The aim of this study was to evaluate the interaction between packaging parameters (transmission of light and oxygen) and storage temperatures (4, 20, 40 °C) on nutrient retention of Spinach (Spinacia oleracea) juice, spray-dried in the absence of an added encapsulant. β-Carotene was more susceptible to degradation compared with lutein and α-tocopherol. Under our experimental conditions, it was observed that excluding low fluorescent light intensity and air by vacuum packaging at 20 °C did not seem to improve nutrient retention loss over time (p > 0.05). The rate of β-carotene, lutein and α-tocopherol loss displayed first order reaction kinetic with low activation energy of 0.665, 2.650 and 13.893 kJ/mol for vacuum, and 1.089, 4.923 and 14.142 kJ/mol for non-vacuum, respectively. The reaction kinetics and half-life for β-carotene, lutein and α-tocopherol at 4 °C and non-vacuumed were 2.2 × 10 -2 , 1.2 × 10 -2 , and 0.8 × 10 -2  day -1 , and 32.08, 58.25 and 85.37 day, respectively., (Crown Copyright © 2019. Mate preferences are important causes of sexual selection. They shape the evolution of sexual ornaments and displays, sometimes maintaining genetic diversity and sometimes promoting speciation. Mate preferences can be challenging to study because they are expressed in animal brains and because they are a function of the features of potential mates that are encountered. Describing them requires taking this into account. We present a method for describing and analysing mate preference functions, and introduce a freely available computer program that implements the method. We give an overview of how the program works, and we discuss how it can be used to visualize and quantitatively analyse preference functions. In addition, we provide an informal review of different methods of testing mate preferences, with recommendations for how best to set up experiments on mate preferences. Although the program was written with mate preferences in mind, it can be used to study any function-valued trait, and we hope researchers will take advantage of it across a broad range of traits., (© 2017 European Society For Evolutionary Biology. The question of why males of many species produce elaborate mating displays has now been largely resolved: females prefer to mate with males that produce such displays. However, the question of why females prefer such displays has been controversial, with an emerging consensus that such displays often provide information to females about the direct fitness benefits that males provide to females and/or the indirect fitness benefits provided to offspring. Alternative explanations, such as production of arbitrarily attractive sons or innate pre-existing female sensory or perceptual bias, have also received support in certain taxa. Here, we describe multivariate female preference functions for male acoustic traits in two chirping species of field crickets with slow pulse rates; our data reveal cryptic female preferences for long trills that have not previously been observed in other chirping species. The trill preferences are evolutionarily pre-existing in the sense that males have not (yet?) exploited them, and they coexist with chirp preferences as alternative stable states within female song preference space. Divergence in mate recognition systems among closely related species is an important contributor to assortative mating and reproductive isolation. Here, we examine divergence in male song traits and female preference functions in three cricket species with songs consisting of long trills. The shape of female preference functions appears to be mostly conserved across species and follows the predictions from a recent model for song recognition. Multivariate preference profiles, combining the pulse and trill parameters, demonstrate selectivity for conspecific pulse rates and high trill duty cycles. The rules for integration across pulse and trill timescales were identical for all three species. Generally, we find greater divergence in male song traits than in associated female preferences. For pulse rate, we find a strong match between divergent male traits and female peak preferences. Preference functions for trill parameters and carrier frequency are similar between species and show less congruence between signal and preference. Differences among traits in the degree of trait-preference (mis)match may reflect the strength of preferences and the potential for linkage disequilibrium, selective constraints and alternative selective pressures, but appear unrelated to selection for mate recognition per se., (© 2015 European Society For Evolutionary Biology. Objective: This study analyzes the relationship between informants' age and their assessment of mental health needs in postconflict society and examines if mental health needs assessment priorities differ depending upon whether or not the informant was exposed to the Liberian civil war., Methods: cross-sectional survey was conducted in March 2009 to obtain data on mental health needs of Liberian children, adolescents and young adults. A total of 171 individuals were interviewed. The data were analyzed using a two- way ANOVA., Results: Elder respondents expressed a preference for young adults to receive services in a church/mosque (F = 4.020, p < .05); for adolescents in volunteer programs (F = 3.987, p < .05) and for children in sports programs (F = 4.396, p < .05). Experiencing conflict did exert some influence on treatment setting preferences. Those who resided outside Liberia during the conflict cited a preference for traditional healers and medical clinics. However, this preference was for the children and young adult age categories. Those who experienced the civil war reported significantly higher preferences for adolescent services to be located in medical clinics, with traditional healers, and in churches/mosques., Conclusion: This study provides additional support for the premise that the utilization of psychiatric services needs to be viewed from the perspective of Liberians and that there are differences in preferences across groups. Our results suggest that service providers and policy makers take into account the age of the patient when deciding where to locate treatment settings for the population.

Poultry, like mammals and other birds, develop fever when exposed to compounds from gram-negative bacteria. Mammals also develop fever when exposed to the constituents of viruses or gram-positive bacteria, and the fevers stimulated by these different pathogenic classes have discrete characteristics. It is not known whether birds develop fever when infected by viruses or gram-positive bacteria. Therefore, we injected Pekin ducks with muramyl dipeptide, the cell walls of heat-killed Staphylococcus aureus, or the viral mimic polyinosinic:polycytidylic acid and monitored their body temperature (T(b)). For comparative purposes we also injected a group of ducks with lipopolysaccharide, the only known pyrogen in birds. We then compared the T(b) invoked by each injection with the T(b) after an injection of saline. Muramyl dipeptide did not affect T(b). The cell walls of heat-killed S. aureus invoked long-lasting, dose-dependent fevers with relatively low magnitudes. Polyinosinic:polycytidylic acid invoked dose-dependent fevers with high febrile peaks. Fever is a well-known clinical sign of infection in mammals, and the results of this study indicate that the pattern of increase in T(b) could serve as an indicator for diverse pathogenic diseases in birds.

