Within the research along the European Fusion Roadmap, water-cooled divertor PFCs are foreseen in the design of a first fusion demonstration power plant (DEMO) in order to provide reliable heat removal capability. In the frame of this concerted attempt, the Max Planck Institute for Plasma Physics is concentrating on the development and testing of composite materials based on tungsten (W, preferred armour material) and copper (Cu, preferred heat sink material). W fibres (W _f ) as monofilaments and yarns as reinforcement play a central role in these investigations due to their extraordinary properties concerning ductility already at room temperature and high tensile strength. Recent investigations on the impact of radiation damage suggest that the fibres retain their ductility upon irradiation. W reinforced with W fibres (W _f /W) allows to overcome the intrinsic brittleness of W. Quantitative mechanical fracture tests of W _f /W confirm the basic mechanisms of fibre reinforcement and the increased resistance to mechanical fatigue. The good wettability of W with liquid Cu and the absence of any metallurgical solubility make up an ideal material pairing for composite production. W fibre-reinforced Cu (W _f /Cu) cooling tubes provide a rather high thermal conductivity (> 250 W mK ^−1 ) and at least twice the strength of CuCrZr in hoop direction in the temperature range up to at least 500 °C. Very recent neutron irradiation experiments confirm the sustainment of ductility of the W _f /Cu composite. Numerical simulations suggest that thermal stresses in W-Cu PFCs could be strongly reduced by tailoring the local W and Cu volume fraction. This ‘freely’ distributed material composition can be achieved by means of additively manufactured W skeletons consecutively infiltrated by Cu. Investigations with W preforms produced by Laser Beam Powder Bed Fusion and infiltrated by Cu demonstrate the feasibility of this approach while testing of specifically prepared specimen is ongoing.

Currently, tungsten fibre-reinforced (Wf) composites are regarded as promising materials for plasma-facing components of future magnetic confinement fusion devices. In this context, tungsten fibre-reinforced tungsten (Wf/W) is being investigated as a pseudo-ductile composite material overcoming the intrinsic brittleness of bulk tungsten while tungsten fibre-reinforced copper (Wf/Cu) is being developed as a high-strength composite heat sink material. In this contribution, we discuss the current development status and the progress that has been achieved recently with respect to characterization and upscaling of the aforementioned materials.In cooperation with industry, upscaling of multifilamentary W yarn fabrication was demonstrated. Multilayered W fibre braids were made from such yarns and used for the manufacturing of 400mm long medium-scale tungsten fibre-reinforced copper heat sink tubes. The maturity of short tungsten fibre-reinforced tungsten composites produced by powder metallurgy allowed the fabrication of flat tile mock-ups. Test procedure and first results of high heat flux tests are shown. Finally, we discuss the challenges and the benefits of these composites for the use in high heat flux components.

Highly loaded divertor regions in fusion experiments are exposed to high particle and power fluxes. Therefore, tungsten with its excellent thermal and physical properties is a preferred plasma facing material for this application. For divertor areas with moderate steady-state heat flux up to 10 MW/m2 the excellent high-temperature resistance of W is not absolutely necessary. 95–97 % W heavy alloys containing Ni, Cu or Fe could be used from the aspects of plasma wall interaction and their thermomechanical properties. In this study we present results of thermo-mechanical characterisations like measurement of thermal conductivity and tensile tests at elevated temperatures as well as results of high heat flux performance tests at adiabatic and steady-state loading. Finally, we discuss results of deuterium retention measurements.

Experiments on ASDEX Upgrade (AUG) in 2021 and 2022 have addressed a number of critical issues for ITER and EU DEMO. A major objective of the AUG programme is to shed light on the underlying physics of confinement, stability, and plasma exhaust in order to allow reliable extrapolation of results obtained on present day machines to these reactor-grade devices. Concerning pedestal physics, the mitigation of edge localised modes (ELMs) using resonant magnetic perturbations (RMPs) was found to be consistent with a reduction of the linear peeling-ballooning stability threshold due to the helical deformation of the plasma. Conversely, ELM suppression by RMPs is ascribed to an increased pedestal transport that keeps the plasma away from this boundary. Candidates for this increased transport are locally enhanced turbulence and a locked magnetic island in the pedestal. The enhanced D-alpha (EDA) and quasi-continuous exhaust (QCE) regimes have been established as promising ELM-free scenarios. Here, the pressure gradient at the foot of the H-mode pedestal is reduced by a quasi-coherent mode, consistent with violation of the high-n ballooning mode stability limit there. This is suggestive that the EDA and QCE regimes have a common underlying physics origin. In the area of transport physics, full radius models for both L- and H-modes have been developed. These models predict energy confinement in AUG better than the commonly used global scaling laws, representing a large step towards the goal of predictive capability. A new momentum transport analysis framework has been developed that provides access to the intrinsic torque in the plasma core. In the field of exhaust, the X-Point Radiator (XPR), a cold and dense plasma region on closed flux surfaces close to the X-point, was described by an analytical model that provides an understanding of its formation as well as its stability, i.e., the conditions under which it transitions into a deleterious MARFE with the potential to result in a disruptive termination. With the XPR close to the divertor target, a new detached divertor concept, the compact radiative divertor, was developed. Here, the exhaust power is radiated before reaching the target, allowing close proximity of the X-point to the target. No limitations by the shallow field line angle due to the large flux expansion were observed, and sufficient compression of neutral density was demonstrated. With respect to the pumping of non-recycling impurities, the divertor enrichment was found to mainly depend on the ionisation energy of the impurity under consideration. In the area of MHD physics, analysis of the hot plasma core motion in sawtooth crashes showed good agreement with nonlinear 2-fluid simulations. This indicates that the fast reconnection observed in these events is adequately described including the pressure gradient and the electron inertia in the parallel Ohm’s law. Concerning disruption physics, a shattered pellet injection system was installed in collaboration with the ITER International Organisation. Thanks to the ability to vary the shard size distribution independently of the injection velocity, as well as its impurity admixture, it was possible to tailor the current quench rate, which is an important requirement for future large devices such as ITER. Progress was also made modelling the force reduction of VDEs induced by massive gas injection on AUG. The H-mode density limit was characterised in terms of safe operational space with a newly developed active feedback control method that allowed the stability boundary to be probed several times within a single discharge without inducing a disruptive termination. Regarding integrated operation scenarios, the role of density peaking in the confinement of the ITER baseline scenario (high plasma current) was clarified. The usual energy confinement scaling ITER98( p,y ) does not capture this effect, but the more recent H20 scaling does, highlighting again the importance of developing adequate physics based models. Advanced tokamak scenarios, aiming at large non-inductive current fraction due to non-standard profiles of the safety factor in combination with high normalised plasma pressure were studied with a focus on their access conditions. A method to guide the approach of the targeted safety factor profiles was developed, and the conditions for achieving good confinement were clarified. Based on this, two types of advanced scenarios (‘hybrid’ and ‘elevated’ q -profile) were established on AUG and characterised concerning their plasma performance.

