Autoimmune diseases represent a complex array of conditions where the body's immune system mistakenly attacks its own tissues. These disorders, affecting millions worldwide, encompass a broad spectrum of conditions ranging from rheumatoid arthritis and multiple sclerosis to lupus and type 1 diabetes. The Aryl hydrocarbon receptor (AhR) translocator, expressed across immune and other cell types, plays crucial roles in immune disorders and inflammatory diseases. With a realm towards natural remedies in modern medicine for disease prevention, this study investigates the electronic properties and behaviors of bioactive compounds from dietary sources, including Apium graveolens L. (Celery), Coriandrum sativum seeds (Coriander), and Mentha longifolia, as AhR modulators. Through comprehensive analysis (HOMO-LUMO, ESP, LOL, and ELF), electron-rich and -poor regions, electron localization, and delocalization are identified, contrasting these compounds with the toxic AhR ligand, TCDD. Evaluation of Absorption, Distribution, Metabolism, Excretion, and Toxicity (ADMET) properties reveals favorable pharmacokinetics without blood-brain barrier penetration, indicating drug-like characteristics. Molecular docking demonstrates stronger interactions of dietary flavonoid ligands with AhR transcription compared to TCDD. Molecular dynamics simulations confirm the stability of complexes and the sustainability of interactions formed. In this study, three different ecotypes of M. longifolia collected from Lorestan province, Iran. three habitats including Khorramabad (ecotype 1), Aleshtar (ecotype 2), and Delfan (ecotype 3) counties (ecotype 3) were selected. In addition to analysing the chemical composition of the essential oils, for the first time, the effects of essential oils of these ecotypes against plant pathogenic bacterial including Erwinia amylovora and Pseudomonas syringae together with Bacillus subtilis , a biocontrol agent, and viability of colon tumour cell (SW742) was evaluated. Results showed that ecotype 1 had the highest concentration of the essential oil. All three bacteria showed the same level of sensitivity to the essential oils of M. longifolia extracted from three different ecotypes. Considering the findings of the effects of essential oils on SW742 tumour cell viability, we can suggest that M. longifolia essential oil obtained from ecotype 3 can be used to reduce the cell viability of SW742 tumour cells. It can be concluded that M. longifolia essential oil can be used to reduce tumour cell viability and their anti-microbial activity against plant pathogens.

The volatile profile of Mentha longifolia was observed using GC-MS, GC-FID, FT-IR and 13 CNMR. Twenty-two constituents were found to be present in this essential oil, accounting 96.04 (%) of total essential oil and oxegenated monoterpenes were major class of compounds. The key constituents of this essential oil were pieritenone oxide (45.9%), piperitone (17.5%), beta caryophyllene (10.2%), and Germacrene D (5.0%). FT-IR showed peak at 1669 and 1707 cm -1 which may be due to the presence of carbonyl groups. Among the tested compounds, Germacrene D showed highest binding affinity value of -6.8 kcal mol -1 and a pKi value of 6.01. The dsc studies revealed that boiling point of this EO is above 200 °C. Microplate Alamar Blue assay (MABA) was carried out for the assessment of antimycobacterial activity using isoniazid and nicotinic as reference compound and oil was found to be active within conc. range of 0.8-1.6 μg/mL against mycobacterium tuberculosis, hence can act as a potential candidate against antituberculosis.

In this study, the efficacy of the essential oil of Mentha longifolia, Achillea arabica and Artemisia absinthium plants were evaluated against important soil-borne fungal pathogens as Verticillium dahliae, Rhizoctonia solani, and Fusarium oxysporum. Essential oils were obtained from plants by hydrodistillation method and the chemical components of essential oils were determined by analyzing by gas chromatography-mass spectrometry. The main components found as piperitone oxide (13.61%), piperitenone oxide (15.55%), pulegone (12.47%), 1-menthone (5.75%), and camphor (5.75%) in M. longifolia, á-selinene 13.38%, camphor 13.34%, L-4-terpineneol 8.40%, (-)-á-Elemene 7.01%, 1,8-cineole 4.71%, and (-)-spathulenol 3.84% in A. arabica, and á-thujone (34.64%), 1,8-cineole (19.54%), pulegone (7.86%), camphene (5.31%), sabinene (4.86%), and germacrene-d (3.67%) in A. absinthium. The antifungal activities of the oils were investigated 0.05, 0.1, 0.25, 0.5, 1.00, and 2.00 μl/ml concentrations with the contact effect method. M. longifolia oil (1.00 and 2.00 μl/ml) has displayed remarkable antifungal effect and provided 100% inhibition on mycelial growth of V. dahliae, R. solani and F. oxysporum. The results obtained from this study may contribute to the development of new alternative and safe methods against soil-borne fungal pathogens.