Although Akt is a determinant of cisplatin (cis-diaminedichloroplatinum (CDDP)) resistance in ovarian cancer cells, which is related in part to its inhibitory action on p53 activation, precisely how Akt confers CDDP resistance is unclear. In this study, we show that CDDP induced p53-dependent Fas-associated death domain-like interleukin-1beta-converting enzyme (FLICE)-like inhibitory protein (FLIP) degradation in chemosensitive ovarian cancer cells but not their resistant counterparts. CDDP induced FLIP-p53-Itch interaction, colocalization and FLIP ubiquitination in chemosensitive but not chemoresistant ovarian cancer cells. Moreover, although activated Akt inhibited CDDP-induced FLIP degradation and apoptosis in sensitive cells, these responses were facilitated by dominant-negative Akt expression in chemoresistant cells. Inhibition of Akt function also facilitated p53-FLIP interaction and FLIP ubiquitination, which were attenuated by p53 silencing. These results suggest that Akt confers resistance, in part, by modulating CDDP-induced, p53-dependent FLIP ubiquitination. Understanding the precise etiology of chemoresistance may improve treatment for ovarian cancer.

Human pharmacoscintigraphic behavior of two tablets and a capsule formulation of a high dose, poorly water soluble, highly permeable, micronized drug (efavirenz) was investigated. The tablets and capsule, prepared with samarium oxide and neutron activated to produce radioactive samarium-153, were evaluated for their in vivo disintegration and gastrointestinal (GI) transit in healthy subjects under fasted condition. Scintigraphic images were acquired to coincide with blood sampling times to assess the plasma concentration-time profile in relation to in vivo disintegration and GI transit. The mean gastric emptying times were approximately the same for all three formulations. Although in vivo dosage form disintegration was faster for Tablet A as compared to Tablet B and was similar between Tablet A and the capsule, Tablet A showed a slower rate and extent of drug absorption than Tablet B and the capsule. The results of this study eliminated the initial hypothesis that the difference in in vivo performance between the two tablet formulations is due to a different rate of in vivo disintegration and suggest that for this drug the in vivo dissolution rate of the drug from its disintegrated dosage form was a more important factor affecting the rate and extent of drug absorption., (Copyright 2007 Wiley-Liss, Inc.)

Spinocerebellar ataxia type 1 (SCA1) is an incurable neurodegenerative disease resulting from loss of Purkinje neurones within the cerebellum. The ubiquitin proteasome pathway (UPP) has been implicated in SCA1 but the role of proteolysis in the disease is still poorly understood. To further investigate this issue in vivo, genetic crosses were performed between an established mouse model of SCA1 and novel strains expressing elevated levels of wild type or mutant isoforms of ubiquitin. The K48R mutant isoform of ubiquitin (a dominant negative inhibitor of proteolysis) was found to significantly delay the deterioration of Purkinje neurones as evidenced by behavioural, morphological, and molecular indicators. This delay was accompanied by stabilization of p300/CBP, transcriptional mediators whose abundance and activity would otherwise decline in the course of the SCA1 disease, and persistence of protein kinase C gamma (PKCgamma), a protein involved in Purkinje cell dendritic development that is mutated in one form of spinocerebellar ataxia. Whereas the stabilization of p300/CBP was found to occur at the post-translational level the modulation of PKCgamma was at the level of transcription. These results are consistent with transcriptional dysregulation as a key mechanism in neurodegeneration through loss of p300/CBP. Further, the results suggest that the UPP is a potentially useful target for the development of novel therapies for the treatment of neurodegenerative disease.

The ubiquitin-proteasome system (UPS) is a central component in the cellular defence against potentially toxic protein aggregates. UPS dysfunction is linked to the pathogenesis of both sporadic and inherited neurodegenerative diseases, including dominantly inherited familial amyotrophic lateral sclerosis (fALS). To investigate the role of the UPS in fALS pathogenesis, transgenic mice expressing mutant G9 3A Cu,Zn superoxide dismutase (SOD1) were crossed with transgenic mice expressing epitope tagged, wild-type or dominant-negative mutant ubiquitin (Ub(K48R)). RNase protection assays were used to confirm expression of the Ub transgenes in spinal cord and ubiquitin transgene levels were estimated to account for 9-12% of total ubiquitin. Mice expressing the G9 3A transgene exhibited neurological symptoms and histopathological changes typical of this model irrespective of ubiquitin transgene status. Impaired rotarod performance was observed in all G9 3A transgenics by 7 weeks of age irrespective of ubiquitin genotype. The presence of wild-type or mutant ubiquitin transgenes resulted in a small but significant delay in the onset of clinical symptoms and mild acceleration of disease progression, without influencing overall survival. These data suggest that relatively small changes in ubiquitin expression can influence the development of neurodegenerative disease and are consistent with a neuroprotective role for the UPS.

The main objective of this study was to determine whether or not the renal outputs of the osmoregulatory hormones arginine vasotocin (AVT) and aldosterone (ALDO) reflect the osmotic status of whitebellied sunbirds (Nectarinia talatala). The birds were fed a range of sucrose concentrations (from 0.07 to 2.5 mol/l, with osmolalities of 70 to approximately 5,800 mosM/kg), and adjusted their intakes so that they drank large volumes of dilute diets and small volumes of concentrated diets. Renal fluid outputs were appropriately regulated so that large volumes of cloacal fluid (CF) were voided on the dilute diets and small volumes on the concentrated diets. Accordingly, plasma AVT concentrations increased with increasing sugar concentration; however, AVT outputs in CF did not change in a similar manner, rather they decreased as dietary concentration increased. It was not possible to measure plasma ALDO concentrations in the small sunbirds because of insufficient blood samples available; however, ALDO outputs in CF did vary with the sucrose diets and renal function, being highest on the most concentrated diet. In addition ALDO output in CF fell markedly when sodium was added to the 0.5 mol/l sucrose diet. We conclude that in sunbirds fed increasingly concentrated sucrose solutions, changes in CF outputs of ALDO, but not AVT, appear to reflect the water flux and hydration state of these birds.

