The aim of the study was to determine the morphological and morphometric features of structural changes in the components of the exocrine part of the pancreas of male rats after withdrawal of the food additive sodium glutamate and their transfer to a normal diet. Materials and methods. 4 series of experiments were conducted. The 1st series – male rats were given 70 mg/kg of sodium glutamate daily for 8 weeks to enhance their taste; the 2nd series – rats were transferred to a standard diet 8 weeks after the use of monosodium glutamate and withdrawn from the experiment after 8 weeks (at week 16); the 3rd series – control group rats up to 8 weeks, the 4th series – control rats up to 16 weeks of the experiment. The pancreas was examined morphologically with morphometric analysis after the rats were withdrawn from the experiment, the material was paraffin-embedded and sections were made, stained with hematoxylin and eosin and azan. Results. In the 1st and 2nd series of the experiment, atrophic and degenerative changes of the acini were preserved, which were discomposed due to edema of the organ, and had small sizes. Connective tissue and areas of adipose tissue, diffuse and focal infiltrates were visualized between the lobules and around the lobular ducts. The ducts were dilated. As in the 1st series, there was edema of the organ parenchyma, vessel walls and ducts with fluid leakage into the intercalated spaces, and exocrinocyte apoptosis. The morphometric study of the exocrine part of the pancreas revealed that the average size of acini was statistically significantly reduced in the 1st and 2nd experimental series by 1.4 and 1.6 times, and the area by 1.2 and 1.5 times compared to control animals of the 3rd and 4th series. In the 1st and 2nd series, acini ranging in size from 71 μm to 90 μm prevailed, accounting for 38 % and 34 %, respectively, in the 1st and 2nd control series – 91–100 μm (46 % and 42 %) and from 111 μm to 149 μm (36 % and 38 %), respectively. In the study of the number of cells in acini in the 1st series, exocrinocytes were present in the amount of 5.52 ± 0.58 vs. control 8.05 ± 0.32 (p < 0.001), in the 2nd series – 5.24 ± 0.47 vs. control 7.33 ± 0.43 (p < 0.001) The area of exocrinocytes in the 2nd series 61.95 ± 1.91 μm2 vs. 78.99 ± 0.98 μm2 (p < 0.05) were reduced. The diameter of the insertional, intra-lobular and inter-lobular ducts was enlarged. Conclusions. After 8 weeks of feeding rats with monosodium glutamate and after its withdrawal and transfer of rats to the standard diet of the rat vivarium in the next 8 weeks, no improvement in the state of the pancreas was recorded, which was confirmed by morphological and morphometric studies.

Abstract Monosodium glutamate (MSG) is used as a common food additive in some foods. However, based on our search and knowledge, no comprehensive study discussed the effect of MSG on the human gut microbiome. In this study, the effects of MSG on the gut microbiome, liver, and kidney were performed. Data were collected from databases including PubMed, Scopus, Web of Science, and ScienceDirect using the search strategy and keywords. Finally, 14 eligible studies were selected for systematic review. This study provides a new perspective on the effects of MSG on the gut flora, shedding light on the potential relationship between MSG intake and human health.

Background: This study examined administration of MSG intraperitoneally to white rats resulted in a decrease in testicular weight, an increase in lipid peroxidase levels The testes, as the site of spermatogenesis, are highly vulnerable to oxidation processes caused by free radicals, which can disrupt spermatogenesis. Free radicals can be countered by antioxidants. One plant believed to have antioxidant activity is red ginger. Purpose: To determine the effect of red ginger against histopathology seminiferous tubules of white rat (Rattus norvegicus) exposed by monosodium glutamate. Methods: This study used 30 rats which were divided into 5 groups. The positve control group (K+) were given only monosodium glutamate 5g/kgBW and CMC Na orally, the treatments groups P1 were given monosodium glutamte 5g/KgBW + red ginger 50mg/rat/day, P2 were given 5g/KgBW + red ginger 100mg/rat/day, and P3 were given 5g/KgBW + red ginger 200mg/rat/day. All treatments were administered for 42 days. After 42 days of treatments, all rats were sacrificed and seminiferous tubules were observed with microscope from pathology laboratory. Results: Administration of red ginger extract can defend seminiferous tubules epithelial thicknes and diameter from exposure of monosodium glutamate with 100mg/KgBW dosage as the most effective dosage. Seminiferous tubules epitelial thickness and diameter were tested using one-way ANOVA and Duncan’s multiple range test. Conclusion: Red Ginger Extract (Zingiber officinale var rubrum) has shown potential effects to counteract Monosodium Glutamat consequences on The Thickness of the epitelial and diameter of seminiferous tubules of white rat (Rattus norvegicus).

Monosodium glutamate (MSG) mediates body weight gain (BWG) and oxidative stress. Azithromycin (AZT), may be abused and co-consumed with MSG to present unknown outcomes on BWG and oxidative stress. This study evaluated the effect of AZT and MSG in rats’ BWG and antioxidant bioindicators. Thirty rats assigned to five groups were orally exposed for seven consecutive days to groups A, control (distilled water, 1 ml/kg), B, MSG (MSG 8000 mg/kg), C, therapeutic AZT, TAZ (AZT 82.5 mg/kg), D, overdose AZT, OAZ (AZT 412.5 mg/kg) and E, TAZ + MSG (AZT 82.5 mg/kg + MSG 8000 mg/kg). MSG-treated rats exhibited a significantly (p < 0.05) increased BWG; serum, liver, kidney and heart reduced glutathione (GSH), glutathione peroxidase (GPX), superoxide dismutase (SOD), and malondialdehyde (MDA) but decreased catalase (CAT) and zinc (Zn) levels compared to control. Co-treated TAZ + MSG rats significantly (p < 0.05) decreased BWG, GSH, GPX, SOD, Zn; increased CAT and non-significantly (p > 0.05) decreased MDA compared to MSG and control. Thus, TAZ significantly mitigated BWG, and malfunction in the metabolism of antioxidant defense bioindicators in MSG rats via probable anorexigenic, anti-inflammatory and antioxidant responses. This suggests that TAZ could be useful in managing MSG-related dysfunction in BWG and metabolic activity of the antioxidant defense apparatus in rats.

Abstract Background Monosodium glutamate (MSG E621) is one of the most popular flavouring agents of modern times and is widely used in many commercially packed food and even in house hold cooking. Previous studies revealed that excessive intake of MSG in diet causes obesity, metabolic defects, nephrotoxicity, neurotoxicity and hepatotoxicity in rats, but no reports are available in the literature about the ecotoxicological assessment of MSG by using fishes as a bioindicators. Since fishes are important consumer in aquatic food chain and directly linked with human health status, the present study was aimed to investigate the impact of MSG in freshwater fish Labeo rohita by using histological biomarkers. Results Ninety-six h-LC50 of MSG to Labeo rohita was determined (1.5 g/L), and fish exposed to sub-lethal concentration of MSG (1/10th of 96 h-LC50 concentration of MSG (150 mg/L)) showed distinguished behavioural changes like erratic movement, loss of appetite and excessive mucous secretion all over the body as an adaptive syndrome to avoid the direct exposure to MSG in the medium. Histopathological analysis clearly depicts severe damages in the vital organs of fish. In gills, epithelial necrosis, hypertrophy, hyperplasia, primary and secondary gill lamellae degeneration, oedema, fusion of adjacent secondary lamellae and rupture of gill epithelium were observed. The intensity of tissue damage was increased as the exposure period was extended. The liver displayed vein congestion, vacuole formation, degeneration in parenchymal cells and bile stagnation, whereas MSG-treated kidney tissue showed high interstitial inflammation. Among the vital organs gill and liver displayed the highest histopathological alterations. Conclusions The present study clearly demonstrated that MSG is toxic to fish and able to cause significant damages in the vital organs as the exposure period was extended. Since the studies on the toxicity of MSG to fish are rare, the present investigation may contribute to the scarce literature on sub-lethal toxicity of MSG to freshwater fishes.