After a long device enhancement phase, scientific operation resumed in 2022. The main new device components are the water cooling of all plasma facing components and the new water-cooled high heat flux divertor units. Water cooling allowed for the first long-pulse operation campaign. A maximum discharge length of 8 min was achieved with a total heating energy of 1.3 GJ. Safe divertor operation was demonstrated in attached and detached mode. Stable detachment is readily achieved in some magnetic configurations but requires impurity seeding in configurations with small magnetic pitch angle within the edge islands. Progress was made in the characterization of transport mechanisms across edge magnetic islands: Measurement of the potential distribution and flow pattern reveals that the islands are associated with a strong poloidal drift, which leads to rapid convection of energy and particles from the last closed flux surface into the scrape-off layer. Using the upgraded plasma heating systems, advanced heating scenarios were developed, which provide improved energy confinement comparable to the scenario, in which the record triple product for stellarators was achieved in the previous operation campaign. However, a magnetic configuration-dependent critical heating power limit of the electron cyclotron resonance heating was observed. Exceeding the respective power limit leads to a degradation of the confinement.

Superconducting (SC) tokamak JT-60SA plays an essential role in fusion research and development by supporting and complementing the ITER project, providing directions to the DEMO design activity and fostering next generation scientists and engineers. Since the short circuit incident at the terminal joints of equilibrium field coil #1 during the integrated commissioning (IC) in March 2021, both EU and JA implementing agencies (IAs) have examined how to ensure safe operation of JT-60SA by mitigating the risk of possible discharge occurrence inside the cryostat. Based on the experience of the global Paschen tests, the IAs have established a strategy of risk mitigation measures, which is a combination of (i) reinforcement of insulation, (ii) avoiding unnecessary voltage application to the coil systems and (iii) immediate de-energization of the coils when deteriorated vacuum conditions are detected. Thanks to the considerable efforts of the Integrated Project Team members, the IC restarted in May 2023. After confirmation of the SC state of the coil systems (TF, EF and CS), the coil energization test and the plasma operation phase 1 (OP-1) started. The first plasma was successfully achieved on 23 October 2023 with a limited value of voltage and current applied to the coils. The plasma configuration control was also confirmed with low plasma current and low auxiliary heating power conditions. Based on the IO–F4E–QST collaboration, activities of JT-60SA have been shared with the IO and provided an important lesson for ITER assembly and commissioning, and will provide an outstanding contribution to fusion research at large. After OP-1, maintenance & enhancement phase 1 (M/E-1) starts from January 2024, in which in-vessel components are installed, and heating and diagnostic systems are extensively upgraded to allow a high power heating experiment planned in OP-2. In order to make the best use of JT-60SA, a newly organized JT-60SA experiment team will refine the research plan for the future high heating power operation phase.

In 2021 JET exploited its unique capabilities to operate with T and D–T fuel with an ITER-like Be/W wall (JET-ILW). This second major JET D–T campaign (DTE2), after DTE1 in 1997, represented the culmination of a series of JET enhancements—new fusion diagnostics, new T injection capabilities, refurbishment of the T plant, increased auxiliary heating, in-vessel calibration of 14 MeV neutron yield monitors—as well as significant advances in plasma theory and modelling in the fusion community. DTE2 was complemented by a sequence of isotope physics campaigns encompassing operation in pure tritium at high T-NBI power. Carefully conducted for safe operation with tritium, the new T and D–T experiments used 1 kg of T (vs 100 g in DTE1), yielding the most fusion reactor relevant D–T plasmas to date and expanding our understanding of isotopes and D–T mixture physics. Furthermore, since the JET T and DTE2 campaigns occurred almost 25 years after the last major D–T tokamak experiment, it was also a strategic goal of the European fusion programme to refresh operational experience of a nuclear tokamak to prepare staff for ITER operation. The key physics results of the JET T and DTE2 experiments, carried out within the EUROfusion JET1 work package, are reported in this paper. Progress in the technological exploitation of JET D–T operations, development and validation of nuclear codes, neutronic tools and techniques for ITER operations carried out by EUROfusion (started within the Horizon 2020 Framework Programme and continuing under the Horizon Europe FP) are reported in (Litaudon et al Nucl. Fusion accepted), while JET experience on T and D–T operations is presented in (King et al Nucl. Fusion submitted).

An overview is presented of the progress since 2021 in the construction and scientific programme preparation of the Divertor Tokamak Test (DTT) facility. Licensing for building construction has been granted at the end of 2021. Licensing for Cat. A radiologic source has been also granted in 2022. The construction of the toroidal field magnet system is progressing. The prototype of the 170 GHz gyrotron has been produced and it is now under test on the FALCON facility. The design of the vacuum vessel, the poloidal field coils and the civil infrastructures has been completed. The shape of the first DTT divertor has been agreed with EUROfusion to test different plasma and exhaust scenarios: single null, double null, X-divertor and negative triangularity plasmas. A detailed research plan is being elaborated with the involvement of the EUROfusion laboratories.

This paper is dealing with the physics basis used for the design of the Divertor Tokamak Test facility (DTT), under construction in Frascati (DTT 2019 DTT interim design report (2019)) Italy, and with the description of the main target plasma scenarios of the device. The main goal of the facility will be the study of the power exhaust, intended as a fully integrated core-edge problem, and eventually to propose an optimized divertor for the European DEMO plant. The approach used to design the facility is described and their main features are reported, by using simulations performed by state-of-the-art codes both for the bulk and edge studies. A detailed analysis of MHD, including also the possibility to study disruption events and Energetic Particles physics is also reported. Eventually, a description of the ongoing work to build-up a Research Plan written and shared by the full EUROfusion community is presented.

The Micrarchaeota lineage includes poorly characterized archaea with reduced genomes that likely depend on host interactions for survival. Here, the authors report a stable co-culture of a member of the Micrarchaeota and its host, and use multi-omic and physiological analyses to shed light on this symbiosis.

Abstract Sessile microorganisms were described as early as the seventeenth century. However, the term biofilm arose only in the 1960s in wastewater treatment research and was adopted later in marine fouling and in medical and dental microbiology. The sessile mode of microbial life was gradually recognized to be predominant on Earth, and the term biofilm became established for the growth of microorganisms in aggregates, frequently associated with interfaces, although many, if not the majority, of them not being continuous “films” in the strict sense. In this sessile form of life, microorganisms live in close proximity in a matrix of extracellular polymeric substances (EPS). They share emerging properties, clearly distinct from solitary free floating planktonic microbial cells. Common characteristics include the formation of synergistic microconsortia, using the EPS matrix as an external digestion system, the formation of gradients and high biodiversity over microscopically small distances, resource capture and retention, facilitated gene exchange as well as intercellular communication, and enhanced tolerance to antimicrobials. Thus, biofilms belong to the class of collective systems in biology, like forests, beehives, or coral reefs, although the term film addresses only one form of the various manifestations of microbial aggregates. The uncertainty of this term is discussed, and it is acknowledged that it will not likely be replaced soon, but it is recommended to understand these communities in the broader sense of microbial aggregates.

Advanced materials such as tungsten fibre-reinforced composites allow to overcome severe weaknesses of the baseline materials for plasma-facing components — copper and tungsten. The effect of the fusion environment on the mechanical properties of these materials, e.g. the embrittlement by neutron irradiation, plays a key role for the development of future fusion reactors. To simulate this effect, high-energy ions are used as a substitute for the displacement damage by neutrons. We propose the use of very fine tungsten wire as a possibility of studying the influence of irradiation damage on the mechanical properties. This is possible as they allow full-depth irradiation of almost the entire volume despite the limited penetration depth of ions. Geometrical size effects are mitigated due to the nanoscale microstructure of the wire. In addition, similar wire is used in tungsten fibre-reinforced composites. Thus, the investigation of irradiated wire can directly be used for the prediction of the bulk composite properties. For the proof of this concept tungsten wire with a diameter of 16 μm was electrochemically thinned to 5 μm and irradiated with 20.5 MeV W6+ions. The mechanical properties were subsequently determined by macroscopic tensile testing. Irradiation to 0.3, 1 and 9 dpa did not lead to a change of the mechanical behaviour. Both strength and ductility, the latter indicated by the reduction of area, were similar to the as-fabricated state.

