Background: The impact of Prader-Willi syndrome (PWS) domain gene expression on the growth of healthy children is not well understood. This study investigated associations between PWS domain gene expression in umbilical cord tissue and prenatal and postnatal growth, considering potential sex differences., Methods: Relative gene expression of paternally expressed MAGEL2, NDN, and SNURF-SNRPN, and the small nucleolar RNAs SNORD116 and SNORD115 were determined by real-time quantitative polymerase chain reaction in umbilical cord tissue from 122 healthy newborns (59 girls and 63 boys). Gene expression levels were correlated with auxological measures at birth, infancy, and childhood (ages 2, 4, and 6 years)., Results: MAGEL2, NDN, SNORD116, and SNORD115 expression in the umbilical cord was negatively associated with birth weight, length, and placental weight (P < 0.001). Postnatally, these genes were positively associated with weight and length at 3 months (P < 0.001) and weight gain from birth to ages 1, 2, and 4 years (P < 0.01). Negative associations at birth were stronger in girls (P < 0.001), while positive associations during infancy and childhood were stronger in boys (P < 0.001). MAGEL2, SNORD116, and SNORD115 expression predicted early-postnatal growth, explaining the higher growth rate in boys compared to girls and accounting for sex differences up to 1.5 kg in weight and 3 cm in height during infancy., Conclusions: Paternally expressed PWS domain gene expression in the umbilical cord was negatively associated with prenatal growth and positively with early-postnatal growth in healthy infants. This gene expression may predict early human postnatal growth and promote the well-known sex dimorphism in growth. Santolina chamaecyparissus L. (cotton-lavender) is receiving increasing attention due to its potential for modern medicine and is considered both a functional food and nutraceutical. In this work, the phytochemical profile of its flower hydromethanolic extract was investigated by gas chromatography-mass spectrometry, and its applications as a biorational for crop protection were explored against Neocosmospora spp., both in vitro and in planta. The phytochemical profiling analysis identified several terpene groups. Among sesquiterpenoids, which constituted the major fraction (50.4%), compounds featuring cedrane skeleton (8-cedren-13-ol), aromadendrene skeleton (such as (-)-spathulenol, ledol, alloaromadendrene oxide, epiglobulol, and alloaromadendrene), hydroazulene skeleton (ledene oxide, isoledene, and 1,2,3,3a,8,8a-hexahydro-2,2,8-trimethyl-,(3a α ,8 β ,8a α )-5,6-azulenedimethanol), or copaane skeleton ( cis -α-copaene-8-ol) were predominant. Additional sesquiterpenoids included longiborneol and longifolene. The monoterpenoid fraction (1.51%) was represented by eucalyptol, (+)-4-carene, endoborneol, and 7-norbornenol. In vitro tests against N. falciformis and N. keratoplastica , two emerging soil phytopathogens, resulted in effective concentration EC 90 values of 984.4 and 728.6 μg·mL -1 , respectively. A higher dose (3000 μg·mL -1 ) was nonetheless required to achieve full protection in the in planta tests conducted on zucchini ( Cucurbita pepo L.) cv. 'Diamant F1' and tomato ( Solanum lycopersicum L.) cv. 'Optima F1' plants inoculated with N. falciformis by root dipping. The reported data indicate an antimicrobial activity comparable to that of fosetyl-Al and higher than that of azoxystrobin conventional fungicides, thus making the flower extract a promising bioactive product for organic farming and expanding S. chamaecyparissus potential applications.

This study presents a novel system for diagnosing and evaluating soccer performance using wearable inertial sensors integrated into players' insoles. Designed to meet the needs of professional podiatrists and sports practitioners, the system focuses on three key soccer-related movements: passing, shooting, and changes of direction (CoDs). The system leverages low-power IMU sensors, Bluetooth Low Energy (BLE) communication, and a cloud-based architecture to enable real-time data analysis and performance feedback. Data were collected from nine professional players from the SD Huesca women's team during controlled tests, and bespoke algorithms were developed to process kinematic data for precise event detection. Results indicate high accuracy rates for detecting ball-striking events and CoDs, with improvements in algorithm performance achieved through adaptive thresholds and ensemble neural network models. Compared to existing systems, this approach significantly reduces costs and enhances practicality by minimizing the number of sensors required while ensuring real-time evaluation capabilities. However, the study is limited by a small sample size, which restricts generalizability. Future research will aim to expand the dataset, include diverse sports, and integrate additional sensors for broader applications. This system offers a valuable tool for injury prevention, player rehabilitation, and performance optimization in professional soccer, bridging technical advancements with practical applications in sports science.

Lysosomes are subcellular compartments characterised by an acidic pH, containing an ample variety of acid hydrolases involved in the recycling of biopolymers. Among these hydrolases, lysosomal proteases have merely been considered as end-destination proteases responsible for the digestion of waste proteins, trafficked to the lysosomal compartment through autophagy and endocytosis. However, recent reports have started to unravel specific roles for these proteases in the regulation of initially unexpected biological processes, both under physiological and pathological conditions. Furthermore, some lysosomal proteases are no longer restricted to the lysosomal compartment, as more novel non-canonical, extralysosomal targets are being identified. Currently, lysosomal proteases are accepted to play key functions in the extracellular milieu, attached to the plasma membrane and even in the cytosolic and nuclear compartments of the cell. Under physiological conditions, lysosomal proteases, through non-canonical, extralysosomal activities, have been linked to cell differentiation, regulation of gene expression, and cell division. Under pathological conditions, these proteases have been linked to cancer, mostly through their extralysosomal activities in the cytosol and nuclei of cells. In this review, we aim to provide a comprehensive summary of our current knowledge about the extralysosomal, non-canonical functions of lysosomal proteases, both under physiological and pathological conditions, with a particular interest in cancer, that could potentially offer new opportunities for clinical intervention.

