Xylose is the raw material for the synthesis of many important platform compounds. At present, xylose is commercially produced by chemical extraction. However, there are still some bottlenecks in the extraction of xylose, including complicated operation processes and the chemical substances introduced, leading to the high cost of xylose and of synthesizing the downstream compounds of xylose. The current market price of xylose is 8× that of glucose, so using low-cost glucose as the substrate to produce the downstream compounds of xylose can theoretically reduce the cost by 70%. Here, we designed a pathway for the biosynthesis of xylose from glucose in Escherichia coli . This biosynthetic pathway was achieved by overexpressing five genes, namely, zwf , pgl , gnd , rpe , and xylA , while replacing the native xylulose kinase gene xylB with araL from B. subtilis , which displays phosphatase activity toward d-xylulose 5-phosphate. The yield of xylose was increased to 3.3 g/L by optimizing the metabolic pathway. Furthermore, xylitol was successfully synthesized by introducing the xyl1 gene, which suggested that the biosynthetic pathway of xylose from glucose is universally applicable for the synthesis of xylose downstream compounds. This is the first study to synthesize xylose and its downstream compounds by using glucose as a substrate, which not only reduces the cost of raw materials, but also alleviates carbon catabolite repression (CCR), providing a new idea for the synthesis of downstream compounds of xylose.

Background: The sugar sensing and carbon catabolite repression in Baker's yeast Saccharomyces cerevisiae is governed by three major signaling pathways that connect carbon source recognition with transcriptional regulation. Here we present a screening method based on a non-invasive in vivo reporter system for real-time, single-cell screening of the sugar signaling state in S. cerevisiae in response to changing carbon conditions, with a main focus on the response to glucose and xylose., Results: The artificial reporter system was constructed by coupling a green fluorescent protein gene (yEGFP3) downstream of endogenous yeast promoters from the Snf3p/Rgt2p, SNF1/Mig1p and cAMP/PKA signaling pathways: HXT1p/2p/4p; SUC2p, CAT8p; TPS1p/2p and TEF4p respectively. A panel of eight biosensors strains was generated by single copy chromosomal integration of the different constructs in a W303-derived strain. The signaling biosensors were validated for their functionality with flow cytometry by comparing the fluorescence intensity (FI) response in the presence of high or nearly depleted glucose to the known induction/repression conditions of the eight different promoters. The FI signal correlated with the known patterns of the selected promoters while maintaining a non-invasive property on the cellular phenotype, as was demonstrated in terms of growth, metabolites and enzyme activity., Conclusions: Once verified, the sensors were used to evaluate the signaling response to varying conditions of extracellular glucose, glycerol and xylose by screening in 96-well microtiter plates. We show that these yeast strains, which do not harbor any recombinant pathways for xylose utilization, are lacking a signaling response for extracellular xylose. However, for the HXT2p/4p sensors, a shift in the flow cytometry population dynamics indicated that internalized xylose does affect the signaling. These results suggest that the previously observed effects of this pentose on the S. cerevisiae physiology and gene regulation can be attributed to xylose and not only to a lack of glucose.