Arcs, a source of dust particles and a localized erosion mechanism of the plasma-facing components, are found in all major fusion plasma devices. Measurements of arcs require diagnostics with high temporal and local resolution, which are not available at arc dominated locations in ASDEX Upgrade (AUG). To understand the erosion by arcing and to allow extrapolation for future fusion devices different materials are used to scan the material properties. In AUG, inserts were installed at the inner baffle region to measure the erosion by arcing. The use of polished inserts allows an accurate determination of the arc traces by depth maps obtained by laser profilometery. It turned out that the melting temperature of the materials is the main parameter for erosion. For tungsten mounted at the inner baffle, a region which is deposition dominated, an erosion rate by arcing of 1.2·1013 at cm−2 s−1 is measured. For Beryllium, 9.5·1013 at cm−2 s−1 is extrapolated from its thermal properties. As martensitic–ferritic low-activation steel is under discussion for the use in DEMO, magnetic steels were also investigated. Comparing stainless steel with magnetic steel, much deeper and wider craters are found in the latter one: they reach a depth of −80 μm. The erosion of magnetic steel by arcs is 40 times higher compared to stainless steel, which has almost the same physical properties.

The preferred plasma-facing material in present-day and future magnetic confinement thermonuclear fusion devices is tungsten. This material is mainly chosen because of its high threshold energy for sputtering by hydrogen isotopes as well as its low retention of tritium within the material. From an engineering point of view, however, tungsten is a challenging material to work with as it is an inherently hard and brittle metal. In this respect, established fabrication technologies for tungsten and tungsten based materials are a limiting factor directly affecting the design of plasma-facing components. Against this background, additive manufacturing technologies could prove very beneficial with regard to plasma-facing component applications as they offer flexibilities beyond the possibilities that conventional manufacturing methods offer. Within the present contribution, we report on recent results regarding the additive manufacturing of tungsten by means of powder-bed based selective laser beam melting. In more detail, investigations on pure tungsten manufactured by using elevated substrate preheating temperatures up to 1000 ∘C are described. In ASDEX Upgrade with full-tungsten wall, boronization is an important tool to control tungsten sources and allow reliable operation at low collisionality. The duration of the beneficial effects is limited by the erosion of the boron layer, in particular on the tungsten ion-cyclotron antenna limiters. We report results from experiments exploring injection boron-rich powders in tokamak plasmas as a way to replenish boron coating and extend the lifetime of boronization effects. Pure boron and boron nitride powders were introduced gravitationally in plasma discharges at rates up to 60 mg/s for durations up to 3 s. Boron injection with cumulative amounts >100 mg appeared to improve wall conditions similarly to boronization, with indications of reduced influx of oxygen and tungsten from the antenna limiter. Moreover, cumulative injection of 340 mg of B appeared sufficient to enable successful execution of low collisionality scenarios critically relying on freshly boronized walls.

One of the main aims of Wendelstein 7-X (W7-X), an advanced stellarator, is to investigate the quasi-steady state operation of magnetic confinement devices for nuclear fusion, for which power exhaust is an important issue. A dominant fraction of the energy leaving from the confined plasma region will be removed by 10 so-called island divertor units, which are designed to sustain a maximum heat flux of up to 10 MWm−2. An essential prerequisite for the safe operation of a steady-state device is automatic detection of hot spots and other abnormal events. Simple temperature limits in infrared (IR) thermographic images will not be enough on their own, because of plasma-generated surface coatings and other effects summarized in the following. To protect divertor elements from overheating, and to monitor power deposition onto the divertor elements, near real-time hotspot detection algorithms for the analysis of carbon plasma facing components (PFCs) were implemented and tested in the GLADIS facility.One of the difficulties in hotspot detection in a carbon-based machine is the deposition of plasma impurities as layers with a reduced thermal connection to the underlying bulk material. We have developed and successfully tested a method to classify surface layers and benchmarked the performance of the method with the Tore Supra IR data operating with actively cooled carbon PFCs. The surface layers can be detected in a steady plasma discharge during the initial rise and decay in temperature when a strike line touches parts of the divertor or wall. It can also be detected by modulating electron cyclotron resonance heating (ECRH) input power. This feature allows detection of overheated areas while reducing false positives. For the recent operational campaign, inertially cooled test divertor units (TDU) were installed to prepare for steady-state operation with water-cooled divertor units. Automatic, near real-time detection of hot spots and identification of surface layers in the W7-X divertor are presented. Results are compared with a best fit estimate of the heat transmission coefficient α which is used to calculate heat flux onto the divertor in the presence of surface layers. Abstract Background Current knowledge of the urinary tract microbiome is limited to urine analysis and analysis of biofilms formed on Foley catheters. Bacterial biofilms on ureteral stents have rarely been investigated, and no cultivation-independent data are available on the microbiome of the encrustations on the stents. Results The typical encrustations of organic and inorganic urine-derived material, including microbial biofilms formed during 3–6 weeks on ureteral stents in patients treated for kidney and ureteral stones, and without reported urinary tract infection at the time of stent insertion, were analysed. Next-generation sequencing of the 16S rRNA gene V3–V4 region revealed presence of different urotypes, distinct bacterial communities. Analysis of bacterial load was performed by combining quantification of 16S rRNA gene copy numbers by qPCR with microscopy and cultivation-dependent analysis methods, which revealed that ureteral stent biofilms mostly contain low numbers of bacteria. Fluorescence microscopy indicates the presence of extracellular DNA. Bacteria identified in biofilms by microscopy had mostly morphogenic similarities to gram-positive bacteria, in few cases to Lactobacillus and Corynebacterium, while sequencing showed many additional bacterial genera. Weddellite crystals were absent in biofilms of patients with Enterobacterales and Corynebacterium-dominated microbiomes. Conclusions This study provides novel insights into the bacterial burden in ureteral stent encrustations and the urinary tract microbiome. Short-term (3–6 weeks) ureteral stenting is associated with a low load of viable and visible bacteria in ureteral stent encrustations, which may be different from long-term stenting. Patients could be classified according to different urotypes, some of which were dominated by potentially pathogenic species. Facultative pathogens however appear to be a common feature in patients without clinically manifested urinary tract infection. Trial registration ClinicalTrials.gov, NCT02845726. Recent achievements in studies of the effects of ICRF (Ion Cyclotron Range of Frequencies) power on the SOL (Scrape-Off Layer) and PWI (Plasma Wall Interactions) in ASDEX Upgrade (AUG), Alcator C-Mod, and JET-ILW are reviewed. Capabilities to diagnose and model the effect of DC biasing and associated impurity production at active antennas and on magnetic field connections to antennas are described. The experiments show that ICRF near-fields can lead not only to E × B convection, but also to modifications of the SOL density, which for Alcator C-Mod are limited to a narrow region near antenna. On the other hand, the SOL density distribution along with impurity sources can be tailored using local gas injection in AUG and JET-ILW with a positive effect on reduction of impurity sources. The technique of RF image current cancellation at antenna limiters was successfully applied in AUG using the 3-strap AUG antenna and extended to the 4-strap Alcator C-Mod field-aligned antenna. Multiple observations confirmed the reduction of the impact of ICRF on the SOL and on total impurity production when the ratio of the power of the central straps to the total antenna power is in the range 0.6

