Background: Malignant Superior Vena Cava Syndrome (SVCS) corresponds to the clinical manifestations due to the restriction of venous return to the right atrium secondary to obstruction of the superior vena cava and/or its main tributaries for a tumor. Endovascular management has proven to be safe, effective and cause a fast symptomatic relief in patients with SVCS. There is limited evidence in factors associated with outcomes in malignant setting for this procedure. Materials and methods: An analytical retrospective study was conducted and included patients that underwent endovascular management for malignant SVCS at the National Cancer Institute of Colombia between May 2016 and May 2021. Clinical and technical variables were analyzed to found associations with outcomes in these patients. Results: 54 patients were analyzed. Successful procedure rate was 94.4 %. At 10 months, the OS of the entire cohort of patients was 25 %. Patients with breast or lung cancer (P = 0.031), unsuccessful procedure (P = 0.011), and also with short time of symptoms to the date of the endovascular procedure (P = 0.027) had worse OS. Multivariate analysis showed that lung cancer [HR = 2.55, 95%IC:(1.21–5.36)] and left internal jugular vein or left Innominate vein distal stent attachment [HR = 3.27, 95%IC:(1.31–8.15)] were independent factors for worst OS. Conclusions: Based in the high success rate of the endovascular management and the better outcome in patients with early and successful procedure, this procedure should be considered as part of the multimodal treatment in patients with SVCS independent of the clinical scenario and the oncological diagnosis.

Shallow geothermal energy systems utilize the upper layers of the ground to provide space heating and cooling, with a relatively high efficiency. The research presented aims to assess the potential harnessing of this source and applying it for indoor conditioning of a precision agriculture facility. An experimental phase was conducted to measure the temperature of the ground at 0.75 m, 1.5 m, 2.25 m, and 3.0 m. Field testing was performed employing two probes; one uncovered and a second insulated with a polyethylene sheet, to evaluate the effect soil water content variations exert over the soil temperature. It was found that, given the high permeability and good drainage of the soil material, water content is not statistically significant, ruling out the presence of microconvective phenomena within the interstices of the undisturbed earth. Based on the temperature profile obtained, achievable heat extraction capacity was computed by modelling a buried pipe system. A parametrization was conducted showing that, of all the involved factors, pipe depth, pitch and their interaction, are the ones that contribute the most to the heat output variability. The computed available heat flow was used to simulate the performance of a geothermal heat pumping system for greenhouse heating based on real operation data, provided by the owners. The results show that, shallower layers of the ground, can provide adequate solutions for immediate thermal needs; a 5-pipe, 100m-length ground loop at 0.75 m, could provide 90% of the total thermal demand, whilst an 11-pipe, 200m-length loop, at 1.5 m depth, reaches 84% of the same demand, with a variability reduction in availability of 10% to 20% depending on the severity of surface conditions. However, for deeper layers, i.e., 2.25 m and 3.0 m, respective maximum heat output amounts to 43% and 49% of the total thermal demand. Nonetheless, these configurations yield the most stable and constant heat flow of the lot, making them particularly suitable for preheating/subcooling processes or even long-term thermal storage. Despite not fulfilling the total thermal demand of the greenhouse, these setups are more than capable of providing support to on-site conditioning devices; requiring a lower upkeep and yielding a higher efficiency, which would reduce overall production costs. Furthermore, from the conducted data analysis, it was found that controlling the agronomic values, associated to indoor agriculture, facilitates the physical aspect of greenhouse energy management, paving the way to more optimized thermal solutions, tailored to different sets of operation conditions.

Se evaluó la eficacia y la residualidad de diferentes insecticidas sistémicos aplicados con la semilla de algodón o mezclado con la misma para el control de los trips Caliothrips phaseoli Hood y Frankliniella schultzei Trybon y del pulgón Aphis gossypii Glover. Los ensayos se realizaron durante los años agrícolas 1999-00 y 2000-01 en el Campo Experimental La María de la EEA del INTA Santiago del Estero. Los insecticidas y dosis evaluadas fueron aldicarb G 10% a 150 g ia ha-1 y 300 g ia ha-1, Tiodicarb 35% LF a 700 g ia 100 k-1 de semilla, Imidacloprid 60% FS a 282 g ia 100 k-1 de semilla, Thiametoxan 35% FS a 210 g ia 100 k-1 de semilla comparados con el testigo de semillas sin tratar. La dosis de Aldicarb de 150 g ia ha-1 se utilizó solamente en el año 2000-01. Los recuentos de trips y pulgones por planta se realizaron a los 15, 20, 30 y 35 días posteriores a la siembra. En ambos años, para el control de trips y pulgones, todos los tratamientos superaron significativamente al testigo, no registrándose diferencias estadísticas entre los distintos insecticidas hasta los 20 días posteriores a la siembra, a excepción del Thiodicarb en el control de pulgones. Las diferencias entre tratamientos se observaron a partir de los 30 días posteriores a la siembra. El Aldicarb fue el producto de mayor eficacia y residualidad para el control de trips y pulgones, seguido de Thiametoxan e Imidacloprid, estos últimos insecticidas presentaron un mejor control para pulgones. Thiodicarb mostró una residualidad de 20 días en el control de los trips.

