With the development of deep learning technology, convolutional neural networks have made great progress in the field of image segmentation. However, for complex scenes and multi-scale target images, the existing technologies are still unable to achieve effective image segmentation. In view of this, an image segmentation model based on residual connection and feature fusion is proposed. The model makes comprehensive use of the deep feature extraction ability of residual connections and the multi-scale feature integration ability of feature fusion. In order to solve the problem of background complexity and information loss in traditional image segmentation, experiments were carried out on two publicly available data sets. The results showed that in the ISPRS Vaihingen dataset and the Caltech UCSD Birds200 dataset, when the model completed the 56th and 84th iterations, respectively, the average accuracy of FRes-MFDNN was the highest, which was 97.89% and 98.24%, respectively. In the ISPRS Vaihingen dataset and the Caltech UCSD Birds200 dataset, when the system model ran to 0.20s and 0.26s, the F1 value of the FRes-MFDNN method was the largest, and the F1 value approached 100% infinitely. The FRes-MFDNN segmented four images in the ISPRS Vaihingen dataset, and the segmentation accuracy of images 1, 2, 3 and 4 were 91.44%, 92.12%, 94.02% and 91.41%, respectively. In practical applications, the MSRF-Net method, LBN-AA-SPN method, ARG-Otsu method, and FRes-MFDNN were used to segment unlabeled bird images. The results showed that the FRes-MFDNN was more complete in details, and the overall effect was significantly better than the other three models. Meanwhile, in ordinary scene images, although there was a certain degree of noise and occlusion, the model still accurately recognized and segmented the main bird images. The results show that compared with the traditional model, after FRes-MFDNN segmentation, the completeness, detail, and spatial continuity of pixels have been significantly improved, making it more suitable for complex scenes., Competing Interests: The authors have declared that no competing interests exist., (Copyright: © 2024 Li et al. Hyperuricemia, a major worldwide burden on public hygiene, is closely connected with dietary habits. However, few studies have evaluated the association of dietary diversity with hyperuricemia. To preliminarily reveal the status of a diversified diet in preventing hyperuricemia based on a neighborhood-based, massive-scale cohort in China, a total of 43,493 participants aged 20-74 years old, with no history of hyperuricemia at baseline, were enrolled in the research from April 2016 to December 2019. The Dietary Diversity Score (DDS) was utilized to evaluate the dietary variety and split the participants into the low-, medium-, and high-DDS groups. Information on participants was connected to regional health information systems that acquired data on hyperuricemia instances up to 28 February 2023. Hazard ratios (HRs) and 95% confidence intervals (CIs) were computed by Cox proportional hazards models. Restricted cubic splines (RCS) were implemented to analyze dose-response correlation. A total of 1460 individuals with newly diagnosed hyperuricemia were observed over a median follow-up period of 5.59 years. Compared to the low-DDS group, HRs for the medium- and high-DDS groups were 0.87 (95% CI 0.76-0.99) and 0.80 (95% CI 0.70-0.91) in the fully adjusted model, respectively. The risk of hyperuricemia incidence was reduced by 5% for each 1 unit of DDS increase. A linear correlation of DDS with hyperuricemia emerged and further revealed that the intake of 8-10 broad categories of food could decrease the incidence of hyperuricemia. Our results validate the dietary principle of "food diversification" recommended in guidelines. Conclusions should be applied with caution considering the paucity of related evidence in additional nations.

Stem is important for assimilating transport and plant strength; however, less is known about the genetic basis of its structural characteristics. In this study, a high-throughput method, "LabelmeP rice" was developed to generate 14 traits related to stem regions and vascular bundles, which allows the establishment of a stem cross-section phenotype dataset containing anatomical information of 1738 images from hand-cut transections of stems collected from 387 rice germplasm accessions grown over two successive seasons. Then, the phenotypic diversity of the rice accessions was evaluated. Genome-wide association studies identified 94, 83, and 66 significant single nucleotide polymorphisms (SNPs) for the assayed traits in 2 years and their best linear unbiased estimates, respectively. These SNPs can be integrated into 29 quantitative trait loci (QTL), and 11 of them were common in 2 years, while correlated traits shared 19. In addition, 173 candidate genes were identified, and six located at significant SNPs were repeatedly detected and annotated with a potential function in stem development. By using three introgression lines (chromosome segment substitution lines), four of the 29 QTLs were validated. LOC_Os01g70200, located on the QTL uq1.4, is detected for the area of small vascular bundles (SVB) and the rate of large vascular bundles number to SVB number. Besides, the CRISPR/Cas9 editing approach has elucidated the function of the candidate gene LOC_Os06g46340 in stem development. The prognosis of hepatocellular carcinoma (HCC) with extrahepatic metastasis (EHM) is extremely poor. This study aimed to identify prognostic factors for systemic chemotherapy of HCC with EHM. Eighty-five patients who received systemic chemotherapy for HCC with EHM between May 2014 and October 2021 were retrospectively evaluated. Patient demographic data and characteristics of hepatic tumors and EHM were assessed to identify factors that were significantly associated with prognosis. Of the 85 patients, 68 (80.0%) had pulmonary metastasis, 11 (12.9%) had abdominal lymph node metastasis, 7 (8.2%) had abdominal metastasis, and 4 (4.7%) had bone metastasis. The median overall survival (OS) was 17.0 months, and the median progression-free survival (PFS) was 5.1 months. Univariate analysis of OS showed that synchronous EHM-HCC, serum albumin level<35 g/l and number of hepatic tumors>1 were significantly associated with poorer OS. The results of the multivariate analysis indicated that the serum albumin level and number of hepatic tumors were independent prognostic factors. Subgroup analysis of patients with 0, 1, or 2 of these independent prognostic factors showed that the median OS was 24.0 months, 16.2 months and 7.7 months and that the ORR was 38.3%, 22.6% and 0, respectively. Systemic chemotherapy is beneficial for well-selected HCC patients with EHM. The number of hepatic tumors and serum albumin level were independent risk factors for prognosis, and the number of risk factors significantly influenced OS. Therefore, these factors need to be considered before administering systemic chemotherapy for HCC patients with EHM.