The present study aimed to explore the phytochemical profile, and evaluate the antioxidant, antimicrobial, and insecticidal properties, of Moroccan Mentha longifolia L. essential oil (ML-EO) using in vitro and in silico assays. Noteworthily, as chromatography (GC-MS/MS) revealed that ML-EO is majorly composed of piperitenone oxide (53.43%), caryophyllene (20.02%), and (-) germacrene D (16.53%). It possesses excellent antioxidant activity with an IC 50 of 1.49 ± 0.00 for DPPH and 0.051 ± 0.06 μg/mL for ABTS. Moreover, the RP and TAC activities were 0.80 ± 0.01 μg/mL and 315.532 ± 0.00 mg EAA/g, respectively. ML-EO exhibited a potent antimicrobial effect, specifically against Pseudomonas aeruginosa . It also exhibited strong antifungal ability, especially against Candida albicans . Regarding insecticidal activity, for ML-EO, a dose of 20 µL/mL produced a complete reduction in fecundity, fertility, and emergence of adult C. maculatus with mortality rates reaching 100%. In silico results showed that the antioxidant activity is mostly attributed to α-Cadinol, the antibacterial efficiency is attributed to piperitenone oxide, and antifungal capacity is related to cis-Muurola-4(15),5-diene and piperitenone oxide. Accordingly, ML-EO has high potential to be used as an alternative for preserving food and stored grain and protecting them against microbes and insect pests in the food and pharmaceutical sectors.

Antibiotic-resistant bacterial strains and the consequent surge in infections caused by them have become major public health concerns. Silver nanoparticles (AgNPs) exhibit antibacterial properties and have wide applications in biomedical sciences. In this study, AgNPs were synthesized in the presence of antibiotics: Ceftazidime (Cft), Cefotaxime (Cef), Ceftriaxone (Cfx), and Cefepime (Cpm), along with the extract of Mentha longifolia. Mentha longifolia-based AgNPs were kept as the control for all experiments. The associated metabolites, structural properties, surface charges, and antibacterial activity of the AgNPs were also evaluated. Overall, a blue-shift of SPR peaks was observed for control AgNPs (λmax = 421 nm, 422 nm, 426 nm, and 406 nm for Cft-AgNPs, Cef-AgNPs, Cfx-AgNPs, and Cpm-AgNPs, respectively), compared to the control (λmax = 438 nm). Fourier-transform infrared spectroscopy showed that antibiotic-based AgNPs had distinct peaks that corresponded to the respective antibiotics, which were not observed in the control. XRD analysis showed that there were observed changes in crystallinity in antibiotic-based AgNPs compared to the control. TEM images revealed that all samples had spherical nanoparticles with different sizes and distributions compared to the control. The Zeta potential for extract-based AgNPs was - 33.6 mV, compared to -19.6 mV for Cft-AgNPs, -2 mV for Cef-AgNPs, -21.1 mV for Cfx-AgNPs, and - 24.2 mV for Cpm-AgNPs. The increase in the PDI value for antibiotic-based AgNPs also showed a highly polydisperse distribution. However, the antibiotic-AgNPs conjugates showed significantly higher activity against pathogenic bacteria. Objective: The integration of nanomaterials and herbal medicine has led to the design of new nanocomposites, which are therapeutically more effective. The purpose of this study was to prepare different zinc oxide (ZnO)-based nanoparticles (NPs) via Mentha longifolia extract based on gauze linen fibre and study its effects on wound healing., Methods: The textural properties, morphology, thermal stability, purity, spectroscopic and phase structure of nanoparticles were investigated. Subsequently, male Wistar rats were subjected to wounds in six different treatment groups: Group I: control; group II: ZnO/W prepared in water (W); group III: ZnO/M synthesised with Mentha longifolia (M) extract; group IV: ZnO/copper(II) oxide (CuO)/M nanocomposite synthesised with M extract; group IV: treated with ZnO/silver (Ag)/M nanocomposite; group V: treated with ZnO/Ag/M nanocomposite; and finally, group VI: treated with ZnO/CuO/Ag/M nanocomposite. In all groups, the wounds were treated for 21 days with prepared samples. Every seven days, after measuring the decreasing rate of the wound size, tissue samples from each group were taken for histopathological analysis. The prepared tissue sections were assessed by haematoxylin and eosin staining for the formation of the epidermis, dermis and muscular tissue, and Masson's Trichrome staining for the formation of collagen fibres., Results: The results showed that the ZnO/CuO/Ag/M nanocomposite was a significantly more effective wound healing material in comparison with other samples (p<0.05)., Conclusion: In this study, the integration of ZnO/CuO/Ag nanocomposites with secondary metabolites of Mentha longifolia gave rise to a superior combination, which could support different phases of wound healing via the regulation of cytokines and growth factors in the course of healing.

