The residual air saturation plays a crucial role in modeling hydrological processes of groundwater and the migration and distribution of contaminants in subsurface environments. However, the influence of factors such as media properties, displacement history, and hydrodynamic conditions on the residual air saturation is not consistent across different displacement scenarios. We conducted consecutive drainage-imbibition cycles in sand-packed columns under hydraulic conditions resembling natural subsurface environments, to investigate the impact of wetting flow rate, initial fluid state, and number of imbibition rounds (NIR) on residual air saturation. The results indicate that residual air saturation changes throughout the imbibition process, with variations separated into three distinct stages, namely, unstable residual air saturation (S gr-u ), momentary residual air saturation (S gr-m ), and stable residual air saturation (S gr ). The results also suggest that the transition from S gr-u to S gr is driven by changes in hydraulic pressure and gradient; the calculated values followed the following trend: S gr  > S gr-u  > S gr-m . An increase in capillary number, which ranged from 1.46 × 10 -7 to 3.07 × 10 -6 , increased S gr-u and S gr-m in some columns. The increase in S gr ranged from 0.034 to 0.117 across all the experimental columns; this consistent increase can be explained by water film expansion at the primary wetting front along with a strengthening of the hydraulic gradient during water injection. Both the pre-covered water film on the sand grain surface and a pore-to-throat aspect ratio of up to 4.42 were identified as important factors for the increased residual air saturation observed during the imbibition process. Initial air saturation (S ai ) positively influenced all three types of residual air saturation, while initial capillary pressure (P ci ) exhibited a more pronounced inhibitory effect on residual air saturation, as it can partly characterized the initial connectivity of the air phase generated under different drying flow rates. Under identical wetting flow rate conditions, S gr was higher during the second imbibition than during the first imbibition due to variations in initial fluid state, involving both fluid distribution and the concentration of dissolved air in the pore water. In contrast, NIR did not have an obvious effect on the three types of residual air saturation. The sluggish redox reaction kinetics and "shuttle effect" of lithium polysulfides (LPSs) impede the advancement of high-performance lithium-sulfur batteries (LSBs). Transition metal phosphides exhibit distinctive polarity, metallic properties, and tunable electron configuration, thereby demonstrating enhanced adsorption and electrocatalytic capabilities towards LPSs. Consequently, they are regarded as exceptional sulfur hosts for LSBs. Moreover, the introduction of a heterogeneous structure can enhance reaction kinetics and expedite the transport of electrons/ions. In this study, a composite of hollow CoP-FeP cubes with heterostructure modified carbon nanotube (CoFeP-CNTs) was fabricated and utilized as sulfur host in advanced LSBs. The presence of carbon nanotubes (CNTs) facilitates enhanced electron and Li + transport. Meanwhile, the active sites within the heterogeneous interface of CoP-FeP suppress the "shuttle effect" and enhance the conversion kinetics of LPSs. Therefore, the CoFeP-CNTs/S electrode exhibited exceptional cycling stability and demonstrated a capacity attenuation of merely 0.051 % per cycle over 600 cycles at 1C. Background: The progression of non-small cell lung cancer (NSCLC) is significantly influenced by circular RNAs (circRNAs), especially in tumor hypoxia microenvironment. However, the precise functions and underlying mechanisms of dysregulated circRNAs in NSCLC remain largely unexplored., Methods: Differentially expressed circRNAs in NSCLC tissues were identified through high-throughput RNA sequencing. The characteristics of circ_0007386 were rigorously confirmed via Sanger sequencing, RNase R treatment and actinomycin D treatment. The effects of circ_0007386 on proliferation and apoptosis were investigated using CCK8, cloning formation assays, TUNEL staining, and flow cytometry assays in vitro. In vivo, xenograft tumor models were used to evaluate its impact on proliferation. Mechanistically, the regulatory relationships of circ_0007386, miR-383-5p and CIRBP were examined through dual luciferase reporter assays and rescue experiments. Additionally, we detected the binding of EIF4A3 to CRIM1 pre-mRNA by RNA immunoprecipitation and the interaction between YAP1 and EIF4A3 under hypoxic conditions by co-immunoprecipitation., Results: Our investigation revealed a novel circRNA, designated as circ_0007386, that was upregulated in NSCLC tissues and cell lines. Circ_0007386 modulated proliferation and apoptosis in NSCLC both in vitro and in vivo. Functionally, circ_0007386 acted as a sponge for miR-383-5p, targeting CIRBP, which influenced NSCLC cell proliferation and apoptosis via the PI3K/AKT signaling pathway. Furthermore, under hypoxic conditions, the interaction between YAP1 and EIF4A3 was