Key Message: We developed an in-house Python-based image analysis pipeline to investigate the movement patterns of Cuscuta. Our analysis unveiled that the coiling and circumnutation movements of Cuscuta are regulated by its intrinsic circadian rhythm. Cuscuta spp., commonly known as dodders, are rootless and leafless stem parasitic plants. Upon germination, Cuscuta starts rotating immediately in a counterclockwise direction (circumnutation) to locate a host plant, creating a seamless vascular connection to steal water and nutrients from its host. In this study, our aim was to elucidate the dynamics of the coiling patterns of Cuscuta, which is an essential step for successful parasitism. Using time-lapse photography, we recorded the circumnutation and coiling movements of C. campestris at different inoculation times on non-living hosts. Subsequent image analyses were facilitated through an in-house Python-based image processing pipeline to detect coiling locations, angles, initiation and completion times, and duration of coiling stages in between. The study revealed that the coiling efficacy of C. campestris varied with the inoculation time of day, showing higher success and faster initiation in morning than in evening. These observations suggest that Cuscuta, despite lacking leaves and a developed chloroplast, can discern photoperiod changes, significantly determining its parasitic efficiency. The automated image analysis results confirmed the reliability of our Python pipeline by aligning closely with manual annotations. This study provides significant insights into the parasitic strategies of C. campestris and demonstrates the potential of integrating computational image analysis in plant biology for exploring complex plant behaviors. We studied the population and reproductive characteristics of the stone crab Hapalogaster dentata (De Haan, 1849) (Anomura: Lithodidae) parasitized by Briarosaccus hoegi (Rhizocephala: Peltogastridae) from Peter the Great Bay (northwestern Sea of Japan) in 2023. The sex structure of the H. dentata population was female-biased and represented by 39.1% males and 60.9% females. The size-frequency distribution differed between males and females. The prevalence of parasitization of H. dentata by B. hoegi reached 6.7%. No relationship between crab sex and parasitization prevalence was found. Most of the parasitized specimens (94.8%) had only a single parasitic externa. The non-parasitized and parasitized crabs did not differ in main carapace length. The pleopods in most of the externa-bearing females were shortened or completely atrophied. H. dentata produced 1 clutch per year with ovigerous females occurring from October to May. An obvious delay in embryonic development was observed in winter. None of the parasitized H. dentata females were ovigerous. Externae with embryos of B. hoegi were found from May to July.

Parasites from the Leishmania genus are the causative agents of leishmaniasis and primarily reside within macrophages during mammalian infection. Their ability to establish intracellular infection provides a secure niche for proliferation while evading detection. However, successful multiplication within mammalian cells requires the orchestration of multiple mechanisms that control host cell viability. In contrast, innate immune cells, such as macrophages, can undergo different forms of cell death in response to pathogenic intracellular microbes. Thus, modulation of these different forms of host cell death is crucial for Leishmaniasis development. The regulation of host cell apoptosis, a form of programmed cell death, is crucial for sustaining parasites within viable host cells. Accordingly, several studies have demonstrated evasion of apoptosis induced by dermotropic and viscerotropic Leishmania species. Conversely, the prevention of pyroptosis, an inflammatory form of cell death, ensures the establishment of infection by silencing the release of mediators that could trigger massive proinflammatory responses. Acroceridae are active parasitoids of mygalomorph spiders. However, little is known of their interaction numbers and biology in the Neotropics. In this study, we present 15 new records of acrocerid-mygalomorph interactions, adding new biological information about spider fly development. Mygalomorph spiders were captured in the field and maintained in captivity, and the development time of acrocerids was followed from the 4th instar to adulthood. Only seven acrocerid flies were able to complete their development. The acrocerids obtained were Arrhynchus stuardoi, Arrhynchus maculatus, and Archipialea sp. The total recorded development time (from the point where the larva leaves its host to the eclosion of the adult fly) of the spider flies was between 20 and 29 days. Otopheidomenidae include 30 species of parasites of insects. These mites occur in areas with warm and hot climates. The only exception is Hemipteroseius adleri, the distribution of which reaches the central European region, which is characterised by a moderate climate with below-zero temperatures in winter. In Europe, H. adleri only parasitises the red firebug (Pyrrhocoris apterus). Field observations indicate that only fertilised females of H. adleri survive winter conditions. The aim of the study therefore was to evaluate the impact of air temperature on the development and survival of H. adleri. A research model was established involving mites parasitising red firebugs in laboratory conditions at 15, 20, 25 and 30 °C. After oviposition, no egg development was possible at 15 °C. Egg development began at 20 °C, however the egg-to-adult period lasted for 13 days and larval mortality was significantly higher at this temperature than at 25 and 30 °C. At the latter tem temperatures, mites matured fastest (five and four days, respectively). The net reproductive rate (R 0 ) was 10.8, the mean generation time was (T) 14.4 (SD = 0.273), the intrinsic rate of population increase (r m ) was 0.165 (SD = 0.003), and the finite rate of population increase (λ) was 1.12. At 25 °C, H. adleri required 4.2 days to double its population. The results indicate that the thermal conditions prevailing in early spring in central Europe are highly unfavourable for the effective reproduction of H. adleri. The fall armyworm (FAW), Spodoptera frugiperda (Smith), is a significant pest threatening crops like maize across Africa, necessitating sustainable pest management alternatives. This study evaluates the efficacy of Trichogramma evanescens as a biological control agent against FAW egg masses in Egypt under laboratory and semi-field conditions. FAW larvae were initially collected from infested maize fields and reared on castor-oil plant leaves. Meanwhile, T. evanescens was propagated using Sitotroga cerealella eggs as hosts. The host eggs, aged 18 to 24 h, were sterilized with UV light to prevent host development while maintaining suitability for parasitism. Using custom-designed parasitoid incubators and hemisphere clip-cages, experiments focused on various egg mass configurations, assessing the effects of scales and layering. Laboratory conditions were controlled at 25 ± 2ºC and 55 ± 5% relative humidity, while semi-field trials used large cages in maize fields to approximate natural conditions. The results showed that in laboratory settings, parasitism rates averaged 5.96%, 2.00%, and 1.56% for non-, average-, and dense-scale egg masses, respectively. For egg masses with varying layers, parasitism rates were 5.24% for single-layer, 3.09% for double-layer, and 1.18% for ple-layer, regardless of scale presence. In semi-field conditions, parasitism rates were 1.01% for single-layer, 1.13% for double-layer, and 0.59% for triple-layer egg masses. Correspondingly, parasitism rates for non-, average-, and dense-scale eggs were 1.85%, 0.60% and 0.27%. The study concludes that T. evanescens shows promise for integrated pest management programs; however, its effectiveness is constrained by physical and environmental variables. Turtle acanthocephalans in the genus Neoechinorhynchus have complex life cycles and are thought to involve an ostracod intermediate host and a turtle definitive host. For Neoechinorhynchus emydis, an additional snail paratenic host is part of its life cycle. However, how snails become infected and whether other species of Neoechinorhynchus can be found in snails remains unclear. This study was conducted to understand the survival, development, and