Polyethylene (PE) microplastic, which is detected in various environmental media worldwide, also inevitably enters wastewater treatment plants, which may have an impact on anaerobic processes in wastewater treatment. In this work, the effect of PE microplastics on anaerobic sulfur transformation was explored. Experimental results showed that PE microplastics addition at 0.1%- 0.5% w/w promoted H 2 S production by 14.8%-27.4%. PE microplastics enhanced the release of soluble organic sulfur and inorganic sulfate, and promoted the bioprocesses of organosulfur compounds hydrolysis and sulfate reduction. Mechanism analysis showed that PE microplastics increased the content of electroactive components (e.g., protein and humic acids) contained in extracellular polymeric substances (EPS). In particular, PE microplastics increased the proportion and the dipole moment of α-helix, an important component involved in electron transfer contained in extracelluar protein, which provided more electron transfer sites and promoted the α-helix mediated electron transfer. These enhanced the direct electron transfer ability of EPSs, which might explain why PE microplastics facilitated the bioprocesses of organosulfur compounds hydrolysis and sulfate reduction. Currently, collaborative distribution models have not reached the optimal state of carbon emissions. The cost of additional low-carbon expenditures and the problem of carbon data verification have led to the lack of motivation for reducing emissions among collaborative distribution enterprises. Therefore, how to incentivize them to adopt the low-carbon model is crucial for achieving low-carbon goal. By relying on a government-led digital platform, this paper designs a dual low-carbon incentive strategy to encourage enterprise-alliance to adopt a low-carbon distribution model. In this paper, we first construct an evolutionary game model of the government, enterprise-alliance and end-users; then we explore the conditions of the three-party equilibrium evolution strategy by solving the model and analyzing the stability; and finally, we conduct simulation validation and results analysis with the help of MATLAB. In summary, we found that government punishment is more effective at regulating enterprise-alliance than reward. End-users' behavior is affected by the costs they need to bear, and they no longer support enterprise-alliance to carry out collaborative low-carbon distribution above a certain threshold., (© 2024. The interaction between coexisting plasmids can affect plasmid-carried resistance gene persistence and spread. However, whether the persistence of the bla CTX-M gene in clinical Enterobacteriaceae is related to the interaction of coresident nonresistance-conferring plasmids has not been reported. This study was initiated to elucidate how a nonresistance-conferring IncI1 plasmid affected the bla CTX-M -bearing IncFII plasmid colocated on the same cell. Herein, we constructed three isogenic derivatives of E. coli C600, designated as C600 FII , C600 I1 , and C600 FII+I1 , which harbored the bla CTX-M -IncFII plasmid and/or the nonresistance-IncI1 one. We discovered that strain C600 FII+I1 conferred higher fitness advantages than strain C600 FII ; also, the stability of the bla CTX-M -IncFII plasmid was noticeably improved in an antibiotic-free environment when it coexisted with the IncI1 plasmid. To further explore why the IncI1 plasmid enhanced the persistence of the bla CTX-M -IncFII plasmid, we assessed the bla CTX-M -IncFII plasmid's copy numbers, conjugation frequencies, and rep gene expressions in strains C600 FII and C600 FII+I1 . The results demonstrated that the rep expressions of the bla CTX-M -IncFII plasmid in strain C600 FII+I1 was greatly decreased, along with the plasmid's copy numbers and mating efficiencies, compared to those in strain C600 FII . Moreover, further study revealed that the intracellular ATP levels of strain C600 FII+I1 were far lower than those of strain C600 FII . Our findings confirmed that coexistence of the nonresistance-IncI1 plasmid can keep the bla CTX-M -IncFII plasmid more stable by increasing the fitness advantages of the host bacteria, which will pose a threat to preventing the long-term presence of the plasmid-carried bla CTX-M gene in clinical Enterobacteriaceae., Importance: So far, plasmid-carried bla CTX-M is still the most common extended-spectrum beta-lactamase (ESBL) genotype in clinical settings worldwide. Except for the widespread use of third-generation cephalosporins, the interaction between coexisting plasmids can also affect the long-term stable existence of the bla CTX-M gene; however, the study on that is still sparse. In the present study, we assess the interaction of coinhabitant plasmids bla CTX-M -IncFII and nonresistance-IncI1. Our results confirmed that the increased fitness advantages of strain C600 FII+I1 were attributable to the cohabitant nonresistance-IncI1 plasmid, which largely reduced the intracellular ATP levels of host bacteria, thus decreasing the rep gene expression of the bla CTX-M -IncFII plasmid, its copy numbers, and mating efficiencies, while the higher fitness advantages of strain C600 FII+I1 enhanced the persistence of the bla CTX-M -IncFII plasmid. The results indicate that the nonresistance-IncI1 plasmid contributes to the long-term existence of the bla CTX-M -IncFII plasmid, implying a potentially new strategy for controlling the spread of resistance plasmids in clinical settings by targeting nonresistance plasmids., Competing Interests: The authors declare no conflict of interest.

Triclocarban (TCC), as a widely used antimicrobial agent, is accumulated in waste activated sludge at a high level and inhibits the subsequent anaerobic digestion of sludge. This study, for the first time, investigated the effectiveness of microbial electrolysis cell-assisted anaerobic digestion (MEC-AD) in mitigating the inhibition of TCC to methane production. Experimental results showed that 20 mg/L TCC inhibited sludge disintegration, hydrolysis, acidogenesis, and methanogenesis processes and finally reduced methane production from traditional sludge anaerobic digestion by 19.1%. Molecular docking revealed the potential inactivation of binding of TCC to key enzymes in these processes. However, MEC-AD with 0.6 and 0.8 V external voltages achieved much higher methane production and controlled the TCC inhibition to less than 5.8%. TCC in the MEC-AD systems was adsorbed by humic substances and degraded to dichlorocarbanilide, leading to a certain detoxification effect. Methanogenic activities were increased in MEC-AD systems, accompanied by complete VFA consumption. Moreover, the applied voltage promoted cell apoptosis and sludge disintegration to release biodegradable organics. Metagenomic analysis revealed that the applied voltage increased the resistance of electrode biofilms to TCC by enriching functional microorganisms (syntrophic VFA-oxidizing and electroactive bacteria and hydrogenotrophic methanogens), acidification and methanogenesis pathways, multidrug efflux pumps, and SOS response.