Aedes aegypti (Diptera: Culicidae) mosquito is an important vector of many disease-causing pathogens. An effective way to escape from these mosquito-borne diseases is to prevent mosquito bites. In the current study, essential oils of Lepidium pinnatifidum, Mentha longifolia, Origanum vulgare, and Agrimonia eupatoria were evaluated for their repellent potential against Ae. aegypti females. Essential oils were extracted using steam distillation from freshly collected aerial parts of the plants and tested against 4-5 day old females of Ae. aegypti through the human bait technique for repellency and repellent longevity assays. The chemical composition of extracted essential oils was explored by gas chromatography coupled with mass spectrometry (GC-MS). The essential oils of L. pinnatifidum, M. longifolia, O. vulgare, and A. eupatoria at a dose of 33 μg/cm 2 showed 100%, 94%, 87%, and 83% mosquito repellent activity, respectively. Furthermore, M. longifolia and O. vulgare essential oils exhibited 100% repellency at a dose of 165 μg/cm 2 , whereas A. eupatoria essential oil showed 100% repellency only at 330 μg/cm 2 . In the time-span bioassay, M. longifolia and O. vulgare essential oils showed protection against Ae. aegypti bites for 90 and 75 min, respectively whereas both A. eupatoria and L. pinnatifidum were found active for 45 min. Phenylacetonitrile (94%), piperitone oxide (34%), carvacrol (20%) and α-pinene (62%) were the most abundant compounds in L. pinnatifidum, M. longifolia, O. vulgare and A. eupatoria essential oils, respectively. Mentha longifolia L (ML) is locally used as an herbal contraceptive. We aim to assess the toxicity profile and validate the antifertility effects of its leaves in male rats. Adult male Sprague Dawley rats ( n  = 10 animals/group) were divided into four groups. The Control group received .9% saline, while groups II, III, and IV received 50, 75, and 100 mg/kg/day ML doses for 28 days. ML induced a dose-dependent decrease in percent fertility as well as the number of pups born in ML preexposed pairs, with maximum change observed in the ML3 group (60%, 3.75 ± 1.25) as compared to control (100%, 8.75 ± .48) group. A significant ( P  < .01) decrease in the rate of daily sperm production was observed in the ML3 group (.86 ± .16) in contrast to the control (2.93 ± .05). The levels of catalase and superoxide dismutase declined significantly in treatment groups (ML1- P  < .001, ML2- P  < .05, and ML3- P  < .01). A significant increase in the production of reactive oxygen species ( P  < .001) was found in all treatment groups. The concentrations of LH ( P  < .001), FSH ( P  < .001), and testosterone ( P  < .001) were significantly reduced in a dose-dependent manner among ML-treated groups. Decreases in spermatogonial populations, mature spermatids, seminiferous tubule diameter, lumen diameter, and epithelial height were noticed at higher doses. The reversibility study depicted a reduction in both pregnancy outcomes and litter size. Besides contraceptive effects, M. longifolia L usage is associated with oxidative stress; therefore, it is important to consume this herb appropriately and its excessive usage should be prohibited., (© The Author(s) 2022. Objective: Mentha (M.) longifolia (L.) is traditionally used for various ailments. The current study was intended to explore the underlying vasorelaxation mechanisms of M. longifolia., Material and Methods: Aqueous-methanol extract from the aerial parts of M. longifolia was prepared and subjected to activity-guided fractionation. The vasorelaxant activity was performed using porcine coronary arteries with intact and denuded endothelium. In-vitro PDE inhibitory activity of the active fraction was carried out using the radio-enzymatic assay. The active fraction was also subjected to GCMS. Docking and molecular dynamic simulation studies were also performed RESULT: We had observed that aqueous-methanolic extract induced relaxation in the coronary artery in a dose-dependent manner when the endothelium was intact and denuded. n-butanol fraction (MLB) has produced a maximum effect, and it was selected for mechanistic studies. MLB has significantly enhanced the relaxation produced by cAMP and cGMP, elevating atrial natriuretic peptide, sodium nitroprusside, isoproterenol, and forskolin. The pre-treatment with MLB inhibited the contractile response produced by KCl, U46619, and CaCl 2 in without endothelium rings. MLB has non-selectively inhibited the PDE isoforms. GCMS analysis of MLB has revealed the presence of menthol, thymol, and carvacrol in the active fraction. Mentha longifolia (L.) Huds., a wild, diploid mint species, has been developed as a model for mint genetic and genomic research to aid breeding efforts that target Verticillium wilt disease resistance and essential oil monoterpene composition. Here, we present a near-complete, chromosome-scale mint genome assembly for M. longifolia USDA accession CMEN 585. This new assembly is an update of a previously published genome draft, with dramatic improvements. A total of 42,107 protein-coding genes were annotated and placed on 12 chromosomal scaffolds. One hundred fifty-three genes