Nectarivores may be required to switch between water conservation and water excretion as rapidly as they change food plants in nature. We examined the rehydration response in Whitebellied Sunbirds (Nectarinia talatala) that had been fed a concentrated sucrose diet (2.5 mol/l) for 2 days and then were switched to a diet 10 x less concentrated (0.25 mol/l) on the morning of the third day. We measured water gain as well as cloacal fluid (CF) volume hourly over 12 h, and analysed CF osmolality and calculated osmotic excretion. CF was also assayed for the osmoregulatory hormone aldosterone (ALDO). As in most water-deprived birds, whitebellied sunbirds cope with water shortage when fed a concentrated sugar diet by almost completely ceasing to void CF. Although osmolality of CF is high, volumes are not sufficient to maintain a steady rate of excretion and the birds retain osmolytes. Immediately upon switching to dilute diet, sunbirds produce copious volumes of CF and osmotic excretion is elevated and maintained at high levels over the first 6 h of rehydration. This stabilises by the afternoon at levels expected for hydrated birds. Some 2-3 h after peak osmotic excretion, there is a peak in the discrepancy between water intake and output, mirrored in an increase in ALDO output. These data suggest that excretion of retained osmolytes is undertaken as soon as water is available, with changes in the body fluid composition occurring subsequently. This study vindicates the use of CF to obtain repeated measurements of changes in the osmoregulatory steroid ALDO in small birds.

The K48R mutant ubiquitin can exert profound in vivo protective effects against a variety of insults, including agents of direct clinical relevance. The manipulation of the ubiquitin/proteasome pathway has enormous potential for clinical benefit, and it is not unreasonable to expect that such benefits will include diseases of aging.

Nectarivorous whitebellied sunbirds, Nectarinia talatala, demonstrate distinct circadian patterns in osmoregulatory parameters. We recorded intake of a 1 mol/l sucrose solution which enabled calculation of total water gain, and collected cloacal fluid for measurements of volume, osmolality and aldosterone concentration. These variables were assessed hourly over 12 h of photophase, and averaged over the 12-h scotophase period. Overnight, when sunbirds were in negative water balance, aldosterone concentrations and outputs were significantly higher than diurnal levels, reflecting a shut-down of cloacal fluid production. Early morning was marked by a high rate of osmotic excretion, disproportionate to water gain or cloacal fluid output, followed by steady intake and cloacal fluid output during the morning and early afternoon. Reduced water flux (decreased feeding and cloacal fluid output) during mid-afternoon was accompanied by a paradoxical decline in osmotic excretion, whilst a significant increase in the discrepancy between water intake and output was recorded as the birds effectively stored water before the scotophase. These patterns of intake and excretion may be informative in explaining drinking and foraging behaviour in the field.

The present study was designed to determine whether the responses of the avian kidney to circulating arginine vasotocin (AVT), under different osmotic conditions, involve an interaction with prostaglandins (PGs). The renal effects of intravenously infused AVT at a dose of 0.1 ng.kg(-1).min(-1) for 45 min were compared in Pekin ducks given maintenance infusions of either 200 mosmol NaCl or glucose at 1 ml.min(-1), with and without PG inhibition by indomethacin. Birds infused with glucose responded with similar degrees of AVT-induced antidiuresis with and without indomethacin, however the urinary excretion of sodium was significantly reduced when PG production was prevented. In ducks receiving saline without indomethacin, the antidiuretic response to AVT was markedly less than that in the glucose-infused birds, however, indomethacin treatment increased the degree of antidiuresis to a level similar to that in the glucose-infused ducks. The results indicate that PGs have important renotropic actions in birds and in particular modulate the antidiuretic effect of AVT in salt- and volume-loaded animals.

The physiological regulation of body water volume and concentration was evaluated in Pekin ducks, Anas platyrhynchos, slowly acclimated to increasingly saline drinking water (six equal 75 mM NaCl increments). Body mass, total body water (TBW), water flux, plasma osmolality (Osm(pl)), and ionic and osmoregulatory hormone concentrations were measured at the end of each increment. The salinity at which each variable deviates from its homeostatic set point was calculated by continuous two-phase linear regression. We hypothesized that, as drinking water salinity increases: (1) body water increases in concentration before it decreases in volume and (2) that regulating variables that help determine homeostatically set values (plasma hormone concentrations and water flux) deviate from values of freshwater ducks at lower drinking water salinities than the variables they regulate (Osm(pl), hematocrit, TBW). Osm(pl) was the first variable for which we could calculate a deviation from its homeostatically controlled value. It increases at much lower drinking water salinity than that at which TBW decreases, supporting our first hypothesis, but not our second hypothesis. We further hypothesized that, because the concentration of Pekin duck salt gland secretion is only slightly higher than that of their drinking water, they increase water flux (drinking) as salinity of drinking water increases, until the latter exceeds the secretion concentration and then they drink less. There was no change in water flux until it decreases when TBW decreases, 329 mM NaCl and 335 mM NaCl, respectively. The results do not support our hypothesis that Pekin ducks increase drinking as the salinity of their drinking water increases, but do indicate that, at tolerable salinities, Pekin ducks maintain body water volume while allowing body water osmolality to increase. At higher salinities, ducks decrease drinking and use body water to get rid of the excess salt.

The compartmentalization of body fluids was measured in individual Pekin ducks ( Anas platyrhynchos) drinking freshwater and after sequential acclimation to 300 mM NaCl and 400 mM NaCl. Total body water, extracellular fluid volume, plasma volume and exchangeable sodium pool were measured using (3)H(2)O, [(14)C]-polyethylene glycol, Evans Blue dye, and (22)Na dilution, respectively. Following acclimation to 300 mM NaCl, body mass decreased, but total body water and total exchangeable sodium pool were unaltered. Na and water were redistributed from the extracellular fluid (interstitial fluid) compartment into the intracellular fluid compartment. Following further acclimation to 400 mM NaCl, body mass, total body water and intracellular fluid volume decreased, but exchangeable sodium pool and extracellular fluid volume were unchanged. Our results suggested that, when Pekin ducks drink high but tolerable salinities, they maintain total body water, but redistribute Na(+) and water from interstitial fluid to the intracellular fluid compartment. When stressed beyond their ability to maintain total body water, they lose water from the intracellular fluid.

There is convincing evidence from studies in yeast that a functional ubiquitin/proteasome pathway is required to degrade misfolded or oxidatively damaged proteins but for technical reasons, it has been difficult to perform comparable studies in mammalian cells. To investigate the possibility that the ubiquitin/proteasome pathway is cytoprotective for mammalian cells, we have introduced epitope-tagged wild-type ubiquitin or dominant-negative mutant versions of ubiquitin into mouse HT4 neuroblastoma cells. Cells expressing mutant versions of ubiquitin were found to be sensitive to cadmium, an agent that causes oxidative damage to cellular components, and to canavanine, an amino acid analog that generates misfolded proteins. The greatest sensitivity to canavanine was observed in cells expressing a mutant version of ubiquitin unable to support the formation of Lys(48) linkages. Substrates of the proteasome were found to accumulate in these cells, suggesting a general deficit in proteolysis. Our data suggest that defects in the ubiquitin-mediated proteolytic system predispose mammalian cells to the toxic effects of abnormal protein.