Extended-spectrum-β-lactamases (ESBLs)-producing Escherichia coli conferred resistance to most β-lactams, except for carbapenems. To date, the transmission mechanism of bla CTX-M , as the most common ESBLs subtype, in E. coli has received sustained attention around the worldwide, but the research on the pathogenicity of bla CTX-M -bearing E. coli is still scarce. The aims of this study were to discern the spread characteristics of ColV (encoding colicin V) plasmids in bla CTX-M -positive E. coli. The multi-drug resistance traits, phylogroups, and ColV plasmid profilings were screened in 76 bla CTX-M -positive E. coli. Thereafter, the genetic profiles of E. coli G12 and GZM7 were determined by whole genome sequencing, conjugation and S1-pulsed-field gel electrophoresis. The median lethal dose was analyzed in E. coli G12 and TG12A, the ColV-plasmid transconjugant of G12. Of all 76 bla CTX-M -bearing E. coli, 67.11% exhibited resistance to at least 2 drugs in addition to ceftiofur, 14.47% carried ColV-positive plasmids, and 53.95% were phylogroup C. Further studies demonstrated that the bla CTX-M -bearing E. coli G12 was assigned to the predominant lineage O78:H4-ST117 of phylogroup G. In addition, its ColV-positive plasmid simultaneously carried multiple resistance genes, and could be independently transferred to confer partial pathogenicity on its host by plasmid mating. E. coli GZM7 was O53:H9-ST23 of phylogroup C, which belonged to another representative lineage of APEC (avian pathogenic E. coli). Its ColV-positive plasmid could complete conjugation with the help of the other coexisting-resistance conjugative plasmid, although it failed to transfer alone. Our findings highlight the flexibly horizontal transfer of ColV plasmids along with multidrug-resistant genes among bla CTX-M -bearing E. coli poses a threat to poultry health and food safety, which contributes to elucidate the concept of "One Health" and deserves particular concern., (Copyright © 2024 The Authors. Microbial elicitors have been shown to boost plant immunity by inducing defense responses to reduce plant disease. However, little is known about the changes in plant microbiome and metabolism in the process of enhancing plant immunity with elicitors. The protein elicitor BAR11, from Saccharothrix yanglingensis Hhs.015, induces defense responses in Arabidopsis thaliana that enhances resistance to pathogens. In this study, bar11 was inserted into Col-0 A. thaliana to obtain BAR11-Trans plant by Agrobacterium-mediated immersion transformation. BAR11-Trans exhibited an elevated defense level against Pseudomonas syringae pv. tomato DC3000 while experiencing a decline in biomass production of above-ground parts. In the process, BAR11-Trans increased the activity of phenylalanine ammonia lyase (PAL) and catalase (CAT), and up-regulated genes related to plant defense pathways. Furthermore, BAR11-Trans decreased root tip reactive oxygen species (ROS) levels while increasing ROS burst in the leaves. Soil transplantation experiments showed that soil planted with BAR11-Trans could enhance the resistance of Col-0 A. thaliana to DC3000. Analysis of A. thaliana rhizosphere soil through 16S rRNA amplified sequencing revealed that BAR11-Trans increased the relative abundance and diversity of the rhizosphere microbial community, leading to the recruitment of more plant probiotics. Additionally, the accumulation of kaempferitrin and robinin in BAR11-Trans influenced the physicochemical properties of rhizosphere soil and the composition of the bacterial community. In summary, BAR11-Trans exhibited heightened defense levels compared to Col-0, leading to increased secretion of secondary metabolites and the recruitment of a greater number of microorganisms to adapt to the environment. A novel Fe/La decorative biochar filled in sodium alginate beads (SA-KBC-Fe/La) was prepared by a simple sol-gel method and applied to adsorb phosphate (P) efficiently from water in this study. The morphology, structure and chemical component of the hydrogel beads were characterized in detail. And the synthesized bead exhibited easy separation and high P uptake of 46.65 mg/g when the Fe: La was of 1: 2 at 298 K with initial P of 100 mg/L, which was much higher than SA gel bead. The adsorption showed that the optimal pH was 6, and the adsorption was met with pseudo-second-order kinetics and Langmuir isothermal models, indicating a chemical adsorption process. The adsorption capacity remained 82 % after 5 cycles of adsorption. The adsorption mechanism of P was mainly of ligand exchange and electrostatic attraction. Compared with other reported adsorbents, the modification of Fe/La could enhance the mechanical property of SA-KBC-Fe/La beads with increasing active sites. Additionally, the involved biochar could lead to excellent thermal stability and hierarchical porous structure of beads with larger specific surface area (54.22 m 2 /g). Objective: To investigate the status of anxiety, depression, and psychological resilience among individuals with COVID-19, and their interrelationships to provide a scientific basis for developing psychological intervention strategies for these patients., Methods: A total of 126 patients with COVID-19 who were admitted to Wuhan Huoshenshan Hospital were recruited in this study. A comprehensive survey was conducted using a general information questionnaire, the Self-Rating Anxiety Scale, the self-rating depression scale, and the Chinese version of the psychological Connor-Davidson resilience scale; a questionnaire-based survey was conducted., Results: Significant differences in anxiety scores were observed among COVID-19 patients with different education levels and the number of immediate family members. The differences in depression scores were noted among patients of different age groups, and marital statuses were also significant. The total psychological resilience score and the scores of all dimensions are negatively correlated with anxiety and depression. Furthermore, the patient's gender, the number of immediate family members, and the psychological resilience dimensions are associated with the severity anxiety of patients. Patient age and psychological resilience are associated with the depression level of patients., Conclusion: Patients with COVID-19 exhibit elevated levels of both anxiety and depression. Emerging evidence suggests that cryptic translation beyond the annotated translatome produces proteins with developmental or physiological functions. However, functions of cryptic non-canonical open reading frames (ORFs) in cancer remain largely unknown. To fill this gap and systematically identify colorectal cancer (CRC) dependency on non-canonical ORFs, we apply an integrative multiomic strategy, combining ribosome profiling and a CRISPR-Cas9 knockout screen with large-scale analysis of molecular and clinical data. Many such ORFs are upregulated in CRC compared to normal tissues and are associated with clinically relevant molecular subtypes. We confirm the in vivo tumor-promoting function of the microprotein SMIMP, encoded by a primate-specific, long noncoding RNA, the expression of which is associated with poor prognosis in CRC, is low in normal tissues and is specifically elevated in CRC and several other cancer types. Mechanistically, SMIMP interacts with the ATPase-forming domains of SMC1A, the core subunit of the cohesin complex, and facilitates SMC1A binding to cis-regulatory elements to promote epigenetic repression of the tumor-suppressive cell cycle regulators encoded by CDKN1A and CDKN2B. Thus, our study reveals a cryptic microprotein as an important component of cohesin-mediated gene regulation and suggests that the 'dark' proteome, encoded by cryptic non-canonical ORFs, may contain potential therapeutic or diagnostic targets., (© 2023. Nitrite (NO 2 - ) accumulation caused by nitrite-oxidizing bacteria (NOB) inhibition in nitrification is a double-edged sword, i.e., a disaster in aquatic environments but a hope for innovating nitrogen removal technology in wastewater treatment. However, little information is available regarding the molecular mechanism of NOB inhibition at the cellular level. Herein, we investigate the response of NOB inhibition on NO 2 - accumulation established by a side-stream free ammonia treatment unit in a nitrifying reactor using integrated metagenomics and metaproteomics. Results showed that compared with the baseline, the relative abundance and activity of NOB in the experimental stage decreased by 91.64 and 68.66%, respectively, directly resulting in a NO 2 - accumulation rate of 88%. Moreover, RNA polymerase, translation factors, and aa-tRNA ligase were significantly downregulated, indicating that protein synthesis in NOB was interfered during NO 2 - accumulation. Further investigations showed that ribosomal proteins and GTPases, responsible for bindings between either ribosomal proteins and rRNA or ribosome subunits, were remarkably downregulated. This suggests that ribosome biogenesis was severely disrupted, which might be the key reason for the inhibited protein synthesis. Our findings fill a knowledge gap regarding the underlying mechanisms of NO 2 - accumulation, which would be beneficial for regulating the accumulation of NO 2 - in aquatic environments and engineered systems.

