Abstract Infections caused by Candida spp. are frequent in critically hospitalized patients, especially among premature neonates, representing one of the most common healthcare-related infections. Although there is considerable production of current knowledge about the mechanisms of immune response, aspects involved in the newborn's innate defense are not fully understood. The aim of this study was to describe the innate immune mechanisms involved in the defense of neonates against invasive candidiasis. This is an integrative literature review from the Scopus, Scifinder, Medline, Web of Science databases and the electronic libraries ScienceDirect and Scielo, in the period between 2002 and 2020, with rescue based on primary descriptor Immunity Innate plus secondary descriptors Candidiasis Invasive AND Infant Newborn. We have observed the involvement of various mechanisms in the neonatal response against invasive candidiasis, including the recognition, signaling, recruitment, and initiation of an effective immune response. These mechanisms encompass the presence of antimicrobial peptides, phagocytosis, synthesis of reactive oxygen species, inflammatory mediators, and complex cell signaling systems mediated by Pattern Recognition Receptors (PRRs). With this study, it is expected to contribute to the expansion of knowledge about the immunological mechanisms involved in the innate immune response of the newborn against disseminated infections caused by Candida species, and in the same sense, highlight the importance of this knowledge as a reflex in the decrease in mortality in the neonatal period. more...

Abstract Background Major depressive disorder (MDD) is a heterogeneous psychiatric disorder. Childhood trauma (CT, emotional/physical/sexual abuse or neglect before the age of 18) is one of the largest and most consistent risk factors for development and poor course of MDD. Overactivity of the HPA-axis and the stress hormone cortisol is thought to play a role in the vulnerability for MDD following exposure to CT. Rodent experiments showed that antagonism of the glucocorticoid receptor (GR) at adult age reversed the effects of early life stress. Similarly, we aim to target MDD in individuals with CT exposure using the GR antagonist mifepristone. Methods The RESET-medication study is a placebo-controlled double-blind randomized controlled trial (RCT) which aims to include 158 adults with MDD and CT. Participants will be randomized (1:1) to a 7-day treatment arm of mifepristone (1200 mg/day) or a control arm (placebo). Participants are allowed to receive usual care for MDD including antidepressants. Measurements include three face-to-face meetings at baseline (T0), day 8 (T1), week 6 (T2), and two online follow-up meetings at 12 weeks (T3) and 6 months (T4). A subgroup of participants (N = 80) are included in a fMRI sub-study (T0, T2). The main study outcome will be depressive symptom severity as measured with the Inventory of Depressive Symptomatology—Self Rated (IDS-SR) at T2. Secondary outcomes include, among others, depressive symptom severity at other time points, disability, anxiety, sleep and subjective stress. To address underlying mechanisms mifepristone plasma levels, cortisol, inflammation, epigenetic regulation and fMRI measurements are obtained. Discussion The RESET-medication study will provide clinical evidence whether GR antagonism is a disease-modifying treatment for MDD in individuals exposed to CT. If effective, this hypothesis-driven approach may extend to other psychiatric disorders where CT plays an important role. Lunar phases remarkably influence the circadian cycle of living beings. Early amphibian studies date back to the 1960s, but only recently has more research been conducted in this field. Much still needs to be understood to establish the behavioral pattern of this group according to the synodic cycle. In the present study, we sought to determine (i) whether the vocalization activity was influenced by the lunar phases and (ii) whether the influence was species-specific or affects the community with equal intensity. We expected a great diversity of behaviors and adaptations. Rayleigh's test was used to verify whether the sample differs significantly from the null hypothesis; Rao's test was used to check sample size sufficiency; and the Kiviat diagram was used to evaluate the activity of the species in relation to the complete synodic cycle. We have observed 1691 individuals of 37 species over 882 d. The lunar cycle influenced 78 % of the species, with 32 % preferring the lower-albedo phases. The activity pattern of each species was established. These results suggest that the lunar phases influence the vocalization activity of most species. Therefore, there is a general pattern of activity related to the synodic cycles; however, the specificities still need to be better understood. more...

Abstract The Neotropical zone stands out for having the greatest richness of amphibians in the world. About 625 species of anurans are known on Atlantic Forest. The southeastern mountainous regions of Brazil called Serra do Mar and Serra da Mantiqueira, are very important areas for the diversity of anurans in the country because they shelter remaining strata of the Atlantic Forest that have several endemic species. The present study aims to expand the knowledge about the Serra da Mantiqueira anurans, specifically from the Rio Preto and Rio Grande upper basins. The study area comprised 10 sample centers. We realized 38 campaigns, lasting two days each. During this period, we used active search as visual and auditory cues. The passive search was comprised of pitfall traps. Bioacoustic analyses were used to determine the cryptic species. The sufficiency of the sampling effort was estimated through the accumulation curve using Bootstrap and Jackknife-1 as estimators. During the study 55 species were found, 40 of them in the basin of the Rio Preto, and 44 in the basin of the Rio Grande. Twenty-nine species were found in both areas. The Hylidae family was predominant, followed by Leptodactylidae. In addition to species of the Atlantic Forest, the place houses typical components of the Cerrado biome and altitudinal swamps with unexpected sympatries. This inventory expands the area of occurrence of Scinax duartei, a species considered threatened. Abstract The sexing of species is an important tool for population management. In tortoises and turtles, sexing is usually done according to secondary characteristics, such as plastron morphology or phallus eversion techniques. In this paper, we present the phallus eversion method and compare its efficiency to other existing techniques. This new method aims at shortening the management time and reducing the physical aftereffects on the animals, fostering the clinical and reproductive management of Geoffroy’s side-necked turtle (Phrynops geoffroanus) and related species. A sample of fifty individuals was taken in the central region of Piauí state, Northeastern Brazil. The proposed method consisted of moving the head from its resting position. The methods achieving a success rate higher than 80% were compared to each other as for the stimulation time required for phallus eversion. The method as here proposed achieved 100% efficiency in the sex determination of specimens with a linear carapace length between 10 and 34 cm. Also, when compared to the second most efficient method, it substantially reduced the time needed for managing the specimens (91%), avoiding possible sequelae in the individuals handled. more...

