The pelagic cephalopod species jumbo flying squid Dosidicus gigas is ecologically and economically important in the Humboldt ecosystem off Peru. This squid species is sensitive to oceanic environmental changes, and regional oceanographical variability is one of the important factors driving its redistribution. Off Peruvian waters, mesoscale eddies are ubiquitous and dominate the biogeochemical processes in this region. This study first explored the role of mesoscale eddies in regulating the environments and their effects on the abundance and habitat distribution of D. gigas off Peru by analyzing squid distribution in eddy-centric coordinates and building a habitat suitability index (HSI) model. Results indicated that the abundance and habitat distribution of D. gigas in mesoscale eddies varied across months, with significant differences observed between anticyclonic eddies (AE) and cyclonic eddies (CE). In AE, a higher abundance and proportion of suitable habitat occurred. While in CE, the abundance was relatively low and the suitable habitat was relatively less, concentrating at the periphery of CE. Based on the HSI model results, sea surface temperature (SST) and 50 m water temperature (T50m) in AE were more favorable for D. gigas, which was 0.3-0.5 °C lower than that in CE, yielding high-quality habitats and higher abundance of D. gigas. Dosidicus gigas (D. gigas) is a pelagic cephalopod of ecological and economic importance widely distributed in the eastern Pacific Ocean. Generally, small-, medium-, and large-sized groups of the squids have been respectively identified on the basis of the mantle length (ML) of adults. Intraspecific feeding variability maximizes the utilization of available food resources by D. gigas. However, the coexistence mechanism of three groups has not been fully understood yet. In our study, based on the analyses of beak morphology and stable carbon and nitrogen isotopes, the feeding strategies and coexistence patterns of large-, medium-, and small-sized groups of D. gigas were investigated. D. gigas had a wide range of 13 C/ 12 C (δ 13 C) and 15 N/ 14 N (δ 15 N) values in muscle tissue, variable feeding behaviors, and wide food sources. The δ 13 C or δ 15 N values showed no significant difference between the small- and medium-sized groups, which shared the same habitats and fed on preys with the similar trophic level. Compared to small- and medium-sized groups, the large-sized group had a smaller range of habitats and consumed more nearshore foods. Both isotopes and feeding apparatus morphology indicated a high degree of niche overlap between the small- and medium-sized groups, whereas the large-sized group differed significantly from other groups. In addition, the niche width of the female was larger than that of the male in all three groups. We inferred that the sex differences in body length and reproductive behavior led to the difference in niche width. The isotopic niche overlap between female and male samples was the most significant in large-sized group and the least significant in the small-sized group, indicating that different feeding strategies were adopted by the three groups. These findings proved that the three groups of D. gigas off Peruvian waters adopted a feeding strategy with inter- and intra-group regulation. Jumbo squid (Dosidicus gigas) is a commercially valuable mollusk in Mexico; 80% of its body is edible. Despite the high protein content (∼18%) and low cholesterol content of this species, its high proteolytic activity (microbial and endogenous enzymes) induces protein degradation and consequent reduction in functional properties from a structural viewpoint. Gelation capacity (texture profile of the gels obtained), solubility, water holding capacity, foaming capacity, emulsification capacity, and emulsion stability were evaluated in protein concentrates obtained by foam-aided pH-shift processing: (A) myofibrillar protein extraction with distilled water and no pH-shifting; (B) alkaline solubilization and isoelectric precipitation; (C) acidic solubilization and isoelectric precipitation; and (D) process A and isoelectric precipitation. Process B showed superior gelation capacity, D had high emulsion stability across a wide range of pH values (4.0-8.0) and C lower plate counts of aerobic mesophilic. Therefore, all three alternative extraction processes showed techno-functional advantages. PRACTICAL APPLICATION: Jumbo squid is an abundant protein source in México, most of which is exported. Functional and physicochemical properties of muscle protein were improved by pH-shift processing. The recovered protein showed modifications of technological properties, using one of the methods described can lead to produce a protein extract with the most desirable attributes, such as foaming, emulsifying, or gelling capacities. The functional and physicochemical properties of protein from squid can be enhanced by selecting a certain pH-shift processing, depending on the desirable use. Directed formation of the structure of the culture of living cells is the most important task of tissue engineering. New materials for 3D scaffolds of living tissue are critical for the mass adoption of regenerative medicine protocols. In this manuscript, we demonstrate the results of the molecular structure study of collagen from Dosidicus gigas and reveal the possibility of obtaining a thin membrane material. The collagen membrane is characterized by high flexibility and plasticity as well as mechanical strength. The technology of obtaining collagen scaffolds, as well as the results of studies of its mechanical properties, surface morphology, protein composition, and the process of cell proliferation on its surface, are shown in the given manuscript. The investigation of living tissue culture grown on the surface of a collagen scaffold by X-ray tomography on a synchrotron source made it possible to remodel the structure of the extracellular matrix. It was found that the scaffolds obtained from squid collagen are characterized by a high degree of fibril ordering and high surface roughness and provide efficient directed growth of the cell culture. The resulting material provides the formation of the extracellular matrix and is characterized by a short time to living tissue sorption.

