The effect of final rolling temperature ranging from 962 to 696 °C on as-rolled high-Mn austenitic low-temperature steel was investigated using scanning electron microscopy, electron backscatter diffraction, transmission electron microscopy, and X-ray diffraction. The microstructure of the experimental steel remained austenite without suffering ferrite phase transformation, even when produced with a final rolling temperature as low as 696 °C. A decrease in the final rolling temperature of the steel was accompanied by a decrease in grain size from 20.2 to 6.0 μm, accompanied by an increase in dislocation density from 3.01 × 1014 to 10.9 × 1014 m−2. In addition, M23C6 carbides precipitated at the grain boundaries when the final rolling temperature was below 782 °C. Grain refinement and increased dislocation density significantly enhanced the strength of the material. However, an increase in the intragranular dislocation density compromised grain deformation ability. Moreover, the precipitation of M23C6 reduced the plasticity and toughness of the material. Considering the balance between yield strength and low-temperature toughness, the optimal final rolling temperature was 898 °C; this produced a material with a yield strength of approximately 507 MPa and a low-temperature impact energy absorption of approximately 143 J.

Fluconazole is a first-line medication for the treatment of pulmonary cryptococcosis, but currently there is a lack of clinical medication experience in obese patients with multiple comorbidities, especially in dose adjustment after treatment failure.Case Introduction: This case report describes the experience of fluconazole in the treatment of pulmonary cryptococcal infection in a 45-year-old Chinese male with obesity, type 2 diabetes, and cirrhosis. The patient had a history of antifungal therapy for two weeks before admission, but the cough and hemoptysis were not improved. The treatment failed. After admission, it was recommended to use a conventional dose of fluconazole as an antifungal regimen according to the guidelines. However, the treatment effect was still unsatisfactory, due to the patients’ cough, hemoptysis, and fever symptoms were not relieved. During this period, it was newly found that the patient had cirrhosis and type 2 diabetes and had not previously controlled blood glucose. Considering the above situation, combined with the pharmacokinetic characteristics of fluconazole and the patient’s weight reaching 113 kg, the team readjusted the fluconazole medication regimen, and ultimately, the pulmonary infection improved without significant adverse reactions.Results: We found that it was more suitable for patients with obesity to calculate the dose of fluconazole by the lean weight. By estimation, the patient was finally given a loading dose of 800 mg fluconazole, and his condition improved significantly. After two weeks of medication, it was adjusted to a maintenance dose of 600 mg until the pulmonary infection in the patient disappeared.Conclusion: This case suggests that fluconazole antifungal therapy for pulmonary cryptococcal infection should fully consider the risk of comorbidities in patients. If necessary, medication dosage can be adjusted according to weight, and it is recommended to use lean bodyweight for evaluation and optimization. Abstract Using e + e − collision data collected by the BESIII detector at BEPCII corresponding to an integrated luminosity of 30 fb −1, we measure Born cross sections and effective form factors for the process e + e − → Ξ 0 Ξ ¯ 0 $$ {e}^{+}{e}^{-}\to {\Xi}^0{\overline{\Xi}}^0 $$ at forty-five center-of-mass energies between 3.51 and 4.95 GeV. The dressed cross section is fitted, assuming a power-law function plus a charmonium(-like) state, i.e., ψ(3770), ψ(4040), ψ(4160), ψ(4230), ψ(4360), ψ(4415) or ψ(4660). No significant charmonium(-like) state decaying into Ξ 0 Ξ ¯ 0 $$ {\Xi}^0{\overline{\Xi}}^0 $$ is observed. Upper limits at the 90% confidence level on the product of the branching fraction and the electronic partial width are provided for each decay. In addition, ratios of the Born cross sections and the effective form factors for e + e − → Ξ 0 Ξ ¯ 0 $$ {e}^{+}{e}^{-}\to {\Xi}^0{\overline{\Xi}}^0 $$ and e + e − → Ξ − Ξ ¯ + $$ {e}^{+}{e}^{-}\to {\Xi}^{-}{\overline{\Xi}}^{+} $$ are also presented to test isospin symmetry and the vector meson dominance model.

Abstract Background Sweet sorghum (Sorghum bicolor) displays an excellent potential to serve as a non-food bioenergy feedstock for bioethanol production in China due to its high potential yield on marginal lands. However, few studies have been conducted on the potential of sweet sorghum yield and appropriate industrial models in different marginal regions in China. This study explored the spatial distribution of potential sweet sorghum production using the Decision Support System for Agrotechnology Transfer (DSSAT) model and proposed three typical industrial models of sweet sorghum industry to calculate their economic and ecological benefits. Results The results indicate that considering the factors of land use, annual precipitation, soil salinity, soil pH, and accumulated temperature, approximately 32.23 million ha of marginal land are suitable for sweet sorghum cultivation in China, and 130 million tonnes (t) of ethanol can be produced. Further, the development of the sweet sorghum industry under the three models can generate 1425.49 billion CNY potential, approximately accounting for 3.57% of industrial added value in China if measured against 2023 levels, and reduce CO2 emissions by 4.68 million t. Conclusions This study provides an innovative perspective for the multi-industry large-scale promotion of sweet sorghum in different marginal lands based on the high spatial resolution Geographic Information System (GIS) data by the DSSAT model with a Life Cycle Assessment (LCA) method, and this applies not only to China but also to the worldwide and other types of energy plants.

Abstract Background Radiation-induced intestinal injury (RIII) interrupts the scheduled processes of abdominal and pelvic radiotherapy (RT) and compromises the quality of life of cancer survivors. However, the specific regulators and mechanisms underlying the effects of RIII remain unknown. The biological effects of RT are caused primarily by DNA damage, and ataxia telangiectasia mutated (ATM) is a core protein of the DNA damage response (DDR). However, whether ATM is regulated by deubiquitination signaling remains unclear. Methods We established animal and cellular models of RIII. The effects of ubiquitin-specific protease 15 (USP15) on DNA damage and radion-induced intestinal injury were evaluated. Mass spectrometry analysis, truncation tests, and immunoprecipitation were used to identify USP15 as a binding partner of ATM and to investigate the ubiquitination of ATM. Finally, the relationship between the USP15/ATM axes was further determined via subsequent experiments. Results In this study, we identified the deubiquitylating enzyme USP15 as a regulator of DNA damage and the pathological progression of RIII. Irradiation upregulates the expression of USP15, whereas pharmacological inhibition of USP15 exacerbates radiation-induced DNA damage and RIII both in vivo and in vitro. Mechanistically, USP15 interacts with, deubiquitinates, and stabilises ATM via K48-linked deubiquitination. Notably, ATM overexpression blocks the effect of USP15 genetic inhibition on DNA damage and RIII progression. Conclusions These findings describe ATM as a novel deubiquitination target of USP15 upon radiation-induced DNA damage and intestinal injury, and provides experimental support for USP15/ATM axis as a potential target for developing strategies that mitigate RIII.

