The diagnosis of chronic fatigue syndrome (CFS) is mainly symptom-based, and the etiology is still unclear. Here, we evaluated the pathological changes in the brain of a mouse model of CFS and studied the effects of Kampo medicine. A mouse model of CFS was established through six repeated injections of Brucella abortus (BA) every two weeks for a period of 12 weeks. Neuroinflammation was measured by estimating interleukin (IL)-1β, IL-6, and interferon-gamma (IFN-γ), and oxidative stress by nitrotyrosine (3-NT) and 4-hydroxynonenal (4-HNE) 6 weeks after the last injection. Hippocampal neurogenesis was evaluated through K i -67, doublecortin (DCX), and 5-bromodeoxyuridine (BrdU) assays. The effects of Kampo medicines (Hochuekkito (TJ-41) and Hachimijiogan (TJ-7)) on neuroinflammation during CFS were studied. The wheel-running activity of mice was decreased by about 50% compared to baseline at 6 weeks after the last BA injection. The levels of IL-1β, IL-6, 3-NT, and 4-HNE were increased in both the cortex and the hippocampus of CFS mice at 6 weeks after the last BA injection. Hippocampal neurogenesis was unchanged in CFS mice. Treatment with TJ-41 and TJ-7 reduced the expressions of IL-1β, IL-6, and IFN-γ in the hippocampus but not in the cortex. The results of the present study indicate that neuroinflammation and oxidative stress play important roles in the pathogenesis of CFS. The data further suggest that treatment with TJ-41 and TJ-7 could help reduce the inflammation associated with CFS in the hippocampus, but failed to improve the symptoms in CFS mice.

Nonalcoholic fatty liver disease (NAFLD) is one of the most common chronic liver diseases worldwide, and its treatment remain a constant challenge. A number of clinical trials have shown that acupuncture treatment has beneficial effects for patients with NAFLD, but the molecular mechanisms underlying its action are still largely unknown. In this study, we established a mouse model of NAFLD by administering a methionine- and choline-deficient (MCD) diet and selected three acupoints (ST36, CV4, and KI1) or nonacupoints (sham) for needling. We then investigated the effects of acupuncture treatment on the progression of NAFLD and the underlying mechanisms. After two weeks of acupuncture treatment, the liver in the needling-nonapcupoint group (NG) mice appeared pale and yellowish in color, while that in the needling-acupoint group (AG) showed a bright red color. Histologically, fewer lipid droplets and inflammatory foci were observed in the AG liver than in the NG liver. Furthermore, the expression of proinflammatory signaling factors was significantly downregulated in the AG liver. A lipid analysis showed that the levels of triglyceride (TG) and free fatty acid (FFA) were lower in the AG liver than in the NG liver, with an altered expression of lipid metabolism-related factors as well. Moreover, the numbers of 8-hydroxy-2'-deoxyguanosine (8-OHdG)-positive hepatocytes and levels of hepatic thiobarbituric acid reactive substances (TBARS) were significantly lower in AG mice than in NG mice. In line with these results, a higher expressions of antioxidant factors was found in the AG liver than in the NG liver. Our results indicate that acupuncture repressed the progression of NAFLD by inhibiting inflammatory reactions, reducing oxidative stress, and promoting lipid metabolism of hepatocytes, suggesting that this approach might be an important complementary treatment for NAFLD.