enhanced, leading to the displacement of EIF4A4 from binding to CRIM1 pre-mRNA. This facilitated the back-splicing of CRIM1 pre-mRNA, increasing the formation of circ_0007386. The circ_0007386/miR-383-5p/CIRBP axis was significantly associated with the clinical features and prognosis of NSCLC patients., Conclusions: Circ_0007386, regulated by YAP1-EIF4A3 interaction under hypoxia conditions, plays an oncogenic role in NSCLC progression via the miR-383-5p/CIRBP axis., (© 2024. When large-scale electric vehicles are connected to the grid for unordered charging, it will seriously affect the stability and security of the power system. To solve this problem, this paper proposes a regional power network optimization scheduling method considering vehicle network interaction. Initially, based on the user behavior characteristics and charging and discharging characteristics of electric vehicles, a charging and discharging behavior model of electric vehicles was established. Based on the Monte Carlo sampling algorithm, the scheduling upper and lower limits of each scheduling cycle of electric vehicles were described, and the scheduling potential of each scheduling cycle of electric vehicles was obtained. Then, the electricity price is then used as an incentive parameter to guide EV users to charge during periods of low electricity prices and participate in discharge during periods of peak electricity prices. Aiming at the highest economic efficiency, the best consumption effect of new energy and the smoothest demand-side power curve of regional power grid, a three-objective optimal dispatching model was established. In the later stage, uncertainty factors are taken into consideration by introducing the concept of interval numbers, and an interval multi-objective optimization dispatching model is established. The two dispatching models are solved by NSGA-II algorithm and improved NSGA-II algorithm, and the Pareto solution set is obtained. Finally, based on the analytic Hierarchy Process (AHP), the optimal scheduling scheme is determined. The Monte Carlo sampling method is used to simulate the user side charging demand, and the effectiveness of this method is verified. This study aimed to investigate the mechanism of the effect of TREM2 on cognitive function in autistic mice. TREM2 overexpression and knockdown viruses were given to autism spectrum disorder (ASD) mice and BV2 microglia cell line. To assess cognitive performance, all groups of mice took part in the open field, new object recognition, Morris water maze, and three-box social experiments. Double immunofluorescence labeling demonstrated co-localization of LC3II and NeuN. Proteins from the PI3K/Akt/mTOR pathway were identified. In vivo, behavior studies revealed that TREM2 could successfully improve ASD mice's social interaction and cognitive performance. Besides, we discovered that TREM2 could increase autophagy in ASD mice. In vitro, overexpressing TREM2 reduced the expression of PI3K/AKT/mTOR pathway proteins, whereas knocking down TREM2 increased the expression of PI3K/AKT/mTOR pathway proteins. In conclusion, TREM2 could inhibit PI3K/Akt/mTOR signaling pathway, enhance autophagy, and improve the social communication ability and cognitive function of ASD mice.

Background: N 6 -methyladenosine (m 6 A) is a common modification in RNA, crucial for various cellular functions and associated with human diseases. Quantification of m 6 A at single-base resolution is of great significance for exploring its biological roles and related disease research. However, existing analysis techniques, such as polymerase chain reaction (PCR) or loop-mediated isothermal amplification (LAMP), face challenges like the requirement for thermal cycling or intricate primer design. Therefore, it is urgent to establish a simple, non-thermal cycling and highly sensitive assay for m 6 A., Results: Leveraging the inhibitory effect of m 6 A on primer elongation and uncomplicated feature of the isothermal exponential amplification reaction (IEXPAR), we have developed an extension-based IEXPAR (E-IEXPAR). This approach requires just a single extension primer and one template, simplifying the design process in comparison to the more complex primer requirements of the LAMP methods. The reactions are conducted at constant temperatures, therby elimiating the use of thermal cycling that needed in PCR methods. By combining IEXPAR with an extension reaction, E-IEXPAR can identify m 6 A in RNA concentrations as low as 4 fM. We have also introduced a new analytical model to process E-IEXPAR results, which can aid to minimize the impact of unmodified adenine (A) on m 6 A measurements, enabling accurate m 6 A quantification in small mixed samples and cellular RNA specimens., Significance and Novelty: E-IEXPAR streamlines m 6 A detection by eliminating the need for intricate primer design and thermal cycling, which are common in current analytical methods. Its utilization of an extension reaction for the initial identification of m 6 A, coupled with a novel calculation model tailored to E-IEXPAR outcomes, ensures accurate m 6 A selectivity in mixed samples. The Ledong Sea Area is located on the southwest side of