Abstract The incidence of patients diagnosed with either breast cancer (BC) or inflammatory bowel disease (IBD) is increasing each year. IBD has been shown to be strongly associated with the development of a variety of solid tumors, but the relationship with breast cancer is not yet definitive. We explored the causative relationship between IBD and BC using a Mendelian randomization (MR) strategy. MR-Egger regression, weighted median (WM), simple median (SM), maximum likelihood (ML), and inverse variance weighting (IVW) methods were among the analytical techniques used in this work. The examination of heterogeneity was conducted by the use of Cochran's Q test and Rucker's Q test. The sensitivity analysis in this study used the IVW and MR-Egger methodologies. The results of our investigation suggested that IBD had a beneficial impact on estrogen receptor-negative (ER-) breast cancer (odds ratio (OR) = 0.92, P = 0.02). The study did not find a significant association between IBD and the risk of developing overall breast cancer (OR = 0.99, P = 0.60), as well as estrogen receptor-positive (ER+) breast cancer (OR = 1.02, P = 0.60) specifically. In addition, our study findings indicated that there was a detrimental association between ER+ breast cancer and IBD as determined using reverse MR analysis (OR = 1.07, P = 0.04). Furthermore, this analysis failed to observe any significant association between overall breast cancer (OR = 1.07, P = 0.07) or ER- breast cancer (OR = 0.99, P = 0.89) and IBD. Our bidirectional MR study yielded a correlation between IBD and some specific hormone receptor status of BC.

Abstract While the tropical Pacific teleconnection to North America has been studied extensively, the impact of the Indian Ocean on North American climate has received less attention. Here, through observational analysis and hierarchy atmospheric model simulations with different complexity, we find that the Indian Ocean plays a crucial role in North American winter climate through a teleconnection termed the Indian Ocean - North America pattern. We show that in the warm Indian Ocean phase, this teleconnection contributes to anomalously cold winters along the west coast of the United States through advection with increased mountain snowfall, while simultaneously leading to warmer conditions over the Great Lakes region. Snow-albedo feedback amplifies these Rossby wave-induced surface anomalies. Remarkably, this teleconnection pattern is at work on both interannual and multi-decadal time scales, with its climatic impact being slightly less pronounced than that induced by tropical Pacific sea surface temperature anomalies. Our findings underscore the significance of the Indian Ocean in both the prediction and future projection of North American climate.

Abstract The procedure of anterior cruciate ligament (ACL) allograft preparation can be divided into fresh-frozen method (FF-allograft) or freeze-dried method (FD-allograft). This study aims to biomechanically and histologically compare the graft to bone tunnel integration between the two allografts. In-vitro results indicated that FF-allograft and FD-allograft showed excellent biocompatibility and biomechanics, while FD-allograft showed a denser collagen fiber arrangement than FF-allograft and autograft. Then, in-vivo preformation of the FF-allograft, FD-allograft, and autograft on bone tunnel integration was evaluated via a canine ACL reconstruction model. In-vivo results indicated that no signs of infection or osteoarthritis were shown in the femur-graft-tibia complexes, but more vascularity and synovitis formed around the implanted FF-allograft. Micro-computed tomography showed that peri-graft bone in the FF-allograft group was significantly increased and remodeled compared with the FD-allograft group; Histologically, the FF-allograft group exhibited similar graft-bone tunnel healing to the FD-allograft group. Tartrate-resistant acid phosphatase (TRAP) staining showed significantly more osteoclasts presented in the FD-allograft group compared to the FF-allograft group. Abstract Autophagy is a highly conserved degradative process that has been linked to various functions, including defending host cells against pathogens. Although the involvement of autophagy in porcine circovirus 2 (PCV2) infection has become apparent, it remains unclear whether selective autophagy plays a critical role in PCV2 restriction. Here we show that retinol-binding protein 4 (RBP4), an adipokine for retinol carrier, initiates the autophagic degradation of PCV2 ORF1 protein. PCV2 infection increases RBP4 protein levels through MAPK-eIF4E axis in living cells. Ectopic expression of RBP4 or recombinant RBP4 treatment promotes the degradation of ORF1 protein. Mechanistically, RBP4 activates TRAF6 to induce K63-linked ubiquitination of ORF1, leading to SQSTM1/p62-mediated selective autophagy for degradation. Consequently, RBP4 deficiency increases viral loads and exacerbates the pathogenicity of PCV2 in vivo. Collectively, these results identify RBP4 as a key host restriction factor of PCV2 and reveal a previously undescribed antiviral mechanism against PCV2 in infected cells.

Abstract Tropical convection plays a critical role in modulating the global climate by influencing climate variability. However, its future projection under climate mitigation scenarios remains uncertain. Here, we found that while the relationship between precipitation intensity and upward motion remains constant regardless of changing CO2 concentrations, the sea surface temperature threshold for tropical convection and the convective zone exhibit hysteretic and irreversible behavior. As the CO2 concentration decreases from its peak (ramp-down), higher tropical ocean temperature leads to higher sea surface temperature thresholds for convection than during the period of increasing CO2 concentration (ramp-up), while convective instability remains the same during both ramp-up and ramp-down. El Niño-like warming during the ramp-down leads to a weakening of the Walker circulation and an expansion of the convective zone in the central to eastern tropical Pacific by a warmer-get-wetter mechanism. Our results suggest that CO2 removal does not guarantee the recovery of tropical convection.

Abstract Background Mitochondrial calcium uniporter (MCU) plays pleiotropic roles in cellular physiology and pathology that contributes to a variety of diseases, but the role and potential mechanism of MCU in the pathogenesis of metabolic dysfunction-associated steatohepatitis (MASH) remain poorly understood. Methods and results Here, hepatic knockdown of MCU in C57BL/6J mice was achieved by tail vein injection of AAV8-mediated the CRISPR/Cas9. Mice were fed a Choline-deficient, L-amino acid-defined high-fat diet (CDAHFD) for 8 weeks to induce MASH and fibrosis. We find that expression of MCU enhanced in MASH livers of humans and mice. MCU knockdown robustly limits lipid droplet accumulation, steatosis, inflammation, and hepatocyte apoptotic death during MASH development both in vivo in mice and in vitro in cellular models. MCU-deficient mice strikingly mitigate MASH-related fibrosis. Moreover, the protective effects of MCU knockdown against MASH progression are accompanied by a reduced level of mitochondrial calcium, limiting hepatic oxidative stress, and attenuating mitochondrial dysfunction. Mechanically, RNA sequencing analysis and protein immunoblotting indicate that knockdown MCU inhibited the Hippo/YAP pathway activation and restored the AMP-activated protein kinase (AMPK) activity during MASH development both in vitro and in vivo. Conclusions MCU is up-regulated in MASH livers in humans and mice; and hepatic MCU knockdown protects against diet-induced MASH and fibrosis in mice. Thus, targeting MCU may represent a novel therapeutic strategy for MASH and fibrosis. Graphical Abstract

