Background: Next-generation sequencing (NGS) for Hepatitis B virus (HBV) antiviral resistance (AVR) testing is a highly sensitive diagnostic method, able to detect low-level mutant subpopulations. Our clinical virology laboratory previously transitioned from DNA hybridization (INNO-LiPA) to NGS, initially with the GS Junior System and subsequently the MiSeq. The Oxford Nanopore Technology (ONT) sequencing system was evaluated for HBV resistance testing, with regards to sequencing accuracy and turn-around time., Methods: We performed amplicon sequencing of the HBV polymerase gene from patient plasma and external quality assessment (EQA) samples on the MiSeq Reagent Nano Kit v2 and GridION ONT with R10.4.1 flowcells. Mutational analysis and genotyping were performed by DeepChek®Assay-HBV (version 2.0)., Results: A total of 49 patient samples and 15 EQA samples were tested on both the MiSeq and ONT. There was high agreement for both patient and EQA samples between the MiSeq and ONT systems, with regards to total drug resistance mutations detected and total patient sample agreement, 68/70 (97 %) and 47/49 (96 %), respectively., Conclusion: The ONT NGS platform provided accurate HBV AVR results, with improved turn-around times. Background: Orthopedic manual therapy is currently considered as an alternative approach for treating hip osteoarthritis. However, studies assessing its efficacy in low-income countries in Sub-Saharan Africa are scarce., Objectives: Investigating the effectiveness of Maitland passive joint mobilization (Maitland PJM) compared to self-rehabilitation in patients with hip osteoarthritis in Benin, a low-income country, in Sub-Saharan Africa., Methods: This was a pragmatic, single-blind, two-arm randomized trial involving 66 participants randomly assigned into two groups (Maitland PJM group, n = 33, and self-rehabilitation group, n = 33). Both interventions took 5 weeks. The primary outcome (pain) was evaluated using numerical rating scale. Secondary outcomes (passive hip range of motion, muscles strength, walking, and quality of life) were assessed using goniometry, medical research council, 10 m walk test, and short-form 36. Participants were assessed at baseline, after interventions and 3-month follow-up., Results: Forty-nine patients (Maitland PJM group, n = 22, self-rehabilitation, n = 27) completed the sessions. Within-group analyses showed a significant improvement in body function, walking speed, and quality of life (p < 0.05) after interventions and at 3-month follow-up within both groups. Photoswitchable materials are of significant interest for diverse applications from energy and data storage to additive manufacturing and soft robotics. However, the absorption profile is often a limiting factor for practical applications. This can be overcome using indirect excitation via complementary photophysical pathways, such as triplet sensitisation or photon upconversion. Here, we demonstrate the use of triplet-triplet annihilation upconversion (TTA-UC) to drive photoswitching of the energy storing photoswitch norbornadiene-quadricyclane (NBD-QC) in the solid-state. A photoswitchable bilayer polymer film, incorporating the TTA-UC sensitiser-emitter pair of platinum octaethylporphyrin (PtOEP) and 9,10-diphenylanthracene (DPA), was used to trigger the photoinduced [2+2] cycloaddition of NBD to form QC using visible instead of UV light. The isolated TTA-UC film showed green-to-blue upconversion, with a competitive upconversion efficiency of (1.9 ± 0.1%) for the solid-state in air. Direct photoswitching of the isolated NBD film was demonstrated with a narrow UV light source (340 nm). However, in the bilayer film, spectral overlap between the upconverted blue emission in the TTA-UC film and the absorbance band of the NBD film resulted in indirect photoswitching using visible green light (532 nm, 1 W cm -2 ), thus extending the spectral operational window of the photoswitching film. Energy transfer between electronically coupled photosynthetic light-harvesting antenna pigments is frequently assisted by protein and chromophore nuclear motion. This energy transfer mechanism usually occurs in the weak or intermediate system-bath coupling regime. Redfield theory is frequently used to describe the energy transfer in this regime. Spectral densities describe vibronic coupling in visible transitions of the chromophores and govern energy transfer in the Redfield mechanism. In this work, we perform finely sampled broadband pump-probe spectroscopy on the phycobilisome antenna complex with sub-10-fs pump and probe pulses. The spectral density obtained by Fourier transforming the pump-probe time-domain signal is used to perform modified Redfield rate calculations to check for vibrational enhancement of energy transfer in a coupled chromophore dimer in the C -phycocyanin protein of the phycobilisome antenna. We find two low-frequency vibrations to be in near-resonance with the interexcitonic energy gap and a few-fold enhancement in the interexcitonic energy transfer rate due to these resonances at room temperature. Our observations and calculations explain the fast downhill energy transfer process in C -phycocyanin. We also observe high-frequency vibrations involving chromophore-protein residue interactions in the excited state of the phycocyanobilin chromophore. We suggest that these vibrations lock the chromophore nuclear configuration of the excited state and prevent the energetic relaxation that blocks energy transfer.

