Life-threatening Candida infections have increased with the COVID-19 pandemic, and the already limited arsenal of antifungal drugs has become even more restricted due to its side effects associated with complications after SARS-CoV-2 infection. Drug combination strategies have the potential to reduce the risk of side effects without loss of therapeutic efficacy. The aim of this study was to evaluate the combination of ent-hardwickiic acid with low concentrations of amphotericin B against Candida strains. The minimum inhibitory concentration (MIC) values were determined for amphotericin B and ent-hardwickiic acid as isolated compounds and for 77 combinations of amphotericin B and ent-hardwickiic acid concentrations that were assessed by using the checkerboard microdilution method. Time–kill assays were performed in order to assess the fungistatic or fungicidal nature of the different combinations. The strategy of combining both compounds markedly reduced the MIC values from 16 µg/mL to 1 µg/mL of amphotericin B and from 12.5 µg/mL to 6.25 µg/mL of ent-hardwickiic acid, from isolated to combined, against C. albicans resistant to azoles. The combination of 1 µg/mL of amphotericin B with 6.25 µg/mL of ent-hardwickiic acid killed all the cells of the same strain within four hours of incubation.

The objective of this study was to evaluate whether pre-weaning heifer calves divergent for residual feed intake (RFI) or residual feed intake and body weight gain (RIG) exhibit differences in thermography, blood, and ruminal parameters. Thirty-two Gyr heifer calves were enrolled in a 63-d trial and classified into 2 feed efficiency (FE) groups based on RFI and RIG (mean ± 0.5 SD). The groups were classified as high efficiency (HE) RFI (HE RFI, n = 9), HE RIG (HE RIG, n = 10), low efficiency (LE) RFI (LE RFI, n = 10), and LE RIG (LE RIG, n = 11). The amount of whole milk provided for each calf was calculated based on their metabolic weight at birth (42% x BW0.75). The liquid diet was divided into two meals at 0700 and 1400 h. The total solid diet (TSD) was composed of 92% concentrate and 8% of Tifton 85 hay chopped in 5-cm lengths, as fed. Intake was measured daily. Blood concentrations of insulin, beta hydroxybutyrate, urea, and glucose, and ruminal pH, N-NH3, and volatile fatty acids (VFA) were evaluated at 14, 28, 42, 56, and 70 days of age. Thermal images of the calves were taken with an infrared camera (FLIR T420, FLIR Systems Inc., Wilsonville, OR) on d 56 (±3) at 0600 h, before the morning feeding. Total VFA concentration and propionate as % of total VFA were 24.2% and 22.2% lower in HE RFI compared to LE RFI calves, respectively. On the other hand, acetate as % of total VFA was 10.6% greater in HE RFI than LE RFI calves. Blood urea concentration tended to be greater in LE RFI than HE RFI calves. High efficiency HE RIG tended to have 6.8% greater acetate and 15.4% lower propionate as % of total VFA than LE RIG. Blood insulin concentration was greater and blood glucose tended to be greater for LE RIG than HE RIG group. Low efficiency RIG group had greater left rib, left flank, and anus surface temperature measured by infrared thermography than the HE RIG group. Differences in ruminal fermentation do not seem to be associated with pre-weaning calves efficiency, while differences in protein metabolism seem to affect RFI during this phase. Infrared thermography appears to be correlated to RIG in pre-weaning heifer calves.

Estudos têm mostrado efeitos tóxicos da enrofloxacina em diversos tecidos. Assim, testou-se a hipótese de que a enrofloxacina pode interferir no desenvolvimento placentário e gerar efeitos adversos ao feto. A enrofloxacina (Baytril(r)) foi administrada em ratas, na dose de 5mg/kg, diariamente, IM, durante toda a gestação. As placentas foram analisadas morfologicamente, morfometricamente e imuno-histoquimicamente aos sete, 14 e 21 dias de prenhez. Os resultados mostraram que a enrofloxacina reduziu o número de sítios de implantação, o peso e a área total do disco placentário aos 14 e 21 dias de desenvolvimento, além dos elementos constituintes da placenta. A análise histoquímica não revelou alterações significativas no teor de fibras colágenas, elásticas e reticulares. O teste de Tunel mostrou atividade apoptótica apenas nas placentas com 14 dias de desenvolvimento de ambos os grupos, sendo mais intensa no grupo tratado. Não foi observado nenhum indício de malformação na cabeça, no tronco e nos membros dos neonatos. No entanto, houve uma redução significativa no número e no peso dos neonatos no grupo tratado, porém sem afetar o seu comprimento. Assim, concluiu-se que a enrofloxacina administrada na dosagem de 5mg/kg durante prenhez em ratas interfere no número de embriões implantados e no desenvolvimento placentário. Isto sugere cautela na administração da enrofloxacina durante a gestação, pois a exposição contínua a esse antibiótico pode ter reflexos na redução do número e do peso da prole.