Abstract Mining safety heavily depends on ventilation, which constitutes a significant portion of the energy costs in operations. Optimizing mine ventilation systems (MVSO) is crucial for minimizing this energy expenditure. However, current algorithms encounter challenges when applied to large-scale mines, primarily due to the complexity of variables and limited attention to optimizing main fans. This study introduces a theoretical knowledge enhanced genetic algorithm for MVSO, incorporating main fan adjustments. The algorithm models changes in the main fan’s operational status and integrates ventilation network equivalent simplification (VNES) and the minimum spanning tree (MST) to reduce the number of variables in the mine ventilation network. Additionally, leveraging mine ventilation sensitivity theory (MVST) enhances the quality of the initial algorithmic population. A simple case and two engineering cases collectively validated that the algorithm consistently provides effective and reliable optimization solutions for mine ventilation systems across varying scales. Specifically, the algorithm reduced energy consumption from 326.94 to 186.99 kW, 433.14 to 239.48 kW, and 520.53 to 324.90 kW across three different scales of mine ventilation systems. Comparative analysis with four other algorithms shows that, although this algorithm has a longer runtime due to the need to identify the minimum spanning tree during iterations, its ability to reduce problem dimensionality and improve population quality results in more stable and superior convergence performance, especially for large-scale mine ventilation systems. Objective Clarifying paraspinal muscle (PM) change in degenerative lumbar scoliosis (DLS) is positive to evaluate the progression of scoliosis. This research compares the characteristic of PM change among different coronal sub‐types of DLS and explores its potential clinical significance. Methods A total of 84 DLS patients between June 2019 to December 2021 were retrospectively analyzed. Patients were classified into three types based on the coronal balance distance (CBD): Type A, CBD 3 cm; Type C: C7PL shifted to the convex side of the curve, and CBD >3 cm. Fat infiltration rates in the multifidus (MS) and erector spinae (ES) at the apex of the main and fractional curves, and spinopelvic parameters were analyzed statistically. Pearson's or Spearman's correlation was applied to analyze the correlation between asymmetric degree of PM change and these parameters in three types. Results There were 62 cases with coronal sub‐Type A, 6 cases with Type B, and 16 cases with Type C. Patients in Type B and C demonstrated higher fat infiltration in MS on the concave side of both the main and fractional curves when compared to those in Type A. The asymmetric degree of ES change was positively correlated with CBD at the apex of the main curve in Type B and at the apex of the fractional curve in Type C respectively, and that of MS was positively correlated with apical vertebral rotation, while negatively strong‐correlated with pelvic incidence and sacral slope in Type C. Conclusion PM fatty infiltration presented difference among varied coronal sub‐types of DLS patients. The CBD in Type B and C patients was correlated with the asymmetric degree of ES change.

Abstract Background The solid pattern is a highly malignant subtype of lung adenocarcinoma. In the current era of transitioning from lobectomy to sublobar resection for the surgical treatment of small lung cancers, preoperative identification of this subtype is highly important for patient surgical approach selection and long‐term prognosis. Methods A total of 1489 patients with clinical stage IA1‐2 primary lung adenocarcinoma were enrolled. Based on patient clinical characteristics and lung imaging features obtained via deep learning, highly correlated diagnostic factors were identified through LASSO regression and decision tree analysis. Subsequently, a logistic model and nomogram were constructed. A restricted cubic spline (RCS) was used to calculate the optimal inflection point of quantitative data and the differences between the groups. Results The three‐dimensional proportion of solid component (PSC), sex, and smoking status was identified as being highly correlated diagnostic factors for solid predominant adenocarcinoma. The logistic model had good prediction efficiency, and the area under the ROC curve was 0.85. Decision curve analysis demonstrated that the application of diagnostic factors can improve patient outcomes. RCS analysis indicated that the proportion of solid adenocarcinomas increased by 4.6 times when the PSC was ≥72%. A PSC of 72% is a good cutoff point. Conclusion The preoperative diagnosis of solid‐pattern adenocarcinoma can be confirmed by typical imaging features and clinical characteristics, assisting the thoracic surgeon in developing a more precise surgical plan.

Purpose: The aim of this study was to evaluate the diagnostic performance of virtual non-calcium imaging using dual-energy computed tomography (CT) for acute sacrococcygeal injuries diagnosed by radiologists with varying levels of experience. Methods: A prospective study was employed and 29 patients presenting with acute coccygeal pain due to trauma between November 2021 and January 2024, involving a total of 174 vertebrae (145 sacral, 29 coccygeal), were enrolled. Both magnetic resonance imaging (MRI) and DECT data were acquired. Three radiologists with 1, 4, and 10 years of experience analyzed conventional CT and VNCa color-coded images, using MRI as the reference standard. Results: Following the application of VNCa imaging, the inter-rater agreement among the three radiologists significantly improved, with Kappa values increasing from 0.452, 0.615, and 0.735 to 0.775, 0.825, and 0.897, respectively. Diagnostic accuracy also notably increased to 92.5%, 94.3%, and 96.6% for the three radiologists, with no statistically significant differences observed among them. Quantitative analysis revealed an average VNCa CT value of (−84.2±20.3) HU for normal bone marrow and (−37.4±18.8) HU for bone marrow edema, showing a significant difference between the two. Using MRI as the reference standard, the area under the curve for distinguishing normal bone marrow from BME based on VNCa CT values was 0.962, with an optimal cutoff of −57.1 HU, yielding a sensitivity and specificity of 91.9％ and 90.2％, respectively. Conclusion: DECT VNCa imaging significantly enhances the diagnostic efficacy of radiologists with varying levels of experience in acute coccygeal injury assessment, particularly benefiting less-experienced physicians. VNCa CT values contribute substantially to diagnostic accuracy in this context.

In the use of Sn58Bi solder alloy, the abnormal enrichment and coarsening of Bi phase in solder joints and the excessive growth of IMC seriously affect its service reliability, among which, the IMC's reaction mechanism and evolution, the influence on the coarsening of the Bi phase within the solder and the coarsening process are not yet clear. The study found that the initial type and morphology evolution of IMC of Sn58Bi/Cu solder joint are influenced by soldering temperature and cooling rate, attributed to the differences in dissolution and diffusion of Cu in Sn58Bi, resulting in different initial appearances of Cu6Sn5 and Cu3Sn; The temperature and cooling rate affect whether Cu6Sn5 and Cu3Sn are directly generated by the reaction of Cu atoms in the substrate or appear through the consumption of Cu3Sn or Cu6Sn5; The cooling rate affects the IMC morphology evolution of top-view; Based on this, it was cleared that Sn58Bi/Cu solder joints follow four interfacial reaction mechanisms. Meanwhile, the evolution of Bi phase in the solder is affected by the growth of IMC in Sn58Bi/Cu, resulting in varying degrees of coarsening. Analysis suggests that the entire coarsening process involves the aggregation, dissolution, and separation of Bi phase. This article elucidates the reaction mechanism of IMC growth at the Sn58Bi/Cu solder joint interface and its influence on the coarsening of Bi phase inside the solder, providing theoretical support for improving the reliability of Sn58Bi solder in the future.