host use of 4 species of turtle acanthocephalans (Neoechinorhynchus chrysemydis, N. emydis, Neoechinorhynchus emyditoides, and Neoechinorhynchus pseudemydis) in their intermediate, paratenic, and definitive hosts. Using eggs recovered from adult acanthocephalans from naturally infected turtle hosts in Oklahoma, experimental infections were conducted in 2 species of ostracods and 1 species of freshwater snail. Ionomes also were generated for these acanthocephalans and their turtle definitive hosts to better understand resource competition. Upon ingestion by ostracods, eggs of all 4 species of acanthocephalans hatched in both ostracod species. However, no further acanthocephalan development occurred in Cypridopsis sp. ostracods, whereas all 4 acanthocephalan species grew and developed into infective juveniles over 28-32 days in Physocypria sp. ostracods. Of the 4 species of acanthocephalans, N. emydis grew faster early in ostracods and was the only species to survive and establish in snail hosts. This finding was supported by the observation of proboscis remnants in the feces of snails that ingested ostracods infected with N. chrysemydis, N. emyditoides, and N. pseudemydis. In contrast, when snails were exposed to eggs of all 4 acanthocephalan species, eggs were passed in the feces, resulting in no infections. The overall ionomic composition of adult acanthocephalans and their turtle hosts suggested that the hosts and parasites share host resources. However, specific elemental differences indicated that turtle acanthocephalans can accumulate heavy metals at concentrations higher than those in their hosts. Results of this study support previous field observations and hypotheses that (1) Physocypria sp. ostracods serve as intermediate hosts for N. chrysemydis, N. emydis, N. emyditoides, and N. pseudemydis; (2) N. emydis uses snail paratenic hosts, but snails are a non-viable route of transmission to turtles for N. chrysemydis, N. emyditoides, and N. pseudemydis; and (3) as adults, these 4 acanthocephalan species utilize their turtle definitive hosts similarly in terms of ionomic composition. The literature about mollusc-parasite interactions is focused on species affecting human health, such as trematodes on freshwater snails as intermediate hosts. Far less attention has been paid to parasite-snail interactions in terrestrial habitats. Here we present the first observation of a sciomyzid larva of Limnia unguicornis, parasitizing a vulnerable, tiny air-breading snail, Vertigo moulinsiana (strictly protected by Polish law). Sciomyzids are almost exclusively malacophagous but their biology and ecology are understudied. Intracellular protozoan parasites are the etiologic agents of important human diseases, like malaria, Chagas disease, toxoplasmosis, and leishmaniasis. Inside host cells, these parasites manipulate the host metabolism and intracellular trafficking for their own benefits and, inevitably, induce several stress response mechanisms. In this review, we discuss autophagy as a stress response mechanism that can be both (i) explored by these intracellular parasites to acquire nutrients and (ii) to restrict parasite proliferation and survival within host cells. We also discuss the immunomodulatory role of autophagy as a strategy to reduce inflammatory-mediated damage, an essential player in the pathophysiology of these parasitic diseases. Parasitoids often exhibit high flexibility in their development depending on stages of their host at the parasitism, yet little is known about the mechanism underlying such flexibility. In the study, we evaluated the larval development time of the parasitoid Exorista sorbillans (Diptera: Tachinidae) on the lepidopteran model insect Bombyx mori (Lepidoptera: Bombycidae). We found that the development duration of E. sorbillans larvae parasitizing on the late-developmental silkworms was significantly shorter than that of the larvae parasitizing on the early-developmental hosts. Intriguingly, the 2nd-3rd instar molting of parasitoid always occurred when the ecdysteroid titers in the host were increased to higher levels. Furthermore, inhibiting the release of ecdysteroids to parasitic abdomen by thorax-abdomen ligation of the host only repressed the 2nd-instar growth and molting of E. sorbillans larvae, but had no effect on their pupation. Meanwhile, the ecdysone synthesis and 20-hydroxyecdysone (20 E) signaling in larval parasitoids were impeded after ligation treatment. Moreover, exogenous 20 E application could largely rescue the defect in 2nd instar growth and molting through stimulating ecdysone synthesis and signaling in E. sorbillans. Our results indicate that the parasitoid requires the host ecdysteroids to stimulate 20 E signaling and the subsequent 2nd-instar growth and molting. The environment is the most important stratum in the epidemiological triad of a parasitic disease and any variations in the environmental factors may decide the dynamic occurrence and existence of different lifecycle stages of these parasites. The present study investigated the correlations between key biometeorological and demographical parameters with the incidence of different gastrointestinal parasites and hemoparasites among goats. Four hundred and thirty-two goats were screened for parasitic infection in a yearlong survey conducted from July 2022 to June 2023 in the Teaching Veterinary Clinical Complex, Mannuthy, Kerala, India. The weather parameters (T max , T min , RH, THI, and bright sunshine hours), air quality parameters [AQI, PM 2.5 , and PM 10 ], and demographic parameters (gender and age) were recorded along with the test positivity of different categories of gastrointestinal parasites and hemoparasites in goats by routine fecal sample examination and blood smear examination, respectively. The infection level was ranked based on the severity of the infection. The mean and daily variations in biometeorological parameters were calculated and the data were statistically analyzed to figure out the pertinent correlations in host-parasite-environment interaction patterns. High levels of parasitic infections with significant month-wise variations and climate-correlated peak infection patterns were noticed. The incidence of parasites was negatively correlated to all parameters except humidity, indicating more severe parasitism during monsoon months. The significant variations in the host-parasite interaction dynamics point towards the need for detailed explorations concerning the lifecycle of each specific parasite with a focus on the possible environment-favourable and resistant lifecycle stages. Parasitic plants have been viewed as pests since they are able to damage agricultural crops and forest trees. They establish vasculature connections with the hosts and withdraw the required nutritive resources. This study aimed to explore the physiological and biochemical effects of the parasitic plant Plicosepalus acacia on the host Tamarix aphylla. It was conducted on young fully expanded leaves from the uninfected and infected trees and the parasitic plant 'in situ'. The parasite had higher net photosynthetic assimilation rate (A), transpiration rate (E) and stomatal conductance (gs) compared to the host. Equally, it had two-fold greater water potential (Ψ) and osmotic potential (Ψs). It accumulated high amount of K, while it avoided accumulation of the most trace and ultratrace elements. Otherwise, parasitism seemed to increase A, WUE, water uptake and accumulation of the most major, trace and ultra-trace elements, however it reduced the accumulation of osmolytes at the infected plants. Based on UPLC-MS approach, P. acacia seemed to use a group of composites to interact with the host, including oleamide as a protector metabolite against host's defense system, carvone to establish vasculature connections with the host, cuminaldehyde to weaken growth and proliferation of the host, and caprolactam to weaken the distal part of the host. In contrast, the host T. aphylla could be used pipecolinic acid and nicotinamide to regulate systemic resistance and to defense against the parasite infection. Understanding the complex interactions between bacteriophages (phages) and bacteria within varied environmental niches is critical yet underexplored for improving microbe-assisted ecological restoration. This study investigates the influence of microhabitat heterogeneity within an abandoned mine on phage-bacteria interaction patterns, focusing on Pseudomonas-enriched bacterial communities. By isolating viral communities and purifying bacteria from soils of three distinct microhabitats, we assessed the regulatory role of environmental factors on these interactions, crucial for bacterial success in environmental applications. We characterized microhabitat variability by analyzing soil particle size fractions, minerals composition, and elemental content using X-ray diffraction and energy-dispersive X-ray analyses. 