Poblaciones de dorado (Pisces) fueron muestreadas en Sinaloa, Nayarit y Baja Califonia Sur, México, en 1997. Unos 3211 organismos fueron analizados. Las tallas máximas fueron de 192 cm y 30 Kg. Hay diferentes tallas asociadas a las zonas y épocas climática (P < 0.05). Las relaciones del peso y la longitud son diferentes (P < 0.05), para totales y sexos. El factor de condición fue alto en Mazatlán y hay gran variación en primavera. La proporción sexual fue constante en Mazatlán, y hubo mayoría de machos en Nayarit, en otoño y primavera y en primavera en Los cabos. Pueden existir diferencias entre poblaciones costeras del Pacífico y Baja CaliforniaDolphin fish population data were recorded from landings in Sinaloa, Nayarit and Baja Califonia Sur, Mexico, in 1997. Of 3 211 organisms only 2 812 produced complete biometric data. The maximal length and weight were 192 cm and 30 Kg. The three sampling sites and the climatic periods also were associated with different size (P < 0.05). The covariate analysis of weight and length relationships indicated significative differences (P < 0.05) for total data and by sex. Condition factor was higher throughout the study period in Mazatlán, and the variation was higher, in the spring. The sexual ratio was in Mazatlán was constant, while there were more males in Nayarit in the fall and the spring and in Los Cabos in the spring. There may be population differences, between the central mexican Pacific Coast and the Peninsula of Baja data

Globally, the tilapia is the second most important group of commercial fish. Polyculture and growth were studied for 181 Nile tilapia, Oreochromis niloticus, in a seawater pond (30-37‰) at a density of 50 fish/m3 for six months (October to April) with 30 shrimp Litopenaeus vannamei at a density of 8 shrimp/m3 in two months (December-February) in an open tank of 3,6 m3. They were fed Camaronina”pellets” at libitum in the morning. The pond bottom was siphoned every other day, was maintained with constant aeration. Water exchange was 80% every two weeks. The temperature varied from 18 to 32oC. The tilapia grew from 72mm to 220mm, and 7g to 232g, survival was 92%. The total biomass was 38,6 kg. The shrimp grew from 6,8 g to 11,5 g and 102mm to 121mm. The survival rate was 100%. Total biomass was 219g. The feed conversion ratio (FCR) was 1,3:1,0 in both species. There was no problem between species; this polyculture is recommended. Acute promyelocytic leukemia (AML M3) is a well-defined subtype of leukemia with specific and peculiar characteristics. Immediate identification of t(15;17) or the PML/RARA gene rearrangement is fundamental for treatment. The objective of the present study was to compare fluorescent in situ hybridization (FISH), reverse transcriptase-polymerase chain reaction (RT-PCR) and karyotyping in 18 samples (12 at diagnosis and 6 after treatment) from 13 AML M3 patients. Bone marrow samples were submitted to karyotype G-banding, FISH and RT-PCR. At diagnosis, cytogenetics was successful in 10 of 12 samples, 8 with t(15;17) and 2 without. FISH was positive in 11/12 cases (one had no cells for analysis) and positivity varied from 25 to 93% (mean: 56%). RT-PCR was done in 6/12 cases and all were positive. Four of 8 patients with t(15;17) presented positive RT-PCR as well as 2 without metaphases. The lack of RT-PCR results in the other samples was due to poor quality RNA. When the three tests were compared at diagnosis, karyotyping presented the translocation in 80% of the tested samples while FISH and RT-PCR showed the PML/RARA rearrangement in 100% of them. Of 6 samples evaluated after treatment, 3 showed a normal karyotype, 1 persistence of an abnormal clone and 2 no metaphases. FISH was negative in 4 samples studied and 2 had no material for analysis. RT-PCR was positive in 4 (2 of which showed negative FISH, indicating residual disease) and negative in 2. When the three tests were compared after treatment, they showed concordance in 2 of 6 samples or, when there were not enough cells for all tests, concordance between karyotype and RT-PCR in one. At remission, RT-PCR was the most sensitive test in detecting residual disease, as expected (positive in 4/6 samples). An incidence of about 40% of 5' breaks and 60% of 3' breaks, i.e., bcr3 and bcr1/bcr2, respectively, was observed.

