Abstract Background Acute kidney injury (AKI) is a common and serious condition, particularly among elderly patients. It is associated with high morbidity and mortality rates, further compounded by the need for continuous renal replacement therapy in severe cases. To improve clinical decision-making and patient management, there is a need for accurate prediction models that can identify patients at a high risk of mortality. Methods Data were extracted from the Dryad Digital Repository. Multivariate analysis was performed using least absolute shrinkage and selection operator (LASSO) logistic regression analysis to identify independent risk factors and construct a predictive nomogram for mortality within 28 days after continuous renal replacement therapy in elderly patients with acute kidney injury. The discrimination of the model was evaluated in the validation cohort using the area under the receiver operating characteristic curve (AUC), and calibration was evaluated using a calibration curve. The clinical utility of the model was assessed using decision curve analysis (DCA). Results A total of 606 participants were enrolled and randomly divided into two groups: a training cohort (n = 424) and a validation cohort (n = 182) in a 7:3 proportion. A risk prediction model was developed to identify independent predictors of 28-day mortality in elderly patients with AKI. The predictors included age, systolic blood pressure, creatinine, albumin, phosphorus, age-adjusted Charlson Comorbidity Index (CCI), Acute Physiology and Chronic Health Evaluation II (APACHE II) score, and sequential organ failure assessment (SOFA) score. These predictors were incorporated into a logistic model and presented in a user-friendly nomogram. In the validation cohort, the model demonstrated good predictive performance with an AUC of 0.799. The calibration curve showed that the model was well calibrated. Additionally, DCA revealed significant net benefits of the nomogram for clinical application. Conclusion The development of a nomogram for predicting 28-day mortality in elderly patients with AKI receiving continuous renal replacement therapy has the potential to improve prognostic accuracy and assist in clinical decision-making.

With the rapid development of intelligent production technology, using the advantage of industrial robots to promote corporate green development has become an inevitable trend. The study conducts an empirical test using 916 Chinese A-share listed companies from 2011 to 2019. Results indicate that industrial robots can effectively reduce firm-level carbon emissions, while technology innovation and internal control promote this effect. Also, robots can suppress corporate carbon emissions by enhancing human capital's level and increasing investment in emission-reduction equipment. Through heterogeneity analysis, this negative effect is more significant in regions with greater carbon reduction pressure and industries with higher capital-to-labor ratios. This paper enriches the related research on intelligent manufacturing and energy saving and provides empirical evidence for the use of intelligent production technology to achieve corporate carbon reduction, which is of great significance in cracking the intrinsic contradiction between emission reduction and development and accelerating the realization of sustainable development.

Abstract In order to investigate the influence of minor Ru on the electrochemical behaviour and structural characteristics of passive films on the surface of Ti-6Al-4V alloys under various oil and gas exploration conditions, electrochemical techniques, X-ray photoelectron spectroscopy (XPS), scanning electron microscope (SEM) and corrosion simulation tests were carried out. The results revealed that the oil and gas exploration conditions had a serious impact on the electrochemical behaviour and corrosion resistance of the tested alloys. The passivation film resistance and corrosion potential of the tested titanium alloys were significantly reduced with increasing acidity and temperature. With the addition of minor ruthenium, the potential of the passive film on the Ti-6Al-4V-0.11Ru alloy surface increased because of the high surface potential of the ruthenium element. The contents of metallic ruthenium and tetravalent titanium oxide TiO2 in the surface film of the Ti-6Al-4V-0.11Ru alloy both increased with increasing temperature, which led to increase the thickness, stability, corrosion resistance and repairability of the passive film on the surface of the Ti-6Al-4V-0.11Ru alloy being better than those qualities of Ti-6Al-4V. These results were also confirmed by corrosion simulation tests.