16S rRNA sequencing and cross-infection assays revealed that although bacterial communities across different microhabitats are taxonomically similar, their interaction patterns with phages are distinct. Phage communities showed nonselective infectivity across soil types, while bacterial communities exhibited selective adaptation, facilitating colonization across diverse microhabitats. Minerals such as mica, kaolinite, and hematite were found to increase phage infectivity, whereas mixed-layer clay correlated with early lysis. Additionally, higher levels of iron (Fe) and potassium (K) were linked to bacterial resistance strategies. Parasitism is a strong selective pressure, and its study is crucial for predicting the persistence of host species. Mantled howler monkeys are infected by the larvae of the bot fly Cuterebra baeri. This parasitosis produces myiasis and may have negative impacts on host health, although systematic information on the dynamics of this host-parasite relationship is very limited. Currently, all available information on infection patterns of C. baeri comes from a single mantled howler monkey population (Barro Colorado Island, Panama). Therefore, in this study we describe temporal variation in infection patterns for a newly mantled howler monkey population and analyze the relationship between climate and infection likelihood. We assessed the presence of C. baeri nodules in 17 adult individuals in Los Tuxtlas for 10 months through direct observation and compiled data on ambient temperature and rainfall. Most subjects had nodules during the study and there were no differences between sexes in the number of nodules. Nodules were usually located in the neck. Prevalence and abundance of nodules peaked thrice during the study (February, April, and September), a pattern that was very similar to that of parasitism intensity (February, April, and August). Incidence closely tracked these peaks, increasing before and decreasing after them. The likelihood of nodule appearance increased when both mean and minimum temperature decreased in the 24-21 prior days to nodule appearance. It also increased with decreased rainfall in the 5-2 prior days to nodule appearance. Although only three of the eight analyzed climate variables had a significant effect on parasitosis, these results suggest that climate may affect pupal development and the access of larvae to hosts. The zoophytophagous mirid predator Nesidiocoris tenuis and the ectoparasitoid Stenomesius japonicus are important biological control agents for several agricultural pests including the invasive leafminer, Phthorimaea absoluta, a destructive pest of Solanaceous crops especially tomato in sub-Saharan Africa. However, little is known about how feeding by N. tenuis can influence the tritrophic interactions in the tomato plant. Here, we tested the hypothesis that N. tenuis phytophagy would influence the tritrophic olfactory interactions between the host plant tomato and pest, predator, and parasitoid. In olfactometer assays, P. absoluta females and N. tenuis adults were both attracted to constitutive volatiles released by the tomato plant. Whereas females of P. absoluta avoided volatiles released by N. tenuis-infested plants, S. japonicus females and N. tenuis adults were attracted to the induced volatiles. In coupled gas chromatography-electroantennographic detection (GC-EAD) recordings of intact and N. tenuis-infested plant volatiles, antennae of P. absoluta and S. japonicus females both detected eight components, whereas N. tenuis adults detected seven components which were identified by GC-mass spectrometry (GC-MS) as terpenes and green leaf volatiles (GLVs). Dose-response olfactometer bioassays revealed that the responses of P. absoluta, N. tenuis, and S. japonicus varied with the composition and concentration of blends and individual compounds tested from N tenuis-induced volatiles. Females of P. absoluta showed no preference for an eight-component blend formulated from the individual repellents including hexanal, (Z)-3-hexenyl butanoate, and δ-elemene identified in the volatiles. On the other hand, S. japonicus females were attracted to an eight-component blend including the attractants (E)-2-hexenal, (Z)-3-hexenol, methyl salicylate, β-phellandrene, and (E)-caryophyllene. Likewise, N. tenuis adults were attracted to a seven-component blend including the attractants β-phellandrene, δ-elemene, and (E)-caryophyllene identified in the volatiles. We studied spatial variation in the effects of environment and network size on species positions and roles in multiple flea-mammal networks from four biogeographic realms. We asked whether species positions (measured as species strength [SS], the degree of interaction specialization [d'], and the eigenvector centrality [C]) or the roles of fleas and their hosts in the interaction networks: (a) are repeatable/conserved within a flea or a host species; (b) vary in dependence on environmental variables and/or network size; and (c) the effects of environment and network size on species positions or roles in the networks depend on species traits. The repeatability analysis of species position indices for 441 flea and 429 host species, occurring in at least two networks, demonstrated that the repeatability of SS, d', and C within a species was significant, although not especially high, suggesting that the indices' values were affected by local factors. The majority of flea and host species in the majority of networks demonstrated a peripheral role. A value of at least one index of species position was significantly affected by environmental variables or network size in 41 and 36, respectively, of the 52 flea and 52 host species that occurred in multiple networks. High species diversity in a community may reduce the risk of infectious disease, termed the dilution effect. However, the generality of the dilution effect in different disease systems remains controversial as both host competence and behaviors of hosts may play roles in dilution or amplification of disease. Using the goldfish (Carassius auratus)-monogenean ectoparasite (Gyrodactylus kobayashii) system, effects of host competence and schooling behavior on parasite transmission were investigated while holding focal host density constant. Following competency tests of 12 fish species as potential hosts for the parasite, infection by G. kobayashii was determined on fins of goldfish mixed with each of three different species based on their level of host competence, including Prussian carp, Carassius gibelio (low competence), grass carp, Ctenopharyngodon idellus (non-competent), swordtail, Xiphophorus helleri (non-competent), and the four species combined. Compared with mean abundance (85.8 ± 25.1) on goldfish in the control group, the mean abundance on goldfish decreased significantly when paired with 10 Prussian carp (30.0 ± 16.5), but did not differ significantly when paired with 10 swordtail (70.0 ± 22.2), 10 grass carp (116.1 ± 33.2), or the multi-species of three Prussian carp, four grass carp and three swordtail (75.9 ± 30.8) during the 11-day experiment. The parasite was also found on the Prussian carp in the Prussian carp group and the multi-species group at a mean abundance of 7.1 and 10.9, respectively. Video recording showed that the school of goldfish mixed well with the Prussian carp, while they maintained separation from the grass carp and swordtail when mixed together. The distance between goldfish increased, and swimming speed and contact time decreased with the additional of other fish species for all groups. The results suggested that the presence of a low-competence host in sufficient numbers was a necessary condition for a dilution effect due to encounter reduction, and the dilution effect may also be enhanced by changes in schooling behavior of goldfish in the presence of low competence hosts. Parasite-mediated selection is widely believed to play a crucial role in maintaining the