Abstract Flue-curing and redrying are important processing stages before tobacco fermentation, closely linked to microbial actions that influence the fermentation process. It is necessary to investigate the effects of flue-curing and redrying on diversity and succession of tobacco fungal communities. It was shown that a total of 9 phyla, 33 classes, 94 orders, 266 families, 646 genera, and 6,396 amplicon sequence variants (ASVs) were identified in the fungi communities of 36 samples from different processing stages (before flue-curing, after flue-curing, before redrying and after redrying) based on high-throughput sequencing technology. Dominant genera shared by tobacco leaves at different stages were Alternaria and Sampaiozyma. About 80% of fungi in stored tobacco leaves after redrying originated from fresh tobacco leaves before flue-curing, while the rest were primarily enriched in the post-harvest processing environment. After flue-curing, major molds like Aspergillus and Penicillium were notably enriched. The distribution of fungal communities suggested that the flue-curing and redrying had a significant impact on fungal composition. Functional annotation of fungal communities at the guild level exhibited differences during processing stages. Main fungal functional groups were identified. In summary, our study elucidated dynamic changes in the composition of fungal communities and highlighted key stages in mold enrichment during tobacco leaf processing, laying groundwork for mildew prevention and control during tobacco leaf fermentation.

Abstract Enterobacter ludwigii has been proven by numerous studies to be an effective plant growth promoter. Enterobacter ludwigii T977 was isolated from leaves of Nicotiana tabacum L. Yunyan 97 which showing high starch degrading ability. The optimal fermentation carbon source of strain T977 was starch, with optimal starch concentration as 2.5 g/L, and the most suitable fermentation nitrogen source for the strain T977 was ammonium acetate, with optimal concentration as 0.25 g/L. The spaying treatment of strain T977 could reduce the starch content of upper leaves from 3.77% to 1.43%, the total sugar and reducing sugar decreased slightly, the starch content of middle leaves decreased from 5.63% to 3.18%, the content of total sugar and reducing sugar increased in middle leaves, and the other chemical components were in the appropriate range. Here, we reported 4.77 MB whole genome of a starch-degrading E. ludwigii T977 that encodes 4501 proteins, 11 α-amylases in GH13 family were identified, and the amylase (GM000159) with signal peptide may play important role in degradation of starch in tobacco leaves. Our study may provide an effective microbiological mean for reducing starch content in tobacco leaves.

Abstract The impact of various organic amendments on soil organic carbon (SOC) have rarely been reported. To address this, a laboratory experiment was designed to scrutinize the effects of different amendments on soil carbon fractions, microbial communities, and the underlying interactive mechanisms. The experiment encompassed a no-amendment control (CK), as well as treatments with corn straw (CS), tobacco stalks (TS), and peanut shell biochar (PB). Over a 70-day incubation, the SOC in plots amended with CS, TS, and PB displayed significant boosts of 13.9%, 17.5%, and 44.8%, respectively, compared to the CK. For soil carbon fractions, amendments with PB, TS, and CS led to a dramatic rise in particulate organic carbon (POC) of 27.4%, 20.2%, and 105.7%, respectively, in contrast to the CK plots. Mantel analysis and structural Equation Modeling uncovered strong interrelationships among the cbbL, cbbM, Bacteroidota, TOC, and POC. Organic amendments enhance soil carbon fractions, modulating the microbial community by increasing Bacteroidetes abundance and suppressing Acidobacteria richness, thereby influencing the abundance of key carbon cycle genes such as cbbL and cbbM. These results suggest that the addition of peanut shell biochar significantly boosted TOC and key carbon fractions, enhancing carbon content and soil fertility.

Based on X ray diffraction and Fourier transform infrared spectroscopy, the microcrystalline structure characteristics and evolution patterns of a total of 6 coal samples from the eastern Junggar, southern Junggar, Tuha, and Kubai coalfields in Xinjiang were investigated. The peak shape of 002 and 100 peaks in low-rank coal is gentle, the peak 002 becomes sharper and the “symmetry” becomes better with the increase of coal rank, and the peak height of the peak 100 gradually increases but the peak width gradually shrinks. The height of the peak 100 gradually increases but the width gradually narrows; with the increase of coal sample metamorphism, the stacking height, elongation and quantity of aromatic laminates increase gradually, the spacing of aromatic laminates decreases on the whole, and the condensation degree of aromatic structures in coal increases gradually; the peak shape of aromatic compounds and oxygen-containing functional groups is sharp, and the peak shape of aliphatic hydrocarbons and hydroxyl functional groups is gentle, which gradually widens and slows down with the rise of coal rank; with the increase of coal rank, the aromaticity, fat chain length, branched chain degree and maturity gradually decrease, while the polycondensation degree and hydrocarbon (oil) generation ability gradually increase. The evolution process of coal is characterized by the shedding of oxygen-containing functional groups, the enrichment of aliphatic hydrocarbons, and the condensation of some aromatics.

Abstract Background Penicillium oxalicum is an important fungal agent in the composting of cattle manure, but the changes that occur in the microbial community, physicochemical factors, and potential functions of microorganisms at different time points are still unclear. To this end, the dynamic changes occurring in the microbial community and physicochemical factors and their correlations during the composting of cattle manure with Penicillium oxalicum were analysed. Results The results showed that the main phyla observed throughout the study period were Firmicutes, Actinobacteria, Proteobacteria, Bacteroidetes, Halanaerobiaeota, Apicomplexa and Ascomycota. Linear discriminant analysis effect size (LEfSe) illustrated that Chitinophagales and Eurotiomycetes were biomarker species of bacteria and eukaryote in samples from Days 40 and 35, respectively. Bacterial community composition was significantly correlated with temperature and pH, and eukaryotic microorganism community composition was significantly correlated with moisture content and NH4 +-N according to redundancy analysis (RDA). The diversity of the microbial communities changed significantly, especially that of the main pathogenic microorganisms, which showed a decreasing trend or even disappeared after composting. Conclusions In conclusion, a combination of high-throughput sequencing and physicochemical analysis was used to identify the drivers of microbial community succession and the composition of functional microbiota during cattle manure composting with Penicillium oxalicum. The results offer a theoretical framework for explaining microecological assembly during cattle manure composting with Penicillium oxalicum.

Abstract Visual adaptive devices have potential to simplify circuits and algorithms in machine vision systems to adapt and perceive images with varying brightness levels, which is however limited by sluggish adaptation process. Here, the avalanche tuning as feedforward inhibition in bionic two-dimensional (2D) transistor is proposed for fast and high-frequency visual adaptation behavior with microsecond-level accurate perception, the adaptation speed is over 104 times faster than that of human retina and reported bionic sensors. As light intensity changes, the bionic transistor spontaneously switches between avalanche and photoconductive effect, varying responsivity in both magnitude and sign (from 7.6 × 104 to −1 × 103 A/W), thereby achieving ultra-fast scotopic and photopic adaptation process of 108 and 268 μs, respectively. By further combining convolutional neural networks with avalanche-tuned bionic transistor, an adaptative machine vision is achieved with remarkable microsecond-level rapid adaptation capabilities and robust image recognition with over 98% precision in both dim and bright conditions.

