The neighborhood polynomial of graph G is the generating function for the number of vertex subsets of G of which the vertices have a common neighbor in G. In this paper, we investigate the behavior of this polynomial under several graph operations. Specifically, we provide an explicit formula for the neighborhood polynomial of the graph obtained from a given graph G by vertex attachment. We use this result to propose a recursive algorithm for the calculation of the neighborhood polynomial. Finally, we prove that the neighborhood polynomial can be found in polynomial-time in the class of k-degenerate graphs.

Abstract Background Sustainable agriculture increasingly emphasizes the importance of microbial communities in influencing plant health and productivity. In viticulture, understanding the impact of management practices on fungal communities is critical, given their role in disease dynamics, grape and wine quality. This study investigates the effects of integrated, organic, and biodynamic management practices on the diversity and function of fungal communities in a vineyard located in Geisenheim, Germany, focusing on above-ground parts such as bark, leaves, and grapes. Results Our findings indicate that while overall fungal species richness did not significantly differ among management systems across various compartments, the composition of these communities was distinctly influenced by the type of management system. In particular, leaf and grape compartments showed notable variations in fungal community structure between integrated and organic/biodynamic management. No differences were observed between organic and biodynamic management. Integrated management demonstrated a significantly higher abundance of mycoparasites in comparison to organic and biodynamic management, primarily attributed to the increased presence of Sporobolomyces roseus, Sporobolomyces ellipsoideus and Rhodotorula glutinis. Conclusions The findings highlight the importance of management practices in shaping fungal community composition and function in vineyards. Although overall species richness remained unaffected, community composition and functional diversity varied, highlighting the potential for strategic microbiome management to enhance vineyard sustainability and plant health.

This paper introduces a trivariate graph polynomial that is a common generalization of the domination polynomial, the Ising polynomial, the matching polynomial, and the cut polynomial of a graph. This new graph polynomial, called the bipartition polynomial, permits a variety of interesting representations, for instance as a sum ranging over all spanning forests. As a consequence, the bipartition polynomial is a powerful tool for proving properties of other graph polynomials and graph invariants. We apply this approach to show that, analogously to the Tutte polynomial, the Ising polynomial introduced by Andrén and Markström in [3], can be represented as a sum over spanning forests.

Abstract Objective Examining the distribution of breast cancer (BC) stage and molecular subtype among women aged below ( 65 years) the recommended screening age range helps to understand the screening program's characteristics and contributes to enhancing the effectiveness of BC screening programs. Methods In this retrospective study, female patients with newly diagnosed BC from 2010 to 2020 were identified. The distribution of cases in terms of TNM stages, severity classes, and subtypes was analysed according to age groups. Results A total of 3282 women diagnosed with BC were included in the analysis. Among these cases 51.4% were detected outside the screening age group, and these were characterized by a higher TNM stage compared to those diagnosed within the screening age band. Ultrasound is an excellent way to acquire data that reveal useful information about systems operating in harsh environments, which may include elevated temperature, ionizing radiation, and aggressive chemicals. The effects of harsh environments on piezoelectric materials have been studied in much more depth than the other aspects of ultrasonic transducers used in pulse-echo mode. Therefore, finite element simulations and laboratory experiments are used to demonstrate the unique characteristics of pulse-echo immersion testing. Using an aluminum nitride piezoelectric element mounted on a vessel wall, characteristics associated with electrode thickness, couplant, backing material, and an acoustic matching layer are investigated. Considering a wave path through a vessel wall and into a fluid containing a target, when the travel distance in the fluid is relatively short, it can be difficult to discern the target echo from the reverberations in the vessel wall. When an acoustic matching layer between the vessel wall and the fluid does not suffice, a simple subtractive signal-processing method can minimize the reverberations, leaving just the target echoes of interest. Simulations and experiments demonstrate that sufficient target echoes are detected to determine the time of flight. Furthermore, a simple disc-like surface anomaly on the target is detectable.