Malaria remains a significant health threat in tropical and subtropical regions. The immune response to Plasmodium falciparum involves both humoral and cellular components, including phagocytosis by neutrophils. However, observing phagocytosis through light microscopy is uncommon. This report presents a rare case of falciparum malaria in a Ghanaian twin, highlighting intracellular P. falciparum forms in neutrophils. Two 7-year-old male twins from Ghana were admitted with fever, vomiting, and abdominal pain. They tested positive for P. falciparum infection via rapid diagnostic tests and blood smears. Child 1 displayed significant neutrophilia and intracellular parasites, including trophozoites, merozoites, and a potential schizont, within neutrophils. Child 2 showed no phagocytosis but exhibited other signs of systemic inflammation and hemolysis. Both children were treated successfully with piperaquine tetraphosphate/artenimol (Eurartesim®). This case provides rare microscopic evidence of P. falciparum phagocytosis by neutrophils, suggesting prior antibody development in the patient. While the findings highlight the role of phagocytosis in parasite clearance, the absence of similar observations in the second twin raises questions about individual immune responses and genetic factors. UV radiation is the main etiological agent of skin cancer. Other factors such as pollution, diet and lifestyle are also contributing factors. Our aim was to investigate the association between melanoma and exposome factors in the Spanish population with a prospective multicenter case-control study. Case group included patients with melanoma while the control group included people who attended the consultations as companions without a past medical history of skin cancer. A total of 73 melanoma patients and 126 controls were included. The former group included more outdoor workers, a history of skin cancer, drug use (acetylsalicylic acid, antidepressants and ACE inhibitors, p<0.05), more sun exposure (p<0.001) and more sunburns (p=0.04). Controls used shade (p=0.04) or clothing (p<0.001) and the sun protection factor (SPF) used 15 years earlier was higher (p=0.04). Melanoma-related exposome factors are associated with sun exposure, drug intake and food. Prevention strategies should target specific populations, such as outdoor workers by promoting sun-safe behaviors and healthy lifestyle habits since childhood., (Copyright © 2024 AEDV. Purpose: The purpose of this study is to examine the long-term effects of a programme called Ser Mayor, un RETO («Being Older, a Challenge») aimed at improving quality of life in older adults over 60 living in the community., Methods: This programme used co-design principles to create sustainable opportunities to improve quality of life, connecting with the community, and empowering older adults to take responsibility for their own health. Ser Mayor, un RETO was based on Bronfenbrenner's bio-social-ecological perspective and the health assets model. Its design was complemented with leisure education and physical activity promotion-based guidelines, identified in the scientific literature. The study design was quasi-experimental, with control group, and was conducted over a period of 20-months (September 2017 to June 2019) in several municipalities near the city of Zaragoza (Spain), with the participation of 213 older adults (n=120 experimental group and n=93 control group). Data were analysed using generalised linear mixed models controlling for age and gender of participants and accounting for initial differences between groups in the study outcomes., Results: The results indicate that the experimental group experienced an improvement in levels of quality of life relative to the control group, which was sustained over the study period, especially in several physical dimensions of quality of life: physical function (P=.014), physical role (P=.027), physical component summary (P=.022) and in emotional role (P=.033)., Conclusion: The Ser Mayor, un RETO programme had a positive effect on the quality of life of older adults, showing potential for sustainability of its effects and for the service providers to develop a sense of ownership over the programme., (Copyright © 2024 SEGG. Introduction: Patient blood management (PBM) adherence in clinical practice is unclear. This real-world practice study assessed the management of patients with gastrointestinal (GI) bleeding after the implementation of the PBM strategy., Methods: This was a nationwide multicenter and prospective study involving consecutive adults with GI bleeding between March 2019 and March 2021. Patients were examined according to hemoglobin (Hb) level at admission (<7 g/dl, n  = 93; 7-8 g/dl, n  = 47; 8-9 g/dl, n  = 61; and >9 g/dl, n  = 249). Study outcomes measures were morbidity and mortality during hospitalization and at 3- and 6-month follow-up. Appropriate anemia or iron deficiency management was considered when adherence to PBM policy was higher than 75%., Results: A total of 450 patients (57.6% men, median age: 74 years, interquartile range: 63-82) were included. Overall, 55.1% and 59.3% of patients received transfusion and iron supplementation, respectively. The rates of appropriate transfusion and iron supplementation adherence were 90.9% (range: 86.9-93.5%, P  = 0.109) and 81.8% (range: 78.5-85.1%, P  = 0.041), depending on Hb level, respectively. No associations were observed between adherence to the PBM strategy and length of stay ( P  = 0.263) or risk of further bleeding ( P  = 0.742). Patients who were transfused [hazard ratio (HR): 0.79, 95% confidence interval (CI): 0.27-0.85] and iron supplemented (HR: 0.91, 95% CI: 0.38-1.41), however, appropriately achieved a lower risk of death. Age (HR: 1.12, 95% CI: 1.12-1.25) and further bleeding (HR: 39.08, 95% CI: 4.01-181.24) were poor prognostic factors. No serious adverse events were reported., Conclusions: In this nationwide study, there is a high level of adherence and safety of PBM for the treatment of GI bleeding. Aim: The aim was to study the effect of two low-calorie, high-protein (HP) diets, with most of the protein coming from animal or plant sources, on glycaemic and other cardiometabolic outcomes in subjects with overweight or obesity and glucose metabolism disorders., Materials and Methods: A total of 117 participants aged >18 years with body mass index over 27.5 kg/m 2 and prediabetes or type 2 diabetes mellitus (T2DM) were randomized to one of two HP low-calorie diets (35% of total calories from protein), in which 75% of the protein was from either plant-based sources (HPP) or animal sources (HPA). For both diets, 30% and 35% of the total calories were from fat and carbohydrates, respectively. The dietary intervention lasted 6 months., Results: Both diets improved body composition to a similar extent, including weight loss (-8.05 ± 5.12 kg for the HPA diet and -7.70 ± 5.47 kg for the HPP diet at 6 months) and fat mass, mainly visceral fat. Both diets had a similar beneficial effect on glucose metabolism, including fasting glucose, insulin, homeostasis model assessment of insulin resistance index and glycated haemoglobin. Other biochemical parameters, including lipid profiles, liver enzymes, adipokines and inflammatory biomarkers, similarly improved in both groups. Harsh early environmental conditions can exert delayed, long-lasting effects on phenotypes, including reproductive traits such as sexual signals. Indeed, adverse early conditions can accelerate development, increasing oxidative stress that may, in turn, impact adult sexual signals. Among signals, colorations produced by red ketocarotenoids seem to depend on mitochondrial functioning. Hence, they could reveal individual cell respiration efficiency. It has been hypothesized that these traits are unfalsifiable "index" signals of condition due to their deep connection to individual metabolism. Since mitochondrial dysfunction is frequently linked to aging, red ketocarotenoid-based ornaments could also be good signals of a critical fitness component: longevity. We tested this red color per longevity correlation in captive zebra finches. In addition, we experimentally decreased the synthesis of glutathione (a critical intracellular antioxidant) during the first days of the birds' life to resemble harsh early environmental conditions (e.g., undernutrition). Longevity was recorded until the death of the last bird (almost 9 years). Males, but not females, exhibiting a redder bill in early adulthood lived longer than males with paler bills, which agrees with some precedent studies. However, such bill redness-longevity connection was absent among males with inhibited glutathione synthesis. These findings may suggest that environmental factors can alter the reliability of red ketocarotenoid-based sexual signals, making them less unfalsifiable than believed., (© 2024 The Author(s). Introduction: Crohn's disease (CD) varies by location, potentially affecting therapy efficacy and surgery risk, although research on this topic is conflicting. This study aims to investigate the independent association between CD location and therapeutic patterns., Methods: We analyzed patients with CD diagnosed from January 2005 to May 2023 registered in the nationwide ENEIDA registry. A univariate Cox regression analysis assessed the association of disease location with biologic use and persistence (with treatment discontinuation as a failure event), as well as the use of intestinal resections. A multivariate model was constructed to evaluate the independent association of disease location with therapeutic patterns, controlling for potential confounders such as sex, age at inclusion and diagnosis, disease duration and behavior, previous surgery or biological therapy, extraintestinal manifestations, and perianal disease., Results: The study included 17,292 patients with a median follow-up period of 6 years (interquartile range 2-10 years). Ileocolonic location was associated with a higher biologic use than colonic location (hazard ratio [HR] 1.30, 95% confidence interval [CI] 1.22-1.38) and ileal disease (HR 1.21, 95% CI 1.16-1.27), independently predicting biologic use (P < 0.001). Ileal location was associated with a lower biologic persistence than ileocolonic location (HR 1.14, 95% CI 1.07-1.21) and colonic disease (HR 1.10, 95% CI 1.01-1.20), independently predicting biologic persistence (P = 0.019). Ileal disease was associated with a higher likelihood of intestinal resections than colonic (HR 2.82, 95% CI 2.45-3.25) and ileocolonic location (HR 1.13, 95% CI 1.05-1.22), independently predicting the use of surgery (P < 0.001)., Discussion: CD location with ileal predominance is associated with a distinct therapeutic pattern, including higher biologic use, lower treatment persistence, and increased rates of intestinal resections., (Copyright © 2024 by The American College of Gastroenterology.)

Treatment options for viral infections are limited and viruses have proven adept at evolving resistance to many existing therapies, highlighting a significant vulnerability in our defenses. In response to this challenge, we explored the modulation of cellular RNA metabolic processes as an alternative paradigm to antiviral development. Previously, the small molecule 5342191 was identified as a potent inhibitor of HIV-1 replication by altering viral RNA accumulation at doses that minimally affect host gene expression. In this report, we document 5342191 as a potent inhibitor of adenovirus, coronavirus, and influenza replication. In each case, 5342191-mediated reduction in virus replication was associated with altered viral RNA accumulation and loss of viral structural protein expression. Interestingly, while resistant viruses were rapidly isolated for compounds targeting either virus-encoded proteases or polymerases, we have not yet isolated 5342191-resistant variants of coronavirus or influenza. As with HIV-1, 5342191's inhibition of coronaviruses and influenza is mediated through the activation of specific cell signaling networks, including GPCR and/or MAPK signaling pathways that ultimately affect SR kinase expression. Together, these studies highlight the therapeutic potential of compounds that target cellular processes essential for the replication of multiple viruses. Not only do these compounds hold promise as broad-spectrum antivirals, but they also offer the potential of greater resilience in combating viral infections.

