Objective: This retrospective follow-up study analyzes the effect of intrauterine postpartum hemorrhage (PPH) therapy on menstrual, reproductive, and mental health outcomes., Methods: All women who delivered at a university hospital between 2016 and 2021 with PPH and who needed intrauterine therapy were included. A questionnaire on well-being, menses, fertility, and reproductive outcomes was mailed to the patients. Those who did not reply were surveyed by telephone., Results: A total of 214 women treated with chitosan-covered gauze (group A) and 46 women treated with a balloon tamponade (group B) were recruited, and their short-term courses were analyzed. For long-term follow-up, 71 women of group A (33%) and 21 women of group B (46%) could be reached. A total of 89% of group A and 95% of group B had regular menstrual bleeding in the most recent 12 months; 27% (group A) and 29% (group B) were trying to conceive again, and all of them did so successfully. There were 12 deliveries, 3 ongoing pregnancies, 3 miscarriages, and 2 terminations of pregnancies (TOP) in group A and 4 deliveries, 1 miscarriage, and 2 TOPs in group B. More than half of our study participants was sorted into grade II or III of the Impact of Events Scale, indicating they experienced clinical impacts in the form of psychological sequelae. One-quarter of patients had symptoms of post-traumatic stress disorder., Conclusion: Chitosan gauze as well as balloon tamponade appear to have few adverse effects on subsequent menstrual and reproductive function. Women after PPH are at increased risk of long-term adverse psychological outcomes.

Colistin resistance in Escherichia coli is of public health significance for its use to treat multidrug-resistant Gram-negative infections. Amino acid variations in PmrB have been implicated in colistin resistance in E. coli . In this cross-sectional study, 288 generic E. coli isolates from surveillance of broiler chicken and feedlot cattle feces, retail meat, wastewater, and well water were whole-genome sequenced. Phylogroup designation and screening for two amino acid substitutions in PmrB putatively linked to colistin resistance (Y358N, E123D) were performed in silico . Three additional data sets of publicly available E. coli assemblies were similarly scrutinized: (i) E. coli isolates from studies identifying the Y358N or E123D substitutions, (ii) colistin-susceptible E. coli isolates reported in the literature, and (iii) a random sampling of 14,700 E. coli assemblies available in the National Center for Biotechnology Information public database. Within all data sets, ≥95% of phylogroup B1 and C isolates have the PmrB Y358N variation. The PmrB E123D amino acid substitution was only identified in phylogroup B2 isolates, of which 94%-100% demonstrate the substitution. Both PmrB amino acid variations were infrequent in other phylogroups. Among published colistin susceptible isolates, colistin minimum inhibitory concentrations (MICs) were not higher in isolates bearing the E123D and Y358N amino acid variations than in isolates without these PmrB substitutions. The E123D and Y358N PmrB amino acid substitutions in E. coli appear strongly associated with phylogroup. The previously observed associations between Y358N and E123D amino acid substitutions in PmrB and colistin resistance in E. coli may be spurious., Importance: Colistin is a critical last-resort treatment for extensively drug-resistant Gram-negative infections in humans. Therefore, accurate identification of the genetic mechanisms of resistance to this antimicrobial is crucial to effectively monitor and mitigate the spread of resistance. Examining over 16,000 whole-genome sequenced Escherichia coli isolates, this study identifies that PmrB E123D and Y358N amino acid substitutions previously associated with colistin resistance in E. coli are strongly associated with phylogroup and are alone not sufficient to confer a colistin-resistant phenotype. This is a critical clarification, as both substitutions are identified as putative mechanisms of colistin resistance in many publications and a common bioinformatic tool. Given the potential spurious nature of initial associations of these substitutions with colistin resistance, this study's findings emphasize the importance of appropriate experimental design and consideration of relevant biological factors such as phylogroup when ascribing causal mechanisms of resistance to chromosomal variations., Competing Interests: The authors declare no conflict of interest.