Fast video data recorded from 2008 to 2012 in full-tungsten ASDEX Upgrade have been analyzed with the TRACE algorithm developed to automatically detect and track dust particles. The purpose of the work presented in this paper is to complete earlier study of the influence of various discharge conditions on dust rates with an investigation of dust motion under most remarkable conditions. The 3D trajectories of intrinsic dust particles have been confronted to simulations carried out with the DUCAD (Dust Characterization And Dynamics) code, based on the Orbital Motion Limited (OML) approach for dust/plasma interaction modelling. The motion of micrometric spherical dust grains is found to be largely dominated by inertia in the considered experimental conditions. The influence of Vertical Displacement Events (VDEs) and of various heating scenarios on dust areas of formation and trajectories is investigated. It is found that VDEs inject dust originating from the PFCs which are heated by contact with the plasma, upward VDEs having a stronger impact on dust rates and transport than downward VDEs. The use of Neutral Beam Injection (NBI) significantly enhances dust production and transport coming from sectors where injectors are installed. On the contrary, Ion Cyclotron Resonance Heating significantly reduces dust rates and transport in the whole plasma volume. The tungsten “heavy alloy” HPM 1850, a liquid-phase sintered composite material with two weight percent Ni and one weight percent Fe, as well as the self-passivating tungsten alloy W-10Cr-0.5Y, a high temperature oxidation resistant alloy with 10 weight percent of Cr and 0.5 weight percent of Y, were investigated with respect to their deuterium retention. The samples were deuterium loaded in an electron cyclotron resonance plasma up to a fluence of 1025m−2. The deuterium retention was then investigated by Nuclear Reaction Analysis and by Thermal Desorption. In HPM 1850 the observed deuterium amount was similar to pure tungsten, however the outgassing behaviour during thermal desorption was considerably faster. In W-10Cr-0.5Y the released deuterium amount during thermal desorption was about one order of magnitude higher; by comparison of nuclear reaction analysis and thermal desorption this was attributed to deeper diffusion of deuterium into the bulk of the material. The design and manufacture of highly heat loaded plasma-facing components (PFCs) represents a major challenge for the realisation of thermonuclear magnetic confinement fusion. The performance of such PFCs is essentially related to the properties of the materials that are used for their design. Currently, tungsten fibre-reinforced metal matrix composites (MMCs) are regarded as promising advanced materials for PFC applications. In this respect, tungsten fibre-reinforced tungsten is being investigated as an advanced pseudo-ductile plasma-facing material while tungsten fibre-reinforced copper is being developed as an advanced heat sink material. The essential ingredients for the abovementioned MMCs are the fibrous reinforcements which are commercially available drawn tungsten fibres.An important aspect regarding the development of the abovementioned MMCs is the effect of the composite material manufacturing process on the properties of these high-strength reinforcements. During composite material manufacturing experiments it has been found that the mechanical properties of the used W fibres can be deteriorated significantly already at process temperatures of approximately 1200 °C.Against this background, dedicated investigations have been conducted on drawn tungsten fibre samples. In more detail, single fibre tensile tests, microstructural investigations as well as chemical composition analyses have been conducted. All in all, the performed investigations indicate that impurities incorporated into the tungsten fibre material are the underlying reason for the observed deterioration of the mechanical properties. For the first wall of a fusion reactor unique challenges on materials in extreme environments require advanced features in areas ranging from mechanical strength to thermal properties. The main challenges include wall lifetime, erosion, fuel management and overall safety. For the lifetime of the wall material, considerations of thermal fatigue due to transient heat loading are crucial as severe mechanical and thermal loads during operation are expected.Tungsten (W) is the main candidate material for the first wall of a fusion reactor as it is resilient against erosion, has the highest melting point of any metal and shows rather benign transmutation behavior under neutron irradiation. However, Tungsten has an issue related to intrinsic brittleness as well as operational embrittlement. To overcome this, a W-fiber enhanced W-composite material (Wf/W) incorporating extrinsic toughening mechanisms has been developed. Recently progress has been made in the powder metallurgical routes towards fully dense multi short-fiber Wf/W. For reasonable performance with respect to mechanical properties and hydrogen retention a fully dense pseudo-ductile Wf/W with is crucial. The properties of the used fibres are crucial. For the composite mechanisms to work a level of strength of the used fibres is required. In this contribution the change in ductility of the fibres is studied.In this contribution it is shown that excluding or minimising the impact of carbon impurities during the sintering process can significantly improve the mechanical properties of the fibres. New test results on the behaviour of PM Wf/W with and without a diffusion barrier during the sintering show a clear benefit as the fibres can retain ductility. Not the grain growth during sintering but the carbon present during sintering is clearly identified as determining the mechanical properties of the fibres.

Use of new 3-strap ICRF antennas with all-tungsten (W) limiters in ASDEX Upgrade results in a reduction of the W sources at the antenna limiters and of the W content in the confined plasma by at least a factor of 2 compared to the W-limiter 2-strap antennas used in the past. The reduction is observed with a broad range of plasma shapes. In multiple locations of antenna frame, the limiter W source has a minimum when RF image currents are decreased by cancellation of the RF current contributions of the central and the outer straps. In JET with ITER-like wall, ITER-like antenna produces about 20% less of main chamber radiation and of W content compared to the old A2 antennas. However the effect of the A2 antennas on W content is scattered depending on which antennas are powered. Experiments in JET with trace nitrogen (N2) injection show that a presence of active ICRF antenna close to the midplane injection valve has little effect on the core N content, both in dipole and in -90° phasing. This indicates that the effect of ICRF on impurity transport across the scape-off-layer is small in JET compared to the dominant effect on impurity sources leading to increased impurity levels during ICRF operation. In 2013 a redesigned solid tungsten divertor, Div-III, was installed in ASDEX Upgrade. In two experimental campaigns more than 2700 plasma discharges with up to 10s duration, 110MJ plasma heating, and reaching a Psep/R of 10MW/m were conducted. Div-III is an adiabatically loaded divertor with tungsten target tiles clamped onto a water cooled structure. The tungsten target tiles were inspected and characterized during the shut-down period in 2014. As a result of the target inspection characteristic modifications of the plasma exposed tiles were detected that were not found during the target qualification in a high heat load test facility. 126 out of 128 tiles reveal deep cracks through the target. A lot of tiles show shallow cracks in the high heat load region and finally protruding tiles with strong local damages were found. We discuss the benefits of installing a pair of in-vessel coils with currents |Ifx| ≲ 50 kAt in the upper divertor of ASDEX Upgrade (AUG) to study a series of ‘alternative’ divertor configurations, like the Snowflake (SF) and the X-divertor (XD), that are currently considered as alternative solutions for the power exhaust problem. The possibility of operating the standard lower single-null (SN) and double-null (DN) would be preserved. Potential effects to reduce the peak parallel- and/or perpendicular heat flux are predicted from a simple geometrical-diffusive model as well as by numerical EMC3-EIRENE simulations for pure deuterium attached conditions with spatially constant diffusion coefficients. Beyond that a series of other potential transport- and radiation related heat flux mitigation effects are identified and could be studied experimentally with the modified upper divertor in the high-power divertor Tokamak AUG. DEMO is the name for the first stage prototype fusion reactor considered to be the next step after ITER. For the realization of fusion energy especially materials questions pose a significant challenge already today. Advanced materials solution are under discussion in order to allow operation under reactor conditions [1] and are already under development used in the next step devices. Apart from issues related to material properties such as strength, ductility, resistance against melting and cracking one of the major issues to be tackled is the interaction with the fusion plasma. Advanced tungsten (W) materials as discussed below do not necessarily add additional lifetime issues, they will, however, add concerns related to erosion or surface morphology changes due to preferential sputtering. Retention of fuel and exhaust species are one of the main concerns. Retention of hydrogen will be one of the major issues to be solved in advanced materials as especially composites and alloys will introduce new hydrogen interactions mechanisms. Initial calculations show these mechanisms. Especially for Helium as the main impurity species material issues arise related to surfaces modification and embrittlement. Solutions are proposed to mitigate effects on material properties and introduce new release mechanisms.

