Purpose: Skin injury, often caused by physical or medical mishaps, presents a significant challenge as wound healing is critical to restore skin integrity and tissue function. However, external factors such as infection and inflammation can hinder wound healing, highlighting the importance of developing biomaterials with antibiotic and wound healing properties to treat infections and inflammation. In this study, a novel photothermal nanomaterial (MMPI) was synthesized for infected wound healing by loading indocyanine green (ICG) on magnesium-incorporated mesoporous bioactive glass (Mg-MBG) and coating its surface with polydopamine (PDA)., Results: In this study, Mg-MBG and MMPI was synthesized via the sol-gel method and characterized it using various techniques such as scanning electron microscopy (SEM), the energy dispersive X-ray spectrometry (EDS) system and X-ray diffraction (XRD). The cytocompatibility of MMPI was evaluated by confocal laser scanning microscopy (CLSM), CCK8 assay, live/dead staining and F-actin staining of the cytoskeleton. The antibacterial efficiency was assessed using bacterial dead-acting staining, spread plate method (SPM) and TEM. The impact of MMPI on macrophage polarization was initially evaluated through flow cytometry, qPCR and ELISA. Additionally, an in vivo experiment was performed on a mouse model with skin excision infected. Objective: To investigate the physicochemical properties, osteogenic properties, and osteogenic ability in rabbit model of femoral condylar defect of acellular dermal matrix (ADM)/dicalcium phosphate (DCP) composite scaffold., Methods: ADM/DCP composite scaffolds were prepared by microfibril technique, and the acellular effect of ADM/DCP composite scaffolds was detected by DNA residue, fat content, and α-1，3-galactosyle (α-Gal) epitopes; the microstructure of scaffolds was characterized by field emission scanning electron microscopy and mercury porosimetry; X-ray diffraction was used to analyze the change of crystal form of scaffold; the solubility of scaffolds was used to detect the pH value and calcium ion content of the solution; the mineralization experiment in vitro was used to observe the surface mineralization. Twelve healthy male New Zealand white rabbits were selected to prepare the femoral condylar defect models, and the left and right defects were implanted with ADM/DCP composite scaffold (experimental group) and skeletal gold ® artificial bone repair material (control group), respectively. Gross observation was performed at 6 and 12 weeks after operation; Micro-CT was used to detect and quantitatively analyze the related indicators [bone volume (BV), bone volume/tissue volume (BV/TV), bone surface/bone volume (BS/BV), trabecular thickness (Tb.Th), trabecular number (Tb.N), trabecular separation (Tb.Sp), bone mineral density (BMD)], and HE staining and Masson staining were performed to observe the repair of bone defects and the maturation of bone matrix., Results: Gross observation showed that the ADM/DCP composite scaffold was a white spongy solid. Compared with ADM, ADM/DCP composite scaffolds showed a significant decrease in DNA residue, fat content, and α-Gal antigen content ( P <0.05). Field emission scanning electron microscopy showed that the ADM/DCP composite scaffold had a porous structure, and DCP particles were attached to the porcine dermal fibers. The porosity of the ADM/DCP composite scaffold was 76.32%±1.63% measured by mercury porosimetry. X-ray diffraction analysis showed that the crystalline phase of DCP in the ADM/DCP composite scaffolds remained intact. Mineralization results in vitro showed that the hydroxyapatite layer of ADM/DCP composite scaffolds was basically mature. The repair experiment of rabbit femoral condyle defect showed that the incision healed completely after operation without callus or osteophyte. Micro-CT showed that bone healing was complete and a large amount of new bone tissue was generated in the defect site of the two groups, and there was no difference in density between the defect site and the surrounding bone tissue, and the osteogenic properties of the two groups were equivalent. There was no significant difference in BV, BV/TV, BS/BV, Tb.Th, Tb.N, and BMD between the two groups ( P >0.05), except that the Tb.Sp in the experimental group was significantly higher than that in the control group ( P <0.05). At 6 and 12 weeks after operation, HE staining and Masson staining showed that the new bone and autogenous bone fused well in both groups, and the bone tissue tended to be mature., Conclusion: The ADM/DCP composite scaffold has good biocompatibility and osteogenic ability similar to the artificial bone material in repairing rabbit femoral condylar defects. It is a new scaffold material with potential in the field of bone repair.

