People are exposed to microplastics (MPs) on a large scale in everyday life. However, it is not clear whether MPs can also be distributed and retained in certain tissues. Therefore, the development of analytical methods capable of detecting MPs in specific human organs/tissues is of utmost importance. In this study, the use and combination of spectroscopic techniques, namely Raman microspectroscopy and laser-induced breakdown spectroscopy (LIBS), was tested for the detection of polyethylene (PE) MPs in human tonsils. Preliminary results showed that Raman microspectroscopy was able to detect MPs down to 1 μm in size and LIBS down to 10 μm. In the next step, human tonsils were spiked with PE MPs, and digested. The filtered particles were analyzed using Raman microspectroscopy and LIBS, and complemented by X-ray fluorescence (XRF). The results showed that Raman microspectroscopy could reliably detect PE MPs in spiked human tonsils, while LIBS and XRF served as a reference analytical method to characterize particles that could not be classified by Raman microspectroscopy for their non-organic origin. The results of this study, supported by a current feasibility study conducted on clinical samples, demonstrated the reliability and feasibility of this approach for monitoring MPs in biotic samples.

Spinal cord injury results in significant sensorimotor deficits, currently, there is no curative treatment for the symptoms induced by spinal cord injury. Basic and pre-clinical research on spinal cord injury relies on the development and characterization of appropriate animal models. These models should replicate the symptoms observed in human, allowing for the exploration of functional deficits and investigation into various aspects of physiopathology of spinal cord injury. Non-human primates, due to their close phylogenetic association with humans, share more neuroanatomical, genetic, and physiological similarities with humans than rodents. Therefore, the responses to spinal cord injury in nonhuman primates most likely resemble the responses to traumatism in humans. In this review, we will discuss nonhuman primate models of spinal cord injury, focusing on in vivo assessments, including behavioral tests, magnetic resonance imaging, and electrical activity recordings, as well as ex vivo histological analyses. Additionally, we will present therapeutic strategies developed in non-human primates and discuss the unique specificities of non-human primate models of spinal cord injury.

Hyperspectral imaging is an emerging technology that has gained significant attention in the medical field due to its ability to provide precise and accurate imaging of biological tissues. The current methods of hyperspectral imaging, such as filter-wheel, snapshot, line-scanning, and push-broom cameras have limitations such as low spatial and spectral resolution, slow acquisition time. New developments on the field of light field cameras show the potential to overcome these limitations. In this paper, we use a novel hyperspectral lightfield camera and try to combine the capability of hyperspectral and 3D analysis. For this purpose we calibrate our system and test it during two ENT-surgeries to show its potential for improving surgical outcomes. The micro-lenses of the camera map 66 spectral sub-images onto the sensor allowing to reconstruct the spectral behavior of the captured scene in the spectral range of 350-1000nm. In addition, we use the sensor data to apply a 3D camera calibration pipeline to allow 3D surface reconstruction. We captured 26 calibration images and achieved calibration results in accordance to stated company data. The best calibration showed a re-projection error of 0.55 px. Further, we tested the camera during a parotidectomy and a neck-dissection. The extracted reflectance spectra of the selected venal and arterial regions correspond perfectly to the spectrum of oxygenated and deoxygenated hemoglobin. For the first time, up to our knowledge, a hyperspectral lightfield camera has been used during a surgery. We were able to continuously capture images and analyze the reconstructed spectra of specific tissue types. Further, we are able to use the sensor data of the micro-lens projections to calibrate the multilens camera system for later intraoperative measurement tasks.

