This study focuses on urban farming practices in Bandung, West Java, known as Buruan SAE, a programme initiated by the Department of Food Security and Agriculture, Bandung. The objectives of this study are to analyse the effects of urbanisation on urban farming and to explore the potential of urban farming in supporting food security. This study uses a qualitative approach. The results of the study show 3 important issues: the diverse production of urban farming is able to meet the food needs of community members and some have already become independent farmers; there has been a change in the attitude of the people of Bandung city, through urban farming they have become ‘farmers’ a completely new activity for them; urban farming provides a source of income for the urban community. These findings suggest that urban farming can enhance the sustainability of the Buruan SAE community.

Background: Online home exercises represent opportunities to increase physical activity levels. However, high dropout rates are commonly reported in such programmes. This study aimed to investigate the predictors of dropping out from an online home exercise programme. Methods: A total of 760 individuals from nine countries participated in this 8-week prospective cohort study derived from a randomised controlled trial. The participants were randomised into "4-week live-streamed exercise –>4-week recorded exercise" or "4-week no intervention –>4-week recorded exercise" group. Repeated measurements using weekly questionnaires were performed. Pain intensity, disability, mental well-being score, exercise motivation, sleep quality, impulsiveness/anxiety, and physical activity level were analysed. Results: A total of 53.8% (95% confidence interval [CI] 50.3%–57.3%) participants dropped out from the programme. The identified predictors of dropping out from the programme were: well-being (odds ratio [OR] 0.94, 95% CI 0.91–0.97) and disability (OR 1.02, 95% CI 1.002–1.04) at baseline considering the first 4 weeks; age (0.98; 95% CI 0.96–1.00) and baseline well-being (0.93; 95% CI 0.89–0.97) considering the entire follow-up (8 weeks); exercise motivation (0.92; 95% CI 0.87 to 0.97) and general impulsiveness/anxiety (1.04; 95% CI 1.01–1.07) repeated measured over time. Conclusion: About half of the participants dropped out from the online home exercise programme. Higher baseline scores in mental well-being and age predicted a reduction in dropping out. Higher baseline disability predicted an increase in dropping out. During the follow-up, higher exercise motivation was associated with a reduction in dropping out, and higher impulsiveness and anxiety were associated with an increase in dropping out.

Abstract “Beyond-GDP” metrics are essential for understanding societal progress. Yet despite their importance, these metrics are scattered across various databases, hindering accessibility and interdisciplinary analysis. Addressing this gap, we present the ‘WISE database’ – the first extensive collection of important Beyond-GDP metrics organized by Wellbeing, Inclusion, and Sustainability (WISE) dimensions. The WISE database consolidates data from a variety of sources, including international institutions and academic publications. It encompasses over one million data points across 244 metrics, covering 218 countries and 61 country groupings, from specific social and environmental indicators to the main Beyond-GDP indexes, and is augmented by essential metadata. The data primarily spans from 1995 to 2015, with some metrics extending back to the 19th century. To improve accessibility and data interpretation, a user-friendly online visualization tool has been developed. The WISE database aims to foster a broader view of societal progress, facilitate research on synergies and trade-offs of wellbeing, inclusion, and sustainability, and provide a foundation for interdisciplinary research.

Background: Internship represents the context for nursing students to acquire professional skills. The mutual relationship between the intern and the tutor, where both parties have the opportunity to grow and develop, contributes to transforming theoretical knowledge into intellectual, psychomotor, and social skills. However, the gap between education and clinical practice has always challenged education experts. This study assesses the gap between the theory learned during lectures and the applied care practice during the internship, highlighting factors opposing knowledge translation. Methods: A prospective observational study was conducted in June and July 2022, involving clinical tutors (48 tutors) and third-year nursing students (107 students) recruited by convenience sampling. Both groups were administered a designed questionnaire built by the researchers, using semi-structured interviews in the related literature and adapted to the Italian context. Kabupaten Klaten mengalami perkembangan pesat dari tahun ke tahun karena adanya perkembangan aspek fisik, politik, ekonomi, sosial dan budaya. Untuk melihat perkembangan kota dari segi fisik maka harus diamati dari kenampakan fisik dari sistem jalan, blok bangunan permukiman serta blok bangunan bukan wilayah permukiman. Bentuk morfologi kota berbeda pada setiap bagiannya sesuai dengan sejarah pembentukan wilayah dapat dikaji dengan Citra Landsat 8. Penelitian ini bertujuan untuk mengetahui arah perkembangan wilayah kawasan perkotaan Kabupaten Klaten melalui pengamatan dengan Citra Landsat-8. Metode yang digunakan dalam penelitian ini adalah metode deskriptif kuantitatif. Metode deskriptif dalam penelitian ini untuk mengkaji interaksi wilayah menggunakan metode perhitungan teori gravitasi. Introduction. In the evolving landscape of anesthetic practices for short-duration urologic procedures, the quest for an anesthetic agent that balances efficacy with minimal side effects remains a significant clinical challenge.The objective was to compare the efficacy and safety of 2% hyperbaric prilocaine and 0.5% hyperbaric bupivacaine in spinal anesthesia.Materials and methods. A comparative analysis was performed in terms of onset and duration of sensory and motor block, incidence of side effects (hypotension and bradycardia), and overall patient outcomes in urologic endoscopy. The research sample was divided into two groups, each consisting of 20 patients. In one case, spinal anesthesia was performed using 2% hyperbaric prilocaine (40 mg) + fentanyl 25 mcg, in the other – 0.5% hyperbaric bupivacaine (10 mg) + fentanyl 25 mcg.Results. Prilocaine offers a faster onset of sensory and motor block and a shorter duration of sensory block compared to bupivacaine. Prilocaine also showed a quicker recovery of full motor function and had a significantly lower incidence of side effects such as hypotension and bradycardia.Conclusion. These results suggest that 2% hyperbaric prilocaine could be an alternative to 0.5% hyperbaric bupivacaine in spinal anesthesia for urologic endoscopy, especially in procedures requiring quick recovery. The promising results of prilocaine in such short-duration surgeries can also prompt a reevaluation of anesthesia protocols across various surgical interventions and lead to enhanced patient outcomes, emphasizing safety, comfort, and efficacy of surgical care.