Genetically modified (GM) proteins in edible tissues of transgenic maize are of intense public concern. We provided a Cre/loxP-based strategy for manipulating the expression of transgenes in green tissues while locking them in nongreen tissues. First, the Cre gene was driven by the green tissue-specific promoter Zm1rbcS to generate transgenic maize KEY. Meanwhile, a gene cassette containing a Nos terminator (NosT) in front of the Cry1Ab/c gene was driven by the strong promoter ZmUbi to generate another transgenic maize LOCK. By crossing KEY and LOCK plants, the expressed Cre recombinase under the control of the Zm1rbcS promoter from KEY maize accurately removed the NosT of LOCK maize. Consequently, the expression of blocked Cry1Ab/c was enabled in specific green tissues in their hybrids. The expression level and concentration of Cry1Ab/c were observed using a strategy with high specific accumulation in green tissues (leaf and stem). Still, only a small or absent amount was observed in root and kernel tissues. Furthermore, we assessed the bioactivity of transgenic maize against 2 common lepidopteran pests, Ostrinia furnacalis and Spodoptera frugiperda, in the laboratory and field. The transgenic plants showed high plant resistance levels against the 2 pests, with mortality rates above 97.2% and damage scales below 2.2 compared with the control group. These findings are significant for exploring novel genetic engineering techniques in GM maize and providing a feasible strategy for transgenes avoiding expression in edible parts. Aim of this study was to investigate the prevalence and genetic environment of the oxazolidinone resistance gene optrA in Streptococcus suis (S. suis) isolates from diseased pigs in China. A total of 178 S. suis isolates were screened for the optrA gene by PCR. The phenotypes and genotypes of optrA-positive isolates were investigated by antimicrobial susceptibility testing, core genome Multilocus Sequence Typing (cgMLST), capsular serotypes determination and whole-genome sequencing (WGS). Fifty-one (28.7%) S. suis isolates were positive for optrA. Phylogenetic analysis indicated that the spread of the optrA among S. suis isolates was primarily due to horizontal transfer. Analysis of S. suis serotypes from diseased pigs revealed substantial diversity. The genetic environment of optrA was complex and diverse and could be divided into 12 different types. Interestingly, we identified a novel integrative and conjugative element ICESsu988S, carrying optrA and erm(T) genes. This is to the best of our knowledge the first report of the optrA and erm(T) co-located on an ICE in S. suis. Our results showed a high prevalence of optrA gene in S. suis isolates in China. Gibberella ear rot (GER) in maize caused by Fusarium graminearum is one of the most devastating maize diseases reducing grain yield and quality worldwide. The genetic bases of maize GER resistance remain largely unknown. Using artificial inoculation across multiple environments, the GER severity of an association panel consisting of 316 diverse inbred lines was observed with wide phenotypic variation. In the association panel, a genome-wide association study using a general linear model identified 69 single-nucleotide polymorphisms (SNPs) significantly associated with GER resistance at the threshold of 2.04 × 10 -5 , and the average phenotypic variation explained (PVE) of these SNPs was 5.09%. We also conducted a genome-wide association study analysis using a mixed linear model at a threshold of 1.0 × 10 -4 , and 16 significantly associated SNPs with an average PVE of 4.73% were detected. A combined general linear model and mixed linear model method obtained 10 co-localized significantly associated SNPs linked to GER resistance, including the most significant SNP (PZE-105079915) with the greatest PVE value, 9.07%, at bin 5.05 following 10 candidate genes. These findings are significant for the exploration of the complicated genetic variations in maize GER resistance. The presence of surfactants in waste activated sludge (WAS) system is generally regarded as beneficial to sludge treatment such as enhancing sludge dewatering and improving value-added fermentation products generation. However, in this study, it was firstly found that sodium dodecylbenzene sulfonate (SDBS, a typical surfactant) obviously increased toxic hydrogen sulfide (H 2 S) gas production from WAS anaerobic fermentation at environmentally relevant concentrations. Experimental results showed that H 2 S production from WAS significantly increased from 53.24 × 10 -3 to 111.25 × 10 -3  mg/g volatile suspended solids (VSS) when SDBS level increased from 0 to 30 mg/g total suspended solid (TSS). It was found that SDBS presence destroyed WAS structure and enhanced sulfur containing organics release. SDBS reduced the proportion of α-helix structure, damaged disulfide bridges and protein conformation, and effectively destroyed protein structure. SDBS promoted sulfur containing organics degradation and provided more readily hydrolyzed micro-molecule organics for sulfide production. Microbial analysis showed that SDBS addition enhanced the abundance of functional genes encoding protease, ATP-binding cassette transporters, and amino acids lyase, enhanced the activities and abundance of hydrolytic microbes, thus increased sulfide production from the hydrolysis of sulfur containing organics. Compared with the control, 30 mg/g TSS SDBS increased organic sulfurs hydrolysis and amino acids degradation by 47.1 % and 63.5 %, respectively. Key genes analysis further showed that SDBS addition promoted sulfate transport system and dissimilatory sulfate reduction. Objectives: Crohn's disease is an incurable disease characterized by unpredictable intestinal symptoms, which unavoidably affect patients' lives and contribute to feelings of stigma. This study aimed to explore the status and its correlates of stigma among patients with Crohn's disease., Methods: Using a convenience sampling, 146 hospitalized patients with Crohn's disease were recruited in a tertiary hospital in Southern China from October 2020 to March 2021. The participants were assessed by demographic and disease-related questionnaires, the Social Impact Scale (SIS), Inflammatory Bowel Disease-Self-Efficacy Scale (IBD-SES), and Social Support Rating Scale (SSRS). Multivariate linear regression analysis was conducted to explore the influencing factors of stigma among patients with Crohn's disease., Results: The mean SIS score was 58.14 ± 10.74, representing a moderate effect of stigma, and 85.6% (125/146) of the patients experienced moderate to high levels of stigma. The multiple linear regression analysis showed that perceived public awareness of the disease, family income, age, and self-efficacy were the main influencing factors of stigma, which could explain 52% of the total variation of stigma among patients with Crohn's disease., Conclusion: Stigma among patients with Crohn's disease is influenced by perceived public awareness of the disease, family income, age, and self-efficacy. Free ammonia (FA), the unionized form of ammonium, is presented in anaerobic fermentation of waste activated sludge (WAS) at high levels. However, its potential role in sulfur transformation, especially H 2 S production, during WAS anaerobic fermentation process was unrecognized previously. This work aims to unveil how FA affects anaerobic sulfur transformation in WAS anaerobic fermentation. It was found that FA significantly inhibited H 2 S production. With an increase of FA from 0.04 to 159 mg/L, H 2 S production reduced by 69.9%. FA firstly attacked tyrosine-like proteins and aromatic-like proteins in sludge EPSs, with CO groups being responded first, which decreased the percentage of α-helix/(β-sheet + random coil) and destroyed hydrogen bonding networks. Cell membrane potential and physiological status analysis showed that FA destroyed membrane integrity and increased the ratio of apoptotic and necrotic cells. These destroyed sludge EPSs structure and caused cell lysis, thus strongly inhibited the activities of hydrolytic microorganisms and sulfate reducing bacteria. Microbial analysis showed that FA reduced the abundance of functional microbes (e.g., Desulfobulbus and Desulfovibrio) and genes (e.g., MPST, CysP, and CysN) involved in organic sulfur hydrolysis and inorganic sulfate reduction. This study was designed to depict prevalence and antimicrobial resistance characteristics of Proteus mirabilis (P. mirabilis) strains in 4 chicken farms and to probe the transfer mechanism of resistance genes. A total of 187 P. mirabilis isolates were isolated from 4 chicken farms. The susceptibility testing of these isolates to 14 antimicrobials showed that the multidrug resistance (MDR) rate was as high as 100%. The β-lactamase resistance genes bla OXA-1 , bla CTX-M-1G , bla CTX-M-9G and colistin resistance gene mcr-1 were highly carried in the P. mirabilis isolates. An MDR strain W47 was selected for whole genome sequencing (WGS) and conjugation experiment. The results showed that W47 carried 23 resistance genes and 64 virulence genes, and an SXT/R391 integrated conjugative elements (ICEs) named ICEPmiChn5 carrying 17 genes was identified in chromosome. ICEPmiChn5 was able to be excised from the chromosome of W47 forming a circular intermediate, but repeated conjugation experiments were unsuccessful. Among 187 P. mirabilis isolates, 144 (77.01%, 144/187) isolates carried ICEPmiChn5-like ICEs, suggesting that ICEs may be the major vector for the transmission of resistance genes among MDR chicken P. mirabilis strains in this study. Municipal sludge biochar (MSBC) can be used to absorb phosphorus in water for waste treatment. Nano-zero-valent zinc (nZVZ) was uniformly attached to MSBC to obtain a highly efficient phosphorus-absorbing composite material, nZVZ-MSBC. Characterization by FTIR, XPS, XRD, and BET showed that nZVZ was uniformly dispersed on the surface of the MSBC. Zinc loading was able to greatly improve the adsorption performance of MSBC for phosphorus. Adsorption experiments illustrated that the adsorption process conformed to the Langmuir model, and the maximum adsorption amount was 186.5 mg/g, which is much higher than that for other municipal sludge biochars. The adsorption process reached 80% of the maximum adsorption capacity at 90 min, and this gradually stabilized after 240 min; adsorption equilibrium was reached within 24 h. The optimum pH for adsorption was 5. The main adsorption mechanism was chemical adsorption, but physical adsorption, external diffusion, internal diffusion, and surface adsorption also played roles. The potential for application as an efficient adsorbent of phosphorus from water was confirmed. In addition, a novel strategy for municipal sludge disposal and resource utilization is provided.

ICEGpa1804 was identified in the genome of a serovar 2, ST279 isolate EHP1804 carrying eight different resistance genes from 200 Glaesserella parasuis strains isolated from swine with lower respiratory tract infection in seven provinces of China. Susceptibility testing for EHP1804 was determined by broth microdilution, and its genetic profile was determined by whole-genome sequencing. The complete ICEGpa1804 was analysed by polymerase chain reaction, conjugation assay and bioinformatics tools. The conjugation assay was performed using EHP1804 as the donor and G. parasuis V43 (rifampicin-resistant) as the recipient. ICEGpa1804 has a size of 71,880 bp and contains 83 genes, including eight resistance genes [tet(B), bla Rob-1 , aphA1, strA, strB, aac(3)-IId, catA3 and sul2]. The conjugation assay showed that ICEGpa1804 could be transferred to G. parasuis V43 with frequencies of 4.3 × 10 -7 . To the best of the authors' knowledge, this is the first study to identify a novel integrative and conjugative element (ICE) carrying eight resistance genes and seven insertion sequence (IS) elements from a G. parasuis isolate. Tn6743, a novel transposon carrying six resistance genes, was identified. Moreover, ISGpa1, a novel IS256 family insertion element, is the first characterized example of a G. parasuis insertion element. Multiple mobile genetic elements involved in resistance genes were located in chromosomal ICEGpa1804, which showed that ICEs may serve as a vital platform for the accumulation of resistance genes., (Copyright © 2023. Gibberella ear rot (GER) caused by Fusarium graminearum (teleomorph Gibberella zeae ) is one of the most devastating maize diseases that reduces grain yield and quality worldwide. Utilization of host genetic resistance has become one of the most suitable strategies to control GER. In this study, a set of 246 diverse inbred lines derived from the intermated B 73 × Mo 17 doubled haploid population (IBM Syn10 DH) were used to detect quantitative trait loci (QTL) associated with resistance to GER. Meanwhile, a GradedPool-Seq (GPS) approach was performed to identify genomic variations involved in GER resistance. Using artificial inoculation across multiple environments, GER severity of the population was observed with wide phenotypic variation. Based on the linkage mapping, a total of 14 resistant QTLs were detected, accounting for 5.11 to 14.87% of the phenotypic variation, respectively. In GPS analysis, five significant single nucleotide polymorphisms (SNPs) associated with GER resistance were identified. Combining QTL mapping and GPS analysis, a peak-value SNP on chromosome 4 from GPS was overlapped with the QTL qGER4.2 , suggesting that the colocalized region could be the most possible target location conferring resistance to GER. Subsequently, seven candidate genes were identified within the peak SNP, linking them to GER resistance. These findings are useful for exploring the complicated genetic variations in maize GER resistance. The genomic regions and genes identified herein provide a list of candidate targets for further investigation, in addition to the combined strategy that can be used for quantitative traits in plant species.