diversity of the major histocompatibility complex (MHC) genes, which is thought to be maintained through heterozygote advantage, rare-allele advantage, and fluctuating selection. However, the relationship between parasite pressure and MHC diversity has yielded inconsistent findings. These inconsistencies may arise from the influence of environmental factors and individual variations in traits on host-parasite interactions. To address these issues, our study extensively investigated populations of striped hamsters inhabiting regions characterized by environmental heterogeneity. The primary objective was to examine the universality of parasite-mediated selection mechanisms. Our observations revealed the presence of multiple parasite infections, accompanied by spatial and temporal variations in parasite communities and infection patterns among individual hamsters. Specifically, the temperature was found to influence all four parasite indices, while the presence of gamasid mites and parasite richness decreased with increasing precipitation. We also noted significant seasonal variation in parasite dynamics. Moreover, a significant sexual dimorphism was observed with males exhibiting a considerably higher parasite burden compared to their female counterparts. Lastly, we identified the maintenance of MHC polymorphism in striped hamsters as being driven by the heterozygote advantage and fluctuating selection mechanisms. The intestinal helminth Ascaris lumbricoides infects over 800 million people. Infections are often chronic and immunity is not sterilizing due to host-immune modulation, therefore reinfection is common after antihelmintic treatment. We have previously demonstrated a role for Ascaris spp. extracellular vesicles (EVs) in host immune modulation but whether EVs are recognized by the adaptive immune system and are present systemically in the host remains unknown. Therefore, we employed a well-established trickle infection model in pigs to mimic natural Ascaris infection in humans. EVs were isolated from adult Ascaris suum followed by immunoblotting of EV and EV-depleted secretory fractions using plasma from infected and uninfected pigs. Next, EVs were isolated from pig plasma at day 56 post first infection and subjected to deep small RNAseq analysis. RNAs were aligned to A. suum and Sus scrofa miRNA complements to detect A. suum EVs and elucidate the host EV micro RNA (miRNA) response to infection, respectively. Infection generates robust antibody responses against A. suum EVs that is distinct from EV-depleted fractions. However, A. suum miRNAs were not detectable in EVs from the peripheral blood. Subsequently, supplemental feeding can affect both the immunological function of individuals and transmission dynamics among individuals and species. The magnitude of the effect supplemental feeding has on disease is likely to vary across time and space with the nutritional demands of hosts. However, the broad temporal or spatial scale effects of supplementation are poorly understood. Recently, Knutie et al. (2024) introduced their citizen science program, the Nest Parasite Community Project, a broadscale coordinated effort by scientists and the public to monitor box nesting wild birds and their ectoparasites across the eastern United States. The authors amassed an impressive 4-year data set with hundreds of nests spanning the entire US breeding range of Eastern bluebirds (Sialia sialis). In the first study to come from the project, the authors demonstrate that the effects of food supplementation on host-parasite interactions vary across time and space and do not consistently influence host-parasite outcomes, highlighting that host-parasite interactions are often context dependent and influenced by many environmental factors (e.g. weather and habitat quality). The authors also found that supplemental feeding increases host fitness, regardless of parasitism. Background: Climate change is driving increased extreme weather events that can impact ecology by moderating host-pathogen interactions. To date, few studies have explored how cold snaps affect disease prevalence and proliferation. Using the Daphnia magna-Ordospora colligata host-parasite system, a popular model system for environmentally transmitted diseases, the amplitude and duration of cold snaps were manipulated at four baseline temperatures, 10 days post-exposure, with O. colligata fitness recorded at the individual level., Results: Cold snaps induced a fivefold increase or a threefold decrease in parasite burden relative to baseline temperature, with complex nuances and varied outcomes resulting from different treatment combinations. Both amplitude and duration can interact with the baseline temperature highlighting the complexity and baseline dependence of cold snaps. Furthermore, parasite fitness, i.e., infection prevalence and burden, were simultaneously altered in opposite directions in the same cold snap treatment., Conclusions: We found that cold snaps can yield complicated outcomes that are unique from other types of temperature variation (for example, heatwaves). Palpita forficifera Munroe, 1959 (Lepidoptera: Crambidae) is the main pest of the olive tree (Olea europaea L., Oleaceae) in Brazil and its management has been difficult, as there are few products recommended for its control. This study aimed to evaluate the population dynamics of P. forficifera and the entomofauna of parasitoid eggs and larvae in three olive orchards under different cultivation systems, in Rio Grande do Sul state, Brazil. The study was carried out from October to May, in the 2017/18 and 2018/19 harvests, in olive orchards of different cultivars in the municipalities of Pelotas and Rio Grande, state of Rio Grande do Sul, Brazil. Population dynamics of P. forficifera varied according to the agricultural season, the months sampled, and the orchards (sites) evaluated. The highest infestation reached around 60% of the shoots in some months, depending on the orchard evaluated. During the pest infestation period, the occurrence of the egg parasitoid Trichogramma foersteri Takahashi, 2021 (Hymenoptera: Trichogrammatidae) was observed along with three larval parasitoids belonging to the genera Dolichogenidea Viereck, 1911, Hymenochaonia Dalla Torre, 1898 (Hymenoptera: Braconidae) and Temelucha Förster, 1869 (Hymenoptera: Ichneumonidae). Temelucha hilux Gauld, 2000 is recorded for the first time in association with a host, and its distribution in the country is extended to the southern region, from Brazil. Due to the scarcity of information on pest management, the natural occurrence of natural enemies in crops is of paramount importance in helping to manage P. forficifera in the field. The population dynamics of P. forficifera is also influenced by the parasitoid community, made up of at least four species of parasitic Hymenoptera. Therefore, strategies aimed at managing P. forficifera must be well developed in order to maintain and increase natural biological control in the field.

To understand the ecoepidemiology of fleas and their interactions with domestic and wild animals, it is necessary to decode the cycles of flea abundance in relation to host species and environmental factors. The province of Misiones, known for its biodiversity and land use changes, provides an excellent study location. We assessed the diversity, prevalence, abundance, and parasitic intensity of fleas collected from 1153 rodents and opossums in various environments in the Upper Paraná Atlantic Forest of Argentina, considering phytogeography and land use. Our findings were consistent with previous studies conducted in humid regions of Argentina and the Atlantic Forest in Brazil. Hosts with high population density exhibited greater flea prevalence and abundance, and widely distributed hosts showed higher flea species richness and diversity. Significant similarities in flea similarity indices among phytogeographic areas, especially in forest. Environments with an anthropogenic gradient exhibited marked dissimilarities, with higher species richness and diversity in less disturbed sites. These parameters may be influenced by the presence of polyxenous Ctenocephalides, Polygenis, and Adoratopsylla fleas co-infesting opossum in urban areas. These findings provide valuable insights into the ecoepidemiology of fleas, their relationships with host species, and the impact of environmental factors in a strategic Atlantic forest ecoregion.

