Two novel web apps for W7-X are introduced: Profile Cooker and Power House. They are designed to streamline the workflow of profile fitting and power balance analysis while offering a graphical user interface that works in any common browser. This allows us to compile a comprehensive database of experimental power balance results. All fitting functions available in Profile Cooker are presented and compared on the basis of example profiles. The power balance equation assumed in Power House is established and its individual terms are discussed. The main focus of the power balance analysis is on the turbulent transport coefficients. A model for quick calculation of neutral beam power deposition based on experimental profiles is presented. Neoclassical root transition poses an issue for power balance analysis due to the uncertainty of the radial electric field. A global, neoclassical simulation with the code EUTERPE is performed for a set of experimental profiles to gain an understanding of the neoclassical root transition., (© 2024 Author(s). Larger fusion experiments require long beam paths for laser diagnostics, which requires mechanical stability and measures to deal with remaining alignment variations. At the same time, due to technical and organizational boundary conditions, calibrations become challenging. The current mid-sized experiments face the same issues, yet on a smaller scale, which makes them ideal testing environments for novel calibration methods, since a comparison with the established best practices is still possible. At the stellarator Wendelstein 7-X, the calibration and operation of the Thomson scattering diagnostic is hampered by beam displacements, coating of windows during operation, and access restrictions while the superconducting coils are active. New calibration techniques were developed to improve the profile quality and reduce calibration time. While positional variations of the laser beam have to be minimized, the remaining displacements can be accounted for during the absolute calibration. An in situ spectral calibration has been developed based on Rayleigh scattering, which calibrates the whole diagnostic, including observation windows. In addition, a less accurate but faster method has been developed, which utilizes stray-light of a tunable OPO to perform spectral calibration within minutes and does not require torus hall access. Finally, a workflow has been established to consider finite linewidths of the calibration source in the spectral calibration. While these methods will be used at W7-X to complement existing calibration techniques, they may also solve some of the aforementioned issues expected for even larger and nuclear experiments, where access restrictions are stringent and calibration becomes even more demanding., (© 2024 Author(s). SUN domain proteins are conserved proteins of the nuclear envelope and key components of the LINC complexes (for 'linkers of the nucleoskeleton and the cytoskeleton'). Previous studies have demonstrated that the testis-specific SUN domain protein SUN4 (also known as SPAG4) is a vital player in the directed shaping of the spermatid nucleus. However, its molecular properties relating to this crucial function have remained largely unknown, and controversial data for the organization and orientation of SUN4 within the spermatid nuclear envelope have been presented so far. Here, we have re-evaluated this issue in detail and show robust evidence that SUN4 is integral to the inner nuclear membrane, sharing a classical SUN domain protein topology. The C-terminal SUN domain of SUN4 localizes to the perinuclear space, whereas the N-terminus is directed to the nucleoplasm, interacting with the spermiogenesis-specific lamin B3. We found that SUN4 forms heteromeric assemblies with SUN3 in vivo and regulates SUN3 expression. We assess the magnetic field configuration in modern fusion devices by comparing experiments with the same heating power, between a stellarator and a heliotron. The key role of turbulence is evident in the optimized stellarator, while neoclassical processes largely determine the transport in the heliotron device. Gyrokinetic simulations elucidate the underlying mechanisms promoting stronger ion scale turbulence in the stellarator. Similar plasma performances in these experiments suggests that neoclassical and turbulent transport should both be optimized in next step reactor designs.