Objective: To determine whether rates of intestinal fluid absorption and renal diuresis can match high rates of fluid ingestion in healthy humans exposed to oral fluid overload, thereby preventing the development of hyponatraemia either by reverse sodium movement across the intestine (the Priestley-Haldane effect) or by expansion of the extracellular fluid volume., Methods: Changes in renal function and in plasma chemical measurements in response to an oral fluid overload (0.9-1.8 l/h x 3 h) were investigated in 6 healthy control subjects at rest, and in a subject with a history of exercise-induced symptomatic hyponatraemia, during both prolonged (160-minute) exercise and at rest., Findings: All control subjects gained weight (2.7 +/- 0.2 kg, mean +/- standard error of mean (SEM)) because the rate of oral fluid intake exceeded the peak rate of urine production (778 +/- 39 ml/h). Blood volume rose by 7.1 (+/- 0.5)% and plasma sodium concentrations fell progressively from 144 +/- 2.6 to 136 +/- 1.1 mmol/l (P < 0.05) in the control subjects. Plasma potassium and angiotensin II concentrations were unchanged and creatinine clearance was normal (approximately 125 ml/min). Free water clearance reached a maximum of 11.2 +/- 0.9 ml/min after 2 hours. The increase in body mass could be accounted for by calculated or measured changes in extra- and intracellular fluid volumes. Similar changes were measured in the subject with a previous history of symptomatic hyponatraemia., Conclusion: The rate of intestinal fluid absorption appeared to match the rate of oral fluid ingestion and there was no evidence of fluid accumulation in the intestine with reverse sodium movement from the extracellular space into intestinal fluid. The results of this study are therefore at variance with the Priestley-Haldane hypothesis and suggest that reverse sodium movement did not contribute to the hyponatraemia induced by oral fluid overload in these subjects. Rather it appears that humans may have a limited capacity to excrete fluid at rates in excess of approximately 900 ml/h in response to higher rates of oral fluid intake. When the rate of intestinal fluid absorption matches the rate of fluid ingestion and exceeds the kidneys' maximum capacity for fluid excretion, the excess fluid accumulates in the extra- and intracellular fluid compartments, inducing the dilutional hyponatraemia of water intoxication. These findings may have relevance to other clinical conditions in which hyponatraemia develops in response to high rates of oral or intravenous fluid provision.

The murine Unp gene encodes a widely expressed ubiquitin-specific protease. The predicted sequence of the UNP protein features motifs common to viral oncoproteins through which these proteins interact with the retinoblastoma gene product pRb, as well as the related 'pocket proteins' p107 and p130. We have explored the possibility that UNP interacts with pocket proteins, and report here that such associations can be detected in vitro and in cells. Associations of UNP and pocket proteins are sensitive to site-directed mutations in a manner directly analogous to those documented in viral oncoproteins. We conclude that within cells UNP does physically associate with pRb, and can also associate with p107 and p130.

The primary pathway for the proteolytic destruction of cellular proteins is through ubiquitin-mediated targeting to the proteasome. This pathway is pivotal not only in the elimination of damaged or misfolded proteins but also in the temporal, developmental, or signal-mediated destruction of normal cellular substrates. The list of known substrates of the ubiquitin/proteasome pathway is long, but most substrates have been identified in yeast or, more recently, in cultured mammalian cells. It is likely that many mammalian substrates with developmental or disease relevance have yet to be identified because their ubiquitination occurs in tissue or organ systems that cannot be adequately modeled in vitro. We have developed a transgenic mouse model that will allow the isolation and identification of these substrates. The human UbC promoter was used to drive expression of a hexahistidine-tagged version of human ubiquitin in a variety of mouse tissues from early embryonic stages, as assessed by a green fluorescent protein marker. Cleavage of the fusion protein by endogenous enzymes produced epitope-tagged ubiquitin that was detected both in monomeric form and conjugated to cellular proteins. This mouse model should facilitate in the analysis of normal and disease-related ubiquitination events in vivo.

This study was designed to further examine the role of plasma ANG II in the excretion of sodium in the Pekin duck, a bird with salt glands. Renal and extrarenal (salt gland) excretion of an intravenously administered isotonic saline load was monitored over a 4-h period in a group of eight birds under two conditions: the control condition, in which isotonic saline infusion decreased endogenous plasma ANG II from 102.6 to 16.5 pg/ml, and the experimental condition, in which ANG II suppression was prevented by intravenous infusion of a 3.5 ng. kg(-1). min(-1) dose of synthetic ANG II. ANG II infusion significantly decreased the total sodium excretion (by 15%), primarily via an inhibition of salt gland output. The results suggest that ANG II suppression facilitates the excretion of an administered sodium load in birds with salt glands.

This study documents growth and dual-energy X-ray absorptiometry (DXA)-determined peak bone mass profiles in the rabbit model, and tests the hypothesis that rabbits show patterns of bone accretion similar to humans and thus may serve as a viable model for human bone physiology. It is also shown that dietary Ca intake affects peak bone mass and the temporal pattern of its attainment. Groups of weaned animals were administered two nutritionally complete but calcium-altered diets (0.5% or 1.0% Ca). We evaluated growth, bone mass accretion, and Ca metabolism from 20 to 56 weeks of age in both the 1.0% Ca and 0.5% Ca groups of rabbits. For each monthly period, we monitored body mass, naso-tail length, food consumption, and fecal output. In addition, we collected blood and 24 h urine samples for biochemical analyses, and measured bone mass variables of the lumbar spine with DXA. The 1.0% Ca group had a lower apparent fractional absorption of Ca and higher urinary Ca excretion, but retained more Ca than the 0.5% Ca group during the growth phase. Furthermore, the 1.0% Ca group had lower parathyroid hormone (PTH) and bone biochemical marker concentrations throughout the study than the 0.5% Ca group. The lower levels of PTH and bone markers of resorption and formation, may have resulted in a reduction in skeletal remodeling, and this physiological mechanism may have contributed to the 10% increase in peak bone mineral density of the lumbar spine in the 1.0% Ca group of rabbits.

