This study sought to better understand how time of day (ToD) or turgor pressure might affect the flexural stiffness of sweet sorghum stalks and potentially regulate stalk lodging resistance. Stalk flexural stiffness measured across a 48 h period in 2019 showed a significant diurnal association with leaf water potential and stalk flexural stiffness. While the correlation between stalk flexural stiffness and this proxy for internal turgor status was statistically significant, it only accounted for roughly 2% of the overall variance in stiffness. Given that turgor status is a dynamic rather than fixed physiological variable like the cellular structure, these data suggest that internal turgor plays a small yet significant role in influencing the flexural stiffness of fully mature stalks prior to a stalk lodging event. The association was assessed at earlier developmental stages across three distinct cultivars and found not to be significant. Panicle weight and stalk basal weight, but not stalk Brix or water content, were found to be better predictors of stalk flexural stiffness than either ToD or turgor status. Observation across three cultivars and four distinct developmental stages ranging from the vegetative to the hard-dough stages suggests that stalk flexural stiffness changes significantly as a function of time. However, neither ToD nor turgor status appear to meaningfully contribute to observed variations in stalk flexural stiffness in either individual stalks or across larger populations. As turgor status was not found to meaningfully influence stalk strength or flexural stiffness at any developmental time point examined in any of the three sweet sorghum cultivars under study, turgor pressure likely offers only inconsequential contributions to the biomechanics underlying sweet sorghum stalk lodging resistance.

Neck shrivel is a fruit disorder of european plum (Prunus domestica L.). We investigate whether an asymmetrical distribution of osmolytes might explain the observations of a turgid stylar end and a flaccid stem end, in a selection of 17 plum cultivars sourced from two sites. The osmotic potential (ΨΠ) of the juices expressed from stem or stylar end fruit samples decreased (became more negative) during development. The cell turgor (ΨP) slightly increased during development up to 352 ± 42 kPa at 78 days after full bloom (DAFB) in the stem end and up to 331 ± 51 kPa at 92 DAFB in the stylar end, and then decreased. At maturity, ΨP averaged 22 ± 3 kPa in the stem end and 22 ± 4 kPa in the stylar end. These values are negligibly low compared with the very negative values of ΨΠ in the stylar end (−3188 ± 73 kPa) and stem end (−3060 ± 74 kPa). There was a transient gradient in ΨΠ between stylar end and stem end that almost disappeared by maturity. Marked differences in ΨΠ and its distribution were measured across 17 cultivars. In the majority (14), ΨΠ was more negative at the stylar end than at the stem end. A more negative ΨΠ in the stem was only detected in ‘Aprimira’, ‘Topfive’, and ‘Tophit’. Our results demonstrate that cell ΨP is very low and is essentially independent of ΨΠ in developing european plums. In most cultivars, ΨΠ in the stylar end is more negative than in the stem end. The absence of an axial gradient in ΨP and the small differences in ΨΠ between the stem and stylar end make both factors unlikely candidates for explaining neck shrivel.

Why this research Matters The inner bark plays important roles in tree stems, including radial exchange of water with the xylem and translocation of carbohydrates. Both processes affect the water content and the thickness of the inner bark on a diurnal basis. For the first time, we simultaneously measured the diurnal variations in the inner bark thickness of hinoki cypress (Chamaecyparis obtusa) by using point dendrometers and those of local xylem potential by using stem psychrometers located next to the dendrometers to determine how these variations were related to each other, to phloem turgor and carbohydrate transport. We also estimated the axial hydrostatic pressure gradient by measuring the osmolality of the sap extracted from the inner bark. The inner bark shrunk during the day and swelled during the night with an amplitude related to day‐to‐day and seasonal variations in climate. The relationship between changes in xylem water potential and inner bark thickness exhibited a hysteresis loop during the day with a median lag of 2 h. A phloem turgor‐related signal can be retrieved from the diurnal variations in the inner bark thickness, which was higher at the upper than at the lower position along the trunk. However, a downward hydrostatic pressure gradient was only observed at dawn, suggesting diurnal variations in the phloem sap flow velocity.

This study deals with the best responses of a diverse collection of grapevine genotypes to osmotic stress associated with source-sink balance responses given by an estimator such as leaf area to fruit ratio. ‘Centennial Seedless’, a drought tolerant cultivar, was selected as control. The cultivars, ‘Victoria’ and ‘Argessis’, were chosen as a repetition from previous research dealing with pollen grain test, two years ago. Ten genotypes were hybrid elites in first and second hybrid generations. Three cultivars ‘Victoria’, ‘Centennial Seedless’, and ‘Argessis’ were grown under field conditions in containers, and in the soil under greenhouse conditions. Significant differences were found between genotypes for both responses to osmotic stress and source-sink balance. ‘Centennial Seedless’ and ‘BP9’ hybrid showed the best responses of induced osmotic adjustment; results confirmed the compensatory potassium uptake theory. ‘Victoria’ and ‘Argessis’ had almost the same average values as ‘Centennial Seedless’ osmotic estimator for induced osmotic adjustment. ‘Victoria’ and ‘HR7’ hybrid showed an increase in osmotic stress in the cell, after application of polyethylene glycol solutions without potassium cation and a lower source-sink ratio, which could be associated with higher photosynthesis rates. No correlations were identified between the mechanisms expressed by the analyzed estimators, indicating that they are activated and functional separately from each other, sometimes only compensatory.