Plant-plant interactions are major determinants of the dynamics of terrestrial ecosystems. There is a long tradition in the study of these interactions, their mechanisms and their consequences using experimental, observational and theoretical approaches. Empirical studies overwhelmingly focus at the level of species pairs or small sets of species. Although empirical data on these interactions at the community level are scarce, such studies have gained pace in the last decade. Studying plant-plant interactions at the community level requires knowledge of which species interact with which others, so an ecological networks approach must be incorporated into the basic toolbox of plant community ecology. The concept of recruitment networks (RNs) provides an integrative framework and new insights for many topics in the field of plant community ecology. RNs synthesise the set of canopy-recruit interactions in a local plant assemblage. Canopy-recruit interactions describe which ("canopy") species allow the recruitment of other species in their vicinity and how. Here we critically review basic concepts of ecological network theory as they apply to RNs. We use RecruitNet, a recently published worldwide data set of canopy-recruit interactions, to describe RN patterns emerging at the interaction, species, and community levels, and relate them to different abiotic gradients. Our results show that RNs can be sampled with high accuracy. The studies included in RecruitNet show a very high mean network completeness (95%), indicating that undetected canopy-recruit pairs must be few and occur very infrequently. Across 351,064 canopy-recruit pairs analysed, the effect of the interaction on recruitment was neutral in an average of 69% of the interactions per community, but the remaining interactions were positive (i.e. facilitative) five times more often than negative (i.e. competitive), and positive interactions had twice the strength of negative ones. Moreover, the frequency and strength of facilitation increases along a climatic aridity gradient worldwide, so the demography of plant communities is increasingly strongly dependent on facilitation as aridity increases. At network level, species can be ascribed to four functional types depending on their position in the network: core, satellite, strict transients and disturbance-dependent transients. This functional structure can allow a rough estimation of which species are more likely to persist. In RecruitNet communities, this functional structure most often departs from random null model expectation and could allow on average the persistence of 77% of the species in a local community. The functional structure of RNs also varies along the aridity gradient, but differently in shrubland than in forest communities. This variation suggests an increase in the probability of species persistence with aridity in forests, while such probability remains roughly constant along the gradient in shrublands. The different functional structure of RNs between forests and shrublands could contribute to explaining their co-occurrence as alternative stable states of the vegetation under the same climatic conditions. Background: Little information is available on the embryology of the structures that connect the lateral meniscus to its nearby structures (proximal tibia, fibular head, and popliteus tendon), which restrict lateral meniscal extrusion., Purpose: To describe the menisco-tibio-popliteus-fibular complex (MTPFC)-conformed by the lateral meniscotibial ligament (LMTL), popliteofibular ligament, meniscofibular ligament, and the 2 popliteomeniscal ligaments (superior and inferior)-and anterolateral ligament (ALL) of the knee in human embryos/fetuses from weeks 9 to 37 of gestation., Study Design: Descriptive laboratory study., Methods: The authors analyzed 36 formalin-fixed embryos/fetus knees, ranging between weeks 9 and 37 of gestation. All had normal external morphological characteristics. A microdissection was performed by meticulously searching the structures of the MTPFC and the ALL, their anatomic relationships, and their appearance over time. Histological sections were taken of the entire knee, the lateral meniscus, the ALL, and the LMTL to observe its structure from its early stages, junctions, and cellularity. In addition, the composition and orientation of the fibers were evaluated., Results: The MTPFC was not found in the youngest knees (9 weeks). The ligaments of this complex could not be seen until week 11 of development. Moreover, the ALL was a consistent finding from the 13th week of development onward. All the ligaments of the MTPFC were seen between developmental weeks 11 and 16 in all the specimens. The histological results showed that the histology of the ALL showed dense connective tissue organized into thick bundles of collagen fibers in its central portion, with a parallel and undulating arrangement and the anterolateral capsule demonstrated 3 easily distinguishable layers corresponding to the fibrous membrane, the subsynovial membrane formed by loose connective tissue, and the synovial membrane., Conclusion: All the structures of the MTPFC were visualized from week 11, with the LMTL being the first to form. Moreover, the ALL was a consistent finding from week 13 of development onward., Clinical Relevance: Understanding the embryological development of the MTPFC and the ALL of the knee is clinically relevant, as it can provide crucial insights into the pathogenesis of knee conditions and inform treatment strategies. Pulmonary aspiration during anaesthesia induction is a serious adverse event that can lead to catastrophic consequences for the patient, including death. Preoperative fasting has so been assessed on the basis of the clinical history and fasting schedules recommended by clinical guidelines. This assessment is not objective, since the presence or absence of gastric contents cannot be guaranteed. Gastric point of care ultrasound (POCUS) is an objective, simple, and rapid method for assessing fasting prior to anaesthesia induction that stratifies risk and aids in decision-making. The aim of this review is to summarize the existing literature on gastric POCUS in the assessment of preoperative fasting. The objective of this study was to evaluate the impact of dietary carob pulp (Cp, Ceratonia siliqua L.) and vitamin E (Vit E) on the quality and shelf-life of light lamb meat stored for a maximum of 15 days under modified atmosphere packaging (MAP). Forty-eight lambs (half females and half entire males, 25.3 ± 0.3 kg of body weight and 83 ± 5.8 days old) were randomly selected and slaughtered after an experimental fattening period with a 2 × 2 factorial design: 0 vs. 20% Cp and 40 vs. 300 IU of Vit E/kg of diet for 41 days. The carcass traits, fatty acid (FA) profile, antioxidant content, colour, lipid oxidation, and microbial count in the meat were evaluated. Scarce interactions between Cp and Vit E were observed on most of the variables studied. The dietary inclusion of Cp did not affect carcass traits ( p > 0.05) but produced minor changes in the content of branched FA and reduced the α-tocopherol content in the meat ( p < 0.05), while no impact was observed on discolouration or lipid oxidation ( p > 0.05). High Vit E supplementation increased the yellowness of caudal fat and the α-tocopherol content, which limited discolouration and lipid oxidation ( p < 0.05). Furthermore, the High Vit E diet (without Cp) controlled the psychrotrophic bacterial count after 15 days of storage. At slaughter, males were heavier than females but had lower carcass dressing ( p < 0.05). The lambs' sex had minor impacts on the quality and shelf-life of meat. This study demonstrated that there is no synergistic interaction between Cp and Vit E on lamb meat quality and shelf-life. Including 20% of Cp in lamb diets is feasible without impairing meat quality. Nevertheless, supplementation with 300 IU of Vit E/kg of feed is essential to extend the shelf-life of meat stored under MAP.

Crude protein (CP) reduction and amino acid (AAs) supplementation is an interesting cost-reducing strategy for environmental compliance. The objective of this work is to study productive performances of heavy Duroc X (Landrace X Large white) male and female pigs during the finishing stage under a 2% CP reduction with crystalline AA supplementation. A total of 60 entire male and 60 female pigs (means ± standard deviations: 109 ± 4.9 days of age; 67.9 ± 7.18 kg of body weight) were distributed in split-sex pens (6 pigs/pen) and fed ad libitum. Two isoenergetic diets were provided (145 and 125 g CP/kg with AA supplementation, control and low-CP) during the final 42 days of finishing period. Growth performance, carcass and meat traits were evaluated. The fixed effects of dietary treatment, sex and their interaction were considered and simple least squares models used. Males were 7.25% heavier ( p < 0.001) and had 16% higher average daily gains ( p < 0.0001) than females in both treatments with no differences in feed intake or feed conversion ratio ( p > 0.05). Nitrogen efficiency was 35% higher in low-CP males. Little difference was found for most carcass and meat traits between treatments. Males had 7-8% higher loin subcutaneous fat thickness in both dietary treatments ( p = 0.011). Males and females of the low-CP diet showed higher fat and saturated fat contents than controls ( p = 0.020). A 20 g/kg CP reduction with AA supplementation is an efficient strategy during the finishing phase without productivity losses in heavy Duroc sire line genotypes of both sexes.