Type I allergic reactions to MSG have not previously been reported. Our patient, a 21-year-old woman, was 14 years old when she first noticed swelling of her tongue (but no oral itching, diarrhea, or abdominal pain) after eating various snack foods. Current skin prick testing elicited a weak positive reaction to MSG. We then performed an oral challenge test during which our patient ingested potato snacks. Subsequent histology showed telangiectasia of the buccal mucosa, interstitial edema in the subepithelial submucosa, and mast cell infiltration. Oral mucosal challenge tests using sodium glutamate confirmed oral swelling in this patient. This report is the first to confirm a case of type 1 allergy to MSG by combining pathology findings with the results of challenge testing.Keywords: food additive, mast cell, monosodium glutamate, food allergy

Abstract Background Komagataella phaffii (a.k.a. Pichia pastoris) harbors a glutamate utilization pathway in which synthesis of glutamate dehydrogenase 2 and phosphoenolpyruvate carboxykinase (PEPCK) is induced by glutamate. Glutamate-inducible synthesis of these enzymes is regulated by Rtg1p, a cytosolic, basic helix-loop-helix protein. Here, we report food-grade monosodium glutamate (MSG)-inducible recombinant protein production from K. phaffii PEPCK promoter (P PEPCK ) using green fluorescent protein (GFP) and receptor binding domain of SARS-CoV-2 virus (RBD) as model proteins. Results P PEPCK -RBD/GFP expression cassette was integrated at two different sites in the genome to improve recombinant protein yield from P PEPCK . The traditional, methanol-inducible alcohol oxidase 1 promoter (P AOX1 ) was used as the benchmark. Initial studies carried out with MSG as the inducer resulted in low recombinant protein yield. A new strategy employing MSG/ethanol mixed feeding improved biomass generation as well as recombinant protein yield. Cell density of 100–120 A600 units/ml was achieved after 72 h of induction in shake flask cultivations, resulting in recombinant protein yield from P PEPCK that is comparable or even higher than that from P AOX1 . Conclusions We have designed an induction medium for recombinant protein production from K. phaffii P PEPCK in shake flask cultivations. It consists of 1.0% yeast extract, 2.0% peptone, 0.17% yeast nitrogen base with ammonium sulfate, 100 mM potassium phosphate (pH 6.0), 0.4 mg/L biotin, 2.0% MSG, and 2% ethanol. Substitution of ammonium sulphate with 0.5% urea is optional. Carbon source was replenished every 24 h during 72 h induction period. Under these conditions, GFP and RBD yields from P PEPCK equaled and even surpassed those from P AOX1 . Compared to the traditional methanol-inducible expression system, the inducers of glutamate-inducible expression system are non-toxic and their metabolism does not generate toxic metabolites such as formaldehyde and hydrogen peroxide. This study sets the stage for MSG-inducible, industrial scale recombinant protein production from K. phaffii P PEPCK in bioreactors.

This study aimed to investigate the effects of MSG consumption on receptor gene expression and quantification of hormones and receptors, as well as oxidative stress biomarkers and other lactogenic parameters in lactating animals. Twenty-four female Wistar rats, nine weeks of age, were randomly assigned to four groups, each containing six rats, at parturition. The rats in groups II, III, and IV were given varying doses of monosodium glutamate (MSG); while, group I was given distilled water and served as the control. Abstract Background Hepatocellular carcinoma (HCC) is the most common type of primary liver cancer, occurring mostly in individuals with chronic liver disease, but biomarkers for therapeutic diagnosis and prognosis are lacking. This study aimed to investigate the possible effect of the common food additive monosodium glutamate (MSG) and tannic acid (TA), a phenolic compound, on the key molecular actors responsible for HCC development. Methods Eight HCC‐related public microarray datasets (GSE84005, GSE14520, GSE25097, GSE57958, GSE22058, GSE84402, GSE54238, and GSE36376) were extracted from the gene expression omnibus (GEO) database and analyzed to identify differentially expressed genes (DEGs). To make sense of the identified biological data and to identify hub genes, protein–protein interaction (PPI) network and enrichment analysis were performed. The mRNA expression profiles of the identified hub genes, expression changes in different stages of HCC, and their prognostic significances in HCC were determined using GEPIA, UALCAN, and Kaplan–Meier Plotter databases, respectively. Finally, mRNA expression changes of identified hub genes in the liver tissues of rats treated with MSG and TA were measured by the quantitative real‐time PCR (qPCR) method. Results Two up‐regulated (AURKA and CCNB2) and two down‐regulated (F9 and CYP2E1) genes were identified between the HCC tumor and adjacent non‐tumor liver tissue samples. qPCR results showed that the mRNA expression of up‐regulated DEGs involved in HCC development increased significantly in rat liver tissues exposed to MSG, while this increase was remarkably suppressed by TA treatment. It was observed that the mRNA expressions of down‐regulated DEGs involved in HCC development decreased markedly in the presence of MSG, while this decrease was alleviated with TA. Conclusion Our results provide new insights into pivotal molecular candidates that should be focused on in future in vivo and in vitro HCC research. Moreover, MSG may play a crucial role in HCC development and progression and TA may be used as a favorable restorative agent in HCC.

The aim of this study is to histologically assess the effect of maternal administration of monosodium glutamate on placental development in Wistar rat using appropriate standard methods after establishing pregnancy in animals with regular cyclicity by pairing them overnight with sexually active males in the ratio 2:1 and placentae harvested for histological studies. Histological studies of the placenta when treated with MSG showed there was varying alterations in the histomorphology of the placental ranging from degenerative changes in glycogen cell island in junctional zone of (gestational day )GD 15, mild congestion of sinusoid in labyrinth zone of GD 17, dilated and congested fetal capillary of labyrinth zone in GD 19, vacuolar degeneration of glycogen cell island in junctional zone of GD 17, dilated and congested spiral artery in junctional zone of GD19, dilated sinusoid in labyrinth zone of GD 19. In conclusion, this study shows that there is evidence of placental toxicity following maternal consumption of monosodium glutamate in Wistar rats. Further studies are recommended to assess the mechanism of MSG-induced placental toxicity, as well the effects of MSG-induced placental toxicity on the overall development of the fetus.

Background & objectives: Pink salt and monosodium glutamate (MSG) are two typical food additives used in cooking to enhance flavour. However, excessive use of them has been associated to a variety of metabolic problems, including weight gain and hyperglycemia. The current study aimed to assess the metabolic changes caused by submaximal dosages of MSG and pink salt in experimental rats. Methods: Twenty-four 120–150 g Wister rats of both sexes were divided into three groups: control, pink salt-treated (0.8 g/kg daily for three weeks), and MSG-treated (3.6 g/kg daily for three weeks). The body weight, amount of food and water consumed, and blood glucose levels of animals were measured and recorded as indicators of their metabolic changes. This study was conducted to investigate the protective effects of Apis dorsata honey on the histopathological changes of the kidney in mice exposed to monosodium glutamate. This study used 25 male mice as the experimental animal which were divided into five groups with five replication, respectively i.e. (C-) was administered basal diet, (C+) was administered 4 mg/g BW MSG, (T1) was administered A. dorsata honey of 53.82 mg/20g BW followed by MSG 4 mg/g BW, (T2) was administered A. dorsata honey of 107.64 mg/20g BW followed by MSG 4 mg/g BW, and (T3) was administered A. dorsata honey of 161.46 g/20g BW followed by MSG 4 mg/g BW. All treatments were carried out orally for 52 days. This study was observed tubular epithelial hydropic degeneration, epithelial necrosis, and glomerular necrosis. Data were analyzed using Kruskal-Wallis test followed by the Mann-Whitney test. As a result, for the epithelial and glomerular necrosis variables, the T3 group showed significant results when compared with C+, T1, and T2 groups. Moreover, T3 was not significantly different from the C- group. It can be concluded that A. dorsata honey in T3 group could protect mice kidneys from the damaging effect of MSG.