Background: Schistosoma mansoni was introduced from Africa to the Americas during the transatlantic slave trade and remains a major public health problem in parts of South America and the Caribbean. This study presents a comprehensive comparative analysis of three S. mansoni strains with different geographical origins-from Liberia, Belo Horizonte and Puerto Rico. We demonstrated significant variation in virulence and host-parasite interactions., Methods: We investigated the phenotypic characteristics of the parasite and its eggs, as well as the immunopathologic effects on laboratory mouse organ systems., Results: Our results show significant differences in worm morphology, worm burden, egg size, and pathologic organ changes between these strains. The Puerto Rican strain showed the highest virulence, as evidenced by marked liver and spleen changes and advanced liver fibrosis indicated by increased collagen content. In contrast, the strains from Liberia and Belo Horizonte had a less pathogenic profile with less liver fibrosis. We found further variations in granuloma formation, cytokine expression and T-cell dynamics, indicating different immune responses., Conclusion: Our study emphasizes the importance of considering intra-specific variations of S. mansoni for the development of targeted therapies and public health strategies. For many years, parasite ecologists have debated the relative importance of phylogeny and ecology as drivers of parasite community structure. Here, we address this issue using data on the metazoan ectoparasite communities of different flatfish species. Twenty species of flatfish were collected along the continental shelf of the Southern Gulf of Mexico and examined for ectoparasites. Eight flatfish species were parasitized by at least one ectoparasite species. In total, 326 ectoparasites, representing 11 species (4 monogeneans, 4 copepods, 1 isopod, 1 branchiurid, and 1 leech) were removed from 1622 hosts examined. The highest prevalence (37.5%) occurred in Bomolochus sp1 from Trinectes maculatus, while the lower (0.1%) for Argulus sp., Gnathia sp. and Trachellobdella lubrica from Cyclopsetta chittendeni. Changes in the ectoparasite community structure per host species and region were evaluated using a Permutational Multivariate Analysis of Variance and represented by a multidimensional scaling analysis. Significant differences in the parasite species composition among regions and hosts were detected, but no significant interaction between regions and hosts occurred. A multivariate pairwise t-test detected significant differences in the parasite infracommunities between the Yucatan Shelf and the other two regions; in addition, significant differences were detected between C. chittendeni and the Syacium species as well as between Ancylopsetta dilecta and Syacium papillosum. Background: India's oilseed economy falls short of self-sufficiency and is supplemented by huge imports every year. Increasing national productivity of the major oilseeds is confronted with yield losses due to diverse biotic and abiotic stresses. The productivity of Indian mustard (Brassica juncea Linnaeus), belonging to the family Brassicaceae, is significantly reduced due to damage caused by mustard aphids (Lipaphis erysimi Kaltenbach, Hemiptera: Aphididae). Rapid colonization by the nymphs makes it difficult to protect the crop through agrochemicals. Aphids release effector molecules to modulate the host-defence responses. Glucosinolates (GSLs) extensively found in Brassicaceae family, are hydrolysed by myrosinase into toxic compounds that deter herbivore insects., Methods: Here, we investigated the differential activation of the glucosinolate-myrosinase pathway in mustard manifesting susceptibility and resistance to different aphid species. Mustard plants were challenged by two different aphid species mustard aphid and cowpea aphid (Aphis craccivora Koch, Hemiptera: Aphididae) leading to complete host-susceptibility in one case and resistance in the other, respectively. Differential regulation of the GSL biosynthetic pathway and myrosinase activity was assessed by gene expression study and ultra-performance liquid chromatography quadrupole time-of-flight tandem mass spectrometry (UPLC- QToF-ESL-MS)., Results: Gene expression study identified selective transcriptional attenuation of the key GSL biosynthetic and myrosinase gene in mustard when challenged with mustard aphid. In contrary, the activation of GSL biosynthetic genes in conjunction with myrosinase at the transcriptional level was profound in mustard, when challenged with cowpea aphid. UPLC-MS analysis showed higher turnover in the hydrolysis of glucosinolates by myrosinase which led to concomitant generation of glucose as byproduct in response to cowpea aphid in mustard plants., Conclusion: GSL-myrosinase pathway is specifically attenuated by the successful aphid species in mustard and thus plays a pivotal role in determining the outcome of the B. juncea-aphid interaction. In both mice and humans, Type II interferon gamma (IFNγ) is crucial for the regulation of Toxoplasma gondii ( T. gondii ) infection, during acute or chronic phases. To thwart this defense, T. gondii secretes protein effectors hindering the host's immune response. For example, T. gondii relies on the MYR translocon complex to deploy soluble dense granule effectors (GRAs) into the host cell cytosol or nucleus. Recent genome-wide loss-of-function screens in IFNγ-primed primary human fibroblasts identified MYR translocon components as crucial for parasite resistance against IFNγ-driven vacuole clearance. However, these screens did not pinpoint specific MYR-dependent GRA proteins responsible for IFNγ signaling blockade, suggesting potential functional redundancy. Our study reveals that T. gondii depends on the MYR translocon complex to prevent parasite premature egress and host cell death in human cells stimulated with IFNγ post-infection, a unique phenotype observed in various human cell lines but not in murine cells. Intriguingly, inhibiting parasite egress did not prevent host cell death, indicating this mechanism is distinct from those described previously. Genome-wide loss-of-function screens uncovered TgIST, GRA16, GRA24, and GRA28 as effectors necessary for a complete block of IFNγ response. GRA24 and GRA28 directly influenced IFNγ-driven transcription, GRA24's action depended on its interaction with p38 MAPK, while GRA28 disrupted histone acetyltransferase activity of CBP/p300. Given the intricate nature of the immune response to T. gondii , it appears that the parasite has evolved equally elaborate mechanisms to subvert IFNγ signaling, extending beyond direct interference with the JAK/STAT1 pathway, to encompass other signaling pathways as well.IMPORTANCE Toxoplasma gondii , an intracellular parasite, affects nearly one-third of the global human population, posing significant risks for immunocompromised patients and infants infected in utero . In murine models, the core mechanisms of IFNγ-mediated immunity against T. gondii are consistently preserved, showcasing a remarkable conservation of immune defense mechanisms. In humans, the recognized restriction mechanisms vary among cell types, lacking a universally applicable mechanism. This difference underscores a significant variation in the genes employed by T. gondii to shield itself against the IFNγ response in human vs murine cells. Here, we identified a specific combination of four parasite-secreted effectors deployed into the host cell nucleus, disrupting IFNγ signaling. Mononuclear phagocytes facilitate the dissemination of the obligate intracellular parasite Toxoplasma gondii . Here, we report how a set of secreted parasite effector proteins from dense granule organelles (GRA) orchestrates dendritic cell-like chemotactic and pro-inflammatory activation of parasitized macrophages. These effects enabled efficient dissemination of the type II T. gondii lineage, a highly prevalent genotype in humans. We identify novel functions for effectors GRA15 and GRA24 in promoting CCR7-mediated macrophage chemotaxis by acting on NF-κB and p38 mitogen-activated protein kinase signaling pathways, respectively, with contributions by GRA16/18 and counter-regulation by effector TEEGR. Furthermore, GRA28 boosted chromatin accessibility and GRA15/24/NF-κB-dependent transcription at the Ccr7 gene locus in primary macrophages. In vivo , adoptively transferred macrophages infected with wild-type T. gondii outcompeted macrophages infected with a GRA15/24 double mutant in migrating to secondary organs in mice. Trichomonas vaginalis causes trichomoniasis, the most common non-viral sexually transmitted disease worldwide. As an extracellular parasite, adhesion to host cells is essential for the development of infection. During attachment, the parasite changes its tear ovoid shape to a flat ameboid form, expanding the contact surface and migrating through tissues. Here, we have identified a novel structure formed at the posterior pole of adherent parasite strains, resembling the previously described uropod, which appears to play a pivotal role as an anchor during the attachment process. Moreover, our research demonstrates that the overexpression of the tetraspanin T. vaginalis TSP5 protein (TvTSP5), which is localized on the cell surface of the parasite, notably enhances the formation of this posterior anchor structure in adherent strains. Finally, we demonstrate that parasites that overexpress TvTSP5 possess an increased ability to adhere to host cells, enhanced aggregation and reduced migration on agar plates. The obligate intracellular parasite Toxoplasma gondii can infect and replicate in any warm-blooded cell tested to date, but much of our knowledge about T. gondii cell biology comes from just one host cell type: human foreskin fibroblasts (HFFs). To expand our knowledge of host-parasite lipid interactions, we studied T. gondii in intestinal epithelial cells, the first site of host-parasite contact following oral infection and the exclusive site of parasite sexual development in feline hosts. We found that highly metabolic Caco-2 cells are permissive to T. gondii growth even when treated with high levels of linoleic acid (LA), a polyunsaturated fatty acid (PUFA) that kills parasites in HFFs. Caco-2 cells appear to sequester LA away from the parasite, preventing membrane disruptions and lipotoxicity that characterize LA-induced parasite death in HFFs. Several aquatic ecosystems present favorable conditions for the establishment of freshwater gastropod populations in Maranhão (Brazil). However, research on these snails in this Brazilian state is more focused on Baixada Maranhense, an endemic area for schistosomiasis in Maranhão. Considering the need to expand information on the occurrence of limnic gastropods in Maranhão, a qualitative study was carried out in the Microregion of Lençóis Maranhenses. The capture of freshwater snails was carried out between June 2016 and November 2019. Were obtained 3,244 specimens of freshwater gastropods: 2,372 specimens of Biomphalaria straminea (Dunker, 1848); 17 specimens of Biomphalaria schrammi (Crosse, 1864); 19 specimens of Drepanotrema lucidum (Pfeiffer, 1939); 115 specimens of (Hydrobiidae; Rissooidea); 24 specimens of Physa marmorata Guilding, 1828; 73 specimens of Pomacea maculata (Perry, 1810); 613 specimens of Pomacea sp. (Perry, 1810). Were also found 11 shells of Biomphalaria glabrata (Say, 1818). The greatest richness of snails was observed in Tutóia. The most abundant species during the study was B. straminea (73,45%). Pomacea sp. was the most representative species with constant frequency (57,15%) in the Lençóis Maranhenses Microregion, present in lentic and lotic ecosystems. The forecast maps indicate the possibility of occurrence and establishment of freshwater gastropods species throughout the investigated region. The presence of gastropods that are hosts of parasites represents a risk to human health, especially in the Lençóis Maranhenses area, which is one of Brazil's tourist destinations. Studies on the occurrence and distribution of limnic gastropods are of great relevance for understanding the conservation status of existing species. Furthermore, they are necessary to monitor snails that act as parasite hosts.