In Chinese zoos, there are usually specially designed bird parks, similar to petting zoos, that allow children and adults to interact with diverse birds. However, such behaviors present a risk for the transmission of zoonotic pathogens. Recently, we isolated eight strains of Klebsiella pneumoniae and identified two bla CTX-M -positive strains from 110 birds, including parrots, peacocks, and ostriches, using anal or nasal swabs in a bird park of a zoo in China. There, K. pneumoniae LYS105A was obtained from a diseased peacock with chronic respiratory diseases by a nasal swab, which harbored the bla CTX-M-3 gene and exhibited resistance to amoxicillin, cefotaxime, gentamicin, oxytetracycline, doxycycline, tigecycline, florfenicol, and enrofloxacin. According to an analysis by whole-genome sequencing, K. pneumoniae LYS105A belongs to serotype ST859 (sequence type 859)-K19 (capsular serotype 19) and contains two plasmids, of which pLYS105A-2 can be transferred by electrotransformation and harbors numerous resistance genes such as bla CTX-M-3 , aac(6')-Ib-cr5 , and qnrB91 . The above-mentioned genes are located in a novel mobile composite transposon, Tn 7131 , which makes horizontal transfer more flexible. Although no known genes were identified in the chromosome, a significant increase in SoxS upregulated the expression levels of phoPQ , acrEF - tolC , and oqxAB , which contributed to strain LYS105A acquiring resistance to tigecycline (MIC = 4 mg/L) and intermediate resistance to colistin (MIC = 2 mg/L). Altogether, our findings show that bird parks in zoos may act as important vehicles for the spread of multidrug-resistant bacteria from birds to humans and vice versa. IMPORTANCE A multidrug-resistant ST859-K19 K. pneumoniae strain, LYS105A, was obtained from a diseased peacock in a Chinese zoo. In addition, multiple resistance genes such as bla CTX-M-3 , aac(6')-Ib-cr5 , and qnrB91 were located in a novel composite transposon, Tn 7131 , of a mobile plasmid, implying that most of the resistance genes in strain LYS105A can be moved easily via horizontal gene transfer. Meanwhile, an increase in SoxS can further positively regulate the expression of phoPQ , acrEF - tolC , and oqxAB , which is the key factor for strain LYS105A to develop resistance to tigecycline and colistin. Taken together, these findings enrich our understanding of the horizontal cross-species spread of drug resistance genes, which will help us curb the development of bacterial resistance.

A magnetic copper ferrite and biochar composite (CuFe 2 O 4 @BC) catalyst was prepared by an improved sol-gel calcination method and initially used for the removal of antibiotics ciprofloxacin (CIP) by activated peroxymonosulfate (PMS). Using CuFe 2 O 4 @BC as the activator, 97.8% CIP removal efficiency could be achieved in 30 min. After a continuous degradation cycle, CuFe 2 O 4 @BC catalyst still exhibited great stability and repeatability and could also be quickly recovered by an external magnetic field. Meanwhile, the CuFe 2 O 4 @BC/PMS system presented good stability for metal ion leaching, which was far less than the leaching of metal ions in the CuFe 2 O 4 /PMS system. Moreover, the effects of various influencing factors, such as initial solution pH, activator loading, PMS dosage, reaction temperature, humic acid (HA), and the inorganic anions were explored. The quenching experiments and the electron paramagnetic resonance (EPR) analysis manifested that hydroxyl radical (•OH), sulfate radical (SO 4 •- ), superoxide radical (O 2 •- ), and singlet oxygen ( 1 O 2 ) were generated in the CuFe 2 O 4 @BC/PMS system, while 1 O 2 and O 2 •- are mainly involved in the degradation process. The synergistic effect between CuFe 2 O 4 and BC enhanced the structural stability and electrical conductivity of the material, which promoted the bonding between the catalyst and PMS, resulting in the enhanced catalytic activity of CuFe 2 O 4 @BC. This indicates that CuFe 2 O 4 @BC activating PMS is a promising remediation technique for CIP-contaminated water.

1-Butyl-3-methylimidazolium chloride (BmimCl), an imidazolium-based ionic liquid, is considered the representative emerging persistent aquatic pollutant, and its environmental toxicity has attracted a growing concern. However, most of the investigations focused on monocultures or a single organism, with little information available on the complex syntrophic consortium that dominates the complex and successional biochemical processes, such as anaerobic digestion. In this study, the effect of BmimCl at environmentally relevant levels on glucose anaerobic digestion was therefore investigated in several laboratory-scale mesophilic anaerobic digesters to provide such support. Experimental results showed that BmimCl at 1-20 mg/L inhibited the methane production rate by 3.50-31.03%, and 20 mg/L BmimCl inhibited butyrate, hydrogen, and acetate biotransformation by 14.29%, 36.36%, and 11.57%, respectively. Toxicological mechanism studies revealed that extracellular polymeric substances (EPSs) adsorbed and accumulated BmimCl through carboxyl, amino, and hydroxyl groups, which destroyed the EPSs' conformational structure, thereby leading to the inactivation of microbial cells. MiSeq sequencing data indicated that the abundance of Clostridium&#95;sensu&#95;stricto&#95;1 , Bacteroides , and Methanothrix decreased by 6.01%, 7.02%, and 18.45%, respectively, in response to 20 mg/L BmimCl. Molecular ecological network analysis showed that compared with the control, the lower network complexity, fewer keystone taxa, and fewer associations among microbial taxa were found in the BmimCl-present digester, indicating the reduced stability of the microbial community.