Urban sprawl has become a concern in development because of its negative impact on sustainable development. Such a special trend is apparent in the transformation of a peri-urban area of Sinduadi Village that is directly affected by the city of Yogyakarta. The village is most likely affected by the urban sprawl phenomenon in the city of Yogyakarta, especially on the morphological aspect of the urban morphology. This study aims to analyze the urban morphology in the Sinduadi Village in 2021 as a result of the urban sprawl of Yogyakarta City. Using quantitative methods, this research is based on a primary data of obtained from field surveys through probability sampling method. The research data came from image interpretation and attribute data processing using ArcGIS 10.3 Software. Secondary data was obtained through Sentinel 2A Imagery while primary data was The data analysis used in this research is quantitative descriptive analysis and spatial analysis using maps. The analysis of the morphological form of the city is carried out by analysis of overlapping maps. This research found that the morphology of the city in the Sinduadi Village in 2021 is linear based on the center of the area on Magelang street This marked by activities in the center of the area are dominated by commercial activities, namely, trade and service. Therefore, regional needs can be accommodated. The center of the area has a high building density (BCR>70%). High density residential land surrounds the center of the area and its development follows a grid and spinal road network pattern. These findings show that Sinduadi village has experienced urban sprawl marked by development trend that trespasses its under bounded city.

Urban farming has been associated with sustainable living in urbanized areas. However, the interconnection between the two need further studies. Alternatives for promoting sustainable living is especially in important in Indonesia because urbanization in the country is expanding, while the area of productive agricultural land is decreasing. The limited spaces become a challenge for urban residents that need land for settlements, planting, green open spaces, spaces for interaction and socializing, and recreation. Therefore, urban farming presents an alternative in answering the problems caused by urbanization. This research aims to explore the interconnection of urban farming and urbanization in an effort to realize sustainable urban development. This research is the initial part of an effort to create an integrated model of urban farming for urbanization for food security for urban residents (2023-2025) in the Innovation Research for Advanced Indonesia (RIIM) scheme. This study uses qualitative methods with informants as a sample data source. Informants were selected using a purposive sampling technique, totaling 30 urban farming actors, with details of 10 informants from Palembang City, 10 from Bandung City, and 10 from Denpasar City. Collection through the stages of observation, in-depth interviews, and documentation. Data analysis uses the Miles and Hubberman Model with three stages, namely data reduction, data display, and drawing conclusions. The data validity test technique uses triangulation with the cross-check method. The results of the analysis found an interrelation between urban farming and urbanization in nine dimensions, namely; economy,social, environment, health, culture, food, education, tourism, and technology. The interrelationship between urban farming and urbanization is positive, which means that the positive impact of urban farming has an indication of minimizing the negative impact of urbanization. In addition, this interrelation is also relevant to the three principles of sustainable development, namely having the function of controlling environmental impacts to a minimum, economic and social functions.

Within Indonesia, the structure of consumption and production differs significantly across provinces. This implies that carbon footprints and intensities between provinces are also diverse. This paper calculates historical consumption- and production-based carbon emissions at the provincial level using a multi-scale input-output (IO) database for 2010, in which an environmentally extended multi-regional IO (EE MRIO) table for 34 Indonesian provinces is integrated in the global EE MRIO EXIOBASE with data for 43 countries and 5 rest of the world regions. Emissions from consumption are detailed by product and their points of origin, while emissions from production are detailed by industry and their destinations. Our results show the heterogeneity of Greenhouse Gas (GHG) emissions under both sides. The Java region is a net importer of carbon emission, while Sumatra and Kalimantan are net exporters. In the global context, the Asia Pacific region plays important role in national GHG emissions. Services product contributed 57.1% of national consumption-based GHG emissions, followed by manufacture (30.6%), and agriculture (12.3%). On the national level, 63.5% of national GHG emissions are related to household consumption. There is a high disparity across provinces in Indonesia in carbon footprints. Provincial average per capita carbon footprints vary from 2 t CO2e/capita in East Nusa Tenggara to 13.84 t CO2e/capita in East Kalimantan. Carbon intensity also varies from 0.83 kt CO2e/M Euro in Jakarta to 2.37 kt CO2e/M Euro in North Kalimantan. Agriculture and food products dominate household carbon footprints, while construction leads in government carbon footprints. Utilities and transportation services play important roles on national carbon intensities. We further correlated the Human Development Index (HDI) with per capita carbon footprints and expenditure, and find that provinces with similar GHG emissions and expenditure per capita income as Java, tend to have a lower HDI. Understanding development status and province-level characteristics is important for selecting policy strategies.

Abstract Background Preliminary studies suggest that moderate ARDS and acute renal failure might benefit from extracorporeal CO2 removal (ECCO2R) coupled with CRRT. However, evidence is limited and potential for this coupled treatment may need to be explored. The aim of the present study was to evaluate whether a protective driving pressure was obtained applying low-flow ECCO2-R plus CRRT in patients affected by moderate ARDS with COVID-19 compared to an historical group without COVID-19. Methods A case-control study has been conducted comparing a group of consecutive moderate ARDS patients presenting AKI and affected by COVID-19, who needed low-flow ECCO2-R plus CRRT to achieve an ultra-protective ventilatory strategy, with historical group without COVID-19 that matched for clinical presentation and underwent the same ultra-protective treatment. In 2011 the Indonesian and the German government initiated the Sustainable Economic Development through Technical and Vocational Education and Training Program (SED-TVET). Within the diplomacy realm, this kind of bilateral strategy has often been utilized to strengthen a country's specific needs. Following the program, Provincial Government in Central Java responds distinctively to the initiative by emphasizing link and match program to promote skilled-labor employment. This paper aims to investigate the effect of these national and local policies on firms’ productivity. Our identification of treatment year is critical as this precedes wider implementation of TVET program at the national level in 2016. To capture the precise treatment effect of the program, we deploy difference-in-difference strategy using the province’s neighbors as the control group. We find that firms in Central Java employed nearly 20% higher share of skilled labor relative to its controls. This correlates with significantly higher productivity and export performance. The results highlight the benefit of well-coordinated bilateral cooperation while also stressing the important role of local leadership in optimizing the impact of the program.

This research explores the impact of developing Accounting Information Systems (AIS) based on cutting-edge technology on company efficiency. We gathered data from questionnaires distributed to companies that have adopted state-of-the-art technology in their AIS. The research results indicate that the use of artificial intelligence (AI) significantly enhances operational efficiency in companies. Companies adopting blockchain technology also achieve higher levels of user satisfaction. User training and company size also have a positive impact on efficiency and better decision-making.In the discussion, we highlight the importance of investing in user training and maintaining state-of-the-art technology. We also acknowledge that implementation costs can be a constraint, particularly for smaller companies. This research has certain limitations, including a sample that may not cover the entire spectrum of industries and company sizes. However, further research can delve deeper through case studies. In the continually evolving business world, the development of AIS based on cutting-edge technology is a key factor in achieving efficiency, better decision-making, and user satisfaction. Companies must understand and address these challenges to attain a competitive edge and long-term success.