Abstract Besides vaccines, the development of antiviral drugs targeting SARS-CoV-2 is critical for preventing future COVID outbreaks. The SARS-CoV-2 main protease (Mpro), a cysteine protease with essential functions in viral replication, has been validated as an effective drug target. Here, we show that Mpro is subject to redox regulation in vitro and reversibly switches between the enzymatically active dimer and the functionally dormant monomer through redox modifications of cysteine residues. These include a disulfide-dithiol switch between the catalytic cysteine C145 and cysteine C117, and generation of an allosteric cysteine-lysine-cysteine SONOS bridge that is required for structural stability under oxidative stress conditions, such as those exerted by the innate immune system. We identify homo- and heterobifunctional reagents that mimic the redox switching and inhibit Mpro activity. The discovered redox switches are conserved in main proteases from other coronaviruses, e.g. MERS-CoV and SARS-CoV, indicating their potential as common druggable sites.

Abstract Background IgG-class autoantibodies to N-Methyl-D-Aspartate (NMDA)-type glutamate receptors define a novel entity of autoimmune encephalitis. Studies examining the prevalence of NMDA IgA/IgM antibodies in patients with Parkinson disease with/without dementia produced conflicting results. We measured NMDA antibodies in a large, well phenotyped sample of Parkinson patients without and with cognitive impairment (n = 296) and controls (n = 295) free of neuropsychiatric disease. Detailed phenotyping and large numbers allowed statistically meaningful correlation of antibody status with diagnostic subgroups as well as quantitative indicators of disease severity and cognitive impairment. Methods NMDA antibodies were analysed in the serum of patients and controls using well established validated assays. We used anti-NMDA antibody positivity as the main independent variable and correlated it with disease status and phenotypic characteristics. Results The frequency of NMDA IgA/IgM antibodies was lower in Parkinson patients (13%) than in controls (22%) and higher than in previous studies in both groups. NMDA IgA/IgM antibodies were neither significantly associated with diagnostic subclasses of Parkinson disease according to cognitive impairment, nor with quantitative indicators of disease severity and cognitive impairment. A positive NMDA antibody status was positively correlated with age in controls but not in Parkinson patients. Conclusion It is unlikely albeit not impossible that NMDA antibodies play a significant role in the pathogenesis or progression of Parkinson disease e.g. to Parkinson disease with dementia, while NMDA IgG antibodies define a separate disease of its own.

Abstract Background The aim of our study was the identification of genetic variants associated with postoperative complications after cardiac surgery. Methods We conducted a prospective, double-blind, multicenter, randomized trial (RIPHeart). We performed a genome-wide association study (GWAS) in 1170 patients of both genders (871 males, 299 females) from the RIPHeart-Study cohort. Patients undergoing non-emergent cardiac surgery were included. Primary endpoint comprises a binary composite complication rate covering atrial fibrillation, delirium, non-fatal myocardial infarction, acute renal failure and/or any new stroke until hospital discharge with a maximum of fourteen days after surgery. Results A total of 547,644 genotyped markers were available for analysis. Ultrasonic transducers are often used in the nuclear industry as sensors to monitor the health and process status of systems or the components. Some of the after-effects of the Fukushima Daiichi earthquake could have been eased if sensors had been in place inside the four reactors and sensed the overheating causing meltdown and steam explosions. The key element of ultrasonic sensors is the piezoelectric wafer, which is usually derived from lead-zirconate-titanate (Pb(Zr, Ti)O3, PZT). This material loses its piezoelectrical properties at a temperature of about 200 °C. It also undergoes nuclear transmutation. Bismuth titanate (Bi4Ti3O12, BiTi) has been considered as a potential candidate for replacing PZT at the middle of this temperature range, with many possible applications, since it has a Curie–Weiss temperature of about 650 °C. The aim of this article is to describe experimental details for operation in gamma and nuclear radiation concomitant with elevated temperatures and details of the performance of a BiTi sensor during and after irradiation testing. In these experiments, bismuth titanate has been demonstrated to operate up to a fast neutron fluence of 5 × 1020 n/cm2 and gamma radiation of 7.23 × 1021 (gamma/cm2). The results offer a perspective on the state-of the-art for a possible sensor for harsh environments of high temperature, Gamma radiation, and nuclear fluence.