Microplastic (MP) ingestion by marine animals has been well documented, but less being known about pelagic squid. Jumbo squid Dosidicus gigas supports the world's largest cephalopod fishery and plays an important ecological role in the Eastern Pacific Ocean. In this study, D. gigas taken from the open sea of the Peruvian Exclusive Economic Zone were selected as research objects. We estimated the abundance and characteristics of MPs in the stomach and intestine of D. gigas and investigated the differences between tissues and sexes. Similar abundance and characteristics of MPs were observed in the same tissue of females and males. However, the stomach had a higher abundance of MPs with larger size than the intestine, while the MP abundance by stomach wet weight was lower than that of the intestine. The MPs were predominantly fiber-shaped, with blue or black color. The most frequent polymers were high-density cellophane and polyacrylic acid. These polymers could sink into deeper sea layers and were available for D. gigas living there during the daytime. Our findings revealed the distribution pattern of MPs in the waters of the Peruvian fishing ground. This study could improve our understanding of the MP contamination level in pelagic squid, and have implications for evaluating the ecological effects of MP on cephalopods.

The growing applications of tissue engineering technologies warrant the search and development of biocompatible materials with an appropriate strength and elastic moduli. Here, we have extensively studied a collagenous membrane (GSCM) separated from the mantle of the Giant squid Dosidicus Gigas in order to test its potential applicability in regenerative medicine. To establish the composition and structure of the studied material, we analyzed the GSCM by a variety of techniques, including amino acid analysis, SDS-PAGE, and FTIR. It has been shown that collagen is a main component of the GSCM. The morphology study by different microscopic techniques from nano- to microscale revealed a peculiar packing of collagen fibers forming laminae oriented at 60-90 degrees in respect to each other, which, in turn, formed layers with the thickness of several microns (a basketweave motif). The macro- and micromechanical studies showed high values of the Young's modulus and tensile strength. No significant cytotoxicity of the studied material was found by the cytotoxicity assay. Thus, the GSCM consists of a reinforced collagen network, has high mechanical characteristics, and is non-toxic, which makes it a good candidate for the creation of a scaffold material for tissue engineering.