For a small satellite, the processor onboard the attitude determination and control system (ADCS) is required to monitor, communicate, and control all the sensors and actuators. In addition, the processor is required to consistently communicate with the satellite bus. Consequently, the processor is unable to ensure all the sensors and actuators will immediately respond to the data acquisition request, which leads to asynchronous data problems. The extended Kalman filter (EKF) is commonly used in the attitude determination process, but it assumes fully synchronous data. The asynchronous data problem would greatly degrade the attitude determination accuracy by EKF. To minimize the attitude estimation accuracy loss due to asynchronous data while ensuring a reasonable computational complexity for small satellite applications, this paper proposes the simplex-back-propagation Kalman filter (SBPKF). The proposed SBPKF incorporates the time delay, gyro instability, and navigation error into both the measurement and covariance estimation during the Kalman update process. The performance of SBPKF has been compared with EKF, modified adaptive EKF (MAEKF), and moving–covariance Kalman filter (MC-KF). Simulation results show that the attitude estimation error of SBPKF is at least 30% better than EKF and MC-KF. Abstract The work aims were to describe the histological and histochemical structure of the gastroesophageal tube of Iguana iguana and verify the occurrence and distribution of immunoreactive serotonin (5-HT) and somatostatin (SS) cells. Fragments of the gastrointestinal tract (GIT) of five iguanas were which underwent standard histological and immunohistochemistry technique. Immunoreactive cells for 5-HT and SS were quantified using the STEPanizer. The oesophagus has ciliated columnar pseudostratified epithelium with staining Alcian blue (AB) + and goblet cells highly reactive to periodic acid Schiff (PAS). In the cervical oesophagus, the numerical density of 5-HT cells per unit area (QA [5-HT cells]/µm2) was 4.6x10-2 ± 2.0 and celomatic oesophagus presented QA = 4.0x10-2 ± 1.0. The epithelium of the stomach is simple columnar, PAS and AB +. The cranial and middle regions of the stomach presented (QA [5-HT cells]/µm2) = 6.18x10-2 ± 3.2 and the caudal region, QA = 0.6x10-2 ± 0.2. The SS cells were only observed in the caudal stomach, with numerical density (QA [SS cells]/µm2) = 1.4x10-2 ± 0.9 In I. iguana, variation was observed in terms of the distribution of mucus secretions and the pattern of occurrence of serotonin and somatostatin-secreting enteroendocrine cells in the TGI, which possibly will result in an interspecific adaptive response. more...

Abstract Vocalizations are an important trait for the identification of cryptic and/or closely related amphibian species. Different vocalizations also contribute to partitioning of the acoustic space by sympatric species. This study aimed to describe the advertisement calls of anurans in a pond of the municipality of Floriano, State of Piauí, Brazil, and infer the acoustic niche partitioning of amphibians. Euclidean distance was used in a cluster analysis approach to infer the acoustic similarities among species. Thirteen species were analysed: Boana raniceps, Dendropsophus nanus, D. rubicundulus , D. minutus, Leptodactylus fuscus, L. troglodytes, L. vastus, Pithecopus nordestinus , Physalaemus cuvieri, P. nattereri, Pleurodema diplolister, Proceratophrys cristiceps and Scinax ruber. From these, six showed more than 90% of acoustic overlap: P. nattereri , P. cuvieri, L. fuscus and L. vastus (Leptodactylidae); and, D. nanus and D. rubicundulus (Hylidae). Despite the acoustic similarities among these six species, the acoustic interference was reduced due to the small number of sympatric species and to distinct features on carrier frequency such as dominant frequency and the degree of modulation in the frequency. Abstract The practice of capture-recapture to estimate the diversity is well known to many animal groups, however this practice in the larval phase of anuran amphibians is incipient. We aimed at evaluating the Lincoln estimator, Venn diagram and Bayes theorem in the inference of population size of a larval phase anurocenose from lotic environment. The adherence of results was evaluated using the Kolmogorov-Smirnov test. The marking of tadpoles for later recapture and methods measurement was made with eosin methylene blue. When comparing the results of Lincoln-Petersen estimator corresponding to the Venn diagram and Bayes theorem, we detected percentage differences per sampling, i.e., the proportion of sampled anuran genera is kept among the three methods, although the values are numerically different. By submitting these results to the Kolmogorov-Smirnov test we have found no significant differences. Therefore, no matter the estimator, the measured value is adherent and estimates the total population. Together with the marking methodology, which did not change the behavior of tadpoles, the present study helps to fill the need of more studies on larval phase of amphibians in Brazil, especially in semi-arid northeast. more...

Background. Subjective well-being incorporates elements of life satisfaction, happiness and optimism. It is increasingly relevant in the assessment of population health and economic development. There are strong continuities in well-being from youth into later life. Despite its significance, few global surveys capture subjective well-being. This paper describes patterns of well-being among young people in five Eastern European countries [Belarus, Bosnia and Herzegovina (BiH), the Former Yugoslav Republic of Macedonia, Serbia and Ukraine] and investigates association between demographic factors and well-being. Methods. Nationally representative household surveys, including large Roma population samples, were conducted as part of UNICEF's Multiple Indicator Cluster Survey programme. Young people aged 15–24 years (N = 11 944) indicated their satisfaction with life, happiness and expectations about the future. Multilevel logistic regressions were conducted to determine the impact of individual-level predictors while accounting for country- and cluster-level variability. Results. Around 40% of young people considered themselves very happy or very satisfied with their life overall. Three quarters reported optimism. Yet well-being varied greatly between countries, with youth in BiH and Ukraine reporting lowest levels of well-being. Current marriage, increasing wealth, higher education, rural residence and not having children were associated with greater well-being. Conclusions. Patterns of well-being in youth vary substantially between countries and are only partly accounted for by standard demographic characteristics. Despite higher rates of adolescent marriage and childbearing, and lower levels of educational attainment and employment, Roma youth had similar levels of well-being to the general population. more...

Several yeast species of Candida genus can colonize the skin as well as the mucous membrane of the vagina and the digestive tract for short or long periods. Depending on the host's immunological state and the yeast's virulence, colonization can become an infection, invading the colonized tissues and also disseminating. AIDS is characterized by the host's intensive and progressive immunodepression which manifests as diverse symptoms, mainly lesions in the mouth. Oral candidiasis is the most prevalent opportunistic infection in individuals infected with human immunodeficiency virus (HIV) and is an important indicator of the disease progress and the immunosuppression increase. Neste trabalho o método do precursor polimérico foi usado para obter a fase Zn2TiO4 dopada com V5+. As fases foram obtidas depois que o pó precursor foi submetido a diferentes temperaturas; a caracterização do composto foi feita por meio de difração de raios X, análise térmica e fotoluminescência. A difícil entrada do vanádio na rede polimérica conduziu à segregação na resina e também no óxido final. Depois da dopagem a temperatura de decomposição térmica foi diminuída de 438 ºC a 420 ºC. Os pós calcinados a 400 °C e 500 °C apresentaram uma solução sólida substitucional entre os íons metálicos Ti4+ e V5+, além de formação de fase secundária. Uma diminuição do volume da cela unitária depois da adição de vanádio foi observada. A cristalinidade relativa e o tamanho de cristalito apresentaram uma variação de tamanho devido a difícil substituição do vanádio pelo titânio. Foram obtidos os melhores resultados de propriedade fotoluminescente para a amostra com 0,2 mol% de vanádio.In this work the polymeric precursor method was used to obtain the Zn2TiO4 doped with V5+. The phases were obtained after calcination the respective precursor powders at different temperatures, and their characterization was accomplished by X ray diffraction, thermal analysis and photoluminescence. It was difficult for vanadium to get into the polymeric net, leading to segregation in the resin and also in the final material. After doping, the thermal decomposition temperature decreased from 438 ºC to 420 ºC. The powders calcined at 400 °C and 500 °C presented a substitutional solid solution between Ti4+ and V5+ metallic ions, besides secondary phase formation. A decrease of the volume unit cell after vanadium addition was observed. Relative crystallinity and crystallite size presented a random variation due to the difficulty for vanadium substituting titanium. The best results of photoluminescent properties were obtained for the sample with 0.2 mol% of vanadium. more...