Foot and mouth disease (FMD) is a highly contagious infection caused by FMD-virus (FMDV) that affects livestock worldwide with significant economic impact. The main strategy for the control is vaccination with FMDV chemically inactivated with binary ethylenimine (FMDVi). In FMDV infection and vaccination, B cell response plays a major role by providing neutralizing/protective antibodies in animal models and natural hosts. Extracellular vesicles (EVs) and small EVs (sEVs) such as exosomes are important in cellular communication. EVs secreted by antigen-presenting cells (APC) like dendritic cells (DCs) participate in the activation of B and T cells through the presentation of native antigen membrane-associated to B cells or by transferring MHC-peptide complexes to T cells and even complete antigens from DCs. In this study, we demonstrate for the first time that APC activated with the FMDVi O1 Campos vaccine-antigens secrete EVs expressing viral proteins/peptides that could stimulate FMDV-specific immune response. The secretion of EVs-FMDVi is a time-dependent process and can only be isolated within the first 24 h post-activation. These vesicles express classical EVs markers (CD9, CD81, and CD63), along with immunoregulatory molecules (MHC-II and CD86). With an average size of 155 nm, they belong to the category of EVs. Studies conducted in vitro have demonstrated that EVs-FMDVi express antigens that can stimulate a specific B cell response against FMDV, including both marginal zone B cells (MZB) and follicular B cells (FoB). These vesicles can also indirectly or directly affect T cells, indicating that they express both B and T epitopes. Additionally, lymphocyte expansion induced by EVs-FMDVi is greater in splenocytes that have previously encountered viral antigens in vivo . The present study sheds light on the role of EVs derived from APC in regulating the adaptive immunity against FMDV. This novel insight contributes to our current understanding of the immune mechanisms triggered by APC during the antiviral immune response. Objective: This study compared clinical and immunological responses to coinfection challenge of beef calves mucosally primed and differentially boosted with commercial combination vaccines containing antigens against bovine coronavirus (BCoV), bovine parainfluenza virus Type 3 (BPIV3), and bovine respiratory syncytial virus (BRSV)., Animals: Nineteen commercial beef heifers., Procedure: At birth, calves were mucosally (IN) primed with modified-live virus (MLV) vaccines, differentially boosted by injection of either combination MLV (IN-MLV) or inactivated virus (IN-KV) vaccines at a mean age of 44 d, and then challenged by coinfection with BCoV, BPIV3, and BRSV at weaning., Results: Both groups were similarly protected from clinical disease and had anamnestic neutralizing antibody responses to all 3 viruses. Objective: To describe the copper and selenium statuses of beef calves at weaning., Animal: Calves ( n = 1998) were sampled from 106 Canadian cow-calf herds in the fall of 2021., Procedure: Serum samples from calves were tested for copper, selenium, and molybdenum concentrations., Results: Although the percentages of calves classified as selenium deficient (< 0.025 ppm) were relatively low (0.5% western Canada, 3% eastern Canada), 53% of calves from western Canada and 77% of calves from eastern Canada were classified as having less than adequate selenium concentrations (< 0.08 ppm). Copper deficiency (< 0.5 ppm) was common in calves from both western (17%) and eastern (14%) Canada. High molybdenum concentrations (> 0.10 ppm) were identified in 6% of calves from western Canada and 7% of calves from eastern Canada., Conclusion: Selenium concentrations were higher in calves from western Canada than from those in eastern Canada ( P < 0.001). Copper and molybdenum concentrations were not significantly different between western and eastern Canada. Less-than-adequate serum copper was the most common deficiency identified in Canadian beef calves at weaning., Clinical Relevance: Trace minerals are important for immune system function in calves at weaning. Selenium concentrations in calves at weaning were lower than in cows from the same herds collected at pregnancy testing 2 y earlier. Copper deficiency was also identified, though less frequently than for mature cows. Bovine respiratory disease (BRD) is an important health and economic burden to the cattle industry worldwide. Three bacterial pathogens frequently associated with BRD (Mannheimia haemolytica, Pasteurella multocida, and Histophilus somni) can possess integrative and conjugative elements (ICEs), a diverse group of mobile genetic elements that acquire antimicrobial resistance (AMR) genes (ARGs) and decrease the therapeutic efficacy of antimicrobial drugs. We developed a duplex recombinase polymerase amplification (RPA) assay to detect up to two variants of ICEs in these Pasteurellaceae. Whole genome sequence analysis of M. haemolytica, P. multocida, and H. somni isolates harbouring ICEs revealed the presence of tnpA or ebrB next to tet(H), a conserved ARG that is frequently detected in ICEs within BRD-associated bacteria. This real-time multiplex RPA assay targeted both ICE variants simultaneously, denoted as tetH&#95;tnpA and tetH&#95;ebrB, with a limit of detection (LOD) of 29 (95% CI [23, 46]) and 38 genome copies (95% CI [30, 59]), respectively. DNA was extracted from 100 deep nasopharyngeal swabs collected from feedlot cattle on arrival. Samples were tested for ICEs using a real-time multiplex RPA assay, and for M. haemolytica, P. multocida, H. somni, and Mycoplasma bovis using both culture methods and RPA. Cow-calf surveillance data provide critical information about changing herd demographics, productivity, health, and management as well as the opportunity to investigate the impact of differing management practices. A national cow-calf network was established to collect baseline information on herd health, production performance measures, and management. Questionnaires were used to collect information on herd attributes as well as data for the calving season, breeding, pregnancy testing, and weaning for herds from across Canada. From 2018 to 2022, a total of 565 calving record questionnaires, representing 110,658 calving female records from 171 herds were returned, along with 543 herd breeding to weaning questionnaires. Suggested benchmark values based on the 25th percentiles from Western and Eastern Canada were determined to be <5% for non-pregnancy, <2% for calf death from birth to 24 h, and < 2% for calf death from 24 h to weaning. Herds from Eastern Canada were more likely to report any assistance at calving ( p  < 0.001) than herds from Western Canada. Participating herds from the east had longer breeding ( p  < 0.001) and calving ( p  < 0.001) seasons than those from the west and were also more likely to use artificial insemination or embryo transfer ( p  < 0.001). Timing of calving season and use of large pastures for calving were not associated with mortality at birth. Herds that started calving before April were more likely to have calves die before 30 d than those that started calving later; however, this difference was not observed between 30 d and weaning. Herds that started to calve earlier were also more likely to report treating more calves for bovine respiratory disease, diarrhea, and navel or joint infections as well as to calf deaths from respiratory disease. Calves from herds in the east were more likely to be treated or die from diarrhea than from the west. Females from herds that started calving later were less likely to be pregnant. Objective: This study addressed the current gap in knowledge of neonatal prime-boost immune responses for the control of bovine coronavirus (BCoV) respiratory disease in weaning-age beef cattle., Animals: Study 1 and Study 2 had 33 and 22 commercial cross neonatal beef calves, respectively., Procedures: Study 1 compared BCoV-neutralizing antibody concentrations of control calves with 3 groups of calves differentially vaccinated with mucosal and/or systemic BCoV modified live virus (MLV) vaccines. Study 2 compared specific and neutralizing antibody concentrations among mucosally BCoV primed groups of calves that were differentially systemically boosted., Results: In Study 1, calves that were mucosally primed and systemically boosted had higher BCoV-neutralizing antibody concentrations than the control group at weaning. Liquid penetration in porous cellulosic materials is crucial in many technological fields. The complex geometry, small pore size, and often fast timescale of liquid uptake makes the process hard to capture. Effects such as swelling, vapor transport, film flow and water transport within cellulosic material makes transport deviate from well-known relations such as Lucas-Washburn and Darcy's Law. In this work it is demonstrated how Ultra-Fast Imaging NMR can be used to simultaneously monitor the liquid distribution and swelling during capillary uptake of water with a temporal- and spatial resolution of 10 ms and 14.5-18 μm respectively. The measurements show that in a cellulose fiber sheet, within the first 65 ms, liquid first penetrates the whole sheet before swelling takes place for another 30 s. Furthermore, it was observed that the liquid front traps 15 v% of air which is slowly replaced by water during the final stage of liquid uptake. Our method makes it possible to simultaneously quantify the concentration of all three phases (solid, liquid and air) within porous materials during processes exceeding 50 ms (5 times the temporal resolution). In the Northern Great Plains, cattle may be exposed to water with an elevated sulfate concentration resulting in ruminal hydrogen sulfide (H2S) production and risk of copper deficiency. There are currently few strategies available to help mitigate effects arising from high-sulfate water (HS). The objective of this study was to evaluate the effects of feeding a moderate-forage diet with or without bismuth subsalicylate (BSS; 0.0% vs. 0.4% DM basis) when provided water with a low- (LS; 346 ± 13) or HS (4,778 ± 263 mg/L) concentration on feed and water intake, ruminal H2S concentration, and liver and serum trace-mineral concentrations. Twenty-four Limousin × Simmental cross beef heifers (221 ± 41 kg) were stratified based on initial liver Cu into a completely randomized block design with a 2 × 2 factorial treatment arrangement. Feed and water intake (measured weekly), ruminal H2S concentration (measured on days 42 and 91), liver (measured on days -13 and 91), and serum trace-mineral concentrations (measured on days 1, 28, 56, and 91) were evaluated. Initial liver trace-mineral concentrations were used as a covariate in the statistical model. Water intake tended to be reduced with the inclusion of BSS (P = 0.095) but was not affected by water sulfate (P = 0.40). Water sulfate and BSS did not affect dry matter intake (DMI; P ≥ 0.89). Heifers consuming HS had a ruminal H2S concentration that was 1.58 mg/L more (P < 0.001) than LS. The inclusion of BSS reduced (P = 0.035) ruminal H2S concentration by more than 44% (1.35 vs. 0.75 mg/L). Regardless of the water sulfate concentration, heifers fed BSS had lesser liver Cu concentration (average of 4.08 mg/kg) than heifers not provided BSS, and when not provided BSS, HS had lesser Cu than LS (42.2 vs. 58.3; sulfate × BSS, P = 0.019). The serum concentration of Cu did not differ over time for heifers not provided BSS; whereas, heifers provided BSS had lesser serum Cu concentration on day 91 than on days 28 and 55 (BSS × time, P < 0.001). The liver concentration of selenium was reduced (P < 0.001) with BSS inclusion but the selenium concentration in serum was not affected by sulfate, BSS, or time (P ≥ 0.16). BSS reduced ruminal H2S concentration, but depleted liver Cu and Se. Background: The magnitude and knowledge gaps regarding antimicrobial use (AMU) and antimicrobial resistance (AMR) have not been summarized for the North American cow-calf production sector, although estimates of AMU and AMR are essential to AMR risk analysis. The objectives of this scoping review were to map AMU and AMR in the beef cow-calf sector in Canada and the United States, summarize published AMU/AMR predictors, and identify research gaps., Methods: An electronic search was conducted of four bibliographic databases and Google Scholar, augmented by a hand-search of captured studies., Results: Twenty-three of three-hundred and forty-three publications screened advanced to data extraction. Of these, 10 were conducted in the USA and 13 in Canada. Thirteen studied AMR and twelve studied AMU, with two reporting both. Of twelve captured AMU studies, nine presented counts of herd AMU by antimicrobial class or specific antimicrobial. Antimicrobial resistance in Escherichia coli ( E. coli ) was reported in nine studies. Risk factors for AMU include herd size, vaccine use, and start date of calving season., Conclusions: Overall, a small number of AMR studies were available for synthesis in primarily one population (cows) reporting E. coli AMR. Additional studies targeting reasons for AMU in calves, the impact of management procedures on AMU, potential environmental AMR sources, and AMR in respiratory pathogens and enteric organisms other than E. coli for pre-weaning calves are required to inform AMR risk mitigation strategies.