Microplastics (MPs) found in marine invertebrates have aroused great concern, but MP ingestion by cephalopods is rare. To evaluate MP contamination in commercially important pelagic squids, we examined the abundance and characteristics of MPs in the gill, intestine, and stomach of jumbo squid Dosidicus gigas (30.9 to 65.0 cm mantle length), collected from the northern Humboldt Current. The average abundance ranged from 4.0 to 7.4 items/individual and 0.2 to 0.7 items/g wet weight for the three tissues. The MPs were sized 80.75 to 4632.27 μm, with larger MPs generally found in the stomach. The majority of MPs were fibrous in shape, blue or black-gray in color, and cellophane in composition. These results revealed the MP distribution in D. gigas and could be driven by its movement pattern and habitat use. AbstractThe limitations that hypoxia imparts on mitochondrial oxygen supply are circumvented by the activation of anaerobic metabolism and prosurvival mechanisms in hypoxia-tolerant animals. To deal with the hypoxia that jumbo squid ( Dosidicus gigas ) experience in the ocean's depth, they depress their metabolic rate by up to 52% relative to normoxic conditions. This is coupled with molecular reorganization to facilitate their daily descents into the ocean's oxygen minimum zone, where they face not only low oxygen levels but also higher pressures and colder frigid waters. Our current study explores the tissue-specific hypoxia responses of three central processes: (1) antioxidant enzymes responsible for defending against oxidative stress, (2) early apoptotic machinery that signals the activation of cell death, and (3) mitogen-activated protein kinases (MAPKs) that act as central regulators of numerous cellular processes. Luminex xMAP technology was used to assess protein levels and phosphorylation states under normoxic and hypoxic conditions in brains, branchial hearts, and mantle muscles. Hypoxic brains were found to activate apoptosis via upregulation of phospho-p38, phospho-p53, activated caspase 8, and activated caspase 9, whereas branchial hearts were the only tissue to show an increase in antioxidant enzyme levels. Hypoxic muscles seemed the least affected by hypoxia. Our results suggest that hypoxic squid do not undergo large dynamic changes in the phosphorylation states of key apoptotic and central MAPK factors, except for brains, suggesting that these mechanisms are involved in squid hypometabolic responses.

Understanding the adaptation of important marine species to environmental changes is critical for evaluating the effects of fisheries and climate change on marine services. The jumbo squid, Dosidicus gigas , is a keystone species in the eastern Pacific, which plays an intermediate role in the marine food web. Better understanding of their responses to climate change would be a big step to understand their population dynamics. In this study, stable isotope and high-throughput 16S rRNA sequencing were used to compare the variation of trophic niche, gut length, and gut microbiota of D. gigas in the eastern equatorial water during normal and El Niño periods. The results showed a significant variation in δ 13 C values for D. gigas in different periods, indicating differences in their food sources. The main phylum-level gut microbiome included Actinobacteria, Bacteroidetes, Cyanobacteria, Firmicutes, Proteobacteria, and Tenericutes. There was no significant difference in the gut microbial composition during normal and El Niño periods, but differences in gut microbial diversity and relative abundance of some phyla bacteria. El Niño events could decrease the trophic niche breadth of D. gigas , and positively impact gut length and gut microbial diversity and richness. Firmicutes, Bacteroidetes, Spirochaetes, WPS-2, and Kiritimatiellaeota had a significant increase in the gut microbiota of D. gigas combined with a more concentrated intraspecific rank of distance during El Niño, suggesting that the changes of habitat and food sources caused by El Niño events could limit the distribution range of D. gigas . D. gigas might change their digestive system to improve the digestive and absorption capacity and enhance their immunocompetence. Such a climate-driven alteration might help D. gigas rapidly adapt to the changes of environmental conditions and food resources under El Niño.

Background: Protein glycation by Maillard reaction is commonly used to improve the functional and bioactive properties of food proteins. It is also known that this glycation method can be accelerated by heat without the need for chemical reagents that could be harmful to health. In this study, glycoconjugates were obtained from a mixture of connective tissue proteins (CTP) from jumbo squid (Dosidicus gigas) and two different sugars, dextran (DEX; 10 kDa) and glucose (GLU), using protein-to-carbohydrate ratios of 1:2 and 1:3, in solution at 50 °C for 6 h. The glycation products were characterized by means of their physicochemical properties and cytotoxic effect., Results: The intensity of the browning measured at A 420nm and A 294nm in glycoconjugates showed no significant difference (P < 0.05). CTP-DEX (1:2) and CTP-DEX (1:3) were those products with the greatest fluorescence related to the intermediate stage in the Maillard reaction, and also with the highest degree of glycation, which was confirmed using o-phthaldialdehyde assay and Fourier transform infrared analysis. The values of cellular viability for CTP-GLU (1:3), CTP-DEX (1:2, 1:3) as well as CTP (0, 6 h) were around 92-103%., Conclusions: The operational parameters used in the glycation process achieved the formation of glycoconjugates from proteins of D. gigas, showing no cytotoxic effect on the HaCaT cell line. Jumbo squid ( Dosidicus gigas ) is one of the largest cephalopods, and represents an important economic fishery in several regions of the Pacific Ocean, from southern California in the United States to southern Chile. Large and considerable discards of this species, such as skin, have been reported to constitute an important source of potential by-products. In this paper, a shotgun proteomics approach was applied for the first time to the characterization of the jumbo squid ( Dosidicus gigas ) skin proteome. A total of 1004 different peptides belonging to 219 different proteins were identified. The final proteome compilation was investigated by integrated in-silico studies, including gene ontology (GO) term enrichment, pathways, and networks studies. Potential new valuable bioactive peptides such as antimicrobial, bioactive collagen peptides, antihypertensive and antitumoral peptides were predicted to be present in the jumbo squid skin proteome. The integration of the global proteomics results and the bioinformatics analysis of the jumbo squid skin proteome show a comprehensive knowledge of this fishery discard and provide potential bioactive peptides of this marine by-product.