The Gerd-Q scores during the postoperative follow-up were collected to determine the incidence of GERD. A logistic regression analysis was performed to explore the correlation of the HREM results and general clinical data with the incidence of GERD after LSG.Results: The percentage of synchronous contractions, lower esophageal sphincter (LES) resting pressure, and history of smoking were correlated with the development of GERD after LSG, with the history of smoking and percentage of synchronous contractions as risk factors and LES resting pressure as a protective factor. The training set showed an area under the ROC curve (AUC) of the nomogram model of 0.847. The validation set showed an AUC of 0.761. The decision and clinical impact curves showed a high clinical value for the prediction model.Conclusion: The HREM results correlated with the development of GERD after LSG, with the percentage of synchronous contractions and LES resting pressure showing predictive value. This study endeavours to establish a rapid, precise, and straightforward method for detecting SLCO1B1 genetic variants utilizing Duplex Fluorescence Melting Curve Analysis (DFMCA).Methods: Whole blood samples were collected from 54 individuals from Meizhou People’s Hospital (2023.01– 2023.03), with a mean age of 58.90 years (SD = 7.86), including 28 men and 26 women. DNA was extracted from these samples and subjected to PCR amplification targeting two allelic regions. Primers, fluorescent probes, and corresponding allelic target sequences were designed specifically for two common SLCO1B1 polymorphisms (rs2306283 and rs4149056). The functionality of the fluorescent probes in binding to their respective allelic targets was verified using melting curve analysis, enabling the identification of distinct melting temperatures for different genotypes. Subsequently, DFMCA was employed to differentiate genotypes based on the melting temperature shifts of the corresponding fluorescent probes. The sensitivity, accuracy, and consistency of the method were evaluated, with sequencing validation performed on a subset of samples.Results: DFMCA facilitated the concurrent detection and accurate genotyping of both polymorphisms within 2 hours, demonstrating concordance with sequencing results from randomly selected samples. Importantly, stable detection performance was achieved for human genomic DNA at concentrations ≥ 3.125 ng. In a cohort comprising Han Chinese individuals from southern China, the allele frequencies for rs2306283 (A: 28.7%, G: 71.3%) and rs4149056 (T: 88.89%, C: 11.11%) concurred well with previous studies in the Han Chinese population.Conclusion: The SNP typing system utilizing DFMCA technology presents advantages in terms of speed, ease of use, accuracy, and cost-effectiveness, making it a suitable tool.Keywords: SLCO1B1, melting curve analysis, gene polymorphism, drug delivery

Therefore the study aimed to evaluate the measurement accuracy of the G.LAB MD41A0 oscillometric automatic upper-arm blood pressure monitor in pregnancy and pre-eclampsia women according to the AAMI/ESH/ISO Universal Standard (ISO 81060– 2:2018).Methods: A total of 45 pregnant women were included in the cross-sectional validation study. The same left-arm sequential method was used for blood pressure measurement, and the blood pressure differences between the test device and the mercury standard reference were assessed according to the AAMI/ESH/ISO Universal Standard.Results: The participants included 15 normotensives, 15 gestational hypertension and 15 pre-eclampsia. The average (mean ± SD) differences between the device and mercury standard was − 0.84± 3.88 mmHg and − 0.58± 3.35 mmHg for systolic and diastolic blood pressure, respectively, for the validation Criterion 1. The SD of the averaged differences between the device and reference readings per participant was 2.92 mmHg and 2.28 mmHg for systolic and diastolic blood pressure, respectively, for the Criterion 2 of the universal standard.Conclusion: The G.LAB MD41A0 automatic upper-arm blood pressure monitor fulfills the criteria of the AAMI/ESH/ISO Universal Standard and can be recommended for clinical use and self-measurement in pregnancy and pre-eclampsia.Keywords: blood pressure monitor, validation, universal standard, pregnancy, pre-eclampsia

Abstract The oral and maxillofacial region comprises a variety of organs made up of multiple soft and hard tissue, which are anatomically vulnerable to the pathogenic factors of trauma, inflammation, and cancer. The studies of this intricate entity have been long-termly challenged by a lack of versatile preclinical models. Recently, the advancements in the organoid industry have provided novel strategies to break through this dilemma. Here, we summarize the existing biological and engineering approaches that were employed to generate oral and maxillofacial organoids. Then, we detail the use of modified co-culture methods, such as cell cluster co-inoculation and air-liquid interface culture technology to reconstitute the vascular network and immune microenvironment in assembled organoids. We further retrospect the existing oral and maxillofacial assembled organoids and their potential to recapitulate the homeostasis in parental tissues such as tooth, salivary gland, and mucosa. Finally, we discuss how the next-generation organoids may benefit to regenerative and precision medicine for treatment of oral-maxillofacial illness.

Abstract To investigate the relationship between Leisure time physical activity (LTPA) patterns and PhenoAgeAccel in patients with Type 2 diabetes (T2D), emphasizing the role of regular LTPA in mitigating biological aging. This study utilized data from the National Health and Nutrition Examination Survey (NHANES) from 1999 to 2018, including 4,134 adults with T2D. Multivariable linear regression models and restricted cubic spline (RCS) methods were employed to assess the relationship between LTPA and Phenotypic age acceleration (PhenoAgeAccel), with segmented likelihood ratio tests to detect nonlinear thresholds. Stratified regression and interaction tests were conducted for robust analysis. Abstract Reliable traffic flow data is not only crucial for traffic management and planning, but also the foundation for many intelligent applications. However, the phenomenon of missing traffic flow data often occurs, so we propose an imputation model for missing traffic flow data to overcome the randomness and instability bands of traffic flow. First, k-means clustering is used to classify road segments with traffic flow belonging to the same pattern into a group to utilize the spatial characteristics of roads fully. Then, the LSTM networks optimized with an attention mechanism are used as the base learner to extract the temporal dependence of the traffic flow. Finally, the AdaBoost algorithm is used to integrate all the LSTM-attention networks into a reinforced learner to impute the missing data. To validate the effectiveness of the proposed model, we use the PeMS dataset for validation, we impute the data with missing data rate from 10 to 60% under three missing modes, and we use multiple baseline models for comparison, which confirms that our proposed model improves the stability and accuracy of imputing the missing data of the traffic flow with different scenarios.