Microplastics are a major environmental issue. In marine systems, these break down into small fragments that may lodge within organisms, but they behave as vectors for chemicals when pollutants such as metals, polycyclic aromatic hydrocarbons (PAHs), pesticides, and pharmaceuticals are absorbed by the particles. The rate and extent of uptake of organic compounds onto microplastics is dependent on a range of factors such as the sites available on the surface, the type of plastic, and the compound being sorbed, with hydrophobicity an important property. Laboratory experiments determined the uptake of pyrene and acenaphthene (and sometimes phenanthrene) onto pellets of polyvinyl chloride (PVC), polyethylene terephthalate (PET), polyoxymethylene (POM), polypropylene (PP) and polystyrene (PS). The kinetics of uptake in the experiments reveals half-lives ~10 h. The extent of partition onto the plastic pellets was similar for pyrene and phenanthrene, though lower for acenaphthene. This was not surprising because the octanol-water distribution coefficient of acenaphthene is about an order of magnitude smaller than that of pyrene, suggesting it is less hydrophobic. The PAHs are distributed most strongly into PP but only weakly into PVC. The fragmentation and oxidation of microplastics means that they sorb increasing amounts of organic material as they age.

A compressive near-field millimeter wave (MMW) imaging algorithm is proposed. From the compressed sensing (CS) theory, the compressive near-field MMW imaging process can be considered to reconstruct an image from the under-sampled sparse data. The Gini index (GI) has been founded that it is the only sparsity measure that has all sparsity attributes that are called Robin Hood, Scaling, Rising Tide, Cloning, Bill Gates, and Babies. By combining the total variation (TV) operator, the GI-TV mixed regularization introduced compressive near-field MMW imaging model is proposed. In addition, the corresponding algorithm based on a primal-dual framework is also proposed. Experimental results demonstrate that the proposed GI-TV mixed regularization algorithm has superior convergence and stability performance compared with the widely used l1-TV mixed regularization algorithm.

Abstract Background Fournier's gangrene (FG), a urological emergency with high mortality, is an infectious necrotizing fasciitis of the perineal and genital regions. The majority of FG is caused by polymicrobial organisms involving mixed aerobes and anaerobes but rarely reveals Actinomyces species. Case presentation We report a healthy 67-year-old Asian male who presented with rapidly progressive painful swelling of the scrotum. Clinically diagnosed with FG, the patient underwent an emergency radical debridement, followed by broad-spectrum antibiotics and negative pressure wound therapy. The identification of the causative microorganisms showed Actinomyces turicensis and the antibiotic treatment was adjusted accordingly. After wound bed preparation, we took split-thickness skin grafts to cover the scrotal wound. Active management to minimize faecal contamination was applied throughout the whole course of treatment and repair. The patient was satisfied with the outcome. This was an extremely rare case of A. turicensis as the main causative pathogen of FG. Conclusions FG due to Actinomyces species is rarely reported, but we should still consider this pathogenic microorganism that has long been neglected.

Radiotherapy is the major treatment of non-small cell lung cancer (NSCLC). The radioresistance and toxicity are the main obstacles that leading to therapeutic failure and poor prognosis. Oncogenic mutation, cancer stem cells (CSCs), tumor hypoxia, DNA damage repair, epithelial-mesenchymal transition (EMT), and tumor microenvironment (TME) may dominate the occurrence of radioresistance at different stages of radiotherapy. Chemotherapy drugs, targeted drugs, and immune checkpoint inhibitors are combined with radiotherapy to treat NSCLC to improve the efficacy. This article reviews the potential mechanism of radioresistance in NSCLC, and discusses the current drug research to overcome radioresistance and the advantages of Traditional Chinese medicine (TCM) in improving the efficacy and reducing the toxicity of radiotherapy.

Abstract Messenger RNA (mRNA) vaccine technology has shown its power in preventing the ongoing COVID-19 pandemic. Two mRNA vaccines targeting the full-length S protein of SARS-CoV-2 have been authorized for emergency use. Recently, we have developed a lipid nanoparticle-encapsulated mRNA (mRNA-LNP) encoding the receptor-binding domain (RBD) of SARS-CoV-2 (termed ARCoV), which confers complete protection in mouse model. Herein, we further characterized the protection efficacy of ARCoV in nonhuman primates and the long-term stability under normal refrigerator temperature. Intramuscular immunization of two doses of ARCoV elicited robust neutralizing antibodies as well as cellular response against SARS-CoV-2 in cynomolgus macaques. More importantly, ARCoV vaccination in macaques significantly protected animals from acute lung lesions caused by SARS-CoV-2, and viral replication in lungs and secretion in nasal swabs were completely cleared in all animals immunized with low or high doses of ARCoV. No evidence of antibody-dependent enhancement of infection was observed throughout the study. Finally, extensive stability assays showed that ARCoV can be stored at 2–8 °C for at least 6 months without decrease of immunogenicity. All these promising results strongly support the ongoing clinical trial.

