Fluctuation-based super-resolution microscopy enhances image resolution using signal fluctuations, yet the inherent fluctuations of fluorophores limit its spatiotemporal resolution. In this work, we reveal a far-field enhancement (FFE) effect via a nanosilver film that significantly boosts fluorescence fluctuations of fluorophores positioned up to 10 μm away. The FFE effect arises from the interference of scattered light from the nanosilver film and photothermal-induced refractive index changes in the imaging medium, which create periodic auxiliary illumination on the sample. This phenomenon enabled the development of far-field enhanced super-resolution microscopy (FFE-SRM), a technique compatible with commonly used fluorophores. FFE-SRM improves temporal resolution up to 10-fold and enhances spatial resolution by about 2-fold over various SRM methods, including stochastic optical fluctuation imaging, super-resolution radial fluctuation, mean-shift super-resolution, and direct stochastic optical reconstruction microscopy. We demonstrated the potential of FFE-SRM by revealing mitochondrial dynamics in live-cell imaging, advancing super-resolution imaging, and cellular process exploration.

The growth and development of maize (Zea mays L.) are significantly impeded by prolonged exposure to high temperatures. Heat stress transcription factors (HSFs) play crucial roles in enabling plants to detect and respond to elevated temperatures. However, the genetic mechanisms underlying the responses of HSFs to heat stress in maize remain unclear. Thus, we aimed to investigate the role of ZmHSFA2B in regulating heat tolerance in maize. Here, we report that ZmHSFA2B has two splicing variants, ZmHSFA2B-I and ZmHSFA2B-II. ZmHSFA2B-I encodes full-length ZmHSFA2B (ZmHSFA2B-I), whereas ZmHSFA2B-II encodes a truncated ZmHSFA2B (ZmHSFA2B-II). Overexpression of ZmHSFA2B-I improved heat tolerance in maize and Arabidopsis thaliana, but it also resulted in growth retardation as a side effect. RNA-sequencing and CUT&Tag analyses identified ZmMBR1 as a putative target of ZmHSFA2B-I. Overexpression of ZmMBR1 also enhanced heat tolerance in Arabidopsis. ZmHSFA2B-II was primarily synthesized in response to heat stress and competitively interacted with ZmHSFA2B-I. This interaction consequently reduced the DNA-binding activities of ZmHSFA2B-I homodimers to the promoter of ZmMBR1. Subsequent investigations indicate that ZmHSFA2B-II limits the transactivation and tempers the function of ZmHSFA2B-I, thereby reducing the adverse effects of excessive ZmHSFA2B-I accumulation. Based on these observations, we propose that the alternative splicing of ZmHSFA2B generates a self-regulatory loop that fine-tunes heat stress response in maize., (© 2024 The Author(s). High-entropy materials (HEMs) are extremely popular for electrochemical energy storage nowadays. However, the detailed effects of four core factors of high entropy on the electrochemical properties of HEMs are still unclear. Here, a high-entropy La 1/4 Ce 1/4 Pr 1/4 Nd 1/4 Nb 3 O 9 (HE-LaNb 3 O 9 ) oxide is prepared through multiple rare-metal-ion substitution in LaNb 3 O 9 , and uses HE-LaNb 3 O 9 as a model material to systematically study the effects of the four core factors of high entropy on electrochemical energy-storage materials. The high-entropy effect lowers the calcination temperature for obtaining pure HE-LaNb 3 O 9 . The lattice distortion in HE-LaNb 3 O 9 leads to its decreased unit-cell-volume variations, which benefits its cyclability. Based on the restrained diffusion arising from the lattice distortion, the Li + diffusivity of HE-LaNb 3 O 9 at room temperature (25 °C) is limited, which causes its lowered rate capability. However, the Li + diffusivity of HE-LaNb 3 O 9 at high temperature (60 °C) becomes faster than that of LaNb 3 O 9 , which is attributed to the alleviated lattice distortion at the high-temperature, resulting in higher rate capability. The cocktail effects in HE-LaNb 3 O 9 enable its larger electronic conductivity, better electrochemical activity, more intensive Nb 5+ ↔ Nb 3+ redox reaction, and larger reversible capacity. Value-based decision-making relies on the striatum, where neural plasticity can be altered by chronic ethanol (EtOH) exposure, but the effects of such plasticity on striatal neural dynamics during decision-making remain unclear. This study investigated the long-term impacts of EtOH on reward-driven decision-making and striatal neurocomputations in male and female rats using a dynamic probabilistic reversal learning task. Following a prolonged withdrawal period, EtOH-exposed male rats exhibited deficits in adaptability and exploratory behavior, with a preference for value updating based on rewards rather than omissions. These behavioral changes were linked to altered neural encoding in the dorsomedial striatum (DMS), where EtOH increased outcome-related signals and decreased choice-related signals. In contrast, female rats showed minimal behavioral changes with distinct EtOH-evoked alterations of neural signals, revealing significant sex differences in the impact of chronic EtOH. Our findings underscore the profound impact of chronic EtOH exposure on adaptive decision-making, revealing enduring changes in neurocomputational processes in the striatum underlying cognitive deficits that differ by sex., Competing Interests: Competing interests: The authors declare that they have no competing interests.

Reward-associated cues serve different functions depending on whether they precede or terminate action sequences. Cues that precede action sequences and signal opportunity for reward could serve as GO signals to initiate the sequence, whereas sequence termination cues could serve as response feedback by signaling reward delivery. Reward expectation during sequence execution depends on these cues and might condition whether behavior is habitual or goal-directed. However, it remains unknown how sequence initiation and termination cues differentially affect reward expectation and contribute to habit learning. Further, while mesolimbic dopamine plays a key role in cue-induced reward expectation and sequence learning, how dynamic changes in dopamine signals differ depending on the response strategy is unclear. Here, we determined how mesolimbic DA signals change over training during cue-mediated sequence learning, depending on the type of cue and the nature of behavioral control. We found sequence initiation and termination cues differentially affect reward expectation during action sequences, with the termination cue contributing to habit and automaticity. Distinct response strategies induced by sequence initiation and termination cues induced differential changes in mesolimbic DA signals that captured variations in reward expectation along sequence execution. Notably, habit-like behavior induced by the sequence termination cue was associated with a rapid shift in DA signals from reward retrieval to the cue. This habit-like behavior was reflected in behavioral inflexibility and attenuated DA reward prediction error signals. Finally, using optogenetics, we provide evidence that phasic DA activity elicited by the sequence termination cue is critical for the development of habit-like behavior.