Background: Photodynamic therapy (PDT) is a widely-used non-surgical treatment for non-melanoma skin cancers, including basal cell carcinoma (BCC), actinic keratoses (AK), and Bowen's disease (BD). PDT has high success rates, but various factors, can influence treatment response. This study investigates the clinical, histological, and molecular factors that affect the efficacy of methyl aminolevulinate PDT (MAL-PDT) for BCC and BD., Methods and Patients: Prospective observational multicentric study performed between May 2019 and January 2021 with 64 patients included. Clinical data such as tumor thickness, location, and histological subtype were recorded. Immunohistochemical analysis was performed on tumor samples to assess the expression of biomarkers including p53, β-catenin, and GLUT1., Results: Tumor thickness was found to be a critical determinant of MAL-PDT response, with thicker nodular BCCs showing reduced response rates compared to thinner, superficial BCCs and BD lesions. Immunohistochemical analysis revealed that p53 positivity was associated with better treatment outcomes, while increased β-catenin and cytoplasmic GLUT1 expression correlated with resistance to PDT. On the other hand, the metabolic profile of the tumors indicated that tumors with higher glycolytic activity were less responsive to treatment, therefore, using metformin, a glycolytic inhibitor, as a potential adjuvant therapy to improve outcomes in resistant tumors should be considered., Conclusion: This study emphasizes the importance of personalized approaches in the use of MAL-PDT, tailoring treatment according to tumor-specific characteristics. Nitrogen (N) fertilization is essential to maximize crop production. However, around half of the applied N is lost to the environment, causing water and air pollution and contributing to climate change. Understanding the natural genetic and metabolic basis underlying plants N use efficiency is of great interest to attain an agriculture with less N demand and thus more sustainable. The study of ammonium (NH4+) nutrition is of particular interest, because it mitigates N losses due to nitrate (NO3-) leaching or denitrification. In this work, we studied Brachypodium distachyon, the model plant for C3 grasses, grown with NH4+ or NO3- supply. We performed gene expression analysis in the root of the B. distachyon reference accession Bd21 and examined the phenotypic variation across 52 natural accessions through analyzing plant growth and a panel of 22 metabolic traits in leaf and root. We found that the adjustment of primary metabolism to NH4+ nutrition is essential for the natural variation of NH4+ tolerance, notably involving NH4+ assimilation and phosphoenolpyruvate carboxylase (PEPC) activity. Additionally, genome-wide association studies (GWAS) indicated several loci associated with B. distachyon growth and metabolic adaptation to NH4+ nutrition. We found that the GDH2 gene was associated with the induction of root glutamate dehydrogenase activity under NH4+ nutrition and that two genes encoding malic enzyme were associated with leaf PEPC activity. Recent studies suggest that only 27.6% of cancer survivors meet PA guidelines. This could partially be attributed to the limited knowledge reported by healthcare professionals (HCP) regarding the appropriate timing, methodology, and suitability of referring cancer survivors to exercise programs or professionals. Background : Oral diseases remain among the most common non-communicable diseases worldwide, affecting almost half of the world's population. This is partly because there has been a separation of the mouth from the rest of the body and human health, and psychological aspects such as personality, thoughts, and emotions are not taken into account in the dental office. The objective was to analyze the relationship between oral health conditions and personality traits in adult patients who underwent dental treatment at the Center for Health Studies and Services. Methods : This was a descriptive, observational, and correlational study, carried out at the Center for Health Studies and Services. A total of 184 patients who attended the dentistry area in the period from October 2022 to May 2023 participated in the study, of which 59.78% were women and 40.21% men. The age of the population was 18 to 79 years, with the age range of 21-40 years being the most prevalent (48.37%). Results : The results show that although the hygiene of the population treated was good (0.77 ± 0.79) and that the perception of oral health was positive (14.34 ± 9.43), the means and percentages of oral pathologies and parafunctional habits were high (i.e., DMFT: 9.98 ± 5.40; attrition: 87.50%; onychophagia: 45.10%). It is noteworthy that both the correlation, network, multiple line regression, and logistic regression analyses showed associations of the personality, gender, and age variables with a history of caries and oral hygiene as well as with parafunctional habits. Conclusions : Therefore, variations in both the personality and the age and gender of the patients treated have repercussions on oral health conditions, which can be used in the prevention of oral diseases and in health promotion.

Background: some patients with inflammatory bowel disease (IBD) treated with antiTNF develop drug-induced psoriasis (antiTNF-IP). Several therapeutic strategies are possible., Aims: to assess the management of antiTNF-IP in IBD, and its impact in both diseases., Methods: patients with antiTNF-IP from ENEIDA registry were included. Therapeutic strategy was classified as continuing the same antiTNF, stopping antiTNF, switch to another antiTNF or swap to a non-antiTNF biologic. IP severity and IBD activity were assessed at baseline and 16, 32 and 54 weeks., Results: 234 patients were included. At baseline, antiTNF-IP was moderate-severe in 60 % of them, and IBD was in remission in 80 %. Therapeutic strategy was associated to antiTNF-IP severity (p < 0.001). AntiTNF-IP improved at week 54 with all strategies, but continuing with the same antiTNF showed the worst results (p = 0.042). Among patients with IBD in remission, relapse was higher in those who stopped antiTNF (p = 0.025). In multivariate analysis, stopping antiTNF, trunk and palms and soles location were associated with antiTNF-IP remission; female sex and previous surgery in Crohn´s disease with IBD relapse., Conclusion: skin lesions severity and IBD activity seem to determine antiTNF-IP management. Forest disturbance regimes across biomes are being altered by interactive effects of global change. Establishing baselines for assessing change requires detailed quantitative data on past disturbance events, but such data are scarce and difficult to obtain over large spatial and temporal scales. The integration of remote sensing with dense time series analysis and cloud computing platforms is enhancing the ability to monitor historical disturbances, and especially non-stand replacing events along climatic gradients. Since the integration of such tools is still scarce in Mediterranean regions, here, we combine dense Landsat time series and the Continuous Change Detection and Classification - Spectral Mixture Analysis (CCDC-SMA) method to monitor forest disturbance in continental Spain from 1985 to 2023. We adapted the CCDC-SMA method for improved disturbance detection creating new spectral libraries representative of the study region, and quantified the year, month, severity, return interval, and type of disturbance (stand replacing, non-stand replacing) at a 30 m resolution. In addition, we characterised forest disturbance regimes and trends (patch size and severity, and frequency of events) of events larger than 0.5 ha at the national scale by biome (Mediterranean and temperate) and forest type (broadleaf, needleleaf and mixed). We quantified more than 2.9 million patches of disturbed forest, covering 4.6 Mha over the region and period studied. Forest disturbances were on average larger but less severe in the Mediterranean than in the temperate biome, and significantly larger and more severe in needleleaf than in mixed and broadleaf forests. Since the late 1980s, forest disturbances have decreased in size and severity while increasing in frequency across all biomes and forest types. Active waveguide Bragg gratings (AWBGs) are promising photonic structures that combine the very efficient reflective properties of a Bragg grating with the power amplification character of rare earths. This combination may lead to a potential monolithic laser under the proper conditions. However, the photonic response of these structures highly depends on the grating design and operating parameters, so modeling its response for their laser performance is a must. In this work, a numerical method is employed to calculate the optical power propagation along an Er/Yb-codoped integrated AWBG. A complete database of the AWBG response as a function of the most relevant operational parameters is obtained. As a result, the critical values of each design and operating parameter to achieve its laser performance have been determined, which represents very useful information for further experimental design, optimization, and fabrication of these photonic structures.

Exercise, despite being a beneficial activity for health, can also be a source of oxidative imbalance, which can lead to a decrease in performance. Furthermore, melatonin is an endogenous molecule that may counteract exercise-induced oxidative stress. The aim of this study was to evaluate the potential ergogenic and antioxidant capacity of melatonin administered for a maximal effort test. A total of 30 rats were divided into three groups-control, exercise, and exercise + melatonin (intraperitoneal administration of 10 mg/kg)-to assess the effects of an exhaustive incremental protocol in the two exercise groups (with and without melatonin) on the treadmill-running performance (final speed reached), lipid and protein oxidation markers (malondialdehyde + 4-hidroxyalkenals and carbonyl content, respectively), and cellular and mitochondrial membranes' fluidity in skeletal muscle, brain, and liver tissues. Our results show an ergogenic effect of melatonin (31 ± 4 vs. 36 ± 4 cm/s), which may be due to its antioxidant properties being significantly stronger than its protective effect when performing increasing exercise on a treadmill until exhaustion. Melatonin reverted the membrane rigidity in the brain caused by exercise (with no effect on muscle or liver), prevented lipid oxidation in muscle, and prevented lipid and protein oxidation in the liver. Differences between tissues' responses to exercise and melatonin need to be investigated in the future to elucidate other possible mechanisms that explain melatonin's ergogenic effect.