Carbon dioxide (CO2) as one of the main factors driving climate change is known to increase grapevine growth and yield and could, therefore, have an impact on the fruit quality of vines. This study reports the effects of elevated CO2 (eCO2) on berry development and bunch structure of two grapevine cultivars (Vitis vinifera L. cvs. Riesling and Cabernet Sauvignon) within the VineyardFACE (Free-Air Carbon Dioxide enrichment) experiment, using must analysis and non-invasive fluorescence sensor technology. Berry development was examined on five dates over three consecutive years by analyzing total soluble solids (TSS), pH, total acidity, organic acids, nutrition status, and non-invasive Multiplex measurements. Before harvest, secondary bunches were collected to examine bunch and berry parameters. Results showed that eCO2 had little impact on berry composition of Riesling and Cabernet Sauvignon during berry development, which could be related to bunch structure or single berry weight within single seasons. Elevated CO2 (eCO2) did not result in modified TSS accumulation during ripening but was directly related to the chlorophyll index SFR_R. Higher single berry weights (SBW), higher malic acid (MA), and lower tartaric acid (TAA) were examined at some stages during development of berries under eCO2 levels. Our study provides evidence that eCO2 did alter some bunch and berry parameters without a negative impact on fruit quality.

Biofuels are expected to play a major role in meeting California's long-term energy needs, but many factors influence the commercial viability of the various feedstock and production technology options. We developed a spatially explicit analytic framework that integrates models of plant growth, crop adoption, feedstock location, transportation logistics, economic impact, biorefinery costs and biorefinery energy use and emissions. We used this framework to assess the economic potential of hybrid poplar as a feedstock for jet fuel production in Northern California. Results suggest that the region has sufficient suitable croplands (2.3 million acres) and nonarable lands (1.5 million acres) for poplar cultivation to produce as much as 2.26 billion gallons of jet fuel annually. However, there are major obstacles to such large-scale production, including, on nonarable lands, low poplar yields and broad spatial distribution and, on croplands, competition with existing crops. We estimated the production cost of jet fuel to be $4.40 to $5.40 per gallon for poplar biomass grown on nonarable lands and $3.60 to $4.50 per gallon for biomass grown on irrigated cropland; the current market price is $2.12 per gallon. Improved poplar yields, use of supplementary feedstocks at the biorefinery and economic supports such as carbon credits could help to overcome these barriers.

ADHD is a psychiatric disorder which is characterized by hyperactivity, impulsivity and attention problems. Due to recent findings of microbial involvement in other psychiatric disorders like autism and depression, a role of the gut microbiota in ADHD pathogenesis is assumed but has not yet been investigated. In this study, the gut microbiota of 14 male ADHD patients (mean age: 11.9 yrs.) and 17 male controls (mean age: 13.1 yrs.) was examined via next generation sequencing of 16S rDNA and analyzed for diversity and biomarkers. We found that the microbial diversity (alpha diversity) was significantly decreased in ADHD patients compared to controls (pShannon = 0.036) and that the composition (beta diversity) differed significantly between patients and controls (pANOSIM = 0.033, pADONIS = 0.006, pbetadisper = 0.002). In detail, the bacterial family Prevotellacae was associated with controls, while patients with ADHD showed elevated levels of Bacteroidaceae, and both Neisseriaceae and Neisseria spec. were found as possible biomarkers for juvenile ADHD. Our results point to a possible link of certain microbiota with ADHD, with Neisseria spec. being a very promising ADHD-associated candidate. This finding provides the basis for a systematic, longitudinal assessment of the role of the gut microbiome in ADHD, yielding promising potential for both prevention and therapeutic intervention.