Monosodium glutamate (MSG), a sodium salt derived from glutamic acid, is widely used in commercial food products to improve taste, quality, and preservation. However, its consumption may have detrimental effects on male reproductive function. Nevertheless, plant extracts, such as Lepidium meyenii (Maca), Trigonella foenum-graecum (Fenugreek), Spirulina platensis (Spirulina), and Tribulus arabica (Tribulus), may ameliorate these adverse effects. To this effect, the phytochemical properties of Lepidium meyenii, Trigonella foenum-graecum, Spirulina platensis, and Tribulus arabica were assessed, and their potential impact on MSG-induced impairment of reproductive parameters was examined. The phytochemical composition (steroids, terpenes, phenols, flavonoids) of the plants was profiled through spectrophotometry and the antioxidant activity was assessed using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay. Thirty-six male Wistar rats were divided into six groups at random: a control group receiving distilled water, and five experimental groups (MSG, Maca, Fenugreek, Spirulina, and Tribulus) receiving 900 mg/kg/day of MSG dissolved in water for 45 days. Subsequently, the animals in the experimental groups were administered 500 mg/kg/day of the respective plant extract via oral gavage for an additional 35 days, while the MSG group continued to receive water only. Following the treatment period, the animals were sacrificed, and their reproductive tract organs were collected, weighed, and subjected to further analysis. Phytochemical analysis revealed the presence of diverse bioactive elements in the plant extracts, including phenolic and flavonoid compounds. Exposure to MSG negatively impacted total and progressive sperm motility, which was ameliorated by Lepidium meyenii treatment. Sperm morphology showed no significant differences among groups. Treatment of the phytochemical agents diminished histomorphometric alternations of the testicular length, germinal epithelium height, and number of cells in seminiferous tubules, which were caused by the initial administration of MSG. Testosterone and LH levels were reduced in the MSG group but improved in extract-treated groups. The study suggests Lepidium meyenii as a potential remedy for reproductive dysfunction. However, further investigation into its mechanisms and human safety and efficacy is warranted.

Oxide copper minerals are commonly extracted via acidic leaching, using acids such as H2SO4, HCl, or HNO3. These strong acids are the most widely used because of their high dissolution kinetics. However, their main concern is the high acid consumption because copper oxide deposits contain large amounts of acid-consuming gangue. This paper proposes using an alternative aqueous alkaline monosodium glutamate (MSG) system to leach copper oxide minerals. Tenorite (CuO) was used as the copper oxide mineral under study. The influence of process variables (such as temperature and glutamate concentration) and kinetics of this system on copper leaching from tenorite were studied. The results showed that temperature has a significant effect on copper dissolution rates. Increased temperature from 15 °C to 60 °C enhanced the copper extraction from 9.1% to 97.7% after 2 h. Leaching kinetics were analyzed using the shrinking core model (SCM) under various conditions, indicating that the leaching rate presented a mixed control. This method, however, fails to describe leaching for broad particle sizes due to its requirement for single-sized solid grains. This study demonstrated that a large particle size distribution in tenorite supported a successful extension of the SCM for leaching it from mixed glutamate solutions. The activation energy for the 15–60 °C temperature range was calculated to be 102.6 kJ/mol for the chemical control.

This study was conducted to investigate the protective effects of Betanin active ingredient in red beetroot plant (Beta vulgaris) in elderly rats exposed to chronic toxicity of monosodium glutamate (MSG). A total of 48 elderly rats were randomly divided into 4 different groups. At the end of the 28–day study, the rats were sacrificed under deep anesthesia. Total antioxidant capacity (TAC), total oxidant capacity (TOC), paraoxonase (PON), thiol, malondialdehyde (MDA), and nitric oxide (NO) levels were investigated in rat blood serum using the spectrophotometric method. Oxidative Stress Index (OSI) was calculated by dividing TOC by TAC. Total bilirubin was measured with the colorimetric method using an ELISA kit. Liver tissues were stained with hematoxylin–eosin (HE) for histopathological examination. The difference in serum levels of TAC, TOC, OSI, PON, MDA, and thiol was statistically significant between the groups (P0.05). The analysis of histopathological findings revealed uncommon mild hydropic degeneration in the MSG group and almost normal histological appearance in the MSG+Betanin group. This study demonstrated that betanin could increase the antioxidant effect and reduce the histopathological damage caused by MSG.

The immune system plays a vital role in controlling liver fibrosis and enhancing the pathogenesis of liver inflammation. Monosodium glutamate is a common flavor-enhancement food additive. This study evaluated the immunomodulatory and hepato-curative effects of the Immuno-Kachiks polyherbal formulation against monosodium glutamate-induced immune suppression and hepatic damage in rats. Monosodium glutamate was given orally at a 2000 mg/kg dose to male Wistar rats for three months to induce liver damage and immune suppression. After three months of successful induction, three groups were separately administered orally with Immuno-Kachiks formula at 400, 800, and 1500 mg/kg/day for 28 days. At the end of the treatment period, liver and blood samples were collected for histological and biochemical analysis. The lymphocyte count remained significantly low while the neutrophil count and the neutrophil-to-lymphocyte ratio increased significantly, despite the cessation of monosodium glutamate ingestion for 28 days. The Immuno-Kachiks formula (IKF) significantly increased the lymphocyte count, reduced the neutrophil-to-lymphocyte ratio, and normalized the neutrophil count. Neither monosodium glutamate nor the IKF significantly caused alpha-fetoprotein levels to rise or fall below normal. High doses (800 and 1500 mg/kg) of the Immuno-Kachiks formula significantly raised serum levels of aspartate aminotransferase, alkaline phosphatase, and total bilirubin. 1500 mg/kg of the IKF caused mild liver inflammation. The IKF restored the liver morphologic alterations observed in monosodium glutamate-induced liver damage in rats. The results suggest that the Immuno-Kachiks herbal formulation is a potential curative agent for early-stage liver damage and could restore suppressed adaptive immunity.

Excessive neuronal excitation by glutamate is a well-established cause of neurotoxicity, leading to severe impairment of brain function. Excitotoxicity is a key factor in numerous neurodegenerative conditions. In this study, we investigated the neuroprotective effects of Danshensu (DSS) against monosodium glutamate (MSG)-induced neurotoxicity in adult mice and their offspring. We randomly divided one hundred 8-week-old Kunming mice (equal number of males and females) into a control group and an experimental group. The experimental group was further subdivided into various treatment groups, including MSG gavage treatment, bwbw DSS treatment group 1 (bwbw DSS treatment group 2, a drug control group, and a normal control group (receiving an equal volume of physiological saline for ten consecutive days). Additionally, another one hundred healthy 8-week-old Kunming mice were similarly divided into groups and treated. These mice were paired randomly (one male and one female) and pregnant females were housed separately to obtain offspring. Subsequently, we conducted histological and behavioral analyses on adult mice and their offspring. MSG treatment induced significant cellular edema and hippocampal damage in both the treated mice and their offspring. However, varying doses of DSS effectively counteracted the neurotoxic effects of MSG, with no adverse impact on brain tissue structure or neural function in either adult mice or their offspring. Behavioral experiments further confirmed that DSS exerted a substantial protective effect against MSG-induced impairment of learning and memory in the treated adult mice and their offspring, in addition to mitigating central nervous system overexcitation and inhibiting exploratory behavior. In conclusion, DSS exerts significant protective effects against MSG-induced neurotoxicity in both adult mice and their offspring.