Tungsten (W) will be used as material for plasma-facing components (PFCs) in the divertor of ITER and interact with Helium (He) ions either from initial He plasma operation or from Deuterium-Tritium (DT) fusion reactions in the active operation phase. Laboratory experiments reported that in a specific operational window of impact energy, ion fluence, and surface temperature (Ein ≥ 20 eV, ϕ ≥ 1 × 1024 He+mBt=2.5T,Ip=0.8MA,Paux≃8.0MW Tsurf ≥ 1000 K) a modification of W surfaces occurs resulting in the formation of He-induced W nanostructures. Experiments in ASDEX Upgrade H-mode plasmas (Ein=37keV T, ϕ≃0.75×1024He0m−2 MA, Paux ≃ 8.0 MW) in He have been carried out to investigate in detail (a) the potential growth of W nanostructures on pre-damaged W samples incorporating He nanobubbles, and (b) the potential ELM-induced erosion of W nanostructure. Both W surface modifications were generated artificially in the GLADIS facility by He bombardment of W samples at ϕ≃1×1024He0m−2 keV (a) to ϕ ≃ 0.75 × 1024 He0mϕ≃1.6×1024He+m−2 at Tsurf ≃ 1800 K and (b) ϕ ≃ 1 × 1024 He0m−2 at Tsurf ≃ 2300 K prior to exposure in the divertor manipulator of ASDEX Upgrade. Though in part (a) conditions of W nanostructure growth with a total He ion fluence of ϕ ≃ 1.6 × 1024 He+m−2 and peak He ion impact energies above 150 eV were met, no growth could be detected. In part (b) lower density plasmas with more pronounced type I ELMs, carrying energetic He ions in the keV range, were executed with the strike-line positioned on 2 µm thick W nanostructure accumulating a fluence of ϕ ≃ 0.8 × 1024 He+m−2. Post-mortem analysis revealed that co-deposition by predominantly W, and Boron (B), eroded at the main chamber wall and transported into the divertor, took place on all W samples. Erosion of W nanostructure or its formation was hindered by the fact that the outer divertor at the location of the samples was turned under these He plasma conditions into a net deposition zone by W, B and Carbon (C) ions. The surface morphology with large roughness and effective surface area act as a catcher for the impinging impurities. Thus, apart from operation in the existence diagram of W nanostructure with respect to Tsurf, ϕ, and Ein, also the impinging impurity flux contribution needs to be considered in predictions concerning the formation of W nanostructures. The European Fusion Roadmap foresees water cooled plasma facing components in a first DEMO design in order to provide enough margin for the cooling capacity and to only moderately extrapolate the technology which was developed and tested for ITER. In order to make best use of the water cooling concept copper (Cu) and copper-chromium-zirconium alloy (CuCrZr) are envisaged as heat sink whereas as armour tungsten (W) based materials will be used. Combining both materials in a high heat flux component asks for an increase of their operational range towards higher temperature in case of Cu/CuCrZr and lower temperatures for W. A remedy for both issues- brittleness of W and degrading strength of CuCrZr- could be the use of W fibres (Wf) in W and Cu based composites. Fibre preforms could be manufactured with industrially viable textile techniques. Flat textiles with a combination of 150/70 µm W wires have been chosen for layered deposition of tungsten-fibre reinforced tungsten (Wf/W) samples and tubular multi-layered braidings with W wire thickness of 50 µm were produced as a preform for tungsten-fibre reinforced copper (Wf /Cu) tubes. Cu melt infiltration was performed together with an industrial partner resulting in sample tubes without any blowholes. Property estimation by mean field homogenisation predicts strongly enhanced strength of the Wf/CuCrZr composite compared to its pure CuCrZr counterpart. Wf /W composites show very high toughness and damage tolerance even at room temperature. Cyclic load tests reveal that the extrinsic toughening mechanisms counteracting the crack growth are active and stable. FEM simulations of the Wf/W composite suggest that the influence of fibre debonding, which is an integral part of the toughening mechanisms, and reduced thermal conductivity of the fibre due to the necessary interlayers do not strongly influence the thermal properties of future components. The development of advanced materials is essential for sophisticated energy systems like a future fusion reactor. Tungsten fibre-reinforced tungsten composites (Wf/W) utilize extrinsic toughening mechanisms and therefore overcome the intrinsic brittleness of tungsten at low temperature and its sensitivity to operational embrittlement. This material has been successfully produced and tested during the last years and the focus is now put on the technological realisation for the use in plasma facing components of fusion devices. In this contribution, we present a way to utilize Wf/W composites for divertor applications by a fabrication route based on the chemical vapour deposition (CVD) of tungsten. Mock-ups based on the ITER typical design can be realized by the implementation of Wf/W tiles. A concept based on a layered deposition approach allows the production of such tiles in the required geometry. One fibre layer after the other is positioned and ingrown into the W-matrix until the final sample size is reached. Charpy impact tests on these samples showed an increased fracture energy mainly due to the ductile deformation of the tungsten fibres. The use of Wf/W could broaden the operation temperature window of tungsten significantly and mitigate problems of deep cracking occurring typically in cyclic high heat flux loading. Textile techniques are utilized to optimise the tungsten wire positioning and process speed of preform production. A new device dedicated to the chemical deposition of W enhances significantly, the available machine time for processing and optimisation. Modelling shows that good deposition results are achievable by the use of a convectional flow and a directed temperature profile in an infiltration process.

To overcome the intrinsic brittleness of tungsten (W), a tungsten fiber-reinforced tungsten-composite material (Wf/W) is under development. The composite addresses the brittleness of W by extrinsic toughening through the introduction of energy dissipation mechanisms. These mechanisms allow the reduction of stress peaks and thus improve the materials resistance against crack growth. They do not rely on the intrinsinc material properties such as ductility. By utilizing powder metallurgy (PM) one could benefit from available industrialized approaches for composite production and alloying routes. In this contribution the PM method of hot isostatic pressing (HIP) is used to produce Wf/W samples containing W fibers coated with an Er2O3 interface. Analysis of the matrix material demonstrates a dense tungsten bulk, a deformed fiber and a deformed, but still intact interface layer. Metallographic analysis reveals indentations of powder particles in the interface, forming a complex 3D structure. Special emphasis is placed on push-out tests of single fiber HIP samples, where a load is applied via a small indenter on the fiber, to test the debonding and frictional properties of the Er2O3 interface region enabling the energy dissipation mechanisms. Together with the obtained experimental results, an axisymmetric finite element model is discussed and compared to existing work. In the HIP Wf/W composites the matrix adhesion is rather large and can dominate the push-out behavior. This is in contrast to the previously tested CVD produced samples.