Background/objectives: Patients with relapsed/refractory (R/R) AML with FLT3 mutation ( FLT3 mut ) have a dismal prognosis. FLT3 mut offers a target for therapy in these patients. Gilteritinib (gilter) and quizartinib (quizar) have demonstrated efficacy as single agents in two phase 3 clinical trials., Methods: We retrospectively analyzed the characteristics, treatments, and outcomes of 50 patients with R/R FLT3 mut AML who received gilter or quizar as monotherapy in 27 Spanish centers before their commercial availability. Forty-four patients were treated with gilter and six with quizar., Results: The median age was 62.5 years, and 52% were women. Most patients presented with FLT3 -ITD mutations (80%); 46% had refractory disease and 54% had relapsed disease at treatment initiation. First-line treatment was chemotherapy in 80% of patients, with 40% of these also receiving midostaurin. Twenty-five patients (50%) had previously received FLT3 inhibitor, and twenty-eight (56%) had received more than one line treatment before starting gilter/quizar. The rates of complete remission (CR), CR without hematological recovery (CRi), and partial remission were 22%, 18%, and 16%, respectively. The median overall survival (OS) and disease-free survival were 4.74 months and 2.99 months, respectively. We observed a significant improvement in OS in patients who had received only one prior line of therapy compared to those who had received two or more therapies (10.77 months vs. 4.24 months, p = 0.016). Multivariate analysis identified failure to achieve CR/CRi, receiving more than one prior line of therapy, age, and white blood cells count as independent prognostic factors for OS. The most common toxicities were febrile neutropenia, liver function abnormalities, and QT interval prolongation., Conclusions: Gilter/quizar monotherapy are effective and tolerable options for patients with R/R FLT3 mut AML in a real-world setting. Response and toxicity rates are similar to those reported in the phase 3 trials, despite the more heterogeneous nature of the study population.

During defatted Tenebrio molitor (TM) larvae powder production, oil is obtained as a by-product, mainly intended for feed enrichment or as a biofuel component. In 2021, EFSA authorized TM as the first insect to be a novel food. Thus, the study aimed to assess the composition, including fatty acids (FAs), tocopherols, carotenoids, phenolics, volatiles, antioxidant capacity, sensory aroma attributes, physical properties, and oxidative and hydrolytic stability of TM oil. The FAs profile was dominated by oleic-C18:1 9c (36.8%) and linoleic-C18:2 9c12c (32,4%) acids, resulting in a PUFA/SFA ratio similar to vegetable oils. Thus, TM oil was characterized by a beneficial Health Promoting Index (HPI) (2.42), which was 10-fold higher than the HPI of common animal fats. TM oil contained bioactive compounds such as carotenoids (13.65 mg/kg), tocopherols (105.8 mg/kg), and phenolic compounds (74 mg GAE/kg). A noticeable amount of apigenin was also noted among nine detected phenolic compounds. The substantial presence of lipophilic and phenolic compounds contributed to antioxidative potential. Sensory estimation revealed the dominance of fried and nutty aromas, probably because of the abundance of Strecker aldehydes and pyrazines in their volatile profile. The results indicated that the technological process needs modification to limit the formation of lipid oxidation volatile compounds such as aldehydes and eliminate some differences between batches. This preliminary study on the composition and properties of TM oil encourages its use as an ingredient for food, pharmaceutical, and cosmetics purposes.

Residency (R) and site fidelity (SF) are important parameters in population ecology, yet their quantification poses challenges in marine mammals. Based on a previous review, this study used simulated and empirical mark-resight data to assess the variations and performance of the most used R ( n  = 8) and SF ( n  = 11) indices in peer-reviewed literature under different scenarios. We applied the Jolly-Seber model to simulate thousands of bottlenose dolphin populations varying resighting ( p ) and survival ( Phi ) probabilities, and performed calibration, sensitivity, and validation analyses. Our results underscore the effects of p and Phi on individual categorization within the diverse simulated conditions, representing the often-overlooked heterogeneity in residency classification for Tursiops populations. All SF indices showed similar and consistent performance (>0.70 Gower's distance) across the simulated scenarios, even when compared to field study data from wild dolphin populations ( i.e., Savannah, USA, and Alvarado, Mexico); thus, SF should be a critical parameter for interstudy comparisons. Conversely, R indices were remarkably different based on their definitions and classification criteria. The different thresholds among definitions largely biased the proportion of residents and transient individuals (or occasional visitors) even leading to counterintuitive outcomes. This emphasizes the importance of considering trade-offs in R index selection aligned with project goals, specific sampling efforts, and population dynamics. For instance, the simplified binomial categorization of R defined by Conway (2017) (https://digitalcommons.coastal.edu/etd/10/) easier to interpret but R indices incorporating temporal components ( e.g., monthly, seasonal, and annual) outperformed (>0.70 Gower's distance) other R indices lacking such criteria. This allowed for a more detailed representation of the temporal structure of the population, and higher consistency and accuracy while classifying individuals. Also, although the residency categories proposed by Möller, Allen & Harcourt (2002) (DOI 10.1071/AM02011) did not perform as well, these seemed to fit better when dealing with data gaps across spatial and temporal scales. The present study aims to analyze the psychological characteristics associated with the performance of amateur athletes, exploring their relationships with key variables such as resilience, harmonious and obsessive passion, and perception of discomfort. The main objective is to establish the relationship between psychological characteristics and each of the variables used (resilience, personal competence, and acceptance of self and life), and to consider whether these are related to passion (harmonious and obsessive) through a structural equation analysis. The sample was composed of 110 persons: 87 adults (22 females, mean age 40.7 ± 9.1 years; 65 males, mean age in years 42.1 ± 11.9) and 23 adolescents (16 females, mean age 14.8 ± 1.3 years; 7 males, mean age 15.3 ± 2.1 years). Psychological characteristics were assessed using the Psychological Characteristics Related to Performance (CPRD) questionnaire, passion using the Passion Scale instrument, and resilience using the Resilience Scale (RS-14) questionnaire. The analyses were based on a structural equation model analysis (PLS-SEM). The results showed adequate coefficients of determination ( R index 2 ) and Stone-Geisser predictive relevance ( Q 2 ) for the personal competence resilience factors ( R 2 = 0.517; Q 2 = 0.218) and acceptance of self and life ( R 2 =.415; Q 2 = 0.231), as well as for the passion dimensions harmonious passion ( R 2 = 0.357; Q 2 = 0.168) and obsessive passion ( R 2 = 0.085; Q 2 = 0.034). In conclusion, a close relationship was demonstrated between psychological characteristics related to sports performance and the variables of resilience and passion (both harmonious and obsessive). In particular, it was confirmed that mental skills and stress management are linked to the dimensions of resilience, and these dimensions influence both types of passion. In addition, team cohesion and personal competence also play a crucial role in the development of passion, highlighting the importance of these factors in the assessment of sports performance and influencing the well-being of amateur athletes in their personal and sports development.

Background: Health Promoting Schools (HPS) have emerged as a powerful framework to promote healthy behaviors in many countries. However, HPS still present several challenges, highlighting the excessive workload involved in the accreditation, design, implementation, and evaluation processes. In this sense, a resource to facilitate the implementation processes may have a positive impact on the support of HPS., Objective: The aim of this study was to describe the co-design processes undertaken and resulting learnings to develop the Red Escuelas Promotoras de Salud (network of health promoting schools; REDEPS)-Gestion platform to facilitate the accreditation, design, implementation procedures, and evaluation processes of the Aragon's Health-Promoting School Network., Methods: The Double Diamond Design Approach was used to co-design this web-platform. The different stakeholders that participated in this co-design, progressed through a 4-stage reflective phase, to discover, define, develop, and deliver the REDEPS-Gestion platform., Results: Participants agreed that the functions of the REDEPS-Gestion platform should permit the management of both the educational centers and the administration such as accreditation processes, definition and review intervention projects, and preparation and review of the different progress reports to evaluate the HPS. Despite co-design being a well-established approach to creative practice, especially within the public sector, some challenges emerged during the co-design process, such as engaging and facilitating stakeholders' participation or the complexity of combining the interests of all stakeholders. This approach allowed us to identify the main barriers for future users and implement platform improvements., Conclusions: We hope that the REDEPS-Gestion platform will therefore be able to contribute to facilitating the implementation of HPS. The Double Diamond Design Approach used to co-design this web platform was an efficient and feasible methodological design approach. The REDEPS-Gestion platform will facilitate HPS implementation in Aragon as well as all the processes involving HPS. Background: Research into key performance factors in trail running, particularly in vertical kilometer (VK) races, is crucial for effective training and periodization. However, recent studies on metabolic and cardiorespiratory responses during VK races, especially using field tests, are limited. Objectives: Therefore, the aim of this study is to evaluate the metabolic and cardiorespiratory responses during a VK field test, identifying differences based on sex and performance level, as well as key performance factors and their deterioration due to fatigue. Fifteen trained trail runners (ten males and five females, 19 to 38 years old) perform a VK race. Methods: The global physiological response is evaluated using the portable gas analyzer Cosmed K5 and the local response using near-infrared spectroscopy technology. Results : In gender comparisons, the ANCOVA test shows significant differences ( p < 0.05) in the ventilation, tidal volume, expiratory time-to-inspiratory time ratio, inspiratory flow rate, end-tidal CO 2 partial pressure, heart rate, oxygen pulse, and total hemoglobin. Additionally, the performance comparison reveals significant differences in the variables' velocity, oxygen consumption, carbon dioxide production, ventilation, dead space-to-tidal volume ratio, total time of the breathing cycle, expiratory time-to-inspiratory time ratio, inspiratory duty cycle, expiratory fractions of CO 2 , quadriceps saturation index, and VE/VCO 2 ratio. Finally, the correlation analysis shows oxygen consumption (r = -0.80 mean; r = -0.72 peak), carbon dioxide production (r = -0.91 mean; r = -0.75 peak), expiratory time-to-inspiratory time ratio (r = 0.68 peak), ventilation (r = -0.58 mean), and quadriceps saturation index (r = 0.54 mean; r = -0.76 coefficient of variation) as the key performance factors in the VK race. Conclusions: Overall, the physiological analysis indicates the importance of local muscular adaptations and respiratory system capacity in this type of short-duration race.

