Abstract External signs of disease are frequently used as indicators of disease susceptibility. However, immune profiling can be a more effective indicator to understand how host responses to infection may be shaped by host, pathogen and environmental factors. To better inform wildlife health assessment and research directions, we investigated the utility of a novel multivariate immunophenotyping approach examining innate and adaptive immune responses in differing climatic, pathogen co-infection and demographic contexts across two koala (Phascolarctos cinereus) populations in New South Wales: the Liverpool Plains (LP), and Southern Highlands to South-west Sydney (SHSWS). Relative to the comparatively healthy SHSWS, the LP had greater and more variable innate immune gene expression (IL-1β, IL-6), and KoRV transcription. During extreme heat and drought, koalas from the LP displayed upregulation of a stress pathway gene and reduced adaptive immune genes expression, haematocrit and plasma protein, suggesting the possibility of environmental impacts through multiple pathways. In those koalas, KoRV transcription status, Chlamydia pecorum infection loads, and visible urogenital inflammation were not associated with immune variation, suggesting that immune markers were more sensitive indicators of real-time impacts than observed disease outcomes.

Abstract Genetic management is a critical component of threatened species conservation. Understanding spatial patterns of genetic diversity is essential for evaluating the resilience of fragmented populations to accelerating anthropogenic threats. Nowhere is this more relevant than on the Australian continent, which is experiencing an ongoing loss of biodiversity that exceeds any other developed nation. Using a proprietary genome complexity reduction‐based method (DArTSeq), we generated a data set of 3239 high quality Single Nucleotide Polymorphisms (SNPs) to investigate spatial patterns and indices of genetic diversity in the koala (Phascolarctos cinereus), a highly specialised folivorous marsupial that is experiencing rapid and widespread population declines across much of its former range. Our findings demonstrate that current management divisions across the state of New South Wales (NSW) do not fully represent the distribution of genetic diversity among extant koala populations, and that care must be taken to ensure that translocation paradigms based on these frameworks do not inadvertently restrict gene flow between populations and regions that were historically interconnected. We also recommend that koala populations should be prioritised for conservation action based on the scale and severity of the threatening processes that they are currently faced with, rather than placing too much emphasis on their perceived value (e.g., as reservoirs of potentially adaptive alleles), as our data indicate that existing genetic variation in koalas is primarily partitioned among individual animals. As such, the extirpation of koalas from any part of their range represents a potentially critical reduction of genetic diversity for this iconic Australian species.

Abstract Scavenging on carrion is critical and often fiercely competitive for a range of vertebrate species, from native apex predators to invasive species and even reptiles. Within Australia, a notable reptilian scavenger is the lace monitor (Varanus varius). In this study, we quantified lace monitor activity at carcasses and compared their use of the resource to common co‐occurring predators that also scavenge; the invasive red fox (Vulpes vulpes) and a native apex predator, the dingo (Canis dingo). To do so, we deployed 80 macropod carcasses equally across seasons (summer and winter) and habitats (open and closed canopy) in a temperate bioregion and monitored vertebrate scavenging with camera traps. Lace monitor activity (visitation at carcass sites inclusive of both non‐scavenging and scavenging events) was 1.67 times higher in summer than in winter, but it did not differ across closed and open habitats. Monitor activity occurred earlier after carcass deployment at sites deployed in summer than winter (1.47‐fold earlier), and at carcasses in open than closed habitats (0.22‐fold earlier). Lace monitors initially discovered carcass sites faster in summer than winter and before both red foxes and dingoes in summer. The species was active diurnally in both summer and winter, differing from the red fox, which was strictly a nocturnal scavenger and the dingo, which was significantly more active at night across both seasons. Finally, we found that lace monitor activity at carcass sites decreased slightly with higher rates of activity for dingoes (0.04‐fold decrease as dingo activity increased), but not with red fox activity. Our results have implications for understanding lace monitor foraging and scavenging and highlight the value of monitoring carcasses to provide important insights into the behaviour of varanid lizards that scavenge.