Abstract Artesunate (ART), a natural product isolated from traditional Chinese plant Artemisia annua, has not been extensively explored for its anti-melanoma properties. In our study, we found that ART inhibited melanoma cell proliferation and induced melanoma cell ferroptosis. Mechanistic study revealed that ART directly targets Ido1, thereby suppressing Hic1-mediated transcription suppression of Hmox1, resulting in melanoma cell ferroptosis. In CD8+ T cells, ART does not cause cell ferroptosis due to the low expression of Hmox1. It also targets Ido1, elevating tryptophan levels, which inhibits NFATc1-mediated PD1 transcription, consequently activating CD8+ T cells. Our study uncovered a potent and synergistic anti-melanoma efficacy arising from ART-induced melanoma cell ferroptosis and concurrently enhancing CD8+ T cell-mediated immune response both in vivo and in vitro through directly targeting Ido1. Our study provides a novel mechanistic basis for the utilization of ART as an Ido1 inhibitor and application in clinical melanoma treatment.

Abstract Background Multidrug-resistant organisms (MDRO) pose a significant threat to public health. Intensive Care Units (ICU), characterized by the extensive use of antimicrobial agents and a high prevalence of bacterial resistance, are hotspots for MDRO proliferation. Timely identification of patients at high risk for MDRO can aid in curbing transmission, enhancing patient outcomes, and maintaining the cleanliness of the ICU environment. This study focused on developing a machine learning (ML) model to identify patients at risk of MDRO during the initial phase of their ICU stay. Methods Utilizing patient data from the First Medical Center of the People’s Liberation Army General Hospital (PLAGH-ICU) and the Medical Information Mart for Intensive Care (MIMIC-IV), the study analyzed variables within 24 h of ICU admission. Machine learning algorithms were applied to these datasets, emphasizing the early detection of MDRO colonization or infection. Model efficacy was evaluated by the area under the receiver operating characteristics curve (AUROC), alongside internal and external validation sets. Results The study evaluated 3,536 patients in PLAGH-ICU and 34,923 in MIMIC-IV, revealing MDRO prevalence of 11.96% and 8.81%, respectively. Significant differences in ICU and hospital stays, along with mortality rates, were observed between MDRO positive and negative patients. In the temporal validation, the PLAGH-ICU model achieved an AUROC of 0.786 [0.748, 0.825], while the MIMIC-IV model reached 0.744 [0.723, 0.766]. External validation demonstrated reduced model performance across different datasets. Key predictors included biochemical markers and the duration of pre-ICU hospital stay. Conclusions The ML models developed in this study demonstrated their capability in early identification of MDRO risks in ICU patients. Continuous refinement and validation in varied clinical contexts remain essential for future applications.

The spatiotemporal variability of chlorophyll-a (Chl-a) in the Arabian Sea (AS) and Persian Gulf (PG) has been widely studied, but long-term trends and influencing factors remain less understood due to data gaps. This study investigates Chl-a variability and trends from 2001 to 2019 using reconstructed MODIS-Terra monthly Chl-a and sea surface temperature (SST) data, employing the Data Interpolating Empirical Orthogonal Functions (DINEOF) method for high-accuracy reconstruction. Results reveal pronounced seasonal variability, with Chl-a peaks exceeding 3 mg m-3 during southwestern monsoons and ranging between 1–3 mg m-3 during northeastern monsoons, with the lowest levels in transitional months. Spatially, the highest Chl-a concentrations were observed in the western and northeastern AS, influenced by summer southwestern (SW) and winter northeastern (NE) monsoons. Trend analysis using Sen’s slope and the Mann-Kendall test indicates significant Chl-a declines (-0.002 to 0) along ASPG coasts, with slight increases (~0.005) in the southeastern AS and southern PG. Rising SST anomalies (SST_A) correlated with reduced Chl-a anomalies (Chl-a_A) in the western AS, while increased wind anomalies (Wind_A) enhanced Chl-a_A in the western AS but decreased it in the southern PG. These findings enhance our understanding of the complex environmental dynamics shaping the ASPG ecosystems.

Abstract Diabetic wounds have become a global healthcare burden owing to impaired angiogenesis and persistent infections. Extracellular vesicles (EVs) can improve diabetic wounds, though their targeting ability is limited. In this study, we investigated the performance of a novel hydrogel dressing comprised of gelatin methacryloyl, glycoengineered EVs, and polylysine in treating infected diabetic wounds. High‐throughput single‐cell RNA sequencing (scRNA‐seq) and immunofluorescence staining revealed that E‐selectin (SELE) levels were higher in diabetic wounds than in non‐diabetic wounds. Mesenchymal stromal cells (MSCs) were transfected with a lentivirus containing fucosyltransferase VII (FUT7) and a CD63‐P19‐Nluc vector to enhance the expression of sialyl Lewis X (sLeX), the ligand of E‐selectin, on the surface of EVs (s‐EVs) derived from transfected MSCs (s‐MSCs). s‐EVs can target human umbilical vein endothelial cells (HUVECs) under lipopolysaccharide stimulation and promote the function of stimulated HUVECs in vitro. To promote and sustain the release of s‐EVs, we fabricated a gelatin methacryloyl (Gel)/poly‐L‐lysine methacryloyl (PL)‐5 hydrogel with good antibacterial ability, biocompatibility and mechanical properties. In a mouse experiment, s‐EV@Gel/PL‐5 exhibited excellent angiogenesis and anti‐inflammatory abilities and further promoted the healing of infected diabetic wounds. Our findings demonstrated the potential of the s‐EV@Gel/PL‐5 hydrogel in the clinical treatment of diabetic infectious wounds.

Abstract Background The use of stem cell-derived exosomes (Exos) as therapeutic vehicles is receiving increasing attention. Exosome administration has several advantages over cell transplantation, thus making exosomes promising candidates for large-scale clinical implementation and commercialization. However, exosome extraction and purification efficiencies are relatively low, and therapeutic heterogeneity is high due to differences in culture conditions and cell viability. Therefore, in this study, we investigated a priming procedure to enhance the production and therapeutic effects of exosomes from human umbilical cord mesenchymal stem cells (hucMSCs). After preconditioning hucMSCs with agonists/inhibitors that target the Wnt/β-catenin pathway, we assessed both the production of exosomes and the therapeutic efficacy of the optimized exosomes in the context of diabetic wound healing, hoping to provide a safer, more stable and more effective option for clinical application. Results The Wnt signalling pathway agonist CHIR99021 increased exosome production by 1.5-fold without causing obvious changes in the characteristics of the hucMSCs or the size of the exosome particles. Further studies showed that CHIR99021 promoted the production of exosomes by facilitating exocytosis. This process was partly mediated by SNAP25. To further explore whether CHIR99021 changed the cargo that was loaded into the exosomes and its therapeutic effects, we performed proteomic and transcriptomic analyses of exosomes from primed and control hucMSCs. The results showed that CHIR99021 significantly upregulated the expression of proteins that are associated with cell migration and wound healing. Animal experiments confirmed that, compared to control hucMSC-derived exosomes, CHIR99021-pretreated hucMSC-derived exosomes (CHIR-Exos) significantly accelerated wound healing in diabetic mice, enhanced local collagen deposition, promoted angiogenesis, and reduced chronic inflammation. Subsequent in vitro experiments confirmed that the CHIR-Exos promoted wound healing by facilitating cell migration, inhibiting oxidative stress-induced apoptosis, and preventing cell cycle arrest. Conclusions The Wnt agonist CHIR99021 significantly increased exosome secretion by hucMSCs, which was partly mediated by SNAP25. Notably, CHIR99021 treatment also significantly increased the exosomal levels of proteins that are associated with wound healing and cell migration, resulting in enhanced acceleration of wound healing. All of these results suggested that pretreatment of hucMSCs with CHIR99021 not only promoted exosome production but also improved the exosome therapeutic efficacy, thus providing a promising option for large-scale clinical implementation and commercialization. In situ temperature monitoring has become extremely imperative in high-temperature harsh environments and polymer-derived ceramics (PDCs) as sensing materials have attracted great attention. However, the stability and oxidation/corrosion resistance of PDCs cannot be simultaneously achieved at the moment, limiting their practical application. Herein, polymer-derived SiAlBCN ceramics were synthesized via polymer conversion method under different pyrolysis temperatures. Their microstructure evolution, high temperature sensing properties, and stability were investigated in detail. The results show that the amorphous SiAlBCN phase grows more orderly and the size of the free carbon phase enlarges with the increasing temperature. The defect concentration displays a decreasing tendency. Concurrently, the SiAlBCN ceramics as sensing materials exhibit a good temperature–resistance property from roo temperature to 1100 ℃. The fabricated SiAlBCN temperature sensor possesses excellent stability, repeatability, and accuracy. Moreover, SiAlBCN ceramics exhibit distinguished oxidation/corrosion resistance after 100 h treatment at 1200 ℃ in a water/oxygen environment, which is attributed to their low corrosive rate constant (0.57 mg/(cm2·h)) and oxidative rate constant (3.43 mg2/(cm4·h)). Therefore, polymer-derived SiAlBCN ceramics as sensing materials, which possess outstanding stability and oxidation/corrosion resistance, have great potential for in-situ monitoring of extreme environmental temperatures in the future.