In this study, we assessed 43 accessions of sorghum from 16 countries across three continents. Our objective was to identify stomatal and photosynthetic traits that could be exploited in breeding programmes to increase photosynthesis without increasing water use under dynamic light environments. Under field conditions, sorghum crops often have limited water availability and are exposed to rapidly fluctuating light intensities, which influences both photosynthesis and stomatal behaviour. Our results highlight a tight coupling between photosynthetic rate (A) and stomatal conductance (gs) even under dynamic light conditions that results in steady intrinsic water use efficiency (Wi). This was mainly due to rapid stomatal responses, with the majority of sorghum accessions responding within ≤5 min. The maintenance of the ratio of the concentration of CO2 inside the leaf (Ci) and the surrounding atmospheric concentration (Ca) over a large range of accessions suggests high stomatal sensitivity to changes in Ci, that could underlie the rapid gs responses and extremely close relationship between A and gs under both dynamic and steady-state conditions. Therefore, sorghum represents a prime candidate for uncovering the elusive mechanisms that coordinate A and gs, and such information could be used to design crops with high A without incurring significant water losses and eroding Wi., (© The Author(s) 2024. Recent research has shown that optimizing photosynthetic and stomatal traits holds promise for improved crop performance. However, standard phenotyping tools such as gas exchange systems have limited throughput. In this work, a novel approach based on a bespoke gas exchange chamber allowing combined measurement of the quantum yield of PSII (Fq'/Fm'), with an estimation of stomatal conductance via thermal imaging was used to phenotype a range of bread wheat (Triticum aestivum L.) genotypes. Using the dual-imaging methods and traditional approaches, we found broad and significant variation in key traits, including photosynthetic CO2 uptake at saturating light and ambient CO2 concentration (Asat), photosynthetic CO2 uptake at saturating light and elevated CO2 concentration (Amax), the maximum velocity of Rubisco for carboxylation (Vcmax), time for stomatal opening (Ki), and leaf evaporative cooling. Anatomical analysis revealed significant variation in flag leaf adaxial stomatal density. Associations between traits highlighted significant relationships between leaf evaporative cooling, leaf stomatal conductance, and Fq'/Fm', highlighting the importance of stomatal conductance and stomatal rapidity in maintaining optimal leaf temperature for photosynthesis in wheat. Additionally, gsmin and gsmax were positively associated, indicating that potential combinations of preferable traits (i.e. inherently high gsmax, low Ki, and maintained leaf evaporative cooling) are present in wheat. This work highlights the effectiveness of thermal imaging in screening dynamic gs in a panel of wheat genotypes. The synthesis of 9 core-functionalised naphthalene diimide (c-NDI) residues is reported via a 3-step synthesis from naphthalenetetracarboxylic dianhydride using only mechanochemical activation. Selective dibromination and subsequent diimidization were achieved for the first time in a vibratory ball mill, resulting in the key structural intermediate, 2,6-dibromonaphthalenediimide (DBND), which is the basis for elaboration into a multitude of organic electronic materials. Our new synthesis of DBND is achieved in just 5 hours reaction time over two steps compared to typical solution state times of 24 hours. Subsequent Heck-type cross coupling reactions, with a range of styrene residues, produced a series of c-NDIs in good yields. Innovations in light technologies (i.e. Light Emitting Diodes; LED) and cover films with specific optical features (e.g. photo-selective, light-extracting) have revolutionized crop production in both protected environments and open fields. The possibility to modulate the light spectra, thereby enriching/depleting cultivated plants with targeted wavebands has attracted increasing interest from both basic and applicative research. Indeed, the light environment not only influences plant biomass production but is also a pivotal factor in shaping plant size, development and metabolism. In the last decade, the strict interdependence between specific wavebands and the accumulation of targeted secondary metabolites has been exploited to improve the quality of horticultural products. Innovation in LED lighting has also marked the improvement of streetlamp illumination, thereby posing new questions about the possible influence of light pollution on urban tree metabolism. In this case, it is urgent and challenging to propose new, less-impacting solutions by modulating streetlamp spectra in order to preserve the ecosystem services provided by urban trees. The present review critically summarizes the main recent findings related to the morpho-anatomical, physiological, and biochemical changes induced by light spectra management via different techniques in crops as well as in non-cultivated species. This review explores the following topics: (1) plant growth in monochromatic environments, (2) the use of greenhouse light supplementation, (3) the application of covering films with different properties, and (4) the drawbacks of streetlamp illumination on urban trees. Cambodia is a rapidly developing country in south-east Asia, a region forecast as an outsized source of plastic pollution into the world's oceans. However, to date there has been no large-scale assessment of plastic pollution in the environment within the country. We conducted a comprehensive assessment of plastic items and hotspots across 243 coastal, river and inland sites along the entire coastline of Cambodia, recording 46,927 items in transects throughout