Hainan Island. In recent years, with the development of industrialization and urbanization, the problem of heavy metals in marine sediments has gradually become a global problem, and research on this topic is of great significance for nearshore environmental protection and coastal management. This paper analysed the heavy metal content of 97 surface sediments in the Ledong Sea, indicating unpolluted to moderately polluted and low to moderate risk. Cu, Zn, Hg, Pb, Cr, and Cd are highly correlated, with similar origins, and originate from rivers carrying industrial wastewater, domestic sewage, and weathered material from the parent rocks, which are subsequently redistributed under the action of ocean dynamics. The distribution of Hg is mainly influenced by feed and biological metabolites during the farming process. Natural ginsenoside needs to be converted into rare ginsenoside before it can be readily absorbed into the bloodstream for action. In this study, an α-l-arabinofuranosidase (α-l-AFase) gene Bsafs2 was cloned from Bacillus subtilis (B. subtilis). Bsafs2 was ligated to the expression vector pET28a(+), and the expression vector was constructed and transformed into Escherichia coli (E. coli) BL21 heterologous recombinant expression to obtain α-l-AFase. α-l-AFase can hydrolyze at the C 20 site of Ginsenoside Rc to obtain rare ginsenoside Rd. Studies on the enzymatic property showed that α-l-AFase had good tolerance to ethanol, glucose, and l-arabinose. The optimum temperature of α-l-AFase was 40 °C and pH = 5.5. Kinetic parameters K m of α-l-AFase for pNPαAraf and Ginsenoside Rc were 1.93 and 8.9 mmol/L, the V max were 26 and 154 μmol/min/mg, the K cat were 24.14 and 1.48 S -1 , respectively. The high activity barriers of Li 2 S nucleation and deposition limit the redox reaction kinetics of lithium polysulfides (LiPSs), meanwhile, the significant shuttle effect of LiPSs hampers the advancement of Li-S batteries (LSBs). In this work, a NiSe 2 /CoSe 2 -rGO (NiSe 2 /CoSe 2 -G) sulfur host with bifunctional catalytic activity was prepared through a hard template method. Electrochemical experiment results confirm that the combination of NiSe 2 and CoSe 2 not only facilitates the bidirectional catalytic function during charge and discharge processes, but also increases the active sites toward LiPSs adsorption. Simultaneously, the highly conductive rGO network enhances the electronic conductivity of NiSe 2 /CoSe 2 -G/S and provides convenience for loading NiSe 2 /CoSe 2 catalysts. Benefitting from the exceptional catalytic-adsorption capability of NiSe 2 /CoSe 2 and the presence of rGO, the NiSe 2 /CoSe 2 -G/S electrode exhibits excellent electrochemical properties. At 1C, it demonstrates a low capacity attenuation of 0.087 % per cycle during 500 cycles. The electrode can maintain a discharge capacity of 927 mAh/g at a sulfur loading of 3.3 mg cm -2 . Background: Non-small cell lung cancer (NSCLC) is a type of lung cancer that occurs in the cells of the respiratory tract, and its development is influenced by the regulation of circular RNAs (circRNAs). However, the role of circRNA carboxypeptidase A4 (circCPA4) in the progression of NSCLC and the underlying mechanism remain relatively clear., Methods: The study utilized both real-time quantitative polymerase chain reaction (RT-qPCR) and western blot techniques to evaluate the levels of circCPA4, microRNA-145-5p (miR-145-5p), alanine, serine, or cysteine-preferring transporter 2 (ASCT2). To assess cell proliferation, cell counting kit-8 (CCK8) and 5-ethynyl-2'-deoxyuridine (EdU) assays were performed. Apoptosis was determined using flow cytometry, while cell migration and invasive capacity were evaluated through transwell and wound-healing assays. Intracellular levels of glutamine, glutamate, and α-KG were measured using specific kits. The relationship between miR-145-5p and circCPA4 or ASCT2 was confirmed using dual-luciferase reporter assay and RNA immunoprecipitation assay., Results: CircCPA4 and ASCT2 RNA levels were elevated, while miR-145-5p was downregulated in both NSCLC tissues and cells. Depletion of circCPA4 significantly inhibited NSCLC cell proliferation, migration, invasion, and intracellular levels of glutamine, glutamate, and α-KG, and promoted apoptosis. Moreover, circCPA4 knockdown delayed tumor growth in vivo. Furthermore, circCPA4 was found to bind to miR-145-5p, thereby regulating the progression of NSCLC in vitro. ASCT2 was also identified as a downstream target of miR-145-5p, and its upregulation rescued the effects of miR-145-5p overexpression on NSCLC cell processes., Conclusion: CircCPA4 knockdown inhibited tumor property of NSCLC cells by modulating the miR-145-5p/ASCT2 axis., (© 2024 The Authors. The present study focuses on investigating 60 strains of yeast isolated from the natural fermentation broth of Vitis labruscana Baily × Vitis vinifera L. These strains underwent screening using lysine culture medium and esculin culture medium, resulting in the identification of 27 local non-Saccharomyces yeast strains exhibiting high β-glucosidase production. Subsequent analysis of their fermentation characteristics led to the selection of four