Ubiquitin can be detected in the neuronal and glial inclusions that are the diagnostic hallmarks of a number of human neurodegenerative diseases. It has been assumed that the presence of ubiquitin signifies the failed attempt of the cell to remove abnormal protein structures, which have been allowed to aggregate. The burden of abnormal protein arising from genetic mutations or cumulative oxidative damage might in the course of time overwhelm the ubiquitin-proteasome pathway (whose responsibility it is to eliminate misfolded or damaged proteins). However, ubiquitin may still serve a protective purpose distinct from its role in proteolysis. The physical properties of ubiquitin are such that a surface coating of ubiquitin should preclude further growth of the aggregate, prevent non-productive interactions, and conceal the contents from detection mechanisms that might ultimately kill the cell. This 'nonstick coating' hypothesis makes predictions about the nature of the conjugated ubiquitin and the consequences of removing it.

Cold acclimation induces very divergent responses in thyroid function in reptiles and mammals reflective of their different thermoregulatory modes. Naked mole-rats, unlike other small mammals, are unable to effectively employ endothermy and are operatively poikilotherms. We therefore investigated changes in their thyroid status with chronic cold exposure. Under simulated burrow conditions, free thyroxine (T(4); 0.39 +/- 0.09 ng/dl) and thyroid stimulating hormone (TSH; 1.12 +/- 0.56 microIU/ml) levels fell within the reptilian range, one order of magnitude lower than mammalian levels. However, cold induced typical mammalian responses: free T(4) levels (0.55 +/- 0.09 ng/dl) and thyroid follicular cell height were significantly greater. Although TSH levels (1.28 +/- 0.83 microIU/ml) were not significantly elevated, thyrotrophs exhibited ultrastructural signs of increased secretory activity. Low thyroid hormone concentrations may contribute substantially to the unusual thermoregulatory mode exhibited by naked mole-rats.

Supercritical fluids can extract components from some matrixes (e.g., fat and flavors from food) as well as infusing additives into synthetic polymer matrixes. To study the feasibility of infusing flavors into matrixes as a potential flavoring mechanism, a wide range of volatile flavor compounds was infused into a well-defined synthetic polymer (low-density polyethylene) using supercritical carbon dioxide. The polymer was then extracted, and the amount of infused compound was determined. The effects of time, temperature, pressure, rate of depressurization, volatile concentration, and volatile properties on the degree of infusion were studied. Infusion with supercritical carbon dioxide achieved much higher loadings of the polymer (0.01 to 6.87 mg/g LDPE, depending on the volatile molecule being infused) compared to those achieved by static diffusion. Forty-five volatiles were infused, from which a model was developed to predict infusion as a function of certain physicochemical properties.

Recent theoretical work has shown that sexual selection may cause speciation under a much wider range of conditions than previously supposed. There are, however, no empirical studies capable of simultaneously evaluating several key predictions that contrast this with other speciation models. We present data on male pulse rates and female phonotactic responses to pulse rates for the field cricket Gryllus texensis; pulse rate is the key feature distinguishing G. texensis from its cryptic sister species G. rubens. We show (i) genetic variation in male song and in female preference for song, (ii) a genetic correlation between the male trait and the female preference, and (iii) no character displacement in male song, female song recognition, female species-level song discrimination, or female song preference. Combined with previous work demonstrating a lack of hybrid inviability, these results suggest that divergent sexual selection may have caused speciation between these taxa.

Male sexual displays provide females with information that is crucial to their reproductive decisions. That same information is available to eavesdroppers, with potential consequences for both signaller and receiver. We present empirical evidence for size-dependent responses to intersexual communication by conspecific rivals. Acoustic features of a male house cricket's (Acheta domesticus) mating call are positively associated with its size, with females preferring the calls of larger males. In order to investigate whether conspecific males make use of the information content of mating calls, we examined their phonotactic responses to call recordings that differ in attractiveness to females. Males of all sizes exhibited positive phonotaxis, with smaller males showing a clear preference for female-preferred calls. Smaller males were also less likely to seek contact with the speaker playing their chosen call. We discuss possible explanations for this size-dependent male behaviour.

The paper describes the relationship between the physicochemical characteristics of model oi-in-water emulsion systems containing oil droplets and soluble fragments (monomers and micelles) and the activity of pure soybean lipoxygenase 1. The method of emulsification, as well as the chemistry and concentration of surfactant and substrate molecules, was manipulated to modify the overall physicochemical properties of ternary emulsions. The effect of these experimental factors on the size and relative concentration of oil droplets and soluble fragments was examined with static light scattering and turbidity measurements. Lipoxygenase 1 activity was assayed polarographically. Experimental factors that increase the critical micelle concentration of the monomers and facilitate their transport out of micelles, as well as across the oil-water interfaces and into the aqueous phase, were shown to improve the activity of the enzyme and vice versa.

The objective of this study was to determine the effect of modulating the plasma concentrations of the avian antidiuretic hormone, arginine vasotocin (AVT), upon the febrile response to lipopolysaccharide (LPS) in Pekin ducks. LPS, intravenously administered into conscious control birds at a dose of 1 microg x kg(-1), caused a monophasic increase in body temperature of 0.85 +/- 0.12 degrees C associated with a Thermal Response Index of 2.5 +/- 0.6 C degrees h. Plasma AVT concentrations in the control birds also increased with the progression of the fever response, more than doubling from their basal values. Ducks in which the circulating level of AVT had either been elevated by the intravenous infusion of the peptide or dehydration, or reduced by the administration of a specific AVT antibody prior to LPS administration, produced body temperature profiles and Thermal Response Index values that did not differ significantly from those of the control birds. The lack of any direct effect of variations in plasma AVT concentrations upon the magnitude of the fever response indicates that the LPS-induced elevation in plasma AVT is not associated with modulating the rise in body temperature obtained in avian fever.

