Toxicological assessment of chemicals is crucial for safeguarding human health and the environment. However, traditional animal experiments are associated with ethical, technical, and predictive limitations in assessing the toxicity of chemicals to the skin. With the recent development of bioengineering and tissue engineering, three-dimensional (3D) skin models have been commonly used as an alternative for toxicological studies. The skin consists of the subcutaneous, dermis, and epidermis. All these layers have crucial functions such as physical and biological protection and thermoregulation. The epidermis is the shallowest layer protecting against external substances and media. Because the skin is the first contact point for many substances, this organ is very significant for assessing local toxicity following skin exposure. According to the classification of the United Nations Global Harmonized System, skin irritation is a major potentially hazardous characteristic of chemicals, and this characteristic must be accurately assessed and classified for enhancing chemical safety management and preventing and reducing chemical accidents. Background: The long-term survival benefit of immune checkpoint inhibitors (ICIs) in neoadjuvant and adjuvant settings is unclear for colorectal cancers (CRC) and gastric cancers (GC) with deficiency of mismatch repair (dMMR) or microsatellite instability-high (MSI-H)., Methods: This retrospective study enrolled patients with dMMR/MSI-H CRC and GC who received at least one dose of neoadjuvant ICIs (neoadjuvant cohort, NAC) or adjuvant ICIs (adjuvant cohort, AC) at 17 centers in China. Patients with stage IV disease were also eligible if all tumor lesions were radically resectable., Results: In NAC (n = 124), objective response rates were 75.7% and 55.4%, respectively, in CRC and GC, and pathological complete response rates were 73.4% and 47.7%, respectively. The 3-year disease-free survival (DFS) and overall survival (OS) rates were 96% (95%CI 90-100%) and 100% for CRC (median follow-up [mFU] 29.4 months), respectively, and were 84% (72-96%) and 93% (85-100%) for GC (mFU 33.0 months), respectively. In AC (n = 48), the 3-year DFS and OS rates were 94% (84-100%) and 100% for CRC (mFU 35.5 months), respectively, and were 92% (82-100%) and 96% (88-100%) for GC (mFU 40.4 months), respectively. Among the seven patients with distant relapse, four received dual blockade of PD1 and CTLA4 combined with or without chemo- and targeted drugs, with three partial response and one progressive disease., Conclusion: With a relatively long follow-up, this study demonstrated that neoadjuvant and adjuvant ICIs might be both associated with promising DFS and OS in dMMR/MSI-H CRC and GC, which should be confirmed in further randomized clinical trials., (© 2024. Background: Alzheimer's disease (AD) is the most common neurodegenerative disease. Studying the effects of drug treatments on multiple health outcomes related to AD could be beneficial in demonstrating which drugs reduce the disease burden and increase survival., Methods: We conducted a comprehensive causal inference study implementing doubly robust estimators and using one of the largest high-quality medical databases, the Oracle Electronic Health Records (EHR) Real-World Data. Our work was focused on the estimation of the effects of the two common Alzheimer's disease drugs, Donepezil and Memantine, and their combined use on the five-year survival since initial diagnosis of AD patients. Also, we formally tested for the presence of interaction between these drugs., Results: Here, we show that the combined use of Donepezil and Memantine significantly elevates the probability of five-year survival. In particular, their combined use increases the probability of five-year survival by 0.050 (0.021, 0.078) (6.4%), 0.049 (0.012, 0.085), (6.3%), 0.065 (0.035, 0.095) (8.3%) compared to no drug treatment, the Memantine monotherapy, and the Donepezil monotherapy respectively. We also identify a significant beneficial additive drug-drug interaction effect between Donepezil and Memantine of 0.064 (0.030, 0.098)., Conclusions: Based on our findings, adopting combined treatment of Memantine and Donepezil could extend the lives of approximately 303,000 people with AD living in the USA to be beyond five-years from diagnosis. Vacuum induction melting is a more energy-efficient process for the preparation of a titanium alloy with good homogeneity and low cost. But the crucial problem for this technology is in developing a crucible refractory with high stability. In the present work, a novel (Ca,Sr,Ba)ZrO 3 crucible was prepared by slip casting and its performance in melting NiTi alloy was studied. The results showed that a single solid solution was formed with a homogeneous distribution of metal elements after sintering at 1500 °C. It was found that the total content of oxygen and nitrogen remaining in the TiNi alloy after melting in the (Ca,Sr,Ba)ZrO 3 crucible was 0.0173 wt.%, which fulfills the ASTM standard on biomedical TiNi alloys. The good resistance of the (Ca,Sr,Ba)ZrO 3 crucible to molten NiTi has a relationship with the sluggish diffusion effect of high-entropy ceramics. This study provides insights into the process of designing highly suitable crucible material for melting a NiTi alloy.