Mycoplasma bovis pneumonia is a highly contagious respiratory infection caused by Mycoplasma bovis. It is particularly prevalent in calves, posing a significant threat to animal health and leading to substantial economic losses. Dang-Shen-Yu-Xing decoction is often used to treat this condition in veterinary clinics. It exhibits robust anti-inflammatory effects and can alleviate pulmonary fibrosis. However, its mechanism of action remains unclear. Therefore, this study aimed to preliminarily explore the molecular mechanism of Dang-Shen-Yu-Xing decoction for treating mycoplasma pneumonia in calves through a combination of network pharmacology, molecular docking, molecular dynamics simulation methods, and experimental validation. The active components and related targets of Dang-Shen-Yu-Xing decoction were extracted from several public databases. Additionally, complex interactions between drugs and targets were explored through network topology, Gene Ontology, and Kyoto Encyclopedia of Genes and Genomes enrichment analyses. Subsequently, the binding affinity of drug to disease-related targets was verified through molecular docking and molecular dynamics simulation. Finally, the pharmacodynamics were verified via animal experiments. The primary network topology analysis revealed two core targets and 10 key active components of Dang-Shen-Yu-Xing decoction against Mycoplasma bovis pneumonia. Kyoto Encyclopedia of Genes and Genomes enrichment analysis showed that the mechanism of Dang-Shen-Yu-Xing decoction for treating mycoplasma bovis pneumonia involved multiple signaling pathways, with the main pathways including PI3K-Akt and IL17 signaling pathways. Moreover, molecular docking predicted the binding affinity and conformation of the core targets of Dang-Shen-Yu-Xing decoction, IL6, and IL10, with the associated main active ingredients. The results showed a strong binding of the active ingredients to the hub target. Further, molecular docking dynamics simulation revealed three key active components of IL10 induced by Dang-Shen-Yu-Xing decoction against Mycoplasma bovis pneumonia. Finally, animal experiments confirmed Dang-Shen-Yu-Xing decoction pharmacodynamics, suggesting that it holds potential as an alternative therapy for treating mycoplasma bovis pneumonia.

Background: Yinqiaosan decoction (YQSD), a traditional Chinese medicinal recipe, has been employed to treat influenza in China for approximately 300 years. Objective: Our study aimed to explore the mechanisms of YQSD against influenza via in vivo and in vitro experimental studies. Study design: and methods UHPLC-Q-TOF-MS/MS was utilized to examine the substances of the YQSD. The chemical components of YQSD detected by UHPLC-Q-TOF-MS/MS were used for network pharmacology analysis. The antiviral effect of YQSD in vivo was investigated. The potential mechanisms of YQSD in combating influenza, which were predicted from network pharmacology analysis, were validated in vitro. Results: By use of UHPLC-Q-TOF-MS/MS, 97 compounds were identified from YQSD. Network pharmacology analysis revealed that the therapeutic effect of YQSD against influenza may be associated with the regulation of T cell receptors (TCR) and Phosphoinositide 3-Kinase (PI3K)- protein kinase B (Akt) signaling pathways. Treatment with YQSD significantly prolonged the mean survival time of the mice and reduced lung injury due to the influenza A virus in vivo. It was discovered that YQSD efficiently inhibited the expression of inflammation-related cytokines. Moreover, YQSD has been found to significantly reduce the expression levels of cluster of differentiation 3 (CD3), monocyte chemoattractant protein-1 (MCP-1), and H1N1 virus nucleoprotein (NP), and prevent the decrease of epithelial cadherin (E-cadherin) protein. In addition, YQSD can inhibit the phosphorylation of the zeta chain of T cell receptor-associated protein kinase 70 (ZAP70) and PI3K proteins in vitro. Conclusion: The capacity of YQSD to suppress viral multiplication and inflammatory response by modulating T cell immunity may explain its effect against influenza viral pneumonia, which may involve the regulation of TCR and PI3K signaling pathways.

Flash sintering (FS) is a novel technique for rapidly densifying silicon carbide (SiC) ceramics. This work achieved a rapid sintering of SiC ceramics by the utilization of ultra-high temperature flash sintering within 60 s. Pyrolysis carbon (PyC) “bridges” were constructed between SiC particles through the carbonisation of phenolic resin, providing a large number of current channels. The incubation time of the flash sintering process was significantly reduced, and the sintering difference between the centre and the edge regions of the ceramics was minimized, with an average particle size of the centre region and edge region being 12.31 and 9.02 μm, respectively. The results showed that the porosity of the SiC ceramics after the flash sintering was reduced to 14.79% with PyC “bridges” introduced, and the Vickers hardness reached 19.62 GPa. PyC “bridges” gradually evolved from amorphous eddy current carbon to oriented graphite carbon, indicating that the ultra-high temperature environment in which the sample was located during the flash sintering was successfully constructed. Ultra-high temperature flash sintering of SiC is expected to be applied to the local repair of matrix damage in SiC ceramic matrix composites.