The pericope of demonic possession in Mark 5 invites us to reject a simplistic aversion to uninterpretable experiences. Engaging with Julia Kristeva’s concept of abjection encourages readers to recognize that embracing heterogeneous otherness can create new opportunities for both individuals and communities. This study critiques the limited and normative conception of mentally diverse bodies as being confined within a single boundary, and it represents the (un)interpretability of these bodies. Through a reading of Mark 5:1–20 that considers perceptions of the demonic, negotiating identity and agency, and a way of representation within a community, this study argues for the importance of mutuality, the experience of becoming a body of abject, and the epistemic acceptance of the uninterpretable in order to engage with the interpret-‘able’.

Latar Belakang: Seksio sesarea (SC) digunakan sebagai solusi ketika persalinan normal tidak memungkinkan karena alasan medis tertentu. Beban global pemulihan bedah obstetri mencakup sekitar 140.000.000 kelahiran setiap tahun dengan perkiraan tingkat sesarea global 23%. Salah satu masalah yang harus diatasi dalam pemulihan seksio sesarea adalah masalah nyeri. Bukti baru menunjukkan potensi terapeutik vitamin B1, B6, dan B12 dalam kondisi nyeri yang berbeda. Tujuan: untuk mengetahui efektivitas vitamin B1, B6, dan B12 terhadap intensitas nyeri pada seksio sesarea. Subjek dan Metode: Penelitian dengan desain eksperimental rancangan acak tersamar ganda pada pasien yang dilakukan seksio sesarea di RS Batara Siang Pangkep, bulan Januari hingga Februari 2024. Sebanyak 26 subyek penelitian dibagi rata dalam dua kelompok: kelompok kontrol tidak mendapat suplemen dan kelompok perlakuan mendapatkan vitamin B1 100 mg, vitamin B6 100 mg, vitamin B12 5000 mcg dalam bentuk sediaan ampul (Neurosanbe®) 1 jam sebelum operasi. Pencatatan intensitas nyeri (numerical rating scale = NRS) pada jam ke 2, 4, 8, 12, 24 pasca seksio sesarea, yaitu berupa nyeri diam dan nyeri gerak. Bladder cancer is a common type of cancer around the world, and the majority of patients are diagnosed with non-muscle-invasive bladder cancer (NMIBC). Although low-risk NMIBC has a good prognosis, the disease recurrence rate and development of treatment-refractory disease remain high in intermediate- to high-risk NMIBC patients. To address these challenges for the treatment of NMIBC, a novel combination therapy composed of an oncolytic adenovirus (oAd) co-expressing interleukin (IL)-12, granulocyte-macrophage colony-stimulating factor (GM-CSF), and relaxin (RLX; HY-oAd) and a clinical-stage glycogen synthase kinase (GSK)-3β inhibitor (9-ING-41; elraglusib) was investigated in the present report. Our findings demonstrate that HY-oAd and 9-ING-41 combination therapy (HY-oAd+9-ING-41) exerted superior inhibition of tumor growth compared with respective monotherapy in a syngeneic NMIBC tumor model. HY-oAd+9-ING-41 induced high-level tumor extracellular matrix (ECM) degradation and a more potent antitumor immune response than the respective monotherapy. In detail, HY-oAd+9-ING-41 induced superior accumulation of intratumoral T cells, prevention of immune cell exhaustion, and induction of tumor-specific adaptive immune response compared to either monotherapy. Collectively, these results demonstrate that the combination of HY-oAd and 9-ING-41 may be a promising approach to elicit a potent antitumor immune response against bladder cancer.

Dendritic cell (DC)-based vaccines have emerged as a promising strategy in cancer immunotherapy due to low toxicity. However, the therapeutic efficacy of DC as a monotherapy is insufficient due to highly immunosuppressive tumor environment. To address these limitations of DC as immunotherapeutic agent, we have developed a polymeric nanocomplex incorporating (1) oncolytic adenovirus (oAd) co-expressing interleukin (IL)-12 and granulocyte-macrophage colony-stimulating factor (GM-CSF) and (2) arginine-grafted bioreducible polymer with PEGylated paclitaxel (APP) to restore antitumor immune surveillance function in tumor milieu and potentiate immunostimulatory attributes of DC vaccine. Nanohybrid complex (oAd/APP) in combination with DC (oAd/APP+DC) induced superior expression level of antitumor cytokines (IL-12, GM-CSF, and interferon gamma) than either oAd/APP or DC monotherapy in tumor tissues, thus resulting in superior intratumoral infiltration of both endogenous and exogenous DCs. Furthermore, oAd/APP+DC treatment led superior migration of DC to secondary lymphoid organs, such as draining lymph nodes and spleen, in comparison with either monotherapy. Superior migration profile of DCs in oAd/APP+DC treatment group resulted in more prolific activation of tumor-specific T cells in these lymphoid organs and greater intratumoral infiltration of T cells. Additionally, oAd/APP+DC treatment led to lower subset of tumor infiltrating lymphocytes and splenocytes being immunosuppressive regulatory T cells than any other treatment groups. Collectively, oAd/APP+DC led to superior induction of antitumor immune response and amelioration of immunosuppressive tumor microenvironment to elicit potent tumor growth inhibition than either monotherapy.

Crash severity prediction is a challenging research area, where the objective is to accurately assess the extent of severity of an injury resulting from road traffic accidents. The main aim of existing studies is to precisely assess the potential severity of crashes under diverse circumstances, such as weather conditions, vehicle attributes, road characteristics and layout, and traffic control factors. This effort aids authorities in establishing effective emergency response systems. The novelty and objective of our work involve contributing to this research area by employing a graph architecture to capture relationships among various crash records to uncover any hidden patterns that traditional ML models might overlook. The current study extends existing knowledge by leveraging Graph Neural Networks (GNN) and comparing their performance to popular ensemble-based models, which include Extreme Gradient Boosting (XGBoost), Random Forest (RF), and Artificial Neural Networks (ANNs). Real data from the United Kingdom (UK) was employed to achieve our goal. The data was obtained from the Department for Transport open data portal. All models underwent training using the training dataset, followed by performance evaluation using diverse metrics such as the accuracy, precision, recall, f1-score, Matthews Correlation Coefficient (MCC), confusion matrix, and computational cost on the test dataset. Overall, our proposed GNN-based model demonstrated better performance when compared to other models. Specifically, the GNN model outperformed all other models across all metrics. For instance, the accuracy of the GNN model was 85.55% as compared to 83.36%, 83.18%, and 83.27% for the XGBoost, RF, and ANN models, respectively. The GNN model assisted in identifying hidden patterns by considering non-linear relationships among crash records. Thus, the model had the potential to improve its ability to predict severe accidents, which could in turn significantly improve emergency response efforts and reduce the likelihood of severe accidents resulting in fatalities.