Introduction: Gastric neuroendocrine carcinoma (GNENs), as a highly malignant gastrointestinal tumor, is characterized by easy postoperative metastasis, and its prognosis has always been poor compared to other gastrointestinal tumors. Due to its rarity, there have been few case reports and studies on it. As of now, there is no clear diagnosis and treatment sequence and guidelines. In this report, we present a case of GNENs that underwent first-line treatment after surgery. The patient achieved an excellent prognosis and survival after a combination of chemotherapy resistance and immunotherapy. This report aims to provide valuable reference and guidance for the treatment of GNENs in the future. Therefore, we conducted a literature review on GNENs., Patient Concerns: A 61-year-old man was admitted to the hospital with complaints of upper abdominal distension and discomfort that had been persistent for 1 month. During the endoscopy examination, a 2.5 cm irregular deep mucosal defect was observed at the center of the gastric angle. The defect appeared covered with dirty moss, had a hard texture, and exhibited a tendency to bleed upon contact., Diagnosis: Biopsy results confirmed the presence of a medium to poorly differentiated adenocarcinoma in the gastric horn. Subsequently, the patient underwent surgery, and the removed specimen was diagnosed as GNENs., Intervention: Postoperative chemotherapy combined with immunotherapy., Outcome: The patient in this case achieved a good prognosis and extremely long survival [overall survival > 3 years+] after receiving first-line treatment, which included chemotherapy, drug resistance and immunotherapy, and is currently in good health condition. The tumor is not sensitive to the standard EP regimen for neuroendocrine carcinoma, but after being replaced with oxaliplatin based regimen combined with immunotherapy, partial response was obtained, indicating a synergistic effect between chemotherapy and immunity. Background: Asthma is a chronic inflammatory disorder with high prevalence in childhood. Airway remodeling, an important structural change of the airways, is resulted from epithelial-mesenchymal transition. Long non-coding RNA non-coding RNA activated by DNA damage (NORAD) has been found to promote epithelial-mesenchymal transition in multiple cancers. This study aimed to analyze the role of NORAD in asthma, mainly focusing on epithelial-mesenchymal transition-mediated airway remodeling, and further explored the NORAD-miRNA-mRNA network., Methods: NORAD expression was analyzed in transforming growth factor-β1-induced BEAS-2B human bronchial epithelial cells and ovalbumin-challenged asthmatic mice. The influences of NORAD on the epithelial-mesenchymal transition characteristics and Wnt/β-catenin pathway activation were analyzed in vitro . The interactions between NORAD and miR-410-3p as well as miR-410-3p and regulator of chromosome condensation 2 were detected by dual-luciferase reporter assay and RNA pull-down assay. Rescue experiments using miR-410-3p antagonist and chromosome condensation 2 overexpression were used to confirm the mechanism of NORAD. Additionally, the role and mechanism of NORAD were further evaluated in asthmatic mice., Results: NORAD expression was elevated in both asthmatic models. Knockdown of NORAD impeded spindle-like morphology changes, elevated E-cadherin expression, decreased N-cadherin expression, suppressed cell migration, and inactivated the Wnt/β-catenin pathway in transforming growth factor-β1-stimulated BEAS-2B cells. NORAD acted as a sponge of miR-410-3p to regulate chromosome condensation 2 expression. Rescue assays demonstrated that silencing of NORAD ameliorated transforming growth factor-β1-induced EMT via miR-410-3p/chromosome condensation 2/Wnt/β-catenin axis. Potato virus Y (PVY) is a highly destructive pathogen that infects Solanum tuberosum vL., commonly known as potato, a crop that produces one of the most crucial food staples of the world. The PVY viral infection can considerably reduce the yield and quality of potatoes, thereby causing significant economic ramifications. Given the unsatisfactory performance of commercially available antiviral agents against PVY, we synthesized a series of novel indole-derived compounds followed by their bioevaluation and investigation of the mechanisms governing their anti-PVY activity. These indole-based derivatives contain dithioacetal as a key chemical moiety, and most of them exhibit promising anti-PVY activities. In particular, compound B2 displays remarkable in vivo protective and inactivating properties, with half-maximal effective concentration (EC 50 ) values of 209.3 and 113.0 μg/mL, respectively, in stark contrast to commercial agents such as ningnanmycin (EC 50 = 281.4 and 136.3 μg/mL, respectively) and ribavirin (EC 50 = 744.8 and 655.4 μg/mL, respectively). The mechanism using which B2 enhances plant immune response to protect plants from PVY is elucidated using enzyme activity tests, real-time quantitative polymerase chain reaction (RT-qPCR), and proteomics techniques. This study aims to pave the way for developing candidate pesticides and related molecules using antiphytoviral activity.

Alzheimer's disease is the most common type of dementia that currently affects over 6.5 million people in the U.S. Currently there is no cure and the existing drug therapies attempt to delay the mental decline and improve cognitive abilities. Two of the most commonly prescribed such drugs are Donepezil and Memantine. We formally tested and confirmed the presence of a beneficial drug-drug interaction of Donepezil and Memantine using a causal inference analysis. We applied doubly robust estimators to one of the largest and high-quality medical databases to estimate the effect of two commonly prescribed Alzheimer's disease (AD) medications, Donepezil and Memantine, on the average number of hospital or emergency department visits per year among patients diagnosed with AD. Our results show that, compared to the absence of medication scenario, the Memantine monotherapy, and the Donepezil monotherapy, the combined use of Donepezil and Memantine treatment significantly reduces the average number of hospital or emergency department visits per year by 0.078 (13.8%), 0.144 (25.5%), and 0.132 days (23.4%), respectively. The assessed decline in the average number of hospital or emergency department visits per year is consequently associated with a substantial reduction in medical costs. As of 2022, according to the Alzheimer's Disease Association, there were over 6.5 million individuals aged 65 and older living with AD in the US alone. If patients who are currently on no drug treatment or using either Donepezil or Memantine alone were switched to the combined used of Donepezil and Memantine therapy, the average number of hospital or emergency department visits could decrease by over 613 thousand visits per year. The misuse and abuse of opioids has become a serious public health threat in the United States. The state of California has been hit particularly hard by the opioid epidemic, with a noticeable increase in opioid-related fatalities and hospitalizations. This brief report paper aims to contribute to the growing literature by conducting a geospatial analysis of opioid dispensing patterns in California in 2021. The primary objective was to identify areas characterized by high-risk opioid dispending patterns and explore possible contributing factors. This retrospective study analyzed data from over 7 million records of opioid and benzodiazepine prescriptions dispensed by outpatient pharmacies in California in 2021. A series of generalized linear regression models was employed to assess the impact of neighborhood characteristics on opioid recipients and high-risk opioid dispensing. The study defined high-risk opioid dispensing behavior as: (1) multiple provider episodes, (2) overlapping opioid prescriptions for seven or more days, (3) overlapping opioid and benzodiazepine prescriptions for seven or more days, and (4) a high standardized dosage of opioid prescriptions per month. The study identified variables associated with high-risk opioid dispensing behaviors, including age, population density, income, and housing-related variables, as well as marital status and family-related variables. The study uncovered that there are noticeable disparities in opioid dispensing among different racial and ethnic groups within California. The findings indicated a correlation of high-risk dispensing indicators with certain demographic and socioeconomic factors. There was a substantial regional variation in opioid dispensing practices, with certain rural areas having higher rates of opioid prescriptions than urban areas.