A acidez potencial tem grande importância pela sua utilização na determinação da saturação por bases e auxílio na recomendação da necessidade de calagem. Este estudo teve por objetivo avaliar a relação entre o teor de H + Al e o pH SMP, visando estabelecer uma equação para estimar a acidez potencial de solos da Região Norte de Minas Gerais. As análises dos teores de H + Al (acetato de cálcio 0,5 mol L-1 a pH 7,0) e dos valores de pH SMP foram realizadas no laboratório de fertilidade do solo da EPAMIG/CTNM em 50 amostras de solo, com valores de pH em água variando de 4,6 a 7,8, teores de carbono orgânico de 3 a 45 g kg-1 e os de argila de 80 a 610 g kg-1. A acidez potencial, expressa em cmol c dm-3, pode ser estimada pelo uso do pH SMP, utilizando para isso a seguinte equação: H + Al = 0,00359 + 1556,5806 e- pHSMP (R² = 0,96**). more...

Resumo Os pigmentos inorgânicos são os mais utilizados industrialmente, pois apresentam maior estabilidade química e térmica e podem ser produzidos com um elevado grau de pureza e uniformidade. Essas características levaram ao desenvolvimento de diferentes cores intensas, estáveis e adequadas ao uso como pigmentos. O presente trabalho tem por objetivo sintetizar materiais cerâmicos com estrutura espinélio para uso como pigmento cerâmico e aplicá-los em fritas cerâmicas. O processo de síntese foi realizado pela rota da gelatina e com os pós obtidos foram conduzidos um estudo de evolução de fases cristalinas por meio de difração de raios X, refinamento das estruturas por meio do método Rietveld, espectroscopia na região do infravermelho, microscopia eletrônica de varredura, espectroscopia na região do UV-visível e colorimetria. Foram observadas nos difratogramas reflexões características da estrutura espinélio. O tamanho médio de cristalito e a cristalinidade aumentou com o aumento da temperatura de calcinação. Patients with gastro‐intestinal stromal tumors (GISTs) undergoing tyrosine kinase inhibitor therapy are monitored with regular computed tomography (CT) scans, exposing patients to cumulative radiation. This exploratory study aimed to evaluate circulating tumor DNA (ctDNA) testing to monitor treatment response and compare changes in ctDNA levels with RECIST 1.1 and total tumor volume measurements. Between 2014 and 2021, six patients with KIT proto‐oncogene, receptor tyrosine kinase (KIT) exon‐11‐mutated GIST from whom long‐term plasma samples were collected prospectively were included in the study. ctDNA levels of relevant plasma samples were determined using the KIT exon 11 digital droplet PCR drop‐off assay. Tumor volume measurements were performed using a semi‐automated approach. In total, 94 of 130 clinically relevant ctDNA samples were analyzed. Upon successful treatment response, ctDNA became undetectable in all patients. At progressive disease, ctDNA was detectable in five out of six patients. Higher levels of ctDNA correlated with larger tumor volumes. Undetectable ctDNA at the time of progressive disease on imaging was consistent with lower tumor volumes compared to those with detectable ctDNA. In summary, ctDNA levels seem to correlate with total tumor volume at the time of progressive disease. Our exploratory study shows promise for including ctDNA testing in treatment follow‐up. more...

Sporotrichosis is a subacute to chronic mycosis caused by species of the genus Sporothrix. While the lymphocutaneous form predominates in most cases, immunocompromised individuals may present with disseminated variants. It is more prevalent in adults and rare in infants, with poorly studied risk factors and epidemiological aspects in this age group. We describe a case of sporotrichosis infection in a 9-month-old child, presenting with two progressively growing nodules, soft in consistency, located in the right cervical and pre-auricular regions, associated with a decline in general condition and fever over a month’s duration. Despite multiple oral antibiotics, there was no lesional improvement. Aspiration of one of the nodules, and culture of the secretion revealed Sporothrix. The patient was then treated with oral itraconazole, leading to regression of the lesions within 2 months. The identification of the transmission agent in this case was not possible. This report aims to broaden the understanding of sporotrichosis manifestation in infants and to provide valuable insights for the enhancement of specific preventive, diagnostic, and therapeutic strategies for this age group of dermatological patients. more...

Two monoterpenoid lactones, loliolide (1) and epi-loliolide (2), were isolated from the crude dichloromethane extract of a microalga, Thalassiosira sp.). The structures of loliolide (1) and epi-loliolide (2) were elucidated by 1D and 2D NMR analysis, as well as a comparison of their 1H or/and 13C NMR data with those reported in the literature. In the case of loliolide (1), the absolute configurations of its stereogenic carbons were confirmed by X-ray analysis, whereas those of epi-loliolide (2) were determined by NOESY correlations. Loliolide (1) and epi-loliolide (2) were tested for their growth inhibitory activity against two Gram-positive (Staphylococcus aureus ATCC 29213, Enterococcus faecalis ATCC 29212) and two Gram-negative (Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853) bacteria, as well as one clinical isolate (E. coli SA/2, an extended-spectrum β-lactamase producer-ESBL) and two environmental isolates, S. aureus 74/24, a methicillin-resistant (MRSA), and E. faecalis B3/101, a vancomycin-resistant (VRE) isolates. The results showed that none of the tested compounds exhibited antibacterial activity at the highest concentrations tested (325 μM), and both revealed low antioxidant activity, with ORAC values of 2.786 ± 0.070 and 2.520 ± 0.319 µmol TE/100 mg for loliolide (1) and epi-loliolide (2), respectively. more...