The overconsumption of conventional plastics has led to several environmental and social-economic issues related to plastic pollution, carbon emissions and resource depletion. Acknowledging these issues, the introduction of alternatives such as bioplastics has been promoted by national, supranational, and international organizations. However, the market for such materials is still niche, where businesses are uncertain about the benefits and costs associated with the use of these innovative materials. Successful (and sustainable) uptake of such alternatives will depend on public acceptance and changes in consumer behavior. Therefore, this study aims to explore how consumers' orientation towards environmental sustainability is related to consumer utilization of alternatives such as bio-based and biodegradable plastics. Consumer knowledge and performance expectations of these materials are also analyzed. This study employed a predominately quantitative research approach, where a self-administered online survey was used to collect the opinions of consumers across society using a snowball sampling technique. Results show continued consumer confusion, unrealistic expectations, and a value-action gap. These elements may have consequences for market uptake and broader implications across the value chain. A key implication is that both policy makers and businesses should address these barriers through enhanced communication of relevant information alongside improved consumer awareness and education.

Virgin and recycled poly(lactic acid) (PLA) based nanocomposite materials were obtained and subjected to microstructural, thermal and mechanical analysis in view of fabricating efficient microwave absorbers. PLA was first exposed to artificial accelerated aging, next was mechanically recycled through grinding followed by reprocessing using melt extrusion and compression molding, resulting in recycled PLA samples (rPLA). Addition of organically modified montmorillonite (OMMT) as nanoclay was performed in a second melt extrusion process in order to obtain virgin and recycled PLA-OMMT nanocomposites. The impact of recycling process and presence of OMMT nanoclay in the host PLA matrix has been studied by FTIR, TGA and DSC, while the mechanical performance has been investigated by micro-hardness test. The dielectric properties were measured in the 26–40 GHz frequency range using a Vector Network Analyzer to assess the performance of virgin and recycled PLA and OMMT-PLA material as microwave absorbers. The FTIR results show that the recycling process generated more C = O groups in the polymer. These polar groups tend to orient themselves in the direction of the applied field and increase the dielectric constant (ε'). Measured electromagnetic absorption index revealed that rPLA-4OMMT with a thickness of 400 µm is able to absorb 20.3% on average of the spectrum with a peak of 36%, while 200 µm-thick films of rPLA-4 wt.% OMMT has a mean absorption index of 14.5%. The overall results show that mechanically recycled polymer can replace virgin polymer in this kind of applications.

This work aims to study the effect of immersion in a ethanolic food simulant in mechanically recycled poly(lactic acid) (PLAR) and its nanocomposites reinforced with halloysite nanotubes (HNT). PLAR was obtained by subjecting PLA to an accelerated ageing process, which includes photochemical, thermal and hydrothermal ageing steps, followed by a final demanding washing step. PLAR was further reinforced with 4 %wt. HNT to improve the properties of the PLAR films. The materials were melt compounded by melt extrusion and processed into films by compression molding. The resulting films were exposed to food simulant D1 (50 %vol. ethanol solution) for 10 days at 40 °C. The intrinsic viscosity, crystallization behavior, thermal stability as well as the mechanical performance were analyzed before and after the contact with the food simulant. The swelling, plasticizing and hydrolyzing effect of the food simulant led to an important decrease of the intrinsic viscosity of all the samples, along with a significant increase of the crystallinity. Thermal stability was negatively affected by the decrease of the molecular weight, while the high crystallinity values resulted in materials with higher Vickers hardness values after the immersion in the food simulant.

Background and aim: Herbal medicines are used to treat a broad number of maladies. However, the pharmacological profile of most remedies is poorly understood. We investigated the effect of herbal remedies from kampo, traditional Chinese medicine (TCM) and other phytotherapies on human two-pore domain potassium channels (KCNK channels; TREK-1, TASK-1 and TASK-3) as well as the human TRPV1 channel. KCNK channels are responsible for the background potassium current of excitable cells, thus essential for the maintenance of the resting membrane potential. Hence, modulators of KCNK channels are of medical significance, e.g. for the treatment of sleep disorders and pain. The transient receptor potential channel TRPV1 is a pain detector for noxious heat. Agonists of this receptor are still used for the treatment of pain in ectopic applications. Experimental procedure: We evaluated the effect of 158 herbal remedies on these channels in a heterologous expression system (Xenopus laevis oocytes) using the two-electrode voltage-clamp technique with the aim of increasing the comprehension of their pharmacological profile. Results and conclusion: Some remedies with modulating effects were identified such as Angelica pubescens (radix), which inhibit TASK-1 and TASK-3 channels. Furthermore, the modulatory effects of the most effective remedies on the two TASK family members TASK-1 and TASK-3 correlate positively, reflecting their close relation. For the TRPV1 channel Terminalia chebula and Alchemilla xanthochlora were identified as potentiators. This study identifies a variety of herbal remedies as modulators of human K2P and TRPV1 channels and gives new insights into the pharmacological profile of these herbal remedies.