Trace mineral supplementation of beef cattle is essential for efficient reproduction and herd health. Understanding regional differences in cow trace mineral status could inform decisions about risks of deficiencies and supplementation management. Cow-calf surveillance projects provided three opportunities to evaluate the trace mineral status of Canadian beef cow herds. Blood samples were collected at pregnancy testing in 2014 from 102 cow-calf herds and in 2016 from 86 cow-calf herds in Western Canada. In 2019, blood samples were collected at pregnancy testing from cows in 163 cow-calf herds from Eastern and Western Canada. Serum samples were analyzed for copper, selenium, and molybdenum concentrations using a plasma mass spectrometer. The prevalence of copper deficient cows sampled from the Western provinces ranged from 24% to 43% across the three periods, and was 20% from Eastern Canada in 2019. The prevalence of selenium deficient cows ranged from 0.2% to 0.4% across the three projects in Western Canada, but was higher in Eastern Canada at 4.6% in 2019. High serum molybdenum was identified in 9.4% to 14% of cows across the three periods in Western Canada and in 15% of cows sampled in Eastern Canada in 2019. Serum copper, selenium, and molybdenum concentrations varied by cow age and month of sample collection. Serum selenium and molybdenum concentrations, but not copper, varied by soil type associated with the location of the farm. A subsample of samples from cows from Western Canadian herds provided body condition score ( BCS ) data, pregnancy status, and calf survival data and were used to estimate updated serum reference values for adequate concentrations. Age-specific values were required for selenium and molybdenum. Reference intervals (80%) were estimated from 2,406 pregnant beef cows from 99 herds with each cow having a BCS ≥ 2.5/5 and a live calf at 3 wk with no retained placenta: copper for all cows (0.379 to 0.717 ppm), selenium for cows <4 yr (0.052 to 0.152 ppm), and selenium for cows ≥4 yr (0.064 to 0.184 ppm). Upper 90% reference limits were also estimated for serum molybdenum for cows <4 yr (>0.104 ppm) and cows ≥4 yr (>0.110 ppm). Hypothesis: The widely used Lucas-Washburn (LW) equation depends on the contact angle as the driving force for liquid penetration. However, the contact angle depends on both, the liquid and the substrate. It would be desirable to predict the penetration into porous materials, without the requirement to measure the solid-liquid interaction. Here, we propose a novel modeling approach for liquid penetration from mutually independent substrate- and liquid properties. For this purpose, the contact angle in the LW-equation is replaced by polar and dispersive surface energies, utilizing the theories of Owens-Wendt-Rabel-Kaelble (OWRK), Wu, or van Oss, Good, Chaudhury (vOGC)., Experiments: The proposed modelling approach is validated exhaustively by measuring penetration speed for 96 substrate-liquid pairings and comparing the results to model predictions based on literature- and measured data., Findings: Liquid absorption is predicted very well (R 2  = 0.8-0.9) with all three approaches, spanning a wide range of penetration speed, substrate- and liquid surface energy, viscosity, and pore size. The models for liquid penetration without measurement of solid-liquid interaction (contact angle) performed well. Objective: This study compares immune and clinical responses of bovine viral diarrhea virus (BVDV)-maternal antibody (MatAb)-positive beef calves primed with intranasal modified-live virus vaccine (MLV) and differentially boosted with a systemic MLV or an inactivated vaccine (KV)., Animal: Eighteen commercial Black Angus steers., Procedure: Calves were mucosally primed at ~24 h of age with an MLV and boosted by injection of a MLV (IN-MLV) or inactivated vaccine (IN-KV) at an average age of 54 d. Objective: To determine the prevalence of Mycobacterium avium spp. paratuberculosis (MAP) in cow-calf herds located in the prairie provinces of Alberta, Saskatchewan, and Manitoba using a serum ELISA test., Animals: Study herds were recruited from the Western Canadian Cow-calf Surveillance Network (WC 3 SN) designed to monitor factors related to the health and productivity of cow-calf herds. Overall, 1791 cows from 92 herds were included in the study., Procedure: Blood samples were collected from 20 cows per herd in a systematic random fashion by private veterinarians in the fall of 2014. A serum ELISA (IDEXX, Westbrook, Maine, USA) test was used for the detection of MAP antibodies in the blood samples., Results: The cow level seroprevalence across all 3 provinces was 1.5%. Alberta had the lowest cow seroprevalence (1.3%) followed by Saskatchewan (1.7%), and Manitoba (2.1%). Herd level data showed that 24% of herds had at least 1 positive animal and 5% had at least 2 positive animals. Seroprevalence estimates varied between geographical regions within each province and with herd size., Conclusions: The apparent prevalence of MAP in prairie cow-calf herds remains low and similar to past estimates for the region. Johne's disease is an insidious infectious disease of ruminants caused by Mycobacterium avium subspecies paratuberculosis (MAP). Johne's disease can have important implications for animal welfare and risks causing economic losses in affected herds due to reduced productivity, premature culling and replacement, and veterinary costs. Despite the limited accuracy of diagnostic tools, testing and culling is the primary option for controlling Johne's disease in beef herds. However, evidence to inform specific test and cull strategies is lacking. In this study, a stochastic, continuous-time agent-based model was developed to investigate Johne's disease and potential control options in a typical western Canadian cow-calf herd. The objective of this study was to compare different testing and culling scenarios that included varying the testing method and frequency as well as the number and risk profile of animals targeted for testing using the model. The relative effectiveness of each testing scenario was determined by the simulated prevalence of cattle shedding MAP after a 10-year testing period. A second objective was to compare the direct testing costs of each scenario to identify least-cost options that are the most effective at reducing within-herd disease prevalence. Whole herd testing with individual PCR at frequencies of 6 or 12 months were the most effective options for reducing disease prevalence. Scenarios that were also effective at reducing prevalence but with the lowest total testing costs included testing the whole herd with individual PCR every 24 months and testing the whole herd with pooled PCR every 12 months. The most effective method with the lowest annual testing cost per unit of prevalence reduction was individual PCR on the whole herd every 24 months. Individual PCR testing only cows that had not already been tested 4 times also ranked well when considering both final estimated prevalence at 10 years and cost per unit of gain. Bovine respiratory disease (BRD), calf diarrhea (CD), and navel infections are the most commonly reported diseases of western Canadian beef calves. The objectives of this study were to estimate the frequency of treatment for these diseases for specific age cohorts and identify potential opportunities for reducing antimicrobial use. Producers representing 89 western Canadian cow-calf herds completed a survey describing calfhood diseases and management. The most common reason for calf treatment before weaning was BRD (4.9%), and BRD treatment was described in 51% of reporting herds before 2 months of age. Calf diarrhea (2.9%) and navel infection (2.0%) were the second and third most common reasons for treatment. Most calves were treated for CD between 6 days and 1 month of age. Almost one in five herds reported routinely administering antimicrobials at birth. Calving heifers and cows together were all associated with an increased treatment risk for BRD in calves from birth to 2 months (OR 3.55, 95%CI 2.13-5.94, P < 0.0001), CD from 1 month to weaning (OR 3.94, 95%CI 1.29-12.0, P = 0.02), and navel infection (OR 4.55, 95%CI 1.78-11.6, P = 0.002). Failure to sort cow-calf pairs out of the calving area was also associated with an increased treatment risk for BRD from 4 months to weaning (OR 4.89, 95%CI 1.96-12.2, P = 0.0006) and CD from 24 h to 5 days (OR 2.82, 95%CI 1.03-7.75, P = 0.04), and not using the Sandhills system was associated with an increased treatment risk for navel infection (OR 4.55, 95%CI 1.78-11.6, P = 0.002). Other potentially modifiable factors associated with an increased risk of BRD in calves from birth to 2 months were winter feeding and calving in one area ( P < 0.0001), heifers calving in a higher density area ( P = 0.01), and an increasing number of times cow-calf pairs were gathered before turn out to summer pasture ( P = 0.0005). The purchase of any cows during the calving or prebreeding period was associated with an increased risk of BRD from birth to 2 months ( P < 0.0001) and from 2 to 4 months ( P < 0.0001). A history of respiratory bacterin vaccines administered to the dams was associated with a decreased risk of BRD in calves from 4 months to weaning ( P = 0.01). Cows calving in a higher density area was associated with an increased risk of CD from 1 month to weaning ( P = 0.02). While Johne's disease (JD) is less common in beef than in dairy herds, consolidation is increasing transmission risk. Estimates of Mycobacterium avium spp. paratuberculosis (MAP) prevalence and test performance in cow-calf herds are needed to inform control programs. Objectives of this study included describing the prevalence of MAP in Canadian cow-calf herds and comparing the relative performance of a serum ELISA, pooled fecal PCR and individual fecal PCR using Bayesian latent class models, and to investigate factors associated with positive MAP tests. Blood and fecal samples ( n = 3,171) were collected from 159 Canadian cow-calf herds. All samples were analyzed using serum ELISA and fecal PCR (pools of five samples) and a subset of 913 fecal samples were also tested with individual PCR. Based on latent class analysis, MAP prevalence was higher in eastern compared to western Canada for both animals {East, 3% [95% Credible Interval (CrI) 1-7%]; West, 1% [95% CrI 0.2-2%]} and herds [East, 15% (95% CrI 2-35%); West, 10% (95% CrI 1-26%), based on one or more positive results]. Sensitivity (Se) and specificity (Sp) for animal level individual PCR were 96% (95% CrI 80-100%) and 98% (95% CrI 96-100%), respectively followed by pooled PCR [Se = 54% (95% CrI 36-72%), Sp > 99.9% (95% CrI 99.8-100%)] and ELISA [Se = 36% (95% CrI 22-52%), Sp = 98% (95% CrI 96-99%)]. Based on 20 samples per herd, the herd level Se of ELISA was 79% (95% CrI 47-100%) (at least one positive sample) compared to 43% (95% CrI 14-94%) for pooled PCR. Herd-level Sp was 99% (95% CrI 96-100%) for pooled PCR and 90% (95% CrI 83-100%) for ELISA. Cows from herds with dairy cattle on farm and cows with symptoms of JD in the past 3 years were more likely to be MAP positive. Herds that had animals with JD symptoms in the previous 3 years and those with more breeding females were most likely to test positive for MAP. While serum ELISA can be effective for herd screening, PCR performed better for animal testing. Background: Bovine respiratory disease (BRD) is an important cause of morbidity and mortality and is responsible for most of the injectable antimicrobial use in the feedlot industry. Traditional bacterial culture can be used to diagnose BRD by confirming the presence of causative pathogens and to support antimicrobial selection. However, given that bacterial culture takes up to a week and early intervention is critical for treatment success, culture has limited utility for informing rapid therapeutic decision-making. In contrast, metagenomic sequencing has the potential to quickly resolve all nucleic acid in a sample, including pathogen biomarkers and antimicrobial resistance genes. In particular, third-generation Oxford Nanopore Technology sequencing platforms provide long reads and access to raw sequencing data in real-time as it is produced, thereby reducing the time from sample collection to diagnostic answer. The purpose of this study was to compare the performance of nanopore metagenomic sequencing to traditional culture and sensitivity methods as applied to nasopharyngeal samples from segregated groups of chronically ill feedlot cattle, previously treated with antimicrobials for nonresponsive pneumonia or lameness., Results: BRD pathogens were isolated from most samples and a variety of different resistance profiles were observed across isolates. The sequencing data indicated the samples were dominated by Moraxella bovoculi, Mannheimia haemolytica, Mycoplasma dispar, and Pasteurella multocida, and included a wide range of antimicrobial resistance genes (ARGs), encoding resistance for up to seven classes of antimicrobials. Genes conferring resistance to beta-lactams were the most commonly detected, while the tetH gene was detected in the most samples overall. Metagenomic sequencing detected the BRD pathogens of interest more often than did culture, but there was limited concordance between phenotypic resistance to antimicrobials and the presence of relevant ARGs., Conclusions: Metagenomic sequencing can reduce the time from sampling to results, detect pathogens missed by bacterial culture, and identify genetically encoded determinants of resistance. Computed tomographic (CT) features of lymph nodes are used for diagnosis, staging, and response evaluation in veterinary patients. The objectives of this study were to describe maximum short- and long-axis transverse diameters (SATD and LATD, respectively), volume, and attenuation of presumed normal canine mandibular lymph nodes (MLNs) and medial retropharyngeal lymph nodes (MRLNs), to investigate the effect of variables on these features, and to describe inter-observer reliability of transverse diameter and attenuation measurement. Computed tomography studies of 161 dogs were retrospectively reviewed. Median values for SATD were 5.2 and 5.4 mm, median values for volume were 0.26 and 0.54 cm 3 , and median values for attenuation were 32.3 and 31.8 Hounsfield units (HU) for MLN and MRLN, respectively. Median LATD was 11 mm for MLNs. Height of the first cervical vertebra (HCV1) and weight were positively associated with transverse diameters of both MLNs and MRLNs ( P < 0.001). Grade of dental disease was negatively associated with transverse diameters of both MLNs ( P = 0.001) and MRLNs ( P < 0.03). The intraclass correlation coefficient (ICC) was 0.94 [95% confidence interval (CI): 0.94 to 0.95] for transverse diameter measurement. Mandibular and medial retropharyngeal lymph nodes are routinely evaluated with CT when staging dogs with oral melanomas. While size alone is considered inadequate for detecting nodal metastasis, it is critical in evaluating treatment response, as clinical decisions are based on changes in size. It is common for different radiologists to measure the size of pre- and posttreatment lymph nodes in the same patient. The objective of this retrospective, observer agreement study was to evaluate the inter- and intraobserver agreement in measuring canine mandibular and medial retropharyngeal lymph nodes by a diverse population of veterinary radiologists and trainees. Fourteen dogs with documented oral melanoma and head CT studies identified from records of a single institution were included in this study. North American veterinary radiologists and trainees were recruited to measure the mandibular and medial retropharyngeal lymph nodes; in triplicate. Prior to performing the study measurements, participants completed a training tool demonstrating the lymph node measurements. Overall, interobserver intraclass correlation coefficient (ICC) was 0.961 (95% confidence interval [CI]: 0.946, 0.972) and intraobserver ICC was 0.977 (95% CI: 0.968, 0.983), indicating excellent agreement (ICC > 0.9 considered excellent). Similar findings were noted following sub-analysis for most variables (experience, size, laterality, axis of measurement). These results suggest that follow-up measurement of the long and short axis of the mandibular lymph nodes and short axis of the medial retropharyngeal lymph nodes in the transverse plane, performed by different veterinary radiologists using the same method of measure, should have minimal impact on clinical decision making., (© 2021 American College of Veterinary Radiology.)

