Passaged cell lines represent currently an integral component in various studies of malignant neoplasms. These cell lines are utilized for drug screening both in monolayer cultures or as part of three-dimensional (3D) tumor models. They can also be used to model the tumor microenvironment in vitro and in vivo through xenotransplantation into immunocompromised animals. However, immortalized cell lines have some limitations of their own. The homogeneity of cell line populations and the extensive passaging in monolayer systems make these models distant from the original disease. Recently, there has been a growing interest among scientists in the use of primary cell lines, as these are passaged directly from human tumor tissues. In this case, cells retain the morphological and functional characteristics of the tissue from which they were derived, an advantage often not observed in passaged cultures. This review highlights the advantages and limitations of passaged and primary cell cultures, their similarities and differences, as well as existing test systems that are based on primary and passaged cell cultures for drug screening purposes.

Rosa centifolia L. and Rosa gallica L. (Rosaceae) are grown as raw materials for valuable essential oils and hydrosols. There are scarce data about the biological activities and the genoprotective potential of the hydrosols of these roses. The aim of the study was to provide information on their cytotoxic/genotoxic activity and anti-cytotoxic/anti-genotoxic capacity against mutagenic N-methyl-N′-nitro-N-nitrosoguanidine (MNNG). The evaluation was performed using classical tests for chromosomal aberrations and micronuclei in the higher plant Hordeum vulgare and human lymphocyte test systems. The experimental schemes included combined hydrosol and mutagen treatment. Both hydrosols (6, 14, 20%) had no cytotoxic effect on barley and showed low genotoxicity in both test systems as the injuries were enhanced to a lesser extent compared to the controls. Lymphocytes were more susceptible than H. vulgare. Under the conditions of combined treatment, it was found that the two hydrosols possessed good anti-cytotoxic and anti-genotoxic potential against MNNG. Both rose products exerted genoprotective potential to a similar extent, decreasing the frequencies of aberrations in chromosomes and micronuclei to a significant degree in both types of cells when non-toxic concentrations of hydrosols were applied before MNNG. This was performed both with and without any inter-treatment time. The observed cytoprotective/genoprotective potential suggests that these hydrosols are promising for further application in phytotherapy and medicine.

By applying the “safety atmosphere” measurement theory to Chinese management companies engaged in road transportation, a corporate and individual safety risk assessment system was established that is consistent with the management and cultural climate in China, thereby reducing the driving safety risk of truck drivers. The system realizes the safety risk assessment of enterprises, fleets and individuals in the form of test scales by constructing a structural model of the enterprise safety atmosphere, including the management, communication, and supervision of enterprises, fleets, and individuals. The safety climate was modeled using a two-level framework, at the organizational level and fleet level, and three dimensions of test items for each level were obtained by exploratory factor analysis. The three dimensions of safety management, safety supervision, and safety priority at the organizational level, and the three dimensions of positive communication, safety awareness, and self-discipline at the fleet level, respectively, passed a valid factorial test (p < 0.01). Finally, the validity of the system evaluation results was verified by relying on the actual in-vehicle monitoring data and accident records of the corporate transportation fleet. The results show that the total test scores at the organizational level and the fleet level are significantly correlated with their driving risk behaviors, and both are linearly and negatively correlated with the number of accidents per thousand kilometers. This indicates a high degree of consistency between the system’s test results and actual risky accidents.

The relevance of the study of coronavirus infection in animals is conditioned upon the lack of sufficient information about the mechanisms of development of this disease, imperfect methods of diagnosis and treatment, and, most importantly, almost 100% of their mortality. The purpose of this study was to identify the age, breed, seasonal, and sexual characteristics of cats' predisposition to coronavirus infection and the development of infectious peritonitis. The paper presents the results of epizoothogolic features of the spread of coronavirus infection among animals of this species and feline infectious peritonitis during 2020-2022 based on the veterinary clinic “White Wolf” (city of Kyiv). During this period, 483 samples were examined from cats with symptoms of coronavirus infection. From these samples, using immunochromatographic analysis, a virus of the Coronaviridae family was detected in 399 animals, and infectious peritonitis was established in 63 animals. This paper highlights the results of a study of the age, breed, and sexual predisposition of cats to coronavirus infection and the development of infectious peritonitis. It was found that cats of any age are susceptible to coronavirus infection, while infectious peritonitis develops in animals aged from 2 months to 3 years. Coronavirus infection was most frequently found in mixed breed cats and British shorthair cats, infectious peritonitis – in Burmese and Bengal breeds. The study also summarised data on the seasonal manifestation of coronavirus infection and infectious peritonitis of cats. Based on the results of epizootological analysis, a nosological profile of infectious diseases was formed in animals of this species that had similar clinical symptoms of coronavirus infection and infectious peritonitis. Eight infections are presented, of which the most frequently recorded diseases were caused by viruses of the Herpesviridae, Caliciviridae, and Parvoviridae families. The results of this study provide new information about the epizootological features of the manifestation and development of coronavirus infection in cats, which allows not only improving the available diagnostic methods, but also developing new ones