Abstract Background Mobile phone addiction has become a social problem that affects the healthy growth of adolescents, and it may be correlated with coping style. The aim of this study was to investigate the relationship between mobile phone addiction and coping style and the influencing factors for adolescents. Methods A meta-analysis was conducted by searching China National Knowledge Infrastructure (CNKI), WANFANG DATA and Chongqing VIP Information Co., Ltd. (VIP), PubMed, Web of Science, Embase, and PsycINFO. Stata 16.0 was used to analyse the overall effect and test the moderating effect. Results Thirty-three studies were included, involving a total of 20,349 subjects. There was no significant correlation between adolescents’ mobile phone addiction and positive coping style (r = − 0.02, 95% CI = − 0.06 to 0.02, P > 0.05), but there was a moderate positive correlation between adolescents’ mobile phone addiction and negative coping style (r = 0.31, 95% CI = 0.26 to 0.36, P

Recently, an algorithm of surface turbulent heat fluxes over snow/sea ice has been developed based on the theory of maximum entropy production (MEP), which is fundamentally different from the bulk flux algorithm (BF) that has been used in sea ice models for a few decades. In this study, we first assess how well the MEP algorithm captures the observed variations of turbulent heat fluxes over Arctic sea ice. It is found that the calculated heat fluxes by the MEP method are in good agreement with in-situ observations after considering the absorption of incoming radiation in a snow/ice surface layer with infinitesimal depth. We then investigate the effects of two different schemes (MEP vs. BF) in the sea ice model of CICE6 on simulated turbulent heat fluxes and sea ice processes in the Arctic Basin. Our results show that the two different schemes give quite different representations of seasonal variations of heat fluxes, particularly for sensible heat fluxes in summer. The heat fluxes simulated by the MEP produce weak cooling effect on the ice surface in summer, whereas the BF generates a warming effect. As a result, compared to the BF, the MEP leads to a reduced seasonal cycle of Arctic sea ice mass flux by modulating snow-to-ice conversion, basal ice growth, surface ice melt and basal ice melt.

Abstract Background Patients with prior illness are more vulnerable to heat stroke-induced injury, but the underlying mechanism is unknown. Recent studies suggested that NLRP3 inflammasome played an important role in the pathophysiology of heat stroke. Methods In this study, we used a classic animal heat stroke model. Prior infection was mimicked by using lipopolysaccharide (LPS) or lipoteichoic acid (LTA) injection before heat stroke (LPS/LTA 1 mg/kg). Mice survival analysis curve and core temperature (TC) elevation curve were produced. NLRP3 inflammasome activation was measured by using real-time PCR and Western blot. Mice hypothalamus was dissected and neuroinflammation level was measured. To further demonstrate the role of NLRP3 inflammasome, Nlrp3 knockout mice were used. In addition, IL-1β neutralizing antibody was injected to test potential therapeutic effect on heat stroke. Results Prior infection simulated by LPS/LTA injection resulted in latent inflammation status presented by high levels of cytokines in peripheral serum. However, LPS/LTA failed to cause any change in animal survival rate or body temperature. In the absence of LPS/LTA, heat treatment induced heat stroke and animal death without significant systemic or neuroinflammation. Despite a decreased level of IL-1β in hypothalamus, Nlrp3 knockout mice demonstrated no survival advantage under mere heat exposure. In animals with prior infection, their heat tolerance was severely impaired and NLRP3 inflammasome induced neuroinflammation was detected. The use of Nlrp3 knockout mice enhanced heat tolerance and alleviated heat stroke-induced death by reducing mice hypothalamus IL-1β production with prior infection condition. Furthermore, IL-1β neutralizing antibody injection significantly extended endotoxemic mice survival under heat stroke. Conclusions Based on the above results, NLRP3/IL-1β induced neuroinflammation might be an important mechanistic factor in heat stroke pathology, especially with prior infection. IL-1β may serve as a biomarker for heat stroke severity and potential therapeutic method.