the study area. Plastic densities along coastlines were more than ten times those on riverbanks and more than twenty times that of inland areas. Averaging 70 plastic items per metre of coastline, Cambodia has among the highest average loads of whole plastic items recorded on coastlines globally. Single-use plastic bottles were the most abundant whole item recorded, overrepresented by drinking water bottles. Polystyrene foam was the most common fragmented item. Accumulation of edible biomass by crop plants relies on maintenance of a high photosynthetic rates across the photoperiod, with assimilation rate ( A ) generally responding to increasing light intensity in a hyperbolic fashion. In natural environments light fluctuates greatly over the course of the day, however in Controlled Environmental Agricultural (CEA) systems, light intensity can be supplemented or precisely controlled using LEDs to create near optimum conditions. In such indoor growth environments light is often delivered as a square wave and recommendations to horticulturalists are given in the form of Daily Light Integrals (DLI). However, this does not take into account the slow photosynthetic induction at the start of the photoperiod and the decline of A towards the end of the photoperiod, which has been demonstrated by several previous studies. Square wave light regimes therefore potentially cause suboptimal photosynthetic utilization of the applied lighting and waste electricity. Here we have adapted light recipes to gradually increase and decrease in intensity to take account of these findings. We demonstrate that, utilising a sinusoidal light regime capped at 250 μmol m -2 s -1 , it is possible to increase edible biomass of rocket (by ca. 20%) compared to square wave delivered at 250 at the same DLI. Additionally, this can be achieved using less electricity (0.6%), therefore reducing energy costs and improving profitability. We suggest that capping maximum light intensity at 250 µmol m -2 s -1 improves the operating efficiency of PSII photochemistry ( Fq'/Fm' ) also known as the photosynthetic efficiency by maintaining A later in the photoperiod. We show that a higher electron transfer rate (ETR) is maintained in these treatments over the photoperiod compared to higher light intensity caps, resulting in a greater Daily Photochemical Integral (DPI). Improving photosynthesis, the fundamental process by which plants convert light energy into chemical energy, is a key area of research with great potential for enhancing sustainable agricultural productivity and addressing global food security challenges. This perspective delves into the latest advancements and approaches aimed at optimizing photosynthetic efficiency. Our discussion encompasses the entire process, beginning with light harvesting and its regulation and progressing through the bottleneck of electron transfer. We then delve into the carbon reactions of photosynthesis, focusing on strategies targeting the enzymes of the Calvin-Benson-Bassham (CBB) cycle. Additionally, we explore methods to increase carbon dioxide (CO2) concentration near the Rubisco, the enzyme responsible for the first step of CBB cycle, drawing inspiration from various photosynthetic organisms, and conclude this section by examining ways to enhance CO2 delivery into leaves. Moving beyond individual processes, we discuss two approaches to identifying key targets for photosynthesis improvement: systems modeling and the study of natural variation. Finally, we revisit some of the strategies mentioned above to provide a holistic view of the improvements, analyzing their impact on nitrogen use efficiency and on canopy photosynthesis., Competing Interests: Conflict of interest statement. Over the last decade, carbon-based metal-free electrocatalysts (C-MFECs) have become important in electrocatalysis. This field is started thanks to the initial discovery that nitrogen atom doped carbon can function as a metal-free electrode in alkaline fuel cells. A wide variety of metal-free carbon nanomaterials, including 0D carbon dots, 1D carbon nanotubes, 2D graphene, and 3D porous carbons, has demonstrated high electrocatalytic performance across a variety of applications. These include clean energy generation and storage, green chemistry, and environmental remediation. The wide applicability of C-MFECs is facilitated by effective synthetic approaches, e.g., heteroatom doping, and physical/chemical modification. These methods enable the creation of catalysts with electrocatalytic properties useful for sustainable energy transformation and storage (e.g., fuel cells, Zn-air batteries, Li-O 2 batteries, dye-sensitized solar cells), green chemical production (e.g., H 2 O 2 , NH 3 , and urea), and environmental remediation (e.g., wastewater treatment, and CO 2 conversion). Furthermore, significant advances in the theoretical study of C-MFECs via advanced computational modeling and machine learning techniques have been achieved, revealing the charge transfer mechanism for rational design and development of highly efficient catalysts. Photon absorption is the first process in light harvesting. Upon absorption, the photon redistributes electrons in the materials to create a Coulombically bound electron-hole pair called an exciton. The exciton subsequently separates into free charges to conclude light harvesting. When two excitons are in each other's proximity, they can interact and undergo a two-particle process called exciton-exciton annihilation. In this process, one electron-hole pair spontaneously recombines: its energy is lost and cannot be harnessed for applications. In this work, we demonstrate the creation of two long-lived excitons