Monosodium glutamate (MSG) is one of the most extensively used flavour enhancers worldwide. Although it is widely regarded as a safe food additive with no recommended daily dosage, its over-consumption has been associated with notably pathophysiological events in various tissues and organs of the body. Previous studies have reported of the neuro- cardio- and hepato- toxic effects of its excessive exposure. Moreover, the food additive instigates metabolic dysfunction. It has been established that MSG damages male reproductive accessory organs like prostate glands and epididymis. In addition, it impairs serum enzymatic activities and serum levels of testosterone, gonadotropin-releasing hormone, luteinizing hormone and cholesterol. Reduced sperm count, sperm motility, sperm morphology, and sperm viability, imbalances in male reproductive hormones, alongside alteration in the histoarchitecture of the testes and other male reproductive tissues have also been connected with excessive exposure to MSG. Literature reports affirm the link between the over-consumption of MSG and reproductive organ weight and male sexual behaviour. This review article addresses the multi-systemic effects of exposure to MSG and the possible mechanism of action of the compound with a focus on the negative implications of the food additive on male reproductive functions and the possible role of natural antioxidants in male reproductive functions. carefully selected keywords were used during the literature search to gather credible and up-to-date information about the subject matter.

Thirty-six male albino Wistar rats 10 weeks old (180 to 200g) were divided into 4 groups: Group I: received distilled water, group II received L-arginine (L-A) 500 mg/kg, group III received 2 mg/kg monosodium glutamate (MSG), group IV received L-arginine 500 mg/kg and 2 mg/kg monosodium glutamate by oral gavage for 10 days. Cognitive performance was assessed using novel object recognition (NOR) and Y-maze tests. The relative brain weight of experimental rats was recorded. The malondialdehyde (MDA) level in the brain homogenate as oxidative stress biomarkers, antioxidants glutathione peroxidase (GPx), superoxide dismutase (SOD), catalase (CAT) activities, and dopamine (DA) levels were estimated. Histopathology and immunohistochemistry to serotonin (S-2A) receptors and caspase-3 were performed. Results revealed that MSG can cause a decline in cognitive functions as evidenced by NOR and y-maze tests. Besides, it has a neurotoxic effect as evidenced by increasing MDA level and GPx activity, decreasing SOD and CAT activities, reduced DA level, histopathological alteration in the brain, decreased S-2A receptors, and increased apoptosis as demonstrated by promoted caspase-3. Treatment with L-A 500 mg/Kg BW ameliorated the neurophysiological effects of MSG through improving memory, decreasing MDA level, GPx activity, increasing SOD and CAT activities, increasing DA level, improving histoarchitecture of the brain, improving S-2A receptors and decreasing apoptosis ameliorating caspase-3 protein in the brain.

Vernonia amygdalina (VA) is popularly consumed as food and as medicine due to its nutritional and bioactive constituents. This study assessed the anti-genotoxic effect of aqueous leaf extract of VA against monosodium (MSG) –induced genotoxicity. Crude extraction and phytochemical analysis were done using standard methods. In silico studies was done using compounds in the extract against Bcl-2, NF-kB 50, DNA polymerase lambda, DNA ligase, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX). Twelve rats were divided into three groups with four rats in each group. Group I was fed on food and water, group II received MSG (4 g/kg) per body weight (pbw) intraperitoneally, group III received MSG (4 g/kg) pbw intraperitoneally followed by oral dose of VA leaf extract (250 mg/kg) per body weight. The number of the micronucleated red blood cells and white blood cells were determined from blood smears microscopically. Results showed that aqueous extract of VA contained in mg/100 g alkaloids (7.04 ± 0.16), saponins (3.91 ± 0.13), flavonoid (1.64 ± 0.16), phenol (3.40 ± 0.12) and tannins (0.07 ± 0.32). In silico studies revealed high binding interaction (ΔG > −8.6) of vernoniosides D and E with all the tested proteins. There was a reduction in the number of micronucleated cells, neutrophils and eosinophils of the treated group compared to the MSG group, while there was an increase in the lymphocyte count. The anti-genotoxic effects of VA leaf extract might be attributed to the synergistic interaction of the various bioactive components in the extract. VA could be a potential plant for the prevention of cancer and other diseases that attenuate the immune system.

Introduction: In the poultry industry, feed efficiency is of great importance in order to reduce the cost of feed by maximizing production efficiency. To achieve this result, the use of synthetic amino acids, such as the amino acids methionine, lysine, arginine, etc. in poultry nutrition can play an effective role in animal performance. Application of monosodium glutamate in animals cause to increased levels of triglycerides, total protein, cholesterol and blood glucose in rats. Addition of 1% monosodium glutamate in broiler diets increased feed intake compared to the control group and also in relation to weight gain at the level of 0.25 and 0.5% monosodium glutamate had a significant decrease compared to the control group. Therefore, considering the role and importance of laying hen performance during the production process and due to the very important role of monosodium glutamate in the occurrence of these changes, the effects of consumption of different levels of monosodium glutamate on performance, egg quality characteristics and blood parameters were investigated.Materials and Methods: In this study, 60 laying hens of "Hy-Line W36" strain at the age of 68 to 74 weeks in 4 treatments and 5 replications and 3 hens per replication were used. The experiment was conducted as a completely randomized design in four periods 14-day for 8 weeks. Experimental treatments included 0, 0.4, 0.8 and 1.2% levels of monosodium glutamate per kg of feed. To better benefit from the data, all measurement factors except body weight gain were sampled and evaluated at the end of every two weeks. Egg mass was also obtained by multiplying the percentage of daily egg production by the average weight of eggs produced on the same day. Egg quality traits including height and diameter of albumin and yolk, relative weight of yolk and shell and albumin, shell thickness, shape and specific gravity were measured. Blood parameters were selected from two cages in each cage every two weeks and blood samples were taken from their wing veins and the concentrations of glucose, cholesterol, triglyceride, HDL and VLDL were measured using a Pars azmon kit and a spectrophotometer. Glutamate is one of the most abundant amino acids in nature, accounting for up to 8-10% of most dietary proteins and peptides and most tissues. Only the free form of glutamate has taste-enhancing properties as a unique umami taste, and when glutamate is bound to proteins, it is tasteless with no umami taste. Fermentation, ageing, ripening and heat cooking are typical natural processes of protein hydrolysis during which free glutamate is released. The food industry most commonly uses it in the form of monosodium glutamate (MSG) in amounts between 0.1 and 0.8% as a flavour enhancer. Since the beginning of the 21st century, its use as a flavour enhancer in meat and meat products with reduced salt content has become more widespread. The sodium content of MSG (12.28 g/100 g) is one-third that of salt (39.34 g/100 g), making MSG a promising salt alternative in sodium reduction strategies. There is no one-sided and conclusive scientific information reporting adverse human health effects of MSG in the general population, and it is still considered safe. However, in the wake of "clean label" initiatives, many consumers would prefer not to have additives or flavour enhancers such as glutamates in their food. This can be overcome by the known synergistic effect of natural 5'-ribonucleotides and glutamates found in seaweed, cheese, fish sauce, yeast extract, soy sauce, fermented soybeans and tomatoes, as they enhance the overall perception of umami flavour.

Modern enzyme biotechnology is a promising and rapidly developing field that requires the latest research on the conditions of enzyme biosynthesis. Optimizing the composition of the nutrient medium depending on the needs of microorganisms and physicochemical factors directly affect the increase in the efficiency of the biosynthesis of amylolytic enzymes, namely the biosynthetic capacity of the strain Streptomyces recifensis var. lyticus 2P-15. Modulation of the biosynthetic activity of strains producing amylolytic enzymes will allow to significantly increase their economic yield. Aim. The purpose of this work is to optimize the biosynthetic capacity of the strain Streptomyces recifensis var. lyticus 2P-15 in terms of the synthesis of amylolytic enzymes and the study of the dynamics of the influence of physical and chemical factors on optimization. Methods. The object of the study is the strain Streptomyces recifensis var. lyticus 2P-15, obtained by three-stage selection of the producer. The simplex method of selecting the composition of the environment was used for the research. The ratio of amylolytic activity to the level of biomass accumulation was taken as the biosynthetic capacity of the strain. A photocolometric method was used to determine amylolytic activity. The level of biomass accumulation was determined by the weight method. Results. It was established that as a result of optimizing the composition of the simplex nutrient medium by the method of mathematical modeling, the biosynthetic capacity increased by 3.63 compared to the control variant. It was also investigated that the optimal concentration of such a component of the nutrient medium as monosodium glutamate С5Н8NO4Na・H2O is 1.5%, which increases the amylolytic activity by 2.63 and increases the accumulation of biomass. Separately, it should be noted the obtained results of the study of the optimal concentrations of heavy metal ions added to the optimized version of the nutrient medium, which allow further research in this aspect to be continued and the use of Co, Mo, Cd ions in the composition of the nutrient medium. With the obtained results, there is an increase in amylolytic activity in the best response by 3.54. The obtained results have theoretical and practical significance for further research in the biotechnology of enzymes. Conclusions. The prospect of further research into the optimization of the biosynthesis of actinomycetes by the simplex method of other aspects of its regulation will be to increase the biosynthetic capacity of the studied strain, which will have a positive effect on the economic output of the production of amylolytic enzyme preparations by obtaining microbial synthesis.