Niobates are promising all-climate Li + -storage anode material due to their fast charge transport, large specific capacities, and resistance to electrolyte reaction. However, their moderate unit-cell-volume expansion (generally 5%-10%) during Li + storage causes unsatisfactory long-term cyclability. Here, "zero-strain" NiNb 2 O 6 fibers are explored as a new anode material with comprehensively good electrochemical properties. During Li + storage, the expansion of electrochemical inactive NiO 6 octahedra almost fully offsets the shrinkage of active NbO 6 octahedra through reversible O movement. Such superior volume-accommodation capability of the NiO 6 layers guarantees the "zero-strain" behavior of NiNb 2 O 6 in a broad temperature range (0.53%//0.51%//0.74% at 25// - 10//60 °C), leading to the excellent cyclability of the NiNb 2 O 6 fibers (92.8%//99.2% // 91.1% capacity retention after 1000//2000//1000 cycles at 10C and 25// - 10//60 °C). This NiNb 2 O 6 material further exhibits a large reversible capacity (300//184//318 mAh g -1 at 0.1C and 25// - 10//60 °C) and outstanding rate performance (10 to 0.5C capacity percentage of 64.3%//50.0%//65.4% at 25// - 10//60 °C). Background: Currently, the conclusions of studies on subthalamic nucleus (STN) deep brain stimulation (DBS) for improving Parkinson's disease (PD) with depression are inconsistent, and the reasons for improvement or deterioration remain unclear., Methods: The aim was to investigate the prognosis of PD with depression after bilateral STN-DBS and the factors related to the improvement in depression. The local and network effects of DBS on depression in PD (DPD) were further explored based on the volume of tissue activation (VTA). The study analyzed 80 primary PD patients who had undergone bilateral STN-DBS, comprising 47 patients with improved depression and 33 patients without improvement. Two groups of clinical profiles and stimulation parameters were compared, and the network models for improving depression were constructed., Results: The improvement in depression was closely associated with improvement in anxiety (odd rate [OR] = 1.067, P = 0.006) and the standardized space left y-coordinate (OR = 0.253, P = 0.005). The VTA overlapping with the left motor STN subregion is most significantly associated with improvement in depression (R Spearman  = 0.53, P < 0.001; R Pearson  = 0.43, P < 0.001). The y-coordinates in the improvement group were closer to the optimal stimulation site for improving motor symptoms. Finally, both the structural and functional network models indicate a positive correlation between depression improvement and the connectivity of the sensorimotor cortex., Conclusion: The amelioration of DPD is primarily attributed to the stimulation of bilateral motor STN, particularly on the left. The construction of heterogeneous microstructure and the selection of multicomponents have turned into a research hotspot in developing ultralight, multifunctional, high-efficiency electromagnetic wave absorbing (EMA) materials. Although aerogels are promising materials to fulfill the above requirements, the increase in functional fillers inevitably leads to the deterioration of intrinsic properties. Tuning the electromagnetic properties from the structural design point of view remains a difficult challenge. Herein, we design customized pore creation strategies via introducing sacrificial templates to optimize the conductive path and construct the discontinuous dielectric medium, increasing dielectric loss and achieving efficient microwave absorption properties. A 3D porous composite (MEM) was crafted, which encapsulated an EVA/FeCoNi (EVA/MNPs) framework with Ti 3 C 2 T x MXene coating by employing a direct heated cross-linking and immersion method. Controllable adjustment of the conductive network inside the porous structure and regulation of the dielectric character are achieved by porosity variation. Eventually, the MEM-5 with a porosity of 66.67% realizes RL min of -39.2 dB (2.2 mm) and can cover the entire X band. Moreover, through off-axis electronic holography and the calculation of conduction loss and polarization loss, the dielectric property is deeply investigated, and the inner mechanism of optimization is pointed out. Thanks to the inherent characteristic of EVA and the porous structure, MEM-5 showed excellent thermal insulating and superior compressibility, which can maintain 60 °C on a 90-100 °C continuous heating stage and reached a maximum compressive strength of 60.12 kPa at 50% strain. Conceivably, this work provides a facile method for the fabrication of highly efficient microwave absorbers applied under complex conditions.

Acetonitrile (AN) is a compelling electrolyte solvent for high-voltage and fast-charging batteries, but its reductive instability makes it incompatible with lithium metal anodes (LMAs). Herein, 1,1,2,2-tetrafluoroethyl-2,2,3,3-tetrafluoropropyl ether (TTE) is used as the diluent to build an AN-based local high-concentration electrolyte (LHCE) to realize dense, dendrite-free, and stable LMAs. Such LHCE exhibits an exceptional electrochemical stability window close to 6 V (vs Li + /Li), excellent wettability, and promising flame retardancy. Compared to a baseline carbonate-based electrolyte, its electrochemical performance is prominent: the overpotential of lithium nucleation is minimal (only 24 mV), the average half-cell coulombic efficiency (CE) reaches 99.5% at 0.5 mA cm -2 , and its practicality in full cells with LiFePO 4 (LFP) and LiNi 0.8 Co 0.1 Mn 0.1 O 2 (NCM811) cathodes is also demonstrated. Compounding factors are identified to decipher the superiority of the AN-based LHCE. From the respect of solvation structures, both the elimination of free AN molecule and the diluent separation would contribute to prevention of anodic AN decomposition. Based on cryogenic electron microscopy (Cryo-EM) characterization and theoretical simulations results, the produced solid-electrolyte interphase (SEI) layer is uniform and compact. Spinel oxides have emerged as a promising candidate in the realm of nanozymes with variable oxidation states, while their limited active sites and low conductivity hinder further application. In this work, we synthesize a series of metal-doped NiCo 2 O 4 nanospheres decorated with Pd, which are deployed as highly efficient nanozymes for the detection of cancer biomarkers. Through meticulous modulation of the molar ratio between NiCo 2 O 4 and Pd, we orchestrated precise control over the oxygen vacancies and electronic structure within the nanozymes, a key factor in amplifying the catalytic prowess. Leveraging the superior H 2 O 2 reduction catalytic properties of Fe-NiCo 2 O 4 @Pd, we have successfully implemented its application in the electrochemical detection of biomarkers, achieving unparalleled analytical performance, much higher than that of Pd/C and other reported nanozymes. This research paves the way for innovative electron modification strategies in the design of high-performance nanozymes, presenting a formidable tool for clinical diagnostic analyses.

Objective: To analyze the factors related to the efficacy of consciousness-regaining therapy (CRT) for prolonged disorder of consciousness., Methods: A retrospective analysis was conducted on the case data of 114 patients with prolonged disorder of consciousness (pDOC) admitted to the Department of Functional Neurosurgery of Tianjin Huanhu Hospital from January 2019 to January 2022 to explore the relevant factors that affect the efficacy of CRT for pDOC. Next, basic information on the cases, data on pDOC disease assessment, CRT methods, and efficacy evaluation were collected., Results: These 114 patients were grouped, and a comparative analysis was done based on the efficacy at the end of treatment. Of these, 61 cases were allotted to the ineffective group and 53 cases to the effective group. There was a lack of statistical difference (P > 0.05) between the 2 groups based on gender, age, etiology, acute cerebral herniation, emergency craniotomy surgery, emergency decompressive craniectomy, time from onset to start of CRT, and CRT duration (P > 0.05). However, secondary hydrocephalus, CRT methods, JFK Coma Recovery Scale-Revised grading before treatment, and extended Glasgow Outcome Scale score at six months after treatment were found to be statistically different. The results of binary logistic regression analysis showed that the type of therapy (OR = 0.169, 95% CI: 0.057-0.508) affected the efficacy of CRT (P < 0.05)., Conclusions: Personalized awakening therapy using various invasive CRT methods could improve the efficacy of therapy for pDOC compared with noninvasive therapy., (Copyright © 2024 Elsevier Inc. Learning causal relationships relies on understanding how often one event precedes another. To investigate how dopamine neuron activity and neurotransmitter release change when a retrospective relationship is degraded for a specific pair of events, we used outcome-selective Pavlovian contingency degradation in rats. Conditioned responding was attenuated for the cue-reward contingency that was degraded, as was dopamine neuron activity in the midbrain and dopamine release in the ventral striatum in response to the cue and subsequent reward. Contingency degradation also abolished the trial-by-trial history dependence of the dopamine responses at the time of trial outcome. This profile of changes in cue- and reward-evoked responding is not easily explained by a standard reinforcement learning model. An alternative model based on learning causal relationships was better able to capture dopamine responses during contingency degradation, as well as conditioned behavior following optogenetic manipulations of dopamine during noncontingent rewards. Our results suggest that mesostriatal dopamine encodes the contingencies between meaningful events during learning.