on the same chromophore site (biexcitons) at room temperature in a strongly coupled H-aggregated zinc phthalocyanine material. We show that exciton-exciton annihilation is suppressed in these H- aggregated chromophores at fluences many orders of magnitudes higher than solar light. When we chemically connect the same aggregated chromophores to allow exciton diffusion, we observe that exciton-exciton annihilation is switched on. Gas exchange measurements enable mechanistic insights into the processes that underpin carbon and water fluxes in plant leaves which in turn inform understanding of related processes at a range of scales from individual cells to entire ecosytems. Given the importance of photosynthesis for the global climate discussion it is important to (a) foster a basic understanding of the fundamental principles underpinning the experimental methods used by the broad community, and (b) ensure best practice and correct data interpretation within the research community. In this review, we outline the biochemical and biophysical parameters of photosynthesis that can be investigated with gas exchange measurements and we provide step-by-step guidance on how to reliably measure them. We advise on best practices for using gas exchange equipment and highlight potential pitfalls in experimental design and data interpretation. The Supporting Information contains exemplary data sets, experimental protocols and data-modelling routines. Background: Molecular syndromic panels can improve rapidity of results and ease clinical laboratory workflow, although caution has been raised for potential false-positive results. Upon implementation of a new panel for infectious diarrhea (BioFire® FilmArray® Gastrointestinal [GI] Panel, bioMérieux) in our clinical laboratory, a higher than expected number of stool samples with norovirus were detected., Objectives: The goal of this study was to investigate positive percent agreement and the false-positive rate of norovirus detected by the multiplex BioFire GI panel compared to a singleplex commercial assay., Study Design: From October 2023 to January 2024, all prospective stool samples with a positive norovirus result by BioFire had melting curves reviewed manually using the BioFire FilmArray Torch System. Stool samples further underwent testing by a supplementary real-time RT-PCR assay (Xpert® Norovirus, Cepheid) for comparative analysis., Results: Of the 50 stool samples with norovirus detected by BioFire, 18 (36 %) tested negative by Xpert (deemed "false-positives"). Furthermore, melting curve analysis revealed nearly all of these samples had atypical melting curve morphologies for the "Noro-1" target on BioFire (16/18, 89 %), which was statistically significant (Odds Ratio 173.2, 95 % CI [22.2, 5326.9], p < 0.0001). Carbon nanodots (CNDs) are nanosized light-harvesters emerging as next-generation photosensitizers in photocatalytic reactions. Despite their ever-increasing potential applications, the intricacies underlying their photoexcited charge carrier dynamics are yet to be elucidated. In this study, nitrogen-doped graphitic CNDs (NgCNDs) are selectively excited in the presence of methyl viologen (MV 2+ , redox mediator) and different electron donors (EDs), namely ascorbic acid (AA) and ethylenediaminetetraacetic acid (EDTA). The consequent formation of the methyl viologen radical cation (MV •+ ) is investigated, and the excited charge carrier dynamics of the photocatalytic system are understood on a 0.1 ps-1 ms time range, providing spectroscopic evidence of oxidative or reductive quenching mechanisms experienced by optically excited NgCNDs (NgCNDs*) depending on the ED implemented. In the presence of AA, NgCNDs* undergo oxidative quenching by MV 2+ to form MV •+ , which is short-lived due to dehydroascorbic acid, a product of photoinduced hole quenching of oxidized NgCNDs. The EDTA-mediated reductive quenching of NgCNDs* is observed to be at least 2 orders of magnitude slower due to screening by EDTA-MV 2+ complexes, but the MV •+ population is stable due to the irreversibly oxidized EDTA preventing a back reaction. In general, our methodology provides a distinct solution with which to study charge transfer dynamics in photocatalytic systems on an extended time range spanning 10 orders of magnitude. Metal-organic frameworks (MOFs) began to emerge over two decades ago, resulting in the deposition of 120 000 MOF-like structures (and counting) into the Cambridge Structural Database (CSD). Topological analysis is a critical step toward understanding periodic MOF materials, offering insight into the design and synthesis of these crystals via the simplification of connectivity imposed on the complete chemical structure. While some of the most prevalent topologies, such as face-centered cubic ( fcu ), square lattice ( sql ), and diamond ( dia ), are simple and can be easily assigned to structures, MOFs that are built from complex building blocks, with multiple nodes of different symmetry, result in difficult to characterize topological configurations. In these complex structures, representations can easily diverge where the definition of nodes and linkers are blurred, especially for cases where they are not immediately obvious in chemical terms. Currently, researchers have the option to use software such as ToposPro, MOFid, and CrystalNets to aid in the assignment of topology descriptors to new and existing MOFs. These software packages are readily available and are frequently used to simplify original MOF structures into their basic connectivity representations before algorithmically matching these condensed representations to a database of underlying mathematical nets. These