Research on magnetic confinement of high-temperature plasmas has the ultimate goal of harnessing nuclear fusion for the production of electricity. Although the tokamak 1 is the leading toroidal magnetic-confinement concept, it is not without shortcomings and the fusion community has therefore also pursued alternative concepts such as the stellarator. Unlike axisymmetric tokamaks, stellarators possess a three-dimensional (3D) magnetic field geometry. The availability of this additional dimension opens up an extensive configuration space for computational optimization of both the field geometry itself and the current-carrying coils that produce it. Such an optimization was undertaken in designing Wendelstein 7-X (W7-X) 2 , a large helical-axis advanced stellarator (HELIAS), which began operation in 2015 at Greifswald, Germany. A major drawback of 3D magnetic field geometry, however, is that it introduces a strong temperature dependence into the stellarator's non-turbulent 'neoclassical' energy transport. Indeed, such energy losses will become prohibitive in high-temperature reactor plasmas unless a strong reduction of the geometrical factor associated with this transport can be achieved; such a reduction was therefore a principal goal of the design of W7-X. In spite of the modest heating power currently available, W7-X has already been able to achieve high-temperature plasma conditions during its 2017 and 2018 experimental campaigns, producing record values of the fusion triple product for such stellarator plasmas 3,4 . The triple product of plasma density, ion temperature and energy confinement time is used in fusion research as a figure of merit, as it must attain a certain threshold value before net-energy-producing operation of a reactor becomes possible 1,5 . Here we demonstrate that such record values provide evidence for reduced neoclassical energy transport in W7-X, as the plasma profiles that produced these results could not have been obtained in stellarators lacking a comparably high level of neoclassical optimization., (© 2021. In nuclear fusion research, the effective ion charge Z eff , which characterizes the overall content of impurities, can be experimentally derived from the plasma electron-ion bremsstrahlung, given the electron density n e and temperature T e . At Wendelstein 7-X, a multichannel near-infrared spectrometer is installed to collect the plasma bremsstrahlung along 27 lines of sight covering more than half the plasma cross section, which provides information on Z eff over the entire plasma radius. To infer spatially resolved Z eff profiles, a Bayesian model is developed in the Minerva framework. Z eff , n e , and T e profiles are modeled as Gaussian processes, whose smoothness is determined by hyperparameters. These profiles are transformed to fields in Cartesian coordinates, given the poloidal magnetic flux surfaces calculated by the variational moments equilibrium code. Given all these physical quantities, the model predicts line-of-sight integrals of near-infrared bremsstrahlung spectra. The model includes the predictive (forward) models of the interferometer, Thomson scattering system, and visible and near-infrared spectrometers. Given the observations of all these diagnostics, the posterior probability distribution of Z eff profiles is calculated and shown as an inference solution. The smoothness (gradient) of the profiles is optimally chosen by Bayesian Occam's razor. Furthermore, wall reflections can significantly pollute the measurements of the plasma bremsstrahlung, which leads to over-estimation of Z eff values in the edge region. In the first results presented in this work, this problem does not appear, and the posterior samples of Z eff profiles are overall plausible and consistent with Z eff values inferred, given the data from the single-channel visible spectrometer.

The Charge Exchange Recombination Spectroscopy (CXRS) diagnostic has become a routine diagnostic on almost all major high temperature fusion experimental devices. For the optimized stellarator Wendelstein 7-X (W7-X), a highly flexible and extensive CXRS diagnostic has been built to provide high-resolution local measurements of several important plasma parameters using the recently commissioned neutral beam heating. This paper outlines the design specifics of the W7-X CXRS system and gives examples of the initial results obtained, including typical ion temperature profiles for several common heating scenarios, toroidal flow and radial electric field derived from velocity measurements, beam attenuation via beam emission spectra, and normalized impurity density profiles under some typical plasma conditions.

For the first time, the optimized stellarator Wendelstein 7-X has operated with an island divertor. An operation regime in hydrogen was found in which the total plasma radiation approached the absorbed heating power without noticeable loss of stored energy. The divertor thermography recorded simultaneously a strong reduction of the heat load on all divertor targets, indicating almost complete power detachment. This operation regime was stably sustained over several energy confinement times until the preprogrammed end of the discharge. The plasma radiation is mainly due to oxygen and is located at the plasma edge. This plasma scenario is reproducible and robust at various heating powers, plasma densities, and gas fueling locations. These experimental results show that the island divertor concept actually works and displays good power dissipation potential, producing a promising exhaust concept for the stellarator reactor line.

At the Wendelstein 7-X stellarator, the X-ray imaging crystal spectrometer provides line integrated measurements of ion and electron temperatures, plasma flows, as well as impurity densities from a spectroscopic analysis of tracer impurity radiation. In order to infer the actual profiles from line integrated data, a forward modeling approach has been developed within the Minerva Bayesian analysis framework. In this framework, the inversion is realized on the basis of a complete forward model of the diagnostic, including error propagation and utilizing Gaussian processes for generation and inference of arbitrary shaped plasma parameter profiles. For modeling of line integrated data as measured by the detector, the installation geometry of the spectrometer, imaging properties of the crystal, and Gaussian detection noise are considered. The inversion of line integrated data is achieved using the maximum posterior method for plasma parameter profile inference and a Markov chain Monte Carlo sampling of the posterior distribution for calculating uncertainties of the inference process. The inversion method shows a correct and reliable inference of temperature and impurity density profiles from synthesized data within the estimated uncertainties along the whole plasma radius. The application to measured data yields a good match of derived electron temperature profiles to data of the Thomson scattering diagnostic for central electron temperatures between 2 and 5 keV using argon impurities.