Abstract Purpose Prolonged exposure to plasma free fatty acids (FFAs) leads to impaired glucose tolerance (IGT) which can progress to type 2 diabetes (T2D) in the absence of timely and effective interventions. High-fat diet (HFD) leads to chronic inflammation and oxidative stress, impairing pancreatic beta cell (PBC) function. While Didymin, a flavonoid glycoside derived from citrus fruits, has beneficial effects on inflammation dysfunction, its specific role in HFD-induced IGT remains yet to be elucidated. Hence, this study aims to investigate the protective effects of Didymin on PBCs. Methods HFD-induced IGT mice and INS-1 cells were used to explore the effect and mechanism of Didymin in alleviating IGT. Serum glucose and insulin levels were measured during the glucose tolerance and insulin tolerance tests to evaluate PBC function and insulin resistance. Next, RNA-seq analysis was performed to identify the pathways potentially influenced by Didymin in PBCs. Furthermore, we validated the effects of Didymin both in vitro and in vivo. Mitochondrial electron transport inhibitor (Rotenone) was used to further confirm that Didymin exerts its ameliorative effect by enhancing mitochondria function. Results Didymin reduces postprandial glycemia and enhances 30-minute postprandial insulin levels in IGT mice. Moreover, Didymin was found to enhance mitochondria biogenesis and function, regulate insulin secretion, and alleviate inflammation and apoptosis. However, these effects were abrogated with the treatment of Rotenone, indicating that Didymin exerts its ameliorative effect by enhancing mitochondria function. Conclusions Didymin exhibits therapeutic potential in the treatment of HFD-induced IGT. This beneficial effect is attributed to the amelioration of PBC dysfunction through improved mitochondrial function.

Apples are one of the most popular fruits in the world and have a significant share in domestic and international fruit production. Drying is a common method used to extend the shelf life of apples. However, it also induces irregular morphological changes in apples, which are essential to maintaining the structural integrity of the material. Therefore, it is necessary to understand the effect of cellular changes at the microscopic level on the macroscopic deformation of the material during drying. In this paper, the evolution of cell wall pectin fractions and viscoelastic properties of apples during freeze drying combined with hot air drying was investigated. The findings indicated that during the HAD stage, a decrease in the relaxation modulus (E1) of the samples was observed in the compression tests when the sample temperature was significantly higher than the glass transition temperature (Tg). As the difference between the two decreased, the samples exhibited increased stiffness and higher E1. The results of the pectin content analysis showed that the HAD process accelerated the loss and degradation of water-soluble pectin in the samples with high moisture content at the transition point. Simultaneously, the esterification degree of chelator-soluble pectin increased, leading to a reduction in the support provided to the cellular structure of the samples, which consequently affected their mechanical properties. These findings may provide valuable information for the application of freeze drying combined with hot air drying in the efficient processing of dried fruit and vegetable products.

Mycoplasma bovis is a global concern for the cattle industry owing to its high rates of infection and resulting morbidity, but there are no reports on the lipid composition and metabolic pathways. This study examined the lipidomics of M. bovis to better investigate the biological characteristics of clinical isolates of M. bovis. The M. bovis strains NX114 and PG45, cultivated to log-phase, underwent untargeted lipidomics via ultra-high-performance liquid chromatography-mass spectrometry for differential analysis. Over 65 lipid types and 1476 lipid molecules were identified. We found that glycerophospholipids and sphingolipids constitute the predominant lipid components of M. bovis, with significant constituents of its cell membrane comprising ceramides, phosphatidylglycerols, sphingomyelins, triacylglycerols, hexosylceramides, ether-linked oxidized phosphatidylcholines, and oxidized phosphatidylglycerols. Secondly, 562 differentially expressed lipid molecules were identified, including 17 lipid subclasses (15 up-regulated and 2 down-regulated) with significant differences in relative content. These findings indicate that distinct M. bovis isolates contain structurally varied lipid molecules, with sphingomyelin, phosphatidylinositol, cardiolipin, and phosphatidylcholine being characteristic lipids. The glycerophospholipid metabolism pathway was identified as a key pathway regulating lipid metabolism in M. bovis by KEGG pathway enrichment analysis. The results indicated alterations in the lipid metabolism of M. bovis, offering insights into its pathogenic mechanisms.

Mycoplasma bovis is a global problem for the cattle industry due to its high infection rates and associated morbidity, although its pathophysiology is poorly understood. In this study, the M. bovis transcriptome and proteome were analyzed to further investigate the biology of clinical isolates of M. bovis. A differential analysis of M. bovis, a clinical isolate (NX114), and an international type strain (PG45) at the logarithmic stage of growth, was carried out using prokaryotic transcriptome and 4D-label-free quantitative non-labeled proteomics. Transcriptomics and proteomics identified 193 DEGs and 158 DEPs, respectively, with significant differences in 49 proteins/34 transcriptomic CDS post-translational protein sequences (15 jointly up-regulated and 21 jointly down-regulated). GO comments indicate membrane, cytoplasmic and ribosome proteins were important components of the total proteins of M. bovis NX114 clinical isolate. KEGG enrichment revealed that M. bovis NX114 is mainly associated with energy metabolism, the biosynthesis of secondary metabolites, and the ABC transporters system. In addition, we annotated a novel adhesion protein that may be closely related to M. bovis infection. Triosephosphate isomerase (TpiA) and Pyruvate kinase (Pyk) genes may be the key enzymes that regulate the growth and maintenance of M. bovis and are involved in the pathogenic process as virulence factors. The results of the study revealed the biology of different isolates of M. bovis and may provide research ideas for the pathogenic mechanism of M. bovis.

BackgroundIn recent epochs, the field of critical medicine has experienced significant advancements due to the integration of artificial intelligence (AI). Specifically, AI robots have evolved from theoretical concepts to being actively implemented in clinical trials and applications. The intensive care unit (ICU), known for its reliance on a vast amount of medical information, presents a promising avenue for the deployment of robotic AI, anticipated to bring substantial improvements to patient care. ObjectiveThis review aims to comprehensively summarize the current state of AI robots in the field of critical care by searching for previous studies, developments, and applications of AI robots related to ICU wards. In addition, it seeks to address the ethical challenges arising from their use, including concerns related to safety, patient privacy, responsibility delineation, and cost-benefit analysis. MethodsFollowing the scoping review framework proposed by Arksey and O’Malley and the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) guidelines, we conducted a scoping review to delineate the breadth of research in this field of AI robots in ICU and reported the findings. The literature search was carried out on May 1, 2023, across 3 databases: PubMed, Embase, and the IEEE Xplore Digital Library. Eligible publications were initially screened based on their titles and abstracts. Publications that passed the preliminary screening underwent a comprehensive review. Various research characteristics were extracted, summarized, and analyzed from the final publications. ResultsOf the 5908 publications screened, 77 (1.3%) underwent a full review. These studies collectively spanned 21 ICU robotics projects, encompassing their system development and testing, clinical trials, and approval processes. Upon an expert-reviewed classification framework, these were categorized into 5 main types: therapeutic assistance robots, nursing assistance robots, rehabilitation assistance robots, telepresence robots, and logistics and disinfection robots. Most of these are already widely deployed and commercialized in ICUs, although a select few remain under testing. All robotic systems and tools are engineered to deliver more personalized, convenient, and intelligent medical services to patients in the ICU, concurrently aiming to reduce the substantial workload on ICU medical staff and promote therapeutic and care procedures. This review further explored the prevailing challenges, particularly focusing on ethical and safety concerns, proposing viable solutions or methodologies, and illustrating the prospective capabilities and potential of AI-driven robotic technologies in the ICU environment. Ultimately, we foresee a pivotal role for robots in a future scenario of a fully automated continuum from admission to discharge within the ICU. ConclusionsThis review highlights the potential of AI robots to transform ICU care by improving patient treatment, support, and rehabilitation processes. However, it also recognizes the ethical complexities and operational challenges that come with their implementation, offering possible solutions for future development and optimization.