The proliferation of high-dimensional data in many advanced database applications is a result of today’s technological advancements. These data points that correspond to objects are often without a precise description, which make their representation uncertain. While the concept of data streaming is not new, its practical uses are only recently emerging. This research focuses on continuous range data—a type of uncertain data common in database applications—that do not have explicit representations of their exact values. Furthermore, the identification of skyline objects—one of the popular database applications—becomes more challenging when skylines are to be identified from a collection of continuously generated input data streams where objects might have imprecise values. This makes it imperative to determine which approach has the optimal accuracy for estimating or predicting the uncertain values and at the same time able to handle a massive streams of data that are continuously generated and analyze them almost instantly to provide accurate and timely responses. Given this, the following techniques are selected—Linear Regression (LR), k-Nearest Neighbour (k-NN), Random Forest (RF), Decision Trees (DT), and Centre and Range Method (CRM) and their effectiveness is evaluated in terms of execution time, precision, recall, F1-score, and root mean square error (RMSE). Additionally, in order to verify the accuracy of each prediction technique, the predicted data derived from its model is used to derive skyline objects, which are subsequently compared to the actual skyline results. An inaccurate prediction of a continuous range value would result in incorrect set of skyline objects.

Pandemi Covid-19 memberikan dampak pada sektor perekonomian sehingga menyebabkan menurunnya aktivitas perekonomian di Desa Sriharjo khususnya bagi UMKM. Dampak yang dirasakan berupa menurunnya pendapatan akibat sepinya pembeli. Penelitian ini bertujuan untuk membahas dan menganalisis perilaku UMKM dalam menghadapi pandemi Covid-19 serta mengkaji strategi perubahan perilaku UMKM. Penelitian ini didasarkan pada focus group Discussion (FGD), wawancara, dan observasi terhadap UMKM di Desa Sriharjo Kabupaten Bantul Yogyakarta. Secara khusus menganalisis perilaku UMKM berupa pengetahuan, sikap dan tindakan pada masa pandemi Covid-19. Hasil penelitian menunjukkan bahwa pengetahuan, sikap, dan tindakan berpengaruh terhadap perilaku UMKM di Sriharjo. Menyikapi kondisi penuh tantangan di masa pandemi ini, para pelaku UMKM menentang keras adanya protokol kesehatan. Perilaku pelaku tercermin dalam penerapan protokol kesehatan dan adaptasi kebiasaan baru di tempat usaha. Strategi untuk mengatasi dampak ekonomi di Desa Sriharjo antara lain dengan membuat platform online, melalui grup WhatsApp dan website bernama pasardesa.id, untuk kegiatan jual beli. Hal ini difasilitasi berupa bantuan langsung dana desa, bantuan tunai Kementerian Sosial, dan BLT UMKM. This research aims to determine the role of the UMY campus in improving the welfare of residents around the campus. The method used was descriptive quantitative and uses a scoring analysis model. This research used several parameters collected through structured interviews and observations. These parameters were analyzed using a scoring/weighting method to determine the socio-economic conditions and level of welfare of residents around the UMY campus. The results show that 64% of the socio-economic conditions in Tamantirto Village are classified as moderate. Likewise, the welfare condition of the population is known to be 56%, including the middle class too. Based on the research results, it can be concluded that the UMY campus has a real role in improving the socio-economic welfare of the peripheral communities around the campus.

Masyarakat Desa Tegaltirto sebagian besar bermata pencaharian di sektor informal yang menjadi komponen paling rentan terhadap dampak Pandemi Covid-19. Ketahanan masyarakat diperlukan dalam menghadapi krisis dan ancaman Pandemi Covid-19. Penelitian ini bertujuan: (1) menganalisis tingkat ketahanan masyarakat menurut aset penghidupannya; dan (2) mengetahui faktor-faktor yang memengaruhi ketahanan masyarakat Desa Tegaltirto. Penelitian ini merupakan penelitian kuantitatif dengan metode survei. Populasi penelitian berdasarkan dusun yang dianggap mewakili Desa Tegaltirto menurut keragaman pekerjaan masyarakat yaitu Dusun Kadisono, Sompilan, dan Semoya. Teknik sampling menggunakan stratified random sampling jenis proporsional sampel sebanding jumlah populasi. Teknik pengumpulan data menggunakan kuesioner dan dokumentasi. Teknik analisis data menggunakan statistik deskritif untuk analisis tingkat ketahanan, dan analisis faktor untuk identifikasi faktor yang memengaruhi ketahanan masyarakat. Hasil penelitian menunjukkan: (1) Ketahanan masyarakat Desa Tegaltirto menurut kepemilikan aset menunjukkan tingkat ketahanan sedang, kategori tingkat ketahanan tinggi terdiri dari 9 rumah tangga, ketahanan sedang terdiri dari 75 rumah tangga, dan ketahanan rendah terdiri dari 16 rumah tangga. (2) Terbentuk 7 faktor yang mempengaruhi ketahanan masyarakat yaitu; faktor pendidikan dan perekonomian, pengetahuan dan akses informasi Covid-19, alam, ketersediaan sarana dan prasarana, pencegahan risiko Covid-19, bantuan dari pihak luar, dan kesehatan dan sosial masyarakat.