approaches often require the use of in-built bond assignment algorithms alongside the simplification and matching rules. Considering the prevalence of ever-changing conditions in the natural world, investigation of photosynthetic responses in C 4 plants under fluctuating light is needed. Here, we studied the effect of dynamic illumination on photosynthesis in totally 10 C 3 , C 3 -C 4 intermediate, C 4 -like and C 4 dicots and monocots at CO 2 concentrations of 400 and 800 μmol mol -1 . C 4 and C 4 -like plants had faster photosynthetic induction and light-induced stomatal dynamics than C 3 plants at 400 μmol mol -1 , but not at 800 μmol mol -1 CO 2 , at which the CO 2 supply rarely limits photosynthesis. C 4 and C 4 -like plants had a higher water use efficiency than C 3 plants at both CO 2 concentrations. There were positive correlations between photosynthetic induction and light-induced stomatal response, together with CO 2 compensation point, which was a parameter of the CO 2 -concentrating mechanism of C 4 photosynthesis. These results clearly show that C 4 photosynthesis in both monocots and dicots adapts to fluctuating light conditions more efficiently than C 3 photosynthesis. Objective: The immunogenic nature of coronavirus disease 2019 (COVID-19) mRNA vaccines led to some initial concern that these could stimulate the HIV reservoir. We analyzed changes in plasma HIV loads (pVL) and reservoir size following COVID-19 mRNA vaccination in 62 people with HIV (PWH) receiving antiretroviral therapy (ART), and analyzed province-wide trends in pVL before and after the mass vaccination campaign., Design: Longitudinal observational cohort and province-wide analysis., Methods: Sixty-two participants were sampled prevaccination, and one month after their first and second COVID-19 immunizations. Vaccine-induced anti-SARS-CoV-2-Spike antibodies in serum were measured using the Roche Elecsys Anti-S assay. HIV reservoirs were quantified using the intact proviral DNA assay; pVL were measured using the cobas 6800 (lower limit of quantification: 20 copies/ml). The province-wide analysis included all 290 401 pVL performed in British Columbia, Canada between 2012 and 2022., Results: Prevaccination, the median intact reservoir size was 77 [interquartile range (IQR): 20-204] HIV copies/million CD4 + T-cells, compared to 74 (IQR: 27-212) and 65 (IQR: 22-174) postfirst and -second dose, respectively (all comparisons P > 0.07). Prevaccination, 82% of participants had pVL <20 copies/ml (max: 110 copies/ml), compared to 79% postfirst dose (max: 183 copies/ml) and 85% postsecond dose (max: 79 copies/ml) ( P  > 0.4). There was no evidence that the magnitude of the vaccine-elicited anti-SARS-CoV-2-Spike immune response influenced pVL nor changes in reservoir size ( P  > 0.6). Uveal melanoma (UM) is an ocular cancer predominantly affecting adults, characterized by challenging diagnostic outcomes. This research endeavors to develop an innovative multifunctional nanocomposite system sensitive to near-infrared (NIR) radiation, serving as both a non-oxygen free-radical generator and a photothermal agent. The designed system combines azobis isobutyl imidazoline hydrochloride (AIBI) with mesoporous copper sulfide (MCuS) nanoparticles. MCuS harnesses NIR laser energy to induce photothermal therapy, converting light energy into heat to destroy cancer cells. Simultaneously, AIBI is activated by the NIR laser to produce alkyl radicals, which induce DNA damage in remaining cancer cells. This distinctive feature equips the designed system to selectively eliminate cancers in the hypoxic tumor microenvironment. MCuS is also beneficial to scavenge the overexpressed glutathione (GSH) in the tumor microenvironment. GSH generally consumes free radicals and hiders the PDT effect. To enhance control over AIBI release in cancer cells, 1-tetradecyl alcohol (TD), a phase-changing material, is introduced onto the surface of MCuS nanoparticles to create the final AMPT nanoparticle system. In vitro and in vivo experiments confirm the remarkable anti-tumor efficacy of AMPT. Objectives: To define consensus entrustable professional activities (EPAs) for neurocritical care (NCC) advanced practice providers (APPs), establish validity evidence for the EPAs, and evaluate factors that inform entrustment expectations of NCC APP supervisors., Design: A three-round modified Delphi consensus process followed by application of the EQual rubric and assessment of generalizability by clinicians not affiliated with academic medical centers., Setting: Electronic surveys., Subjects: NCC APPs ( n = 18) and physicians ( n = 12) in the United States with experience in education scholarship or APP program leadership., Interventions: None., Measurements and Main Results: The steering committee generated an initial list of 61 possible EPAs. The panel proposed 30 additional EPAs. A total of 47 unique nested EPAs were retained by consensus opinion. The steering committee defined six core EPAs addressing medical knowledge, procedural competencies, and communication proficiency which encompassed the nested EPAs. All core EPAs were retained and subsequently met the previously described cut score for quality and structure using the EQual rubric. Most clinicians who were not affiliated with academic medical centers rated each of the six core EPAs as very important or mandatory. Entrustment expectations did not vary by prespecified groups., Conclusions: Expert consensus was used to create EPAs for NCC APPs that reached a predefined quality standard and were important to most clinicians in different practice settings. Four-dimensional printing with embedded photoluminescence is emerging as