Background: In advanced Parkinson's disease (PD), axial symptoms are common and can be debilitating. Although deep brain stimulation (DBS) significantly improves motor symptoms, conventional high-frequency stimulation (HFS) has limited effectiveness in improving axial symptoms. In this study, we investigated the effects on multiple axial symptoms after DBS surgery with three different frequency programming paradigms comprising HFS, low-frequency stimulation (LFS), and variable-frequency stimulation (VFS)., Methods: This study involved PD patients who had significant preoperative axial symptoms and underwent bilateral subthalamic nucleus (STN) DBS. Axial symptoms, motor symptoms, medications, and quality of life were evaluated preoperatively (baseline). One month after surgery, HFS was applied. At 6 months post-surgery, HFS assessments were performed, and HFS was switched to LFS. A further month later, we conducted LFS assessments and switched LFS to VFS. At 8 months after surgery, VFS assessments were performed., Results: Of the 21 PD patients initially enrolled, 16 patients were ultimately included in this study. Regarding HFS, all axial symptoms except for the Berg Balance Scale (p < 0.0001) did not improve compared with the baseline (all p > 0.05). As for LFS and VFS, all axial symptoms improved significantly compared with both the baseline and HFS (all p < 0.05). Moreover, motor symptoms and medications were significantly better than the baseline (all p < 0.05) after using LFS and VFS. Additionally, the quality of life of the PD patients after receiving LFS and VFS was significantly better than at the baseline and with HFS (all p < 0.0001)., Conclusion: Our findings indicate that HFS is ineffective at improving the majority of axial symptoms in advanced PD. However, both the LFS and VFS programming paradigms exhibit significant improvements in various axial symptoms., (© 2024. The ultrathin multi-nanolayered structure with ultrathin monolayer thickness (<10 nm) and certain interlayer spacing can significantly shorten Li + paths and alleviate the volume effect for Li + -storage materials. However, unlike layered materials such as MXene and MoS 2 , shear ReO 3 -type niobates have difficulty forming ultrathin multi-nanolayered structures due to their crystal structures, which still remains a challenge. Herein, by a polyvinylpyrrolidone (PVP)-assisted solvothermal method, we first synthesize ultrathin multi-nanolayered Cu 2 Nb 34 O 87-x with oxygen vacancies composed of ultrathin nanolayers (2-10 nm in thickness) and interlayer spacing (1-5 nm). Oxygen vacancies can radically enhance the inherent electronic/ionic conductivity and Li + diffusion coefficient of this material. The PVP-induced formation mechanism of this material is expounded in detail. The well-preserved ultrathin multi-nanolayered structure and excellent multi-electron electrochemical reversibility (Nb 5+  ↔ Nb 4+  ↔N b 3+ and Cu 2+  ↔ Cu + ) of this material during cycling are fully verified. Based on an ultrathin multi-nanolayered structure and oxygen vacancies, this material as the anode of lithium-ion batteries is highly competitive among reported shear ReO 3 -type Cu-Nb-O anodes, displaying a high reversible capacity (315.3 mAh g -1 after 300 cycles at 1 C), durable cycling stability (85.7 % capacity retention after 1000 cycles at 10 C), and outstanding rate performance. Moreover, the application of this material to lithium-ion capacitors generates a large energy density (97.9 Wh kg -1 at 87.5 W kg -1 ) and a high power density (17,500 W kg -1 at 12.6 Wh kg -1 ), thus further indicating its fast faradaic pseudocapacitive behavior for practical applications. Diabetic foot ulcers (DFUs) are one of the most common and challenging complications of diabetes, yet our understanding of their pathogenesis remains limited. We collected gene expression data of DFU patients from public databases. Bioinformatics tools were applied for systematic analysis, including the identification of differentially expressed genes (DEGs), weighted gene co-expression network analysis (WGCNA) and enrichment analysis. We further used single-cell RNA sequencing to identify the distribution of different cell populations in DFU. Finally, key results were validated using reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and flow cytometry. We identified 217 DEGs between ulcerated and healthy skin, and 37 DEGs between healing ulcers and ulcers. WGCNA revealed that the cyan module had the highest positive correlation with healthy skin and negative correlation with ulcers. The black module had the highest negative correlation with healthy skin and positive correlation with ulcers. Enrichment analysis showed that the genes in the cyan module were mainly associated with complement and coagulation cascades, while the genes in the black module were mainly associated with the IL-17 signalling pathway. In addition, CD8 T cells were significantly lower in ulcers than in healthy and healing ulcers. By comparing marker genes of CD8 T cells, we identified key genes in the cyan and black modules and validated their expression using RT-qPCR. The proportion of CD8 T cells was increased in healing ulcers. Flow cytometry detected increased levels of CD8 T, B and natural killer cells in healing ulcers. CD8 T cells and related key genes play an important role in the healing process of DFU. Impulse control disorders (ICDs), a wide spectrum of maladaptive behaviors which includes pathological gambling, hypersexuality and compulsive buying, have been recently suggested to be triggered or aggravated by treatments with dopamine D 2/3 receptor agonists, such as pramipexole (PPX). Despite evidence showing that impulsivity is associated with functional alterations in corticostriatal networks, the neural basis of the exacerbation of impulsivity by PPX has not been elucidated. Here we used a hotspot analysis to assess the functional recruitment of several corticostriatal structures by PPX in male rats identified as highly (HI), moderately impulsive (MI) or with low levels of impulsivity (LI) in the 5-choice serial reaction time task (5-CSRTT). PPX dramatically reduced impulsivity in HI rats. Assessment of the expression pattern of the two immediate early genes C-fos and Zif268 by in situ hybridization subsequently revealed that PPX resulted in a decrease in Zif268 mRNA levels in different striatal regions of both LI and HI rats accompanied by a high impulsivity specific reduction of Zif268 mRNA levels in prelimbic and cingulate cortices. PPX also decreased C-fos mRNA levels in all striatal regions of LI rats, but only in the dorsolateral striatum and nucleus accumbens core (NAc Core) of HI rats. Structural equation modeling further suggested that the anti-impulsive effect of PPX was mainly attributable to the specific downregulation of Zif268 mRNA in the NAc Core. Lithium-ion batteries (LIBs) are very popular electrochemical energy-storage devices. However, their applications in extreme environments are hindered because their low- and high-temperature electrochemical performance is currently unsatisfactory. In order to build all-climate LIBs, it is highly desirable to fully understand the underlying temperature effects on electrode materials. Here, based on a novel porous-microspherical yttrium niobate (Y 0.5 Nb 24.5 O 62 ) model material, this work demonstrates that the operation temperature plays vital roles in electrolyte decomposition on electrode-material surfaces, electrochemical kinetics, and crystal-structure evolution. When the operation temperature increases, the reaction between the electrolyte and the electrode material become more intensive, causing the formation of thicker solid electrolyte interface (SEI) films, which decreases the initial Coulombic efficiency. Meanwhile, the electrochemical kinetics becomes faster, leading to the larger reversible capacity, higher rate capability, and more suitable working potential (i.e., lower working potential for anodes and higher working potential for cathodes). Additionally, the maximum unit-cell-volume change becomes larger, resulting in poorer cyclic stability. Background: The research findings on the effects of subthalamic nucleus (STN) deep brain stimulation (DBS) in Parkinson's disease (PD) with Rapid Eye Movement Sleep Behavior Disorder (RBD) are inconsistent, and there is a lack of research on DBS electrode sites and their network effects for the explanation of the differences. Our objective is to explore the optimal stimulation sites (that is the sweet spot) and the brain network effects of STN-DBS for RBD in PD., Methods: In this study, among the 50 PD patients who underwent STN-DBS treatment, 24 PD patients with RBD were screened. According to clinical scores and imaging data, the sweet spot of STN-DBS was analyzed in PD patients with RBD, and the optimal structure and functional network models of subthalamic stimulation were constructed., Results: Bilateral STN-DBS can effectively improve the symptoms of RBD and other non-motor symptoms in 24 PD patients with RBD. RBD Questionnaire-Hong Kong (RBDQ-HK) score was 41.33 ± 17.45 at baseline and 30.83 ± 15.83 at 1-year follow-up, with statistical significance between them (P < 0.01). However, the MoCA score was an exception with a baseline of 22.04 ± 4.28 and a 1-year follow-up of 21.58 ± 4.33, showing no statistical significance (P = 0.12). Lithium (Li) metal is considered as the "holy grail" of anode materials for next-generation high energy batteries. However, notorious dendrite growth and interfacial instability could induce irreversible capacity loss and safety issues, limiting the practical application of Li metal anodes. Herein, we develop a novel approach to construct a borate-based artificial solid-electrolyte interphase (designated as B-SEI) through the reaction of metallic Li with triethylamine borane (TEAB). According to our cryogenic electron microscopy (Cryo-EM) characterization results, the artificial SEI adopts a glass-crystal bilayer structure, which facilitates uniform Li-ion transport and inhibits dendrite growth during Li plating. Benefiting from such an artificial SEI, the Li anode delivers an improved rate performance and prolonged cycle life. The symmetric Li/B-SEI||Li/B-SEI cell can maintain stable cycling for 700 h at a high current density of 3 mA cm -2 . The full-cell pairing Li/B-SEI with LiFePO 4 only exhibits minimal capacity decay after 500 cycles in a conventional carbonate-based electrolyte. This work demonstrates the feasibility of building a boride-based artificial SEI to stabilize the Li metal anode based on microscopic characterization results and comprehensive electrochemical data, which represents a promising avenue to develop practical Li metal batteries.