Suicidal and self-injurious incidents in correctional settings deplete the institutional and healthcare resources, create disorder and stress for staff and other inmates. Traditional statistical analyses provide some guidance, but they can only be applied to structured data that are often difficult to collect and their recommendations are often expensive to act upon. This study aims to extract information from medical and mental health progress notes using AI algorithms to make actionable predictions of suicidal and self-injurious events to improve the efficiency of triage for health care services and prevent suicidal and injurious events from happening at California's Orange County Jails. The results showed that the notes data contain more information with respect to suicidal or injurious behaviors than the structured data available in the EHR database at the Orange County Jails. Using the notes data alone (under-sampled to 50%) in a Transformer Encoder model produced an AUC-ROC of 0.862, a Sensitivity of 0.816, and a Specificity of 0.738. Incorporating the information extracted from the notes data into traditional Machine Learning models as a feature alongside structured data (under-sampled to 50%) yielded better performance in terms of Sensitivity (AUC-ROC: 0.77, Sensitivity: 0.89, Specificity: 0.65). Purpose: To investigate the efficacy of PD-1/PD-L1 inhibitors plus chemotherapy versus anti-PD-1/PD-L1 monotherapy in advanced microsatellite instability (MSI)/mismatch repair-deficient (dMMR) gastrointestinal cancers., Methods: We retrospectively recruited patients with MSI/dMMR gastrointestinal cancer who received anti-PD-1/PD-L1 with or without chemotherapy and compared objective response rate (ORR), disease control rate (DCR), progression-free survival (PFS), and overall survival (OS) of PD-1/PD-L1 inhibitor plus chemotherapy (chemo-anti-PD-1/PD-L1 group) and PD-1/PD-L1 inhibitor alone (anti-PD-1/PD-L1 group). Propensity score-based overlap weighting analysis was conducted to adjust the baseline covariable imbalance. Sensitivity analysis was performed to confirm the stability of the results by propensity score matching and multivariable Cox and logistic regression models., Results: A total of 256 patients were eligible, with 68 and 188 receiving chemo-anti-PD-1/PD-L1 and anti-PD-1/PD-L1, respectively. The chemo-anti-PD-1/PD-L1 group showed significant improvements versus the anti-PD-1/PD-L1 group in ORR (61.8% v 38.8%; P = .001), DCR (92.6% v 74.5%; P = .002), PFS (median PFS [mPFS], not reached [NR] v 27.9 months; P = .004), and OS (median OS [mOS], NR v NR; P = .014). After overlap weighting, the improvements tended to be more significant with chemo-anti-PD-1/PD-L1 versus anti-PD-1/PD-L1 in ORR (62.5% v . 38.3%; P < .001), DCR (93.8% v 74.2%; P < .001), PFS (mPFS, NR v 26.0 months; P = .004), and OS (mOS, NR v NR; P = .010). These results were solidified through sensitivity analysis., Conclusion: Chemo-anti-PD-1/PD-L1 is superior to anti-PD-1/PD-L1 in MSI/dMMR gastrointestinal cancers with improved efficacy.

Antimicrobial peptides (AMPs) have attracted considerable interest in the past decade due to their advantages for tackling antibiotic resistance. They exhibit potential antimicrobial activity through unique cell membrane destruction mechanism based on their net charge, hydrophobic properties, and α-helix. In this work, a series of HJH peptides was rationally designed and synthesized. The antimicrobial activity and cytotoxicity assays indicated that HJH-5 and HJH-6 containing hydrophobic residues and helices displayed prominent antimicrobial activity and mild cytotoxicity, respectively. TiO 2 /Au/BDD composites with a Z-scheme structure was prepared by orderly depositing gold (Au) and titanium dioxide (TiO 2 ) on the surface of a boron-doped diamond (BDD) film using sputtering and electrophoretic deposition methods. It was found that the introduction of Au between TiO 2 and the BDD, not only could reduce their contact resistance, to increase the carrier transport efficiency, but also could improve the surface Hall mobility of the BDD electrode. Meanwhile, the designed Z-scheme structure provided a fast channel for the electrons and holes combination, to promote the effective separation of the electrons and holes produced in TiO 2 and the BDD under photoirradiation. The electrochemical characterization elucidated that these modifications of the structure obviously enhanced the electrocatalytic performance of the electrode, which was further verified by the simulated wastewater degradation experiments with reactive brilliant red X-3B. In addition, it was also found that the photoirradiation effectively enhanced the pollution degradation efficiency of the modified electrode, especially for the TiO 2 /Au/BDD-30 electrode.

Ethylicin (ET) was reported to be promising in the control of rice bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae ( Xoo ). The detailed mechanism for this process remains unknown. Disclosed here is an in-depth study on the action mode of ET to Xoo . Through plant physiological and biochemical analysis, it was found that ET could inhibit the proliferation of Xoo by increasing the content of defense enzymes and chlorophyll in rice ( Oryza sativa ssp. Japonica cv. Nipponbare). Label-free quantitative proteomic analysis showed that ET affected the rice abscisic acid (ABA) signal pathway and made the critical differential calcium-dependent protein kinase 24 (OsCPK24) more active. In addition, the biological function of OsCPK24 as a mediator for rice resistance to Xoo was determined through the anti- Xoo phenotypic test of OsCPK24 transgenic rice and the affinity analysis of the OsCPK24 recombinant protein in vitro and ET. This study revealed the molecular mechanism of ET-induced resistance to Xoo in rice via OsCPK24 , which provided a basis for the development of new bactericides based on the OsCPK24 protein.