The purpose of this study was to determine the potential of administration robusta coffee extract as an antioxidant on the renal histopathology in mice exposed to monosodium glutamate (MSG) orally. This study used 20 adult male rats with Balb C strain divided into 5 experimental groups, namely K + (MSG 0.12 mg / head and Vitamin C 6 mg/mice), K- (MSG 0.12 mg/mice and Sodium carboxymethyl cellulose (CMC Na) 0,1 ml/mice), P1 (MSG 0.12 mg/mice and Robusta coffee extract 0.1 mg/mice), P2 (MSG 0.12 mg/mice and Robusta coffee extract 0.2 mg/mice), and P3 (MSG 0.12 mg/mice and 0.4 mg Robusta coffee extract mg/mice). In most current farm operations, lactating sows need to overcome reproductive and environmental stresses that have resulted in poor sow production performance and piglet growth. Therefore, this study aimed to investigate the effects of in-feed supplementation of monosodium glutamate (MSG) in sows during late gestation lactation in regard to litter performance. The study subjects were 12 multi-parity sows (Landrace × Large White), farrowing sows with an average parity of four (three with three parities, seven with four parities, and two with five parities). They were randomly divided into the following two diet groups: the basal diet as a control (CON) group based on corn and soybean meal; and the basal diet + 2% MSG group. The experimental time ranged from 109 days before delivery to 21 days after delivery. There were six sows in each group, and each sow served as the experimental unit. There were no significant differences (p > 0.05) in body weight (BW), back fat (BF) thickness and estrus interval between sows supplemented with 2% MSG in their diets before and after farrowing and during weaning (p > 0.05). However, MSG-treated sows tended to increase BW loss at farrowing more than the CON group (p = 0.093) but lost less weight during lactation than the CON group (p = 0.019). There were no significant differences in the body condition scores (BCSs) and BF loss of the two groups of sows before and after farrowing and at weaning (p > 0.05). There was no significant difference in the weight of newborn piglets between the two groups of sows (p > 0.05). The weaning weight (p = 0.020) and average daily gain (ADG) (p = 0.045) of suckling piglets were higher in the MSG treated group compared to the CON group. The daily milk production of sows in the MSG treatment group was higher compared to the CON group (p = 0.045). The protein concentration of milk at week 3 (p = 0.060) and fat concentration of milk at week 5 (p = 0.095) of the MSG-supplemented sows tended to increase more than the CON group. In summary, the dietary inclusion of MSG supplementation had a beneficial effect on the late gestating sows and their piglet’s growth and milk production. Our research has shown that the addition of 2% MSG in late gestation and lactation diet would be beneficial for both sow and piglet production.

Current study examined the protective impact of Rhodiola rosea (R. rosea) extract on hepatic toxicity markers caused by MSG. MSG induced significant increases in hepatic stress biomarkers. There were significant declines in GSH, catalase and SOD levels, which were accompanied by substantial elevations in IL-1β, IL-6, and TNF-α. There was hepatic cellular damage, and upregulation of caspase-3 and a decrease in Bcl-2 expression. The hepatotoxic impacts of MSG were normalized by Rhodiola supplementation. R. rosea downregulated caspase-3 and upregulated HO-1 and Nrf2 mRNA expression. R. rosea regulated the immunoreactivity of COX-2, TGF-β1, and NFkB genes associated with hepatic toxicity. R. rosea extract normalized histopathological changes induced by MSG and restored hepatic architecture. R. rosea extract exhibited anti-apoptotic, antioxidant, and anti-inflammatory properties. In conclusion, R. rosea extract is a promising medication against hepatic toxicity induced by MSG, which works by regulating the different signaling pathways of inflammation-, oxidative-stress-, and apoptosis-associated markers.

Strategies to prevent the health abnormalities associated with the extensive use of MSG (monosodium glutamate) as a flavoring booster are badly needed. The current study was conducted to investigate oxidative stress, inflammation, and abnormal lipid profile as the main risk factors of neurotoxicity in MSG-exposed female albino rats. Besides, the effect of concurrent consumption of Zingiber officinale rhizomes powder was studied at low doses. Twenty rats (total) were split into 4 separate groups. The 1st group was a negative control group (without any treatment), while the others received 6 mg MSG/kg. The 2nd group was left untreated, whereas the 3rd and 4th groups were given a regular laboratory diet that included ginger rhizome powder supplements (GRP, 0.5 & 1%, respectively) for six weeks. In brain tissue homogenates, exposure to MSG caused a significant depletion of gamma-aminobutyric acid (GABA) and total protein levels, while triglycerides and cholesterol contents were significantly elevated. Moreover, a noteworthy upsurge in oxidative load and inflammation markers was also noticed associated with a marked reduction of antioxidant levels, which histopathological staining verified further. The rat diet formulated with GRP, with a dose-dependent effect, resulted in increased GABA and total protein contents and attenuated inflammation, oxidative stress, abnormal lipid profile, and marked histological changes in cerebral cortical neurons of MSG-administered animals. Therefore, this study reveals that GRP shields rats against the neurotoxicity that MSG causes. The anti-inflammatory as well as antioxidant, and lipid-normalizing properties of rhizomes of ginger may be accountable for their observed neuroprotective action.

This systematic review analyzes monosodium glutamate (MSG) in the Alzheimer’s disease-like condition to enhance translational research. Our review seeks to understand how MSG affects the brain and causes degenerative disorders. Due to significant preclinical data linking glutamate toxicity to Alzheimer’s disease and the lack of a comprehensive review or meta-analysis, we initiated a study on MSG’s potential link. We searched PubMed, ScienceDirect, ProQuest, DOAJ, and Scopus for animal research and English language papers without time constraints. This study used the PRISMA-P framework and PICO technique to collect population, intervention or exposure, comparison, and result data. It was registered in PROSPERO as CRD42022371502. MSG affected mice’s exploratory behaviors and short-term working memory. The brain, hippocampus, and cerebellar tissue demonstrated neuronal injury-related histological and histomorphometric changes. A total of 70% of MSG-treated mice had poor nesting behavior. The treated mice also had more hyperphosphorylated tau protein in their cortical and hippocampus neurons. Glutamate and glutamine levels in the brain increased with MSG, and dose-dependent mixed horizontal locomotor, grooming, and anxiety responses reduced. MSG treatment significantly decreased phospho-CREB protein levels, supporting the idea that neurons were harmed, despite the increased CREB mRNA expression. High MSG doses drastically lower brain tissue and serum serotonin levels. In conclusion, MSG showed AD-like pathology, neuronal atrophy, and short-term memory impairment. Aim:Although monosodium glutamate is widely used in the food industry, it has effects in terms of teratogenicity, especially during pregnancy and embryogenesis. Our study analyses the effect on the development of the notochord by giving monosodium glutamate to chicken egg embryos.Methods:The incubation and embryonic development follow-up of our study were held in the experimental animals laboratory of University of Health Sciences Turkey, Istanbul Bakirkoy Prof. Dr. Mazhar Osman Mental Health and Neurological Diseases Training and Research Hospital, and the preparation and microscopy of the samples were carried out in the Pathology Department of the Cerrahpasa Faculty of Medicine. 120 fertile, pathogen-free eggs were incubated at 75% humidity and 37.4±0.2 °C until embryos reached Hamburger and Hamilton stages 9-10 (30th hour). Eggs were divided into four groups. Group 1 consisted of uninjected eggs, group 2 consisted of eggs injected with saline (10 nL 0.9 NaCl), group 3 consisted of eggs injected with 15 mg/kg monosodium glutamate (MSG); and group 4 consisted of eggs injected at regular intervals.Results:The correlation values between height, dilatation, autolysis, chest, and lumbar diameters of embryos in all groups were examined. The mean embryo length was 1.0017±0.36 cm. The mean chest diameter was 20.70±14.45 cm. The mean lumbar diameter was 10.31±14.34 cm. When the groups were compared in terms of embryo length, it was observed that the groups given MSG were significantly shorter than the control and SF groups (p=0.00).Conclusion:The amount of MSG taken with food is important because it can affect the organs. Depending on the amount consumed, MSG may adversely affect notochord development.