an exciting area in additive manufacturing. Slim polymer films patterned with three-dimensional lattices of multimode cylindrical waveguides (waveguide-encoded lattices, WELs) with enhanced fields of view can be fabricated by localizing light as self-trapped beams within a photopolymerizable formulation. Luminescent WELs have potential applications as solar cell coatings and smart planar optical components. However, as luminophore-photoinitiator interactions are expected to change the photopolymerization kinetics, the design of robust luminescent photopolymer sols is nontrivial. Here, we use model photopolymer systems based on methacrylate-siloxane and epoxide homopolymers and their blends to investigate the influence of the luminophore Lumogen Violet ( LV ) on the photolysis kinetics of the Omnirad 784 photoinitiator through UV-vis absorbance spectroscopy. Initial rate analysis with different bulk polymers reveals differences in the pseudo-first-order rate constants in the absence and presence of LV , with a notable increase (∼40%) in the photolysis rate for the 1:1 blend. Fluorescence quenching studies, coupled with density functional theory calculations, establish that these differences arise due to electron transfer from the photoexcited LV to the ground-state photoinitiator molecules. We also demonstrate an in situ UV-vis absorbance technique that enables real-time monitoring of both waveguide formation and photoinitiator consumption during the fabrication of WELs. The in situ photolysis kinetics confirm that LV -photoinitiator interactions also influence the photopolymerization process during WEL formation. In 2021, Svante, in collaboration with BASF, reported successful scale up of CALF-20 production, a stable MOF with high capacity for post-combustion CO 2 capture which exhibits remarkable stability towards water. CALF-20's success story in the MOF commercialisation space provides new thinking about appropriate structural and adsorptive metrics important for CO 2 capture. Here, we combine atomistic-level simulations with experiments to study adsorptive properties of CALF-20 and shed light on its flexible crystal structure. We compare measured and predicted CO 2 and water adsorption isotherms and explain the role of water-framework interactions and hydrogen bonding networks in CALF-20's hydrophobic behaviour. Furthermore, regular and enhanced sampling molecular dynamics simulations are performed with both density-functional theory (DFT) and machine learning potentials (MLPs) trained to DFT energies and forces. From these simulations, the effects of adsorption-induced flexibility in CALF-20 are uncovered. Stomatal conductance (g s ) determines CO 2 uptake for photosynthesis (A) and water loss through transpiration, which is essential for evaporative cooling and maintenance of optimal leaf temperature as well as nutrient uptake. Stomata adjust their aperture to maintain an appropriate balance between CO 2 uptake and water loss and are therefore critical to overall plant water status and productivity. Although there is considerable knowledge regarding guard cell (GC) osmoregulation (which drives differences in GC volume and therefore stomatal opening and closing), as well as the various signal transduction pathways that enable GCs to sense and respond to different environmental stimuli, little is known about the signals that coordinate mesophyll demands for CO 2 . Furthermore, chloroplasts are a key feature in GCs of many species, however, their role in stomatal function is unclear and a subject of debate. In this review we explore the current evidence regarding the role of these organelles in stomatal behaviour, including GC electron transport and Calvin-Benson-Bassham (CBB) cycle activity as well as their possible involvement correlating g s and A along with other potential mesophyll signals. Disorders of consciousness are neurological conditions characterized by impaired arousal and awareness of self and environment. Behavioural responses are absent or are present but fluctuate. Disorders of consciousness are commonly encountered as a consequence of both acute and chronic brain injuries, yet reliable epidemiological estimates would require inclusive, operational definitions of the concept, as well as wider knowledge dissemination among involved professionals. Whereas several manifestations have been described, including coma, vegetative state/unresponsive wakefulness syndrome and minimally conscious state, a comprehensive neurobiological definition for disorders of consciousness is still lacking. The scientific literature is primarily observational, and studies-specific aetiologies lead to disorders of consciousness. Despite advances in these disease-related forms, there remains uncertainty about whether disorders of consciousness are a disease-agnostic unitary entity with a common mechanism, prognosis or treatment response paradigm. Our knowledge of disorders of consciousness has also been hampered by heterogeneity of study designs, variables, and outcomes, leading to results that are not comparable for evidence synthesis. The different backgrounds of professionals caring for patients with disorders of consciousness and the different goals at different stages of care could partly explain this variability. The Prospective Studies working group of the Neurocritical Care Society Curing Coma Campaign was established to create a platform for observational studies and future clinical trials on disorders of consciousness and coma across the continuum of care. In this narrative review, the author panel presents limitations of prior observational clinical research and outlines practical considerations for future