Background: To examine the association between daily physical activity (PA) and major adverse cardiovascular events (MACEs) in northwest China., Methods: The data in this analysis were part of the baseline survey of the Regional Ethnic Cohort Study in Northwest China from June 2018 to May 2019 in Shaanxi Province. This study used standardized self-reported total physical activity (continuous and categorical variables) and self-reported outcomes of MACEs. All analyses were conducted using the logistic regression model and stratified by age, sex, body mass index (BMI), and region. The dose-response relationships were assessed with a restricted cubic spline., Results: The average level of total PA was 17.60 MET hours per day (MET-h/d). Every increase of four MET-h/d of total PA was associated with a lower risk of MACEs [adjusted OR = 0.95 (95% CI, 0.93~0.98)]. Compared with participants in the bottom quartile of total PA, a lower risk of MACEs was observed in the top quartile group [≥23.3 MET-h/d, 0.68 (0.55~0.83)]. Stratified analyses showed similar results in males, females, participants over 45 years old, participants in the rural region, and normal weight range participants (BMI < 24 kg/m 2 ). Total participants also observed a dose-response relationship after adjusting for socioeconomic and lifestyle factors., Conclusions: A higher level of PA was associated with a lower MACE risk. Peracetic acid (PAA), a widely used organic peroxide with strong disinfection and oxidizing effect, has recently attracted research interest in waste activated sludge (WAS) treatment to achieve sludge reduction and resource utilization. However, its impact on hydrogen accumulation from WAS dark fermentation has not been documented. This study therefore is intended to fill in this knowledge gap and clarify the underlying mechanism of PAA-promoted hydrogen generation. Batch experiments revealed that when raised PAA dosage from 0 to 8 mg/g TSS (total suspended solids), cumulative hydrogen production within 168 h fermentation increased from 1.3 to 14.2 mL/g VSS (volatile suspended solids), however, further increase PAA dosage to 10 mg/g TSS resulted in a slight decrease in hydrogen yield. Mechanism studies revealed that PAA was beneficial to sludge disintegration (10 mg/g TSS PAA increased SCOD (soluble chemical oxygen demand) by 254 %). Although PAA inhibited the activity of all microorganism involved in dark fermentation, the inhibitory effect on hydrogen consumers were much more serious than that on hydrogen producers (-45.8 % versus -5.1 % and - 7.3 %). The fermentation was found to shift from propionate type to acetate and butyrate type, favoring hydrogen production. Moreover, the methane production process was effectively inhibited by PAA, which meant less hydrogen consumption. Microbial community analysis results unveiled that PAA increased the abundances of hydrolytic bacteria (e.g., norank&#95;f&#95;&#95;Saprospiraceae) and hydrogen producers (e.g., Clostridium&#95;sensu&#95;stricto&#95;10). Synthetic cannabinoids, a class of psychoactive compounds, are controlled as new psychoactive substances (NPSs) identified by the early warning system (EWS) of the European Monitoring Centre for Drugs and Drug Addiction (EMCDDA). At present, several new synthetic cannabinoids have appeared in the illegal drug market, including 4-methylnaphthalen-1-yl-(1-pentylindol-3-yl) methanone (JWH-122), methyl (1-(5-fluoropentyl)-1H-indazole-3-carbonyl)-L-valinate (5F-AMB), and methyl 2-(1-(4-fluorobenzyl)-1Hindazole-3-carboxamido)-3-methylbutanoate (AMB-FUBINACA). A convenient, rapid, and highly sensitive analytical method was developed to determine three synthetic cannabinoids in rat plasma and urine. The liquid chromatography tandem mass spectrometry (LC-MS/MS) method was optimized and validated to analyze the three synthetic cannabinoids in rat plasma and urine. The method identified intra-assay precision (1.3-9.0% and 2.8-6.7%), inter-assay precision (3.0-8.6% and 3.9-8.8%), limits of detection (0.003-0.004 ng/mL and 0.00125-0.002 ng/mL) and quantification (0.012-0.016 ng/mL and 0.003-0.005 ng/mL), recovery (95.4-106.8% and 92.0-106.8%) for rat plasma and urine, and the matrix effect (93.4-118.0%) for rat urine, and the correlation coefficients were above 0.99 in the linear range. The established LC-MS/MS method was successfully used to simultaneously detect the JWH-122 and 5F-AMB in rat plasma and JWH-122, 5F-AMB, and AMB-FUBINACA in rat urine. The present study provides methodological support for internal exposure assessment of three synthetic cannabinoids and promotes the quantitative analysis and technical supervision of synthetic cannabinoids.

The treatment and disposal of sludge is a complex environmental problem because of the high moisture content. Herein, We reported the process of Fe(II) activating Urea hydrogen peroxide (UHP) to improve waste activated sludge (WAS) dewaterability for the first time. Fe(II)/UHP was proven to significantly improve WAS dewaterability. Specifically, under the optimal conditions with 60/35-Fe(II)/UHP mg/g TSS, the CST, SRF, and WC SC of WAS reduced from 215.3 ± 7.5s, 9.2 ± 0.32 (× 10 12 m/kg), and 92.2 ± 0.7% (control) to 62.3 ± 4.3s, 2.8 ± 0.09 (× 10 12 m/kg), and 70.4 ± 0.4%, respectively. Further analysis revealed that •OH was generated in the Fe(II)/UHP system and played the dominant role in enhancing WAS dewaterability. •OH was found to attack extracellular polymeric substances (EPSs) and cells, causing EPSs fragmentation and decomposition part of EPSs into micro-molecule organics or even inorganics, and leading to cell destruction, thus liberating the EPSs-bound and cells-bound water. •OH also degraded the protein in centrifugal liquor (CL) into micro-molecule organics such as amino acids, which could reduce the viscosity and electronegativity of CL. The above facts ultimately reduced solid-liquid interface interaction but increased hydrophobicity, flocculation, and flowability of WAS. Meanwhile, the broken WAS flocs were then re-flocculated via adsorption bridging and charge neutralization induced by Fe(II) and Fe(III). Moreover, Fe(II)/UHP treatment achieved the reduction and stabilization of heavy metals of dewatered sludge, which further enabled its land application. Glycosaminoglycan from Apostichopus japonicus (AHG) and its depolymerized fragments (DAHGs) are anticoagulant fucosylated chondroitin sulfate. The aim of this study was to further evaluate the anticoagulant and antithrombic activity of AHG and DAHGs, as well as reveal the dynamic relationship between exposure and effect in vivo. The results demonstrated that AHG100 (Mw~100 kDa), DAHG50 (Mw~50 kDa), and DAHG10 (Mw~10 kDa) exhibited potent anticoagulant activity by inhibiting intrinsic factor Xase complex (FXase) as well as antithrombin-dependent factor IIa (FIIa) and factor Xa (FXa). These glycosaminoglycans markedly prevented thrombosis formation and thrombin-induced platelet aggregation in a dose- and molecular weight-dependent manner in vitro and in vivo. The further bleeding time measurement indicated that DAHG10 exhibited obviously lower hemorrhage risks than native AHG100. Following oral administration, DAHG10 could be absorbed into blood, further dose-dependently prolonging activated partial thromboplastin time (APTT) and thrombin time (TT) as well as inhibiting FXa and FIIa partially through FXase. Anticoagulant activity was positively associated with plasma concentration following oral administration of DAHG10. Our study proposed a new point of view to understand the correlation between effects and exposure of fucosylated chondroitin sulfate as an effective and safe oral antithrombotic agent.

A palladium-catalyzed coupling reaction of alkynylsulfones with alkenes has been described, which provides an efficient and practical entry to various functionalized dichlorinated vinyl sulfones. This method features excellent regio- and stereoselectivities, good functional group compatibility, as well as mild reaction conditions. Mechanistic studies suggest that the reaction goes through sequential syn -chloropalladation, alkene insertion, and C(sp 3 )-Cl bond formation processes, and the sulfonyl group is crucial to the stereoselectivity control of the reaction.