superior strains (Z-6, Z-11, Z-25, and Z-58) with excellent β-glucosidase production and fermentation performance. Notably, these selected strains displayed a dark coloration on esculin medium and exhibited robust gas production during Duchenne tubules' fermentation test. Furthermore, all four non-Saccharomyces yeast strains demonstrated normal growth under specific conditions including SO 2 mass concentration ranging from 0.1 to 0.3 g/L, temperature between 25 and 30 °C, glucose mass concentration ranging from 200 to 400 g/L, and ethanol concentration at approximately 4%. Background: Autism spectrum disorder (ASD) encompasses a diverse range of neurodevelopmental disorders, but the precise underlying pathogenesis remains elusive. This study aim to explore the potential mechanism of TREM2 in regulating microglia function in ASD., Materials and Methods: The offspring rat model of ASD was established through prenatal exposure to valproic acid (VPA), and the behavioral symptoms of the ASD model were observed. On postnatal day (PND) 7 and PND 28, the effects of prenatally exposure to VPA on synaptic development and microglia phenotype of offspring rats were observed. Primary microglia were cultured in vitro. Lentivirus and adenovirus were utilized to interfere with TREM2 and overexpress TREM2., Results: Prenatally VPA exposure induced offspring rats to show typical ASD core symptoms, which led to abnormal expression of synapse-related proteins in the prefrontal cortex of offspring rats, changed the phenotype of microglia in offspring rats, promoted the polarization of microglia to pro-inflammatory type, and increased inflammatory response. The experimental results in vitro showed that overexpression of TREM2 could increase the expression of Gephyrin, decrease the content of CD86 protein and increase the content of CD206 protein. In addition, after the expression of TREM2 was interfered, the content of p-P38 MAPK protein increased and the content of p-ELK-1 protein decreased., Conclusion: The protective influence of TREM2 on the VPA-induced ASD model is attributed to its inhibition of the P38 MAPK pathway, this protective effect may be achieved by promoting the polarization of microglia to anti-inflammatory phenotype and improving the neuronal synaptic development., (© 2024. Quantification of microRNAs (miRNAs) at the single-molecule level is of great significance for clinical diagnostics and biomedical research. The challenges lie in the limits to transforming single-molecule measurements into quantitative signals. To address these limits, here, we report a new approach called a Single Microbead-based Space-confined Digital Quantification (SMSDQ) to measure individual miRNA molecules by counting gold nanoparticles (AuNPs) with localized surface plasmon resonance (LSPR) light-scattering imaging. One miRNA target hybridizes with the alkynyl-modified capture DNA probe immobilized on a microbead (60 μm) and the azide-modified report DNA probe anchored on AuNP (50 nm), respectively. Through the click reaction between the alkynyl and azide group, a single microbead can covalently link the AuNPs in the confined space within the view of the microscope. By digitally counting the light-scattering spots of AuNPs, we demonstrated the proposed approach with single-molecule detection sensitivity and high specificity of single-base discrimination. We developed a method for quantifying N 6 -methyladenosine at one-nucleotide resolution based on double blocking gap-filling-ligation and cascade isothermal amplification. This proposed method can detect as low as 1 fM target RNA, achieving selectivity up to approximately 100-fold between m 6 A and A, and has been successfully applied to the analysis of m 6 A at specific sites in cell samples.

The cytosine base editor (CBE) has shown promise as a gene editing tool for gene therapy, as it can convert cytidine to thymidine. Adeno-associated virus (AAV) has been widely used for in vivo gene therapy, but its limited 4.7 kb packing capacity presents challenges in delivering CBE by a single AAV. To address this, one feasible solution is to split CBE into two sections for dual-AAV delivery. In this study, we utilized BE3 as an example and constructed 22 potential split-BE3 pairs with the combination of 11 splitting sites and two split-inteins (Npu and Rma). These split-BE3 pairs were initially screened in the green fluorescent protein (GFP) reporter system, with six split-BE3 pairs selected for further evaluation. The subsequent screening of split-BE3 pairs was performed at two endogenous sites in 293T and HeLa cells, revealing that the split-BE3-Rma674, split-BE3-Rma713, and split-BE3-Rma1005 displayed effective C-to-T conversion after transfection. The effectiveness of dual-AAV split-BE3 was further validated in culture cells and adult mouse eyes. Of note, the split-BE3-Rma674 demonstrated the most efficient C-to-T editing after AAV infection, with a maximal editing efficiency of 23.29% ± 10.98% in the mouse retinal pigment epithelium cells in vivo . Overall, our study presents a novel split-BE3 system with effective C-to-T conversion, which could be applied to CBE-based in vivo gene therapy.