Introduction. Here we discuss the problem of timely diagnostics and prevention of Rh-immunization of pregnant women as well as fetal hemolytic disease, which remains currently relevant, despite the existence of proven diagnostic, therapeutic and preventive methods.Aim: to assess the medico-economic efficiency of non-invasive prenatal diagnostics of using fetal Rh factor (rhesus D antigen, RhD) in maternal blood – a fetal RhD-genotyping.Materials and Methods. A retrospective observational study was conducted to analyze determining fetal Rh-factor in the blood samples from 4109 Rh-negative pregnant women observed in the 41 medical facilities of the Ulyanovsk region in the years 2018–2020. The fetal RhD level was determined by polymerase chain reaction at gestational age of ≥ 10 weeks. To assess testrelated medical effectiveness, there were examined sensitivity, specificity, predictive value of positive and negative data as well as diagnostic accuracy. The data collected during the study were compared with those obtained after delivery. To assess the economic efficiency, the difference between the cost of immunization and the cost of determining the fetal Rh factor level was determined.Results. A positive and negative fetal Rh-factor was detected in 67.26 % (n = 2793) and 32.74 % (n = 1316) cases, respectively. Diagnostic accuracy of the test system "Test-RhD" was 99.40 %, sensitivity – 99.84 %, specificity – 97.51 %, the prognostic value of a positive result was 99.43 %, the predictive value of a negative result – 99.28 % with low rate of false positive and false negative data. It was shown that our study allows to avoid unnecessary immunization costs for all Rh-negative pregnant women.Conclusion. Analysis of the diagnostic characteristics and cost-effectiveness of the RhD test evidences about high medical significance of the method described and allows to recommend its wider application.

Nowadays, specialized literature uses different test systems to verify their proposed models and methodologies regarding reconfiguration and operation of distribution networks. However, none of these research works include various test systems with enough information in order to endorse studies that approach distribution system reconfiguration and operation issues. This paper contains several test systems including the load data, network configuration, line characteristics, maximum current of branches, and nominal powers and voltages. The main objective is to provide all the details and information required to evaluate models and methods developed for reconfiguration and operation of radial distribution systems. Verification of all presented data and information has been performed by solving the network reconfiguration problem using established techniques. Moreover, in order to check the radiality of test systems and correctness of their given data, a data visualization technique is employed to online depict network topologies based on the electrical distance between buses.

The purpose of the research is to determine the main clinical signs of dirofilariosis in dogs, taking into account the peculiarities of animal exploitation and to analyze the efficacy and convenience of using some diagnostic techniques. Materials and methods. The main clinical signs of dirofilariosis in dogs on the farms of Armenia have been studied. The presence of microfilariae was determined in native smears and using a modified Knott method. We also used the Asan Easy Test Heartworm and SNAP 4Dx Plus Test one-step cassette rapid tests for visual detection of Dirofilaria spp. antigen in blood serum. Results and discussion. 8.5% of dogs in the farms of the Republic of Armenia are infected with Dirofilaria sp. The most infected were dogs aged 5–8 years (75%). The main symptomatic signs of dirofilariosis in dogs were established in the specific conditions of the studied region and the specifics of the specific exploitation of animals. It has been established that the immunochromatographic test systems Asan Easy Test Heartworm and SNAP 4Dx Plus Test for visual detection of canine Dirofilaria spp. antigen in blood serum are the most effective for the diagnosis of dirofilariosis.

Aim: the study was aimed to develop a reagent kit for the real-time RT-PCR diagnostics of virus causing COVID-19.Materials and Methods. Three target sites were chosen in the genome SARS-CoV-2. The testing included 220 samples, 48 artificially created positive samples (made from patients’ biomaterial) and 172 clinical samples (scrapes from nasal and pharyngeal cavities, bronchoalveolar lavage, expectoration, endotracheal/nasopharyngeal aspirate, feces, post-mortem material), obtained from two medical centers. Preliminary, the obtained biomaterial was analyzed with a reagent kit of comparison. The evaluation was performed with a confidential interval CI 95%. The calculation of CI for the sensitivity and specificity was made based on the distribution of χ2.Results. The authors developed a technology of novel coronavirus infection (COVID-19) real-time RT-PCR diagnostics for the application in practical healthcare and proposed the variants of testing at all the stages (preanalytical, analytical, and post-analytical, including automated results processing). The proposed reagent kit meets the requirements of the World Health Organization and the Ministry of Healthcare of the Russian Federation. The study results demonstrated high sensitivity and specificity. The sensitivity was 100% (95% CI) 95.6–100%; the specificity was 100% (95% CI) 96.7–100%.Conclusion. The proposed reagent kit was registered in the RF as a medical product; the registration certificate No. RZN 2020/9948 dated 01.04.2020. The application of the reagent kit in network laboratories will provide patients with access to testing for the virus causing COVID-19 and contribute to quick differential diagnostics, improvement of pandemic control, and accurate statistics on the spread of the virus.