Oxidative stress is currently considered to be the main cause of brain aging. Astaxanthin can improve oxidative stress under multiple pathological conditions. It is therefore hypothesized that astaxanthin might have therapeutic effects on brain aging. To validate this hypothesis and investigate the underlying mechanisms, a mouse model of brain aging was established by injecting amyloid beta (Aβ)25–35 (5 μM, 3 μL/injection, six injections given every other day) into the right lateral ventricle. After 3 days of Aβ25–35 injections, the mouse models were intragastrically administered astaxanthin (0.1 mL/d, 10 mg/kg) for 30 successive days. Astaxanthin greatly reduced the latency to find the platform in the Morris water maze, increased the number of crossings of the target platform, and increased the expression of brain-derived neurotrophic factor, synaptophysin, sirtuin 1, and peroxisome proliferator-activated receptor-γ coactivator 1α. Intraperitoneal injection of the sirtuin 1 inhibitor nicotinamide (500 μM/d) for 7 successive days after astaxanthin intervention inhibited these phenomena. These findings suggest that astaxanthin can regulate the expression of synaptic proteins in mouse hippocampus through the sirtuin 1/peroxisome proliferator-activated receptor-γ coactivator 1α signaling pathway, which leads to improvements in the learning, cognitive, and memory abilities of mice. Marine plastic debris is an environmental problem, and its degradation into microplastics (1-5000 μm) introduces them into the food chain. In this study, small polyoxymethylene (global production ~3000 Tg per year) pellets were exposed in terrestrial and simulated marine environments to heat and light, resulting in cracking during decay with increasing IR absorption (OH-bonds). Furthermore, sunlight over three years reduced pellet mass and diameter (~10% and ~40%), initially yielding 100-300 μm fragments. Changes under UV irradiation were smaller as it could not penetrate into particle interiors. Background Indigo-containing plant tissues change blue after a freezing treatment, which is accompanied by changes in indigo and its related compounds. Phaius flavus is one of the few monocot plants containing indigo. The change to blue after freezing was described to explore the biosynthesis of indigo in P. flavus. Methods In this study, we surveyed the dynamic change of P. flavus flower metabolomics and transcriptomics. Results The non-targeted metabolomics and targeted metabolomics results revealed a total of 98 different metabolites, the contents of indole, indican, indigo, and indirubin were significantly different after the change to blue from the freezing treatment. A transcriptome analysis screened ten different genes related to indigo upstream biosynthesis, including three anthranilate synthase genes, two phosphoribosyl-anthranilate isomerase genes, one indole-3-glycerolphosphate synthase gene, five tryptophan synthase genes. In addition, we further candidate 37 cytochrome P450 enzyme genes, one uridine diphosphate glucosyltransferase gene, and 24 β-D-glucosidase genes were screened that may have participated in the downstream biosynthesis of indigo. This study explained the changes of indigo-related compounds at the metabolic level and gene expression level during the process of P. flavus under freezing and provided new insights for increasing the production of indigo-related compounds in P. flavus. In addition, transcriptome sequencing provides the basis for functional verification of the indigo biosynthesis key genes in P. flavus.

Glioma is the most common malignant tumor of the central nervous system, and it isdifficult to cure. The growth of malignant glioma is invasive. In recent decades, the strategy of surgicalresection combined with concurrent radiotherapy and chemotheraphy, and immunocheckpoint inhibitorshave been made progress. However, the therapeutic effect is still not ideal, which may be related to the lowimmunogenicity of tumor cells and the immunosuppression of tumor microenvironment (TME). In the TME,immune cells compete with glioma cells and other proliferating cells for nutrients, while metabolites in theTME affect the differentiation and functions of immune cells. This article summarizes the research progressof glioma metabolism regulation and tumor immunosuppressive microenvironment in recent years, whichprovides new ideas for the study of the mechanism of tumor progression and glioma of treatment. The tumor microenvironment is essential for the formation and development of tumors. Cytokines in the microenvironment may affect the growth, metastasis and prognosis of tumors, and play different roles in different stages of tumors, of which transforming growth factor β (TGF-β) and tumor necrosis factor α (TNF-α) are critical. The two have synergistic and antagonistic effect on tumor regulation. The inhibition of TGF-β can promote the formation rate of tumor, while TGF-β can promote the malignancy of tumor. TNF-α was initially determined to be a natural immune serum mediator that can induce tumor hemorrhagic necrosis, it has a wide range of biological activities and can be used clinically as a target to immune diseases as well as tumors. However, there are few reports on the interaction between the two in the tumor microenvironment. This paper combs the biological effect of the two in different aspects of different tumors. We summarized the changes and clinical medication rules of the two in different tissue cells, hoping to provide a new idea for the clinical application of the two cytokines.