With the development of information technology, a mass of data are generated every day. Collecting and analysing these data help service providers improve their services and gain an advantage in the fierce market competition. K-means clustering has been widely used for cluster analysis in real life. However, these analyses are based on users’ data, which disclose users’ privacy. Local differential privacy has attracted lots of attention recently due to its strong privacy guarantee and has been applied for clustering analysis. However, existing K-means clustering methods with local differential privacy protection cannot get an ideal clustering result due to the large amount of noise introduced to the whole dataset to ensure the privacy guarantee. To solve this problem, we propose a novel method that provides local distance privacy for users who participate in the clustering analysis. Instead of making the users’ records in-distinguish from each other in high-dimensional space, we map the user’s record into a one-dimensional distance space and make the records in such a distance space not be distinguished from each other. To be specific, we generate a noisy distance first and then synthesize the high-dimensional data record. We propose a Bounded Laplace Method (BLM) and a Cluster Indistinguishable Method (CIM) to sample such a noisy distance, which satisfies the local differential privacy guarantee and local dE-privacy guarantee, respectively. Furthermore, we introduce a way to generate synthetic data records in high-dimensional space. Our experimental evaluation results show that our methods outperform the traditional methods significantly.

Abstract Ionizing radiation (IR) causes a wide variety of DNA lesions, of which DNA double-stranded breaks (DSBs) are the most deleterious. Homologous recombination (HR) is a crucial route responsible for repairing DSBs. RecQ-mediated genome instability protein 1 (RMI1) is a member of an evolutionarily conserved Bloom syndrome complex, which prevents and resolves aberrant recombination products during HR, thereby promoting genome stability. However, little is known about the role of RMI1 in regulating the cellular response to IR. This study aimed to understand the cellular functions and molecular mechanisms by which RMI1 maintains genomic stability after IR exposure. Here, we showed IR upregulated the RMI1 protein level and induced RMI1 relocation to the DNA damage sites. We also demonstrated that the loss of RMI1 in cells resulted in enhanced levels of DNA damage, sustained cell cycle arrest, and impaired HR repair after IR, leading to reduced cell viability and elevated genome instability. Taken together, our results highlighted the direct roles of RMI1 in response to DNA damage induced by IR and implied that RMI1 might be a new genome safeguard molecule to radiation-induced damage.

Abstract Metabolic dysfunction-associated fatty liver disease (MAFLD) is frequently linked to type 2 diabetes mellitus (T2DM), and both conditions exacerbate the progression of the other. However, there is currently no standardized treatment or drug for MAFLD. In this study, A MAFLD animal model through a high-fat diet (HFD) along with administration of streptozotocin (STZ), and palmitic acid (PA)-induced AML12 cells were treated by puerarin. The objective of this study was to assess the therapeutic effect of puerarin, a flavonoid substance that possesses various pharmacological properties, on MAFLD. The results showed that puerarin administration enhanced glucose tolerance and insulin sensitivity, while also mitigating liver dysfunction and hyperlipidemia in MAFLD mice. Moreover, puerarin attenuated oxidative stress levels and inflammation in the liver. Transmission electron microscopy and Western blot analysis indicated that puerarin inhibited ferroptosis in vivo. Further mechanistic investigations revealed that puerarin upregulated SIRT1 expression, increased nuclear factor erythroid 2-related factor 2 (Nrf2) protein levels, and facilitated translocation into the nucleus. The protective effect of puerarin on PA-induced AML12 cells was diminished by the utilization of EX-527 (a SIRT1 inhibitor) and Nrf2 siRNA. Overall, the results demonstrate that puerarin ameliorates MAFLD by suppressing ferroptosis and inflammation via the SIRT1/Nrf2 signaling pathway. The results emphasize the possible medicinal application of puerarin for managing MAFLD.

Abstract The preoperative diagnosis of thyroid nodules now routinely includes BRAF V600E genetic testing in most provincial and municipal hospitals in China. This study identified the most suitable patients of thyroid nodule for BRAFV600E genetic testing. We retrospectively collected data of patients from the Hospital Information System that had undergone fine needle aspiration biopsy (FNAB) from May 2019 to December 2021. Data of FNAB, BRAF V600E genetic testing, and post-surgical pathological diagnosis were collected. A total of 12,392 patients were included in this study. Among them, 7,010 patients underwent solely FNAB, while 5,382 patients had both FNAB and BRAF V600E genetic testing. In the FNAB group, 2,065 thyroid nodules were surgically removed, with a 93.12% malignancy rate. In the FNAB + BRAF group, 2,005 nodules were dissected, and the malignancy rate was 98.20%. The continuous deepening of coal-seam extraction has sharply increased both gas pressure and content. The use of viscoelastic surfactant fracturing fluids (VESFFs) has been demonstrated to effectively improve coal-seam permeability and mitigate the occurrence of gas disasters. After injection into coal, VESFFs interact with the coal and affect its surface characteristics. In this study, to characterize changes in zeta potential, oxygen-containing functional groups, and the microcrystalline structure of hard and soft coal surfaces under the influence of VESFFs with different formulations, zeta potential measurements and Fourier-transform infrared and Raman spectroscopies were performed. The VESFFs enhanced the electrostatic repulsion between the pore wall and coal particles, which is favorable for the removal of coal particles from hard and soft coal surfaces. The combination of cationic with zwitterionic viscoelastic surfactants (VESs) in the VESFFs exposed more hydrophilic functional groups on the surfaces of hard and soft coal, increasing wettability and affecting nanometer pores. A VESFF based on anionic and zwitterionic VESs as the primary agents could enhance the extension of the aromatic layer (La) of the aromatic crystal nuclei and reduce the interlayer spacing (d002) of hard and soft coal, thereby increasing the volume of micropores. This research offers theoretical guidance for optimizing the primary components of VESFFs and elucidates the mechanism through which VESFFs act on nanopores in coal from a microscopic perspective.

Abstract Soil bacterial communities regulate nutrient cycling and plant growth in forests. Although these bacterial communities vary with soil nutrients and plant traits, the variation and degree with soil horizons in different forest types remain unclear. Here, bacterial communities of 44 soil samples from organic horizon (O horizon) and mineral horizon (M horizon) of three forest types (Cunninghamia, broad-leaved and Pinus forests) in subtropical forests of Dabie Mountain, China were analyzed based on amplicon sequencing. We assessed the effects of soil horizons and forest types on bacterial communities. The results showed that the bacterial richness and diversity were significantly higher in the O horizon than in the M horizon. Furthermore, the bacterial community composition and functions were also remarkably different between the two soil horizons. Furthermore, forest types could affect bacterial community composition but not for diversity and functions. Moreover, soil organic matter, including the total organic carbon, available phosphorus, total organic nitrogen, available potassium, ammonium nitrogen, and pH were main drivers for bacterial community composition. The results propose robust evidence that soil horizons strongly driven bacterial community composition and diversity, and suggest that microhabitat of soil bacterial communities is important to maintain the stability of forest ecosystem.