contained conserved sequence domains consistent with nucleotide binding site-leucine-rich-repeat plant disease resistance genes. Homologs of genes implicated in Verticillium wilt resistance in other plant species were also identified. Multiple paralogs of genes putatively involved in p-menthane monoterpenoid biosynthesis were identified and several cases of gene clustering documented. Heterologous expression of candidate genes, purification of recombinant target proteins, and subsequent enzyme assays allowed us to identify the genes underlying the pathway that leads to the most abundant monoterpenoid volatiles. Mentha longifolia is an important medicinal and aromatic perennial herb that exhibits wide distribution range from sub-tropical to temperate regions. In the present study, agro-morphological traits and genetic differences in 19 different populations of M. longifolia were studied to evaluate the level and extent of its diversity. Analysis of variance (ANOVA) showed that the different phenotypic characters show considerable differences among various populations and was significant at p < 0.05. Molecular diversity analysis performed by using arbitrary amplified eleven ISSR primers generated a total of 121 amplicons that range within the size of 200-2500 base pairs (bp). Each primer on average generated 11 amplicons with percentage polymorphism being 100. The analysis of molecular variance (AMOVA) showed more (64%) among population genetic diversity and less (36%) within the populations. Greater genetic differentiation (Gst = 0.6852) among these populations occurs due to low gene flow ( Nm  = 0.2297) and greater habitat variability. Geographic and genetic distances were positively correlated according to Mantel's test. In order to remove any kind of biases, we used R software to perform cluster and redundancy analysis to analyse the extent of relatedness among studied populations. In terms of morphological and molecular aspects, the populations were grouped into four and five clusters respectively based on hierarchical clustering method. Diabetic kidney disease is one of the most common microvascular complications of diabetes mellitus with consequences of diabetic nephropathy. Here we amined to evaluate the nephroprotective potential of methanolic Mentha longifolia (MML) against serotonin-induced hypoglycemia allied toxicity in the rat model of diabetes. Diabetes was induced in rats via alloxan administration and validated by blood glucose level measurement. After that, the animals were treated with serotonin and methanolic extract of Mentha longifolia. Surprisingly, serotonin treatment significantly reduced the glucose levels to hypoglycemic conditions, accompanied by impaired redox defense system, abnormal kidney histopathology, dyslipidemia, and altered level of liver toxicity markers. Interestingly these changes were rescued by the methanolic extract of M. longifolia. The present study suggests that impaired serotonin levels during diabetic conditions may accelerate hypoglycemic allied free radical-dependent abnormalities; however, medicinal plants like M. longifolia can reduce these deleterious effects by scavenging free radicals and their associated toxicity., (© 2021. Terpenoids are a wide variety of natural products and terpene synthase (TPS) plays a key role in the biosynthesis of terpenoids. Mentha plants are rich in essential oils, whose main components are terpenoids, and their biosynthetic pathways have been basically elucidated. However, there is a lack of systematic identification and study of TPS in Mentha plants. In this work, we genome-widely identified and analyzed the TPS gene family in Mentha longifolia , a model plant for functional genomic research in the genus Mentha . A total of 63 TPS genes were identified in the M. longifolia genome sequence assembly, which could be divided into six subfamilies. The TPS-b subfamily had the largest number of genes, which might be related to the abundant monoterpenoids in Mentha plants. The TPS-e subfamily had 18 members and showed a significant species-specific expansion compared with other sequenced Lamiaceae plant species. The 63 TPS genes could be mapped to nine scaffolds of the M. longifolia genome sequence assembly and the distribution of these genes is uneven. Tandem duplicates and fragment duplicates contributed greatly to the increase in the number of TPS genes in M. longifolia . The conserved motifs (RR(X)8W, NSE/DTE, RXR, and DDXXD) were analyzed in M. longifolia TPSs, and significant differentiation was found between different subfamilies. Adaptive evolution analysis showed that M. longifolia TPSs were subjected to purifying selection after the species-specific expansion, and some amino acid residues under positive selection were identified. Furthermore, we also cloned and analyzed the catalytic activity of a single terpene synthase, MlongTPS29 , which belongs to the TPS-b subfamily. MlongTPS29 could encode a limonene synthase and catalyze the biosynthesis of limonene, an important precursor of essential oils from the genus Mentha . This study provides useful information for the biosynthesis of terpenoids in the genus Mentha .