Cyclic arginine-glycine-aspartic (RGD) peptides that specifically bind to integrin α ν β 3 have been developed for drug delivery, tracers, and imaging for tumor diagnosis and treatment. Herein, a series of polycyclic RGD peptides containing dual, tri, and tetra rings were designed and synthesized through sortase A-mediated ligation. The ageing of the population has resulted in an increase in the demand for long-term care services for older people, but with limited resources, its challenges have also been highlighted. Although factors affecting the willingness of older people to engage with long-term care services have been widely reported, however, systematic review has not synthesised the evidence, and its associated factors remain unclear. This systematic review aims to study and synthesise the best available evidence on the potential factors related to the willingness of older people to engage with long-term care services. Eight electronic databases were comprehensively searched from inception to January 2021: the Cochrane Library, PubMed, Web of Science, CINAHL, Wanfang Database, China Knowledge Resource Integrated Database (CNKI), Weipu Database (VIP), and Chinese Biomedical Literature Database (CBM). The original literatures were screened according to selection criteria. Two researchers used 11-item checklist recommended by the Agency for Healthcare Research and Quality to evaluate the quality of the included literatures. And the review used narrative synthesis to integrate factors related to the willingness of older people to engage with long-term care services. In total, 7794 studies were screened and 35 studies were included in this review. Among them, 23 studies were rated as moderate quality and 12 was high quality. Data synthesis identified that age, education, number of children, living arrangements, the relationship with children, primary caregivers, place of residence, social support, household income, medical insurance, activities of daily living ability and spiritual comfort should all be taken into account when establishing the appropriate long-term care service model or formulating relevant policies. However, many factors remain undetermined and require more rigorous original literature support. CsPbX 3 (X = Cl, Br, I) semiconductor nanocrystals (NCs) have excellent optical and photoelectric properties, and are potential core materials for various photoelectric devices. However, the toxicity of Pb and instability have been the key limitations to application of NCs. Herein, using MnBr 2 and MnBr 2 ·4H 2 O as manganese sources, heavy Mn-doped CsPbBr 3 (Mn:CsPbBr 3 ) NCs are synthesized by high-energy ball grinding, which avoids high temperature, a large number of polar solvents and atmosphere protection required in traditional liquid phase methods. However, when MnBr 2 ·4H 2 O is used as the raw material, infinite solid solution doping can be achieved, and the synthesized Mn:CsPbBr 3 NCs show smaller particle size, stronger PL intensity and stability. The reason is that presence of crystal water plays a similar role to wet milling in the ball milling process, and can promote the passivation effect of oleylamine (OAm) on nanocrystal defects and the connection between them. In addition, a simple, easy-operating and beneficial to commercial production method for the preparation of Mn:CsPbBr 3 NCs/EVA flexible films is proposed, which can effectively improve the stability of Mn:CsPbBr 3 NCs. Background: Discharge medical notes written by physicians contain important information about the health condition of patients. Many deep learning algorithms have been successfully applied to extract important information from unstructured medical notes data that can entail subsequent actionable results in the medical domain. This study aims to explore the model performance of various deep learning algorithms in text classification tasks on medical notes with respect to different disease class imbalance scenarios., Methods: In this study, we employed seven artificial intelligence models, a CNN (Convolutional Neural Network), a Transformer encoder, a pretrained BERT (Bidirectional Encoder Representations from Transformers), and four typical sequence neural networks models, namely, RNN (Recurrent Neural Network), GRU (Gated Recurrent Unit), LSTM (Long Short-Term Memory), and Bi-LSTM (Bi-directional Long Short-Term Memory) to classify the presence or absence of 16 disease conditions from patients' discharge summary notes. We analyzed this question as a composition of 16 binary separate classification problems. The model performance of the seven models on each of the 16 datasets with various levels of imbalance between classes were compared in terms of AUC-ROC (Area Under the Curve of the Receiver Operating Characteristic), AUC-PR (Area Under the Curve of Precision and Recall), F1 Score, and Balanced Accuracy as well as the training time. The model performances were also compared in combination with different word embedding approaches (GloVe, BioWordVec, and no pre-trained word embeddings)., Results: The analyses of these 16 binary classification problems showed that the Transformer encoder model performs the best in nearly all scenarios. In addition, when the disease prevalence is close to or greater than 50%, the Convolutional Neural Network model achieved a comparable performance to the Transformer encoder, and its training time was 17.6% shorter than the second fastest model, 91.3% shorter than the Transformer encoder, and 94.7% shorter than the pre-trained BERT-Base model. The BioWordVec embeddings slightly improved the performance of the Bi-LSTM model in most disease prevalence scenarios, while the CNN model performed better without pre-trained word embeddings. In addition, the training time was significantly reduced with the GloVe embeddings for all models., Conclusions: For classification tasks on medical notes, Transformer encoders are the best choice if the computation resource is not an issue. MXenes represent an emerging family of two-dimensional materials of transition metal carbides/carbonitrides terminated with functional groups like -O, -OH, and -F on the chemically active surface of MX slabs. As a member of the family, Nb 2 CT x exhibits superior lithium storage capacity over most of the other MXenes as anode materials in lithium-ion batteries (LIBs). However, an in-depth understanding of the charge storage mechanism is still lacking so far. Here, through combining complementary experiments and density functional theory calculations, we provide insights into the (de)lithiation process. Specifically, Nb 2 CT x with dominant -O functional groups stores charge as a result of changes in the oxidation states of both transition metals Nb and O, which is supported by Bader charge analysis showing a significant change in the oxidation states of Nb and O upon lithiation. As monitored by ex situ X-ray diffraction, the interlayer spacing of Nb 2 CT x changes slightly upon lithium ion (de)intercalation, corresponding to a volume change of only 2.3% with a near zero-strain feature. By coupling with a LiFePO 4 /C cathode, the full cell presents superior rate capability and cycling stability as well. The insights into the charge storage mechanism of Nb 2 CT x in this work provide useful guidance for the rational design of MXene-based anode materials for high-performance LIBs.