Global climate warming and frequent extreme low-temperature events have made it essential to investigate the impact of low temperatures on parasitic wasps to protect and strengthen farmland biodiversity, which in turn enhances the biological control potential of natural enemies such as parasitic wasps. We systematically examined how low-temperature stress affects the parasitic functional response of Trichopria drosophilae to Drosophila suzukii (Diptera: Drosophilidae) pupae. Our findings indicate that the parasitic behavior of T. drosophilae towards D. suzukii pupae aligns with the Holling II functional response model following exposure to different temperatures. Within the temperature range of 8 °C to -8 °C, lower temperatures correlated decreased instantaneous attack rate of T. drosophilae and an increase in processing time. The search constant Q initially increased and then decreased with declining temperatures. Short-term low-temperature stress negatively impacted the parasitic and searching abilities of T. drosophilae but did not alter its parasitic functional response model. Notably, short-term low-temperature stress had minimal effects on the water content, protein content, and total sugar content of male and female T. drosophilae adults. However, as temperatures decreased, the activities of key enzymes, including GAPDH, SOD, T-AOC, and malondialdehyde (MDA), exhibited an initial increase followed by a decrease. Conversely, the activities of LDH and HOAD decreased, while the activities of CAT and POD increased. Further study on the effect of short-term low temperature on T. drosophilae can provide a research basis for the large-scale production and low-temperature refrigeration technology of T. drosophilae, and provide a scientific basis for its efficient use in the field., (© The Author(s) 2024. Trichogramma spp. wasps are egg parasitoids with a long history of mass rearing for augmentation biocontrol programs in field crop and orchard landscapes. Supplementary nutrition can improve the longevity, fecundity, and biocontrol efficacy of parasitoids. To improve the production efficiency and parasitism performance of Trichogramma dendrolimi Matsumura (Hymenoptera: Trichogrammatidae), the present study screened and examined the potential supplementary nutrients for this biological control agent. Dietary supplementation with a 10% sucrose solution significantly increased wasp longevity and parasitism potential of T. dendrolimi on host eggs, but provision of pollen did not provide additional benefits. Laboratory and greenhouse cage tests demonstrated that wasp access to soybean aphid Aphis glycines Matsumura (Hemiptera: Aphididae) honeydew, comprised primarily of melezitose and trehalose, improved T. dendrolimi longevity and parasitism. In conclusion, provision of a 10% sucrose solution to adult wasps will enhance the mass-rearing efficiency of T. dendrolimi; furthermore, field release of T. dendrolimi by plant vectors bearing honeydew-producing aphids holds promise for improving the biocontrol efficacy of T. dendrolimi., (© The Author(s) 2024. The stink bugs Acrosternum arabicum Wagner and Brachynema germari Kolenati (Hemiptera: Pentatomidae) damage developing pistachio nuts in Iran. Ooencyrtus egeria Huang and Noyes (Hymenoptera: Encyrtidae) parasitizes eggs of both species, alternating between them seasonally. We compared the biological and life history attributes of O. egeria reared for 3 generations on each host species with those that had been transferred to the alternate host for 2 generations. We also tested female responses to host egg density. Eggs of B. germari were parasitized at higher rates and yielded faster wasp development, heavier female progeny with greater fecundity, and a more female-biased sex ratio, than eggs of A. arabicum. Rearing on A. arabicum did not diminish wasp fitness on B. germari, and the F1 progeny of wasps switched to A. arabicum inherited some beneficial parental effects from rearing on B. germari. Regardless of host species, females parasitized fewer eggs in 8 h as density increased from 10 to 30 eggs, but progeny sex ratio (% female) increased. The anomalous response to host density appears to arise from a combination of behavioral and egg load constraints. Females mature only 10-12 eggs per day. Carefully inspect egg clusters before selecting the most suitable eggs for oviposition, a more time-consuming process for large clusters. Our results indicate that rearing O. egeria on eggs of A. arabicum, which are more convenient and cost-effective to produce than those of B. germari, will not diminish its performance on B. germari following augmentative release, beyond a slight reduction in female fecundity., (© The Author(s) 2024. Physiological mechanisms underlying relationships between the trematode parthenitae and their molluscan hosts are poorly understood. In this study, we estimated the cardiac function of gastropods Littorina littorea L. infected with Himasthla elongata and Cryptocotyle lingua under laboratory conditions and in situ. The heart rate (HR) of the infected periwinkles was significantly lower than in the uninfected ones. HR of uninfected periwinkles in the natural environment followed a clear tidal pattern, with high values at high tide and low values at low tide. However, HR in most of the infected molluscs did not follow this rhythm. The decrease in HR was more pronounced in the periwinkles infected with C. lingua than with H. elongata. Thus, infection with C. lingua had a greater effect on the physiology and metabolic rate of periwinkles than infection with H. elongata.