Alzheimer's disease (AD) is an irreversible neurodegenerative disorder manifested by cognitive deterioration where the available treatments failed to delay its progression. The objective of this study was to investigate the neuroprotective activity in an aluminum chloride (AlCl 3 )-induced AD in vivo model and phytochemical profile of the traditional Egyptian herb Mentha longifolia (Ml). Male albino rats were injected with Ml fractions and essential oil for 15 days followed by AlCl 3 for 30 days. Oxidative stress and neuroinflammatory markers were measured namely: malondialdehyde (MDA), reduced glutathione (GSH), nitric oxide (NO), and nuclear factor-κB (NF-κB). Furthermore, cholinesterase activity was tested and analysis of brain neurotransmitters using HPLC was performed. Results showed that methylene chloride and ethyl acetate fractions were able to reverse the AlCl 3 mediated MDA increase, GSH decrease and exhibited anticholinesterase activity. EaFr reversed the increased levels of NF-κB and NO. Ml fractions and oil counteracted the AlCl 3 effect on brain neurotransmitters. Forty metabolites were tentatively characterized in the bioactive fractions using UPLC-PDA-ESI-MS. 5,6,4'-trihydroxy-3',7,8-trimethoxy flavone was isolated from Ml as a first report, in addition to 5,6-dihydroxy-3',4',7,8-tetramethoxy flavone and rosmarinic acid. These findings suggest that Ml is a promising nutraceutical and source of lead compounds halting AD progression. PRACTICAL APPLICATIONS: The results presented in this paper unravels the neuroprotective effect of Mentha longifolia fractions and oil by acting as anti-inflammatory, antioxidant agents, and regulating the levels of neurotransmitters. Background: Fungi growing on wood cause deterioration of stored food materials or discoloration of the wood itself, and the search for new and safe bioagents is recently needed., Methods: Essential oils (EOs) from aerial parts from Mentha longifolia L. and Citrus reticulata L., analyzed by gas chromatography-mass spectrometry (GC-MS), were tested for their antifungal activity by the vapor method against four common fungi, Aspergillus flavus , A. niger , A. fumigatus , and Fusarium culmorum , and confirmed by SEM examination as the oils applied on wood samples., Results: The most abundant compounds identified in the EO from M. longifolia were menthone and eucalyptol; in C. reticulata EO, they were β-caryophyllene, β-caryophyllene oxide, and β-elemene. EOs from M. longifolia and C. reticulata , at 500 and 250 µL/mL, showed potent antifungal activity against A. flavus and A. fumigatus , with 100% fungal mycelial inhibition growth (FMIG). C. reticulata and M. longifolia EOs, at 125 µL/mL, observed FMIG values of 98% and 95%, respectively, against A. fumigatus . M. longifolia EO, at 500 and 250 µL/mL, showed potent activity against A. niger , with 100% FMIG. F. culmorum completely inhibited (100% FMIG) EOs from M. longifolia and C. reticulata applied at 500 µL/mL. Pinus roxburghii Sarg. Wood, treated with M. longifolia at 125 µL/mL, showed inhibition zone values of 7.33 and 21.33 mm against A. flavus and A. niger , respectively., Conclusions: Both oils possessed good wood-biofungicide activity with the vapor method, as clearly shown by the SEM examination. These activities suggest their possible use as natural wood preservatives.

The use of chemical pesticides to preserve food commodities is a global issue of concern due to their negative effect on the environment and public health. In recent years, the European Union is trying to reduce their use, favoring alternative or complementary approaches based on natural products. In this scenario, plant-borne essential oils (EOs) represent valid options for Integrated Pest Management (IPM) programs. In the present study, the insecticidal effect of eight EOs obtained from plants from different parts of the world, namely Mentha longifolia, Dysphania ambrosioides, Carlina acaulis, Trachyspermum ammi, Pimpinella anisum, Origanum syriacum, Cannabis sativa and Hazomalania voyronii, were evaluated against two stored-product insect species of economic importance, Prostephanus truncatus and Trogoderma granarium. Simulating a small-scale stored-product conservation environment, an AG-4 airbrush was used to spray maize and wheat with 500 and 1000 ppm of EOs, then T. granarium and P. truncatus were exposed to the stored products and mortality was evaluated over selected time intervals (4, 8, and 16 h, and 1, 2, 3, 4, 5, 6, and 7 days). The EO of C. acaulis exhibited high efficacy against P. truncatus adults at both tested concentrations by killing > 97% of the individuals exposed to treated maize within 3 days at 500 ppm. The EO of D. ambrosioides eliminated all T. granarium adults exposed to 1000 ppm-treated wheat 2 days post-exposure. At this exposure interval, 91.1% of the exposed T. granarium adults died on wheat treated with 1000 ppm of C. acaulis EO. The EO of M. longifolia at both