Background: The secondary structure of a protein determines its functional properties, such as its gelling capacity. The α-helix and β-sheet comprise its main structures. Myofibrillar proteins from jumbo squid are composed mainly of the actomyosin-paramyosin complex; this complex contains a high percentage of α-helix, because actin, paramyosin, and myosin constitute 30%, 100%, and 55% of the α-helix, respectively. It is important to elucidate the role of the secondary structures in the gelation of giant squid proteins as they form gel. The role of the secondary structures in the gelation of giant squid proteins is therefore very important. For this reason, the objective of this work was to evaluate the effect of temperature on the structural behavior of actomyosin-paramyosin isolate (API) from Dosidicus gigas., Results: The unfolding of the API system, which is composed of the actomyosin-paramyosin complex, was clarified by studying surface hydrophobicity and viscosity. Three characteristic peaks were found, associated with myosin, paramyosin, and actin. Infrared and circular dichroism corroborated the view that API undergoes major structural changes, because it proceeds from mostly an α-helix structure to 100% β-sheet., Conclusion: The structural rearrangement favors gelation by cross-linking, generating new protein-protein and water-protein interactions, which create a more stable structure compared to mantle proteins (MP). Likewise, the presence of sarcoplasmic and stromal proteins in D. gigas muscle prevents the unfolding of myofibrillar proteins, favoring gelation by agglomeration, decreasing the ability to trap water and thus its gelling capacity. © 2019 Society of Chemical Industry., (© 2019 Society of Chemical Industry.)

Background: The giant squid (Dosidicus gigas) has been proposed as raw material to obtain myofibrillar protein concentrates. However, it has been observed that colloidal systems formed from squid proteins have limited stability. Therefore, the isolation and characterization of the actomyosin-paramyosin isolated (API) complex were performed, because they are the main proteins to which functionality has been attributed., Results: Densitogram analysis revealed 45% of actin, 38% of myosin and 17% of paramyosin. The amino acid profile indicates a higher proportion of acidic amino acids, which gives a higher negative charge; this was supported by the zeta potential. Total sulfhydryl (TSH) content was lower compared with proteins of other aquatic species., Conclusion: The higher percentage of actin in relation to myosin, the presence of paramyosin, as well as the low content of sulfhydryl groups, could comprise the main causes of the low technological functional property of proteins from D. gigas mantle. © 2017 Society of Chemical Industry., (© 2017 Society of Chemical Industry.)