Fucus spp. seaweeds thrive in the cold temperate waters of the northern hemisphere, specifically in the littoral and sublittoral regions along rocky shorelines. Moreover, they are known to be a rich source of bioactive compounds. This study explored the valorization of Fucus spiralis through the extraction of bioactives and polysaccharides (PSs) for food applications and biostimulant use. The bioactives were extracted using microwave hydrodiffusion and gravity (MHG), where the condition of 300 W for 20 min resulted in the highest total phenolic content and antioxidant activity of the extract. Cellular assays confirmed that the extract, at 0.5 mg/mL, was non-cytotoxic to HaCat cells. Polysaccharides (PSs) were extracted from the remaining biomass. The residue from this second extraction contained 1.5% protein and 13.35% carbohydrates. Additionally, the free amino acids and minerals profiles of both solid residues were determined. An edible film was formulated using alginate (2%), PS-rich Fucus spiralis extract (0.5%), and F. spiralis bioactive-rich extract (0.25%). The film demonstrated significant antioxidant properties, with ABTS and DPPH values of 221.460 ± 10.389 and 186.889 ± 36.062 µM TE/mg film, respectively. It also exhibited notable physical characteristics, including high water vapor permeability (11.15 ± 1.55 g.mm.m−2.day−1.kPa−1) and 100% water solubility. The residues from both extractions of Fucus spiralis exhibited biostimulant (BS) effects on seed germination and seedling growth. BSs with PSs enhanced pea germination by 48%, while BSs without PSs increased the root dry weight of rice and tomato by 53% and up to 176%, respectively, as well as the shoot dry weight by up to 38% and up to 74%, respectively. These findings underscore the potential of Fucus spiralis within the framework of a circular economy, wherein both extracted bioactives and post-extraction by-products can be used for sustainable agriculture and food applications. more...

The use of by-products as a source of bioactive compounds with economic added value is one of the objectives of a circular economy. The olive oil industry is a source of olive pomace as a by-product. The olive pomace used in the present study was the exhausted olive pomace, which is the by-product generated from the air drying and subsequent hexane extraction of residual oil from the olive pomace. The objective was to extract bioactive compounds remaining in this by-product. Various types of green extraction were used in the present study: solvent extraction (water and hydroalcoholic); ultrasound-assisted extraction; Ultra-Turrax-assisted extraction; and enzyme-assisted extraction (cellulase; viscoenzyme). The phenolic profile of each extract was determined using HPLC-DAD and the total phenolic content (TPC) and antioxidant activity (ABTS, DPPH, and ORAC) were determined as well. The results showed significant differences in the yield of extraction among the different methods used, with the enzyme-assisted, with or without ultrasound, extraction presenting the highest values. The ultrasound-assisted hydroethanolic extraction (USAHE) was the method that resulted in the highest content of the identified phenolic compounds: 2.021 ± 0.29 mg hydroxytyrosol/100 mg extract, 0.987 ± 0.09 mg tyrosol/100 mg extract, and 0.121 ± 0.005 mg catechol/100 mg extract. The conventional extraction with water at 50 °C produced the best results for TPC and antioxidant activity of the extracts. The extracts from the USAHE were able to inhibit Gram-positive bacteria, especially Bacillus cereus, showing 67.2% inhibition at 3% extract concentration. more...

Observational meteorological data from the field experiment GoAmazon 2014/15 and data from numerical simulations with the cloud-resolving model (CRM) called the System for Atmospheric Modeling (SAM) are used to study the interaction between the cloudiness–radiation as well as the atmospheric dynamics and thermodynamics variables for a site located in the central Amazon region (−3.2∘ S, −60.6∘ W) during the wet and dry periods. The main aims are to (a) analyze the temporal series of the integrated cloud fraction, precipitation rate and downward shortwave flux as well as (b) to determine the relationship between the integrated cloud fraction, radiative fluxes and large-scale variable anomalies as a function of the previous day's average. The temporal series of the integrated cloud fraction, precipitation rate and downward shortwave flux from SAM simulations showed physical consistency with the observations from GoAmazon 2014/15. Shallow and deep convection clouds show to have a meaningful impact on radiation fluxes in the Amazon region during wet and dry periods. Anomalies of large-scale variables (relative to the previous day's average) are physically associated with cloud formation, evolution and dissipation. SAM consistently simulated these results, where the cloud fraction vertical profile shows a pattern very close to the observed data (cloud type). Additionally, the integrated cloud fraction and large-scale variable anomalies, as a function of the previous day's average, have a good correlation. These results suggest that the memory of the large-scale dynamics from the previous day can be used to estimate the cloud fraction as well as the water content, which is a variable of the cloud itself. In general, the SAM satisfactorily simulated the interaction between cloud–radiation as well as dynamic and thermodynamic variables of the atmosphere during the periods of this study, being able to obtain atmospheric variables that are impossible to obtain in an observational way. more...

The present review paper focuses on recent developments in edible films and coatings made of base compounds from biological sources, namely plants, animals, algae, and microorganisms. These sources include by-products, residues, and wastes from agro-food industries and sea products that contribute to sustainability concerns. Chitosan, derived from animal biological sources, such as crustacean exoskeletons, has been the most studied base compound over the past three years. Polysaccharides typically constitute no more than 3–5% of the film/coating base solution, with some exceptions, like Arabic gum. Proteins and lipids may be present in higher concentrations, such as zein and beeswax. This review also discusses the enrichment of these bio-based films and coatings with various functional and/or bioactive compounds to confer or enhance their functionalities, such as antimicrobial, antioxidant, and anti-enzymatic properties, as well as physical properties. Whenever possible, a comparative analysis among different formulations was performed. The results of the applications of these edible films and coatings to fruit and vegetable products are also described, including shelf life extension, inhibition of microbial growth, and prevention of oxidation. This review also explores novel types of packaging, such as active and intelligent packaging. The potential health benefits of edible films and coatings, as well as the biodegradability of films, are also discussed. Finally, this review addresses recent innovations in the edible films and coatings industry, including the use of nanotechnologies, aerogels, and probiotics, and provides future perspectives and the challenges that the sector is facing. more...

Lactic acid bacteria (LAB) have been documented as potential vitamin B12 producers and may constitute an exogenous source of cobalamin for the microalga Chlorella vulgaris, which has been described as being able to perform vitamin uptake. Hence, there is an interest in discovering novel B12-producing probiotic LAB. Therefore, the purpose of the current work was to perform a phenotype–genotype analysis of the vitamin B12 biosynthesis capacity of LAB isolated from C. vulgaris bioreactors, and investigate their probiotic potential. Among the selected strains, Lactococcus lactis E32, Levilactobacillus brevis G31, and Pediococcus pentosaceus L51 demonstrated vitamin B12 biosynthesis capacity, with the latter producing the highest (28.19 ± 2.27 pg mL−1). The genomic analysis confirmed the presence of pivotal genes involved in different steps of the biosynthetic pathway (hemL, cbiT, cobC, and cobD). Notably, P. pentosaceus L51 was the only strain harboring cobA, pduU, and pduV genes, which may provide evidence for the presence of the cobalamin operon. All strains demonstrated the capability to withstand harsh gastrointestinal conditions, although P. pentosaceus L51 was more resilient. The potential for de novo cobalamin biosynthesis and remarkable probiotic features highlighted that P. pentosaceus L51 may be considered the most promising candidate strain for developing high-content vitamin B12 formulations. more...