Oxygen reduction reaction (ORR) occupies a pivotal position in fuel cell applications, and it is a challenge to obtain highly durable ORR catalysts. Herein, porous cobalt oxide microsphere growing at the surface of on nitrogen-doped graphitized-nano-diamond (CoO x /N-GND) was prepared using hydrothermal and subsequent heat treatment process. Porous cobalt oxide of high specific surface area could expose more surface Co 2+ that act as active sites than bulk one does. The doping of nitrogen also promotes the catalytic activity. Besides, nano-diamond (ND) of sp 3 hybrid structure was used as an electronic conduction carriers of ultrahigh stability to improve the durability of catalytic composite. Prepared CoO x /N-GND shows a satisfactory half-wave potential of 0.82 V (versus RHE), which is close to that of Pt/C (0.85 V), an excellent methanol tolerance and a lower activity loss after 5000 cycles. These merits inspire the application of CoO x /N-GND as the cathode of Zn-air battery and the battery performance was evaluated in this work. Photocatalytic degradation of pollutants has been proved to be an effective strategy for wastewater treatment. Herein, TiO 2 nanoparticles were synthesized on a Ti 3 C 2 matrix by in situ growth, forming Z-scheme TiO 2 @Ti 3 C 2 /Cd 0.5 Zn 0.5 S (TO/CZS) multilevel structured nanocomposites via one-step hydrothermal route. The effects of hydrothermal temperature and Cd 0.5 Zn 0.5 S content on microstructure and properties of composites were assessed. TO/CZS nanocomposites were probed into phase composition, morphological and optical properties with x-ray diffractometer, infrared radiation, scanning electron microscope and UV-vis reflective spectra. Following the hydrothermal reaction at 160 °C for 12 h, TiO 2 nanoparticles of 30 nm in diameter were generated in situ on Ti 3 C 2 lamina and Cd 0.5 Zn 0.5 S particles were evenly distributed on the Ti 3 C 2 matrix. The photocatalytic activity of TO/CZS composites were evaluated, which found that degradation rate constant (k = 0.028 min -1 ) of TO/CZS-40 on Rhodamine B was 5.19 times that of pure TiO 2 and 4.48 times that of Cd 0.5 Zn 0.5 S. Through anchoring Ti 3 C 2 as an electron transition mediator and combination with TiO 2 and Cd 0.5 Zn 0.5 S, the new Z-scheme between TiO 2 oxidized by Ti 3 C 2 and Cd 0.5 Zn 0.5 S establishes a multilevel structure of separating electron-hole pairs. This work demonstrates a valid way to control electrons and hole transfer directions efficiently through designing multilevel semiconductor structural designs.

Purpose: To investigate the correlation between metastasis of colon cancer and the single nucleotide polymorphism (SNP) rs2555639 in nicotinamide adenine dinucleotide (NAD)+-dependent 15-hydroxyprostaglandin dehydrogenase (15-PGDH) (rs2555639)., Methods: We investigated the genotyping of peripheral blood genomic DNA in patients using the TaqMan probe method. The relationship between the genotype of 15-PGDH (rs2555639) and metastasis of colon cancer was analyzed., Results: We noticed that rs2555639 TT polymorphism was significantly correlated with the susceptibility to colon cancer metastasis. Also, in the stratified analysis, we found similar results., Conclusion: Our data suggested that the rs2555639 T allele is associated with increased risk of metastasis of colon cancer, which can be used as an indicator for colon cancer metastasis.

MXenes have emerged as promising high-volumetric-capacitance supercapacitor electrode materials, whereas their voltage windows are not wide. This disadvantage prevents MXenes from being made into aqueous symmetric supercapacitors with high energy density. To attain high energy density, constructing asymmetric supercapacitors is a reliable design choice. Here, we propose a strategy to achieve high energy density of hydrogen ion aqueous-based hybrid supercapacitors by integrating a negative electrode of Ti 3 C 2 T x MXene and a positive electrode of redox-active hydroquinone (HQ)/carbon nanotubes. The two electrodes are separated by a Nafion film that is proton permeable in H 2 SO 4 electrolyte. Upon charging/discharging, hydrogen ions shuttle back and forth between the cathode and anode for charge compensation. The proton-induced high capacitance of MXene and HQ, along with complementary working voltage windows, simultaneously enhance the electrochemical performance of the device. Specifically, the hybrid supercapacitors operate in a 1.6 V voltage window and deliver a high energy density of 62 Wh kg -1 , which substantially exceeds those of the state-of-the-art aqueous asymmetric supercapacitors reported so far. Additionally, the device exhibits excellent cycling stability and the all-solid-state planar hybrid supercapacitor displays exceptional flexibility and integration for bipolar cells to boost the capacitance and voltage output. These encouraging results provide the possibility of designing high-energy-density noble-metal-free asymmetric supercapacitors for practical applications.

BACKGROUND Bronchial epithelial cells proliferation plays a pivotal role in airway remodeling in children with severe asthma. MicroRNAs (miRNAs) have gained great attention for many diseases, including asthma. The purpose of this study was to explore the mechanisms that underlie miR-744 modulating bronchial epithelial cells proliferation in severe asthma in children. MATERIAL AND METHODS Bronchial epithelial cells were isolated from bronchial biopsies of normal controls and asthmatic subjects. miR-744 and transforming growth factor-ß1 (TGF-ß1) expressions were measured by quantitative reverse transcription PCR (qRT-PCR). Proliferating cell nuclear antigen (PCNA), phosphorylation or total of mothers against decapentaplegic homolog3 (Smad3) and production of Smad anchor for receptor activation (SARA) were measured via Western blot analysis. A link between miR-744 and TGF-ß1 was probed by luciferase activity and RNA immunoprecipitation. Cell proliferation was evaluated using the Cell Proliferation Assay Kit. RESULTS Severe asthma showed a significantly elevated cell proliferation rate and reduced abundance of miR-744, which in turn inhibited cell proliferation of bronchial epithelial cells. In particular, TGF-ß1 might be a candidate target of miR-744, and enrichment of miR-744 lowered the expression of TGF-ß1 at mRNA and protein levels. Indeed, overexpression of miR-744 lowered the proliferation rate of bronchial epithelial cells via driving TGF-ß1. Moreover, addition of miR-744 limited phosphorylation of Smad3 but reversed SARA protein abundance by regulating TGF-ß1. CONCLUSIONS The presence of miR-744 repressed bronchial epithelial cells proliferation through mediating the Smad3 pathway by modulating TGF-ß1, providing a promising therapeutic approach for epithelial function in severe asthma.