The reinforcement of voluntary alcohol-seeking behavior requires dopamine-dependent long-term synaptic plasticity in the striatum. Specifically, the long-term potentiation (LTP) of direct-pathway medium spiny neurons (dMSNs) in the dorsomedial striatum (DMS) promotes alcohol drinking. However, it remains unclear whether alcohol induces input-specific plasticity onto dMSNs and whether this plasticity directly drives instrumental conditioning. In this study, we found that voluntary alcohol intake selectively strengthened glutamatergic transmission from the medial prefrontal cortex (mPFC) to DMS dMSNs in mice. Importantly, mimicking this alcohol-induced potentiation by optogenetically self-stimulating mPFC→dMSN synapse with an LTP protocol was sufficient to drive the reinforcement of lever pressing in operant chambers. Conversely, induction of a post-pre spike timing-dependent LTD at this synapse time-locked to alcohol delivery during operant conditioning persistently decreased alcohol-seeking behavior. Our results establish a causal relationship between input- and cell-type-specific corticostriatal plasticity and the reinforcement of alcohol-seeking behavior. A stable solid electrolyte interphase (SEI) layer is crucial for lithium metal anode (LMA) to survive in long-term cycling. However, chaotic structures and chemical inhomogeneity of natural SEI make LMA suffering from exasperating dendrite growth and severe electrode pulverization, which hinder the practical application of LMAs. Here, we design a catalyst-derived artificial SEI layer with an ordered polyamide-lithium hydroxide (PA-LiOH) bi-phase structure to modulate ion transport and enable dendrite-free Li deposition. The PA-LiOH layer can substantially suppress the volume changes of LMA during Li plating/stripping cycles, as well as alleviate the parasitic reactions between LMA and electrolyte. The optimized LMAs demonstrate excellent stability in Li plating/stripping cycles for over 1000 hours at an ultra-high current density of 20 mA cm -2 in Li||Li symmetric cells. A high coulombic efficiency up to 99.2 % in Li half cells in additive-free electrolytes is achieved even after 500 cycles at a current density of 1 mA cm -2 with a capacity of 1 mAh cm -2 ., (© 2023 The Authors. High-concentrated non-flammable electrolytes (HCNFE) in lithium metal batteries prevent thermal runaway accidents, but the microstructure of their solid electrolyte interphase (SEI) remains largely unexplored, due to the lack of direct imaging tools. Herein, cryo-HRTEM is applied to directly visualize the native state of SEI at the atomic scale. In HCNFE, SEI has a uniform laminated crystalline-amorphous structure that can prevent further reaction between the electrolyte and lithium. The inorganic SEI component, Li 2 S 2 O 7 , is precisely identified by cryo-HRTEM. Niobate Li + -storage anode materials with shear ReO 3 crystal structures have attracted intensive attention due to their inherent safety and large capacities. However, they generally suffer from limited rate performance, cyclic stability, and temperature adaptability, which are rooted in their insufficient interlayer spacings. Here, sodium niobate (NaNb 13 O 33 ) micron-sized particles are developed as a new anode material owning the largest interlayer spacing among the known shear ReO 3 -type niobates. The large interlayer spacing of NaNb 13 O 33 enables very fast Li + diffusivity, remarkably contributing to its superior rate performance with a 2500 to 125 mA g -1 capacity percentage of 63.2%. Moreover, its large interlayer spacing increases the volume-accommodation capability during lithiation, allowing small unit-cell-volume variations (maximum 6.02%), which leads to its outstanding cyclic stability with 87.9% capacity retention after as long as 5000 cycles at 2500 mA g -1 . Its cyclic stability is the best in the research field of niobate micron-sized particles, and comparable to that of "zero-strain" Li 4 Ti 5 O 12 . At a low temperature of -10 °C, it also exhibits high rate performance with a 1250 to 125 mA g -1 capacity percentage of 65.6%, and even better cyclic stability with 105.4% capacity retention after 5000 cycles at 1250 mA g -1 . Addictive substance use impairs cognitive flexibility, with unclear underlying mechanisms. The reinforcement of substance use is mediated by the striatal direct-pathway medium spiny neurons (dMSNs) that project to the substantia nigra pars reticulata (SNr). Cognitive flexibility is mediated by striatal cholinergic interneurons (CINs), which receive extensive striatal inhibition. Here, we hypothesized that increased dMSN activity induced by substance use inhibits CINs, reducing cognitive flexibility. We found that cocaine administration in rodents caused long-lasting potentiation of local inhibitory dMSN-to-CIN transmission and decreased CIN firing in the dorsomedial striatum (DMS), a brain region critical for cognitive flexibility. Moreover, chemogenetic and time-locked optogenetic inhibition of DMS CINs suppressed flexibility of goal-directed behavior in instrumental reversal learning tasks. Notably, rabies-mediated tracing and physiological studies showed that SNr-projecting dMSNs, which mediate reinforcement, sent axonal collaterals to inhibit DMS CINs, which mediate flexibility. Background: Prolonged disorders of consciousness (pDOC) are common in neurology and place a heavy burden on families and society. This study is aimed at investigating the characteristics of brain connectivity in patients with pDOC based on quantitative EEG (qEEG) and extending a new direction for the evaluation of pDOC., Methods: Participants were divided into a control group (CG) and a DOC group by the presence or absence of pDOC. Participants underwent magnetic resonance imaging (MRI) T1 three-dimensional magnetization with a prepared rapid acquisition gradient echo (3D-T1-MPRAGE) sequence, and video EEG data were collected. After calculating the power spectrum by EEG data analysis tool, DTABR (( δ + θ )/( α + β ) ratio), Pearson's correlation coefficient (Pearson r ), Granger's causality, and phase transfer entropy (PTE), we performed statistical analysis between two groups. Finally, receiver operating characteristic (ROC) curves of connectivity metrics were made., Results: The proportion of power in frontal, central, parietal, and temporal regions in the DOC group was lower than that in the CG. The percentage of delta power in the DOC group was significantly higher than that in the CG, the DTABR in the DOC group was higher than that in the CG, and the value was inverted. The Pearson r of the DOC group was higher than that of CG. The Pearson r of the delta band ( Z = -6.71, P < 0.01), theta band ( Z = -15.06, P < 0.01), and alpha band ( Z = -28.45, P < 0.01) were statistically significant. Granger causality showed that the intensity of directed connections between the two hemispheres in the DOC group at the same threshold was significantly reduced ( Z = -82.43, P < 0.01). The PTE of each frequency band in the DOC group was lower than that in the CG. The PTE of the delta band ( Z = -42.68, P < 0.01), theta band ( Z = -56.79, P < 0.01), the alpha band ( Z = -35.11, P < 0.01), and beta band ( Z = -63.74, P < 0.01) had statistical significance., Conclusion: Brain connectivity analysis based on EEG has the advantages of being noninvasive, convenient, and bedside. Nonflammable electrolytes are critical for the safe operation of high-voltage lithium-ion batteries (LIBs). Although organic phosphates are effective flame retardants, their poor electrochemical stability with a graphite (Gr) anode and Ni-rich cathodes would lead to the deterioration of electrode materials and fast capacity decay. Herein, we develop a safe and high-performance electrolyte formulation for high-voltage (4.6 V-class) LIBs using flame-retarding ethoxy(pentafluoro) cyclotriphosphazene (PFPN) as a non-solvating diluent for the high-concentration carbonate-ether hybrid electrolyte. In contrast to conventional nonflammable additives with restricted dosage, the high level of PFPN (69% mass ratio in our electrolyte design) could significantly increase the electrolyte flash point and protect the favored anion-rich inner solvation sheath because of its non-solvating feature, thus preventing solvent co-intercalation and structural damage to the Gr anode. The nonflammable electrolyte could also form a stable LiF-rich cathode electrolyte interphase (CEI), which enables superior electrochemical performances of Gr‖LiNi 0.8 Mn 0.1 Co 0.1 O 2 (NMC811) full cells at high voltages (∼82.0% capacity retention after 1000 cycles at 4.5 V; 89.8% after 300 cycles at 4.6 V) and high temperatures (50 °C). Rechargeable solid-state Na metal batteries (SSNMB) can offer high operational safety and energy density. However, poor solid-solid contact between the electrodes and the electrolyte can dramatically increase interfacial resistance and Na dendrite formation, even at low current rates. Therefore, we developed a carbon-fiber-supported liquid Na-K alloy anode that ensures close anode-electrolyte contact, enabling superior cycle stability and rate capability. We then demonstrated the first cryogenic transmission electron microscopy (cryo-TEM) characterization of an SSNMB, capturing the evolution of solid-electrolyte interphase (SEI) and revealing both crystalline and amorphous phases, which could facilitate ion transport and prevent continuous side reactions. By enhancing contact between the Na-K alloy and solid-state electrolyte, these symmetric cells are capable of cycling for over 800 h without notable increased polarization and enable an unprecedented critical current density (CCD) at 40 mA cm -2 . Our liquid Na-K alloy approach offers a promising strategic avenue toward commercial SSNMBs.