Background: Truffle cultivation is evolving rapidly and new agronomic practices such as 'truffle nests' (localized peat amendments of the orchard soil) are being developed. Truffle nests improve the shape of truffles and their depth in the soil and reduce the occurrence of insect damage but have also raised concerns about their impact on the ripeness and maturity of the harvested truffles. In this study, the effect of the nests on the volatile organic compounds profile and the aromatic profile of black truffles was evaluated, as well as the existence of perceptible sensorial differences in truffles. For this, truffles growing in nests were compared with truffles growing in the bulk soil of the same host tree., Results: Gas chromatography showed that nest truffles had a less complex volatile organic compound profile than bulk-soil truffles. Olfactometry indicated that nest truffles were associated with higher modified frequency values of odorants corresponding to sulfur-containing compounds. Despite this, sensory evaluation with consumers could not clearly show that nest truffles can be distinguished sensorially from bulk-soil truffles., Conclusion: The results prove that soil conditions can influence the aromatic profile of truffles and thus suggest the possibility of managing truffle aroma using agronomic practices. © 2024 The Author(s). Background: The efficacy of ustekinumab and vedolizumab for treating complex perianal fistula in Crohn's disease has been barely studied. We aimed to assess treatment persistence, clinical remission, and safety of these drugs in this context., Methods: Crohn's disease patients who had received ustekinumab or vedolizumab for the indication of active complex perianal fistula, were included. Clinical remission was defined according to Fistula Drainage Assessment Index (no drainage through the fistula upon gentle pressure) based on physicians' assessment., Results: Of 155 patients, 136 received ustekinumab, and 35 vedolizumab (16 received both). Median follow-up for ustekinumab was 27 months. Among those on ustekinumab, 54 % achieved remission, and within this group, 27 % relapsed during follow-up. The incidence rate of relapse was 11 % per patient-year. Multivariate analysis found no variables associated with treatment discontinuation or relapse. Median follow-up time for patients receiving vedolizumab was 19 months. Remission was achieved in 46 % of the patients receiving vedolizumab, and among them, 20 % relapsed during follow-up. The incidence rate of relapse was 7 % per patient-year. Background: Ustekinumab is approved for ulcerative colitis (UC)., Aims: To assess the durability of ustekinumab in patients with UC and its short-term effectiveness, durability and tolerability in clinical practice., Methods: Retrospective, multicentre study of patients who had received their first ustekinumab dose at least 16 weeks before inclusion. Patients were followed until treatment discontinuation or last visit. Only patients with active disease at the start of ustekinumab treatment were considered in the effectiveness analysis. Patients who stopped ustekinumab before their last visit were considered not to be in subsequent remission., Results: We included 620 patients; 155 (25%) discontinued ustekinumab during follow-up (median 12 months). Rate of discontinuation was 20% per patient-year of follow-up. Anaemia at baseline (hazard ratio, HR 1.5; 95% confidence interval [CI] 1.1-2.1), steroids at baseline (HR 1.5; 95% CI 1.06-2.08) and more severe clinical activity at baseline (HR 1.5; 95% CI 1.09-2.06) were associated with higher risk of discontinuation. At the end of induction, 226 (40%) patients were in steroid-free clinical remission. Moderate-severe vs mild disease activity at baseline (odds ratio [OR] 0.3; 95% CI 0.2-0.5), male sex (OR 0.5; 95% CI 0.4-0.8), and increased number of previous biologics (OR 0.6; 95% CI 0.6-0.8) were associated with lower likelihood of steroid-free clinical remission at week 16. One hundred and seventy-six patients (28%) had at least one adverse event. We observed no negative impact of ustekinumab on extraintestinal manifestations and/or immune-mediated diseases., Conclusions: Ustekinumab durability in UC was relatively high, and treatment was effective in highly refractory patients. Background and Aims: The size and shape of reproductive structures is especially relevant in evolution because these characters are directly related to the capacity for pollination and seed dispersal, a process that plays a basic role in evolutionary patterns. The evolutionary trajectories of reproductive phenotypes in gymnosperms have received special attention in terms of pollination and innovations related to the emergence of the spermatophytes. However, variability of reproductive structures, evolutionary trends and the role of environment in the evolution of cycad species have not been well documented and explored. This study considered this topic under an explicitly phylogenetic and evolutionary approach that included a broad sampling of reproductive structures in the genus Ceratozamia., Methods: We sampled 1400 individuals of 36 Ceratozamia species to explore the evolutionary pattern and identify and evaluate factors that potentially drove their evolution. We analysed characters for both pollen and ovulate strobili within a phylogenetic framework using different methods and characters (i.e. molecular and both quantitative and qualitative morphological) to infer phylogenetic relationships. Using this phylogenetic framework, evolutionary models of trait evolution for strobilar size were evaluated. In addition, quantitative morphological variation and its relation to environmental variables across species were analysed., Key Results: We found contrasting phylogenetic signals between characters of pollen and ovulate strobili. These structures exhibited high morphological disparity in several characters related to size. Results of analyses of evolutionary trajectories suggested a stabilizing selection model. With regard to phenotype-environment, the analysis produced mixed results and differences for groups in the vegetation type where the species occur; however, a positive relationship with climatic variables was found., Conclusions: The integrated approach synthesized reproductive phenotypic variation with current phylogenetic hypotheses and provided explicit statements of character evolution. The characters of volume for ovulate strobili were the most informative, and could provide a reference for further study of the evolutionary complexity in Ceratozamia. Background and Aims: Inflammatory pouch disorders exhibit a heterogeneous clinical spectrum and therapeutic requirements have not been properly studied., Methods: This retrospective, multicentre study included ulcerative colitis patients with ileal pouch construction and were later diagnosed with an inflammatory pouch disorder between 1995 and 2020. Classifications, behaviour and therapies applied were recorded and compared in the long-term., Results: Overall, 338 patients were recruited. The most common disorders were pouchitis (n = 258, 76%), Crohn's disease of the pouch (n = 55, 16%) and cuffitis (n = 25, 7%). Pouchitis presented mainly as chronic (65.2%) and recurrent (87%) forms. Crohn's disease manifested as stricturing/penetrating in 53% of cases and perianal disease in 42%. Patients received multiple therapies: 86% antibiotics, 42% steroids, 27% immunosuppressants, 43% biologics and 27% surgery. Compared with pouchitis, Crohn's disease of the pouch was characterised by a later diagnosis (99 vs. 55 months, p < 0.001) and greater needs for immunosuppressants (OR 3.53, 1.79-6.94, p < 0.0001), biologics (OR 5.45, 2.78-10.6, p < 0.0001) and surgeries (OR 2.65, 1.43-4.89, p < 0.001)., Conclusions: Chronic pouchitis is the most common pouch disorder presentation. Background: Gait variables assessed by inertial measurement units (IMUs) show promise as screening tools for aging-related diseases like sarcopenia. The main aims of this systematic review were to analyze and synthesize the scientific evidence for screening sarcopenia based on gait variables assessed by IMUs, and also to review articles that investigated which gait variables assessed by IMUs were related to sarcopenia., Methods: Six electronic databases (PubMed, SportDiscus, Web of Science, Cochrane Library, Scopus and IEEE Xplore) were searched for journal articles related to gait, IMUs and sarcopenia. The search was conducted until December 5, 2023. Titles, abstracts and full-length texts for studies were screened to be included., Results: A total of seven articles were finally included in this review. Despite some methodological variability among the included studies, IMUs demonstrated potential as effective tools for detecting sarcopenia when coupled with artificial intelligence (AI) models, which outperformed traditional statistical methods in classification accuracy. The findings suggest that gait variables related to the stance phase such as stance duration, double support time, and variations between feet, are key indicators of sarcopenia., Conclusions: IMUs could be useful tools for sarcopenia screening based on gait analysis, specifically when artificial intelligence is used to process the recorded data. Background and Aims: Familial inflammatory bowel disease (IBD) history is a controversial prognostic factor in IBD. We aimed to evaluate the impact of a familial history of IBD on the use of medical and surgical treatments in the biological era., Methods: Patients included in the prospectively maintained ENEIDA database and diagnosed with IBD after 2005 were included. Familial forms were defined as those cases with at least one first-degree relative diagnosed with IBD. Disease phenotype, the use of biological agents, or surgical treatments were the main outcomes., Results: A total of 5263 patients [2627 Crohn's disease (CD); 2636 ulcerative colitis (UC)] were included, with a median follow-up of 31 months. Of these, 507 (10%) corresponded to familial forms. No clinical differences were observed between familial and sporadic IBD forms except a lower age at IBD diagnosis and a higher rate of males in familial forms of UC. In CD, the proportions of patients treated with thiopurines (54.4% vs 46.7%; P = .015) and survival time free of thiopurines (P = .009) were lower in familial forms. No differences were found regarding the use of biological agents. Concerning surgery, a higher rate of intestinal resections was observed in sporadic CD (14.8% vs 9.9%, P = .027). No differences were observed in UC., Conclusions: In the era of biological therapies, familial and sporadic forms of IBD show similar phenotypes and are managed medically in a similar way; whether these is due to lack of phenotypical differences or an effect of biological therapies is uncertain. What is already known on this topic: IBD's etiopathogenesis points to an interaction between environmental and genetic factors, being familial history a controversial prognostic factor. Biological agents use and need for surgery regarding familial or sporadic forms of IBDs present conflicting results. What this study adds: Familial and sporadic forms of IBD have similar phenotypes and are managed medically and surgically in a similar way. Background: Polyploidisation often results in genome rearrangements that may involve changes in both the single-copy sequences and the repetitive genome fraction. In this study, we performed a comprehensive comparative analysis of repetitive DNA, with a particular focus on ribosomal DNA (rDNA), in Brachypodium hybridum (2n = 4x = 30, subgenome composition DDSS), an allotetraploid resulting from a natural cross between two diploid species that resemble the modern B. distachyon (2n = 10; DD) and B. stacei (2n = 20; SS). Taking advantage of the recurrent origin of B. hybridum, we investigated two genotypes, Bhyb26 and ABR113, differing markedly in their evolutionary age (1.4 and 0.14 Mya, respectively) and which resulted from opposite cross directions. To identify the origin of rDNA loci we employed cytogenetic and molecular methods (FISH, gCAPS and Southern hybridisation), phylogenetic and genomic approaches., Results: Unlike the general maintenance of doubled gene dosage in B. hybridum, the rRNA genes showed a remarkable tendency towards diploidisation at both locus and unit levels. While the partial elimination of 35S rDNA units occurred in the younger ABR113 lineage, unidirectional elimination of the entire locus was observed in the older Bhyb26 lineage. Additionally, a novel 5S rDNA family was amplified in Bhyb26 replacing the parental units. The 35S and 5S rDNA units were preferentially eliminated from the S- and D-subgenome, respectively. Thus, in the more ancient B. hybridum lineage, Bhyb26, 5S and 35S rRNA genes are likely expressed from different subgenomes, highlighting the complexity of polyploid regulatory networks., Conclusion: Comparative analyses between two B. hybridum lineages of distinct evolutionary ages revealed that although the recent lineage ABR113 exhibited an additive pattern of rDNA loci distribution, the ancient lineage Bhyb26 demonstrated a pronounced tendency toward diploidisation manifested by the reduction in the number of both 35S and 5S loci. Background: Community-acquired pneumonia (CAP) is associated with high morbidity and hospitalization rate. In infectious diseases, host genetics plays a critical role in susceptibility and immune