Abstract Chlamydiosis is a significant disease affecting Eastern Australian koala (Phascolarctos cinereus) populations, impacting individual animal welfare and fecundity and therefore influencing population dynamics. The aim of this study was to investigate the effect of a synthetic peptide vaccine based on 4 components of the Chlamydia pecorum major outer membrane protein (MOMP), over an 18-month period in a koala population severely impacted by chlamydiosis. Wild koalas were recruited into a vaccination or a placebo treatment group on a random allocation, then followed through a period of 18 months, with recapture at 6 monthly intervals. Vaccination did not alter clinical disease expression or chlamydial shedding from the ocular or urogenital sites. Vaccination did not stimulate a significant plasma anti-MOMP IgG response, when compared to the placebo group. There was no significant effect of vaccination on IFN-γ and IL-17A mRNA expression of peripheral blood lymphocytes when stimulated with rMOMP. We have demonstrated that a synthetic peptide vaccination against chlamydiosis is not an effective management tool in a koala population with a high prevalence of C. pecorum infection and related disease. The lack of antigenic response found in this study suggests that further research utilising a larger, full-length antigen is an avenue worth investigation if we are to consider vaccination as a part of a management strategy in diseased koala populations.

Abstract Expert elicitation can be valuable for informing decision‐makers on conservation and wildlife management issues. To date, studies eliciting expert opinions have primarily focused on identifying and building consensus on key issues. Nonetheless, there are drawbacks of a strict focus on consensus, and it is important to understand and emphasize dissent, too. This study adopts a dissensus‐based Delphi to understand conflict among dingo experts. Twenty‐eight experts participated in three rounds of investigation. We highlight disagreement on most of the issues explored. In particular, we find that disagreement is underpinned by what we call “conflict over values” and “conflict over evidence.” We also note the broader role played by distrust in influencing such conflicts. Understanding and recognizing the different elements shaping disagreement is critical for informing and improving decision‐making and can also enable critique of dominant paradigms in current practices. We encourage greater reflexivity and open deliberation on these aspects and hope our study will inform similar investigations in other contexts.

Abstract Shortly after the enactment of restrictions aimed at limiting the spread of COVID-19, various local government and public health authorities around the world reported an increased sighting of rats. Such reports have yet to be empirically validated. Here we combined data from multi-catch rodent stations (providing data on rodent captures), rodent bait stations (providing data on rodent activity) and residents’ complaints to explore the effects of a six week lockdown period on rodent populations within the City of Sydney, Australia. The sampling interval encompassed October 2019 to July 2020 with lockdown defined as the interval from April 1st to May 15th, 2020. Rodent captures and activity (visits to bait stations) were stable prior to lockdown. Captures showed a rapid increase and then decline during the lockdown, while rodent visits to bait stations declined throughout this period. There were no changes in the frequency of complaints during lockdown relative to before and after lockdown. There was a non-directional change in the geographical distribution of indices of rodent abundance suggesting that rodents redistributed in response to resource scarcity. We hypothesize that lockdown measures initially resulted in increased rodent captures due to sudden shortage of human-derived food resources. Rodent visits to bait stations might not show this pattern due to the nature of the binary data collected, namely the presence or absence of a visit. Relocation of bait stations driven by pest management goals may also have affected the detection of any directional spatial effect. We conclude that the onset of COVID-19 may have disrupted commensal rodent populations, with possible implications for the future management of these ubiquitous urban indicator species.

In addition to feeding on animal remains, many scavengers also function as predators. Carcasses may therefore affect local animal communities by attracting facultative scavengers and increasing predation risk for other species in the vicinity of the carcasses. This risk may be elevated in low productivity environments, especially where humans increase carcass production and where facultative scavengers include invasive species. In June and October 2018, we monitored experimentally placed red kangaroo (Osphranter rufus) carcasses and artificial bird nests in two different habitats in the Simpson Desert, Australia, to identify the nest predators attracted to the carcasses, and to determine how carcasses affect overall and predator-specific nest predation. We modelled our nests to approximate those of the ground nesting little buttonquail (Turnix velox) and the endangered night parrot (Pezoporus occidentalis). Native Corvus spp. and then invasive red foxes (Vulpes vulpes) were the top carcass visitors and nest egg predators. Carcass presence and open habitat increased overall nest predation and fewer artificial parrot nest eggs were depredated compared to those of quail. Open habitat and carcass presence only increased predator-specific nest predation by foxes, but corvid nest predation was highest in June 2018, and for the artificial quail nest types. Foxes were the main predator of eggs from night parrot nests. Our study shows that carcass provisioning by humans may have indirect, deleterious effects on ground nesting birds, and indicates that foxes might pose a greater threat to night parrot populations than previously recognised.