To release sugars effectively from sweet sorghum bagasse (SSB), a cellulose solvent and organic solvent-based lignocellulose fractionation pretreatment approach was studied using response surface methodology (RSM). Based on RSM's central composite design, a batch experimental matrix was set up to determine the effects of reaction time (20-60 min) and temperature (40-60 °C) on delignification, total reducing sugar yield, glucan digestibility, and overall glucose yields following a pretreatment-hydrolysis process. The optimum pretreatment conditions of 50 °C and 40 min led to 51.4% delignification, 86% overall glucose yield, and 61% overall xylose yield. An effort has also been made to obtain predictive models to illustrate the correlation between independent and dependent variables using RSM. The significance of the correlations and adequacy of these models were statistically tested for the selected objective functions. The optimum pretreatment condition predicted by the model was 49.1 °C and 39.2 min which matched the experimental data well. Wheat straw was pretreated and fermented to ethanol. Two strains, which had been mutated from the genetically modified Saccharomyces cerevisiae TMB3400, KE6-12 and KE6-13i, have been used in this study and the results of performance were compared to that of the original strain. The glucose and xylose co-fermentation ability was investigated in batch fermentation of steam-pretreated wheat straw (SPWS) liquid (undiluted, and diluted 1.5 and 2 times). Both strains showed improved xylose uptake in diluted SPWS liquid, and increased ethanol yields compared with the original TMB3400 strain, although xylitol formation also increased slightly. In undiluted SPWS liquid, however, only KE6-13i performed better than the original strain regarding xylose utilization. Fed-batch fermentation of 1.5 and 2 times diluted liquid was performed by adding the glucose-rich hydrolysates from enzymatic hydrolysis of the solid fraction of SPWS at a constant feed rate after 5 h of fermentation, when the glucose had been depleted. The modified strains showed improved xylose conversion; however, the ethanol yield was not significantly improved due to increased glycerol production. Lignocellulose and other carbohydrates are being studied extensively as potential renewable carbon sources for liquid biofuels and other valuable chemicals. In the present study, a simple, sensitive, selective, and reliable HPLC method using a photodiode array (PDA) detector and an evaporative light scattering detector (ELSD) was developed for the simultaneous determination of important sugars (D(+)-cellobiose, glucose, xylose, and arabinose), furfural and 5-hydroxymethylfurfural (5-HMF) in lignocellulose hydrolysate. The analysis was carried out on an Aminex HPX-87H column (250 mm × 4.6 mm, 5 μm particle size). Ultra-pure water with 0.00035 M H(2)SO(4) was used as the mobile phase with a flow rate of 0.6 mL/min. The temperature of the ELSD drift tube was kept at 50 °C, the carrier gas pressure was 350 kPa, and the gain was set at 7. Furfural and 5-HMF were quantified on a PDA detector at 275 nm and 284 nm, respectively. The sugar concentrations were determined by ELSD. This method was validated for accuracy and precision. The regression equation revealed a good linear relationship (r(2) = 0.9986 ± 0.0012) within the test ranges. The method showed good reproducibility for the quantification of six analytes in corncob hydrolysate, with intra- and inter-day variations less than 1.12%. Fourier transform infrared, attenuated total reflectance (FTIR-ATR) spectroscopy combined with partial least squares (PLS) regression accurately predicted 72-h glucose and xylose conversions (g sugars/100 g potential sugars) and yields (g sugars/100 g dry solids) from cellulase-mediated hydrolysis of alkali-pretreated lignocellulose. Six plant biomasses that represent a variety of potential biofuel feedstocks--two switchgrass cultivars, big bluestem grass, a low-impact, high-diversity mixture of 32 species of prairie biomasses, mixed hardwood, and corn stover--were subjected to four levels of low-temperature NaOH pretreatment to produce 24 samples with a wide range of potential digestibility. PLS models were constructed by correlating FTIR spectra of pretreated samples to measured values of gluose and xylose conversions and yields. Variable selection, based on 90% confidence intervals of regression-coefficient matrices, improved the predictive ability of the models, while simplifying them considerably. Final models predicted sugar conversions with coefficient of determination for cross-validation (Q(2)) values of 0.90 for glucose and 0.89 for xylose, and sugar yields with Q(2) values of 0.92 for glucose and 0.91 for xylose. The sugar-yield models are noteworthy for their ability to predict enzymatic saccharification per mass dry solids without a priori knowledge of the composition of the solids. All peaks retained in the final regression coefficient matrices were previously assigned to chemical bonds and functional groups in lignocellulose, demonstrating that the models were based on real chemical information. Water extract of steam-exploded corn stalk (SECS) was detoxified and used as feed for acetone-butanol-ethanol (ABE) fermentation using Clostridium beijerinckii. Utilization of water extract improved the total ABE yield (g ABE/g dry SECS). Separated fermentation showed higher fermentability (0.078 g ABE/g dry SECS) over typical fermentation (0.058 g ABE/g dry SECS). Furthermore, the final ABE yields (g ABE/g utilized sugar) from water extract neutralized by Ca(OH)(2), NaOH, and Na(2)SO(3) were 0.16, 0.1 and 0.07, respectively, suggesting that Ca(OH)(2) had the best detoxification effect.

The effect of corn stover pretreatment on glucose quantitation in hydrolysate using Raman spectroscopy is evaluated. Dilute sulfuric-acid pretreatment results in a 20 mg mL(-1) glucose limit of detection in hydrolysate. Soaking in aqueous ammonia pretreatment produces a 4 mg mL(-1) limit of detection. Water, ethanol or hexane extraction of corn stover reduces the spectral background that limits glucose detection in dilute acid hydrolysate. Additionally, a Raman spectroscopy multi-peak fitting method is presented to simultaneously measure glucose and xylose concentration in hydrolysate. This method yields a 6.1% average relative standard error at total saccharide concentrations above 45 mg mL(-1). When only cellulase is present, glucose and xylose yield were measured by Raman spectroscopy to be 32 ± 4 and 7.0 ± 0.8 mg mL(-1), respectively. When both cellulase and hemicellulase were present, xylose yield increased to 18.0 ± 0.5 mg mL(-1). A room temperature operated pulsed external-cavity (EC) quantum cascade laser (QCL) was used for mid-infrared (mid-IR) transmission measurements of glucose and lactate in aqueous solution. The high spectral power density of the EC-QCL (ranging from 1-350 mW) over a wide tuning range (1030-1230 cm(-1)) allowed transmission measurements through optical paths of 130 μm and more. This is a significant improvement in terms of robustness of the measurement setup, especially when samples containing cells or other particles, as is the case for biofluids, are to be analyzed. The broad tuning range furthermore permitted multi-analyte detection based on multivariate calibrations. Promising results on the simultaneous determination of glucose (c = 0-800 mg dL(-1)) and sodium-lactate (c = 0-224 mg dL(-1)) in aqueous solutions in the presence of the interferents maltose and xylose are reported. A partial least squares (PLS) calibration model was calculated which was able to predict the glucose concentration with a root mean square error of prediction (RMSEP) of 9.4 mg dL(-1), as proved by external validation. Due to their small size and room temperature operation, EC-QCLs offer an attractive alternative regarding the way mid-IR measurements are carried out. This may be of special importance for new reagent-free bedside monitoring systems.