Infectious diseases stem from disrupted interactions among hosts, parasites, and the environment. Both abiotic and biotic factors can influence infection outcomes by shaping the abundance of a parasite's infective stages, as well as the host's ability to fight infection. However, disentangling these mechanisms within natural ecosystems remains challenging. Here, combining environmental DNA analysis and niche modelling at a regional scale, we uncovered the biotic and abiotic drivers of an infectious disease of salmonid fish, triggered by the parasite Tetracapsuloides bryosalmonae. We found that the occurrence and abundance of the parasite in the water-i.e., the propagule pressure- were mainly correlated to the abundances of its two primary hosts, the bryozoan Fredericella sultana and the fish Salmo trutta, but poorly to local abiotic environmental stressors. In contrast, the occurrence and abundance of parasites within fish hosts-i.e., proxies for disease emergence-were closely linked to environmental stressors (water temperature, agricultural activities, dams), and to a lesser extent to parasite propagule pressure. These results suggest that pathogen distribution alone cannot predict the risk of disease in wildlife, and that local anthropogenic stressors may play a pivotal role in disease emergence among wild host populations, likely by modulating the hosts' immune response. Many factors affect an organism's microbiome including its environment, proximity to other organisms, and physiological condition. As filter feeders, bivalves have highly plastic microbiomes that are especially influenced by the surrounding seawater, yet they also maintain a unique core set of microbes. Using 16S ribosomal RNA sequencing, we characterized the bacterial microbiomes of four species of bivalves native to the Mid-Atlantic East Coast of North America: Crassostrea virginica, Macoma balthica, Ameritella mitchelli , and Ischadium recurvum and assessed the impact of their external environment, internal parasites, and size on their microbial communities. We found significant differences in bacterial amplicon sequence variants (ASVs) across species, with each species harboring a core ASV present across all individuals. We further found that some C. virginica co-cultured with I. recurvum had high abundances of the I. recurvum core ASV. We identified ASVs associated with infection by the parasites Perkinsus marinus and Zaops ostreum as well others associated with bivalve size. Several of these ASV are candidates for further investigation as potential probiotics, as they were found positively correlated with bivalve size and health. This research represents the first description of the microbiomes of A. mitchelli, I. recurvum , and M. balthica . Sleep is a complex and conserved biological process that affects several body functions and behaviors. Evidence suggests that there is a reciprocal interaction between sleep and immunity. For instance, fragmented sleep can increase the probability of parasitic infections and reduce the ability to fight infections. Moreover, viral and bacterial infections alter the sleep patterns of infected individuals. However, the effects of macro-parasitic infections on sleep remain largely unknown, and measuring sleep in non-model organisms remains challenging. In this study, we investigated whether macro-parasite infections could alter sleep-like behavior of their hosts. We experimentally infected three-spined sticklebacks (Gasterosteus aculeatus), a freshwater fish, with the tapeworm Schistocephalus solidus and used a hidden Markov model to characterize sleep-like behavior in sticklebacks. One to four days after parasite exposure, infected fish showed no difference in sleep-like behavior compared with non-exposed fish, and fish that were exposed-but-not-infected only showed a slight reduction in sleep-like behavior during daytime. Twenty-nine to 32 days after exposure, infected fish showed more sleep-like behavior than control fish, while exposed-but-not-infected fish showed overall less sleep-like behavior. Using brain transcriptomics, we identified immune- and sleep-associated genes that potentially underlie the observed behavioral changes. These results provide insights into the complex association between macro-parasite infection, immunity, and sleep in fish and may thus contribute to a better understanding of reciprocal interactions between sleep and immunity., (© 2024. Telenomus remus (Hymenoptera: Scelionidae) is an efficient parasitoid of Spodoptera eggs. However, biological control programs require taxonomic, bioecological and behavioral studies of biological agents. Although the performance of T. remus in pest control has been evaluated, little is known about its behavioral aspects that can influence IPM tactics. The aim of this study was therefore to study the parameters related to the oviposition behavior of T. remus on Spodoptera frugiperda (Lepidoptera: Noctuidae) eggs. The experiment was conducted in the laboratory, where 17 females were transferred individually to arenas containing a mass of S. frugiperda eggs. Oviposition behavior was recorded for 30 minutes. The average walking speed of the females was 0.116 mm.s-1 and the total distance covered was 203.3 mm. The females remained on the egg masses for an average of 16.7 min, which corresponds to more than half of the total time. The average number of parasitized eggs was 15.1 per female in 30 minutes. Females with larger antennae had lower values for average speed and total distance covered. The results of the parameters evaluated show the standard oviposition behavior of T. remus females on S. frugiperda eggs and help us to better understand the species.

This study investigated the role of host traits, habitat, and sampling season on the prevalence and intensity of parasites in three species of platycephalid fishes. Three host species sampled were dusky flathead (Platycephalus fuscus, n = 3), blue-spotted flathead (Platycephalus caeruleopunctatus, n = 38), and tiger flathead (Platycephalus richardsoni, n = 59). A total of 14 metazoan parasite species were collected over 15 months, between July 2020 and September 2021. The parasites found included a chondracanthid copepod, adult hemiuroidean trematodes, cestode plerocercoids, larval and adult acanthocephalans, larval and adult nematodes, and an unknown species of helminth. General linear models were used to assess the relationship between host traits and sampling season with parasite infection intensity. The infection intensity of an unidentified plerocercoid species in P. caeruleopunctatus was found to be significantly associated with both sampling season and the interaction of host mass with sex. In P. richardsoni the infection intensity of the acanthocephalan, Corynosoma sp. was found to correlate with sampling season. Insect gut microbes play important roles in digestion, metabolism, development, and environmental adaptation. Parasitoid wasps are one of the most important biological control agents in pest control, while the gut microbial species compositions and the associated functions have been poorly investigated. Two endoparasitoid wasps, Cotesia vestalis and Diadromus collaris , parasitize the larval stage and pupal stage of the diamondback moth, Plutella xylostella , respectively. Using whole-genome shotgun metagenomic sequencing, we characterized the gut microbial composition, diversity, and potential functional roles associated with the two parasitoid wasp larvae. The results reveal that Proteobacteria and Firmicutes are the dominant phyla in the gut of C. vestalis and D. collaris larvae, with Rhizobium and Enterococcus being the dominant genera. The putative microbial functions associated with the two parasitoid wasps might play a virtual role in assisting in consuming the host's nutritional composition. The enriched CAZymes family genes are primarily involved in the degradation and synthesis of chitin. Despite the richness of microbial species and communities, the microbes species and the microbial community structure exhibit significant similarity between the two parasitoid wasps and between the parasitoid wasp and the host P. xylostella . Notably, the prevalence of the genus Enterococcus shared among them suggests a possible link of gut microbes between the host and their associated parasitoids. Our study offers insights into the gut microbe-based interactions between the host and parasitoid wasps for the first time, potentially paving the way for the development of an ecologically friendly biocontrol strategy against the pest P. xylostella .IMPORTANCEEndoparasitoid wasps spend the majority of their lifespan within their host and heavily rely on the host's nutrition for survival. There is limited understanding regarding the composition and physiological impacts of gut microbial communities in parasitoid wasps, particularly during the larval stage, which is directly linked to the host. Based on a thorough characterization of the gut microbe and comprehensive comparative analysis, we found the microbial species of the larval parasitoid wasp Cotesia vestalis and the pupal parasitoid wasp Diadromus collaris were similar, sharing 159 genera and 277 species, as were the microbial community structure. Certain of the dominant microbial strains of the two parasitoid wasps were similar to that of their host Plutella xylostella larvae, revealing host insect may affect the microbial community of the parasitoid wasps. Leishmaniasis represents a severe global health problem. In the last decades, there have been significant challenges in controlling this disease due to the unavailability of licensed vaccines, the high toxicity of the available drugs, and an unrestrained surge of drug-resistant parasites, and human immunodeficiency virus (HIV)-Leishmania co-infections. Leishmania spp. preferentially invade macrophage lineage cells of vertebrates for replication after subverting cellular functions of humans and other mammals. These early events in host-parasite interactions are likely to influence the future course of the disease. Thus, there is a continuing need to discover a simple cellular model that reproduces the in vivo pathogenesis. Acanthamoeba spp. are non-mammalian phagocytic amoeba with remarkable similarity to the cellular and functional aspects of macrophages. We aimed to assess whether the similarity reported between macrophages and Acanthamoeba spp. is sufficient to reproduce the infectivity of Leishmania spp. Herein, we analyzed co-cultures of Acanthamoeba castellanii or Acanthamoeba polyphaga with Leishmania infantum, Leishmania amazonensis, Leishmania major, and Leishmania braziliensis. Light and fluorescence microscopy revealed that the flagellated promastigotes attach to the A. castellanii and/or A. polyphaga in a bipolar and or random manner, which initiates their uptake via pseudopods. Once inside the cells, the promastigotes undergo significant changes, which result in the obligatory amastigote-like intracellular form. There was a productive infection with a continuous increase in intracellular parasites. However, we frequently observed intracellular amastigotes in vacuoles, phagolysosomes, and the cytosol of Acanthamoeba spp. Our findings corroborate that Leishmania spp. infects Acanthamoeba spp. and replicates in them but does not cause their rapid degeneration or lysis. Overall, the evidence presented here confirms that Acanthamoeba spp. have all prerequisites and can help elucidate how Leishmania spp. infect mammalian cells. Parasitism involves diverse evolutionary strategies, including adaptations for blood feeding, which provides essential nutrients for growth and reproduction. Sparicotyle chrysophrii (Polyopisthocotyla: Microcotylidae), an ectoparasitic flatworm, infects the gills of gilthead seabream ( Sparus aurata ), significantly affecting fish health, welfare and Mediterranean cage farm profitability. Despite its impact, limited information exists on its feeding behaviour. This study demonstrates the presence of blood and exogenous haem groups in S. chrysophrii and explores its digestive tract using light and electron microscopy, elucidating its internal morphology and spatial arrangement. Elemental analysis of the digestive haematin cells shows residual oxidized haem depots as haematin crystals. Additionally, we studied the impact of the blood feeding on the host by estimating the average volume of blood intake for an adult parasite (2.84 ± 2.12µl·24h -1 ) and we described the significant drop of the plasmatic free iron levels in infected hosts. Overall, we demonstrate the parasite's reliance on its host blood, the parasite's buccal and digestive morphological adaptations for blood feeding and the provoked effect on the fish host's health.