In the commercial production of phalaenopsis orchids, the cultivation time after deflasking is used to describe the plant age and maturity. Carbon-to-nitrogen (C/N) ratio is often used as an indicator of plant growth and flowering potential. High C/N ratios are considered to promote reproductive growth, and low C/N ratios are associated with the early vegetative growth or even inhibiting flowering. This study investigated how plant age and maturity affected flowering ability and flower quality of phalaenopsis and their relationship to C/N ratio. The plant materials of various ages were the purple, small-flowered Phalaenopsis Sogo Lotte ‘F2510’ and white, large-flowered P. Sogo Yukidian ‘V3’, which were 2 to 7 months and 10 to 20 months after deflasking, respectively. Plants were placed under 25/20 °C for 4 months to force flowering and investigate the flowering-related parameters. The leaf C/N ratio of both varieties increased in general with the increase of plant age. The spiking (flower-stalk emergence) rate of P. Sogo Lotte ‘F2510’ 2 months after deflasking was only 42%, which indicates that these plants were not completely out of their juvenile phase, whereas that of those 3 to 7 months after deflasking was 100%, indicating that plants had acquired full flowering ability. No linear correlation was found between the C/N ratio and days to spiking, to first visible bud, to first flower open, and to 90% flower opening in the white, large-flowered P. Sogo Yukidian ‘V3’. However, there was a positive correlation between the C/N ratio and inflorescence length, flower-stalk diameter, first flower diameter, and flower count. Thus, the C/N ratio is feasible to be used as an indicator for assessing the flowering quality in phalaenopsis.

Direct mass spectrometry (MS) analysis of human tissue at the molecular level could gain insight into biomarker discovery and disease diagnosis. Detecting metabolite profiles of tissue sample play an important role in understanding the pathological properties of disease development. Because the complex matrices in tissue samples, complicated and time-consuming sample preparation processes are usually required by conventional biological and clinical MS methods. Direct MS with ambient ionization technique is a new analytical strategy for direct sample analysis with little sample preparation, and has been proven to be a simple, rapid, and effective analytical tools for direct analysis of biological tissues. In this work, we applied a simple, low-cost, disposable wooden tip (WT) for loading tiny thyroid tissue, and then loading organic solvents to extract biomarkers under electrospray ionization (ESI) condition. Under such WT-ESI, the extract of thyroid was directly sprayed out from wooden tip to MS inlet. In this work, thyroid tissue from normal and cancer parts were analyzed by the established WT-ESI-MS, showing lipids were mainly detectable compounds in thyroid tissue. The MS data of lipids obtained from thyroid tissues were further analyzed with MS/MS experiment and multivariate variable analysis, and the biomarkers of thyroid cancer were also investigated.

Training models with semi- or self-supervised learning methods is one way to reduce annotation effort since they rely on unlabeled or sparsely labeled datasets. Such approaches are particularly promising for domains with a time-consuming annotation process requiring specialized expertise and where high-quality labeled machine learning datasets are scarce, like in computational pathology. Even though some of these methods have been used in the histopathological domain, there is, so far, no comprehensive study comparing different approaches. Therefore, this work compares feature extractors models trained with state-of-the-art semi- or self-supervised learning methods PAWS, SimCLR, and SimSiam within a unified framework. We show that such models, across different architectures and network configurations, have a positive performance impact on histopathological classification tasks, even in low data regimes. Moreover, our observations suggest that features learned from a particular dataset, i.e., tissue type, are only in-domain transferable to a certain extent. Finally, we share our experience using each method in computational pathology and provide recommendations for its use.

Development of a novel in vivo lung perfusion (IVLP) procedure allows localized delivery of high-dose doxorubicin (DOX) for targeting residual micrometastatic disease in the lungs. However, DOX delivery via IVLP requires careful monitoring of drug level to ensure tissue concentrations of this agent remain in the therapeutic window. A small dimension nitinol wire coated with a sorbent of biocompatible morphology (Bio-SPME) has been clinically evaluated for in vivo lung tissue extraction and determination of DOX and its key metabolites. The in vivo Bio-SPME-IVLP experiments were performed on pig model over various (150 and 225 mg/m2) drug doses, and during human clinical trial. Two patients with metastatic osteosarcoma were treated with a single 5 and 7 μg/mL (respectively) dose of DOX during a 3-h IVLP. In both pig and human cases, DOX tissue levels presented similar trends during IVLP. Human lung tissue concentrations of drug ranged between 15 and 293 μg/g over the course of the IVLP procedure. In addition to DOX levels, Bio-SPME followed by liquid chromatography-mass spectrometry analysis generated 64 metabolic features during endogenous metabolite screening, providing information about lung status during drug administration. Real-time monitoring of DOX levels in the lungs can be performed effectively throughout the IVLP procedure by in vivo Bio-SPME chemical biopsy approach. Bio-SPME also extracted various endogenous molecules, thus providing a real-time snapshot of the physiology of the cells, which might assist in the tailoring of personalized treatment strategy.