A dexametasona é utilizada em casos de gestação com risco de prematuridade; porém, doses suprafisiológicas podem afetar a embriogênese. A melatonina tem demonstrado prevenir efeitos deletérios dos glicocorticoides. Assim, avaliamos a influência da melatonina sobre efeitos da dexametasona em ratas prenhes através dos seguintes parâmetros: 1. Hemograma e perfil glicídico; 2. Níveis de progesterona; e 3. Histomorfometria e histoquímica. Foram utilizadas 20 ratas divididas nos grupos: I - ratas prenhes que receberam placebo (Controle); II - ratas prenhes tratadas com dexametasona (0,8mg/kg); III - ratas prenhes tratadas com melatonina (0,5mg/kg); IV - ratas prenhes tratadas com dexametasona e melatonina. Todos os tratamentos foram iniciados 10 dias após confirmação do acasalamento até o final da prenhez. O sangue foi coletado no 7º, 14º e 21º dia de prenhez. As dosagens de carboidratos e progesterona foram realizadas pelo método antrona e ELISA, respectivamente. O fígado, rins e adrenais foram analisados histoquímica e morfometricamente. No 7º dia de prenhez, não houve alteração significativa nos parâmetros analisados. Porém, no 14º dia de prenhez, houve aumento significativo do volume de hematócrito, redução do número total de hemácias e leucócitos, neutrofilia, linfopenia, eosinopenia e redução do diâmetro das hemácias nas matrizes tratadas com dexametasona. Esses efeitos permaneceram no 21º dia de prenhez, exceto para o hematócrito, o qual reduziu. Verificou-se ainda redução significativa dos níveis de glicose (21º dia de prenhez) e progesterona (14º e 21º dia de prenhez). Não houve alteração nos parâmetros morfométricos e histoquímico no fígado, rins e adrenais. A dexametasona na dosagem de 0,8mg/kg, administrada a partir do terço médio da prenhez, produz alterações hematológicas, bioquímicas e hormonais em ratas, sendo prevenidas pela melatonina; porém não afeta o fígado, rins e adrenais quanto aos parâmetros morfométricos e histoquímicos.

Concluiu-se que a melatonina materna é importante para a modulação do hemograma em ratas prenhes e para o desenvolvimento normal do baço e do timo dos filhotes.We investigated the influence of melatonin on the hemogram of pregnant rats and puppies, and on the histogenesis and morphology of the spleen and thymus of puppies. Melatonin was administered at a dose 0.5mg/kgbody weight, dissolved in 0.1mL of ethanol and diluted in 0.3mL of saline. For hematocrit analysis, total erythrocyte count and total and differential leukocyte count, blood samples were collected on the 7th, 14th and 21st days of pregnancy and the offspring at 10 days of birth. Histological sections of spleen and thymus of the offspring were used for histochemistry and morphometry. The absence of melatonin promoted changes in blood count only in the final third of gestation, without interfering with the hemogram of the puppies, and induced morphological and morphometric changes in the thymus and spleen in the first days of life of the puppies. It was concluded that maternal melatonin is important for the modulation of the blood count in pregnant rats and the normal development of the spleen and thymus of the offspring.

Avaliou-se o efeito da administração de dexametasona no início e no meio da gestação de ratas, sobre os perfis glicídicos e hematológicos materno e da prole. Os animais foram submetidos aos seguintes tratamentos: dexametasona do primeiro ao sétimo dia e placebo do oitavo ao 14º dia; placebo do primeiro ao sétimo dia e dexametasona do oitavo ao 14º dia; dexametasona do primeiro ao 14º dia e placebo do primeiro ao 14º dia de gestação. A dexametasona foi administrada por via intraperitoneal, na dose de 0,8mg/kg. Foram coletadas amostras de sangue no sétimo, 14º e 21º dias de gestação, e de sangue e tecido hepático da prole no quinto, 10º e 15º dias pós-natal. Para a verificação das reservas de glicogênio hepático da prole, cortes histológicos foram corados pelo ácido periódico de Schiff. Os resultados apontam para um efeito tempo-dependente da administração de dexametasona durante a gestação, levando a alterações temporais distintas na hematologia e na concentração plasmática de carboidratos nas matrizes e na prole.

Diterpenes have been identified as active compounds in several medicinal plants showing remarkable biological activities, and some isolated diterpenes are produced at commercial scale to be used as medicines, food additives, in the synthesis of fragrances, or in agriculture. There is great interest in developing methods to obtain derivatives of these compounds, and biotransformation processes are interesting tools for the structural modification of natural products with complex chemical structures. Biotransformation processes also have a crucial role in drug development and/or optimization. The understanding of the metabolic pathways for both phase I and II biotransformation of new drug candidates is mandatory for toxicity and efficacy evaluation and part of preclinical studies. This review presents an overview of biotransformation processes of diterpenes carried out by microorganisms, plant cell cultures, animal and human liver microsomes, and rats, chickens, and swine in vivo and highlights the main enzymatic reactions involved in these processes and the role of diterpenes that may be effectively exploited by other fields.