The strong innate fear response shown by laboratory rodents to predator cues could provide powerful and innovative tools for pest management. Predator cues are routinely used to induce fear and anxiety in laboratory rodents for pharmacological studies. However, research on the fear response induced by predator cues in different species of rodents in the wild has been inconclusive with results often contradictory to laboratory experiments. Potential explanations for this inconsistency include the prey's: (i) physiological state; (ii) parasite load; (iii) differential intensity of perceived threats; (iv) fear learning and habituation; and (v) information gathering. In this review, we first explore current knowledge on the sensory mechanisms and capabilities of rodents, followed by the discussion of each of these explanations within the context of their implications for the use of antipredator response as a pest rodent management tool. Finally, we make recommendations on potential solutions and strategies to resolve issues in rodent management related to these hypotheses.

Biological invasions can massively disrupt ecosystems, but evolutionary and ecological adjustments may modify the magnitude of that impact through time. Such post-colonisation shifts can change priorities for management. We quantified the abundance of two species of giant monitor lizards, and of the availability of their mammalian prey, across 45 sites distributed across the entire invasion trajectory of the cane toad (Rhinella marina) in Australia. One varanid species (Varanus panoptes from tropical Australia) showed dramatic population collapse with toad invasion, with no sign of recovery at most (but not all) sites that toads had occupied for up to 80 years. In contrast, abundance of the other species (Varanus varius from eastern-coastal Australia) was largely unaffected by toad invasion. That difference might reflect availability of alternative food sources in eastern-coastal areas, perhaps exacerbated by the widespread prior collapse of populations of small mammals across tropical (but not eastern) Australia. According to this hypothesis, the impact of cane toads on apex predators has been exacerbated and prolonged by a scarcity of alternative prey. More generally, multiple anthropogenically-induced changes to natural ecosystems may have synergistic effects, intensifying the impacts beyond that expected from either threat in isolation.

Arboreal folivores are particularly vulnerable to the impacts of extreme climate change-driven heatwaves and droughts as they rely on leaf moisture to maintain hydration. During these increasingly frequent and intense weather events, leaf water content may not be enough to meet their moisture requirements, potentially leading to large-scale mortality due to dehydration. Water supplementation could be critical for the conservation of these animals. We tested artificial water stations for a threatened arboreal folivore, the koala (Phascolarctos cinereus), as a potential mitigation measure during hot and dry weather in New South Wales, Australia. We provided ground and tree drinkers to koalas and investigated changes in use with season, environmental conditions and foliar moisture. Our study provides first evidence of the regular use of free water by koalas. Koalas used supplemented water extensively throughout the year, even during cooler months. Time spent drinking varied with season and depended on days since last rain and temperature. The more days without rain, the more time koalas spent drinking. When temperature was high, visits to water stations were more frequent, indicating that in hot weather koalas need regular access to free water. Our results suggest that future changes in rainfall regimes and temperature in Australia have the potential to critically affect koala populations. Our conclusions can be applied to many other arboreal folivorous mammals worldwide which rely on leaves for their nutritional and water requirements. Artificial water stations may facilitate the resilience of vulnerable arboreal folivores during heat and drought events and may help mitigate the effects of climate change.

Carnivore predation on livestock often leads people to retaliate. Persecution by humans has contributed strongly to global endangerment of carnivores. Preventing livestock losses would help to achieve three goals common to many human societies: preserve nature, protect animal welfare, and safeguard human livelihoods. Between 2016 and 2018, four independent reviews evaluated >40 years of research on lethal and nonlethal interventions for reducing predation on livestock. From 114 studies, we find a striking conclusion: scarce quantitative comparisons of interventions and scarce comparisons against experimental controls preclude strong inference about the effectiveness of methods. For wise investment of public resources in protecting livestock and carnivores, evidence of effectiveness should be a prerequisite to policy making or large-scale funding of any method or, at a minimum, should be measured during implementation. An appropriate evidence base is needed, and we recommend a coalition of scientists and managers be formed to establish and encourage use of consistent standards in future experimental evaluations.