Low rainfall in Northeast Brazil is a limiting factor for animal production. Forages that present crassulacean acid metabolism, such as forage cactus, are adapted to the edaphoclimatic conditions of this region, as they lose little water through the stomata. Thus, the objective was to evaluate the physical and chemical quality, fatty acid profile and sensory acceptance of the meat from goats fed forage cactus as a substitute for Tifton 85 hay. Twenty-one uncastrated mixed-breed goats with a mean body weight of 18 ± 0.86 kg and 7 ± 1 months of age were used. A completely randomized design with three treatments and seven replications per treatment was performed. The inclusion of 0 (control), 25 and 55% of forage cactus in substitution of Tifton 85 hay in the diet of the goats was evaluated. The lipid content in the meat of animals fed 25 and 55% of forage cactus was 1.33% and 1.26%, respectively, and was lower (p < 0.05) in relation to the meat of animals that received the control diet (1.56%). The inclusion of 55% of forage cactus provided an increase (p < 0.05) in the content of monounsaturated fatty acids in the meat (52.71%) in relation to the control meat (37.75%). Sensory analysis differed (p < 0.05) between treatments. We recommend replacing Tifton 85 hay with 55% forage cactus, as it presents greater sensory acceptance, and provides lower lipid content and higher content of monounsaturated fatty acids in goat meat. more...

The micro- and nanoencapsulation of bioactive compounds has resulted in a large improvement in the food, nutraceutical, pharmaceutical, and agriculture industries. These technologies serve, on one side, to protect, among others, vitamins, minerals, essential fatty acids, polyphenols, flavours, antimicrobials, colorants, and antioxidants, and, on the other hand, to control the release and assure the delivery of the bioactive compounds, targeting them to specific cells, tissues, or organs in the human body by improving their absorption/penetration through the gastrointestinal tract. The food industry has been applying nanotechnology in several ways to improve food texture, flavour, taste, nutrient bioavailability, and shelf life using nanostructures. The use of micro- and nanocapsules in food is an actual trend used mainly in the cereal, bakery, dairy, and beverage industries, as well as packaging and coating. The elaboration of bio capsules with high-value compounds from agro-industrial by-products is sustainable for the natural ecosystem and economically interesting from a circular economy perspective. This critical review presents the principal methodologies for performing micro- and nanoencapsulation, classifies them (top-down and/or bottom-up), and discusses the differences and advantages among them; the principal types of encapsulation systems; the natural plant sources, including agro-industrial by-products, of bioactive compounds with interest for the food industry to be encapsulated; the bioavailability of encapsulates; and the main techniques used to analyse micro- and nanocapsules. Research work on the use of encapsulated bioactive compounds, such as lycopene, hydroxytyrosol, and resveratrol, from agro-industrial by-products must be further reinforced, and it plays an important role, as it presents a high potential for the use of their antioxidant and/or antimicrobial activities in food applications and, therefore, in the food industry. The incorporation of these bioactive compounds in food is a challenge and must be evaluated, not only for their nutritional aspect, but also for the chemical safety of the ingredients. This study examined the effect of the COVID- 19 pandemic and related events on the use of e-learning, as well as other key determinants of it. The data were collected from 1039 university students in Sri Lanka. To examine the influence of the COVID-19 pandemic, which was viewed through the lens of precipitating events, on the intention–behaviour relationship, we employed the Technology Acceptance Model (TAM) with the inclusion of a moderating variable. While the findings indicated that the COVID-19 pandemic had clearly increased the usage of e-learning, we found no evidence to establish a moderating impact on the intention–behaviour relationship. The empirical model, however, was well fitted to the data, and the key components of the TAM were likewise adequately described by the relevant predictors. Furthermore, attitudes toward e-learning and perceived ease of use emerged as the most important factors in explaining behavioural intention, whereas relevance and experience were shown to be more significant in relation to perceived usefulness and perceived ease of use. Our work is significant because it adds to the existing empirical evidence on e-learning and supports the relevance of TAM in understanding the usage of e-learning even in extreme situations such as the COVID-19 pandemic. RESUMOA pesquisa de produtos naturais benéficos à saúde humana vem crescendo nos últimos 20 anos. Considerando que as plantas de Aloe são amplamente utilizadas pela população humana, em geral de maneira terapêutica, o objetivo deste estudo foi avaliar os efeitos de Aloearborescens Miller e Aloe barbadensis Miller, sobre o desenvolvimento vegetativo de linhagens normais e mutantes de Aspergillus nidulans. Conídios da linhagem biA1methG1, MSE e CLB3 de A. nidulans, foram inoculados em meio completo sem (Controle) e com extratos das duas espécies incubados por 2, 4, 6 e 8 horas a 37ºC, no escuro. Foi analisado em microscópio óptico, 200 conídios de cada tratamento. Para o desenvolvimento das colônias, as linhagens foram inoculadas no centro das placas juntamente com o meio de cultura sólido e sobre a membrana de diálise, visando a medição do diâmetro e do peso. A análise estatística foi baseada no teste de Tukey e todos os procedimentos experimentais foram conduzidos em triplicata. Todas as linhagens apresentaram interferências positivas quando expostas às plantas de Aloe, porém, de maneira variada. Ambas as espécies aceleraram a germinação em todas as linhagens testadas e atuaram na redução significativa de conídios mortos e/ou malformados. Em relação ao desenvolvimento vegetativo, todos os dados referentes ao peso úmido e diâmetro corrigido dos tratamentos demonstraram progressos, contudo, a razão diâmetro/peso apresentou somente na linhagem MSE, ação favorável dos tratamentos naturais. As informações deste estudo sugerem benefícios de A. arborescens e A. barbadensis, justificando a importância e continuidade da investigação, para melhor elucidar os mecanismos de ação dessas plantas. more...