Abstract Double-atom catalysts (DACs) have opened distinctive paradigms in the field of rapidly developing atomic catalysis owing to their great potential for promoting catalytic performance in various reaction systems. However, increasing the loading and extending the service life of metal active centres represents a considerable challenge for the efficient utilization of DACs. Here, we rationally design asymmetric nitrogen, sulfur-coordinated diatomic iron centres on highly defective nitrogen-doped carbon nanosheets (denoted A-Fe2S1N5/SNC, A: asymmetric), which possess the atomic configuration of the N2S1Fe-FeN3 moiety. The abundant defects and low-electronegativity heteroatoms in the carbon-based framework endow A-Fe2S1N5/SNC with a high loading of 6.72 wt%. Furthermore, A-Fe2S1N5/SNC has a low overpotential of 193 mV for the oxygen evolution reaction (OER) at 10 mA cm−2, outperforming commercial RuO2 catalysts. In addition, A-Fe2S1N5/SNC exhibits extraordinary stability, maintaining > 97% activity for over 2000 hours during the OER process. This work provides a practical scheme for simultaneously balancing the activity and stability of DACs towards electrocatalysis applications.

Hesperidin is a dihydroflavonoids, accounting for more than 50% of the total flavonoids in Citri Reticulatae Pericarpium (CRP) of traditional Chinese medicine. It is an effective antioxidant and free radical scavenger that has anti-inflammatory, antiviral and hypoglycemic properties. The latest studies reported that hesperidin has a potential for novel coronavirus resistance. However, little is known about the synthesis regulation and accumulation site of hesperidin in plants. In this study, hesperidin synthase gene Crc1,6RhaT was cloned, and the protein can be completely transformed flavanone-7-O-glucoside into hesperidin in vitro and in vivo. Studies on biological characteristics of ovary walls and exocarps showed that the relative expression levels of the Crc1,6RhaT gene and protein decreased gradually with the development of citrus fruits, and the relative content of hesperidin firstly increased, then sequentially decreased. In situ hybridization results further revealed that Crc1,6RhaT transcription was mainly concentrated in the secretory cavity cells, which are revealed to be the site of flavonoid synthesis. Immunocytochemistry localization results showed that the Crc1,6RhaT was mainly located in the endoplasmic reticulum, nucleus and vacuole of secretory cells. We inferred that the Crc1,6RhaT was synthesized in the endoplasmic reticulum, then was transported into the vacuoles through enlarged vesicles at the end of the endoplasmic reticulum. Our results not only revealed that Crc1,6RhaT may be involved in the synthesis of hesperidin of the main bioactive substance in the medicinal plant Citrus reticulata ‘Chachi’ fruit, but also provided novel insights into the main subcellular sites of hesperidin biosynthesis in vacuoles.

Graft healing involves a series of cytological and molecular events including wound responses, callus formation and vascular bundle remodelling. Hormones are important signalling molecules regulating plant development and responses to environmental stimuli. However, the detailed dynamics of phytohormones in graft healing remain elusive. In this research, internodes above and below the graft site were harvested from 0 to 168 h after grafting (HAG), and liquid chromatography tandem mass spectrometry (LC-MS/MS) was used to determinate jasmonic acid, auxin, cytokinin, ethylene, salicylic acid, abscisic acid and gibberellin levels during the graft healing process. Uniform manifold approximation and projection (UMAP) and k-means analyses were performed to explore hormone spatio-temporal dynamics. We found the stage-specific and asymmetric accumulation of phytohormones in the tomato graft healing process. At the early healing stage (before vascular bundle reconnection), IAA, cZ, ABA, JA and SA mainly accumulated above the graft site, while tZ and ACC mainly accumulated below the graft site. MEIAA, ICAld and IP mainly accumulated at the later stage. Comminated with the healing process, we suggested that JA is mainly involved in wound responses, IAA is beneficial to the formation of callus and vascular cell development, tZ promotes cell division, and IP is linked to vascular bundle remodelling. In addition, expression of JA-related genes SlMYC2 and SlJAZ2, IAA-related gene SlIAA1, tZ-related genes SlHP2 and SlRR8, and IP-related gene SlRR9 correlated with hormone accumulation. The findings provide important information about the hormones and genes involved in the tomato graft healing process.

AIM: To explore the predictive value of the combination of triglyceride-glucose(TyG)index, nesfatin-1, and retinol-binding protein 4(RBP4)for diabetic retinopathy, and provide evidence for early prediction of DR.METHODS: The clinical data of 164 patients with type 2 diabetes mellitus(T2DM)who admitted to the hospital between February 2022 and December 2023 were retrospectively collected. Based on the fundus examination results, these patients were divided into two groups: the DR group(n=43), including proliferative DR(PDR, n=19)and non-proliferative DR(NPDR, n=24), and the T2DM without DR group(n=121). The TyG index and the level of nesfatin-1 and RBP4 were measured after admission.RESULTS:T2DM patients with DR had a longer disease duration compared with T2DM patients without DR, and the DR group had higher fasting blood glucose, glycosylated hemoglobin, triglyceride, total cholesterol, low-density lipoprotein, TyG index, and RBP4 levels, while lower high-density lipoprotein and nesfatin-1 levels(all P

The compressive creep performance of Ti alloy is critical for the application of submersibles due to the pressure hull of deep-diving equipment mainly subjected to the seawater pressure during its service. The compressive creep behaviors and the corresponding deformation mechanism of Ti–6Al–4V ELI alloy under different applied stress amplitudes were investigated. It was found that the stress threshold for Ti–6Al–4V ELI alloy produced significant compressive creep damage was 0.8 times yield strength (Rp0.2), and the primary mechanism was {101‾0}⟨112‾0⟩ prismatic slip. Specifically, a large number of prismatic slips were preferentially activated in the αp grains with a relative high Schmid factor when the applied stress amplitude was greater than 0.8Rp0.2. As the applied stress amplitude increases, the prismatic slips were also activated in the αlath. Meanwhile, the Burgers relationship between αlath and βlamellae gradually destroyed due to the dislocation pile-up at the αlath/βlamellae interface. This investigation provides data support for the compressive creep performance evaluation of Ti–6Al–4V ELI pressure hull.