Wetland vegetation biomass is an essential indicator of wetland health, and its estimation has become an active area of research. Zizania latifolia ( Z. latifolia ) is the dominant species of emergent vegetation in Honghu Wetland, and monitoring its aboveground biomass (AGB) can provide a scientific basis for the protection and restoration of this and other wetlands along the Yangtze River. This study aimed to develop a method for the AGB estimation of Z. latifolia in Honghu Wetland using high-resolution RGB imagery acquired from an unoccupied aerial vehicle (UAV). The spatial distribution of Z. latifolia was first extracted through an object-based classification method using the field survey data and UAV RGB imagery. Linear, quadratic, exponential and back propagation neural network (BPNN) models were constructed based on 17 vegetation indices calculated from RGB images to invert the AGB. The results showed that: (1) The visible vegetation indices were significantly correlated with the AGB of Z. latifolia . The absolute value of the correlation coefficient between the AGB and CIVE was 0.87, followed by ExG (0.866) and COM2 (0.837). (2) Among the linear, quadratic, and exponential models, the quadric model based on CIVE had the highest inversion accuracy, with a validation R 2 of 0.37, RMSE and MAE of 853.76 g/m 2 and 671.28 g/m 2 , respectively. (3) The BPNN model constructed with eight factors correlated with the AGB had the best inversion effect, with a validation R 2 of 0.68, RMSE and MAE of 732.88 g/m 2 and 583.18 g/m 2 , respectively. ​Compared to the quadratic model constructed by CIVE, the BPNN model achieved better results, with a reduction of 120.88 g/m 2 in RMSE and 88.10 g/m 2 in MAE. Lakes are important natural resources and carbon gas emitters and are undergoing rapid changes worldwide in response to climate change and human activities. A detailed global characterization of lakes and their long-term dynamics does not exist, which is however crucial for evaluating the associated impacts on water availability and carbon emissions. Here, we map 3.4 million lakes on a global scale, including their explicit maximum extents and probability-weighted area changes over the past four decades. From the beginning period (1984-1999) to the end (2010-2019), the lake area increased across all six continents analyzed, with a net change of +46,278 km 2 , and 56% of the expansion was attributed to reservoirs. Interestingly, although small lakes (<1 km 2 ) accounted for just 15% of the global lake area, they dominated the variability in total lake size in half of the global inland lake regions. The identified lake area increase over time led to higher lacustrine carbon emissions, mostly attributed to small lakes. Skin cutaneous melanoma has high morbidity and mortality. Identification of reliable and quantitative melanoma biomarkers could facilitate an early diagnosis and improve survival and morbidity rates. CD96 has a significant role in adjusting immune function. Although the abnormal expression of CD96 has been reported to participate in carcinogenesis in many human types of cancer, the bioinformatics role of the CD96 in melanoma is unknown. Expression degrees and their underlying functions were first studied by this study. According to TCGA, GTEx, and gene expression profile interaction analysis dataset in this paper, compared with normal skin tissues, CD96 was expressed at higher levels in human cutaneous melanoma skin tissues. Meanwhile, we detected the relative CD96 expression levels by immunohistochemistry. Gene functional enrichment analyses were applied through cBioPortal database analysis. CD96 was clearly upregulated in skin cutaneous melanoma patients and carried out its effects through regulating several signaling pathways, containing the JAK-STAT, PI3K-Akt, and MAPK. Hierarchically porous carbons (HPCs), with large specific surface area, abundant porous channels and adequate anchor points, act as one type of ideal carbon supports for the preparation of single-atom electrocatalysts. In this study, the blood plasma-derived HPC with an interconnected porous framework is constructed via a generated-template method, with the formation of ZnS nanoparticles from the abundant disulfide bonds (-S-S-) in serum albumin. After the thermal activation with heme-containing molecules (also from the bovine-blood biowaste), the HPC exhibits high-exposure and low-spin-state Fe(ii)-N 4 atomic active sites, and thereby presents a superior oxygen reduction reaction activity (the half wave potential of 0.87 V) and excellent stability (a 4 mV negative shift after 3000 potential cycles), even comparable with the benchmark Pt/C. Many Gram-negative bacterial pathogens utilize the type III secretion system (T3SS) to inject virulence factors, named effectors, into host cells. These T3SS effectors manipulate host cellular signaling pathways to facilitate bacterial pathogenesis. Death receptor signaling plays an important role in eukaryotic cell death pathways. NleB from enteropathogenic Escherichia coli (EPEC) and SseK1/3 from Salmonella enterica serovar Typhimurium ( S . Typhimurium) are T3SS effectors. They are defined as a family of arginine GlcNAc transferase to modify a conserved arginine residue in the death domain (DD) of the death receptor TNFR and their corresponding adaptors to hijack death receptor signaling. Here we identified that these enzymes, NleB, SseK1, and SseK3 could catalyze auto-GlcNAcylation. Residues, including Arg13/53/159/293 in NleB, Arg30/158/339 in SseK1, and Arg153/184/305/335 in SseK3 were identified as the auto-GlcNAcylation sites by mass spectrometry. Mutation of the auto-modification sites of NleB, SseK1, and SseK3 abolished or attenuated the capability of enzyme activity toward their death domain targets during infection. Loss of this ability led to the increased susceptibility of the cells to TNF- or TRAIL-induced cell death during bacterial infection. BACKGROUND Spinal cord injury (SCI) is a severe devastating condition associated with serious disability and neurologic deficits. Aberrant micro RNA (miRNA) expression has been related to a variety of central nervous system diseases including SCI. In the present study, we aimed to discover the role of miR-129-5p on SCI. MATERIAL AND METHODS An acute SCI rat model was induced, following the modified Allen method. A total of 36 rats were randomly assigned into 4 groups (n=9 in every group): Sham group; Model group (SCI+saline); SCI+NC group; and SCI+miR-129-5p group (100 nm solution, every 2 days). Basso-Beattie-Bresnahan (BBB) locomotor rating score was carried out to determine functional recovery. TUNEL (terminal dUTP nick-end labeling) staining was used to evaluate cell apoptosis. Hematoxylin and eosin staining was performed to assess the pathological state of spinal cord. Furthermore, western blot assay was conducted to measure the calpain1 and calpain2 expression. RESULTS Our data suggested that the expression level of miR-129-5p was markedly reduced in rats after SCI. Then miR-129-5p mimic was injected into the vertebral canal. We found that the SCI+miR-129-5p group had a high score in the BBB test compared with the SCI+NC group and the Model group. The overexpression of miR-129-5p obviously reduced tissue loss, damaged cells, and the number of TUNEL positive cells. Moreover, western blot assay exhibited that overexpression of miR-129-5p decreased calpain1, calpain2, and cleaved caspase-3 expression. CONCLUSIONS Our findings suggested that overexpression of miR-129-5p improved neurological function by promoting functional recovery, reducing tissue loss and cell apoptosis in rats in an SCI model, possibly through downregulation of calpain1 and calpain2.

Enclosure fisheries have accommodated the widespread expansion of aquaculture in many lakes throughout the Yangtze Plain (YP), China, for over four decades. Such practices have increased food provision but have also triggered various detrimental environmental consequences. To restore ecosystem functions, the Chinese government recently implemented specific regulations to remove enclosure fences from lakes throughout the YP. However, little information is available on the spatial and temporal distributions of the enclosure fences, particularly in relation to the enforcement of recent policy changes. Using synthetic aperture radar (SAR) satellite images taken between 2002 and 2018, we conducted the first comprehensive assessment of the interannual changes in enclosure fences in 17 large lakes throughout the YP. Consistent decreases in fence density were found in most lakes after 2015; 15 lakes had >50% of their fences removed, while 9 lakes had >90% removed. The timing and implementation of the development and destruction of enclosure fisheries were related to government policy; before 2015, regional dynamics in enclosure fisheries were attributed to provincial policies, whereas the nearly ubiquitous fence demolition after 2015 was likely a response to national policy. This study represents remotely sensed evidence that demonstrates the importance of both local and national environmental policies and their effectiveness in mitigating ongoing human impacts on vulnerable and valuable natural resources. These findings provide valuable baseline information for future lake environmental monitoring and restoration in the YP region, and the methods used here could be applied to other lacustrine and coastal regions experiencing similar aquaculture activities., (Copyright © 2019 Elsevier B.V. Bone nonunion remains a challenge during the treatment of bone defect accompanied with infection. Epimedii herba is a widely used medicine in clinic to enhance bone healing. Epimedin C and icariin are the major flavonoid glycosides from Epimedii herba. Although the effects of these compounds have been reported, their different absorption and utilisation by osteoblasts remain unclear. In the present study, lipopolysaccharide (LPS)-induced osteoblast was adopted as the model cell to evaluate the effects of epimedin C and icariin. The intracellular and extracellular drug concentrations within 24 h were assayed by pipette tip solid-phase extraction and high-performance liquid chromatography, respectively. MTT, alkaline