Abstract Background T1 mapping using modified Look-Locker inversion recovery (MOLLI) provides quantitative information on myocardial tissue composition. T1 results differ between sites due to variations in hardware and software equipment, limiting the comparability of results. The aim was to test if Z-scores can be used to compare the results of MOLLI T1 mapping from different cardiovascular magnetic resonance (CMR) platforms. Methods First, healthy subjects (n = 15) underwent 11 combinations of native short-axis T1 mapping (four CMR systems from two manufacturers at 1.5 T and 3 T, three MOLLI schemes). Mean and standard deviation (SD) of septal myocardial T1 were derived for each combination. T1 maps were transformed into Z-score maps based on mean and SD values using a prototype post-processing module. Second, Z-score mapping was applied to a validation sample of patients with cardiac amyloidosis at 1.5 T (n = 25) or 3 T (n = 13). Results In conventional T1 analysis, results were confounded by variations in field strength, MOLLI scheme, and manufacturer-specific system characteristics. Z-score-based analysis yielded consistent results without significant differences between any two of the combinations in part 1 of the study. In the validation sample, Z-score mapping differentiated between patients with cardiac amyloidosis and healthy subjects with the same diagnostic accuracy as standard T1 analysis regardless of field strength. Conclusions T1 analysis based on Z-score mapping provides consistent results without significant differences due to field strengths, CMR systems, or MOLLI variants, and detects cardiac amyloidosis with the same diagnostic accuracy as conventional T1 analysis. Z-score mapping provides a means to compare native T1 results acquired with MOLLI across different CMR platforms.

Interpreting tissue architecture plays an important role in gaining a better understanding of healthy tissue development and disease. Novel molecular detection and imaging techniques make it possible to locate many different types of objects, such as cells and/or mRNAs, and map their location across the tissue space. In this review, we present several methods that provide quantification and statistical verification of observed patterns in the tissue architecture. We categorize these methods into three main groups: Spatial statistics on a single type of object, two types of objects, and multiple types of objects. We discuss the methods in relation to four hypotheses regarding the methods' capability to distinguish random and non-random distributions of objects across a tissue sample, and present a number of openly available tools where these methods are provided. We also discuss other spatial statistics methods compatible with other types of input data.

Histological changes in tissue are of primary importance in pathological research and diagnosis. Automated histological analysis requires ability to computationally separate pathological alterations from normal tissue. Conventional histopathological assessments are performed from individual tissue sections, leading to the loss of three-dimensional context of the tissue. Yet, the tissue context and spatial determinants are critical in several pathologies, such as in understanding growth patterns of cancer in its local environment. Here, we develop computational methods for visualization and quantitative assessment of histopathological alterations in three dimensions. First, we reconstruct the 3D representation of the whole organ from serial sectioned tissue. Then, we proceed to analyze the histological characteristics and regions of interest in 3D. As our example cases, we use whole slide images representing hematoxylin-eosin stained whole mouse prostates in a Pten+/- mouse prostate tumor model. We show that quantitative assessment of tumor sizes, shapes, and separation between spatial locations within the organ enable characterizing and grouping tumors. Further, we show that 3D visualization of tissue with computationally quantified features provides an intuitive way to observe tissue pathology. Our results underline the heterogeneity in composition and cellular organization within individual tumors. As an example, we show how prostate tumors have nuclear density gradients indicating areas of tumor growth directions and reflecting varying pressure from the surrounding tissue. The methods presented here are applicable to any tissue and different types of pathologies. This work provides a proof-of-principle for gaining a comprehensive view from histology by studying it quantitatively in 3D.