In addition to conventional therapies, several new strategies have been proposed for modulating autoimmune diseases, including the adoptive transfer of immunological cells. In this context, dendritic cells (DCs) appear to be one of the most promising treatments for autoimmune disorders. The present study aimed to evaluate the effects of adoptive transfer of DCs obtained from both naïve and ovalbumin (OVA)-tolerant mice on the severity of TNBS induced colitis and analyze the eventual protective mechanisms.To induce oral tolerance, BALB/c mice were fed 4mg/mL OVA solution for seven consecutive days. Spleen DCs were isolated from tolerant (tDC) and naïve (nDC) mice, and then adoptively transferred to syngeneic mice. Three days later, colitis was induced in DC treated mice by intrarectal instillation of 100μg2,4,6-trinitrobenzenesulfonic acid (TNBS) dissolved in 50% ethanol. Control subjects received only intrarectal instillation of either TNBS solution or a vehicle. Five days later, mice from all groups were euthanized and examined for physiological and immunological parameters. Regarding the phenotype, we observed that the frequencies of CD11+ MHC II+ and CD11+ MHCII+ CD86+ cells were significantly lower in DCs isolated from tolerant mice than in those from naive mice. However, pretreatment with both types of DCs was able to significantly reduce clinical signs of colitis such as diarrhea, rectal prolapse, bleeding, and cachexia, although only treatment with tDCs was able to prevent weight loss from instillation of TNBS. In vitro proliferation of spleen cells from mice treated with either type of DCs was significantly lower than that observed in splenic cell cultures of naïve mice. Although no significant difference was observed in the frequencies of Treg cells in the experimental groups, the frequency of Th17+CD4+cellsand the secretion of IL-17 were more reduced in the cultures of spleen cells from mice treated with either type of DCs. The levels of IL-9 and IFN-γ were lower in supernatants of cells from mice treated with nDCs.The results allow us to conclude that the adoptive transfer of cells expressing CD11c is able to reduce the clinical and immunological signs of drug-induced colitis. Adoptive transfer of CD11c+DC isolated from both naive and tolerant mice altered the proliferative and T cell responses. The market of functional foods has experienced a huge growth in the last decades due to the increased consumers’ awareness in a healthy lifestyle. Dried fruits constitute good snacks, in alternative to salty or sweet ones, and food ingredients due to their taste and nutritional/health benefits. Bioactive molecules are interesting sources to develop functional foods, as they play a major role in improving the health status and minimizing disease risks. The bioactive compounds most widely discussed in literature are presented in this review, for example, polyphenols, phytosterols, and prebiotics. Different technologies to dry bioproducts for producing functional foods or ingredients are presented. New drying techniques for the preservation of bioactive compounds are proposed, focusing more specifically on dielectric drying. A discussion on the techniques that can be used to optimize drying processes is performed. An overview on dehydrated plant based foods with probiotics is provided. The microorganisms used, impregnation procedures, drying methods, and evaluated parameters are presented and discussed. The principal bioactive compounds responsible for nutritional and health benefits of plant derived dried food products—fruits and vegetables, fruits and vegetables by-products, grains, nuts, and algae—are presented. INTRODUCTION:Literature data have shown that the consumption of dietary proteins may cause modulatory effects on the host immune system, process denominated oral tolerance by bystander suppression. It has been shown that the bystander suppression induced by dietary proteins can improve inflammatory diseases such as experimental arthritis. Here, we evaluated the effects of oral tolerance induced by ingestion of ovalbumin (OVA) on TNBS-induced colitis in mice, an experimental model for human Crohn's disease. METHODS AND RESULTS:Colitis was induced in BALB/c mice by instilling a single dose of TNBS (100 mg/kg) in ethanol into the colon. Tolerized mice received OVA (4mg/mL) dissolved in the drinking water for seven consecutive days, prior to or concomitantly with the intrarectal instillation. Control groups received protein-free water and ethanol by intrarectal route. We observed that either the prior or concomitant induction of oral tolerance were able to reduce the severity of colitis as noted by recovery of body weight gain, improvement of clinical signs and reduction of histological abnormalities. The in vitro proliferation of spleen cells from tolerant colitic mice was lower than that of control mice, the same as the frequencies of CD4+ T cells secreting IL-17 and IFN-γ. The frequencies of regulatory T cells and T cells secreting IL-10 have increased significantly in mice orally treated with OVA. The levels of inflammatory cytokines (IL-17A, TNF-α, IL-6 and IFN-γ) were lower in supernatants of cells from tolerant colitic mice, whereas IL-10 levels were higher. CONCLUSION:Our data show that the modulation of immune response induced by oral tolerance reduces the severity of experimental colitis. Such modulation may be partially attributed to the increase of Treg cells and reduction of pro-inflammatory cytokines in peripheral lymphoid organs of tolerant mice by bystander suppression. more...

The rice weevil Sitophilus oryzae is considered the major problem in stored rice in Sri Lanka. Due to the adverse effects of pesticide usage, research on the re-evaluation and use of many ethnobotanicals as alternative storage pest control agents has been intensified. Although plant materials with insecticidal properties provide small-scale farmers with a locally available, eco-friendly and inexpensive method of control of storage insect pests, lack of understanding and knowledge prevent their widespread application. The present study was therefore undertaken to investigate the effectiveness of the botanical Olax zeylanica in controlling infestations of the rice weevil with the view of fulfilling this lack.In two separate bioassays, contact/feeding and fumigant toxicity of powdered leaves of O. zeylanica were tested against 1-7 days old adults under laboratory conditions. All experiments were conducted using a no-choice bioassay apparatus. Contact/feeding toxicity was tested by directly exposing weevils to 1.0, 3.0, 5.0 and 7.5g of leaf powder mixed with 100g of rice grains while fumigant toxicity was evaluated by using the same doses where weevils were exposed to fumes emitted from the leaf powders.In both bioassays 100% mortality of the weevils was observed within 18 hours of exposure to 3.0, 5.0 and 7.5g doses of leaf powder. Percentage weevil mortality in treated rice tested with three doses of leaf powder at all the time intervals (except for 1.0g) was significantly higher (p < 0.05) than that of the corresponding control. No Contact/feeding toxicity was recorded when weevils were directly exposed to 1.0g leaf powder whereas only 14% weevil mortality was observed even after 24hours of exposure to fumes of leaf powder. Results also revealed that weevil mortality increased both with increasing dose and time of exposure. It is of interest to note that in both bioassays a 100% weevil mortality was obtained after 18 hours of exposure to a lower amount of leaf powder such as 3.0g, when the adults were either directly exposed to the leaf powder or to its fumes. Moreover, LD50 values of 2.55g and 2.08g for leaf powders obtained after 12 hours of exposure to insects in contact/feeding toxicity test and fumigation test respectively indicated that leaf powder is more toxic to weevils when they were in direct contact with it. Findings of the present study bears out the exceptionally high efficacy of O. zeylanica leaves applied directly mixed with the food material or introduced as a fumigant to suppress weevil infestations in stored grains and strengthen the possibility of using this plant as an alternative to synthetic chemicals in storage pest management. more...