The current demographic development of our society results in an increasing number of elderly patients with chronic diseases being treated in the intensive care unit. A possible long-term consequence of such a treatment is that patients remain dependent on certain invasive organ support systems, such as long-term ventilator dependency. The main goal of this project is to define the transition process between in-hospital and out of hospital (ambulatory) ventilator support. A further goal is to identify evidence-based quality indicators to help define and describe this process.This project describes an ideal sequence of processes (process chain), based on the current evidence from the literature. Besides the process chain, key data and quality indicators were described in detail. Due to the limited project timeline, these indicators were not extensively tested in the clinical environment.The results of this project may serve as a solid basis for proof of feasibility and proof of concept investigations, optimize the transition process of ventilator-dependent patients from a clinical to an ambulatory setting, as well as reduce the rate of emergency re-admissions.

In field applications currently used for health monitoring and nondestructive testing, ultrasonic transducers primarily employ PZT5-H as the piezoelectric element for ultrasound transmission and detection. This material has a Curie−Weiss temperature that limits its use to about 210 °C. Some industrial applications require much higher temperatures, i.e., 1000−1200 °C and possible nuclear radiation up to 1020 n/cm2 when performance is required in a reactor environment. The goal of this paper is the survey and review of piezoelectric elements for use in harsh environments for the ultimate purpose for structural health monitoring (SHM), non-destructive evaluation (NDE) and material characterization (NDMC). The survey comprises the following categories: 1. High-temperature applications with single crystals, thick-film ceramics, and composite ceramics, 2. Radiation-tolerant materials, and 3. Spray-on transducers for harsh-environment applications. In each category the known characteristics are listed, and examples are given of performance in harsh environments. Highlighting some examples, the performance of single-crystal lithium niobate wafers is demonstrated up to 1100 °C. The wafers with the C-direction normal to the wafer plane were mounted on steel cylinders with high-temperature Sauereisen and silver paste wire mountings and tested in air. In another example, the practical use in harsh radiation environments aluminum nitride (AlN) was found to be a good candidate operating well in two different nuclear reactors. The radiation hardness of AlN was evident from the unaltered piezoelectric coefficient after a fast and thermal neutron exposure in a nuclear reactor core (thermal flux = 2.12 × 1013 ncm−2; fast flux 2 (>1.0 MeV) = 4.05 × 1013 ncm−2; gamma dose rate: 1 × 109 r/h; temperature: 400−500 °C). Additionally, some of the high-temperature transducers are shown to be capable of mounting without requiring coupling material. Pulse-echo signal amplitudes (peak-to-peak) for the first two reflections as a function of the temperature for lithium niobate thick-film, spray-on transducers were observed to temperatures of about 900 °C. Guided-wave send-and-receive operation in the 2−4 MHz range was demonstrated on 2−3 mm thick Aluminum (6061) structures for possible field deployable applications where standard ultrasonic coupling media do not survive because of the harsh environment. This approach would benefit steam generators and steam pipes where temperatures are above 210 °C. In summary, there are several promising approaches to ultrasonic transducers for harsh environments and this paper presents a survey based on literature searches and in-house laboratory observations.

The Ca2+-sensor synaptotagmin-1 that triggers neuronal exocytosis binds to negatively charged membrane lipids (mainly phosphatidylserine (PtdSer) and phosphoinositides (PtdIns)) but the molecular details of this process are not fully understood. Using quantitative thermodynamic, kinetic and structural methods, we show that synaptotagmin-1 (from Rattus norvegicus and expressed in Escherichia coli) binds to PtdIns(4,5)P2 via a polybasic lysine patch in the C2B domain, which may promote the priming or docking of synaptic vesicles. Ca2+ neutralizes the negative charges of the Ca2+-binding sites, resulting in the penetration of synaptotagmin-1 into the membrane, via binding of PtdSer, and an increase in the affinity of the polybasic lysine patch to phosphatidylinositol-4,5-bisphosphate (PtdIns(4,5)P2). These Ca2+-induced events decrease the dissociation rate of synaptotagmin-1 membrane binding while the association rate remains unchanged. We conclude that both membrane penetration and the increased residence time of synaptotagmin-1 at the plasma membrane are crucial for triggering exocytotic membrane fusion.