Consumers can influence the competitive outcomes of prey species in various ways. Modern coexistence theory predicts that consumers can promote prey coexistence by preferably targeting a competitively superior one, thereby reducing fitness differences. However, previous studies yielded mixed conclusions. In this study, we tested the hypothesis that a parasitic annual plant, Cuscuta campestris, facilitates the coexistence of two common annual plants. We performed field surveys and parasitism experiments to parameterize a plant competition dynamics model. The model suggested a competition-defence tradeoff: the legume Lespedeza striata was a better competitor than the grass Setaria faberi, while it was more susceptible to the parasite. Moreover, an empirical host-parasite dynamics model, extended from the plant competition model, predicted their coexistence within broad, biologically reasonable ranges of parameters. We identified two gall-inducing wasp species infesting eucalypts leaves, including an undescribed species, Ophelimus cracens sp. nov., and Epichrysocharis burwelli , which is a new record for Taiwan. The major hosts of O. cracens were Eucalyptus grandis , Eu. urophylla , and Eu. camaldulensis (Myrtaceae) . We observed failed galls of O. cracens at an early stage on Eu. amplifolia and Corymbia maculata . All O. cracens adults discovered on infected leaves were females, and four parasitoid species were identified in samples collected from two cities, three counties, and four municipalities across Taiwan. Epichrysocharis burwelli was exclusively found on C. citriodora in Hsinchu, Nantou, and Tainan Cities. This marks the first record of Ep. burwelli in Asia, accompanied by the identification of one parasitoid species. The adult longevity of adults which emerged from their major hosts, when provided with honey-water solution, was 5.5 days for O. cracens and 5.7 days for Ep. burwelli . Recognizing the potential damage by these wasps on Eucalyptus production areas in the absence of parasitoids, further investigations of their biology and control are warranted.

Two key epidemiological parameters, prevalence and mean intensity of infection, together capture the abundance of macroparasite populations, the strength of density-dependent effects they experience, their potential impact on host population dynamics and the selective pressures they exert on their hosts. Yet, the drivers of the extensive variation observed in prevalence and mean intensity of infection, even among related parasite taxa infecting related hosts, remain mostly unknown. We performed phylogenetically grounded Bayesian modelling across hundreds of amphibian populations to test the effects of various predictors of prevalence and intensity of infection by six families of helminth parasites. We focused on the potential effects of key host traits and environmental factors pertinent to focal host populations, i.e. the local diversity of the amphibian community and local climatic variables. Our analyses revealed several important determinants of prevalence or intensity of infection in various parasite families, but none applying to all families. Our study uncovered no universal driver of parasite infection levels, even among parasite taxa from the same phylum, or with similar life cycles and transmission modes. Although local variables not considered here may have effects extending across taxa, our findings suggest the need for a taxon-specific approach in any attempt to predict disease dynamics and impacts in the face of environmental and climatic changes.

Drosophila suzukii is a pest of global concern due to its great impact on several crops. The entomopathogenic nematode Steinernema carpocapsae was highly virulent to the larvae of the fly although some immune mechanisms were triggered along the infection course. Thus, to understand the gene activation profile we performed a comparative transcriptome of D. suzukii larvae infected with S. carpocapsae and Xenorhabdus nematophila to map the differentially expressed genes involved in the defence response. The analysis exposed the induction of genes involved in the humoral response such as the antimicrobial peptides and pattern-recognition receptors while there was a suppression of the cellular defence. Besides, genes involved in melanisation, and clot formation were downregulated hindering the encapsulation response and wound healing. After the infection, larvae were in a stress condition with an enrichment of metabolic and transport functionalities. Concerning the stress response, we observed variations of the heat-shock proteins, detoxification, and peroxidase enzymes. These findings set a genetical comprehensive knowledge of the host-pathogen relation of D. suzukii challenged with S. carpocapsae which could support further comparative studies with entomopathogenic nematodes.

The egg parasitoid Anastatus japonicus is a key natural enemy in the biological control of various agricultural and forestry pests. It is particularly used against the brown marmorated stink bug Halyomorpha halys and the emerging defoliator pest Caligula japonica in East Asia. It has been proved that the eggs of Antheraea pernyi can be used as a factitious host for the mass production of A. japonicus . This study systematically documented the parasitic behaviour and developmental morphology exhibited by A. japonicus on the eggs of A. pernyi . The parasitic behaviour of A. japonicus encompassed ten steps including searching, antennation, locating, digging, probing, detecting, oviposition, host-feeding, grooming, and resting. Oviposition, in particular, was observed to occur in three stages, with the parasitoids releasing eggs during the second stage when the body remained relatively static. Among all the steps of parasitic behaviour, probing accounted for the longest time, constituting 33.1% of the whole time. It was followed by digging (19.3%), oviposition (18.5%), antennation (9.6%), detecting (7.4%), and the remaining steps, each occupying less than 5.0% of the total event time. The pre-emergence of adult A. japonicus involves four stages: egg (0 to 2nd day), larva (3rd to 9th day), pre-pupa (10th to 13th day), pupa (14th to 22nd day), and subsequent development into an adult. Typically, it takes 25.60 ± 0.30 days to develop from an egg to an adult at 25℃. This information increases the understanding of the biology of A. japonicus and may provide a reference for optimising reproductive devices.