OBJETIVO: Descrever o processo de revisão e de integração curricular de um programa de pediatria por meio da criação de uma matriz de competências referenciada nas Diretrizes Curriculares Nacionais. MÉTODOS: Estudo quali-quantitativo de intervenção que avaliou a percepção de estudantes e docentes em relação ao currículo existente (grupos focais e entrevistas semiestruturadas). Discutiram-se os resultados em oficinas de desenvolvimento docente, o que propôs uma matriz baseada em competências para todo o programa de pediatria do 3º ao 6º ano. O novo currículo foi aprovado, implementado e reavaliado após 6 meses. RESULTADOS: Doze estudantes (12%) do 3º ao 6º ano participaram dos grupos focais, e 11 dos 14 professores (78,5%) responderam ao questionário. A maioria referiu falta de integração entre as disciplinas, desconhecimento dos objetivos de aprendizagem dos estágios, poucas oportunidades de práticas e avaliação predominantemente teórica. Nas oficinas de capacitação, foi criada uma matriz curricular integrada por competências após a pactuação entre professores da pediatria e da saúde coletiva. A matriz destacava a competência geral, os objetivos de aprendizagem, oportunidades disponíveis para aprendê-los e o sistema de avaliação. Após 6 meses, 93% (104/112) dos alunos e 79% (11/14) dos professores relataram que percebiam maior integração do programa e destacaram a incorporação da avaliação de desempenho clínico. CONCLUSÃO: A construção coletiva da matriz curricular por competências levou à maior satisfação de docentes e discentes com a nova proposta que, após a implementação, foi percebida como integradora de conteúdos e práticas de ensino da pediatria, tendo qualificado a avaliação de desempenho clínico. more...