Black peppercorns (Piper nigrum L.) elicit a pungent and tingling oral impression. Their pungency is partially explained by the agonist activity of some of their active principles, especially piperine, on TRP channels. However, we recently showed that piperine, as well as other pungent compounds, also possess a marked effect on two-pore domain (KCNK, K2P) K+ channels. Members of this family play a key role in maintaining the resting membrane potential of excitable cells. Interestingly, tingling compounds have been shown to induce neuronal excitation by inhibiting KCNK channels. We addressed the question of whether it was plausible that KCNK channels could constitute a physiologically relevant target for the sensory active compounds present in black peppercorns. Because previous studies have demonstrated that mouse trigeminal neurons respond to several pungent compounds, to which humans are also sensitive, we used a primary culture of mouse trigeminal neurons to investigate whether the effect of piperine on these cell types could also be mediated by KCNK channels. We observed that even in the presence of classical TRP-antagonists, piperine was still able to activate a fraction of trigeminal neurons. Furthermore, our results showed that piperine is capable of inducing neuronal depolarization by a mechanism that does not require extracellular Na+ or Ca2+. This depolarization was mediated by the inhibition of a background K+ conductance, most likely corresponding to the KCNK channels of the TASK subfamily. We then performed a screening with 12 other pungent and/or tingling chemosensates isolated from black peppercorns. These compounds were evaluated on Xenopus laevis oocytes expressing the human orthologues of KCNK3, KNCK9 and KCNK18, which we previously showed to be inhibited by piperine. Remarkably, almost all of the isolated chemosensates inhibited the basal activity of hKCNK3, with 1-(octadeca-2E,4E,13/12Z-trienoyl)pyrrolidine acting as one of the most potent natural blockers for hKCNK3 found to date. Our results suggest that KCNK channels, especially KCNK3, are likely to play a complementary role to TRP channels in the complex orosensory impression elicited by black peppercorns, while they also help to expand the pharmacological knowledge of KCNK channels.

The enterotoxigenic Escherichia coli strains lead to diarrhoea in humans due to heat-labile and heat-stable (STa) enterotoxins. STa increases Cl-release in intestinal cells, including the human colonic carcinoma T84 cell line, involving increased cGMP and membrane alkalization due to reduced Na+/H+ exchangers (NHEs) activity. Since NHEs modulate intracellular pH (pHi), and NHE1, NHE2, and NHE4 are expressed in T84 cells, we characterized the STa role as modulator of these exchangers. pHi was assayed by the NH4Cl pulse technique and measured by fluorescence microscopy in BCECF-preloaded cells. pHi recovery rate (dpHi/dt) was determined in the absence or presence of 0.25 μmol/L STa (30 minutes), 25 μmol/L HOE-694 (concentration inhibiting NHE1 and NHE2), 500 μmol/L sodium nitroprusside (SNP, spontaneous nitric oxide donor), 100 μmol/L dibutyryl cyclic GMP (db-cGMP), 100 nmol/L H89 (protein kinase A inhibitor), or 10 μmol/L forskolin (adenylyl cyclase activator). cGMP and cAMP were measured in cell extracts by radioimmunoassay, and buffering capacity (ßi) and H+ efflux (JH+) was determined. NHE4 protein abundance was determined by western blotting. STa and HOE-694 caused comparable reduction in dpHi/dt and JH+ (~63%), without altering basal pHi (range 7.144-7.172). STa did not alter ßi value in a range of 1.6 pHi units. The dpHi/dt and JH+ was almost abolished (~94% inhibition) by STa + HOE-694. STa effect was unaltered by db-cGMP or SNP. However, STa and forskolin increased cAMP level. STa-decreased dpHi/dt and JH+ was mimicked by forskolin, and STa + HOE-694 effect was abolished by H89. Thus, incubation of T84 cells with STa results in reduced NHE4 activity leading to a lower capacity of pHi recovery requiring cAMP, but not cGMP. STa effect results in a causal phenomenon (STa/increased cAMP/increased PKA activity/reduced NHE4 activity) ending with intracellular acidification that could have consequences in the gastrointestinal cells function promoting human diarrhoea.