Tsunamis are natural hazards that have the potential to cause significant damage and losses to the South Coast of Java. As the initial foundation for local spatial planning and risk reduction, preparing a tsunami susceptibility mapping is imperative to minimize the disaster's impact. This study aims to identify the spatial distribution of tsunami susceptibility in Watukarung Village, Pacitan Regency, using the Spatial Multi-Criteria Evaluation (SMCE) method. The variables involved in the modeling include landform, elevation, slope, distance from the shoreline, and distance from the river. The value and weight of each factor were determined using the pairwise technique in the SMCE framework. The research results indicated that the tsunami susceptibility in Watukarung comprises four classes: safe (598.40 ha), low (9.68 ha), moderate (23.92 ha), and high (25.13 ha). The areas most prone to tsunamis are generally identified in the southern part of Watukarung, which is generally associated with beach or alluvial plain landforms, very close to shore, and low land elevation. Ironically, human settlements and coastal tourism are overgrowing in the tsunami-prone zone, highlighting that risk reduction measures must be implemented optimally to anticipate tsunami hazards. As a recommendation, further research must be carried out to comprehensively represent the tsunami risk in the Watukarung coastal region. Background The landscape of stroke care has shifted from stand-alone hospitals to cooperative networks among hospitals. Despite the importance of these networks, limited information exists on their characteristics and functional attributes.Methods We extracted patient-level data on acute stroke care and hospital connectivity by integrating national stroke audit data with reimbursement claims data. We then used this information to transform interhospital transfers into a network framework, where hospitals were designated as nodes and transfers as edges. Using the Louvain algorithm, we grouped densely connected hospitals into distinct stroke care communities. The quality and characteristics in given stroke communities were analysed, and their distinct types were derived using network parameters. The clinical implications of this network model were also explored.Results Over 6 months, 19 113 patients with acute ischaemic stroke initially presented to 1009 hospitals, with 3114 (16.3%) transferred to 246 stroke care hospitals. These connected hospitals formed 93 communities, with a median of 9 hospitals treating a median of 201 patients. Derived communities demonstrated a modularity of 0.904, indicating a strong community structure, highly centralised around one or two hubs. Three distinct types of structures were identified: single-hub (n=60), double-hub (n=22) and hubless systems (n=11). The endovascular treatment rate was highest in double-hub systems, followed by single-hub systems, and was almost zero in hubless systems. The hubless communities were characterised by lower patient volumes, fewer hospitals, no hub hospital and no stroke unit.Conclusions This network analysis could quantify the national stroke care system and point out areas where the organisation and functionality of acute stroke care could be improved.

Abstract Background Neuroacanthocytosis (NA) denotes a heterogeneous group of diseases that are characterized by nervous system abnormalities in association with acanthocytosis in the patients' blood. The 4.1R protein of the erythrocyte membrane is critical for the membrane-associated cytoskeleton structure and in central neurons it regulates the stabilization of AMPA receptors on the neuronal surface at the postsynaptic density. We report clinical, biochemical, and genetic features in four patients from four unrelated families with NA in order to explain the cause of morphological abnormalities and the relationship with neurodegenerative processes. Case presentation All patients were characterised by atypical NA with a novel alteration of the erythrocyte membrane: a 4.1R protein deficiency. The 4.1R protein content was significantly lower in patients (3.40 ± 0.42) than in controls (4.41 ± 0.40, P < 0.0001), reflecting weakened interactions of the cytoskeleton with the membrane. In patients IV:1 (RM23), IV:3 (RM15), and IV:6 (RM16) the 4.1 deficiency seemed to affect the horizontal interactions of spectrin and an impairment of the dimer self-association into tetramers was detected. In patient IV:1 (RM16) the 4.1 deficiency seemed to affect the skeletal attachment to membrane and the protein band 3 was partially reduced. Conclusion A decreased expression pattern of the 4.1R protein was observed in the erythrocytes from patients with atypical NA, which might reflect the expression pattern in the central nervous system, especially basal ganglia, and might lead to dysfunction of AMPA-mediated glutamate transmission.

Urbanization is the process of increasing the population migrating to urban areas, and urban farming is a farming practice carried out within the city. Both concepts have significant impacts on urban life and the environment. This research aims to analyze the interrelation of the positive impacts of urban farming and the negative impacts of urbanization. The analysis focuses on food and environmental aspects as alternative problem-solving methods in the cities of Bandung, Denpasar, and Palembang. This research uses qualitative method. Data collection uses field interview techniques with 30 informants, as well as a review of several scientific articles, previous research results, and reports from data-providing agencies such as the Indonesian Central Bureau of Statistics. Data analysis used the Miles and Huberman Model, with the stages of reduction, display, and conclusion drawing. The results of the analysis show that there is an interrelation between the positive impacts of urban farming and the negative impacts of urbanization in the cities of Bandung, Denpasar, and Palembang. In terms of food aspect, urban farming can provide various types of easily accessible food for urban dwellers, such as vegetables, fruits, fisheries, and small-scale livestock. The food produced is relatively close to residential areas, so the quality is still fresh and healthy and the price is affordable as well. In the environmental aspect, urban farming can optimize the use of limited or vacant land in residential areas into productive land, such as for green spaces, farming activities, gardening, fisheries, and livestock. Other functions include enhancing the beauty of the environment, reducing household waste, and preserving organism biodiversity.

AbstractThe irrigated rice fields are multifunctional in sustainable development but are vulnerable to climate change. The current study is intended to analyze traditional irrigated rice field farmers’ adaptive capacity and formulate the strategies needed to strengthen it. The study was conducted in subaks located in Badung Regency, Bali’s urban and rural areas.The sample of the subak areas and the sample of the farmers were determined by multistage sampling.The data were obtained through a semi-structured and questionnaire-guided interview and were analyzed using the sequential explanatory mixed methods. Index-indicated numeric and local specific variables and dimensions measured the adaptive capacity of the farmers. The results of the study show that the adaptive capacity of the farmers in the rural area is better (high level) than that of the farmers in the urban area. The critical node of the adaptive capacity of the farmers in the rural area is situated on the economic and human dimensions. In contrast, the critical node of the adaptive capacity of the farmers in the urban area is situated in the cultural, social, human, and physical-natural dimensions. Five strategies and several activities relevant to what to do to strengthen the adaptive capacity of the farmers in each area are formulated. The commitment and role of the government as the policymakers, and the support from the other stakeholders, is crucial to strengthen the farmers’ adaptive capacity.

This study aims to implement practical management accounting on nipah water by looking for inputs and outputs for other commodities. The main problem in this study is how accounting can raise environmental problems in the form of green accounting practices. The review is seen from management accounting to determine cross production between nipah water production as capital input for other commodities. Environmental accounting practices dealing with the problem of pricing agricultural products become increasingly complex in agricultural locations. The data source is primary data taken directly from 30 nipah farmers located on the banks of the Moncongloe Lappara River, Moncongloe District, Maros Regency, South Sulawesi during 2020. The analytical method used is to measure input-output commodities with the Social Accounting Matrix. The results of this study indicate that the total contribution index of sap water production from Nipah is 456.66 to other commodities, while commodity B is 1236.04 and commodity C is 851.66. With this contribution, farmers can manage their own sources of funds sourced from nipa palm water in a sustainable manner, not only farmers in the Lappara Moncongloe River area, but also the condition of nipa palm farmers in other parts of Indonesia

The orphanage is a place where neglected children receive their physical needs, such as a place to live, as well as their spiritual needs, which are met by foster parents or caregivers. Caregivers can act as parents by providing neglected children with attention, affection, and a sense of security at the Pekanbaru City Social Care Center (PSPA), which is fully supervised by the Riau Province Social Service. Parenting neglected children is based on empathic communication. The purpose of this research is to determine the affective and cognitive aspects of empathic communication that occurs between caregivers and neglected children in the formation of an attitude of independence at the UPT Panti Social Caregivers (PSPA) Pekanbaru City. This is a qualitative research method with a humanistic perspective. Data for this study were gathered through observation, interviews, and documentation. Data reduction, data presentation, and conclusions were used in technical data analysis. Participation extension and triangulation are used in the data validity technique. The finding showed that the affective aspect of empathic communication used by caregivers to foster the independence attititude of abandoned children was founded on deep feelings, sympathy and empathy, and a sense of care. Cognitive aspects of empathic communication between caregivers and abandoned children are formed through good relationships and the nature of similarity. Caregivers have a big responsibility in protecting abandoned children in social institutions so that they can be independent.