Monosodium glutamate is a non-essential amino acid in the human body that has been widely used as a food additive since 1909 due to its unique umami taste. Typical food containing monosodium glutamate include tomatoes, yeast extracts, fermented soy products, and cheese. After entering the human body, it is mainly absorbed in the small intestine by passive diffusion. Subsequent binding to TAS1R1-TAS1R3 promotes the release of intracellular calcium that ultimately activates the brain’s taste center, recognized as an umami taste of monosodium glutamate. However, more and more preclinical and clinical data indicates that monosodium glutamate exerts adverse effects on humans, including metabolic syndrome, neurotoxicity, renal toxicity, cardiovascular disease, infertility and fetal underdevelopment, cancer, immune malfunction and so on. In addition, it has been found that medicinal plants and nutraceuticals could mitigate or treat the adverse effects of monosodium glutamate adverse. Taken together, we hope to raise the correct awareness of monosodium glutamate and improve the understanding of monosodium glutamate quality approaches to maximize their use.

We hypothesized whether 10% praying-mantis-egg-cake (10% PMEC) can be applied against inflammatory-erectile-dysfunction and whether it could be linked to NO-cGMP-dependent PKG signaling cascade. Ninety male albino-rats were randomly distributed into nine (n = 10) groups. Group I was given distilled water. Group II and III were pre-treated with 80 mg/kg NaCl and 75 mg/kg MSG, respectively. Group IV was pre-treated with 80 mg/kg NaCl + 75 mg/kg MSG. Group V was administered with 80 mg/kg NaCl+ 3 mg/kg Amylopidin. Group VI was given 80 mg/kg NaCl + 10% PMEC. Group VII was treated with 75 mg/kg MSG + 10% PMEC. Group VIII was treated with 80 mg/kg NaCl+ 75 mg/kg MSG + 10% PMEC. Group IX was post-treated with 10% PMEC for 14 days. Penile PDE-51, arginase, ATP hydrolytic, cholinergic, dopaminergic (MAO-A) and adenosinergic (ADA) enzymes were hyperactive on intoxication with NaCl and MSG. The erectile dysfunction caused by inflammation was linked to alteration of NO-cGMP-dependent PKG signaling cascade via up-regulation of key cytokines and chemokine (MCP-1). These lesions were prohibited by protein-rich-cake (10% PMEC). Thus, protein-rich-cake (10% PMEC) by a factor of 4 (25%) inhibited penile cytokines/MCP-1 on exposure to mixture of salt-intake through NO-cGMP-PKG dependent-NF-KB signaling cascade in rats

Monosodium glutamate (MSG) is a worldwide food flavour enhancer commonly used by the food industry. This feed additive may cause male infertility. Nigella sativa seeds (NSS) is a widely used in herbal medicine as it has many biological benefits and could provide a solution. This work was designed to investigate the histological effects of NSS on rats ingesting MSG. To achieve this aim, adult male albino rats (2- 3 months old) were randomly and equally assigned into three experimental groups. For a period of 21 days, control group received no treatment, MSG group received MSG as 30 g/kg feed, and MSG + NSS group received MSG as 30 g/kg feed and NSS as 30 g/kg feed. Seminal vesicle histopathology in MSG group showed mild seminal vesiculitis with degeneration of smooth muscle fibers in tunica muscularis. In addition, there was an increase in the amount of connective tissue and apoptotic cells count. Periodic Acid Schiff stain indicated irregular and interrupted epithelial basement membranes. Glutathione reductase (GR), superoxide dismutase 2 (SOD2), and caspase-3 immuno-expressions increased in MSG group. It was found that there was an increase in the number of apoptotic cells, intraepithelial lymphocytes and dendritic cells in MSG group. However, treatment with NSS ameliorated these disturbances. NSS mitigated MSG-induced seminal vesicle damage by its histoprotective, cytoprotective and anti-apoptotic activities.

MSG use has significantly increased over the past 30 years, its global demand stands huge at over three million metric tons which is worth over $4.5 billion. Asia was responsible for more than three quarter of world MSG consumption with the country China also leading in global consumption as well as production and export to other countries. Prior to year 2020, global demand for MSG increased by almost four percent each year with the highest significant increase in demand for MSG predicted to rise in Thailand, Indonesia, Vietnam and China, followed by Brazil and Nigeria. However, several researches featured in this review has identified MSG consumption as a major contributor to the development and progression of some metabolic disorders such as obesity, which is a risk factor for other metabolic syndromes like hypertension, diabetes mellitus and cancer initiation. The mechanism by which MSG induce obesity involves induction of hypothalamic lesion, hyperlipidemia, oxidative stress, leptin resistance and increased expression of peroxisome proliferator-activated receptors (PPARs) Gamma and Alpha. Similarly for induction of diabetes mellitus, MSG consumption resulted in decreased pancreatic beta cell mass, increased oxidative stress and metabolic rates, reduced glucose and insulin transport to adipose tissue and skeletal muscles, insulin insensitivity, reduced insulin receptors and induced severe hyperinsulinemia. Dietary salt, an active component of MSG is also found to be a major risk factor for high blood pressure (which may lead to hypertension). MSG is used to enhance the taste of tobacco, causing smokers to consume the product in excess and thereby increasing the risk of cancer development. Depending on the amount consumed, MSG has both positive and negative effects. Despite the controversy surrounding MSG's safety and its probable contribution to risk of development and progression of metabolic disorders, its global consumption is still very high. Therefore, this article will sensitize the public on the need for cautious use of MSG in foods and also aid regulatory agencies to further review the daily MSG consumption limit based on metabolic toxicities observed at the varied dosages reported in this review.

Monosodium glutamate (MSG) is found in refined foods. Apocynin (APO) is a selective NADPH oxidase (NOX) inhibitor. The aim of this experimental study was to investigate possible effects of MSG and the curative effects of APO in rats. Twenty-eight male Sprague-Dawley rats were randomly divided into four groups (Normal control, APO, MSG and MSG + APO, n:7 for each group). The MSG and MSG + APO groups received 120 mg/kg MSG solution orally for 28 consecutive days. The APO and MSG + APO groups received 25 mg/kg APO solution orally for 5 days until the end of the experiment. At the end of the experiment, all rats were sacrificed and liver tissue and blood samples were taken for histological, ultrastructural, and biochemical analyses. In the MSG group, vacuolization and loss in glycogen content in the hepatocytes, leukocyte infiltration and fibrosis in the liver parenchyme and portal triads, were observed. Terminal deoxynucleotidyl transferase dUTP (TUNEL)-positivity and NADPH oxidase (NOX)-2-positivity were higher in the MSG group compared with the other experimental groups. The concentrations of alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP), total bilirubin, malondialdehyde (MDA), and myeloperoxidase (MPO) were higher, whereas albumin, glutathione (GSH), and superoxide (SOD) levels were lower in the MSG group. All these data has been reversed in MSG + APO group. The histological and biochemical criteria indicated the prominent ameliorating effect of APO on MSG -induced liver injury.