The ability of plants to absorb CO2 for photosynthesis and transport water from root to shoot depends on the reversible swelling of guard cells that open stomatal pores in the epidermis. Despite decades of experimental and theoretical work, the biomechanical drivers of stomatal opening and closure are still not clearly defined. We combined mechanical principles with a growing body of knowledge concerning water flux across the plant cell membrane and the biomechanical properties of plant cell walls to quantitatively test the long-standing hypothesis that increasing turgor pressure resulting from water uptake drives guard cell expansion during stomatal opening. To test the alternative hypothesis that water influx is the main motive force underlying guard cell expansion, we developed a system dynamics model accounting for water influx. This approach connects stomatal kinetics to whole plant physiology by including values for water flux arising from water status in the plant .

Pre-anthesis drought is expected to greatly increase yield losses in wheat (Triticum aestivum L.), one of the most important crops worldwide. Most studies investigate the effects of pre-anthesis drought only at maturity. The physiology of the plant before anthesis and how it is affected during drought is less studied. Our study focused on physiological patterns in wheat plants during pre- and post-anthesis drought. To this end, we measured leaf xylem water potential, osmotic potential and water content in different plant parts at a high temporal frequency: every 3 days, three times a day. The experiment started just before booting until 2 weeks after flowering. Drought stress was induced by withholding irrigation with rewatering upon turgor loss, which occurred once before and once after anthesis. The goal was to investigate the patterns of osmotic adjustment, when it is used for protection against drought, and if the strategy changes during the phenological development of the plant. Our data gave no indication of daily osmotic adjustment, but did show a delicate control of the osmotic potential during drought in both leaves and stem. Under high drought stress, osmotic potential decreased to avoid further water loss. Before anthesis, rewatering restored leaf water potential and osmotic potential quickly. After anthesis, rewatering restored water potential in the flag leaves, but the osmotic potential in the stem and flag leaf remained low longer. Osmotic adjustment was thus maintained longer after anthesis, showing that the plants invest more energy in the osmotic adjustment after anthesis than before anthesis. We hypothesize that this is because the plants consider the developing ear after anthesis a more important carbohydrate sink than the stem, which is a carbohydrate sink before anthesis, to be used later as a reserve. Low osmotic potential in the stem allowed turgor maintenance, while the low osmotic potential in the flag leaf led to an increase in leaf turgor beyond the level of the control plants. This allowed leaf functioning under drought and assured that water was redirected to the flag leaf and not used to refill the stem storage.

Cell division and cell elongation are fundamental processes for growth. In contrast to animal cells, plant cells are surrounded by rigid walls and therefore loosening of the wall is required during elongation. On the other hand, vacuole size has been shown to correlate with cell size and inhibition of vacuolar expansion limits cell growth. However, the specific role of the vacuole during cell elongation is still not fully resolved. Especially the question whether the vacuole is the leading unit during cellular growth or just passively expands upon water uptake remains to be answered. Here, we review recent findings about the contribution of the vacuole to cell elongation. In addition, we also discuss the connection between cell wall status and vacuolar morphology. In particular, we focus on the question whether vacuolar size is dictated by cell size or vice versa and share our personnel view about the sequential steps during cell elongation.

For more than ten years, Central Chile has faced drought conditions, which impact crop production and quality, increasing food security risk. Under this scenario, implementing management practices that allow increasing water use efficiency is urgent. The study was carried out on kiwifruit trees, located in the O’Higgins region, Chile for season 2018–2019 and 2019–2020. We evaluate the time-series of nine vegetation indices in the VNIR and SWIR regions derived from Sentinel-2 (A/B) satellites to establish how much variability in the canopy water status there was. Over the study’s site, eleven sensors were installed in five trees, which continuously measured the leaf’s turgor pressure (Yara Water-Sensor). A strong Spearman’s (ρ) correlation between turgor pressure and vegetation indices was obtained, having −0.88 with EVI and −0.81 with GVMI for season 2018–2019, and lower correlation for season 2019–2020, reaching −0.65 with Rededge1 and −0.66 with EVI. However, the NIR range’s indices were influenced by the vegetative development of the crop rather than its water status. The red-edge showed better performance as the vegetative growth did not affect it. It is necessary to expand the study to consider higher variability in kiwifruit’s water conditions and incorporate the sensitivity of different wavelengths.