The rabbit could be a superior animal model to use in bone physiology studies, for the rabbit does attain true skeletal maturity. However, there are neither normative bone mineral density (BMD) data on the rabbit nor are there any validation studies on the use of dual-energy X-ray absorptiometry (DXA) to measure spinal BMD in the rabbit. Therefore, our aim was twofold: first, to investigate whether DXA could be used precisely and accurately to determine the bone mineral content (BMC). bone area (BA). and BMD of the rabbit lumbar spine: Second. to evaluate the new generation fan-beam DXA (Hologic QDR-4500) with small animal software by comparing two DXA methodologies QDR-1000 and QDR-4500 with each other, as well as against volumetric bone density (VBMD) derived from Archimedes principle. As expected. there was a magnification error in the QDR-4500 (BMC, BA. and BMD increased by 52%. 38%. and 10%, respectively, when the vertebrae were positioned flat against the scanning table). With the magnification error kept constant (vertebrae positioned 10 cm above the scanning table to match the height in vivo). there were no differences among the mean BMC. BA. and BMD of the rabbit vertebrae (Ll-L7) in vivo and in vitro using the QDR-4500 (p > 0.05). BMC, BA, and BMD differed between QDR-1000 and QDR-4500 in vitro because of a magnification error when the vertebrae were flat on the table (p <0.0001). and, consequently. the machines did not correlate with one another (p > 0.05). However, the BMC, BA, and BMD of the two DXAs did significantly correlate with each other in vivo and in vitro when the magnification error was compensated for (r = 0.44 and 0.52. i2 = 0.45 and 0.63. and 12 = 0.41 and 0.60. respectively. p < 0.05-0.008). The BMC and BMD (in vivo and in vitro) of the rabbit vertebrae measured by QDR-4500 was significantly correlated with VMBD, ash weight, and mineral content (,2 = 0.67-0.90,j <0.01-0.0001). Therefore, the QDR-4500 can be used to yield precise and accurate measurements of the rabbit spine.

Many indoor household pesticides are efficient and useful tools for a variety of functions necessary to maintain clean, sanitary, and pleasant homes and institutional facilities, and to provide significant public health benefits. They do so by incorporating active ingredients and formulation technology that have not been associated with significant environmental impact in use or when disposed in landfills. Chemical and environmental fate properties, toxicological characteristics, and use patterns of indoor household pesticides that distinguish them from other categories of pesticides which have been associated with environmental contamination should be recognized when HHW policy is debated and established by governmental agencies. Most indoor household pesticides as defined here should not be considered hazardous waste or HHW because those relatively few containers, often no longer full, that have been disposed with MSW over the years have not been associated with environmental contamination. The tiny amounts of those product residues that will reach MSW landfills have been shown, in general, not to have chemical or environmental fate characteristics that would make them susceptible to leaching. Those that do have the potential to leach based on these characteristics, in most cases, do not represent a threat to human health based on toxicological considerations. However, compounds such as propoxur, which are very mobile and relatively persistent in soil and in addition have been associated with significant potential health effects, may be targeted by the screening process as described here and could be selected for further investigation as candidates for special waste management status (such as HHW). Our analysis and recommendations have not been extended to the many types of lawn and garden pesticides that are commonly used by homeowners and are frequently brought to HHW programs. However, their potential for groundwater contamination could also be judged using the same technical considerations as applied in this review to indoor household pesticides. In light of the very high costs of diverting wastes from the MSW stream and into HHW programs, it is recommended that, as a matter of public policy, all categories of household waste that might be considered as HHW be carefully and objectively evaluated for their potential to harm public health or the environment after disposal at MSW landfills.

Background: There are no large published studies of the prevalence of seasonal affective disorder (SAD) among UK populations., Aim: To determine the prevalence of SAD among patients attending a general practitioner (GP)., Method: Patients aged 16-64 consulting their GPs in Aberdeen during January were screened with the Seasonal Pattern Assessment Questionnaire (SPAQ). SPAQs were also mailed to 600 matched patients, who had not consulted their GP during January. Surgery attenders who fulfilled SPAQ criteria for SAD were invited for interview to determine whether they met criteria for SAD in DSM-IV and the Structured Interview Guide for the Hamilton Rating Scale for Depression--Seasonal Affective Disorder Version (SIGH-SAD)., Results: Of 6161 surgery attenders, 4557 (74%) completed a SPAQ; 442 (9.7%) were SPAQ cases of SAD. Rate of caseness on the SPAQ did not differ between surgery attenders and non-attenders. Of 223 interviewed SPAQ cases of SAD, 91 (41%) also fulfilled DSM-IV and SIGH-SAD criteria., Conclusions: There is a high prevalence of SAD among patients attending their GPs in January in Aberdeen; this is likely to reflect a similar rate in the community.

The interferon-inducible, double-stranded RNA-dependent protein kinase PKR has been implicated in anti-viral, anti-tumor, and apoptotic responses. Others have attempted to examine the requirement of PKR in these roles by targeted disruption at the amino terminal-encoding region of the Pkr gene. By using a strategy that aims at disruption of the catalytic domain of PKR, we have generated mice that are genetically ablated for functional PKR. Similar to the other mouse model of Pkr disruption, we have observed no consequences of loss of PKR on tumor suppression. Anti-viral response to influenza and vaccinia also appeared to be normal in mice and in cells lacking PKR. Cytokine signaling in the type I interferon pathway is normal but may be compromised in the erythropoietin pathway in erythroid bone marrow precursors. Contrary to the amino-terminal targeted Pkr mouse, tumor necrosis factor alpha-induced apoptosis and the anti-viral apoptosis response to influenza is not impaired in catalytic domain-targeted Pkr-null cells. The observation of intact eukaryotic initiation factor-2alpha phosphorylation in these Pkr-null cells provides proof of rescue by another eukaryotic initiation factor-2alpha kinase(s).

1. The objective of this study was to determine the changes in plasma concentrations of the hormones arginine vasotocin (AVT) and angiotensin II (AII) associated with lipopolysaccharide (LPS)-induced fever in Pekin ducks. 2. LPS, intravenously administered into conscious birds at doses of 1, 10 and 100 microgram kg-1, caused dose-dependent and monophasic increases in body temperature, with fever index values of 3.5, 7.0 and 10.6, respectively. 3. Plasma AVT concentrations also increased with the progression of the fever, with the largest elevation (from 8.4 +/- 1. 6 to 25.2 +/- 3.2 pg ml-1; means +/- s.e.m., n = 7) being caused by the highest dose of LPS. 4. Plasma AII concentrations did not significantly change from basal values (mean of 45.5 +/- 6.3 pg ml-1 for all groups) during the acute phase of the fever response. 5. The osmotic status of the birds, as indicated by plasma osmolality and electrolyte values, did not significantly change in any of the experimental animals. 6. The elevation of AVT in avian fever leads to speculation about a possible antipyretic action of this hormone, which would have particular relevance to understanding the evolution of fever.