Electric microgrid is counted with the emerging technologies recently identified as being required for revolutionizing Africa within the space of one decade. The place of innovative research and development on various components of microgrid systems is also found pivotal and indispensable to the envisioned massive deployment of the technology across the sub-Saharan region of the continent. It has, however, become a global practice to employ specially developed experimental microgrid test systems as dedicated research tools for establishing design criteria and evaluating operational performances of microgrids in efficient way. This study therefore presents a survey on the availability and deployment of microgrid test systems across the nations of the sub-Saharan Africa. Available literatures published on the concept, though very scanty and not readily accessible, are thoroughly reviewed and reported in this survey. It is found that while there are few instances of works done on development and application of on-the-site demonstration microgrid as experimental facilities, laboratory-based microgrid emulators as test systems does not have much popularity despite being comparatively cost effective in design and development. This discovery could influence researchers, utility providers, national governments, regional organizations, research funders, as well as other stakeholders across the region into embracing the deployment of microgrid test systems in order to enhance seamlessness in the adaptation of the technology to the African context for its efficient application across the African sub-Sahara.

The present article summarizes the materials from literary sources dedicated to the issues of laboratory diagnosis of anthrax. It shows the priority of the development of new methods for the elaboration of highly sensitive test systems and preparations designed for the detection and identification of anthrax bacteria. It outlines the role of express-diagnostics of infectious diseases, which is very important when investigating bioterrorism cases, outbreaks and sporadic cases in humans and animals. Providing with the analysis of different methods for detection of anthrax, the article at the same time focuses on modern molecular diagnostic technologies. It is shown that the basic parameters of diagnostic test systems are sensitivity, specificity and reproducibility of the results. The article presents data on the development of culture media for isolation and identification of anthrax, as well as the possibility of using anthrax bacteriophage for phage-based indication and identification of bacteria.

A major feature specific to commissioning of NPPs in Russia and of Russian-designed NPPs in foreign countries is a great deal of measurement involved at different pre-commissioning stages, which differs from foreign experience where emphases is placed on computational justification and the scope of in-situ commissioning testing is smaller. The use of various measuring systems in pre-commissioning operations requires the involvement of a large number of personnel from different organizations, infrequently with no data acquisition coordination among them. This leads to both measuring channels and the information gathered being multiply duplicated (assembly, installation, adjustment), which is one of the sources for its misrepresentation, and to a greatly increased cost of operations. At the same time, most modern power units are equipped with test systems (TS) [1–4] consisting of heterogeneous measuring channels. The current TS commissioning practice required systems to be adjusted and started up only after the 100% unit power level was reached which did not make it possible to use test systems at pre-commissioning and power ascension stages. This study is an attempt to create an integrated information system composed of heterogeneous local systems to enable the largest possible number of standard channels to be used simultaneously with temporarily installed channels for pre-commissioning measurements to provide sound and high-quality information on the state of the unit.

This paper presents our empirical observations related to the evolution of a large automated test system. The system observed is used in the industry as a test tool for complex telecommunication systems, itself consisting of more than one million lines of source code. This study evaluates how different changes during the development have changed the number of observed Code Smells in the test system. We have monitored the development of the test scripts and measured the code quality characteristics over a five years period.

The development of solution algorithms for power system problems is based on hypothetical test systems and test cases. These systems are very scarce, and the degree of variability is relatively low. The constant development of the economic analysis in electrical power systems denotes the need to obtain standardized systems and cases. In this study, the creation of standardized test cases based on a hybrid model using Lévy alpha stable distributions and generalized additive models is proposed. The objective of the work is to present a methodological proposal for the creation of test environments for optimization models based on general information about the operation of particular power systems. The simulation of random values based on Lévy alpha stable distributions lets capturing the series impulsivity and demand peaks, and the use of generalized additive models permits capturing non-linearity in the behavior of the demand for electrical energy. The hybrid model will tolerate simulating as many instances as necessary, with a coherent behavior attached to the reality of the operation of the analyzed electrical systems.

This article provides a framework for the design and development of intelligent information system of knowledge evaluation using an open form tasks, the “open” (without variants of answers) test tasks are considered as efective. Student’s answer entered in a free form gives an opportunity to present student’s views on the subject more accurately. And we evaluate his knowledge, and not based on a guessed answer to the question, which proves the efectiveness of the proposed method. This article presents an approach for text analysis in natural language and organization of query in the knowledge based on the ontology.

Detergent surfactantscan be found in wastewater in relevant concentrations. Most of them are known as ready degradable under aerobic conditions, as required by European legislation. Far fewer surfactants have been tested so far for biodegradability under anaerobic conditions. The natural environment is predominantly aerobic, but there are some environmental compartments such as river sediments, sub-surface soil layer and anaerobic sludge digesters of wastewater treatment plants which have strictly anaerobic conditions. This review gives an overview on anaerobic biodegradation processes, the methods for testing anaerobic biodegradability, and the anaerobic biodegradability of different detergent surfactant types (anionic, nonionic, cationic, amphoteric surfactants).