To investigate the clinical value of changes in the subtypes of peripheral blood lymphocytes and levels of inflammatory cytokines in patients with COVID-19, the total numbers of lymphocytes and CD4+ lymphocytes and the ratio of CD4+/CD8+ lymphocytes were calculated and observed in different groups of patients with COVID-19. The results show that the lymphocytopenia in patients with COVID-19 was mainly manifested by decreases in the CD4+ T lymphocyte number and the CD4+/CD8+ ratio. The decreased number of CD4+ T lymphocytes and the elevated levels of TNF-α and IL-6 were correlated with the severity of COVID-19 disease.

This paper puts forward the concept of incentive utility, which converts the value of each attribute in multi-attribute decision-making into a unified index for measurement, thus simplifying multi-attribute decision-making. In this paper, a comprehensive evaluation model based on the TOPSIS of incentive utility is established. Through the establishment of incentive index, the utility of incentive and the calculation of ideal point of utility, the disadvantage of "ideal point is not ideal" in the TOPSIS is overcome. It can better synthesize the sub-attribute value and realize the subjective multi-attribute decision-making. The example shows that the model can apply subjective factors to decision-making scientifically, and the result is more reasonable and effective.

Non-expressor of pathogenesis-related genes 1 (NPR1) plays a significant role in the defense responses of plants to pathogens by regulating the expression of defense-related genes. In the present study, we isolated two NPR1 genes from Vitis aestivalis cv. Norton and Vitis vinifera cv. Cabernet Sauvignon, which were referred to as VaNPR1.1 and VvNPR1. 1-CS, respectively. They encode a protein of 584 amino acids with a predicted molecular weight of 64.8 kDa and a theoretical isoelectric point (pI) of 5.74. The predicted amino acid sequences of VaNPR1.1 and VvNPR1.1-CS differ by only one amino acid. Over-expression of VaNPR1.1 gene in Arabidopsis npr1-1 mutant plants restores the transcriptional expression of AtPR-1 gene, though not to the full scale. This result demonstrated that a grapevine VaNPR1.1 possesses a similar function to the Arabidopsis NPR1 in the regulation of defense-related genes. Over-expression of VaNPR1.1 in transgenic Arabidopsis plant increased tolerance to salinity, but had no effect on the drought tolerance. We conclude that VaNPR1.1 is a functional ortholog of AtNPR1 and also involved in grapevine's response to the salt stress.

Experiment was conducted at the Gongzhuling Experimental Station of Institute of Crop Sciences, Chinese Academy of Agricultural Sciences, Jilin Province, China, during 2009–2010. Six representative varieties of maize (Baihe in the 1950s, Jidan 101 in the 1960s, Zhongdan 2 in the 1970s, Yedan 13 in the 1980s, Zhengdan 958 in the 1990s, and Xianyu 335 in the 2000s) were each planted under two different densities (52 500 and 82 500 plants ha−1) and two different nitrogen application levels (150 and 300 kg ha−1). Root characteristics and distribution among soil layers were studied by the field root digging method. The results showed that root mass increased with the process of the growth and development of the plant, and it peaked at kernel filling stage, and decreased at maturity due to the root senesces. Root mass of different maize varieties from the 1950s to 1980s had a trend of increase, while it decreased for the modern varieties. Root length and root surface areas had the similar changing trend. The study suggested that early maize varieties may have root redundancy, and reducing root redundancy may be a direction for variety improvement for high yield. Root characteristics were affected by nitrogen application level and density; modern varieties were more suitable for higher fertilizer application level and density conditions. Root characteristics distribution among soil layers decreased by an exponent equation, but the regression coefficients of different varieties were different. Though the root length density (RLD) of every soil layer of different varieties also decreased by an exponent equation, there were large variations of RLD in every part of a layer.

