg plant -δ 26 Mg nutrient ; 1.90 ‰ to 2.06 ‰), which suggests that Mg enters the root cells mainly via Mg-specific transporters rather than non-selective diffusion. The decreased Δ 26 Mg plant-nutrient values with increasing Zn supply can be explained by the competition between Zn and Mg, both of which combine with same transporters in roots. In contrast, the shoots (stem and leaf) display much lower δ 26 Mg values than roots, which suggests that the transport of Mg from roots to aerial biomass is mainly via free Mg ions, during which Zn cannot competitively inhibit the movement of Mg. A sequential chromatographic separation procedure for subsequent high-precision isotopic analysis of Mg and Ca via multi-collector ICP-mass spectrometry (MC-ICP-MS) from a single aliquot of sample was developed and used for a variety of animal/human biofluids and tissues. The procedure consists of a one-stage Mg isolation protocol (for most of the sample types) and a three-stage isolation protocol for Ca. AG50W-X8 strong cation exchange resin was used for the isolation of Mg and Ca, while Sr-resin was used to additionally purify the Ca fraction from Sr. Potential effects on the Mg isotope ratio measurement results caused by the possible presence of concomitant matrix elements (Cu, Fe, Zn, Ca) were systematically evaluated. δ 26 Mg values were biased for a Fe/Mg ratio > 0.13 and a Ca/Mg ratio > 1.5, resulting in a shift towards a lighter Mg isotopic composition. It was shown that the Mg isotope ratio data for Mg standards, the isotopic reference materials ERM-AE143 and IRMM 009 and the biological samples investigated are located on a mass-dependent fractionation line. Biological reference materials and commercially available serum samples were analyzed for both their Mg and Ca isotope ratios. For some of the biomaterials analyzed, the Ca isotope ratio data as obtained using MC-ICP-MS were further validated via their determination using double-spike thermal ionization mass spectrometry (DS-TIMS). The expanded uncertainty for δ 26 Mg was ≤ 0.12‰ and for δ 44 /42 Ca ≤ 0.29‰. Biological fluids and tissues of mice were analyzed to characterize the body distribution of the stable isotopes of Mg and Ca. The isotopic variability among the body compartments was about 1.5‰ for Mg and 1.0‰ for Ca. We undertook 87 Sr/ 86 Sr analyses for a range of carbonate bearing geological reference materials, and combined these with δ 26 Mg for a subset of samples. Following chemical purification in a series of chromatographic extractions, isotope ratios were measured by Multi-Collector-ICP-MS using a Plasma II (Nu instruments, Wrexham, UK). To validate efficient sample digestion procedures of carbonate fractions, total samples were treated with either 3 mol l -1 HNO 3 and 0.5 mol l -1 HCl, respectively. Results of both leaching procedures are identical within reproducibility. Reference values for SRM 88A (formerly NBS 88A), SRM 1B (formerly NBS 1B), SARM 40, SARM 43, JDo-1, JLs-1, and San Carlos olivine range from 0.70292 to 0.73724 in 87 Sr/ 86 Sr and from -2.80 to -0.41 ‰ for δ 26 Mg, respectively. This set of geological reference materials can be used for sedimentary rock material with different carbonate mineral and matrix composition as quality control measurements of combined stable Mg and radiogenic Sr isotope analyses.•We present a protocol that facilitates the chemical separation of Mg and Sr in carbonate bearing geological reference materials including 87 Sr/ 86 Sr and δ 26 Mg of certified reference materials., (© 2020 The Author(s).)

To better understand the mechanism of Mg isotopic variation in magma systems, here we report high precision Mg isotopic data of 17 bulk rock samples including dunite, clinopyroxenite, hornblendite and gabbro and 10 pairs of dunite-hosted olivine and chromite separates from the well-characterized Alaskan-type Xiadong intrusion in NW China, which formed by continuous and high degree of lithological differentiation from mafic magmas. Chromite separates have highly variable δ 26 Mg values from -0.10‰ to 0.40‰, and are consistently heavier than coexisting olivine separates (-0.39‰ to -0.15‰). Both mineral δ 26 Mg values and the degrees of inter-mineral fractionation are well correlated with geochemical indicators of magma differentiation, indicating that these inter-sample and inter-mineral Mg isotope fractionations are caused by magma evolution. The δ 26 Mg values range from -0.20‰ to -0.02‰ in the dunite, -0.43‰ in the clinopyroxenite, -0.43‰ to -0.28‰ in the hornblendite, 0.18‰ in the chromite-bearing hornblendite, and -0.56‰ to -0.16‰ in the gabbro. The Mg isotopic variations in different types of rocks are closely related to fractional crystallization and accumulation of different proportions of oxides vs. silicates. Chromite crystallization and accumulation is the most important factor in controlling Mg isotope fractionation during the formation of the Xiadong intrusion. Compared to basaltic and granitic magmas, differentiation of the Alaskan-type intrusions occurs at a relatively high oxygen fugacity, which favors chromite crystallization and consequently significant Mg isotope fractionations at both mineral and whole-rock scales.