Background: Lonicera japonica has been studied extensively by scholars at home and abroad, a number of compounds have been isolated from it, which mainly include organic acids and flavonoids. Pharmacological studies have shown that Lonicera japonica has antibacterial and gall bladder-protective effects., Objective: To study the active constituents in Lonicera japonica and the mechanism of their anti-upper respiratory tract infection action in children., Methods: Compounds were identified by chromatographic methods, and the mechanism of anti-pediatric upper respiratory tract infection action of Lonicera japonica decoction was studied using experimental animals., Results: A total of four compounds were isolated, after injection of egg white, toe edema in rats in the control group was very obvious, different test concentrations of Lonicera japonica decoction all inhibited toe edema in rats to some extents, the edema was the mildest in the Lonicera japonica decoction high-dose group, which had the strongest inhibitory effect on the development of inflammation, the Lonicera japonica decoction showed certain dose-effect relationship with toe edema in rats. In the rat body temperature control experiment, while body temperature of rats in the blank group had already risen, other groups were still able to lower the body temperature of rats under the action of test drugs. The severity of ear edema in mice in the blank control group was obvious, with increased thickness which showed significant difference between left and right ears. Under test doses, three Lonicera japonica decoction groups all inhibited xylene-induced ear edema in mice., Conclusion: Lonicera japonica has an anti-upper respiratory tract infection action in children.

By depositing a graphene layer on the metallic film with subwavelength hole arrays, the tunable extraordinary transmission property based on the metal-dielectrics-graphene (MDG) structure has been investigated in the terahertz (THz) and near-infrared (NIR) regimes. The influences of operation frequency, composed materials, and the Fermi level of the graphene layer have been taken into account. The results show that by varying the Fermi level of the graphene layer, the transmission of the MDG structure can be tuned in a wide range and the modulation depth of the peak value of the transmission can reach more than 50%. But the tunable mechanisms in the THz and NIR regimes are quite different. In the infrared (THz) regime, the graphene behaves like the dielectric (metallic) layer; its dielectric constant decreases (increases) with the increase of Fermi level, resulting in the transmission increasing (decreasing). Compared with the metallic structure, the transmission of the semiconductor structure can also be modulated by using the doping or varying temperature; its peak position can also be changed in a much broader range. The results are very useful to understand the mechanism of the graphene plasmonic devices and to design novel filters, switchers, modulators, and sensors.

Ectopic expression of reprogramming factors has been widely adopted to reprogram somatic nucleus into a pluripotent state (induced pluripotent stem cells [iPSCs]). However, genetic aberrations such as somatic gene mutation in the resulting iPSCs have raised concerns regarding their clinical utility. To test whether the increased somatic mutations are primarily the by-products of current reprogramming methods, we reprogrammed embryonic fibroblasts of inbred C57BL/6 mice into either iPSCs (8 lines, 4 previously published) or embryonic stem cells (ESCs) with somatic cell nuclear transfer (SCNT ESCs; 11 lines). Exome sequencing of these lines indicates a significantly lower mutation load in SCNT ESCs than iPSCs of syngeneic background. In addition, one SCNT-ESC line has no detectable exome mutation, and two pairs of SCNT-ESC lines only have shared preexisting mutations. In contrast, every iPSC line carries unique mutations. Our study highlights the need for improving reprogramming methods in more physiologically relevant conditions.

Thymus transplantation has great clinical potential for treating immunological disorders, but the shortage of transplant donors limits the progress of this therapy. Human embryonic stem cells (hESCs) are promising cell sources for generating thymic epithelial cells. Here, we report a stepwise protocol to direct the differentiation of hESCs into thymic epithelial progenitor-like cells (TEPLCs) by mimicking thymus organogenesis with sequential regulation of Activin, retinoic acid, BMP, and WNT signals. The hESC-derived TEPLCs expressed the key thymic marker gene FOXN1 and could further develop in vivo into thymic epithelium expressing the functional thymic markers MHC II and AIRE upon transplantation. Moreover, the TEPLC-derived thymic epithelium could support mouse thymopoiesis in T-cell-deficient mice and promote human T cell generation in NOD/SCID mice engrafted with human hematopoietic stem cells (hHSCs). These findings could facilitate hESC-based replacement therapy and provide a valuable in vitro platform for studying human thymus organogenesis and regeneration., (Copyright © 2013 Elsevier Inc. Narciclasine (NCS) is a plant growth inhibitor isolated from the secreted mucilage of Narcissus tazetta bulbs. It is a commonly used anticancer agent in animal systems. In this study, we provide evidence to show that NCS also acts as an agent in inducing programmed cell death (PCD) in tobacco Bright Yellow-2 (TBY-2) cell cultures. NCS treatment induces typical PCD-associated morphological and biochemical changes, namely cell shrinkage, chromatin condensation and nuclear DNA degradation. To investigate possible signaling events, we analyzed the production of reactive oxygen species (ROS) and the function of mitochondria during PCD induced by NCS. A biphasic behavior burst of hydrogen peroxide (H(2)O(2)) was detected in TBY-2 cells treated with NCS, and mitochondrial transmembrane potential (MTP) loss occurred after a slight increase. Pre-incubation with antioxidant catalase (CAT) and N-acetyl-L-cysteine (NAC) not only significantly decreased the H(2)O(2) production but also effectively retarded the decrease of MTP and reduced the percentage of cells undergoing PCD after NCS treatment. In conclusion, our results suggest that NCS induces PCD in plant cells; the oxidative stress (accumulation of H(2)O(2)) and the MTP loss play important roles during NCS-induced PCD., (Copyright © Physiologia Plantarum 2011.)