Background: This study aims to provide a theoretical basis for the prevention of oral cancer by analyzing the epidemiological trends of oral cancer., Materials and Methods: The data on oral cancer from 1990 to 2019 were extracted from the Global Burden of Disease 2019 database. The incidence, mortality, disability-adjusted life years (DALYs), and age-standardized rate as well as attributable risk factors of oral cancer were used for the analysis. Estimated annual percentage change (EAPC) was calculated to describe the changes in age-standardized incidence rate (ASIR), age-standardized mortality rate (ASMR), and age-standardized DALYs rate (ASDR)., Results: The global ASIR of oral cancer showed an increasing trend from 1990 to 2019. ASIR in high SDI regions showed a decreasing trend during the studied period, with high SDI regions having the lowest ASMR in 2019. In 2019, the highest ASIR, ASMR, and ASDR were detected in South Asia. At the national level, Pakistan had the highest ASMR and ASDR in 2019. The increasing disease burden was observed in younger populations aged below 45 during the studied period. Smoking and alcohol use still exerted profound impacts on the oral cancer burden, with South Asia having the greatest increase in the percentage of deaths due to oral cancer attributable to chewing tobacco from 1990 to 2019., Conclusion: In conclusion, there is a large variability in the temporal and spatial burden of oral cancer, and it is essential for priority countries to take targeted intervention policies and measures to reduce the disease burden of oral cancer. The target-dependent endonuclease activity (also known as the trans -cleavage activity) of CRISPR-Cas systems has stimulated great interest in the development of nascent sensing strategies for nucleic acid diagnostics. Despite many attempts, the majority of the sensitive CRISPR-Cas diagnostics strategies mainly rely on nucleic acid preamplification, which generally needs complex probes/primers designs, multiple experimental steps, and a longer testing time, as well as introducing the risk of false-positive results. In this work, we propose the CRISPR-Cas-Driven Single Micromotor (Cas-DSM), which can directly detect the nucleic acid targets at a single-molecule level with high specificity. We have demonstrated that the Cas-DSM is a reliable and practical method for the quantitative detection of DNA/RNA in various complex clinical samples as well as in individual cells without any preamplification processes. Due to the excellent features of the CRISPR/Cas system, including constant temperature, simple design, high specificity, and flexible programmability, the Cas-DSM could serve as a simple and universal platform for nucleic acid detection. More importantly, this work will provide a breakthrough for the development of next-generation amplification-free CRISPR/Cas sensing toolboxes.

The machining process of aluminum alloy usually produces built-up edge and tool sticking problems due to their low hardness and large plastic deformation, which may further affect the machined surface quality and tool life. This paper aims to investigate the influence of different cutting fluids on the machined surface quality and tool life during the milling process of 7050 aluminum alloy. A novel cutting fluid (QC-2803) was considered in the study, which is synthesized by addition of alkyl alcohol amide and chlorinated polyolefin, and the traditional cutting fluid (CCF-10) was used as the control group. The physical and chemical properties of two cutting fluids were characterized. The milling process of 7050 aluminum alloy was carried out under two different cutting fluid conditions. The machined surface morphology, cutting force and tool wear morphology were observed during the process. Results show that the surface tension of the novel cutting fluid is significantly lower than that of the traditional cutting fluid, which makes it easier to produce a lubricating film between the aluminum alloy and tool, and further benefits the machined surface quality and tool life. As a result, the surface roughness and cutting force are reduced by ~20.0% and ~42.9%, respectively, and the tool life is increased by 25.6% in the case of the novel cutting fluid (QC-2803). The results in this paper revealed the important laws of cutting fluid with metal surface quality, cutting performance and tool wear, which helps to control the machined surface quality and tool life by the selection of cutting fluid during metal milling.