Hydrophobization of cellulosic materials and particularly paper products is a commonly used procedure to render papers more resistant to water and moisture. Here, we explore the hydrophobization of unsized paper sheets via the gas phase. We employed three different compounds, namely palmitoyl chloride (PCl), trifluoroacetic anhydride/acetic anhydride (TFAA/Ac 2 O)) and hexamethyldisilazane (HMDS) which were vaporized and allowed to react with the paper sheets via the gas phase. All routes yielded hydrophobic papers with static water contact angles far above 90° and indicated the formation of covalent bonds. The PCl and TFAA approach negatively impacted the mechanical and optical properties of the paper leading to a decrease in tensile strength and yellowing of the sheets. The HMDS modified papers did not exhibit any differences regarding relevant paper technological parameters (mechanical properties, optical properties, porosity) compared to the non-modified sheets. XPS studies revealed that the HMDS modified samples have a rather low silicon content, pointing at the formation of submonolayers of trimethylsilyl groups on the fiber surfaces in the paper network. This was further investigated by penetration dynamic analysis using ultrasonication, which revealed that the whole fiber network has been homogeneously modified with the silyl groups and not only the very outer surface as for the PCl and the TFAA modified papers. Bovine respiratory disease (BRD) is an economically important cause of morbidity and mortality in beef calves. Control of BRD is most often addressed through "homologous" vaccination utilizing the same injectable modified-live viral (MLV) vaccine for both priming and boosting. Heterologous prime-boosting uses different routes and antigenic forms for priming and boosting. Three vaccine protocols were compared: an injectable (IJ) MLV (IJ-MLV) group (IJ-MLV priming at ~48 days and boosted with IJ-MLV at weaning), intranasal (IN) MLV (IN-MLV) group (intranasal priming with MLV at ~24 hours, boosted twice with an IJ-MLV), and intranasal killed viral (IN-KV) group (primed with an IN-MLV at ~24 hours, boosted twice with an IJ-KV). Serum antibody concentrations determined by enzyme-linked immunosorbent assays (ELISAs) were compared and the IN-KV group had significantly higher BRSV-specific antibody concentrations after boosting compared with the 2 homologous groups. Data generated from Statistics Canada's 2016 Census of Agriculture and Census of Population were used to describe the adoption of 8 technologies by the Canadian dairy industry: computer/laptop, smartphone/tablet, auto-steering, auto-feeding, auto-environment, robotic milking, global positioning systems (GPS), and geographical information systems (GIS). Logistic regression was used to analyze the adoption of each technology by geographical region, operators' gender, operators' age, herd size, and number of operators per farm. Gender and age were marginally related to the level of adoption of each technology, whereas the number of operators per dairy farm and farm size were associated with increased adoption of most technologies. Bovine viral diarrhea virus (BVDV) is an important pathogen causally associated with morbidity and mortality, and production losses in both suckling and weaned beef calves. Vaccination for protection against disease caused by BVDV is challenging because of the inhibitory effect of maternal antibodies; however, it is most convenient for beef producers to vaccinate calves before moving herds to summer pastures. We compared modified live and inactivated vaccines used for priming and boosting beef calves and found that neither type of vaccine results in strong antibody responses in the face of maternal antibodies. Ultrasonic liquid penetration (ULP) measurements of porous sheets have been applied for a variety of purposes ranging from determining liquid absorption dynamics to surface characterization of substrates. Interpretation of ULP results, however, is complex as the ultrasound signal can be affected by several mechanisms: (1) air being replaced by the liquid in the substrate pores, (2) air bubbles forming during penetration, and (3) structural changes of the substrate due to swelling of the substrate material. Analyzing tailored liquids and substrates in combination with contact angle measurements we are demonstrating that the characteristic shape of the ULP measurement curves can be interpreted in terms of the regime of liquid uptake. A fast and direct decline of the curve corresponds to capillary penetration, the slope of the curve indicates the penetration speed. A slow decline after a previous maximum in the signal can be related to diffusive liquid transport and swelling of the substrate material.