response, and the immune pathways involved are highly dependent on the microorganism and its route of infection. Here we aimed to identify genetic risk loci for CAP using a case-control genome-wide association study (GWAS)., Methods: We performed a GWAS on 3,765 Spanish individuals, including 257 adult patients hospitalized with CAP and 3,508 population controls. Pneumococcal CAP was documented in 30% of patients; the remaining 70% were selected among patients with unidentified microbiological etiology. We tested 7,6 million imputed genotypes using logistic regressions. UK Biobank GWAS of bacterial pneumonia were used for results validation. Subsequently, we prioritized genes and likely causal variants based on Bayesian fine mapping and functional evidence. Imputation and association of classical HLA alleles and amino acids were also conducted., Results: Six independent sentinel variants reached the genome-wide significance (p < 5 × 10 -8 ), three on chromosome 6p21.32, and one for each of the chromosomes 4q28.2, 11p12, and 20q11.22. Only one variant at 6p21.32 was validated in independent GWAS of bacterial and pneumococcal pneumonia. Our analyses prioritized C4orf33 on 4q28.2, TAPBP on 6p21.32, and ZNF341 on 20q11.22. Interestingly, genetic defects of TAPBP and ZNF341 are previously known inborn errors of immunity predisposing to bacterial pneumonia, including pneumococcus and Haemophilus influenzae. The Mediterranean Basin has experienced substantial land use changes as traditional agriculture decreased and population migrated from rural to urban areas, which have resulted in a large forest cover increase. The combination of Landsat time series, providing spectral information, with lidar, offering three-dimensional insights, has emerged as a viable option for the large-scale cartography of forest structural attributes across large time spans. Here we develop and test a comprehensive framework to map forest above ground biomass, canopy cover and forest height in two regions spanning the most representative biomes in the peninsular Spain, Mediterranean (Madrid region) and temperate (Basque Country). As reference, we used lidar-based direct estimates of stand height and forest canopy cover. The reference biomass and volume were predicted from lidar metrics. Landsat time series predictors included annual temporal profiles of band reflectance and vegetation indices for the 1985-2023 period. Additional predictor variables including synthetic aperture radar, disturbance history, topography and forest type were also evaluated to optimize forest structural attributes retrieval. The estimates were independently validated at two temporal scales, i) the year of model calibration and ii) the year of the second lidar survey. The final models used as predictor variables only Landsat based metrics and topographic information, as the available SAR time-series were relatively short (1991-2011) and disturbance information did not decrease the estimation error. Model accuracies were higher in the Mediterranean forests when compared to the temperate forests (R 2  = 0.6-0.8 vs. 0.4-0.5). Between the first (1985-1989) and the last (2020-2023) decades of the monitoring period the average forest cover increased from 21 ± 2% to 32 ± 1%, mean height increased from 6.6 ± 0.43 m to 7.9 ± 0.18 m and the mean biomass from 31.9 ± 3.6 t ha -1 to 50.4 ± 1 t ha -1 for the Mediterranean forests. In temperate forests, the average canopy cover increased from 55 ± 4% to 59 ± 3%, mean height increased from 15.8 ± 0.77 m to 17.3 ± 0.21m, while the growing stock volume increased from 137.8 ± 8.2 to 151.5 ± 3.8 m 3  ha -1 . Normal aging presents subtle cognitive changes that can be detected before meeting the criteria for Mild cognitive impairment (MCI). Older people with low cognitive reserve and who receive limited cognitive stimulation are at greater risk of deterioration. In this regard, cognitive stimulation (CS) has been identified as an intervention that reduces this risk, provided that its design takes into account the differences in the level of cognitive reserve (CR) acquired throughout life and the baseline level of cognitive functioning. The general objective of this study is to evaluate, through a randomized clinical trial, the effectiveness of a computerized cognitive stimulation program, designed and adapted from Occupational Therapy based on the level of cognitive reserve in older adults in Primary Care. 100 participants will be randomized in a stratified manner according to the level of cognitive reserve (low/moderate/high), assigning 50 participants to the control group and 50 participants to the intervention group. The intervention group will carry out a computerized cognitive stimulation intervention designed and adapted from occupational therapy according to the level of cognitive reserve, through the "stimulus" platform. The main result expected to be achieved is the improvement of higher brain functions. As secondary results, we expect that those cognitive aspects most vulnerable to aging will decrease more slowly (in areas such as memory, executive function, attention and processing speed), and that the cognitive reserve of the participants will increase, in addition to being able to balance gender differences in these aspects. Objective: To compare spinal misalignment and execution time of 3 rescue maneuvers for casualties in traffic accidents., Materials and Methods: Biomechanical analysis using inertial sensors to measure the range of spinal misalignment and execution time of 3 maneuvers: 1) standard manual rescue (SMR); 2) rapid extrication device (RED); 3) extrication device (ED). The sample consisted of 117 rescue professionals (firefighters) from 14 Autonomous Communities in Spain. The total range of motion and the execution time of maneuvers were compared using ANOVA., Results: The highest range of cervical flexion-extension movement was recorded with RED (30° ± 9°). There were no statistically significant differences between SMR (21° ± 9°) and ED (21° ± 10°). In dorsal flexion-extension, the highest range of movement was with RED (36° ± 10°), followed by ED (30° ± 7°), with the lowest found for SMR (26° ± 11°). RED presented the least restriction of cervical (p < 0.001) and dorsal movement (p < 0.001). Introduction: Male breast cancer accounts for 1% of all breast cancers. Its low frequency leads to a lack of awareness, resulting in significant diagnostic delays. Additionally, this limits the available evidence, which primarily uses diagnostic-therapeutic algorithms based on women., Objectives: To analyze the prevalence, clinical presentation, anatomical and pathological characteristics, and prognosis of male breast cancer using one of the largest series available. Secondarily, to compare our data with studies conducted in women., Materials and Methods: A multicenter, observational, descriptive, retrospective study was conducted in the autonomous community of Aragon, Spain, from 1995 to 2022 including men with a pathological diagnosis of breast cancer., Results: A total of 148 patients were included, with a prevalence of 1%. The most common clinical presentation was a palpable retroareolar mass. Invasive ductal carcinoma was the most frequent type (88.89%), and luminal B was the predominant subtype (47.76%). Surgery was the most utilized treatment; mastectomy was performed in 90.34% and AL in 46.89%. At diagnosis, 52.46% had extramammary involvement. The recurrence rate was 24.1%, and the mortality attributed to the disease was 14.6%., Conclusions: There is a high rate of metastatic involvement at diagnosis, a high percentage of mutilating surgeries, and a high number of recurrences compared to available studies on males. Additionally, a worse prognosis is observed compared to breast cancer in women, despite these tumors having a less aggressive molecular subtype. Brachypodium stacei is the most ancestral lineage in the genus Brachypodium, a model system for grass functional genomics. B. stacei shows striking and sometimes contradictory biological and evolutionary features, including a high selfing rate yet extensive admixture, an ancient Miocene origin yet with recent evolutionary radiation, and adaptation to different dry climate conditions in its narrow distribution range. Therefore, it constitutes an ideal system to study these life history traits. We studied the phylogeography of 17 native circum-Mediterranean B. stacei populations (39 individuals) using genome-wide RADseq SNP data and complete plastome sequences. Nuclear SNP data revealed the existence of six distinct genetic clusters, low levels of intra-population genetic diversity and high selfing rates, albeit with signatures of admixture. Coalescence-based dating analysis detected a recent split between crown lineages in the Late Quaternary. Plastome sequences showed incongruent evolutionary relationships with those recovered by the nuclear data, suggesting interbreeding and chloroplast capture events between genetically distant populations. Demographic and population dispersal coalescent models identified an ancestral origin of B. stacei in the western-central Mediterranean islands, followed by an early colonization of the Canary Islands and two independent colonization events of the eastern Mediterranean region through long-distance dispersal and bottleneck events as the most likely evolutionary history. Climate niche data identified three arid niches of B. stacei in the southern Mediterranean region. Background: The response to SARS-CoV-2 vaccination decreases in inflammatory bowel disease (IBD) patients, specially under anti-TNF treatment. However, data on medium-term effectiveness are limited, specially using new recommended seroconversion rate (>260BAU/mL). Our aim was to evaluate the 6-month>260 BAU-seroconversion rate after full vaccination and after booster-dose., Methods: VACOVEII is a Spanish multicenter, prospective study promoted by GETECCU. IBD patients full vaccinated against SARS-CoV-2 and without previous COVID-19 infection, treated or not with immunosuppressants, were included. The booster dose was administered 6 months after the full vaccination. Seroconversion was set at 260BAU/mL, according to most recent recommendations and was assessed 6 months after the full vaccination and 6 months after booster-dose., Results: Between October 2021 and March 2022, 313 patients were included (124 no treatment or mesalazine; 55 immunomodulators; 87 anti-TNF; 19 anti-integrin; and 28 ustekinumab). Most patients received mRNA-vaccines (86%). Six months after full vaccination, overall seroconversion rate was 44.1%, being significantly lower among patients on anti-TNF (19.5%, p<0.001) and ustekinumab (35.7%, p=0.031). The seroconversion rate after booster was 92%. Again, anti-TNF patients had a significantly lower seroconversion rate (67%, p<0.001). mRNA-vaccine improved seroconversion rate (OR 11.720 [95% CI 2.26-60.512])., Conclusion: The full vaccination regimen achieves suboptimal response in IBD patients, specially among those anti-TNF or ustekinumab. The booster dose improves seroconversion rate in all patients, although it remains limited in those treated with anti-TNF. Objective: To quantify the incremental impact that population dispersion has on the number of health personnel in Primary Care in Alto Aragón, using a reproducible method., Method: Descriptive observational study that compares health the number of health personnel (family medicine, pediatrics and nursing) in EAP and PA emergencies in 2019 in an unpopulated and dispersed territory such as Huesca, with the number that would correspond to it by applying population ratios per professional of hypothetical constructs with different population densities., Results: Huesca, with respect to the national average, has 39% more PA health personnel. There are 239 additional professionals (112 in family medicine, 2 in pediatrics and 115 in nursing), 130 in emergencies and 109 in EAP. With the average of the five most densely populated provinces, it would reduce this staff by 49%, and with the average of the five least densely populated provinces, it would increase it by 12%., Conclusions: There is a relationship between low population density and a greater number of family medicine and PC nurses, but not with pediatrics. The powerful incremental effect that dispersion has on health care spending gives it a relevant role in the regional financing system. Pseudolipomatosis (PL) is a rare, benign, iatrogenic condition first described in colorectal mucosa. This condition is thought to arise from gas penetration into the mucosa due to mechanical or chemical injury during endoscopy. Despite these associations and its characteristic appearence, the definitive pathogenesis remains unclear and it continues to be underrecognized by some pathologists and endoscopists, which is crucial to avoid any further unnecessary treatments. Immunohistochemistry techniques play a role in differentiating PL from similar conditions such as cystic intestinal pneumatosis, intestinal lipomatosis, or lymphangiectasia. A 70-year-old man with a family history of colon adenocarcinoma was diagnosed with duodenal pseudolipomatosis after a routine endoscopic assessment. To the best of our knowledge, there have been only two reports of PL diagnosed in the duodenum. A review of over 500 duodenal biopsies at our centre revealed that no other instances were identified, emphasizing the unusual occurrence of this particular location.