Essential plant nutrients are needed at crop-specific concentrations to obtain optimal growth and yield. Foliar tissue analysis is the standard method for assessing nutrient levels in plants. Symptoms of nutrient deficiency or toxicity occur when the foliar tissue values become too low or high. Diagnostic nutrient deficiency criteria for Brassica rapa var. Chinensis (bok choy) is lacking in the current literature. In this study, green (‘Black Summer’) and purple (‘Red Pac’) bok choy plants were grown in silica sand culture, with control plants receiving a complete modified Hoagland’s all-nitrate solution, and nutrient-deficient plants induced by using a complete nutrient formula withholding a single nutrient. Tissue samples were collected at the first sign of visual disorder symptoms and analyzed for dry weight and nutrient concentrations of all plant essential elements. Six weeks into the experiment, the newest matured leaves were sampled for chlorophyll a, b, and total carotenoids concentrations for both cultivars, and total anthocyanin concentration in ‘Red Pac’. Compared to control plants, the dry weight of ‘Black Summer’ green bok choy was significantly lower for nitrogen (N), phosphorus (P), calcium (Ca), or boron (B) deficiency treatments, and nutrient concentrations were lower for all variables except iron (Fe) deficiency. Dry weight was less in ‘Red Pac’ plants grown without N, potassium (K), Ca, B, or molybdenum (Mo), and nutrient concentrations were lower for all except Mo-deficiency compared to controls. Total chlorophyll and total carotenoid concentrations were lower in leaves from N−, Fe-, and manganese- (Mn) deficient plants of both cultivars. Leaf anthocyanin concentration was lower only for K-, Ca-, and B-deficiencies in ‘Red Pac’. Our results indicate that visual symptoms of nutrient deficiency are well correlated with nutrient disorders. In contrast, changes in dry weight, chlorophyll, and anthocyanin did not show consistent changes across nutrient disorders.

Saffron is traditionally cultivated in soil as a semi-perennial crop, although the feasibility of crop production is today constrained in Europe due to both agronomic and socioeconomic factors. Accordingly, interest has been increasing concerning its possible cultivation within protected environments through adoption of soilless cultivation technologies. The aim of the present study was to optimize nutrient solution features in the soilless cultivation of saffron corms. The trial was conducted in a greenhouse at Almeria University. Saffron was grown in 15-L pots filled with perlite. Three fertigation treatments were used, obtained by a linear increase of all nutrients of one standard in order to reach an electrical conductivity (EC) of 2.0 (control, EC2.0), 2.5 (EC2.5) and 3.0 (EC3.0) dS·m−1. Measurements included determinations of shoot length, corm yield, as well as nutrient uptake from the nutrient solution and concentrations within plant tissues. The nutrient solution with the highest EC (EC3.0) allowed obtaining three to five times more corms above 25-mm diameter. The increasing EC had a significant effect on the increase of macronutrient uptake, except for NO3− and NH4+ and resulted in a general increase of nutrient concentrations in tissues, such as corms and roots. Both macronutrient uptake and accumulation in plant tissues were highest under EC3.0. Nutrient uptake was significantly correlated with production of larger corms due to higher horizontal diameter.

In vivo solid-phase microextraction (SPME) has been recently proposed for the extraction, clean-up and preconcentration of analytes of biological and clinical concern. Bioanalysis can be performed by sampling exo- or endogenous compounds directly in living organisms with minimum invasiveness. In this context, innovative and miniaturized devices characterized by both commercial and lab-made coatings for in vivo SPME tissue sampling have been proposed, thus assessing the feasibility of this technique for biomarker discovery, metabolomics studies or for evaluating the environmental conditions to which organisms can be exposed. Finally, the possibility of directly interfacing SPME to mass spectrometers represents a valuable tool for the rapid quali- and quantitative analysis of complex matrices. This review article provides a survey of in vivo SPME applications focusing on the extraction of tissues, cells and simple organisms. This survey will attempt to cover the state-of- the-art from 2014 up to 2019.

In vivo imaging techniques can be integrated with classical histochemistry to create an actual histochemistry of water. In particular, Magnetic Resonance Imaging (MRI), an imaging technique primarily used as diagnostic tool in clinical/preclinical research, has excellent anatomical resolution, unlimited penetration depth and intrinsic soft tissue contrast. Thanks to the technological development, MRI is not only capable to provide morphological information but also and more interestingly functional, biophysical and molecular. In this paper we describe the main features of several advanced imaging techniques, such as MRI microscopy, Magnetic Resonance Spectroscopy, functional MRI, Diffusion Tensor Imaging and MRI with contrast agent as a useful support to classical histochemistry.