Background: Narciclasine (NCS) is an Amaryllidaceae alkaloid isolated from Narcissus tazetta bulbs. NCS has inhibitory effects on a broad range of biological activities and thus has various potential practical applications. Here we examine how NCS represses plant root growth., Results: Results showed that the inhibition of NCS on cell division in Arabidopsis root tips and its effects on cell differentiation are concentration-dependent; at low concentrations (0.5 and 1.0 μM) NCS preferentially targets mitotic cell cycle specific/cyclin complexes, whereas at high concentration (5.0 μM) the NCS-stimulated accumulation of Kip-related proteins (KRP1 and RP2) affects the CDK complexes with a role at both G1/S and G2/M phases., Conclusions: Our findings suggest that NCS modulates the coordination between cell division and differentiation in Arabidopsis root tips and hence affects the postembryonic development of Arabidopsis seedlings.

Plant development displays an exceptional plasticity and adaptability that involves the dynamic, asymmetric distribution of the phytohormone auxin. Polar auxin flow, which requires transport facilitators of the PIN family, largely contributes to the establishment and maintenance of auxin gradients and mediates multiple developmental processes. Here, we report the effects of narciclasine (NCS), an Amaryllidaceae alkaloid isolated from Narcissus tazetta bulbs, on postembryonic development of Arabidopsis roots. Arabidopsis seedlings grown on NCS showed defects in root gravitropism which correlates with a reduction in auxin transport in roots. Expressions of auxin transport genes were affected and the polar localization of PIN2 protein was altered under NCS treatment. Taken together, we propose that NCS modulates auxin transport gene expression and PIN2 localization, and thus affects auxin transport and auxin distribution necessary for postembryonic development of Arabidopsis roots., (Copyright © 2011 Elsevier GmbH. The role of ethylene and hydrogen peroxide (H₂O₂) in the induction of the alternative respiratory pathway (AP) in calluses from wild-type (WT) Arabidopsis and ethylene-insensitive mutant etr1-3 under salt stress was investigated. The capacity and the contribution of the AP to the total respiration were significantly induced by 100 mM sodium chloride (NaCl) in WT calluses but only slightly induced in etr1-3 calluses. Ethylene emission was enhanced in WT calluses under salt stress. Application of 1-aminocyclopropane-1-carboxylic acid (an ethylene precursor) further increased the AP capacity in WT calluses but not in etr1-3 calluses under salt stress. Reduction of ethylene production by aminooxyacetic acid (AOA, an ethylene biosynthesis inhibitor) in WT calluses eliminated the NaCl-induced increase of ethylene emission and inhibited AP induction under salt stress, suggesting that ethylene is required for AP induction. H₂O₂ enhanced ethylene production while ethylene reduced H₂O₂ generation in WT calluses under salt stress. In addition, ethylene and H₂O₂ modulated NaCl-induced alternative oxidase gene (AOX1a) expression and the increase in pyruvate content in WT calluses. Inhibition of the AP by salicylhydroxamic acid in WT calluses under salt stress resulted in severe cellular damage as indicated by the high content of H₂O₂, malondialdehyde and more electrolyte leakage. Taken together, ethylene and H₂O₂ are involved in the salt-induced increase of the AP, which plays an important role in salt tolerance in WT calluses, and ethylene may be acting downstream of H₂O₂.

Chemical sensors based on semiconducting metal oxide nanocrystals are of academic and practical significance in industrial processing and environment-related applications. Novel alcohol response sensors using two-dimensional WO(3) nanoplates as active elements have been investigated in this paper. Single-crystalline WO(3) nanoplates were synthesized through a topochemical approach on the basis of intercalation chemistry (Chen et al 2008 Small 4 1813). The as-obtained WO(3) nanoplate pastes were coated on the surface of an Al(2)O(3) ceramic microtube with four Pt electrodes to measure their alcohol-sensing properties. The results show that the WO(3) nanoplate sensors are highly sensitive to alcohols (e.g., methanol, ethanol, isopropanol and butanol) at moderate operating temperatures (260-360 degrees C). For butanol, the WO(3) nanoplate sensors have a sensitivity of 31 at 2 ppm and 161 at 100 ppm, operating at 300 degrees C. For other alcohols, WO(3) nanoplate sensors also show high sensitivities: 33 for methanol at 300 ppm, 70 for ethanol at 200 ppm, and 75 for isopropanol at 200 ppm. The response and recovery times of the WO(3) nanoplate sensors are less than 15 s for all the test alcohols. A good linear relationship between the sensitivity and alcohol concentrations has been observed in the range of 2-300 ppm, whereas the WO(3) nanoparticle sensors have not shown such a linear relationship. The sensitivities of the WO(3) nanoplate sensors decrease and their response times become short when the operating temperatures increase. The enhanced alcohol-sensing performance could be attributed to the ultrathin platelike morphology, the high crystallinity and the loosely assembling structure of the WO(3) nanoplates, due to the advantages of the effective adsorption and rapid diffusion of the alcohol molecules.

To explore the molecular mechanisms involved in asexual spore development in Phytophthora sojae, the zoospores of strain PS26 were treated with ultraviolet (UV) irradiation. After selection, a mutant progeny, termed PS26-U03, was obtained and demonstrated to exhibit no oospore production. A suppression subtractive hybridization (SSH) approach was developed to investigate differences in gene expression between PS26 and PS26-U03 during asexual sporogenesis. Of the 126 sequences chosen for examination, 39 putative unigenes were identified that exhibit high expression in PS26. These sequences are predicted to encode proteins involved in metabolism, cell cycle, protein biosynthesis, cell signalling, cell defence, and transcription regulation. Seven clones were selected for temporal expression analysis using RT-PCR based on the results of the dot-blot screens. Three of the selected genes, developmental protein DG1037 (UB88), glycoside hydrolase (UB149) and a hypothetical protein (UB145), were expressed only in PS26, whereas the transcripts of phosphatidylinositol-4-phosphate 5-kinase (UB36), FAD-dependent pyridine nucleotide-disulphide oxidoreductase (UB226) and sugar transporter (UB256) were expressed at very low levels in PS26-U03 but at high levels in PS26.