This study aims to identify the histological basis for the extraordinary, fast movement of the style in Marantaceae. Although this explosive pollination mechanism was subject of many studies, quantitative measurements to document volumetric changes have never been conducted. Based on physical parameters and limitations (poroelastic time), the movement itself is by far too fast to be explained by turgor changes solely. Therefore, we address the hypothesis that the style contains elastic structures to store energy allowing the fast movement. We provide an experimental approach in Goeppertia bachemiana to identify histological differences of styles in various states, i.e., steady, unreleased, and released state. Cross and longitudinal sections were used to reconstruct the cell volume in different sectors of the style. Histological data were discussed with respect to a putative water shift (turgor movement) and elastic instabilities that were proposed to explain the style movement of Marantaceae. Current data show, that the upper epidermis is under tensile stress in the unreleased state. After style release, the lower side of the style revealed an enormous water up-take. According to our results, we hypothesize that the fast style movement of G. bachemiana is likely based on an elastically stretched upper epidermis, whereas a “soaking tissue” at the lower side presumably mediates the up curling of the style. The experimental data show that at least for G. bachemiana, physical limitations such as the poroelastic time are suitable parameters to predict movements that are based on elastic instabilities.

Vascular epiphytes are a major biomass component of forests across the globe and they contribute to 9% of global vascular plant diversity. To improve our understanding of the whole-plant response of epiphytes to future climate change, we investigated for the first time both individual and combined effects of elevated CO2 (560 ppm) and light on the physiology and growth of two epiphyte species [Tillandsia brachycaulos (CAM) and Phlebodium aureum (C3)] grown for 272 days under controlled conditions. We found that under elevated CO2 the difference in water loss between the light (650 μmol m-2s-1) and shade (130 μmol m-2s-1) treatment was strongly reduced. Stomatal conductance (gs) decreased under elevated CO2, resulting in an approximate 40–45% reduction in water loss over a 24 h day/night period under high light and high CO2 conditions. Under lower light conditions water loss was reduced by approximately 20% for the CAM bromeliad under elevated CO2 and increased by approximately 126% for the C3 fern. Diurnal changes in leaf turgor and water loss rates correlated strong positively under ambient CO2 (400 ppm) and high light conditions. Future predicted increases in atmospheric CO2 are likely to alter plant water-relations in epiphytes, thus reducing the canopy cooling potential of epiphytes to future increases in temperature.

Results of researches of various cultivars and hybrids of tangerine growing in the conditions of the humid subtropics of Russia and possessing valuable physiological and biochemical markers are presented in article. The stress factors limiting cultivation of tangerine culture in Krasnodar region are defined. The assessment of the water saving and enzymes activity in tangerine leaves is given. Changes of physiological parameters at influence of a stressful factor are shown. The analysis of water deficiency is revealed existence of essential distinctions between cultivars and hybrids. Change of thickness of a leaf blade of tangerine is the important diagnostic characteristic testifying the ability of leaf tissues to accumulate and keep water. Researches are showing that difference between catalase action of cultivars and hybrids are insignificant. Strengthening of enzyme activity can testify to bigger violation of a functional state of plant cultivars in comparison with the hybrides. Water deficiency and drought resistance coefficient can be used for diagnostics of stability of tangerine to hydrothermal factors. Correlation coefficients between the studied markers were calculated and regression models of their interrelation were given. So, there was a high correlation between thickness of a leaf blade and water deficiency index; close correlation between water deficiency and thickness of a leaf blade, as well as dependence of average degree on water deficiency and catalase activity were noted. There were revealed tangerine cultivars and hybrids which are stable in in the changing environmental conditions and have high biochemical value of fruits.

The Candida albicans RHR2 gene, which specifies a glycerol biosynthetic enzyme, is required for biofilm formation in vitro and in vivo. Prior studies indicate that RHR2 is ultimately required for expression of adhesin genes, such as ALS1. In fact, RHR2 is unnecessary for biofilm formation when ALS1 is overexpressed from an RHR2-independent promoter. Here, we describe two additional biological processes that depend upon RHR2: invasion into an abiotic substrate and pathogenicity in an abdominal infection model. We report here that abiotic substrate invasion occurs concomitantly with biofilm formation, and a screen of transcription factor mutants indicates that biofilm and hyphal formation ability correlates with invasion ability. However, analysis presented here of the rhr2Δ/Δ mutant separates biofilm formation and invasion. We found that an rhr2Δ/Δ mutant forms a biofilm upon overexpression of the adhesin gene ALS1 or the transcription factor genes BRG1 or UME6. However, the biofilm-forming strains do not invade the substrate. These results indicate that RHR2 has an adhesin-independent role in substrate invasion, and mathematical modeling argues that RHR2 is required to generate turgor. Previous studies have shown that abdominal infection by C. albicans has two aspects: infection of abdominal organs and persistence in abscesses. We report here that an rhr2Δ/Δ mutant is defective in both of these infection phenotypes. We find here that overexpression of ALS1 in the mutant restores infection of organs, but does not improve persistence in abscesses. Therefore, RHR2 has an adhesin-independent role in abdominal infection, just as it does in substrate invasion. This report suggests that RHR2, through glycerol synthesis, coordinates adherence with host- or substrate-interaction activities that enable proliferation of the C. albicans population.