JUSTIFICATIVA E OBJETIVOS: Adição de clonidina subaracnóidea (±-agonista) prolonga a ação analgésica da combinação sufentanil e bupivacaína isobárica em analgesia combinada para o trabalho de parto ¹. O objetivo deste estudo foi comparar a qualidade de analgesia e a prevalência de efeitos colaterais após a adição de clonidina subaracnóidea à solução anestésica em gestantes durante trabalho de parto. MÉTODO: Após aprovação da Comissão de Ética, 22 gestantes em trabalho de parto receberam aleatoriamente no espaço subaracnóideo 2,5 mg de bupivacaína hiperbárica 0,5% (grupo CLON/HIPER; n = 11) ou 2,5 mg de bupivacaína isobárica 0,5% (grupo CLON/ISO; n = 11) em associação ao sufentanil 2,5 µg e à clonidina 30 µg. A dor avaliada pela Escala Analógica Visual, a freqüência cardíaca e a pressão arterial média foram estudadas a cada 5 minutos nos primeiros 15 minutos e, a seguir, a cada 15 minutos até o nascimento. Foi avaliada a prevalência de efeitos colaterais (náusea, vômito, prurido e sedação). O estudo foi encerrado no momento em que foi necessária complementação analgésica peridural (dor > 3 cm) ou ao nascimento. A análise estatística foi realizada pelos testes t de Student, Qui-quadrado, Fisher e ANOVA de duas vias para medidas repetidas, considerando como significativo p < 0,05. RESULTADOS: Os grupos CLON/HIPER e CLON/ISO foram semelhantes com relação a dados antropométricos, duração da analgesia (70,9 ± 32,9 vs. 85,4 ± 39,5), freqüência cardíaca, ocorrência de prurido, sedação, náusea e vômitos. No grupo CLON/ISO houve diminuição significativa da pressão arterial média com relação ao grupo CLON/HIPER nos momentos 15, 30 e 45 minutos (p < 0,05). CONCLUSÕES: Nas condições estudadas, a adição de clonidina em baixa dose (30 ¼g), associada ao sufentanil, determinou maior ocorrência de hipotensão quando administrada com soluções isobáricas de anestésico local. El análisis estadístico fue realizado a través de los tests t de Student, Chi-cuadrado, Fisher y ANOVA de dos vías para medidas repetidas, considerando como significativo p < 0,05. RESULTADOS: Los grupos CLON/HIPER y CLON/ISO fueron similares con relación a los datos antropométricos, duración de la analgesia (70,9 ± 32,9 vs. 85,4 ± 39,5), frecuencia cardíaca, incidencia de prurito, sedación, náusea y vómitos. En el grupo CLON/ISO hubo una disminución significativa de la presión arterial promedio con relación al grupo CLON/HIPER en los momentos 15, 30, y 45 minutos (p < 0,05). CONCLUSIONES: En las condiciones estudiadas, la adición de clonidina en baja dosis (30 ¼g), asociada al sufentanil, determinó una mayor incidencia de hipotensión cuando se administró con soluciones isobáricas de anestésico local. Con relación a los demás efectos colaterales, las soluciones hiperbáricas e isobáricas se comportan de manera similar.BACKGROUND AND OBJECTIVES: The addition of subarachnoid clonidine (α-agonist) prolongs the analgesia produced by the combination of sufentanil and isobaric bupivacaine in combined labor analgesia¹. The objective of this study was to compare the quality of analgesia and the prevalence of side effects after the addition of subarachnoid clonidine to the anesthetic solution in labor analgesia. METHODS: After approval by the Ethics Commission, 22 pregnant women in labor were randomly assigned to the subarachnoid administration of either 2.5 mg of 0.5% hyperbaric bupivacaine (CLON/HYPER Group; n = 11) or 2.5 mg of 0.5% isobaric bupivacaine (CLON/ISO Group; n = 11) associated with 2.5 µg of sufentanil and 30 µg of clonidine. Pain, evaluated by the Visual Analogue Scale, heart rate, and mean arterial pressure were assessed every 5 minutes during the first 15 minutes, and then every 15 minutes afterwards until delivery. The prevalence of side effects (nausea, vomiting, pruritus, and sedation) was evaluated. The study was terminated whenever the patient needed supplemental epidural analgesia (pain > 3) or upon delivery of the fetus. The Student t test, Chi-square test, Fisher exact test, and two-way ANOVA for repeated measurements were used in the statistical analysis and a p < 0.05 was considered significant. RESULTS: Anthropometric data, duration of analgesia (70.9 ± 32.9 vs. 85.4 ± 39.5), heart rate, and the incidence of pruritus, sedation, nausea, and vomiting were similar in both groups. Mean arterial pressure was significantly lower in the CLON/ISO Group than in the CLON/HYPER Group at 15, 30, and 45 minutes (p < 0.05). CONCLUSIONS: Under the conditions of the present study, the association of a small dose of clonidine (30 µg) with sufentanil caused a higher incidence of hypotension when the isobaric solution of the local anesthetic was used. Macrophages harvested from the peritoneal cavities of rats release a neutrophil chemotactic factor (MNCF) in response to stimulation with Gram-negative bacterial lipopolysaccharide (LPS). MNCF has been shown to be active in rats treated with dexamethasone, a glucocorticoid that usually inhibits the neutrophil migration induced in this species by interleukin (IL)-1, tumour necrosis factor alpha (TNFα), IL-8, C5a and leukotriene B4 (LTB4). Here we report that macrophages harvested from peritoneal cavities of mice, and stimulated in vitro with LPS, also release a factor that induces neutrophil migration in dexamethasone-treated animals. This chemotactic activity was neutralized by the incubation of the LPS-stimulated macrophage supernatants with a purified polyclonal IgG anti-mouse TNFα. In addition, significant amounts of TNF were detected in the supernatants. The neutrophil migration induced by intraperitoneal administration of recombinant murine TNFα was also unaffected by pretreatment of the mice with dexamethasone. Among the cereals, rice is the most important staple food supplying energy requirements for mostof the worlds’ population. However during storage a loss of about 10-20% rice grains occurs due to storedgrain pests. Repellents are considered as the best source of protection against insect attack upon storedproducts as they have potential for the exclusion of stored product pests from grain, and therebypreventing insect feeding and oviposition on food materials. Various plant materials have been utilizedeffectively through time as safe and ecofriendly insect pest control measures due to their repellentactivity. The aim of the present study was to investigate the potential of powdered leaves and leaf extractsof Olax zeylanica as repellents against the rice weevil, Sitophilus oryzae. All the experiments were carriedout under laboratory conditions using 1-7 day old unsexed adults. Four different doses (1.0g, 3.0g, 5.0gand 7.0g).of powdered leaves were tested for fumigant repellency in a dual-choice bio-assay apparatus.Repellent action of leaf extracts was evaluated by means of an area preference test using methanol,ethanol and n- hexane as solvents. Repellent effect of powdered leaves against the adult rice weevils wasfound to be significantly high (P< 0.05) at all doses. The highest repellent effect was produced by 7.0g ofleaf powder resulting in repellency of 97%, while the lowest dose (1.0g) also elicited more than 50%repellency in weevils indicating a very strong repellent action of the powdered leaves. In comparison,methanol extract of leaves produced the highest repellent effect (96%) on weevils whereas n-hexaneextract elicited the lowest. Nevertheless, at higher concentrations all three extracts produced more or lesssignificantly similar repellent effect on the weevils. The findings of the present study suggest that certainactive materials of Olax zeylanica leaves have potential to act as a grain protectant and may be exploitedfor the control of Sitophilus oryzae in rice storage in an environment-friendly way. The molecular mechanisms of plant recognition, colonization, and nutrient exchange between diazotrophic endophytes and plants are scarcely known. Herbaspirillum seropedicae is an endophytic bacterium capable of colonizing intercellular spaces of grasses such as rice and sugar cane. The genome of H. seropedicae strain SmR1 was sequenced and annotated by The Paraná State Genome Programme--GENOPAR. The genome is composed of a circular chromosome of 5,513,887 bp and contains a total of 4,804 genes. The genome sequence revealed that H. seropedicae is a highly versatile microorganism with capacity to metabolize a wide range of carbon and nitrogen sources and with possession of four distinct terminal oxidases. The genome contains a multitude of protein secretion systems, including type I, type II, type III, type V, and type VI secretion systems, and type IV pili, suggesting a high potential to interact with host plants. H. seropedicae is able to synthesize indole acetic acid as reflected by the four IAA biosynthetic pathways present. A gene coding for ACC deaminase, which may be involved in modulating the associated plant ethylene-signaling pathway, is also present. Genes for hemagglutinins/hemolysins/adhesins were found and may play a role in plant cell surface adhesion. BACKGROUND: Valproic acid (VPA) is a potent anticonvulsant that inhibits histone deacetylases. Because of this inhibitory action, we investigated whether VPA would affect chromatin supraorganization, mitotic indices and the frequency of chromosome abnormalities and cell death in HeLa cells. METHODOLOGY/PRINCIPAL FINDINGS: Image analysis was performed by scanning microspectrophotometry for cells cultivated for 24 h, treated with 0.05, 0.5 or 1.0 mM VPA for 1-24 h, and subjected to the Feulgen reaction. TSA-treated cells were used as a predictable positive control. DNA fragmentation was investigated with the TUNEL assay. Chromatin decondensation was demonstrated under TSA and all VPA treatments, but no changes in chromosome abnormalities, mitotic indices or morphologically identified cell death were found with the VPA treatment conditions mentioned above, although decreased mitotic indices were detected under higher VPA concentration and longer exposure time. The frequency of DNA fragmentation identified with the TUNEL assay in HeLa cells increased after a 24-h VPA treatment, although this fragmentation occurred much earlier after treatment with TSA. CONCLUSIONS/SIGNIFICANCE: The inhibition of histone deacetylases by VPA induces chromatin remodeling in HeLa cells, which suggests an association to altered gene expression. Under VPA doses close to the therapeutic antiepileptic plasma range no changes in cell proliferation or chromosome abnormalities are elicited. Inhibitory effect upon neutrophil migration to the inflammatory focus was previously detected in the cell-free incubation fluid of lipopolysaccharide (LPS)-stimulated macrophage monolayers. In the present study we showed that the neutrophil recruitment inhibitory activity from this supernatant was mainly detected in a fraction (P2) obtained by gel filtration chromatography on Sephacryl S-300. P2 fraction was able to inhibit ‘in vivo’ neutrophil emigration induced by different inflammatory stimuli, but it did not affect ‘in vitro’ neutrophil chemotaxis induced by FMLP. When injected intravenously, P2 inhibited oedema induced by carrageenin or immunological stimulus but not the oedema induced by dextran, thus affecting cell-dependent inflammatory responses. It was observed that P2 also induced neutrophil migration when injected locally in peritoneal cavities. This activity was significantly reduced by pretreatment of the animals with dexamethasone. Cytokines, such as IL-8 and TNF-α that are known to exhibit inhibitory effect upon neutrophil migration, were not detected in P2 fraction by highly sensitive assays. Este trabalho teve por objetivo avaliar o efeito da idade e da suplementação oral com acetato de DL-alfa-tocoferol sobre os níveis séricos de vitamina E e sobre o metabolismo oxidativo dos neutrófilos de bezerros da raça Jersey do nascimento aos 45 dias de idade. Foram utilizados 14 bezerros divididos em dois grupos de sete animais cada. O grupo tratamento recebeu 2g de acetato de DL-alfa-tocoferol, por via oral, diariamente, até os 45 dias de vida e o controle não recebeu nenhum tipo de suplementação para esta vitamina. Foram colhidas amostras de soro sanguíneo ao nascimento, aos cinco, 10, 20, 30 e 45 dias de idade, para: determinação sérica da vitamina E e para a avaliação da função dos neutrófilos. Concluímos que a suplementação oral com acetato de DL-alfa-tocoferol, na dose de 2 g/dia/VO, do nascimento aos 45 dias de idade aumentou significativamente (p≤ 0,05) os níveis séricos de vitamina E em bezerros. Os menores percentuais de neutrófilos redutores foram detectados logo após o nascimento e os maiores aos 45 dias de idade. A suplementação com vitamina E não alterou significativamente a função oxidativa dos neutrófilos, mas os valores superiores obtidos para os animais do grupo tratado, para o teste estimulado, sugerem uma ação neutrófilica mais potente diante uma infecção natural nesses animais. more...