The use of metaphylactic antimicrobials to control bovine respiratory disease (BRD) is under increased scrutiny; therefore, effective immunization is becoming a priority. Stimulation of an effective immune response is challenging because calves are often primed with injectable (IJ) vaccines, in the face of maternal antibodies which can interfere with the immune response. Maternal antibody interference may be bypassed with use of intranasal (IN) vaccines. Between 2011 and 2016, the number of dairy operations in Canada decreased by 13.3%. Mean herd size increased from 65 to 73 animals per farm. The ratio of older (> 60 years old) to younger (< 31 years old) producers also increased. Vaccination against putative causal organisms is a frequently used and preferred approach to controlling bovine respiratory disease complex (BRD) because it reduces the need for antibiotic use. Because approximately 90% of feedlots use and 90% of beef cattle receive vaccines in the USA, information about their comparative efficacy would be useful for selecting a vaccine. We conducted a systematic review and network meta-analysis of studies assessing the comparative efficacy of vaccines to control BRD when administered to beef cattle at or near their arrival at the feedlot. We searched MEDLINE, MEDLINE In-Process, MEDLINE Daily Epub Ahead of Print, AGRICOLA, Cambridge Agricultural and Biological Index, Science Citation Index, and Conference Proceedings Citation Index - Science and hand-searched the conference proceedings of the American Association of Bovine Practitioners and World Buiatrics Congress. We found 53 studies that reported BRD morbidity within 45 days of feedlot arrival. The largest connected network of studies, which involved 17 vaccine protocols from 14 studies, was included in the meta-analysis. Consistent with previous reviews, we found little compelling evidence that vaccines used at or near arrival at the feedlot reduce the incidence of BRD diagnosis.

We conducted a systematic review and network meta-analysis to determine the comparative efficacy of antibiotics used to control bovine respiratory disease (BRD) in beef cattle on feedlots. The information sources for the review were: MEDLINE®, MEDLINE In-Process and MEDLINE® Daily, AGRICOLA, Epub Ahead of Print, Cambridge Agricultural and Biological Index, Science Citation Index, Conference Proceedings Citation Index - Science, the Proceedings of the American Association of Bovine Practitioners, World Buiatrics Conference, and the United States Food and Drug Administration Freedom of Information New Animal Drug Applications summaries. The eligible population was weaned beef cattle raised in intensive systems. The interventions of interest were injectable antibiotics used at the time the cattle arrived at the feedlot. The outcome of interest was the diagnosis of BRD within 45 days of arrival at the feedlot. The network meta-analysis included data from 46 studies and 167 study arms identified in the review. The results suggest that macrolides are the most effective antibiotics for the reduction of BRD incidence. Injectable oxytetracycline effectively controlled BRD compared with no antibiotics; however, it was less effective than macrolide treatment. Because oxytetracycline is already commonly used to prevent, control, and treat BRD in groups of feedlot cattle, the use of injectable oxytetracycline for BRD control might have advantages from an antibiotic stewardship perspective.