With the increasing demand for large-scale, lightweight, and highly safe ocean transportation vessels and drilling equipment, as well as the growing need for environmentally friendly steels, the demand for high-strength, high-toughness, and weldable thick steel plates for ships and ocean engineering is becoming increasingly significant. However, as the strength and thickness of steel plates increase, the low-temperature toughness of the center of high-strength and extra-thick steel plates has become a significant challenge. Owing to low rolling reduction and central segregation in ultra-heavy plate steel, the poor low-temperature toughness of the central region poses a major challenge that limits the application of high-strength ultra-heavy steels. This work systematically investigated the effects of a two-step intercritical heat treatment on regulating the multiphase microstructure and properties of 100 mm EH500 marine engineering steel with severe central segregation. The results showed that after intercritical annealing in the 740 ℃ two-phase region, the experimental steel exhibited a yield strength of 540 MPa and a tensile strength of 869 MPa. However, the elongation and low-temperature toughness at −40 ℃ were relatively low, at only 5.1% and 14 J, respectively. Subsequent tempering at 600 ℃, 660 ℃, and 680 ℃ did not significantly alter the yield strength of the experimental steel, which remained within the range of 528 MPa to 551 MPa, while the tensile strength decreased to between 687 MPa and 730 MPa. The elongation and low-temperature toughness of the experimental steel initially increased and then decreased with the tempering temperature. At a tempering temperature of 660 ℃, the plasticity and toughness were optimized, with an elongation of 30.6% and a Charpy impact energy of 163 J at −40 ℃. Microstructure characterization results indicated that the experimental steel annealed at 740 ℃ consisted of intercritical ferrite (IF) and martensite (M). After further tempering at 600 ℃, a multi-phase microstructure comprising IF and tempered martensite (TM) with fine carbides was obtained. At a tempering temperature of 660℃, the microstructure of the experimental steel consisted of IF, TM, and fine retained austenite (RA). The RA content in the central segregation region was significantly higher than that in the matrix, resulting in a significant improvement in the plastic toughness of the experimental steel. With the further increase in the tempering temperature to 680 ℃, the experimental steel realized an IF and TM structure, with a small proportion of RA and a significant fraction of martensite/austenite (M/A) constituents in the central segregation zone. The large fraction of M/A constituents could substantially deteriorate the plasticity and toughness of the experimental steel.

This study investigates the effects of cold rolling and annealing on the microstructure, texture, and mechanical properties of FeCoCrNiMn high-entropy alloys. Utilizing vacuum induction melting, the alloy was initially cast into ingots and hot-rolled into plates, which were subsequently cold-rolled to various thicknesses and annealed at different temperatures. Microstructural analyses were conducted using X-ray diffraction and electron backscatter diffraction techniques, revealing a persistent face-centered cubic structure across all conditions. The texture evolution demonstrated a shift from Copper and S components to dominant Goss and Brass components as cold rolling intensified, suggesting the formation of Brass-type texture in FeCoCrNiMn at high deformation. Mechanical testing showed that the alloy's yield and tensile strengths significantly increased with cold rolling, reaching optimum values at ∼66 % reduction. Annealing at 750 °C enhanced both strength and ductility, primarily through grain refinement and the formation of Σ3 annealing twin boundaries, which dominated the microstructure of recrystallized grains. The study confirms that the low stacking fault energy of the alloy facilitates the activation of twinning and transformation-induced plasticity mechanisms, crucial for the observed enhancements in mechanical properties.

The significant effect of Ni content (0.92, 1.94 and 2.94 wt%) on ductile-brittle transition temperature (DBTT) and microstructure in a 1000 MPa grade high-strength low-alloy (HSLA) steel was studied. Using scanning electron microscopy (SEM), electron backscatter diffraction (EBSD), transmission electron microscopy (TEM), Charpy impact test and low-temperature tensile test to study the fundamental reasons for the effect of Ni content on toughness. The results indicated that increasing the Ni content can reduce the DBTT of HSLA steel and improve the impact toughness at low temperatures. EBSD data post-processing analysis revealed that the key reason for the increase in low-temperature toughness is the refinement of the microstructural crystallographic structure, specifically the significant increase in the boundary density of the block and packet. With the increase of Ni content, the density of grain boundary with an orientation difference greater than 5° between two adjacent {110} crystal planes was higher, which can form a higher density of dislocation pile-up group, thus better reducing local stress concentration. Meanwhile, the stacking fault energy (SFE) increases with the increase of Ni content, which made the screw dislocation more prone to cross slip at low temperature, resulting in an increase in plasticity at low temperatures. These observed phenomena and reasons provided a theoretical explanation for the role of Ni content in reducing DBTT and enhancing the toughness of the core in heavy gauge plates.

Lanthanum lithium titanate is one of the promising electrolytes for solid-state lithium-ion batteries due to its high bulk ionic conductivity up to ∼10−3 S/cm. However, the practical application of this material has been bottlenecked by high grain boundary (GB) resistance, while the underlying mechanism is still under debate. Here we report a comprehensive study with direct evidence to reveal the origin of high GB resistance in La2/3–xLi3xTiO3 (LLTO). Atomic-scale observations via advanced scanning transmission electron microscopy show that the GBs are uniformly subject to subsurface segregation of La atoms to compensate for the excess surface charges. The La segregation leads to an abrupt decrease of charge carrier concentration neighboring GBs and hence is supposed to have deleterious effect on GB conductivity. The findings suggest a novel mechanism of space-charge-induced cation segregation, which shed lights on the intrinsic origin of low GB ionic conductivity in LLTO.

Objective To investigate the efficacy and safety of 4-week cluster immunotherapy with house dust mite extracts in patients allergic to house dust mites. Methods From August 2022 to May 2024， 80 patients with house dust mite-induced allergic rhinitis， complicated with or without asthma， were recruited from multiple centers （including Peking Union Medical College， et al） to undergo 4-week dose-escalation subcutaneous cluster immunotherapy with house dust mites. Patient symptoms， medication use， quality of life， asthma control test （ACT） results， as well as local and systemic adverse reactions were recorded at 0， 1， 6， and 12 months， respectively. Results Significant differences were observed in the daily total symptom scores （P < 0.001）， daily nasal scores （P < 0.001）， daily ocular scores （P = 0.009）， and total symptom and medication scores （P = 0.003） across different time points during cluster immunotherapy， respectively. Significant differences were also found in the activity RQLQ （P < 0.001）， sleep RQLQ （P = 0.002）， non-eye/nose symptom RQLQ （P = 0.032）， practical problems RQLQ （P < 0.001）， nasal symptom RQLQ （P < 0.001）， emotional RQLQ （P = 0.011）， total RQLQ （P < 0.001）， and children’s ACT scores （P < 0.001）， respectively. The longer the time， the lower the score. During the dose-escalation period， 22 patients （27.5%） experienced local adverse reactions， and 5 patients （6.3%） experienced systemic adverse reactions. A total of 35 （3.6%） injections resulted in adverse reactions， including 27 （2.8%） injections with local reactions and 8 （0.8%） injections with systemic reactions. Conclusion Four-week cluster immunotherapy with house dust mites shows significant efficacy in patients allergic to house dust mites.

Abstract Insulation deterioration, which is mainly caused by partial discharge (PD) occurring inside power transformers, is one of the prime reasons to cause transformer faults. Therefore, an effective diagnosis of PD is crucial to ensure the safe and stable operation of transformers. To extract more effective features that characterise transformers PD signals and enhance the recognition accuracy, a novel transformer PD fault diagnosis model based on improved adaptive local iterative filtering (ALIF) and bidirectional long short‐term memory (BILSTM) neural network is proposed. Addressing the issue of predetermined decomposition levels and accuracy in ALIF decomposition, the golden jackal optimisation (GJO) algorithm is introduced to optimise the parameters. The proposed fault diagnostic model extracts dominant PD features employing the improved ALIF and Refined Composite Multi‐Scale Dispersion Entropy and improves the diagnostic accuracy with the optimised BILSTM by introducing GJO. Experimental data evaluates the performance of support vector machine, long short‐term memory and BILSTM. The results verify the effectiveness and superiority of the proposed model.