Biooxidation of reduced inorganic sulfur compounds (RISCs) by thermoacidophiles is of particular interest for the biomining industry and for environmental issues, e.g., formation of acid mine drainage (AMD). Up to now, interfacial interactions of acidophiles with elemental sulfur as well as the mechanisms of sulfur oxidation by acidophiles, especially thermoacidophiles, are not yet fully clear. This work focused on how a crenarchaeal isolate Acidianus sp. DSM 29099 interacts with elemental sulfur. Analysis by Confocal laser scanning microscopy (CLSM) and Atomic force microscopy (AFM) in combination with Epifluorescence microscopy (EFM) shows that biofilms on elemental sulfur are characterized by single colonies and a monolayer in first stage and later on 3-D structures with a diameter of up to 100 μm. The analysis of extracellular polymeric substances (EPS) by a non-destructive lectin approach (fluorescence lectin-barcoding analysis) using several fluorochromes shows that intial attachment was featured by footprints rich in biofilm cells that were embedded in an EPS matrix consisting of various glycoconjugates. Wet chemistry data indicate that carbohydrates, proteins, lipids and uronic acids are the main components. Attenuated reflectance (ATR)-Fourier transformation infrared spectroscopy (FTIR) and high-performance anion exchange chromatography with pulsed amperometric detection (HPAE-PAD) indicate glucose and mannose as the main monosaccharides in EPS polysaccharides. EPS composition as well as sugar types in EPS vary according to substrate (sulfur or tetrathionate) and lifestyle (biofilms and planktonic cells). This study provides information on the building blocks/make up as well as dynamics of biofilms of thermoacidophilic archaea in extremely acidic environments.

Due to the current dependence on biocidal antifouling coatings for biofouling control, there is a continuing international challenge to develop more environmentally acceptable antifouling systems. Fluctuating the pH values on paint surfaces is one of these approaches. We developed an antifouling test device to investigate algal biofilms on conductive paints by using a flume with electrochemically working test panels and subsequent confocal laser scanning microscopy (CLSM) of biofilms. By employing a pole reversal of direct current, fluctuating pH values on the paint surface were generated. As a consequence of the resulting pH stress, colonization of the paint surface by diatoms decreased substantially. The density of biofilm algae decreased with increasing pH fluctuations. However, breaks between electrochemical treatments should not exceed one hour. Overall, we established an experimental setup for testing the antifouling capabilities of electrodes based on conductive paints, which could be used for further development of these varnishes.

A global heat flux model based on a fractional derivative of plasma pressure is proposed for the heat transport in fusion plasmas. The degree of the fractional derivative of the heat flux, α, is defined through the power balance analysis of the steady state. The model was used to obtain the experimental values of α for a large database of the Joint European Torus (JET) carbon-wall as well as ITER like-wall plasmas. Although methanol has frequently been used as an inexpensive supplementary carbon source to support treatment processes, knowledge of the resultant microbial biofilms, their 3D architecture, microenvironments, exopolymer chemistry and populations remains limited. We supplied methanol as a supplementary carbon source to biofilms developing in rotating annular reactors. Analysis of circulation waters (1.0 l d−1) indicated that dissolved organic carbon was reduced by 25%, NO3-nitrogen by 95%, and total phosphorus by 70%. Analyses of populations using culture based techniques and fluorescence in situ hybridization indicated enrichment of nitrifiers, denitrifiers, and methylotrophic bacteria relative to reference biofilms not receiving methanol. The biofilms that developed were up to 4,000 μm thick. Staining with fluor conjugated lectins in combination with nucleic acid stains, revealed the presence of discrete bacterial cells inside complex globular polymeric structures. These structures were in turn surrounded by an interstitial polymer containing a variety of bacterial cell types. The globular structures bound FITC-conjugated lectins, from Canavalia ensiformis and Ulex europeaus. The FITC-lectin of Phaseolus vulgaris bound the surface of the globular structures and more generally within the matrix. Chemical analyses of the polymer paralleled the results of lectin analyses indicating that the dominant neutral sugars were glucose, galactose, mannose, rhamnose, with fucose and ribose as minor constituents. Amino sugars were not detected. Dual channel imaging with pH sensitive probes indicated that pH gradients from pH 4 to 7 occurred across the globular microcolonies. Critically for the maintenance of aerobic conditions throughout the thick biofilm it was extensively penetrated by a fine fissure network revealed by the location of fluorescent latex microbeads as detected by confocal laser scanning microscopy. Microelectrode studies confirmed the absence of any detectable Eh gradients within the biofilm. However, mobility of various size-fractionated fluorescent probes indicated that the basal region was only penetrated by the lowest molecular weight probes with a hydrated radius of 2.2 nm or less. These observations indicate the selection of a unique, thick (>4,000 μm) microbial community in which a self-organized architecture promotes the maintenance of optimal conditions and metabolism throughout the biofilm community.

Anaerobic granulation at elevated salinities has been discussed in several analytical and engineering based studies. They report either enhanced or decreased efficiencies in relation to different Na+ levels. To evaluate this discrepancy, we focused on the microbial and structural dynamics of granules formed in two upflow anaerobic sludge blanket (UASB) reactors treating synthetic wastewater at low (5 g/L Na+) and high (20 g/L Na+) salinity conditions. Granules were successfully formed in both conditions, but at high salinity, the start-up inoculum quickly formed larger granules having a thicker gel layer in comparison to granules developed at low salinity. Granules retained high concentrations of sodium without any negative effect on biomass activity and structure. 16S rRNA gene analysis and Fluorescence in Situ Hybridization (FISH) identified the acetotrophic Methanosaeta harundinacea as the dominant microorganism at both salinities. Fluorescence lectin bar coding (FLBC) screening highlighted a significant shift in the glycoconjugate pattern between granules grown at 5 and 20 g/L of Na+, and the presence of different extracellular domains. The excretion of a Mannose-rich cloud-like glycoconjugate matrix, which seems to form a protective layer for some methanogenic cells clusters, was found to be the main distinctive feature of the microbial community grown at high salinity conditions.

Filamentous macroscopic algae were observed in slightly acidic to circumneutral (pH 5.9–6.5), metal-rich stream water that leaked out from a former uranium mining district (Ronneburg, Germany). These algae differed in color and morphology and were encrusted with Fe-deposits. To elucidate their potential interaction with Fe(II)-oxidizing bacteria (FeOB), we collected algal samples at three time points during summer 2013 and studied the algae-bacteria-mineral compositions via confocal laser scanning microscopy (CLSM), scanning electron microscopy (SEM), Fourier transform infrared (FTIR) spectra, and a 16S and 18S rRNA gene-based bacterial and algae community analysis. Surprisingly, sequencing analysis of 18S rRNA gene regions of green and brown algae revealed high homologies with the freshwater algae Tribonema (99.9–100 %). CLSM imaging indicated a loss of active chloroplasts in the algae cells, which may be responsible for the change in color in . Fe(III)-precipitates on algal cells identified as ferrihydrite and schwertmannite by FTIR were associated with microbes and extracellular polymeric substances (EPS)-like glycoconjugates. SEM imaging revealed that while the green algae were fully encrusted with Fe-precipitates, the brown algae often exhibited discontinuous series of precipitates. This pattern was likely due to the intercalary growth of algal filaments which allowed them to avoid detrimental encrustation. 16S rRNA gene-targeted studies revealed that Gallionella-related FeOB dominated the bacterial RNA and DNA communities (70–97 and 63–96 %, respectively), suggesting their capacity to compete with the abiotic Fe-oxidation under the putative oxygen-saturated conditions that occur in association with photosynthetic algae. Quantitative PCR (polymerase chain reaction) revealed even higher Gallionella-related 16S rRNA gene copy numbers on the surface of green algae compared to the brown algae. The latter harbored a higher microbial diversity, including some putative predators of algae. A loss of chloroplasts in the brown algae could have led to lower photosynthetic activities and reduced EPS production, which is known to affect predator colonization. Collectively, our results suggest the coexistence of oxygen-generating algae Tribonema sp. and strictly microaerophilic neutrophilic FeOB in a heavy metal-rich environment.