Abstract Background Monosodium glutamate is a widely used flavor enhancer/food additive in meat, fish, milk and cheese or vegetable origins. Our present study aimed to assess the effect of utilization monosodium glutamate on the cardiac, splenic, hepatic and renal organs in mice. Thirty animals were divided into three groups: control group; glutamate 10 treated groups and glutamate 15 treated group. The experimental period was two successive weeks. Results Oral administrations of monosodium glutamate for 10 days induced moderate histopathological changes in cardiac, splenic, hepatic and renal tissues and also resulted in significant increase of the nuclear factor-kB expression depend on time of administration as compared to the control group, while treatment with monosodium glutamate for 14 days resulted in severe histopathological changes as well as highly significant increase of the nuclear factor-kB expression as compared to the control group. Conclusions It could be concluded that oral administration of monosodium glutamate induced histopathological alterations, and cytotoxicity in cardiac, splenic, hepatic and renal tissues dependent on time.

Objectives This study aimed to evaluate the potential mitigating effect of fisetin on monosodium glutamate (MSG)-induced testicular toxicity and investigate the possible involvement of silent mating type information regulation 2 homolog 1 (SIRT1) in this effect.Methods Forty male rats were divided into normal control, fisetin-treated, MSG-treated, and fisetin + MSG-treated groups. Testosterone, GnRH, FSH, and LH were measured in plasma, as well as SIRT1 and phosphorylated AMP-activated protein kinase (pAMPK) levels in testicular tissues using ELISA. Hydrogen peroxide (H2O2), nitric oxide (NO), and reduced glutathione (GSH) were measured colorimetrically, while Nicotinamide adenine dinucleotide phosphate oxidase 4 (NOX4) expression was relatively quantified using RT–PCR in testicular tissues.Results After 30 days, fisetin could ameliorate MSG-induced testicular toxicity by acting centrally on the hypothalamic-pituitary-gonadal axis, increasing plasma levels of GnRH, FSH, LH, and testosterone. Peripheral actions of fisetin on the testis were indicated as it increased testicular SIRT1 and pAMPK. Furthermore, it antagonized glutamate-induced oxidative stress by significantly lowering H2O2, NO, and relative NOX4 expression while significantly increasing reduced GSH levels. It also improved the architecture of the seminiferous tubules, reduced sperm abnormality, and increased sperm count.Discussion Fisetin ameliorates MSG-induced testicular toxicity via central and peripheral mechanisms making it a promising therapeutic target for male infertility.

Over the last century the people’s mode of life and eating habits has dramatically changed: the people of developed countries began to consume fast food, and also started disorderly and frequent snacking. The production of dietary meals and the increase of food assortment, including food produced from low-quality ingredients, led to the manufacturer’s necessity to use a large number of functional ingredients, i. e. those that improve taste of the food. Monosodium glutamate (MSG) is one of the widely used additives. Monosodium L-Glutamate (E621) is the sodium salt of glutamic acid found in all protein foods; it is used throughout the world as a food flavor enhancer. The legislation of the Russian Federation limits the content of monosodium glutamate, or additive E621, in a food product. Due to the fact that the glutamic acid takes the major weight in the monosodium glutamate molecule, which molecule is naturally present in almost all food products, the weight of the molecule of the E621 additive was determined by content of this amino acid expressed in terms of monosodium glutamate. In connection with the foregoing, it became necessary to develop a method for the quantitative determination of the mass fraction of monosodium glutamate introduced into food during the production of food products. Within the framework of this research a new method for determining the share of added monosodium glutamate is proposed, which is not associated with the natural content of glutamic acid. The authors have developed a method for determining the mass fraction of monosodium glutamate in food products with the help of high performance liquid chromatography with precolumn derivatization. This research presents metrological assessment of the developed methodology, determines accuracy rates and reproducibility factors in two concentrations ranges. For a range of 0.1 to 1%, the reproducibility is set at 17% and the accuracy rate is set at 30%. For the range of 1–10%, the reproducibility is 6%, the accuracy rate is 10% respectively. Also, during the development of the method, the lower limits for the quantitative determination (Limit of Detection — LOD) and qualitative determination (Limit of Quantification — LOQ) of the method were calculated. LOQ was equal to 0.01% and LOD accounted for 0.1%. The method has successfully passed the metrological certification and is included in the Register of Measurement Methods of the Russian Federation. It can be used by accredited laboratories for assessment and control of food quality.

As the relevance between left and right brain neurons when transmitting electrical signals of umami taste is unknown, the aim of this work was to investigate responsive regions of the brain to the umami tastant monosodium glutamate (MSG) by using scalp-electroencephalogram (EEG) to identify the most responsive brain regions to MSG. Three concentrations of MSG (0.05, 0.12, 0.26 g/100 mL) were provided to participants for tasting while recoding their responsive reaction times and brain activities. The results indicated that the most responsive frequency to MSG was at 2 Hz, while the most responsive brain regions were T4CzA2, F8CzA2, and Fp2CzA2. Moreover, the sensitivity of the brain to MSG was significantly higher in the right brain region. This study shows the potential of using EEG to investigate the relevance between different brains response to umami taste, which contributes to better understanding the mechanism of umami perception.

Background: Monosodium Glutamate (MSG), used widely in the food industry, is a threat to the public health. We investigated whether the MSG administration depletes non-enzymatic antioxidants, i.e., vitamins C and E in the liver of Wistar albino rats. We also examined the restorative effect of the ethanolic extract of Phyllanthus emblica (P. emblica). Methods: Wistar albino rats (n=42) were adapted and then randomly divided into seven groups of: 1) control, 2, 3, 4) MSG treatment, and 5, 6, 7) combined MSG and P. emblica extract treatment. Objective(s): Inflammation is the major progenitor of obesity and associated metabolic disorders. The current study investigated the modulatory role of saroglitazar on adipocyte dysfunction and associated inflammation in monosodium glutamate (MSG) obese Wistar rats.Materials and Methods: The molecular docking simulation studies of saroglitazar and fenofibrate were performed on the ligand-binding domain of NLRP3 and NF- κB. Under in vivo study, neonatal pups received normal saline or MSG (4 g/kg, SC) for 7 alternate days after birth. After keeping for 42 days as such, animals were divided into seven groups: Normal control; MSG control; MSG + saroglitazar (2 mg/kg); MSG + saroglitazar (4 mg/kg); saroglitazar (4 mg/kg) per se; MSG + fenofibrate (100 mg/kg); fenofibrate (100 mg/kg) per se. Drug treatments were given orally, from the 42nd to 70th day. On day 71, blood was collected and animals were sacrificed for isolation of liver and fat pads. Results: In silico study showed significant binding of saroglitazar and fenofibrate against NLRP3 and NF- κB. Saroglitazar significantly reduced body weight, body mass index, Lee’s index, fat pad weights, adiposity index, decreased serum lipids, interleukin-1β (IL-1β), tumor necrosis factor-α(TNF-α), interleukin-6 (IL-6), leptin, insulin, blood glucose, HOMA-IR values, oxidative stress in the liver and increased hepatic low-density lipoprotein receptor levels. Histopathological analysis of the liver showed decreased inflammation and vacuolization, and reduced adipocyte cell size. Immunohistochemical analysis showed suppression of NLRP3 in epididymal adipocytes and NF- κB expression in the liver. Conclusion: Saroglitazar ameliorated obesity and associated inflammation via modulation of NLRP3 inflammasome and NF- κB in MSG obese Wistar rats.