False detection of SARS-CoV-2 is detrimental to epidemic prevention and control. The scalar nature of the detected signal and the imperfect target recognition property of developed methods are the root causes of generating false signals. Here, we reported a collaborative system of CRISPR-Cas13a coupling with the stabilized graphene field-effect transistor, providing high-intensity vector signals for detecting SARS-CoV-2. In this collaborative system, SARS-CoV-2 RNA generates a "big subtraction" signal with a right-shifted feature, whereas any untargets cause the left-shifted characteristic signal. Thus, the false detection of SARS-CoV-2 is eliminated. High sensitivity with 0.15 copies/μL was obtained. In addition, the wide concerned instability of the graphene field-effect transistor for biosensing in solution environment was solved by the hydrophobic treatment to its substrate, which should be a milestone in advancing it's engineering application. With the increasing requirement for high capacity energy storage systems, a large amount of recent work has focused on the development of zinc-iodine batteries (ZIBs) on account of high energy density, fast redox kinetics, and excellent reversibility. Nevertheless, low electron conductivity, the shuttle effect, and highly soluble iodine species (I 2 , I - , and I 3 - ) have impeded their widespread application. In this study, metal organic framework-5 (MOF-5)-derived mesoporous carbon (MPC) loaded iodine (MPC/I 2 ) cathode and the single-sided ketjen black modified cotton fiber (KB@CF) separator are designed to solve the problems mentioned above. That is, the double fixation strategy using MPC and KB@CF separators for iodine species suppresses the shuttle effect. Therefore, the ZIBs constructed with the MPC/I 2 cathode and the KB@CF separator can exhibit excellent electrochemical performance. At the current density of 0.1 A g -1 , a high discharge specific capacity of 137 mAh g -1 is still available after 300 cycles. Digital counting individual nucleic acid molecule is of great significance for fundamental biological research and accurate diagnosis of genetic diseases, which is hard to achieve with existing single-molecule detection technologies. Herein, we report a novel one-by-one single-molecule counting method for digital quantification of SARS-Cov-2 RNA. This method uses one fluorescent micromotor functionalized with peptide nucleic acids (PNAs) to specially capture one target RNA molecule. The RNA-micromotors can be propelled by the electric field to target district and accurately counted. In order to prepare high ionic conductivity and robust mechanical properties of alkaline solid polyelectrolyte (ASPE) for applications in flexible wearable devices, a co-continuous structure membrane was designed using in-situ polymerization to introduce cross-linked polyacrylic acid (N-PAA) into the cellulose network constructed by regenerated degreasing cotton (RDC). The resultant ASPE membrane showed high ionic conductivity (430 mS·cm -1 at 25 °C), strong mechanical properties, and excellent alkaline stabilities, proving the viability of cellulose for use in energy storage systems. Surprisingly, the sandwich-shaped zinc-air battery assembled using RDC/N-PAA/KOH membranes as electrolytes exhibits superior values of cycling stability, discharge time, specific capacity (731.5 mAh·g -1 ), peak power density (40.25 mW·cm -2 ), and mechanical flexibility. A novel selenium-electrocatalytic intramolecular cyclization of 2-vinylanilides for synthesis of functionalized indoles and azaindoles has been developed. In contrast to the previous synthetic methods, this sustainable protocol enabled unparalleled broad substrates scope for viable indoles with highly functional and sensitive groups by employing recyclable selenium catalyst, under mild, metal- and external-oxidant-free conditions. The approach can be used to the late-stage modification of complex bioactive molecular system, thereby setting the stage for versatile syntheses of decorated indoles with peptide labeling. Oxidative stress contributes to the pathogenesis of various neurodegenerative diseases and induction of the Kelch-like ECH-associated protein 1 (Keap1)-nuclear factor erythroid 2-related factor 2 (Nrf2) pathway is a validated neuroprotective strategy. Synthetically-derived samples of members of the ribisin class of natural product together with a range of analogues were evaluated for their neuroprotective capacities. Four of the twenty-four compounds tested were found to strongly stimulate antioxidant response element-dependent transcriptional activity in human-derived SH-SY5Y cells. Further, in rat pheochromocytoma PC12 cells and mouse brain cortical cultures these compounds upregulated levels of nuclear factor erythroid 2-related factor 2 (Nrf2) and its downstream target gene products, namely heme oxygenase (HO-1) and NAD(P)H quinone reductase 1 (NQO1). Functionally speaking, the compounds conferred protection in these cell models challenged with H 2 O 2 . In silico molecular modeling suggests that certain of the ribisins can dock in the Nrf2-binding Kelch domain in Keap1, while cysteine labeling by biotinylated iodoacetamide suggest that cysteine residues within Keap1 react with the ribisins. Rechargeable aqueous zinc-iodine batteries (ZIBs) with low environmental impacts and abundant natural reserves have emerged as promising electrochemical energy storage devices. However, the shuttle effect and low conductivity of the iodine species cause poor electrochemical performance and hinder their practical application. Herein, we propose a ZIF-8 derived porous carbon (ZPC) for iodine species immobilization in ZIBs. The rich porous structure and highly conductive framework of ZPC provide efficient iodine loading and allow the fast transmission of electrons. In addition, the presence of N, Zn and ZnO in the carbon framework can build chemical anchoring with the iodine species to mitigate the shuttle effect. Thus, the ZPC/I 2 cathode exhibits a reversible capacity of 156 mAh g -1 after 100 cycles at 100 mA g -1 and a long-term stability of 1000 cycles at a high rate. Objective: To analyze variants of TSC1 and TSC2 genes in a Chinese patient with tuberous sclerosis complex (TSC)., Methods: Peripheral blood samples were collected from the patient and her parents with informed consent. Following extraction of genomic DNA, potential variants of the TSC1 and TSC2 genes was detected by using targeted capture next-generation sequencing (NGS) and Sanger sequencing., Results: The patient was found to harbor a de novo mosaicism variant c.3295_3298delG (Val1100CysfsTer3) of the TSC2 gene, with the proportion of the mutant allele determined as 13.4%, which was confirmed by Sanger sequencing. Based on the guidelines from the American College of Medical Genetics and Genomics (ACMG), the c.3295_3298delG (Val1100CysfsTer3) variant was predicted to be pathogenic (PVS1+PS2+PM2)., Conclusion: The mosaicism heterozygous variant of c.3295_3298delG of the TSC2 gene, as detected by both NGS and Sanger sequencing, probably underlay the TSC2 in this patient.