Stress-associated proteins (SAPs), a class of A20/AN1 zinc finger proteins, play vital roles in plant stress response. However, investigation of SAPs in maize has been very limited. Herein, to better trace the evolutionary history of SAPs in maize and plants, 415 SAPs were identified in 33 plant species and four species of other kingdoms. Moreover, gene duplication mode exploration showed whole genome duplication contributed largely to SAP gene expansion in angiosperms. Phylogeny reconstruction was performed with all identified SAPs by the maximum likelihood (ML) method and the SAPs were divided into five clades. SAPs within the same clades showed conserved domain composition. Focusing on maize, nine ZmSAPs were identified. Further promoter cis-elements and stress-induced expression pattern analysis of ZmSAPs indicated that ZmSAP8 was a promising candidate in response to drought stress, which was the only AN1-AN1-C2H2-C2H2 type SAP in maize and belonged to clade I. Additionally, ZmSAP8 was located in the nucleus and had no transactivation activity in yeast. Overexpressing ZmSAP8 enhanced the tolerance to drought stress in Arabidopsis thaliana , with higher seed germination and longer root length. Our results should benefit the further functional characterization of ZmSAPs .

WRKY transcription factors present unusual research value because of their critical roles in plant physiological processes and stress responses. Taraxacum kok-saghyz Rodin (TKS) is a perennial herb of dandelion in the Asteraceae family. However, the research on TKS WRKY TFs is limited. In this study, 72 TKS WRKY TFs were identified and named. Further comparison of the core motifs and the structure of the WRKY motif was analyzed. These TFs were divided into three groups through phylogenetic analysis. Genes in the same group of TkWRKY usually exhibit a similar exon-intron structure and motif composition. In addition, virtually all the TKS WRKY genes contained several cis -elements related to stress response. Expression profiling of the TkWRKY genes was assessed using transcriptome data sets and Real-Time RT-PCR data in tissues during physiological development, under abiotic stress and hormonal treatments. For instance, the TkWRKY18 , TkWRKY23 , and TkWRKY38 genes were significantly upregulated during cold stress, whereas the TkWRKY21 gene was upregulated under heat-stress conditions. These results could provide a basis for further studies on the function of the TKS WRKY gene family and genetic amelioration of TKS germplasm.

The high specific capacity of alkalic metal (Li, Na, and K) anodes has drawn widespread interest; however, the practical applications of alkalic metal anodes have been hampered by dendrite growth and interfacial instability, resulting in performance deterioration and even safety issues. Here, we describe a simple method for building tunable fluoride-based artificial solid-electrolyte interphase (SEI) from the fluorination reaction of alkali metals with a mild organic fluorinating reagent. Comprehensive characterization by advanced electron microscopes shows that the LiF-based artificial SEI adopts a crystal-glass structure, which enables efficient Li ion transport and improves structural integrity against the volume changes that occur during Li plating/stripping. Compared with bare Li anode, the ones with artificial SEI exhibit decreased voltage hysteresis, enhanced rate capability, and prolonged cycle life. This method is also applied to generate fluoride-based artificial SEI on Na and K metal anodes that brings significant improvement in battery performance.

The 3D nanocomposite structure of plated lithium (Li Metal ) and solid electrolyte interphases (SEI), including a polymer-rich surficial passivation layer (SEI exoskeleton) and inorganic SEI "fossils" buried inside amorphous Li matrix, is resolved using cryogenic transmission electron microscopy. With ether-based DOLDME-LiTFSI electrolyte, LiF and Li 2 O nanocrystals are formed and embedded in a thin but tough amorphous polymer in the SEI exoskeleton. The fast Li-stripping directions are along [ 1 ¯ 10 ] or [ 12 1 ¯ ] , which produces eight exposed {111} planes at halfway charging. Full Li stripping produces completely sagging, empty SEI husks that can sustain large bending and buckling, with the smallest bending radius of curvature observed approaching tens of nanometers without apparent damage. In the 2nd round of Li plating, a thin Li BCC sheet first nucleates at the current collector, extends to the top end of the deflated SEI husk, and then expands its thickness. The apparent zero wetting angle between Li BCC and the SEI interior means that the heterogeneous nucleation energy barrier is zero. Due to its complete-wetting property and chemo-mechanical stability, the SEI largely prevents further reactions between the Li metal and the electrolyte, which explains the superior performance of Li-metal batteries with ether-based electrolytes. However, uneven refilling of the SEI husks results in dendrite protrusions and some new SEI formation during the 2nd plating. Co-existence of meningioma and breast carcinoma metastasis in the type of collision tumor is uncommon in the brain. A 65-year-old female with a history of breast carcinoma suffered from a mass in posterior fossa of brain. After a craniotomy, the pathological specimen showed a collision tumor with breast cancer metastasis and meningioma. In the clinical setting of previously vague diagnosis in imaging, craniotomy for removal of the mass is more recommended if conditions permit. Only the rare tumor is identified in pathology for further follow-up and research.