The extracellular matrix is a poorly studied, yet important component of dental biofilms. Fluorescence lectin-binding analysis (FLBA) is a powerful tool to characterize glycoconjugates in the biofilm matrix. This study aimed to systematically investigate the ability of 75 fluorescently labeled lectins to visualize and quantify extracellular glycoconjugates in dental biofilms. Lectin binding was screened on pooled supragingival biofilm samples collected from 76 subjects using confocal microscopy. FLBA was then performed with 10 selected lectins on biofilms grown in situ for 48 h in the absence of sucrose. For five lectins that proved particularly suitable, stained biovolumes were quantified and correlated to the bacterial composition of the biofilms. Additionally, combinations of up to three differently labeled lectins were tested. Of the 10 lectins, five bound particularly well in 48-h-biofilms: Aleuria aurantia (AAL), Calystega sepiem (Calsepa), Lycopersicon esculentum (LEA), Morniga-G (MNA-G) and Helix pomatia (HPA). No significant correlation between the binding of specific lectins and bacterial composition was found. Fluorescently labeled lectins enable the visualization of glycoconjugates in the dental biofilm matrix. The characterization and quantification of glycoconjugates in dental biofilms require a combination of several lectins. For 48-h-biofilms grown in absence of sucrose, AAL, Calsepa, HPA, LEA, and MNA-G are recommendable.

The bovine milk protein osteopontin (OPN) may be an efficient means to prevent bacterial adhesion to dental tissues and control biofilm formation. This study sought to determine to what extent OPN impacts adhesion forces and surface attachment of different bacterial strains involved in dental caries or medical device–related infections. It further investigated if OPN’s effect on adhesion is caused by blocking the accessibility of glycoconjugates on bacterial surfaces. Bacterial adhesion was determined in a shear-controlled flow cell system in the presence of different concentrations of OPN, and interaction forces of single bacteria were quantified using single-cell force spectroscopy before and after OPN exposure. Moreover, the study investigated OPN’s effect on the accessibility of cell surface glycoconjugates through fluorescence lectin-binding analysis. OPN strongly affected bacterial adhesion in a dose-dependent manner for all investigated species (Actinomyces naeslundii, Actinomyces viscosus, Lactobacillus paracasei subsp. paracasei, Staphylococcus epidermidis, Streptococcus mitis, and Streptococcus oralis). Likewise, adhesion forces decreased after OPN treatment. No effect of OPN on the lectin-accessibility to glycoconjugates was found. OPN reduces the adhesion and adhesion force/energy of a variety of bacteria and has a potential therapeutic use for biofilm control. OPN acts upon bacterial adhesion without blocking cell surface glycoconjugates.

Despite the ubiquity of Mn oxides in natural environments, there are only a few observations of biological Mn(II) oxidation at pH < 6. The lack of low pH Mn-oxidizing bacteria (MOB) isolates limits our understanding of how pH influences biological Mn(II) oxidation in extreme environments. Here, we report that a novel MOB isolate, Mesorhizobium australicum strain T-G1, isolated from an acidic and metalliferous uranium mining area, can oxidize Mn(II) at both acidic and neutral pH using different enzymatic pathways. X-ray diffraction (XRD), Raman spectroscopy, and scanning electron microscopy with energy dispersive X-ray spectroscopy (SEM-EDS) revealed that T-G1 initiated bixbyite-like Mn oxide formation at pH 5.5 which coincided with multi-copper oxidase (MCO) expression from early exponential phase to late stationary phase. In contrast, reactive oxygen species (ROS), particularly superoxide, appeared to be more important for T-G1 mediated Mn(II) oxidation at neutral pH. ROS was produced in parallel with the occurrence of Mn(II) oxidation at pH 7.2 from early stationary phase. Solid phase Mn oxides did not precipitate, which is consistent with the presence of a high amount of H2O2 and lower activity of catalase in the liquid culture at pH 7.2. Our results show that M. australicum T-G1, an acid tolerant MOB, can initiate Mn(II) oxidation by varying its oxidation mechanisms depending on the pH and may play an important role in low pH manganese biogeochemical cycling.

Powders of metallurgically prepared precursors expand to a foam differently depending on how fast they are heated to their foaming temperature. The foaming behavior of four alloys was studied with heating rates varying from 0.1 K/s to 18 K/s. It was found that each alloy shows its own non-linear behavior of expansion values. Alloys not containing magnesium exhibit a maximum at intermediate heating rates, whereas Mg-containing alloys tend to expand more for higher heating rates.

Microbial biofilm systems are defined as interface-associated microorganisms embedded into a self-produced matrix. The extracellular matrix represents a continuous challenge in terms of characterization and analysis. The tools applied in more detailed studies comprise extraction/chemical analysis, molecular characterization, and visualisation using various techniques. Imaging by laser microscopy became a standard tool for biofilm analysis, and, in combination with fluorescently labelled lectins, the glycoconjugates of the matrix can be assessed. By employing this approach a wide range of pure culture biofilms from different habitats were examined using the commercially available lectins. From the results, a binary barcode pattern of lectin binding can be generated. Furthermore, the results can be fine-tuned and transferred into a heat map according to signal intensity. The lectin barcode approach is suggested as a useful tool for investigating the biofilm matrix characteristics and dynamics at various levels, e.g. bacterial cell surfaces, adhesive footprints, individual microcolonies, and the gross biofilm or bio-aggregate. Hence fluorescence lectin bar-coding (FLBC) serves as a basis for a subsequent tailor-made fluorescence lectin-binding analysis (FLBA) of a particular biofilm. So far, the lectin approach represents the only tool for in situ characterization of the glycoconjugate makeup in biofilm systems. Furthermore, lectin staining lends itself to other fluorescence techniques in order to correlate it with cellular biofilm constituents in general and glycoconjugate producers in particular.

Aerobic granular sludge is based on self-granulated flocs forming mobile biofilms with a gel-like consistence. Bacterial and structural dynamics from flocs to granules were followed in anaerobic-aerobic sequencing batch reactors fed with synthetic wastewater, namely a bubble column (BC-SBR) operated under wash-out conditions for fast granulation, and two stirred-tank enrichments of Accumulibacter (PAO-SBR) and Competibacter (GAO-SBR) operated at steady-state. In the BC-SBR, granules formed within two weeks by swelling of Zoogloea colonies around flocs, developing subsequently smooth zoogloeal biofilms. However, Zoogloea predominance (37-79%) led to deteriorated nutrient removal during the first months of reactor operation. Upon maturation, improved nitrification (80-100%), nitrogen removal (43-83%), and high but unstable dephosphatation (75-100%) were obtained. Proliferation of dense clusters of nitrifiers, Accumulibacter, and Competibacter from granule cores outwards resulted in heterogeneous bioaggregates, inside which only low abundance Zoogloea (

We investigated the ability of bacterial communities to colonize and dissolve two biogenic carbonates (Foraminifera and oyster shells). Bacterial carbonate dissolution in the upper water column is postulated to be driven by metabolic activity of bacteria directly colonising carbonate surfaces and the subsequent development of acidic microenvironments. We employed a combination of microsensor measurements, scanning electron microscopy (SEM), confocal laser scanning microscopy (CLSM) and image analysis and molecular documentation of colonising bacteria to monitor microbial processes and document changes in shell surface topography. Bacterial communities rapidly colonised shell surfaces, forming dense biofilms with extracellular polymeric substance (EPS) deposits. Despite this, we found no evidence of bacterially mediated carbonate dissolution. Dissolution was not indicated by Ca²⁺ microprofiles, nor was changes in shell surface structure related to the presence of colonizing bacteria. Given the short time (days) settling carbonate material is actually in the twilight zone (500-1000 m), it is highly unlikely that microbial metabolic activity on directly colonised shells plays a significant role in dissolving settling carbonates in the shallow ocean.