Rhamnolipid (RL), one representative biosurfactant, is widely regarded as an economically feasible and environmentally beneficial additive to improve fermentation efficiency and resource recovery from waste activated sludge (WAS). However, its potentially detrimental impact on WAS fermentation such as H 2 S generation was overlooked previously. This study therefore aims to fill the gap through exploring whether and how the presence of RL affects H 2 S generation from WAS anaerobic fermentation. Experimental results showed that when RL increased from 0 to 40 mg/g total suspended solids (TSS), the cumulative H 2 S yield enhanced from 323.6 ×  10 -4 to 620.3 ×  10 -4  mg/g volatile suspended solids (VSS). Mechanism analysis showed that RL reduced WAS surface tension, which benefited transformations of organic sulfurs (e.g., aliphatic-S and sulfoxide) and inorganic sulfate from solid to liquid phase. The presence of RL not only reduced the ratio of α-helix/(β-sheet + random coil) and damaged the hydrogen bonding networks of organic sulfurs but also promoted substrate surface charges and cell membrane permeability. These facilitated the contact between hydrolase and organic sulfurs, thereby increasing sulfide production from organic sulfurs hydrolysis. Further investigations showed that RL promoted the expression of key genes (e.g., aprA/B and dsrA/B) involved in the dissimilatory sulfate reduction, which accelerated the reaction of adenosine 5'-phosphosulfate (APS)→ sulfite→ sulfide. Meanwhile, RL inhibited the corresponding key genes such as CysH, and Sir, responsible for assimilatory sulfate reduction (APS→3'-phosphoadenosine-5'phosphosulfate→organosulfur), which reduced substrate competition in favor of H 2 S production from dissimilatory sulfate reduction. In the present study, pyrophosphate (PP) was used to activate peroxymonosulfate (PMS) for acid orange 7 (AO7) removal under neutral pH conditions. The removal rate of AO7 (20 mg/L) was 84% within the reaction time with a rate constant value of 0.0165 min -1 under optimum conditions. Additionally, the effects of the concentrations of PMS and PP in solutions with various pH values and the coexisting inorganic anions on AO7 removal were measured. In addition, the performance of phosphate (P(V)) on PMS activation was compared with that of phosphite (P(III)) species. In contrast to P(III), the concentration of P(V) showed a positive correlation with the efficiency of AO7 decolorization. PMS activation in different types of buffer solutions was also examined, and the results indicated that the decolorization efficiency of AO7 induced by PP addition, and the buffer solution also contributed to PMS self-decomposition. Singlet oxygen ( 1 O 2 ) might be the primary reactive oxygen species (ROS) in the PP/PMS system in which AO7 is decolorized at an initial pH of 7.06, as indicated by quenching experiments and electron spin resonance (ESR) tests. Discovery of disease biomarker based on untargeted metabolomics is informative for pathological mechanism studies and facilitates disease early diagnosis. Numerous of metabolomic strategies emerge due to different sample properties or experimental purposes, thus, methodological evaluation before sample analysis is essential and necessary. In this study, sample preparation, data processing procedure and metabolite identification strategy were assessed aiming at the discovery of biomarker of breast cancer. First, metabolite extraction by different solvents, as well as the necessity of vacuum-dried and re-dissolution, was investigated. The extraction efficiency was assessed based on the number of eligible components (components with MS/MS data acquired), which was more reasonable for metabolite identification. In addition, a simplified data processing procedure was proposed involving the OPLS-DA, primary screening for eligible components, and secondary screening with constraints including VIP, fold change and p value. Such procedure ensured that only differential candidates were subjected to data interpretation, which greatly reduced the data volume for database search and improved analysis efficiency. Furthermore, metabolite identification and annotation confidence were enhanced by comprehensive consideration of mass and MS/MS errors, isotope similarity, fragmentation match, and biological source confirmation. On this basis, the optimized strategy was applied for the analysis of serum samples of breast cancer, according to which the discovery of differential metabolites highly encouraged the independent biomarkers/indicators used for disease diagnosis and chemotherapy evaluation clinically. Gut microbiome disturbances have been widely implicated in major depressive disorder (MDD), although the identity of causal microbial species and the underlying mechanisms are yet to be fully elucidated. Here we show that Bacteroides species enriched in the gut microbiome from MDD patients differentially impact the susceptibility to depressive behaviors. Transplantation of fecal microbiome from MDD patients into antibiotic-treated mice induced anxiety and despair-like behavior and impaired hippocampal neurogenesis. Colonization of Bacteroides fragilis, Bacteroides uniformis, and, to a lesser extent, Bacteroides caccae, but not Bacteroides ovatus, recapitulated the negative effects of MDD microbiome on behavior and neurogenesis. The varying impacts of Bacteroides species were partially explained by differential alternations of tryptophan pathway metabolites and neurotransmitters along the gut-brain axis. Notably, an intensified depletion of cerebral serotonin concurred with the enhanced susceptibility to depression. LncRNA have been increasingly shown that plays pivotal roles in the development of various diseases, including renal fibrosis. Nevertheless, the pathological function of Long non-coding RNA KCNQ1OT1 (KCNQ1OT1) in the renal fibrosis remains obscure. Unilateral ureteral obstruction (UUO) was used to induce renal fibrosis. We detected the expression levels of KCNQ1OT1 in the TGF-β1-induced HK-2 cells via RT-qPCR analysis. The functions of KCNQ1OT1 on the progression of renal fibrosis were examined by CCK-8, EdU, dual-luciferase reporter, and immunofluorescence analyses. In the present study, we found that sh-KCNQ1OT1 obviously attenuated UUO-induced renal fibrosis. Moreover, production of extracellular matrix (ECM), including α-SMA and Fibronectin levels, was significantly increased in kidney and HK-2 cells after UUO or TGF-β stimulation. Knockdown of KCNQ1OT1 inhibited cell proliferation and inhibits the α-SMA and Fibronectin expression of TGF-β1-induced HK-2 cells. In addition, bioinformatics analysis and dual-luciferase reporter assay indicated that miR-124-3p was a target gene of KCNQ1OT1. Mechanistically, silencing miR-124-3p abolished the repressive effects of KCNQ1OT1 on TGF-β1-induced HK-2 cells. In conclusion, KCNQ1OT1 knockdown plays an anti-fibrotic effect through promotion of miR-124-3p expression in renal fibrosis, which provides a promising therapeutic target for the treatment of renal fibrosis.

Lignin, a biological resource with great potential, can be as high as ∼16% of the total organics in the waste activated sludge (WAS). This work therefore aims to fill the knowledge gap about the effect of lignin on short-chain fatty acids (SCFAs) production from anaerobic fermentation of sludge. Experimental results showed that lignin promoted rather than inhibited SCFAs production. Specifically, the presence of 15% lignin promoted the SCFAs production from 129.1 ± 6.5 to 223.14 ± 7.8 mg COD/g VSS compared with the control, and the proportion of acetic increased by 61.8%, while that of propionic decreased by 44.9%. Mechanism exploration revealed that lignin improved the solubilization of biodegradable substrates due to its hydrophobic characteristics. In addition, lignin enhanced the acidogenesis process, possibly by perfecting the electron transfer chain in the fermentation system, and the quinone structure in lignin may compete electrons with methanogens to inhibit the consumption of SCFAs. Microbiological analysis showed that the abundance of microorganisms related to acidogenesi, especially the acetogenesis, including Proteiniclasticum sp., Acetoanaerobium sp., in the fermenter with lignin increased, which caused the community to shift towards specialized and diverse SCFAs production., (Copyright © 2022 Elsevier Ltd. Two-dimensional calcium terephthalate flakes with a thickness of 100-300 nm have been synthesized via adjusting the water : ethanol volume ratio in the solvothermal method. As anodes for sodium-ion batteries, 2D sheet electrodes possess fast kinetics and thus display superior cycling capacity and high-rate capability (195 mA h g -1 at 1000 mA g -1 ).

Producing methane from anaerobic co-digestion of kitchen waste and waste activated sludge has been widely implemented in real-world situations. However, the fate and impact of allyl isothiocyanate (AITC), a main active component in cruciferous vegetables, in the anaerobic co-digestion has never been documented. This study therefore aims to provide such supports. Experimental results exhibited that AITC was degraded completely by microorganisms and served as a substrate to produce methane. As AITC increased from 0 to 60 mg/L, the maximum methane production decreased from 285.1 to 35.8 mL/g VS, and the optimum digestion time was also prolonged. The mechanism study demonstrated that AITC induced cell apoptosis by modifying the physicochemical properties of cell membrane, which resulted in inhibitions to the procedure of anaerobic co-digestion. The high-throughput sequencing showed that AITC enriched the microorganism for degradation of complex organic compounds such as Bacillus, but lessened anaerobes involved in hydrolysis, acidogenesis, and methanogenesis., (Copyright © 2021 Elsevier Ltd. Two diverse conjugative plasmids can interact within bacterial cells. However, to the best of our knowledge, the interaction between bla CTX-M -bearing IncFII plasmid and mcr-1 -carrying IncI2 plasmid colocated on the same bacterial host has not been reported. This study was initiated to explore the interaction and to analyze the reasons that these two plasmids are often coresident in multidrug-resistant Escherichia coli. To assess the interactions on plasmid stabilities, fitness costs, and transfer rates, we constructed two groups of isogenic derivatives, C600 FII , C600 I2 , and C600 FII+I2 of E. coli C600 and J53 FII , J53 I2 , and J53 FII+I2 of E. coli J53, respectively. We found that carriage of FII and I2 plasmids, independently and together, had not impaired the growth of the bacterial host. It was difficult for the single plasmid FII or I2 in E. coli C600 to reach stable persistence for a long time in an antibiotic-free environment, while the stability would be striking improved when they coresided. Meanwhile, plasmids FII and I2, whether together or apart, could notably enhance the fitness advantage of the host; moreover, E. coli coharboring plasmids FII and I2 presented more obvious fitness advantage than that carrying single plasmid FII. Coresident plasmids FII and I2 could accelerate horizontal cotransfer by conjugation. The transfer rates from a strain carrying coresident FII and I2 plasmids increased significantly when it mated with a recipient cell carrying one of them. Our findings highlight the advantages of coinhabitant FII and I2 plasmids in E. coli to drive the persistence and spread of plasmid-carried bla CTX-M and mcr-1 genes, although the molecular mechanisms of their coresidence warrant further study. IMPORTANCE More and more Enterobacteriaceae carry both bla CTX-M and mcr-1 , which are usually located on IncFII-type and IncI2-type plasmids in the same bacterial host, respectively. However, the study on advantages of coresident plasmids in bacterial host is still sparse. Here, we investigated the stability, fitness cost, and cotransfer traits associated with coresident IncFII-type and IncI2-type plasmids in E. coli. Our results show that coinhabitant plasmids in E. coli are more stable, confer more fitness advantages, and are easier to transfer and cotransfer than a single plasmid IncFII or IncI2. Our findings confirm the advantages of coresident plasmids of bla CTX-M -bearing IncFII and mcr-1 -bearing IncI2 in clinical E. coli, which will pose a serious threat to clinical therapy and public health.