Monosodium glutamate (MSG) is the sodium salt of glutamic acid (GLA), used as a flavour enhancer. MSG is considered a controversial substance. It is incriminated in disturbing the antioxidant system, but also has beneficial effects, as GLA metabolism plays a crucial role in homeostasis. This study highlights which positive or negative aspects of MSG sub-chronic consumption are better reflected in subjects potentially affected by advanced age. Daily doses of MSG were administered to four groups of two-year-old Wistar rats for 90 days: (I) 185 mg/kg bw, (II) 1500 mg/kg bw, (III) 3000 mg/kg bw and (IV) 6000 mg/kg bw, compared to a MSG non-consumer group. Aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, direct and total bilirubin, total cholesterol, triglycerides, creatinine and urea levels were analysed; stomach, liver and kidney samples were subjected to histopathological analysis. Although, in most cases, there were no statistical differences, interesting aspects of the dose–effect relationship were observed. After MSG sub-chronic consumption, the positive aspects of GLA seem to be reflected better than the negative ones. The hormesis effect, with low-level reactive oxygen species’ protective effects and GLA metabolism, may represent the hypothesis of a potential defence mechanism triggered by MSG sub-chronic consumption in ageing rats.

Purpose: This study aims to examine the effect of ileal transposition (IT) on plasma levels of the total antioxidant status (TAS), total oxidant status (TOS), Oxidative Stress Index (OSI), Superoxide dismutase (SOD), Nicotinamide adenine dinucleotide phosphate oxidase (NOX), Catalase (CAT), Reduced Glutathione (GSH) in both rats with Metabolic syndrome (MetS) and healthy controls. Materials and Methods: In the MetS model, newborn male Wistar albino rats were given MSG (4 g/mg) on days 0, 2, 4, 6, 8, and 10. The control group was injected only saline. In the 5th month, sham and IT animals underwent selected surgery. 2 months after surgery TOS, TAS, OSI, SOD, NOX, CAT, and GSH levels were assessed in the plasma. Results: IT procedure significantly increased SOD and CAT levels in MetS + IT group when compared to the MetS group (SOD; MetS 1.75 ± 0.04, MetS+IT 2.1 ± 0.15, CAT; MetS 32.02 ± 1.73, MetS+IT 41.64 ± 1.18,). As expected, while GSH levels was increased in MetS+IT rats compared to MetS rats, but the difference was not significant (MetS 243.31 ± 6.36, MetS+IT 269.76 ± 9.17). The NOX activity was significantly lower in MetS+IT group than MetS and MetS+S groups (MetS 610.35 ± 26.25, MetS+IT 348.86 ± 14.12). Conclusion: These data revealed the healing effect of IT surgery against oxidative stress associated with MetS. The available data endorses IT surgery as an effective strategy to reduce oxidative damage in rats with MetS by modulating systemic oxidant and antioxidant responses.

Monosodium glutamate (MSG) is an additive which is substantially applied in commercially processed foods in order to increase the flavor and sapidity and make a unique flavor which cannot be provided by any other ingredient. Since the discovery of endogenous amino acid glutamate (as a neurotransmitter) in human body, the possible toxicity of exogenous glutamic acid has attracted the attention of numerous scholars. Accordingly, various animal studies have been documented on toxic impacts of MSG on different parts of the body including central nervous system, liver, adipose tissue, reproductive organs, and other systems. Thus, since that time, the safety of MSG has repeatedly been checked and reaffirmed within the scientific communities due to the contradict results. This literature review article specifically aimed to discuss the probable safety of dietary MSG for central nervous system and also provide an integrated information from several studies documented on possible neurotoxic effects of monosodium glutamate on glutamate receptors of Central Nervous System in order to elevate the public awareness about it. Collecting the results of all studied articles seems to supports the hypothesis of safety. In fact, it seems that MSG as a food additive within the limited amounts as well as natural levels of glutamic acid which is present in food supplies provides no serious hazard to the human CNS.

Background: The toxicological effects of monosodium glutamate (MSG), widely used as a food additive, are a major public health challenge. This research aimed to investigate the nephroprotective effects of methanol and n-hexane extracts of Piper nigrum (MEPN and HEPN, respectively) against MSG-induced renal toxicity in rats. Materials and methods: The phytochemicals, vitamins, and in vitro antioxidant activities were determined using standard protocols. Acute toxicity was determined with thirty-two Swiss albino mice, while thirty male Wistar albino rats were used for the nephroprotective study. The rats were randomly distributed into 6 experimental groups (n = 5). Group 1 (baseline) was not induced or treated; groups 2–6 were given 2 g/kg body weight of MSG. Additionally, group 2 received normal saline; groups 3–6 were given 250 and 500 mg/kg body weight of MEPN and HEPN, respectively. MSG was administered alone for 21 days, after which MSG was concomitantly administered with the extracts for additional 28 days. After treatment, biochemical tests were done on the serum, and a histopathological examination was determined with the kidney. Results: Both extracts scavenged 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and reduced ferric ions to ferrous ions in the Ferric Reducing Antioxidant Power (FRAP) assay, with MEPN having higher efficacy. The extracts also ameliorated lipid peroxidation and increased the activities of superoxide dismutase and catalase, as well as the level of reduced glutathione. In addition, intervention with MEPN and HEPN provided nephroprotective effects by decreasing urea and creatinine levels and also improved the histo-architecture of the kidney. This efficacy could be due to the rich phytochemicals and antioxidant vitamins previously identified in the plant. Conclusions: These findings are critical in focusing attention on abolishing the use of MSG in our food sector and highlights the health benefits of incorporating P. nigrum into our food to avert food and drug toxicity.

Background: This study aimed to investigate the role of spirulina against Monosodium glutamate (MSG) induced hepatic dysfunction as an in vivo study in a rat model. Methods: In this study, 28 rats were divided into four groups (i) controls, (ii) spirulina, (iii) MSG and (MSG + spirulina). Each group consists of 7 rats. The first group is considered as a normal group and consists of the availability of food and water without any restrictions. The second group involves 1000 mg/kg bw spirulina extract daily for ten days. The third group is the positive intoxicated group, given orally 4 g/kg bw monosodium glutamate (MSG) for ten days. The final group is orally administered Spirulina and MSG for ten days. Within each of the four groups, serum, qPCR, histology, and immunohistochemistry were examined. Results: Biochemical and antioxidant analysis showed a significant association with MSG and MSG + spirulina, and molecular analysis revealed a significant association with Nrf2, HO-1, BCl2, and β-actin genes. Histological analysis showed modest congestion of the central vein, intact hepatocytes organized in cords, and mild sinusoidal dilatation in the Centro lobular area of the liver of the spirulina-treated group. The H&E stain was documented to be 50–200 m bar. The liver section analysis in NF-κB, TGF-β and COX-2 showed a protective effect. Conclusion: This study concludes as the combination of MSG and spirulina has a protective effect. However, MSG has shown an elevated role among all groups.

Abstract Primary eosinophilic colitis (PEC) is a primary eosinophilic gastrointestinal disorder, and immunoglobulin/histamine complex (IHC) may be an effective therapeutic for PEC. IHC has a nonallergen‐specific antinociceptive effect in the treatment of histamine‐mediated pain.

The study aimed to assess the growth performance, haematology, serum biochemistry, gonadal and extragonadal sperm reserves of two breeds of rabbit bucks fed dietary monosodium glutamate (MSG) at varying inclusion levels (0.00, 0.25, 0.50 and 0.75 g/kg diet). A total of 320 sexually mature New Zealand White Bucks and Dutch Belted Bucks aged 8 to 10 mo with average weight ranging from 1.34 to 1.96 kg were used for the study, which lasted 8 wk. The bucks were weighed and distributed to the four treatment diets. Each treatment was replicated 10 times with four bucks per replicate in a 2×4 factorial experiment. At the end of the feeding trial, 2 bucks per replicate were euthanised. Blood samples were collected from the jugular veins for haematological and serum analyses and their reproductive tracts were dissected.