The murine Unp gene encodes a ubiquitin-specific protease, a member of a family of enzymes that includes the product of the human tre-2 oncogene. The Unp gene has previously been mapped to chromosome 9. We have cloned in bacteriophage a 50 kilobase region of chromosome 9 containing the Unp gene, and have determined the nucleotide sequence of the gene. The gene has 22 exons, distributed over 47.4 kb. A processed ribosomal S2 pseudogene was identified in the third intron of the Unp gene. Expression of Unp is driven by a GC-rich, 'housekeeping' type promoter., (Copyright 1998 Elsevier Science B.V. Background: Little is known about the prevalence or the consultation patterns of patients with seasonal affective disorder (SAD) in primary care settings., Methods: Patients aged 16-64 years consulting a general practitioner (G.P.) during a three week period in January were screened with the Seasonal Pattern Assessment Questionnaire (SPAQ). Interview-confirmed cases of SAD (N = 33) were matched by age and sex to controls without seasonal morbidity and primary care consultation patterns were compared over a 5 year period., Results: Of 692 patients screened, 64 (9.3%) satisfied SPAQ criteria for winter SAD. Patients with SAD showed no winter excess of consultations but were high year round consulters, averaging 1.8 times more consultations than controls., Conclusions: High levels of SAD caseness on the SPAQ were found among patients consulting in primary care during January. Possible explanations for the high non-seasonal consultation patterns among SAD patients include somatisation and diagnostic inaccuracies., Limitations: Diagnostic criteria for SAD, and the SPAQ in particular, may be over-inclusive. The SPAQ completion rate (about 39% of eligible patients consulting a G.P.) was relatively low., Clinical Relevance: Patients with SAD (using SPAQ criteria) commonly consult their G.P.s in winter but frequency, rather than seasonality, of consultation may be a better guide to diagnosing SAD in primary care.

We measured body temperature in Pekin ducks for 22 h after intravenous injection of the lipopolysaccharide (LPS) of gram negative bacteria at doses of 0, 1, 10, and 100 micrograms.kg body mass (-1). The ducks developed monophasic fevers showing increases in peak temperature reached and duration of fever with increases in dose of LPS. Body temperatures of unrestrained telemetered ducks without access to food and water were similar to those of saline-injected controls in the fever experiments, but were lower in the morning than when the same birds had access to food and water. This nocturnal hypothermia may have resulted from energy restriction imposed by lack of food and water. The dose of LPS required to elicit a fever of over 18 h duration (100 micrograms.kg-1) will elicit a biphasic fever of 5 h duration in rats. Pekin ducks did not exhibit biphasic fever even at the highest LPS dose administered, indicating that while fever is superficially similar in the two homeothermic classes, there may be differences in details of the mechanism. The similarities of the dose/response characteristics to that of mammals lends support to the theory that fever in vertebrates has a common phyletic origin.

Ubiquitin is a small eukaryotic protein that is synthesized naturally as one of several fusion proteins, which are processed by ubiquitin-specific proteases to release free ubiquitin. The expression of heterologous proteins as fusions to ubiquitin in either prokaryotic or eukaryotic hosts often dramatically enhances their yield, and allows the exposure of any amino acid following cleavage of ubiquitin. The single exception is when proline is the amino acid immediately following ubiquitin; the ubiquitin-proline bond is poorly cleaved by presently studied ubiquitin-specific proteases. We show that the mouse ubiquitin-specific protease Unp, and its human homolog Unph, can efficiently cleave the ubiquitin-proline bond in ubiquitin fusion proteins of different sizes. N-terminal sequencing of the cleavage products reveals that cleavage occurs precisely at the ubiquitin-proline junction. The biological significance of this cleavage activity is unclear, as ubiquitin-proline fusions do not occur naturally. However, it may indicate a different catalytic mechanism for these ubiquitin-specific proteases and/or that they can cleave ubiquitin-like proteins. Unp and Unph thus represent versatile ubiquitin-specific proteases for cleaving ubiquitin-fusion proteins in biotechnology and basic research, regardless of both the amino acid immediately following ubiquitin, and the size of the fusion partner.

Polyclonal antibodies raised in a rabbit against avian atrial natriuretic peptide (ANP) were used to investigate the role of endogenous plasma ANP in the control of salt gland function of conscious, saltwater-adapted Pekin ducks. Salt gland secretion was initiated and maintained either by a hypervolemic (290 mosmol/kg NaCl i.v. at 2 ml/min) or hyperosmotic (1,000 mosmol/kg NaCl i.v. at 0.4 ml/min) stimulus. Both experimental conditions caused significant elevations in endogenous plasma ANP concentrations. At steady states of secretion driven by hypervolemia, the administration of ANP antiserum (anti-ANP), which reduced plasma ANP concentrations by 90%, caused an immediate 30% reduction in fluid secretion rate and sodium excretion that lasted for 20-30 min. The activity of salt glands driven by hyperosmolality was not changed by anti-ANP. The results show that the high circulating concentrations of endogenous ANP associated with conditions of sustained volume expansion promote salt gland secretion.

The Mpv 20 transgenic mouse strain was created by infection of embryos with a defective retrovirus. When Mpv 20 heterozygous animals were crossed, no homozygous neonatal mice or midgestation embryos were identified. When embryos from heterozygous crosses were cultured in vitro, approximately one quarter arrested as uncompacted eight-cell embryos, indicating that proviral insertion resulted in a recessive lethal defect whose phenotype was manifest very early in development. Molecular cloning of the Mpv 20 insertion site revealed that the provirus had disrupted the Npat gene, a gene of unknown function, resulting in the production of a truncated Npat mRNA. Expression of the closely linked Atm gene was found to be unaffected by the provirus.