Purple carrots are rich in anthocyanins which are interesting as natural dyes in food and beverages. It is, thus, relevant to increase the concentration of anthocyanins by agricultural practices. We tested whether the combination of reduced nitrogen (N) supply and extended harvesting periods maximized the anthocyanin concentration of purple carrot roots, ideally without reducing their yield. The carrot variety ‘Deep Purple’ was grown with total N supplies of 220 kg N ha−1 (controls) and 73 kg N ha−1 (reduced N), respectively. Upon harvests in September, October and November, root yield and quality were assessed. Concentrations of chlorophylls (leaves) and anthocyanins (roots and leaves) were determined by spectroscopic and chemical analyses, and carbon and N content were quantified. Reduced N supply neither affected leaf or root biomass nor their chemical composition. Later harvests did not impact the yield of roots, but increased their diameter by 8.5–20%. Additionally, the anthocyanin concentrations of the roots increased by 40–50% in the controls, but not in N-limited plants, at late harvests. Consequently, extending the harvesting period might increase the anthocyanin concentration in roots of ‘Deep Purple’. Moreover, N supply might be reduced for this carrot variety without negative effects on root yield.

The main objective of this study was to determine growth, yield and fruit quality of grapevines under organic and biodynamic management in relation to integrated viticultural practices. Furthermore, the mechanisms for the observed changes in growth, yield and fruit quality were investigated by determining nutrient status, physiological performance of the plants and disease incidence on bunches in three consecutive growing seasons. A field trial (Vitis vinifera L. cv. Riesling) was set up at Hochschule Geisenheim University, Germany. The integrated treatment was managed according to the code of good practice. Organic and biodynamic plots were managed according to Regulation (EC) No 834/2007 and Regulation (EC) No 889/2008 and according to ECOVIN- and Demeter-Standards, respectively. The growth and yield of the grapevines differed strongly among the different management systems, whereas fruit quality was not affected by the management system. The organic and the biodynamic treatments showed significantly lower growth and yield in comparison to the integrated treatment. The physiological performance was significantly lower in the organic and the biodynamic systems, which may account for differences in growth and cluster weight and might therefore induce lower yields of the respective treatments. Soil management and fertilization strategy could be responsible factors for these changes. Yields of the organic and the biodynamic treatments partially decreased due to higher disease incidence of downy mildew. The organic and the biodynamic plant protection strategies that exclude the use of synthetic fungicides are likely to induce higher disease incidence and might partially account for differences in the nutrient status of vines under organic and biodynamic management. Use of the biodynamic preparations had little influence on vine growth and yield. Due to the investigation of important parameters that induce changes especially in growth and yield of grapevines under organic and biodynamic management the study can potentially provide guidance for defining more effective farming systems.

Ancillary to decline in cognitive abilities, patients with Alzheimer's disease (AD) frequently suffer from behavioural and psychological symptoms of dementia (BPSD). Hypothalamic polypeptides such as melanin-concentrating hormone (MCH) and hypocretin-1 (HCRT-1, orexin-A) are promoters of sleep-wake regulation and energy homeostasis and are found to impact on cognitive performance. California Agriculture - All Issues

Biomass constitutes a major renewable energy resource for California, with more than 30 million tons per year of in-state production estimated to be available on a sustainable basis for electricity generation, biofuels production and other industrial processing. Annually, biofuel production from these resources could exceed 2 billion gallons of gasoline equivalent, while providing opportunities for agricultural and rural economic development. Continuing research and large-scale demonstrations now under way will test alternative technologies and provide much-needed information regarding costs and environmental performance. Biomass can help meet state goals for increasing the amounts of electricity and fuels from renewable resources under the Renewable Portfolio Standard (RPS) and the Low Carbon Fuel Standard (LCFS), and can similarly help meet national biofuel targets under the federal Renewable Fuel Standard (RFS). Internationally consistent sustainability standards and practices are needed to inform policy and provide direction and guidance to industry.