Natural killer (NK) cells are the effectors of innate immunity to act as the first line of defense against viruses and tumors. Granzyme H (GzmH) is predicted to evolve from GzmB and constitutively expressed at a high level in human NK cells. It indicates GzmH plays a pivotal role in NK cell mediated cytolysis. However GzmH is defined as an orphan granzyme and its function has less been defined. Here we demonstrate GzmH can induce rapid apoptosis of target cells, which is dependent on caspase activation and mitochondrial damage. GzmH-induced death is characterized by phophatidylserine externalization, nuclear condensation, DNA fragmentation, caspase activation and cytochrome c release that are hallmarks of typical apoptosis. GzmH can directly cleave ICAD to unleash CAD for DNA fragmentation. Moreover, GzmH directly processes Bid to produce the active form tBid leading to cytochrome c release. Therefore, GzmH may play an essential role in caspase-dependent pathogen clearance in the innate immunity that may complement the proapoptotic function of GzmB in human NK cells.

Objectives: Ovarian cancer has the highest mortality of all the gynecologic cancers. The antitumor agent paclitaxel has been proved to be efficient in the treatment of ovarian cancer. Our study is to develop a polymeric drug delivery system for paclitaxel and determine whether paclitaxel nanoparticle can inhibit growth of ovarian carcinoma xenografts in Fisher344 (F344) rats by intraperitoneal administration. The mechanism of paclitaxel nanoparticles in rats bearing ovarian cancer has been investigated in this study., Methods: Synthesize paclitaxel loading nanoparticle (PLA) by ultrasonic emulsification; MTT analysis identified cytotoxic activity of paclitaxel nanoparticle in vitro; rat ovarian carcinoma cells were injected into the peritoneal cavity of F344 rats. The antitumor effect of paclitaxel nanoparticle in vivo has been evaluated by measuring tumor weight and ascite volume. At the end of the procedure rats were sacrificed; tumors were excised and processed for PCNA staining, tissue terminal deoxynucleotide transferase-mediated dUTP nick and labeling assay and RT-PCR to evaluate the proliferative and apoptotic changes and cancer transfer-related gene expression induced by PLA. Paclitaxel concentration in plasma, pelvic lymph nodes, liver, and heart were determined by high-performance liquid chromatography., Results: Paclitaxel nanoparticle and PTX (Cremophor) showed equivalent cytotoxic activity in vitro. In rats implanted carcinoma cells, paclitaxel nanoparticles significantly reduced tumor weight and ascites volume, and induced apoptosis of tumor cells. PLA also inhibited cell proliferation and matrix metalloproteinase 9 mRNA expression. The paclitaxel concentration of pelvic lymph nodes in PLA treated animals was 20-fold higher than that of free PTX treated animals at 48 h after intraperitoneal administration., Conclusion: The intraperitoneal administration of paclitaxel nanoparticle can significantly inhibit the progression of ovarian carcinoma in peritoneal cavity of female F344 rat. The paclitaxel nanoparticle is safe and lymphatic targeting.

Granzyme (Gzm)M is constitutively highly expressed in NK cells that may play a critical role in NK cell-mediated cytolysis. However, the function of GzmM has been less defined. Just one report showed GzmM induces a caspase-independent death pathway. In this study, we demonstrate a protein transfection reagent Pro-Ject can efficiently transport GzmM into target cells. GzmM initiates caspase-dependent apoptosis with typical apoptotic nuclear morphology. GzmM induces DNA fragmentation, not DNA nicking. GzmM can directly degrade inhibitor of caspase-activated DNase to release the nuclease activity of caspase-activated DNase for damaging DNA. Furthermore, GzmM cleaves the DNA damage sensor enzyme poly(ADP-ribose) polymerase to prevent cellular DNA repair and force apoptosis.

Objective: To probe into the possible impact on T cell immunity in children with obstructive sleep apnea syndrome (OSAS)., Method: Lymphocytes in peripheral blood vessel of children with OSAS vs normal controls were checked by APAAP method and analyzed statistically., Result: The comparison with the controls showed that CD3+ had no significant difference (P>0.05), CD4+, CD4+/CD8+ were significantly lower, CD8+ was significantly higher in children with OSAS (P<0.01), and the analysis of the LSaO2 and period of history showed that the lower the LSaO2, and the longer the period of history, the more effect to CD4+, CD8+, CD3+/CD8+ (P<0.05)., Conclusion: T cell immunity are impaired in children with OSAS.

Objective: To explore the relationship of the ACE gene insertion/deletion (I/D) polymorphism and the rennin-angiotensin system in the pathogenesis of hypertension in OSAHS., Method: Gene DNA was extracted from blood samples and amplified by polymerase chain reaction (PCR). The distribution of.the ACE gene I/D allele and genotypes were analyzed in 30 OSAHS with hypertension patients, 30 normotensive OSAHS patients and 30 healthy control group without cardiovascular diseases. The serum level of angiotensin II were also measured in all subjects by angiotensin II enzyme immunoassay kit., Result: The frequency of II genotype and I allele were significantly higher in the OSAHS accompanied hypertension patients than those in the healthy controls (chi2=9.88, chi2=16.13, P<0.01, respectively) and in the normotensive OSAHS patients (chi2=5.67, P<0.05, chi2=8.61, P<0.01). The frequency of II genotype and I allele had no significantly different between the normotensive OSAHS patients and the healthy controls (P>0.05). The serum level of angiotensin II was significantly higher in both OSAHS patients with and without hypertension than that in the healthy control (t=3.66, t=3.23, respectively P<0.01).