tested concentrations was the most effective against T. granarium larvae, leading to 97.8% mortality at 500 ppm after 3 days of exposure, and 100% mortality at 1000 pm 2 days post-exposure. At 1000 ppm, the EOs of D. ambrosioides and P. anisum led to 95.6 and 90% mortality, respectively, to larvae exposed to treated wheat for 7 days. Background: Acinetobacter baumannii is an opportunistic pathogen that causes nosocomial infections especially in patients in intensive care units (ICUs). Accordingly, the aim of our study was to detection of adeABC efllux pump encoding genes and antimicrobial effect of the essential oil of Mentha longifolia and Menthol on the minimum inhibitory concentration (MIC) of imipenem and ciprofloxacin in clinical isolates of A. baumannii., Methods: A total of 75 clinical isolates of A. baumannii were collected. The presence of efflux pump genes was detected by polymerase chain reaction (PCR). The minimum inhibitory concentration (MIC) of the essential oil of Mentha longifolia and Menthol and their combined effect with antibiotics were measured by microbroth dilution method and fractional inhibitory concentration (FIC) index., Results: The frequency of adeA, adeB, and adeC genes in clinical isolates of A. baumannii were 86.7, 90.7, and 92%, respectively. When the essential oil of Mentha longifolia was combined with ciprofloxacin and imipenem, MICs decreased 4- and 8-fold, respectively. In the combination of menthol with imipenem, the resistance to imipenem was reduced from 0- to 16-fold in 90% (63/70) of the isolates., Conclusion: The presence of efflux pump genes in more than 90% of A. baumannii isolates indicates its potential role in inducing imipenem- and ciprofloxacin-resistance in this bacterium. Menthol has an antimicrobial effect as an active ingredient in Mentha longifolia. In the future, the combination of medicinal plants with antibiotics can be used as a complement in treating diseases caused by drug-resistant bacteria such as A. baumannii infections.

Mint [ Mentha longifolia (L.) Hudson] is an aromatic plant that belongs to Lamiaceae family. It is traditionally used as herbal tea in Europe, Australia and North Africa and shows numerous pharmacological effects, such as spasmolytic, antioxidant, antimicrobial and anti-hemolytic. Recently, its antiproliferative role has been suggested in a small number of tumor cell models, but no data are available on adrenocortical carcinoma, a malignancy with a survival rate at 5 years of 20%-30% which frequently metastasize. This work aimed to study the effects of Mentha longifolia L. crude extract (ME) on two adrenocortical tumor cell models (H295R and SW13 cells). Chemical composition of ME was assessed by gas-chromatography/mass spectrometry and NMR spectroscopy analysis. Brine shrimp lethality assay showed ME effects at >0.5 µg/µl (p < 0.05). Cell viability and vitality were determined by MTT, SRB, and trypan blue assays in H295R and SW13 cells. The anti-proliferative effects of ME were more evident in SW13 cells at 72 h (ME > 0.5 µg/µl, p < 0.05). Combination of ME with mitotane (approved drug for adrenocortical carcinoma) seemed not to reinforce the efficacy of the herb. As control, human fibroblasts were treated with ME with no effect on cell viability. Clonogenic assay was concordant with previous cell viability tests (ME > 0.5 µg/µl, p < 0.05), while Wright staining demonstrated the presence of both necrotic and apoptotic cells. Cell cycle analysis showed a strong increase in subG0/G1 phase, related to cell death. Furthermore, MAPK and PI3k/Akt pathways were modulated by Western blot analysis when treating cells with ME alone or combined with mitotane. The crude methanolic extract of wild mountain mint can decrease cell viability, vitality and survival of adrenocortical tumor cell models, in particular of SW13 cells. Background: The aim of this study was to evaluate the efficacy of the herbal tea based on Foeniculum vulgare, on inducing regular bleeding in women with oligomenorrhea and secondary amenorrhea( oligo/amenorrhea)., Methods: Forty women aged 18- 40 with oligo/amenorrhea were enrolled in this randomized controlled clinical trial and were allocated to two groups equally. The women in the first group were treated by Fomentex (Foeniculum vulgare / Mentha longifolia / Vitex agnus-castus) herbal tea 11.2 g/day in 2 divided doses for 2 weeks and the second group were treated by medroxy progesterone acetate (MP)10 mg/day for the last 10 days of their menstrual cycles. The intervention was repeated in three cycles of menstruation in both groups. Bleeding pattern was documented by the patient on diary cards. The occurrence (yes/no) of bleeding, the regularity of bleeding pattern, the interval of cycles, the duration of bleeding, the volume of blood flow, the hormonal parameters (total testosterone, free testosterone luteinizing hormone and follicle-stimulating hormone), and the endometrial thickness in sonography before