investigations. A narrative review format was selected to ensure that the full breadth of study design considerations could be addressed and to facilitate a future consensus-based statement (e.g. via a modified Delphi) and series of recommendations. The panel convened weekly online meetings from October 2021 to December 2022. Research considerations addressed the nosographic status of disorders of consciousness, case ascertainment and verification, selection of dependent variables, choice of covariates and measurement and analysis of outcomes and covariates, aiming to promote more homogeneous designs and practices in future observational studies. The goal of this review is to inform a broad community of professionals with different backgrounds and clinical interests to address the methodological challenges imposed by the transition of care from acute to chronic stages and to streamline data gathering for patients with disorders of consciousness. Background: Multiplex real-time RT-PCR assays for respiratory pathogens are valuable tools to optimize laboratory workflow and turnaround time. At a time when resurgence of influenza and respiratory syncytial virus (RSV) cases have been widely observed along with continued transmission of SARS-CoV-2, timely identification of all circulating respiratory viruses is crucial. This study evaluates the detection of low viral loads of SARS-CoV-2 by four multiplex molecular assays: Roche cobas 6800/8800 SARS-CoV-2 & Influenza A/B Test, Cepheid Xpert Xpress SARS-CoV-2/Flu/RSV, cobas Liat SARS-CoV-2 & Influenza A/B, and a laboratory-developed test (LDT)., Methods: Retrospective upper respiratory tract specimens positive for various respiratory viruses at a range of cycle threshold (Ct) values (18-40) were tested by four multiplex assays. Positive and negative percent agreement (PPA and NPA) with validated RT-PCR assays were calculated., Results: A total of 82 samples were assessed, with discordant results observed in a portion of the samples (10/82, 12.2%) where Ct values were >33. The majority of the discordant results (6/10, 60%) were false negatives. Overall, PPA was 100% (58/58) for cobas 6800, 97.4% (38/39) for GeneXpert, 100% (17/17) for Liat, and 90.5% (57/63) for the LDT. PPA for the LDT increased to 92.1% after manual review of amplification curves., Conclusions: Commercial multiplex respiratory virus assays have good performance for samples with medium to high viral loads (Ct values <33). Hyperspectral imaging is a remote sensing technique that enables remote, noninvasive measurement of plant traits. Here, we outline the procedures for camera setup, scanning, and calibration, along with the acquisition of black and white reference materials, which are the key steps in collecting hyperspectral imagery. We also discuss the development of predictive models such as partial least-squares regression, using both large and small datasets, which are used to predict plant traits from hyperspectral data. To ensure practical applicability, we provide code examples that allow readers to immediately implement these techniques in real-world scenarios. Measurements of in vivo photosynthesis are powerful tools that probe the largest fluxes of carbon and energy in an illuminated leaf, but often the specific techniques used are so varied and specialized that it is difficult for researchers outside the field to select and perform the most useful assays for their research questions. The goal of this chapter is to provide a broad overview of the current tools available for the study of photosynthesis, both in vivo and in vitro, so as to provide a foundation for selecting appropriate techniques, many of which are presented in detail in subsequent chapters. This chapter will also organize current methods into a comparative framework and provide examples of how they have been applied to research questions of broad agronomical, ecological, or biological importance. This chapter closes with an argument that the future of in vivo measurements of photosynthesis lies in the ability to use multiple methods simultaneously and discusses the benefits of this approach to currently open physiological questions. Introduction. BK polyomavirus (BKPyV) quantitative testing is an important screening tool post-transplantation, although interpretation can be challenging due to lack of standardization, assay heterogeneity and variability of BKPyV DNA over time (in urine). Methods. Remnant clinical EDTA plasma and urine samples were tested by the cobas BKV test and a validated laboratory-developed test (LDT). Accuracy [positive and negative percent agreement (PPA and NPA), Pearson's correlation, Bland-Altman analysis] and reproducibility were evaluated. To assess BKPyV DNA stability in urine, prospective urine samples were maintained at two different storage temperatures and tested in triplicate over 7 days. Results. Overall PPA was 95.6 % (43/45) and NPA was 94.4 % (170/180). For plasma, Pearson's correlation (0.950) and Bland-Altman analysis (0.113±0.22 log 10 IU ml -1 ) showed high agreement. For neat urine, Pearson's correlation (0.842) and Bland-Altman analysis (0.326±0.80 log 10 IU ml -1 ) showed somewhat higher variability. Reproducibility was high for the cobas BKV versus the LDT. BKPyV DNA levels in neat urine remained relatively stable over 7 days at both storage temperatures, although outlier results were intermittently detected. Conclusion. The cobas BKV test showed high agreement and reproducibility compared to the reference LDT. BKPyV viral load testing in urine has known limitations, but neat urine can be processed by the cobas BKV.

