Abstract

A number of pathways have been identified involving glucose transporter expression and insulin mediation in L6 muscle cells. The enzymes 70-kDa ribosomal protein S6 kinase and p38 mitogen-activated protein kinase (MAPK), unlike the enzymes extracellular signal-regulated protein kinase (EPK) and MAPK/ERK kinase, affect glucose transporter expression (GLUT). No such mechanism has been identified for 2,4-dinitrophenol.

Abstract

Protein kinase Cs (cPKCs) translocate to plasma membranes and display 200% increase in kinase activity when the L6 skeletal muscle cells are treated with acute 2,4-dinitrophenol (DNP). The DNP-simulated glucose uptake was partially inhibited by the overnight treatment of 4-phorbol 12-myristate 13-acetate, but the treatment has no effect on the insulin-simulated uptake. Moreover, the selective PKC-beta inhibitor LY-379196 also partially inhibits DNP effects on glucose uptake.