It is very important to analyze and monitor agricultural drought to obtain high temporal-spatial resolution soil moisture products. To overcome the deficiencies of passive microwave soil moisture products with low resolution, we construct a spatial fusion downscaling model (SFDM) using Moderate Resolution Imaging Spectroradiometer (MODIS) data. To eliminate the inconsistencies in soil depth and time among different microwave soil moisture products (Advanced Microwave Scanning Radiometer on the Earth Observing System (AMSR-E) and its successor (AMSR2) and the Soil Moisture Ocean Salinity (SMOS)), a time series reconstruction of the difference decomposition (TSRDD) method is developed to create long-term multisensor soil moisture datasets. Overall, the downscaled soil moisture (SM) products were consistent with the in situ measurements (R > 0.78) and exhibited a low root mean square error (RMSE < 0.10 m 3 /m 3 ), which indicates good accuracy throughout the time series. The downscaled SM data at a 1-km spatial resolution were used to analyze the spatiotemporal patterns and monitor abnormal conditions in the soil water content across North East China (NEC) between 2002 and 2018. The results showed that droughts frequently appeared in western North East China and southwest of the Greater Khingan Range, while drought centers appeared in central North East China. Waterlogging commonly appeared in low-terrain areas, such as the Songnen Plain. Seasonal precipitation and temperature exhibited distinct interdecadal characteristics that were closely related to the occurrence of extreme climatic events. Abnormal SM levels were often accompanied by large meteorological and natural disasters (e.g., the droughts of 2008, 2015, and 2018 and the flooding events of 2003 and 2013). The spatial distribution of drought in this region during the growing season shows that the drought-affected area is larger in the west than in the east and that the semiarid boundary extends eastward and southward.

Porcine reproductive and respiratory syndrome (PRRS) is the most significant infectious disease currently affecting the swine industry worldwide. Several inactivated and modified live vaccines (MLV) have been developed to curb PRRSV infections. However, the efficacy and safety of these vaccines are unsatisfactory, and hence, there is a strong demand for the development of new PRRS universal vaccines. Virus-like particle (VLP)-based vaccines are gaining increasing acceptance compared to subunit vaccines, as they present the antigens in a more veritable conformation and are readily recognized by the immune system. Hepatitis B virus core antigen (HBcAg) has been successfully used as a carrier for more than 100 viral sequences. In this study, hybrid HBcAg VLPs were generated by fusion of the conserved protective epitopes of PRRSV and expressed in E. coli. An optimized purification protocol was developed to obtain hybrid HBcAg VLP protein from the inclusion bodies. This hybrid HBcAg VLP protein self-assembled to 23-nm VLPs that were shown to block virus infection of susceptible cells when tested on MARC 145 cells. Together with the safety of non-infectious and non-replicable VLPs and the low cost of production through E. coli fermentation, this hybrid VLP could be a promising vaccine candidate for PRRS.

The behavior ecology of copulation and oviposition of Apis cerana cerana Fab. was studied. The results showed that temperature influence on the cell-covered time of Apis cerana cerana queen and drone induced the remarkable difference in their newborn weight and mature time (P < 0.05). Queen and drone fly frequency for discerning cell was 1.23-1.31 and 1.08-1.13, and duration was 0.12-0.13 h and 0.16-0.20 h. Their copulation fly frequency was 1.10-1.12 and 1.10-1.05, and duration was 0.22-0.23 h and 0.18-0.23 h. The most suitable temperature for copulation was 20-28 degrees C. The number of semen in side--oviduct of the queen in each copulation fly was 3.37 x 10(6)-4.15 x 10(6), and in natural copulation, semen number in spermathecal sac was 3.55 x 10(6)-3.62 x 10(6). There was a direct relationship between queen newborn weight and ovulation number. The annual effect of climate and flower fertility on the queen's ovulation number was obvious.

Systematical observations and researches were conducted on the population size dynamics of Apis mellifera Ligustica Spi. and Apis cerana cerana Feb. in Wanzhong, Wanxi and Wannan mountainous area in Anhui Province in 1997-1999. The results showed that the bee population size was influenced by climate and flower fertility, which was higher in Spring and Autumn, and lower in Summer and Winter. The propagation and renewal of A. mellifera in Autumn were quicker than those of A. cerana cerana, while the effect of overcoming Summer was inferior to that of Apis cerana cerana. The sex ratio of A. mellifera was (314.4 +/- 289.9):1-(329.4 +/- 305.8):1, and that of A. cerana cerana was (334.2 +/- 235.5):1-(413.1 +/- 377.2):1. The birth of drones was seasonal, and the age structure of each bee population was variable.