Laboratory and field experiments were performed to assess the ecological roles of natural products produced by the Brazilian red seaweed Laurencia obtusa. Laboratory assays revealed that the natural concentration of the crude organic extract of L. obtusa significantly inhibited feeding by two herbivores: the crab Pachygrapsus transversus and the sea urchin Lytechinus variegatus. It was verified that this chemically defensive action was due to halogenated sesquiterpenoid elatol, found to be the major natural product of this red seaweed. In addition, it was verified that the antifouling property of the chemicals produced by L. obtusa could make this red alga less attractive for fish grazing. Direct protection against two herbivore species and indirect protection against herbivory by fouling inibition constitute evidence that the major natural product from Brazilian L. obtusa plays multiple environmental roles, thereby increasing the adaptive value of these metabolites. On the other hand, the evidence reinforces the idea that marine natural products may have different functions in the sea.

In this paper, we prove that if a Nemytskii operator maps Lp(omega, E) into Lq(omega, F), for p, q greater than 1, E, F separable Banach spaces and F reflexive, then a sequence that converge weakly and a.e. is sent to a weakly convergent sequence. We give a counterexample proving that if q = 1 and p is greater than 1 we may not have weak sequential continuity of such operator. However, we prove that if p = q = 1, then a weakly convergent sequence that converges a.e. is mapped into a weakly convergent sequence by a Nemytskii operator. We show an application of the weak continuity of the Nemytskii operators by solving a nonlinear functional equation on W1,p(omega), providing the weak continuity of some kind of resolvent operator associated to it and getting a regularity result for such solution.Neste artigo, provamos que se um operador de Nemytskii aplica Lp(ômega, E) no Lq(ômega , F), para p, q maiores do que 1, E, F espaços de Banach separáveis e F reflexivo, então uma seqüência que converge fracamente e q.t.p. é transformada em uma seqüência fracamente convergente. Fornecemos um contra-exemplo mostrando que se q = 1 e p é maior do que 1, podemos não ter continuidade seqüêncial de tal operador. Contudo provamos que se p = q = 1, então seqüências fracamente convergentes que convergem q.t.p. são aplicadas em seqüências fracamente convergentes por um operador de Nemytskii. Mostramos uma aplicação da continuidade fraca dos operadores de Nemytskii resolvendo uma equação funcional não linear no W1,p(ômega), provando a continuidade fraca de um tipo de operador resolvente associado ao operador de Nemytskii e obtendo um resultado de regularidade de tal solução.

The present research describes the histology of the midgut, gastric caeca, and pyloric valve of Tropidacris collaris (Stoll, 1813), (Orthopetera: Romaleidae). We used light microscopy, staining (Gomori's trichrome and periodic acid-Schiff (PAS)), and a routine histological analysis method (hematoxilin-eosin). The insects were obtained from, and also bred in, the Laboratory of Entomology, Department of Biology, of the Rural Federal University of Pernambuco (UFRPE). The collected material was fixed in alcoholic Boüin and embedded in paraplast. The results demonstrated that the midgut wall is composed of an inner epithelial layer and two outer layers of striate muscles: one internal (circular) and the other external (longitudinal), with connective tissue between the muscle fibers. The epithelium is single-layered, with two cell types: regenerative and elongated columnar. The gastric caeca presents muscle layers similar to those of the midgut. Simple columnar epithelium lines the gastric caeca, which presents villi and projects towards the lumen. The pyloric valve is of striate muscle tissue, covered by a single epithelial-cell layer.

Seaweed preference by the Brazilian endemic gastropod Astraea latispina was examined in the laboratory to evaluate the role of secondary metabolites in determining food choice. Of three species of seaweeds examined, Plocamium brasiliense was highly preferred; less so were Sargassum furcatum and Dictyota cervicornis were preferred less. Extracts and/or pure major metabolites of the two potentially chemically-defended seaweeds (P. brasiliense and D. cervicornis) were tested as feeding deterrents against A. latispina. Algal extract assays demonstrated that three concentrations of crude organic extract of the red alga P. brasiliense (50%, 100%: natural concentration, and 200% of dry weight: dw) did not affect feeding of this gastropod. In contrast, the three concentrations of crude organic extract of the brown alga D. cervicornis (50%, 100% and 200% dw) inhibited feeding by A. latispina. The chemical deterrent property of D. cervicornis extract against the gastropod A. latispina occurred due to a mixture of the secodolastane diterpenes isolinearol/linearol (4:1 -- 0.08% dry weight). This is the first report showing that Dictyota cervicornis produces a chemical defense against herbivores using secodolastane diterpenoid. In addition, these results widen the action spectrum of secondary metabolites found in seaweed belonging to this brown algal genus.