and after the intervention were evaluated and compared as outcomes., Results: The number of women with bleeding during the first cycle was in the Fomentex group and the MP group 83.3% and 94.1% respectively (p = 0.61). The regularity of bleeding did not significantly differ in patients treated with Fomentex from those given MP (66.7% vs. 94.1%; p = 0.088). Mean interval of cycles decreased in both groups after intervention (P<0.001). Mean duration decreased significantly in MP group after the intervention but it was not different in patients treated with Fomentex. The difference between 2 groups was not significant (P=0.705). Volume of blood flow, with regard to Pictorial Blood Assessment Chart (PBAC), increased significantly in MP group after the intervention (P=0.001) and it was not different in patients treated with Fomentex (P=0.757); however, difference between 2 groups was not significant (P=0.063). The percentage of patients with on time menstruation in the next (drug-free) episode, was higher in the Fomentex group compared with the MP group (50% vs. 23.5%; p = 0.105). Secondary outcomes such as dysmenorrhea, acne and hirsutism reduced in the Fomentex group (P≤0.05), while they increased in the MP group (P=0.007). At the end of the treatment, there was a significant decrease in luteinizing hormone, total testosterone and free testosterone in patients taking Fomentex. The decrease of endometrial thickness, was significant in both groups after the intervention (P=0.001), but the difference between 2 groups was not significant (P=0.58). No notable complication or side effect was reported in relation to Fomentex., Conclusion: Fomentex herbal tea is a safe, well-tolerated, and effective choice in inducing bleeding and maintaining regular bleeding in women with oligo/amenorrhea., (Copyright© Bentham Science Publishers; For any queries, please email at epub@benthamscience.net.)

Hypertension is highly prevalent among the Lebanese adult population and is indeed the major cause of mortality in Lebanon. Traditional use of antihypertensive medicinal plants has long been practiced. The aim of this study is to document this traditional knowledge and clinically test the antihypertensive capacity of three of the most commonly used wild plant species Mentha longifolia , Viola odorata and Urtica dioica . Ethno-pharmacological data was collected by personal interviews with herbalists and traditional healers using a semi structured survey questionnaire and assessing relative frequency of citation (RFC). The clinical study was conducted by a randomized, blind, placebo-controlled trial in 29 subjects with mild hypertension distributed in four groups, three plant extract treatments and one placebo. Systolic (SBP) and diastolic blood pressures (DBP) as well as mean arterial blood pressures (MAP) were monitored at weeks 4, 8, 12 and 16 during the treatment with 300 mL/day of plant extract. Results showed that M. longifolia , U. dioica and V. odorata exhibited the highest values of RCF (0.95) followed by Allium ampeloprasum (0.94), Apium graveolens (0.92) and Crataegus azarolus (0.90). The clinical trial revealed dose- and duration-dependent significant reductions in SBP, DBP and MAP of subjects treated with M. longifolia , U. dioica or V. odorata . Our findings indicate that extracts of these plants present an effective, safe and promising potential as a phyto-therapuetical approach for the treatment of mild hypertension. More research on the phytochemistry, pharmacological effects and the underlying mechanisms is necessary.

Objective: This study aimed to determine the antibiofilm activity of Mentha longifolia essential oil (EO) against biofilms forming on in-vitro implant surfaces., Materials and Methods: Enterococcus faecalis, Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Klebsiella pneumoniae, and Candida albicans biofilms were used. Stainless steel and titanium samples were grouped as control, water diluted, no EO addition, and reducing amounts of EO doses. The six microorganisms included in the study were investigated to examine if there were differences between the doses on the implant surfaces. The eradication effect of the EO in samples investigated with electron microscope was classified as 0: none, 1: mild, 2: moderate, and 3: severe. The chemical composition of the EO was determined with gas chromatography., Results: In terms of biofilm formation, no difference was observed between implant surfaces. While S. aureus and C. albicans were observed to be the most susceptible, P. aeruginosa was identified as the most resistant. According to gas chromatography, M. longifolia EO comprised 61.40% carvacrol and 0.28% thymol., Conclusion: In vitro, M. longifolia EO was shown to be effective against gram negative/positive and fungal biofilms forming on the surface of stainless steel and titanium implants., Competing Interests: Conflict of Interest: The authors have no conflict of interest to declare.