Exposure to addictive substances impairs flexible decision making. Cognitive flexibility is mediated by striatal cholinergic interneurons (CINs). However, how chronic alcohol drinking alters cognitive flexibility through CINs remains unclear. Here, we report that chronic alcohol consumption and withdrawal impaired reversal of instrumental learning. Chronic alcohol consumption and withdrawal also caused a long-lasting (21 days) reduction of excitatory thalamic inputs onto CINs and reduced pause responses of CINs in the dorsomedial striatum (DMS). CINs are known to inhibit glutamatergic transmission in dopamine D1 receptor-expressing medium spiny neurons (D1-MSNs) but facilitate this transmission in D2-MSNs, which may contribute to flexible behavior. We discovered that chronic alcohol drinking impaired CIN-mediated inhibition in D1-MSNs and facilitation in D2-MSNs. Importantly, in vivo optogenetic induction of long-term potentiation of thalamostriatal transmission in DMS CINs rescued alcohol-induced reversal learning deficits. These results demonstrate that chronic alcohol drinking reduces thalamic excitation of DMS CINs, compromising their regulation of glutamatergic transmission in MSNs, which may contribute to alcohol-induced impairment of cognitive flexibility. These findings provide a neural mechanism underlying inflexible drinking in alcohol use disorder.

Background: Stereoelectroencephalography (SEEG) has become a common diagnostic method in epilepsy surgery and is found to be safe for a wide range of clinical applications. SEEG combined with radiofrequency thermocoagulation (RF-TC) not only reveals the seizure onset zone by hypothesis, but also acts as a treatment option without any additional cost to institutions and patients. Thus, we analyzed the treatment of the SEEG-guided RF-TC., Methods: This retrospective study analyzed seventeen epileptic patients treated with RF-TC between April 2019 and December 2020. All patients underwent a single round of SEEG-guided RF-TC treatment after more than three habitual seizures were recorded. The demographic characteristics of the patients were retrospectively reviewed. Outcomes were assessed using the Engel classification system., Results: All patients underwent SEEG-guided RF-TC without catastrophic functional damage. Follow-up data of all patients were complete. The number of contacts per patients where RF-TC was applied ranged from 9 to 43 (mean: 17.7±10.2). After RF-TC, the types of anti-epileptic drugs used reduced from 2.4±0.7 to 1.6±0.7. With RF-TC alone, four (23.5%) patients achieved Engel Ia, two (11.8%) patients achieved Engel Ib, one patient underwent resection without seizure at the 5-month follow-up, five patients had a relapse after 3-10 months of seizure freedom, and five patients had recurrence after 1 month. After RF-TC, six patients underwent secondary interventions followed by resection. Overall, 12 patients achieved Engel Ia or Ib, three patients achieved Engel IIa or IIb, and two patients achieved Engel IIIa. There were no Engel IV cases., Conclusions: SEEG-guided RF-TC performed in our institution was found to be a safe ablation procedure for the treatment of drug-resistant focal epilepsy. All patients experienced a reduction in the frequency of seizures after receiving RF-TC. RF-TC can be used as a palliative treatment option for patients with epilepsy who refuse surgery or cannot undergo resection surgery. Background: Sevoflurane (Sev) is a rapidly acting, potent inhalation anesthetic with rapid uptake and elimination. But many studies have shown that Sev could result in cognition dysfunction in adolescent or aging patients. Now, therapeutic targeting with IL-17A antibody has shown a promising effect on Sev-induced cognition impairment. In the study we report that P300 inhibition could act as an alternative approach to decrease IL-17A activity., Methods: SHSY5Y cells were treated with Sev and cell apoptosis was evaluated by Annexin V-FITC/PI staining. The expression of P300 and IL-17A were assessed by Western blotting. Water maze tests were conducted in order to assess the cognitive function., Results: We found that P300 and IL-17A were activated in SHSY5Y cells treated with Sev. P300 inhibitor C646 could reduce the apoptosis induced by Sev through decreasing IL-17A avtivity. Furthermore, IL-17A expression was upregulated after neurons were transfected with P300 expression plasmid and IL-17A expression was downregulated after neurons were incubated with P300 inhibitor C646. P300 overexpression could upregulate the promoter activity of IL-17A. Finally, in a rat model treated with Sev, we also found C646 to significantly improve the cognition impairment through the IL-17A pathway., Conclusions: These data show that P300 will potentially be a new drug target for the therapy of cognition impairment caused by Sev., (Copyright © 2021 The Authors. Purpose: To compare the efficacy and safety between endoscopic submucosal dissection (ESD) and conventional surgical treatment in the treatment of colorectal cancer (CRC) and the precancerous lesions., Methods: A retrospective analysis was performed on the clinical data of 65 patients with CRC or precancerous lesions (ESD group) and another 65 patients receiving surgical treatment at the same period (Surgery group). The surgical indicators, incidence of complications, and quality of life score were compared between the two groups, and the survival and tumor progression were followed up and recorded., Results: The rate of en bloc tumor resection was 89.2% (58/65) and 100% (65/65) and the rate of tumor curative resection was 92.3% (60/65) and 100% (65/65) in ESD group and Surgery group. Moreover, ESD group had markedly shorter operation time and mean hospital stay. After treatment, ESD group had higher scores of emotional functioning, fatigue, constipation, and diarrhea symptoms and general quality of life on the European Organization for Research and Treatment of Cancer quality of life questionnaire Core 30 (EORTC QLQ-C30) than Surgery group. The follow-up results showed no statistically significant difference in the 5-year recurrence rate between ESD group and Surgery group (7.7% vs. 0%, p=0.208)., Conclusion: ESD and surgery have similar long-term clinical efficacy in treating early CRC and precancerous lesions, but ESD is more minimally invasive and safer, and is superior in accelerating postoperative recovery and improving the overall survival of patients.

Uncontrolled drug-seeking and -taking behaviors are generally driven by maladaptive corticostriatal synaptic plasticity. The orbital frontal cortex (OFC) and its projections to the dorsomedial striatum (DMS) have been extensively implicated in drug-seeking and relapse behaviors. The influence of the synaptic plasticity of OFC projections to the DMS (OFC→DMS) on drug-seeking and -taking behaviors has not been fully characterized. To investigate this, we trained rats to self-administer 20% alcohol and then delivered an in vivo optogenetic protocol designed to induce long-term potentiation (LTP) selectively at OFC→DMS synapses. We selected LTP induction because we found that voluntary alcohol self-administration suppressed OFC→DMS transmission and LTP may normalize this transmission, consequently reducing alcohol-seeking behavior. Importantly, ex vivo slice electrophysiology studies confirmed that this in vivo optical stimulation protocol resulted in a significant increase in excitatory OFC→DMS transmission strength on day two after stimulation, suggesting that LTP was induced in vivo. Rat alcohol-seeking and -taking behaviors were significantly reduced on days 1-3, but not on days 7-11, after LTP induction. Striatal synaptic plasticity is modulated by several critical neurotransmitter receptors, including dopamine D1 receptors (D1Rs) and adenosine A2A receptors (A2ARs). We found that delivery of in vivo optical stimulation in the presence of a D1R antagonist abolished the LTP-associated decrease in alcohol-seeking behavior, whereas delivery in the presence of an A2AR antagonist may facilitate this LTP-induced behavioral change. These results demonstrate that alcohol-seeking behavior was negatively regulated by the potentiation of excitatory OFC→DMS neurotransmission. The posterior dorsomedial striatum (pDMS) is mainly composed of medium spiny neurons (MSNs) expressing either dopamine D1 receptors (D1Rs) or D2Rs. Activation of these two MSN types produces opposing effects on addictive behaviors. However, it remains unclear whether pDMS D1-MSNs or D2-MSNs receive afferent inputs from different brain regions or whether the extrastriatal afferents express distinct dopamine receptors. To assess whether these afferents also contained D1Rs or D2Rs, we generated double transgenic mice, in which D1R-expressing and D2R-expressing neurons were fluorescently labeled. We used rabies virus-mediated retrograde tracing in these mice to perform whole-brain mapping of direct inputs to D1-MSNs or D2-MSNs in the pDMS. We found that D1-MSNs preferentially received inputs from the secondary motor, secondary visual, and cingulate cortices, whereas D2-MSNs received inputs from the primary motor and primary sensory cortices, and the thalamus. We also discovered that the bed nucleus of the stria terminalis (BNST) and the central nucleus of the amygdala (CeA) contained abundant D2R-expressing, but few D1R-expressing, neurons in a triple transgenic mouse model. Remarkably, although limited D1R or D2R expression was observed in extrastriatal neurons that projected to D1-MSNs or D2-MSNs, we found that cortical structures preferentially contained D1R-expressing neurons that projected to D1-MSNs or D2-MSNs, while the thalamus, substantia nigra pars compacta (SNc), and BNST had more D2R-expressing cells that projected to D2-MSNs. During the COVID-19 outbreak, some patients with COVID-19 pneumonia also suffered from acute abdomen requiring surgical treatment; however, there is no consensus for the treatment of such patients. In this study, we retrospectively reviewed 34 patients with acute abdomen who underwent emergency surgery during the COVID-19 outbreak. Among the 34 patients with acute abdomen, a total of six cases were found with COVID-19 pneumonia (clinical classification for COVID-19 pneumonia: all were the common type). On the premise of similar demographics between both groups, patients with COVID-19 pneumonia had worse indicators of liver and coagulation function. Compared with acute abdomen patients without COVID-19, patients with COVID-19 pneumonia had a longer hospital stay, but there were no significant differences in postsurgical complications (P = 0.58) or clinical outcomes (P = 0.56). In addition, an obvious resolution of lung inflammation after surgery was observed in five COVID-19 patients (83.3%). No new COVID-19 cases occurred during the patients' hospital stays. Therefore, for the common type of COVID-19 pneumonia, emergency surgery could not only improve the outcomes of COVID-19 pneumonia patients with acute abdomen, but also benefit the resolution of pulmonary inflammation.