Background: An advanced strategy for chilled fish preservation, based on the inclusion in ice of an extract of jumbo squid (Dosidicus gigas) skin (JSS), is proposed. Aqueous solutions including acetic acid-ethanol extracts of JSS were tested at two different concentrations as icing media, with the effects on the quality evolution of chilled hake (Merluccius merluccius) being monitored., Results: A significant inhibition (P < 0.05) of microbial activity (aerobes, psychrotrophs, Enterobacteriaceae, proteolytic bacteria; pH, trimethylamine) was obtained in hake corresponding to the icing batch including the highest JSS concentration. Additionally, fish specimens from such icing conditions showed an inhibitory effect (P < 0.05) on lipid hydrolysis development, while no effect (P > 0.05) was depicted for lipid oxidation. Sensory analysis (skin and mucus development; eyes; gills; texture; external odour; raw and cooked flesh odour; flesh taste) indicated a shelf life extension of chilled hake stored in ice including the highest JSS concentration., Conclusion: A profitable use of JSS, an industrial by-product during jumbo squid commercialisation, has been developed in the present work, which leads to a remarkable microbial inhibition and a significant shelf life extension of chilled hake. Squids are active and opportunistic predators that feed on a wide range of prey. Their active movements in the water column and their voracity promote a high consumption of food. In the pelagic environment off Ecuador, marine pollution is characterized by plastic debris with a mainland origin, including plastics trash of fishing gears. The objective of this work was to describe the presence of plastic remains in the stomach contents of Dosidicus gigas caught off the coast of Ecuador. Results demonstrated that 12% of the stomachs contained plastic remains. These plastics were identified as multifilament of polyethylene lines and polyvinyl chloride remains. Findings of this work could be related to an increase in the discharge of solid materials in the water column, increasing the probability to be ingested by the jumbo squid., (Copyright © 2016 Elsevier Ltd. Chymotrypsin was purified from jumbo squid hepatopancreas (HP) with 2.4-fold and yield 1.9%, and characterized with a molecular weight of 31 kDa, as estimated by sodium dodecyl sulfatepolyacrylamide gel electrophoresis (SDS-PAGE). Chymotrypsin effect over collagen extracted from the mantle, fins and arms of the jumbo squid was evaluated. The enzyme exhibited the maximum activity at pH 7 and 65°C using Suc-Ala-Ala-Pro-Phe-p-nitroanilide (SAAPNA) as a substrate and it was identified using the specific inhibitors N-tosyl-L-phenylalaninechloromethyl ketone (TPCK) and phenyl methyl sulfonyl fluoride (PMSF), showing residual activities of 6% and 0%, respectively. Furthermore, high activity was observed in the pH range of 4.0 to 8.0. Purified enzyme showed a moderate in vitro activity using muscle collagen as a substrate. Although further research is needed, the results suggest that the enzyme has a potential application where acidic or slightly alkaline conditions are needed.

Squid are the largest jet propellers in nature as adults, but as paralarvae they are some of the smallest, faced with the inherent inefficiency of jet propulsion at a low Reynolds number. In this study we describe the behavior and kinematics of locomotion in 1 mm paralarvae of Dosidicus gigas, the smallest squid yet studied. They swim with hop-and-sink behavior and can engage in fast jets by reducing the size of the mantle aperture during the contraction phase of a jetting cycle. We go on to explore the general effects of a variable mantle and funnel aperture in a theoretical model of jet propulsion scaled from the smallest (1 mm mantle length) to the largest (3 m) squid. Aperture reduction during mantle contraction increases propulsive efficiency at all squid sizes, although 1 mm squid still suffer from low efficiency (20%) because of a limited speed of contraction. Efficiency increases to a peak of 40% for 1 cm squid, then slowly declines. Squid larger than 6 cm must either reduce contraction speed or increase aperture size to maintain stress within maximal muscle tolerance. Ecological pressure to maintain maximum velocity may lead them to increase aperture size, which reduces efficiency. This effect might be ameliorated by nonaxial flow during the refill phase of the cycle. Our model's predictions highlight areas for future empirical work, and emphasize the existence of complex behavioral options for maximizing efficiency at both very small and large sizes.

Jumbo (or Humboldt) squid, Dosidicus gigas, is a large jet-propelled top oceanic predator off the Eastern Pacific. The present study reports, for the first time, concentrations of V, Cr, Mn, Co, Ni, Cu, Zn, As, Se, Cd and Pb in gills, mantle and digestive gland of this powerful pelagic squid in the Gulf of California. Zinc and Cu were the most abundant elements. All elements, with the exception of As, were largely stored in digestive gland; particularly Cd that reached concentrations between 57 and 509 µg g(-1). Significant relationships between tissues were found for Co (digestive gland-gills), As (gills-mantle) and Cd (digestive gland-mantle). Proportionality of Cd concentrations between mantle and digestive gland suggested that detoxification capacity by digestive gland was insufficient to avoid the transfer of this element to mantle and other tissues. Nonetheless, Cd concentrations in the mantle were always below the regulatory limit and, therefore lack of constraints for human consumption.