This paper discusses the differences in the microstructure, mechanical properties, wear resistance, and corrosion behavior of nickel-aluminum bronze (NAB) alloys fabricated via wire arc additive manufacturing (WAAM) versus casting. The grain size of the WAAM-NAB alloy is smaller than that of the as-cast NAB alloys; furthermore, the precipitation of the large size κⅠ phase is completely suppressed while the remaining precipitated κ phases are limited and reduced in size. Compared with the cast alloy, the WAAM-NAB alloy indicates a higher bending and tensile strength by 46% and 18%, respectively. The coefficient of friction of WAAM-NAB alloy is 56% lower than cast alloys. Additionally, the WAAM-NAB alloy shows greater corrosion resistance against seawater, in that it only exhibited residual β phase corrosion, whereas the cast alloy sustained severe selective-phase corrosion.

By employing high-throughput first-principles calculations, the segregation capacity of fifteen widely used metallic alloying elements (viz., Be, Mn, Co, Cr, Ni, Al, Mo, W, Mg, Ta, Nb, Sb, Sn, Zr, and Bi) at ∑3 grain boundary in low alloy ferritic steel, as well as their impact on grain boundary stability, interfacial separation work, and other properties, were systematically investigated. The findings reveal that, for alloying atoms Sb, Sn, Bi, Nb, and Zr, whose size is notably larger than that of the matrix Fe atoms, the effect of strain energy minimization in segregation is comparable to that of chemical energy minimization. Furthermore, the impact of strain energy minimization is closely related to the volume of the alloying atoms both at the solid solution sites in the crystal and at the segregation sites at the grain boundary. Thus, the segregation of large alloy atoms on the grain boundaries can be predicted by atomic volume of each segregation site, which can provide valuable insights for the development of new alloys and for grain boundary engineering.

Abstract During the processing of tobacco leaves, flue-curing and redrying can affect the structure of bacterial community, having an effect on the aging quality of tobacco leaves. In order to characterize the effects of flue-curing and redrying on the bacterial community of tobacco leaves, the bacterial community of samples at different processing stages (before flue-curing, after flue-curing, before redrying and after redrying) was analyzed using Illumina sequencing. A total of 33 phyla, 79 classes, 195 orders, 344 families, 826 genera and 7922 ASVs were obtained from 36 samples. There was no significant difference in the core bacterial groups of tobacco leaf at four processing stages. Proteobacteria dominated at the phylum level. Sphingomonas, Pseudomonas and Methylobacterium were the main genera shared by all samples. The functional prediction by PICRUSt showed an increase in the relative abundance of pathway related to metabolism after flue-curing and pathway related to environmental information processing after redrying. This study, we analyzed the changes of bacterial community and structural composition of tobacco leaves from flue-curing to redrying, and found that flue-curing had a greater effect on the microbial community than redrying. This is conducive for the exploration of microbial resources and improvement of tobacco leaf quality.

Abstract Microorganisms play an important role in the tobacco aging process. Before the aging process, raw tobacco leaves must be threshed and redried. In order to explore the differences of microbial community structure of threshed and redried tobacco leaves from different origins at home and abroad, 14 groups of tobacco leaves from 8 different countries were tested by high-throughput DNA sequencing and microbiology analysis. Then, through amplicon sequence variants (ASV) cluster analysis, Venn diagram and species labeling and other microbial diversity analysis, the dominant bacteria and fungi on the surface of threshed and redried tobacco leaves were obtained. The results showed that there were significant differences in the composition of tobacco bacteria and fungi after threshing and redrying from different geographical areas. The relative abundance of Microbacterium and Sphingomonas in domestic tobacco leaves was significantly higher than that of foreign tobacco leaves. The relative abundance of Pseudomonas in foreign tobacco bacterial colonies was significantly higher than that of domestic tobacco leaves. In terms of fungi, the relative abundance of Aspergillus and Alternaria in domestic tobacco leaves was significantly higher than that of foreign tobacco leaves. Septoria, Sampaiozyma, Cladosporium and Phoma account for significantly higher proportions of foreign tobacco leaves. These microorganisms may be indispensable in aging process to form different flavors of tobacco leaves. It provides an important theoretical basis for the further use of microorganisms to promote tobacco leaf aging.

In this work, the SiC ceramics were fabricated by stereolithography (SLA) combined with reactive melt infiltration (RMI), and the microstructure and mechanical properties of SiC ceramics were optimized by adjusting the solid content and impregnating phenolic resin. The bulk density and bending strength of SiC ceramics were positively correlated with the proportion of SiC content. The SiC phase content was increased from 48.10% to 61.42% with the solid content increased from 30 vol% to 45 vol%. When the solid content was 40 vol%, the bulk density and bending strength of SiC ceramics attained the highest value of 2.85 g/cm3 and 234.8 MPa. Meanwhile, the pyrolysis shrinkage of SiC ceramics along the non-additive direction and the additive direction was 4.83% and 1.21%, respectively. This research has successfully demonstrated the direct correlation of microstructure and content of SiC with the performance parameters of SiC ceramics fabricated by SLA.

Abstract In recent years, 2D reconfigurable phototransistors (RPTs) have been applied in broadband convolutional processing, retinomorphic hardware devices, and non‐volatile memorizers. However, there has been a lack of investigation into all‐2D Schottky junctions used in RPT with polarity control behavior. Herein, a vertically stacked multilayered WS2/WTe2 Schottky RPT is reported. The semimetal characteristics of 1T’‐WTe2 is designed to form a built‐in electric field of 69 meV across the heterojunction and WS2 exhibits gate‐tunable characteristics. Therefore, reconfigurable rectifying behavior and self‐driven bidirectional photo response can be achieved. The phototransistor possesses a gate‐tunable rectification ratio ranging from 10−2 to 105, and the corresponding logic half‐wave rectifier shows excellent switchable rectifying states. Under 635 nm illumination, the responsivity can be adjusted from −1325 to 430 mA W−1 with reversed signs. Meanwhile, the maximum power conversion efficiency is 2.84%, and the specific detectivity is 1.47 × 1012 Jones. The device shows both negative and positive responsivity with linear gate dependence within a voltage window of 10 V. Impressively, nonvolatile photovoltaic performance can be demonstrated by reversing short‐circuit current and open‐circuit voltage by applying and releasing pulsed gate voltage. Finally, reconfigurable polarization behavior, single‐pixel imaging, and the optical logic circuit are applicable to the heterostructure.

Abstract Background Continuation of bevacizumab plus second‐line chemotherapy has significantly improved overall and progression‐free survival in patients with metastatic colorectal cancer (mCRC). However, the cost‐effectiveness of such high cost therapy is still uncertain in China; so this analysis was performed to evaluate the cost‐effectiveness of these treatment options from the Chinese health care system perspective. Methods A cost‐effectiveness analysis was conducted using data from the ML18147 trial (ClinicalTrials.gov identifier NCT00700102) by modeling a partitioned survival model. Main evaluation indicators were quality‐adjusted life years (QALYs) and incremental cost‐effectiveness ratio (ICER) with a willingness to pay (WTP) threshold of $38,201 per QALY. One‐way and probabilistic sensitivity analyses were conducted to assess the robustness and stability of the model. Subgroup and scenario analyses were also performed to make our study more relevant. Results Bevacizumab plus chemotherapy increased 0.12 QALYs and an incremental cost of $22,761.62 compared with chemotherapy, resulting in an ICER of $188,904.09 per QALY. The model was most sensitive to the utility of progression‐free survival and the cost of bevacizumab. Compared with chemotherapy, bevacizumab plus chemotherapy had a 0% cost‐effectiveness probability, and no cost‐effectiveness in subgroups at the WTP threshold of $38,201 per QALY. The scenario analysis found that bevacizumab biosimilar gained an ICER of $126,397.38 per QALY when assuming the cost of drugs was calculated at the most affordable price. Conclusions At the WTP threshold of $38,201 per QALY, continuation of bevacizumab plus chemotherapy is unlikely considered cost‐effective for patients after first progression of mCRC.