To quantify the renal excretion of angiotensin II (AII) in birds, synthetic AII was intravenously infused into conscious Pekin ducks at rates of 5, 15 and 45 ng/kg/min for 30 min and the relationships between plasma and urine AII concentrations were monitored by radioimmunoassay. The infusions of AII produced dose-dependent elevations in the plasma concentrations of the hormone, however, urinary concentrations and excretion rates did not change significantly from the basal values of 11.6 +/- 1.2 pg/ml and 4.3 +/- 1.0 pg/min, respectively. The metabolic clearance rate of AII remained constant at each infusion dose (approximately 120 ml/min/kg). The urinary clearance rates (< 0.3 ml/min/kg) indicated that the fraction of AII cleared from the blood via the urine was less than 0.2%. Clearly, excretion of intact AII via this route is not a quantitatively important means of elimination for this hormone in birds.

A promoter-trap retrovirus has been constructed in which a promoterless polyomavirus middle T antigen gene was inserted in the U3 region of the long terminal repeat of a replication-competent Moloney murine leukemia virus. The resulting virus, designated PyT, was used to infect mouse mammary glands in situ. As expected, mammary tumors appeared in some infected animals. These tumors were found to contain PyT proviruses of the predicted structure. From one such tumor, the PyT provirus and surrounding sequences from the integration site were cloned. The provirus was found to have integrated adjacent to the promoter of a novel mouse gene (TRAP1) that was expressed at low levels in various mouse tissues. These data show that the PyT retrovirus provides a sensitive means of detecting active promoters in vivo.

To quantify the renal excretion of atrial natriuretic peptide (ANP) in birds, synthetic ANP was infused intravenously into conscious Pekin ducks at rates of 10, 30 and 90 ng/kg per min for 30 min and the relationships between plasma and urine ANP concentrations were monitored by RIA. The infusions of ANP produced dose-dependent elevations in the plasma concentrations of the hormone; however, urinary concentrations and excretion rates did not change significantly from the basal values of 142.2 +/- 21.5 pg/ml and 26.4 +/- 4.5 pg/min per kg respectively. The metabolic clearance rate of ANP remained constant at each infusion dose (approximately 200 ml/min per kg). The urinary clearance rates (< 0.3 ml/min per kg) indicated that the fraction of ANP cleared from the blood via the urine was less than 0.2%. Clearly, excretion of intact ANP via this route is not a quantitatively important means of elimination for this hormone in birds.

To quantify the renal excretion of arginine vasotocin (AVT) in birds, synthetic AVT was infused into conscious Pekin ducks at rates of 0.03, 0.10, and 0.30 ng/kg/min for 30 min and the relationships between plasma and urine AVT concentrations were monitored by radioimmunoassay. As plasma AVT concentrations increased (from a basal value of 7.8 +/- 1.1 pg/ml to levels of 11.5 +/- 1.9, 17.6 +/- 2.2, and 24.2 +/- 2.5 pg/ml, respectively) the urinary concentrations and excretion rates increased linearly from basal levels of 26.0 +/- 3.1 pg/ml and 14.9 +/- 1.8 pg/min to 215.6 +/- 36.6 pg/ml and 39.2 +/- 3.7 pg/min, respectively. The metabolic clearance rate of AVT (approximately 23 ml/min/kg) and urinary clearance (approximately 0.91 ml/min/kg) remained constant at each infusion dose and were therefore independent of plasma AVT levels. Over the range of plasma AVT concentrations tested, a constant fraction of about 5% of the AVT cleared from the blood was excreted intact in the urine.

The murine Unp proto-oncogene encodes a nuclear ubiquitin protease whose overexpression leads to oncogenic transformation of NIH3T3 cells. We have cloned cDNAs originating from the human homolog of this gene, designated herein as Unph (Unp, human), and have used these cDNAs to map the gene to 3p21.3, a region frequently rearranged in human tumor cells. Unph mRNA levels are consistently elevated in small cell tumors and adenocarcinomas of the lung, suggesting a possible causative role for the gene in the neoplastic process.

Eight endurance-trained cyclists rode as far as possible in 1 h on a stationary cycle simulator in a moderate environment (20 degrees C, 60% relative humidity, 3 m.s-1 wind speed) while randomly receiving either no fluid (NF) or attempting to replace their approximate 1.71 sweat loss measured in a previous 1-h familiarisation performance ride at approximately 85% of peak oxygen uptake with artificially sweetened, coloured water (F). During F, the cyclists drank mean 1.49 (SEM 0.14) 1 of which mean 0.27 (SEM 0.08) 1 remained in the stomach at the end of exercise and mean 0.20 (SEM 0.05) 1 was urinated after the trial. Thus, only mean 1.02 (SEM 0.12) 1 of the ingested fluid was available to replace sweat losses during the 1-h performance ride. That fluid decreased the mean average heart rate from 166 (SEM 3) to 157 (SEM 5) beats.min-1 (P < 0.0001) and reduced the final mean serum [Na+] and osmolalities from 143 (SEM 0.6) to 139 (SEM 0.6) matom.l-1 (P < 0.005) and from 294 (SEM 1.7) to 290 (SEM 1.9) mosmol.l-1 (P = 0.05), respectively. Fluid ingestion did not significantly attenuate rises in plasma anti-diuretic hormone and angiotensin concentrations, or decrease the approximate-15% falls in estimated plasma volume in the F and NF trials. Nor did fluid ingestion significantly affect the approximate 1.71.h-1 sweat rates, the rises in rectal temperature (from 36.6 degrees to 38.3 degrees C) or the ratings of perceived exertion in the two trials.(ABSTRACT TRUNCATED AT 250 WORDS)

A mutant Moloney murine leukemia virus was created in which the binding site for the embryonal long terminal repeat binding protein had been replaced with a synthetic linker sequence. The virus was found to replicate normally in murine 3T3 fibroblasts, and the mutation was stably transmitted throughout repeated passages. The binding site is therefore dispensable with respect to viral replication. Titers of the mutant virus were comparable to those of wild-type preparations, even when supernatants were prepared from undifferentiated P19 embryonal carcinoma cells. Elimination of the repressor binding site therefore had little effect on viral replication in embryonal carcinoma cells. Elimination of the repressor binding site therefore had little effect on viral replication in embryonal cells, an unexpected result in light of the reported activity of the binding protein as a transcriptional repressor.