Social media occupies increasingly important roles in disaster response management. However, there are still doubts regarding the effectiveness of social media-based disaster communication. This paper aims to understand the discourse that emerge in Twitter containing facts or opinion regarding the flood disaster communication in DKI Jakarta under the different leadership of Basuki Tjahaja Purnama (2014- 2017) and Anies Rasyid Baswedan (2017-2020). This research used the methods of Corpus Assisted Discourse Study (CADS) and Actor Network Theory (ANT). These methods collected data on the discourse that is happening on Twitter and analyzes the network of actors who are active in discussions about floods in DKI Jakarta at different periods of leadership. This research found that the discourse that has developed on Twitter about floods in DKI Jakarta covers the personal problems of the leaders, the personality of the leaders, and the political position of the leaders, offered little messages that leads to solving the problem of flooding in DKI Jakarta. The active actor network is dominated by the local government of Jakarta as the responsibility holder or the main actor in delivering in information is only played by BPBD Jakarta.

Yogyakarta has excellent human resources, but informal sector workers are threatened with low income. This article aims to examine how the influence of worker and business characteristics on informal sector workers’ income in Yogyakarta. Characteristic of workers consist of education, work or business experience, status in the household, marital status, gender, and age. Then business characteristics consist of types of business fields, working days, hours of work, length of work, use of internet technology, and financial bookkeeping. The study was conducted quantitatively using secondary data sources obtained from the National Labor Force Survey (Sakernas) in August 2018. The data were analyzed using binary logistic regression. The findings show that both partially and simultaneously, each factor of the two characteristics has a significant influence on the income of informal sector workers in Yogyakarta.

Health investment has an important role in economic development and major capital in order to be able to do work. Lack of health insurance can be a poverty trap. This is the fundamental reason why health insurance receives the most attention in the SDG’s. Based on SAKERNAS data in 2017 the percentage of informal sector workers in Indonesia is still quite high, reaching 57.03 percent and of that number only 57.3 percent of informal sector workers have health insurance which means that almost half of the informal sector workers work without health insurance protection . This study aims to determine the factors that influence the ownership of health insurance for informal sector workers in Indonesia. The analysis was conducted inferentially using chi-square and binary logistic regression using SUSENAS data in March 2018. Based on the results of the analysis using chi-square obtained the results that all independent variables have a meaningful relationship with health insurance ownership so the analysis is continued with logistic regression. From the results of logistic regression it is found that the factors that influence the health insurance ownership of informal sector workers in Indonesia are the area of residence, marital status, status in the household, gender, age, NIK ownership, education level, business field, health complaints, status economics, and self-medication.

Background: The addition of adjuvants to local anesthetics in the peribulbar block might be a method to elevate block quality. This study aimed to determine the effectiveness of peribulbar local anesthesia adjuvants using dexmedetomidine 20 mcg and dexamethasone 5 mg. Patients and Methods: This study was a single-blind randomized clinical trial. Patients were divided into three groups who received peribulbar group: Group C (4 ml of levobupivacaine 0.5% and 4 ml of 2% lidocaine added with 1 ml of normal saline), Group D (4 ml of levobupivacaine 0.5% and 4 ml of 2% lidocaine added with 1 ml of dexamethasone 5 mg), and Group M (received 4 ml of levobupivacaine 0.5% and 4 ml of 2% lidocaine added with 1 ml of dexmedetomidine 20 mcg). Sensory and motor block onsets and durations were assessed. Results: There were 42 patients included. The sensory block onset was 70.7 s in Group M and the slowest in Group C (152.1 s). The longest sensory block duration was found in Group D (259.3 min) and the shortest in Group C (143.2 min). Conclusion: Dexamethasone 5 mg adjuvant affected sensory block onset, sensory, and motor block duration but did not affect motor block onset, while dexmedetomidine 20 mcg adjuvant affected sensory and motor block onsets as well as sensory and motor block durations.

The study set out to analyse the differences in the characteristics and spatial distribution of urban beggars in Sanglah, Denpasar City. It employed survey research for data collection by observation, structured interviews with a questionnaire, and documentation. These data were later processed and analysed in a quantitative descriptive manner. The results show that, based on religion, there were two groups of beggars, namely Muslim and Hindu. Spatially, the Hindu beggars disregarded the religion of the people they implored, whereas the Muslim beggars would only target the houses and shops owned by Muslims. Those two class of beggars differ in languages, bagging duration, daily-earning, clothing, and sympathy-drawing strategy during the panhandling.

Immunotherapy holds enormous promise to create a new outlook of cancer therapy by eliminating tumors via activation of the immune system. In immunotherapy, polymeric systems play a significant role in improving antitumor efficacy and safety profile. Polymeric systems possess many favorable properties, including magnificent biocompatibility and biodegradability, structural and component diversity, easy and controllable fabrication, and high loading capacity for immune-related substances. These properties allow polymeric systems to perform multiple functions in immunotherapy, such as immune stimulants, modifying and activating T cells, delivery system for immune cargos, or as an artificial antigen-presenting cell. Among diverse immunotherapies, immune checkpoint inhibitors, chimeric antigen receptor (CAR) T cell, and oncolytic virus recently have been dramatically investigated for their remarkable success in clinical trials. In this report, we review the monotherapy status of immune checkpoint inhibitors, CAR-T cell, and oncolytic virus, and their current combination strategies with diverse polymeric systems.