The roles that top predators play in regulating the structure and function of ecosystems have long been controversial. This is particularly the case when predators pose adverse risks for human life and/or economic interests. The critique of literature on dingoes and their ecological roles in Australia provided by Allen et al. (2011) shows that top predators remain a potentially polarising issue. In opposition to Allen et al. we argue that these widespread patterns of species’ abundances, attributed to the effects of dingoes and evident at scales ranging from the foraging behaviour of individuals through to continental scale patterns of species abundances, constitute strong support for the mesopredator release hypothesis and provide evidence that dingoes benefit biodiversity conservation by inducing community wide trophic cascades. Harnessing the positive ecological effects of dingoes while at the same time minimising their impacts on agriculture is a major socio-political challenge in Australia [Current Zoology 57 (5): 668-670].

In open, arid environments with limited shelter there may be strong selection on small prey species to develop behaviors that facilitate predator avoidance. Here, we predicted that rodents should avoid predator odor and open habitats to reduce their probability of encounter with potential predators, and tested our predictions using a native Australian desert rodent, the spinifex hopping-mouse (Notomys alexis). We tested the foraging and movement responses of N. alexis to non-native predator (fox and cat) odor, in sheltered and open macro- and microhabitats. Rodents did not respond to predator odor, perhaps reflecting the inconsistent selection pressure that is imposed on prey species in the desert environment due to the transience of predator-presence. However, they foraged primarily in the open and moved preferentially across open sand. The results suggest that N. alexis relies on escape rather than avoidance behavior when managing predation risk, with its bipedal movement probably allowing it to exploit open environments most effectively.

A detailed understanding of how extensively animals move through the landscape, and the habitat features upon which they rely, can identify conservation priorities and thus inform management planning. For many endangered species, information on habitat use either is sparse, or is based upon studies from a small part of the species' range. The broad-headed snake (Hoplocephalus bungaroides) is restricted to a specialized habitat (sandstone outcrops and nearby forests) within a small geographic range in south-eastern Australia. Previous research on this endangered taxon was done at a single site in the extreme south of the species' geographic range. We captured and radio-tracked 9 adult broad-headed snakes at sites in the northern part of the species' distribution, to evaluate the generality of results from prior studies, and to identify critical habitat components for this northern population. Snakes spent most of winter beneath sun-warmed rocks then shifted to tree hollows in summer. Thermal regimes within retreat-sites support the hypothesis that this shift is thermally driven. Intervals between successive displacements were longer than in the southern snakes but dispersal distances per move and home ranges were similar. Our snakes showed non-random preferences both in terms of macrohabitat (e.g., avoidance of some vegetation types) and microhabitat (e.g., frequent use of hollow-bearing trees). Despite many consistencies, the ecology of this species differs enough between southern and northern extremes of its range that managers need to incorporate information on local features to most effectively conserve this threatened reptile.

Invasive predators can impose strong selection pressure on species that evolved in their absence and drive species to extinction. Interactions between coexisting predators may be particularly strong, as larger predators frequently kill smaller predators and suppress their abundances. Until 3500 years ago the marsupial thylacine was Australia's largest predator. It became extinct from the mainland soon after the arrival of a morphologically convergent placental predator, the dingo, but persisted in the absence of dingoes on the island of Tasmania until the 20th century. As Tasmanian thylacines were larger than dingoes, it has been argued that dingoes were unlikely to have caused the extinction of mainland thylacines because larger predators are rarely killed by smaller predators. By comparing Holocene specimens from the same regions of mainland Australia, we show that dingoes were similarly sized to male thylacines but considerably larger than female thylacines. Female thylacines would have been vulnerable to killing by dingoes. Such killing could have depressed the reproductive output of thylacine populations. Our results support the hypothesis that direct killing by larger dingoes drove thylacines to extinction on mainland Australia. However, attributing the extinction of the thylacine to just one cause is problematic because the arrival of dingoes coincided with another the potential extinction driver, the intensification of the human economy.