Silicone rubber (SR) is a common soft tissue filler material used in plastic surgery. However, it presents a poor surface for cellular adhesion and suffers from poor biocompatibility. In contrast, hydroxyapatite (HA), a prominent component of animal bone and teeth, can promote improved cell compatibility, but HA is an unsuitable filler material because of the brittleness in mechanism. In this study, using a simple and economical method, two sizes of HA was applied to coat on SR to counteract the poor biocompatibility of SR. Surface and mechanical properties of SR and HA/SRs confirmed that coating with HA changes the surface topology and material properties. Analysis of cell proliferation and adhesion as well as measurement of the expression levels of adhesion related molecules indicated that HA-coated SR significantly increased cell compatibility. Furthermore, mass spectrometry proved that the biocompatibility improvement may be related to elongation factor 1-beta (EF1β)/γ-actin adjusted cytoskeletal rearrangement.

Medical device implants are drawing increasing amounts of interest from modern medical practitioners. However, this attention is not evenly spread across all such devices; most of these implantable devices can cause adverse reactions such as inflammation, fibrosis, thrombosis, and infection. In this work, the biocompatibility of silicone rubber (SR) was improved through carbon (C) ion implantation. Scanning electron microscopy (SEM), atomic force microscopy (AFM), X-ray photoelectron spectroscopy (XPS), and X-ray diffraction (XRD) results confirmed that these newly generated carbon-implanted silicone rubbers (C-SRs) had large, irregular peaks and deep valleys on their surfaces. The water contact angle of the SR surface decreased significantly after C ion implantation. C ion implantation also changed the surface charge distribution, silicone oxygen rate, and chemical-element distribution of SR to favor cell attachment. The dermal fibroblasts cultured on the surface C-SR grew faster and showed more typical fibroblastic shapes. The expression levels of major adhesion proteins, including talin-1, zyxin, and vinculin, were significantly higher in dermal fibroblasts cultured on C-SR coated plates than in dermal fibroblasts cultured on SR. Those same dermal fibroblasts on C-SRs showed more pronounced adhesion and migration abilities. Osteopontin (OPN), a critical extracellular matrix (ECM) protein, was up-regulated and secreted from dermal fibroblasts cultured on C-SR. Matrix metalloproteinase-9 (MMP-9) activity was also increased. These cells were highly mobile and were able to adhere to surfaces, but these abilities were inhibited by the monoclonal antibody against OPN, or by shRNA-mediated MMP-9 knockdown. Together, these results suggest that C ion implantation significantly improves SR biocompatibility, and that OPN is important to promote cell adhesion to the C-SR surface.

Mitochondrial membrane permeability has received considerable attention recently because of its key role in apoptosis and necrosis induced by physiological events such as hypoxia. The manner in which mitochondria interact with other molecules to regulate mitochondrial permeability and cell destiny remains elusive. Previously we verified that hypoxia-induced phosphorylation of microtubule-associated protein 4 (MAP4) could lead to microtubules (MTs) disruption. In this study, we established the hypoxic (1% O(2)) cell models of rat cardiomyocytes, H9c2 and HeLa cells to further test MAP4 function. We demonstrated that increase in the pool of MAP4 could promote the stabilization of MT networks by increasing the synthesis and polymerization of tubulin in hypoxia. Results showed MAP4 overexpression could enhance cell viability and ATP content under hypoxic conditions. Subsequently we employed a yeast two-hybrid system to tag a protein interacting with mitochondria, dynein light chain Tctex-type 1 (DYNLT1), by hVDAC1 bait. We confirmed that DYNLT1 had protein-protein interactions with voltage-dependent anion channel 1 (VDAC1) using co-immunoprecipitation; and immunofluorescence technique showed that DYNLT1 was closely associated with MTs and VDAC1. Furthermore, DYNLT1 interactions with MAP4 were explored using a knockdown technique. We thus propose two possible mechanisms triggered by MAP4: (1) stabilization of MT networks, (2) DYNLT1 modulation, which is connected with VDAC1, and inhibition of hypoxia-induced mitochondrial permeabilization.

The asymmetric unit of the title compound, C14H14N4·2H2O consists of two half-molecules of the main molecule, each situated on an inversion center, and two molecules of water.