Objectives: To explore tuberculosis (TB) incidence in Canada and the United States from 1953 to 2015. In the most recent decade, the US incidence was lower than that of Canada. Since both countries are high income and have low TB incidence with similar TB surveillance programs, we hypothesized that rates should be similar., Methods: TB incidence data from 1953 to 2015 were retrieved for both countries. Joinpoint regression was performed to identify change points in the trend, and direct standardization of US rates using Canadian ethnic population distribution was calculated. Adjusted rate and average annual percent change (AAPC) were estimated., Results: Canada rates/100,000 were higher from 1953 to 1974 and similar from 1975 to 1985. This coincided with a change in US case definition in 1975. US rates were higher from 1986 to 1996. HIV/TB coinfection in the USA was 10.2% compared to that of Canada, 1.6%. Rates were similar from 1997 to 2004. Canada rates were again higher from 2005 to 2015. The Canada average AAPC rate in 1975-2015 was lower, - 2.9%, compared to that of the USA, - 4.1%. Foreign-born and Indigenous population proportions were 20.2% and 4.2% for Canada and 12.9% and 1.7% for the USA. The US rate adjusted to the Canada ethnic composition was 4.8 compared to the Canadian rate of 4.7., Conclusion: Case definition change and HIV coinfection contributed to the 1980 US rate increase. TB rates decreased in both countries from 1997, but more rapidly in the USA. The Canada proportion of foreign-born and Indigenous populations was higher. When US rates were standardized by Canada ethnic distribution, the national rates were similar. Further exploration of factors contributing to differences between these countries is needed.

Rural communities dependent on unregulated drinking water are potentially at increased health risk from exposure to contaminants. Perception of drinking water safety influences water consumption, exposure, and health risk. A community-based participatory approach and probabilistic Bayesian methods were applied to integrate risk perception in a holistic human health risk assessment. Tap water arsenic concentrations and risk perception data were collected from two Saskatchewan communities. Drinking water health standards were exceeded in 67% (51/76) of households in Rural Municipality #184 (RM184) and 56% (25/45) in Beardy's and Okemasis First Nation (BOFN). There was no association between the presence of a health exceedance and risk perception. Households in RM184 or with an annual income >$50,000 were most likely to have in-house water treatment. The probability of consuming tap water perceived as safe (92%) or not safe (0%) suggested that households in RM184 were unlikely to drink water perceived as not safe. The probability of drinking tap water perceived as safe (77%) or as not safe (11%) suggested households in BOFN contradicted their perception and consumed water perceived as unsafe. Integration of risk perception lowered the adult incremental lifetime cancer risk by 3% to 1.3 × 10 -5 (95% CI 8.4 × 10 -8 to 9.0 × 10 -5 ) for RM184 and by 8.9 × 10 -6 (95% CI 2.2 × 10 -7 to 5.9 × 10 -5 ) for BOFN. Highlights: Comparably low accuracy of dietary recall is present after 7 and 18 days. Experiencing gastrointestinal symptoms did not affect accuracy of dietary recall. Repeatedly recalling food exposures leads to a higher rate of false positives. Prevalence of exposure was associated with higher sensitivity and lower specificity. Recall inaccuracies can be quantified and accounted for in public health practice.

Exosomes are 60-150 nm small extracellular vesicles (EVs) released by most cells. Tumor-cell-derived exosomes, used as a vaccine, elicit a specific cytotoxic response against tumor cells, usually with a greater immunogenicity than tumor-cell lysates. However, the number of exosomes isolated from culture cells is limited. In recent studies, it was observed that cells respond to different stressor stimuli such as cytotoxic drugs, hypoxia, acidosis, or radiation by increasing the release of EVs. In this study, using the murine LBC T-cell lymphoma, we found that cyclophosphamide significantly increased EVs yield. These EVs express exosome marker proteins such as TSG-101, CD9, CD81, and CD63. Furthermore, similar humoral and cellular immune responses were induced in vivo by EVs isolated from LBC-tumor cells whether they were grown under normal culture conditions (EVs C) or in the presence of cyclophosphamide (EVs CTX). Mice vaccinated either with EVs C or EVs CTX were similarly protected against an intraperitoneal challenge with LBC tumor cells. CD4+ and CD8+ IFN-γ secreting cells were induced in immunized mice and a specific cytotoxic cellular immune response was elicited in vitro. These results demonstrate that a Th1 response was induced by immunization with the EVs. Our findings suggest that treatment of tumor cells with cyclophosphamide is a useful method to enhance the secretion of EVs in sensitive cell lines without altering their antitumor properties and thus may be used to produce antigens for future design of cancer vaccines., (Copyright © 2019 Elsevier Ltd. Statistics Canada's 2016 census data were analyzed to determine the proportion of Canadian cow-calf producers who had adopted the use of 7 different technologies and 2 different grazing/feeding management practices, collectively referred to as "management tools." The 4 most commonly used management tools were rotational grazing, in-field winter grazing/feeding, smartphones/tablets, and computers/laptops. Differences in the adoption of these technologies by geographical region, number of producers/operations, herd size, operator gender, and operator age were examined using logistic regression. Estimates of the mean proportion of producers in eastern (65%) and western (60%) Canada using rotational grazing were similar ( P = 0.24). However, a greater proportion of producers in western Canada versus eastern Canada were using in-field winter grazing/feeding ( P < 0.001), smartphones/tablets ( P < 0.001), and computers/laptops ( P = 0.002). Adoption of all 4 tools was higher on farm operations with ≥ 2 operators versus those with 1 operator ( P < 0.001). Larger herd size was associated with higher adoption rates across all 4 management tools. The effect of gender on adoption rates was equivocal.

A commonly used therapeutic strategy for type 2 diabetes mellitus (DM) in humans involves the use of synthetic incretin hormone-based therapies including exenatide, a glucagon-like pepetide-1 hormone agonist. Glucagon-like pepetide-1 agonists can be used alone or as an ancillary therapy with other agents, including insulin and oral antihyperglycemics. Little is known about the role of these therapies for DM in cats. Therefore, the primary objective of this study was to evaluate the safety and efficacy of short-acting exenatide combined with insulin, as compared to placebo and insulin for the treatment of DM in cats. Treatment with exenatide was well tolerated; only 2 cats developed side effects requiring dose reduction. Two cats (25%) went into diabetic remission while receiving exenatide and insulin, whereas remission was not reported during placebo treatment. The average change in the daily exogenous insulin dose was significant (β = -0.56 U/kg, 95% confidence interval, -0.96 to -0.15, P = 0.007), and the dose of insulin administered was lower during exenatide treatment. The average weight loss experienced on exenatide was significantly higher than on placebo (β = 0.65 kg, 95% confidence interval, 0.09-1.21, P = 0.02). There was no significant difference in any of the hormone concentrations evaluated for cats on exenatide vs placebo treatments. Overall, the treatment of diabetic cats with insulin and a fixed dose of exenatide was found to be safe. The weight loss and decreased exogenous insulin requirement experienced with exenatide treatment could be a significant benefit for overweight diabetic cats and warrants further evaluation., (Copyright © 2018 Elsevier Inc. Four gauges (14G, 16G, 18G, 20G) of single use hypodermic needles were evaluated for sharpness by measuring the force required to puncture rehydrated bovine leather. The needles began to dull after 1 use with maximum bluntness occurring within 4 to 5 uses.