Magnetic resonance imaging (MRI) is increasingly used to diagnose focal and diffuse liver disorders. Despite their enhanced efficacy, liver-targeted gadolinium-based contrast agents (GBCAs) raise safety concerns owing to the release of toxic Gd3+ ions. A π-conjugated macrocyclic chelate, Mn-NOTA-NP, was designed and synthesized as a non-gadolinium alternative for liver-specific MRI. Mn-NOTA-NP exhibits an r1 relaxivity of 3.57 mM−1 s−1 in water and 9.01 mM−1 s−1 in saline containing human serum albumin at 3 T, which is significantly greater than the clinically utilized Mn2+-based hepatobiliary drug, Mn-DPDP (1.50 mM−1 s−1), and comparable with that of GBCAs. Furthermore, the in vivo biodistribution and MRI enhancement patterns of Mn-NOTA-NP were similar to those of the Gd3+-based hepatobiliary agent, Gd-DTPA-EOB. Additionally, a 0.05 mmol/kg dose of Mn-NOTA-NP facilitated high-sensitivity tumor detection with tumor signal enhancement in a liver tumor model. Ligand-docking simulations further indicated that Mn-NOTA-NP differed from other hepatobiliary agents in their interactions with several transporter systems. Collectively, we demonstrated that Mn-NOTA-NP could be a new liver-specific MRI contrast agent.

A patients Because of the past 3 decades’ extensive research, several disease modifying therapies became available, thus a paradigm change is multiple sclerosis care was necessary. In 2018 a therapeutic guideline was created recommending that treatment of persons with multiple sclerosis should take place in specified care units where the entire spectrum of disease modifying therapies is available, patient monitoring is ensured, and therapy side effects are detected and treated promptly. In 2019 multiple sclerosis care unit criteria were developed, emphasizing personnel and instrumental requirements to provide most professional care. However, no survey was conducted assessing the real-world adaptation of these criteria. Objective To assess whether Hungarian care units fulfil international criteria. Methods A self-report questionnaire was assembled based on international guidelines and sent to Hungarian care units focusing on 3 main aspects: personnel and instrumental background, disease-modifying therapy use, number of people living with multiple sclerosis receiving care in care units. Data on number of persons with multiple sclerosis were compared to Hungarian prevalence estimates. Descriptive statistics were used to analyse data. Results Out of 27 respondent care units, 3 fulfilled minimum requirements and 7 fulfilled minimum and recommended requirements. The least prevalent neighbouring specialties were spasticity and pain specialist, and neuro-ophthalmologist and oto-neurologist. Only 15 centres used all available disease modifying therapies. A total number of 7213 people with multiple sclerosis received care in 27 respondent centres. Compared to prevalence estimates, 2500 persons with multiple sclerosis did not receive multiple sclerosis specific care in Hungary. Conclusion Less than half of Hungarian care units provided sufficient care for people living with multiple sclerosis. Care units employing fewer neighbouring specialties, might have difficulties diagnosing and providing appropriate care for persons with multiple sclerosis, especially for people with progressive disease course, contributing to the reported low number of persons living with multiple sclerosis.

Although we live in a diasporic world, we need interpretive strategies for reading the Bible through the eyes of history and the experiences of marginalized groups, most of whom are still on the margins. Attempts to bring diverse voices without exclusion, discrimination, and disregard into biblical interpretation in the diasporic world have been linked to the study of epistemic injustice. I consider signifiers of those norms that make the power/knowledge of ‘others’ inferior through the contemporary hospitality perspectives. I consider how reading and interpreting the Bible, without dualistic language, plays a role in creatively participating in new situations, and not claiming absolute ownership of fixed spaces, discourses, and movements. In particular, by reading Lot’s story of biblical hospitality as a discourse on hostility and hospitality, this study will examine how an interpretive lens of contemporary hospitality can expand and apply to encourage epistemic justice in a more relational and plentiful way.

In this study, we designed, synthesized, and evaluated gadolinium compounds conjugated with flavonoids as potential theranostic agents for the treatment of inflammation. These novel theranostic agents combine a molecular imaging agent and one of three flavonoids (galangin, chrysin, and 7-hydroxyflavone) as anti-inflammatory drugs as a single integrated platform. Using these agents, MR imaging showed contrast enhancement (>10 in CNR) at inflamed sites in an animal inflammation model, and subsequent MR imaging used to monitor the therapeutic efficacy of these integrated agents revealed changes in inflamed regions. The anti-inflammatory effects of these agents were demonstrated both in vitro and in vivo. Furthermore, the antioxidant efficacy of the agents was evaluated by measuring their reactive oxygen species scavenging properties. For example, Gd-galangin at 30 μM showed a three-fold higher ROS scavenging of DPPH. Taken together, our findings provide convincing evidence to indicate that flavonoid-conjugated gadolinium compounds can be used as potentially efficient theranostic agents for the treatment of inflammation.

Current technologies based on inertial measurement units (IMUs) are considered valid and reliable tools for monitoring barbell velocity in strength training. However, the extracted outcomes are often limited to a few velocity metrics, such as mean or maximal velocity. This study aimed at validating a single IMU-based methodology to automatically obtain the barbell velocity full profile as well as key performance metrics during maximal Paralympic bench press. Seven Paralympic powerlifters (age: 30.5 ± 4.3 years, sitting height: 71.6 ± 6.8 cm, body mass: 72.5 ± 16.4 kg, one-repetition maximum: 148.4 ± 38.6 kg) performed four attempts of maximal Paralympic bench press. The barbell velocity profile and relevant metrics were automatically obtained from IMU linear acceleration through a custom-made algorithm and validated against a video-based reference system. The mean difference between devices was 0.00 ± 0.04 m·s−1 with low limits of agreement (−1) and moderate-to-good reliability (ICC: 0.55–0.90). Linear regression analysis showed large-to-very large associations between paired measurements (r: 0.57–0.91, p < 0.003; SEE: 0.02–0.06 m·s−1). The analysis of velocity curves showed a high spatial similarity and small differences between devices. The proposed methodology provided a good level of agreement, making it suitable for different applications in barbell velocity monitoring during maximal Paralympic bench press.

The purpose of this research is to develop ecotourism development and planning model, especially the relationship between ecotourism and sustainable community development through green entrepreneurship development strategy by highlighting local uniqueness as the main weapon to enhance the attractiveness of ecotourism destination in NTB Province. Results of primary and secondary data collection have obtained SWOT picture which is useful to make the draft of village tourism policy in lombok NTB. Green Village based tourism entreprenuership and local wisdom to enhance tourist attraction to Lombok NTB: Output, governance system of green entrepreneur development and Tourism Village in NTB province, consisting of (a) Policy of developing green entrepreneur and tourism village for ecotourism destination development (b) Action Plan green entrepreneur and green product (c) Planning Indicators and (d) Monitoring and evaluation system.