Circadian leaf movements are widely known in plants, but nocturnal movement of tree branches were only recently discovered by using terrestrial laser scanning (TLS), a high resolution three-dimensional surveying technique. TLS uses a pulsed laser emitted in a regular scan pattern for rapid measurement of distances to the targets, thus producing three dimensional point cloud models of sub-centimeter resolution and accuracy in a few minutes. Here, we aim to gain an overview of the variability of circadian movement of small trees across different taxonomic groups, growth forms and leaf anatomies. We surveyed a series of 18 full scans over a 12-h night period to measure nocturnal changes in shape simultaneously for an experimental setup of 22 plants representing different species. Resulting point clouds were evaluated by comparing changes in height percentiles of laser scanning points belonging to the canopy. Changes in crown shape were observed for all studied trees, but clearly distinguishable sleep movements are apparently rare. Ambient light conditions were continuously dark between sunset (7:30 p.m.) and sunrise (6:00 a.m.), but most changes in movement direction occurred during this period, thus most of the recorded changes in crown shape were probably not controlled by ambient light. The highest movement amplitudes, for periodic circadian movement around 2 cm were observed for Aesculus and Acer, compared to non-periodic continuous change in shape of 5 cm for Gleditschia and 2 cm for Fargesia. In several species we detected 2–4 h cycles of minor crown movement of 0.5–1 cm, which is close to the limit of our measurement accuracy. We present a conceptual framework for interpreting observed changes as a combination of circadian rhythm with a period close to 12 h, short-term oscillation repeated every 2–4 h, aperiodic continuous movement in one direction and measurement noise which we assume to be random. Observed movement patterns are interpreted within this framework, and connections with morphology and taxonomy are proposed. We confirm the existence of overnight “sleep” movement for some trees, but conclude that circadian movement is a variable phenomenon in plants, probably controlled by a complex combination of anatomical, physiological, and morphological factors.

Morphogenesis in plants is directly linked to the mechanical elements of growing tissues, namely cell wall and inner cell pressure. Studies of these structural elements are now often performed using indentation methods such as atomic force microscopy. In these methods, a probe applies a force to the tissue surface at a subcellular scale and its displacement is monitored, yielding force-displacement curves that reflect tissue mechanics. However, the interpretation of these curves is challenging as they may depend on not only the cell probed, but also on neighboring cells, or even on the whole tissue. Here, we build a realistic three-dimensional model of the indentation of a flower bud using SOFA (Simulation Open Framework Architecture), in order to provide a framework for the analysis of force-displacement curves obtained experimentally. We find that the shape of indentation curves mostly depend on the ratio between cell pressure and wall modulus. Hysteresis in force-displacement curves can be accounted for by a viscoelastic behavior of the cell wall. We consider differences in elastic modulus between cell layers and we show that, according to the location of indentation and to the size of the probe, force-displacement curves are sensitive with different weights to the mechanical components of the two most external cell layers. Our results confirm most of the interpretations of previous experiments and provide a guide to future experimental work.

Asexual spores of the rice blast fungus germinate to produce a specialized and melanized infection structure, the appressorium, which is pivotal to successful plant penetration. To investigate whether Magnaporthe grisea counteracts the toxic burst of H2O2 localized beneath the site of attempted invasion, we examined the temporal expression of five candidate antioxidant genes. Of these, the putatively secreted large subunit catalase CATB gene was 600-fold up-regulated in vivo, coincident with penetration, and moderately up-regulated in vitro, in response to exogenous H2O2. Targeted gene replacement of CATB led to compromised pathogen fitness; the catB mutant displayed paler pigmentation and accelerated hyphal growth but lower biomass, poorer sporulation, fragile conidia and appressoria, and impaired melanization. The catB mutant was severely less pathogenic than Guy 11 on barley and rice, and its infectivity was further reduced on exposure to H2O2. The wild-type phenotype was restored by the reintroduction of CATB into the catB mutant. We found no evidence to support a role for CATB in detoxification of the host-derived H2O2 at the site of penetration. Instead, we demonstrated that CATB plays a part in strengthening the fungal wall, a role of particular importance during forceful entry into the host.