This paper reports about a novel approach to the absolute intensity calibration of an electron cyclotron emission (ECE) spectroscopy system. Typically, an ECE radiometer consists of tens of separated frequency channels corresponding to different plasma locations. An absolute calibration of the overall diagnostic including near plasma optics and transmission line is achieved with blackbody sources at LN 2 temperature and room temperature via a hot/cold calibration mirror unit. As the thermal emission of the calibration source is typically a few thousand times lower than the receiver noise temperature, coherent averaging over several hours is required to get a sufficient signal to noise ratio. A forward model suitable for any radiometer calibration using the hot/cold method and a periodic switch between them has been developed and used to extract the voltage difference between the hot and cold temperature source via Bayesian analysis. In contrast to the classical analysis which evaluates only the reference temperatures, the forward model takes into account intermediate effective temperatures caused by the finite beam width and thus uses all available data optimally. This allows the evaluation of weak channels where a classical analysis would not be feasible, is statistically rigorous, and provides a measurement of the beam width. By using a variance scaling factor, a model sensitive adaptation of the absolute uncertainties can be implemented, which will be used for the combined diagnostic Bayesian modeling analysis.

A new method for in situ spectral calibration of Thomson scattering diagnostics is proposed. The idea of the method is to apply a wavelength tunable optical parametric oscillator for measurements of Rayleigh scattering at different wavelengths, from which relative sensitivities can be calculated. This extends the usual approach where Rayleigh scattering is used only at a single wavelength for the absolute calibration and spectral sensitivities are obtained separately. With the new approach, the full diagnostic setup is spectrally calibrated at once. Such a calibration can be repeated at regular intervals during an experimental campaign since it does not require a break of the vacuum. In this paper, the Rayleigh scattering calibration is tested in a laboratory setup with a sample Wendelstein 7-X (W7-X) polychromator. It is shown that relative sensitivities of spectral channels can be recovered with a sufficient resolution even under conditions of significant stray light. The stray light is overcome by measuring the linear dependence of the scattered signal on the gas pressure. Good results of laboratory tests motivate the installation of the new calibration system for the Thomson scattering diagnostic at W7-X.

Electron temperature gradient (ETG)-driven turbulence, despite its ultrafine scale, is thought to drive significant thermal losses in magnetic fusion devices-but what role does it play in stellarators? The first numerical simulations of ETG turbulence for the Wendelstein 7-X stellarator, together with power balance analysis from its initial experimental operation phase, suggest that the associated transport should be negligible compared to other channels. The effect, we argue, originates essentially from the geometric constraint of multiple field periods, a generic feature of stellarators.

This paper presents the approach of the dual-laser wavelength Thomson scattering (TS) system for the Wendelstein 7-X stellarator. The dual-laser wavelength TS method is based on two lasers with different wavelengths being fired quasi-simultaneously. This method has two advantages compared to a single laser wavelength TS system. First, the dual laser availability allows an in situ spectral calibration, and second, higher electron temperatures can be measured without any change in the spectral filter setup of the polychromators. The W7-X dual-laser wavelength TS concept is based on high power lasers: a set of standard Nd:YAG lasers with λ = 1064 nm wavelength and a Nd:YAG laser with λ = 1319 nm wavelength newly developed for this application. This laser uses a different transition line with 34% efficiency compared to the main 1064 nm Nd:YAG line. Simulations of the expected performance of the new dual-laser wavelength system show that electron temperatures up to T e = 15 keV can be measured compared to the original design parameter up to Te = 10 keV. The in situ spectral calibration can be performed using a range of temperatures from 1 keV to 10 keV using TS measurements of the 1064 nm versus 1319 nm TS simultaneously.