The 2019 novel coronavirus disease is spreading all over the world. Pregnant women and infants require particular concern, owing to the special immune conditions. A case of a pregnant woman who was exposed to SARS-CoV-2 at 34+1 weeks gestation and chose to continue pregnancy is reported. Without obvious symptoms or signs, the woman did not receive any treatment before delivery, and gave birth at 37+5 weeks to a neonate with positive immunoglobulin G for SARS-CoV-2 and negative nucleic acid tests. The mother was given anti-infection, oxytocin, and fluid rehydration treatment after delivery. Both mother and infant recovered well after a three-month follow-up. Continued expectation to deliver at term instead of preterm can decrease the potential risk of severe perinatal and infant complications and is beneficial to the development of the neonate. In this study, a type of alkaline solid polyelectrolyte (ASPE) membrane was developed via the introduction of microcrystalline cellulose (MCC) and its modified product (QMCC) into the polyvinyl alcohol (PVA) matrix. In this process, green NaOH/urea-based solvent was used to achieve a good dispersion of MCC in the PVA matrix; meanwhile, the OH - groups in the NaOH/urea-based solvent provided an alkaline environment for good ion conductivity. Compared to the MCC-incorporated ASPE, further improved conductivity was achieved when the MCC was modified with quantitative quaternary ammonium salt. TGA showed that the addition of QMCC improved the water retention of the matrix, which was beneficial to the OH - conduction in the system. Compared to the control (50 mS cm -1 ), a maximum conductivity of 238 mS cm -1 was obtained after the incorporation of QMCC in the PVA matrix. Moreover, the tensile strength of the polymer electrolyte were also significantly increased with the addition of QMCC. Finally, this developed ASPE membrane was used in assembling a flexible Zn-air battery and showed a promising potential in the development of flexible electronic devices.

A highly sensitive HpaII-mediated ligation PCR assay that can precisely discriminate 5hmC from 5mC and C is developed for the quantitative determination of site-specific 5hmC in genomic DNA samples.

Herein, a metal-organic framework (MOF)/polythiophene (PTh)-derived S-doped carbon is successfully designed and prepared employing zeolitic imidazolate frameworks (ZIF-8/ZIF-67) and thiophene (Th) as precursors. The S-doped carbon presents a neuronlike three-dimensional network structure (3DSC). The 3DSC delivers extra-high capacities (225 mAh/g at 5000 mA/g after 3000 cycles) and excellent endurance ability of current changes when applied in Na-ion batteries (SIBs). Moreover, when the 3DSC-700 anode is coupled with a sodium vanadium phosphate cathode to construct a Na-ion full cell, after 50 cycles, a high capacity of ∼229.64 mAh/g is obtained at 100 mA/g. Electrochemical impedance spectroscopy analysis, density functional theory calculations, and pseudocapacitance contributions are adopted to investigate the excellent sodium storage mechanism of the 3DSC electrode. A new idea has been provided in this work to open up the possibility of MOF materials and carbon-based materials applications in SIBs in the future.

Long non-coding RNA (lncRNA) SNHG14 is previously found to be overexpressed in several types of cancers. However, the clinical significance and biological function of SNHG14 in non-small cell lung cancer (NSCLC) are still elusive. In the present study, we found that SNHG14 was aberrantly up-regulated in NSCLC tissues from patients and cell lines compared with their normal counterparts. Increased SNHG14 expression was closely associated with aggressive tumor progression and poor clinical outcome of NSCLC patients. Knockdown of SNHG14 inhibited NSCLC cell proliferation through inducing cell cycle arrest and apoptosis, whereas SNHG14 overexpression exerted the opposite effects. Animal experiment further revealed that down-regulated SNHG14 greatly inhibited NSCLC tumor growth in vivo Further studies demonstrated that SNHG14 might serve as a competing endogenous RNA (ceRNA) by sponging miR-340 in NSCLC cells. Taken together, our study demonstrated that SNHG14/ miR-340 axis might play a novel role in regulating the malignant behaviors of NSCLC, which provided a new potential diagnostic and therapeutic strategy for this malignant disease., (© 2019 The Author(s).)