An attractive method for assembling densely substituted pyridine derivatives from 1-methyl-1,3-(ar)enynes and nitriles via a formal [4+2] cycloaddition has been established. The well-balanced affinities of two alkali metal salts enable C(sp 3 )-H bond activation and excellent chemo- and regioselectivities. Experimental studies revealed that nitrile functions only as a partial nitrogen source for pyridine synthesis, and the addition of a metalated imine intermediate to an intramolecular alkyne is the rate-limiting step.

Precipitation may increase or decrease by different intensities, but the pattern and mechanism of soil microbial community assembly under various precipitation changes remain relatively underexplored. Here, although ±30% precipitation caused a small decrease (∼19%) in the within-treatment taxonomic compositional dissimilarity through the deterministic competitive exclusion process in a steppe ecosystem, ±60% precipitation caused a large increase (∼35%) in the dissimilarity through the stochastic ecological drift process (random birth/death), which was in contrast with the traditional thought that increasing the magnitude of environmental changes (e.g., from +30% to +60%) would elevate the importance of deterministic relative to stochastic processes. The increased taxonomic dissimilarity/stochasticity under ±60% precipitation translated into functional dissimilarity/stochasticity at the gene, protein, and enzyme levels. Overall, our results revealed the distinctive pattern and mechanism of precipitation changes affecting soil microbial community assembly and demonstrated the need to integrate microbial taxonomic information to better predict their functional responses to precipitation changes., Competing Interests: The authors declare that they have no competing interests., (© 2022 The Author(s).)

Sodium dodecylbenzene sulfonate (SDBS), a typical surfactant being widely used in various applications, was highly accumulated in waste activated sludge. To date, however, its effect on hydrogen production from dark fermentation of sludge has not been documented. The work therefore aimed to explore whether and how SDBS affects hydrogen production. Experimental results showed that with an increase of SDBS from 0 to 30 mg/g TSS, the maximal hydrogen yield increased from 2.47 to 10.73 mL/g VSS (without any treatment) and from 13.05 to 23.51 mL/g VSS (under free ammonia pretreatment). Mechanism exploration showed that SDBS lowered surface tension, facilitated organics transfer from solid to liquid. SDBS also destroyed hydrogen bonding networks of protein, promoted macromolecular organics degradation. Besides, SDBS improved the electric charge in organics, then weakened the mutual repulsion, improved adsorb, interact and promoted the availability of reaction sites between anaerobes and organic substances. Enzyme activity analysis showed that SDBS not only improved the activities of enzymes related to hydrolysis and acidification processes, but also inhibited the activities of homoacetogens and methanogens. SDBS presence lowered sludge ORP and created an environment which was helpful to the growth of butyric-type bacteria, thus enhanced butyric-type fermentation, which contributed hydrogen production largely. Microbial community analysis revealed that SDBS existence affected distributions of microbial populations, and increased the abundances of hydrogen producing microorganisms (e.g., unclassified&#95;f&#95;Synergistaceae). The expanding use of chitosan in sewage and sludge treatment processes raises concerns about its potential environmental impacts. However, investigations of the impacts of chitosan on sewage sludge anaerobic digestion where chitosan is present at substantial levels are sparse. This study therefore aims to fill this knowledge gap through both long-term and batch tests. The results showed that 4 g/kg total suspended solid (TSS) chitosan had no acute effects on methane production, but chitosan at 8-32 g/kg TSS inhibited methane production by 7.2-30.3%. Mass balance and metabolism of organic analyses indicated that chitosan restrained the transfer of organic substrates from solid phase to liquid phase, macromolecules to micromolecules, and finally to methane. Further exploration revealed that chitosan suppressed the secretion of extracellular polymeric substances of anaerobes by occupying the connection sites of indigenous carbohydrates and increased the mass transfer resistance between anaerobes and substrates, which thereby lowered the metabolic activities of anaerobes. Although chitosan could be partly degraded by anaerobes, it is much more persistent to be degraded compared with indigenous organics in sludge. Microbial community and key enzyme encoding gene analyses further revealed that the inhibition of chitosan to CO 2 -dependent methanogenesis was much severer than that to acetate-dependent methanogenesis.

The aims of this study were to elucidate the role of IS 1294 in plasmid reorganization and to analyze biological characteristics of cointegrates derived from different daughter plasmids. The genetic profiles of plasmids in Escherichia coli strain C21 and its transconjugants were characterized by conjugation, S1 nuclease pulsed-field gel electrophoresis (S1-PFGE), Southern hybridization, whole-genome sequencing (WGS) analysis, and PCR. The traits of cointegrates were characterized by conjugation and stability assays. bla CTX-M-55 -bearing IncI2 pC21-1 and nonresistant IncI1 pC21-3, as conjugative helper plasmids, were fused with nonconjugative rmtB -bearing IncN-X1 pC21-2, generating cointegrates pC21-F1 and pC21-F2. Similarly, pC21-1 and pC21-3 were fused with nonconjugative IncF33:A-:B- pHB37-2 from another E. coli strain to generate cointegrates pC21-F3 and pC21-F4 under experimental conditions. Four cointegrates were further conjugated into the E. coli strain J53 recipient at high conjugation frequencies, ranging from 2.8 × 10 -3 to 3.2 × 10 -2 . The formation of pC21-F1 and pC21-F4 was the result of host- and IS 1294- mediated reactions and occurred at high fusion frequencies of 9.9 × 10 -4 and 2.1 × 10 -4 , respectively. Knockout of RecA resulted in a 100-fold decrease in the frequency of plasmid reorganization. The phenomenon of cointegrate pC21-F2 and its daughter plasmids coexisting in transconjugants was detected for the first time in plasmid stability experiments. IS 26 - orf - oqxAB was excised from cointegrate pC21-F2 through a circular intermediate at a very low frequency, which was experimentally observed. To the best of our knowledge, this is the first report of IS 1294 -mediated fusion between plasmids with different replicons. This study provides insight into the formation and evolution of cointegrate plasmids under different drug selection pressures, which can promote the dissemination of MDR plasmids. IMPORTANCE The increasing resistance to β-lactams and aminoglycoside antibiotics, mainly due to extended-spectrum β-lactamases (ESBLs) and 16S rRNA methylase genes, is becoming a serious problem in Gram-negative bacteria. Plasmids, as the vehicles for resistance gene capture and horizontal gene transfer, serve a key role in terms of antibiotic resistance emergence and transmission. IS 26 , present in many antibiotic-resistant plasmids from Gram-negative bacteria, plays a critical role in the spread, clustering, and reorganization of resistance determinant-encoding plasmids and in plasmid reorganization through replicative transposition mechanisms and homologous recombination. However, the role of IS 1294 , present in many MDR plasmids, in the formation of cointegrates remains unclear. Here, we investigated experimentally the intermolecular recombination of IS 1294 , which occurred with high frequencies and led to the formation of conjugative MDR cointegrates and facilitated the cotransfer of bla CTX-M-55 and rmtB , and we further uncovered the significance of IS 1294 in the formation of cointegrates and the common features of IS 1294 -driven cointegration of plasmids.