This work presents the pretreatment of oil palm fronds (OPF) using hot compressed water (HCW) to enhance sugar recovery in enzymatic hydrolysis. A central, composite rotatable design was used to optimize the effect of reaction temperature, reaction time and liquid-solid ratio on the pretreatment process. All variables were found to significantly affect the glucose yield. A quadratic polynomial equation was used to model glucose yield by multiple regression analysis, using response surface methodology (RSM). Using a 10 bar pressurized reactor, the optimum conditions for pretreatment of OPF were found at 178 degrees C, 11.1 min and a liquid-solid ratio of 9.6. The predicted glucose yield was 92.78 wt.% at the optimum conditions. An ammonia pressurization/depressurization process was investigated to evaluate the potential of producing reducing sugars from dwarf elephant grass, a warm-season forage. Moisture, temperature, and ammonia loading affected sugar yield (p < 0.0001). At optimal conditions, ammonia processing solubilized 50.9% of the hemicellulose and raised the sugar yield (percentage of theoretical) from 18 to 83%. Glucose and xylose production were increased 3.2- and 8.2-fold, respectively. The mild processing conditions of the ammonia treatment (90-100 degrees C, 5 min), the low enzyme loading (2 international filter paper units/g), and the short hydrolysis time (24 h), greatly enhance the potential of using forages to produce sugars valuable for several applications.

An enzyme electrode was constructed for amperometric determination of xylose and glucose. The electrode is based on the PQQ-dependent membrane-bound aldose dehydrogenase (ALDH) from Gluconobacter oxydans. ALDH was covalently immobilized on a graphite electrode. Immobilized dimethylferrocene, soluble ferrocene carboxylic acid and phenazine methosulphate were used as electron transfer mediators. When xylose was measured electrochemically using an electrode modified with ALDH and dimethylferrocene, the linear measurement range extended to 100 mM. For glucose measurement the linear measurement range was about one-tenth of that for xylose. The electrode showed fairly good stability; 50% of the original electrode response was still obtained after 5 days of intermittent use. The effect of possible leakage of adsorbed mediator was determined by measuring the response of an electrode with soluble mediator as a function of time. The reproducibility of the electrode was good, the standard deviation of the electrode response in ten measurements with the same electrode being only 2.7%.

1. The structures of the alpha(2)- and alpha(3)-azopigments, prepared by diazotization of dog bile with ethyl anthranilate, were shown by mass spectrometry and g.l.c. to correspond to azobilirubin beta-d-xylopyranoside and azobilirubin beta-d-glucopyranoside respectively. 2. Both azopigments consist of a mixture of two methyl vinyl isomers having structures (IIIa) and (IIIb) for the alpha(2)-azopigment and structures (IVa) and (IVb) for the alpha(3)-azopigment. Separation of methyl vinyl isomers was obtained by t.l.c. or column chromatography performed on the acetylated azopigments. Hydrolysis of the less polar acetates derived from components (IIIa) and (IVa) gave rise to the azopigment (Ia), whereas hydrolysis of the more polar acetates derived from components (IIIb) and (IVb) gave rise to the azopigment acid (Ib). The positions of methyl and vinyl substituents in compounds (Ia) and (Ib) were assigned on the basis of their n.m.r. spectra. 3. Molecular ions in the mass spectra of the trimethylsilyl and acetyl derivatives of the azopigments indicated the presence of a pentose and a hexose conjugating sugar. 4. The ester functions linking the sugars to the propionic acid side chain of azobilirubin were demonstrated by ammonolysis and identification of the amide of azobilirubin as the aglycone derivative. 5. The sugar moieties were shown to occur as xylopyranose (alpha(2)) and glucopyranose (alpha(3)), bound at C-1, by application of a sequence of reactions performed on a micro-scale. The sugar hydroxyl groups were acetylated and the 1-acyl aglycone removed selectively by treatment with hydrogen bromide in acetic acid. Hydrolysis of the 1-bromo sugar acetates followed by acetylation afforded the alpha- and beta-xylopyranose tetra-acetates and alpha- and beta-glucopyranose penta-acetates, identified by a combination of g.l.c. and mass spectrometry. 6. The validity of this degradation scheme was confirmed (a) by g.l.c.-mass spectrometry identification of the alpha- and beta-1-propionyl derivatives of glucopyranose tetra-acetate, obtained from the alpha(3)-azopigment after final reaction with propionic anhydride; (b) by subjecting the acetates of alphabeta-glucopyranose, alphabeta-xylofuranose and alphabeta-glucofuranose to the same sequence of reactions.