IntroductionThe sensitivity of mammography screening is lower in women with dense breast. Increasing the efficacy of breast cancer screening have received special attention recently. The automated breast ultrasound (ABUS) shows promising results to complement mammography. Our aim was to expand the existing breast cancer screening protocol with ABUS within a Hungarian pilot project.MethodsFirst, we developed a protocol for the screening process focusing on integrating ABUS to the current practice. Consensus among clinical experts was achieved considering information from the literature and the actual opportunities of the hospital. Then we developed a protocol for evaluation that ensures systematic data collection and monitoring of screening with mammography and ABUS. We identified indicators based on international standards and adapted them to local setting. We considered their feasibility from the data source and timeframe perspective. The protocol was developed in a partnership of researchers, clinicians and hospital managers.ResultsThe process of screening activity was described in a detailed flowchart. Human and technological resource requirements and communication activities were defined. We listed 23 monitoring indicators to evaluate the screening program and checked the feasibility to calculate these indicators based on local data collection and other sources. Partnership between researchers experienced in planning and evaluating screening programs, interested clinicians, and hospital managers resulted in a locally implementable, evidence-based screening protocol.DiscussionThe experience and knowledge gained on the implementation of the ABUS technology could generate real-world data to support the decision on using the technology at national level.

Atmospheric carbon dioxide (CO2) concentration has continuously increased since pre-industrial times and has currently reached an average growth rate of 2.3 ppm per year. For the majority of plant species elevated CO2 (eCO2) improves photosynthesis and thus plant biomass production. To investigate the effects of eCO2 on leaf physiology and morphological leaf characteristics two Vitis vinifera L. cultivars, Riesling and Cabernet Sauvignon, grown in the VineyardFACE (Free Air Carbon dioxide Enrichment) system were used. The VineyardFACE is located at Geisenheim, Rheingau comparing future atmospheric CO2-concentrations (eCO2, predicted for the mid-21st century) with current ambient CO2-conditions (aCO2). Experiments were operated under rain-fed conditions for two consecutive years (2015 and 2016). For both varieties and CO2 treatments, leaf gas exchange measurements were performed as well as measures of epidermal flavonoid (Flav) and leaf chlorophyll (Chl) indices by using a portable leaf clip. Furthermore, leaves were sampled for spectrophotometric analysis of the leaf pigments chlorophyll a (Chl a), chlorophyll b (Chl b) and carotenoid (Car). Additionally, leaf cross-sections were produced as permanent preparations to investigate morphological characteristics of the leaf structure. Both cultivars did not differ in leaf chlorophyll meter readings or leaf pigments between the two CO2 treatments while net assimilation was highly stimulated under elevated CO2 for both seasons. Differences found in leaf cross-sections were detected in palisade parenchyma and epidermal thickness of Cabernet Sauvignon under eCO2, whereas Riesling net assimilation increased by 40% under a 20% CO2 enrichment while remaining unaffected in different leaf layer thickness. The observed results within grapevine leaf tissues provide insights to seasonal adaptation strategies of grapevines under elevated CO2 concentrations predicted in future.

Climate change and rising atmospheric carbon dioxide concentration are a concern for agriculture, including viticulture. Studies on elevated carbon dioxide have already been conducted on grapevines, mainly taking place in greenhouses using potted plants or using field-grown vines under instant and higher CO2 enrichment, i.e., > 650 ppm. The VineyardFACE, located at Hochschule Geisenheim University, is an open field Free Air CO2 Enrichment (FACE) experimental set-up designed to study the effects of elevated carbon dioxide using adapted, field-grown vines (Vitis vinifera L. cv. Cabernet-Sauvignon). As the carbon dioxide fumigation started in 2014, the long-term effects of elevated carbon dioxide treatment can be investigated on berry ripening parameters and fruit metabolic composition. The present study investigates the effect on fruit composition under a moderate increase (+20 %; eCO2) of carbon dioxide concentration, as predicted for 2050 on Cabernet-Sauvignon. Berry growth, ripening dynamics and composition were determined and primary (sugars, organic acids, amino acids) and secondary metabolites (anthocyanins) were analysed. Compared to previous results of the early adaptive phase of the vines (Wohlfahrt et al., 2020), our results show little effects of eCO2 treatment on primary metabolites composition in berries. However, total anthocyanins concentration in berry skin was lower for eCO2 treatment in the hot and dry season of 2020, although the ratio between anthocyanins derivatives did not differ.