Abstract In order to further improve the output torque and stability of the external rotor permanent magnet synchronous motor, a double‐layer non‐uniform Halbach external rotor permanent magnet synchronous motor (DNH) structure is proposed and optimised. Finite element models of DNH and other structures are established to compare the electromagnetic characteristics of these motors and verify the superiority of the proposed structure. Secondly, the multi‐objective whale optimisation algorithm is improved, and the algorithm is tested by test function to verify the improvement effect. Further, the Spearman correlation of the parameters is calculated, and the parameters are layered. The first layer parameters are optimised by the improved multi‐objective whale optimisation algorithm, and the second layer parameters are optimised by parameter scanning. Finally, the motor performance of the initial scheme, single layer optimisation scheme, and hierarchical optimisation scheme is compared by simulation. The simulation results show that the hierarchical optimisation method is superior to the single layer optimisation method. By comparing with other schemes, the performance advantages of hierarchical optimisation DNH in terms of average output torque, cogging torque, torque ripple, permanent magnet cost and other parameters are proved, as well as the effectiveness of the optimisation method.

This study aims to investigate the crack propagation mechanism of directional hydraulic fracturing of coal mine roof and explore the relationship between fracture criteria. By drawing on fracture mechanics, we propose a hydraulic fracturing mechanical model of drilling and derive the calculation formulas of crack initiation angle and critical water pressure under different fracture criteria to analyze the impact of different variables on crack initiation angle and critical water pressure. Results show that: ① the increase of stress difference leads to a gradual increase of the crack initiation angle; ② the effect of prefab crack length and borehole radius on crack initiation angle is caused by two factors. The crack initiation angle increases with the decrease in the ratio of these two factors; ③ Poisson's ratio has limited effect on crack initiation angle; ④ under high stress difference and low prefab crack inclination angle, the critical water pressure decreases with the increase of stress difference and inclination angle, and vice versa; ⑤ the increase of borehole radius and prefabricated crack length would significantly reduce the critical water pressure. The two criteria yield different calculation results as ① prefab crack length and borehole radius would impact crack initiation angle; ② principal stress difference and prefab crack angle would impact critical water pressure. The theoretical calculation results and the experimental results show a good fit. The present study offers reference to the hydraulic fracturing site construction.

Abstract Diboson production in association with jets is studied in the fully leptonic final states, pp → (Z/γ *)(Z/γ *) + jets → 2ℓ2ℓ′ + jets, (ℓ, ℓ′ = e or μ) in proton-proton collisions at a center-of-mass energy of 13 TeV. The data sample corresponds to an integrated luminosity of 138 fb −1 collected with the CMS detector at the LHC. Differential distributions and normalized differential cross sections are measured as a function of jet multiplicity, transverse momentum p T, pseudorapidity η, invariant mass and ∆η of the highest-p T and second-highest-p T jets, and as a function of invariant mass of the four-lepton system for events with various jet multiplicities. These differential cross sections are compared with theoretical predictions that mostly agree with the experimental data. However, in a few regions we observe discrepancies between the predicted and measured values. Further improvement of the predictions is required to describe the ZZ+jets production in the whole phase space.

BackgroundMercury, as a global heavy metal pollutant, poses a serious threat to human health. The toxicity of mercury depends on its chemical form. Distinguishing the forms of mercury in the environment is of great significance for mercury management and reducing human mercury exposure risks. ObjectiveTo establish a non-targeted metallomics method based on synchrotron radiation X-ray fluorescence (SRXRF) spectroscopy combined with machine learning to screen inorganic mercury (IHg) or methylmercury (MeHg) exposed rice plants. MethodsRice seeds were exposed to ultra-pure water (control group), 0.1 mg·L−1 IHg (IHg group) or MeHg (MeHg group) solutions, respectively. After germination, the seedlings were cultured for 21 d, and rice leaves were collected, dried, weighed, and pressed. The content of metallome in rice leaves was determined by SRXRF. Machine learning models including soft independent modeling cluster analysis (SIMCA), partial least squares discriminant analysis (PLS-DA), and logistic regression (LR) were used to classify the SRXRF full spectra of different groups and find the best model to distinguish rice exposed to IHg or MeHg. Besides, characteristic elements were selected as input parameters to optimize the model by improving computing speed and reducing model calculation. ResultsThe SRXRF spectral intensities of the control group, IHg group, and MeHg group were different, indicating that exposure to IHg and MeHg can interfere the homeostasis of metallome in rice leaves. The results of principal component analysis (PCA) of SRXRF spectra showed that the control group could be well distinguished from the mercury exposed groups, but the IHg group and the MeHg group were mostly overlapped. The accuracy rates of the three models (PLS-DA, SIMCA, and LR) were higher than 98% for the training set, higher than 95% for the validation set, and higher than 94% for the cross-validation set. Besides, the accuracy of the LR model was higher than that of the PLS-DA model and the SIMCA model. Furthermore, the accuracy was 92.05% when using characteristic elements K, Ca, Mn, Fe, and Zn selected by LR to distinguish the IHg group and the MeHg group. Compared with the full spectra model, although the prediction accuracy of the characteristic spectral model decreased, the input parameters of the model decreased by 99.51%, and precision, recall, and F1 score were above 84.48%, indicating that the model could distinguish rice exposed to different mercury forms. ConclusionNon-targeted metallomics method based on SRXRF and machine learning can be applied for high-throughput screening of rice exposed to different forms of mercury and thus decrease the risks of people being exposed to mercury.

Improving the accuracy of solar power forecasting is crucial to ensure grid stability, optimize solar power plant operations, and enhance grid dispatch efficiency. Although hybrid neural network models can effectively address the complexities of environmental data and power prediction uncertainties, challenges such as labor-intensive parameter adjustments and complex optimization processes persist. Thus, this study proposed a novel approach for solar power prediction using a hybrid model (CNN-LSTM-attention) that combines a convolutional neural network (CNN), long short- term memory (LSTM), and attention mechanisms. The model incorporates Bayesian optimization to refine the parameters and enhance the prediction accuracy. To prepare high-quality training data, the solar power data were first preprocessed, including feature selection, data cleaning, imputation, and smoothing. The processed data were then used to train a hybrid model based on the CNN-LSTM-attention architecture, followed by hyperparameter optimization employing Bayesian methods. The experimental results indicated that within acceptable model training times, the CNN-LSTM-attention model outperformed the LSTM, GRU, CNN-LSTM, CNN-LSTM with autoencoders, and parallel CNN-LSTM attention models. Furthermore, following Bayesian optimization, the optimized model demonstrated significantly reduced prediction errors during periods of data volatility compared to the original model, as evidenced by MRE evaluations. This highlights the clear advantage of the optimized model in forecasting fluctuating data.