A nutrição mineral da videira constitui-se em importante fator para a qualidade dos vinhos. Devido a isso, avaliou-se o efeito de porta-enxertos no teor de nutrientes em diferentes tecidos da videira 'Cabernet Sauvignon' (Vitis vinifera L.) na Serra Gaúcha. o experimento foi conduzido durante o ciclo vegetativo de 2004/2005, com os porta-enxertos Rupestris du lot, 101-14, 3309, 420A, Kober 5BB, 161-49, So4 e Paulsen 1103, enxertados em 1993 com a cv. 'Cabernet Sauvignon'. o delineamento experimental foi em blocos ao acaso, com oito tratamentos e três repetições, sendo quatro plantas/parcela. Coletaram-se folhas - separando-se os pecíolos dos limbos -, cachos - separando-se as bagas das ráquis - e ramos, os quais foram posteriormente secados em estufa e pesados. Analisaram-se os nutrientes n, P, K, Ca e Mg. os resultados mostram que houve efeito significativo do porta-enxerto nos teores de N, P, K, Ca e Mg no limbo, pecíolo, ráquis e baga da videira 'Cabernet Sauvignon' e que este efeito variou em função do nutriente e do tecido considerado. Entretanto, não houve efeito significativo do porta-enxerto no teor desses nutrientes no ramo da videira. Além disso, a ordem de grandeza do teor dos nutrientes variou em função do tecido avaliado. Assim, os teores de n e de Ca foram maiores no limbo; os de P e K, na ráquis; e o de Mg, no pecíolo.Grapevine mineral nutrition is an important factor influencing wine quality. For this, the effect of rootstocks on the nutrient content in different tissues of 'Cabernet Sauvignon' grapevines (Vitis vinifera L.) grown in the Serra Gaúcha region was evaluated. The experiment was carried out during the 2004/2005 vegetative cycle with the rootstocks Rupestris du Lot, 101-14, 3309, 420A, Kober 5BB, 161-49, SO4, and Paulsen 1103. The experimental design was in randomized blocks, with eight treatments, three replicates, four plants/plot. leaves - petioles were separated from the limbs -, clusters - berries were separated from the rachis -, and shoots were collected, dried in an oven, and weighted. N, P, K, Ca, and Mg were analyzed. Results show that there was a significant effect of the rootstock on the contents of N, P, K, Ca, and Mg in limb, petiole, rachis and berry of the 'Cabernet Sauvignon' grapevine. This effect varied as a function of the analyzed nutrient and tissue. But there was not a significant effect on the nutrients present in the shoot. Besides this, nutrient content varied according to the kind of tissue analyzed. In this way, contents of N and Ca were higher in limb; contents of P and K, in rachis; and contents of Mg, in petiole.

The methods of grapevine nutritional diagnosis used in Michigan, California, and South Africa were evaluated with the objective to assess and adjust values of these methods for the Brazilian conditions. The study was carried out in 53 vineyards in the Serra Gaúcha viticultural region, State of Rio Grande do Sul, Brazil, from 1995/96 to 1997/98 (25 of these vineyards were Cabernet Sauvignon and 28 Italie Riesling). All these vineyards were conducted in the pergola system. Data of yield, °Brix, total titratable acidity, density, and pH of the grape must, the concentration of macro- and micro nutrients in the leaves as well as data from soil analyses were assessed. The yield of each vineyard and the concentration of the mineral nutrients in the tissue were correlated. The final adjustments were made by means of the method of the critical threshold using the NCRIz criterion. The method of California was the less precise as it neither establishes defined border values for the sufficiency level nor takes some nutrients into account. The method of Michigan needed few adjustments because of its origin; for this it may be considered viable for the nutritional diagnosis, mainly for American cultivars. Out of the three methods, the South African proved to be the most appropriate for diagnosing the nutritional status of the grapevine cultivated in the Serra Gaúcha, because it did not need adjustments for most nutrients.