Aims: Bacillus subtilis, a typical plant growth-promoting rhizobacteria, can benefit plant through promoting growth and reducing disease. The colonization intensity of B. subtilis in rhizosphere is a key factor for improving their effectiveness of field application. In this study, we developed a rapid and sensitive method for detecting B. subtilis in rhizosphere via TaqMan qPCR and droplet digital PCR (ddPCR) methods., Methods and Results: The primers/probe set targeting gyrB gene could successfully distinguish B. subtilis from its close-related species. Both the TaqMan qPCR and ddPCR methods exhibited a good linear relationship in the sensitivity assay, suggesting the developed method was specific, effective and reliable. Finally, the two methods were used to detect the colonization dynamic of B. subtilis within Arabidopsis rhizosphere. Both of them showed a consistent trend compared with the traditional cultivation-based and microscopy-based methods., Conclusions: The TaqMan qPCR and droplet digital PCR (ddPCR) methods we developed could be used to rapidly detect B. subtilis in rhizosphere., Significance and Impact of the Study: The TaqMan qPCR and ddPCR methods developed in this study can be applied to rapid quantitative detection of B. subtilis populations, and will be helpful to evaluate their effectiveness of field application., (© 2019 The Society for Applied Microbiology.)

Lithium metal anodes (LMAs) are critical for high-energy-density batteries such as Li-S and Li-O 2 batteries. The spontaneously formed solid electrolyte interface on LMAs is fragile, which may not accommodate the cyclic Li plating/stripping. This usually will result in a low coulombic efficiency (CE), short cycle life, and potential safety hazards induced by the uncontrollable growth of lithium dendrites. In this study, we fabricate a Li alginate-based artificial SEI (ASEI) layer that is chemically stable and allows easy Li ion transport on the surface of LMAs, thus enabling the stable operation of lithium metal anodes. Compared to bare LMAs, the ASEI layer-protected LMAs exhibit a more stable Li plating/stripping behavior and present effective dendrite suppression. The symmetric Li∥Li cells with the ASEI layer-protected LMAs can stably run for 850 and 350 h at current densities of 0.5 and 1 mA cm -2 , respectively. Additionally, the LiFePO 4 ∥Li full cell with the ASEI layer-protected LMA exhibits a capacity retention of about 94.0% coupled with a CE of 99.6% after 1000 cycles at 4 C. We believe that this study of engineering an ASEI brings a new and promising approach to the stabilization of LMAs for high-performance lithium metal batteries.

Dopaminergic modulation of corticostriatal transmission is critically involved in reward-driven behaviors. This modulation is mainly mediated by dopamine D1 receptors (D1Rs) and D2Rs, which are highly expressed in medium spiny neurons (MSNs) of the dorsomedial striatum (DMS), a brain region essential for goal-directed behaviors and addiction. D1Rs and D2Rs are also present at presynaptic cortical terminals within the DMS. However, it is not known how addictive substances alter the glutamatergic strength of striatal synapses expressing presynaptic dopamine receptors. Using cell type-specific Cre mice in combination with optogenetic techniques, we measured glutamatergic transmission at D1R- or D2R-expressing afferents to DMS MSNs. We found larger excitatory postsynaptic currents at the synapses between the extra-striatal D2R-expressing afferents and D1R-expressing MSNs (D2→D1), as compared with those observed at the other tested synapses (D1→D1, D1→D2, and D2→D2). Additionally, excessive alcohol consumption induced a long-lasting potentiation of glutamatergic transmission at the corticostriatal D2→D1 synapse. Furthermore, we demonstrated that activation of postsynaptic, but not presynaptic, D2Rs inhibited corticostriatal transmission in an endocannabinoid-dependent manner. Taken together, these data provide detailed information on the mechanisms underlying dopamine receptor-mediated modulation of brain reward circuitry.

Several novel techniques were developed recently to explore neural circuit mechanisms of neuropsychiatric disorders. These techniques include the Designer Receptors Exclusively Activated by Designer Drugs (DREADD)-based chemogenetic tools, which represent valuable platforms for selective and non-invasive control of neural activity with a high degree of spatial resolution. Among all variants, Gq- and Gi-DREADDs are widely used by neuroscientists to dissect out the circuitry and cellular signals. This review is focused on strategies to access a specific neuronal population or circuit using the DREADD technique and summarizes the current knowledge of the DREADDs' application in alcohol use disorder research and therapeutics., (Copyright © 2018 Elsevier Inc. Nuclear factor I/B (NFIB) is a widely studied transcription factor that participates in tumor progression; nevertheless, studies on NFIB in colorectal cancer (CRC) are limited. In our study, Western blot and RT-PCR analyses showed that NFIB was overexpressed in CRC tissues and cell lines, which was consistent with our bioinformatic analysis results. Furthermore, NFIB expression was closely related to the TNM stage of CRC. NFIB promoted cell proliferation and migration and inhibited cell apoptosis in vitro. Meanwhile, we discovered that NFIB accelerated xenograft tumor growth in vivo. In addition, NFIB weakened the sensitivity of CRC cells to 5-fluorouracil (5-FU). NFIB induced epithelial-mesenchymal transition (EMT) by upregulating snail expression, which was accompanied by decreased E-cadherin and Zo-1 expression and increasedd Vimentin expression. Because the Akt pathway plays an important role in CRC progression, we examined whether there was a correlation between NFIB and the Akt pathway in cell proliferation and migration. Our results showed that NFIB promoted cell proliferation and increased 5-FU resistance by activating the Akt pathway. In summary, our findings suggested that NFIB induced EMT of CRC cells via upregulating snail expression and promoted cell proliferation and 5-FU resistance by activating the Akt pathway., (© 2018 The Authors. Background: Prenatal alcohol exposure (PAE) is a leading cause of hyperactivity in children. Excitation of dopamine D1 receptor-expressing medium spiny neurons (D1-MSNs) of the dorsomedial striatum (DMS), a brain region that controls voluntary behavior, is known to induce hyperactivity in mice. We therefore hypothesized that PAE-linked hyperactivity was due to persistently altered glutamatergic activity in DMS D1-MSNs., Methods: Female Ai14 tdTomato reporter mice were given access to alcohol in an intermittent access, 2-bottle choice paradigm before pregnancy, and following mating with male D1-Cre mice, through the pregnancy period, and until postnatal day (P) 10. Locomotor activity was tested in juvenile (P21) and adult (P133) offspring, and alcohol-conditioned place preference (CPP) was measured in adult offspring. Glutamatergic activity in DMS D1-MSNs of adult PAE and control mice was measured by slice electrophysiology, followed by measurements of dendritic morphology., Results: Our voluntary maternal alcohol consumption model resulted in increased locomotor activity in juvenile PAE mice, and this hyperactivity was maintained into adulthood. Furthermore, PAE resulted in a higher alcohol-induced CPP in adult offspring. Glutamatergic activity onto DMS D1-MSNs was also enhanced by PAE. Finally, PAE increased dendritic complexity in DMS D1-MSNs in adult offspring., Conclusions: Our model of PAE does result in persistent hyperactivity in offspring. In adult PAE offspring, hyperactivity is accompanied by potentiated glutamatergic strength and afferent connectivity in DMS D1-MSNs, an outcome that is also consistent with the observed increase in alcohol preference in PAE offspring. Rechargeable lithium metal anodes (LMAs) with long cycling life have been regarded as the "Holy Grail" for high-energy-density lithium metal secondary batteries. The skeleton plays an important role in determining the performance of LMAs. Commercially available copper foam (CF) is not normally regarded as a suitable skeleton for stable lithium storage owing to its relatively inappropriate large pore size and relatively low specific surface area. Herein, for the first time, we revisit CF and address these issues by rationally designing a highly porous copper (HPC) architecture grown on CF substrates (HPC/CF) as a three-dimensional (3D) hierarchically bicontinuous porous skeleton through a novel approach combining the self-assembly of polystyrene microspheres, electrodeposition of copper, and a thermal annealing treatment. Compared to the CF skeleton, the HPC/CF skeleton exhibits a significantly improved Li plating/stripping behavior with high Coulombic efficiency (CE) and superior Li dendrite growth suppression. The 3D HPC/CF-based LMAs can run for 620 h without short-circuiting in a symmetric Li/Li@Cu cell at 0.5 mA cm -2 , and the Li@Cu/LiFePO 4 full cell exhibits a high reversible capacity of 115 mAh g -1 with a high CE of 99.7% at 2 C for 500 cycles. These results demonstrate the effectiveness of the design strategy of 3D hierarchically bicontinuous porous skeletons for developing stable and safe LMAs.