This paper reports on the design and the performance of the recently upgraded X-ray imaging spectrometer systems, X-ray imaging crystal spectrometer and high resolution X-ray imaging spectrometer, installed at the optimized stellarator Wendelstein 7-X. High resolution spectra of highly ionized, He-like Si, Ar, Ti, and Fe as well as H-like Ar have been observed. A cross comparison of ion and electron temperature profiles derived from a spectral fit and tomographic inversion of Ar and Fe spectra shows a reasonable match with both the spectrometers. The also measured impurity density profiles of Ar and Fe have peaked densities at radial positions that are in qualitative agreement with the expectations from the He-like impurity fractional abundances, given the measured temperature profiles. Repeated measurements of impurity decay times have been demonstrated with an accuracy of 1 ms via injection of non-recycling Ti, Fe, and Mo impurities using a laser blow-off system.

A passive phased array Doppler reflectometry system has recently been installed in the Wendelstein-7X stellarator. In contrast to conventional Doppler reflectometry systems, the microwave beam can be steered on short time scales in the measurement plane perpendicular to the magnetic field in the range of ±25° without mechanical steering components. This paper characterizes the design and properties of the phased array antenna system and presents the first measurement results from the latest OP1.2a campaign.

Wendelstein 7-X, a superconducting optimized stellarator built in Greifswald/Germany, started its first plasmas with the last closed flux surface (LCFS) defined by 5 uncooled graphite limiters in December 2015. At the end of the 10 weeks long experimental campaign (OP1.1) more than 20 independent diagnostic systems were in operation, allowing detailed studies of many interesting plasma phenomena. For example, fast neutral gas manometers supported by video cameras (including one fast-frame camera with frame rates of tens of kHz) as well as visible cameras with different interference filters, with field of views covering all ten half-modules of the stellarator, discovered a MARFE-like radiation zone on the inboard side of machine module 4. This structure is presumably triggered by an inadvertent plasma-wall interaction in module 4 resulting in a high impurity influx that terminates some discharges by radiation cooling. The main plasma parameters achieved in OP1.1 exceeded predicted values in discharges of a length reaching 6 s. Although OP1.1 is characterized by short pulses, many of the diagnostics are already designed for quasi-steady state operation of 30 min discharges heated at 10 MW of ECRH. An overview of diagnostic performance for OP1.1 is given, including some highlights from the physics campaigns.

This paper describes the design of the Thomson scattering system at the Wendelstein 7-X stellarator. For the first operation campaign we installed a 10 spatial channel system to cover a radial half profile of the plasma cross section. The start-up system is based on one Nd:YAG laser with 10 Hz repetition frequency, one observation optics, five fiber bundles with one delay line each, and five interference filter polychromators with five spectral channels and silicon avalanche diodes as detectors. High dynamic range analog to digital converters with 14 bit, 1 GS/s are used to digitize the signals. The spectral calibration of the system was done using a pulsed super continuum laser together with a monochromator. For density calibration we used Raman scattering in nitrogen gas. Peaked temperature profiles and flat density profiles are observed in helium and hydrogen discharges.

LINC complexes are evolutionarily conserved nuclear envelope bridges, physically connecting the nucleus to the peripheral cytoskeleton. They are pivotal for dynamic cellular and developmental processes, like nuclear migration, anchoring and positioning, meiotic chromosome movements and maintenance of cell polarity and nuclear shape. Active nuclear reshaping is a hallmark of mammalian sperm development and, by transducing cytoskeletal forces to the nuclear envelope, LINC complexes could be vital for sperm head formation as well. We here analyzed in detail the behavior and function of Sun4, a bona fide testis-specific LINC component. We demonstrate that Sun4 is solely expressed in spermatids and there localizes to the posterior nuclear envelope, likely interacting with Sun3/Nesprin1 LINC components. Our study revealed that Sun4 deficiency severely impacts the nucleocytoplasmic junction, leads to mislocalization of other LINC components and interferes with the formation of the microtubule manchette, which finally culminates in a globozoospermia-like phenotype. An overview of the diagnostics which are essential for the first operational phase of Wendelstein 7-X and the set of diagnostics expected to be ready for operation at this time are presented. The ongoing investigations of how to cope with high levels of stray Electron Cyclotron Resonance Heating (ECRH) radiation in the ultraviolet (UV)/visible/infrared (IR) optical diagnostics are described.

The critical issues in the development of diagnostics, which need to work robust and reliable under quasi-steady state conditions for the discharge durations of 30 min and which cannot be maintained throughout the one week duration of each operation phase of the Wendelstein 7-X stellarator, are being discussed.