Biomass and tissue elemental differences were quantified for lettuce grown in deep-water conventional hydroponic conditions at two pH and alkalinity conditions. Nutrient solutions were created using inorganic salts and either reverse osmosis (RO) water or municipal water with high alkalinity. Three treatments were evaluated: (a) nutrient solution created with reverse osmosis (RO) water and maintained at pH 5.8 (H5); (b) same as H5 but maintained at pH 7.0 (H7); and (c) nutrient solution created using municipal water and maintained at pH 7.0, referred to as HA7. Averaged across three trials, the HA7 and H7 treatments produced 26% less shoot fresh weight (FW) than the H5 treatment with an 18% reduction in dry weight (DW). The H5 treatment had the least biomass in root FW and DW. In tissue elemental analyses, both the pH 7.0 treatments showed lower concentrations than H5 in Cu, N, Mo, and Sr, and increased concentrations in Ba, Mg, Na, and Zn. There were no differences in Al, C, Ca, Fe, K, Mn, Ni, P, S, and Si concentrations among treatments (p = 0.05). The results from this experiment can be used to isolate the effects of pH and alkalinity in aquaponic conditions where pH and alkalinity will mimic HA7 conditions.

Spinach (Spinacia oleracea cv. Carmel) was grown in a conventional glass greenhouse under three different nutrient solution treatments. Lighting and temperature conditions were identical. Six growing systems were used to provide a duplicate trough system for each of these three treatments. Six trials were harvested from each system over a two month time period. Two treatments received hydroponic nutrient inputs, with one treatment at pH 7.0 (referred to as H7) and the other at pH 5.8 (H5), and the third treatment was aquaponic (A7), receiving all of its nutrients from a single fish tank with koi (Cyprinus carpio) except for chelated iron. System pH was regulated by adding K2CO3 to aquaponic systems and KOH to hydroponic systems. Comparisons made between treatments were total yield, leaf surface area, tissue elemental content, and dry weight to fresh weight ratio. Dry weight biomass yield values were not different in pairwise comparisons between treatments (A7 vs. H5: p = 0.59 fresh weight, p = 0.42 dry weight). Similarly, surface area results were not different between treatments. The important comparison was that A7 achieved the same growth as H5, the conventional pH with a complete inorganic nutrient solution, despite unbalanced and less than “ideal” nutrient concentrations in the A7 condition.

Effect of growing media on macro- and microelements, as well as on chlorophyll concentration in Phalaenopsis orchids were investigated. In the years 2006–2008, an experiment on Phalaenopsis orchids cultivation was carried out. The plants were planted into New Zealand sphagnum moss, mixture of expanded clay pellets and New Zealand sphagnum moss (v:v = 1:1), or into expanded clay pellets only. Orchids were grown in translucent plastic pots. Total macro and micro elements concentration in leaves and roots were determined. Furthermore concentrations of chlorophyll a and chlorophyll b in leaves and roots were measured. Growing media exert a significant influence on the nutritional status of Phalaenopsis orchids. The highest concentration of magnesium, iron and copper was found in plants grown in expanded clay, less in orchids grown in the mixed medium and the least in plants grown in sphagnum moss. A higher concentration of nitrogen, phosphorus, potassium, calcium and manganese was found in leaves. Inversely, in roots, there was a higher concentration of magnesium, iron and zinc. Growing media did not affect chlorophyll concentration in leaves and roots of Phalaenopsis.

Abstract The present study aimed to compare the use of leaf and flower analysis and to verify the most appropriate organ for the diagnosis of the nutritional status of guava trees. The evaluation was carried out using the CND (Compositional Nutrient Diagnosis) methodology, using yield and nutritional contents of the leaf and flower of ‘Paluma’ guava trees in 48 plots of an irrigated orchard. The contents of flower and leaf nutrients were submitted to variance analysis, by the Tukey test at 5%, and the nutrient sufficiency ranges in organs were calculated, as well as the correlations between multi-nutrient variables and yield. Significant differences between contents of nutrients in flower and leaf, as well as greater variability in the nutritional contents in leaf, were observed. The sufficiency ranges of flower proved to be shorter compared to leaf. The number of correlations between nutrients and yield was higher in the flower. The greater sensitivity of the leaf analysis in expressing interactions between nutrients and yield indicates that this organ can also be used for the nutritional diagnosis of guava trees.