Arsenic main inorganic compound is arsenic trioxide (ATO) presented in solution mainly as arsenite. ATO increases intracellular pH (pHi), cell proliferation and tumor growth. Sodium-proton exchangers (NHEs) modulate the pHi, with NHE1 playing significant roles. Whether ATO-increased cell proliferation results from altered NHEs expression and activity is unknown. We hypothesize that ATO increases cell proliferation by altering pHi due to increased NHEs-like transport activity. Madin-Darby canine kidney (MDCK) cells grown in 5 mmol/L D-glucose-containing DMEM were exposed to ATO (0.05, 0.5 or 5 µmol/L, 0-48 hours) in the absence or presence of 5-N,N-hexamethylene amiloride (HMA, 5-100 µmol/L, NHEs inhibitor), PD-98059 (30 µmol/L, MAPK1/2 inhibitor), Gö6976 (10 µmol/L, PKCα, βI and μ inhibitor), or Schering 28080 (10 µmol/L, H(+)/K(+)ATPase inhibitor) plus concanamycin (0.1 µmol/L, V type ATPases inhibitor). Incorporation of [(3)H]thymidine was used to estimate cell proliferation, and counting cells with a hemocytometer to determine the cell number. The pHi was measured by fluorometry in 2,7-bicarboxyethyl-5,6-carboxyfluorescein loaded cells. The Na(+)-dependent HMA-sensitive NHEs-like mediated proton transport kinetics, NHE1 protein abundance in the total, cytoplasm and plasma membrane protein fractions, and phosphorylated and total p42/44 mitogen-activated protein kinases (p42/44(mapk)) were also determined. Lowest ATO (0.05 µmol/L, ~0.01 ppm) used in this study increased cell proliferation, pHi, NHEs-like transport and plasma membrane NHE1 protein abundance, effects blocked by HMA, PD-98059 or Gö6976. Cell-buffering capacity did not change by ATO. The results show that a low ATO concentration increases MDCK cells proliferation by NHEs (probably NHE1)-like transport dependent-increased pHi requiring p42/44(mapk) and PKCα, βI and/or μ activity. This finding could be crucial in diseases where uncontrolled cell growth occurs, such as tumor growth, and in circumstances where ATO, likely arsenite, is available at the drinking-water at these levels.

PURPOSE: Transient receptor potential melastatin-8 (TRPM8) is an ion channel expressed extensively in sensory nerves, human prostate and overexpressed in a variety of cancers including prostate, breast, lung, colon and skin melanomas. It is activated by innoxious cooling and chemical stimuli. TRPM8 activation by cooling or chemical agonists is reported to induce profound analgesia in neuropathic pain conditions. Known TRPM8 agonists like menthol and icilin cross-activate other thermo-TRP channels like TRPV3 and TRPA1 and mutually inhibit TRPM8. This limits the usefulness of menthol and icilin as TRPM8 ligands. Consequently, the identification of selective and potent ligands for TRPM8 is of high relevance both in basic research and for therapeutic applications. In the present investigation, a group of menthol derivates was characterized. These ligands are selective and potent agonists of TRPM8. Interestingly they do not activate other thermo-TRPs like TRPA1, TRPV1, TRPV2, TRPV3 and TRPV4. These ion channels are also nociceptors and target of many inflammatory mediators. METHODS: Investigations were performed in a recombinant system: Xenopus oocytes microinjected with cRNA of gene of interest were superfused with the test substances after initial responses of known standard agonists. Evoked currents were measured by two-electrode voltage clamp technique. RESULTS: The newly characterized ligands possess an up to six-fold higher potency (EC50 in low µM) and an up to two-fold increase in efficacy compared to the parent compound menthol. In addition, it is found that chemical derivatives of menthol like CPS-368, CPS-369, CPS-125, WS-5 and WS-12 are the most selective ligands for TRPM8. The enhanced activity and selectivity seems to be conferred by hexacyclic ring structure present in all ligands as substances like WS-23 which lack this functional group activate TRPM8 with much lower potency (EC50 in mM) and those with pentacyclcic ring structure (furanone compounds) are totally inactive. CONCLUSION: The new substances activate TRPM8 with a higher potency, efficacy and specificity than menthol and will thus be of importance for the development of pharmacological agents suitable for treatment and diagnosis of certain cancers and as analgesics. STATEMENT OF NOVELTY: The new compounds have an unmatched specificity for TRPM8 ion channels with additional display of high potency and efficacy. Thus these substances are better pharmacological tools for TRPM8 characterization then known compounds and it is suggested that these menthol-derivates may serve as model substances for the development of TRPM8 ligands.
