Humans have a unique ability to use language for social communication. The neural architecture for language comprehension and production may have prominently emerged in the brain areas that were originally involved in social cognition. Here, we directly tested the fundamental link between language and social processing using functional magnetic resonance data (MRI) data from over 1,000 human subjects. Cortical activations in language and social tasks showed a striking similarity with a complementary hemispheric lateralization. Within core language areas, left-lateralized activations in the language task were mirrored by right-lateralized activations in the social task. Outside these areas, the activations were left lateralized in both tasks, perhaps indicating multimodal integration of social and semantic information. Structural connections between cortical areas form an intricate network with a high degree of specificity. Many aspects of this complex network organization in the adult mammalian cortex are captured by an architectonic type principle, which relates structural connections to the architectonic differentiation of brain regions. In particular, the laminar patterns of projection origins are a prominent feature of structural connections that varies in a graded manner with the relative architectonic differentiation of connected areas in the adult brain. Here we show that the architectonic type principle is already apparent for the laminar origins of cortico-cortical projections in the immature cortex of the macaque monkey. We find that prenatal and neonatal laminar patterns correlate with cortical architectonic differentiation, and that the relation of laminar patterns to architectonic differences between connected areas is not substantially altered by the complete loss of visual input. Moreover, we find that the degree of change in laminar patterns that projections undergo during development varies in proportion to the relative architectonic differentiation of the connected areas. Hence, it appears that initial biases in laminar projection patterns become progressively strengthened by later developmental processes. These findings suggest that early neurogenetic processes during the formation of the brain are sufficient to establish the characteristic laminar projection patterns. This conclusion is in line with previously suggested mechanistic explanations underlying the emergence of the architectonic type principle and provides further constraints for exploring the fundamental factors that shape structural connectivity in the mammalian brain.

Fragile X syndrome (FXS) is a neurodevelopmental disorder caused by silencing of the FMR1 gene and subsequent loss of its protein product, fragile X retardation protein (FMRP). One of the most robust neuropathological findings in post-mortem human FXS and Fmr1 KO mice is the abnormal increase in dendritic spine densities, with the majority of spines showing an elongated immature morphology. However, the exact mechanisms of how FMRP can regulate dendritic spine development are still unclear. Abnormal dendritic spines can result from disturbances of multiple factors during neurodevelopment, such as alterations in neuron numbers, position and glial cells. In this study, we undertook a comprehensive histological analysis of the cerebral cortex in Fmr1 KO mice. They displayed significantly fewer neuron and PV-interneuron numbers, along with altered cortical lamination patterns. In terms of glial cells, Fmr1 KO mice exhibited an increase in Olig2-oligodendrocytes, which corresponded to the abnormally higher myelin expression in the corpus callosum. Iba1-microglia were significantly reduced but GFAP-astrocyte numbers and intensity were elevated. Using primary astrocytes derived from KO mice, we further demonstrated the presence of astrogliosis characterized by an increase in GFAP expression and astrocyte hypertrophy. Our findings provide important information on the cortical architecture of Fmr1 KO mice, and insights towards possible mechanisms associated with FXS.

Navigation engages many cortical areas, including visual, parietal, and retrosplenial cortices. These regions have been mapped anatomically and with sensory stimuli and studied individually during behavior. Here, we investigated how behaviorally driven neural activity is distributed and combined across these regions. We performed dense sampling of single-neuron activity across the mouse posterior cortex and developed unbiased methods to relate neural activity to behavior and anatomical space. Most parts of the posterior cortex encoded most behavior-related features. However, the relative strength with which features were encoded varied across space. Therefore, the posterior cortex could be divided into discriminable areas based solely on behaviorally relevant neural activity, revealing functional structure in association regions. Multimodal representations combining sensory and movement signals were strongest in posterior parietal cortex, where gradients of single-feature representations spatially overlapped. Comparison and integration of neuroimaging data from different brains and populations are fundamental in neuroscience. Over the past decades, the neuroimaging field has largely depended on image registration to compare and integrate neuroimaging data from individuals in a common reference space, with a basic assumption that the brains are similar. However, the intrinsic neuroanatomical complexity and huge interindividual cortical folding variation remain underexplored. Here we focus on a specific cortical convolution pattern, termed 3-hinge gyral folding, which is the conjunction of gyri from multiple orientations and has unique and consistent anatomically, structurally, and functionally connective patterns across subjects. By developing a novel shape descriptor and a two-stage clustering pipeline, we devise an automatic method to identify 3-hinges in the Human Connectome Project Q3 868 human brains, and further parameterize the complexity of such a pattern and quantify its regularity and variation in terms of 3-hinge number, position, and morphology. Our results not only exhibit the huge interindividual variations, but also reveal regular relationship between gyral hinges and other factors, such as their locations and cortical morphologies. It is found that "line-shape" cortices have relatively more consistent 3-hinge shape pattern distributions, and certain types of 3-hinge patterns favor particular cortical morphologies. In addition, more 3-hinges are found on "line-shape" cortices while their numbers vary more across subjects than those on "non-line-shape" cortices. In the last few years, several studies have revisited long-held assumptions in the field of brain development and evolution providing us with a fundamentally new vision on the mechanisms controlling its size and shape, hence function. Among these studies, some described hitherto unforeseeable subtypes of neural progenitors while others reinterpreted long-known observations about their cell cycle in alternative new ways. Most remarkably, this knowledge combined has allowed the generation of mammalian model organisms in which brain size and folding has been selectively increased giving us the means to understand the mechanisms underlying the evolution of the most complex and sophisticated organ.