Aerogels have an impressive thermal insulation performance at room temperature. However, with high infrared transmittance, pure aerogels have no advantages in thermal insulation at high temperature. Opacifiers can reduce infrared transmittance to make a low thermal conductivity with sacrifice of that at room temperature. Herein, VO2 powders undergoing reversible insulator to metal transition at 67.8 °C as opacifier with temperature-switch were introduced into silica aerogel composites to prepare aerogel reinforced by VO2@SiO2 fibers paper (AVSP). At 25 °C, insulator VO2 made little contribution to thermal conductivity (0.025 W/(m·K)) so that it was close to aerogel reinforced by pure SiO2 fibers papers without opacifier (0.023 W/(m·K)). When temperature rose to 500 °C, metal VO2 made thermal conductivity of AVSP (0.051 W/(m·K)) much lower than that of aerogel reinforced by pure SiO2 fibers papers without opacifier (0.076 W/(m·K)). Meanwhile, SiO2 fibers papers with three-dimensional network structure improved flexibility of aerogel. The tensile strength of AVSP was 2.1 MPa with density of 0.186 g/cm3. With 152° contact angle and flame resistance, AVSP showed a great potential in high temperature thermal insulation in a nonplanar condition. Thereby, this work provides a novel strategy to improve thermal insulation property at high temperature with little contribution to that at room temperature.

Abstract Background Diabetes and obesity are associated with muscle atrophy that reduces life quality and lacks effective treatment. Mesenchymal stromal cell (MSC)‐based therapy can ameliorate high fat‐diet (HFD) and immobilization (IM)‐induced muscle atrophy in mice. However, the effect of MSCs on muscle atrophy in type 2 diabetes mellitus (T2DM) and the potential mechanism is unclear. Here, we evaluated the efficacy and explored molecular mechanisms of human umbilical cord MSCs (hucMSCs) and hucMSC‐derived exosomes (MSC‐EXO) on diabetes‐ and obesity‐induced muscle atrophy. Methods Diabetic db/db mice, mice fed with high‐fat diet (HFD), mice with hindlimb immobilization (IM), and C2C12 myotubes were used to explore the effect of hucMSCs or MSC‐EXO in alleviating muscle atrophy. Grip strength test and treadmill running were used to measure skeletal muscle strength and performance. Body composition, muscle weight, and muscle fibre cross‐sectional area (CSA) was used to evaluate muscle mass. RNA‐seq analysis of tibialis anterior (TA) muscle and Western blot analysis of muscle atrophy signalling, including MuRF1 and Atrogin 1, were performed to investigate the underlying mechanisms. Results hucMSCs increased grip strength (P = 0.0256 in db/db mice, P = 0.012 in HFD mice, P = 0.0097 in IM mice), running endurance (P = 0.0154 in HFD mice, P = 0.0006 in IM mice), and muscle mass (P = 0.0004 in db/db mice, P = 0.0076 in HFD mice, P = 0.0144 in IM mice) in all models tested, with elevated CSA of muscle fibres (P

Abstract Background Malaria is still one of the major infectious diseases affecting human health, and the World Health Organization (WHO) has attached special importance to malaria-related technical training for its global elimination efforts. The Jiangsu Institute of Parasitic Diseases (JIPD), designated as a WHO Collaborating Centre for Research and Training on Malaria Elimination, has conducted numerous international malaria training programmes during the last 2 decades. Methods A retrospective analysis of international training programmes organized and facilitated by JIPD in China since 2002 was conducted. A web-based questionnaire was designed to gather respondents’ basic information, evaluation of course topics, methodology, trainers, and facilitators, course impact, and suggestions for future trainings. Individuals who participated in the training courses from 2017 to 2019 were invited to participate in this assessment. Results Since 2002, JIPD has conducted 62 malaria-related international trainings attended by 1935 participants from 85 countries, covering 73% of malaria endemic countries. Of 752 participants enrolled, 170 responded to the online survey. A majority of respondents (160/170, 94.12%) gave a high evaluation of the training, with an average score of 4.52 (5 maximum score). Also, survey respondents gave a 4.28 score on “knowledge and skills gained in the training useful for the national malaria programme”, 4.52 on “topics appropriate to their professional needs”, and 4.52 on “knowledge and skills gained in the training useful to their career”. Surveillance and response was the most important topic discussed and field visit was the most effective method of training. For future training programmes, with increasing length of training, more field visits and demonstration, improving language barrier, and sharing experience were what the respondents requested most. Conclusion JIPD, as a professional institute for malaria control, has conducted a great quantity of training in the past 20 years, providing training opportunities to both malaria and non-malaria endemic countries globally. For future training, survey respondents’ suggestions will be considered to provide a more effective capacity building activity to better contribute to global malaria elimination.

Abstract Background Imported malaria cases remains a key health concern, especially during the COVID-19 pandemic. Providing accurate health information is important to improving people’s awareness of malaria. WeChat is an excellent social media tool for health information dissemination, especially during the pandemic. This study explored the effect of malaria knowledge dissemination via a WeChat public account. Methods A questionnaire for data collection was constructed using the online survey tool Sojump. Questionnaires were sent to users who followed the Jiangsu institute of Parasitic Disease WeChat public account during the National Malaria Day 2021 period. A small incentive (WeChat Red Packet) was distributed to everyone who answered the questionnaire correctly on time. Results A total of 13,169 valid questionnaires were collected during the China National Malaria Day period. Questions in which participants focused mainly on information pertaining to themselves, such as infection, symptoms, and epidemic areas, reached highest accuracy (above 90%). Questionnaires were submitted through smartphones and most of them were completed during the period of 4 days from April 23 to April 26, 2021 when a WeChat Red Packet was offered. The accuracy of responses was related to bolded words and location and number of knowledge points that were shown at the beginning of the questionnaire. The number of users of the WeChat public account in question increased from 5961 to 12,339 in just 4 days of the activity. Conclusion A WeChat public account is a convenient and accessible tool for spreading malaria-related health information to the public. Distribution of incentives (Red Packets) can effectively increase public attention to popular science and health information and activities.