Regional disparity was a common problem in developing countries. Different regional potentials result in disparity between regions. This study aims to analyze regional disparity in Subosukwonosraten region during 2001-2016 and to determine factors that affect regional disparity in Subosukawonosraten. Analysis techniques used in this study were entropy theil index and panel data regression. The result showed that regional disparity in Subosukawonosraten tends to increase during 2001-2016. Showed by the results of the total theil index in 2001 was 8.8133 to 12.4879 in 2016. The results of panel data regression fixed effect with Weighted Least Squares were share indsutry sector and share agriculture sector have the negative and significant impact on regional disparity. Enrollment rate, morbidity, and population growth have the positive and significant impact on regional disparity.

There has been much discussion among academics on how pupils may be taught online while yet maintaining a high degree of learning efficiency, in part because of the worldwide COVID-19 pandemic in the previous two years. Students may have trouble focusing due to a lack of teacher–student interaction, yet online learning has some advantages that are unavailable in traditional classrooms. The architecture of online courses for students is integrated into a system called the Adaptive and Intelligent Education System (AIES). In AIESs, reinforcement learning is often used in conjunction with the development of teaching strategies, and this reinforcement-learning-based system is known as RLATES. As a prerequisite to conducting research in this field, this paper undertakes the consolidation and analysis of existing research, design approaches, and model categories for adaptive and intelligent educational systems, with the hope of serving as a reference for scholars in the same field to help them gain access to the relevant information quickly and easily.

Here, we describe the synthesis, characterization, and in vitro biological evaluation of a series of transition metal complexes containing benzothiazole aniline (BTA). We employed BTA, which is known for its selective anticancer activity, and a salen-type Schiff-based ligand to coordinate several transition metals to achieve selective and synergistic cytotoxicity. The compounds obtained were characterized by NMR spectroscopy, mass spectrometry, Fourier transform infrared spectroscopy, and elemental analysis. The compounds L, MnL, FeL, CoL, and ZnL showed promising in vitro cytotoxicity against cancer cells, and they had a lower IC50 than that of the clinically used cisplatin. In particular, MnL had synergistic cytotoxicity against liver, breast, and colon cancer cells. Moreover, MnL, CoL, and CuL promoted the production of reactive oxygen species in HepG2 tumor cell lines. The lead compound of this series, MnL, remained stable in physiological settings, and docking results showed that it interacted rationally with the minor groove of DNA. Therefore, MnL may serve as a viable alternative to platinum-based chemotherapy.

The use of cancer-derived exosomes has been studied in several cancer types, but the cancer-targeting efficacy of glioma-derived exosomes has not been investigated in depth for malignant glioblastoma (GBM) cells. In this study, exosomes were derived from U87MG human glioblastoma cells, and selumetinib, a new anticancer drug, was loaded into the exosomes. We observed the tropism of GBM-derived exosomes in vitro and in vivo. We found that the tropism of GBM-derived exosomes is in contrast to the behavior of non-exosome-enveloped drugs and non-GBM-specific exosomes in vitro and in vivo in an animal GBM model. We found that the tropism exhibited by GBM-derived exosomes can be utilized to shuttle selumetinib, with no specific targeting moiety, to GBM tumor sites. Therefore, our findings indicated that GBM-derived exosomes loaded with selumetinib had a specific antitumor effect on U87MG cells and were non-toxic to normal brain cells. These exosomes offer improved therapeutic prospects for glioblastoma therapy.

Penelitian ini bertujuan untuk mengetahui hasil kontribusi kinerja guru pendidikan jasmani olahraga dan kesehatan terhadap motor ablity dan hasil belajar pada murid SD Negeri di Kabupaten Pinrang. Penelitian ini adalah jenis penelitian korelasional. Populasinya adalah siswa putra SD Negeri di Kabupaten Pinrang dengan sampel berjumlah 100 orang siswa dengan teknik cluster random sampling. Teknik analisis data yang digunakan adalah korelasi ganda. Berdasarkan hasil tersebut dapat disimpulkan bahwa (1) Ada kontribusi kinerja guru pendidikan jasmani olahraga dan kesehatan terhadap motor ability pada murid SD Negeri di Kabupaten Pinrang sebesar 57,1%, Hasil ini menunjukkan bahwa kinerja guru pendidikan jasmani olahraga dan kesehatan memiliki kontribusi terhadap motor ability murid SD Negeri di Kabupaten Pinrang, dan (2) Ada kontribusi kinerja guru pendidikan jasmani olahraga dan kesehatan terhadap hasil belajar pada murid SD Negeri di Kabupaten Pinrang sebesar 71,5%. Hasil ini menunjukkan analisa bahwa kinerja guru pendidikan jasmani olahraga dan kesehatan memiliki kontribusi terhadap hasil belajar murid SD Negeri di Kabupaten Pinrang.

Adenoviruses (Ads) are attractive nonviral vectors and show great potential in cancer gene therapy. However, inherent properties of Ads, including immunogenicity, nonspecific toxicity, and coxsackie and adenovirus receptor (CAR)-dependent cell uptake, limit their clinical use. To surmount these issues, we developed a pH- and glutathione-responsive poly(ethylene glycol)-poly(ꞵ-aminoester)-polyethyleneimine (PPA) for conjugation with Ad. The pH sensitivity of the PPA copolymer was elegantly tuned by substitution with different amino acids (arginine, histidine, and tryptophan), piperazines (Pip1, Pip2, and Pip3), and guanidine residues in the backbone of the PPA conjugate. PPA copolymer was further functionalized with short-chain cross-linker succinimidyl 3-(2-pyridyldithio)propionate) (SPDP) to obtain PPA-SPDP for facile conjugation with Ad. The PPA-conjugated Ad (PPA-Ad) conjugate was obtained by reacting PPA-SPDP conjugate with thiolated Ad (Ad-SH). Ad-SH was prepared by reacting Ad with 2-iminothiolane. The size distribution and zeta potential results of PPA-Ad conjugate showed an increasing trend with an increase in copolymer dose. From in vitro test, it was found that the transduction efficiency of PPA-Ad conjugate in CAR-positive cells (A549 and H460 cells) was remarkably increased at the acidic pH condition (pH 6.2) when compared with PPA-Ad conjugate incubated under the physiological condition (pH 7.4). Interestingly, the increase in transduction efficiency was evidenced in CAR-negative cells (MDA-MB-231 and T24 cells). These results demonstrated that biocompatible and biodegradable PPA copolymers can efficiently cover the surface of Ad and can increase the transduction efficiency, and hence PPA copolymers can be a useful nanomaterial for viral vector delivery in cancer therapy.