Objective To explore the application of ALA photodynamic fluorescence combined with dermatoscopy in the diagnosis of facial cancerization and precancerous lesions. The fall webworm (Hyphantria cunea) poses a significant threat to agriculture, as its larvae feed on leaves and form silken webs, which can severely impact plant growth. However, the lack of specific image datasets for the larvae’s webs hinders the use of image recognition technologies in pest prevention and control. To address this issue, an enhancement method is proposed here based on an improved Deep Convolutional Generative Adversarial Network (DCGAN). This method generates a diverse set of high-quality web images, significantly expanding the existing dataset. Experimental results demonstrated that this enhanced dataset improved the robustness of recognition networks, enabling better automatic identification and precision spraying to control Hyphantria cunea. This approach not only advances automated pest monitoring in agriculture but also offers new possibilities for applying similar technologies to the identification of other plant pests.

COVID-19 posed a major challenge to the healthcare system and resources worldwide. The popularization of vaccines and the adoption of numerous prevention and control measures enabled the gradual end of the COVID-19 pandemic. However, successive occurrence of autoimmune diseases in patients with COVID-19 cannot be overlooked. Long COVID has been the major focus of research due to the long duration of different symptoms and the variety of systems involved. Autoimmunity may play a crucial role in the pathogenesis of long COVID. Here, we reviewed several autoimmune disorders occurring after COVID-19 infection and the pathogenesis of long COVID.

Abstract Introduction The role of complement system in late stage of IgA nephropathy (IgAN) remains unknown. We therefore investigated the effects of complement system on worsening kidney function in advanced (stage 4 CKD) IgAN. Methods Renal specimens of 69 IgAN patients who underwent renal biopsy during stage 4 CKD between 2010 and 2021, were stained using immunofluorescence (IF) and immunohistochemistry (IHC) for glomerular complement components. The primary outcome was progression to end-stage renal disease (ESRD). Associations of complement components with baseline clinicopathological characteristics and outcomes were assessed using multivariable Cox regression and Spearman analyses. Results During a median follow-up of 18.0 months, 26 (37.7%) patients progressed to ESRD and none died. C1q and C3 deposition were detected in 12 and 66 patients, respectively. Abstract Many observational studies and experiments have found a strong association between lipid levels and adipokines and multiple myeloma (MM), but the causal relationship between lipid levels, adipokines and MM remains to be determined. We performed a two-sample and multivariate MR analysis to investigate the causal relationship between lipid levels, adipokines and MM. Total cholesterol(TC), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), and triglycerides (TG) were used to represent lipid levels, and adiponectin, leptin, and resistin were used to represent adipokines. Genetic data for each index and MM were obtained from the Integrated Epidemiology Unit (IEU) Genome-Wide Association Study (GWAS) database, and two-sample MR analyses were performed, as well as multivariate MR analyses of adipokines for causality of MM using BMI as an adjusting factor. In the analyzed results, no significant causal association was found between adipokines, lipid levels and multiple myeloma, and after adjusting for BMI, an association between adipokines and MM was still not found. The results of this MR study do not support an association between genetically predicted adipokines, lipid levels, and risk of MM, but we cannot rule out the existence of a weak association. The mechanisms need to be further investigated.

Abstract Saposhnikovia divaricata is commonly used in clinical practice as a heat-clearing and detoxifying traditional Chinese medicine. Current research on Saposhnikovia divaricata has mostly focused on chromones, while studies on its lipophilic parts remain scarce. In this study, samples of the lipophilic components of Saposhnikovia divaricata (SDL) from different regions in China were collected to establish an HPLC fingerprint. The quality of SDL was evaluated through chemical pattern recognition and multi-component content determination. Furthermore, using the Caenorhabditis elegans (C. elegans) model, the anti-aging ability of SDL was preliminarily studied. In the HPLC fingerprints of 11 batches of SDL, 19 common peaks were identified, and 4 components were characterized as follows: peak 14, linolenic acid (ALA); peak 15, docosahexaenoic acid (DHA); peak 16, linoleic acid (LA); peak 17, oleic acid (OA). The similarity of the fingerprint ranged from 0.704 to 0.929. The results indicate that the content of ALA, DHA, LA, and OA are 0.040–0.440 mg/g, 0.003–0.116 mg/g, 1.047–6.460 mg/g, and 0.040–0.294 mg/g, respectively. HCA analysis divided 11 batches of SDL into two categories, and PCA analysis extracted 6 principal components with a cumulative contribution rate of 85.91%. The anti-aging ability of the SDL was proved by measuring the lifespan, reproductive capacity, body length, mobility, lipofuscin content, and heat stress resistance of C. elegans. The combination of high-performance liquid chromatography fingerprint and chemical pattern recognition for multi-component content determination is a reliable, comprehensive, and promising evaluation of the quality of SDL. The confirmation of SDL’s anti-aging ability provides the possibility for the research of Saposhnikovia divaricata in the field of healthy food and cosmetics.

Abstract Background: The use of immune checkpoint inhibitors (ICIs) as neoadjuvant therapy is a promising novel approach in resectable non-small-cell lung cancer (NSCLC). This study aimed to investigate the prognostic value of PD-L1 in patients with NSCLC receiving neoadjuvant immune checkpoint inhibitor plus chemotherapy (CT). Materials and methods: Several databases (PubMed, Embase, and cochrane central register of controlled trials [CENTRAL]) were systematically searched. Randomized controlled trials (RCTs) investigating pathological and survival outcomes with neoadjuvant ICI + CT versus CT alone in NSCLC were analyzed. Abstract Aims Night blindness (NB), an important manifestation of VA deficiency, may be associated with the odds of diabetes. The aim of this study was to explore the probable association between NB history and diabetes in Chinese community-dwelling adults. Methods This multi-center, cross-sectional study enrolled a total of 5664 participants aged 18–82 years from eight sites in China. Information on demographics and medical history was collected using a standardized questionnaire. Diabetes was diagnosed based on the oral glucose tolerance test or a self-reported history. NB history was ascertained by a face-to-face interview with reference to the recommendation by the World Health Organization. Logistic regression analysis was used to evaluate the association between NB history and the odds of diabetes. Results A total of 5049 participants were finally included, with 252 ascertained with NB history and 1076 with diabetes. The mean age of included participants was 52.9 years, and the percentage of participants with NB history was significantly higher in participants with diabetes than those without (7.0% vs. 4.5%). The multivariable adjusted odds ratio for diabetes was 1.41 (95% confidence interval 1.06, 1.89) in participants with NB history compared with those without. Furthermore, mediation analysis showed that obesity, as assessed by waist-height ratio, partially mediated the relationship between NB history and increased odds of diabetes. Conclusions The results suggest that NB history might be associated with increased odds of diabetes in Chinese community-dwelling adults.