From 2011 to 2016, both the number of cow-calf producers and farm operations in Canada decreased by 12.4%. Furthermore, the current demographics portend a similar or greater loss in the number of producers and farms by the next census (2021). This level of consolidation has implications with respect to veterinary education and how practitioners will service the cow-calf sector.

Emergence and dissemination of antimicrobial resistance (AMR) in food-borne bacteria is a public health issue. Retail meat is considered an important carrier for antimicrobial-resistant Escherichia coli. Currently, resistance of E. coli strains to third generation cephalosporins are of particular concern, with significant potential animal and public health consequences. Resistance to tetracyclines is also a concern, due to high prevalence of this resistance and important co-resistance patterns. However, the actual likelihood of exposure to antimicrobial-resistant bacteria via the consumption of retail meats, beyond the simple frequencies of resistance found in samples of meat at the grocery store, in Canada remains to be investigated. This study was conducted to estimate the potential human exposure to ceftriaxone- and tetracycline-resistant E.coli (CREC and TREC; the hazards of interest) through the consumption of ground beef in Western Canada. Our exposure assessment simulation model included five modules: 1) estimation of prevalence and concentration of the hazards of interest in retail ground beef samples collected by the Canadian Integrated Program for Antimicrobial Resistance Surveillance (CIPARS), 2011-2014; 2) potential growth of the hazards during transportation from retail to home; 3) potential growth or reduction of the hazards during home storage; 4) thermal inactivation of the hazards due to cooking; and 5) consumption. The outputs of the model were presented as the probability of consumers' exposure to various threshold levels of the hazards [10 to 10 6 colony forming units (CFU)] in one serving of cooked ground beef. Overall, the probabilities of exposure to high threshold levels of the hazards (>4 log 10  CFU/serving) were estimated to be lower than 0.12% in the study population (2011-2014). The mean probabilities of exposure to at least 1 log 10 CFU CREC and TREC in a single meal containing cooked ground beef were 0.067% (SD: 0.001%) and 1.20% (SD: 0.02%), respectively. This probability substantially decreased as the threshold level of exposure increased to ≥6 log 10 CFU. The probability of exposure to TREC was consistently greater than that for CREC. Cooking led to a prominent drop in the mean concentration of the hazards (4.7 log 10  CFU/g). The findings from this research could inform the policy-making process and provide suggestions for adjustments in future retail surveillance plans. Objective: To evaluate the incidence of hypochloremic metabolic alkalosis (HCMA) in dogs and cats in the ICU that had intermittent nasogastric tube (NGT) aspiration for up to 36 hours., Design: Prospective cohort study (December 2013 to October 2014)., Setting: Privately owned emergency and referral teaching hospital., Animals: Forty-nine client-owned dogs and 16 client-owned cats., Interventions: Patients wherein NGT placement was recommended and client consent was obtained were included in the interventional group. Those with an NGT placed (NGT group) had the NGT aspirated every 4 hours. Patients for whom placement of a NGT was declined by the owner served as a reference group (NoNGT). Venous blood gas and electrolyte values were obtained every 12 hours., Measurements and Main Results: Thirty-five dogs and cats had an NGT placed. Thirty dogs and cats did not have an NGT placed. The serum venous blood gas and electrolyte changes were compared over time within the NGT group and between the NGT and NoNGT groups. No cases developed HCMA. In the NGT group, blood pH increased over time. There was no significant difference between the NGT and the NoNGT group in the average value of pH, HCO 3 - , base excess, chloride, or corrected chloride. Serum venous blood gas, chloride, and corrected chloride changes were not associated with the volumes of gastric fluid aspirated over time., Conclusions: In this small population of dogs and cats, intermittent NGT aspiration was not associated with the development of HCMA over a period of up to 36 hours after NGT placement., (© Veterinary Emergency and Critical Care Society 2018.)

This study determined whether mineral concentrations in the hooves of cattle with toe tip necrosis syndrome (cases) differed from those of cattle dying of all other causes (controls). Samples were collected over a 2-year period from 16 different feedlots and analyzed for 8 minerals [cobalt (Co), copper (Cu), iron (Fe), magnesium (Mg), manganese (Mn), molybdenum (Mo), selenium (Se), and zinc (Zn)]. Mineral concentrations in the hoof wall and solar horn of the same hoof were poorly correlated; Se was the most correlated (ρ = 0.865; P < 0.001), while Mg (ρ = 0.465; P < 0.001) and Zn (ρ = 0.157; P = 0.053) were the least correlated. The cases had significantly lower Mg concentrations in both the hoof wall and solar horn tissue compared to the control subjects. For every 10 ppm decrease in Mg, the odds of a diagnosis of toe tip necrosis syndrome (TTNS) increased by 1.13 times for the hoof wall ( P = 0.002) and 1.21 times for solar horn ( P < 0.001).

Objective: The objective of this study was to examine the association between area level deprivation and vehicle collisions resulting in either property damage or injury., Methods: A multilevel observational study was conducted using the 2000 to 2010 Saskatchewan Traffic Accident Information System (TAIS) (n = 72,234) and 2006 Census data at the Dissemination Area level (n = 337) for the city of Saskatoon., Results: Total area level deprivation was associated with severity of traffic collisions, but the association varied based on time of day and road repair status. Collisions were more likely to result in injury from the most deprived (Q5) versus the least deprived quintile (Q1) at all times of day; the difference was greatest in the evening (OR 1.7, 95% CI 1.3 to 2.3). However, there was no other evidence of a monotonic increase in risk associated with area level deprivation. When there were faded markings or potholes, the odds of a collision involving injury were 2.6 (95% CI 1.5 to 4.4) times greater for the most deprived quintile compared to the least deprived quintile. There were no significant differences in the risk of injury between area level deprivation quintiles when road conditions were good., Conclusion: While the association between area level deprivation and whether vehicle collisions result in injury in Saskatoon varies based on time of day and road repairs, under many circumstances the most deprived areas report more injuries from collisions compared to the least deprived.

The objective of this study was to provide contemporary data on the epidemiology of gastrointestinal nematode parasite infections of grazing yearling beef cattle in the province of Saskatchewan. Fecal samples ( n = 1290) were collected over 4 time periods during the summer grazing season from 21 separately managed groups of cattle. Fecal egg counts (FEC) were estimated using generalized estimating equations with a negative binomial distribution with log link function, adjusting for clustering of samples within each herd for each time period. Nematodirus spp. and Trichuris spp. eggs were enumerated separately and were detected in 5.7% (73/1290) and 1.7% (22/1290) of samples, respectively. One or more strongyle-type eggs were detected in 79.5% (1025/1290) of the samples and FEC increased by 2.8 times over the summer grazing season. Interestingly, FEC were ~3.4 times higher on pastures located in dark brown versus brown soil zones, a finding that warrants further investigation.