phosphatase (ALP) and calcified nodule staining were performed to identify and evaluate the effects of epimedin C and icariin on LPS-induced osteoblasts. The regulatory roles of epimedin C and icariin in the bone morphogenetic protein-2 (BMP-2)/Runt-related transcription factor 2 (Runx2) signalling pathway were investigated. The results revealed that epimedin C and icariin were not efficiently absorbed by LPS-induced osteoblasts. Nevertheless, they still had high utilisation efficiency after entering the cells. ALP activity, mineralisation and osteoblasts proliferation were enhanced by a high concentration of epimedin C and icariin. The suppressed expression of BMP-2 and Runx2 in LPS-induced osteoblasts was up-regulated significantly after treatment with epimedin C and icariin. These findings firstly illustrated the behavior of epimedin C and icariin from Epimedii herba on LPS-induced osteoblasts and the regulatory property on the expression of key genes and proteins of the BMP-2/Runx2 signalling pathway, which might be helpful for better understanding flavonoids' mechanism to enhance bone repair and improving their future application., (Copyright © 2019 The Authors. High tibial osteotomy (HTO) has been widely used for clinical treatment of osteoarthritis of the medial compartment of the knee, and both opening-wedge and closing-wedge HTO are the most commonly used methods. However, it remains unclear which technique has better clinical and radiological outcomes in practice. To systematically evaluate this issue, we conducted a comprehensive meta-analysis by pooling all available data for the opening-wedge HTO and closing-wedge HTO techniques from the electronic databases including PubMed, Embase, Wed of Science and Cochrane Library. A total of 22 studies encompassing 2582 cases were finally enrolled in the meta-analysis. There was no significant difference regarding surgery time, duration of hospitalization, knee pain VAS, Lysholm score and HSS knee score (clinical outcomes) between the opening-wedge and closing-wedge HTO groups (P > 0.05). However, the opening-wedge HTO group showed wider range of motion than the closing-wedge HTO group (P = 0.003). Moreover, as for Hip-Knee-Ankle angle and mean angle of correction, no significant difference was observed between the opening-wedge and closing-wedge HTO groups (P > 0.05), while the opening-wedge HTO group showed greater posterior tibial slope angle (P < 0.001) and lesser patellar height than the closing-wedge HTO group (P < 0.001). A two-dimensional (2D) water quality model was established to determine the response of water quality variables and submerged aquatic vegetation biomass to load reduction from watershed inflows and enclosure aquaculture in Honghu Lake in China. Results showed that the total nitrogen (TN) and total phosphorus (TP) loads from upstream discharge were the major external loads in the lake, accounting for 70% and 63% of the total loads, respectively. Scenario simulation results indicated that 93.2% of the lake area in summer (August) and 89.5% in autumn (November) could reach the protective targets (TN<1.0mg/L) under 50% reduction of inflow TN loads. Meanwhile, 58.7% of the lake area in summer and 63.1% in autumn could reach the protective targets (TP<0.05mg/L) under 50% reduction of aquaculture areas. The mass budget results of TN and TP showed that TP immobilisation was larger than TN immobilisation. The immobilisations for TN and TP from July to September were higher than those of other months under the combined impacts of increasing runoff during the wet period, phytoplankton bloom and water residence time. The 2D water quality model provided a relevant example for assessing the effects of runoff and aquaculture activities and served as scientific support for lake management to improve water quality in large shallow macrophytic lakes., (Copyright © 2016. Basin-scale water volumes of lakes and reservoirs are difficult to obtain due to a number of challenges. In this study, area-based water storage estimation models are proposed for large lakes and reservoirs in the Yangtze River Basin (YRB). The models are subsequently applied to Moderate Resolution Imaging Spectroradiometer (MODIS) observations of 128 large lakes and 108 reservoirs between 2000 and 2014, and the first comprehensive map of the temporal and spatial dynamics of water storage in large water bodies in the YRB is provided. The results show that 53.91% of the lakes experienced significant decreasing trends in water storage during this period, and the total water storage in lakes showed a decreasing trend of 14 million m 3 month -1 . By contrast, a monthly mean increase of 177 million m 3 was observed for water storage in reservoirs. Our analysis revealed that the pronounced increase in reservoirs was primarily due to the rapid water level increase in the Three Gorges Reservoir in recent years, while understanding the water loss in lakes requires additional studies. The long-term data presented in this study provide critical baseline information for future water resource monitoring and regulation in the YRB and China.