Many multicellular organisms specify germ cells during early embryogenesis by the inheritance of ribonucleoprotein (RNP) granules known as germplasm. However, the role of complex interactions of RNP granules during germ cell specification remains elusive. This study characterizes the interaction of RNP granules, Buc, and zebrafish Vasa (zfVasa) during germ cell specification. We identify a novel zfVasa-binding motif (Buc-VBM) in Buc and a Buc-binding motif (zfVasa-BBM) in zfVasa. Moreover, we show that Buc and zfVasa directly bind in vitro and that this interaction is independent of the RNA. Our circular dichroism spectroscopy data reveal that the intrinsically disordered Buc-VBM peptide forms alpha-helices in the presence of the solvent trifluoroethanol. Intriguingly, we further demonstrate that Buc-VBM enhances zfVasa ATPase activity, thereby annotating the first biochemical function of Buc as a zfVasa ATPase activator. Collectively, these results propose a model in which the activity of zfVasa is a central regulator of primordial germ cell (PGC) formation and is tightly controlled by the germplasm organizer Buc.

Biomass constitutes a major renewable energy resource for California, with more than 30 million tons per year of in-state production estimated to be available on a sustainable basis for electricity generation, biofuels production and other industrial processing. Annually, biofuel production from these resources could exceed 2 billion gallons of gasoline equivalent, while providing opportunities for agricultural and rural economic development. Continuing research and large-scale demonstrations now under way will test alternative technologies and provide much-needed information regarding costs and environmental performance. Biomass can help meet state goals for increasing the amounts of electricity and fuels from renewable resources under the Renewable Portfolio Standard (RPS) and the Low Carbon Fuel Standard (LCFS), and can similarly help meet national biofuel targets under the federal Renewable Fuel Standard (RFS). Internationally consistent sustainability standards and practices are needed to inform policy and provide direction and guidance to industry.

Besides transketolase (TKT), a thiamin-dependent enzyme of the pentose phosphate pathway, the human genome encodes for two closely related transketolase-like proteins, which share a high sequence identity with TKT. Transketolase-like protein 1 (TKTL1) has been implicated in cancerogenesis as its cellular expression levels were reported to directly correlate with invasion efficiency of cancer cells and patient mortality. It has been proposed that TKTL1 exerts its function by catalyzing an unusual enzymatic reaction, a hypothesis that has been the subject of recent controversy. The most striking difference between TKTL1 and TKT is a deletion of 38 consecutive amino acids in the N-terminal domain of the former, which constitute part of the active site in authentic TKT. Our structural and sequence analysis suggested that TKTL1 might not possess transketolase activity. In order to test this hypothesis in the absence of a recombinant expression system for TKTL1 and resilient data on its biochemical properties, we have engineered and biochemically characterized a "pseudo-TKTL1" Δ38 deletion variant of human TKT (TKTΔ38) as a viable model of TKTL1. Although the isolated protein is properly folded under in vitro conditions, both thermal stability as well as stability of the TKT-specific homodimeric assembly are markedly reduced. Circular dichroism and NMR spectroscopic analysis further indicates that TKTΔ38 is unable to bind the thiamin cofactor in a specific manner, even at superphysiological concentrations. No transketolase activity of TKTΔ38 can be detected for conversion of physiological sugar substrates thus arguing against an intrinsically encoded enzymatic function of TKTL1 in tumor cell metabolism.