Objective: Mentha longifolia L. Hudson has been used in folk medicine for various purposes especially for its anti-inflammatory effects. Lymphocytes play a central role in development of inflammation. In the present study, we investigated the immunomodulatory effects of different extracts of M. longifolia on human peripheral blood lymphocytes (PBLs), as main players in development of inflammation., Materials and Methods: PBLs stimulated with phytohemagglutinin (PHA) were cultured in the presence of the plant extracts. The effects of the extracts on activation of cells were determined by BrdU assay. The viability of cells was examined by flow cytometry using propidium iodide staining. Also, IFN-γ (T helper 1, TH1) and IL-4 (TH2) secretion was measured by ELISA., Results: Except for the water extract which had a weak inhibitory effect, treatment of cells with more than 1μg/ml of butanol, hexane, ethyl acetate and dichloromethane extracts resulted in strong inhibition of cells proliferation (IC 50 4.6-9.9 µg/ml). Flow cytometry analysis showed that these extracts at ≤10μg/ml were non-cytotoxic. Dichloromethane and ethyl acetate extracts at 10 μg/ml decreased IFN-γ production in a dose-dependent manner from 919±91.1 pg/ml in PHA-only-treated cells to 568±22.6 pg/ml (in dichloromethane-treated cells) and 329±12.3 pg/ml (in ethyl acetate-treated cells) (p<0.001). At 10 μg/ml, the ethyl acetate extract increased IL-4 secretion compared to PHA-only-treated cells (p<0.05). The hexane extract decreased IFN-γ level but did not affectIL-4 production., Conclusion: Reduction of IFN-γ and augmentation of IL-4 secretion induced by the extracts suggested the potential of M. longifolia to inhibit TH1 inflammatory responses toward a TH2 dominant response., Competing Interests: The authors declare no conflicts of interest.

Ethnopharmacological Relevance: Mentha longifolia L (Wild Mint or Habak) (ML) is used in traditional medicine in treatment of many gastrointestinal disorders., Aim of the Study: This study aimed to evaluate potential protecting effect of ML and its major constituent, eucalyptol, against acetic acid-induced colitis in rats, a model of human inflammatory bowel disease (IBD)., Materials and Methods: Rats were divided into ten groups (n=8) given orally for three days (mg/kg/day) the following: normal control, acetic acid-induced colitis (un-treated, positive control), vehicle (DMSO), sulfasalazine (500), ML extract (100, 500, 1000), and eucalyptol (100, 200, 400). After 24h-fasting, two ML of acetic acid (3%) was administered intrarectally. On the fifth day, serum and colonic biochemical markers, and histopathological changes were evaluated., Results: Colitis significantly increased colonic myeloperoxidase activity and malonaldehyde level, and serum tumor necrosis factor-α, interleukin-6, and malonaldehyde levels while significantly decreased colonic and serum glutathione levels. All treatments (except ML 100, ML 1000, and eucalyptol 100) significantly reversed these changes where eucalyptol (400) showed the highest activity in a dose-dependent manner. The colitis-induced histopathological changes were mild in sulfasalazine and eucalyptol 400 groups, moderate in ML 500 and eucalyptol 200 groups, and severe in ML 100, ML 1000, and eucalyptol 100 groups nearly similar to colitis-untreated rats., Conclusion: ML (in moderate doses) and eucalyptol (dose-dependently) exerted protective effects against acetic acid-induced colitis in rats possibly through antioxidant and antiinflammatory properties suggesting a potential benefit in treatments of IBD. Mentha longifolia (wild mint) is a popular folk remedy. Some parts of this plant have been used in traditional medicine of Iran and other countries. Many studies have shown various pharmacological and therapeutic effects of the plant. Our aim in preparing this study was to review the traditional uses of M. longifolia together with the pharmacological and therapeutic effects of its entire extract and major compounds. Mentha longifolia is an herb with a wide range of pharmacological properties such as antimicrobial, gastrointestinal, and nervous system effects. Pulegone is the main compound of the plant responsible for most of its pharmacological effects followed by menthone, isomenthone, menthol, 1, 8-cineole, borneol, and piperitenone. Moreover, the plant may dose-dependently exert toxic effects in different systems of the body. Based on the review of various studies, it can be concluded that M. longifolia is a potential natural source for the development of new drugs. However, further studies are required to determine the precise quality and safety of the plant to be used by clinicians.

Lamiaceae is an important plant family that has been investigated for its medicinal properties due to its large amounts of phenolic acids and flavonoids. Flavonoids have been shown to have antioxidant and antimutagenic activities in different test systems, but their certain mechanisms are still unclear. This study was designed to evaluate the mutagenic and antimutagenic activities of apigenin 7-O-rutinoside, a flavonoid isolated from Mentha longifolia (L.) Huds. ssp. longifolia. The possible antimutagenic potential of apigenin 7-O-rutinoside (A7R) was examined against mutagens ethyl methanesulfonate (EMS) and acridine (AC) in a eukaryotic cell system Saccharomyces cerevisiae RS112. The results showed that A7R has different inhibition rates against EMS and AC-induced mutagenicity. Thus, the properties of A7R are of great pharmacological importance and might be beneficial for reducing the risk of reactive oxygen species-related diseases.