Stomata can be distributed exclusively on the abaxial or adaxial leaf surface, but they are most commonly found on both leaf surfaces. Variations in stomatal arrangement, patterning, and the impact on photosynthesis can be measured using an infrared gas exchange system. However, when using standard gas exchange techniques, both surfaces are measured together and averaged to provide leaf-level values. Employing an innovative gas exchange apparatus with two infrared gas analyzers, separate gaseous flux from both leaf surfaces can be quantified simultaneously and independently. Photosynthesis is increasingly becoming a recognized target for crop improvement. Phenotyping photosynthesis-related traits on field-grown material is a key bottleneck to progress here due to logistical barriers and short measurement days. Many studies attempt to overcome these challenges by phenotyping excised leaf material in the laboratory. To date there are no demonstrated examples of the representative nature of photosynthesis measurements performed on excised leaves relative to attached leaves in crops. Here, we tested whether standardized leaf excision on the day prior to phenotyping affected a range of common photosynthesis-related traits across crop functional types using tomato (C3 dicot), barley (C3 monocot), and maize (C4 monocot). Potentially constraining aspects of leaf physiology that could be predicted to impair photosynthesis in excised leaves, namely leaf water potential and abscisic acid accumulation, were not different between attached and excised leaves. We also observed non-significant differences in spectral reflectance and chlorophyll fluorescence traits between the treatments across the three species. However, we did observe some significant differences between traits associated with gas exchange and photosynthetic capacity across all three species. Augmented reality (AR) is an emerging technique used to improve visualization and comprehension of complex 3D materials. This approach has been applied not only in the field of chemistry but also in real estate, physics, mechanical engineering, and many other areas. Here, we demonstrate the workflow for an app-free AR technique for visualization of metal-organic frameworks (MOFs) and other porous materials to investigate their crystal structures, topology, and gas adsorption sites. We think this workflow will serve as an additional tool for computational and experimental scientists working in the field for both research and educational purposes.

This work introduces a novel multifunctional system called UPIPF (upconversion-polydopamine-indocyanine-polyethylene-folic) for upconversion luminescent (UCL) imaging of cancer cells using near-infrared (NIR) illumination. The system demonstrates efficient inhibition of human hepatoma (HepG2) cancer cells through a combination of NIR-triggered photodynamic therapy (PDT) and enhanced photothermal therapy (PTT). Initially, upconversion nanoparticles (UCNP) are synthesized using a simple thermal decomposition method. To improve their biocompatibility and aqueous dispersibility, polydopamine (PDA) is introduced to the UCNP via a ligand exchange technique. Indocyanine green (ICG) molecules are electrostatically attached to the surface of the UCNP-polydopamine (UCNP@PDAs) complex to enhance the PDT and PTT effects. Moreover, polyethylene glycol (PEG)-modified folic acid (FA) is incorporated into the UCNP-polydopamine-indocyanine-green (UCNP@PDA-ICGs) nanoparticles to enhance their targeting capability against cancer cells. The excellent UCL properties of these UCNP enable the final UCNP@PDA-ICG-PEG-FA nanoparticles (referred to as UPIPF) to serve as a potential candidate for efficient anticancer drug delivery, real-time imaging, and early diagnosis of cancer cells. Furthermore, the UPIPF system exhibits PDT-assisted PTT effects, providing a convenient approach for efficient cancer cell inhibition (more than 99% of cells are killed). Optical monitoring and screening of photocatalytic batch reactions using cuvettes ex situ is time-consuming, requires substantial amounts of samples, and does not allow the analysis of species with low extinction coefficients. Hollow-core photonic crystal fibers (HC-PCFs) provide an innovative approach for in situ reaction detection using ultraviolet-visible absorption spectroscopy, with the potential for high-throughput automation using extremely low sample volumes with high sensitivity for monitoring of the analyte. HC-PCFs use interference effects to guide light at the center of a microfluidic channel and use this to enhance detection sensitivity. They open the possibility of comprehensively studying photocatalysts to extract structure-activity relationships, which is unfeasible with similar reaction volume, time, and sensitivity in cuvettes. Here, we demonstrate the use of HC-PCF microreactors for the screening of the electron transfer properties of carbon dots (CDs), a nanometer-sized material that is emerging as a homogeneous light absorber in photocatalysis. The CD-driven photoreduction reaction of viologens (XV 2+ ) to the corresponding radical monocation XV •+ is monitored in situ as a model reaction, using a sample volume of 1 μL per measurement and with a detectability of <1 μM. A range of different reaction conditions have been systematically studied, including different types of CDs ( i.e. , amorphous, graphitic, and graphitic nitrogen-doped CDs), surface chemistry, viologens, and electron donors. Furthermore, the excitation irradiance was varied to study its effect on the photoreduction rate. The findings are correlated with the electron transfer properties of CDs based on their electronic structure characterized by soft X-ray absorption spectroscopy.