Extreme-ultraviolet to x-ray free-electron lasers (FELs) in operation for scientific applications are up to now single-user facilities. While most FELs generate around 100 photon pulses per second, FLASH at DESY can deliver almost two orders of magnitude more pulses in this time span due to its superconducting accelerator technology. This makes the facility a prime candidate to realize the next step in FELs—dividing the electron pulse trains into several FEL lines and delivering photon pulses to several users at the same time. Hence, FLASH has been extended with a second undulator line and self-amplified spontaneous emission (SASE) is demonstrated in both FELs simultaneously. FLASH can now deliver MHz pulse trains to two user experiments in parallel with individually selected photon beam characteristics. First results of the capabilities of this extension are shown with emphasis on independent variation of wavelength, repetition rate, and photon pulse length.

The mechanisms responsible for hyperfiltration in diabetes mellitus (DM) as well as for the initiation and progression of diabetic nephropathy are not fully elucidated. Enhanced prostaglandin E2 (PGE2) production has been invoked in the former and thromboxane (TXB2) and hyperlipidemia in the latter. Fish oil (FO)-enriched diets can favorably alter eicosanoid synthesis and serum lipid profiles. We therefore examined the effects of a FO-enriched diet on glomerular filtration (GFR), proteinuria, glomerular eicosanoid production, and serum lipids in rats with streptozotocin-induced DM (STZ-DM). Groups of 5-8 rats with STZ-DM were maintained on low insulin and then pair-fed with isocaloric diets enriched with either FO (20% w/w) or beef tallow (BT; 20% w/w). GFR was determined in the same animals at onset of diet and after 8 and 20 weeks on the respective diets by [14C]inulin clearance using implanted osmotic minipumps each time. Significant hyperfiltration was present initially and GFR did not change on either diet for 20 weeks, in spite of a significant and greater than 50% decrease in all prostaglandins (PGE2, TXB2, PGF2 alpha, 6-keto, PGF1 alpha) produced by glomeruli isolated from DM/FO as compared to DM/BT or control rats. FO diet completely corrected the hypertriglyceridemia of diabetes and significantly reduced the mild and early proteinuria of DM. The decrease in proteinuria and the correction of hyperlipidemia of DM by a FO-enriched diet may be beneficial in the long term not only for the development of diabetic glomerulopathy, but also for the accelerated atherosclerosis of DM.

Using liquid metals confined in capillary porous structures (CPSs) as a plasma-facing component (PFC) could prolong the lifetime of the divertor in the high heat flux area. However, the high atomic number of tin (Sn) limits its acceptable fraction in the main plasma. Therefore, a crucial step in developing this concept is to test it in a tokamak environment, particularly in the diverted plasma region, e.g. ASDEX Upgrade (AUG). In this paper, the design of liquid tin module (LTM) is explained, and the testing in the high heat flux device GLADIS before its use in AUG is presented. The LTM was additively manufactured using selective laser melting, consisting of a 1.5mm porous layer tungsten (W) directly attached to a solid W bulk. The LTM has a plasma-facing area of 16×40mm2 and was filled with 1.54g of Sn. In GLADIS, the module was exposed to power loads between 2 and 8MWm−2 for 1 up to 10s, first unfilled and later filled with Sn. The surface temperature was monitored with infrared imaging and pyrometry. The thermal response was used to compare with simulations in Ansys Mechanical, enabling a determination of the module’s effective thermal properties. Sn droplets could be observed on the infrared camera, until a surface temperature of about a 1000°C was reached. The enhanced wetting of tin on the plasma-facing surface, which was observed by a visible camera, suggests that there is a conditioning of the surface, possibly due to the removal of impurities and oxides. Subsequent examinations of the adjacent tile revealed minor Sn leakages emanating from the module’s edge. Furthermore, the module showed no indication of mechanical failure. Therefore, these results indicated that the LTM qualifies for the heat fluxes expected in ASDEX Upgrade.

Wendelstein 7-X, the world’s largest superconducting stellarator in Greifswald (Germany), started plasma experiments with a water-cooled plasma-facing wall in 2022, allowing for long pulse operation. In parallel, a project was launched in 2021 to develop a W based divertor, replacing the current CFC divertor, to demonstrate plasma performance of a stellarator with a reactor relevant plasma facing materials with low tritium retention. The project consists of two tasks: Based on experience from the previous experimental campaigns and improved physics modelling, the geometry of the plasma-facing surface of the divertor and baffles is optimized to prevent overloads and to improve exhaust. In parallel, the manufacturing technology for a W based target module is qualified.This paper gives a status update of project. It focusses on the conceptual design of a W based target module, the manufacturing technology and its qualification, which is conducted in the framework of the EUROfusion funded WPDIV program. A flat tile design in which a target module is made of a single target element is pursued. The technology must allow for moderate curvatures of the plasma-facing surface to follow the magnetic field lines. The target element is designed for steady state heat loads of 10 MW/m2 (as for the CFC divertor). Target modules of a similar size and weight as for the CFC divertor are assumed (approx.

Cold gas spraying for the production of thick tungsten (W) coatings has been investigated for use at plasma facing components in fusion devices. Since the brittle nature of W strongly impedes its deposition, a systematic study was performed using mixtures of tungsten and tantalum (Ta) powders. Whereas the use of 100% W powder was not successful yet, 2 mm thick coatings on steel were produced by using a W/Ta powder mixture with 90 vol% W, yielding a W content in the coating of 70 vol%. The coatings show negligible porosity and very good adhesion to the substrate. High heat flux experiments on samples with the size 80 × 80 mm2 were performed in order to investigate the behaviour under low (≤4 MW/m2) steady state loads and high power (∼40 MW/m2) transients. During the pulses with low power density, being typical for applications at the main chamber first wall, no defects were observed and a thermal conductivity close to that of the bulk materials was found. During the high power transients lasting for 200 ms cracks parallel to the surface appeared inside the coating.

Solid tungsten tiles exposed at the outer strike point module of the ASDEX Upgrade divertor show cracks parallel to surface in the bulk and humps at the surface. At ASDEX Upgrade these horizontal cracks were observed for the first time. About 10 % of all tiles exhibit protrusions originated by horizontal cracks about 3–5 mm below the surface. The position of this damage coincidence with the location of highest deposited energy, which varies for the different tiles due to the mounting. The tungsten at the locations of the horizontal cracks exhibits recrystallization and grain growth, presumably due to the worse thermal contact caused by the horizontal cracks. Although, the operation of AUG was not hampered by these damages, these tiles had to be exchanged preventing progression of these damages. High thermal stress due to temperature gradients is discussed to originate these cracks. Additionally, humps are found at the surface at the position of high heat flux. Their occurrence seems to be related to the increased number of high power discharges. Focused ion beam prepared cross sections indicate pores and cavities in and below these humps. This finding is discussed in the view of He bubble formation in tungsten.