Newly developed adsorbents, poly(tannin-hexamethylendiamine) (PTHA), were fabricated by varying the mole ratio of tannin (TA) and hexamethylendiamine (HA) under one-pot reaction. The specific forming process of the adsorbent which had undergone the transition from hydrogen bonds to covalent bonds was subsequently explored. Based on the efficiency of Cr(VI) removal from aqueous solution over all prepared adsorbents, the PTHA-4 (mole ratio of TA/HA = 1:12.5) exhibited an excellent adsorption behavior. Adsorption experiments affected by contact time and ionic strength have been conducted successively by PTHA-4, and the equilibrium was reached at 24 h. The kinetic data revealed that the adsorption was good agreement with pseudo-second order model and needed to undergo the rate-controlling step. The maximum adsorption capacity was 283.29 mg/g at 30 °C, relying on the isothermal curve suitably described by Langmuir model. Furthermore, toxic Cr(VI) had been reduced to the low toxic Cr(III) during adsorption process. The structures and adsorption performance of adsorbent were confirmed by means of SEM, FT-IR, XPS etc. Thus, the cheap-sustainable adsorbents have a superior feature for Cr(VI)-wastewater purification in future., (Copyright © 2018 Elsevier B.V. N 6 -Methyladenosine (m 6 A) is the most frequent post-transcriptional modification in RNA, and it plays a critical role in biological processes. The functions of m 6 A remain largely unexplored due to a lack of highly sensitive methods to quantitatively determine the m 6 A modification fraction at a precise location. Here, we first reveal that T3 DNA ligase has significant selectivity towards the m 6 A modification. On the basis of the new finding, we establish an ultrasensitive quantitation assay for accurately determining m 6 A at one-nucleotide resolution in RNA. With the proposed assay, as low as 4 fM RNA containing m 6 A can be determined and the selectivity is up to 54.1-fold to discriminate m 6 A against unmodified adenosine (A). The sensitivity has been improved about 10 6 -fold so the proposed method can be successfully employed to accurately determine m 6 A in real biological samples, even in low abundance RNA.

The influence of the concentration and type of salt on the physical gelation of gelatin at early stages was explored by polarimeter, rheometer and differential scanning calorimetry (DSC). With the increase of salt concentration, both the triple helix content and the storage modulus of gelatin increased slightly and then decreased in CaCl 2 solutions, whereas they decreased monotonically in NaCl and CrCl 3 solutions. The addition of CaCl 2 at low concentrations facilitated the coil-helix transition of gelatin, which could be significantly inhibited by high salt concentrations. The melting temperature of gels was nearly unchanged with varying salt concentration, but the melting enthalpy decreased monotonically with salt addition. This result indicates that salt additions mainly affect the nucleation of triple helices, but had minor effect on its growth. Introduction: Cervical cancer is the second leading cause of cancer morbidity and mortality for women around the world. Long non-coding RNAs (lncRNAs) have been investigated as a new class of regulators of cellular processes, such as cell growth, apoptosis, and carcinogenesis. Although downregulation of lncRNA GAS5 in several cancers has been studied, its role in cervical cancer remains unknown. The aim of this study is to investigate the expression, clinical significance and biological role in cervical cancer., Methods: Expression of GAS5 was analyzed in cervical cancer tissues by quantitative Real-time PCR (qRT-PCR). And its association with overall survival of patients was analyzed by statistical analysis. Small interfering RNA (siRNA) was used to suppress GAS5 expression in cervical cancer cells. In vitro assays were performed to further explore the biological functions of GAS5 in cervical cancer., Results: We found that GAS5 expression was markedly downregulated in cervical cancer tissues than in corresponding adjacent normal tissues. Decreased GAS5 expression was significantly correlated with FIGO stage, vascular invasion and lymph node metastasis. Moreover, cervical cancer patients with GAS5 lower expression have shown significantly poorer overall survival than those with higher GAS5 expression. And GAS5 expression was an independent prognostic marker of overall survival in a multivariate analysis. In vitro assays our data indicated that knockdown of GAS5 promoted cell proliferation, migration, and invasion., Conclusions: Our study presents that lncRNA GAS5 is a novel molecule involved in cervical cancer progression, which provide a potential prognostic biomarker and therapeutic target.