Abstract Background Coronavirus disease 2019 (COVID-19) caused a series of biological changes in cancer patients which have rendered the original treatment ineffective and increased the difficulty of clinical treatment. However, the clinical treatment for cancer patients infected with COVID-19 is currently unavailable. Since bioinformatics is an effective method to understand undiscovered biological functions, pharmacological targets, and therapeutic mechanisms. The aim of this study was to investigate the influence of COVID-19 infection in cancer patients and to search the potential treatments. Methods Firstly, we obtained the COVID-19-associated genes from seven databases and analyzed the cancer pathogenic genes from Gene Expression Omnibus (GEO) databases, respectively. The Cancer/COVID-19-associated genes were shown by Venn analyses. Moreover, we demonstrated the signaling pathways and biological functions of pathogenic genes in Cancer/COVID-19. Results We identified that Go-Ichi-Ni-San complex subunit 1 (GINS1) is the potential therapeutic target in Cancer/COVID-19 by GEPIA. The high expression of GINS1 was not only promoting the development of cancers but also affecting their prognosis. Furthermore, eight potential compounds of Cancer/COVID-19 were identified from CMap and molecular docking analysis. Conclusion We revealed the GINS1 is a potential therapeutic target in cancer patients infected with COVID-19 for the first time, as COVID-19 will be a severe and prolonged pandemic. However, the findings have not been verified actually cancer patients infected with COVID-19, and further studies are needed to demonstrate the functions of GINS1 and the clinical treatment of the compounds.

The creation of van der Waals (vdW) ferromagnets with tunable Curie temperature (TC) and magnetic anisotropy is essential in developing vdW magnet-based devices. Here, we report an effective and reliable method for modulating the magnetic properties of vdW Fe5GeTe2 by site-specific Ga+ implantation. In this study, we report an easy axis in the ab-plane for bulk Fe5GeTe2 (TC = 310 K) and an axis out of the plane for thin Fe5GeTe2 flakes (TC = 290 K). Combining element-resolved photoemission electron microscopy and spatially resolved magneto-optic Kerr microscopy, we find that the implantation of a tiny amount of 10−3 Ga+·Å−3 in Fe5GeTe2 greatly enhances the TC from 290 to 360 K and switches the magnetic easy axis from the out-of-plane c axis to the ab-plane. The room-temperature x-ray magnetic circular dichroism signal is enhanced from 0% to 9% at an implantation level of 10−2 Ga+·Å−3. These results provide new opportunities for tailoring the magnetic properties of vdW materials beyond room temperature.

Background: Regular physical activity is an effective means to enhance university students’ subjective well-being. However, current research needs to understand how physical activity enhances the subjective well-being of Chinese university students. Therefore, the study investigated the mechanism of physical activity’s impact on university students’ subjective well-being and the mediating roles of cognitive reappraisal and resilience in this mechanism. Methods: The physical activity scale, subjective well-being scale, cognitive reappraisal scale and resilience scale were used to investigate 1350 university students, and the relationship between physical activity, cognitive reappraisal, resilience and university students’ subjective well-being was verified through correlation analysis, regression analysis and a Bootstrap method. Results: (1) There is a significant positive correlation between physical activity, cognitive reappraisal, resilience and university students’ subjective well-being (p < 0.01); (2) physical activity, cognitive reappraisal and resilience all have a significant positive effects on university students’ subjective well-being (p < 0.01); (3) cognitive reappraisal and resilience have significant mediating roles in the process of physical activity affecting university students’ well-being, with mediating-effect values of 0.052 and 0.285; (4) the chain-mediating role of cognitive reappraisal and resilience in the process of physical activity affecting university students’ well-being is significant, with the chain-mediating effect value of 0.062. Conclusion: Promoting university students’ participation in physical activity not only directly enhances university students’ subjective well-being but also indirectly improves university students’ subjective well-being through cognitive reappraisal and resilience.

Salt stress is one of the most important factors limiting rice growth and yield increase. Salt tolerance of rice at the bud burst (STB) stage determines whether germinated seeds can grow normally under salt stress, which is very important for direct seeding. However, reports on quantitative trait loci (QTLs) and candidate genes for STB in rice are very limited. In this study, a natural population of 130 indica and 81 japonica rice accessions was used to identify STB-related QTLs and candidate genes using a genome-wide association study (GWAS). Nine QTLs, including five for relative shoot length (RSL), two for relative root length (RRL), and two for relative root number (RRN), were identified. Five of these STB-related QTLs are located at the same site as the characterized salt tolerance genes, such as OsMDH1, OsSRFP1, and OsCDPK7. However, an important QTL related to RSL, qRSL1-2, has not been previously identified and was detected on chromosome 1. The candidate region for qRSL1-2 was identified by linkage disequilibrium analysis, 18 genes were found to have altered expression levels under salt stress through the RNA-seq database, and 10 of them were found to be highly expressed in the shoot. It was also found that, eight candidate genes (LOC_Os01g62980, LOC_Os01g63190, LOC_Os01g63230, LOC_Os01g63280, LOC_Os01g63400, LOC_Os01g63460, and LOC_Os01g63580) for qRSL1-2 carry different haplotypes between indica and japonica rice, which exactly corresponds to the significant difference in RSL values between indica and japonica rice in this study. Most of the accessions with elite haplotypes were indica rice, which had higher RSL values. These genes with indica-japonica specific haplotypes were identified as candidate genes. Rice accessions with elite haplotypes could be used as important resources for direct seeding. This study also provides new insights into the genetic mechanism of STB.

Abstract This study investigates the effect of microstructure on short-term and long-term oxidation behaviours of GH3536 superalloy fabricated by laser powder bed fusion (LPBF), in which the superalloy is isothermally oxidised at 950 °C for 6 h and 500 h in air. The LPBF sample exhibits improved oxidation resistance compared with a wrought counterpart after long-term exposure. The effect of microstructure diversity between LPBF and wrought samples on oxidation behaviour is discussed. The cellular structure produced during the LPBF process acts as a rapid diffusion path to accelerate the formation of a protective film in the initial stage, leading to an enhancement in oxidation resistance for extended exposure.

Environmental perception plays a fundamental role in decision-making and is crucial for ensuring the safety of autonomous driving. A pressing challenge is the online evaluation of perception uncertainty, a crucial step towards ensuring the safety and the industrialization of autonomous driving. High-definition maps offer precise information about static elements on the road, along with their topological relationships. As a result, the map can provide valuable prior information for assessing the uncertainty associated with static elements. In this paper, a method for evaluating perception uncertainty online, encompassing both static and dynamic elements, is introduced based on the high-definition map. The proposed method is as follows: Firstly, the uncertainty of static elements in perception, including the uncertainty of their existence and spatial information, was assessed based on the spatial and topological features of the static environmental elements; secondly, an online assessment model for the uncertainty of dynamic elements in perception was constructed. The online evaluation of the static element uncertainty was utilized to infer the dynamic element uncertainty, and then a model for recognizing the driving scenario and weather conditions was constructed to identify the triggering factors of uncertainty in real-time perception during autonomous driving operations, which can further optimize the online assessment model for perception uncertainty. The verification results on the nuScenes dataset show that our uncertainty assessment method based on a high-definition map effectively evaluates the real-time perception results’ performance.

This research paper employed a multi-session framework to present an innovative approach to map monitoring within the domain of high-definition (HD) maps. The proposed methodology uses a machine learning algorithm to derive a confidence level for the detection of specific map elements in each frame and tracks the position of the element in subsequent frames. This creates a virtual belief system, which indicates the existence of the element on the HD map. To confirm the existence of the element and ensure the credibility of the map data, a reconstruction and matching technique was implemented. The notion of an expected observation area is also introduced by strategically limiting the vehicle’s observation range, thereby bolstering the detection confidence of the observed map elements. Furthermore, we leveraged data from multiple vehicles to determine the necessity for updates within specific areas, ensuring the accuracy and dependability of the map information. The validity and practicality of our approach were substantiated by real experimental data, and the monitoring accuracy exceeded 90%.