A blinded randomized controlled trial was used to evaluate a multi-modal therapeutic regime for treatment of beef bulls infected with Campylobacter fetus subsp. venerealis (Cfv) . Treatment included 2 doses of a commercially available monovalent vaccine and long-acting oxytetracycline applied twice at a 2-week interval with treatment completed 2 weeks before post-treatment observation. Fifteen confirmed Cfv infected bulls were randomly allocated to control ( n = 8) or treatment groups ( n = 7). Preputial scrapings were collected each week from before infection to 11 weeks following the last treatment. When the polymerase chain reaction (PCR) results for both culture and preputial scrapings were interpreted in parallel, there were no significant differences between treated and untreated bulls. Regardless of the type of diagnostic testing considered, treatment with 2 label doses of this regime did not stop shedding of Cfv in all treated bulls and is, therefore, not recommended as an effective management strategy.

Background: While a new generation of computational statistics algorithms and availability of data streams raises the potential for recurrently regrounding dynamic models with incoming observations, the effectiveness of such arrangements can be highly subject to specifics of the configuration (e.g., frequency of sampling and representation of behaviour change), and there has been little attempt to identify effective configurations., Methods: Combining dynamic models with particle filtering, we explored a solution focusing on creating quickly formulated models regrounded automatically and recurrently as new data becomes available. Given a latent underlying case count, we assumed that observed incident case counts followed a negative binomial distribution. In accordance with the condensation algorithm, each such observation led to updating of particle weights. We evaluated the effectiveness of various particle filtering configurations against each other and against an approach without particle filtering according to the accuracy of the model in predicting future prevalence, given data to a certain point and a norm-based discrepancy metric. We examined the effectiveness of particle filtering under varying times between observations, negative binomial dispersion parameters, and rates with which the contact rate could evolve., Results: We observed that more frequent observations of empirical data yielded super-linearly improved accuracy in model predictions. We further found that for the data studied here, the most favourable assumptions to make regarding the parameters associated with the negative binomial distribution and changes in contact rate were robust across observation frequency and the observation point in the outbreak., Conclusion: Combining dynamic models with particle filtering can perform well in projecting future evolution of an outbreak. Most importantly, the remarkable improvements in predictive accuracy resulting from more frequent sampling suggest that investments to achieve efficient reporting mechanisms may be more than paid back by improved planning capacity. The robustness of the results on particle filter configuration in this case study suggests that it may be possible to formulate effective standard guidelines and regularized approaches for such techniques in particular epidemiological contexts. Most importantly, the work tentatively suggests potential for health decision makers to secure strong guidance when anticipating outbreak evolution for emerging infectious diseases by combining even very rough models with particle filtering method.

Safe drinking water is a global challenge for rural populations dependent on unregulated water. A scoping review of research on human health risk assessments (HHRA) applied to this vulnerable population may be used to improve assessments applied by government and researchers. This review aims to summarize and describe the characteristics of HHRA methods, publications, and current literature gaps of HHRA studies on rural populations dependent on unregulated or unspecified drinking water. Peer-reviewed literature was systematically searched (January 2000 to May 2014) and identified at least one drinking water source as unregulated (21%) or unspecified (79%) in 100 studies. Only 7% of reviewed studies identified a rural community dependent on unregulated drinking water. Source water and hazards most frequently cited included groundwater (67%) and chemical water hazards (82%). Most HHRAs (86%) applied deterministic methods with 14% reporting probabilistic and stochastic methods. Publications increased over time with 57% set in Asia, and 47% of studies identified at least one literature gap in the areas of research, risk management, and community exposure. HHRAs applied to rural populations dependent on unregulated water are poorly represented in the literature even though almost half of the global population is rural., Competing Interests: The authors have no actual or potential competing financial interests.

Background: Saskatchewan has one of the highest rates of gonorrhea among the Canadian provinces-more than double the national rate. In light of these high rates, and the growing threat of untreatable infections, improved understanding of gonorrhea transmission dynamics in the province and evaluation of the current system and tools for disease control are important., Methods: We extracted data from a cross-sectional sample of laboratory-confirmed gonorrhea cases between 2003 and 2012 from the notifiable disease files of the Regina Qu'Appelle Health Region. The database was stratified by calendar year, and social network analysis combined with statistical modeling was used to identify associations between measures of connection within the network and the odds of repeat gonorrhea and risk of coinfection with chlamydia at the time of diagnosis., Results: Networks were highly fragmented. Younger age and component size were positively associated with being coinfected with chlamydia. Being coinfected, reporting sex trade involvement, and component size were all positively associated with repeat infection., Conclusions: This is the first study to apply social network analysis to gonorrhea transmission in Saskatchewan and contributes important information about the relationship of network connections to gonorrhea/chlamydia coinfection and repeat gonorrhea. This study also suggests several areas for change of systems-related factors that could greatly increase understanding of social networks and enhance the potential for bacterial sexually transmitted infection control in Saskatchewan.

Avian influenza virus (AIV) occurrence and transmission remain important wildlife and human health issues in much of the world, including in North America. Through Canada's Inter-Agency Wild Bird Influenza Survey, close to 20,000 apparently healthy, wild dabbling ducks (of seven species) were tested for AIV between 2005 and 2011. We used these data to identify and evaluate ecological and demographic correlates of infection with low pathogenic AIVs in wild dabbling ducks (Anas spp.) across Canada. Generalized linear mixed effects model analyses revealed that risk of AIV infection was higher in hatch-year birds compared to adults, and was positively associated with a high proportion of hatch-year birds in the population. Males were more likely to be infected than females in British Columbia and in Eastern Provinces of Canada, but more complex relationships among age and sex cohorts were found in the Prairie Provinces. A species effect was apparent in Eastern Canada and British Columbia, where teal (A. discors and/or A. carolinensis) were less likely to be infected than mallards (A. platyrhynchos). Risk of AIV infection increased with the density of the breeding population, in both Eastern Canada and the Prairie Provinces, and lower temperatures preceding sampling were associated with a higher probability of AIV infection in Eastern Canada. Our results provide new insights into the ecological and demographic factors associated with AIV infection in waterfowl.

Background: Circulating l-lactate concentration is commonly measured in hospitalized horses by sampling from indwelling intravenous (IV) catheters. However, there are no published evidence-based recommendations to prevent contamination by lactated Ringer's solution (LRS)., Hypothesis: Withdrawing 10 mL of blood from the LRS-containing extension set connected to the IV catheter before obtaining the sample for analysis should be adequate to obtain accurate measurement of blood lactate concentration (BLC)., Animals: Thirty-three adult hospitalized horses receiving constant rate infusion of LRS., Methods: Immediately after disconnecting the LRS, 5 sequential 5 mL blood samples were obtained by aspiration from an extension set connected to an indwelling IV catheter, followed by 3 samples collected by direct venipuncture of the contralateral jugular vein. Samples were analyzed with 1 portable blood lactate analyzer. A linear mixed model was used to examine differences in lactate concentrations among samples collected from the catheter and by direct venipuncture., Results: After considering differences in age, breed, sex, and reason for hospitalization, BLCs were higher (P < .001) in the first and second 5 mL samples collected through the extension set/catheter than in all other extension set/catheter samples or the direct venipuncture samples. The largest difference observed between the third and subsequent catheter or venipuncture samples was 0.34 mmol/L with an upper 95% CI of 1.12 mmol/L., Conclusions and Clinical Importance: Withdrawing 15 mL of blood from a LRS-containing extension set connected to an IV catheter (5.9 mL total volume capacity) before obtaining the sample for blood lactate analysis is suggested to optimize accuracy of BLC measurements., (Copyright © 2016 The Authors.