Background: Allergic diseases begin early in life and are often chronic, thus creating an inflammatory environment that may precede or exacerbate other pathologies. In this regard, allergy has been associated to metabolic disorders and with a higher risk of cardiovascular disease, but the underlying mechanisms remain incompletely understood., Methods: We used a murine model of allergy and atherosclerosis, different diets and sensitization methods, and cell-depleting strategies to ascertain the contribution of acute and late phase inflammation to dyslipidemia. Untargeted lipidomic analyses were applied to define the lipid fingerprint of allergic inflammation at different phases of allergic pathology. Expression of genes related to lipid metabolism was assessed in liver and adipose tissue at different times post-allergen challenge. Also, changes in serum triglycerides (TGs) were evaluated in a group of 59 patients ≥14 days after the onset of an allergic reaction., Results: We found that allergic inflammation induces a unique lipid signature that is characterized by increased serum TGs and changes in the expression of genes related to lipid metabolism in liver and adipose tissue. Alterations in blood TGs following an allergic reaction are independent of T-cell-driven late phase inflammation. On the contrary, the IgG-mediated alternative pathway of anaphylaxis is sufficient to induce a TG increase and a unique lipid profile. Lastly, we demonstrated an increase in serum TGs in 59 patients after undergoing an allergic reaction., Conclusion: Overall, this study reveals that IgG-mediated allergic inflammation regulates lipid metabolism., (© 2024 The Author(s).