Calcium/calmodulin-dependent protein kinase II (CaMKII) has been linked to neuronal plasticity associated with long-term potentiation as well as structural synaptic plasticity. Previous work in adult honeybees has shown that a single CaMKII gene is strongly expressed in the mushroom bodies (MBs), brain centers associated with sensory integration, and learning and memory formation. To study a potential role of CaMKII in synaptic plasticity, the cellular and subcellular distribution of activated (phosphorylated) pCaMKII protein was investigated at various life stages of the honeybee using immunocytochemistry, confocal microscopy, and western blot analyses. Whereas at pupal stages 3-4 most parts of the brain showed high levels of pCaMKII immunoreactivity, the protein was predominantly concentrated in the MBs in the adult brain. The results show that pCaMKII is present in a specific subpopulation of Kenyon cells, the noncompact cells. Within the olfactory (lip) and visual (collar) subregion of the MB calyx neuropil pCaMKII was colocalized with f-actin in postsynaptic compartments of microglomeruli, indicating that it is enriched in Kenyon cell dendritic spines. This suggests a potential role of CaMKII in Kenyon cell dendritic plasticity. Interestingly, pCaMKII protein was absent in two other types of Kenyon cells, the inner compact cells associated with the multimodal basal ring and the outer compact cells. During adult behavioral maturation from nurse bees to foragers, pCaMKII distribution remained essentially similar at the qualitative level, suggesting a potential role in dendritic plasticity of Kenyon cells throughout the entire life span of a worker bee., (Copyright © 2011 Wiley-Liss, Inc.)

The status of the diagnostic developments for the quasistationary operable stellarator Wendelstein 7-X (maximum pulse length of 30 min at 10 MW ECRH heating at 140 GHz) will be reported on. Significant emphasis is being given to the issue of ECRH stray radiation shielding of in-vessel diagnostic components, which will be critical at high density operation requiring O2 and OXB heating.

The stellarator Wendelstein 7-X will allow for quasicontinuous operation with the duration only being limited to two 30 min discharges per day, at a continuous heating power of 10 MW electron cyclotron resonance heating (ECRH) at 140 GHz, by the capacity of the cooling water reservoir. This will result in high thermal loads on all plasma facing components of 50-100 kW/m(2) from radiation alone and of up to about 500 kW/m(2) on components additionally exposed to convective loads. In high density scenarios toroidally varying ECRH stray radiation levels of 50-200 kW/m(2) need to be coped with, requiring careful material selection and different shielding and hardening techniques. Furthermore, a gradual buildup of coatings on plasma facing optical components, which without any measures being taken, would lead to high transmission losses already within a few days of long pulse operation (equivalent to about 1 year of operation in pulsed devices like JET or ASDEX-upgrade) and therefore needs to be prevented as much as possible. In addition in situ cleaning as well as absolute calibration techniques need to be developed for all plasma facing optical systems. Here we report about some of our efforts to find, for various types of diagnostics, ways to cope with these adverse effects. Moreover, we give a few examples for individual diagnostic specific issues with respect to quasicontinuous operation, such as the development of a special integrator for the magnetic diagnostics as well as special interferometer types which can cope with unavoidable vibrations and slow path length changes due to, e.g., thermal expansion of the plasma vessel.

A promising new plasma operational regime on the Wendelstein stellarator W7-AS has been discovered. It is extant above a threshold density and characterized by flat density profiles, high energy and low impurity confinement times, and edge-localized radiation. Impurity accumulation is avoided. Quasistationary discharges with line-averaged densities n(e) to 4 x 10(20) m(-3), radiation levels to 90%, and partial plasma detachment at the divertor target plates can be simultaneously realized. Energy confinement is up to twice that of a standard scaling. At B(t) = 0.9 T, an average beta value of 3.1% is achieved. The high n(e) values allow demonstration of electron Bernstein wave heating using linear mode conversion.

The plasma rotation in a linear magnetic configuration has been investigated by means of high-resolution Doppler-spectroscopy. The effects of external device parameters and ion mass as well as the radial profiles have been studied. A simple model, based on the conservation of the total angular momentum, explains the main empirically determined dependences of the average angular velocity. The results are compared with the common multifluid description developed for hollow cathode discharges.