Addiction is proposed to arise from alterations in synaptic strength via mechanisms of long-term potentiation (LTP) and depression (LTD). However, the causality between these synaptic processes and addictive behaviors is difficult to demonstrate. Here we report that LTP and LTD induction altered operant alcohol self-administration, a motivated drug-seeking behavior. We first induced LTP by pairing presynaptic glutamatergic stimulation with optogenetic postsynaptic depolarization in the dorsomedial striatum, a brain region known to control goal-directed behavior. Blockade of this LTP by NMDA-receptor inhibition unmasked an endocannabinoid-dependent LTD. In vivo application of the LTP-inducing protocol caused a long-lasting increase in alcohol-seeking behavior, while the LTD protocol decreased this behavior. We further identified that optogenetic LTP and LTD induction at cortical inputs onto striatal dopamine D1 receptor-expressing neurons controlled these behavioral changes. Our results demonstrate a causal link between synaptic plasticity and alcohol-seeking behavior and suggest that modulation of this plasticity may inspire a therapeutic strategy for addiction.

Dopamine signals mainly through D1 receptors (D1Rs) and D2 receptors (D2Rs); D1R-expressing or D2R-expressing neurons contribute to distinct reward and addictive behaviors. Traditionally, transgenic mice expressing green fluorescent protein (GFP) under D1R or D2R promoters are used for fluorescent verification in electrophysiology studies, whereas Cre mice are employed for behavioral research. However, it is unknown whether the same neuronal populations are targeted in GFP and Cre mice. Additionally, while D1Rs and D2Rs are known to be expressed in different striatal neurons, their expression patterns outside the striatum remain unclear. The present study addressed these two questions by using several transgenic mouse lines expressing fluorescent proteins (GFP or tdTomato) or Cre under the control of D1R or D2R promoters. We found a high degree of overlap between GFP-positive and Cre-positive neurons in the striatum and hippocampus. Additionally, we discovered that D1Rs and D2Rs were highly segregated in the orbitofrontal cortex, prefrontal cortex, dorsal and ventral hippocampus, and amygdala: ~4-34 percent of neurons co-expressed these receptors. Importantly, slice electrophysiological studies demonstrated that D1R-positive and D1R-negative hippocampal neurons were functionally distinct in a mouse line generated by crossing Drd1a-Cre mice with a Cre reporter Ai14 line. Lastly, we discovered that chronic alcohol intake differentially altered D1R-positive and D2R-positive neuron excitability in the ventral CA1. These data suggest that GFP and Cre mice target the same populations of striatal neurons, D1R-expressing or D2R-expressing neurons are highly segregated outside the striatum, and these neurons in the ventral hippocampal may exert distinct roles in alcohol addiction., (© 2017 Society for the Study of Addiction.)

In this work, three-dimensional flower-like and petal-like copper hydroxyphosphate Cu 5 (OH) 4 (PO 4 ) 2 (CHP) based on the self-assembly of numerous nanosheets has been successfully fabricated on a copper foil by a mild one-pot wet-chemical method without ligand assistance. This research contributes to the development of the method to change the morphology of the CHP active material by varying the degree of substrate oxidation. The two different CHP architectures were used to photocatalytically degrade rhodamine 6G (Rh 6G) under solar light, which can absorb wide-range light wavelength from the UV to the near-infrared region. They all exhibit high photocatalytic activity and good durability, which are potential candidates for high performance and recyclable wide wavelength photocatalysis.

Objective: One of the critical mechanisms of gastrointestinal cancer pathogenesis is the silencing of death associated protein kinase 1 (DAPK1), which could be caused by aberrant methylation of the promoter. However, the relationship between DAPK1 methylation and the risk of gastrointestinal cancer is still controversial. Hence, we conducted this study to determine the potential correlation., Methods: Eligible publications were searched in the Pubmed, Embase, and Cochrane Library through November 2016 according to the inclusion criteria and exclusion criteria. Revman 5.3 and Stata 12.0 software were used to analyze the relevant data regarding the association between the frequency of DAPK1 methylation and gastrointestinal cancer., Results: A total of 22 studies with 2406 patients were included in this meta analysis. Methylation of DAPK1 was positively related with the risk of gastrointestinal cancer (odds ratio [OR] = 5.35, 95% confidence interval [CI]: 2.76-10.38, P<0.00001, random effects model). The source of heterogeneity was analyzed by sensitivity analysis and subgroup analysis. After omitting one heterogeneous study, the I2 decreased and the OR increased in pooled analysis. Also, the heterogeneity decreased most significantly in the subgroup of studies that had a sample size of less than 60 cases. Then, the correlations between DAPK1 methylation and clinicopathological features of gastrointestinal cancer were assessed. DAPK1 methylation was positively correlated with the lymph node (N) stage (positive vs. negative, OR = 1.45, 95%CI: 1.01-2.06, P = 0.04, fixed effects model) and poor differentiation (OR = 1.55, 95%CI: 1.02-2.35, P = 0.04, fixed effects model) in gastric cancer, and the association was significant among Asian patients. However, among cases of gastrointestinal cancer, the association between DAPK1 methylation and tumor (T) stage, N stage, distant metastasis (M) stage, and cancer differentiation were not statistically significant., Conclusions: DAPK1 methylation is a potential biomarker for the early diagnosis of gastrointestinal cancer. Further analysis of the clinicopathological features indicated that aberrant methylation of DAPK1 is positively associated with the tumorigenesis of gastrointestinal cancer, and metastasis of gastric cancer.
